    Beschreibung: We investigated the high frequency attenuation of S-waves in Southeastern Alps and Northern External Dinarides using waveforms from 331 earthquakes (3.0〈 Mw〈 6.5). The spectral decay parameter, k, was computed using 1345 three component high quality records, collected by the Italian Strong Motion Network (RAN) and by the Short-Period Seismometric Network of North-Eastern Italy (NEI) in the period 1976-2007. Weak motion data from 11 stations of the NEI network and strong motion data collected by 5 accelerometers of the RAN were analyzed. The k parameter was estimated in the 0-250 Km distance range, in a frequency band extending from the corner frequency of the event up to 25 or 45 Hz, using the amplitude acceleration Fourier spectra of S-waves. The observed record-to-record variability of k was modeled by applying a generalized inversion procedure, using both parametric and non-parametric approaches. Our results evidence that k is independent on earthquake size, while it shows both site and distance dependence. Stations of the NEI network present the same increase of k with epicentral distance, Re, and show values of the zero-distance k parameter, k0(S), between 0.017 and 0.053 s. For the whole region, the k increase with distance can be described through a linear model with slope dk/dRe = (1.4±0.1)x10^(-4) s/Km. Assuming an average S-wave velocity, 〈Vs〉=3.34 Km/s between 5 and 15 Km depth, we estimate an average frequency independent quality factor, 〈Qi〉=2140, for the corresponding crustal layer. The non-parametric approach evidences a weak positive concavity of the curve that describes the k increase with Re at about 90 Km distance. This result can be approximated through a piecewise linear function with slopes of 1.0x10^(-4) s/Km and 1.7x10^(-4) s/Km, in accordance with a three layers model where moving from the intermediate to the bottom layer both 〈Qi〉 and 〈Vs〉 decrease. Two regional dependences were found: data from earthquakes located westward to the NEI network evidence weaker attenuation properties, probably because of S-wave reflections from different part of the Moho discontinuity under the eastern Po Plain, at about 25-30 Km depth, while earthquakes located eastward (in western Slovenia), where the Moho deepens up to 45-50 Km, evidence a higher attenuation. Moreover, the k estimates obtained with data from earthquakes located in the area of the 1998 (Mw=5.7) and 2004 (Mw=5.2) Kobarid events are 0.017 s higher than the values predicted for the whole region, probably because of the high level of fracturing that characterizes fault zones. The comparison between measured and theoretical values of k, computed at a few stations with available S-wave velocity profiles, reveals that the major contribution to the total k0(S) is due to the sedimentary column (from surface to 800 m depth). The hard rock section contribution is limited to 0.005 s, in accordance with a maximum contribution of 0.010 s predicted by the non-parametric inversion.

    Beschreibung: We present a high-resolution palaeomagnetic and rock magnetic study of two cores, MS06 and MS06-SW (6.7 and 1.1 m long, respectively), collected at 72 m below sea level in the Augusta Bay shelf (Eastern Sicily, Ionian Sea, Italy) about 2.3 kmfrom the coastline. Geophysical surveying carried out in the sampling area highlighted the presence of a homogeneous sedimentary sequence that most likely was deposited after the Last Glacial Maximum and was not affected by anthropogenic disturbances. The two cores penetrated a monotonous mud sedimentary sequence, interrupted at ∼3 m depth by a 3–4-cm-thick volcanic sandy layer that is correlated with the tephra fallout deposit produced by the 122 BC plinian eruption of Mt Etna. This tephra, along with radiocarbon dating of nine marine shells and with radioactive tracers for the uppermost 0.3 m (210Pb and 137Cs), provide the chronological constraints for the stratigraphic sequence that resulted younger than 4500 yr BP. Palaeomagnetic and rock magnetic data show that the sample sequence is magnetically homogeneous. A single peak of high magnetic mineral concentration is present and corresponds to the volcanic sandy layer. Palaeomagnetic data allowed the identification of a well-defined characteristic remanent magnetization that provides a high-resolution record of palaeosecular variation (PSV) at the sampling site. The reconstructed PSV curve is in good agreement with the available regional reference PSV curves and with the prediction from recent PSV modelling for Europe. The palaeomagnetic data obtained in this study on the one hand support and refine the age model for the cores, derived from other independent constraints, and on the other hand provide an original high-resolution PSV curve that can serve as a reference for the central Mediterranean over the last 4 ka.

    Beschreibung: The genus Alexandrium (Halim) is perhaps the most intensively studied among toxic marine dinoflagellates. This is largely attributable to the devastating consequences of toxigenic blooms of this genus, with human poisonings from contaminated seafood, primarily from shellfish and more rarely from finfish; socio–economic losses to the aquaculture and fisheries industries; marine faunal mortalities; and food web disruptions common in coastal waters throughout the world. Members of this genus are globally distributed from the Arctic to the tropics, and in both hemispheres from sub–polar through temperate to sub–tropical to tropicalwaters. At least four distinct groups of marine phycotoxins are associated with various Alexandrium species, along with poorly characterized bioactive compounds (allelochemicals) that may affect species interactions among the plankton. According to the most recent iteration of the IOC–UNESCO reference list of toxic microalgae, there are now more than 30 recognized morphological species of Alexandrium, posing a daunting challenge for risk assessment and accurate identification in toxic phytoplankton monitoring programs.

    Beschreibung: Understanding how the Antarctic ice sheet will respond to global warming relies on knowledge of how it has behaved in the past. The use of numerical models, the only means to quantitatively predict the future, is hindered by limitations to topographic data both now and in the past, and in knowledge of how subsurface oceanic, glaciological and hydrological processes interact. Incorporating the variety and interplay of such processes, operating at multiple spatio-temporal scales, is critical to modeling the Antarctic’s system evolution and requires direct observations in challenging locations. As these processes do not observe disciplinary boundaries neither should our future research.

    Beschreibung: Natural dissolved organic matter (DOM) comprises a broad range of dissolved organic molecules in aquatic systems and is among the most complex molecular mixtures known. Here we show, by comparing detailed structural fingerprints of individual molecular formulae in DOM from a set of four marine and one freshwater environments, that a major component of DOM is molecularly indistinguishable in these diverse samples. Molecular conformity was not only apparent by the co-occurrence of thousands of identical molecular formulae, but also by identical structural features of those isomers that collectively represent a molecular formula. The presence of a large pool of compounds with identical structural features in DOM is likely the result of a cascade of degradation processes or common synthetic pathways that ultimately lead to the formation of a universal background, regardless of origin and history of the organic material. This novel insight impacts our understanding of long-term turnover of DOM as the underlying mechanisms are possibly universal.

    Beschreibung: Traditionally, the description of microorganisms starts with their isolation from an environmental sample. Many environmentally relevant anaerobic microorganisms grow very slowly, and often they rely on syntrophic interactions with other microorganisms. This impedes their isolation and characterization by classic microbiological techniques. We developed and applied an approach for the successive enrichment of syntrophic hydrocarbon-degrading microorganisms from environmental samples. We collected samples from microbial mat-covered hydrothermally heated hydrocarbon-rich sediments of the Guaymas Basin and mixed them with synthetic mineral medium to obtain sediment slurries. Supplementation with defined substrates (i.e., methane or butane), incubation at specific temperatures, and a regular maintenance procedure that included the measurement of metabolic products and stepwise dilutions enabled us to establish highly active, virtually sediment-free enrichment cultures of actively hydrocarbon-degrading communities in a 6-months to several-years' effort. Using methane as sole electron donor shifted the originally highly diverse microbial communities toward defined mixed cultures dominated by syntrophic consortia consisting of anaerobic methane-oxidizing archaea (ANME) and different sulfate-reducing bacteria. Cultivation with butane at 50 °C yielded consortia of archaea belonging to Candidatus Syntrophoarchaeum and Candidatus Desulfofervidus auxilii partner bacteria. This protocol also describes sampling for further molecular characterization of enrichment cultures by fluorescence in situ hybridization (FISH), and transcriptomics and metabolite analyses, which can provide insights into the functioning of hydrocarbon metabolism in archaea and resolve important mechanisms that enable electron transfer to their sulfate-reducing partner bacteria.

    Beschreibung: The construction of high capacity data sharing networks to support increasing government and commercial data exchange has highlighted a key roadblock: the content of existing Internet-connected information remains siloed due to a multiplicity of local languages and data dictionaries. This lack of a digital lingua franca is obvious in the domain of human food as materials travel from their wild or farm origin, through processing and distribution chains, to consumers. Well defined, hierarchical vocabulary, connected with logical relationships—in other words, an ontology—is urgently needed to help tackle data harmonization problems that span the domains of food security, safety, quality, production, distribution, and consumer health and convenience. FoodOn (http://foodon.org) is a consortium-driven project to build a comprehensive and easily accessible global farm-to-fork ontology about food, that accurately and consistently describes foods commonly known in cultures from around the world. FoodOn addresses food product terminology gaps and supports food traceability. Focusing on human and domesticated animal food description, FoodOn contains animal and plant food sources, food categories and products, and other facets like preservation processes, contact surfaces, and packaging. Much of FoodOn’s vocabulary comes from transforming LanguaL, a mature and popular food indexing thesaurus, into a World Wide Web Consortium (W3C) OWL Web Ontology Language-formatted vocabulary that provides system interoperability, quality control, and software-driven intelligence. FoodOn compliments other technologies facilitating food traceability, which is becoming critical in this age of increasing globalization of food networks.

    Beschreibung: The connection between Tropical Pacific and North Pacific variability is investigated in a state-of-the art coupled ocean-atmosphere model, comparing two 20th century simulations at T30 and T106 atmospheric horizontal resolutions. Despite a better simulation of the frequency and the spatial distribution of the Tropical Pacific anomalies associated with the El Nino Southern Oscillation (ENSO) in the high-resolution experiment, the response in the North Pacific is scarcely different from the low-resolution experiment where the ENSO variability is weaker and at higher than observed frequency. In the North Pacific, the response of surface atmospheric fields to the variability in the Tropical Pacific appears to be affected by local coupling processes significantly different in the two experiments. The coupling between sea level pressure (SLP) and sea surface temperature (SST) in the North Pacific as well as the influence of the Tropical Pacific SST has been measured here by means of the ‘coupled manifold’ technique. In the low-resolution case the SLP variances linked to the fraction of North Pacific SST not influenced by the Tropical Pacific are weak suggesting that the remote influence is strong, consistently with the observations. On the contrary, in the high-resolution experiment the fractions and the patterns of the SLP variances due to the Tropical Pacific SST and those linked to the North Pacific SST are comparable. In the latter case, model systematic errors in the northwestern Pacific influences the local coupling processes thus triggering the remote response. We conclude that an increased atmospheric horizontal resolution does not reduce the coupled model systematic errors in the representation of the teleconnection between the North and the Tropical Pacific and that the validation of coupled models has to consider both remote and local processes.

    Beschreibung: Edifice instability, that can result in catastrophic flank collapse, is a fundamental volcanic hazard. The subvolcanic basement can encourage such instability, especially if it is susceptible to mechanical weakening by devolatilization reactions near magmatic temperatures. For this reason, understanding how the physical and chemical properties of representative lithologies deteriorate at high temperatures is potentially highly relevant for volcanic hazard mitigation. This is particularly true for sedimentary rock, commonly found underlying volcanic edifices worldwide, that undergo rapid deterioration even under modest temperatures. Therefore, here we present the first experimental study of devolatilization reactions, induced by magmatic temperatures, on sedimentary rock comprising a subvolcanic basement. Our results show that, for a marly limestone representative of the basement at Mt Etna, devolatilization reactions, namely the dehydroxylation of clay minerals and the decarbonation of calcium carbonate, result in a dramatic reduction of mechanical strength and seismic velocities. These temperature-driven reactions can promote volcanic instability at stresses much lower than previously estimated.

