    Notizen: Abstract The effect of cereal leaf surface wax on Diuraphis noxia (Mordvilko), the Russian wheat aphid, probing behavior and nymphoposition was evaluated. Ultrastructure of leaf epicuticular wax from wheat (Triticum aestivum L.) c.v. ‘Arapahoe’ and ‘Halt’ was different from barley (Hordeum vulgare L.) c.v. ‘Morex’, and oat (Avena sativa L.) c.v. ‘Border’. Both wheat cultivars had similar rod-shaped epicuticular wax, while barley and oat plants had flakes. The chemical composition comparison of gas chromatograms also indicated that the extract of the two wheat cultivars had similar pattern of peaks, while the barley and oat leaves had similar peaks. Cereal variety significantly affected aphid probing behavior (P 〈 0.05), but wax removal using ethyl ether swab did not (P 〈 0.05). Aphids initiated significantly more probes on Border oat leaves than on Morex barley irrespective of wax removal, although total probing duration per aphid was not significantly different among the four cereals examined. Accumulative salivation duration per aphid on oat leaves with wax was significantly longer than other cereal leaves with wax, while accumulative ingestion duration per aphid on Arapahoe wheat and Morex barley was significantly longer than on oat. Nymphoposition of D. noxia on cereal leaves maintained on the benzimidazole-agar medium showed that aphids produced a greater number of nymphs on Morex barley and less on Border oat leaves, although wax removal did not affect aphid nymphoposition. Removal of leaf epicuticular waxes from the 4 cereal genotypes using ethyl ether swab indicated that the influence of wax on plant resistance to D. noxia probing and reproduction was limited. Morex barley was the most favorable, while Border oat was the least favorable cereal host of D. noxia.

    Notizen: Abstract Four DNA extraction protocols were compared for ability to produce DNA from the leaves or needles of several species: oak, elm, pine, fir, poplar and maize (fresh materials) and rhododendron (silica dried or frozen material). With the exception of maize and poplar, the species are known to be difficult for DNA extraction. Two protocols represented classical procedures for lysis and purification, and the other two were a combination of classical lysis followed by anion exchange chromatography. The DNA obtained from all procedures was quantified and tested by PCR and Southern hybridisation.Test results indicated superiority of one of the four protocols; a combination of CTAB lysis followed by anion exchange chromatography which enabled DNA extraction from all seven species. A second protocol also produced DNA from leaves or needles of all species investigated and was well suited for PCR applications but not Southern hybridisations.     Notizen: Abstract Samples of wheat (n = 25) and maize (n = 30) for animal consumption, collected in 1997 after harvest from western Romania, were analyzed by enzyme immunoassays for mycotoxin contamination. Toxins analyses included deoxynivalenol (DON), 3-acetylDON, 15- acetylDON, fusarenone X (FX), T-2 Toxin (T-2), diacetoxyscirpenol (DAS), zearalenone (ZEA), fumonisin B1 (FB1), aflatoxin B1 (AFB1), ochratoxin A (OA), and citrinin (CT). DON and acetylDONs were the major contaminants in wheat (100%) and maize (46%). Median values for DON, 3-acetylDON, and 15-acetylDON were 880 μg kg-1, 66 μg kg- 1, and 150 μg kg-1 in wheat, and 890 μg kg-1, 180 μg kg-1, and 620 μg kg- 1 in maize, respectively. Additionally, 3,15-diacetylDON was detected in some samples by HPLC-EIA analysis. All samples were negative for FX (〈150 μg kg-1). T-2 was found in wheat (n = 6) and maize (n = 1) at levels between 13 and 63 μg kg- 1. DAS (2.6 μg kg-1) was found in one maize sample. ZEA occurred in all wheat and in four maize samples, median values were 10 μg kg-1 and 250 μg kg-1, respectively. One maize sample contained FB1 (140 μg kg-1). All samples were AFB1-negative (〈4 μg kg-1). OA was found in one wheat sample (37 μg kg- 1), CT was found in one maize sample (580 μg kg- 1). This first reported natural occurrence of a range of mycotoxins in Romanian feeding stuff shows that DON and acetyl DONs may be present at levels which may affect animal production.

    Notizen: Abstract A single-seed DNA extraction method was developed for rapid identification of plant genotype. The method was applied to 12 plant species, including the oil seeds sesame and soybean. The results were comparable to those obtained for oil-less seeds such as rice. This method will be useful for genotypic selection which requires rapid screening of large populations. It can also be used to identify varietal purity of seed stocks by PCR and RFLP analysis. The method includes two major steps, (i) treatment by proteinase K in an SDS extraction buffer, and (ii) grinding of a single half seed in the buffer after incubation. About 1.5–2 µg of DNA per half seed (the endosperm part) of rice was obtained and more than 200 half seed samples could be handled by one person in a day. The DNA could be used for fingerprinting and detection of target genes in a transgenic plant by PCR. The amplified PCR products from the half seed DNA exhibited the same banding patterns as those from leaf DNA. Yield and quality of DNA extracted from half seeds of rice was also sufficient for RFLP analysis. The remnant half seeds containing the embryo can be maintained for later germination of selected genotypes.

    Notizen: Abstract The recent growth of conservation biology has demanded that faster and more effective measures of biodiversity be utilized. Arthropods, due to high levels of diversity and their relative ease of capture, are often the subject of such surveys. The vacuum sampler, used quite often in the context of agricultural arthropod surveys, has never been adequately evaluated or compared to more traditional collection techniques in relatively complex ecosystems. The purpose of this study was to determine whether the vacuum sampler was more or less effective than a sweep-net in measuring arthropod biodiversity in California coastal sage scrub. The results show that significantly more individuals were collected by the vacuum sampler per unit effort for three out of six orders of arthropods examined. In addition, the vacuum sampler collected a significantly greater number of arthropod species than the sweep-net technique for two out of the six orders sampled. There were no significant differences in the number of species collected for the remaining four orders. We feel these findings are important for arthropod biodiversity studies utilized for conservation efforts as the vacuum sampler can attain a level of efficiency and sensitivity (with regard to species detection) that sweep-net techniques cannot. © Rapid Science Ltd. 1998

    Notizen: Abstract Songs of Orthoptera can be used for inventorying and monitoring of individual species and communities. Acoustic parameters such as carrier frequency and pulse rates allow the definition of recognizable taxonomic units (RTUs) which help to overcome the taxonomic impediment due to our scanty knowledge, particularly of tropical faunas. Bioacoustic diversity is a first estimate for species richness and provides baseline data which can be a prerequisite for conservation. Additional ecological and behavioural information such as habitat preference and singing schedules can be inferred. Many Orthoptera are sensitive indicator species for habitat quality in temperate and tropical ecosystems. Examples are given for evaluation of habitat quality and deterioration by acoustic detection of Orthoptera.

    Notizen: Abstract It is estimated that only 10–15% of the world's insect fauna has been described and named. Efforts to inventory insect biodiversity are hampered by this taxonomic impediment, which is compounded by the logistical problems of an insufficient taxonomic workforce and their remote location in museums thousands of miles from the areas of highest biodiversity. Compared to most other invertebrate groups however, the taxonomic impediment is relatively benign in the order Orthoptera. This is a small to medium-sized order (approximately 20 000 described species) which is well known taxonomically, owing to the group's agricultural importance worldwide. Furthermore, orthopteran taxonomists are now fortunate to have a published up-to-date catalogue of all known species, which has just become accessible as a regularly updated database on the World Wide Web. Whilst new information technology, in the form of e-mail networks, World Wide Web sites and CD-ROM information archives, is already enhancing communication between specialists and helping to reduce the logistical problems of documenting orthopteran biodiversity, a major reinvestment in basic taxonomic research is needed if we are to reduce the existing taxonomic impediment significantly. There is general agreement that an internationally coordinated approach will be necessary and priorities must be set to tackle the biodiversity/systematics crisis. In the future, the Orthoptera can make an important contribution to invertebrate faunal surveys and have potential as an indicator taxon. Furthermore, the Orthoptera Species File establishes a taxonomic framework which could be readily enlarged to include geographic data and phenology of species from existing museum specimens.

    Notizen: Abstract This essay deals with the allocation of genetic resources with regard to property-rights theory. In addition to this, different arrangements of property rights will be examined as to their allocation and distribution effects. The result will show that property rights on biological diversity can only cover patrimonial rights which contain the right to use the resource but not to destroy it. Bioprospecting conventions, such as the well known Merck-INBio agreement, will be examined as mechanisms to establish property rights in terms of biological diversity. Moreover, the identification of these agreements will be critically questioned for their applicability as a form of the Coase-theorem.

    Notizen: Abstract The development of a specific, reliable and noninvasive method for measuring oxidative stress in humans is essential for establishing the role of free radicals in human diseases. Currently, accurate techniques to assess oxidant injury in vivo are extremely limited although a number of approaches are being investigated. Of these, the measurement of specific products of nonenzymatic lipid peroxidation, the F2-isoprostanes (F2-IsoPs), appears to be a more accurate marker of oxidative stress in vivo in humans than other available methods. The purpose of this brief review is to acquaint the reader with the IsoPs from a biochemical perspective and to provide information regarding the utility of quantifying these compounds as indicators of oxidant stress.

    Notizen: Abstract  The ultrastructure of egg cells in Abies alba was examined to elucidate the lack of maternal inheritance of plastids. Before fertilization, maternal plastids are absent in the perinuclar zone containing mainly mitochondria and smooth endoplasmic reticulum. During egg cell development the maternal plastids are transformed into large inclusions which are situated mostly towards the periphery of the egg cell, and finally disintegrate. As a consequence, they do not participate in zygote formation. RFLP analysis of cpDNA of parental trees and their F1 interspecific hybrids (A. alba×A. numidica, A. alba×A. nordmanniana, A. nordmanniana×A. Alba) using HindIII and BamHI showed a paternal mode of cpDNA inheritance. Paternal inheritance has also been found with PCR/RFLP analysis of cpDNA from parental trees and their hybrids (A. alba×A. pinsapo, A. pinsapo×A. alba, A. pinsapo×A. numidica) using ApaI and HaeIII digests, as well as in the crosses of A. cephalonica×A. nordmanniana, A. nordmanniana×A. cephalonica, A. cephalonica×A. numidica using TagI digests.

    Notizen: Abstract The ecological concept of resilience has begun to inform analysis of change in economy-environment systems. The linkages between resilience and the stability of dynamical systems are discussed, along with its role in understanding of the evolution of such systems. Particular linkages discussed include those between resilience, biodiversity and the sustainability of alternative states. Recent developments in modelling the resilience of joint economy-environment systems suggest the advantages of analysing change in the system as a Markov process, the transition probabilities between states offering a natural measure of the resilience of the system in such states. It is argued that this ‘emergent property’ of the collaboration between ecology and economics has far-reaching implications for the way we think about, model and manage the environmental sustainability of economic development.

    Notizen: Abstract The social and economic implications of atmospheric change on biodiversity need to be seen in a global context of major shifts in the conceptualization and management of our relationship with nature. Traditionally, we have conceptualized the atmosphere and the other creatures of the biosphere as separate from the human, but their quasi-autonomy is now becoming subject to more and more human management. This raises not only economic issues, but social, political, and ethical concerns that will have substantial influence on public policy. Among these are the commodification of genetic material; the privatization of traditional knowledge; and the management of information. In this broader context, the paper examines an array of current and proposed strategies of response to changes in biodiversity as a result of climatic and other stresses.

    Notizen: Abstract With a recent media-fueled transition from a scientific to a political perspective, biodiversity has become an issue of ethics and ensuing values, beyond its traditional ecological roots. More fundamentally, the traditional perspective of biodiversity is being challenged by the emergence of a post-normal or systems-based approach to science. A systems-based perspective of living systems rests on the central tenets of complexity and uncertainty, and necessitates flexibility, anticipation and adaptation rather than prediction and control in conservation planning and management. What are the implications of this new perspective? This paper examines these challenges in the context of biodiversity conservation planning. The new perspectives of biodiversity are identified and explored, and the emergence of a new ecological context for biodiversity conservation is discussed. From the analysis, the challenges and implications for conservation planning are considered, and a systems-based or post-normal approach to conservation planning and management is proposed. In light of the new perspectives for biodiversity, conservation planning and management approaches should ultimately reflect the essence of living systems: they should be diverse, adaptive, and self-organizing, accepting the ecological realities of change.

    Notizen: Abstract Overall, the greatest threats to Canadian and global biodiversity are associated with conversions of natural ecosystems to anthropogenic ones, and over-exploitation of biological resources. This circumstance does not, however, trivialize the importance of atmospheric influences. Although scientific understanding of the risks is incomplete, it is nevertheless clear that anthropogenic changes in atmospheric stressors are potentially damaging to biodiversity and other ecological values over medium- and longer-term scales. It is important that greater investments be made in support of longer-term monitoring and research designed to understand the effects of atmospheric and other environmental stressors on the biodiversity and structure and function of Canadian ecosystems.

