Chaque pied est contaminé au moment de l'émission du pollen, par une ooplaque d'O. nubilalis Hübn. (ECB) de race européenne E. L'installation, la colonisation et le développement des chenilles sont notés lors de 12 périodes de prélèvements destructifs (4 par température). La vitesse de développement d'O. nubilalis est affectée par la température, main non par l'humidité du sol. Les 4 niveaux d'humidité du sol n'ont aucun effet sur la teneur en eau des tiges de maïs. En fait, les feuilles de maïs présentent une senescence précoce lorsqu'il y a déficit en eau dans le sol. La teneur en eau du sol agit sur l'installation, sur la distribution verticale, la dispersion et le lieu d'alimentation des chenilles; mais ces effets sont légers et ne modifient pas la vitesse de développement. L'environnement larvaire dans la tige de maïs est efficacement isolé des variations externes par l'aptitude de la plante à maintenir la teneur en eau des tiges relativement élevée et stable. Ainsi, des changements importants au niveau du sol n'ont pratiquement pas d'effets sur le développement d'O. nubilalis, malgré les conséquences brutales pour la plante. Cette étude montre que la vitesse de développement d'O. nubilalis est relativement insensible aux modifications de la teneur en eau du sol ainsi qu'aux effets de ce stress de sécheresse sévère sur le pied de maïs. La discussion porte sur l'importance de ces résultats pour la modélisation de la dynamique de l'insecte, la physiologie de la culture et les interactions entre insecte et plante.
    Notes: Abstract Maize plants were grown under four moisture regimes (wet to extreme deficit) and three constant temperatures (20°, 25° & 30°C) in a phytotron. Each plant was infested with one E-race European corn borer [Ostrinia nubilalis (Hubn.)] (ECB) egg mass at pollen shed. ECB development, location, and establishment were recorded over the course of 12 destructive sample dates (4/temperature). ECB developmental rates were not significantly affected by soil moisture treatments, but were significantly affected by temperature. In spite of successful establishment of four distinctly different soil moisture regimes, the maize stalk tissue water levels were not significantly different among soil water treatments. Instead, the maize plants exhibited accelerated leaf senescence in response to the water deficit conditions. Among the soil water treatments, differences were found in larval establishment, vertical distribution and dispersion, and feeding site selection; however, those effects were slight and could not explain the similarity in ECB developmental rates observed in these treatments. In maize, the larval environment within the stalk was effectively insulated from changes in the external environment by the plant's ability to maintain a relatively high and stable stalk tissue water content. Thus, large changes to the soil environment had essentially no effect on ECB development, though drastic consequences for the plant. This study indicates that ECB rates of development are relatively insensitive to changes in the soil water environment as well as the associated changes in the maize plant that accompany severe drought stress. The significance of these findings to insect modelling, crop physiology, and insect-crop interactions is discussed.

    Description / Table of Contents: Résumé L'étude a porté sur les effets d'une température constante (23 °C), de la qualité de l'aliment,-B = aliment naturel, c'est-à-dire de l'écorce de chêne pulvérisée; B+C = aliment naturel enrichi avec de la poudre de biscuits sucrés-, et de la saison sur le développement larvaire de M. funereus L. La température de 23 °C a été défavorable aux larves récoltées en septembre dans des souches de chênes et élevées sur aliment B: les larves étaient mortes en 30 jours. Avec aliment B+C, l'effet défavorable a été neutralisé et le poids des larves augmenté de 543%, 897% et 1179% en 1, 2 et 3 mois. Des larves néonates d'été ou d'hiver se sont bien développées sur B+C, mais toutes les larves d'hiver étaient mortes sur B en 40 j. Des larves de mêmes parents, écloses à différentes époques de l'année, ont présenté des différences saisonnières du taux de survie et de la vitesse de développement lors de leur élevage à 23 °C sur B+C. Le développement le plus rapide et la meilleure survie ont été obtenus avec des larves écloses au début ou au milieu de l'été; tandis que le développement le plus lent et la plus mauvaise survie ont été obtenus avec les larves écloses à la fin de l'été. Les changements saisonniers synchrones des adultes et de leurs descendants, exprimés à différents niveaux d'organisation biologique, suggèrent l'existence d'un rythme annuel endogène qui dépend de l'expression de différents gènes au cours du cycle annuel.
    Notes: Abstract The effects of constant temperature (23 °C), food quality (B, natural diet i.e. powdered oak bark; B+C, natural diet enriched with powdered sweet crackers) and season on larval development of the cerambycid Morimus funereus L. were studied. The temperature of 23 °C exerted an unfavourable effect on larvae collected from oak stumps in September and bred on B; i.e. the larvae died within 30 days. When the larvae consumed B+C the unfavourable effect of temperature was abolished and the larvae increased their weight by 543%, 897% and 1179% in 1, 2 and 3 months, respectively. Newly hatched summerand winter-larvae developed successfully on B+C, while all the winter-larvae died within 40 days on B. Larvae of an identical parentage hatched in different phases of the annual cycle and showed seasonal differences in the rate of development and survival when reared on B+C at 23 °C. The fastest development and the highest survival rate were observed in larvae which emerged in early- and midsummer, whereas the slowest development and the lowest survival rate were in those hatched in late summer. The synchronized seasonal changes of adults and their offspring, as expressed at different levels of biological organization, suggest the existence of an endogenous annual rhythm which is dependent upon the expression of different genes in the course of the annual cycle.

La longévité des adultes mâles et femelles a été déterminée. Les vitesses de développement de la ponte à la formation du cocon et de celleci à l'émergence de E. diversicornis Howard, encyrtide parasite de P. herreni ont été déterminées à 18°, 20°, 25° et 30°C. Il en a été de même pour un second encyrtide parasite A. coccois Smith à 20°, 25° et 30°C. Les équations polynomiales des derniers carrés dérivés et les équations logistiques ont été ajustées pour chaque lot de données (à l'exception de A. coccois) de façon à ce qu'elles aient pu être interpolées pour analyser l'impact de ces parasites sur la dynamique de population de P. herreni. Les résultats de cette analyse sont présentés à part.
    Notes: Abstract Developmental rates for Phenacoccus herreni Cox & Williams were determined at 18°, 20°, 22°, 25°, 30° and 35°C for the egg, all juvenile male and all juvenile female stadia. Longevity was determined for adult females and adult males. Developmental rates for the P. herreni parasitoid Epidinocarsis diversicornis (Howard) were determined at 18°, 20°, 25° and 30°C for the oviposition-to-mummy-formation period and the mummy-formation-to-adult-eclosion period. Developmental rates were determined for the P. herreni parasitoid Acerophagus coccois Smith for the same two life stages at 20°, 25° and 30°C. Least-squares-derived polynomial equations or logistic equations were fitted to each data set (except for A. coccois) so that rates could be interpolated for temperatures between observed points for use in an analysis of the impact of these parasitoids on population dynamics of P. herreni. Results of this analysis are presented separately.

    Description / Table of Contents: Résumé La diapause des oeufs de Zeiraphera canadensis Mutuura et Freeman (Lepidoptera: Tortricidae) a été complétée après 15 semaines d'élevage à 0,5°C. A ce stade de leur développement, une température supérieure à 30°C leur est létale. Les relations entre la température et le développement des larves sur l'épinette blanche, Picea glauca (Moench) Voss, et des pupes ont été observées au laboratoire. Les temps de développement les plus courts ont été observés à 28°C. Les larves sont tolérantes à la chaleur et se développent à 32 C, une température qui est létale aux pupes. II y a peu de variabilité des taux de développement de tous les stades immatures. Ceci explique le haut degré de synchronie observé au sein de populations de cet insecte. Un modèle simulant la phénologie a été mis au point et calibré pour prédire la fréquence des divers stades du cycle vital sous des conditions de terrain. La processus de calibration a révélé que la température dans le microhabitat des oeufs est très semblable à celle de l'air, mais que les larves, retrouvées sous l'enveloppe des bourgeons, sont exposées à des températures pouvant excéder celle-ci de plusieurs degrés. Pour des fins de planification du synchronisme entre le développement de l'insecte et des programmes de suivi et de lutte, le nombre de degré-jours nécessaires pour atteindre 10% d'éclosion des oeufs ou 75% d'émergence des adultes ont été estimés à partir d'une température seuil de 4,4°C.
    Notes: Abstract Diapause requirements of eggs of Zeiraphera canadensis Mutuura and Freeman (Lepidoptera: Tortricidae) are satisfied by 15 weeks of storage at 0.5°C. Temperatures above 30°C are lethal for this life stage. Temperature-dependent development of larvae fed on white spruce, Picea glauca (Moench) Voss, and of pupae was observed under laboratory conditions. The shortest development times were observed at 28°C. Larvae are tolerant to heat, and developed at 32°C, a lethal temperature for pupae. There was little variation of development rates in all stages of the life cycle, explaining the high degree of phenological synchrony reported in populations of this species. A phenological model was developed and calibrated to simulate the relative abundance of the various immature stages under field conditions. The calibration process suggested that temperature in the microhabitat of eggs may be very close to air temperature, but that temperatures under bud caps where larvae are feeding may rise several degrees above air temperature. For the purposes of planning the timing of survey and control practices, degree-day requirements for 10% egg hatch and 75% adult emergence were estimated based on a 4.4°C threshold temperature.