    Beschreibung: With the continued growth in availability of DInSAR and GPS data, space based geodesy has been widely applied to image the coseismic displacement field and to retrieve the static dislocation over the fault plane for almost all the significant earthquakes of the past two decades. This is performed by linear data inversion over a set of subfaults, generally characterized by a constant and predefined or manually adjusted dimensions. In this paper we propose a new algorithm to automatically retrieve an optimized fault subdivision in the linear inversion of coseismic geodetic data. The code iteratively keeps the parameter resolution close to a predefined high value. We first discuss the rationale supporting our algorithm and, after a detailed description of its implementation, we analyze the advantages of its introduction in the data inversion. The algorithm was tested against an exhaustive range of synthetic and real datasets and fault mechanisms. Among them, we present the results for the Mw 6.2, 2009 L’Aquila (Central Italy) earthquake and compare the new and previously published slip distributions showing the disappearance of misleading slip pattern and the increased resolution for shallower zones.

    Beschreibung: The 2009 April 6, Mw= 6.3 L’Aquila earthquake occurred within a complex system of NW–SE trending normal faults in the Abruzzi Central Apennines (Italy). We analyse the coseismic deformation as measured by 〉70 global positioning system (GPS) stations, both from continuous and survey-mode networks, providing unprecedented details for a moderate normal faulting earthquake in Italy from GPS measurements. We use rectangular, uniform-slip, dislocations embedded in an elastic, homogeneous and isotropic half-space and a constrained, non-linear optimization algorithm, to solve for the best-fitting rectangular dislocation geometry and coseismic-slip distribution. We use a bootstrap approach to investigate uncertainties in the model parameters and define confidence bounds for all the inverted parameters. The rupture occurred on a N129°E striking and 50° southwestward dipping normal fault, in agreement with geological observations of surface breaks along the Paganica fault. Our distributed slip model exhibits a zone of relatively higher slip (〉60 cm) between ∼1.5 and ∼11 km depth, along a roughly downdip, NW–SE elongated patch, confined within the fault plane inverted assuming uniform-slip. The highest slip, of the order of ∼1 m, occurred on a ∼16 km2 area located at ∼5 km depth, SE of the mainshock epicentre. The analysis of model resolution suggests that slip at depth below ∼5 km can be resolved only at a spatial scale larger than 2 km, so a finer discretization of different asperities within the main patch of coseismic-slip is not allowed by GPS data. We compute the coseismic Coulomb stress changes in the crustal volume affected by the major aftershocks, and compare the results obtained from the uniform-slip and the heterogeneous-slip models. We find that most of the large aftershocks occurred in areas of Coulomb stress increase of 0.2–13 bar and that a deepening of the slip distribution down to a depth greater than 6 km in the SE part of the fault plane, in agreement with the inverted slip model, can explain the deepest, April 7, Mw 5.3 aftershock.

    Beschreibung: The MW 8.8 mega-thrust earthquake and tsunami that occurred on February 27, 2010, offshore Maule region, Chile, was not unexpected. A clearly identified seismic gap existed in an area where tectonic loading has been accumulating since the great 1835 earthquake experienced and described by Darwin during the voyage of the Beagle. Here we jointly invert tsunami and geodetic data (InSAR, GPS, land-level changes), to derive a robust model for the co-seismic slip distribution and induced co-seismic stress changes, and compare them to past earthquakes and the pre-seismic locking distribution. We aim to assess if the Maule earthquake has filled the Darwin gap, decreasing the probability of a future shock . We find that the main slip patch is located to the north of the gap, overlapping the rupture zone of the MW 8.0 1928 earthquake, and that a secondary concentration of slip occurred to the south; the Darwin gap was only partially filled and a zone of high pre-seismic locking remains unbroken. This observation is not consistent with the assumption that distributions of seismic rupture might be correlated with pre-seismic locking, potentially allowing the anticipation of slip distributions in seismic gaps. Moreover, increased stress on this unbroken patch might have increased the probability of another major to great earthquake there in the near future.

    Beschreibung: We analyzed a broad region around L’Aquila in search of seismogenic faults similar to that responsible for the 6 April 2009 earthquake (Mw 6.3). Having the lessons learned from this earthquake in mind, we focused on adjacent areas displaying similar morphotectonic, geological and structural evidence. The basin running from Barisciano to Civitaretenga-Navelli, notably located near the southeastern edge of the 2009 aftershock pattern, appears to be one of such areas. We collected morphotectonic and structural data indicating that this basin is underlain by a major active normal fault (San Pio Fault). All the observations are very much reminiscent of the morphotectonic, geological and structural setting of area struck by the L’Aquila earthquake, suggesting that the newly identified fault has the potential for a Mw 6.2-6.4 shock.

    Beschreibung: Our study area is a ~50 km long section of the central-southern Apennines tectonic belt that includes the Pergola-Melandro basin and the Agri valley. This region is located between the areas interested by the 1980 Ms=6.9 Irpinia and the 1857 M=7.0 Val d’Agri earthquakes and is characterized by rare historical events and very low and sparse background seismicity. In this study we provide new seismological and geophysical information to identify the characteristics of the seismotectonics in the area, as the prevailing faulting mechanism and the fit of local to regional stress field. These data concern focal mechanisms from waveform modeling and P-wave polarities, analyses of borehole breakouts and detailed investigation of two seismic sequences. All the data cover a significantly broad range of magnitudes and depths and suggest that no important local variation in stress orientation seems to affect this area, which shows a NE-SW direction of extension consistent with that regionally observed in Southern Italy. Such local homogeneity in the stress field pattern is peculiar of the study area; the variations of orientation and/or type of stress observed in the northern Apennines or only less than 100 km toward the northwest within the same tectonic belt are absent here. Furthermore, there is a suggestion for a northeastward sense of dip of the seismogenic faults in the region, an interesting constraint to the characterization of seismic sources

    Beschreibung: The aerosol climatology at the coastal Antarctic Neumayer Station (NM) was investigated based on continuous, 25 years long observations of biogenic sulfur components (methanesulfonate and non sea salt sulfate), sea salt and nitrate. Whilst significant long-term trends could only be detected for nitrate (-3.6±2.5% per year between 1983 and 1993 and +4.0±3.2% per year from 1993-2007), non-harmonic periodicities between 2 and 5 years were typical for all species. Dedicated time series analyses revealed that relations to sea ice extent and various circulation indices are weak at best or not significant. In particular, no consistent link between sea ice extent and sea salt loadings was evident suggesting only a rather local relevance of the NM sea salt record. Nevertheless, a higher Southern Annular Mode index tended to entail a lower biogenic sulfur signal. In examining the spatial uniformity of the NM findings we contrasted them to respective 17 years records from the coastal Dumont d’Urville Station (DDU). We found similar long term trends for nitrate, indicating an Antarctic-wide but not identifiable atmospheric signal, though any significant impact of solar activity or pollution could be ruled out. No inter-site variability on the multi annual scale was evident for the other ionic compounds.

    Beschreibung: We investigated the variability of 210Pb, 7Be and 10Be in coastal Antarctica aerosol samples based on continuous, monthly and annually resolved time series which we obtained from Neumayer Station over the period 1983 to 2008. Clear seasonal cycles peaking in the local summer half year stands out in being common to all three radionuclide records. However, MC-SSA time series analyses suggest that significant multi-annual changes are confined to a 4-5 years periodicity resembling that of the SAM circulation index in case of 210Pb and to the expected solar decadal cycle in case of the cosmogenic Be-isotopes. Both, changes in the meridional transport and surface inversion strength appear to drive the seasonal 210Pb cycle which generally peaks in November. In contrast, stratospheric air mass intrusion are proved to be the main reason for the Be-isotopes seasonality as revealed by enhanced 10Be/ 7Be ratios occurring broadly concurrently with the individual Be-isotopes and the 7Be/210Pb ratios during late summer/early autumn. Although both, the (annual) 10Be and the monthly 7Be records reflects the decadal solar modulated production signal the detailed pattern of their decadal changes essentially deviate for unknown reasons, though a substantial excess 7Be production by solar energetic particles could be ruled out.

    Beschreibung: Warm subtropical-origin Atlantic water flows northward across the Greenland-Scotland Ridge into the Nordic Seas, where it relinquishes heat to the atmosphere and gradually transforms into dense Atlantic-origin water. Returning southward along east Greenland, this water mass is situated beneath a layer of cold, fresh surface water and sea ice. Here we show, using measurements from autonomous ocean gliders, that the Atlantic-origin water was re-ventilated while transiting the western Iceland Sea during winter. This re-ventilation is a recent phenomenon made possible by the retreat of the ice edge toward Greenland. The fresh surface layer that characterises this region in summer is diverted onto the Greenland shelf by enhanced onshore Ekman transport induced by stronger northerly winds in fall and winter. Severe heat loss from the ocean offshore of the ice edge subsequently triggers convection, which further transforms the Atlantic-origin water. This re-ventilation is a counterintuitive occurrence in a warming climate, and highlights the difficulties inherent in predicting the behaviour of the complex coupled climate system.

    Beschreibung: Intertidal inhabitants are exposed to the 24-hour solar day, and the 12.4 hour rising and falling of the tides. One or both of these cycles govern intertidal organisms’ behaviour and physiology, yet little is known about the molecular clockworks of tidal rhythmicity. Here, we show that the limpet Cellana rota exhibits robust tidally rhythmic behaviour and gene expression. We assembled a de-novo transcriptome, identifying novel tidal, along with known circadian clock genes. Surprisingly, most of the putative circadian clock genes, lack a typical rhythmicity. We identified numerous tidally rhythmic genes and pathways commonly associated with the circadian clock. We show that not only is the behaviour of an intertidal organism in tune with the tides, but so too are many of its genes and pathways. These findings highlight the plasticity of biological timekeeping in nature, strengthening the growing notion that the role of ‘canonical’ circadian clock genes may be more fluid than previously thought, as exhibited in an organism which has evolved in an environment where tidal oscillations are the dominant driving force.

    Beschreibung: The kakapo is a critically endangered, herbivorous parrot endemic to New Zealand. The kakapo hindgut hosts a dense microbial community of low taxonomic diversity, typically dominated by Escherichia fergusonii, and has proven to be a remarkably stable ecosystem, displaying little variation in core membership over years of study. To elucidate mechanisms underlying this robustness, we performed 16S rRNA gene-based co-occurrence network analysis to identify potential interactions between E. fergusonii and the wider bacterial community. Genomic and metagenomic sequencing were employed to facilitate interpretation of potential interactions observed in the network. E. fergusonii maintained very few correlations with other members of the microbiota, and isolates possessed genes for the generation of energy from a wide range of carbohydrate sources, including plant fibres such as cellulose. We surmise that this dominant microorganism is abundant not due to ecological interaction with other members of the microbiota, but its ability to metabolise a wide range of nutrients in the gut. This research represents the first concerted effort to understand the functional roles of the kakapo microbiota, and leverages metagenomic data to contextualise co-occurrence patterns. By combining these two techniques we provide a means for studying the diversity-stability hypothesis in the context of bacterial ecosystems.

    Beschreibung: Satellite-tracking of mature white sharks (Carcharodon carcharias) has revealed open-ocean movements spanning months and covering tens of thousands of kilometers. But how are the energetic demands of these active apex predators met as they leave coastal areas with relatively high prey abundance to swim across the open ocean through waters often characterized as biological deserts? Here we investigate mesoscale oceanographic variability encountered by two white sharks as they moved through the Gulf Stream region and Sargasso Sea in the North Atlantic Ocean. In the vicinity of the Gulf Stream, the two mature female white sharks exhibited extensive use of the interiors of clockwise-rotating anticyclonic eddies, characterized by positive (warm) temperature anomalies. One tagged white shark was also equipped with an archival tag that indicated this individual made frequent dives to nearly 1,000 m in anticyclones, where it was presumably foraging on mesopelagic prey. We propose that warm temperature anomalies in anticyclones make prey more accessible and energetically profitable to adult white sharks in the Gulf Stream region by reducing the physiological costs of thermoregulation in cold water. The results presented here provide valuable new insight into open ocean habitat use by mature, female white sharks that may be applicable to other large pelagic predators.

