    Notes: Abstract A field experiment with Drosophila melanogasterrevealed that when flies encounter a particular food type soon after emergence, the probability of their subsequently being attracted to such a resource is increased. In this experiment, the length of time flies experienced their postemergence environments was under the control of the flies themselves. The experiment thus realistically mimicked one form of experiential effect that may be important in nature. A theoretical model is developed which shows that enhanced adult preferences for the types of resources fed on as larvae can substantially increase the degree of host-based genetic subdivision within a polyphagous population.

Les mâles eux aussi ont révélé une activité sexuelle réduite aux photophases courtes, c'est-à-dire qu'ils sont entrés comme les femelles en diapause reproductive. Les photophases critiques pour l'induction de la diapause des mâles et des femelles n'ont pas présenté de différences. Les diapause des mâles et des femelles s'achèvent même sous courtes photophases, mais la diapause mâle était quelque peu plus faible que la diapause femelle. La photophase critique et le taux de diapause ont varié en fonction de l'origine géographique dans l'espèce actuelle. Lors de croisements entre lignées diapausantes et non-diapausantes, la photophase critique et la durée de la diapause ont été héritées quantitativement. A partir de ces expériences et d'expériences précédentes de croisements (Kimura, 1983), quelques modèles de méchnisme d'induction de la diapause de cette espèce sont proposés.
    Notes: Abstract In Drosophila triauraria Bock & Wheeler (Diptera: Drosophilidae) of which females were known to enter reproductive diapause at short daylengths, males also showed reduced mating activity at short daylengths, i.e., males as well as females entered reproductive diapause. The critical daylength for diapause induction did not differ between females and males. Both male and female diapause ended even under short daylengths, but the male diapause was somewhat weaker than the female diapause. The critical daylength and the diapause rate varied geographically in this species. In the cross between diapausing and non-diapausing strains, the critical daylength and the diapause duration inherited in a quantitative manner. On the basis of the present and previous crossing experiments, some models are proposed on the mechanism of diapause induction of this species.

    Notes: Summary Because natural populations ofDrosophila melanogaster are polymorphic for different allozymes of alcohol dehydrogenase (ADH) and becauseD. melanogaster is more tolerant to the toxic effects of ethanol than its sibling speciesD. simulans, information regarding the sensitivities of the different forms of ADH to the products of ethanol degradation are of ecological importance. ADH-F, ADH-S, ADH-71k ofD. melanogaster and the ADH ofD. simulans were inhibited by NADH, but the inhibition was relieved by NAD+. The order of sensitivity of NADH was ADH-F〈ADH-71k, ADH-S〈ADH-simulans with ADH-F being about four times less sensitive than theD. melanogaster enzymes and 12 times less sensitive than theD. simulans enzyme. Acetaldehyde inhibited the ethanolto-acetaldehyde activity of the ADHs, but at low acetaldehyde concentrations ethanol and NAD+ reduced the inhibition. ADH-71k and ADH-F were more subject to the inhibitory action of acetaldehyde than ADH-S and ADH-simulans, with ADH-71k being seven times more sensitive than ADH-S. The pattern of product inhibition of ADH-71k suggests a rapid equilibrium random mechanism for ethanol oxidation. Thus, although the ADH variants only differ by a few amino acids, these differences exert a far larger impact on their intrinsic properties than previously thought. How differences in product inhibition may be of significance in the evolution of the ADHs is discussed.

    Notes: Abstract A series of fourteen Pacific North-west Douglas-fir installations, ranging in age from 6 to 26 years were analysed with respect to site factors, foliage nutrients, and growth response to applied fertilizer. Unfertilized basal area increment ranged from 1.2 to 3.1 m2 ha−1 yr−1 with no apparent relationship with soil, stand age or site index. Basal area increment was correlated with foliage N and a critical level for N was calculated as 1.7%. Applications of 220 kg N ha−1 as urea increased growth between 0 and 95% of the unfertilized basal area growth, with an average of 24.9%. Response could be predicted from foliage N and unfertilized basal area increment. When the same relationships were applied to previously older stand data, results were more variable as elements such as B and S showed evidence of being limiting.

    Notes: Abstract The investigation studied the effect of fertilization with four nitrogen levels (0, 100, 150 and 200 kg/ha), four potassium levels (0, 100, 150 and 200 kg/ha) and four irrigation treatments (0, 3, 7 and 14-day interval) on the growth and yield of smooth Cayenne pineapples. Increasing irrigation frequency increased growth parameters of number of leaves, D-leaf length and days to 50% flowering. Fruit weight was highest at N = 150 kg/ha, K = 200 kg/ ha and irrigation once a week. Higher N levels (beyond 150 kg/ha) applied after the plant crop has been harvested did not further enhance yields of the ratoon crop. For K = 200 kg/ha potential and harvested yields were reduced in all irrigation treatments when N 〉 150 kg/ha.

    Notes: Abstract Field experiments with rice-wheat rotation were conducted during five consecutive years on a coarse-textured low organic matter soil. By amending the soil with 12t FYM ha−1, the yield of wetland rice in the absence of fertilizers was increased by 32 per cent. Application of 80 kg N ha−1 as urea could increase the grain yield of rice equivalent to 120 kg N ha−1 on the unamended soil. Although the soil under test was low in Olsen's P, rice did not respond to the application of phosphorus on both amended and unamended soils. For producing equivalent grain yield, fertilizer requirement of maize grown on soils amended with 6 and 12 t FYM ha−1 could be reduced, respectively to 50 and 25 per cent of the dose recommended for unamended soil (120 kg N + 26.2 kg P + 25 kg K ha−1). Grain yield of wheat grown after rice on soils amended with FYM was significantly higher than that obtained on unamended soil. In contrast, grain yield of wheat which followed maize did not differ significantly on amended or unamended soils.

    Notes: Abstract The investigation evaluated the productivity of plantain intercropped with cassava, cocoyam and yam, fertilized annually with 0, 320 and 480 kg N ha−1 respectively. Yields from nitrogen fertilized intercrops were higher than those of unfertilized treatments. In plantain + cassava intercrop receiving 480 kg N ha−1 plantain growth was suppressed. Plantain intercropped with yam and fertilized with 320 kg N ha−1 matured early and produced better bunches than other treatments. Plantain + yam or cocoyam intercropping systems fertilized with 320 kg N ha−1 were recommended because of improved plantain establishment and increased combined crop yields.

    Notes: Summary The phenotype of rotund (rn) null alleles is described, and compared to wild type. The mutants are expressed zygotically and cause position specific defects in certain imaginal discs (antenna, legs, wing, haltere and proboscis) and their corresponding adult derivatives. In the discs, specific folds are absent in rn mutants compared to wild type. Clonal analysis shows that the rn + gene is partially autonomous in its expression in cells destined to form certain distal parts of the adult appendages. The results are consistent with the idea that the rn + gene is required for normal morphogenesis of specific distal parts of the adult appendages.

The N2-CO2 near-resonant system in theD andE regions is taken into account. We describe our understanding of the methods necessary to find the vibrational populations of N2 and CO2 (asymmetric mode of CO2). The calculations of the vibrational temperatures in theD, E, andF regions for the mid-latitude ionosphere and an aurora are presented. The connection between the excited species and the 4.26-μm radiation intensities is considered.     Notes: Summary Transformations of tumorous-head Drosophila melanogaster were examined in order to investigate whether head structures were replaced by specific abdominal structures. Heads selected for the presence of genital structures were analyzed in detail. Female abnormalities included any combination of vaginal teeth, vulvar papillae, sensilla trichodea, abdominal tergites 6 (T6), 7 (T7), 8 (T8) and anal plate. Anal plate was observed in the prefrons and rostral membrane, while all other genital structures were intimately associated with modified shingle cuticle. Male abnormalities included transformation of antennal structures to penis, clasper teeth, lateral plate, anal plate and eye to T6. The distribution of each type of homeotic structure was confined to general regions of the eye-antenna, with no precise dividing lines between them. However, the spatial sequence of homeotic structures in the eye-antenna was generally the same as the sequence of the same structures in the posterior abdomen.

    Notes: Summary In Drosophila melanogaster the neuroblasts separate from epidermoblasts to give rise to the neural primordium. This process is under the control of several genes. The group of the so-called neurogenic genes is required for epidermal development; other genes, comprising those of the achaete-scute complex and daughterless, are required for neural development. We have studied the relationships between both groups of genes in two different ways. We have analyzed the phenotype of double-mutant embryos and our results show that the neural hyperplasia caused by neurogenic mutations can be partially prevented if a mutation in one of the other genes is present in the same genome. Only the neural cells that do not require the function of a particular gene of the achaete-scute complex in the wild-type seem to develop to a neural fate in the double mutant embryos. At least some of the genetic interactions affect the transcriptional level, as shown by in situ hybridization, since the territories of transcription of the achaetescute genes are expanded in neurogenic mutants. All cells of the neurogenic region of the double mutants apparently initiate neural development. However, during later development some of these cells switch their fate either to epidermogenesis or to cell death and this leads to the final phenotype of the double mutants. We discuss these results with respect to the events of early neurogenesis.

    Notes: Summary The temperature-sensitive mutation shibire (shi) in Drosophila melanogaster is thought to disrupt membrane recycling processes, including endocytotic vesicle pinch-off. This mutation can perturb the development of nerves and muscles of the adult escape response. After exposure to a heat pulse (6 h at 30° C) at 20 h of pupal development, adults have abnormal flight muscles. Wing depressor muscles (DLM) are reduced in number from the normal six to one or two fibers, and are composed of enlarged fibers that appear to represent fiber fusion; large spaces devoid of muscle fibers suggested fiber deletion. The normal five motor axons are present in the peripheral nerve PDMN near the ganglion. However, while some motor axons pass dorsally to the extant fibers, other motor axons lacking end targets pass into an abnormal posterior branch and terminate in a neuroma, i.e., a tangle of axons and glia without muscle target tissue. Hemisynapses are common in axons of the proximal PDMN and within the neuroma, but they are rarely seen in control (no heat pulse) shi or wild-type flies. All surviving muscle fibers are innervated; no muscle tissue exists without innervation. Fibrillar fine structure and neuromuscular synapses appear normal. Fused fibers have dual innervation, suggesting correct and specific matching of target tissue and motor axons. Motor axons lacking target fibers do not innervate erroneous targets but instead terminate in the neuroma. These results suggest developmental constraints and rules, which may contribute to the orderly, stereotyped development in the normal flight system. The nature of the anomalies inducible in the flight motor system in shi flies implies that membrane recycling events at about 20 h of pupal development are critical to the formation of the normal adult nerve-muscle pattern for DLM flight muscles.