    Notes: Summary Bacteria of the genus Azospirillum are extensively studied for their plant-growth promoting effect following inoculation. Physiological and biochemical studies of these diazotrophic bacteria are now benefiting from recent breakthroughs in the development of genetic tools for Azospirilum. Moreover, the identification and cloning of Azospirillum genes involved in N2 fixation, plant interaction, and phytohormone production have given new life to many research projects on Azospirillum. The finding that Azospirillum genes can complement specific mutations in other intensively studied rhizosphere bacteria like Rhizobia will certainly trigger the exploration of new areas in rhizosphere biology. Therefore a review of the Azospirillum-plant interactions is particularly timely.

    Notes: Summary Damage caused by Rivellia angulata larvae to pigeonpea root nodules at the ICRISAT center in India was greater in the crop grown on Vertisols (up to 86%) compared to that on Alfisols (20%). Attempts to quantify the field effects of nodule damage on growth and yield of pigeonpea in a Vertisol, involving many heavy applications of soil insecticides (aldrin and hexachlorocyclohexane) failed because the insecticides did not control the pest and adversely affected the growth of the pigeonpea and the subsequent crop of sorghum (Sorgorum bicolor L. Moench). The impact of nodule damage on pigeonpea growth, yield and nutrient uptake was successfully studied in greenhouse-grown plants at three N levels. In this pot study, artificial inoculation with Rivellia sp. led to substantial nodule damage (70%). The results of this damage were a significant overall reduction in nodule dry weight (46%), acetylene reduction activity (31%), total leaf area (36%), chlorophyll content of leaves (39%) and shoot dry weight (23%) 68 days after sowing. At maturity, Rivellia sp. infestation caused significant reductions in top dry weight (22%), root and nodule dry weight (27%), seed dry weight (14%), and total N (29%) and P uptake (19%). The problems and prospects of manipulating nodule damage so as to reduce N losses in pigeonpea are discussed.

The possibility of selecting superior N2-fixingFrankia-Ceanothus spp. actinorhizal associations was examined. Greenhouse tests were used to expose various soil-borne microsymbiont andCeanothus sp. population accessions in reciprocal combination. The acetylene reduction rate was used as a measure of N2-fixation capacity. There was no significant interaction between host and microsymbiont regardless of source for all variables measured. The acetylene reduction rate, nodule number and mass, plant biomass, and root: shoot ratio were significantly different among soil sources. The acetylene reduction rate was not significantly different amongCeanothus sp. accessions. Neither was it strongly correlated with other variables. It was concluded that the N2-fixation rate is more a function ofFrankia sp. than the hostCeanothus sp. in actinorhizal associations. It appears possible to select soil sources with superior N2-fixing microsymbiont populations.

    Description / Table of Contents: Zusammenfassung Untersuchungen zur Bioverfügbarkeit essentieller Mikronährstoffe (Vitamine, Mineralstoffe, Spurenelemente) aus Lebensmitteln wie auch aus Arzneimittelzubereitungen sind von entscheidender Bedeutung bei der Beurteilung des jeweiligen Beitrages zur Nährstoffversorgung bzw. zur Abschätzung eines möglichen Therapieerfolges. Die Durchführung derartiger Prüfungen erfolgt in der Praxis nicht selten nach unterschiedlichen Kriterien. Deshalb werden zunächst die Grundsätze erläutert, die bei entsprechenden Prüfungen für eine valide Bewertung von Nähr- bzw. Wirkstoffgemischen oder Monopräparaten bedeutsam sind. Abschließend wird die Ermittlung der Bioverfügbarkeit von Folsäure an einem Beispiel demonstriert. Zur Testung werden die postresorptiven Kurvenintegrale ermittelt, wobei die Daten auf das basale Ausgangsniveau bezogen werden. Zur Beurteilung der Bioverfügbarkeit findet neben dem maximalen Vitaminanstieg im Serum sowie der Zeit bis zum Konzentrationsmaximum insbesondere der Bioverfügbarkeitsquotient von Test- und Referenzzubereitung mit seinem 95%-VB Berücksichtigung. Die Ergebnisse zeigen, daß die exemplarisch eingesetzte Testsubstanz, 5-Formyl-Tetrahydrofolsäure, eine sehr gute Bioverfügbarkeit aufweist und daß unter Anwendung der beschriebenen Rahmenbedingungen eine objektive Bewertung der Bioverfügbarkeit möglich ist.
    Notes: Summary Bioavailability studies of micronutrients (vitamins, minerals) from different food stuffs, as well as from pharmaceutical preparations are of great importance for the calculation of nutrient intake and/or the evaluation of the therapeutic value. In practice, bioavailability studies are not generally performed on a standardized level. For this reason basic criterions are presented and discussed. Finally, the investigation of the bioavailability of folate is demonstrated by an example. The parameters investigated are: the maximal concentration (cmax), the time to reach this maximum (tmax), and especially the bioavailability ratio determined from the area under the curve (AUC) after administration of the folic acid containing test and reference preparation. The results indicate that 5-formyl-tetrahydrofolic acid given in the tested form has nearly the same bioavailability (ratio: 1.02) as folic acid (pteroylmonoglutamic acid) in aqueous solution.

    Notes: Abstract Sr/Ca ratios in modern brachiopod shells reflect variations in ambient seawater, whereas their Na contents show no relationship with water depth or habitat. Their Mn and Fe contents are controlled, in part, by leaching of these elements from oxide coatings or the low input/sedimentation rate of detrital material into depositional areas such as Quatsino Sound. For most Carboniferous brachiopods from North America, the Mn and Fe contents are similar to those recorded by their Recent counterparts. The high Mn and Fe contents in the brachiopods from shales suggest several possibilities for these levels. One possibility is the leaching of Mn and Fe from oxide coatings/matrix which was not completely removed in the cleaning process, or the high levels in part reflect unusual depositional conditions (some degree of anoxia) for the local shaly environments. The Sr/Ca ratio of brachiopods and, by inference, complementary seawater, did not vary significantly during the Carboniferous. The Sr/Ca minimum observed in brachiopods of Mississippian age coincides with a dip in the 87Sr/86Sr curve and correlates with the Hercynian orogeny. This is attributed to the cycling of seawater through mid-ocean ridge basalts, and postulated exchange reactions account for variation in the composition of seawater-Ca. The unidirectional trend of heavier δ13C values from the Devonian to the Permian is intricately coupled with the evolution of the terrestrial biomass. In addition to expansion of terrestrial plants, burial of reduced carbon in the form of coal (organic matter) contributed to the observed shift. The start of the Permo-Pennsylvanian glaciation is marked by a negative excursion of the secular carbon trend, which is linked to weathering of reduced carbon and its return to the oceanic reservoir with its oxidized carbon. The oxygen isotope values reflect the unidirectional trend towards higher values of the carbon data with decreasing geologic age. Negative excursions of the trend may be related to extensive weathering of terrestrial and submarine rocks, whereas positive excursions may be related to hydrothermal alteration of submarine rocks and dehydration of oceanic crust during times of active sea-floor spreading. Oxygen-calculated water temperatures of unaltered brachiopod material are unrealistically high for all of the Devonian, and the Chesterian-Meramecian, Desmoinesian-Missourian, and Artinskian Epochs. During these times maximum water temperatures of 42° to 56°C are well above the thermal threshold of protein denaturation. This process, which is lethal to most higher organisms, demands an adjustment in oxygen of -2.5%. for samples older than Missourian, and of -1.250%. for samples spanning the Missourian-Artinskian interval. With these adjustments and salinity considerations made prior to calculations, water temperatures become reasonable for the Late Paleozoic epeiric, tropical seas of North America.

Hoffm., a nitrogen-fixing epiphyte common in mesic temperate forests, were collected in a Douglas-fir (Pseudotsuga menziesii Franco) forest near Corvallis, Oregon, and maintained for 20 to 40 days in controlled-environment chambers with atmospheric CO2 concentrations of 374 and 700 μll-1. Nitrogenase activity, which was assayed by the acetylene reduction method, was approximately doubled in the lichen maintained in elevated CO2. Increases in nitrogen fixation by lichens may be an important part of the integrated ecosystem response to rising CO2.