    Beschreibung: Foraminifera in sediments exposed to gas-hydrate dissociation are not expected to have cellular adaptations that facilitate inhabitation of chemosynthesis-based ecosystems because, to date, there are no known endemic seep foraminifera. To establish if foraminifera inhabit sediments impacted by gas-hydrate dissociation, we examined the cellular ultrastructure of Melonis barleeanus (Williamson, 1858) from the Vestnesa gas hydrate province (Arctic Ocean, west of Svalbard at ~79 °N; ~1200-m depth; n = 4). From sediments with gas hydrate indicators, living M. barleeanus had unusual pore plugs composed of a thick, fibrous meshwork; mitochondria were concentrated at the cell periphery, under pore plugs. While there was no evidence of endosymbioses with prokaryotes, most M. barleeanus specimens were associated with what appear to be Type I methanotrophic bacteria. One foraminifer had a particularly large bolus of these microbes concentrated near its aperture. This is the first documented instance of bona fide living M. barleeanus in gas-hydrate sediments and first documentation of a foraminifer living in close association with putative methanotrophs. Our observations have implications to paleoclimate records utilizing this foundational foraminiferal species.

    Beschreibung: Voltage-sensing (VSD) and cyclic nucleotide-binding domains (CNBD) gate ion channels for rapid electrical signaling. By contrast, solute carriers (SLCs) that passively redistribute substrates are gated by their substrates themselves. Here, we study the orphan sperm-specific solute carriers SLC9C1 that feature a unique tripartite structure: an exchanger domain, a VSD, and a CNBD. Voltage-clamp fluorimetry shows that SLC9C1 is a genuine Na+/H+ exchanger gated by voltage. The cellular messenger cAMP shifts the voltage range of activation. Mutations in the transport domain, the VSD, or the CNBD strongly affect Na+/H+ exchange, voltage gating, or cAMP sensitivity, respectively. Our results establish SLC9C1 as a phylogenetic chimaera that combines the ion-exchange mechanism of solute carriers with the gating mechanism of ion channels. Classic SLCs slowly readjust changes in the intra- and extracellular milieu, whereas voltage gating endows the Na+/H+ exchanger with the ability to produce a rapid pH response that enables downstream signaling events.

    Beschreibung: Glomeruli are the functional units of olfactory information processing but little remains known about their individual unit function. This is due to their widespread activation by odor stimuli. We expressed channelrhodopsin-2 in a single olfactory sensory neuron type, and used laser stimulation and simultaneous in vivo calcium imaging to study the responses of a single glomerulus to optogenetic stimulation. Calcium signals in the neuropil of this glomerulus were representative of the sensory input and nearly identical if evoked by intensity-matched odor and laser stimuli. However, significantly fewer glomerular layer interneurons and olfactory bulb output neurons (mitral cells) responded to optogenetic versus odor stimuli, resulting in a small and spatially compact optogenetic glomerular unit response. Temporal features of laser stimuli were represented with high fidelity in the neuropil of the glomerulus and the mitral cells, but not in interneurons. Increases in laser stimulus intensity were encoded by larger signal amplitudes in all compartments of the glomerulus, and by the recruitment of additional interneurons and mitral cells. No spatial expansion of the glomerular unit response was observed in response to stronger input stimuli. Our data are among the first descriptions of input-output transformations in a selectively activated olfactory glomerulus.

    Beschreibung: Subduction zones impose an important control on the geochemical cycling between the surficial and internal reservoirs of the Earth. Sulphur and carbon are transferred into Earth’s mantle by subduction of pelagic sediments and altered oceanic lithosphere. Release of oxidizing sulphate- and carbonate-bearing fluids modifies the redox state of the mantle and the chemical budget of subduction zones. Yet, the mechanisms of sulphur and carbon cycling within subduction zones are still unclear, in part because data are typically derived from arc volcanoes where fluid compositions are modified during transport through the mantle wedge. We determined the bulk rock elemental, and sulphur and carbon isotope compositions of exhumed ultramafic and metabasic rocks from Syros, Greece. Comparison of isotopic data with major and trace element compositions indicates seawater alteration and chemical exchange with sediment-derived fluids within the subduction zone channel. We show that small bodies of detached slab material are subject to metasomatic processes during exhumation, in contrast to large sequences of obducted ophiolitic sections that retain their seafloor alteration signatures. In particular, fluids circulating along the plate interface can cause sulphur mobilization during several stages of exhumation within high-pressure rocks. This takes place more pervasively in serpentinites compared to mafic rocks.

    Beschreibung: The relative importance of north–south migrations of the intertropical convergence zone (ITCZ) versus El Niño-Southern Oscillation and its associated Pacific Walker Circulation (PWC) variability for past hydrological change in the western tropical Pacific is unclear. Here we show that north–south ITCZ migration was not the only mechanism of tropical Pacific hydrologic variability during the last millennium, and that PWC variability profoundly influenced tropical Pacific hydrology. We present hydrological reconstructions from Cattle Pond, Dongdao Island of the South China Sea, where multi-decadal rainfall and downcore grain size variations are correlated to the Southern Oscillation Index during the instrumental era. Our downcore grain size reconstructions indicate that this site received less precipitation during relatively warm periods, AD 1000–1400 and AD 1850–2000, compared with the cool period (AD 1400–1850). Including our new reconstructions in a synthesis of tropical Pacific records results in a spatial pattern of hydrologic variability that implicates the PWC.

    Beschreibung: Paleoclimate reconstructions are only as good as their chronology. In particular, different chronological assumptions for marine sediment cores can lead to different reconstructions of ocean ventilation age and atmosphere−ocean carbon exchange history. Here we build the first high-resolution chronology that is free of the dating uncertainties common in marine sediment records, based on radiocarbon dating twigs found with computed tomography scans in two cores from the Eastern Equatorial Pacific (EEP). With this accurate chronology, we show that the ventilation ages of the EEP thermocline and intermediate waters were similar to today during the Last Glacial Maximum and deglaciation, in contradiction with previous studies. Our results suggest that the glacial respired carbon pool in the EEP was not significantly older than today, and that the deglacial strengthening of the equatorial Pacific carbon source was probably driven by low-latitude processes rather than an increased subsurface supply of upwelled carbon from high-latitude oceans.

    Beschreibung: Hurricanes passing over the ocean can mix the water column down to great depths and resuspend massive volumes of sediments on the continental shelves. Consequently, organic carbon and reduced inorganic compounds associated with these sediments can be resuspended from anaerobic portions of the seabed and re-exposed to dissolved oxygen (DO) in the water column. This process can drive DO consumption as sediments become oxidized. Previous studies have investigated the effect of hurricanes on DO in different coastal regions of the world, highlighting the alleviation of hypoxic conditions by extreme winds, which drive vertical mixing and re-aeration of the water column. However, the effect of hurricane-induced resuspended sediments on DO has been neglected. Here, using a diverse suite of datasets for the northern Gulf of Mexico, we find that in the few days after a hurricane passage, decomposition of resuspended shelf sediments consumes up to a fifth of the DO added to the bottom of the water column during vertical mixing. Despite uncertainty in this value, we highlight the potential significance of this mechanism for DO dynamics. Overall, sediment resuspension likely occurs over all continental shelves affected by tropical cyclones, potentially impacting global cycles of marine DO and carbon.

    Beschreibung: The mechanisms of transfer of crustal material from the subducting slab to the overlying mantle wedge are still debated. Mélange rocks, formed by mixing of sediments, oceanic crust, and ultramafics along the slab-mantle interface, are predicted to ascend as diapirs from the slab-top and transfer their compositional signatures to the source region of arc magmas. However, the compositions of melts that result from the interaction of mélanges with a peridotite wedge remain unknown. Here we present experimental evidence that melting of peridotite hybridized by mélanges produces melts that carry the major and trace element abundances observed in natural arc magmas. We propose that differences in nature and relative contributions of mélanges hybridizing the mantle produce a range of primary arc magmas, from tholeiitic to calc-alkaline. Thus, assimilation of mélanges into the wedge may play a key role in transferring subduction signatures from the slab to the source of arc magmas.

    Beschreibung: The poor regenerative capacity of descending neurons is one of the main causes of the lack of recovery after spinal cord injury (SCI). Thus, it is of crucial importance to find ways to promote axonal regeneration. In addition, the prevention of retrograde degeneration leading to the atrophy/death of descending neurons is an obvious prerequisite to activate axonal regeneration. Lampreys show an amazing regenerative capacity after SCI. Recent histological work in lampreys suggested that GABA, which is massively released after a SCI, could promote the survival of descending neurons. Here, we aimed to study if GABA, acting through GABAB receptors, promotes the survival and axonal regeneration of descending neurons of larval sea lampreys after a complete SCI. First, we used in situ hybridization to confirm that identifiable descending neurons of late-stage larvae express the gabab1 subunit of the GABAB receptor. We also observed an acute increase in the expression of this subunit in descending neurons after SCI, which further supported the possible role of GABA and GABAB receptors in promoting the survival and regeneration of these neurons. So, we performed gain and loss of function experiments to confirm this hypothesis. Treatments with GABA and baclofen (GABAB agonist) significantly reduced caspase activation in descending neurons 2 weeks after a complete SCI. Long-term treatments with GABOB (a GABA analogue) and baclofen significantly promoted axonal regeneration of descending neurons after SCI. These data indicate that GABAergic signalling through GABAB receptors promotes the survival and regeneration of descending neurons after SCI. Finally, we used morpholinos against the gabab1 subunit to knockdown the expression of the GABAB receptor in descending neurons. Long-term morpholino treatments caused a significant inhibition of axonal regeneration. This shows that endogenous GABA promotes axonal regeneration after a complete SCI in lampreys by activating GABAB receptors.

    Beschreibung: Tectonic landforms reveal that the West Antarctic Ice Sheet (WAIS) lies atop a major volcanic rift system. However, identifying subglacial volcanism is challenging. Here we show geochemical evidence of a volcanic heat source upstream of the fast-melting Pine Island Ice Shelf, documented by seawater helium isotope ratios at the front of the Ice Shelf cavity. The localization of mantle helium to glacial meltwater reveals that volcanic heat induces melt beneath the grounded glacier and feeds the subglacial hydrological network crossing the grounding line. The observed transport of mantle helium out of the Ice Shelf cavity indicates that volcanic heat is supplied to the grounded glacier at a rate of ~ 2500 ± 1700 MW, which is ca. half as large as the active Grimsvötn volcano on Iceland. Our finding of a substantial volcanic heat source beneath a major WAIS glacier highlights the need to understand subglacial volcanism, its hydrologic interaction with the marine margins, and its potential role in the future stability of the WAIS.

