    Notes: Abstract A mutant of Saccharomyces cerevisiae lacking phosphoenolpyruvate carboxykinase (E.C. 4.1.1.32) was isolated. The mutant did not grow on gluconeogenic sources except glycerol. The mutation was recessive and apparently affected the structural gene of the enzyme. Intracellular levels of metabolites related to the metabolic situation of the enzyme were not significantly affected after transfer of the mutant from a medium with glycerol to a medium with ethanol as carbon source. In these conditions only AMP decreased 3 to 5 times. A search for mutants affected in the other gluconeogenic enzyme, fructose 1,6 bisphosphatase, remained unsuccessful.

    Notes: Abstract Protoplasts of Saccharomyces cerevisiae synthesized and degraded trehalose when they were incubated in a medium containing traces of glucose and acetate. Such protoplasts were gently lyzed by the polybase method and a particulate and soluble fraction was prepared. Trehalose was found in the soluble fraction and the trehalase activity mostly in the particulate fraction which also contained the vacuoles besides other cell organelles. Upon purification of the vacuoles, by density gradient centrifugation, the specific activity of trehalase increased parallel to the specific content of vacuolar markers. This indicates that trehalose is located in the cytosol and trehalase in the vacuole. It is suggested that trehalose, in addition to its role as a reserve may also function as a protective agent to maintain the cytosolic structure under conditions of stress.

    Notes: Abstract Tryptophan was found to be degraded in Saccharomyces cerevisiae mainly to tryptophol. Upon chromatography on DEAE-cellulose two aminotransferases were identified: Aromatic aminotransferase I was constitutively synthesized and was active in vitro with tryptophan, phenylalanine or tyrosine as amino donors and pyruvate, phenylpyruvate or 2-oxoglutarate as amino acceptors. The enzyme was six times less active with and had a twenty times lower affinity for tryptophan (K m=6 mM) than phenylalanine or tyrosine. It was postulated thus that aromatic aminotransferase I is involved in vivo in the last step of tyrosine and phenylalanine biosynthesis. Aromatic aminotransferase II was inducible with tryptophan but also with the other two aromatic amino acids either alone or in combinations. With tryptophan as amino donor the enzyme was most active with phenylpyruvate and not active with 2-oxoglutarate as amino acceptor; its affinity for tryptophan was similar as for the other aromatic amino acids (K m=0.2–0.4 mM). Aromatic aminotransferase II was postulated to be involved in vivo mainly in the degradation of tryptophan, but may play also a role in the degradation of the other aromatic amino acids. A mutant strain defective in the aromatic aminotransferase II (aat2) was isolated and its influence on tryptophan accumulation and pool was studied. In combination with mutations trp2 fbr, aro7 and cdr1-1, mutation aat2 led to a threefold increase of the tryptophan pool as compared to a strain with an intact aromatic aminotransferase II.

    Notes: Abstract When α pheromone-pretreated cells of an inducible a strain of Saccharomyces cerevisiae carrying the inducible gene saa1 were incubated in a growth medium at 28°C, induction of sexual agglutinability began after a 10 min lag period. If the cells were incubated at 38°C during the lag period, no induction occurred even after incubation at 28°C. Contrary to this, if the cells were incubated at 28°C during the lag period, almost complete induction occurred, even after transfer to 38°C. Temperature shift experiments revealed that 5 min incubation at 28°C was necessary for the initiation of the temperature-sensitive period and further 5 min incubation for the completion of the period. The temperature-sensitive period was sensitive to phenylmethylsulfonyl fluoride.

    Notes: Abstract 31P NMR spectra were obtained from suspensions of Candida utilis, Saccharomyces cerevisiae and Zygosaccharomyces bailii grown aerobically on glucose. Direct introduction of substrate into the cell suspension, without interruption of the measurements, revealed rapid changes in pH upon addition of the energy source. All 31P NMR spectra of the yeasts studied indicated the presence of two major intracellular inorganic phosphate pools at different pH environments. The pool at the higher pH was assigned to cytoplasmic phosphate from its response to glucose addition and iodoacetate inhibition of glycolysis. After addition of substrate the pH in the compartment containing the second phosphate pool decreased. A parallel response was observed for a significant fraction of the terminal and penultimate phosphates of the polyphosphate observed by 31P NMR. This suggested that the inorganic phosphate fraction at the lower pH and the polyphosphates originated from the same intracellular compartment, most probably the vacuole. In this vacuolar compartment, pH is sensitive to metabolic conditions. In the presence of energy source a pH gradient as large as 0.8 to 1.5 units could be generated across the vacuolar membrane. Under certain conditions net transport of inorganic phosphate across the vacuolar membrane was observed during glycolysis: to the cytoplasm when the cytoplasmic phosphate concentration had become very low due to sugar phosphorylation, and into the vacuole when the former concentration had become high again after glucose exhaustion.

