    Notes: Abstract Feeding behaviour of five species of cereal aphids in wheat seedlings differing in hydroxamic acid (Hx) levels, was monitored via electrical penetration graphs (EPG). Aphid species could be grouped as sensitive to the feeding deterrent effect of Hx in the seedlings (Rhopalosiphum padi, Schizaphis graminum, Sitobion avenae, andMetopolophium dirhodum) or insensitive to them (Rhopalosiphum maidis). However, when feeding behaviour was studied in artificial diets containing Hx, all species were equally sensitive to Hx. The behavour ofR. maidis was further compared with that ofR. padi through detailed EPG analysis. It was found that the insensitivity ofR. maidis to Hx in seedlings may be due to a feeding strategy avoiding contact with the compounds by decreasing the number of cellular punctures in live tissues other than sieve elements during its way to the phloem.

    Notes: Abstract Hessian flyMayetiola destructor (Say) larvae are able to obtain food from their host plant without inflicting mechanical damage to the plant surface, apparently by secreting substances which elicit release of nutrients from plant cells surrounding the feeding site. Cells of fully susceptible plants retain their normal appearances, while in resistant plants extensive areas of cellular collapse occur. These responses indicate that hypersensitivity is the basis of wheat's resistance to the Hessian fly. The fly's feeding mechanism more closely resembles that of a pathogen than of a phytophagous insect; correspondingly, both the genetic relationship and resistance mechanism of the host plant to the parasite are of the sorts commonly associated with bacterial and fungal pathogens.

    Notes: Abstract The effect of experience on the responsiveness of the aphidiid parasitoidLysiphlebus testaceipes (Cresson) (Hymenoptera: Aphidiidae) to host-associated cues was investigated using a wind-tunnel bioassay. Naive females were able to discriminate between uninfested wheat (Triticum aestivum L.) and wheat infested withSchizaphis gramimum (Rondani) (Homoptera: Aphididae), but oviposition experience significantly increased the parasitoid's propensity to respond to aphid-infested plants with upwind, targeted flight. The behavioural change associated with such experience was acquired rapidly (within five minutes) and persisted for at least 24 h. The parasitoid could be successfully conditioned to associate a novel odour with the presence of hosts, suggesting that the increase in response to aphid-infested plants which occurs as a result of experience is probably due to associative learning of olfactory cues from the plant-aphid complex.

    Notes: Abstract The Russian wheat aphid, Diuraphis noxia (Mordvilko), is one of the most important aphid pests of wheat, Triticum aestivum L., worldwide. Among the various pest management options, plant resistance is an economical management tactic to control D. noxia in cereal crops such as wheat. Researchers have identified D. noxia resistant germplasm and it has been incorporated into wheat. This study compared D. noxia resistance between the ‘Betta’ wheat isolines Betta-Dn1, Betta-Dn2, and Betta-Dn5 and their corresponding donor gene plant introduction (PI) lines PI 137739 (Dn1), PI 262660 (Dn2), and PI 294994 (Dn5). Although the Betta isolines and PI lines showed D. noxia resistance when compared with Betta wheat, the degree of resistance in the isolines to D. noxia was different from their corresponding PI donors. Aphid number, aphid fecundity, and biomass per aphid were not different between Betta-Dn1 and PI 137739 or Betta-Dn2 and PI 262660; however, the same parameters were significantly lower on PI 294994 compared with Betta-Dn5. This indicated that aphid resistance in PI 137739 and PI 262660 was probably governed by a single dominant gene, while the resistance in PI 294994 was controlled by more than one gene. Additionally, plant biomass reduction was aphid density dependent, which suggested that use of appropriate aphid infestation level is important when using plant biomass reduction as an indicator of resistance. Plant resistance categorization showed that there was no detectable difference in antixenosis among the seven lines evaluated. However, the higher aphid fecundity observed on PI 262660 compared with PI 137739 and PI 294994, in addition to no significant differences among the three PIs in plant biomass reduction, suggested PI 262660 was a tolerant line, while PI 137739 and PI 294994 were antibiotic lines. Plant tolerance could not be elucidated among the three Betta isolines using aphid fecundity and plant biomass reduction as indicators.

    Notes: Abstract The effect of cereal leaf surface wax on Diuraphis noxia (Mordvilko), the Russian wheat aphid, probing behavior and nymphoposition was evaluated. Ultrastructure of leaf epicuticular wax from wheat (Triticum aestivum L.) c.v. ‘Arapahoe’ and ‘Halt’ was different from barley (Hordeum vulgare L.) c.v. ‘Morex’, and oat (Avena sativa L.) c.v. ‘Border’. Both wheat cultivars had similar rod-shaped epicuticular wax, while barley and oat plants had flakes. The chemical composition comparison of gas chromatograms also indicated that the extract of the two wheat cultivars had similar pattern of peaks, while the barley and oat leaves had similar peaks. Cereal variety significantly affected aphid probing behavior (P 〈 0.05), but wax removal using ethyl ether swab did not (P 〈 0.05). Aphids initiated significantly more probes on Border oat leaves than on Morex barley irrespective of wax removal, although total probing duration per aphid was not significantly different among the four cereals examined. Accumulative salivation duration per aphid on oat leaves with wax was significantly longer than other cereal leaves with wax, while accumulative ingestion duration per aphid on Arapahoe wheat and Morex barley was significantly longer than on oat. Nymphoposition of D. noxia on cereal leaves maintained on the benzimidazole-agar medium showed that aphids produced a greater number of nymphs on Morex barley and less on Border oat leaves, although wax removal did not affect aphid nymphoposition. Removal of leaf epicuticular waxes from the 4 cereal genotypes using ethyl ether swab indicated that the influence of wax on plant resistance to D. noxia probing and reproduction was limited. Morex barley was the most favorable, while Border oat was the least favorable cereal host of D. noxia.

    Notes: Abstract Extended sieve element salivation (E1 waveform in the electrical penetration graph) is a characteristic activity during early sieve element punctures, particularly in resistant plants. In order to explore a chemically-mediated mechanism of resistance associated with sieve element salivation, we compared the pattern of feeding behaviour of the aphid, Sitobion fragariae (Walker), on two cultivars of the wheat Triticum aestivum L., with different concentrations of hydroxamic acids (Hx). During 24 h of electronic monitoring, aphids dedicated over 50% of the total time to phloem ingestion from the sieve elements. Total time allocated to E1 in the experiment, time to first E1 within the experiment, time allocated to E1 before a sustained phloem ingestion (E2) and the contribution of sieve element salivation to the phloem phase (E1/[E1+E2]) were significantly higher in the high-Hx cultivar. The increased salivation in plants with higher contents of Hx suggests the existence, at least in this system, of a chemically-mediated sieve element constraint.

    Notes: Abstract We have used hybridization analysis to detect ancient DNA in wheat seeds collected from three archaeological sites in Europe and the Middle East. One of these samples, carbonizedT. spelta dated to the first millennium BC, has yielded PCR products after amplification with primers directed at the leader regions of the HMW (high molecular weight) glutenin alleles. Sequences obtained from these products suggest that the DNA present in the Danebury seeds is chemically damaged, as expected for ancient DNA, and also indicate that it should be possible to study the genetic variability of archaeological wheat by ancient DNA analysis. Finally, we describe a PCR-based system that enables tetraploid and hexaploid wheats to be distinguished.

    Description / Table of Contents: Zusammenfassung 32 abgesetzte männliche Albinoratten wurden in 4 Gruppen eingeteilt, um die Wirkung einer lysinarmen Weizendiät (AW), einer Weizendiät mit 0,4 % Lysin (LW) oder 0,2 % Carnitin (CW) sowie einer Caseindiät auf den Stoffwechsel von Lipiden in verschiedenen Geweben zu untersuchen. Nach 8 Wochen Fütterung der verschiedenen Diäten unter Anwendung der „paired feeding technique“ wurden Veränderungen in den Gesamtlipiden, den Lipidbestandteilen, den einzelnen Fettsäuren und dem Lipidgehalt der Mitochondrien des Herzens, der Skelettmuskeln, der Lungen und des Fettgewebes der Tiere bestimmt. Die lysinarme Weizendiät (AW) bewirkte eine Lipidanreicherung (vor allem an Acylglyzerinen) im Herzen, in der Leber, den Skelettmuskeln sowie eine Lipidabnahme im Fettgewebe. Die Diäten mit 0,4 % Lysin (LW) oder 0,2 % Carnitin (CW) zeitigten die entgegengesetzte Wirkung, wobei CW wirkungsvoller erschien als LW. LW und CW erhöhten die relativen Anteile von C14∶0-, C16∶0- und C16∶1-Fettsäuren und verkleinerten diejenigen von C18∶1-, C18∶2- und C18∶3-Fettsäuren, während die Anteile unter der AW- und Caseindiät ab- bzw. zunahmen. Die Fettsäurezusammensetzung des Fettgewebes war bei allen Gruppen gleich. Die AW-Diät vergrößerte die relativen Anteile von C14∶0- und C20∶4- und verringerte die von C16∶0-, C16∶1- und C18∶3-Fettsäuren in den Lungen. Die AW-Ergänzungsdiäten verringerten die relativen Anteile von C16∶0-, C16∶1-, C18∶3-sowie auch der C18∶1-Fettsäuren. Der Lipidgehalt der Mitochondrien von Leber, Herz, Skelettmuskeln und Lunge verringerte sich unter der AW-Diät und erhöhte sich unter den LW- und CW-Diäten.
    Notes: Summary 32 weanling male albino rats were divided into 4 groups to study the effects of lysine-deficient wheat diet (AW) and AW supplemented with either 0.4 % lysine (LW) or 0.2 % carnitine (CW) as compared to casein diet on metabolism of lipids in various tissues. LW, CW and casein diet groups were pair-fed with AW group. Changes in total lipids, lipid components, individual fatty acids, mitochondrial content in liver, heart, skeletal muscles, lungs and adipose tissue were determined after 8 weeks of feeding. AW diet resulted in accumulation of lipids (mainly acylglycerols) in heart, liver, skeletal muscles and depletion in adipose tissue. The LW and CW diets reversed the effects of AW diet, the CW being more effective than LW diet. The LW and CW diets increased the relative proportion of C 14∶0, C 16∶0, C 16∶1 and decreased that of C18∶1, C18∶2, C18∶3 fatty acids which were decreased and increased, respectively, on the AW and casein diets. The fatty acids composition of adipose tissue was the same in all the groups. The AW diet increased the relative proportions of C 14∶0, C 20∶4 and decreased that of C 16∶0, C 16∶1, C 18∶3 fatty acids in the lungs. Supplemented AW diet decreased the relative proportions of the former group and increased that of the later group including C 18∶1 fatty acid also. The mitochondrial content of liver, heart, skeletal muscles and lungs was decreased on AW and reversed on LW and CW diets.

    Notes: Abstract Wheat (cv Chinese Spring) tissues were transformed using Agrobacterium tumefasciens and a new plasmid modular vector, pMVTBP. We constructed pMVTBP with unique restriction sites connecting (1) the CaMV 35S promoter, (2) a Kozak sequence, (3) the FLAG epitope, (4) the (His)6 epitope, (5) a coding region (for wheat TATA Binding Protein, wTBP) and (6) the CaMV 35S 3′UTR. This vector thus allows easy exchange of different regulatory or coding sequences. Explants of either germinating mature seeds, or immature embryos, were induced to callus for up to two weeks, treated with virulence-induced bacteria for one hour, then regenerated into plantlets. Transient expression of a GUS reporter gene, assayed at about one week, occurred in 10–12% of calluses. Expression of the FLAG-tagged wTBP was also detected, by immunostaining. Stable expression, by selective growth on geneticin, and by GUS expression at about six weeks, occurred in 1–2% of calluses, quite comparable to that achieved by other methods.