    Beschreibung: The abundance of organic carbon (OC) in vegetation and soils (~2,600 PgC) compared to carbon in the atmosphere (~830 PgC) highlights the importance of terrestrial OC in global carbon budgets. The residence time of OC in continental reservoirs, which sets the rates of carbon exchange between land and atmosphere, represents a key uncertainty in global carbon cycle dynamics. Retention of terrestrial OC can also distort bulk OC- and biomarker-based paleorecords, yet continental storage timescales remain poorly quantified. Using “bomb” radiocarbon (14C) from thermonuclear weapons testing as a tracer, we model leaf-wax fatty acid and bulk OC 14C signatures in a river-proximal marine sediment core from the Bay of Bengal in order to constrain OC storage timescales within the Ganges-Brahmaputra (G-B) watershed. Our model shows that 79–83% of the leaf-waxes in this core were stored in continental reservoirs for an average of 1,000–1,200 calendar years, while the remainder was stored for an average of 15 years. This age structure distorts high-resolution organic paleorecords across geologically rapid events, highlighting that compound-specific proxy approaches must consider storage timescales. Furthermore, these results show that future environmental change could destabilize large stores of old - yet reactive - OC currently stored in tropical basins.

    Beschreibung: Agricultural intensification offers potential to grow more food while reducing the conversion of native ecosystems to croplands. However, intensification also risks environmental degradation through emissions of the greenhouse gas nitrous oxide (N2O) and nitrate leaching to ground and surface waters. Intensively-managed croplands and nitrogen (N) fertilizer use are expanding rapidly in tropical regions. We quantified fertilizer responses of maize yield, N2O emissions, and N leaching in an Amazon soybean-maize double-cropping system on deep, highly-weathered soils in Mato Grosso, Brazil. Application of N fertilizer above 80 kg N ha−1 yr−1 increased maize yield and N2O emissions only slightly. Unlike experiences in temperate regions, leached nitrate accumulated in deep soils with increased fertilizer and conversion to cropping at N fertilization rates 〉80 kg N ha−1, which exceeded maize demand. This raises new questions about the capacity of tropical agricultural soils to store nitrogen, which may determine when and how much nitrogen impacts surface waters.

    Beschreibung: Haynesina germanica, an ubiquitous benthic foraminifer in intertidal mudflats, has the remarkable ability to isolate, sequester, and use chloroplasts from microalgae. The photosynthetic functionality of these kleptoplasts has been demonstrated by measuring photosystem II quantum efficiency and O2 production rates, but the precise role of the kleptoplasts in foraminiferal metabolism is poorly understood. Thus, the mechanism and dynamics of C and N assimilation and translocation from the kleptoplasts to the foraminiferal host requires study. The objective of this study was to investigate, using correlated TEM and NanoSIMS imaging, the assimilation of inorganic C and N (here ammonium, NH4+) in individuals of a kleptoplastic benthic foraminiferal species. H. germanica specimens were incubated for 20 h in artificial seawater enriched with H13CO3− and 15NH4+ during a light/dark cycle. All specimens (n = 12) incorporated 13C into their endoplasm stored primarily in the form of lipid droplets. A control incubation in darkness resulted in no 13C-uptake, strongly suggesting that photosynthesis is the process dominating inorganic C assimilation. Ammonium assimilation was observed both with and without light, with diffuse 15N-enrichment throughout the cytoplasm and distinct 15N-hotspots in fibrillar vesicles, electron-opaque bodies, tubulin paracrystals, bacterial associates, and, rarely and at moderate levels, in kleptoplasts. The latter observation might indicate that the kleptoplasts are involved in N assimilation. However, the higher N assimilation observed in the foraminiferal endoplasm incubated without light suggests that another cytoplasmic pathway is dominant, at least in darkness. This study clearly shows the advantage provided by the kleptoplasts as an additional source of carbon and provides observations of ammonium uptake by the foraminiferal cell.

    Beschreibung: In mammals, a complex array of oral sensors assess the taste, temperature and haptic properties of food. Although the representation of taste has been extensively studied in the gustatory cortex, it is unclear how the somatosensory cortex encodes information about the properties of oral stimuli. Moreover, it is poorly understood how different oral sensory modalities are integrated and how sensory responses are translated into oral motor actions. To investigate whether oral somatosensory cortex processes food-related sensations and movements, we performed in vivo whole-cell recordings and motor mapping experiments in rats. Neurons in oral somatosensory cortex showed robust post-synaptic and sparse action potential responses to air puffs. Membrane potential showed that cold water evoked larger responses than room temperature or hot water. Most neurons showed no clear tuning of responses to bitter, sweet and neutral gustatory stimuli. Finally, motor mapping experiments with histological verification revealed an initiation of movements related to food consumption behavior, such as jaw opening and tongue protrusions. We conclude that somatosensory cortex: (i) provides a representation of the temperature of oral stimuli, (ii) does not systematically encode taste information and (iii) influences orofacial movements related to food consummatory behavior.

    Beschreibung: Subduction zone magmas are more oxidised on eruption than those at mid-ocean ridges. This is attributed either to oxidising components, derived from subducted lithosphere (slab) and added to the mantle wedge, or to oxidation processes occurring during magma ascent via differentiation. Here we provide direct evidence for contributions of oxidising slab agents to melts trapped in the sub-arc mantle. Measurements of sulfur (S) valence state in sub-arc mantle peridotites identify sulfate, both as crystalline anhydrite (CaSO4) and dissolved SO42− in spinel-hosted glass (formerly melt) inclusions. Copper-rich sulfide precipitates in the inclusions and increased Fe3+/∑Fe in spinel record a S6+–Fe2+ redox coupling during melt percolation through the sub-arc mantle. Sulfate-rich glass inclusions exhibit high U/Th, Pb/Ce, Sr/Nd and δ34S (+ 7 to + 11‰), indicating the involvement of dehydration products of serpentinised slab rocks in their parental melt sources. These observations provide a link between liberated slab components and oxidised arc magmas.

    Beschreibung: Tidal wetlands produce long-term soil organic carbon (C) stocks. Thus for carbon accounting purposes, we need accurate and precise information on the magnitude and spatial distribution of those stocks. We assembled and analyzed an unprecedented soil core dataset, and tested three strategies for mapping carbon stocks: applying the average value from the synthesis to mapped tidal wetlands, applying models fit using empirical data and applied using soil, vegetation and salinity maps, and relying on independently generated soil carbon maps. Soil carbon stocks were far lower on average and varied less spatially and with depth than stocks calculated from available soils maps. Further, variation in carbon density was not well-predicted based on climate, salinity, vegetation, or soil classes. Instead, the assembled dataset showed that carbon density across the conterminous united states (CONUS) was normally distributed, with a predictable range of observations. We identified the simplest strategy, applying mean carbon density (27.0 kg C m−3), as the best performing strategy, and conservatively estimated that the top meter of CONUS tidal wetland soil contains 0.72 petagrams C. This strategy could provide standardization in CONUS tidal carbon accounting until such a time as modeling and mapping advancements can quantitatively improve accuracy and precision.

    Beschreibung: The oceans are warming and coral reefs are bleaching with increased frequency and severity, fueling concerns for their survival through this century. Yet in the central equatorial Pacific, some of the world’s most productive reefs regularly experience extreme heat associated with El Niño. Here we use skeletal signatures preserved in long-lived corals on Jarvis Island to evaluate the coral community response to multiple successive heatwaves since 1960. By tracking skeletal stress band formation through the 2015-16 El Nino, which killed 95% of Jarvis corals, we validate their utility as proxies of bleaching severity and show that 2015-16 was not the first catastrophic bleaching event on Jarvis. Since 1960, eight severe (〉30% bleaching) and two moderate (〈30% bleaching) events occurred, each coinciding with El Niño. While the frequency and severity of bleaching on Jarvis did not increase over this time period, 2015–16 was unprecedented in magnitude. The trajectory of recovery of this historically resilient ecosystem will provide critical insights into the potential for coral reef resilience in a warming world.

    Beschreibung: During recent years, rapid seasonal growth of macroalgae covered extensive areas within the Yellow Sea, developing the world’s most spatially extensive “green tide”. The remarkably fast accumulation of macroalgal biomass is the joint result of high nitrogen supplies in Yellow Sea waters, plus ability of the macroalgae to optionally use C4 photosynthetic pathways that facilitate rapid growth. Stable isotopic evidence shows that the high nitrogen supply is derived from anthropogenic sources, conveyed from watersheds via river discharges, and by direct atmospheric deposition. Wastewater and manures supply about half the nitrogen used by the macroalgae, fertiliser and atmospheric deposition each furnish about a quarter of the nitrogen in macroalgae. The massive green tides affecting the Yellow Sea are likely to increase, with significant current and future environmental and human consequences. Addressing these changing trajectories will demand concerted investment in new basic and applied research as the basis for developing management policies.

    Beschreibung: Sunlight is the dominant control on phytoplankton biosynthetic activity, and darkness deprives them of their primary external energy source. Changes in the biochemical composition of phytoplankton communities over diel light cycles and attendant consequences for carbon and energy flux in environments remain poorly elucidated. Here we use lipidomic data from the North Pacific subtropical gyre to show that biosynthesis of energy-rich triacylglycerols (TAGs) by eukaryotic nanophytoplankton during the day and their subsequent consumption at night drives a large and previously uncharacterized daily carbon cycle. Diel oscillations in TAG concentration comprise 23 ± 11% of primary production by eukaryotic nanophytoplankton representing a global flux of about 2.4 Pg C yr−1. Metatranscriptomic analyses of genes required for TAG biosynthesis indicate that haptophytes and dinoflagellates are active members in TAG production. Estimates suggest that these organisms could contain as much as 40% more calories at sunset than at sunrise due to TAG production.

    Beschreibung: The orbital-scale timing of South Asian monsoon (SAM) precipitation is poorly understood. Here we present new SST and seawater δ18O (δ18Osw) records from the Bay of Bengal, the core convective region of the South Asian monsoon, over the past 1 million years. Our records reveal that SAM precipitation peaked in the precession band ~9 kyrs after Northern Hemisphere summer insolation maxima, in phase with records of SAM winds in the Arabian Sea and eastern Indian Ocean. Precession-band variance, however, accounts for ~30% of the total variance of SAM precipitation while it was either absent or dominant in records of the East Asian monsoon (EAM). This and the observation that SAM precipitation was phase locked with obliquity minima and was sensitive to Southern Hemisphere warming provides clear evidence that SAM and EAM precipitation responded differently to orbital forcing and highlights the importance of internal processes forcing monsoon variability.

    Beschreibung: Conduit geometry affects magma ascent dynamics and, consequently, the style and evolution of volcanic eruptions. However, despite geological evidences support the occurrence of conduit widening during most volcanic eruptions, the factors controlling conduit enlargement are still unclear, and the effects of syn-eruptive variations of conduit geometry have not been investigated in depth yet. Based on numerical modeling and the application of appropriate stability criteria, we found out a strong relationship between magma rheology and conduit stability, with significant effects on eruptive dynamics. Indeed, in order to be stable, conduits feeding dacitic/rhyolitic eruptions need larger diameters respect to their phonolitic/trachytic counterparts, resulting in the higher eruption rates commonly observed in dacitic/rhyolitic explosive events. Thus, in addition to magma source conditions and viscosity-dependent efficiency for outgassing, we suggest that typical eruption rates for different magma types are also controlled by conduit stability. Results are consistent with a compilation of volcanological data and selected case studies. As stability conditions are not uniform along the conduit, widening is expected to vary in depth, and three axisymmetric geometries with depth-dependent radii were investigated. They are able to produce major modifications in eruptive parameters, suggesting that eruptive dynamics is influenced by syn-eruptive changes in conduit geometry.

