    Description / Table of Contents: Zusammenfassung Der Umsatz der Lipide vonSaccharomyces carlsbergensis ATCC 9080 wurde nach Vormarkierung mit3H-Ölsäure und14C-Palmitinsäure untersucht. Inositversorgte Zellen zeigen eine Verschiebung der Fettsäuren von den Triacylglycerinen in die Phospholipide, im besonderen in Phosphatidylcholin und Phosphatidylinosit. Eine verstärkte Übertragung der Fettsäuren von Triacylglycerinen auf Phospholipide konnte festgestellt werden, wenn vormarkierte Zellen auf ein Nährmedium, welches Cerulenin enthielt, übertragen wurde. Cerulenin inhibiert die Fettsäuresynthese und ruft in wachsenden Zellen Fettsäuremangel hervor. Inositdefiziente Hefezellen, welche einen erhöhten Triacylglycerinspiegel aufweisen, verwenden diese Triacylglycerine unter Fettsäuremangelbedingungen ebenfalls für die Synthese von Phospholipiden, besonders von Phosphatidylcholin, Phosphatidyläthanolamin und Phosphatidylserin. Da aus früheren Arbeiten bekannt ist, daß inSaccharomyces carlsbergensis praktisch keine β-Oxidation existiert, können die Triacylglyerine in diesem Hefestamm als Speicher für Fettsäuren angesehen werden, welche zur Synthese von Phospholipiden dienen.
    Notes: Abstract The turnover of lipids was studied in the yeast,Saccharomyces carlsbergensis ATCC 9080, after prelabeling of the cells with [3H] oleic acid and [14C] palmitic acid. In inositol supplemented cells, a redistribution of fatty acids from triacylglycerols to phospholipids (mainly phosphatidylcholine and phosphatidylinositol) could be demonstrated. An increased transfer of fatty acids from triacylglycerols to phospholipids was observed when prelabeled cells were transferred to a growth medium containing cerulenin, which inhibits fatty acid synthesis and thus induces fatty acid deficiency in the growing cells. Inositol deficient cells contain increased levels of triacylglycerols, which are equally well utilized for phospholipid (mainly phosphatidylcholine, phosphatidylethanolamine and phosphatidylserine) synthesis under conditions of fatty acid deficiency. The present results together with the previous finding that β-oxidation is practically absent inSaccharomyces carlsbergensis suggest that in this yeast triacylglycerols function as storage of fatty acids which can be mobilized for phospholipid biosynthesis.

    Description / Table of Contents: Résumé La cinétique de la formation et de la transformation des précipités de phosphate de calcium, obtenus en mélangeant de volumes égaux de solutions à 6×10−3 M de calcium total et/ou phosphate total est étudiée à 25°C. Les solutions de phosphate sont préajustées à un pH de 7.4. Les changements de pH et de turbidité des solutions sont suivis simultanément en fonction du temps. Les précipités sont isolés à des intervalles de temps variables et caractérisés par diverses méthodes physico-chimiques. Initialement un précipité avec un rapport molaire Ca/P de 1.5, amorphe aux rayons X et en diffraction électronique, est formé. Le spectre IR indique la présence de PO 4 3− et de HPO 4 2− . Après une période métastable, on observe la précipitation d'un matériel cristallin dans ou sur la phase amorphe. Vingt quatre heures après préparation de l'échantillon les précipités présentent surtout les caractères du phosphate octocalcique.
    Abstract: Zusammenfassung Die Kinetik der Bildung und Transformation von Calciumphosphat-Niederschlägen wurde bei 25°C untersucht. Es wurden dazu gleiche Volumen von Lösungen gemischt, bei einer Konzentration von 6×10−3M totales Calcium und/oder totales Phosphat. Die Phosphatlösungen wurden zuerst auf pH 7,4 eingestellt. Veränderungen des pH und Trübung der Lösungen wurden gleichzeitig als eine Funktion der Zeit aufgezeichnet. Niederschläge wurden in verschiedenen Zeitintervallen isoliert und mit verschiedenen physiko-chemischen Methoden charakterisiert. Am Anfang wurde ein Niederschlag mit einem molaren Ca/P-Verhältnis von 1,5, im Röntgenbild und in der Elektronendiffraktion amorph, gebildet. Infrarotspektren deuteten die Anwesenheit von PO 4 3− - und HPO 4 2− -Ionen an. Nach einer metastabilen Periode erfolgte ein Niederschlag aus kristallinem Material innerhalb oder auf der amorphen Substanz. 24 Std nach der Herstellung der Proben zeigten die Niederschläge in der Hauptsache die Charakteristiken von Octocalciumphosphat.
    Notes: Abstract The kinetics of the formation and transformation of calcium phosphate precipitates obtained by mixing equal volumes of solutions, 6×10−3 M in total calcium and/or total phosphate was investigated at 25°. The phosphate solutions were preadjusted to pH 7.4. Changes of the pH and turbidity of the solutions were followed simultaneously as a function of time. Precipitates were isolated at various time intervals and characterized by different physicochemical methods. Initially a precipitate with a molar Ca/P ratio of 1.5, amorphous to X-ray and electron diffraction was formed. IR spectra indicated the presence of PO 4 3− and HPO 4 2− ions. After a period of metastability, precipitation of a crystalline material within or upon the amorphous matter occurred. Twenty four hours after sample preparation the precipitates showed mainly the characteristics of octacalcium phosphate.

    Description / Table of Contents: Résumé Les surfaces d'émail dentaire sont confrontées avec des solutions de calcium et phosphate, qui y recirculent. Les réactions cinétiques au niveau des surfaces en calcium, phosphate, fluorure et ions hydrogène sont déterminées dans des solutions à concentrations en ions fluor constantes (0.012, 0.025, 0.050, 0.250 ou 0.500 mM/l) et/ou à pH constant (6.8, 7.0, 7.2 ou 7.4). Les vitesses de réactions des ions calcium, phosphate, hydrogène et fluorures augmentent dans les conditions suivantes: 1) augmentation de la surface d'émail, 2) augmentation des vitesses d'écoulement au niveau des surfaces, 3) augmentation du pH de la solution et 4) augmentation de la concentration en ion fluorure. Les vitesses presque proportionnelles des réactions mesurées simultanément indiquent la formation de minéraux apatitiques à la surface de l'émail pendant tout le temps de contact solide-solution.
    Abstract: Zusammenfassung Oberflächen von Zahnschmelz wurden mit konstant zirkulierenden Lösungen, welche Calcium und Phosphate enthielten, equilibriert. Die kinetischen Oberflächenreaktionen von Calcium-, Phosphat-, Fluori- und Wasserstoff-Ionen wurden in Lösungen mit konstanten Fluoridkonzentrationen (0,012, 0,025, 0,050, 0,250 oder 0,500 mM/l) und/oder konstantem pH (6,8, 7,0, 7,2 oder 7,4) bestimmt. Die Reaktionsgeschwindigkeiten dieser Ionen an der Grenze zwischen fester und flüssiger Phase erhöhten sich unter folgenden Bedingungen: 1. Vergrößerung der Schmelzoberfläche; 2. Erhöhung der Geschwindigkeit der der Oberfläche zugeführten Lösungen; 3. Erhöhung des pH der Lösung; 4. Erhöhung der Fluoridionenkonzentration in der Lösung. Die beinahe proportionalen Geschwindigkeiten der gleichzeitig gemessenen Reaktionen deuteten darauf hin, daß auf den Schmelzoberflächen während der ganzen Equilibrierungszeit Apatitminerale gebildet wurden.
    Notes: Abstract Surfaces of tooth enamel were interfaced with recirculating solutions containing calcium and phosphate. The kinetic interfacial reactions of calcium, phosphate, fluoride, and hydrogen ions were determined in solutions of constant fluoride ion concentrations (0.012, 0.025, 0.050, 0.250, or 0.500 mM/l) and /or constant pH (6.8, 7.0, 7.2, or 7.4). The rates of the calcium, phosphate, fluoride, and hydrogen ion reactions at the solid-solution interface increased under the following conditions: (1) increase of the enamel surface area; (2) increase of the rates of solution flow to the interface; (3) increase of solution pH; and (4) increase of the solution fluoride ion concentration. The nearly proportional rates of the concurrently measured reactions indicated the formation of apatitic minerals on the enamel surfaces throughout the time of solid-solution interface.