    Notes: Abstract Samples of wheat (n = 25) and maize (n = 30) for animal consumption, collected in 1997 after harvest from western Romania, were analyzed by enzyme immunoassays for mycotoxin contamination. Toxins analyses included deoxynivalenol (DON), 3-acetylDON, 15- acetylDON, fusarenone X (FX), T-2 Toxin (T-2), diacetoxyscirpenol (DAS), zearalenone (ZEA), fumonisin B1 (FB1), aflatoxin B1 (AFB1), ochratoxin A (OA), and citrinin (CT). DON and acetylDONs were the major contaminants in wheat (100%) and maize (46%). Median values for DON, 3-acetylDON, and 15-acetylDON were 880 μg kg-1, 66 μg kg- 1, and 150 μg kg-1 in wheat, and 890 μg kg-1, 180 μg kg-1, and 620 μg kg- 1 in maize, respectively. Additionally, 3,15-diacetylDON was detected in some samples by HPLC-EIA analysis. All samples were negative for FX (〈150 μg kg-1). T-2 was found in wheat (n = 6) and maize (n = 1) at levels between 13 and 63 μg kg- 1. DAS (2.6 μg kg-1) was found in one maize sample. ZEA occurred in all wheat and in four maize samples, median values were 10 μg kg-1 and 250 μg kg-1, respectively. One maize sample contained FB1 (140 μg kg-1). All samples were AFB1-negative (〈4 μg kg-1). OA was found in one wheat sample (37 μg kg- 1), CT was found in one maize sample (580 μg kg- 1). This first reported natural occurrence of a range of mycotoxins in Romanian feeding stuff shows that DON and acetyl DONs may be present at levels which may affect animal production.

    Notes: Abstract Eleven-kilogram parcels of HY-320 wheat, a cultivar of the new Canada Prairie Spring class, were kept at 15 and 19% initial moisture contents (IMC) in simulated storage in a Manitoba farm granary for 60 weeks to determine biotic and abiotic changes and mycotoxin production. Ochratoxin A reached a maximum of 0.24 ppm by week 20 in the 19% IMC wheat, but was absent in the 15% IMC wheat; no other mycotoxins were detected. Temperature, moisture content, O2 and CO2 levels, fat acidity values, seed germination, microfloral incidence and abundance, and the presence of other mycotoxins were monitored. Principal component analysis of all variables showed that the first principal components accounted for 32–41% of the system variability, and contained the ochratoxin A variable. Ochratoxin A was produced in moist grain that had decreased seed germination andAltermaria activity, and high fungal activity byPenicillium andAspergillus versicolor. Compared to other stored cereals previously studied, HY-320 wheat would be ranked in a low-risk category for mycotoxin formation, based on the ochratoxin A levels observed.

    Notes: Abstract Sixty-four wheat samples from Spanish flour factories were screened for patulin and patulin-producing moulds. None of them was found to contain any patulin, whereas samples experimentally contaminated with this toxin proved it to be highly unstable. On the other hand, Penicillium griseofulvum was the only in vitro patulin-producing species found (19 samples). Mould growth in the samples was investigated by using yeast-sucrose medium (YES) and high-performance liquid chromatography (HPLC) to measure the amounts of toxin produced during 40 day's incubation at 20 and 28°C. The highest yield rate of patulin was obtained between the 20th and 30th day of incubation; such a rate, however, was very low throughout the vigorous growth phase, during the first 20 days of incubation. The more appropriate temperature for incubation and patulin production was 28 °C. We also investigated the influence of other incubation conditions in the yield and found stationary dark cultures to be more efficient that shaken or fermentation cultures in YES medium. The best patulin yield achieved was 11.9 mg in the culture broth and 6.3 mg in the mycelium from 100 ml of medium.

    Notes: Abstract Wheat for human consumption (140 samples) was collected after harvest from all regions of Bulgaria. The 1995 crop year was characterized by heavy rainfall in the spring and summer months. The internal mycoflora of wheat samples was dominated by Fusarium spp. and Alternaria spp., and storage fungi were rarely present. The samples were analysed for contamination with Fusarium mycotoxins deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-AcDON), 15-acetyldeoxynivalenol (15-AcDON), T-2 Toxin (T-2), diacetoxyscirpenol (DAS), and zearalenone (ZEA), using enzyme immunoassay methods. DON and ZEA were the predominant toxins, with a contamination frequency of 67% and 69%, respectively. The average levels of these toxins in positive samples were 180 μg/kg (DON) and 17 μg/kg (ZEA), maximum concentrations were 1800 μg kg−1 and 120 μg kg−1, respectively. Acetyl derivatives of DON, namely 3-AcDON and 15-AcDON, were found in 2.1 % and 0.7% of the samples, at at maximum level of about 100 μg kg−1. Only one sample was positive for T-2 (55 μg/kg), DAS was not detected. This is the first report about the natural occurrence of a range of Fusarium mycotoxins in wheat for human consumption in Bulgaria.

    Notes: Abstract In many developing countries, fertilizer recommendations must be made in the absence of plant and soil analyses. When the region is variable in terms of soils, weather, and magnitude of response to fertilizer, a recommendation is likely to involve a high degree of risk for the farmer. Quantification of such risk is key to developing appropriate recommendations for the farmer. However, most methodologies generally used in analyzing fertilizer trials do not allow adequate quantification, especially as a continuous function, of the risk associated with a given recommendation. Three years of on-farm trials conducted in the High Valley of Mexico were used to evaluate different methodologies for generating N fertilization recommendations and their associated risk for wheat (Triticum aestivum) production in the absence of soil tests. When the traditional approach, using average yield responses or separate trial results, was used, an economic optimum was identified, but it was not possible to quantify the associated risk. In contrast, however, by using a combination of response surface methodology and simple probability analysis, the risk associated with any given recommendation was developed, even under the highly variable conditions of the study zone. The approach uses a treatment difference matrix (developed using average yield differences between a treatment and the zero N (0N) check) and its associated standard deviation over locations. From the matrix, an equation (being a function of N rate and relative grain:N price ratios) was developed that shows the probability of outperforming the 0N check for the economic optimum rate.

    Notes: Abstract In a field experiment on deep, yellow, sandy soil near Badgingarra, Western Australia, the residual value of superphosphate applied one and two years previously was measured relative to freshly-applied superphosphate using yields of narrow-leafed lupin (Lupinus angustifolius), barley and wheat. In addition, soil samples were collected for measurement of bicarbonate-extractable soil P. This was also used to estimate the residual value of the superphosphate. For lupins and wheat, and for bicarbonate-extractable soil P, the residual value decreased with increasing level of application. For barley grain, the residual value was not significantly affected by the level of application. The decrease in residual value of superphosphate with increasing level of application is attributed to increased leaching of applied phosphorus (P) down the profile of the sandy soils as the level of application increases. This may reduce subsequent plant yields due to the delay in seedling roots reaching the P in the soil during the crucial early stages of plant growth. For lupins, the relationship between yield and the level of superphosphate applied was markedly sigmoidal. The relationship for wheat and barley was exponential. Consequently, at suboptimal levels of P application, lupins required about two to three times more P than wheat or barley to produce the same yield. However, lupins required less P to achieve near-maximum yield.

    Notes: Abstract In a field experiment on a deep pale-yellow sand in a 600 mm per annum rainfall Mediterranean environment of south-western Australia, six levels of phosphorus (P) as superphosphate (O up to 546 kg P ha−1) were applied once only, to the soil surface, before sowing lupins (Lupinus angustifolius). The lupins were grown in a continuous arable cropping rotation with, in successive years, oats (Avena sativa), wheat (Triticum aestivum), lupins. Five such rotations were started in the experiment from 1985 to 1989. The experiment continued until the end of 1990. The relationship between lupin seed (grain) yields and the level of P applied was measured in the year of P application for five successive years (1985 to 1989). The relationship had the same general form but it varied between years, largely due to different maximum yields (yield plateaux) in each year. The residual value of superphosphate applied three years previously was measured for lupins on two occasions (1988 and 1989) relative to superphosphate applied in the current year. The residual values was different in the two years. The superphosphate applied three years previously was about 30% as effective as freshly applied superphosphate in 1988, and 12% as effective in 1989. At each harvest, the relationship between grain yield and the P concentration in the grain differed for different species. However, for each species at each harvest, the relationship was similar regardless of when the P was applied in the previous years. Thus each species had the same internal efficiency of P use curve, and yields varied only with P concentration in tissue. Bicarbonate-extractable soil P was determined on soil samples taken in mid-July of 1989 and 1990. These soil test values were related to grain yields at harvest. The relationship between yield and soil test values had the same general form but varied for different species within years and for each species between years. It also varied for each species within years depending on the year the P was applied.

    Notes: Abstract In a field experiment on a sandplain soil in a low rainfall (326 mm per annum) Mediterranean environment of south-western Australia, the effectiveness of superphosphate applied in 1986 was measured in three subsequent years relative to freshly-applied superphosphate each year, using grain (seed) yields of wheat (Triticum aestivum) and lupins (Lupinus angustifolius). The wheat and lupins were grown in rotation and both crops were grown each year starting in 1986. Bicarbonate-soluble phosphorus was determined on soil samples taken in mid June from where the P treatment was applied in 1986 only. These soil test values were related to the grain yields produced that year. For each level of superphosphate applied in 1986, soil test values decreased with increasing time from application. The relationship between grain yield and soil test values had the same general form within each year for both plant species, but varied between years. For both species, the effectiveness of superphosphate decreased by about 70–80% between the year of application and the first and second years after application, and by a further approximate 10% in the third year. The relationship between grain yield and the level of superphosphate applied became sigmoidal by 1989.

    Notes: Abstract The objective of the investigation was to examine whether there exist relationships between the optimum nitrogen fertilizer rate for winter wheat and soil nitrogen fractions extracted by electroultrafiltration (EUF) from autumn samples of the upper soil layer (0–30 cm). Optimum nitrogen fertilizer rates were derived from grain yield curves of field trials carried out with increasing nitrogen fertilizer rates on 19 different sites in 1985/86 and 1986/87. Most soils were luvisols derived from loess, two soils were brown earths and one a pararendzina. Total Nitrogen fertilizer rates were 0, 40, 80, and 120 kg N/ha applied twice before ear emergence. The final nitrogen rate at ear emergence was the same for all treatments, namely 60 kg N/ha. Optimum nitrogen fertilizer rates were derived from the grain yield curve fitted to a modified Mitscherlich equation. The optimum nitrogen fertilizer rates were correlated with the nitrogen fractions extracted by EUF. The regression equation thus obtained showed that NO 3 - , the organic N fraction (EUF Norg), and the EUF Norg-quotient each had a highly significant impact on the optimum nitrogen fertilizer rate. The higher the amounts of EUF-N extracted the lower the optimum nitrogen rate. Substituting the EUF Norg-fraction for total nitrogen concentration in the upper soil layer gave a poorer relationship between the optimum nitrogen fertilizer rate and the soil data. In absolute terms the EUF Norg-fraction had by far the greatest impact on calculating the optimum nitrogen fertilizer rate. The investigation shows that the EUF method is a suitable technique for the determination of available soil nitrogen from which optimum nitrogen fertilizer rates can be derived for winter wheat cultivated under soil and climatic conditions typical for cereal growing areas in central Europe.