    Beschreibung: During the MW 5.7 and 6.0 Umbria–Marche earthquakes of 1997 September 26, the historical centre of Nocera Umbra suffered MCS intensity VII–VIII. The zone is located on the top of a hill, a condition potentially favourable to ground motion amplification. However, also vulnerability is higher on the hill because of the ancient age of buildings. A temporary array of eight seismological stations was installed across the hill to quantify the amplification effect due to topography.Waveforms of 14 aftershocks (2.6〈ML 〈4.1) are selected for the analysis. During each earthquake the largest amplitudes are observed on the hilltop, spectral ratios are computed using rotated horizontal components to search for directional effects. Amplifications are found in two separate frequency bands: one in the range 2–4 Hz, where the increase of amplitude is moderate (never exceeding a factor of 4) and the polarization is transversal to the hill major axis; the second above 10 Hz, where amplifications are larger and reach values as high as 25 Hz. High-frequency polarization varies for different sites and frequencies suggesting that smaller-scale complexities control the high frequency response. Synthetic seismograms of 2-D models confirm the occurrence of amplification, although not all details are fit by numerical simulations and the agreement between observations and models is significant only in terms of the fundamental resonance frequency, around 3 Hz. In the models, amplifications are much smaller than the observed ones. We conclude that topography could have been responsible for a small increase of damage in the hill zone but the most significant role on damage was played by the locally higher vulnerability.

    Beschreibung: The shallow subsurface structure of the 2009 April 6 Mw 6.3 L’Aquila earthquake surface rupture at Paganica has been investigated with ground penetrating radar to study how the surface rupture relates spatially to previous surface displacements during the Holocene and Pleistocene. The discontinuous surface rupture stepped between en-echelon/parallel faults within the overall fault zone that show clear Holocene/Pleistocene offsets in the top 10 m of the subsurface. Some portions of the fault zone that show clear Holocene offsets were not ruptured in 2009, having been bypassed as the rupture stepped across a relay zone onto a fault across strike. The slip vectors, defined by opening directions across surface cracks, indicate dip-slip normal movement, whose azimuth remained constant between 210◦ and 228◦ across the zone where the rupture stepped between faults. We interpret maximum vertical offsets of the base of the Holocene summed across strike to be 4.5 m, which if averaged over 15 kyr, gives a maximum throw-rate of 0.23–0.30 mm yr–1, consistent with throw-rates implied by vertical offsets of a layer whose age we assume to be ∼33 ka. This compares with published values of 0.4 mm yr–1 for a minimum slip rate implied by offsets of Middle Pleistocene tephras, and 0.24 mm yr–1 since 24.8 kyr from palaeoseismology. The Paganica Fault, although clearly an important active structure, is not slipping fast enough to accommodate all of the 3–5 mm yr–1 of extension across this sector of the Apennines; other neighbouring range-bounding active normal faults also have a role to play in the seismic hazard.

    Beschreibung: Six velocity sections straddling Cape Hatteras show a deep counterflow rounding the Cape wedged beneath the poleward flowing Gulf Stream and the continental slope. This counterflow is likely the upper part of the equatorward-flowing Deep Western Boundary Current (DWBC). Hydrographic data suggest that the equatorward flow sampled by the shipboard 38 kHz ADCP comprises the Upper Labrador Sea Water (ULSW) layer and top of the Classical Labrador Sea Water (CLSW) layer. Continuous DWBC flow around the Cape implied by the closely-spaced velocity sections here is also corroborated by the trajectory of an Argo float. These findings contrast with previous studies based on floats and tracers in which the lightest DWBC constituents did not follow the boundary to cross under the Gulf Stream at Cape Hatteras but were diverted into the interior as the DWBC encountered the Gulf Stream in the crossover region. Additionally, our six quasi-synoptic velocity sections confirm that the Gulf Stream intensified markedly at that time as it approached the separation point and flowed into deeper waters. Downstream increases were observed not only in the poleward transport across the sections but also in the current’s maximum speed.

    Beschreibung: Efforts to estimate the physical and economic impacts of future climate change face substantial challenges. To enrich the currently popular approaches to impact analysis—which involve evaluation of a damage function or multi-model comparisons based on a limited number of standardized scenarios—we propose integrating a geospatially resolved physical representation of impacts into a coupled human-Earth system modeling framework. Large internationally coordinated exercises cannot easily respond to new policy targets and the implementation of standard scenarios across models, institutions and research communities can yield inconsistent estimates. Here, we argue for a shift toward the use of a self-consistent integrated modeling framework to assess climate impacts, and discuss ways the integrated assessment modeling community can move in this direction. We then demonstrate the capabilities of such a modeling framework by conducting a multi-sectoral assessment of climate impacts under a range of consistent and integrated economic and climate scenarios that are responsive to new policies and business expectations.

    Beschreibung: Despite concerted international effort to track and interpret shifts in the abundance and distribution of Adélie penguins, large populations continue to be identified. Here we report on a major hotspot of Adélie penguin abundance identified in the Danger Islands off the northern tip of the Antarctic Peninsula (AP). We present the first complete census of Pygoscelis spp. penguins in the Danger Islands, estimated from a multi-modal survey consisting of direct ground counts and computer-automated counts of unmanned aerial vehicle (UAV) imagery. Our survey reveals that the Danger Islands host 751,527 pairs of Adélie penguins, more than the rest of AP region combined, and include the third and fourth largest Adélie penguin colonies in the world. Our results validate the use of Landsat medium-resolution satellite imagery for the detection of new or unknown penguin colonies and highlight the utility of combining satellite imagery with ground and UAV surveys. The Danger Islands appear to have avoided recent declines documented on the Western AP and, because they are large and likely to remain an important hotspot for avian abundance under projected climate change, deserve special consideration in the negotiation and design of Marine Protected Areas in the region.

    Beschreibung: Mutations in Fused in Sarcoma/Translocated in Liposarcoma (FUS) cause familial forms of amyotrophic lateral sclerosis (ALS), a neurodegenerative disease characterized by progressive axonal degeneration mainly affecting motor neurons. Evidence from transgenic mouse models suggests mutant forms of FUS exert an unknown gain-of-toxic function in motor neurons, but mechanisms underlying this effect remain unknown. Towards this end, we studied the effect of wild type FUS (FUS WT) and three ALS-linked variants (G230C, R521G and R495X) on fast axonal transport (FAT), a cellular process critical for appropriate maintenance of axonal connectivity. All ALS-FUS variants impaired anterograde and retrograde FAT in squid axoplasm, whereas FUS WT had no effect. Misfolding of mutant FUS is implicated in this process, as the molecular chaperone Hsp110 mitigated these toxic effects. Interestingly, mutant FUS-induced impairment of FAT in squid axoplasm and of axonal outgrowth in mammalian primary motor neurons involved aberrant activation of the p38 MAPK pathway, as also reported for ALS-linked forms of Cu, Zn superoxide dismutase (SOD1). Accordingly, increased levels of active p38 MAPK were detected in post-mortem human ALS-FUS brain tissues. These data provide evidence for a novel gain-of-toxic function for ALS-linked FUS involving p38 MAPK activation.

    Beschreibung: The ocean’s role in global climate change largely depends on its heat transport. Therefore, understanding the oceanic meridional heat transport (MHT) variability is a fundamental issue. Prevailing observational and modeling evidence suggests that MHT variability is primarily determined by the large-scale ocean circulation. Here, using new in situ observations in the eastern subpolar North Atlantic Ocean and an eddy-resolving numerical model, we show that energetic mesoscale eddies with horizontal scales of about 10–100 km profoundly modulate MHT variability on time scales from intra-seasonal to interannual. Our results reveal that the velocity changes due to mesoscale processes produce substantial variability for the MHT regionally (within sub-basins) and the subpolar North Atlantic as a whole. The findings have important implications for understanding the mechanisms for poleward heat transport variability in the subpolar North Atlantic Ocean, a key region for heat and carbon sequestration, ice–ocean interaction, and biological productivity.

    Beschreibung: In mammals, spinal cord injury (SCI) leads to dramatic losses in neurons and synaptic connections, and consequently function. Unlike mammals, lampreys are vertebrates that undergo spontaneous regeneration and achieve functional recovery after SCI. Therefore our goal was to determine the complete transcriptional responses that occur after SCI in lampreys and to identify deeply conserved pathways that promote regeneration. We performed RNA-Seq on lamprey spinal cord and brain throughout the course of functional recovery. We describe complex transcriptional responses in the injured spinal cord, and somewhat surprisingly, also in the brain. Transcriptional responses to SCI in lampreys included transcription factor networks that promote peripheral nerve regeneration in mammals such as Atf3 and Jun. Furthermore, a number of highly conserved axon guidance, extracellular matrix, and proliferation genes were also differentially expressed after SCI in lampreys. Strikingly, ~3% of differentially expressed transcripts belonged to the Wnt pathways. These included members of the Wnt and Frizzled gene families, and genes involved in downstream signaling. Pharmacological inhibition of Wnt signaling inhibited functional recovery, confirming a critical role for this pathway. These data indicate that molecular signals present in mammals are also involved in regeneration in lampreys, supporting translational relevance of the model.

    Beschreibung: Because microbial plankton in the ocean comprise diverse bacteria, algae, and protists that are subject to environmental forcing on multiple spatial and temporal scales, a fundamental open question is to what extent these organisms form ecologically cohesive communities. Here we show that although all taxa undergo large, near daily fluctuations in abundance, microbial plankton are organized into clearly defined communities whose turnover is rapid and sharp. We analyze a time series of 93 consecutive days of coastal plankton using a technique that allows inference of communities as modular units of interacting taxa by determining positive and negative correlations at different temporal frequencies. This approach shows both coordinated population expansions that demarcate community boundaries and high frequency of positive and negative associations among populations within communities. Our analysis thus highlights that the environmental variability of the coastal ocean is mirrored in sharp transitions of defined but ephemeral communities of organisms.

    Beschreibung: Sediments in deep ocean trenches may contain crucial information on past earthquake history and constitute important sites of carbon burial. Here we present 14C data on bulk organic carbon (OC) and its thermal decomposition fractions produced by ramped pyrolysis/oxidation for a core retrieved from the 〉7.5 km-deep Japan Trench. High-resolution 14C measurements, coupled with distinctive thermogram characteristics of OC, reveal hemipelagic sedimentation interrupted by episodic deposition of pre-aged OC in the trench. Low δ13C values and diverse 14C ages of thermal fractions imply that the latter material originates from the adjacent margin, and the co-occurrence of pre-aged OC with intervals corresponding to known earthquake events implies tectonically triggered, gravity-flow-driven supply. We show that 14C ages of thermal fractions can yield valuable chronological constraints on sedimentary sequences. Our findings shed new light on links between tectonically driven sedimentological processes and marine carbon cycling, with implications for carbon dynamics in hadal environments.

    Beschreibung: The assembly of membranous extensions such as microvilli and cilia in polarized cells is a tightly regulated, yet poorly understood, process. Peptidylglycine α-amidating monooxygenase (PAM), a membrane enzyme essential for the synthesis of amidated bioactive peptides, was recently identified in motile and non-motile (primary) cilia and has an essential role in ciliogenesis in Chlamydomonas, Schmidtea and mouse. In mammalian cells, changes in PAM levels alter secretion and organization of the actin cytoskeleton. Here we show that lack of Pam in zebrafish recapitulates the lethal edematous phenotype observed in Pam−/− mice and reveals additional defects. The pam−/− zebrafish embryos display an initial striking loss of microvilli and subsequently impaired ciliogenesis in the pronephros. In multiciliated mouse tracheal epithelial cells, vesicular PAM staining colocalizes with apical actin, below the microvilli. In PAM-deficient Chlamydomonas, the actin cytoskeleton is dramatically reorganized, and expression of an actin paralogue is upregulated. Biochemical assays reveal that the cytosolic PAM C-terminal domain interacts directly with filamentous actin but does not alter the rate of actin polymerization or disassembly. Our results point to a critical role for PAM in organizing the actin cytoskeleton during development, which could in turn impact both microvillus formation and ciliogenesis.