    Notes: Summary Prednisolone absorption and bioavailability of 10 mg enteric-coated (EC) and plain (uncoated) tablets were investigated after fasting and heavy meals (EC only) consumed to satiety in normal healthy volunteers. The same volunteers had also received 16 mg of prednisolone intravenously. In fasted subjects, the absolute bioavailability fraction, as normalised for intravenous doses, of prednisolone from plain tablets was 1.055 and from EC tablets was 0.996. The peak concentrations after plain and EC tablets were 309 and 249 ng/ml attained at 0.98 and 5.14 h, respectively. The means plasma elimination half-lives following the plain, EC tablets and intravenous administration in fasting conditions were 3.73, 3.89 and 3.78 h, respectively. Food interfered with both the absorption and the pharmacokinetics of prednisolone after EC tablets resulting in variability in its plasma levels. In some cases absorption of prednisolone was delayed for 12 h and remained at a measurable level for 24 h. In other cases, a normal absorption pattern was observed. This inter- and intrasubject variability of the effect of food appears to be related to its quantity, constituents and also the subjects physiological characteristics. It is concluded that enteric-coated prednisolone tablets should be administered at least 2 h between meals. However, for more predictable corticosteroid absorption (perhaps thus avoiding the therapeutic failure), plain prednisolone tablets are preferable.

    Notes: Summary The pharmacokinetics and clinical efficacy of a once-daily sustained-release formulation of naproxen (sodium salt) have been compared with those of conventional-release agents. In a single dose pharmacokinetic study, the rate of absorption of the sustained-release preparation was less than that of a conventional-release preparation but the extent of absorption was the same. As is the case with conventional-release naproxen, food decreased the rate but not the extent of absorption of the sustained-release formulation. On multiple dose administration for 7 days, the AUC and average concentrations of the sustained release preparation (1 g daily) were the same as those for conventional release preparations of naproxen sodium (250 mg four times daily) and naproxen free acid (500 mg daily). The conventional-release sodium salt was absorbed more quickly with no differences in bioavailability. A double-blind clinical comparison in patients with osteoarthritis showed the sustained-release preparation (1 g daily) to be equivalent in efficacy to conventional naproxen capsules (500 mg twice daily) but to have a significantly lower incidence of gastrointestinal side-effects. The results suggest that sustained-release naproxen sodium has potential for use as a once-daily treatment for inflammatory disease.

    Notes: Summary The pharmacokinetics of the antiarrhythmic drug diprafenone have been investigated in 6 healthy volunteers following single intravenous (50 mg) and oral doses (50 and 150 mg). Diprafenone was mainly eliminated by metabolism in the liver. Following i.v. infusion of 50 mg diprafenone, the terminal half-life of elimination was 1.50 h, the volume of distribution at steady-state was 1.23 l·kg−1, and the free fraction in plasma was 1.68%. Mean total plasma clearance was 741 ml·min−1·70 kg−1, which approaches normal liver blood flow after correction for the blood/plasma concentration ratio. Thus, diprafenone can be classified as a high extraction drug. Following oral administration, a dose-dependent increase in bioavailability from 10.9 (50 mg dose) to 32.5% (150 mg dose) was observed. The data suggest that diprafenone is subject to saturable hepatic first-pass metabolism.

    Notes: Summary A cross-over study of kinetics has been undertaken in 12 healthy adults volunteers using two sustained-release theophylline products that allow once a day dosing (Theo-Dur tablets and Dilatrane A.P. bead filled capsules) to compare the i.v. pharmacokinetic profiles when taken with an hyperlipidic meal and a balanced standard meal. Each subject took part in four phases in randomised order, corresponding to all possible combinations of the products and the types of meal. Each phase involved a single dose of 9 to 11 mg·kg−1 theophylline administered at 20.00 h, at the beginning of the meal, with 100 ml water. The two formulations were found to be bioequivalent with both types of meal. Taken with a balanced meal, the mean parameters were similar; for Theo-Dur and Dilatrane A.P. they were respectively: Cmax: 11.32 mg·l−1 which plateaued from 8 to 10 h after dosing and 10.9 mg·l−1, which plateaued after 6 to 10 h; AUC 230 mg·h·l−1 and 220 mg·h·l−1; and MRT 18.2 h and 17.7 h. After the hyperlipidic meal the values for Theo-Dur and Dilatrane A.P. respectively, were: Cmax 10.9 mg·l−1 at 12 h and 11.3 mg·l−1 at 10 h; AUC 237 mg·h·l−1 and 227 mg·h·l−1; and MRT 19.2 h and 18.9 h. In spite of a decrease in the absorption rate, which led to a shift to the right of about 2 h of the plasma concentration-time curve, the bioavailability of both formulations were not significantly modified by a hyperlipidic meal as compared to a balanced meal. The shift of the curve with fatty food was not clinically important, as there was no dumping effect. The main difference between the two formulations was seen during the absorption phase, which was linear and less variable with Dilatrane A.P. and sigmoidal with Theo-Dur. This was observed with both types of meal.

    Notes: Abstract For Azospirillum brasilense Sp7, the energy transformation efficiencies were measured in anaerobic respirations with either nitrate, nitrite or nitrous oxide as respiratory electron acceptors by determining the maximal molar growth yields and the H+-translocations using the oxidant pulse method. In continuous cultures grown with malate limiting, the maximal molar growth yields (Y s max -values) were essentially the same with O2 or N2O but were 1/3 and 2/3 lower with NO 2 - or NO 3 - , respectively, as respiratory electron acceptors. Both the maximal molar growth yields and the maintenance energy coefficients were surprisingly high when Azospirillum was grown with nitrite as the sole electron acceptor and source for N-assimilation. Growth under N2-fixing conditions drastically reduced the Y s max -values in the N2O and O2-respiring cells. In the H+-translocation measurements, the $$\vec H^ + $$ /oxidant ratios were 5.6 for O2→H2O, 2.5–2.8 for NO 3 - →NO 2 - , 2.2 for NO 2 - →N2O and 3.1 for N2O→N2 respirations when the cells were preincubated with valinomycin and K+. All the values were enhanced when the experiments were performed with valinomycin plus methyltriphenylphosphonium (=TPMP+) cation. The uncoupler carbonyl cyanide-m-chlorophenyl-hydrazone diminished the H+-excretion indicating that this translocation was due to vectorial flow across the membrane. In the absence of any ionophore, nitrate and nitrite respirations were accompanied by a H+-uptake $$(NO_3^ - \to N_2 = - 2.9 \vec H^ + /NO_3^ - and NO_2^ - \to N_2 = - 2.5 \vec H^ + /NO_2^ - )$$ . Any significant H+-translocation could not be detected in N2O- and O2-respirations under these conditions. It is concluded that nitrate reduction proceeds inside the cytoplasmic membrane, whereas nitrite is reduced extramembraneously. The data are not conclusive for the location of nitrous oxide reductase. The maximal molar growth yield determinations and the absence of any H+-uptake in untreated cells indicate a cytoplasmic orientation of the enzyme similar to the terminal cytochrome oxidase of respiration. The low H+-extrusion values for N2O-respiration compared to O2-respiration in cells treated with valinomycin plus TPMP+ are, however, not in accord with such an interpretation.

    Notes: Abstract The distribution of sex pheromone induced aggregation substance was studied on the cell surface of various Enterococcus faecalis strains. In the accompanying paper we have shown that the aggregation substance appears as a layer of hairlike structures. Using direct and indirect immunogold technique, transmission electron microscopy and high resolution scanning electron microscopy we investigated the appearance and distribution of the aggregation substance. The “hairs” increase in number with increasing exposure to sex pheromones (maximum density: 1300/μm2). We show that these structures are unequally distributed over the cell surface, even if the cells were induced by sex pheromones for a long period of time. Statistical analysis of the unequal distribution indicates that aggregation substance is incorporated into pre-existing “old” cell-walls and that this incorporation shows a saturation ca. 40 min after addition of sex pheromones.