    Notes: Abstract A technique for the application of the15N isotope dilution technique for the quantification of plant associated biological nitrogen fixation (BNF) was tested and applied to quantify the BNF contribution to two genotypes ofPhaseolus vulgaris. The technique makes use of sequential measurements of the15N enrichment of soil mineral N, and the uptake of labelled N by the “N2-fixing” plant, to simulate its uptake of soil N (the “soil to plant simulation” technique). The test was made with two non-N2-fixing crops (non-nodulating beans and wheat) and two bean genotypes (PR 923450 and Puebla 152), at two levels of N fertilizer addition (10 and 40 kg N ha−1), to compare the actual N uptake with that simulated from the soil and crop15N data. The simulation of the soil N uptake by the non-nod bean crop using this “soil to plant simulation” technique underestimated by 20 to 30% the true N uptake, suggesting that the mineral N extracted from soil samples taken from the 0–15cm layer had a higher15N enrichment than that N sampled by the roots of this crop. In the case of the wheat crop the simulation resulted in a much greater underestimation of actual N uptake. In general the results using this technique suggested that BNF inputs to the bean cultivars was higher than would be expected from the nodulation and acetylene reduction data, except for the early PR beans in the 40 kg N ha−1 treatment. In this case the total N and simulated soil N accumulation were well matched suggesting no BNF inputs. An allied technique (the “plant to plant simulation technique”) was proposed where the15N enrichrnent of soil mineral N was simulated from the data for total N and labelled N accumulation taken from sequential harvests of either of the non-N2 -fixing control crops. This was then utilized in combination with the labelled N uptake data of the other crop to simulate its soil N uptake. However, the results using either technique indicated that the wheat and non-nod or nodulating beans exploited pools of N in the soil with completely different15N enrichments probably due to differences in exploitation of the soil N with depth.

    Notes: Abstract Data was assembled from experiments on the fate of15N-labelled fertilizer applied to wheat (Triticum spp.) grown in different parts of the world. These data were then ranked according to the annual precipitation-evaporation quotient for each experimental location calculated from the average long-term values of precipitation and potential evaporation. Percentage recovery of15N fertilizer in crop and soil varied with location in accordance with the precipitation-evaporation quotient. In humid environments more15N fertilizer was recovered in the crop than in the soil, while in dry environments more15N fertilizer was recovered in the soil than in the crop. Irrespective of climatic differences between locations 20% (on average) of the15N fertilizer applied to wheat crops was unaccounted for at harvest. Most of the15N fertilizer remaining in the soil was found in the 0–30 cm layer. The most likely explanation of these differences is that wheat grown in dry environments has a greater root:shoot ratio than wheat grown in humid environments and, further, that the residue of dryland crops have higher C/N ratios. Both factors could contribute to the greater recovery of15N fertilizer in the soil in dry environments than in humid ones.

    Notes: Abstract Long-term field experiments play an important role in understanding the complex interactions of plants, soils, climate and management and their effects on sustainable crop production. A long-term fertilizer experiment with maize-wheat-cowpea (fodder) is in progress since 1971 at Punjab Agricultural University farm Ludhiana, India. The experimental result for the first 21 years showed that application of N alone or in combination with P did not produce as much maize and wheat grains as the application of N, P and K together. Eight years after the start of the experiment, the optimal levels of N, P and K application (100% NPK) were unable to sustain the similar (maize) yield level as before because of Zn deficiency. Whereas in FYM amended plots the Zn deficiency did not appear and the higher crop yields could be sustained. The chemical control of weeds could not sustain the maize productivity at the same level as the manual removal of weeds. It was concluded that the high level of crop production can be sustained with the application of N, P and K under intensive cropping system provided deficiency of any of the micronutrient does not crop up. The deficiency of Zn is most likely to occur in semi-arid light textured alluvial soils under intensive cropping without the addition of farm yard manure/organic manures. In maize based cropping systems, manual control of weeds may be preferred to the chemical one. Addition of FYM in conjunction with 100% NPK is most beneficial both from bio-physical and economic point of view.

    Notes: Abstract A greenhouse study was conducted to determine if soil pH affects the requirement for water-soluble P and the tolerance of water-insoluble impurities in TSP fertilizers. Two commercial TSP fertilizers were selected to represent a range in phosphate rock sources and impurities. Phosphate fertilizer impurities were isolated as the water-washed fraction by washing whole fertilizers with deionized water. TSP fertilizers with various quantities of water-soluble P (1.2 to 99% water-soluble P) were simulated by mixing the water-washed fertilizer fractions or dicalcium phosphate (DCP) with reagent-grade monocalcium phosphate (MCP). The fertilizers were applied to supply 40 mg AOAC available P kg−1 to a Mountview silt loam (fine-silty, siliceous, thermic Typic Paleudults). Wheat (Triticum aestivum (L.)) was harvested at 49 and 84 days after planting. Soil pH values at the final forage harvest were 5.4±0.16 and 6.4±0.15. At a soil pH of 5.4, the TSP fertilizers required only 37% water-soluble P to reach maximum yields while at pH 6.4 the fertilizers required 63% water-soluble P. Results of this study show that higher levels of water -insoluble P can be tolerated in TSP fertilizers when applied to acid soils. Phosphorus uptake was not affected by soil pH, but for the mixtures containing the fertilizer residues the source having the lowest level of Fe and Al had a higher relative agronomic effectiveness.

    Notes: Abstract In land use plans, fertilizer recommendations are indispensable to avoid soil nutrient depletion or soil water pollution. Nutrient relations of five cereals have been evaluated on the basis of a literature review with the aim of arriving at such fertilizer recommendations at regional level. Nutrients considered were nitrogen, phosphorus and potassium for millet, sorghum, maize, rice and wheat. The relevant nutrient relations are fertilizer nutrient application to nutrient uptake, and nutrient uptake to crop yield. In addition, post-anthesis nutrient uptake is considered. Subsequently, obtained results are used in simulation modelling exercises to calculate the time required to attain an equilibrium nutrient balance and to investigate the effect of erosion control and straw recycling. Although fertilizer requirements could be assessed for each of the five cereals, monitoring of nutrient supply from natural sources remains necessary. Moreover, research on fertilizer use should focus on improvement of fertilizer recoveries and multiperiod models for both N and P uptakes by crops to allow quantitative land use planning where the time scale is included.

    Notes: Abstract Winter wheat crops were grown with ostensibly adequate supplies of all soil nutrients in 1990 and 1991 with the aim of testing if late foliar supplements of K and N, applied at key development stages, could improve grain yield and grain N content. Foliar sprays of KNO3 solution, supplying up to 40 kg K ha−1 in total, at flag leaf unfolded, inflorescence completed and the watery-ripe stage of grain filling, had no effect on yield, yield components or grain N. Urea, supplying 40 kg N ha−1 at flag leaf unfolded, had no effects on grain yield and grain N in 1990, but in 1991 grain N was increased by 0.14% whilst yield was reduced by up to 0.6 t ha−1. Urea scorched flag leaf tips in both years. In 1990, the spring was very dry and foliar supplements might have been expected to have had an effect, but on this highly fertile soil all crop K and N requirements were met from the soil.

    Notes: Abstract Seven chemical extractants were tested for their relative performance to predict the response of wheat to Mn application in coarse textured alkaline soils of semi-arid region. Five out of the seven extractants were found to be promising for the estimation of critical level of available Mn in these soils, as the amount of Mn extracted by these extractants was positively and significantly correlated with relative grain yield as well as Mn uptake. The critical deficiency level of soil available Mn with 0.005 M DTPA, 0.02% hydroquinone, 0.02 N sodium pyrophosphate, 0.1N H3PO4 and 0.05N HCl+0.025N H2SO4 was 3.1, 13.8, 23.5, 5.3 and 17.8 mg kg-1 soil, respectively. The 1N ammonium acetate and 0.01M CaCl2 were found to be unsuitable extractants for these soils. Further field trials at eight locations with varying levels of Mn deficiency showed successive increase in the grain yield of wheat with foliar Mn application, emphasizing the need for Mn fertilization when wheat is grown on Mn deficient soils.

Soil application of 5.6 kg Zn ha−1 significantly increased the grain yield in deficient soils. Soil extractable Zn was significantly related with per cent grain response and absolute grain yield. Both the graphical and statistical methods of Cate and Nelson indicated the critical level to be 0.75 mg kg−1 soil of DTPA extractable Zn. This level gave a predictability value of 82 per cent.

    Notes: Abstract Three factorial experiments with four replications were conducted in a greenhouse to examine the effectiveness of gypsum, elemental sulfur (ES powder) and three S containing N fertilizers, viz., ammonium sulfate (AS), urea + ES, and Ureas (20% AS and 80% urea). All experiments were conducted twice in different years. In the first experiment with uncropped soil, the effects of soil type, leaching rate (2.3 and 6.9 mm water per day) and urea addition on sulfate leaching losses were studied. Leaching losses decreased in the order Ureas 〉 ammonium sulfate (AS) 〉 gypsum ≫ urea + ES. Increasing the leaching rate greatly increased sulfate losses from both soils. Losses were greater in the sandy Typic Hapludoll than in the clayey Oxic Paleustalf. Sulfate adsorption was found to decrease strongly with rising the pH in both soils. Hydrolysis of urea temporarily raised the pH of the soil, thereby increasing the sulfate leaching losses. In the second experiment the effects of S rate (0–65 mg per kg soil), split application and leaching rate (0 and 2.3 mm per day) on sulfate leaching losses and ‘apparent S recovery’ (ASR) by three successive cuts of ryegrass (Lolium perenne L.) were studied. Herbage yield more than doubled when S was applied. The effectiveness of the sulfate fertilizers was greater when S was split-applied than given all at once. With split applications the ASR decreased in the order: Ureas 〉 AS 〉 gypsum 〉 urea + ES 〉 ES powder. ES fertilizers were least effective, because the oxidation rate of ES to sulfate was clearly too slow. In the third experiment the effects of S rate (0–40 mg per kg soil) and split application on sulfate leaching losses and ASR in the grain of wheat (Triticum aestivum L.) were studied under leaching conditions (2.3 mm per day). Grain yield increased strongly due to S application. Split application greatly increased the effectiveness of the sulfate fertilizers and appeared to be an effective tool in satisfying the S need of the crop under leaching conditions. Again, ES fertilizers were least effective, because the oxidation rate of ES was too slow to meet the S demand of the crop. In all experiments leaching losses of sulfate from the ES fertilizers were smaller than from the sulfate fertilizers.

    Notes: Abstract In a field experiment on a sandplain soil in a low rainfall (326mm per annum) Mediterranean environment of south-western Australia, seven levels of single superphosphate, 0, 7.5, 10, 14, 19.5, 30 and 39 kg P ha−1, were placed at either 3, 5, 7, 9, 11 or 13 cm depth before sowing wheat (Triticum aestivum) at 3 cm. In a separate treatment, superphosphate was drilled with the seed (the normal practice). In the second year, the plots were sown with lupins (Lupinus angustifolius) at 3 cm depth with no additional superphosphate. In three separate treatments, superphosphate at 0, 14 and 39 kg P ha−1, was drilled with the lupin seed (the normal practice) on plots that had received no superphosphate in the first year. Yields of wheat and lupins were used as a measure of the effectiveness of the superphosphate placement treatments relative to the effectiveness of superphosphate drilled with seed of wheat (year 1) or lupins (year 2), to give relative effectiveness (RE) values in each of the two years. In the first year the RE of superphosphate was increased by about 20% when the fertilizer was placed 5 to 9 cm deep in the soil. In the second year, the RE of superphosphate for producing lupin grain was increased by about 30–60% where the fertilizer had been placed 5–13 cm deep in the previous year compared with freshly drilled 3 cm deep. The yield of wheat or lupins was closely related to the P content of plant tissue; each relationship was independent of the depth or year of superphosphate application.