    Notizen: Abstract  We mapped and characterized quantitative trait loci (QTLs) for resistance to Ustilago maydis and investigated their consistency across different flint-maize populations. Four independent populations, comprising 280 F3 lines (A×BI), 120 F5 lines (A×BII), 131 F4 lines (A×C) and 133 F4 lines (C×D), were produced from four European elite flint inbreds (A, B, C, D) and genotyped at 89, 151, 104, and 122 RFLP marker loci, respectively. All Fn lines were evaluated in field trials with two replications in five German environments. Genotypic variances were highly significant for the percentage of U. maydis infected plants (UST) in all populations, and heritabilities exceeded 0.69. Between five and ten QTLs were detected in individual populations by composite interval mapping, explaining between 39% and 58% of the phenotypic variance. These 19 different QTLs were distributed over all ten chromosomes without any clustering on certain chromosomes. In most cases, gene action was dominant or overdominant. Fourteen pairs of the detected QTLs for UST displayed significant digenic epistatic interactions, but only two of them did so after arcsin √UST/100 transformation. Significant QTL× environment interactions occurred frequently. Between two to four QTLs were common between pairs of populations. Population C×D was also grown in Chartres, a location with a high U. maydis incidence. Two out of six QTLs identified for Chartres were in common with QTLs detected across five German environments for C×D. Consequently, marker-assisted or phenotypic selection based on results from natural infection seem to be suitable breeding strategies for improving the resistance of maize to U. maydis.

    Notizen: Abstract  An RFLP-based map consisting of 160 loci was constructed in an intervarietal cross of foxtail millet [Setaria italica (L.) P. Beauv.], Longgu 25×Pagoda Flower Green. The map comprises nine linkage groups, which were aligned with the nine foxtail millet chromosomes using trisomic lines, and spans 964 cM. The intraspecific map was compared to an interspecific map, constructed in a S. italica×S. viridis cross. Both the order of the markers and the genetic distances between the loci were highly conserved. Deviations from the expected 1 : 2 : 1 Mendelian segregation ratios were observed in both the intra- and inter-specific populations. The segregation data indicate that chromosome VIII in the Longgu 25×Pagoda Flower Green cross carries a gene that strongly affects gamete fertility.

    Notizen: Abstract  Genetic and physical maps, consisting of a large number of DNA markers for Arabidopsis thaliana chromosomes, represent excellent tools to determine the organization of related genomes such as those of Brassica. In this paper we report the chromosomal localization and physical analysis by pulsed-field gel electrophoresis (PFGE) of a well-defined gene complex of A. thaliana in the Brassica nigra genome (B genome n=8). This complex is approximately 30 kb in length in A. thaliana and contains a cluster of six genes including ABI1 (ABA-responsive), RPS2 (resistance against Pseudomonas syringae, a bacterial disease), CK1 (casein kinase I), NAP (nucleosome-assembly protein), X9 and X14 (both of unknown function). The Arabidopsis chromosomal complex was found to be duplicated and conserved in gene number at different levels in the Brassica genome. Linkage group B1 had the most-conserved arrangement carrying all six genes tightly linked. Group B4 had an almost complete complex except for the absence of RPS2. Other partial complexes of fewer members were found on three other chromosomes. Our studies demonstrate that by this approach it is possible to identify ancestrally related chromosome segments in a complex and duplicated genome, such as the genome of B. nigra, permitting one to draw conclusions as to its origin and evolution.

    Notizen: Abstract  DNA from pooled leaf samples of 11 true major mangrove, three true minor mangrove, two mangrove associate, two mangrove parasite, three terrestrial and one cultivated species were isolated for the present study. In total, 198 random amplified polymorphic DNAs (RAPDs) and 180 restriction fragment length polymorphism (RFLP) loci were scored by using ten primers and 14 enzyme-probe combinations respectively. The polymorphism observed for these markers revealed a high degree of genetic diversity in mangroves at both inter-specific or inter-generic levels. A dendrogram, constructed after pooling both RAPD and RFLP data, using a similarity index was analysed for genome relationships among these species. The dendrogram showed clustering of all the major mangroves, except for Nypa fruticans (Arecaceae), into one group. All species under the tribe Rhizophorae formed a sub-cluster, to which Xylocarpus granatum was found to be the most closesly related species. The clustering pattern implied that Excoecaria agallocha and Acanthus ilicifolius should be considered as true minor mangroves. The present study also provided molecular data favouring the separation of Avicennia spp. from the Verbenaceae to create a monotypic family the Avicenniaceae. The separation of Viscum orientale into the Viscaceae was also favoured.

    Notizen: Abstract  A genetic linkage map of tetraploid wheat [Triticum turgidum (L.) Thell.] was constructed using segregation data from a population of 65 recombinant inbred lines (RILs) derived from a cross between the durum wheat cultivar Messapia and accession MG4343 of T. turgidum (L.) Thell. ssp dicoccoides (Korn.) Thell. A total of 259 loci were analysed, including 244 restriction fragment length polymorphisms (RFLPs), one PCR (polymerase chain reaction) marker (a sequence coding for a LMW (low-molecular-weight) glutenin subunit gene located at the Glu-B3 locus), seven biochemical (six seed-storage protein loci and one isozyme locus) and seven morphological markers. A total of 213 loci were mapped at a LOD≥3 on all 14 chromosomes of the A and B genomes. The total length of the map is 1352 cM and the average distance between adjacent markers is 6.3 cM. Forty six loci could not be mapped at a LOD≥3. A fraction (18.6%) of the markers deviated significantly from the expected Mendelian ratios; clusters of loci showing distorted segregation were found on chromosomes 1B, 3AL, 4AL, 6AL and 7AL. The durum wheat map was compared with the published maps of bread wheat using several common RFLP markers and general features are discussed. The markers detected the known structural rearrangements involving chromosomes 4A, 5A and 7B as well as the translocation between 2B-6B, but not the deletion on 2BS. This map provides a useful tool for analysing and breeding economically important quantitative traits and for marker-assisted selection, as well as for studies of genome organisation in small grain cereal species.

    Notizen: Abstract  A genetic map of the long arm of chromosome 6R of rye was constructed using eight homoeologous group-6 RFLP clones and five PCR markers derived from the rye-specific dispersed repetitive DNA family, R173. The map was developed using a novel test-cross F1 (TC-F1) population segregating for resistance to the cereal cyst nematode. Comparisons were made between the map generated with other rye and wheat group-6 chromosome maps by the inclusion of RFLP clones previously mapped in those species. Co-linearity was observed for common loci. This comparison confirmed a dramatic reduction in recombination for chromosome 6R in the TC-F1 population. The CreR locus was included in the linkage map via progeny testing of informative TC-F1 individuals. CreR mapped 3.7 cM distal from the RFLP locus, XksuF37. Comparative mapping should allow the identification of additional RFLP markers more closely linked to the CreR locus.

    Notizen: Abstract  One hundred and thirty nine restriction fragment length polymorphisms (RFLPs) were used to construct a soybean (Glycine max L. Merr.) genetic linkage map and to identify quantitative trait loci (QTLs) associated with resistance to corn earworm (Helicoverpa zea Boddie) in a population of 103 F2-derived lines from a cross of ‘Cobb’ (susceptible) and PI229358 (resistant). The genetic linkage map consisted of 128 markers which converged onto 30 linkage groups covering approximately 1325 cM. There were 11 unlinked markers. The F2-derived lines and the two parents were grown in the field under a plastic mesh cage near Athens, Ga., in 1995. The plants were artificially infested with corn earworm and evaluated for the amount of defoliation. Using interval-mapping analysis for linked markers and single-factor analysis of variance (ANOVA), markers were tested for an association with resistance. One major and two minor QTLs for resistance were identified in this population. The PI229358 allele contributed insect resistance at all three QTLs. The major QTL is linked to the RFLP marker A584 on linkage group (LG) ‘M’ of the USDA/Iowa State University public soybean genetic map. It accounts for 37% of the total variation for resistance in this cross. The minor QTLs are linked to the RFLP markers R249 (LG ‘H’) and Bng047 (LG ‘D1’). These markers explain 16% and 10% of variation, respectively. The heritability (h2) for resistance was estimated as 64% in this population.

    Notizen: Abstract  RFLP (restriction fragment length polymorphism) and GISH (genomic in situ hybridization) analyses were employed to identify the chloroplast and nuclear genomes of the somatic hybrids and progeny between tomato ‘Ohgata zuiko’ and Solanum lycopersicoides (‘LA 2386’). A random distribution of the chloroplast genotype was determined using a cloned 19.6-kb BamHI fragment (Ba1) of tobacco chloroplast DNA. Eight selected hybrids were analyzed for their chromosomal compositions; 4 were tetraploids (2n=48) with an equal number of chromosomes derived from each parent as accurately determined by GISH, and the other 4 were hexaploids, containing an average of two sets of tomato chromosomes and one set from the wild parent. RFLP analysis with six tomato nuclear probes of known chromosomal locations revealed no major variation among the 44 hybrid plants surveyed. However, it also showed the presence of both parent-specific alleles and the loss of some and the presence of a few non-parental alleles, indicating rearrangement and/or recombination of the nuclear DNA. The relevance of the molecular and cytological methods and the potential use of somatic hybrids for plant breeding are demonstrated.

    Notizen: Abstract  The gene Ry adg that confers resistance to potato Y potyvirus (PVY) in the cultivated potato [Solanum tuberosum subsp. andigena, line 2x(v-2)7] is located on chromosome XI in a segment that contains three other known resistance genes in other syntenic solanaceous species. One of them is the gene N that controls resistance to tobacco mosaic tobamovirus in tobacco and has previously been isolated and sequenced. Three sequence-related, resistance gene-like (RGL) DNA fragments (354–369 bp) highly homologous to the gene N were PCR-amplified from the potato line 2x(v-2)7. Two RGL fragments (79 and 81% homologous to the N gene) co-segregated with Ry adg among the 77 F1 progeny tested. These RGLs may originate from a resistance gene family on chromosome XI. The potato line 2x(v-2)7 also expressed resistance to potato A potyvirus (PVA), which was controlled by another locus on chromosome XI mapped ca. 6.8 cM distal to Ry adg .

    Notizen: Abstract  Bulked segregant analysis and comparative mapping were applied to identify molecular markers linked to the Rfo restorer gene used for the Ogu-INRA cytoplasmic male-sterility system in rapeseed. These markers were then used to localise the radish introgression on the B. napus genetic map constructed from the cross ‘Darmor.bzh’ x ’Yudal’. The introgression mapped on the DY15 linkage group. From the comparison of this latter group to the linkage group constructed on a F2 progeny segregating for the radish introgression, it was concluded that the introgression had occurred through homoeologous recombination, that it was not distal and that it had replaced a B. napus region of around 50 cM. A QTL involved in aliphatic seed glucosinolate content was located on the DY15 linkage group at a position corresponding to one end of the introgression. The DNA markers identified in this study are being used in map-based cloning of the Rfo gene and in marker-assisted selection.

    Notizen: Abstract  Three mutations determining self-fertility at the S, Z and S5 self-incompatibility loci on chromosomes 1R, 2R and 5R of rye, respectively, were mapped using three different F2 populations. There was a close linkage of one isozyme and four RFLP markers, and no recombinant plants were detected. These markers are Prx7, Xiag249 and Xpsr634 for the S locus (1R), Xbcd266 for the Z locus (2R) and Xpsr100 for the S5 locus (5R). Linkage data for markers associated to the self-fertility mutations at the S, Z and S5 loci were calculated and compared with genetic maps computed by MAPMAKER multipoint analysis.

    Notizen: Abstract  An RFLP map constructed using 150 single seed descent (SSD) lines from a cross between two hexaploid wheat varieties (‘Schomburgk’בYarralinka’) was used to identify loci controlling flour colour. Flour colour data were obtained from field trials conducted over two seasons at different sites. The estimated heritability of this trait was calculated as 0.67. Two regions identified in the preliminary analysis on chromosomes 3A and 7A, accounted for 13% and 60% of the genetic variation respectively. A detailed analysis of the major locus on 7A was conducted through fine mapping of AFLP markers identified using bulked segregant analysis (BSA). Seven additional markers were identified by the BSA and mapped to the region of the 7A locus. The applicability of these markers to identify wheat lines with enhanced flour colour is discussed.

The gene was linked to the two flanking markers Xmwg2264 and Xmwg916 by 6.7 and 13.0 cM, respectively. Compared to Ppd-H1 (chromosome 2H) and Ppd-H2 (chromosome 1H), ea 7 determines the strongest effect on flowering time with 55 and 18 days difference compared to photoperiod sensitive genotypes grown under short and long photoperiods, respectively. Allelic and homoeologous relationships to major genes and quantitative trait loci controlling flowering time in barley and wheat are discussed.

    Notizen: Abstract  We report the molecular mapping of the py-1 gene for resistance to corky root rot [Pyrenochaeta lycopersici (Schneider and Gerlach)] in tomato using RAPD and RFLP marker analysis. DNA from near-isogenic lines (NILs) of tomato differing in corky root rot resistance was screened with 575 random oligonucleotide primers to detect polymorphic DNAs linked to py-1. Three primers (OPW-04, OPC-02, OPG-19) revealed polymorphisms between the NILs. Twelve resistant and eight susceptible DNA pools derived from segregating F3 families were used to confirm that the RAPD markers were linked to the py-1 gene. Two of the linked amplified fragments, corresponding to OPW-04 and OPC-02, were subsequently cloned and mapped on the tomato molecular linkage map as RFLPs. These clones were located between TG40 and CT31 on the short arm of chromosome 3. Further analysis with selected RFLP markers showed that 7% (8.8 cM) of chromosome 3 of the resistant line ‘Moboglan’ was introgressed from the L. peruvianum donor parent. Three RFLP markers (TG40, TG324, and TG479) from the introgressed part of chromosome 3 were converted to cleaved amplified polymorphism (CAP) markers for use in a polymerase chain reaction (PCR) assay. These PCR markers will allow rapid large-scale screening of tomato populations for corky root rot resistance.