    Notes: Abstract Random and site-directed Tn5-induced mutagenesis of Bradyrhizobium japonicum yielded two mutations, one in strain 2960 and the other in strain 2606::Tn5-20, which mapped close to each other but in separate genes. The corresponding wild-type genes were cloned, and their approximate location on the cloned DNA was determined. Mutant 2960 was Fix- and formed green nodules on soybean, whereas strain 2606::Tn5-20 had ca. 4% of wild-type Fix activity and formed white nodules. Cytochrome oxidase assays (Nadi tests) showed a negative reaction with both mutants, indicating a functional deficiency of cytochrome c or its terminal oxidase or both. However, the mutants grew well under aerobic conditions on minimal media with different carbon sources. Furthermore, mutant 2960 had a reduced activity in hydrogen uptake, was unable to grow anaerobically with nitrate as the terminal electron acceptor and 2960-infected soybean nodules contained little, if any, functional leghemoglobin. Southern blot analysis showed that a B. japonicum heme biosynthesis mutant [strain LO505: O'Brian MR, Kirshbom PM, Maier RJ (1987) Proc Natl Acad Sci USA 84: 8390–8393] had its mutation close to the Tn5 insertion site of our mutant 2606::Tn5-20. This finding, combined with the observed phenotypes, suggested that the genes affected in mutants 2960 and 2606::Tn5-20 were involved in some steps of heme biosynthesis thus explaining the pleiotropic respiratory deficiencies of the mutants. Similar to strain LO505, the mutant 2606::Tn5-20 (but not 2960) was defective in the activity of protoporphyrinogen IX oxidase which catalyzes the penultimate step in the heme biosynthesis pathway. This suggests that one of the two cloned genes may code for this enzyme.

    Notes: Abstract A number of in-frame deletions have been constructed in the Klebsiella pneumoniae regulatory gene nifL. The effects of each nifL mutation on NifA-mediated expression from the nifH promoter of K. pneumoniae have then been assessed with respect to both nitrogen and oxygen control. These experiments indicate that, in contrast to the situation with the homologous regulatory proteins NtrB and NtrC, NifA activity is not impaired in the absence of NifL. We conclude that the only function of NifL is to inactivate NifA in response to an increase in the nitrogen or oxygen status of the cell.

    Notes: Abstract The two isoenzymes of NADH-dependent glutamate synthase (NADH-GOGAT; EC 1.4.1.14), previously identified in root nodules of Phaseolus vulgaris L., have both been shown to be located in root-nodule plastids. The nodule specific NADH-GOGAT II accounts for the majority of the activity in root nodules, and is present almost exclusively in the central tissue of the nodule. However about 20% of NADH-GOGAT I activity is present in the nodule cortex, at about the same specific activity as this isoenzyme is found in the central tissue. Glutamine synthetase (GS; EC 6.3.1.2) occurs predominantly as the γ polypeptide in the central tissue, whereas in the cortex, the enzyme is represented mainly by the β polypeptide. Over 90% of both GS and NADH-GOGAT activities are located in the central tissue of the nodule and GS activity exceeds NADH-GOGAT activity by about twofold in this region. Using the above information, a model for the subcellular location and stoichiometry of nitrogen metabolism in the central tissue of P. vulgaris root nodules is presented.

The time constant of the on-response atE=4 mV,τ on, was strongly temperature dependent, whereas the amount of displaced charge atE=39 mV, Qon, was only slightly temperature dependent. The mean Q10 forτ on -1 was 2.54, the mean Q10 for Qon was 1.07. The time constant of charge immobilization,τ i , atE=4 mV varied significantly (α=0.001) with temperature. The mean Q10 forτ i -1 was 2.71±0.38. The time constants of immobilization of gating charge and of fast inactivation of Na permeability were similar in the temperature range from 6 to 22°C. The Qoff/Qon ratio forE=4 mV pulses of 0.5 msec duration decreased with increasing temperature. The temperature dependence of the time constant of the off-response could not be described by a single Q10 value, since the Q10 depended on the duration of the test pulse. Increasing temperature shifted Qon (E) curves to more negative potentials by 0.51 mVK −1, but shiftedP Na (E) curves andh ∞ (E) curves to more positive potentials by 0.43 and 0.57 mV K−1, respectively.h ∞ (E=−70 mV) increased monotonously with increasing temperature. The present data indicate that considerable entropy changes may occur when the Na channel molecule passes from closed through open to inactivated states.

    Notes: Sumary The objective of this work was to know the behaviour and variability of Rhizobium leguminosarum after irradiation. The induced variation was tested under greenhouse conditions on the variety JV 3 of broad beans (Vicia faba) in six replications. Induced genetic variabilty was observed for strain, parent and mutant versus parent. Out of 24 irradiated strains, strain 93-32 performed better with a greater number of nodules and higher dry weight of nodules per plant and biological yield. Environment played an important role in the expression of characters observed. High heritability and genetic advance of these traits indicated that the nitrogen fixation ability of Rhizobium can easily be improved by selection.

    Notes: Summary The spontaneous dwarf rat is a novel experimental model animal on the study of pituitary dwarfism. The fine structure of the anterior pituitary cells was studied in the immature and mature dwarf rats. Pituitary glands were removed from 5-, 10-, 20-day-old immature dwarfs, adult (45 days-16 weeks) dwarfs and normal 3-month-old rats and processed for electron-microscopic observation. In the control animals, growth hormone cells were readily identified by their ultrastructural characteristics, such as the presence of numerous electron-dense secretory granules, 300–350 nm in diameter, well developed rough endoplasmic reticulum and a prominent Golgi complex. In contrast, growth hormone cells were not found in the anterior pituitary gland of the spontaneous dwarf rat at any age examined. Other pituitary cell types, i.e., luteinizing hormone/ follicle stimulating hormone, thyroid stimulating hormone, adrenocorticotropic hormone and prolactin cells, appeared similar in their fine structure to those found in the control rats. In the pituitary gland of dwarf rats, a number of polygonal cells were observed either with no or relatively few secretory granules. The rough endoplasmic reticulum was arranged in parallel cisternae and the Golgi complex was generally prominent in these cells. In addition, many were found to have abundant lysosomes. A few minute secretory granules were occasionally observed; however, the immunogold technique failed to localize growth hormone or prolactin in the granules. The nature of these cells remained obscure in this study. Since their incidence and fine structural features, other than the secretory granules, were quite similar to those of the growth hormone cells in normal rats, we postulate that these cells are dysfunctional growth hormone cells. These results suggest that the cause of the growth impairment in the spontaneous dwarf rat is due to a defect in the functional growth hormone cells in the pituitary gland, and since other pituitary cell types appeared normal, the disorder seems to be analogous to the isolated growth hormone deficiency in the human.

    Notes: Summary There is increasing evidence that, in the rat, a serotonin-mediated neural input may have an inhibitory influence on the secretory activity of the subcommissural organ (SCO). In the present investigation the rat SCO was studied 7, 30 and 90 days after transplantation under the kidney capsule, an area devoid of local serotonin-containing nerves. The grafted tissue was examined by use of immunocytochemistry employing a series of primary antisera, lectin histochemistry and transmission electron microscopy. The grafted SCO survived transplantation and contained, in addition to secretory ependymal and hypendymal SCO-cells, also elements immunoreactive with antisera against glial fibrillary acidic protein or S-100 protein. In transplants, SCO-cells produced a material displaying the characteristic immunocytochemical and lectin-binding properties of SCO-cells observed under in-situ conditions. The ependymal cells lined 1–3 small cavities, which contained secretory material. A fully developed structural equivalent of Reissner's fiber was, however, never found. The immunocytochemical and ultrastructural study of the grafted SCO showed an absence of nerve fibers within the graft and suggested a state of enhanced secretory activity. A network of protruding basal lamina structures connected the secretory cells to the newly formed capillaries revascularizing the SCO. One week after transplantation, long-spacing collagen started to appear in expanded areas of such laminar networks and also in the perivascular space. It is suggested (i) that the formation of long-spacing forms of collagen is triggered by factors provided by the SCO-secretory cells, and (ii) that secretory material of the ependymal and hypendymal cells may reach the reticular extensions of the basal lamina. In contrast to the SCO in situ, the grafted SCO-cells showed a positive immunoreaction for neuron-specific enolase. They became surrounded by a S-100-immunoreactive glial sheath that separated them from other transplanted cell types and the adjacent kidney tissue of the host.

    Notes: Summary It is generally accepted that variations in membrane cholesterol content affect the fluidity of the bilayer, thus altering its permeability. In the biological membranes, in physiological conditions, a high cholesterol content rigidifies the bilayer decreasing its permeability, a lower cholesterol content induces the opposite effect by increasing the permeability. Since differences in the epithelial permeability for short chain fatty acids have previously been demonstrated in the proximal and distal colon of the guinea pig, these two regions were investigated to establish whether differences in membrane cholesterol content of the absorbing cells can be demonstrated. Freeze-fracture replicas of filipin-treated colonic tissue were used. The results show that in the proximal colon the density of filipin cholesterol complexes located on the luminal plasma membrane of the columnar absorbing cells was significantly higher (about twice) than in the distal colon. Therefore the lower amount of cholesterol present in the membrane of the absorbing cells in the distal colon indicates a greater fluidity of the membranes of the epithelial cells in this region. Such fluidity could be correlated to the higher absorption rates of shortchain fatty acids characteristic of this region.