    Notes: Summary Cell-cell interactions are involved in mediating developmental fate. An example is the decision of the neuroectodermal cells of Drosophila to develop as neural or epidermal progenitors, where cellular interactions participate in the process of acquisition of either cell fate. The results of heterochronic cell transplantations we describe here suggest that both neuroblasts and epidermoblasts are not irreversibly committed to a particular developmental fate. Rather, they retain the ability to interact with neighbouring cells and, under our experimental conditions, are capable of switching their fate during a relatively long period of time, i.e. until the end of embryonic stage 11.

This transformation suggests a reduced activity of the Sex combs reduced (Scr) gene on the basis of gene dosage studies, as well as enhanced expression of the phenotypes of the weak Scr − alleles in Rg-bx − larvae. Morphological observations of adult flies doubly heterozygous for Rg-bx and Scr mutations also suggest the enhancement of an aspect of Scr adult phenotypes. I conclude that the Rg-bx + gene function is required for the optimal expression of the Scr gene in larval and imaginai cells.

    Notes: Summary Random insertions of a promotor fused to a reporter gene, such as Lac-Z, reveal regulatory sequences that confer temporal and spatial patterns of gene expression in eukaryotes. These patterns may reflect the activity of a neighbouring gene and thus lead to the isolation of new genes essential for normal development. Here, we demonstrate that this hypothesis is true for an insertion into the well characterized segmentation gene, hairy, in Drosophila. The insertion is homozygous lethal and fails to complement other hairy alleles, giving the phenotype described for hairy mutations. The insertion is located at 66D on the polytene chromosome map, is within 300–600 bp 5′ to the first hairy exon, and is orientated in the same sense (5′-3′) as the hairy transcription unit. Expression of β-galactosidase (β-gal), deriving from the insertion, follows closely the spatio-temporal patterns of expression of hairy gene product during embryogenesis. In addition, other sites of β-galactosidase expression are shown in the third larval instar stage and in the adult ovary. The results show that some insertions, giving restricted patterns of reporter gene expression, will reflect the temporo-spatial activity of a nearby gene.

    Notes: Summary Drosophila melanogaster embryos were injected before the blastoderm stage with conditioned media from several male Burkitt's lymphoma human cell lines and the Daudi cell line. Such injections do not have any effect on the male genital apparatus or on the female tract. The Daudi conditioned medium modifies the ovarian morphogenesis of the flies and the rudimentary ovaries obtained look like nymphal gonads. Moreover, they have a drastically reduced number of germ cells. The ovaries that looked functional contain numerous necrotic germ cells and the mean number of ovarioles per fly is significantly smaller than that of the controls. The abnormalities observed resemble the results of experimental and genetic lack of germ cells. They disappear at very high dilution (1×10−6).

Positive molecules are able to migrate through intercellular bridges from the oocyte to the nurse cells, but are unable to migrate detectably from nurse cells to the oocyte. Their negatively charged counterparts can move from the nurse cells to the oocyte, but are unable to traverse the intercellular bridges in the counter direction. This charge-dependent movement of molecules is accompanied by an electrical potential difference, focused across the nurse cell-oocyte bridges, which makes the nurse cells negatively charged to the oocyte. The addition of insect hemolymph to the physiological salt solution in which the experiments were performed resulted in only a small increase in the transmembrane resistance, but enhanced the potential difference between oocyte and nurse cells from 0.2±0.3 (SE) mV (nurse cells negative) to 2.3±0.45 (SE) mV (nurse cells negative).

    Notes: Summary Most viable alleles of homeotic genes cause partial transformations within given lineages in a topographically specific fashion. We study this phenomenon as a way to understand the normal mechanisms involved in the spatial regulation of homeotic gene expression. To this end we have investigated the distribution of Ultrabithorax (Ubx) proteins in imaginai discs mutant for hypomorphic and neomorphic alleles of Ubx and alleles of trans-acting genes. We find that the morphological discontinuities observed in the adult transformations are caused by corresponding new patterns of the Ubx proteins in the imaginai anlagen. These novel patterns of Ubx proteins are understood as a consequence of a process of reinforcement-extinction of Ubx expression. The evidence suggesting that this process results from a positive feed-back loop and cell-cell interactions is discussed.

    Notes: Summary The phenotypic effects in imaginal hypodermal tissue of a number of Abdominal-B mutations of the bithorax complex are described. Evidence is given from complementation analysis that the phenotypic heterogeneity in both the spatial limits and the nature of the homeotic transformations produced is not an arbitrary classification of allelic differences that we find. We have used genetic mosaic analysis to support the interpretation that the Abdominal-B genetic unit can exist in a number of alternative functional states of expression during development and that individual Abdominal-B mutations may abolish some states whilst leaving others unaffected.

    Notes: Summary Fused is a segmentation gene belonging to the segment-polarity class. Mutations at thefused locus are known to display pleiotropic effects, causing zygotically determined anomalies of ovaries and of some adult cuticular structures, and maternally determined embryonic segmentation defects. In order to determine the amorphic phenotype offused and to study the genetical basis of its pleiotropy, newfused alleles (18 viable and 11 lethal) were isolated. The phenotype of these mutants and of others already known are described, taking into account zygotic and maternal effects. The main results provided by this analysis are as follows. Firstly, allfused alleles show the whole complex fused phenotype, and a good correlation is observed between the strength of the wing and segmentation defects, suggesting that a single function is involved in both processes. Secondly, all embryonic and larval lethals carry deficiencies which allow us to localizefused between the 17C4 and 17D2 bands of the X-chromosome. Thirdly, the 24 viable and 2 pupal lethals examined behave as point mutants, as shown cytologically or by Southern blot analysis. However, only one of them, the pupal lethalfu mH63 was proven to carry a nullfused allele, since it displays in germ-line clones a strong maternal phenotype and a very low zygotic rescue, similar to those of the small deficiencyDf(1)fu z4. The phenotype of the amorphic mutant indicates that zygotic ezpression offused is required for normal metamorphosis, while maternal expression is necessary for a normal segmentation pattern, since a complete loss offused expression during oogenesis cannot be compensated zygotically.

    Notes: Summary The gastrulation defective (gd) locus is a maternally expressed gene in Drosophila required for normal differentiation of structures along the embryonic dorso-ventral axis. Cuticular defects of the offspring from females with different combinations of gd alleles comprised a phenotypic continuum. Complementation among several alleles produced normal offspring while progressively more severe mutations produced a graded loss of structures from ventral, and then lateral, blastoderm cells. The most severely affected embryos consisted entirely of structures derived from dorsal blastoderm cells. Histological examination of staged siblings from selected allelic combinations showed that internal tissues were similarly affected. The tissues observed in amorphic embryos support new, more dorsal, assignments of fate map positions for blastoderm precursors of the cephalopharyngeal apparatus, hindgut and ventral nerve cord. The loss of ventral and lateral structures did not occur through cell death and appeared to involve a change in blastoderm cell fate. A direct effect of the mutations on blastoderm cell determination, however, was insufficient to explain the development of the dorsalized embryos. Intermediate phenotypes suggested that cell interactions or movements associated with morphogenesis are required for the determination of some cell fates in the dorsoventral axis. Thus, the developmental fate of all blastoderm cells may not be fixed at the time of blastoderm formation.

    Notes: Summary A systematic search for X chromosome loci showing a dominant maternal interaction with the segmentation genes Krüppel, hunchback, knirps and hairy was performed using deficiencies spanning 65% of the X chromosome. No interaction with the knirps gene was observed, but five regions of the X chromosome showed a maternal dominant interaction with the Krüppel gene. Two of these regions also show a maternal dominant interaction with either hunchback (region 10A7–10A8) or hairy (region 10E1–10F3). In all of these interactions dead embryos were observed which showed the same defects as embryos homozygous for the segmentation gene tested. These results suggest that a complex repartition of maternal products necessary for subsequent segmentation may occur in the Drosophila egg.

    Notes: Summary At two locations in the Sonoran Desert, yeasts were sampled from species of Drosophila, the flies' cactus hosts, and other neighboring sources of cactophilic yeasts to determine the relation between the yeasts vectored by the fly and the yeasts found in their breeding sites. D. mojavensis, D. nigrospiracula, and D. mettleri vectored yeast assemblages significantly more similar to the yeast species found on the rot from which the flies were collected than to the yeasts found on other rots from the flies host cactus or other rotting cactus at the same site. Rots with Drosophila had fewer yeast species than those without flies, suggesting that flies were associated with younger rots. Rots with flies and the Drosophila also had more yeast species with the capability to produce ethyl acetate than rots without flies. The results support the contention that cactophilic Drosophila feed on a subset of the yeasts available in an area, and may act to maintain differences among the yeast communities found on different species of cactus.

    Notes: Summary Drosophila C virus (DCV) has a considerable impact on ovarian morphogenesis inDrosophila melanogaster host populations. This virus also affects the developmental time and the fresh weight of infected females. In order to investigate the hypothesis that DCV may play a role in the dynamics ofDrosophila populations, the fertility and embryonic and larvo-pupal death rates of a host population and that of five DCV-free populations were determined. A comparison of two populations, one of them DCV-free, the other infected, suggested that the fertility of the DCV-infected flies was higher than that of uninfected flies, despite a greater larvo-pupal death rate. Fertility of the infected flies was greater among the infected population than for the DCV-free populations. The DCV-free populations originated from five different localities. The virus clearly does have an impact on the biotic potential of its host population. This paper reports for the first time a positive interaction between a viral population and a host population as it increases certain parameters of host population dynamics.

On the basis of their immunological similarity, parallel dosage responses, and cosegregation of electrophoretic mobility differences, it is concluded that both isozymes are the product of a single gene. The consequences of this in relation to the regulation and evolution of this unusual gene-enzyme system are discussed. It is inferred that the origin of the phosphagen shuttle must predate the divergence of invertebrates and vertebrates.