    Notizen: Abstract  There are both economic and environmental reasons for reducing the use of herbicides for weed control in soybean [Glycine max (L.) Merr.] fields. Optimizing crop competitiveness can reduce reliance on chemical weed control. Fast and vigorous early growth and rapid canopy development can be effective in suppressing weed infestation of crop plants. The purposes of this study were to identify and molecularly map the quantitative trait loci (QTLs) conditioning soybean plant height and canopy width during the early vegetative stages of soybean growth. A restriction fragment length polymorphism (RFLP) linkage map was created using 142 markers and 116 F2-derived lines from a cross of ‘S100’בTokyo’. The parents and the 116 F2-derived lines were evaluated in the greenhouse and in the field at Athens, Ga., in 1996 and 1997. Combined over environments, Tokyo averaged 41 and 17% taller plants than S100 at the V7 and V10 stages of development. Transgressive segregation was observed among the progeny at both stages. Based on single-factor analysis of variance (ANOVA), three and four independent RFLP loci were associated with plant height at the V7 and V10 stages, respectively. All three loci detected [on linkage groups (LGs) C2 and F, and unlinked] at the V7 stage were also detected at the V10 stage along with one additional independent locus on LG E. The Tokyo allele contributed to increased plant height at all loci except at the unlinked locus. Three QTLs (on LGs C2, E, and F) were consistent across environments, three (on LGs C2 and F, and unlinked) were consistent across stages of plant development, and two (on LGs C2 and F) were consistent both across environments and stages of plant development. Within each stage of development, there was no interaction among the independent loci, and the respective loci together explained most of the variation in the traits. Three independent RFLP loci were associated with canopy width at the V10 stage, of which one was unique to the trait, while the remaining loci (on LGs C2 and F) were in common with the independent loci for plant height. Canopy width had a strong correlation (r=0.87) with plant height at the V10 stage. However, mature plant height, lodging, or seed weight had no phenotypic or QTL association with early plant height or canopy width.

    Notizen: Abstract  Consensus linkage maps were constructed for all seven rye chromosomes using 12 basic RFLP maps. The maps presented contain a total of 413 markers. The number of markers per chromosome varies from 41 (chromosome 3R) to 83 (chromosome 1R). In addition to 374 RFLP and 24 isozyme markers 15 gene loci were incorporated, determining the traits reduced plant height, self fertility, male sterility restoration, vernalization response, resistance against powdery mildew, chlorophyll deficiency, hairy leaf sheath, hairy peduncle, waxy endosperm, waxless plant and absence of ligules. The maps presented allow the selection of markers for the fine mapping of certain regions of the rye genome. In terms of the known chromosomal rearrangements within the Triticeae its utilization can also be extended for mapping in wheat and barley.

Batch] was constructed in order to identify molecular markers linked to economically important agronomic traits that would be particularly useful for long-lived perennial species. An intraspecific F2 population was generated from self-pollinating a single F1 plant from a cross between a flat non-acid peach, ‘Ferjalou Jalousia®’ and an acid round nectarine ‘Fantasia’. Mendelian segregations were observed for 270 markers including four agronomic characters (peach/nectarine, flat/round fruit, acid/non-acid fruit, and pollen sterility) and 1 isoenzyme, 50 RFLP, 92 RAPD, 8 inter-microsatellite amplification (IMA), and 115 amplified fragment length polymorphism (AFLP) markers. Two hundred and forty-nine markers were mapped to 11 linkage groups covering 712 centiMorgans (cM). The average density between pairs of markers is 4.5 cM. For the four agronomic characters studied, molecular markers were identified. This map will be used for the detection of QTL controlling fruit quality in peach and, particularly, the acid and sugar content.

    Notizen: Abstract  The adaptability of Triticum aestivum to a large range of environments is partially due to genetic differences in sensitivity to vernalization. The most potent gene reducing the vernalization requirement in hexaploid wheat is Vrn-A1. An orthologous vernalization gene, designated Vrn-A m 1, was mapped in the diploid wheat Triticum monococcum between RFLP markers Xwg908 and Xabg702 on the long arm of chromosome 5AmL. The orthology of VrnA m 1 with Vrn-A1 (5A wheat, originally Vrn1), Vrn-D1 (5D wheat, originally Vrn3), Vrn-R1 (5R rye, originally Sp1) and Vrn-H1 (5H barley, originally Sh2) was shown by mapping RFLP markers linked to these vernalization genes on the T. monococcum linkage map. A second vernalization gene, designated Vrn-A m 2, was found in the distal region of chromosome 5AmL within a segment translocated from homoeologous group 4. This gene is completely linked to RFLP marker Xbcd402 and located between the same RFLP markers (Xβ-Amy-1 and Xmwg616) as the Vrn-H2 (originally Sh) locus in Hordeum vulgare.

    Notizen: Abstract  Chinese accessions of Triticum tauschii and T. aestivum L. from the Sichuan white (SW), Yunnan hulled (YH), Tibetan weedrace (TW), and Xinjiang rice (XR) wheat groups were subjected to RFLP analysis. T. tauschii and landraces of T. aestivum from countries in Southwest Asia were also evaluated. For T. tauschii, a west to east gradient was apparent where the Chinese accessions exhibited less diversity than those from Southwest Asia. Compared to the Southwest Asian gene pool, the Chinese T. tauschii was highly homogeneous giving a low frequency of polymorphic bands (16%) and banding patterns (1.33 per probe) with 75 RFLP probe-HindIII combinations. Accessions of T. tauschii from Afghanistan and Pakistan were genetically more similar to the Chinese T. tauschii than those from Iran. Of 368 bands found for 39 Chinese hexaploid wheat accessions with 63 RFLP probe-HindIII combinations, 28.3% were polymorphic with an average of 2.6 banding patterns per probe and 5.0 bands per genotype. The individual Chinese landrace wheat groups revealed less variation than those from Afghanistan, Iran, and Turkey. When classified into country based groups, however, the diversity level over all Chinese landraces was greater than that of some Southwest Asian landraces, especially those from Afghanistan and Iran . The XR wheat group was genetically distinct from the other three Chinese landrace groups and was more related to the Southwest Asian landraces. The TW group was genetically similar to, but more diverse than, the SW and YH groups. The Chinese landraces had a higher degree of genetic relatedness to the Southwest Asian T. tauschii, particularly to accessions from Iran, rather than to the Chinese T. tauschii. ‘Chinese Spring’ was most related to ‘Chengdu-guang-tou’, a cultivar from the SW wheat group.

    Notizen: Abstract  The genetic constitutions of chromosome 2M of Aegilops comosa and the derived wheat-Ae. comosa translocations were analyzed by molecular cytogenetic techniques. Hybridization of 15 RFLP markers covering the entire length of the group-2 chromosomes revealed that chromosome 2M was structurally rearranged compared to the homoeologous chromosomes of wheat by either a pericentric inversion or a terminal intrachromosomal translocation. The breakpoint of the rearrangement was located in a region between the loci Xpsr131 and Xcdo405, resulting in the translocation of 47% of 2MS to 2ML. This aberrant structure of 2M allowed homoeologous recombination between 2M and its wheat counterpart only in the translocated segment on 2ML. C-banding and genomic in situ hybridization analyses confirmed that all translocation chromosomes consisted of the complete 2MS arm, a large part of 2ML, and very small distal segments derived from 2AS or 2DS, as expected from the aberrant structure of chromosome 2M. Thus, the translocation in the line 2A-2M?4/2 can be described as T2AS-2M?1L ⋅ 2M?1S and the translocations in the lines Compair and 2D-2M?3/8 as T2DS-2M?1L ⋅ 2M?1S. RFLP analysis determined the breakpoints in these translocation chromosomes to be within the telomeric 16% of the wheat chromosome arms. The breakpoint of the 2A/2M translocation was between Xbcd348 and Xcdo783, and that of the 2D/2M translocation was between Xcdo783 and Xpsr666. Because the translocation chromosomes retain the structural aberration found in chromosome 2M, further exploitation of the wheat-Ae. comosa translocations for cultivar improvement is questionable.

    Notizen: Abstract  Relationships among 88 accessions representing 45 Citrus species, three man-made hybrids, and six related genera were examined for restriction fragment length polymorphisms (RFLP). Thirty-two Citrus and three Microcitrus accessions were also examined by random amplified polymorphic DNA (RAPD) analysis. A measure of relative heterozygosity was estimated based on the mean of the number of fragments per individual per probe-enzyme combination (PEC) divided by total number of fragments per PEC for all non-hybrid Citrus individuals. The presence in a Citrus species of a rare band found also in a related genus was taken as an indication of possible introgression, while the presence of several fragments unique to 1 species was used to indicate non-involvement of that species in hybridization events. Most species that have been described in the literature as hybrids had high heterozygosity indices and no unique fragments. Distance matrices and dendrograms were generated using simple matching coefficient and neighbor-joining cluster analysis. RFLP and RAPD data gave approximately the same results. These data showed C. maxima was affiliated with the papedas C. hongheensis and C. latipes. C. medica clustered with C. indica when only non-hybrid taxa were examined, or among limes, lemons, and relatives when all species were considered. Mandarins did not show strongly supported groupings among themselves, nor with other species. These data showed that several accessions were probably assigned to the wrong species.

    Notizen: Abstract  The line IvP35 of the diploid (2n=2x=24) cultivated potato species Solanum phureja (family Solanaceae) expresses hypersensitive resistance (H) to potato X potexvirus (PVX). In this study, a diploid potato population was produced using IvP35 as the male parent and a diploid line of S. tuberosum (87HW13.7) as the female parent and tested for resistance to PVX. Data indicated that H to PVX in IvP35 is a dominant, monogenically inherited trait controlled by a single gene, named Nx phu , that is in a simplex condition (Nxnx). RFLP analysis carried out on the progeny lines revealed 4 markers (CT220, TG328, CT112 and TG424) from the long arm of chromosome IX that were linked to the hypersensitive phenotype; the closest linkage was observed with the marker TG424. Previous authors have shown that the same region of chromosome IX contains the gene Sw-5 for resistance to tomato spotted wilt tospovirus in Lycopersicon peruvianum (Solanaceae).

    Notizen: Abstract  Sugarcane mosaic virus (SCMV) causes considerable damage to maize (Zea mays L.) in Europe. The objective of the present study was to determine the genetic basis of resistance to SCMV in European maize germplasm and to compare it with that of U.S. inbred Pa405. Three resistant European inbreds D21, D32, and FAP1360A were crossed with four susceptible inbreds F7, KW1292, D408, and D145 to produce four F2 populations and three backcrosses to the susceptible parent. Screening for SCMV resistance in parental inbreds and segregating generations was done in two field trials as well as under greenhouse conditions. RFLP markers umc85, bnl6.29, umc10, umc44, and SSR marker phi075 were used in F2 populations or F3 lines to locate the resistance gene(s) in the maize genome. Segregation in the F2 and backcross generations fitted to different gene models depending on the environmental conditions and the genotype of the susceptible parent. In the field tests, resistance in the three resistant European inbreds seems to be controlled by two to three genes. Under greenhouse conditions, susceptibility to SCMV in D32 appears to be governed by one dominant and one recessive gene. Allelism tests indicated the presence of a common dominant gene (denoted as Scm1) in all three resistant European inbreds and Pa405. Marker analyses mapped two dominant genes: Scm1 on chromosome 6S and Scm2 on chromosome 3.

    Notizen: Abstract  Wild soybean (Glycine soja Sieb. et Zucc.), regarded as the progenitor of cultivated soybean [G. max (L.) Merr.], is widely distributed in East Asia. We have collected 1097 G. soja plants from all over Japan and analyzed restriction fragment length polymorphisms (RFLPs) of mitochondrial DNA (mtDNA) in them. Based on the RFLPs detected by gel-blot analysis, using coxII and atp6 as probes, the collected plants were divided into 18 groups. Five mtDNA types accounted for 94% of the plants examined. The geographic distribution of mtDNA types revealed that, in many regions, wild soybeans grown in Japan consisted of a mixture of plants with different types of mtDNA, occasionally even within sites. Some of the mtDNA types showed marked geographic clines among the regions. Additionally, some wild soybeans possessed mtDNA types that were identical to those widely detected in cultivated soybeans. Our results suggest that the analysis of mtDNA could resolve the maternal lineage among plants of the genus Glycine subgenus Soja.