    Notes: Abstract Using soils with an acidic subsurface layer, three glasshouse experiments were carried out to evaluate the effect of placement method and application rate of triple superphosphate (TSP) and North Carolina phosphate rock (NCPR) on dry matter (DM) yields. Time of application of NCPR on DM yield response of wheat was also studied. For Experiment 1, soil was collected in depth intervals of 0–2; 4–6; 6–8; and 8–10 cm from a red earth (chromic luvisol). The treatments included two P sources (TSP and NCPR), three placement methods (broadcast, banded or mixed into the subsurface layer, 6–8 cm), and six application rates. In this P deficient soil with an acidic subsurface layer, there was relatively little effect of application method of TSP on wheat yield responses. The maximum dry matter yield responses for broadcast, band and mix application methods was 30, 42 and 50 %, respectively. Responses to NCPR broadcast, band and mix methods were 20, 9 and 44 %, respectively. Mixing NCPR into to acidic subsurface layer produced yields similar to those from TSP although a higher application rate of P as NCPR was needed to achieve this outcome. Treatments for Experiments 2 and 3 were time of application of NCPR (0 and 30 days before sowing) and rate of application of NCPR (0 and 40 mg P/pot). In Experiment 2 (same soil as Experiment 1) application of NCPR prior to sowing, resulted in higher Colwell P concentration than when applied at sowing, but time of application had no effect on final DM yields. Experiment 3 used a red podzolic (chromic luvisol) soil which had a lower P-status, was more acid and had a lower exchangeable Ca2+ concentration than the red earth. Application of NCPR prior to sowing resulted in lower DM yield than when it was applied prior to sowing.

    Notes: Abstract Using15 N tracer technique, the fate and efficiency of nitrogen in supergranules of urea as compared with that in powdered urea were studied in rice fields. The results obtained show that supergranules of urea were characterized by the slight N loss and high N recovery as well as by delayed but long lasting fertilization effects. It follows that the supergranules should be applied earlier and at a lower rate as compared with powdered urea.

    Notes: Abstract Wheat was grown continuously in soil amended with 5 levels of superphosphate and with 4 levels of urea at 3 sites. The incidence and severity of take-all, caused byGaeumannomyces graminis var.tritici, declined with increasing rates of application of both superphosphate and urea. In both years, the severity of take-all on plants receiving neither superphosphate nor urea was about 40% while at the highest level of superphosphate and urea supply the take-all severity was approximately halved at 22%. There was an increase in grain yield in response to applied superphosphate and urea to the highest level of each nutrient. There was also an increase in the 1,000-kernal weights with superphosphate and urea fertilizer application.

    Notes: Abstract The phosphorus (P) requirement for grain production of different crop species (oats (Avena sativa), barley (Hordeum vulgare), triticale (xTriticosecale), narrow-leafed lupins (Lupinus angustifolius), and sandplain lupins (L. cosentinii) was compared with wheat (Triticum aestivum) in five field experiments on different lateritic soils in south-western Australia. Seven or eight levels of superphosphate were applied at the start of each experiment. The amount of P required to produce 70% (four experiments) or 90% (one experiment) of the maximum yield was used to compare P requirements. Large differences in the P requirements of the species were obtained. On P deficient soil in 3 experiments, oats required from 50 to 70% less P than wheat, but required 40% more P on a soil with a long history of superphosphate applications. Compared with wheat, in the year of P application, barley required 50% less P in one experiment, had similar P requirements in two experiments, and required 80% more P in another experiment. In the years after P application, barley required 20% less P in two experiments. On an acidic soil triticale required from 50% to 70% less P than wheat, but on less acidic soil it required 100% more P. In the year of P application, narrow-leafed lupins required 800% more P than wheat in one experiment, and 30% more P in the other experiment. In the year of P application, sandplain lupins required 70% less P than wheat in one experiment.

    Notes: Abstract Nineteen soils from the south east of the Province of Buenos Aires (Argentina) that had been fertilized with moderate amounts of P (10–40 kgP/ha) during the last 10 years were used to investigate the effect of time on the decline of P availability as measured by three soil tests (Bray 1, Bray 2, Olsen) and the null-point method. Differences in rates of P decline among soils and chemical methods were characterized by an exponential coefficient for time (b 2) in equations which describe the changes of the added P retained by the soil (Pr =ac b1 t b2). The rate of decline of P for the nineteen soils calculated for the soil test methods was ordered decreasingly as: null-point 〉 Olsen 〉 Bray 1 〉 Bray 2. The ability of the chemical methods for assessing the residual value of P for wheat growth (RV) was tested in a pot experiment on seven of the soils that differed in their individual rates of reaction with P. Differences between soils in the rate of reaction with P as measured in the laboratory by the null-point method and by the Olsen test were reflected in different residual values for P fertilizer for wheat plants. Thus the value ofb 2 for these methods was well correlated with the observed residual values. The soil properties commonly associated with the retention of P were not related to the value ofb 2 suggesting that more than one soil property may be involved in the measure ofb 2. The exponent for timeb 2 may be used as an index of the ability of the soil test to reflect the decline of P availability with time.

    Notes: Abstract The residual effectiveness of copper (Cu) applied 18 to 21 years previously was estimated for grain yield of wheat. In one field experiment, current levels of Cu fertiliser were applied and its effectiveness was compared to that of the same level of Cu applied previously. The effects of nitrogen (N) fertiliser on the Cu concentration in the youngest emerged blade and in the grain, as well as the effects of N levels on the grain yield of wheat, were also studied. Where the recommended level of Cu fertiliser had been applied previously, its residual effectiveness depended on the soil type. On the grey sands over clay and gravelly sands over clay, the residual Cu would last approximately 20 years where wheat is grown in rotation with a legume crop (Lupinus augustifolius L.) and where N fertiliser is applied at high levels (92 kg N ha−1). On the yellow brown sandy earths of the Newdegate district, the residual value was in excess of 30 years. When Cu levels in the soil are marginal, high levels of N applied to wheat crops grown on stubbles of legume crops (high soil N) could suffer from induce Cu deficiency which could reduce grain production. Critical concentrations of Cu in the youngest emerged blade of less than 1.2 mg Cu kg−1 at Gs50–59 would indicate Cu deficiency. Cu concentrations of less than 1.1–1.2 mg Cu kg−1 in the grain suggest that the wheat crop is marginally supplied with Cu. In both situations, Cu fertiliser needs to be applied before the next crop.

    Notes: Abstract A strain ofPenicillium bilaji Chalabuda (PB) has recently been commercially released as a seed inoculant to increase phosphorus (P) uptake by wheat (Triticum aestivum L.). The purpose of this study was to compare the effects of drill applied P (15 kg P ha−1) with PB seed inoculation on early growth, development, P uptake, and grain yield of ‘Stoa’ spring wheat at four sites in North Dakota. Fertilization with P consistently enhanced early season growth, main stem development, tillering and P uptake. Seed inoculation with PB had little or no effect on these traits. Phosphorus fertilization tended to increase common root rot severity (CRR, incited byCochliobolus sativus (Ito & Kurib) Drechs.), while PB inoculation had no effect. Grain yields were significantly increased by P fertilization at one location. Inoculation with PB also increased grain yield at this location. The reason why PB inoculation increased yield at this location is not evident, as plant growth and P uptake were not enhanced earlier in the season. Averaged across all four sites, PB inoculation increased wheat yields 66 kg ha−1, which is similar to averaged yield responses reported from the Prairie Provinces of Canada.

    Notes: Abstract A static model that predicts the nitrogen (N) fertilizer requirement of grain sorghum or wheat crops is described. Inputs required by the model are soil nitrate-N (kg ha−1) in the profile at sowing, total N (%) in the plough layer, available water in the profile at sowing (mm) plus rainfall during the growing season (mm). Output includes fertilizer N required for both maximum yield and optimum economic yield. The model was tested by using published field data from Nebraska and Kansas (U.S.A.), South Australia and Northern Territory (Australia), and Saskatchewan (Canada). The model was accurate when no fertilizer N was required, and when large amounts were required, but quantitative prediction of moderate requirements was only fair. Predictions for grain sorghum were better than for winter wheat, probably because total water use was a better predictor of yield potential for grain sorghum than for winter wheat. Further refinement for specific environments should make the model practical for dryland cereal crops.

    Notes: Abstract During primary infection by conidia ofErysiphe graminis f. sp.tritici, three mechanisms of resistance operate in first leaves of 8-day-old seedlings of both resistant and susceptible wheats. The first mechanism, operating at the penetration site, is responsible for the failure of penetrations attempted by primary germ tubes (PGT). The second mechanism is concerned with the abortion of haustoria in normal-appearing host cells. The third mechanism relates to the abortion of haustoria and the hypersensitivity of the penetrated host cells. With the inoculum-level of 19–24 conidia/mm2, the three mechanisms together prevented 89.3 % of the attempted penetrations by PGT from producing normal haustoria in resistant wheat Purdue 5752C1-7-5-1 and 37.4 % in the susceptible wheat Vermillion. The first mechanism accounted for the prevention of 73.3 % of the attempted PGT penetrations on Purdue 5752C1-7-5-1 and 36 % on Vermillion. The second mechanism was responsible for stopping 19 % of all the successful penetrations in Purdue 5752C1-7-5-1 and 0.8 % in Vermillion. The third mechanism accounted for the failure of 41 % of all the successful penetrations in Purdue 5752C1-7-5-1 and 1.4% in Vermillion. Thirty-six hours after inoculation, 10.7% of all the attempted PGT penetrations appeared to be developing normally in first leaves of 8-day-old seedlings of resistant wheat Purdue 5752C1-7-5-1 as compared to 62.6 % in the susceptible wheat Vermillion. This appears to be the first report showing the relative effectiveness of various mechanisms of resistance concerning any powdery mildew fungus.

    Notes: Abstract A field experiment was conducted for three consecutive winter crop seasons commencing in 1979–80 on the Typic Ustochrept of Pura to evaluate iron pyrites as S fertilizer. Four crops viz, wheat, chickpea, mustard and Egyptian clover were tested for their responsiveness to added pyrites. All the crops responded significantly to added pyrites. Mustard proved most sensitive to S deficiency in soil and wheat the least. Between the two legumes, Egyptian clover was more sensitive to S stress than chickpea. Average biomass production by Egyptian clover was highest followed by wheat, mustard and chickpea. Mustard and Egyptian clover required more S to achieve maximum biomass production compared with wheat and chickpea but they also recovered from the soil a large proportion of added S than wheat and chickpea. Addition of pyrites increased availability of S in soil. Pyrites enhanced mobilization of soil P and its utilization by the crops.