Seven major and at least four minor glue protein fractions were recognized. Six of the major fractions are glycosylated. They migrate as three prominent doublets (〉100, 43, and 30/28 kd). The synthesis of traceable amounts of these major fractions begins already during the second as well as during the early stages of the third larval instar. The 43-kd and the 30/28-kd fractions are coded by X-chromosomal genes. They are probably clustered within the huge puff of division 10, which is the most prominent X-chromosomal puff in the polytene chromosomes of the third larval instar. Complex posttranslational modification of all but one major glue protein fraction (14 kd) leads to the formation of about 15 different protein fractions in the final glue product. The amount of glue protein produced byD. n. nasuta larvae (in relation to the total saliva proteins) is nearly twice the amount produced byD. melanogaster larvae (ca. 55 and 32%, respectively).

    Notes: Summary The zen protein is encoded by the zerknullt gene required for normal early development inDrosophila. Like many regulatory proteins of this type, zen contains a 60 amino acid homeobox sequence. We have purified the zen protein and studied its solution behavior and its interaction with DNA. The zen protein exists as a monomer in solution with a molecular weight of about 40000. It binds specifically to a site about 900 bases upstream from thezen gene. Within this binding site DNase protection experiments indicate that binding is confined to two regions approximately 11 and 14 bases in length that are separated by about 30 base pairs. The protein concentration dependence of the binding curve suggests that protein binding is non cooperative.

    Notes: Abstract Although many experiments on laboratory stocks ofDrosophila have suggested that mate choice is a major feature of sexual selection in this organism, few attempts have been made to measure its extent in wild populations. In this study, a crossing design was used to obtain a set of 13 genetically identical independent lines representative of genotypes from an African population ofDrosophila melanogaster. They were tested for variation in sexual behavior using dyadic tests. Significant variation in orientation and vibration latencies was found for males, and in mating speed and copulation duration for both sexes. No evidence of assortative mating, either positive or negative, was found. The absence of a correlation in mating speed between males and females sharing the same genotype leads us to doubt the applicability of the notion of “male eagerness” and “female reluctancy” inDrosophila and the importance of “vigor” as a factor in mating speed. The absence of mate choice in natural populations ofDrosophila seems to us the most likely hypothesis on the basis of both theory and empirical evidence.

    Notes: Abstract Digital signal processing methods have revealed spectral components inDrosophila melanogaster's andD. simulans' male courtship songs that had gone undetected in previous studies. We found that a bout of courtship hum (“sine song”) inD. simulans typically consists of a narrowband fundamental frequency, accompanied by second and third harmonics that can comprise a major fraction of the power in the signal. The pulse song spectra consisted of single broad-band peaks of highly variable frequencies, which, nevertheless, are characteristically different in these two species. Genetic elements of the newly discovered song components were examined by analysis of theD. melanogaster/D. simulans hybrid. Such males were found to be intermediate in production of sine song harmonics as well as in other parameters of courtship song, except for sine song and intrapulse frequency bandwidths, for which there may be dominant factors inD. simulans.

    Notes: Summary The relative net fitness of a compound chromosome strain of Drosophila melanogaster was about 0.05, compared with the chromosomally normal strain from which it was derived. Based on meiotic considerations alone, the expected relative fitness was about 0.25. There were no significant differences in fertility between the compound and normal strains; the compound strain produced about 28% as many offspring as the normal strain and developed faster than the normal strain in two replicates, and slower in one replicate. The low relative fitness of the compound strain was apparently due to assortative mating, in which normal females discriminated strongly against compound males. Implications for pest control projects are dicussed.

    Notes: Summary The topography of oxytocin (OT)- and vasopressin (VP)-containing axons of the hypothalamo-neurohypophyseal system was studied in the neurohypophysis of the Djungarian hamster (Phodopus sungorus) by means of immunohistochemistry. Compared with other mammalian species, the neurohypophysis of Phodopus shows some peculiarities. Accumulations of OT-immunoreactivity around the distal vessels of the primary portal plexus can be observed in the distal median eminence and neural stem. This staining pattern indicates that OT is secreted into portal blood. In the neural lobe, OT- and VP-immunopositive fibers terminate in different areas. The vast majority of the OT-containing axons is distributed in the dorsal part of the neural lobe. In contrast, VP-containing axons are mainly found in the centre of the neural lobe up to the pars intermedia.

    Notes: Summary The distribution of VIP- and TRH-immunoreactivity in neurons and processes within the hypothalamus of the pigeon was investigated with light-microscopic immunocytochemical techniques. Most of the VIP-containing neurons are concentrated in the middle and caudal parts of the hypothalamus, with the greatest concentration of perikarya occurring in the medial and lateral part of the ventromedial hypothalamic nucleus and the infundibular nucleus. These cells give rise to axons that seem to extend into the median eminence. An extensive network of VIP-immunoreactive fibers and varicosities occupy the external layer of the median eminence. The majority of TRH-containing neurons is found in the anterior hypothalamus with the greatest concentration of cells in the magnocellular preoptic, medial preoptic, suprachiasmatic and paraventricular nuclei. TRH-immunoreactive fibers and varicosities form a dense arborization in the external layer of the median eminence. Lactation seems to induce substantial changes in VIP as well as in TRH-immunostaining in the median eminence and other hypothalamic regions as compared to control, sexually active animals. Furthermore, TRH-immunoreactivity decreased in the median eminence following 60-min exposure to cold. These results suggest that VIP- and TRH-containing pathways in the pigeon hypothalamus are involved in the mediation of neuroendocrine responses.

In the mesencephalic auditory center (MLD) about 20% of the cells reveal immunoreactivity. In contrast, the thalamic relay station nucleus ovoidalis is devoid of immunostained somata. This nucleus contains a high density of punctate immunoreactive structures presumed to be GABAergic axon terminals. In the auditory forebrain center field L and the auditory portions of the hyperstriatum ventrale, up to 8% of the cells were immunopositive. These neurons were significantly smaller than estimated from measurements of the overall cell population in these nuclei. From the two-dimensional arrangement of immunopositive neurons it is suggested that the GABAergic system in the avian auditory telencephalon consists of two separate groups of neurons: one subgroup mediating local inhibitory interactions, the other responsible for lateral inhibition between different frequency representations.

    Notes: Summary The innervation and myocardial cells of the human atrial appendage were investigated by means of immunocytochemical and ultrastructural techniques using both tissue sections and whole mount preparations. A dense innervation of the myocardium, blood vessels and endocardium was revealed with antisera to general neuronal (protein gene product 9.5 and synaptophysin) and Schwann cell markers (S-100). The majority of nerve fibres possessed neuropeptide Y immunoreactivity and were found associated with myocardial cells, around small arteries and arterioles at the adventitial-medial border and forming a plexus in the endocardium. Subpopulations of nerve fibres displayed immunoreactivity for vasoactive intestinal polypeptide, somatostatin, substance P and calcitonin gene-related peptide. In whole-mount preparations of endocardium, substance P and calcitonin gene-related peptide immunoreactivities were found to coexist in the same varicose nerve terminals. Ultrastructural studies revealed the presence of numerous varicose terminals associated with myocardial, vascular smooth muscle and endothelial cells. Neuropeptide Y immunoreactivity was localised to large electron-dense secretory vesicles in nerve terminals which also contained numerous small vesicles. Atrial natriuretic peptide immunoreactivity occurred exclusively in myocardial cells where it was localised to large secretory vesicles. The human atrial appendage comprises a neuroendocrine complex of peptidecontaining nerves and myocardial cells producing ANP.

    Notes: Summary The transmitter content of identified inhibitory interneurons in the flight system of the locust, Locusta migratoria, has been characterized using antibodies raised against protein-conjugated gamma aminobutyric acid. Identified flight neurons were filled with the fluorescent dye, Lucifer Yellow. Serial sections of dye-filled neurons were incubated with an antibody to gamma aminobutyric acid which was subsequently tagged with a fluorescent marker. Excitatory motoneurons to wing muscles and 13 flight interneurons (3 excitatory, 7 inhibitory, and 3 with unknown synaptic effect) were examined. Neither the moto-neurons nor any of the 3 excitatory interneurons contained immunoreactive material. Six of the 7 inhibitory interneurons did contain immunoreactive material. All the neurons which contained immunoreactive material and whose synaptic effect is known were inhibitory. We conclude that most of the inhibitory flight interneurons which have been described use gamma aminobutyric acid as their transmitter. Interestingly, at least 1 set of interneurons known to be inhibitory does not use gamma aminobutyric acid. We predict that the 2 interneurons which do contain immunoreactive material and whose synaptic effect is not yet known will be found to have inhibitory roles in the operation of the flight circuitry.

    Notes: Summary The distribution of somatostatin-immunoreactive cells and processes throughout the human entorhinal region and subjacent white matter was examined either by the unlabelled antibody-enzyme method or by the avidin-biotin method. The brain slices were obtained at autopsy with a short post-mortem delay. The majority of somatostatin immunoreactive nerve cells was found in the inner principal layer and subjacent white matter. In addition, individually scattered immunoreactive neurons were observed in both the outer principal layer and lamina dissecans. The immunoreactive perikarya varied in shape and ranged in size from 10 to 30 μm. Without exception the neurons could be classified as belonging to the group of non-pyramidal neurons. Each neuron gave rise to a few thick dendrites and a thin axon with a beaded appearance. In the adult human brain, the pattern formed by lipofuscin granules deposited in the nerve cells can be considered characteristic for the type of the neuron. Therefore, immunoreactive perikarya were documented, destained of chromogen and restained to demonstrate lipofuscin pigment and basophilic substance. It became evident from these studies that the previously immunoreactive cells were characterized by a large rounded and eccentrically located nucleus, sparse basophilic substance and, in most cases, a lack of lipofuscin granules. A few of the immunoreactive cells were laden with coarse pigment granules. The findings permit classification of entorhinal somatostatin-immunoreactive neurons as either non-pigmented or pigment-laden non-pyramidal neurons.

    Notes: Summary The presence of 28 kDa calbindin in human odontoblasts was studied by use of specific antibodies raised against chick duodenal 28 kDa calbindin, in immunofluorescence, immuno-peroxidase, and electron-microscopic labelling experiments. The calbindin-like protein was detected mainly in the cytoplasm of odontoblast cell bodies, in their processes and occasionally in their nuclei. Correspondingly, at the ultrastructural level, immunoreactive material was associated with the cytosol, microfilaments and cilia. These findings suggest that human odontoblasts express a 28 kDa vitamin D-dependent calcium-binding protein, unlike those of rats and mice in which ameloblasts are the only cells immunoreactive for the protein.