    Notes: Summary An ultrastructural morphometric study of the endocrine cells of the oxyntic mucosa of the stomach in gastric biopsies collected from five male and five female healthy volunteers aged 19–31 was performed. No sex-related differences were disclosed. Endocrine cells accounted for 1.2±0.4% of the epithelial volume and 0.9±0.4% of the mucosal volume, i.e., including the lamina propria. After classification of the specific endocrine cell types according to the ultrastructural morphology of secretory granules, the volume densities of ECL, P and D cells (30±9%, 24±7%, and 22±4% of the entire endocrine cell mass, respectively) were higher than those of other endocrine cell types. In particular, EC cells contributed less than 10% and X cells represented a very low proportion of the total cells. Non-granulated profiles of cells which in all other respects appeared to be endocrine were also found with a volume density of 8±4%. D cells were distinguished by the high fraction of cytoplasm occupied by secretory granules (31±5%). Subdivision of the whole mucosa into four horizontal segments revealed the endocrine cells to be mostly distributed in the three lower, with virtually no endocrine cells in the superficial segment. The quantitative ultrastructural analysis of the endocrine cell population of the normal human oxyntic mucosa provided by this study may allow a better evaluation of physiological and pharmacological variations of the endocrine cell population.

    Notes: Summary The cavernous body of green monkeys contains many unmyelinated and few myelinated axons. The unmyelinated axons form terminals in the adventitia of the arteries, between trabecular muscle cells, in the interstitium, and close to endothelium cells of the sinuses. All terminals displayed predominantly “small clear vesicles” and very few “large granular vesicles”; “small granular vesicles” were not seen. However, in rabbit penises, terminals with many large granular vesicles are prominent. Immunohistochemistry (PAP technique) showed a dense network of VIP- and NPY-reactive fibres around the arteries and around trabecular muscles. The density of nerve fibres was particularly high around the subendothelial cushions of the helicine arteries. Double staining for NPY and VIP revealed that both peptides were colocalized. Immunocytochemistry (preembedding PAP technique) showed VIP- and NPY-reactivity in terminals with small clear vesicles; the reaction product was bound to the cytoplasmic face of different membrane types. Although the intracellular localization of the reaction product is probably due to artefactual displacement during preparation, the uniformity of the terminals questions the view that large and small granular vesicles in all species characterize peptidergic and noradrenergic terminals, respectively. The essential findings can be summarized as (1) a high degree of uniformity of nerve terminals, (2) colocalization of VIP and NPY, (3) heavy innervation of the subendothelial cushions of the helicine arteries, and (4) possible innervation of endothelial cells.

    Notes: Summary N-acetylglucosaminidase (NAG), acid phosphatase (ACP) and alkaline phosphatase (AKP) were localised histochemically in fixed cells from the 37-day-old rat epididymis grown in static monolayer culture for 2–8 days. ACP and NAG were cytosolic enzymes found in perinuclear positions, whereas staining of AKP was consistent with a membranous position. These enzymes were also examined in frozen tissue sections of the epididymis, from rats of the equivalent age, where NAG had intense activity in both supra- and infra-nuclear cytoplasm and ACP was more active apically. For the first time AKP was localised along basolateral membranes of the epithelium and in the lumen of the mid-caput region. The monolayer in culture was of principal cells only and they maintained their polarity and ultrastructural characteristics, but the height of the cells was reduced compared to that obtained in situ.

    Notes: Summary Granulated epithelial cells at the vascular pole of the renal corpuscle, peripolar cells, have been found in the kidneys of five species of elasmobranchs, the little skate (Raja erinaced), the smooth dogfish shark (Mustelus canis), the Atlantic sharpnose shark (Rhizoprionodon terraenovae), the scalloped hammerhead shark (Sphryna lewini), and the cow-nosed ray (Rhinoptera bonasus). In a sixth elasmobranch, the spiny dogfish shark (Squalus acanthias), the peripolar cells could not be identified among numerous other granulated epithelial cells. The peripolar cells are located at the transition between the parietal epithelium of Bowman's capsule and the visceral epithelium (podocytes) of the glomerulus, thus forming a cuff-like arrangement surrounding the hilar vessels of the renal corpuscle. These cells may have granules and/or vacuoles. Electron microscopy shows that the granules are membrane-bounded, and contain either a homogeneous material or a paracrystalline structure with a repeating period of about 18 nm. The vacuoles are electron lucent or may contain remnants of a granule. These epithelial cells lie close to the granulated cells of the glomerular afferent arteriole. They correspond to the granular peripolar cells of the mammalian, avian and amphibian kidney. The present study is the first reported occurrence of peripolar cells in a marine organism or in either bony or cartilagenous fish.

    Notes: Summary In the hamster, the weight of the adrenal glands increases during the course of gestation, with the highest value at day 5. In comparison to non-pregnant control animals, there were no changes in the volume of the zona glomerulosa (ZG) and zona fasciculata (ZF), while the volume of the zona reticularis (ZR) increased notably. The average volume of ZG-cells rose at day 5 of pregnancy and thereafter gradually decreased to that of control hamsters. A marked drop in the volume of ZF-cells was seen at days 5 and 10 of pregnancy, whereas at day 15 the cells were larger than in controls. At day 5 of pregnancy, a conspicuous increase in the cell volume was found in ZR, followed by lower values at day 10 and again higher than in control hamsters at day 15. The total number of parenchymal cells in hamster adrenal cortex increased at day 10 of gestation, then underwent a marked decrease, reaching the control value at the final day of pregnancy; this drop was mainly due to a reduction in the number of ZF-cells. The changes in the cell volume were paralleled by rather proportional changes in the volume of the mitochondrial compartment and in the quantity of smooth endoplasmic reticulum. The volume of the lipid-droplet compartment significantly rose in the course of gestation in both ZF- and ZR-cells. The cortisol output by adrenal homogenates gradually decreased during pregnancy.

    Notes: Summary Two types of mammosomatotropes (MS), the small-granule and vesicle-granule MS, were detected in mouse adenohypophysis by electron microscopy and immunohistochemistry. Both cell-types were immunoreactive to prolactin (PRL) and growth hormone (GH) antisera. The small-granule MS contained small, round, solid secretory granules about 100 nm in diameter, and were smaller than the classical GH and PRL cell-types. The vesicle-granule MS contained secretory granules like cored vesicles, and were larger than classical GH and PRL cells. Small-granule MS were immunoreactive to both PRL and GH antisera in the same region of the cell cytoplasm; the vesicle-granule MS, however, were immunoreactive to only PRL antiserum in most cytoplasmic areas, and a positive response to both PRL and GH antisera was confined to only certain small areas.

    Notes: Summary The structure and organization of paired lymphoid tissue in the nasal mucosa, situated in the transitional zone on both sides of the septal opening to the pharyngeal duct, of conventionally-housed rats was examined by light microscopy and scanning and transmission electron microscopy. Each lymphoid structure consisted of follicles containing T- and B-cell areas, and was covered with specialized epithelium. This epithelium consisted of cuboidal ciliated cells with oval nuclei parallel to the basal lamina. Goblet cells were sparse. Occasionally, islands of microvilli-bearing cells (so called membraneous or M cells) covered the lymphoid structures. M Cells were also found as single cells among the ciliated cells. The morphological characteristics and the particular localization justify the conclusion that the nasal lymphoid tissue described belongs to the mucosa-associated lymphoid tissue. It is therefore suggested that this nasal structure be designated nasal lymphoid tissue.

    Notes: Summary Neurointermediate lobes from adult or 10-dayold rats were implanted by a stereotaxic procedure into the third ventricle of adult male rats, in an area close to the paraventricular nucleus. They were examined, using immunocytochemical and ultrastructural techniques, at times ranging from 1 week to 8 months. All grafts were recovered in a healthy condition although some rejection of the tissue was detected at the 1and 2-week stages. In the neural lobe, clusters of pituicytes were scattered among the loose network of capillaries, most of which had a fenestrated endothelium. The intermediate lobe remained organized in compact avascular lobules. Axons similar to those projecting into the neurointermediate lobe in situ, but also axons of other types (e.g., somatostatinergic, enkephalinergic) penetrated the grafts. Synapses with melanotrophic cells in the intermediate lobe and neurohaemal contacts in the neural lobe were frequent from 2 1/2 months after transplantation. Immunocytochemical and ultrastructural characteristics indicated intense secretory stimulation of the melanotrophic cells in the early stages. All cells enclosed in a same glandular lobule reacted in a similar manner. In later stages, when re-innervation occurred, the cells recovered their initial characteristics. The overall effect of the re-innervation of the intermediate lobe grafted in this location is inhibitory, as in the lobe in situ.