    Notizen: Abstract  A genetic linkage map of rye composed of 91 loci (88 RFLP, two morphological and one isozyme markers) has been developed using two reciprocal crosses. The RFLP loci covering all seven chromosomes were detected by a selection of rye, wheat, barley and oat cDNA and genomic DNA probes. The level of polymorphism was dependent on the source of the clones, with a ranking of rye〉wheat〉barley〉oat. Distorted segregations were detected in linkage groups of chromosomes 1R, 4R, 5R and 7R. When the recombination of the two reciprocal crosses was compared, no systematic increase or decrease in one or the other direction was observed suggesting that a combination of populations of reciprocal crosses is possible.

The objective of the present study was to refine the placement of the centromere using secondary trisomics. Two independently isolated secondary trisomics (having an isochromosome plus two normal homologs) for 4S were recovered. RFLP analysis of populations segregating for them placed the centromere of chromosome 4 between bnl15.45 and bnl7.20, two RFLP loci that are 5.4-cM apart on the UMC map and 11.5-cM apart on the BNL map.

    Notizen: Abstract  Selection for high specific leaf weight (SLW) in soybean [Glycine max (L) Merr.] may increase apparent photosynthetic rate per unit leaf area (AP), which in turn may improve seed yield. In general, the SLW and leaf size are negatively correlated in soybean. To maximize total photosynthetic performance, and perhaps the seed yield, of a soybean cultivar, it would be necessary to establish a large leaf area rapidly while maintaining a high SLW. The objective of the present study was to identify quantitative trait loci (QTLs) conditioning SLW and leaf size in soybean. One hundred and twenty F4-derived lines from a ‘Young’×PI416937 population were evaluated using restriction fragment length polymorphism (RFLP) markers. The genetic map consisted of 155 loci on 33 linkage groups (LGs) covering 973 cM of map distance. The phenotypic data were collected from two different environments – a greenhouse at Athens, Ga. and a field site at Windblow, N.C. The SLW and leaf-size measurements were made on leaves from the 8th and 9th node of soybean plants at the V12 stage of development. Combined over environments, six putative independent RFLP markers were associated with SLW, and four of these loci were consistent across environments. Individually, the six markers each explained between 8 and 18% of the phenotypic variation among lines for SLW. The Young alleles contributed to a greater SLW at four of the six independent marker loci, and transgressive segregation occurred among the progeny for SLW. Three putative independent RFLP markers were associated with leaf size, each explaining between 6 to 11% of the phenotypic variation in the trait, and one of these markers was identified in both environments. There was no correlation between SLW and leaf size in this population. Similarly, none of the six QTLs conditioning SLW were linked to any of the three QTLs for leaf size. In this soybean population, it is possible to select for progeny lines with greater SLW than either parent perhaps without affecting the leaf size. It is feasible to pyramid all of the desirable alleles for greater SLW and large leaf size in a single genetic background.

    Notizen: Abstract  Prediction of the means and genetic variances in segregating generations could help to assess the breeding potential of base populations. In this study, we investigated whether the testcross (TC) means and variances of F3 progenies from F1 crosses in European maize can be predicted from the TC means of their parents and F1 crosses and four measures of parental genetic divergence: genetic distance (GD) determined by 194 RFLP or 691 AFLPTM 1 markers, mid-parent heterosis (MPH), and absolute difference between the TC means of parents (∣P1−P2∣). The experimental materials comprised six sets of crosses; each set consisted of four elite inbreds from the flint or dent germplasm and the six possible F1 crosses between them, which were evaluated for mid-parent heterosis. Testcross progenies of these materials and 20 random F3 plants per F1 cross were produced with a single-cross tester from the opposite heterotic group and evaluated in two environments. The characters studied were plant height, dry matter content and grain yield. The genetic distance between parent lines ranged between 0.17 and 0.70 for RFLPs and between 0.14 and 0.57 for AFLPs in the six sets. Testcross-means of parents, F1 crosses, and F3 populations averaged across the six crosses in a particular set generally agreed well for all three traits. Bartlett’s test revealed heterogeneous TC variances among the six crosses in all sets for plant height, in four sets for grain yield and in five sets for dry matter content. Correlations among the TC means of the parents, F1 crosses, and F3 populations were highly significant and positive for all traits. Estimates of the TC variance among F3 progenies for the 36 crosses showed only low correlations with the four measures of parental genetic divergence for all traits. The results demonstrated that for our material, the TC means of the parents or the parental F1 cross can be used as predictors for the TC means of F3 populations. However, the prediction of the TC variance remains an unsolved problem.

    Notizen: Abstract  A gene determining the restoration of cytoplasmic genic male sterility (CMS) caused by the Gülzow (G)-type cytoplasm was mapped by analyzing an F2 and F3 population comprising 140 and 133 individual plants, respectively. The target gene, designated Rfg1, was mapped on chromosome 4RL distally to three RFLP (Xpsr119, Xpsr167, Xpsr899) and four RAPD (XP01, XAP05, XR11, XS10) loci. Xpsr167 and Xpsr899 are known to be located on the segment of chromosome 4RL which was ancestrally translocated and is homoeologous to the distal end of other Triticeae 6S chromosomes. It is suggested that Rfg1 may be allelic to the gene determining the restoration of rye CMS caused by the Pampa (P) cytoplasm (chromosome 4RL) and to Rfc4 that on rye addition lines of chromosome 4RL restores male fertility of hexaploid wheat with T. timopheevi cytoplasm. Homoeoallelism to two loci for cytoplasmic-male-sterility restoration on chromosomes 6AS and 6BS in hexaploid wheat is also suggested.

    Notizen: Abstract  We report a strategy for developing codominant PCR-based genetic markers by using sequenced cDNA clones from loblolly pine (Pinus taeda L.). These clones were previously used as probes for detecting restriction fragment length polymorphisms (RFLPs) to generate linkage maps. After assessing the complexity of banding patterns from Southern blots, we selected clones representing relatively simple gene families, and then determined nucleotide sequences for about 200 bp at each end of the cDNA inserts. Specific PCR primers were designed to amplify samples of genomic DNA derived from two loblolly pine mapping populations. Polymorphisms were detected after digesting the amplified DNA fragments with a battery of restriction endonucleases, and most polymorphisms were inherited in a Mendelian fashion. These newly identified genetic markers are codominant and relatively simple to use. By assaying DNA from individuals used to construct RFLP maps, we show that most of these markers map to the same position as the RFLP loci detected using their corresponding cDNAs as probes, implying that these markers have been converted from RFLP to PCR-based methods. These PCR-based markers will be useful for genome mapping and population genetics.

We were able to target the Rfp1 locus as efficiently by comparing NILs as by bulked segregant analysis, and it was demonstrated in this instance that double-screening strategies could significantly improve the overall targeting efficiency. The chance occurrence of shared homozygosity at specific unlinked chromosomal regions in the bulks was found to limit the efficiency of bulked segregant analysis, while the efficiency of NIL comparison was limited by residual DNA from the donor cultivar at scattered sites throughout the genome of the NILs.

    Notizen: Abstract  Polymorphism in the lengths of restriction fragments at 53 single-copy loci, the rRNA locus Nor3, and the high-molecular-weight glutenin locus Glu1 was investigated in the D genome of hexaploid Triticum aestivum and that of Aegilops tauschii, the source of the T. aestivum D genome. The distribution of genetic variation in Ae. tauschii suggests gene flow between Ae. tauschii ssp. strangulata and ssp. tauschii in Iran but less in Transcaucasia. The “strangulata” genepool is wider than it appears on the basis of morphology and includes ssp. strangulata in Transcaucasia and southeastern (SE) Caspian Iran and ssp. tauschii in north-central Iran and southwestern (SW) Caspian Iran. In the latter region, Ae. tauschii morphological varieties ‘meyeri’ and ‘typica’ are equidistant to ssp. strangulata in Transcaucasia, and both belong to the “strangulata” genepool. A model of the evolution of Ae. tauschii is presented. On the geographic region basis, the D genomes of all investigated forms of T. aestivum are most closely related to the “strangulata” genepool in Transcaucasia, Armenia in particular, and SW Caspian Iran. It is suggested that the principal area of the origin of T. aestivum is Armenia, but the SW coastal area of the Caspian Sea and a corridor between the two areas may have played a role as well. Little genetic differentiation was found among the D genomes of all investigated free-threshing and hulled forms of T. aestivum, and all appear to share a single D-genome genepool, in spite of the fact that several Ae. tauschii parents were involved in the evolution of T. aestivum.

Thirty-one Triticeae homoeologous group-6 DNA probes were used to map RFLP loci in the deletion lines and the wheat-H. villosa disomic substitution (DS) line 6V?2(6A). Nine probes failed to detect polymorphism between Chinese Spring and DS6V?2(6A). Ten of sixteen polymorphic short-arm loci were not detected in del6V?2S-1. Thus, the loci are located in the deleted distal chromosome region. Six RFLP markers were mapped in the proximal 58% of 6VS. Of 20 DNA markers specific for 6VL, six mapped in the distal 36% of the long arm, and nine mapped in the proximal 64% of 6VL. The breakpoint of the short arm of 6V?2 occurs between Xpsr106 and Xcdo270, and that of the long arm between Xpsr915 and Xmwg934. The powdery mildew resistance gene Pm21 is located on the short arm of chromosome 6V?2. Pm21 is present in del6V?2S-1, and can be further mapped in the proximal 58% of 6V?2S.

How their work influenced the development of mycology will be demonstrated. It is often forgotten that the authorities attached to fungal names, including those we use in mycology on a daily basis, often belong to amateurs. The true professions of these amateurs, the net-work of correspondents they developed, etc. will be revealed and discussed in the context of an overall understanding of fungi as organisms. In addition the formation of mycological societies, and how they have become a focus of amateur activity and a source of accurate and disciplined information valuable to professional scientists will be plotted. The kind of work undertaken by amateurs I refer to will be demonstrated by documenting the activities of the British Mycological Society which celebrated its centenary in 1996. The continuing work of this band of workers and their counterparts throughout the world in the closing years of the millennium will be described. It is argued that they are nationally important and necessary resources, despite many governments or their advisers, especially the most influential ones, being under the delusion that systematics is not cutting edge science. It is also emphasized that amateurs will have an even more important role to play in the future as custodians of knowledge—until hopefully opinions change.

    Notizen: Abstract The genus Dunaliella comprises 28 species defined primarily by morphological and physiological criteria, which vary considerably depending on growth conditions. Concomitantly, the taxonomic status of various species is uncertain. To confirm the taxonomic identity and to better understand the relationship within Dunaliella, seven taxa ( D. salina, D. bardawil, D. tertiolecta, D. parva, D. viridis, D. lateralis, D. peircei) were compared using RFLP analysis of the nuclear rDNA repeats, specifically the internal transcribed spacer regions, including the 5.8S rRNA gene. Volvox aureus was used as an outgroup. A single ITS PCR amplification product was obtained for each taxon. An ITS fragment of ca. 640 bp was present in all the taxa within the subgenus Dunaliella, except for D. salina CCMP 1303 (ca. 540 bp) and D. lateralis (subgenus Pascheria) (ca. 600 bp). A cluster analysis based on the presence or absence of bands generated by digestion of the PCR product with 8 restriction endonucleases (DpnI, HhaI, EcoRI, PvuII, TaqI, HaeIII, MspI, StyI) revealed no correlation between the genetic relationship inferred from the ITS-RFLP data and the morpho-physiological attributes used for taxonomy. In addition, differences in morphology, physiology and in the length and restriction fragment patterns of the ITS region of D. salina CCMP 1303 suggest that this strain does not belong to Dunaliella.

    Notizen: Abstract Restriction fragment length polymorphism and isoenzyme markers were used to investigate quantitative trait loci involved in sunflower resistance to mycelial extension of Sclerotinia sclerotiorum on leaves and capitula. Seed weight, oil content and flowering data were also evaluated. Four quantitative trait loci were demonstrated for leaf resistance and two for capitulum resistance. One of these zones appears involved in resistance to both types of S. sclerotiorum attack while the others appear specific for resistance of one part of the plant. Two quantitative trait loci were detected for seed weight, three for oil content and three for flowering date. Individual quantitative trait loci explained 9% to 48% of the phenotypic variability, confirming the polygenic basis of the quantitative traits studied. Overall, the quantitative trait loci explain 60% of the genetic variation for leaf resistance and 38% for capitulum resistance to S. sclerotiorum. One linkage group is particularly interesting since it includes quantitative trait loci for all the five quantitative traits measured. Hypotheses for linkage versus pleiotropy and consequences of all the results in resistance breeding are discussed.

    Notizen: Abstract In Dutch forests the species density of vascular plants ranges from 1 to 61 species per 300 m2. The vascular plant species density is high in the coastal dunes, southern Limburg, river valleys, and fen areas. With the exception of southern Limburg, these areas constitute the Holocene part of the Netherlands. Low species densities occur in the sandy centre of the country. To a large extent, the areas of high species density of vascular plants follow the main river valleys. The bryophyte species densities range from 0 to 21 species per 300 m2. High bryophyte species densities occur mainly in the sandy centre and in the north-eastern part of the Netherlands. The highest species densities occur in fen woodlands and derelict coppices. Bryophyte species density is low in the coastal dunes and the very young woodlands in the recently reclaimed areas (polders). The species density contour maps of vascular plants and bryophytes in the Netherlands have little in common.