    Notes: Summary Results from in vitro works suggest that 1,25- and 24,25-dihydroxyvitamin D3 (1,25-(OH)2D3 and 24,25-(OH)2D3) act on bone via different mechanisms. The present investigation was performed to study the effect of these two metabolites and of their precursor 25-hyxdroxyvitamin D3 (25-(OH)D3) on bone cAMP content in vitro. Rats' paired half calvaria were incubated under sterile conditions with one vitamin D3 derivative (10−13 to 10−9 M) or with ethanol (0.005 ml for 15 min to 24 h in 1 ml medium containing 0, 0.2, 1, 2, or 3 mM calcium. In some experiments: (a) cycloheximide (10−5M) was added simultaneously with the vitamin D3 metabolites; (b) 1–84 bPTH (5 × 10−8 M) was added for 5 or 15 min at the end of the 24 h incubation. Calvaria were immersed in 1 ml TCA 5% 4°C and homogenized. The cAMP was extracted with diethylether and measured by a competitive protein binding assay. Results bring further evidence for a particular effect of low doses of 24,25-(OH)2D3 (10−9 to 10−12M) and of 25-(OH)D3 (10−9 to 10−11M) on bone, different from that of 1,25-(OH)2D3: cAMP content was higher in 24,25-(OH)2D3- or 25-(OH)D3-treated and lower in 1,25-(OH)2D3-treated calvaria than in ethanol-treated ones with 1 mM calcium. The 1,25-(OH)2D3 effect persisted in calcium-free medium whereas 25-(OH)D3 and 24,25-(OH)2D3 effects could not be observed with 0 mM nor with 3 mM calcium. The required duration of the preincubation (over 1 h) as well as the inhibitory action of cycloheximide may suggest an involvement of protein synthesis in the vitamin D3 metabolites effects. Neither 1,25-(OH)2D3 nor 24,25-(OH)2D3 affected the PTH-induced increase in bone cAMP content.

    Notes: Summary In a well-defined in vitro perifusion system, the effects of extracellular magnesium concentration (Mg) on parathyroid hormone (PTH) secretion by bovine parathyroid tissue were examined. At Mg less than 0.8 mM, the ability of the glands to secrete hormone maximally in response to low calcium (Ca) stimulation was progressively impaired. Low Mg also impaired the ability of isoproterenol, dibutyryl cyclic AMP and theophylline to stimulate hormone release. The defect in hormone release at low Mg observed in vitro was analogous to the well-documented inhibition of secretion observed in vivo. Increases in Mg from 0 to 0.8 mM rapidly repaired the defect in hormone secretion. At Mg above 1.0 mM there was a Ca-like effect on hormone release, with a progressive decrease in secretion at increased Mg. Although its mechanism is not yet clear, the low Mg effect appears to impair principally the process of hormone release rather than its biosynthesis or storage.

    Notes: Summary Recombinational analysis of oxil mutants was performed using a and a mutant strains with the same mitochondrial and nuclear backgrounds, derived from strain 777-3A. In spite of minor inconsistencies the overall map of oxi1 mutations can be constructed on the basis of wild-type recombinant frequencies in the two-point oxi1 − − x oxi1 − crosses. The frequencies of wild-type recombinants varied in a wide range from 0.003% to 16%, reaching the maximal values expected for unlinked mitochondria) markers. No distinct clusters of mutants were observed. The analysis of translation products of oxil mutants showed that all but one of the oxil mutants studied are connected with the conspicuous changes of the polypeptide band corresponding to subunit 11 of cytochrome c oxidase in electrophoresis on polyacrylamide gels. The exceptional G565 mutant showed no conspicuous change in subunit II, but lacked subunit I of cytochrome c oxidase. Various oxi1 mutants seemed to carry premature chain termination mutations. Most of them show a correlation between the length of the putative fragment of subunit II synthesized and the position on the genetic map. The direction of translation is from the V2 to the V60 mutation. The V2 mutation is proximal to cap and V60 proximal to the par locus.

    Notes: Summary Axenomycin inhibits protein synthesis in vivo and in vitro in Saccharomyces cerevisiae. The antibiotic acts by binding to ribosomes, most probably to the large ribosomal subunit. Mutant strains resistant to axenomycin appear to contain ribosomes that are not inhibited by the antibiotic. The responsible gene has been mapped on the VII chromosome between the centromere and the leu1 gene.

    Notes: Summary We have developed a system for assaying pyrimidine dimers in the 2 ⇐m DNA plasmid of Saccharomyces cerevisiae, using Micrococcus luteus UV endonuclease to nick dimer-containing plasmid molecules and measuring percentages of nicked and covalently closed circles on agarose gels. UV-irradiation induced dimers in plasmid DNA, in vivo, at the same rate as in chromosomal DNA. After a dose of 20 Joules·m−2, approximately 86% of plasmid molecules had. at least one dimer. After 3 h incubation under normal growth conditions only 4% still retained dimers in a wild-type strain. In a rad1 (excision-defective) mutant 81% of plasmid molecules still had dimers after 3 h, suggesting that excision repair operates to remove dimers from plasmid DNA in wild-type yeast. Dimers can be removed from 2 ,um DNA in a rad1 mutant by photoreactivation.

    Notes: Summary Two-dimensional gel electrophoresis was used in a comparative study of ribosomal proteins from various strains of Saccharomyces cerevisiae. The results demonstrated a case of dimorphism of L8 protein of 60S ribosomal subunit. Of eight strains examined, two strains were one type and six were the other type. The former, which was tentatively designated as altered (A) type, was more acidic than the latter, common (C) type, as shown by mobility difference in pH gradient gel. Heterozygous (A/C) diploid cells contained both types of L8 protein and gave rise to tetrads of 2:2 segregation for A and C types, indicating that the difference of mobility was reflection of the allelic difference of the gene coding for L8 protein.

    Notes: Summary A spontaneous high-level cycloheximide-resistant mutant of the yeast Saccharomyces cerevisiae (strain cy32) is found to have an altered protein of the large subunit (60S) of cytoplasmic ribosomes, namely protein L29. The resistance character segregates together with this biochemical defect and is semidominant in heterozygous diploids. Judged from in vitro susceptibility to inhibition by cycloheximide there are at least 50% resistant ribosomes present in such diploid strains. From these results it is concluded that cycloheximide resistance of mutant cy32 is caused by mutation of a single gene and that it is the structural gene for L29 which is affected. Preliminary genetic mapping data are also reported. They indicate a location of cyhx-32 marker on chromosome 7 near met13.

    Notes: Summary Newly synthesised DNA molecules the same size as replicons (7 million-60 million daltons) accumulate in yeast cells treated with hydroxyurea. During prolonged incubation in low concentrations of the drug, there is a large accumulation of these molecules without any corresponding increase in their molecular weight. On release from the inhibtion the molecules are converted to large molecular weight DNA. These observations are consistent with an inhibition by hydroxyurea of the joining of completed replicons. In addition, newly synthesised DNA molecules the size of yeast Okazaki fragments also accumulate in cells treated with hydroxyurea.

    Notes: Summary [2 μm+ and [2μm°] yeast were transformed to stable leucine prototrophy with the hybrid yeast — E. coli plasmid, pJDB219. This plasmid contains the entire sequence of the endogenous 2 μm yeast DNA plasmid in addition to the yeast nuclear LEU2 + gene and the Co1E1 derivative, pMB9. In the [2 μm+] transformants, a new wholly yeast LEU2 + plasmid, pYX, was generated, probably by a recombination event between pJDB219 and 2 μm DNA. The plamid, pYX, in the absence of 2 μm DNA, was found to exist in equimolar amounts of two forms, A and B, which probably arise by intramolecular recombination across the inverted repeat sequences of the 2 μm DNA portion of the plasmid. pJDB219 was found to require the presence of 2 μm DNA to undergo this intramolecular recombination. The results suggest that 2, μm DNA and pYX code for a gene product required in this recombination event which pJDB219 cannot produce.

    Notes: Abstract Changes in the soil phosphorus and potassium status on three farming systems in the Northeast Polder during 25 years are related to the fertilizer regime over this period. The changes in the nutrient status of the soil are reflected in fertilizer policy. High rates of phosphate dressings markedly increased the phosphate status of the soil and built up a reserve of phosphate. In contrast, the potassium status decreased by about half, because only potatoes in the rotation were dressed with potassium. Methods of fertilization are discussed and suggestions are made with regard to possible improvements.

    Notes: Abstract Powerful VHF radars are capable of almost continuously monitoring the threedimensional velocity vector and the distribution of turbulence in the middle atmosphere, i.e. the stratosphere and mesosphere. Methods of radar investigations of the middle atmosphere are outlined and the basic parameters, mean and fluctuating velocities as well as reflectivity and persistency of atmospheric structures, are defined. Results of radar investigations are described which show that the tropopause level as well as a criterion on the stability of the lower stratosphere can be deduced. Besides mean wind velocities, VHF radars can measure instantaneous velocities due to acoustic gravity waves. The interaction of gravity waves with the background wind is discussed, and it is shown that cumulus convection is an effective source of gravity waves in the lower stratosphere. The vertical microstructure of the stratosphere, manifesting itself in thin stratified sheets in which temperature steps occur, is investigated by applying knowledge from investigations of the oceanic thermocline. Possible origins, like shear generation and lateral convection of the microstructure of the stratosphere, are discussed. Observations of gravity waves in the mesosphere are reviewed and their connection with turbulence structures is pointed out. Finally, some open questions which could be answered by further VHF radar investigations are summarized.