    Beschreibung: The rock-hosted subseafloor crustal aquifer harbors a reservoir of microbial life that may influence global marine biogeochemical cycles. Here we utilized metagenomic libraries of crustal fluid samples from North Pond, located on the flanks of the Mid-Atlantic Ridge, a site with cold, oxic subseafloor fluid circulation within the upper basement to query microbial diversity. Twenty-one samples were collected during a 2-year period to examine potential microbial metabolism and community dynamics. We observed minor changes in the geochemical signatures over the 2 years, yet the microbial community present in the crustal fluids underwent large shifts in the dominant taxonomic groups. An analysis of 195 metagenome-assembled genomes (MAGs) were generated from the data set and revealed a connection between litho- and autotrophic processes, linking carbon fixation to the oxidation of sulfide, sulfur, thiosulfate, hydrogen, and ferrous iron in members of the Proteobacteria, specifically the Alpha-, Gamma- and Zetaproteobacteria, the Epsilonbacteraeota and the Planctomycetes. Despite oxic conditions, analysis of the MAGs indicated that members of the microbial community were poised to exploit hypoxic or anoxic conditions through the use of microaerobic cytochromes, such as cbb3- and bd-type cytochromes, and alternative electron acceptors, like nitrate and sulfate. Temporal and spatial trends from the MAGs revealed a high degree of functional redundancy that did not correlate with the shifting microbial community membership, suggesting functional stability in mediating subseafloor biogeochemical cycles. Collectively, the repeated sampling at multiple sites, together with the successful binning of hundreds of genomes, provides an unprecedented data set for investigation of microbial communities in the cold, oxic crustal aquifer.

    Beschreibung: Temperate coastal marine environments are replete with complex biotic and abiotic interactions that are amplified during spring and summer phytoplankton blooms. During these events, heterotrophic bacterioplankton respond to successional releases of dissolved organic matter as algal cells are lysed. Annual seasonal shifts in the community composition of free-living bacterioplankton follow broadly predictable patterns, but whether similar communities respond each year to bloom disturbance events remains unknown owing to a lack of data sets, employing high-frequency sampling over multiple years. We capture the fine-scale microdiversity of these events with weekly sampling using a high-resolution method to discriminate 16S ribosomal RNA gene amplicons that are 〉99% identical. Furthermore, we used 2 complete years of data to facilitate identification of recurrent sub-networks of co-varying microbes. We demonstrate that despite inter-annual variation in phytoplankton blooms and despite the dynamism of a coastal–oceanic transition zone, patterns of microdiversity are recurrent during both bloom and non-bloom conditions. Sub-networks of co-occurring microbes identified reveal that correlation structures between community members appear quite stable in a seasonally driven response to oligotrophic and eutrophic conditions.

    Beschreibung: An influence of solar irradiance variations on Earth’s surface climate has been repeatedly suggested, based on correlations between solar variability and meteorological variables1. Specifically, weaker westerly winds have been observed in winters with a less active sun, for example at the minimum phase of the 11-year sunspot cycle2, 3, 4. With some possible exceptions5, 6, it has proved difficult for climate models to consistently reproduce this signal7, 8. Spectral Irradiance Monitor satellite measurements indicate that variations in solar ultraviolet irradiance may be larger than previously thought9. Here we drive an ocean–atmosphere climate model with ultraviolet irradiance variations based on these observations. We find that the model responds to the solar minimum with patterns in surface pressure and temperature that resemble the negative phase of the North Atlantic or Arctic Oscillation, of similar magnitude to observations. In our model, the anomalies descend through the depth of the extratropical winter atmosphere. If the updated measurements of solar ultraviolet irradiance are correct, low solar activity, as observed during recent years, drives cold winters in northern Europe and the United States, and mild winters over southern Europe and Canada, with little direct change in globally averaged temperature. Given the quasiregularity of the 11-year solar cycle, our findings may help improve decadal climate predictions for highly populated extratropical regions

    Beschreibung: The anaerobic oxidation of methane (AOM) with sulphate, an area currently generating great interest in microbiology, is accomplished by consortia of methanotrophic archaea (ANME) and sulphate-reducing bacteria1, 2. The enzyme activating methane in methanotrophic archaea has tentatively been identified as a homologue of methyl-coenzyme M reductase (MCR) that catalyses the methane-forming step in methanogenic archaea3, 4. Here we report an X-ray structure of the 280 kDa heterohexameric ANME-1 MCR complex. It was crystallized uniquely from a protein ensemble purified from consortia of microorganisms collected with a submersible from a Black Sea mat catalysing AOM with sulphate4. Crystals grown from the heterogeneous sample diffract to 2.1 Å resolution and consist of a single ANME-1 MCR population, demonstrating the strong selective power of crystallization. The structure revealed ANME-1 MCR in complex with coenzyme M and coenzyme B, indicating the same substrates for MCR from methanotrophic and methanogenic archaea. Differences between the highly similar structures of ANME-1 MCR and methanogenic MCR include a F430 modification, a cysteine-rich patch and an altered post-translational amino acid modification pattern, which may tune the enzymes for their functions in different biological contexts.

    Beschreibung: Biological N2 fixation is an important part of the marine nitrogen cycle as it provides a source of new nitrogen that can support biological carbon export and sequestration. Research in the past decade has focused on determining the patterns of distribution and abundance of diazotrophs, defining the environmental features leading to these patterns and characterizing the factors that constrain marine N2 fixation overall. In this Review, we describe how variations in the deposition of iron from dust to different ocean basins affects the limiting nutrient for N2 fixation and the distribution of different diazotrophic species. However, many questions remain about marine N2 fixation, including the role of temperature, fixed nitrogen species, CO2 and physical forcing in controlling N2 fixation, as well as the potential for heterotrophic N2 fixation.

    Beschreibung: To enable quality control of measurement procedures for determinations of Mg isotope amount ratios, expressed as δ26Mg and δ25Mg values, in Earth-surface studies, the δ26Mg and δ25Mg values of eight reference materials (RMs) were determined by inter-laboratory comparison between five laboratories and considering published data, if available. These matrix RMs, including river water SLRS-5, spring water NIST SRM 1640a, Dead Sea brine DSW-1, dolomites JDo-1 and CRM 512, limestone CRM 513, soil NIST SRM 2709a and vegetation NIST SRM 1515 apple leaves, are representative for a wide range of Earth-surface materials from low-temperature environments. The inter-laboratory variability, 2s (twice the standard deviation), of all eight RMs ranges from 0.05 to 0.17‰ in δ26Mg. Thus, it is suggested that all these materials are suitable for validation of δ26Mg and δ25Mg determinations of Earth-surface geochemical studies.

    Beschreibung: The Pacific sector of Antarctica, including both the Antarctic Peninsula and continental West Antarctica, has experienced substantial warming in the past 30 years. An increase in the circumpolar westerlies, owing in part to the decline in stratospheric ozone concentrations since the late 1970s, may account for warming trends in the peninsula region in austral summer and autumn. The more widespread warming in continental West Antarctica (Ellsworth Land and Marie Byrd Land) occurs primarily in austral winter and spring, and remains unexplained. Here we use observations of Antarctic surface temperature and global sea surface temperature, and atmospheric circulation data to show that recent warming in continental West Antarctica is linked to sea surface temperature changes in the tropical Pacific. Over the past 30 years, anomalous sea surface temperatures in the central tropical Pacific have generated an atmospheric Rossby wave response that influences atmospheric circulation over the Amundsen Sea, causing increased advection of warm air to the Antarctic continent. General circulation model experiments show that the central tropical Pacific is a critical region for producing the observed high latitude response. We conclude that, by affecting the atmospheric circulation at high southern latitudes, increasing tropical sea surface temperatures may account for West Antarctic warming through most of the twentieth century.

    Beschreibung: Teleost fish grow continuously throughout their lifespan, and this growth includes visual system components: eyes, optic nerves, and brain. As fish grow, the optic nerve lengthens and neural signals must travel increasing distances from the eye to the optic tectum along thousands of retinal ganglion cell (RGC) axons. Larger fish have better vision that enhances their ability to capture prey, but they are faced with the potential computational problem of changes in the relative timing of visual information arriving at the brain. Optic nerve conduction delays depend on RGC axon conduction velocities, and velocity is primarily determined by axon diameters. If axon diameters do not increase in proportion to body length, then absolute and relative conduction delays will vary with fish size. We have measured optic nerve lengths and axon diameter distributions in different sized zebrafish (Danio rerio) and goldfish (Carassius auratus) and find that, as both species of fish grow, axon diameters increase to reduce average conduction delays by about half and to keep relative delays constant. This invariance of relative conduction delays simplifies computational problems faced by the optic tectum.

    Beschreibung: As sessile filter feeders, sponges rely on a highly efficient fluid transport system. Their physiology depends on efficient water exchange, which is performed by the aquiferous system. This prominent poriferan anatomical character represents a dense network of incurrent and excurrent canals on which we lack detailed 3D models. To overcome this, we investigated the complex leucon‐type architecture in the demosponge Tethya wilhelma using corrosion casting, microtomography, and 3D reconstructions. Our integrative qualitative and quantitative approach allowed us to create, for the first time, high‐resolution 3D representations of entire canal systems which were used for detailed geometric and morphometric measurements. Canal diameters lack distinct size classes, and bifurcations are non‐uniformly ramified. A relatively high number of bifurcations show previously unknown and atypical cross‐sectional area ratios. Scaling properties and topological patterns of the canals indicate a more complex overall architecture than previously assumed. As a consequence, it might be more convenient to group canals into functional units rather than hierarchical clusters. Our data qualify the leucon canal system architecture of T. wilhelma as a highly efficient fluid transport system adapted toward minimal flow resistance. Our results and approach are relevant for a better understanding of sponge biology and cultivation techniques.

    Beschreibung: The tephrostratigraphy of lake sediments in the Endinger Bruch provides the first robust age model for the Lateglacial palynological records of Vorpommern (north-east Germany). Cryptotephra investigations revealed six tephra layers within sediments spanning from Open vegetation phase I (∼Bølling, ∼15 ka) to the Early Holocene Betula/Pinus forest phase (∼Pre-boreal, ∼10.5 ka). Four of these layers have been correlated with previously described tephra layers found in sites across Europe. The Laacher See Tephra (Eifel Volcanic Field) is present in very high concentrations within sediments of the Lateglacial Betula (/Pinus) forest phase (∼Allerød). The Vedde Ash (Iceland) lies midway through Open vegetation phase III (∼Younger Dryas). The Hässeldalen and the Askja tephras (Iceland) lie in the Early Holocene Betula/Pinus forest phase (∼Preboreal). These tephra layers have independently derived age estimates, which have been imported into the Endinger Bruch record. Furthermore, the layers facilitate direct correlation of the regional vegetation record with other palaeoenvironmental archives, which contain one or more of the same tephra layers, from Greenland to Southern Europe. In doing this, localized variations are confirmed in some aspects of the pollen stratigraphy; however, transitions between the main vegetation phases appear to occur synchronously (within centennial errors) with the equivalent environmental transitions observed in sites across the European continent.

    Beschreibung: Climate warming at the end of the last glaciation caused ice caps on Icelandic volcanoes to retreat. Removal of surface ice load is thought to have decreased pressures in the underlying mantle, triggering decompression melting, enhanced magma generation and increased volcanic activity1–3. Present-day climate change could have the same effect, although there may be a time lag of hundreds of years between magma generation and eruption4,5. However, in addition to increased magma generation, pressure changes associated with ice retreat should also alter the capacity for storing magma within the crust. Here we use a numerical model to evaluate the effect of the current decrease in ice load on magma storage in the crust at the Kverkfjöll volcanic system, located partially beneath Iceland’s largest ice cap. We compare the model results with radar and global positioning system measurements of surface displacement and changes in crustal stress between 2007 and 2008, during the intrusion of a deep dyke at Upptyppingar. We find that although the main component of stress recorded during dyke intrusion relates to plate extension, another component of stress is consistent with the stress field caused by the retreating ice cap. We conclude that the retreating ice cap led to enhanced capture of magma within the crust. We suggest that ice-cap retreat can promote magma storage, rather than eruption, at least in the short term.