    Notizen: Abstract Forests in the Netherlands are heavily under stress. Recent surveys suggest that about one-third of the forest area in the Netherlands is affected by desiccation. Generally, plant species of moist situations decline, whereas drought tolerant species tend to increase. Besides desiccation, adverse ecological effects of acidification and nitrogen deposition also occur. Their combined action is held responsible for, among others, the decline of oligotrophic vascular plants, lichens and mycorrhizal fungi. At the same time, N-demanding species increase, which is partly caused by nitrogen deposition, and is partly a secondary effect of desiccation through aeration and concomitant mineralization. Nutrient balance of trees is disrupted. Effects on animals also occur: small snails in forest on acid soil decrease, causing Ca deficiency in birds. Measures to reduce these impacts include restoration of the former hydrology, liming, fertilization and removal of N-saturated littler layers.

    Notizen: Abstract Based on a transect consisting of 19 identical trap stations in cultivated areas and seminatural habitats, the correlation of species numbers of higher taxonomic groups with total species numbers of flowering plants and arthropods per trap site was calculated. A total of 191214 invertebrate specimens and 2221 species of plants and animals were analysed. Considering the value of the correlation coefficient R2 as well as the effort for sorting and identification, a ‘top twenty’ list of indicator groups favours Heteroptera, flowering plants, Symphyta and aculeate Hymenoptera as the best choice for biodiversity evaluation. In general, flight traps rated better than pitfall traps. In most taxonomic groups, diversity indices such as the Shannon and the Simpson index were only weakly correlated with local species diversity.

    Notizen: Abstract Polychaetes are well represented in most marine and estuarine environments, both in terms of number of individuals and species, and they typically contribute a significant percentage of the total macrofaunal diversity. They exhibit considerable variations in recruitment in both time and space, which is then often reflected in adult distributions. Whilst families and genera of polychaetes have wide distributions, normally species have discrete distributions. Polychaetes are often classified according to their diverse feeding guilds. They play a major role in the functioning of benthic communities, in terms of recycling and reworking of benthic sediments, bioturbating sediments and in the burial of organic matter. Some species form dense tubiculous colonies which can radically change recruitment patterns of other infaunal organisms. Polychaetes, by their burrowing and feeding activity, may considerably enhance various sedimentary processes. However, much remains to be learnt as to how benthic communities function, and how they may change in function as they are increasingly being impacted especially in coastal waters adjacent to centres of population.

    Notizen: Abstract More than 6500 species of Collembola are known from throughout the world and these are only a small part of the still undescribed species. There are many checklists and catalogues of Collembola for smaller territories and entire continents. Biogeographical analyses have been made for some genera and smaller territories. The most serious problems for a global biogeographical analysis is the lack of enough records from immense territories of all continents. Local biodiversity of Collembola can be very high, reaching over 100 species in small mountain ranges. Sampling methods do not impede documenting biodiversity on a global scale. Collembola have well differentiated ecomorphological life-forms and feeding guilds which enable the functional role that Collembola play in ecosystems to be recognised in some degree. Collembola play an important role in plant litter decomposition processes and in forming soil microstructure. They are hosts of many parasitic Protozoa, Nematoda, Trematoda and pathogenic bacteria and in turn are attacked by different predators. They utilise as food Protozoa, Nematoda, Rotatoria, Enchytraeidae, invertebrate carrion, bacteria, fungi, algae, plant litter, live plant tissues, and some plant pathogens. Soil acidification, nitrogen supply, global climate change and intensive farming have greatly impacted collembolan diversity.

    Notizen: Abstract The purpose of this article is to pull together various elements from current knowledge regarding the natural history of free-living protozoa in fresh waters. We define their functional role, set the likely limits of ‘biodiversity’, and explore how the two may be related. Protozoa are unicellular, phagotrophic organisms, and 16 phyla of protists contain free-living freshwater protozoan species. They are the most important grazers of microbes in aquatic environments and the only grazers of any importance in anoxic habitats. In sediments, ciliates are usually the dominant protozoans. Benthic ciliate biomass accounts for slightly less than 10% of total benthic invertebrate biomass, but ciliate production may equal or even exceed invertebrate production. Freshwater protozoan species are probably ubiquitous, although many may persist locally for long periods in a cryptic state – as ‘potential’ rather than ‘active’ biodiversity. As protozoa are among the largest and most complex of micro-organisms, it follows that bacteria and all other smaller, more numerous microbes are also ubiquitous. The number of protozoan species recorded in local surveys (232) is about 10% of the estimated global species richness (2390). The 'seedbank’ of protozoan (and microbial) species ensures that local microbial diversity is never so impoverished that it cannot play its full part in ecosystem functions such as carbon fixation and nutrient cycling.

    Notizen: Abstract Ants are important components of ecosystems not only because they constitute a great part of the animal biomass but also because they act as ecosystem engineers. Ant biodiversity is incredibly high and these organisms are highly responsive to human impact, which obviously reduces its richness. However, it is not clear how such disturbance damages the maintenance of ant services to the ecosystem. Ants are important in below ground processes through the alteration of the physical and chemical environment and through their effects on plants, microorganisms, and other soil organisms. This review summarizes the information available on ant biodiversity patterns, how it can be quantified, and how biodiversity is affected by human impacts such as land use change, pollution, invasions, and climate change. The role of ants in ecosystems is discussed, mainly from the perspective of the effects of ground-dwelling ants on soil processes and function, emphasizing their role as ecosystem engineers. Some lines of research are suggested after demonstrating the gaps in our current information on ant-soil interactions.

    Notizen: Abstract Conservation of the vegetation, analysis of the knowledge of the diversity of fungi, new biological methods and estimates of the numbers of species of fungi that grow in Mexico are discussed. The great forest destruction that is occurring and the loss of fungal diversity in the country is seriously depleting total biodiversity, mainly in the tropics. Methods to establish the number of Mexican species of fungi are proposed based on an average of: (1) the numbers of fungal obligates of vascular plants and animals and of the saprobic species; (2) extrapolating the number of the British fungi to Mexico, taking the British mycota as the best known in the world; and (3) extrapolating to the country the number of species known to occur in the State of Veracruz (Mexico), the best known in the country. The average of these three calculations yields about 200 000 species of fungi for Mexico. Of these, only 3.5% are known, despite considerable progress having been made in the inventory of the country's fungi in the last 30 years. It is concluded that it is necessary to place more emphasis on alpha taxonomy, to train more specialists and to prepare monographs of several groups of fungi.

    Notizen: Abstract While tropical lowland peatswamp ecosystems are well-represented among the major forest formations of Borneo, they are rare outside South-East Asia. They are richly endowed with biological diversity, including the crocodile (Crocodylus porosus), which is a threatened species, and are characterized by their organic (histosol) soils that have developed recently, approximately within the past 5000 years. Their unique ecological features, as well as logistical difficulties in accessing and utilizing peatswamps, have not, however, deterred exploitation. Some of the tallest, most commercially desirable timber species, such as ramin (Gonystylus bancanus) have been harvested routinely in the peatswamp forests of Sarawak. Also, due to its primary location in coastal lowlands in close proximity to human activity, drained and reclaimed peatswamp land is desirable for development of highways, housing, and industrial and plantation estates. Ecological information is needed urgently by decision-makers and land-owners, who may be contemplating conversion of natural peatswamp ecosystems to other land uses, to undergird conservation policy and best management practices for the wise stewardship and sustainable development of this ecosystem. The paper provides an overview of peatswamp ecology with recommendations for avoiding or reducing potential ecological impacts associated with peatswamp development in Borneo, which include a moratorium on further development until a Borneo peatswamp survey and inventory as well as a holistic, multi-sectoral, integrated environmental assessment and planning procedure are completed; gazetting of pristine peatswamp forest parcels as totally protected areas; implementing highly restricted multiple-use policy that allows development only in degraded areas, and insuring protection of the critical hydrological role of peatswamps in providing fresh water supply for human use as an overriding criterion in assessing any development scheme.

    Notizen: Abstract Marine fungi are an ecological rather than a taxonomic group and comprise an estimated 1500 species, excluding those that form lichens. They occur in most marine habitats and generally have a pantropical or pantemperate distribution. Marine fungi are major decomposers of woody and herbaceous substrates in marine ecosystems. Their importance lies in their ability to aggressively degrade lignocellulose. They may be important in the degradation of dead animals and animal parts. Marine fungi are important pathogens of plants and animals and also form symbiotic relationships with other organisms. The effect of disturbances on marine fungi is poorly investigated. Keystone marine species may exist, especially in mutualistic symbioses. However, as many saprophytes appear to carry out the same function simultaneously, they may be functionally redundant. The need for a concerted effort to investigate the biodiversity and role of marine fungi globally and on as many substrata as possible is presented.

    Notizen: Abstract Resistance to maize streak virus (MSV) is an essential trait of improved maize varieties in sub-Saharan Africa. We mapped quantitative trait loci (QTL) for resistance to MSV in a population of 196 F2:3 lines derived from a cross between the maize inbred lines CML202 (resistant) from CIMMYT-Zimbabwe and Lo951 (susceptible) from Italy. Field tests were planted at two locations in Zimbabwe, inoculated with viruliferous leaf hoppers (Cicadulina mbila), and scored twice (21 and 83 days after infesting, DAI) on a 1–5 scale. The mean final streak intensity (score 2) of the parent lines was 2.2 (CML202) and 4.8 (Lo951). Genotype × location interaction was large for score 1 but negligible for score 2. Consequently, the heritability was higher for score 2 (0.93) than for score 1 (0.62). By composite interval mapping across locations, using a linkage map with 110 RFLP loci, four significant (LOD ≥3.0) QTL were identified for score 1 on chromosomes (C) 1, 2, 3, and 4, respectively. All four were contributed by CML202. For score 2, only the QTL on C 1 was significant (LOD =37), explaining 59% of the phenotypic and 64% of the genotypic variance. The QTL's partially dominant gene action was consistent with the nearly intermediate resistance of the F1 generation (relative heterosis for resistance 12%). The presence of one major QTL is consistent with the bimodal frequency distribution of the mapping population showing a clear 3:1 segregation. This gene seems to be allelic or identical to Msv1, a major resistance gene which was previously identified in the same genomic region in Tzi4, an inbred line from IITA. Inbred CML202 had lower final disease ratings than Tzi4.     Notizen: Abstract In 1993–1996, the virulence of regional populations of the wheat powdery mildew pathogen (Erysiphe graminis DC f. sp. tritici Marchal) from the Czech Republic, Austria, Hungary and Slovakia against 13 resistance genes was investigated. The populations differed mainly at the regional level. Populations from the Czech Republic, mainly from the western regions, showed higher values of virulence against the Pm4b gene. Lower frequency of virulence against Pm4b was found in Austria, and the lowest value was observed in Hungary. The differences in frequencies of virulence against Pm4a and Pm4b showed a similar geographic pattern across the four countries: a continuous decline from west to east and from north to south. Virulence against Pm2 decreased in all countries considered; virulence to pm5, Pm6, Pm8 and Mli was high throughout. Genes and gene combinations that can ensure a relatively effective biological protection against this pathogen across Central Europe at present are Pm3b, Pm2+Mld and Pm1+2+9. Czech and Slovak populations were the most complex: virulence complexity reached a maximum in Slovakia in 1994. A similar evolution, though less significant, was observed in the Czech Republic. Data on complexity of isolates suggest that Central European populations of wheat powdery mildew tend to reach an intermediate level representing the optimal number of virulence genes. This process is probably a consequence of stabilizing selection.

    Notizen: Abstract Meiosis was examined in pollen mother cells of F1 hybrids made from crosses between wheat (Triticum aestivum) and lymegrass (Leymus arenarius and L. mollis). Fluorescence genomic in situ hybridization detected pairing between wheat and lymegrass chromosomes during prophase I and metaphase I. Such pairing, when resulting in bivalent formation, was likely to yield correct disjunction, and hence intergenomic recombination could be incorporated into the gametes. Bivalents in these hybrids, however, were more frequently formed between chromosomes of the same parental origin. Univalents were common, whereas multivalents were not clearly detected. Meiotic behaviour in some cells was not totally aberrant, and this may have accounted for the presence of normal pollen. The results are discussed in relation to intergenomic pairing, meiotic behaviour in wide-hybrids and genome relationships, including the Leymus genome origin.

    Notizen: Abstract In plants, several types of receptor-like kinases (RLK) have been isolated and characterized based on the sequence of their extracellular domains. Some of these RLKs have been demonstrated to be involved in plant development or in the reaction to environmental signals. Here, we describe a RLK gene family in wheat (wlrk, wheat leaf rust kinase) with a new type of extracellular domain. A member of this new gene family has previously been shown to cosegregate with the leaf rust resistance gene Lr10. The diversity of the wlrk gene family was studied by cloning the extracellular domain of different members of the family. Sequence comparisons demonstrated that the extracellular domain consists of three very conserved regions interrupted by three variable regions. Linkage analysis indicated that the wlrk genes are specifically located on chromosome group 1 in wheat and on the corresponding chromosomes of other members of the Triticeae family. The wlrk genes are constitutively expressed in the aerial parts of the plant whereas no expression was detected in roots. Protein immunoblots demonstrated that the WLRK protein coded by the Lrk10 gene is an intrinsic plasma membrane protein. This is consistent with the hypothesis that WLRK proteins are receptor protein kinases localized to the cell surface. In addition, we present preliminary evidence that other disease resistance loci in wheat contain genes which are related to wlrk.