Les lapins soumis à un régime pauvre en calcium sont plus sensibles à l'intoxication fluorée, ainsi que le démontre les exostoses et la rétention plus élvée de fluor du fémur. La matrice collagène de l'os et la synthèse de l'hexosamine, étudiée par l'incorporation de proline (U)14C dans le collagène et de glucose (1)14C dans l'hexosamine, ne semblent pas affecter par le fluor et par la concentration de calcium dans le régime des lapins.
    Abstract: Zusammenfassung Skelettfluorose wurde bei Kaninchen hervorgerufen, welche ausreichende und niedrige Calciumdiäten erhielten. Kaninchen mit einem niedrigen Calciumgehalt in der Nahrung entwickelten eine stärkere Fluoridintoxikation, was sich durch Exostosebildung und höhere Fluoridretention in den Femora zeigte. Die Synthese des Knochenmatirx-Collagens und der Hexosamine [untersucht mittels Einbau von Prolin (U)14C in Collagen und Glukose (1)14C in Hexosamin] wurde weder durch Fluorid noch durch den Calciumgehalt der Kaninchen-Nahrung beeinflußt.
    Notes: Abstract Skeletal fluorosis was induced in rabbits fed adequate and low calcium diets. Rabbits on a low calcium diet were found to suffer more from fluoride intoxication as evidenced by exostosis formation and higher retention of fluoride in the femora. Bone matrix collagen and hexosamine synthesis assessed by14C-proline (U) incorporation into collagen and14C-glucose (1) into hexosamine were found to be unaffected by the fluoride as well as by the level of calcium in the diets of rabbits.

    Notes: Summary Quantitative measurements were made of the ion fluxes of calcium and phosphate into and from calvaria (mouse or rat) when clamped in specially designed micro-Ussing chambers. The effects of varying concentrations of calcium were examined on the influx and efflux of calcium and of its counterion, phosphate. A comparable series of experiments was performed with varying phosphate concentrations. Both ions, as their concentrations increased, depressed their own influx, increased their own efflux, and significantly increased the equilibrium concentration, E/K, supported by the calvaria. Similarly, both ions, as their concentrations increased, affected the influx or efflux of their counterion only slightly but did depress the counterion's equilibrium level, E/K, significantly. In spite of these changes it was shown that calvaria effectively buffered the medium at physiological concentrations of calcium and phosphate. The buffering capacity, however, was small, and the balance, E/K, was modified by small uptake or loss of either ion. The small size of the interacting mineral pool was confirmed by direct measurement of the rapidly exchanging fractions of both calcium or phosphate. They were only ∼1% of the total ions present. The significance of these findings is discussed.

Le fragment obtenu contient 10% de produit minéral, sous forme de complexe calcium-phosphate, et des protéines et lipides. Des quantités importantes de phospholipides et RNA sont notées, ainsi que des quantités faibles d'hexosamine et d'acide sialique. Les essais enzymatiques et la microscopie électronique indiquent que la fraction contient plusieurs types différents de particules cytophoniques, mais leur contenu en calcium n'a pu être déterminé.
    Abstract: Zusammenfassung Es wurde festgestellt, daß 95% des gesamten Calcium-und Phosphatgehaltes in Zellhomogenaten, welche mechanisch aus Knochen isoliert wurden, mit subzellulären Teilchen in einem Zentrifugenfeld von 15000×G sedimentieren. Zur Isolation und teilweisen Reinigung dieser calciumreichen Fraktion wurde eine Methode entwickelt, die auf enzymatischer Verdauung mit Desoxyribonuclease und Ultrazentrifugation mittels 70% Sucrose basiert. Der entstandene Niederschlag enthielt 10% Mineral, der in Form eines Calciumphosphat-Komplexes vorlag und einen hohen Gehalt an Protein und Lipid aufwies. Es fanden sich ferner signifikante Mengen von Phospholipid und RNS, dagegen nur kleine Mengen von Hexosamin und Sialsäure. Enzymbestimmungen und Elektronenmikroskopie zeigten, daß die gefundene Fraktion mehrere, sich voneinander unterscheidende Typen von Cytoplasmapartikeln enthielt; ihr Calciumgehalt konnte jedoch anhand der verfügbaren Resultate nicht bestimmt werden.
    Notes: Abstract Ninety-five percent of the total calcium and phosphate in homogenates of cells isolated mechanically from bone was found to sediment with subcellular particles in a centrifugal field of 15000×G. A method was devised for the isolation and partial purification of this calcium-rich fraction by enzymatic digestion with deoxyribonuclease and ultracentrifugation through 70% sucrose. The resultant pellet was 10% mineral, present as some form of calcium-phosphate complex, and was rich in protein and lipid. Significant amounts of phospholipid and RNA were present also, but only small amounts of hexosamine and sialic acid. Enzyme assays and electron microscopy indicated that the fraction contained several different types of cytoplasmic particles but those which contained calcium could not be determined from available data.

Lorsque Na2PO3F et CaCluc sont ingérés simultanément, dans la proportion pondérale de F/Ca=1/50, on observe un parallelisme de l'absorption et de la concentration squelettique de F et Ca. Ce fait s'explique partiellement par un effet prolongé de l'absorption de F sous l'influence de CaGluc. L'hydrolyse des ions PO3F est faible dans l'estomac, presque totale dans l'intestin, et apparemment totale dans le plasma sanguin. L'ingestion combinée de Na2PO3F-CaCluc donne une élévation comparativement faible, modérée et prolongée de la concentration plasmatique de F, alors qu'aucune action n'est observée pour le Ca plasmatique chez les rats normaux. L'absorption et la concentration squelettique de F et Ca sont augmentées chez des rats déficients en Ca et diminuées chez des rats en hypervitaminose D2.
    Abstract: Zusammenfassung Die Absorption, die Konzentration in Blut und die Aufnahme im Skelet von Fluorid (F) und Calcium (Ca), zugeführt als Natriummonofluorphosphat (Na2PO3F) und als Calciumgluconat (CaGluc), wurden bei Ratten unter verschiedenen Bedingungen untersucht. Die hauptsächlichsten Ergebnisse waren folgende: Wurden Na2PO3F und CaGluc in einem Gewichtsverhältnis von F∶Ca=1∶50 gleichzeitig eingenommen, so verliefen die Absorption und die Aufnahme im Skelet von F und Ca parallel. Dies konnte teilweise durch eine verlangsamte F-Absorption unter dem Einfluß von CaGluc erklärt werden. Die Hydrolyse der PO3F-Ionen war im Magen spärlich, im Darm beinahe vollständig und scheinbar vollständig im Blutplasma. Die kombinierte Einnahme von Na2PO3F und CaGluc ergab eine verhältnismäßig langsame, gemäßigte und verzögerte Erhöhung der F-Konzentration im Plasma, während bei normalen Ratten keine Beeinflussung des Plasma-Calciums gefunden wurde. Die F- und Ca-Absorption und deren Aufnahme im Skelet waren bei calciumarmen Ratten erhöht und bei mit Vitamin D2 überdosierten Tieren erniedrigt.
    Notes: Abstract Absorption, blood levels and skeletal uptake of fluorine (F) and calcium (Ca) from combinations of sodium monofluorophosphate (Na2PO3F) and calcium gluconate (CaGluc) were studied in rats under varying conditions. When Na2PO3F and CaGluc were ingested simultaneously in a ratio by weight F/Ca=1/50, absorption and skeletal uptake of F and Ca were parallel. This was partly due to the prolonged absorption of F under the influence of CaGluc. Hydrolysis of PO3F ions was weak in the stomach, nearly complete in the intestine and apparently complete in the blood plasma. Combined Na2PO3F-CaGluc ingestion gave a comparatively slow, moderate but prolonged elevation of the plasma F levels, while no influence on plasma Ca was found in normal rats. F and Ca absorption and skeletal uptake were increased in Ca-deficient rats and decreased in vitamin D2-intoxicated rats.

    Notes: Summary The effects of a bone resorptive protein isolated from human cancer ascites fluid on bone cell calcium and cyclic AMP were studied with fetal rat cells. The osteoclast-activating factor increased bone cell calcium uptake at 37°C and 4°C with no direct effects on calcium efflux. Concentrations of the resorptive factor that increased in vitro bone resorption and cell calcium uptake had no effect on cyclic AMP. The effects of the protein on calcium uptake were not specific for bone cells, and large increases were also observed in isolated fetal rat skin cells. These studies suggest that increases in the permeability of the cell membrane to calcium are involved in the mechanism of action of the ascites fluid resorptive protein.