    Notes: Summary Certain secretory cells in the hypophysial pars tuberalis of the Djungarian hamster display marked circannual structural alterations. The present investigation deals with the immunohistochemical properties of this cell group. A distinct TSH-like immunoreactivity was found in secretory cells of this type in the pars tuberalis of animals exposed to long photoperiods, whereas under short photoperiods the TSH-like immunoreactivity was nearly absent. In the pars distalis, the number and distribution of TSH-positive cells did not differ significantly between animals maintained under long and under short photoperiods. LH-and FSH-positive cells could not be detected in the pars tuberalis, but they are clearly present in the pars distalis of both groups of hamsters. Our immunocytochemical results suggest that photoperiodic stimuli influence the secretory activity of TSH-like immunoreactive cells in the pars tuberalis. A connection with the neuroendrocrine-thyroid axis is discussed.

    Notes: Summary We used immunohistochemistry to identify cells present in pituitary allografts in the hamster. Hypophyses removed from neonatal hamsters or adenohypophyses removed from adult females were placed beneath renal capsules of hypophysectomized adult females. Serum PRL, LH, and GH concentrations were measured at two, five, and eight weeks after placement of allografts. Allografts were removed after eight weeks and stained for cells containing PRL, LH, FSH, GH, or ACTH. Allografts did not release LH or GH. Those of adult adenohypophyseal tissue released significantly more PRL. The morphology of allografts of neonatal hypophyseal tissue resembled that of the adult adenohypophysis in situ. Lactotrophs, corticotrophs, somatotrophs and LH-cells were observed; very few FSH-cells were present. Allografts of adult adenohypophyseal tissue contained pituitary cells, numerous cavities, often enclosing lymphoid cells, and fibrous tissue. Atypical lactotrophs were the numerically dominant cells in these allografts; all other cells were present. The LH-cells outnumbered FSH-cells. These observations suggest that: (a) development of normal adenohypophyseal morphology can occur in an ectopic position; (b) intracellular hormones are present in cells in an ectopic site; (c) development and retention of intracellular FSH is more dependent on occupation of the normal position of the adenohypophysis than is retention of intracellular LH; and (d) release of PRL occurs from atypical cells in allografts of adult adenohypophyseal tissue.

    Notes: Summary Snell dwarf mice display remarkable retardation of growth after birth and are known to lack prolactin (PRL), thyroid stimulating hormone (TSH) and growth hormone (GH). The aim of this study was to determine the reason for these hormonal deficiencies. We examined the fine structure of the gland and its immunohistochemical staining pattern with respect to antisera raised against PRL, TSH, GH, adrenocorticotrophic hormone (ACTH) and luteinizing hormone (LH). The gland of control mice reacted immunohistochemically against all antisera used, whereas only ACTH-producing cells (ACTH cells) and LH-producing cells (LH cells) were distinguished in the dwarf mice. ACTH cells in dwarf mice varied in cell shape, although they were similar in size to those of controls. The distribution of secretory granules in the cytoplasm varied from cell to cell. LH cells in the dwarf mice showed immature features, having poorly developed rough endoplasmic reticulum and Golgi apparatus. The cells were about half the size of controls, and secretory granules were smaller. In dwarf mice, non-granulated cells were encountered in addition to granulated ACTH and LH cells. Some of them formed small clusters, characteristic cell junctions being found between the cells; they thus appeared to be follicular cells. The above results suggest that hormone deficiency in Snell dwarf mice is a result of a defect in the hormoneproducing cells in the gland.

    Notes: Summary The distribution and origin of neurokinin A (NKA)-like immunoreactivity were investigated in feline dental pulp by an indirect immunofluorescence method. NKA-containing nerve fibres with varicosities, which entered the dental pulp via apical foramen, were distributed throughout this tissue. Many NKA-containing nerve fibres were localized around blood vessels, but some were observed apart therefrom. At the odontoblastic layer, thin NKA-containing nerve fibres were observed running straight toward the pulp-predentinal border between odontoblasts. After inferior alveolar nerve section, all NKA-containing nerve fibres disappeared in the dental pulp, while the removal of the superior cervial ganglion resulted in no change in the distribution of these fibres. The correlation of NKA-like immunoreactivity and substance P (SP)-like immunoreactivity was also investigated by double-immunofluorescence technique. The distribution of NKA-containing nerve fibres was very similar to that of SP-containing nerve fibres; it appeared that all NKA-containing nerve fibres contained SP.

    Notes: Summary Nervous elements in the periodontal ligament and dental pulp of rat incisors were investigated by means of immunohistochemistry for neurofilament protein (NFP) and glia-specific S-100 protein. The periodontal ligament in the incisors was densely innervated by NFP-immunoreactive nerve fibers; the distribution of the nerve fibers and their terminations differed markedly from those in molars. NFP-positive, thick nerve bundles entered the lingual periodontal ligament through slits located in the mid-region of the alveolar socket, and immediately formed numerous Ruffini-like corpuscles. In the labial periodontal ligament, all of the NFP-immunoreactive nerve fibers terminated in free endings. The restricted location of the stretch receptor, Ruffini-like corpuscle, in the lingual periodontal ligament appears to be an essential element, because this region is regularly extended during mastication. The nervous elements were restricted to the alveolar half of the periodontal ligament in every region; they avoided the dental half of the periodontal ligament, which presumably moves continuously with the tooth. Pulpal nerve fibers in incisors also showed a characteristic distribution different from those in molars; individual nerve fibers with beaded structures ran in the center of the pulp toward the incisai edge, and did not form the subodontoblastic nerve plexus of Raschkow. Immunostaining for S-100 protein revealed a distribution pattern of nervous elements similar to that for NFP, suggesting that the nerves supplying the periodontal ligament and dental pulp were mostly covered by a Schwann sheath.

    Notes: Summary Chronically stimulated fast-twitch muscles of the rabbit were histochemically and immunohistochemically analyzed in serial cross sections (1) for percentages of fiber types, and (2) for the presence of myosin heavy chain isoforms during fast-to-slow transformation. By four weeks of stimulation the number of type-I fibers had increased more than fourfold, while only about 6% of the original IIB fibers remained. Type-IC and -IIC fibers transiently rose to 20% of the total fiber population. After 16 weeks, the number of type-I fibers had increased to 42%. With prolonged stimulation fewer fibers reacted with antibodies against embryonic and neonatal myosins and more with the antibody against slow myosin. The reaction for embryonic myosin was most often detected in the C fibers (IC, IIC). Immunohistochemical subtypes were observed for each fiber type in the stimulated muscles. The greatest number was seen in type-IIC fibers, which, in addition to their reaction for fast/neonatal and slow myosins, might also react with the antibodies against neonatal/embryonic and embryonic myosins. These findings indicated that the transforming fibers temporarily expressed myosin heavy chain isoforms normally not detectable in adult skeletal muscle. Myotubes reacted strongly with the antibodies against fast/neonatal and embryonic myosins, and some of them also with the antibody against slow myosin. Thus, it appears that under the influence of the low frequency stimulus pattern some of the newly formed myotubes developed into type-I fibers.

    Notes: Summary The neuropeptide content of nerve fibers associated with submucosal arteries in the small intestine of guinea pigs was studied in whole-mount preparations using immunohistochemical methods. Tissues were obtained from normal animals or animals in which the small intestine had been extrinsically denervated. In normal animals, submucosal arteries are innervated by extrinsic sensory nerve fibers which contain both substance P and calcitonin gene-related peptide, and by sympathetic noradrenergic nerve fibers. In preparations obtained from animals 5–9 days after denervation, nerve fibers which contained substance P without detectable calcitonin gene-related peptide were associated with a few submucosal arteries. Nerve fibers which contained vasoactive intestinal peptide were also associated with some arteries. By 42–48 days after extrinsic denervation, substance P-containing fibers (without calcitonin gene-related peptide) and vasoactive intestinal peptide-containing fibers were associated with nearly every blood vessel. The extrinsic sympathetic nerve fibers did not regenerate during the course of this study. The nerve fibers associated with submucosal arteries in denervated tissues were not sensitive to capsaicin treatment. The alteration in the innervation of submucosal arterioles that follows extrinsic denervation of the gut may reflect either an increase in the neuropeptide content of the fibers, synthesis of a new peptide, or an increase in the number of fibers as a result of axonal sprouting.

    Notes: Summary The formaldehyde-induced fluorescence technique had shown 5-hydroxytryptamine-containing enteric neurons in the intestine of the teleost Platycephalus bassensis, but did not reveal such neurons in the intestine of Tetractenos glaber or Anguilla australis. Re-examination of these animals with 5-hydroxytryptamine immunohistochemistry showed immunoreactive enteric neurons in the intestine of all three teleost species. The 5-hydroxytryptamine-containing enteric neurons showed essentially the same morphology in all species examined: the somata were situated in the myenteric plexus, extending down into the circular muscle layer, but none were found in the submucosa; processes were found in the myenteric plexus, the circular muscle layer and the lamina propria. It was concluded that the neurons may innervate the muscle layers or the mucosal epithelium, but were unlikely to be interneurons. In a range of teleosts, enterochromaffin cells were found in the intestine of only those species in which the formaldehyde technique did not visualize neuronal 5-hydroxytryptamine. Available evidence suggests that, in vertebrates, 5-HT-containing enterochromaffin cells are lacking only where there is an innervation of the gut mucosa by nerve fibres containing high concentrations of 5-HT.

    Notes: Summary The distribution and localization of the calciumbinding protein, calbindin-D 28K (CaBP28K), in the spinal cord motoneurons of larvae of the teleost fish, Apteronotus leptorhynchus (Gymnotidae) and Pollimyrus isidori (Mormyridae), and in the adult goldfish, Carassius auratus (Cyprinidae), were determined by means of immunohistochemistry. Sections of whole larvae and goldfish spinal cord were reacted with a polyclonal antibody to rat renal CaBP28K. CaBP28K was located by the PAP technique (Sternberger). It was found in the soma, dendrites, axons and axon terminals of spinal motoneurons but not in those of electromotoneurons of Apteronotus leptorhynchus, whereas it occurred in both motoneurons and electromotoneurons of the larval electric organ of Pollimyrus isidori. In these species CaBP28K was also present in the electromotoneuron axon terminals that make synaptic contacts with the pedicles of the electrocytes. In adult Carassius auratus, CaBP28K was found in the soma, dendrites and axons of certain spinal motoneurons. The results indicate that, in teleosts, the motoneurons containing CaBP28K may represent a well-defined population within the spinal cord; the role of this protein in these cells remains to be determined.