    Notizen: Abstract The effect of 24 treatment combinations of cultures of Streptomyces costaricanus sp. nov. (ATCC55274), Bacillus thuringiensis (ATCC55273) and a strain of Paecilomyces marquandii, nematicide (cadusaphos), and/or wheat mash on growth and response of potted banana plants (Musa AAA) and populations of Radopholus similis, Helicotylenchus multicinctus and free living nematodes were studied in Río Frío, Costa Rica. The best plant responses (height, leaf numbers, healthy root weight), lowest numbers of plant parasitic nematodes and highest numbers of free living nematodes were observed for treatments containing wheat as a component. Two treatments, viz. wheat + Streptomyces costaricanus (200-ml culture) and wheat + P. marquandii (200-ml culture), gave the overall best results. Numbers of free living nematodes increased up to 1500-fold only for treatments containing wheat. Significant positive correlations existed between numbers of free living nematodes and shoot weight, healthy root biomass, plant height, and leaf numbers. Non-wheat treatments, including nematicide only, gave the poorest responses in general. Observations of nematodes sampled 50 days following planting in wheat-containing treatments showed most of the free-living nematodes (≈ 90%) to be infected by nematophagous fungi (species not recorded). The results show that an organic amendment to soil, with or without a microbial component, can be an effective inducer of processes that regulate plant-parasitic nematode populations in soil.

    Notizen: Abstract Concern regarding the ecological impact of rapid global warming has encouraged research on climate-induced changes in biological systems. Critical problems, still poorly understood, are the potential for rapid adaptive responses and their genetic costs to populations. The O chromosomal polymorphisms of Drosophila subobscura have been monitored at a southern Palearctic locality experiencing sustained climate warming since the mid-1970s. Observations suggest that the population is rapidly evolving in response to the new environmental conditions, and has lost a significant amount of chromosomal diversity (18.3% in 16 years). These findings are consistent with results from another population of D. subobscura, which is also undergoing climate warming, and are in accord with what would be expected from latitudinal and seasonal patterns of the various inversions. In addition, data on the O chromosomal polymorphisms from other localities throughout t he range of this species suggest that other populations vary similarly.

    Notizen: Abstract The residual effects of Zn supplied (+Zn: 1 mg kg−1 soil, -Zn: no Zn added) in subsoil to four oilseed rape genotypes (Zhongyou 821, Xinza 2 and Narendra: Brassica napus type, and CSIRO-1: Brassica juncea type) were studied on the following wheat in a glasshouse experiment in pots (100 cm long, 10.5 cm diameter). The topsoil (upper 20 cm soil in pots) was supplied with Zn (0.5 mg kg−1 soil) in all treatments whereas Zn (1 mg Zn kg−1 dry soil) was either supplied or omitted from the subsoil at time of sowing of oilseed rape genotypes. Oilseed rape plants were harvested at maturity and wheat (variety Songlen) was grown to evaluate the residual effects of subsoil Zn and oilseed rape genotypes. Subsoil residual Zn significantly improved the root growth, yield attributes, grain and straw yield of wheat. Wheat grain Zn concentration was 4 times higher in +Zn subsoil residual than under -Zn subsoil residual. Oilseed rape genotypes did not vary in their residual effect on grain and straw yield of wheat, but reflected a differential effect on Zn concentration and Zn content of wheat grain. Zinc uptake per wheat plant (grain + shoot + root Zn uptake) was about 4 times higher in +Zn subsoil residual than -Zn subsoil residual. Out of total Zn uptake per plant, 77%, 15% and 8% of Zn was loaded in wheat grains, shoots and roots respectively in +Zn subsoil residual. However in -Zn subsoil residual, distribution of Zn in grains, shoots and roots was 55%, 29% and 16% respectively. Zinc loading in grains of wheat was significantly higher in wheat grown after Narendra and Xinza 2 compared with wheat grown after Zhongyou 821 (Zn-inefficient genotype). These results indicate that subsoil residual Zn and Zn-efficient genotypes of oilseed rape have the potential to increase the Zn loading in wheat grains.

    Notizen: Abstract A recently developed bean RFLP linkage map was used to identify genetic elements affecting quantitative trait loci (QTLs) in two contrasting common bean genotypes, BAT-93 and Jalo EEP558, under two levels of mineral nitrogen: low – 0.25 mM NH4NO3 and a high – 6 mM NH4NO3. QTLs affecting nodule number (NN) and response to Xanthomonas campestris bv. phaseoli, which causes common bacterial blight (CBB) were identified and mapped. Analyses of 70 F2-derived F3 families, using the F1, the two parents, and a nodulation-defective mutant (Nod-) inoculated with R. tropici UM1899 under both levels of N showed significant differences (P#60;0.0001) among the F3 families for NN. Under low N, three genomic regions influenced both traits, with seven linked markers. In three of the six regions influencing NN, higher NN was associated with the Jalo EEP-558 allele, whereas in only two regions was the BAT-93 allele associated with higher NN. One-way analysis of variance, with each marker as the independent variable and NN as the dependent variable, and interval mapping analysis identified four QTLs, which accounted for 45% of the total variation, and two additional QTLs near to yet unassigned loci. In linkage group D7, one QTL mapped to the same region as a QTL for CBB. Under high N, three additional regions were linked to NN, one where the BAT-93 allele was closely associated with CH18 (chitinase), and the others where the Jalo EEP-558 allele was associated with CHS (chalcone synthetase) and PAL-1 (phenylalanine ammonia lyase). Four regions for CBB were mapped adjacent to or in the same region as a QTL for NN. Thus, N showed dual and opposite effects on the expression of NN and CBB. Analysis of these RFLP markers revealed these ‘hidden’ favorable alleles and can serve as an indirect selection tool to increase NN and resistance to CBB.

Sunn pest, known in Turkey since 1927, is one of the most harmful pests of wheat in this region where the infested area has reached 0.3 million ha and the cost of chemical application has reached US$ 2 million per year. Two species of Eurygaster, E. maura and E. austriaca, are prevalent in the region. Observations of tests carried out on different genotypes under natural field conditions and controlled greenhouse tests showed variable response of wheat to sunn pest.

    Notizen: Abstract A method for efficient genetic transformation of wheat has been developed using immature embryos as targets for microprojectile-mediated gene transfer and a helium driven particle delivery system. Screening and selection of transgenic cells, somatic embryos and regenerated plants are performed with the gus-gene and the phosphinothricin acetyl transferase (bar) gene coding for Basta-resistance as the selectable marker. On average, one fertile transgenic plant can be obtained from about 100 microprojectile treated, immature embryos. The number of integrated copies of the transferred gene ranges from 1 up to about 10. Stable integrated genes are inherited in most of the transgenic lines in a normal mendelian fashion segregating 3:1 in the F2. Homozygous, as well as heterozygous, lines have been followed and analysed genetically at the molecular level and up to F5. Apart from normal stable gene expression, examples have also been found which showed a loss of gene activity or unexpected segregation pattern. For applied aspects, different genes are transferred aiming for improved disease resistance, modification of quality, or other characteristics. First results from these transgenic lines are reported, and problems still existing with the production of stable transgenic wheat lines are discussed.

    Notizen: Abstract A study of the genetic diversity in coconut by RFLP analysis was performed in 100 individuals representing 10 Tall and seven Dwarf local populations or 'ecotypes' from various geographical origins. Nine cDNA clones from rice, one mitochondrial DNA clone (CoxI) and one genomic clone (rDNA) from wheat were used as probe for southern hybridization. The distribution of the 40 polymorphic bands revealed by rice cDNA clones was studied using a multivariate analysis and allowed to identify two main genetical groups. The first one includes the ecotypes from the Far East and from the South Pacific, whereas the other one comprises the ecotypes from India, Sri Lanka and Western Africa. The rDNA and the CoxI probes confirm this distinction. The Far East and the Pacific regions which were the most likely center of origin also exhibit the widest polymorphism. The associations between the Panama Tall and the Pacific group and between the West African Tall and the Indian Ocean group reflect their likely origin. The Comoro Tall appears to be intermediate between the two main groups, and could reflect the old migration route between Indonesia and Madagascar. All Dwarf varieties belong to the first group, even those collected in West Africa. Those were probably introduced from Asia and Pacific at the beginning of the century. The cross-pollinating Tall ecotypes were generally more polymorphic than the self pollinating Dwarf ecotypes. The legitimacy of two hybrids between ecotypes was confirmed and maternal inheritance of mitochondrial genome was observed.

    Notizen: Abstract Substitution lines with reciprocal substitutions of chromosomes containing recessive alleles of the homoeologous group 5 chromosomeVrn genes between varieties of winter wheat with high vernalisation requirement (‘Mironovskaya 808’) and low vernalisation requirements (‘Bezostaya 1’) have been created. On this basis the genetic determination of vernalisation requirement was established. Substitution lines Mironovskaya 808 (Bezostaya 1 5A), Mironovskaya 808 (Bezostaya 1 5B), Mironovskaya 808 (Bezostaya 1 5D) and reciprocal substitution lines Bezostaya 1 (Mironovskaya 808 5A), Bezostaya 1 (Mironovskaya 808 5B) and Bezostaya 1 (Mironovskaya 808 5D) were grown under different durations of vernalisation (3, 4, 5, 6, 7 and 8 weeks) and their response was evaluated. Photoperiodic sensitivity of the original parental genotypes was also determined. Reciprocal substitution lines of the same chromosome that carries the same vrn allele responded differently to vernalisation deficit. Differences have been shown between all group 5 reciprocal substitutions. Lines carrying chromosomes 5A and 5D of Mironovskaya 808 had a high vernalisation requirement whereas lines carrying chromosome 5B of Bezostaya 1 (vrn2B) had a low vernalisation requirement. The reciprocal lines had a reverse requirement. This explains the different vernalisation requirements of the original varieties: Mironovskaya 808 with a high vernalisation requirement carries two alleles (vrn1M and vrn3M) in its genotype that increase the vernalisation requirement, whereas Bezostaya 1 with a lower requirement for vernalisation contains only one such allele (vrn2B). By combination of the alleles in the lines with the substitution of chromosome 5B carrying vrn2 allele that in both original genotypes work inversely to the other alleles, transgressive genotypes have been formed: genotype vrn1M vrn2B vrn3M determines a higher vernalisation requirement than original variety Mironovskaya 808, and genotype vrn1B vrn2M vrn3B determines a lower vernalisation requirement than the original Bezostaya 1. An incomplete vernalisation requirement prolonged the time to heading, with exponential dependence on the vernalisation deficit, or prevented heading altogether. The original varieties further differed in photoperiodic sensitivity (Mironovskaya 808 sensitive, Bezostaya 1 less sensitive) that also influenced the background of substitution lines. The impact of the background on the heading time showed itself by about one week difference between Mironovskaya 808 and Bezostaya 1 grown under 8 weeks vernalisation and normal photoperiod. The difference between the lines with Mironovskaya 808 background and the lines with Bezostaya 1 background was approximately the same and was not significantly changed in different vernalisation variants of the lines. This difference may be caused by different photoperiodic sensitivity of the original varieties, but also by other genes, such as genes of earliness per se.

    Notizen: Abstract In recent years, hybrids with levels of resistance to sorghum midge (Stenodiplosis sorghicola Coquillett) have become available to Australian sorghum producers. These hybrids have been readily accepted to the extent that more than 80% of the sorghum growing area was planted to hybrids with some level of midge resistance by 1995. Since selection for resistance to sorghum midge is one of the primary objectives of Australian sorghum breeding programs, the relationship between resistance and genetic diversity was investigated. Genetic diversity and heterozygosity were assessed using restriction fragment length polymorphism analysis among 26 grain sorghum hybrids grown commercially in Australia. The genetic distances between each sorghum hybrid and a standard highly resistant hybrid were found to be strongly negatively correlated to hybrid midge resistance ratings (r = - 0.77, p 〈 0.001). In addition, the average heterozygosity of each hybrid was correlated with their midge resistance ratings (r = - 0.54, p 〈 0.01). The results indicate that the move to midge resistant hybrids has been associated with a narrowing of the genetic diversity and average heterozygosity of commercial sorghum hybrids. Repeated use of particular elite parent lines, linkage drag and genetic drift are likely to have contributed to this decline. This reduction in genetic diversity may have implications for the genetic vulnerability of sorghum in Australia and the rate of progress in breeding for yield.

Agrobacterium T-DNA vectors containing the C1/Lc anthocyanin-biosynthesis regulatory genes, the gusA gene or a synthetic green fluorescent protein gene (sgfp-S65T) were constructed from original binary vectors. The visual T-DNA markers were used as cell-autonomous reporters of early Agrobacterium-mediated transformation events in the wheat and barley cells. This localization of the transformed cells revealed a non-random distribution throughout each embryo and callus piece.