    Notes: Abstract A multisite field experiment was conducted to study the effect of topdressed Se-enriched Ca(NO3)2 (CN) and basal applied NPK on the selenium (Se) concentration in spring wheat (Triticum aestivum L.). Selenium was applied either through CN (at the rates of 0, 6.45, and 12.91 g Se ha−1) or NPK (5.83 g Se ha−1). Selenium concentration in wheat grains increased consistently with increasing rate of Se-enriched CN or NPK. However, the superiority of Se-enriched CN over NPK in raising the Se concentration in wheat grain depended on location and growth conditions. At the same rate both methods of Se-application were found to be equally effective in raising the Se concentration of wheat grains. The Se concentration of grain was generally higher in the light textured soils than in the medium to heavy textured soils. Without Se application, the Se-concentration in wheat grain was about 16µg kg−1 which is regarded insufficient to meet the Se requirement for Se in animal and human. Calcium nitrate enriched with 25 mg Se kg−1 (6.45 g Se ha−1) increased the Se concentration in wheat grain to a desired level.

    Notes: Abstract The bioluminescently marked Pseudomonas fluorescens strain 5RL, has been used previously to follow colonisation of soy bean roots (De Weger et al. [1991] Appl. Environ. Microbiol. 57:36-41). In the present paper the method has been further developed and optimized for wheat roots and it is used to get a quick overview of the colonisation patterns of many different root systems at the same time. Colonisation was followed on wheat plants grown in our gnotobiotic sand system (Simons et al., 1996. Mol Plant Microbe Interact 9: 600–607) and the following results were obtained. (i) A spatio-temporal analysis of the colonisation of wheat roots showed that 4 days after planting the highest bacterial activity was observed at the upper part of the root. After 6 days the high bacterial activity at the upper part was further increased, whereas spot-like activities were observed on the lower root parts, possibly due to micro-colonies. (ii) Bacterial mutations causing lack of motility or auxotrophy for amino acids resulted in impaired colonisation of the lower root parts, indicating that motility and prototrophy for the involved amino acid(s) are important factors for wheat root colonisation by strain 5RL. (iii) Coinoculation of strain 5RL with other wild type Pseudomonas strains on the root influenced the colonisation pattern observed for strain 5RL. Colonisation was not visually affected when the competing strain was a poor root coloniser, but was severely reduced when the competing strain was a good root coloniser. The results show that the spatio-temporal colonisation of wheat root by P. fluorescens strain 5RL and derivatives is similar to that of strain WCS365 on tomato. The advantage of the use of lux-marked strains is that the results are obtained much quicker than when conventional methods are used and that the result is supplied as an image of the colonisation pattern of many different roots.

    Notes: Abstract The influence of prey density, within-field vegetation, and the composition and patchiness of the surrounding landscape on the abundance of insect predators of cereal aphids was studied in wheat fields in eastern South Dakota, USA. Cereal aphids, aphid predators, and within-field vegetation were sampled in 104 fields over a three year period (1988–1990). The composition and patchiness of the landscape surrounding each field were determined from high altitude aerial photographs. Five landscape variables, aggregated at three spatial scales ranging from 2.6 km2 to 581 km2, were measured from aerial photographs. Regression models incorporating within-field and landscape variables accounted for 27–49% of the variance in aphid predator abundance in wheat fields. Aphid predator species richness and species diversity were also related to within-field and landscape variables. Some predators were strongly influenced by variability in the composition and patchiness of the landscape surrounding a field at a particular spatial scale while others responded to variability at all scales. Overall, predator abundance, species richness, and species diversity increased with increasing vegetational diversity in wheat fields and with increasing amounts of non-cultivated lands and increasing patchiness in the surrounding landscape.

    Notes: Abstract Management of stored-grain insect pests by farmers or elevator managers should be based upon a knowledge of the grain storage environment and the ecology of insect pests. Grain storage facilities and practices, geographical location, government policies, and marketing demands for grain quality are discussed as factors influencing stored-grain insect pest management decisions in the United States. Typical practices include a small number of grain samples designed to provide grain quality information for segregation, blending and marketing. This low sampling rate results in subjective evaluation and inconsistent penalties for insect-related quality factors. Information on the efficacy of insect pest management practices in the United States, mainly for farm-stored wheat, is discussed, and stored-grain integrated pest management (IPM) is compared to field-crop IPM. The transition from traditional stored-grain insect pest control to IPM will require greater emphasis on sampling to estimate insect densities, the development of sound economic thresholds and decision-making strategies, more selective use of pesticides, and greater use of nonchemical methods such as aeration. New developments in insect monitoring, predictive computer models, grain cooling by aeration, biological control, and fumigation are reviewed, their potential for improving insect pest management is discussed, and future research needs are examined.

Chinese Spring immature embryos was analysed. This chromosome arm was found to stimulate both embryogenesis and the rate of growth of calli. Recombinant lines carrying segments of 1RS were used to delineate the regions of 1RS responsible for the tissue culture effects. The enhancement of embryogenesis and the stimulation of growth were shown to be associated with two distinct genetic regions of the chromosome arm; the former is located between the centromere and the Sec 1 locus, while the latter is situated in the immediate vicinity of the Sec 1 locus.

    Notes: Abstract The three enzymes, α-amylase, peroxidase, and acid phosphatase, are among those released by the aleurone of bread wheat, Triticum aestivum, as a direct response to gibberellins (GA) from the embryo during germination. Aneuploid genotypes are used to investigate the chromosomal location, nature, and extent of genetic control of the release of these enzymes in endosperms after induction by exogenous GA. Ditelosomics demonstrate the effect of removal of known chromosome arms, and tetrasomics show the effect of duplication of chromosome pairs. Quantitative analysis demonstrates complex control systems over and above those previously found using zymogram techniques. For α-amylase, chromosome arms with net promoter effects and chromosomes with net inhibitor effects were found. These effects were not chromosome-dosage responsive, unlike the promoter-inhibitor system found for acid phosphatase control. Peroxidase levels in the endosperms were generally high in both types of aneuploids. With one exception, all chromosomes showing involvement as ditelosomics showed a similar effect as tetrasomics, indicating that in the euploid a balanced state restricting, rather than promoting, peroxidase levels existed.

    Notes: Abstract Zymogram analysis was used to identify the barley chromosomes that carry the structural genes for particular isozymes. Wheat, barley, and wheatbarley hybrid derivative lines (which contained identified barley chromosomes) were tested by gel electrophoresis for isozymes of particular enzymes. It was found that barley chromosome 4 carries structural genes for acid phosphatase and β amylase isozymes, barley chromosome 5 carries genes for phosphoglucose isomerase and malate dehydrogenase isozymes, and that barley chromosome 2 carries a gene for at least one glucose-6-phosphate dehydrogenase protomer. These results reinforce previous conclusions that barley chromosome 4 shows homoeology with wheat chromosome group 4 and that barley chromosome 5 shows homoeology with wheat chromosome group 1.

    Notes: Abstract Putative allelochemicals found in the soil of no-tillage and conventional-tillage wheat plots near Stillwater, Oklahoma, were obtained by a mild alkaline aqueous extraction procedure, bioassayed to determine their biological activity, purified, and analyzed with a capillary gas chromatography-mass spectrometry-data analysis system. The most significant inhibition was found in bioassays of extracts from soil collected immediately after harvest in June, July, and August. No-tillage soils produced significant inhibition during the rest of the year also. Mass spectrometry showed fatty acids as the most abundant compounds. However, when bioassayed authentic samples of the five free fatty acids showed no significant biological activity toward wheat.

    Notes: Abstract The primary objective of this research was to determine if soil extracts could be used directly in bioassays for the detection of allelopathic activity. Here we describe: (1) a way to estimate levels of allelopathic compounds in soil; (2) how pH, solute potential, and/or ion content of extracts may modify the action of allelopathic compounds on germination and radicle and hypocotyl length of crimson clover (Trifolium incarnatum L.) and ivyleaved morning glory (Ipomoea hederacea L. Jacquin.); and (3) how biological activity of soil extracts may be determined. A water-autoclave extraction procedure was chosen over the immediate-water and 5-hr EDTA extraction procedures, because the autoclave procedure was effective in extracting solution and reversibly bound ferulic acid as well as phenolic acids from wheat debris. The resulting soil extracts were used directly in germination bioassays. A mixture of phenolic acids similar to that obtained from wheat-no-till soils did not affect germination of clover or morning glory and radicle and hypocotyl length of morning glory. The mixture did, however, reduce radicle and hypocotyl length of clover. Individual phenolic acids also did not inhibit germination, but did reduce radicle and hypocotyl length of both species. 6-MBOA (6-methoxy-2,3-benzoxazolinone), a conversion product of 2-o-glucosyl-7-methoxy-1,4-benzoxazin-3-one, a hydroxamic acid in living wheat plants, inhibited germination and radicle and hypocotyl length of clover and morning glory. 6-MBOA, however, was not detected in wheat debris, stubble, or soil extracts. Total phenolic acids (FC) in extracts were determined with Folin and Ciocalteu's phenol reagent. Levels of FC in wheat-conventionaltill soil extracts were not related to germination or radicle and hypocotyl length of either species. Levels of FC in wheat-no-till soil extracts were also not related to germination of clover or morning glory, but were inversely related to radicle and hypocotyl length of clover and morning glory. FC values, solute potential, and acidity of wheat-no-till soil extracts appeared to be independent (additive) in action on clover radicle and hypocotyl length. Radicle and hypocotyl length of clover was inversely related to increasing FC and solute potential and directly related to decreasing acidity. Biological activity of extracts was determined best from slopes of radicle and hypocotyl length obtained from bioassays of extract dilutions. Thus, data derived from the water-autoclave extraction procedure, FC analysis, and slope analysis for extract activity in conjunction with data on extract pH and solute potential can be used to estimate allelopathic activity of wheat-no-till soils

    Notes: Abstract Diaeretiella rapae (M'Intosh) (Hymenoptera: Aphidiidae) is a parasitoid of several aphid species, including the Russian wheat aphid (RWA),Diuraphis noxia (Mordvilko), and the cabbage aphid (CA).Brevicoryne brassicae (L.). The response of matedD. rapae females to odors from wheat, cabbage, and plant-host complexes was investigated using a four-choice olfactometer. Experienced parasitoids, but not inexperienced females, responded positively to odors of the wheat-RWA complex in a no-choice test. In choice tests, experienced parasitoids did not respond to odors of uninfested cabbage and wheat leaves, but did respond positively to aphid-infested plants and to aphids alone. The response ofD. rapae to the cabbage-CA complex and to CA alone was significantly greater than to the wheat-RWA complex and RWA alone, suggesting an innate odor preference for crucifer-feeding aphids.

    Notes: Abstract Hydroxamic acids (Hx) are natural products of Gramineae that are associated with cereal resistance to pests. We aimed at characterizing the induction of Hx accumulation in seedlings of wheat,Triticum aestivum, by short-term infestation of the cereal aphid,Rhopalosiphum padi. A load of 25 aphids increased significantly the Hx levels in the infested primary leaf in comparison with control levels. Lower loads did not increase Hx concentration. Aphid infestation lasting 16 hr did not elicit induction of Hx, even after a time-lag of 32 hr to allow the expression of any induced response. Forty-eight hours was the minimum duration of aphid infestation required to trigger Hx induction. The age of the infested tissue (the primary leaf) did not affect induction. Similar increases of Hx were found in unfolding, expanding, and totally expanded primary leaves. It was determined that the regime of nutrient supply (N-intensive nutritive solutions at low and high concentration) to wheat seedlings had no effect on the magnitude of the aphid-induced Hx (N-based secondary metabolites). Results obtained are discussed in the framework of general theories of plant defense allocation.