    Notes: Summary Nervous elements supplying the filiform papillae of the tongue of cattle and rats were investigated using immunohistochemistry against neurofilament protein (NFP) and glia-specific S-100 protein. The rod-shaped bovine filiform papillae were heavily keratinized along their entire length and lacked the connective tissue core that occurs in other mammals. Instead, the core was located posterior to the filiform papilla. The base of the bovine filiform papillae was invaded vertically by laminar connective tissue papillae. The core contained a large number of NFP-positive nerve fibers, most of them terminating as free endings in its anterior margin. NFP-positive nerves gathered around the anterior ridge of the epithelium at the base of the core and occasionally penetrated into the epithelium. The laminar connective tissue papillae at the base of the filiform papilla also contained NFP-positive nerve fibers. The core contained S-100-immunoreactive lamellated corpuscles, which were identified as “simple corpuscles” in electron micrographs. The structure and innervation of the bovine filiform papilla suggest that they represent a specialized sensory apparatus. The pyramidal filiform papillae of the rat were smaller, each containing a simple connective tissue core. Few NFP-positive nerve fibers from the nerve plexus entered the core. Filiform papillae are thus less specialized in rats than in cattle.

    Notes: Summary An antiserum against glutamate decarboxylase (GAD) of the rat brain was used to locate GAD activity in sections of the nervous system of the cockroach, Periplaneta americana. The sixth abdominal ganglion was chosen because electrophysiological evidence suggests the presence of GABAergic inhibitory synapses in the cereal-giant interneuron system. Groups of somata and numerous fibres and tracts were positively labelled by the GAD antiserum. A posterior group of labelled somata could be identified close to the entry of the cereal nerves. A line of somata clusters lay along a ventro-lateral furrow. Another discrete row of GAD-like cells was located dorso-laterally. Some small cells among the dorsal unpaired neurons were labelled. A small central group appeared under these cells. An abundance of GAD-like processes and transversal tracts were found within the neuropile. The different systems of GABAergic inhibitors in the ganglion are discussed; in particular we show that the fibres of cereal nerve X are not labelled. This demonstrates that the latter act on the giant fibres via interneurons. We suggest that the group that sends axons into the overlapping region between the cereal nerve and the giant fibre could be the inhibitory interneurons involved in this system.

    Notes: Summary Calcitonin gene-related peptide immunoreactivity was localized immunohistochemically in nerve fibers innervating the biliary pathway and liver of the guinea-pig. Immunoreactive fibers are present in all layers of the gallbladder and biliary tract and are particularly numerous around blood vessels. In the liver, immunoreactive processes are usually restricted to the interlobular space and porta hepatis, and only a few, very thin, beaded processes were observed in the hepatic parenchyma. A rich innervation is also associated with the vena portae. Positive ganglion cell bodies were not visualized within the ganglionated plexus of the biliary system, whereas they were found in the myenteric and submucosal plexus in the cranial portion of the duodenum corresponding to the sphincter of Oddi. The vast majority, if not all, of calcitonin gene-related peptide-immunoreactive fibers contain substance P immunoreactivity; however, there are some substance P-containing fibers lacking calcitonin gene-related peptide immunoreactivity. The lack of co-occurrence of calcitonin gene-related peptide and substance P immunoreactivities in intrinsic ganglion cells suggests that these two peptides are coexpressed in the extrinsic component of the innervation of the hepatobiliary system.

    Notes: Summary It has long been disputed whether mammalian enterochromaffin (EC-) cells contain a peptide in addition to serotonin. Previous immunohistochemical studies have provided evidence for the presence of enkephalins in EC-cells. These findings, however, are equivocal. Therefore, the problem of opioid peptides in EC-cells has been re-examined in the gastro-intestinal mucosa of dog, guinea-pig and man. A battery of antisera against derivatives of pro-opiomelanocortin, pro-enkephalin and pro-dynorphin have been applied to semithin serial sections of the tissues, in combination with fluorescence histochemistry and serotonin immunocytochemistry. Our findings indicate that EC-cells of the investigated species contain pro-dynorphin-related peptides, i.e. dynorphin A and α-neo-endorphin, but no derivatives from pro-opiomelanocortin or pro-enkephalin. Since remarkable interspecies variations occur with respect to the number and staining characteristics of opioid immunoreactive EC-cells, it is concluded that pro-dynorphin shows specific routes of post-translational processing depending upon the species and the gastro-intestinal segment investigated. Future studies should focus on the mutual relationships between serotonin and dynorphins and on the physiological significance of these peptides in the gastrointestinal tract.

    Notes: Summary The topographical localization of substance P (SP) and methionine-enkephalin-octapeptide (Enk-8) was examined immunohistochemically in the surface layer of the dorsal horn of rat cervical spinal cord. Although a few neurons were immunoreactive for Enk-8 in the intact animals, after an intracisternal administration of colchicine, immunoreactive Enk-8 neurons were numerous, and half of them indicated immunoreactivity also for SP. Some immunoreactive SP neurons appeared to show no immunoreactivity for Enk-8. Immuno-reactive nerve fibers, on the other hand, were numerous, and many of them contained both peptides. Electron-microscopic examination of the nerve fibers in tissue prepared by a freeze-drying procedure and stained by a postembedding procedure, revealed the costoring of both peptides in the same cored vesicles. The physiological significance of this costoring is discussed.

    Notes: Summary Immunoperoxidase methods were used to localize secretory component, immunoglobulin A and immunoglobulin G1 in mammary tissue from dairy cows. In lactating tissue, immunostaining for immunoglobulin A and secretory component was observed primarily in the luminal contents of alveoli. By day 2 of involution, alveolar epithelial cells stained for both immunoglobulin A and secretory component. Staining of alveolar epithelial cells for immunoglobulin A and secretory component continued throughout the period of mammary involution. No staining for secretory component was observed in the interalveolar stromal area. Immunoglobulin G1 immunostaining was localized primarily in the interalveolar areas in lactating tissue, but was localized at the apical and basolateral surface of alveolar cells on day 2 of involution. In contrast to immunoglobulin A, immunoglobulin G1 staining of epithelial cells did not persist and was primarily in the interalveolar areas by day 4. These results suggest that an increased localization of immunoglobulin G1 in bovine mammary epithelial cells may occur transiently in early involution, while an increase in immunoglobulin A and secretory component localization in epithelial cells persists throughout involution.

    Notes: Summary Corticotropin releasing factor (CRF)-immunoreactive (IR) perikarya, visualized by the indirect immunoperoxidase method in colchicine-pretreated cats, were localized in many discrete regions of the medulla oblongata. They were found mainly in the dorsal aspect and midline of the medulla oblongata, and more rostrally in the ventrolateral portion. Our results also demonstrated CRF-IR neurons in the rostrocaudal extent of the inferior olive, probably projecting to the cerebellar cortex via thick axons visualized along the lateral edge of the medulla. CRF-IR olivary cells were also found in the pontine cat from which the forebrain was removed, but neither in hypophysectomized nor adrenalectomized cats.

    Notes: Summary Two cytochemical techniques were used at the ultrastructural level to study the distribution of specific axon types to different intrarenal structures in the dog. Using the chromaffin reaction to distinguish catecholaminergic fibres from other axon populations, it was found that the renal cortex of the dog is supplied only by catecholaminergic nerves. Immunostaining for tyrosine hydroxylase (TH) labelled all of the intracortical nerves, and 20% to 25% of these profiles also contained dopa decarboxylase (DDC)-immunoreactivity, indicating they were dopaminergic rather than noradrenergic. Both DDC-positive and DDC-negative axons were seen in close association (∼80 nm) with blood vessels and juxtaglomerular cells as well as tubular epithelial cells. The distribution of TH- and DDC-immunoreactive nerves in the renal cortex is compatible with existing functional evidence indicating that both dopaminergic and noradrenergic nerves are involved in the regulation of renal blood flow, tubular reabsorption and renin release.

    Notes: Summary The ultrastructure of porcine ventricular tissue was studied by electron microscopy and immunocytochemical techniques. Electron-dense specific granules were found in both Purkinje fibers and transitional cells in the ventricular walls, and were positively stained by the immunogold staining method using an antiserum against atrial natriuretic polypeptide (ANP). This suggests that both the Purkinje fibers and transitional cells display the same specific granules as atrial cardiocytes containing ANP. These results demonstrate that Purkinje fibers and two types of transitional cells, in addition to the ordinary ventricular cardiocytes, can be identified in porcine ventricular wall tissue.

    Notes: Summary Immunocytochemical methods were used to investigate the occurrence and distribution of sauvagine, corticotropin-releasing factor-, or urotensin I-like immunoreactivities (SVG-ir, CRF-ir, UI-ir, respectively) in the bullfrog (Rana catesbeiana) brain, using specific antisera raised against non-conjugated SVG, ovine CRF, rat/human CRF, and UI. In the hypothalamus, SVG-ir was found in the magnocellular perikarya, in the dorsal and ventral regions of the preoptic nucleus, and in the hypothalamo-hypophyseal projections to the external zone as well as the internal zone of the median eminence, to pars nervosa, and in fibres running from the pars nervosa to the pars intermedia of the pituitary. In contrast, CRF-ir was found only in parvocellular perikarya, mainly localized in the rostro-ventral part of the preoptic nucleus, with fine processes protruding through the ependyma of the third ventricle, fibre projections terminating in the anterior preoptic area and in the neuropil of the periventricular gray, and a caudal projection to the external zone of the median eminence. No CRF-ir staining was seen in the pars nervosa and pars intermedia. The use of UI-specific antisera failed to give a positive response in the frog brain. It is concluded that, in the frog brain, two anatomically different CRF-like (or SVG-like) systems co-exist, comparable to the reported co-existence of UI-ir and CRF-ir neuronal systems in fish brain.

    Notes: Summary The distribution of FMRFamide-like immunoreactive neurons in the nervous system of the slug Limax maximus was studied using immunohistochemical methods. Approximately one thousand FMRFamide-like immunoreactive cell bodies were found in the central nervous system. Ranging between 15 μm and 200 μm in diameter, they were found in all 11 ganglia of the central nervous system. FMRFamide-like immunoreactive cell bodies were also found at peripheral locations on buccal nerve roots. FMRFamide-like immunoreactive nerve fibres were present in peripheral nerve roots and were distributed extensively throughout the neuropil and cell body regions of the central ganglia. They were also present in the connective tissue of the perineurium, forming an extensive network of varicose fibres. The large number, extensive distribution and great range in size of FMRFamide-like immunoreactive cell bodies and the wide distribution of immunoreactive fibres suggest that FMRFamide-like peptides might serve several different functions in the nervous system of the slug.