    Notizen: Abstract A rifampicin-resistant isolate of Burkholderia (Pseudomonas) cepacia (A3R) reduced crown rot (Fusarium graminearum Group 1) symptoms significantly (P ≤ 0.05) in wheat in glasshouse and field experiments and increased grain yield significantly (P ≤ 0.05) in one of two field experiments. In glasshouse experiments, applying the bacteria as a soil drench (2.5 × 109 cfu/g soil) was more effective than coating the bacteria on wheat seed (3.4 × 107cfu/seed). In field experiments, the bacteria were applied as a soil drench at the rate of 1.8 x 1010 cfu/m row. In both the glasshouse and the field, disease severity in the bacteria-inoculated treatments was significantly less in a silt loam than in a sandy loam. The silt loam had a large proportion of fine clay and silt particles (51.7%), which may have favoured the biocontrol activity and survival of the introduced B. cepacia. In a glasshouse experiment, control by B. cepacia was significantly greater in the silt loam than in the sandy loam, which in turn was greater than in a loamy sand. The loamy sand appeared to favour crown rot development but not the activity or survival of the bacterial antagonist. The latter was reflected by the relative populations of the rifampicin-resistant bacteria re-isolated from the various soils during a 5-week period after application of the bacteria (silt loam 〉 sandy loam 〉 loamy sand). This study further confirms that soil type can influence the populations and the level of biocontrol activity of some bacterial antagonists.

    Notizen: Abstract Sterility has emerged as a widespread problem for wheat (Triticum aestivum) production in South and South-east Asia. Whilst boron (B) deficiency is commonly associated with sterility in wheat, the expression of sterility is complicated in rainfed conditions by a number of environmental factors including water stress. A field experiment was conducted to examine the effect of water stress on B response of wheat genotypes on a low B soil (0.087 mg B kg-1soil) at Chiang Mai, Thailand (18°45′ N, 99° E) during the dry season from November to March. The experiment consisted of three factors arranged in a split-split plot design with two levels of irrigation: water stress (I−) and full irrigation (I+) in main plots, two levels of B: 0 kg (B0) and 1 kg B ha-1 (B+) as borax in sub plots and four wheat genotypes: SW 41, BL 1022, UP 262 and Sonora 64 in sub–sub plots. Water stress was applied by discontinuing irrigation in I− treatments after the double ridge stage. In all genotypes, above ground biomass was decreased by I−, but not by B deficiency. Significant B×genotype interactions were detected for reproductive growth. SW 41 and BL 1022 strongly responded to added B with relief from B deficiency symptoms at anthesis and improved grain set index (GSI), grains ear-1, ears with grain and grain yield at maturity. By contrast, Sonora 64 could set grain well at B0 and did not show any response to added B with respect to these parameters. Grains ear-1 of SW 41 and BL 1022 was not affected by full irrigation at B0, but were significantly increased when fully irrigated with added B. In all genotypes, B concentration of the flag leaf and the ear at booting and at anthesis was significantly higher in B+, but was not affected by irrigation. Boron × irrigation interactions detected in this study indicate the possibility of the influence of water stress on the severity of wheat sterility in South and South-east Asia.

    Notizen: Abstract The estimation of soil water reserves is essential for irrigation management. The usual way of calculating these reserves, held between the soil moisture content at field capacity and the classical limit of −1.5 MPa considered as the lower limit of available water, over the rooting depth of the crop, does not correspond with the real behaviour of crops as regards their ability to extract soil water and should be only considered as the apparent available water (AAW). Measurements of moisture profiles made using a neutron probe soil moisture meter from 1970 until 1991 on unirrigated crops at the INRA Agronomy Station at Toulouse-Auzeville, France, on a deep silty clay soil with a high water holding capacity have enabled us to define the water extraction capacities of maize ( Zea mays L.), sunflower (Helianthus annuus L.), sorghum (Sorghum bicolor L. Moench), soya bean (Glycine max L. Merr.), and winter wheat (Triticum aestivum L.). The results show, not only that all the crops can extract soil water from beyond −1.5 MPa in the surface layers to varying degrees and depths, depending on the crop, but also that deeper down, AAW is not fully used, as the moisture profile gradually returns to field capacity. Of the five crops studied, maize extracts the most water from the top 0.5 m, removing 150% of AAW. This amount falls rapidly lower down, reaching nil at 1.6 m. Conversely sunflower extracts less near the surface, but uses all AAW up to 1.2 m, and still extracts 85% of AAW at 1.6 m. Sorghum is somewhat comparable to sunflower, but with a lower use over the entire profile. Soya bean exhibits strong extraction to 1.0 m, and then much less at depth. As to wheat, its extraction capability is quite high near the surface, and then falls steadily with depth where it is still 30% of AAW at 1.6 m. Soil moisture measurements realised on a bare soil during several successive years were used to fix the maximum soil evaporation and to suggest the contribution of crops in soil water depletion from uppermost layers. The water extraction capacities have been modelled and introduced into the model EPICphase, a modified version of the model EPIC, adapted for irrigation management. Four parameters have been introduced to simulate: (1) the rooting pattern of the crop (parameter α), (2) the degree of involvement of deep layers (parameter p), (3) the fraction of AAW beyond which crop transpiration is affected (parameter t) and (4) the intensity of extraction beyond the limit of −1.5 MPa as a function of soil depth (parameter d). Calibrated on the basis of the driest year since 1970 for each crop, the model was then validated under unirrigated conditions, and then tested on irrigated maize plots. Under unirrigated conditions, the simulations correctly reproduced the water extraction by the five crops, both in an extremely dry year and in a wet year. The observed differences between simulations and observations were found mostly at about 0.1 m depth, and were due to lack of precision of moisture measurements with the neutron probe. From 0.2 to 0.6 m the simulations have a tendency to overestimate the extraction. These differences are explained by water fluxes which are especially high in these layers because of the processes of evaporation from the soil and plant transpiration, which are difficult to simulate with precision. Below 0.6 m, a more stable zone where water movements are of minor importance, the simulations are very precise. For irrigated maize, the results show a very good fit between simulation and measurement, indicating that these water extraction capacity figures could be used for irrigation management provided that the rules for exploitation of the water reserves are well established.

    Notizen: Abstract The effect of varied light intensity (50 – 600 μmol m-2 s-1) on the rate of phytosiderophore release was studied under zinc (Zn) deficiency using a bread (Triticum aestivum L. cv. Aroona) and a durum wheat cultivar (Triticum durum Desf. cv. Durati) differing in zinc (Zn) efficiency and under iron (Fe) deficiency using a barley cultivar (Hordeum vulgare L. Europe). Plants were grown under controlled environmental conditions in nutrient solution for 15 days (wheat plants) or 11 days (barley plants). Phytosiderophore release was determined by measuring capacity of root exudates to mobilize copper (Cu) from a Cu-loaded resin. With increasing light intensity visual Zn deficiency symptoms such as whitish-brown lesions on leaf blade developed rapidly and severely in wheat, particularly in the durum cultivar Durati. In wheat plants supplied well with Zn, increases in light intensity from 100 to 600 μmol m-2 s-1 did not clearly affect the rate of phytosiderophore release. However, under Zn deficiency increases in light intensity markedly enhanced release of phytosiderophores in Zn-deficient Aroona, but not in Zn-inefficient Durati. When Fe-deficient barley cultivar Europe was grown first at 220 μmol m-2 s-1 and then exposed to 600 μmol m-2 s-1 for 24 and 48 h, the rate of release of phytosiderophores was enhanced about 4-fold and 7-fold, respectively. Transfer of Fe-deficient plants from 600 to 50 μmol m-2 s-1 for 48 h reduced the rate of release of phytosiderophores by a factor of 7. The effect of light on phytosiderophore release was similar regardless of whether the rate of phytosiderophore release was expressed per plant or per unit dry weight of roots. The results demonstrate a particular role of light intensity in phytosiderophore release from roots under both Zn and Fe deficiency. It is suggested that in the studies concerning the role of phytosiderophore release in expression of Zn or Fe efficiency among and within cereals, a special attention should be given to the light conditions.

    Notizen: Abstract Denitrification and total N losses were quantified from an irrigated field cropped to maize and wheat, each receiving urea at 100 kg N ha-1. During the maize growing season (60 days), the denitrification loss measured directly by acetylene inhibition-soil cover method amounted 2.72 kg N ha-1 whereas total N loss measured by 15N balance was 39 kg ha-1. Most (87%) of the denitrification loss under maize occurred during the first two irrigation cycles. During the wheat growing season (150 days), the denitrification loss directly measured by acetylene inhibition-soil cover and acetylene inhibition-soil core methods was 1.14 and 3.39 kg N ha-1, respectively in contrast to 33 kg N ha-1 loss measured by 15N balance. Most (70-88%) of the denitrification loss under wheat occurred during the first three irrigation cycles. Soil moisture and NO 3 - -N were the major factors limiting denitrification under both crops. Higher N losses measured by 15N balance than C2H2 inhibition method were perhaps due to underestimation of denitrification by C2H2 inhibition method and losses other than denitrification, most probably NH3 volatilization.

    Notizen: Abstract The effects of NaCl salinity (100 or 150 mol m-3) and hypoxia on seedlings of several wheat varieties (Lyallpur-90, SARC-1, Pato, Tchere, Pb-85, Siete Cerros, Chinese Spring and a Chinese Spring × Thinopyrum elongatum amphidiploid) were studied in solution culture. In vivo studies of activities of different enzymes (alcohol dehydrogenase (ADH), lactate dehydrogenase (LDH) and cytochrome oxidase (COase)) extracted from Pato and Pb-85 included the effect of salinity with and without hypoxia, while during in vitro studies, NaCl, glycinebetaine and proline were added to the assay mixture. The extent of aerenchyma formation was also determined in Pato, Chinese Spring and a Chinese Spring × Thinopyrum elongatum amphidiploid. Imposition of hypoxia greatly induced ADH and LDH activity in roots of wheat seedlings after a week-long exposure. However, exposure of roots to salinity also slightly increased LDH and ADH activity compared with the non-saline control. On a relative basis, Pato had higher ADH activity under hypoxic (21×) or saline-hypoxic stress (20×) than in aerated conditions. Hypoxia alone or in the presence of salts decreased COase activity in both Pato and Pb-85. The in vitro studies revealed that NaCl (on an equimolar basis at up to 500 mol m-3) is more disruptive than glycinebetaine or proline. LDH was more sensitive to NaCl than ADH. Aerenchyma development was higher near the root-shoot interface compared to near the root tip. Salinity under hypoxic conditions significantly reduced aerenchyma development near the root tip and root-shoot interface compared to hypoxia alone. Neither enzyme activity nor aerenchyma formation could account for varietal differences in tolerance to hypoxia alone or in combination with salinity.

    Notizen: Abstract In order to evaluate wheat response to yellow rust, 25 advanced, promising and commercial bread wheat cultivars were tested as seedlings in greenhouse conditions in Karaj, Iran, and as adult plants in field conditions at four locations. Five pathotypes of yellow rust, 14E176A+, 134E142A+, 6E210A+, 4E128A- and 64E146A+ prevailing in field test locations, were used in the seedling tests. The results showed that some of the cultivars have seedling or overall resistance to the pathotypes and some have adult plant resistance. Cultivars M-70-4 and MV17 were resistant to all pathotypes as seedlings and showed good adult plant resistance.

    Notizen: Abstract The most important breeding objectives in crop improvement are improving grain yield, grain quality, and resistances to various biotic and abiotic stresses. The objectives of our study were to compare two crossing and four selection schemes for grain yield, yield traits, and slow rusting resistance to leaf rust (Puccinia recondita) based on additive genes in wheat (Triticum aestivum), and to identify the most efficient crossing and selection methodologies in terms of genetic gains and cost efficiency. Segregating populations were derived from 18 simple crosses and the same number of top (three-way) crosses. Half of the crosses were derived from Yecora 70 and the other half from Veery #10 as the common leaf rust susceptible parents. The four selection schemes were: pedigree, modified bulk (F2 and F1-top as pedigree, selected lines in F3, F4, F2-top, F3-top as bulk; and pedigree in F5 and F4-top populations), selected bulk (selected plants in F2, F3, F4, F1-top, F2-top and F3-top as bulk; and pedigree in F5 and F4-top populations), and nonselected bulk (bulk in F2, F3, F4, F1-top, F2-top and F3-top; and pedigree in F5 and F4-top populations). A total of 320 progeny lines, parents and checks were tested for grain yield, other agronomic traits and leaf rust resistance during the 1992/93 and 1993/94 seasons in Ciudad Obregon (Sonora State, Mexico) which represents a typical high yielding irrigated site. The influence of the type of cross and the selection scheme on the mean grain yield and other traits of the progenies was minimal. The selection of parents was the most important feature in imparting yield potential and other favourable agronomic traits. Moreover, the highest yielding lines were distributed equally. Progeny lines derived from Veery #10 crosses had significantly higher mean grain yield compared to those derived from the Yecora 70 crosses. Furthermore, a large proportion of the highest yielding lines also originated from Veery #10 crosses. Mean leaf rust severity of the top cross progenies was lower than that of the simple cross progenies possibly because two parents contributed resistance to top cross progenies. Mean leaf rust severity of the nonselected bulk derivatives was twice that of lines derived from the other three schemes. Selected bulk appears to be the most attractive selection scheme in terms of genetic gains and cost efficiency.