    Notes: Summary Pyrophosphate and diphosphonate ions have been said to diminish the dissolution of hydroxyapatite crystals, because they lower the equilibrium concentrations of calcium and phosphate ions in the bulk solution around hydroxyapatite crystals in a closed system. However, in a closed system these effects are not necessarily due to an effect on dissolution alone. In this paper we have used a continuous flow system to study the effects of pyrophosphate and two diphosphonates, ethane-1-hydroxy-1,1-diphosphonate and dichloromethane diphosphonate, on the dissolution of hydroxyapatite. All three compounds decreased markedly the rate of dissolution of hydroxyapatite as well as the exchangeable pools of calcium and phosphate ions around the cystals.

    Notes: Summary Calcium homeostasis and bone pathology were studied in weanling rats fed a low (70 ppm) magnesium diet for 2–21 days. The rats developed significant, progressive hypercalcemia after 6 days on the diet. The increase in blood calcium was accompanied by progressive hypoactivity of the parathyroid gland (PTG), as determined by histologic and morphometric analyses. Thus hyperactivity of the PTG could not have been responsible for the hypercalcemia observed. Histologic examination of femora and humeri from magnesium-deficient rats showed progressive subperiosteal hyperplasia, consisting of undifferentiated osteoprogenitor cells and fibrous tissue, after 7 days of deficiency. The presence of unmineralized osteoid tissue in the metaphyses indicated that mineralization was not proceeding normally. The alterations in differentiation of osteoprogenitor cells, together with the failure of mineralization, resulted in significantly lower rates of bone formation (as measured by fluorochrome labeling) in the magnesium-deficient rats. Basophilic cementing lines and inactive osteocytes in the cortices of bones from magnesium-deficient rats indicated that bone resorption was also severely reduced in magnesium deficiency. We postulate that bone magnesium depletion (66% by day 21) has a direct negative effect on osteoblastic and osteocytic activity, and may explain, in part, the decreased responsiveness of bone to parathyroid hormone (PTH) that has been observed in magnesium-deficient animals.

    Notes: Summary We have described differences in the aspects of biological rhythms for calcium and sulfur deposition on the labial and lingual sides of the growing rabbit incisor, where growth occurs along a spiral axis. The calcium oscillations appear to be smoother on the labial side than on the lingual side. The lingual side is characterized by high-frequency rhythms with high amplitudes which possess the greatest percent of the power (Fourier analysis). These observations also reflect a difference in behavior of the mean Ca concentration across the labial and lingual sides. Sulfur rhythms on the labial side have higher amplitudes than those on the lingual side, but systematic differences in distribution of power between high and low frequencies is not as pronounced as in the case of Ca. The differences in Ca rhythms reflect differences in the growth rates of incisors on either side of the spiral axis. The labial side grows slightly faster than the lingual side, and its odontoblasts secrete Ca along the spiral axis and toward the pulp cavity at the same time. Thus the resultant direction of growth is more nearly opposite the extension of the occlusal end on the labial side, and Ca is consequently deposited over a wider area relative to that on the lingual surfaces. On the lingual side, Ca is deposited within a more limited area, and growth must therefore be continuous at high frequencies. The distribution of Ca on both sides of the tooth reflects these differences in growth rate and periodicity in two ways. First, given a unit area of tooth, the calcium concentration on the labial side is less than that of the lingual side. Second, whereas the calcium concentration on the labial side declines rapidly from the enamel-dentin junction to the pulp cavity, it is uniformly high across the lingual side because its growth is more continuous at high frequencies.

    Notes: Summary The timed sequence of events following the oral administration of 1α-hydroxy vitamin D3 (1αOHD3) to rachitic chicks was compared to that following a comparable dose of 1α,25-dihydroxyvitamin D3 (1,25(OH)2D3). RNA polymerase activity was maximally increased 20% by 1αOHD3 within 1 to 2 h and returned to control values after 8 h. Alkaline phosphatase activity was stimulated by 4 h and was maximal (3- to 5-fold increase) at 24 h. Calcium binding protein (CaBP) was detected initially within epithelial cells at the proximal end of the villus (just above the crypt) 6 to 8 h after 1αOHD3 administration, in epithelial cells lining the proximal half of the villus by 24 h, and in epithelial cells along nearly the entire villus by 48 h. At no time did goblet cells contain CaBP. Serum calcium concentrations were significantly elevated in 2 h and maximal by 12 h (an increase of 3.6 mg/dl). Calcium accumulation by the intestinal mucosa in vitro was increased by 6 to 8 h and maximal (60% increase over controls) at 24 h. Phosphate accumulation by the intestinal mucosa in vitro was increased by 6 h and maximal (105% increase over controls) between 8 and 24 h. 1,25(OH)2D3 increased CaBP and calcium accumulation by 4 h, 2 h sooner than did 1αOHD3. 1,25(OH)2D3 decreased serum calcium levels and increased serum phosphate levels at 2 h unlike 1αOHD3. No difference in the effects of these compounds on alkaline phosphatase activity, RNA polymerase activity, and phosphate accumulation could be demonstrated. These results are consistent with the possibility that 1αOHD3 may not require conversion to 1,25(OH)2D3 for all of its biological effects.

    Notes: Summary An in vitro perifusion system was used to study parathyroid hormone (PTH) secretion in response to calcium (Ca) and beta-adrenergic agents. Perifused parathyroid tissue responded to changes in Ca within the physiologic range during experiments up to 5 h. There was rapid secretory stimulation after exposure to low Ca, with the maximum response being observed at 20 min. Normal bovine glands showed a Ca-independent nonsuppressible component of PTH release at concentrations of Ca above physiologic. 1-isoproterenol produced rapid stimulation of PTH release, the response being blocked by a beta antagonist. The maximum secretory response to either low Ca (0.5 mM) or 1-isoproterenol (10−5 M) was enhanced when the two stimuli were applied simultaneously. The response to isoproterenol was blocked by raising Ca to 2.5 mM. Although d,l-propranolol (10−4 M) caused mild suppression of PTH release at a Ca of 1.25 mM, it did not cause additional suppression at 2.5 mM Ca nor did it decrease the response to 0.5 mM Ca stimulation. The secretory response of the gland to low Ca was sustained at a level more than double the baseline rate. The response to isoproterenol was more transient, with a return to or toward baseline secretion within 60 min. These results suggest that beta agonists and low Ca have separate but related mechanisms for stimulating PTH release and may affect different pools of hormone. The perifusion system described is a relatively simple technique for assessing the kinetics and interactions of various stimulators of PTH secretion.

    Notes: Summary We have identified a variety of biological rhythms involved in the apposition and mineralization of dentin in the rabbit incisor. Animals were injected during the day or night with lead acetate at 2-week intervals—to provide biological time markers in forming dentin—and transverse undecalcified sections of the lower incisors were prepared for electron microprobe analysis. The positions of the lead markers were identified, and the continuous distribution of calcium and sulfur was measured at 1 µm intervals between the markers. In thin sections stained with hematoxylin after decalcification, the widths of a series of structural increments (bands) were measured with an ocular micrometer. Fourier analysis of the data revealed spectra of structural and compositional rhythms with a range of periodicities which extended from a matter of hours [ultradian (〈24 h)] to days [infradian (〉24 h) and circadian (approximately 24 h)]. The structural and compositional rhythms appeared to be independent to the extent that they did not necessarily have the same periods, or amplitudes. Nor were there simple phase relationships between all of the rhythms. At some times, Ca and S fluctuations are inversely proportional (180° out of phase), but in other cases they are directly proportional or out of phase by varying degrees other than 180°. The analyses thus suggest that calcium and sulfur deposition (representing mineral and glycosaminoglycan deposition, respectively) are not simply inversely proportional, and that the hematoxylin-stained structural increments did not solely reflect differences in the distribution of the mineral components in dentin.

Le calcium semble intracellulaire. La majorité (90% ou plus) sédimente avec des particules intracellulaires au cours de la centrifugation et le reste se situe dans le liquide cellulaire. Des études cinétiques à l'aide de45Ca indiquent un passage rapide du calcium, en rapport avec la température, à la fois dans les réserves solubles et les particules de calcium d'homogénats de cellules osseuses. Ils indiquent aussi un échange direct entre la phase liquide et les particules, d'une part, et le milieu extracellulaire, d'autre part. Ces échanges se font à une vitesse différente et, probablement, selon des mécanismes différents, la portion comportant les particules étant la plus rapide. Les résultats indiquent que ces dernières pourraient correspondre aux petites granules denses identifiées récemment en microscopie électronique et l'acquisition, le stockage et la libération de calcium de ces compartiments pourraient être importants pour le métabolisme squelettique du calcium.
    Abstract: Zusammenfassung Der mittlere Calciumgehalt einer gemischten Population von lebensfähigen, frisch aus der verkalkten Matrix gewonnenen Knochenzellen betrug beim schwein 194,5±7,3 mmol/kg Naßgewicht und bei der Ratte 176,6±8,7 mmol/kg. Das Calcium scheint intracellulär zu liegen, wobei der größte Teil (90% oder mehr) in einem Zentrifugalfeld leicht mit intracellulären Partikeln sedimentiert, während die Zellflüssigkeit im Gleichgewicht bleibt. Kinetische Studien mittels45Ca zeigen einen raschen, temperaturabhängigen Umbau von Calcium sowohl im löslichen als auch im unlöslichen Calcium-Pool von Knochenzellhomogenaten, ferner einen direkten Austausch zwischen jedem Pool und dem extracellulären Medium, wobei jeder mit verschiedenen Geschwindigkeiten und wahrscheinlich durch verschiedene Mechanismen abläuft, wobei der unlösliche Pool der raschere ist. Diese Resultate lassen vermuten, daß der unlösliche Pool den kleinen dichten Granula entsprechen könnte, welche kürzlich durch Elektronnmikroskopie in Knochenzellen identifiziert wurden, und daß die Aufnahme, Lagerung und Abgabe des Calciums aus diesen Speichern für den Skelet-Calcium-Metabolismus wichtig sein könnte.