    Notes: Summary The ultrastructure of rat glomerular epithelial cells (podocytes) in kidney slices in vitro was examined using qualitative and quantitative electron microscopy. The kidney slices were cultured in Medium 199 with Hanks' salts in a 5% CO2/95% O2 environment for up to 14 days. Few changes in podocyte ultrastructure occurred in the first 12 h of culture, but by 24 h cell bodies were rounded, microvilli were present on all podocyte surfaces, and some foot processes had been replaced by flattened expanses of cytoplasm. These changes were more pronounced by 3 days, when some podocytes had developed pseudopodal extensions and appeared to be migrating from glomeruli onto the slice surface. Podocytes could still be identified after 8, 10 and 14 days of culture, although relatively few glomeruli remained at 14 days. Morphometric methods were used to analyse podocyte shape, volume and surface area during the first 4 days of culture. The most significant change involved loss of foot processes: the number of filtration slits per 100 μm of basement membrane decreased from 211.8 ± 15.0 (mean ± SD) at the commencement of culture, to 55.3 ± 22.6 after 2 days (P 〈 0.001). These data provide baseline information for in vitro studies on the effects of nephrotoxins on podocytes.

    Notes: Abstract Paleobotanical studies indicate that several isolated and systematically depauperate groups of extant woody dicotyledons originated in the Mid Cretaceous. TheChloranthaceae had probably differentiated into insect-pollinated (Chloranthus andSarcandra) and wind-pollinated (Ascarina andHedyosmum) forms by the end of the Albian, and leaves referable to theTrochodendrales are known from the Albian and Cenomanian. In the latest Cretaceous and Early Tertiary, extinct representatives of theTrochodendrales includedNordenskioldia and theJoffrea-Nyssidium complex. ThePlatanaceae also differentiated before the end of the Albian and initially had insect-pollinated, unisexual flowers with five carpels or stamens. Some of these features persisted in the platanoid lineage until the Early Tertiary, and during the Paleocene and Eocene thePlatanaceae included forms with elliptical, palmate and pinnate foliage. The history of thePlatanaceae suggests that several features of the reproductive morphology of extant taxa may have arisen in association with a trend toward wind pollination. In the Mid Cretaceous, platanoid foliage partially intergrades with pinnateSapindopsis and pedateDebeya-Dewalquea leaves suggesting a close relationship betweenPlatanaceae andRosidae andFagaceae respectively. TheChloranthaceae, Trochodendrales, andPlatanaceae all occupy a somewhat intermediate position between theMagnoliidae andHamamelidae and are of considerable interest with respect to their role in the initial radiation of nonmagnoliid (“higher”) dicotyledons.

    Notes: Abstract New investigations on the flower and fruit structure of extantHamamelidaceae and other LowerHamamelididae together with new finds of fossil flowers and seeds from the Upper and Lower Cretaceous provide the outline of an increasingly more differentiated picture of the early evolution of the subclass. Three patterns of valvate anther dehiscence are recognized in the subfamilyHamamelidoideae (and the subclassHamamelididae). The basic (plesiomorphic) type within theHamamelididae has 2 valves per theca. The type with 1 valve but 2 pollen sacs per theca is both consistent and exclusive for the 5 southern genera of theHamamelidaceae. They seem to be the remnants of a homogeneous group that originated before the Upper Cretaceous. This is supported by fossil hamamelidaceous flowers from the Upper Cretaceous that have thecae with 1 valve. Since several-seededHamamelidaceae predate one-seeded forms in the fossil seed record (in Europe) and the systematic structure of the one-seeded group is relatively more homogeneous, several-seeded groups are considered to be more ancient. Several parallel evolutionary trends are recognized within theHamamelidaceae as well as within the LowerHamamelididae: anther dehiscence with 2 valves per theca → 1 slit or 1 valve; pollen sacs per theca 2 → 1; pollen tricolpate → polyforate; exine coarsely reticulate → finely reticulate; loss of perianth (tepals or petals and sepals) and concomitant loss of fixed number of floral organs; differentiation of exposed nectaries.

    Notes: Abstract The major radiation of theJuglandaceae occurred during the early Tertiary as recorded by the proliferation of juglandaceous pollen and the appearance of fruits representing extinct and extant genera of the family. Juglandaceous pollen types of the Paleocene were predominantly triporate and exhibited a greater diversity in patterns of exinous thinning than occurs in the family today. Analyses of in situ pollen from early Tertiary juglandaceous inflorescences confirms the taxonomic value of certain patterns of exinous thinning. Data from co-occurring fruits and pollen indicate that relatively unspecialized, isopolar triporate pollen of the type presently confined to the tribeEngelhardieae also occurred in other tribes of the family during the Paleocene. Pollination has been mostly anemophilous throughout the Tertiary. Both wind and animal fruit-dispersal syndromes were established early in the radiation of the family but a greater diversity of wind-dispersed genera has prevailed.

    Notes: Abstract In theEudesmieae B eucalypts and inEucalyptus caesia, the perianth of the mature flower consists of a single, anatomically continuous, opercular structure that is crowned by the tips of the original free and separate calycine and corolline whorls. Ontogenetic and comparative evidence supports the hypothesis that this operculum is mostly corolline in composition, and that the calycine parts have been elevated distally onto the dorsal surface. In theEudesmieae B eucalypts this condition appears to be due to precocious initiation of the corolline primordia, followed by expansion and continuity of their growth centres which incorporates the areas at or below the base of the still differentiating calycine whorl. InEucalyptus caesia the corolline primordia are not precocious, but a similar situation is effected by a seemingly retarded increase in receptacle diameter relative to lateral expansion of the corolline growth centre(s). In these two examples the same final perianth form apparently derives from two different sets of growth processes which, nonetheless, result in the same allometric relationships within the developing flower. Although identical at maturity, the operculum form in these two taxa is best described in terms of convergence, rather than homology.

    Notes: Abstract Eucharis candida andE. formosa are two often sympatric species of bulbous geophytes restricted to neotropical rain forest understory. The species are most common in eastern Ecuador, and are the only two east Ecuadorean species of the genus found north of the Pastaza valley. Data from phenetic, karyotypic, and preliminary isozyme electrophoretic analyses of both species are represented. The species are distinguishable phenetically and karyologically, but isozyme-based relationships are more complex. Phenetic resolution of the isozyme phenotypes supports recognition of two species in Ecuador. A Peruvian isolate ofE. formosa, though not morphologically distinct, shows both allozyme and chromosomal divergence from Ecuadorean populations. Cladistic relationships based on overall allozyme data do not support species distinction, but a novel electrophoretic phenotype for glutathione reductase is shared only by individuals ofE. candida. An apparent geographic component within the monophyletic groups resolved in the cladogram suggests that some degree of gene flow between these two species has been maintained without the complete loss of morpholgoical species identity. This may have been mediated either by artificial population structures due to a probable long history of cultivation, or via Pleistocene refugia effects. Both species may have originated in eastern Ecuador from a common ancestral population which has since radiated outward, perhaps several times.

    Notes: Abstract The influence of different pollen donors on seed formation was investigated in three populations ofEspeletia schultzii that differ in environmental conditions and life history characteristics. Self pollen and pollen from different donors (〈 15m apart) within each population was used in a diallel design in order to test the genetic base of seed set variation. Three measures of seed formation were used: (1) achene number; (2) proportion of filled achenes (fruits) that distinguishes between achenes with seeds and empty achenes; (3) proportion of aborted seeds that distinguishes between viable and aborted seeds. Self-pollinations resulted in empty achenes. Achene number did not vary between the different pollen donors. A bimodal pattern of filled achenes was found in two populations in two consecutive years. On the other hand, a unimodal pattern was found in crosses between more distant donors (〉 30m). These patterns seems to be the results of a sporophytic incompatibility system. Seed abortion was highest at the higher elevations and seems to be correlated with elevation rather than with any genetic effect.

    Notes: Abstract The genetic variability of five natural populations ofNajas marina L., i.e. one diploid of subsp.marina (Europe), two of subsp.intermedia (Europe) and both a diploid (C. Africa) and a tetraploid (Middle East) of subsp.armata, has been estimated by means of electrophoretic studies. These populations differ in their morphology and karyotype. Emphasis is placed on the characteristics and status of a tetraploid cytotype from Merkaz Sappir (Israel). Almost all the variation observed is expressed in seed alcohol dehydrogenase (ADH). The differences are in a unique allele of theAdh-2 locus and in the formation of novel heteromeric isozymes.Adh genes in seeds can be used as a marker for the autotetraploid character. The other enzyme systems tested failed in this respect. The genetic variability based on 23 loci is rather low. Nevertheless, the autotetraploid population has a higher or equal ratio of polymorphic loci than the related diploids. Cluster analysis illustrated not only thatNajas marina subsp.marina has diverged much from subsp.intermedia and subsp.armata, but also showed the difference between the latter two taxa, as well as the intermediate position of the autotetraploid population.