    Notes: Abstract Open-top chambers ventilated with ambient or chiarcoal-filtered air were used to assess the impact of air pollution on the yield of local cultivars of wheat and rice, at a site on the outskirts of Lahore. At this location, 6-h mean O3 concentrations reach 60 ppb in certain months, and annual mean NO2 concentrations are 20–25 ppb. The experiments showed significant yield reduction in two successive seasons which ranged from 33% to 46% in wheat and from 37% to 51% in rice. The major yield parameter affected was the number of ears or panicles per plant, although there was also evidence of small effects on 1000 grain weight and on the number of grains per ear/panicle. These results have significance in terms of the maintenance of agricultural yields as pollution emissions rise in south and south-east Asia.

    Notes: Abstract In closed-chamber fumigation experiments dry matter partitioning and chlorophyll fluorescence of wheat were studied, analysing the effects of ozone during different stages of plant development. Ozone causes enhanced leaf senescence, leading to a loss of green leaf area and, consequently to a decreased supply of assimilates, affecting (in increasing order of severeness) stem, ear and grain productivity because of reduced storage pools for translocation. Leaves of plants before shooting stage were most sensitive but the lack of green leaf area after ear emergence had the most pronounced effects on grain yield. Measurements of photochemical capacity showed that evidence for negative ozone effects could be found in changes of chlorophyll fluorescence parameters in leaf sections not yet showing visible ozone injury. Negative effects on photosynthesis were more distinct with increasing accumulated ozone dose, with increasing age of leaf tissue and with increasing ozone sensitivity of the cultivar. The changes in chlorophyll fluorescence are most likely to be explained by a decreased pool size of plastoquinones caused by ozone.

    Notes: Abstract In Britain wheat is an important crop accounting for 41% of the total cereal production. In this study ozone concentrations for 1989 estimated as described in Part 1 of the paper are integrated with the estimated wheat distribution to derive a detailed estimate of the impact of ozone on wheat yields at a fine spatial scale (1km × 1km). These data provide estimates for calculating regional and national yield losses. The methodology can be applied to other crop species. Recent research on a range of crops has established relationships between the economic yield loss for certain crops, including wheat, and ozone exposure. Exposure is described as the accumulated exposure above a threshold experienced during the daylight hours (AOT). Critical AOT values are derived from yield exposure relationships which show linear reductions of yield loss with increasing ozone concentrations. This study has made use of land cover data from remotely sensed imagery at 25m resolution and nationally collected agricultural statistics for counties. These data were combined using an areal interpolation technique to provide more spatially articulate estimates of the location and intensity of wheat production. The results demonstrate the economic importance of ozone as a pollutant. Wheat yield losses attributed to ozone vary between different parts of the country but, for years when ozone levels are high, yield losses are likely to be significant in some areas.

    Notes: Abstract Inheritance of gliadin components unique to three different varieties of common wheat (Triticum aestivum L.) was studied in F1 and F2 seeds of intervarietal crosses using protein patterns obtained by polyacrylamide gel electrophoresis in aluminum lactate buffer (pH 3.2). The patterns of F1 seeds of the crosses Cheyenne × Justin and INIA 66R × Justin evidenced all the bands present in the patterns of the parents; band intensities reflected gene dosage levels dependent on whether the contributing parent was maternal or paternal in accordance with the triploid nature of endosperm tissue. Most of the gliadin components examined segregated in accordance with control by a single dominant gene, but in two instances single bands in the one-dimensional electrophoretic patterns segregated in the F2 as expected if controlled by two genes. A method of two-dimensional electrophoresis was developed that resolved these apparently single bands into two components each, which could segregate independently. Linkage analysis provided evidence of codominant alleles and closely linked genes coding for gliadin protein components in both coupling and repulsion situations. The gliadin protein components seem to be coded for by clusters of genes located on chromosomes of homoeologous groups 1 and 6 in hexaploid wheats.

    Notes: Abstract The primary structure of the insect α-amylase inhibitor CMa of barley seeds was deduced from a full-length cDNA clone pc43F6. Analysis of RNA from barley endosperm shows high levels 15 and 20 days after flowering. The cDNA predicts an amino acid sequence of 119 residues preceded by a signal peptide of 25 amino acids. Ala and Leu account for 55% of the signal peptide. CMa is 60–85% identical with α-amylase inhibitors of wheat, but shows less than 50% identity to trypsin inhibitors of barley and wheat. The 10 Cys residues are located in identical positions compared to the cereal inhibitor family with a Pro-X-Cys motif present in all.

    Notes: Abstract Covalent attachment of ubiquitin to other cellular proteins has been implicated in a multitude of diverse physiological processes in eukaryotes including selective protein degradation. This attachment is carried out by a multi-enzyme pathway consisting of three classes of enzymes: ubiquitin-activating enzymes (E1s), ubiquitin-conjugating enzymes (E2s), and ubiquitin-protein ligases (E3s). E2s accept activated ubiquitin from E1 and conjugate it to target proteins with or without the participation of specific E3s. Previously, we have isolated wheat cDNAs encoding 16 and 23 kDa E2s, TaUBC1 and TaUBC4, respectively. TaUBC1 shows structural homology to the yeast RAD6 E2 that is essential for DNA repair whereas TaUBC4 is related to the yeast ScUBC8 E2, both of which effectively conjugate ubiquitin to histones in vitro but as yet are without a known in vivo function. Here, we report the isolation of genomic and cDNA homologues of these genes from Arabidopsis thaliana. In Arabidopsis, both of these E2s are encoded by three member gene families. Members of the AtUBC1 gene family, comprising AtUBC1, 2 and 3, encode 150–152 amino acid proteins that are 83–99% identical to each other and TaUBC1 and contain four introns that are conserved with respect to position. Members of the AtUBC4 gene family, comprising AtUBC4, 5 and 6, encode 187–191 amino acid proteins that are 73–88% identical to each other and TaUBC4 and contain five introns that are conserved with respect to position. In contrast, AtUBC1-3 gene products are only 31–36% identical to those derived from AtUBC4-6. mRNA for each family was detected in Arabidopsis roots, leaves, stems, and flowers indicating that members of each family are expressed in most if not all tissues.

    Notes: Abstract A wheat gene (A121) encoding a protein with sequence similarity to mammalian cathepsin B is regulated by gibberellic acid (GA) in aleurone layers of germinating grains. To analyse the mechanism of A121 regulation, its promoter was fused to the β-glucuronidase reporter gene (GUS) and introduced by micro-projectile bombardment into aleurone layers of oat. With 2.3 kb of promoter sequence, the GUS expression was enhanced by GA treatment. This effect was reversed by abscisic acid (ABA). This result showed for A121, like the α-amylase genes, that the regulation by GA and ABA was at the level of transcription. The GA responsiveness of the promoter was retained with as little as 276 bp of promoter sequence. Sequence comparison with a GA responsive promoter of an α-amylase gene identified the conserved element GCAACGGCAACGATGG which is required intact for full expression of both promoters. However, there was no identifiable similarity in the cathepsin-like promoter with the GA-responsive element of α-amylase promoters with the consensus sequence TAACAAA, suggesting that GA affects more than one mechanism of transcriptional control.

    Notes: Abstract WIS-2-1A, a 8624 bp insertion in the Glu-1A-2 locus of chromosome 1A of wheat, consists of two 1755 bp long terminal repeats enclosing a 5114 bp internal region. No long open reading frames could be found, but inspection of the predicted amino acid sequence showed regions with homology to retrotransposon structures, including a methionine tRNA initiator binding site, a nucleotide binding domain, a protease, an integrase and a polymerase. DNA replication errors have resulted in frame-shifts in the protein coding region, suggesting that retrotransposition of WIS-2-1A, if it occurs, must be mediated by trans-acting factors.

    Notes: Abstract Polymerase chain reaction (PCR) was used to amplify low-molecular-weight (LMW) glutenin sequences from genomic DNA extracted from a single germinating seed of several durum wheat genotypes. Electrophoretic analysis of PCR reactions showed the presence of amplified products characteristic of durum wheat cultivars with good and poor technological properties. This PCR-based approach is proposed as a very efficient and safe alternative to standard procedures for selecting durum wheat genotypes with good qualitative characteristics.

    Notes: Abstract The amino acid sequence of wheatwin1, a monomeric protein of 125 residues isolated from wheat kernel (variety S. Pastore), is reported. Wheatwin1 is highly homologous (95%) to barwin, a protein from barley seed, which was shown to be related to the C-terminal domain of two proteins encoded by the wound-induced geneswin1 andwin2 in potato and to a protein encoded by the same domain of the hevein gene (hev1) in rubber tree. Similarly to barwin, wheatwin1 contains six cysteine residues all linked in disulfide bridges and the N-terminal residue is pyroglutamate. Moreover, structural studies performed on wheatwin1 andwin1 protein by predictive methods demonstrated that these proteins and barwin are closely related in the secondary structure also. The high level of homology found with the product ofwin1,win2, andhev1 genes strongly suggests that barwin and wheatwin1 play a common role in the mechanism of plant defence.

    Notes: Abstract We have recently described the properties of a wheat mitochondrial extract that is able to process, accurately and efficiently, artificial transcripts containing wheat mitochondrial tRNA sequences, with the production of mature tRNAs (P.J. Hanic-Joyce and M.W. Gray, J. Biol. Chem., in press). Such processing involves 5′-endonucleolytic, 3′-endonucleolytic, and TRNA nucleotidyltransferase activities. Here we show that this system also acts on transcripts containing sequences corresponding to an unusual class of short repeats (‘t-elements’) in wheat mtDNA. These repeats are theoretically capable of assuming a tRNA-like secondary structure, although stable transcripts corresponding to them are not detectable in vivo. We find that t-element sequences are processed with the same specificity and with comparable efficiency as are authentic tRNA sequences. Because known t-elements are located close to and in the same transcriptional orientation as active genes (18S-5S, 26S, tRNAPro) in wheat mtDNA, our results raise the question of whether t-elements play a role in gene expression in wheat mitochondria.

    Notes: Abstract The wheat monomeric inhibitor WMAI-1 (syn. 0.28) produced inEscherichia coli using the pT7-7 expression ventor has the correct N-terminal sequence and the same electrophoretic mobility and specific activity towards the α-amylase from the insectTenebrio molitor as the native WMAI-1 isolated from wheat. This confirms that the native inhibitor is not glycosylated and contradicts claims that a putative glycosyl moiety was essential for inhibition. Thirteen mutants have been obtained at six different sites. Substitution of the highly conserved N-terminal S by the sequence ARIRAR increased the pre-incubation time required for maximum activity. A similar result was obtained by insertion of GPRLPW after position 4, while insertion of EPRAPW at the same position rendered the inhibitor inactive. The substitution D/EGPRL and insertions DGP or D, at position 58, produced complete inactivation. All other mutations had only minor effects on activity.

    Notes: Abstract A cDNA clone, pMA1949, detects two mRNA species in wheat seedling tissue that are late embryogenesis-abundant (LEA) and dehydration stress-inducible. Sequence analysis of the pMA1949 clone shows it to be a 991 bp partial cDNA encoding a polypeptide of 317 amino acids with homology to two group 3 LEA proteins, carrot (DC8) and a soybean protein encoded by pGmPM2 cDNA. Molecular analysis of the deduced protein reveals a 33 kDa acidic and extremely hydrophilic protein with potential amphiphilic α-helical regions. In addition, the protein contains eleven similar, contiguous repeats of 11 amino acids, which are separated by 118 amino acids from two additional and unique repeats of 36 residues each at the carboxyl end of the protein. Comparisons of sequences of reported group 3 LEA proteins revealed that there are two types, separable by sequence similarity of the 11 amino acid repeating motifs and by the presence or absence of a certain amino acid stretch at the carboxyl terminus. Based on resuls from these comparisons, we propose a second type of group 3 LEA proteins, called group 3 LEA (II).