The calcium appears to be intracellular, the major portion (90% or more) sedimenting readily with intracellular particles in a centrifugal field and the balance being largely in the cell sap. Kinetic studies using45Ca indicate rapid temperature-dependent turnover of the calcium in both soluble and particulate calcium pools of bone cell homogenates and a direct exchange between each pool and the extracellular medium, each occurring at a different rate and possibly by different mechanisms, the particulate pool being the more rapid. These data suggest that the particulate pool may correspond to the small dense granules recently identified in bone cells by electron microscopy and that the uptake, storage and release of calcium from these stores may be important in skeletal calcium metabolism.

    Notes: Summary The potassium pyroantimonate technique was utilized for the selective subcellular localization of calcium in the mandibular condylar cartilage of 1-day-old rats. Electron dense calcium pyroantimonate precipitates were localized principally in mitochondria and at the cell membrane of the chondrocytes. In addition, small intracellular vesicles 0.1–0.2µm in diameter were observed in proximity to the cell membrane of chondrocytes of the mid-hypertrophic zone. The results suggest that these vesicles were being extruded from the cell into the extracellular matrix. Energy-dispersive analysis by X-rays confirmed that calcium is the principal cation of the electron-dense precipitates.

Ils sont analysés pour leur contenu en calcium, phosphate inorganique et protéines. Chez les rats hypercalcémiques, la concentration en calcium du fluide interstitiel est de 4.5% inférieur à celle du sérum. La concentration en calcium de l'ultrafiltrat interstitiel est, de façon inexplicable, différente dans les deux groupes d'animaux; l'un montre une valeur élevée (11,43 mg/100 ml); l'autre présente une valeur faible (4,91 mg/100 ml). La concentration du phosphate inorganique dans le fluide interstitiel ne diffère pas significativement du P sérique, alors que la fraction ultrafiltrable de phosphate inorganique est de 38% inférieur à celle du fluide interstitiel. La concentration protéique dans le fluide interstitiel est de 50% inférieur à la valeur dans le sérum. Chez les rats hypocalciques, la concentration du calcium est nettement inférieure dans le sérum, aussi bien que dans le fluide interstitiel et l'ultrafiltrat. Les rapports entre la quantité de calcium du sérum, du fluide interstitiel et de l'ultrafiltrat, sont similaires à ceux des animaux témoins.     Abstract: Zusammenfassung Es wurden Ratten durch die Verabreichung hoher Vitamin D-Dosen hypercalcaemisch oder durch Parathyroidektomie hypocalcaemisch gemacht und ihre Interstitialflüssigkeit und deren Ultrafiltrat auf Calcium, anorganisches Phosphat und Protein untersucht; dazu wurden Diffusions- und Dialysierkammern subkutan implantiert. Bei den hypercalcaemischen Ratten war die Calciumkonzentration in der Interstitialflüssigkeit nur 4,5% niedrige als in Serum. Im Ultrafiltrat der Interstitialflüssigkeit war die Calciumkonzentration der beiden Tiergruppen unerklärlich verschieden; die eine Gruppe wies einen hohen Wert (11,43 mg/100 ml), die andere einen niedrigen Wert (4,91 mg/100 ml) auf. Die Konzentration des anorganischen Phosphats in der Interstitialflüssigkeit unterschied sich nicht signifikant von derjenigen im Serum, während der ultrafiltrierbare Anteil des anorganischen Phosphats 38% tieferlag als in der Interstitialflüssigkeit.     Notes: Abstract Interstitial fluid and its ultrafiltrate was sampled from rats made hypercalcaemic by administration of high doses of vitamin D or hypocalcaemic by parathyroidectomy, in subcutaneously implanted diffusion and dialyzing chambers, and analyzed for calcium, inorganic phosphate and protein. In the hypercalcaemic rats the concentration of calcium in the interstitial fluid was only 4.5% lower than serum-Ca. The calcium concentration in the interstitial ultrafiltrate was inexplicably different in the two groups of animals, one showing a high value (11.43 mg/100 ml), the other showing a low value (4.91 mg/100 ml). The concentration of inorganic phosphate in the interstitial fluid did not differ significantly from the serum-P, while the ultrafiltrable fraction of inorganic phosphate was 38% lower than the amount found in the interstitial fluid. The concentration of protein in the interstitial fluid was 50% of the serum value. In the hypocalcaemic rats, the concentration of calcium was considerably lowered in serum, as well as in interstitial fluid and ultrafiltrate. Des mesures de diffractions électronique et en rayons X et de spectrophotométrie infra-rouge confirment la nature amorphe de chaque précipité. Le microscope électronique par transmission et par balayage démontre que les particules élémentaires des 5 produits synthétiques sont de forme sphérique, quelque soit la méthode de préparation. Quatre des précipités contiennent de petites sphères de taille identique (200 à 1200 Å de diamètre), alors que le dernier précipité, riche en carbonate, contient des particules plus grandes (700 à 2000 Å de diamètre). Ces sphères sont de densité électronique uniforme lorsqu'une intensité de courant, la moins élevée, est utilisée. Un faisceau d'électrons, d'intensité plus élevée, permet de mettre en évidence de nombreux centres, transparents aux électrons, dans chaque particule. In chauffage sous vide à 600° ne modifie pas la nature amorphe des précipités ou la morphologie et la taille des particules élémentaires, bien que la presque totalité de l'eau, contenu dans ces précipités, soit éliminée. L'échauffement ne provoque pas la formation de centres transparents aux électrons. Au contraire, les sphères deviennent résistantes à des dommages, causés par le faisceau. Il semble que le faisceau d'électrons agit sur les atomes d'oxygène et d'hydrogène de la phase aqueuse du phosphate de calcium amorphe et que cette réaction initiale est suivie par le déplacement et/ou la perte d'autres atomes.
    Abstract: Zusammenfassung Fünf nicht-kristalline Calciumphosphat-Niederschläge, welche aus Lösungen mit variierendem pH, Übersättigungsgrad und Karbonatgehalt gewonnen wurden, wurden mit physikochemischen und morphologischen Methoden untersucht. Röntgendiffraktion, Infrarot-Spektrophotometrie und Elektronendiffraktionsmessungen bestätigten die amorphe Natur jedes Niederschlages. Sowohl Durchstrahlungs-, als auch Raster-Elektronenmikroskopie zeigten, daß die nativen Elementarpartikel aller 5 synthetischen Produkte von kugeliger Form und unabhängig von den angewandten Präparativ-Techniken waren. Vier der Niederschläge enthielten Kügelchen von der gleichen Größe (200–1200 Å Durchmesser), während jener mit relativ hohem Karbonatgehalt viel größere Partikel aufwies (700–2000 Å Durchmesser). Die Kügelchen waren für den Elektronenstrahl von einheitlicher Dichte, wenn mit der Stromdichte bei niedrigster brauchbarer Stufe betrachtet. Wurden die Proben stärkeren Elektronenströmen ausgesetzt, so entwickelten sich sogleich in jedem Teilchen zahlreiche elektronendurchlässige Zentren. Erhitzen in Vakuum bis zu Temperaturen von 600° veränderte weder die amorphe Beschaffenheit der Niederschläge noch die Morphologie und Größe ihrer elementaren Partikel, obwohl beinahe alles Wasser, welches in den ursprünglich vorliegenden Proben enthalten war, durch dieses Vorgehen entfernt wurde. Die Hitzebehandlung verursachte keine Bildung von elektronendurchlässigen Zentren sondern machte die Kügelchen im Gegenteil in zunehmendem Maße widerstandsfähig gegenüber Strahlenschädigungen. Diese Befunde lassen vermuten, daß sich die ausgestrahlten Elektronen und die Sauerstoff- oder Wasserstoffatome der wäßrigen Komponente des amorphen Calciumphosphates gegenseitig beeinflussen und daß diese anfängliche Reaktion von einer Verdrängung und/oder einem Verlust anderer Atome gefolgt wird.
    Notes: Abstract Five non-crystalline calcium phosphate precipitates prepared from solutions varying in pH, degree of supersaturation and carbonate content were examined by physicochemical and morphological methods. X-ray diffraction, infrared spectrophotometry and electron diffraction measurements confirmed the amorphous nature of each precipitate. Both transmission and scanning electron microscopy demonstrated that the native elemental particles of all 5 synthetic products were spherical in shape and independent of the preparative procedures employed. Four of the precipitates contained spherules of similar sizes (200–1200 Å in diameter) while the one relatively rich in carbonate exhibited much larger particles (700–2000 Å in diameter). The spherules were uniformly dense to the electron beam when viewed with the current density at its lowest useful level. Exposure to higher electron currents immediately resulted in the development of numerous electron-lucent centers in each particle. Heating in vacuum to temperatures up to 600° did not change the amorphous nature of the precipitates or the morphology and size of their elemental particles even though almost all of the water contained in the original precipitates was removed by this process. Heat treatment did not induce formation of electron-lucent centers. On the contrary it, made the spherules increasingly resistant to beam damage. These findings suggest that the beam electrons interact with the oxygen or hydrogen atoms of the aqueous constituent of the amorphous calcium phosphate and that this initial reaction is followed by displacement and/or loss of other atoms.