    Notes: Summary The distribution of atrial natriuretic polypeptide (ANP) was immunohistochemically surveyed in the rat heart and lung using an antiserum raised against α-human ANP. The ANP-immunoreactive cells were seen to be distributed in the atrial walls and proximal portions of the pulmonary vein and venae cavae, but were absent from the aorta, pulmonary arteries, trachea, bronchus, and alveolar cells. The immunoreactive cells were present in a narrow region just beneath the endothelium of the pulmonary vein and vena cavae, and, ultrastructurally and immunocytochemically, were seen to be striated muscle cells with ANP-containing specific granules similar to those seen in atrial cardiocytes. A radioimmunoassay for ANP revealed a content of 604±51 pg/mg wet weight in the pulmonary vein, and 3343±1620 pg/mg wet weight in the venae cavae. In addition to the atrial wall, the proximal portion of both the pulmonary vein and venae cavae are suggested to be constituents of an ANP-producing organ.

    Notes: Summary Serotonin-like immunoreactivity was investigated in the pineal complex of the golden hamster by use of the indirect immunohistochemical technique. The superficial and deep portions of the pineal gland, and also the pineal stalk exhibited an intense cellular immunoreaction for serotonin. In addition, perivascular serotonin-immunoreactive nerve fibers were observed. Some serotonin-immunoreactive processes of the pinealocytes terminated on the surface of the ventricular lumen in the pineal and suprapineal recesses, indicating a receptive or secretory function of these cells. Several serotonin-immunoreactive processes connected the deep pineal with the habenular area. One week after bilateral removal of both superior cervical ganglia the serotonin immunoreaction of the entire pineal complex was greatly decreased. However, some cells in the pineal complex, of which several exhibited a neuron-like morphology, remained intensively stained after ganglionectomy. This indicates that the indoleamine content of some cells in the pineal complex of the golden hamster is independent of the sympathetic innervation.

    Notes: Summary The distribution of 5-hydroxytryptamine in the gut of several species of birds and reptiles, and of a prototherian mammal, the platypus, was studied using a monoclonal antibody. 5-Hydroxytryptamine-like immunoreactivity was found in enterochromaffin cells and, in birds, in thrombocytes. Immunoreactivity was not found in enteric neurons fixed immediately after dissection. A detailed study was made on one avian species, the budgerigar. Following incubation of intestine in physiological solution, immunore-activity was found in nerve fibres in the gut wall that was more marked after incubation with the monoamine oxidase inhibitor pargyline. These fibres took up exogenous 5-hydroxytryptamine. Similar fibres were found in the intestinal nerves and in perivascular plexuses on mesenteric arteries. Both the uptake of 5-hydroxytryptamine and the appearance of neuronal immunoreactivity after incubation were inhibited by the amine uptake inhibitors desmethylimipramine or fluoxetine. Fibres taking up 5-hydroxytryptamine were damaged by pretreatment with 6-hydroxydopamine. It was concluded that the fibres showing immunoreactivity after incubation were adrenergic fibres that had taken up 5-hydroxytryptamine released in vitro from enterochromaffin cells or thrombocytes. These, and more limited observations made on the other species, suggest that birds, reptiles and prototherian mammals lack enteric neurons that use 5-hydroxytryptamine as a transmitter substance.

    Notes: Summary Ecdysteroid-like material was demonstrated by means of immunhistochemistry in the anterior body region of Lithobius forficatus with the use of an antiserum against an ecdysone-methoxim-BSA-conjugate in conjunction with a modified PAP-method (Sternberger and Joseph 1979). This material is restricted to a tissue formed by podocytes loosely surrounding the salivary glands. Earlier ultrastructural, experimental and biochemical in vitro investigations indicated that this tissue represents the ecdysial glands; this interpretation is now strengthened by immunohistochemical evidence. Reactivity within the cells occurs predominantly in cytosomes.

    Notes: Summary Immunohistochemical localization of cytochrome P-450 in the colonic mucosa of 3-methylcholanthrene-pretreated and untreated rats was studied by indirect fluorescent antibody staining technique. A polyclonal antibody for cytochrome P-450MC purified from hepatic microsomes of 3-methylcholanthrene-pretreated rats was used for this experiment. A strong immunofluorescence was found to be localized in the cytoplasm of the surface epithelium of the mucosa in the colon of 3-methylcholanthrene-pretreated rats. A faint immunofluorescence was also observed in the epithelium of untreated rats. 7-Ethoxycoumarin O-deethylase activity of colonic microsomes was significantly enhanced by 3-methylcholanthrene-pretreatment in parallel with an increase in the intensity of immunostaining for cytochrome P-450MC in Western blotting analysis. This is the first report on the localization of cytochrome P-450 in the colonic mucosa.

    Notes: Summary By use of the indirect immunofluorescence technique, the cellular localization of thyrotropin-releasing hormone (TRH) was studied in the gastrointestinal tract of rats and guinea pigs of different ages. TRH-like immunoreactivity (LI) was observed in many pancreatic islet cells of young rats and guinea pigs but only in single cells of 6-month-old rats. In aged guinea pigs, a reduction in the number of TRH-positive cells was evident; however, numerous strongly fluorescent cells were still present. In the guinea pig, TRH-LI was in addition observed in gastrin cells in the stomach. TRH-positive nerve fibers occurred in the myenteric plexus of the oesophagus, stomach and intestine of the rat, and in the muscle layers of the guinea pig. These results suggest a functional role of TRH both as hormone and neuroactive compound in various portions and sites of the gastro-intestinal tract of the rat and guinea pig

    Notes: Summary Immunohistochemistry with monoclonal and polyclonal antibodies revealed the presence of cytokeratins in epithelial cells of Rathke's cysts in the pars intermedia of the human pituitary gland. With monoclonal antibodies specific for individual cytokeratins, the expression of CK 18, CK 8, CK 7, and CK 19 could be shown in these cells. Within the hypophysis, CK 19 and CK 7 were restricted to Rathke's cysts and a few epithelial cell clusters in the pars tuberalis, whereas other cytokeratins were also present in endocrine cells of the pars distalis. Furthermore, vimentin and, focally, glial fibrillary acidic protein (GFAP) were detected in the cystic epithelia. By double labelling, coexpression of cytokeratin and vimentin, GFAP and cytokeratin, and GFAP and vimentin could be demonstrated. Compiled data of all known cases of coexpression of cytokeratin and vimentin in normal cells reveal physiological correlations and suggest a functional significance of this rare type of coexpression of intermediate filament proteins.

    Notes: Summary The morphology of atrial-specific granules, which contain atrial natriuretic polypeptide (ANP), was studied in the cardiac tissue of untreated controls and water-deprived rats by means of conventional and immunoelectron microscopy. Immature secretory vesicles or granules appeared to become buded off from the Golgi cisternae and then fused to form specific A-granules. An electron-dense plate with a fuzzy coat was frequently found on the limiting membrane at the end of such fusion. Pale specific B-granules, which were less electron-dense, larger, and more granular than A-granules, were found in small numbers in the left atrial cardiocytes, but rarely in the right ones. Very pale granules with a less granular matrix, considered to be B-type granules which had lost their electron-density, and which had less immunoreactivity for ANP, were numerous in the cardiac tissue after water deprivation. This morphological change, which is interpreted as an indication of granule degradation, was in agreement with the noted increase of natriuretic activity in the atrial tissue of water-deprived specimens.

    Notes: Summary Nerves in the uterine cervix of the rat were examined with regard to co-existence of markers for noradrenaline and neuropeptide Y, and differential tissue innervation by nerves containing different combinations of these markers. Immunohistochemical labeling of single and adjacent serial cryostat sections, and double labeling was employed. Some animals were treated with the noradrenergic neurotoxin, 6-hydroxydopamine. In control animals neuropeptide Y-immunoreactive fibers were numerous in the myometrium and around arteries; noradrenergic fibers were few in the myometrium and moderate in number around arteries. Myometrial neuropeptide Y-immunoreactive fibers were not decreased, but apparently increased, in 6-hydroxydopamine-treated rats; in contrast, perivascular neuropeptide Y-immunoreactive fibers were markedly reduced, but not totally absent. Noradrenergic fibers were absent in the myometrium and around arteries following 6-hydroxydopamine treatment. Labeling of adjacent sections and double labeling revealed coincident labeling of markers for neuropeptide Y and noradrenaline in perivascular, but not myometrial, nerves. We concluded that most myometrial neuropeptide Y-immunoreactive nerves did not contain noradrenaline since they were not sensitive to 6-hydroxydopamine and did not stain doubly; however, perivascular neuropeptide Y-immunoreactive fibers which degenerated after 6-hydroxydopamine treatment and did label doubly must co-store noradrenaline. Some neuropeptide Y-immunoreactive perivascular fibers may contain neuropeptide Y but not noradrenaline. Thus, it appears there is a differential innervation of tissues in the cervix by neuropeptide Y/noradrenergic nerves; this could reflect a differential regulation of tissues innervated by these nerves.

    Notes: Summary The presence, distribution and development of vasoactive intestinal polypeptide (VIP)-like immunoreactivity in the gastro-entero-pancreatic system of a cartilaginous fish Scyliorhinus stellaris (L.) was investigated by immunohistochemical methods utilizing mammalian VIP antisera. In the gut VIP-like immunoreactivity was observed in both nerves and endocrine cells. Endocrine cells with VIP-like material were only detected in the intestinal epithelium while nerve fibres containing VIP-like material were noted along the whole gastro-entero-pancreatic system, being more numerous in the pyloric sphincter and in the intestinal portion. Immunoreactive nerve cell bodies were encountered in the stomach and intestinal portions localized in the submucosa and in the myenteric plexus. Intestinal immunoreactive endocrine cells were already present in the first developmental stage considered (embryos aged 4 months). They grow in number and before birth reach a frequency higher than in adults. Nerves and cell bodies showing VIP-like immunoreactivity, appear later, before birth, as a few elements in the smooth muscular layer, but only after birth their distribution and frequency are similar to those found in adults. The faint immunofluorescence shown by the immunoreactive endocrine cells and their developmental pattern, which is always different from that observed in nervous elements, suggest the presence of at least two VIP-like substances in the gastro-entero-pancreatic system of S. stellaris.