It is the major biotin-dependent carboxylase present in wheat chloroplasts. Active acetyl-CoA carboxylase purified from wheat germ is a homodimer with an apparent molecular mass of ca. 500 kDa. The enzyme from wheat germ or from wheat chloroplasts is sensitive to the herbicide haloxyfop at micromolar levels. The incorporation of 14C-acetate into fatty acids in freshly cut wheat seedling leaves provides a convenient in vivo assay for both acetyl-CoA carboxylase and haloxyfop.

    Notes: Abstract A (1→3, 1→4)-β-glucan 4-glucanohydrolase [(1→3, 1→4)-β-glucanase, EC 3.2.1.73] was purified to homogeneity from extracts of germinated wheat grain. The enzyme, which was identified as an endohydrolase on the basis of oligosaccharide products released from a (1→3, 1→4)-β-glucan substrate, has an apparent pI of 8.2 and an apparent molecular mass of 30 kDa. Western blot analyses with specific monoclonal antibodies indicated that the enzyme is related to (1→3, 1→4)-β-glucanase isoenzyme EI from barley. The complete primary structure of the wheat (1→3, 1→4)-β-glucanase has been deduced from nucleotide sequence analysis of cDNAs isolated from a library prepared using poly(A)+ RNA from gibberellic acid-treated wheat aleurone layers. One cDNA, designated λLW2, is 1426 nucleotide pairs in length and encodes a 306 amino acid enzyme, together with a NH2-terminal signal peptide of 28 amino acid residues. The mature polypeptide encoded by this cDNA has a molecular mass of 32085 and a predicted pI of 8.1. The other cDNA, designated λLW1, carries a 109 nucleotide pair sequence at its 5′ end that is characteristic of plant introns and therefore appears to have been synthesized from an incompletely processed mRNA. Comparison of the coding and 3′-untranslated regions of the two cDNAs reveals 31 nucleotide substitutions, but none of these result in amino acid substitutions. Thus, the cDNAs encode enzymes with identical primary structures, but their corresponding mRNAs may have originated from homeologous chromosomes in the hexaploid wheat genome.

    Notes: Abstract A full-length cDNA clone from hexaploid bread wheat, encoding the small subunit of ADP-glucose pyrophosphorylase, has been isolated from an endosperm cDNA library. The cDNA insert has an open reading frame which encodes a protein of 473 amino acids (52.1 kDa). The presence of a chloroplast/amyloplast transit peptide of 22 amino acids is proposed. The deduced amino acid sequence exhibits a high degree of homology with the small subunit ADP-glucose pyrophosphorylase proteins from rice (with 90% of identical amino acids) and potato (with 86% of identical amino acids) and contains conserved sequence elements which are thought to represent the substrate binding and allosteric activator sites. The genes are organised as single-copy loci on chromosomes 7A, 7B and 7D in the wheat genome and are highly expressed during grain development. Homologous transcripts are expressed in leaves and roots.

    Notes: Abstract Isolation of cDNAs encoding individual members of a gene family is essential for assessing their role in a biological phenomenon. However, this process is often laborious and slow due to highly conserved protein-coding region that interferes with the isolation of the individual members. Identification of gene-specific probes from 3′ non-coding regions of different members can assist in the fast retrieval and characterization of individual members of a multigene family. We used the recent technique of differential display for the same purpose. As an example of a multigene family in plants, we selected a heat shock protein gene family, HSP16.9 from wheat, with estimated 12 members. We modified the original differential display technique for selective amplification of the 3′ non-coding regions of different wheat HSP16.9 genes by replacing the random 10-mer in the original method with a conserved HSP16.9 gene family-specific primer. Sixteen cDNA fragments from these experiments were sequenced and they represent 8 different members of a 12 member gene family. Our succes can be attributed to shorter 3′ non-coding regions that are typical of higher-plant genes and use of highly conserved gene family-specific primer in these experiments. This modified differential display technique can be of general application to other plant systems where cloning of the different members of a gene family is desired.

    Notes: Abstract To isolate genes specifically expressed at the initiation of plant embryo development we have applied a sensitive subtractive hybridization technique for three isogenic wheat lines of the so-called ‘Salmon system’ with either zygotic or autonomous embryo development. Here we present a gene sequence showing a high homology to grass pollen allergens of type II/III thought to be expressed in pollen tissue only. Surprisingly, the pollen allergen-like sequence, designated Tri a III, is also expressed in gynoecia of the sexual, male fertile wheat line ‘(aestivum)-Salmon’, whereas the two parthenogenetic and male sterile wheat lines ‘(caudata)-Salmon’ and ‘(kotschyi)-Salmon’ completely lack any Tri a III transcript. Our data suggest a positive correlation between the expression of this clone and the manifestation of male fertility. Northern and in situ hybridization analysis revealed that, in addition to its presence in pollen, Tri a III is expressed in the parenchymatous tissue of ‘(aestivum)-Salmon’ ovaries exclusively at the day of anthesis. This precise temporal and spatial expression pattern suggests a more general function of the pollen allergen-like sequence Tri a III not limited to the exhibition of allergens in pollen grains.

    Notes: Abstract In wheat (Triticum aestivum L.), water deficit during meiosis in the microspore mother cells (MMCs) induces pollen abortion, resulting in the failure of fertilization and a reduction in grain set. In stressed plants, meiosis in MMCs proceeds normally but subsequent pollen development is arrested. Unlike normal pollen grains, which accumulate starch during the late maturation phase, stress-affected anthers contain pollen grains with little or no starch. Stress also alters the normal distribution of starch in the anther wall and connective tissue. To determine how starch biosynthesis is regulated within the developing anthers of stressed plants, we studied the expression of ADP-glucose pyrophosphorylase (AGP), which catalyzes the rate limiting step of starch biosynthesis. Two partial-length cDNAs corresponding to the large subunit of AGP were amplified by RT-PCR from anther RNA, and used as probes to monitor AGP expression in developing anthers of normal and water-stressed plants. These clones, WAL1 and WAL2, had identical deduced amino acid sequences and shared 96% sequence identity at the nucleic acid level. In normal anthers, AGP expression was biphasic, indicating that AGP expression is required for starch biosynthesis both during meiosis and later during pollen maturation. AGP expression in stressed anthers was not affected during the first phase of starch accumulation, but was strongly inhibited during the second phase. We conclude from these results that the reduced starch deposition later in the development of stressed pollen could be the result of a lower expression of AGP. However, this inhibition of AGP expression is unlikely to be the primary cause of male sterility because anatomical symptoms of pollen abortion are observed prior to the time when AGP expression is inhibited.

    Notes: Abstract The sequence of a trypsin inhibitor, isolated from wheat endosperm, is reported. The primary structure was obtained by automatic sequence analysis of the S-alkylated protein and of purified peptides derived from chemical cleavage by cyanogen bromide and digestion withStaphylococcus aureus V8 protease. This protein, named wheat trypsin inhibitor (WTI), which is comprised of a total of 71 amino acid residues, has 12 cysteines, all involved in disulfide bridges. The primary site of interaction (reactive site) with bovine trypsin has been identified as the dipeptide arginyl-methionyl at positions 19 and 20. WTI has a high degree of sequence identity with a number of serine proteinase inhibitors isolated from both cereal and leguminous plants. On the basis of the findings presented, this protein has been classified as a single-headed trypsin inhibitor of Bowman-Birk type.

    Notes: Abstract We have characterized three cDNA clones corresponding to proteins CM1, CM3 and CM16, which represent the three types of subunits of the wheat tetrameric inhibitor of insect α-amylases. The deduced amino acid sequences of the mature polypeptides are homologous to those of the dimeric and monomeric α-amylase inhibitors and of the trypsin inhibitors. The mature polypeptides are preceded by typical signal peptides. Southern blot analysis of appropriate aneuploids, using the cloned cDNAs as probes, has revealed the location of genes for subunits of the CM3 and of the CM16 type within a few kb of each other in chromosomes 4A, 4B and 4D, and those for the CM1 type of subunit in chromosomes 7A, 7B and 7D. Known subunits of the tetrameric inhibitor corresponding to genes from the B and D genomes have been previously characterized. No proteins of this class have been found to be encoded by the A genome in hexaploid wheat (genomes AA, BB, DD) or in diploid wheats (AA) and no anti α-amylase activity has been detected in the latter, so that the A-genome genes must be either silent (pseudogenes) or expressed at a much lower level.

    Notes: Abstract We report here the complete amino acid sequence of a pathogen-induced putative peroxidase from wheat (Triticum aestivum L.) as deduced from cDNA clones representing mRNA from leaves infected with the powdery mildew fungus Erysiphe graminis. The protein consists of 312 amino acids, of which the first 22 form a putative signal sequence, and has a calculated pI of 5.7. Sequence comparison revealed that the putative wheat peroxidase is most similar to the turnip (Brassica rapa) peroxidase, with which it shares 57% identical and 13% conserved amino acids.

    Notes: Abstract We have used a cDNA clone encoding a pathogen-induced putative wheat peroxidase to screen a genomic libary of wheat (Triticum aestivum L. cv. Cheyenne) and isolated one positive clone, lambda POX1. Sequence analysis revealed that this clone contains a gene encoding a putative peroxidase with a calculated pI of 8.1 which exhibits 58% and 83% sequence identity to the amino acid sequence of the turnip (Brassica rapa) peroxidase and a pathogen-induced putative wheat peroxidase, respectively. The two introns in the wheat gene are at the same positions as introns in the peroxidase genes of tomato and horseradish. Results of S1-mapping experiments suggest that this gene is neither pathogen-nor wound-induced in leaves but is constitutively expressed in roots.

Antibodies to a peptide corresponding to a part of the encoded amino acid sequence of orf256 detect a 7 kDa protein on western blots of mitochondrial proteins from cytoplasmic male-sterile (cms) wheat (T. aestivum nucleus, T. timopheevi mitochondria) but not in mitochondrial proteins from T. aestivum, T. timopheevi, or cms plants restored to fertility by introduction of nuclear genes for fertility restoration. The 7 kDa protein appears to serve as a marker for cms wheat. Its occurrence as an integral protein of the inner membrane may indicate a cms effect through an influence on mitochondrial membrane function.

    Notes: Abstract Amino acid sequences for three members (CMx1, CMx2, and CMx3) of a new subfamily of trypsin/α-amylase inhibitors in wheat have been deduced from the nucleotide sequences of the corresponding cDNAs. A cDNA clone encoding CMx1 was selected from a wheat developing endosperm library using a probe that encoded barley trypsin inhibitor BTI-CMe at low stringency. Sequences corresponding to CMx2 and CMx3 were obtained from cDNA amplified by the polymerase chain reaction. The three CMx sequences contain a premature stop codon after 363 nt, as well as a second stop codon at the same position as in BTI-CMe (nt 439–441). Southern analysis of DNAs from diploid, tetraploid, and hexaploid wheats, as well as from aneuploid lines, indicate that there is a single CMx locus in each of the three genomes of hexaploid wheat, respectively associated with chromosomal arms 4AS, 4BS, and 4DL. These genes are expressed early during endosperm development and not expressed at detectable levels in other tissues. Evolutionary implications are discussed.