    Beschreibung: Populations of fishes provide valuable services for billions of people, but face diverse and interacting threats that jeopardize their sustainability. Human population growth and intensifying resource use for food, water, energy and goods are compromising fish populations through a variety of mechanisms, including overfishing, habitat degradation and declines in water quality. The important challenges raised by these issues have been recognized and have led to considerable advances over past decades in managing and mitigating threats to fishes worldwide. In this review, we identify the major threats faced by fish populations alongside recent advances that are helping to address these issues. There are very significant efforts worldwide directed towards ensuring a sustainable future for the world's fishes and fisheries and those who rely on them. Although considerable challenges remain, by drawing attention to successful mitigation of threats to fish and fisheries we hope to provide the encouragement and direction that will allow these challenges to be overcome in the future.

    Beschreibung: There have been decades, such as 2000–2009, when the observed globally averaged surface-temperature time series shows little increase or even a slightly negative trend1 (a hiatus period). However, the observed energy imbalance at the top-of-atmosphere for this recent decade indicates that a net energy flux into the climate system of about 1 W m−2 (refs 2, 3) should be producing warming somewhere in the system4,5. Here we analyse twenty-first-century climate-model simulations that maintain a consistent radiative imbalance at the top-of-atmosphere of about 1 W m−2 as observed for the past decade. Eight decades with a slightly negative global mean surface-temperature trend show that the ocean above 300 m takes up significantly less heat whereas the ocean below 300 m takes up significantly more, compared with non-hiatus decades. The model provides a plausible depiction of processes in the climate system causing the hiatus periods, and indicates that a hiatus period is a relatively common climate phenomenon and may be linked to La Niña-like conditions.

    Beschreibung: Mass fractions of Sn and In were determined in sixteen geological reference materials including basaltic/mafic (BCR‐2, BE‐N, BHVO‐1, BHVO‐2, BIR‐1, OKUM, W‐2, WS‐E), ultramafic (DTS‐2b, MUH‐1, PCC‐1, UB‐N) and felsic/sedimentary reference materials (AGV‐2, JA‐1, SdAR‐M2, SdAR‐H1). Extensive digestion and ion exchange separation tests were carried out in order to provide high yields (〉 90% for Sn, 〉 85% for In), low total procedural blanks (~ 1 ng for Sn, 〈 3 pg for In) and low analytical uncertainties for the elements of interest in a variety of silicate sample matrices. Replicate analyses (n = 2–13) of Sn‐In mass fractions give a combined measurement uncertainty of 2u that are generally 〈 3% and in agreement with literature data, where available. We present the first high precision In data for reference materials OKUM (32.1 ± 1.5 ng g−1), DTS‐2b (2.03 ± 0.25 ng g−1), MUH‐1 (6.44 ± 0.30 ng g−1), and PCC‐1 (3.55 ± 0.35 ng g−1) as well as the first Sn data for MUH‐1 (0.057 ± 0.010 μg g−1) and DTS‐2b (0.623 ± 0.018 μg g−1).

    Beschreibung: Hydrothermal vents emit sulphur and metals to the ocean1. Particular attention has been paid to hydrothermal fluxes of iron2–4, a limiting micronutrient of marine primary production5. Vent-derived ironwas previously thought to rapidly oxidize and precipitate around vents6. However, organic matter can bind to and stabilize dissolved and particulate iron in hydrothermal plumes7–9, facilitating its dispersion into the open ocean10. Here, we report measurements of the chemical speciation of sulphide and iron in high-temperature fluids emanating from vents in the East Pacific Rise and the Eastern Lau Spreading Center. We show that pyrite nanoparticles—composed of iron and sulphur—account for up to 10% of the filterable iron (less than 200nm in size) in these fluids. We suggest that these particles form before the discharge of the vent fluid. We estimate that pyrite nanoparticles sink more slowly than larger plume particles, and are more resistant to oxidation than dissolved Fe(II) and FeS.We suggest that the discharge of iron in the form of pyrite nanoparticles increases the probability that vent-derived iron will be transported over long distances in the deep ocean.

    Beschreibung: In this study, we present a single-cell genomics approach for the functional characterization of the candidate phylum Poribacteria, members of which are nearly exclusively found in marine sponges. The microbial consortia of the Mediterranean sponge Aplysina aerophoba were singularized by fluorescence-activated cell sorting, and individual microbial cells were subjected to phi29 polymerase-mediated 'whole-genome amplification'. Pyrosequencing of a single amplified genome (SAG) derived from a member of the Poribacteria resulted in nearly 1.6 Mb of genomic information distributed among 554 contigs analyzed in this study. Approximately two-third of the poribacterial genome was sequenced. Our findings shed light on the functional properties and lifestyle of a possibly ancient bacterial symbiont of marine sponges. The Poribacteria are mixotrophic bacteria with autotrophic CO(2)-fixation capacities through the Wood-Ljungdahl pathway. The cell wall is of Gram-negative origin. The Poribacteria produce at least two polyketide synthases (PKSs), one of which is the sponge-specific Sup-type PKS. Several putative symbiosis factors such as adhesins (bacterial Ig-like domains, lamininin G domain proteins), adhesin-related proteins (ankyrin, fibronectin type III) and tetratrico peptide repeat domain-encoding proteins were identified, which might be involved in mediating sponge-microbe interactions. The discovery of genes coding for 24-isopropyl steroids implies that certain fossil biomarkers used to date the origins of metazoan life on earth may possibly be of poribacterial origin. Single-cell genomic approaches, such as those shown herein, contribute to a better understanding of beneficial microbial consortia, of which most members are, because of the lack of cultivation, inaccessible by conventional techniques.

    Beschreibung: Salinity is a major factor controlling the distribution of biota in aquatic systems, and most aquatic multicellular organisms are either adapted to life in saltwater or freshwater conditions. Consequently, the saltwater–freshwater mixing zones in coastal or estuarine areas are characterized by limited faunal and floral diversity. Although changes in diversity and decline in species richness in brackish waters is well documented in aquatic ecology, it is unknown to what extent this applies to bacterial communities. Here, we report a first detailed bacterial inventory from vertical profiles of 60 sampling stations distributed along the salinity gradient of the Baltic Sea, one of world's largest brackish water environments, generated using 454 pyrosequencing of partial (400 bp) 16S rRNA genes. Within the salinity gradient, bacterial community composition altered at broad and finer-scale phylogenetic levels. Analogous to faunal communities within brackish conditions, we identified a bacterial brackish water community comprising a diverse combination of freshwater and marine groups, along with populations unique to this environment. As water residence times in the Baltic Sea exceed 3 years, the observed bacterial community cannot be the result of mixing of fresh water and saltwater, but our study represents the first detailed description of an autochthonous brackish microbiome. In contrast to the decline in the diversity of multicellular organisms, reduced bacterial diversity at brackish conditions could not be established. It is possible that the rapid adaptation rate of bacteria has enabled a variety of lineages to fill what for higher organisms remains a challenging and relatively unoccupied ecological niche.

    Beschreibung: The Denmark Strait overflow water is the largest dense water plume from the Nordic seas to feed the lower limb of the Atlantic Meridional Overturning Circulation. Its primary source is commonly thought to be the East Greenland Current. However, the recent discovery of the North Icelandic Jet—a deep-reaching current that flows along the continental slope of Iceland—has called this view into question. Here we present high-resolution measurements of hydrography and velocity north of Iceland, taken during two shipboard surveys in October 2008 and August 2009. We find that the North Icelandic Jet advects overflow water into the Denmark Strait and constitutes a pathway that is distinct from the East Greenland Current. We estimate that the jet supplies about half of the total overflow transport, and infer that it is the primary source of the densest overflow water. Simulations with an ocean general circulation model suggest that the import of warm, salty water from the North Icelandic Irminger Current and water-mass transformation in the interior Iceland Sea are critical to the formation of the jet. We surmise that the timescale for the renewal of the deepest water in the meridional overturning cell, and its sensitivity to changes in climate, could be different than presently envisaged.

    Beschreibung: Mussel aquaculture is a growing industry in Scotland, dominated by the production of Mytilus edulis. However, Mytilus trossulus, Mytilus galloprovincialis, and their hybrids with M. edulis have recently been detected in Scotland. At some farms, M. trossulus have been associated with significant losses in production because of presenting low meat yields and fragile shells. In the present study, M. edulis, M. trossulus, and their hybrids were examined from three cultivation areas in order to investigate shell and meat characteristics and advise on future management of mixed-species areas in Scotland. Results suggest that M. edulis is likely to consistently outperform M. trossulus and hybrids in mixed-species areas, but that M. trossulus meat yields from different areas can vary considerably. The ability to classify mussels into two (M. edulis and non-edulis) groups suggests that separation of M. trossulus genotypes could potentially be achieved by grading the harvested mussels. In the most affected areas, however, the fallowing of cultivation sites and importation of unispecific M. edulis spat appear as an alternative. Management of this problem in Scotland is likely to involve the implementation of area-specific measures, and the establishment of clear guidance on good practice aiming at preventing further distribution of M. trossulus.

    Beschreibung: We analyse the fluid flow regime within sediments on the Eastern levee of the modern Mississippi Canyon using 3D seismic data and downhole logging data acquired at Sites U1322 and U1324 during the 2005 Integrated Ocean Drilling Program (IODP) Expedition 308 in the Gulf of Mexico. Sulphate and methane concentrations in pore water show that sulphate–methane transition zone, at 74 and 94 m below seafloor, are amongst the deepest ever found in a sedimentary basin. This is in part due to a basinward fluid flow in a buried turbiditic channel (Blue Unit, 1000 mbsf), which separates sedimentary compartments located below and above this unit, preventing normal upward methane flux to the seafloor. Overpressure in the lower compartment leads to episodic and focused fluid migration through deep conduits that bypass the upper compartment, forming mud volcanoes at the seabed. This may also favour seawater circulation and we interpret the deep sulphate–methane transition zones as a result of high downward sulphate fluxes coming from seawater that are about 5–10 times above those measured in other basins. The results show that geochemical reactions within shallow sediments are dominated by seawater downwelling in the Mars-Ursa basin, compared to other basins in which the upward fluid flux is controlling methane-related reactions. This has implications for the occurrence of gas hydrates in the subsurface and is evidence of the active connection between buried sediments and the water column.

    Beschreibung: This paper contains the results of an extensive isotopic study of United States Geological Survey GSD-1G and MPI-DING reference glasses. Thirteen different laboratories were involved using high-precision bulk (TIMS, MC-ICP-MS) and microanalytical (LA-MC-ICP-MS, LA-ICP-MS) techniques. Detailed studies were performed to demonstrate the large-scale and small-scale homogeneity of the reference glasses. Together with previously published isotopic data from ten other laboratories, preliminary reference and information values as well as their uncertainties at the 95% confidence level were determined for H, O, Li, B, Si, Ca, Sr, Nd, Hf, Pb, Th and U isotopes using the recommendations of the International Association of Geoanalysts for certification of reference materials. Our results indicate that GSD-1G and the MPI-DING glasses are suitable reference materials for microanalytical and bulk analytical purposes.