    Notes: Summary Immunohistochemical techniques were used to study the distribution of gamma-amino butyric acid (GABA)-like immunoreactive neurons in the nervous system of the slug Limax maximus. Approximately 170 GABA-like immunoreactive cell bodies were found in the central nervous system. These were located in the cerebral, buccal and pedal ganglia. Most GABA-like immunoreactive neurons had small cell bodies, which were aggregated into discrete clusters within the cerebral and pedal ganglia. Three pairs of longer, uniquely identifiable, GABA-like immunoreactive cells were found in the cerebral ganglion. GABA-like immunoreactive nerve fibres were also found in all of the central ganglia but were absent from peripheral nerves. These results suggest that GABA acts as a central neurotransmitter in the slug. The possible roles of GABA-ergic neurotransmission in the slug are discussed.

    Notes: Summary The purpose of this study was to examine the development and distribution of neuropeptide Y-immunoreactive fibers in the neurohypophysis of the rhesus monkey (Macaca mulatta) throughout life and the relationship of these fibers to the hypothalamo-hypophyseal portal vasculature. In rhesus monkeys, which varied in age from fetal life to 34 years, neuropeptide Y-immunoreactive fibers were present at all ages examined. In adult monkeys, varicose neuropeptide Y-labeled fibers were concentrated in the upper infundibular stem in association with capillary loops of the portal vasculature and the long portal vessels. Other fibers travelled down the infundibular stem and were distributed at the junction of the lower infundibular stem and infundibular process in the vicinity of the short portal vessels. In the infundibular process, neuropeptide Y-immunoreactive fibers were concentrated along the border of the intermediate lobe. Other stained fibers were sparsely distributed in the infundibular process and were often associated with small vessels. Neuropeptide Y-immunoreactivity was also located in a few fibers and cells of the intermediate lobe. Very few labeled fibers were seen in the fetal neurohypophysis, but their number increased gradually during the first postnatal year. At two years of age, a high density of stained fibers was observed, especially in the infundibular process. The number of axons in the infundibular process was lower at 12 years and continued to decline until 34 years of age. Neuropeptide Y may modulate hormone release at these sites and may also be released directly into vessels in the infundibular process. The close association of neuropeptide Y-labeled fibers with capillaries of the portal vasculature strongly suggests that neuropeptide Y is released into the portal blood of monkeys throughout life and may influence hormone secretion from the anterior pituitary gland.

    Notes: Summary In Drosophila β tubulins are encoded by a small gene family and the four members of this family are differentially expressed. mRNAs transcribed from two of these genes, namely the β1 and β3 tubulin genes, are abundant during embryogenesis. While the β1 tubulin gene is constitutively expressed during development, β3 mRNA is restricted to two distinct phases: mid embryogenesis and metamorphosis. The transcription initiation sites are identical in both these stages and comparison of presumptive promoter regions reveals no extensive homologies between the genes. In situ localization shows β1 tubulin mRNA to be maternally expressed in the nurse cells of the egg chambers and evenly distributed during early embryogenesis. In contrast, during later stages of embryogenesis β1 tubulin transcripts are predominantly expressed in neural derivatives. The β3 tubulin gene expression is also spatially regulated, β3 mRNA being restricted to the mesoderm.

    Notes: Summary Mutations at the cin gene display drastically lowered levels of the molybdoenzymes, xanthine dehydrogenase (XDH) and aldehyde oxidase (AO), and lack pyridoxal oxidase (PO) and sulfite oxidase (SO) activities. Certain mutations at cin also display varying degrees of female sterility, which is maternally affected. Here we characterize five new cin alleles with respect to the molybdoenzyme activities as well as the molybdenum cofactor, commonly required for molybdoenzyme activity. In complementing cin heterozygotes we find that, in addition to the previously reported unusually high levels of XDH and AO activities, there are unusually elevated levels of SO activity, as well as complementation for PO activity. The levels of immunologically crossreacting material in such heterozygotes indicate that the elevated levels of molybdoenzyme activities cannot be due to increases in the number of enzyme molecules. Measurements of the level of molybdenum cofactor activity normally present in XDH, AO, PO, and SO point to the possibility that a larger fraction of the enzyme molecules are active in these heterozygotes. The possible role of SO with respect to cinnamon's female sterility is also discussed.

    Notes: Summary A series of P-element insertion mutations at one site in the vestigial (vg) locus was tested for cytotype dependent effects on vg expression. The mutant phenotypes for four P-element vg alleles were suppressed when the alleles were stabilized in the P-cytotype. The suppression was observed whenever repressor-producing P-elements were present in the genome. Genetic and molecular analysis indicated that the suppression is not due to excision or other irreversible alterations of the inserts.     Notes: Summary Six kinds of autonomously replicating sequences (ARSs) derived from Drosophila or tobacco were inserted into the vector pDSV, constructed with pSV2-gpt and the copia long terminal repeat (LTR). The resulting ARS-containing plasmids, pDSV-ARSs, were transfected into the cultured Drosophila cells of GM1 S1cl1. Most of the plasmids remained for about 2 weeks and some for about 1 month in these cells. The retention time of the plasmid was not directly correlated with autonomously replicating activity of ARSs detected in the yeast. Two plasmids, one carrying ARS of Drosophila nuclear DNA and the other carrying tobacco DNA, showed the longest retention time in transformed cells and replication was confirmed in these cells. Some of these long lived plasmids were recovered, however, as modified forms. Other plasmids had disappeared 1 month after transfection. Two months following transfection, none of plasmids were recovered but they were detected in nuclear DNA as the integrated form. The integration patterns in all the cells transformed by different kinds of ARS-containing plasmids were similar to each other, and to the distribution pattern of copia LTR in the genome. These results suggest that copia LTR sequences contained in the pDSV-ARSs may participate in the integration process of these plasmids into Drosophila DNA.

    Notes: Summary The technique of chromosome walking was used to isolate approximately 60 kb of DNA from the region containing the complementation group uncoordinated of Drosophila melanogaster, located in that part of the X chromosome which spans the euchromatin-heterochromatin junction. The cloned DNA can be divided into two distinct regions. The first contains sequences that are low copy number or unique and are largely conserved between strains. The second region is characterized by units repeated in tandem arrays and is polymorphic within, and between, strains. Each repetitive unit is separated by a member of an abundant sequence family, part of which is homologous to the ribosomal type 1 insertion sequence of D. melanogaster. The molecular organization of the cloned DNA was compared with that of sequences isolated from regions of intercalary heterochromatin and also with genes which have been characterized from more conventional euchromatic regions.

    Notes: Abstract An estuary such as the Westerschelde is a highly dynamic environment, both on an ecological time scale where climatic and hydrodynamic forces, mainly the tides, shape a very variable environment and on a geological, evolutionary time scale, since estuaries are young and very unstable habitats. Low species diversity and high adaptability of the resident animal and plant populations are characteristic of estuarine habitats where large fluctuations in submersion, salinity, temperature etc. occur. The existing biota are therefore resilient to environmental stress and effects of the important influx of anorganic and organic pollutants from the river Schelde and its tributaries on the biota in the estuary are not easy to detect. Although water movement and sedimentation patterns in the Westerschelde are relatively well known, there exists little information on important ecological processes such as primary production and heterotrophic metabolism in the estuary.

    Notes: Abstract Barley grains (9 samples from 7 cultivars) with nitrogen contents (N) ranging from 1.45 to 4.01% of dry matter were analysed for their amino acid (AA) composition with high accuracy from six different hydrolysates per sample. AA levels in grain increased as linear functions ofN with correlation coefficients close to unity. A comparison with literature data confirmed that the AA composition of any grain sample of normal barley can be predicted from itsN for all phenotypes and genotypes. AAs in grain protein changed as hyperbolic functions ofN which increased for Phe, Pro and Glx but more or less strongly decreased for the other AAs. By plotting AA scores againstN, barley proteins were shown to be always richer than wheat and rye in Val and Phe + Tyr; sometimes richer than both other species forN〈2 (Lys); 2.2 (Leu and Ile); 3.4 (Thr); sometimes intermediate to wheat and rye above the latterN values. They were also intermediate in sulphur AAs forN〈1.9 and drastically poorer forN〉1.9. However, they were richer than both other species in Trp forN〉1.6. The hyperbolic variations of non-protein nitrogen and nitrogen-to-protein conversion factors were determined as a function ofN and also compared with those of wheat and rye.

    Notes: Summary The composition of mitochondrial DNA (mtDNA) was analyzed in single female flies that developed from fertilized Drosophila melanogaster eggs, into which germ plasm of D. simulans had been introduced. HpaII cleavage patterns showed that all 12 individual female flies examined had developed from eggs in which 37%–71% of the total mtDNA was D. simulans mtDNA (Ds mtDNA) and the rest was D. melanogaster mtDNA (Dm mtDNA). The stability of this heteroplasmic state in these isofemale lines was monitored for seven generations at both individual and population levels. Results showed that the heteroplasmy of Dm and Ds mtDNAs was stably transmitted for at least three generations at the population level, but showed stochastic segregation at the individual level. After 4–6 generations, all individuals lost Ds mtDNA. The mechanisms of preferential loss of Ds mtDNA and of transmission of heteroplasmic mtDNA to descendants are discussed.

    Notes: Summary Minute loci represent a class of about 50 different Drosophila genes that appear to be functionally related. These genes may code for components of the protein synthetic apparatus. While one Minute locus has been recently shown to code for a ribosomal protein, it is not yet known whether any of the other Minute loci also code for ribosomal proteins. We have addressed this question by a combined molecular and genetic approach. In this report, a cloned DNA encoding the ribosomal protein rp21 is partially characterized. The rp21 gene maps to the same region (region 80 of chromosome 3L) as the temperature-sensitive Minute QIII gene. Using P-element mediated transformation, the rp21 gene was transformed into the germline of Drosophila. RNA blot experiments revealed that the transformed gene is expressed in transgenic flies. However, genetic complementation analysis indicated that the QIII locus and the rp21 gene are not identical. Implications of these findings for the relationship between Minutes and ribosomal protein genes are discussed.