    Notes: Abstract Ultrastructure of the mating tube formed in yeast haplont of the heterobasidiomycete Tremella mesenterica was studied by electron microscopy. Cell wall of the mating tube emerged as evagination of the inner layers, rupturing outer layers of the mother cell wall. Comparison with budding cells suggested that the tube emergence place at bud scar and the process of tube emergence was the same as that of bud emergence. Electron transparent vesicles of 0.1 μm diameter were scattered in the cytoplasm of the mating tube. Nucleus-associated organelle was located at one side of the nuclear envelope which extended towards the mating tube. A few microtubules were detected in the mating tube, but their association with a nucleus was not clear. The cytoplasmic structure of the mating tube was discussed in comparison with that of hyphae of the filamentous fungi.

    Notes: Abstract Volatile constituents of leaves of acceptable and unacceptable plants evoke multiunit responses from the antennal olfactory sensilla of larvae of the eastern tent caterpillar. Electrophysiological investigation of the activities of three of the sixteen antennal olfactory receptors revealed that different combinations of the three responded to the various plants. In those instances where the same combination responded to more than one species of plant, the ratios of responses measured as frequency of action potentials were often different. Thus, discrimination of different plants theoretically could be based upon the absolute number of cells responding and/or the ratios of frequencies.

    Notes: Abstract Males of several sphecid wasps apply volatile secretion on specific sites, perches. The odor is supposed to act in premating behavior as an attracting pheromone. There are strong indications that the scenting material is produced by the mandibular glands. One philanthine species,Philanthus triangulum, and three nyssonine ones,Argogorytes fargei, A. mystaceus, andNysson spinosus, the latter cleptoparasite onArgogorytes wasps, have been analyzed by gas chromatography-mass spectrometry. The major compound in the mandibular gland secretion of the four species was tentatively identified as 2,5-dimethyl-3-isopentylpyrazine. A few other pyrazines not yet identified were also found. Preliminary tests withP. triangulum show that alkylpyrazines influence male behavior.

    Notes: Abstract Sequential aerial photography was used to detect ponderosa pine trees killed by successive generations of the western pine beetle (WPB),Dendroctonus brevicomis Lec., over a three-year period during a study to evaluate the effectiveness of attractive pheromones for the suppression and survey of WPB. The total number of WPB-killed trees were estimated for each generation, using probability-proportional-to-size sampling procedures. Infested trees at the beginning of the suppression treatment totaled 283. Attacks by three successive WPB generations in 1970 killed 90,83, and 91 trees, respectively. The first generation in 1971 killed 47 trees and the two subsequent generations combined killed a total of 49 trees. During the suppression treatment, tree mortality was concentrated into the suppression plots in comparison to the check plots and the surrounding area. By 1972, tree mortality distribution returned to its original pattern, but at one-tenth the original level, as shown by maps.     Notes: Summary A combination of qualitative and quantitative studies show that there is not an abrupt release of calcitonin at the time of parturition in the ewe; these results differ from those obtained by several authors in the parturient cow. At lambing time, ewes are neither hypocalcemic nor hypophosphatemic; thyroidal calcitonin content is similar in parturient and in dry non-pregnant ewes. In electron microscopic studies, synthesis and release of calcitonin from C-cells in ewes, slaughtered in the hours just after lambing, appear to be as usual. These results are discussed with regard to the syndrome of parturient paresis in ruminants.

    Notes: Summary The structures of the eyes of two Staphylinid beetles, one large (Creophilus erythrocephalus—18 mm in length) and one small (Sartallus signatus—3 mm in length), were compared. 1. The gross structure of the two eyes is the same: a) biconvex corneal lens, b) crystalline cone of “acone type”, c) narrow crystalline thread and thickening of cone cell extensions just above the rhabdom, d) seven retinula cells plus one basal cell, and e) almost the same corneal refractive index and optical properties of the corneal cuticle. Considerable differences exist in size, shape and arrangement of rhabdom, retinula cells and their axons. The smaller size of the eye of Sartallus is caused by reduction of number of facets rather than cell size, and, in fact, retinula cells and rhabdom have been found to be larger in the smaller beetle. 2. Structural changes during dark-light-adaptation affect crystalline cone, position of screening pigment and size of intercellular spaces between the retinula cells. In the dark-adapted state the cone retracts a little and the crystalline tract becomes wider. A thickening of cone cell extensions occurs just above the rhabdom. Screening pigment migrates to a more distal position and intercellular spaces between the retinula cells considerably increase in size as compared with the light-adapted eye. 3. Both eyes are rich in irregularities. An attempt to classify anomalies of compound eyes has been made. The number of cone cells and principal pigment cells varied, in some cases, from 1–5 and 0–3 respectively. Basal retinula cells did not always contribute to the rhabdom. 4. Interferometrical observations reveal the extreme optical homogeneity of the entire corneal cuticle of the eyes of both species. No layers of different refractive index or optical separation of adjacent ommatidia can be found. The refractive index of the cornea of Creophilus is 1.469; that of Sartallus is 1.488. 5. Functional consequences of the changes which occur during dark-light-adaptation are discussed. Exposure to intense light appears to cause a rapid aging of retinula cells, which is indicated by an increase of onion- and multivesicular bodies.

    Notes: Summary The structure of the compound eyes of adult Cicindela tranquebarica Herbst was examined by use of light, scanning, and transmission electron microscopy. Each ommatidium of these photopic eyes is eucone and has a “subcorneal layer” situated between the corneal lens and crystalline cone. A distal rhabdomere consisting only of microvilli from retinula cell seven, a more proximal, rectangular, fused rhabdom formed from six retinula cells, and a basal, eighth retinula cell with a spherical rhabdomere comprise the light sensitive portions of the ommatidium. The “subcorneal layer” consists of lamellae of endocuticular microfibrils and, in surface view, shows 11 concave polygons. Proximal extensions of the crystalline thread form inter-retinular fibres containing microtubules between retinula cells 1/2, 3/4, 5/6, and 7/1. The primary pigment cells are devoid of pigment granules, but are rich in rough endoplasmic reticulum. Proximal to each retinula cell nucleus are two basal bodies, one perpendicular to the other. The more proximal basal body extends two fibrillar feet proximally which fuse to form a horizontally-banded ciliary rootlet extending the length of the retinula peripheral to the rhabdom. Each ommatidium is surrounded by 16 secondary pigment cells. Interfacetal mechanoreceptors between some adjacent lenses each have a single bipolar neuron, with a dendritic sheath, tubular body, cilium, outer and inner sheath cells, and an axon surrounded by a neurilemma sheath cell.

    Notes: Summary Cytological changes in the pars intermedia of the goldfish were investigated after adding calcium to deionized water (DW). In fish maintained in DW, the PAS-positive cells are highly stimulated in comparison to cells of fish kept in fresh water (FW). In DW supplemented with calcium at the same concentration as in FW (2 mM/l), the hyperactivity of the PAS-positive cells is prevented. When calcium ions are added 60 h before the animals are sacrificed, the PAS positive cells start to show signs of regression and their granules are stored: the release of the granular material appears to be suppressed by calcium. In the goldfish, the PAS-positive cells, homologous to a similar cell type in the eel, react only very weakly with the PAS technique. The name “calcium-sensitive cells” appears to be more appropriate in the goldfish for this particular cell type, secreting an unknown factor. This factor, different from the prolactin produced in the rostral pars distalis of the hypophysis, might be an equivalent of a “hypercalcin”.