    Notizen: Abstract Due to their broad population diversity, old wheat varieties or landraces play an important role in increasing the genetic variability of agronomic traits. On these grounds, an analysis was made of the high molecular weight (HMW) glutenin subunit composition of the old Hungarian wheat variety Bánkúti 1201. It was found that several genotypes with differing breadmaking qualities can be distinguished for this character. When using old varieties in breeding, it is possible to broaden the genetic background of characters responsible for breadmaking quality by separating the populations. A more detailed analysis of the protein composition of germplasm created in this way will be required to obtain a better understanding of this complex character for its conscious introduction into breeding programmes.

    Notizen: Abstract The effect of agronomic practices and cultivars on grain yield, grain protein and small grain sievings was examined in field experiments over four years in the winter rainfall wheatbelt of Western Australia. Rotation with legume crops and pastures was the main factor responsible for increasing grain protein percent. Grain proteins were increased by 4-5% for crops grown in good legume pasture rotations compared to continuous wheat rotations, but only by 1-2% by factors such as delayed sowing time, applied nitrogen, cultivar or grass weed control. In legume based rotations, wheat crops sown at their highest yielding times produced proteins in the appropriate ranges for premium paying grades. Applying N fertilisers up to the optimum rates for yield did not result in proteins below the levels required for premium paying grades, except for hard wheats at 〉11.5% grain protein. Legume rotations and appropriate soil types were required for hard wheats to exceed 11.5% at economic N rates. The yield penalty often associated with high quality cultivars has been reduced or eliminated in the modern cultivars used in the experiments. Some longer season cultivars only produced grain proteins 〉10% if sown after their optimum time for yield, but sowing at optimum time reduced the probability of producing small grain sievings. Some cultivars were more susceptible than others to producing excessive sievings, especially those with inherently smaller than average seed size. Seed rates up to the optimum for grain yield did not result in excessive small grain sievings except where the site was highly fertile, where the crop was sown too late for optimum yield or where too much N fertiliser was used.

    Notizen: Abstract This article describes the impact of plant genetic resources on wheat breeding. It defines the important contribution of N.I. Vavilov Institute (St. Petersburg, Russia) to broadening the genetic diversity of new wheat cultivars. Special attention is given to conducting a comprehensive evaluation of intraspecific variability for valuable characters, including: 1) formation of special subcollections, consisting of accessions with useful characters and accessions representing intraspecies diversity for a given character; 2) revealing genetic differences among phenotypically superior accessions; 3) determining a genetic system of intraspecific variability for a given character; 4) formation of a genetic collection; 5) revealing and developing the donors of useful characters. An example using plant height shows that the proposed research approach adequately determines the genetic potential of species and reveals the most effective genes for practical uses. It is assumed that the main sources of genes for breeding Triticum aestivum L. in order of importance will remain: 1) intraspecific diversity of T. aestivum itself; 2) other Triticum species; 3) other genera of the Triticeae Dum. tribe (particularly Aegilops L.); 4) more remote genera of the Poaceae Barnh. botanical family. It is stressed that existing diversity of T. aestivum has been poorly investigated genetically and only partially used by breeders. Properly evaluated, it can provide multiple solutions for traditional and new problems of wheat improvement.

    Notizen: Abstract RFLP analysis of a PCR amplified 3.2-kbp region of cpDNA bounded by the conserved sequences in rbc L and ORF 106 was performed in eggplant (Solanum melongena), its related Solanum species, S. incanum, S. virginianum (= S. surattense), S. torvum, S. aethiopicum (= S. gilo), S. aethiopicum (= S. integrifolium), S. violaceum (= S. indicum), S. violaceum (= S. sanitwongsei) and S. mammosum and the reciprocal hybrids between S. aethiopicum (= S. integrifolium) and S. melongena 'Uttara'. The target region of cpDNA was amplified correctly by PCR. The amplified products were digested with each of 10 restriction enzymes (Alu I, Ase I, BamH I, Hinf I, Msp I, Rsa I, ScrF I, Sty I, Taq I and Xba I). Variations of restriction patterns among the species were recognized after digesting the amplified products with each of the seven restriction enzymes, Taq I, Alu I, Rsa I, Sty I, Ase I, Hinf I and Xba I. The restriction patterns divided the examined nine species into the following five clusters, 1) S. melongena and S. incanum, 2) S. virginianum (= S. surattense), 3) S. torvum, 4) S. aethiopicum (= S. gilo), S. aethiopicum (= S. integrifolium), S. violaceum (= S. indicum) and S. violaceum (= S. sanitwongsei) and 5) S. mammosum. The restriction pattern with Alu I in each of the reciprocal hybrids between S. melongena 'Uttara' and S. aethiopicum (= S. integrifolium) was identical with that of seed parent. The present study demonstrated the availability of the PCR-RFLP analysis of cpDNA for assessing taxonomic relationships and identifying cytoplasmic parentage of interspecific hybrids in eggplant and related Solanum species.

    Notizen: Abstract The location and effects of genomic regions for rust resistance in sorghum were determined. One hundred and sixty recombinant inbreds, which derived from a cross between QL39 and QL41, were used as a segregating population for genome mapping and rust resistance evaluation. Phenotypic data were collected in replicated field trials in two years. Interval mapping and non-parametric mapping identified four regions, each in a separate linkage group, associated with rust resistance. The region with the largest effect on rust resistance is on linkage group 10; it accounted for 40% of the total phenotypic variation.

    Notizen: Abstract Stripe rust is one of the most serious diseases of wheat in China. Two new stripe rust physiological strains CYR30 (Inter. name: 175 E 191) and CYR31 (Inter. name: 239 E 175) have become the dominant and epidemic physiological strains since 1994 in China. Resistance to these strains of 48 Ae. tauschii accessions was evaluated at the seedling and adult stages. Out of 48, 28 accessions displayed seedling and adult resistance. All of the Chinese Ae. tauschii accessions were susceptible at seedling and adult stages. Seedling resistance was highly related to adult resistance in Ae. tauschii. The new genetic resources of resistance to CYR30 and CYR31 could be incorporated into commercial wheat varieties for wheat resistance breeding by direct hybridization.

    Notizen: Abstract Polymorphism of chloroplast DNA of 106 accessions of Pyrus (mainly east Asian accessions) was examined. Four haplotypes were observed with the combination of three independent restriction site mutations digested by EcoO109 I, Sal I, and Xba I, respectively. In the occidental pear accessions only the most plesiomorphic type was observed, whereas all four types appeared in the oriental pear accessions. This suggests that the oriental species of Pyrus and occidental ones may have evolved independently. The distribution of four haplotypes in the east Asian pear was quite incongruent with the species or infrageneric classification using mainly morphological characters. Considering the high crossability and much occurrence of suspected interspecific hybrids in wild populations, this disaccord is inferred to be the results of the hybridization and introgression between species.

    Notizen: Abstract This paper focuses on the value of landraces (traditional and local crop varieties) to farmers in centers of agricultural diversity. Additional information on the factors contributing to the private value which farmers assign to landraces may help to identify a strategy for ensuring the conservation of the crop genetic resources (CGRs) which are embodied in landraces while at the same time minimizing the costs. Economic and ethnobotanical approaches for examining the value of landraces complement one another. A formal economic approach establishes a framework for quantitative analysis while ethnobotanical methods provide qualitative data for assessing the likelihood that particular farmers or farm sectors will maintain landraces. Our research synthesizes the two approaches in order to examine farmer selection of local wheat landraces in relation to that of modern varieties in three provinces in western Turkey. Multiple farmer concerns (e.g. yield, risk, quality), environmental heterogeneity, and missing markets contribute to the persistence of landraces. Household characteristics informing variety choice will also affect the household's perceptions of the importance and value of landraces.

    Notizen: Abstract Microsatellite (SSR) profiles from 65 wheats and 135 barleys have been generated, involving 14 and 22 loci, respectively. The wheat and barley varieties were chosen to represent the bulk of the area sown to these crops in the UK over the past 70 years. The profiling identified genotypic mixtures in some seed samples. Null alleles were common in wheat, but rare in barley. We describe attempts to increase the efficiency of data acquisition. High resolution agarose gel electrophoresis was unable to satisfactorily resolve 1–2 repeat unit differences in the common size range for SSR loci, and was therefore unsuitable for mass screening of allelic variants. Multiplex PCR was very dependent on the choice of primer combinations and seldom produced amplifications as consistently as when primer pairs were used individually. Background (non-specific) amplification was common to many primer pairs, and this hindered the use of both multiplex PCR and multiple sample loading. Sequential sample loading was the most effective strategy, although this was the least time-efficient of the measures used.

    Notizen: Abstract Field experiments were conducted for three years to investigate the effect of direct, residual and cumulative P application on the grain yield, P removal and P use efficiency of pigeonpea and wheat grown in rotation. Four levels of P, i.e. 0, 13, 26, and 39 kg P ha-1 were applied to either pigeonpea (Cajanus cajan L., cv. AL 15) or wheat (Triticum aestivum, cv. WL 711) or to both crops. Maximum increase in wheat grain yield ranged from 1.5 to 1.9 t ha-1 and in pigeonpea from 0.28 to 0.36 t ha-1. On average, the grain yield of pigeonpea was 8–15% higher in the residual P treatment than by direct application. In wheat, cumulative P application (to both crops) resulted in highest grain yield, but it did not differ significantly from the direct effect. However, the residual effect of P (applied to previous pigeonpea) on wheat proved to be significantly inferior to direct application. Percent recovery of P from applied fertilizer decreased with P level. Total recovery in the complete rotation was higher where P was directly applied to wheat compared to pigeonpea. The P use efficiency was also highest in the treatment comprising direct application to wheat. Pigeonpea could utilize the residual P more efficiently, indicating that fresh application to this crop can be omitted without any reduction in yield.

    Notizen: Abstract This paper presents the findings of a case-study carried out on the Maasai herbalists of Arusha town, whose social and economic situation, both in their villages of origin and in town is being analysed. Special emphasis is laid on the struggle of women herbalists to upgrade their low economic and social status in their home villages and, at the same time, satisfy their most basic needs in town. In this, they depend on their asset of indigenous ethnomedical knowledge, which they commercialise. They do not consider themselves professional experts among their own people, but they become ones when they are in town. A major aim of this paper is also to illustrate the wealth of indigenous knowledge concerning the biodiversity of people's environment. A survey of the medical plants most frequently sold by the herbalists is presented with the intention of identifying those species which are endangered, although the herbalists themselves do not yet perceive the seriousness of the situation.

    Notizen: Abstract A survey of 11 freshwater habitats in the floodplain of the River Nan, northern Thailand was carried out during April and September 1996. The rotifer samples were collected qualitatively from paddy fields, ponds, canals and reservoirs, using a 60 μm mesh net. One hundred and eighteen species were identified, four (Lepadella quinquecostata (Lucks), Macrochaetus danneeli Koste & Shiel, Testudinella ahlstromi Hauer and T greeni Koste) of which are new to Thailand and one (L. quinquecostata) is new to Asia. The numbers of species found in two localities are relatively high, with 86 and 73 rotifer taxa. Most of the species recorded are common, cosmopolitan or pantropical and warm-stenotherms. The occurrence of a species previously considered endemic to Australia, M. danneeli provides more evidence illustrating a relation between the rotifer faunas of southeast Asia and Australia. Comments are presented on some insufficiently known taxa in particular on the new records for Thailand.

    Notizen: Abstract In this paper we review previous studies on sympatric Brachionus populations in Torreblanca Marsh as a model of evolutionary and ecological relationships between closely related species. The marsh is a wetland on the Mediterranean coast of Spain with high spatial and temporal heterogeneity. Allozyme and morphometric analysis showed that Brachionus group plicatilis (formerly, Brachionus plicatilis and currently split into B. plicatilis and B. rotundiformis) was composed of three groups of genotypes with no evidence of gene flow between them (B. plicatilis, B. rotundiformis SM and B. rotundiformis SS). Correlations between seasonal and spatial distributions, on one hand, and temperature and salinity, on the other hand, were consistent with the results of experimental studies on population dynamics. Accordingly, B. plicatilis is a euryhaline, low temperature group; B. rotundiformis SM is adapted to high temperature and low salinity conditions; and B. rotundiformis SS is adapted to high temperature and high salinity conditions. The groups had different mictic responses to density, salinity and temperature, which can be explained to some extent as an adaptive escape response, given their different ecological preferences. These differences imply a partial ecological barrier to male–female encounter between groups. Mating experiments showed that most copulations occurred within a group. B. plicatilis has a mating recognition system different from those of either B. rotundiformis SM or SS, whereas the two B. rotundiformis groups had partially differentiated mating preferences. Cross-mating experiments performed in the laboratory failed to produce any detectable hybrids. We conclude that three sympatric sibling species inhabit Torreblanca Marsh. The remarkable association between genetic differences among clonal groups and their ecological preferences, mixis response and mating behavior is hypothesized to play a role in stabilizing sympatry, and gives insight into the evolution of genetic divergence and speciation in rotifers.

This has included the development of appropriate test methods for the laboratory manufacture and quality assessment of wheat-based food products. Products considered have included white salted and yellow alkaline noodles, instant noodles, steamed bread and dumplings. The research has also been aimed at the wheat quality requirements of these products to assist in defining appropriate quality objectives for Australian wheat breeding programs.