    Notes: Abstract Sesbania sesban was evaluated as green manure crop for lowland rice in the Dry Zone of Sri Lanka. The legume was grown during a fallow period before lowland rice (Oryza sativa) and ploughed under just before transplanting. Weight loss and nitrogen content in litterbags containing leaves, stems and roots of the legume were monitored. Comparisons were made between rice yields from 20 m2 plots after green manuring in combination with different nitrogen fertilizer levels (0, 2.4, 4.8 and 7.2 gm−2) and nitrogen fertilizer (9.6 gm−2) alone. Above-ground biomass ofS. sesban was 440 gm−2 (dry wt) when ploughed under after 84 days growth. N-content in leaves, stems and roots was 3.76%, 0.41% and 0.73%, respectively. This gave a N-input fromS. sesban of 9.2 gm−2 (8.3 g from above-ground parts and 0.9 g from roots). The corresponding K and P inputs were 7.3 and 0.6 gm−2 respectively. The nitrogen rich leaves, which contained 88% of the nitrogen in the above-ground parts, decomposed and released its nitrogen much more rapidly than the stems and roots. After only four days the leaves had released 5.3 g Nm−2 and after 14 days they had released 6.4 g Nm−2. The highest rice yield (505 gm−2) was obtained usingS. sesban and 4.8 gm−2 of N-fertilizer. The yields with only N-fertilizer or onlyS. sesban were 442 gm−2 and 396 gm−2, respectively. Due to the rapid decomposition of the nitrogen rich leaves,S. sesban did not behave as a slow release fertilizer. Thus, it is not necessary to apply nitrogen fertilizers as a basal dose.

    Notes: Abstract The mineralization of C, N, and S from residues of three different crop species (wheat, lentil, and rape) grown under diverse nutritional regimes was measured over a 12-week incubation period under controlled conditions. The rate of decomposition, as measured by CO2 evolution, varied considerably among treatments and appeared to be controlled almost entirely by N content of the residue (R2=0.98). Similarly, N mineralization was strongly tied to N concentration. The critical N concentration, below which significant immobilization of N occurred, declined over time, ranging from 1.9% at day 14 to 1.1% at day 84. Mineralization of S was positively correlated with initial S concentration (R2=0.95) and negatively related to N concentration, apparently because of a dilution effect. The results demonstrate that decomposition and N and S mineralization of crop residues, under conditions prevalent in the experiment, are primarily a function of their nutrient concentrations rather than biochemial composition related to crop species. As a result, it should be possible to enhance rate of residue decomposition, increase quantities of N and S mineralized, and avert detrimental immobilization losses in the following year by governing the nutritional regime under which the crop is grown.

    Notes: Abstract The effect of the initial lignin and nitrogen contents of the leaves of five tree species, on their decomposition dynamics, was studied in the sub-tropical forest ecosystem existing at Shiroy hills. The decomposition dynamics of litter materials are described by inverse linear relationships between the percentage of the original biomass remaining and the nitrogen concentration in the residual material. Initial lignin and nitrogen were highly correlated with remaining biomass (r=0.94 and 0.77). The rate constants (K), for the annual leaf mass loss, ranged from −0.18 to −0.56. The values for initial lignin (%), initial nitrogen (%), the ratio between initial lignin, initial nitrogen and the annual decomposition rate constants were compared with similar results from other studies in various climatic zones of the world.

    Notes: Abstract Anin situ resin bag technique was used to measure the relative availabilities of N and P along a chronosequence of soils in southern New Mexico, and was compared to two more common indices of nutrient availability. Accumulations of N and P during 10-week intervals over an 18 month period were separable into wet season (September–January) and dry season (February–August) groups, with wet season values significantly greater than dry season values. Only accumulations during the wet season showed significant differences among sites, thus stressing the role of field water regime in interpreting resin accumulation results. Total mineral N (NO3+NH4) sorbed by resins was significantly correlated to laboratory N mineralization rates. Although accumulation patterns of N and P were similar to patterns of %N and %P in shrub species growing along the chronosequence, these similarities were not statistically significant. A laboratory experiment demonstrated that bicarbonate-form anion resins are preferable to hydroxyl-form resins, as long as standards are made from solutions extracted by resins to account for variable ion recovery efficiencies.

    Notes: Abstract A calcareous soil was treated with an organic fertilizer mixture of grape debris and peat, or with pig manure, chicken manure, city refuse or with aerobic or anaerobic sewage sludges. Changes in different fractions of carbon and nitrogen were followed by analysis of the soil, immediately after these additions, after they had decomposed for six months in the soil, after a maize harvest, and after a subsequent barley harvest. The various forms of carbon had decreased after six months. The rate of decrease varied with the nature of the organic materials. In all samples, the fulvic acids fraction decreased most. The main transformations happened during the first six months whether there was a crop growing or not, but the crops had no influence on the breakdown of the organic materials. The ratio of humic acids/fulvic acids had increased after six months of humification and, in general terms, the final values of the ratio oxidizable carbon/extractable carbon were lower than the initial ones, indicating a higher degree of humification in the organic matter by the end of the experiment. The total nitrogen level remained practically constant after the first six months of humification even after the second crop. Part of the organic nitrogen of the soil samples amended with chicken manure, city refuse or the two sludges was transformed into mineral nitrogen during the humification process. The extractable nitrogen values increased because of the fertilizer applied during the pot trials.

    Notes: Abstract Nodules were collected from 14 legume species from the Indonesian Islands of South Sulawesi, Java and Sumatra. Their rhizobia were isolated and growth characteristics, nodulation ability and nitrogen fixing effectiveness were assessed against recommended commercially available Australian strains. The test legumes wereMacroptilium atropurpureum Urb. cv. Siratro,Vigna unguiculata (L.) Walp. cv Eureka,Centrosema pubescens Benth cv. Belalto andDesmodium heterocarpon (L) DC. A significant portion of the native rhizobial isolates were of the fast growing type. Dry matter and total nitrogen production forM. atropurpureum andV. unguiculata was highest when inoculated with native strains while the commerical strains produced superior dry matter production forC. pubescens andD. heterocarpon. However the total nitrogen production of native and commercial strains was not significantly different for the latter two legumes. The study indicated that a potential exists for developing inocula from local Rhizobium strains.

    Notes: Abstract Lucerne plants inoculated with vesicular-arbuscular mycorrhizal fungi were grown in pots in a sandy soil amended or not amended with cellulose. Whatever the endophyte or cellulose sources used, the rate of VAM colonization was lower in amended soil. The inhibition of VAM infection increased with the cellulose concentration in the soil. Sequential harvest experiments showed a clear reduction of root colonization from the early stages of plant growth. The effectiveness of cellulose in reducing VAM colonization was influenced by the addition of N fertilizer.

    Notes: Abstract In two experiments, wheat plants growing in solutions of different nitrogen concentration were subjected to root pruning. In higher concentrations of nitrogen the growth rate was higher, and the proportional allocation of growth to shoot higher, but pruning did not affect the allocation of growth at either level of nitrogen. This result gives no support to Thornley's source-sink model of the control of shoot: root ratio.

Seventy five percent of the biomass, 70 percent carbon, 50 percent nitrogen and 30 percent phosphorus remained after 139 days of decomposition. Rice straw decomposition furnished 33% N and 8% P of the total nitrogen and phosphorus provided by man.

    Notes: Abstract Present methods for determining critical nutrient concentrations in plants and soils are unsatisfactory if concentrations change with time while the crop is growing. In such cases the critical concentration can only be applied in relation to growth rate at any given moment. For interpreting field experiments this introduces considerable difficulties: two possible approaches to these problems are suggested, one of which uses a simple simulation model. Results from a Brussels sprout nitrogen experiment are used to show how, using this approach, a single critical sap nitrate concentration (380 mg NO3 N.l−1 for 95% of potential growth rate) may serve to explain the results at all growth stages in three seasons.

    Notes: Abstract Microcosms were used to study the effects of two inorganic nitrogen sources (ammonia and nitrate) and two organic nitrogen sources (urea and glutamic acid) on the growth of algae and bacteria found in the Great Salt Lake, Utah. Ammonia, nitrate and urea stimulated bacterial growth indirectly through increased algal production of unknown organic substances. Glutamic acid, representing readily available organic carbon and nitrogen, stimulated the bacteria directly. No nitrification was observed in the microcosms although nitrite was found when the microcosms were supplemented with nitrate. Lake sediment contained a number of anaerobic bacteria producing hydrogen sulfide, methane and other gases. Production of these gases was stimulated in the columns with high algal and bacterial activity.

    Notes: Abstract Seasonal variation through one year in total nitrogen (TN), total phosphorus (TP), phytoplankton biomass, phytoplankton species composition and other environmental factors were examined in Lake Sonachi, a tropical meromictic soda lake. Mean concentrations of TN and TP were 11 000 µg N l-1 and 100 µg P l-1, respectively. Maximum concentrations of TN and TP occurred in the monimolimnion. Phytoplankton biomass ranged from 350 to 1260 mg m-3. Synechococcus bacillaris, a small coccoid cyanophyte, dominated the phytoplankton. The mean chlorophyll a concentration of 37 mg · m-3 was a modest value when compared with those of other tropical soda lakes. High TN:TP ratios indicated phosphorus limitation in the lake.

    Notes: Abstract The deep lakes in central Alberta are characterized by relatively high total nitrogen (TN) to total phosphorus (TP) ratios (33 to 64, by weight) and high TP (10 to 100 µg 1−1) in the euphotic zone in summer. These same lakes show indications of nitrogen limitation: (1) blue-green algae are relatively important in summer (7 to 88% of total biomass) and a large proportion of the filaments contain heterocysts and (2) inorganic nitrogen levels are extremely low in the surface waters (NO inf2 sup− + NO inf3 sup− + NH inf4 sup+ − N 〈 10 µg 1−1). In 1985 and 1986 inorganic nitrogen (25 to 1200 µg 1−1 − N) and/or phosphorus (12 to 100 µg 1−1 − P) was added to water in 31 m3 limnocorrals or 21 bottles from four deep prairie lakes. In most cases, phytoplankton biomass (estimated as chlorophyll a) was stimulated only by phosphorus additions. However, for a short period (8 d) after an unusual storm, phytoplankton biomass was stimulated by nitrogen additions. After this storm, TP increased by up to 75% in the epilimnion of the study lakes whereas TN did not increase. The algae which increased in these experiments tended to be the dominant algae in the control vessels, which varied with the experiment and the lake. Thus, a large proportion of blue-green algae and low inorganic nitrogen levels are not indicative per se of nitrogen limitation in lakes.