    Notes: Summary The dynamics of Ca++ during induced pinocytosis were studied in Amoeba proteus using chlorotetracycline (CTC). The fluorescence of the Ca++ -CTC-complex was monitored by an image intensification system, which has certain advantages over standard equipment: (1) Living cells are not subjected to the damaging influence of intensive microscopic illumination, (2) fluorescent probes are not bleached during observation, and (3) the rapid dynamics of Ca++-fluxes can be recorded using short exposure times. The results demonstrate the existence of Ca++ bound to intracellular and extracellular sites of the cell membrane complex in normal locomoting and pinocytoting Amoeba proteus. The application of cations inducing pinocytosis causes a rapid decrease in the external CTC-fluorescence probably due to a release of Ca++ from the mucous layer. The degree of fluorescence intensity is correlated with the capacity of pinocytotic channel formation, i.e., the fluorescence decreases as the number of channels increases. During the phase of vesiculation a distinct fluorescence mainly restricted to the basal region of the channels is observed. Intracellular Ca++ was detected in close vicinity to the plasma membrane after both microinjection and external application of CTC. The internal CTC-fluorescence is slightly decreased during the induction phase of pinocytosis. The observations are in good agreement with previous results on the localization of Ca++-binding sites at the plasma membrane of Amoeba proteus and demonstrate the important role of Ca++-fluxes for the process of pinocytosis.

Each sensillum is innervated by a single bipolar sensory cell. The sensilla ofHydrobius are associated with three enveloping cells, which enclose an inner and outer receptor lymph cavity. A single enveloping cell only is found in the completely differentiated sensilla ofAgabus. These sensilla do not form an outer lymph cavity. The area beneath the hair base is filled by the distal process of the enveloping cell and by extensions of epidermal cells. Only one extra-cellular space exists, which seems to be homologous to an inner receptor lymph cavity. The outer dendritic segment surrounded by a dendritic sheath runs to the tip of the hair shaft. In the hair shaft the outer dendritic segment divides into several branches. The poreless hair shaft does not rise over the surface of the cuticle, but it is positioned in a narrow shallow groove. Special socket structures or a tubular body do not exist. The digiti-form sensilla possess neither the typical feature of mechanosensitive, nor gustatory or olfactory sensilla. The functional significance of the structural divergences in the sensilla of both species and the presumed function of the sensilla are discussed referring to hygro- and thermo-receptors.

    Notes: Summary The effect of acid irrigation on the growth of rooted cuttings ofPicea abies (L.) Karst, was investigated in a pot experiment lasting 3 years. It involved two clones of Norway spruce, H 253 Bogstad I and H 254 Bogstad II. Irrigation water of pH 5.4, 4.0, 3.0 and 2.5 was used. Liming was included in the experiment. After the experimental period, the plants of all treatments were growing reasonably well. However, those plants irrigated at pH 2.5 were slightly discoloured. The plant mortality was only 3% throughout the experiment, and was not connected to acid irrigation. The limiting growth factor was N. All other nutrient elements measured in the plants were close to optimal concentration. Plants irrigated at pH 2.5, and to some extent at pH 3, contained excessively high concentrations of Al, t-S and SO4. The total amount of Ca, Fe and Mn taken up by the plants decreased with increasing soil acidity. The increased growth of clone H 254 relative to H 253, produces a corresponding impression on soil characteristics. Soil acidity is governed by acid irrigation and CaCO3 application, but the clonal effects are also of importance. Norway spruce appears to be tolerant to Al concentrations as high as 50 mmol/kg in the needles.

    Notes: Summary Soils influenced by acid mine drainage (pH〈5.0) are characterized by low concentrations of essential nutrients and increased solubility of heavy metals. The conditions typically reduce plant establishment and growth. However, river birch (Betula nigra L.) is commonly found along low pH streams in southeastern Ohio. The objective of this study was to determine the concentration of Al, Mn, Ca and Mg inB. nigra tissues. The results indicate Al and Mn are accumulating inB. nigra when compared to other species. Within river birch, Al concentrations are highest in roots; Mn concentrations are highest in leaves. There is not a concomitant reduction in Ca and Mg concentrations as suggested by soil levels.

    Notes: Abstract Mixtures of (Z)-5- and (Z)-7-dodecenyl acetates in ratios 〉100∶1 are powerful, specific sex attractants for redbacked cutworm male moths. Bioactive lures were produced by admixture of these pure compounds or through syntheses involving catalytic bond shifting. Best field trapping was obtained with 200∶1 (Z)-5∶(Z)-7 ratio, at lure doses (on rubber) of 0.1–2.0 mg, using cone swivel traps with 10- to 13-mm orifices, or Pherocon® 1-CP sticky traps, at a height of 50–90 cm. The pheromone traps were more efficient than large blacklight traps and captured the various morphological forms of the moth in similar ratios.

Highest trap catches were obtained when these compounds were dispensed in the relative proportions observed in the female gland.     Notes: Abstract Methods are presented for mathematically determining dispersal of a vapor in still air, in turbulent air of zero average velocity, and in turbulent or laminar air currents of constant, nonzero average velocity. The methods are combined with several assumptions about insect behavior to derive an insect attraction model that predicts: (1) In a warehouse a searching insect is likely to be attracted to a calling insect if it comes within an attraction sphere, 0.4–2.5 m in radius. (2) The attraction spaces of typical sex pheromone-baited traps that emit pheromone at rates greater than 0.01 ng/sec extend beyond the boundaries of a 10 × 10 × 10-m warehouse. (3) The searching behavior of an attracted insect is likely to be altered from an extensive to an intensive pattern if it comes within an altered-behavior sphere, 6–60 cm from a calling insect or within 10 m of a trap emitting 0.76 ng/sec. (4) Pheromone does not sink unless it is emitted along with a large amount of a high-vapor pressure solvent. The model is used in support of several hypotheses, including: (1) The effect of an adsorptive surface on the vapor concentration after an extended period of emission is negligible except at positions near the surface. (2) Sex pheromone-baited traps with sources of small dimensions have greater trapping efficiency than otherwise identical traps with sources of large dimensions.

    Notes: Abstract Pheromone washes from calling female moths of redbacked cutworm,Euxoa ochrogaster (Guenée), contained the following acetates that are structurally similar to those of known lepidopteran pheromones (%): decanyl (8.7), dodecanyl (8.5), (E)-5-dodecenyl (3.3), (Z)-5-dodecenyl (76.4), (Z)-7-dodecenyl (3.1), and (Z)-9-dodecenyl (trace〈0.5%). This is the first time that (Z)-5-dodecenyl acetate has been identified as a pheromone component. Three types of specific receptor cells were found in the male antennae, and they responded to (Z)-5-decenyl acetate, (Z)-5- and (Z)-7-dodecenyl acetates, respectively. Strong electroantennographic detector responses were also recorded for these three acetates and for (Z)-5-undecenyl acetate. The evidence for the presence of (Z)-5-decenyl acetate in the pheromone washes was inconclusive. The presence of (Z)-7- and the absence of (Z)-8-dodecenyl acetates were confirmed by a special electroantennographic detector technique in which the detector antennae were from males of other species that were known to have strong responses to these acetates. This is a very useful technique. Field results show that low concentrations (0.1–1.3%) of (Z)-5-decenyl acetate were synergistic when tested in a previously reported blend, but 6% was inhibitory. Similarly, (Z)-7-dodecenyl acetate at 2% or less may be essential for the attraction of males, but in previous tests at 14% it also was inhibitory. Species-specific attractant blends for redbacked cutworm males are described.

Response byI. pini-ID to (−)-ipsdienol in the laboratory was inhibited by (+)-ipsdienol, while the field response byI. pini-NY was optimal to a 50:50 mixture of (+) and (−)-ipsdienol. This differential production and response to enantiomers of ipsdienol is sufficient to account for populational specificity observed in earlier experiments and confirmed by our current work. Reciprocal hybrids of the NY and IDI. pini populations did not differ in their patterns of attraction and response; those of both types of F1 were similar to NY beetles. Thus, it appears that (+)-ipsdienol is produced by the hybrids and is necessary for their maximum response, but this supposition was not tested. The genes controlling the pheromone biology ofI. pini are not sex linked. Since previous whole-antenna and single-cell electrophysiological data show that the receptor systems of the two populations are virtually identical, response behavior seems to be governed by the central nervous system rather than by the characteristics of the olfactory receptors.

    Notes: Abstract The mandibular gland secretion of the mutillid wasp,Dasymutilla occidentalis, possesses three short-chained ketones-4-methyl-3-heptanone (4MH), 4,6-dimethyl-3-nonanone (4,6DMN), 4,6-dimethyl-3-octanone (4,6DMO)—and several unidentified compounds. This is the first report of 4,6DMN as a natural product and its synthesis is described. These ketones, which are either known to be ant alarm pheromones or are structurally very similar to ant alarm pheromones, appear to function as allomones against ants, major potential predators of mutillid wasps. The major secretory component, 4-methyl-3-heptanone, which was identified in females and/ or males of the species analyzed within the generaDasymutilla, Timulla, Traumatomutilla, andPappognatha, appears to constitute a chemical character of the defensive secretions of these genera.