    Notes: Abstract A genomic clone encoding the precursor of wheat leaf ferredoxin has been isolated and characterised. The uninterrupted PetF gene encodes a polypeptide of 143 amino acid residues, consisting of an N-terminal presequence of 46 amino acid residues and a mature polypeptide of 97 amino acid residues. Southern blot analysis suggests that six copies of the PetF gene are present in the wheat haploid genome. Northern blot analysis has shown that the genes are both developmentally and light regulated in wheat seedlings and provides evidence that a circadian rhythm regulates the steady-state levels of ferredoxin transcripts. The intact wheat gene and several chimeric constructs, containing portions of the 5′-upstream region fused to the β-glucuronidase reporter gene, have been introduced into tobacco plants, but levels of β-glucuronidase activity above background were not detected, suggesting that the 5′-upstream region is unable to function as a promoter in tobacco plants.

    Notes: Abstract The mitochondrial outer membrane of eukaryotic cells contains voltage-dependent anion channels (VDAC) also termed porins. Three cDNAs from wheat (Triticum aestivum) were isolated and sequenced (Tavdac 1–3). They share 65% similarity of their amino acid sequences, and therefore they probably represent isoforms. The deduced amino acid sequence of one of the cDNAs was found to be identical to the purified VDAC protein from wheat mitochondria [8]. Secondary structure analysis of the deduced amino acid sequences of the three vdac cDNAs revealed a characteristic α helix at their N-terminal and β-barrel cylinders characteristic of VDAC channels. The Tavdac cDNAs are differentially expressed in meristematic tissues. The transcript levels of Tavdac 1 in all wheat tissues is at least 2.5-fold higher than Tavdac 2 and Tavdac 3. Tavdac 2 has a low level of expression in all floral tissues whereas Tavdac 3 is highly expressed in anthers. This is the first report on differential expression of vdac genes in plants. The Tavdac genes have been mapped on the wheat genome. Tavdac 1 is located on the long arm of chromosome 5, Tavdac 2 on the long arm of chromosome 1 and Tavdac 3 on the long arm of chromosome 3. A phylogenetic reconstruction indicates that vdac genes underwent numerous duplication events throughout their evolution. All duplications occurred after the separation of plants from animals and fungi, and no orthologous genes are shared among phyla. Within plants, some of the vdac gene duplications probably occurred before the monocotydelon-dicotydelon split.

    Notes: Abstract A novel cDNA encoding for a peptidyl-prolyl-cis-trans-isomerase (PPIase) belonging to the FK506-binding protein (FKBP) family was isolated from wheat. It contains an open reading frame of 559 amino acids and it represents the first plant FKBP-PPIase to be cloned. It possesses a unique sequence which is composed of three FKPB-like domains, in addition to a putative tetratricopeptide repeat (TPR) motif and a calmodulin-binding site. The recombinant FKBP-PPIase expressed in and purified from Escherichia coli exhibits PPIase activity that is efficiently inhibited by the immunosuppressive drugs FK506 and rapamycin. Northern blot analysis showed that wheat FKBP was found mainly in young tissues. Polyclonal antibodies revealed the presence of cross-reacting proteins in embryos, roots and shoots. The unique structural features, the enzymatic activity and the presence of putative isoforms in wheat tissues indicate the possibility of the involvement of wheat PPIase in essential biological functions, similar to other members of the FKBP gene family.

    Notes: Abstract Dynamical aspects of three chloroplast promoters responding to change in light condition were examined in mature chloroplasts of wheat (Triticum aestivum) by in vitro transcription. The wheat psbD/C operon has four distinct promoters, two of which named as D/C-3 and D/C-4 promoters dominantly function in mature chloroplasts to produce the mRNAs encoding D2/CP43 and CP43 alone, respectively. Activity of the D/C-3 promoter in mature chloroplasts was reduced to less than 30% by 24 h dark adaptation and recovered by re-illumination to the original level within 30 to 60 min. The activation of the D/C-3 promoter which requires de novo cytoplasmic protein synthesis was induced by low fluence of light (e.g. 16 µE m-2 s-1), but the extent of activation increased with increasing light fluence. The accumulation of mRNAs from the D/C-3 promoter saturated at 2- to 3-fold higher level within 2 h when the dark-adapted seedlings were transferred to the lig at 72 µE m-2 s-1, concomitant with the increase in rate of D2 synthesis, suggesting that synthesis of D2 in mature chloroplasts is controlled via the D/C-3 promoter activity in a light-dependent way. Activity of the D/C-4 promoter slightly increased in the dark and decreased in the light. Effect of light on the psbA promoter activity was not observed at all in mature chloroplasts. In vitro transcriptional analysis of the D/C-3 promoter with 5′ deletion mutations revealed that at least two cis elements which are located within the sequences of -78 to -47 and -46 to -29 of the transcription initiation site, respectively, act as enhancing elements in the D/C-3 promoter. The light-switching element of the transcription, however, was suggested to be located in the core promoter sequence downstream of the -35 element.

    Notes: Abstract The wheat mitochondrial orf25 nucleotide sequence of 576 pb has been determined. Its derived protein sequence shares 88% and 75% amino acid identity with those of maize and tobacco mitochondria, respectively. The wheat and tobacco orf25 sequences lack four inserts, of 6 bp to 36 bp, that are present in the maize homologue. The wheat orf25 gene is actively transcribed and is preceded by a regulatory sequence block very similar to those located upstream of the wheat coxII and atp6 genes. Our observations support the view that orf25 sequences encode a functional polypeptide in plant mitochondria.

    Notes: Abstract A cDNA (WPRP1) encoding a wheat proline-rich protein has been isolated and sequenced. The amino acid composition shows 45% proline, with high levels of methionine, lysine and glutamic acid. The derived 378 residue amino acid sequence has a highly repetitive structure which is unlike those of other proline-rich proteins. The WPRP1 cDNA clone was used to determine the copy number and chromosomal location of the WPRP1 gene by restriction fragment length polymorphism analysis of wheat inbred lines. Although WPRP1 is encoded by a single-copy gene it is also a representative of a larger family of related sequences. RNA gel blot analysis showed that expression of WPRP1 is highest in rapidly growing tissue which together with its amino acid composition suggests a structural role for the encoded protein.

    Notes: Abstract The promoter region (−524 to −46) of the wheat α/β-gliadin seed storage protein gene was analyzed for interactions with nuclear proteins from developing wheat seeds. Six complexes were detected within the first 165 bp upstream of the transcriptional start site. One of the proteins was a non-sequence specific AT-binding protein. The remaining five proteins bound in a sequence specific manner. One (CABP) mapped to a conserved CA-rich element at −134 to −112 while another (PalBP) mapped to an adjacent, palindromic sequence at −112 to −106. Three proteins (CTBPs 1–3) formed complexes at two, independent homologous sites. The activities of four of the binding proteins, CTBPs 1–3 and CABP, exhibited similar patterns of expression during seed development: they first appeared at early to mid stages, reached a maximum at mid stage and subsequently decreased, paralleling the pattern of gliadin mRNA accumulation. The non-specific AT-binding protein was detected at relatively high levels only at mid development. PalBP activity, on the other hand, first appeared at mid stage and was present at a constant level throughout later stages of development. The results suggest that the binding proteins may regulate gliadin expression in an antagonistic manner.

    Notes: Abstract We have characterised a DNA-binding interaction common to the upstream sequences of the wheat fructose-1,6-bisphosphatase (FBPase) and sedoheptulose-1,7-bisphosphatase (SBPase) genes. The recognition site for this sequence-specific binding activity, designated wheat FBPase factor (WF-1), is located within 125 bp of the transcription start site of each gene. Within these regions there are no sequence motifs similar to those shown to be important for light-regulated expression in other species. The binding activity was not detected in wheat root nuclear extracts, or in pea leaf extracts. There was a higher level of binding activity in light-grown than in dark-grown wheat leaves. The level was also found to decline when light-grown plants were given an extended dark treatment, but could be reinduced by light. Utilising the gradient of developmental maturity which exists within the wheat leaf it was found that WF-1 activity increases during leaf development.

    Notes: Abstract We isolated a complementary DNA (cDNA) encoding the TATA-binding factor ‘TFIID’ from a wheat seedling cDNA library. The wheat TFIID transcript of 1.2 kb poly(A)+ RNA was expressed at a low level early in germination, but gradually increased as the seedlings developed. In vitro binding experiments showed that the bacterially expressed wheat TFIID protein could specifically bind to the TATA boxes of the cauliflower mosaic virus (CaMV) 35S, wheat histone H3 and adenovirus major late genes with different affinity. A comparison with Arabidopsis TFIID showed the presence of a plant-specific region consisting of 13 amino acids at the divergent amino terminus and a conserved region (182 amino acids) at the carboxy terminus longer than that observed in yeasts (180 amino acids) and animals (181 amino acids).

    Notes: Abstract We report the sequence of a 7.2 kilobase pair DNA fragment containing a copy of the wheat mitochondrial gene (rrn26) that encodes the mitochondrial large-subunit ribosomal RNA (26S rRNA). The mature 26S rRNA was determined by direct RNA sequencing to be 3467 nucleotides long, and to share a 5′-terminal pentanucleotide (5′-AUCAU), thought to be important in post-transcriptional processing, with the wheat mitochondrial small-subunit (18S) rRNA. Two other prominent features of the sequence were noted. First, upstream of rrn26 are located two tandem copies of a 70 base pair element containing a putative mitochondrial promoter motif (TCGTATAAAAA). Second, downstream of rrn26 is a sequence element that, if transcribed, would produce and RNA with a secondary structure resembling that of tRNAs but differing sufficiently from the latter structure to preclude any transcript from functioning normally in translation. These upstream and downstream sequence elements may play a role in the expression of rrn26 in wheat mitochondria.

    Notes: Abstract The S10 ribosomal protein gene (rps10), which has not been previously reported in any angiosperm mitochondrial genome, was identified by sequence analysis in the potato mitochondrial DNA. This gene is found downstream of a truncated non-functional apocytochrome b (cob) pseudogene, and is expressed as multiple transcripts ranging in size from 0.8 to 5.0 kb. Southern hybridization analysis indicates that rps10-homologous sequences are not present in the wheat mitochondrial genome. Sequence analysis of a single-copy region of the pea mitochondrial genome located upstream of cox1 [11] shows that a non-functional rps10 pseudogene is present in this species. These results suggest that the functional genes coding for wheat and pea mitochondrial RPS10 polypeptides have been translocated to the nucleus.

    Notes: Abstract The time course of the accumulation of the transcripts from 13 psb genes encoding a major part of the proteins composing photosystem II during light-induced greening of dark-grown wheat seedlings was examined focusing on early stages of plastid development (0.5 h through 72 h). The 13 genes can be divided into three groups. (1) The psbA gene is transcribed as a single transcript of 1.3 kb in the dark-grown seedlings, but its level increases 5- to 7-fold in response to light due to selective increase in RNA stability as well as in transcription activity. (2) The psbE-F-L-J operon, psbM and psbN genes are transcribed as a single transcript of 1.1 kb, two transcripts of 0.5 and 0.7 kb and a single transcript of 0.3 kb, respectively, in the dark-grown seedlings. The levels of accumulation of every transcript remain unchanged or rather decrease during plastid development under illumination. (3) The psbK-I-D-C gene cluster and psbB-H operon exhibit fairly complicated northern hybridization patterns during the greening process. When a psbC or psbD gene probe was used for northern hybridization, five transcripts differing in length were detected in the etioplasts from 5-day old dark-grown seedlings. After 2 h illumination, two new transcripts of different length appeared. Light induction of new transcripts was also observed in the psbB-H operon.