    Notes: Abstract Leporella fimbriata is a self compatible orchid of southern Australia. It is dependant across its range on unique pollination by sexually attracted male winged antsMyrmecia urens, which pseudocopulate with the flower. Typical pollination sequences began with an initial circling then zig-zag flight to the flower. Vectors usually alighted on the inflorescence stem and quickly crawled to the flower where they adopted a copulatory position sideways along the wide labellum, pseudocopulatory probing immediately followed. In this position pollen carried on the thorax was deposited on the stigma. Departure from the labellum usually resulted in pollinium removal. Pollinator movements were restricted and the distribution leptokurtic with a mean of 3.141 ± 4.59 m. Pollination was widespread but variable from site to site and season to season with a maximum of 70% of all flowers being pollinated. Pollinator limitation is indicated. Traits essential for this pollination interaction include the coincidence of orchid and ant geographic distributions and the coincidence of flowering with the flight period of the ant. The production of pheromonelike substances and the distinctive floral morphology are also essential for attraction and manipulation of male ants. The ant mating system which the orchid can exploit is also important.

This inhibition of the repressive activity of NIFL was reversed by the addition of ferrous or manganous ions to the medium but was unaffected by other transition metals. The dependence on metal ions for NIFL activity was observed when NIFL was overexpressed and when cultures were exposed to oxygen or high levels of fixed nitrogen. Immunochemical evidence suggests that NIFL and NIFA associate to form a functional protein complex. Metal ions are apparently not required for the formation of this complex.

    Notes: Summary Rhodobacter capsulatus genes homologous to Klebsiella pneumoniae nifE, nifN and nifX were identified by DNA sequence analysis of a 4282 bp fragment of nif region A. Four open reading frames coding for a 51188 (NifE), a 49459 (NifN), a 17459 (NifX) and a 17472 (ORF4) dalton protein were detected. A typical NifA activated consensus promoter and two imperfect putative NifA binding sites were located in the 377 bp sequence in front of the nifE coding region. Comparison of the deduced amino acid sequences of R. capsulatus NifE and NifN revealed homologies not only to analogous gene products of other organisms but also to the α and β subunits of the nitrogenase iron-molybdenum protein. In addition, the R. capsulatus nifE and nifN proteins shared considerable homology with each other. The map position of nifX downstream of nifEN corresponded in R. capsulatus and K. pneumoniae and the deduced molecular weights of both proteins were nearly identical. Nevertheless, R. capsulatus NifX was more related to the C-terminal end of NifY from K. pneumoniae than to NifX. A small domain of approximately 33 amino acid residues showing the highest degree of homology between NifY and NifX was also present in all nifB proteins analyzed so far. This homology indicated an evolutionary relationship of nifX, nifY and nifB and also suggested that NifX and NifY might play a role in maturation and/or stability of the iron-molybdenum cofactor. The open reading rame (ORF4) downstream of nifX in R. capsulatus is also present in Azotobacter vinelandii but not in K. pneumoniae. Interposon-induced insertion and deletion mutants proved that nifE and nifN were necessary for nitrogen fixation in R. capsulatus. In contrast, no essential role could be demonstrated for nifX and ORF4 whereas at least one gene downstream of ORF4 appeared to be important for nitrogen fixation.

    Notes: Abstract The bean weevilBruchidius bimaculatus was found in fruits of 68 of 217 populations of theMedicago truncatula — M. littoralis complex of Israel, Greece, Italy and Spain. Infestation was higher in fruits of somewhat larger than mean size, and in the more pubescent and tightly coiled fruits. Bean weevil size proved to be independent of fruit size, so that the “preference” for larger fruits does not seem due to adaptation to a larger food source or a larger domicile. Fruit size has been used to delimit the host “species”M. truncatula andM. littoralis, but the character proved to be unimodal, and it is apparent that the taxonomy of the plant complex requires further clarification. Of the 11 472 fruits examined, less than 4% were infested. Given that the plants have evolved indehiscent legumes in which typically only one of the approximately six seeds survives to reproduce, and that very few seeds of a fruit are destroyed, the bruchid's infestation appears to be non-harmful, and indeed the association may be mutualistic.

    Notes: Abstract C-band patterns are described for 20Lilium spp. distributed across six sections. All species have a similar basic karyotype (n = 12) but C-bands differ markedly between them. The patterns are characterized by a dispersed scattering of thin intercalary bands as well as centric and NOR bands. Only one species,L. canadense, shows a clear equilocal pattern with intercalary C-bands occurring proximally in all of the longer chromosome arms. Comparing species, similar patterns are revealed forL. regale andL. sulphureum, forL. formosanum andL. longiflorum (all in sect.Leucolirion) and to a lesser extent forL. hansonii, L. martagon, andL. tsingtauense (sect.Martagon). The pattern forL. henryi (previously classed in sect.Sinomartagon) matches those ofL. regale andL. sulphureum quite well and its transfer to sect.Leucolirion is proposed. This is consistent with results from interspecies hybrids betweenL. henryi andL. regale (and related species) which are reportedly fertile. No other clear similarities in C-band patterns were seen across species. It seems that C-band patterns change rapidly inLilium and hence their usefulness in classification will be restricted to identifying closely related species.

    Notes: Abstract Nucleolar activity was analyzed in two samples of plants of the wild onion speciesAllium cernuum, one from Canada (2n=14) and one from the United States of America (2n=14+3B), using phase contrast analysis and C- and Ag-NOR-banding. Three chromosome pairs of the standard A-chromosome complement show small telomeric satellites, which corresponds to the maximum number of Ag-NOR-bands at metaphase and to the telomeric C-heterochromatin. However, the maximum number of nucleoli observed at interphase was eight, indicating that another chromosome pair possesses nucleolar activity. Corresponding results were obtained for the A-chromosome complement of plants from the United States of America, however, these plants further possess three small metacentric supernumerary chromosomes, which show telomeric satellites on both arms, corresponding to two telomeric Ag-NOR-bands observed after silver staining. The presented data clearly indicate that the B-chromosomes ofAllium cernuum possess rDNA gene clusters which show strong nucleolar activity.

It is argued that the Asian subg.Melo with x = 7 is derived from the African subg.Cucumis with x = 12; the latter contains all the polyploid species and has the most common basic chromosome number of theCucurbitaceae. No rearrangement was detected for the 113 chromosomes of sexuals, or for 41 chromosomes of two agamospermous individuals. In the other three agamospermous individuals, 3/16, 5/50, and 5/20 chromosomes showed evidence of chromosomal rearrangement. The majority of rearrangement events (10/13) occurred to the satellite rather than to the body of the NOR-chromosome. It is considered that such high levels of somatic chromosomal rearrangement in agamospermousTaraxacum may be the result of activity by transposable genetic elements. This recombination may be of selective advantage to asexual plants which cannot generate genetic variability through the sexual process.

Bees rarely visit more than one pollination unit per flower. Transferral of pollen to the bee is passive and nototribic although all bees collected on the flowers were female and 55% of the bees carried pollen loads with 2–5 pollen taxa in their scopae.Moraea brevistyla flowers are nectariferous but lack scent and last two days. They are visited infrequently by bees and only one femaleLasioglossum spec. carried the pollen ofM. brevistyla. Unlike flowers ofM. inclinata those ofM. brevistyla deposit pollen only on the head and thorax. Bee-mediated autogamy in both species is avoided due to the erratic foraging patterns of the bees and the flexibility of each stigma lobe as the bee backs out of the flower. Approximately 2–4 flowers in the inflorescences of both species (6–8 flowers/infloresence) develop into capsules.