    Notes: Abstract The effect of different concentrations of the sex pheromone (Z,E)-9,12-tetradecadien-1-ol acetate on the upwind anemotactic behavior of the malePlodia interpunctella (Hübner) was measured at 23 ± 1 ° C and 34 ± 1 ° C. The stimulus-response regression lines were analyzed by a new procedure that accounts both for control responses in the absence of pheromone and also for peak responses below 100% in the presence of concentrations considerably above the normal physiological levels. From the regression line for each temperature, the upwind anemotactic thresholds were calculated to be 1.34 × 106 molecules/cm3 at 23 ° C and 1.65 × 104 molecules/cm3 at 34 ° C, similar to other thresholds reported in the literature. Since departures from the two lines occurred at the highest concentrations tested, near 108 molecules/cm3, the upwind anemotactic behavior may change qualitatively above an altered-behavior threshold that is about two orders of magnitude higher than the upwind anemotactic threshold. The lower response at 23 ° C suggests that cool temperatures inhibit flight in response to pheromonal stimulation.

    Notes: Summary Ginger, cassava, maize, wheat, french bean and tomato were grown for periods up to six weeks in continuously flowing nutrient solutions at seven constant pH values ranging from 3.3 to 8.5. All species achieved maximum or near-maximum growth in the pH range 5.5 to 6.5. However, there were substantial differences in the ability of species to grow outside this range. Ginger and cassava were the most tolerant species to low solution pH, while ginger and tomato were the only species to show no yield depression at the highest solution pH. Roots of all species at pH 3.3 and some species at pH 4.0 exhibited symptoms of hydrogen ion injury. In addition, the concentrations of magnesium in the tops of all six species, of nitrogen in the tops of tomato and cassava, and of manganese in the tops of maize at these pH values were inadequate for optimal growth. Growth depression at high solution pH was associated with iron deficiency in maize and wheat and with nitrogen and/or copper deficiency in cassava. The relevance of the present results to crop growth under field conditions is discussed. The complex interplay of plant and soil characteristics militates against precise definition of an optimum pH range for the growth of a particular crop unless the soil is also specified.

    Notes: Summary Dry matter and chemical changes in decomposing spruce needles were investigated after 16 and 38 weeks in laboratory lysimeters treated with distilled water or distilled water acidified to pH 3 or 2 with sulphuric acid. The water was added twice weekly in quantities equal to 100 or 200 mm month−1. The CO2 evolution and leaching of P, K, Mg, Mn, and Ca was followed together with pH measurements of the leachate. The loss of dry matter was approximately 25% during the first 16 weeks and approximately 37% after 38 weeks. At the first samling, 16 weeks, the amount of material decomposed was greater from the lysimeters given 100 mm month−1 of water. At this water quantity dilute sulphuric acid increased the decomposition. After 38 weeks sulphuric acid at pH 3 and 2 had decreased the decomposition at 200 mm month−1. However, the effects of acid application were small. The effect of treatment using acidified water on the content of monosaccharides was not consistent, whereas there was an indication of reduced decomposition of lignin when treated with 200 mm water month−1 at pH 3 and 2. Nitrogen was conserved in the lysimeters with small differences between the various treatments. The order of mobility of metal elements was K〉Mg〉Mn〉Ca. Increasing the quantity of water increased the leaching of K especially, whereas addition of dilute sulphuric acid increased the leaching of Mg, Mn and particularly Ca. During the first 16 weeks of the experiment, sulphuric acid reduced the leaching of P while later on this treatment increased the leaching. The pH of the leachate from the lysimeters treated with distilled water was initially 4.0–4.6 increasing to approximately 6.6 after 22 weeks. The pH of the decomposed needle material was 4.6 and approximately 5.2 after 16 and 38 weeks respectively. When treated with water at pH 3 the pH of the leachate was between 4 and 5, and the pH of the needles 4.2–5.1. Treatment with water at pH 2 gave a leachate with pH just above 2 and decreased the pH of the needles that had received 200 mm ‘rain’ month−1 to 2.9. The effect of the artificial acid rain appears to be more pronounced on the leaching of metal elements than on the biological activity and the dynamics of N and P. The treatments must be considered extreme when compared with the acidity of natural rain.

Aluminum increased the concentrations of P and K in the roots and K in the tops of most of the cultivars tested. A1 tolerant triticale retained a lower concentration of Mg in the roots and tops than the A1 sensitive triticale, when subjected to A1 stress. In addition, A1 treatments resulted in smaller increases in root P for the A1 tolerant triticale than for the A1 sensitive cultivars. The concentration of root Ca and P of the A1 tolerant wheat cultivars were significantly below that of the more sensitive plants. Aluminum tolerance in rye appeared to be associated with lower Ca and higher Mg concentrations in the tops. The accumulation of P and A1 in the roots was characteristic of sensitivity in triticale, wheat and rye.

    Notes: Synopsis Diets deficient in vitamin D effected a significant increase in plasma triiodothyronine (T3) concentration in raibow trout (Salmo gairdneri); different levels of dietary calcium exerted no effect on plasma T3 levels. These effects of vitamin D deficiency on plasma T3 levels appeared to be reversible, vitamin D supplementation after a period of vitamin D deficiency lowered T3 levels. Vitamin D3, vitamin D2 and the metabolites 25(OH)-D3 and 1, 25(OH)2D3 were all effective in lowering plasma T3 levels; vitamin D3 appeared to be more effective than vitamin D2. There appeared to be a correlation between weight gain and plasma T3 concentration in the groups fed different types and levels of vitamin D supplementation suggesting that the increased T3 levels may be a compensatory increase to the reduced weight gain of the vitamin D deficient fish. Plasma T4 levels were not affected by dietary vitamin D deficiency.

    Notes: Abstract During successful courtship in sulfur butterflies, virgin females respond to males by assuming a stationary posture and extending the abdomen ventrally from between the hindwings, thereby permitting copulation. In the clouded sulfur,Colias philodice Godard, this response was used in a laboratory bioassay to confirm the existence of a male chemical signal demonstrated by a previous worker and to document the signal's behavioral function and source. The male scent is shown to be required, in part, to reliably elicit abdominal extension and to be emitted from a patch of cells and scales associated with the dorsal surface of the male hind wing near the wing base. Experiments also show that evaporation of the signal is reduced when the source is covered by the forewing as it is at rest and in flight. These data, coupled with other information on male chemical signals in sulfur butterflies and other Lepidoptera, suggest that in sulfurs the morphology and chemistry of the scent glands along with the behavior of the male are structured in a way that minimizes the evaporative loss of scent from the wings when the male is not courting females.

    Notes: Abstract The release rates of (Z)-9-tetradecen-1-ol formate (Z9TDF) from four sizes of polyethylene tubing were measured periodically for 50 days by collecting the Z9TDF vapors on a polymeric adsorbent. The resulting release rate curves had shapes that were determined by the tubing's diameter and wall thickness. One tubing size that combined good longevity with adequately high release rates was tested in the field to determine whether it could disrupt pheromone communication of the corn earworm,Heliothis zea (Boddie). We observed a low percentage of disruption, but release rate measurements made concurrently showed that the tubing was releasing Z9TDF normally. In a second field test, a larger quantity of Z9TDF was released, and 87% disruption was attained for 66 days.

    Notes: Summary A polytrophic ovariole of the queen honeybee, Apis mellifera, is composed of a linear series of increasingly mature egg chambers, each consisting of an oocyte, an interconnected cluster of nurse cells, and a covering layer of follicle cells. This study describes changes in the volume of each of these components, as a function of the position of the egg chamber in the ovariole. An oocyte increases in volume from approximately 8.9 × 103 μm3 to approximately 9.6 × 106 μm3 over an average series of 20 egg chambers.

    Notes: Summary The effects of removing Na+ from the incubation medium on basal and secretagogue induced zymogen release by pancreatic fragments and isolated pancreatic acini were studied by both morphological evaluation and measurement of amylase release. In both fragments and isolated acini, removal of Na+ led to an increased basal secretion of zymogen granule contents from acinar cells via exocytosis; secretory material, however, accumulated in acinar and ductular lumina as a result of the lack of fluid secretion necessary to wash out the enzymes. In studies with fragments, after Na+ removal there was no significant increase in amylase release into the medium; isolated acini, in contrast, showed an increased amylase release consistent with the shorter distance from the acinar lumen to the bathing medium. Stimulation with either bethanechol or caerulein led to a further depletion of zymogen granules in both preparations; in the absence of Na+ secretory product accumulated in intracellular lakes as well as in duct lumens. The hypothesis that Na+ influx is important in stimulus-secretion coupling to release intracellular Ca2+ was directly tested by measuring 45Ca2+ efflux. No effect of removing Na+ on 45Ca2+ efflux was seen. It was concluded, therefore, that while Na+ is essential for pancreatic fluid secretion, it is not necessary for the secretion of zymogen granule contents into acinar lumina.