    Beschreibung: Fluids entering the subduction zone play a key role in the subduction process. They cause changes in the dynamics and thermal structure of the subduction zone1, and trigger earthquakes when released from the subducting plate during metamorphism. Fluids are delivered to the subduction zone by the oceanic crust and also enter as the oceanic plate bends downwards at the plate boundary. However, the amount of fluids entering subduction zones is not matched by that leaving through volcanic emissions4 or transfer to the deep mantle, implying possible storage of fluids in the crust. Here we use magnetotelluric data to map the entire hydration and dehydration cycle of the Costa Rican subduction zone to 120 km depth. Along the incoming plate bend, we detect a conductivity anomaly that we interpret as sea water penetrating down extensional faults and cracks into the upper mantle. Along the subducting plate interface we document the dehydration of sediments, the crust and mantle. We identify an accumulation of fluids at ~20–30 km depth at a distance of 30 km seaward from the volcanic arc. Comparison with other subduction zones5–14 indicates that such fluid accumulation is a global phenomenon. Although we are unable to test whether these fluid reservoirs grow with time, we suggest that they can account for some of the missing outflow of fluid at subduction zones.

    Beschreibung: Lavas erupted within plate interiors above upwelling mantle plumes have chemical signatures that are distinct from midocean ridge lavas. When a plume interacts with a mid-ocean ridge, the compositions of both their lavas changes, but there is no consensus as to how this interaction occurs1–3. For the past 15 Myr, the Pacific–Antarctic mid-ocean ridge has been approaching the Foundation hotspot4 and erupted lavas have formed seamounts. Here we analyse the noble gas isotope and trace element signature of lava samples collected from the seamounts. We find that both intraplate and on-axis lavas have noble gas isotope signatures consistent with the contribution from a primitive plume source. In contrast, nearaxis lavas show no primitive noble gas isotope signatures, but are enriched in strontium and lead, indicative of subducted former oceanic lower crust melting within the plume source5–7. We propose that, in a near-ridge setting, primitive, plumesourced magmas formed deep in the plume are preferentially channelled to and erupted at the ridge-axis. The remaining residue continues to rise and melt, forming the near-axis seamounts. With the deep melts removed, the geochemical signature of subduction contained within the residue becomes apparent. Lavas with strontium and lead enrichments are found worldwide where plumes meet mid-ocean ridges6–8, suggesting that subducted lower crust is an important but previously unrecognised plume component.

    Beschreibung: Experiments have shown that ocean acidification due to rising atmospheric carbon dioxide concentrations has deleterious effects on the performance of many marine organisms(1-4). However, few empirical or modelling studies have addressed the long-term consequences of ocean acidification for marine ecosystems(5-7). Here we show that as pH declines from 8.1 to 7.8 (the change expected if atmospheric carbon dioxide concentrations increase from 390 to 750 ppm, consistent with some scenarios for the end of this century) some organisms benefit, but many more lose out. We investigated coral reefs, seagrasses and sediments that are acclimatized to low pH at three cool and shallow volcanic carbon dioxide seeps in Papua New Guinea. At reduced pH, we observed reductions in coral diversity, recruitment and abundances of structurally complex framework builders, and shifts in competitive interactions between taxa. However, coral cover remained constant between pH 8.1 and similar to 7.8, because massive Porites corals established dominance over structural corals, despite low rates of calcification. Reef development ceased below pH 7.7. Our empirical data from this unique field setting confirm model predictions that ocean acidification, together with temperature stress, will probably lead to severely reduced diversity, structural complexity and resilience Of Indo-Pacific coral reefs within this century

    Beschreibung: The Southern Ocean is thought to be one of the most energetic regions in the world’s oceans. As a result, it is a location of vigorous diapycnal mixing of heat, salt and biogeochemical properties1, 2, 3. At the same time, the Southern Ocean is poorly sampled, not least because of its harsh climate and remote location. Yet the spatial and temporal variation of diapycnal diffusivity in this region plays an important part in the large-scale ocean circulation and climate4, 5, 6. Here we use high-resolution hydrographic profiles from Argo floats in combination with the Iridium communications system to investigate diapycnal mixing in the Southern Ocean. We find that the spatial distribution of turbulent diapycnal mixing in the Southern Ocean at depths between 300 and 1,800 m is controlled by the topography, by means of its interaction with the Antarctic Circumpolar Current. The seasonal variation of this mixing can largely be attributed to the seasonal cycle of surface wind stress and is more pronounced in the upper ocean over flat topography. We suggest that additional high-resolution profiles from Argo floats will serve to advance our understanding of mixing processes in the global ocean interior.

    Beschreibung: The origin of the Christmas Island Seamount Province in the northeast Indian Ocean is enigmatic. The seamounts do not form the narrow, linear and continuous trail of volcanoes that would be expected if they had formed above a mantle plume1, 2. Volcanism above a fracture in the lithosphere3 is also unlikely, because the fractures trend orthogonally with respect to the east–west trend of the Christmas Island chain. Here we combine 40Ar/39Ar age, Sr, Nd, Hf and high-precision Pb isotope analyses of volcanic rocks from the province with plate tectonic reconstructions. We find that the seamounts are 47–136 million years old, decrease in age from east to west and are consistently 0–25 million years younger than the underlying oceanic crust, consistent with formation near a mid-ocean ridge. The seamounts also exhibit an enriched geochemical signal, indicating that recycled continental lithosphere was present in their source. Plate tectonic reconstructions show that the seamount province formed at the position where West Burma began separating from Australia and India, forming a new mid-ocean ridge. We propose that the seamounts formed through shallow recycling of delaminated continental lithosphere entrained in mantle that was passively upwelling beneath the mid-ocean ridge. We conclude that shallow recycling of continental lithosphere at mid-ocean ridges could be an important mechanism for the formation of seamount provinces in young ocean basins.

    Beschreibung: Three new 22-membered macrolactone antibiotics, atacamycins A-C, were produced by Streptomyces sp. C38, a strain isolated from a hyper-arid soil collected from the Atacama Desert in the north of Chile. The metabolites were discovered in our HPLC-diode array screening and isolated from the mycelium by extraction and chromatographic purification steps. The structures were determined by mass spectrometry and NMR experiments. Atacamycins A, B and C exhibited moderate inhibitory activities against the enzyme phosphodiesterase (PDE-4B2), whereas atacamycin A showed a moderate antiproliferative activity against adeno carcinoma and breast carcinoma cells

    Beschreibung: Ostreococcus is a marine picophytoeukaryote for which culture studies indicate there are 'high-light' and 'low-light' adapted ecotypes. Representatives of these ecotypes fall within two to three 18S ribosomal DNA (rDNA) clades for the former and one for the latter. However, clade distributions and relationships to this form of niche partitioning are unknown in nature. We developed two quantitative PCR primer-probe sets and enumerated the proposed ecotypes in the Pacific Ocean as well as the subtropical and tropical North Atlantic. Statistical differences in factors such as salinity, temperature and NO 3 indicated the ecophysiological parameters behind clade distributions are more complex than irradiance alone. Clade OII, containing the putatively low-light adapted strains, was detected at warm oligotrophic sites. In contrast, Clade OI, containing high-light adapted strains, was present in cooler mesotrophic and coastal waters. Maximal OI abundance (19 555±37 18S rDNA copies per ml) was detected in mesotrophic waters at 40 m depth, approaching the nutricline. OII was often more abundant at the deep chlorophyll maximum, when nutrient concentrations were significantly higher than at the surface (stratified euphotic zone waters). However, in mixed euphotic-zone water columns, relatively high numbers (for example, 891±107 18S rDNA copies per ml, Sargasso Sea, springtime) were detected at the surface. Both Clades OI and OII were found at multiple euphotic zone depths, but co-occurrence at the same geographical location appeared rare and was detected only in continental slope waters. In situ growth rate estimates using these primer-probes and better comprehension of physiology will enhance ecological understanding of Ostreococcus Clades OII and OI which appear to be oceanic and coastal clades, respectively. © 2011 International Society for Microbial Ecology.     Beschreibung: Sediment core PS1904 reveals continuous records of planktic and benthic foraminiferal stable isotopes (δ18O/δ13C) from the north-eastern Greenland continental margin. The data show good comparability with other records from the Nordic Seas, allowing the stratigraphic range of PS1904 to be dated to Marine Isotope Stage (MIS) 6. Focusing on MIS 5 reveals light δ18O values during MIS 5a compared to the last interglacial peak (MIS 5e) which indicates that surface and bottom water layers were strongly affected by freshwater during the former event. We present two possible scenarios explaining the origin and routing of the freshwater: (i) drainage of a Eurasian proglacial lake coupled with the collapse of the Kara Sea Ice Sheet at the MIS 5b/a boundary, and (ii) destabilization and melting of the nearby Greenland Ice Sheet. Although both scenarios could have acted simultaneously, sediment records from the Eurasian sector of the Arctic Ocean hint at the proglacial lake system in north-western Siberia as the largest freshwater source. Regardless of the actual source, the freshwater lowered the surface ocean salinity causing water column stratification and sea ice expansion. Increased sea-ice abundance led to a higher albedo and probably contributed to the cooling and global ice sheet growth that occurred subsequently during MIS 4.

    Beschreibung: The Atlantic Ocean receives warm, saline water from the Indo-Pacific Ocean through Agulhas leakage around the southern tip of Africa. Recent findings suggest that Agulhas leakage is a crucial component of the climate system and that ongoing increases in leakage under anthropogenic warming could strengthen the Atlantic overturning circulation at a time when warming and accelerated meltwater input in the North Atlantic is predicted to weaken it. Yet in comparison with processes in the North Atlantic, the overall Agulhas system is largely overlooked as a potential climate trigger or feedback mechanism. Detailed modelling experiments—backed by palaeoceanographic and sustained modern observations—are required to establish firmly the role of the Agulhas system in a warming climate.

    Beschreibung: Infections arising from multidrug-resistant pathogenic bacteria are spreading rapidly throughout the world and threaten to become untreatable. The origins of resistance are numerous and complex, but one underlying factor is the capacity of bacteria to rapidly export drugs through the intrinsic activity of efflux pumps. In this Review, we describe recent advances that have increased our understanding of the structures and molecular mechanisms of multidrug efflux pumps in bacteria. Clinical and laboratory data indicate that efflux pumps function not only in the drug extrusion process but also in virulence and the adaptive responses that contribute to antimicrobial resistance during infection. The emerging picture of the structure, function and regulation of efflux pumps suggests opportunities for countering their activities.

    Beschreibung: Climate variability in the tropical Atlantic Ocean is determined by large-scale ocean–atmosphere interactions, which particularly affect deep atmospheric convection over the ocean and surrounding continents1. Apart from influences from the Pacific El Niño/Southern Oscillation2 and the North Atlantic Oscillation3, the tropical Atlantic variability is thought to be dominated by two distinct ocean–atmosphere coupled modes of variability that are characterized by meridional4, 5 and zonal6, 7 sea-surface-temperature gradients and are mainly active on decadal and interannual timescales, respectively8, 9. Here we report evidence that the intrinsic ocean dynamics of the deep equatorial Atlantic can also affect sea surface temperature, wind and rainfall in the tropical Atlantic region and constitutes a 4.5-yr climate cycle. Specifically, vertically alternating deep zonal jets of short vertical wavelength with a period of about 4.5 yr and amplitudes of more than 10 cm s−1 are observed, in the deep Atlantic, to propagate their energy upwards, towards the surface10, 11. They are linked, at the sea surface, to equatorial zonal current anomalies and eastern Atlantic temperature anomalies that have amplitudes of about 6 cm s−1 and 0.4 °C, respectively, and are associated with distinct wind and rainfall patterns. Although deep jets are also observed in the Pacific12 and Indian13 oceans, only the Atlantic deep jets seem to oscillate on interannual timescales. Our knowledge of the persistence and regularity of these jets is limited by the availability of high-quality data. Despite this caveat, the oscillatory behaviour can still be used to improve predictions of sea surface temperature in the tropical Atlantic. Deep-jet generation and upward energy transmission through the Equatorial Undercurrent warrant further theoretical study.