Troll's typology of inflorescences which aims at more or less rigid, well defined types, this investigation accentuates the processes that constitute the evolutionary transformations leading from one typical form to another.Troll divided the inflorescences into the two types of monotelic and polytelic synflorescences, the first with a terminal flower on the main axis, the latter with a homogeneous florescence on the indeterminate axis. Both forms are enriched by proximal branches which repeat the structure of the main axis (paracladia). The evolutionary processes leading from the more primitive monotelic type to the advanced polytelic type are truncation (loss of the terminal flower) and homogenization of the distal branches, which thus form a homogeneous florescence. A closer survey of the polytelic groups reveals the fact that, usingTroll's criteria, the same distinction can be found within these groups themselves. Loss of the terminal florescence (truncation of 2nd and higher degree) as well as homogenization of the distal paracladia may lead to florescence-like units of higher complexity. Examples can be found inAsteraceae (Figs. 1 and 2),Fabaceae (Fig. 3 a),Mimosaceae (Fig. 3 b),Acanthaceae, and also in Monocots, as exemplified by theMarantaceae (Figs. 4 and 5). The so-called racemization (inversion of efflorescences from basipetalous to acropetalous) may be mentioned as a third element of transformation, emphasizing the unity of the florescences.—In consequence, there are more organizational levels than reflected in the twoTroll types. The polytelic type comprises several degrees of truncation and homogenization, the basis for a reasonable organizational analysis should therefore be the degree of ramification of flowering branches rather than the mere question of a terminal flower on the main shoot axis (Fig. 6). On the other hand the three processes of truncation, homogenization and racemization are evolutionary transformations that may occur independently from one another, thus giving rise to a large number of variations, which can not be satisfactorily interpreted by exactly defined types. On the basis of these considerations the question of homologous parts in inflorescences is reviewed. The homology of partial florescences and paracladia is accentuated contrary toTroll's interpretation (Fig. 7). Homogenization as an evolutionary trend may transform paracladia of different degree of ramification, leading to one-flowered units on the one side and to highly complex structures as in theMarantaceae on the other.

    Notes: Summary Nitrogen-fixing peanut root nodules are characterized by their unique structural organization, distinct from other legume nodules. The focus of this study has been in and around the hostsymbiont interface, where the bacterioid and the host cell surface (peribacteroid membrane envelope) interact during symbiosis. The infected nodule cells have revealed the presence of lipid bodies (oleosomes) in intimate association with the peribacteroid membrane, which encloses the large spherical bacteroids with a relatively narrow peribacteroid space. Electron dense structures, referred to as dense bodies have been found attached to the bacteroid outer membranes at the host-symbiont interface. The dense bodies are osmiophilic, amorphous and 3,3′-diaminobenzidine positive. The isolated intact bacteroids with dense bodies attached to their cell wall showed significant catalase activity. Many microbodies showing DAB-positive reaction have been found in the host cytoplasm, associated closely with the peribacteroid membrane. These ultrastructural and cytochemical characteristics of peanut root nodules suggest that lipids are utilized during symbiosis and the dense bodies and microbodies may be involved in the catabolic process.

Photosynthetic responses were measured under a series of irradiances (P-I curves) and temperatures to determine immediate responses of the plants before, during and after completion of the transfers. The Philippine variety did not show acclimation to 18 °C either after gradual transfer from the initial culture temperature of 25 °C or when abruptly transferred from 25 to 18 °C. The Florida variety did show acclimation to 25 °C when gradually transferred from 18 to 22 to 25 °C over the 6-week period, but not if abruptly transferred from 18 to 25 °C.     Notes: Abstract Wheat plants were grown in columns of soil until early stem elongation at a wide range of constant root temperatures. Two light environments were imposed and three levels of nitrogen fertilizer added at sowing. Shoot and root development and growth were measured by destructive sampling to investigate the combined effects of temperature and changing nutrient and assimilate supply. Both mainstem leaf and root axis production were linearly related to thermal time above a base temperature of 0°C. Low irradiance affected the appearance of mainstem tillers and associated nodal root axes. Nitrogen had little effect on shoot or root development but increased shoot area between 6 and 8 mainstem leaves. Higher temperatures and supplementary light resulted in larger root systems when compared at equivalent times after sowing. Total root length and root dry weight increased exponentially with thermal time, based on the mean of 4 cm soil and 2 cm air temperatures, but no single relation existed for all temperature and light treatments. Total plant dry matter, root length and root dry weight increased linearly with accumulated, intercepted, photosynthetically active radiation. Root growth responded less than the shoot to supplementary light. Increasing temperature reduced the proportion of root weight to total plant weight.

    Notes: Abstract Winter wheat was sown on 2 dates with 3 levels of nitrogen fiertiliser (0, 50 and 200 kg N ha−1) in one year and on 2 sites in a followign season. Shoot and root development and growth were measured between emergence and anthesis in the first season and emergence and 7 mainstem leaves in the second. Differences in temperature and light regime led to significant differences in shoot and root development and growth between sowing dates. A thermal time-scale, based on soil surface or air temperatures, with a base of 0°C, adequately described the production of mainstem leaves and nodal root axes over all treatments. Autumn applied nitrogen had little effect on development. Shoot growth and green area index increased exponentially with thermal time prior to spring nitrogen application and the completion of canopy development. Early-sown crops had larger root systems than late-sown crops prior to winter and this divergence was retained until anthesis. The relationship between root growth and thermal time was little better than with days after sowing and was not improved by either varying the site of temperature measurement or the base temperature used for calculation. Differences in soil texture and drainage, between sites, led to significant changes in root length distribution. Although spring applied nitrogen generally increased root length, its effects were inconsistent. There was a curvilinear relation between root length and the amount of photosynthetically active radiation (PAR) intercepted; this relation was unaffected by sowing date or nitrogen treatment. The amount of root produced per unit PAR decreased as the season progressed, reflecting the decrease in the proportion of total dry matter partitioned to the root system.

    Notes: Abstract TEM observations were carried out on 40 taxa of the familyVerbenaceae and 35 taxa of the familyOleaceae, in order to ascertain distribution, ultrastructure and morphology of the intranuclear proteinic inclusions in the mesophyll parenchymatic cells. The investigated genera amount to some 25% and 60% respectively of the genera of the two families. Inside theVerbenaceae, lamellar inclusions (L-type) occur in 6 out of 23 investigated genera: they are mostly present inside the tribesCitharexyleae andVerbeneae (both belonging toVerbenoideae), while they are absent in other subfamilies. All of the investigatedOleaceae genera show intranuclear crystalline inclusions (C 1-type) of three different shapes. Among theAsteridae this is a character peculiar toOleaceae. They appear to be a well defined natural group, including the controversial genusNyctanthes.

    Notes: Abstract S. cilicica Siehe andS. morrisii Meikle are two little known sibling species of theScilla siberica alliance with previously difficult taxonomy, especially with regard to specific delimitation and geographic distribution. Quantitative C-band karyotyping and DNA content determinations were performed in four provenances, including the type localities. Both species (2n = 12) differ strikingly in their banding patterns despite of similar DNA content. Karyotype diversification involved changes in eu- and heterochromatin quantities and resulted in some changes of chromosome form. Despite this, actual phylogenetic relatedness is indicated by a chromosomal marker (NOR-position) of rare occurrence in the alliance. Karyological results and circumstantial evidence from the literature suggest thatS. morrisii is an endemic, exclusive to Cyprus as a member of theS. siberica alliance.S. cilicica seems to be restricted to continental S. Anatolia opposite to Cyprus. Its occurrence on Cyprus is questionable.

    Notes: Abstract Didymocarpus geitleri spec. nov. is described from Sungai Pandan, Kuantan distr., Pahang, Peninsular Malaysia. Distinctive features include: leaden-grey leaves with long, erect hairs on the upper side and whitish, campanulate flowers with a long exserted, bright yellow style. The colouring of the latter is caused by densely arranged glands. Together with two yellow blotches in front of the filament insertion, the style apparently acts as an anther (pollen) dummy.

    Notes: Abstract Representative specimens of the EuropeanGladiolus species (excl.G. palustris) were used to study variability and relationships. 25 characters were measured for cluster, principal component and Wells hybrid analyses (Figs. 1–3). Three (with PCA) respective five (cluster analysis) main groups are revealed. Bulb characteristics appear to be very useful for indentification. The subspecific status ofG. communis subsp.byzantinus is confirmed. Hybrids of four different parental combinations were detected in our collection. Most species include polyploid series. Partly 3x, 4x, 6x, 8x, 12x and aneuploid cytotypes but no diploids were found. Polyploids as pioneers and hybridization of sympatric species are apparently linked to migrations during the glacial periods.

    Notes: Abstract In the early stages of ontogeny, the corolline parts of theEudesmia eucalypts develop as compound structures directly comparable to the early stages of the petals ofAngophora and the bloodwood eucalypts, but with the onset of androecial formation a marked difference takes place. Rather than forming on the floral apex, the stamen primordia arise on the basal adaxial components of the young corolline parts; this basal component develops into the staminophore of the mature flower. The operculum consists only of the dorsal components of the corolline parts, the homologues of the “dorsal keels” of theAngophora petals. If the corolline parts remain more or less free in their early developmental stages, corresponding groups of stamens are produced. Early corolline continuity leads to a continuous ring of stamens. The staminophore is not an organ sui generis, but a derivative of the corolla. The bundles of stamens in some species are best referred to as epipetalous groups, not antepetalous fascicles.