    Notes: Abstract Mitochondria, isolated from the ubiquinone-deficient nuclear mutant ofSaccharomyces cerevisiae E3-24, are practically unable to oxidize exogenous substrates. Respiratory activity, coupled to ATP synthesis, can, however, be reconstituted by the simple addition of ethanolic solutions of ubiquinones. A minimal length of the isoprenoid side chain (≥3) was required for the restoration. Saturation of the reconstitution required a large amount of exogeneous ubiquinone, in excess over the normal content present in the mitochondria of the wild type strain. A similar pattern of reconstituted activities could be also obtained using sonicated inverted particles. Mitochondria and sonicated particles are also able to carry out a dye-mediated electron flow coupled to ATP synthesis in the absence of added ubiquinone, using ascorbate or succinate as electron donor. This demonstrates that the energy conserving mechanism at the third coupling site of the respiratory chain is fully independent of the presence of the large mobile pool of ubiquinone in the membrane.

    Notes: Abstract A new magnetic discharge stabilization technique for coaxial laser systems is described. The approach utilizes crosses electric and magnetic fields to create and maintain a large and rapidly rotating plasma volume which does not experience glow-to-arc transitions. Very high cw specific discharge power loadings have been achieved even without the benefit of external gas cooling or circulation. Performance is insensitive to gas composition and pressure such that high power coaxial discharges have been run in CO2 laser gas mixtures up to several hundred torr. Stable cw discharges have also been obtained in mixtures containing several torr of SF6. The technique appears to be readily scalable to give very large excited volumes in systems with comparatively small overall physical dimensions.

    Notes: Abstract We have designed and tested a pulsed single-mode dye laser for spectroscopy experiments. The dye laser is pumped by a 6 kHz copper vapor laser to produce up to 230 mW average power at over 5% efficiency in a nearly diffraction limited beam. The dye laser pulse closely resembles the 30 ns pump pulse. Using an electronic servo loop, the 60 MHz wide line can be scanned, without mode hopping, through the 16 GHz permitted by the travel on the piezo driven mirror mount.

The line tunability of sealed-off FIR NH3 laser was almost achieved in 90, 115, 150, and 220 μm wavelength regions by the selective line tuning of the pumping IR NH3 laser.

    Notes: Abstract The cw output power of the uv CuII laser has been optimized with respect to the hollow cathode geometry, the discharge current, the fill gas pressure and the resonator mirror parameters. A maximum laser output power of 900 mW for multiline operation at 248.6, 259.1, 260.0, and 270.3 nm was achieved with 100 A discharge current, 260 V voltage and 16 mbar fillgas pressure, when a hollow cathode of 1.2 m length and 2×6 mm2 cross section was employed. The single-pass pain gl has been estimated to 7%. A hollow cathode cross section of 1.5×4.5 mm2 is suggested as an optimum geometry. In addition, some investigations on the mechanism of the laser power decay in Ne−Cu-discharges are presented.

    Notes: Abstract A theoretical treatment of the influence of a saturable absorber in close contact with a highly reflecting mirror on a light pulse is given. The shaping of a Gaussian pulse during a single passage through such an absorber is calculated for various absorber lengths. The influence of absorber position and length on the pulse parameters and the stability of a continuously pumped dye laser are investigated. The shortest pulses and the most favourable operation conditions of the laser are to be expected in the case of a short contacted saturable absorber.

    Notes: Abstract Single-mode selection with Michelson selectors in cw standing-wave dye lasers is analyzed. The application of single and double Michelson mode selectors for compensation of spatial hole burning effects is described in detail. Upper limits for the pumping are considered. With a proper resonator length very weak sideband modes on each side of the strong mode produce a feedback signal to a piezoelectric pusher which controls the resonator length.     Notes: Abstract Simultaneous laser operation at the wavelengths of λ1=637 and λ2=717 in Pr0.5La0.5P5O14 single crystals was observed. The crystals were excited by nanosecond publses from a Coumarin 2 dye laser (λ=450 nm). Laser emissions and their interdependence were studied both in crystals without an external resonator and in crystals placed within a plane parallel resonator. The energetic, time and frequency characteristics for both laser actions were examined. The effect of bifrequential emission was also found in crystals of different ratios of Pr and La ions concentration and in pure praseodymium pentaphosphate.

    Notes: Abstract We report infrared laser emission in the region of 3 to 5 μm from sodium vapour optically pumped by a pulsed dye laser with wavelengths ranging from 585 to 610nm. Twophoton excitation processes are believed to be responsible for the primary excitation. Both molecular transitions (4 to 5 μm) between high lying states, and atomic transitions (52 S 1/2−42 P 3/2,1/2 at 3.41 μm) have been identified.

    Notes: Abstract The presented theoretical model for a mode-locked Nd-glass laser simultaneously takes into account dynamics of the mode-locking dye, amplification saturation and radiation background. A systematic variation of laser parameters gives insight into the pulse formation process and allows to improve the laser design. The calculations show that it should be possible to decrease considerably the duration of light pulses of a mode-locked Nd-glass laser. Using a new mode-locking dye with a switching time of τ=2.7×10−12 s we obtained stable laser operation and a pulse duration of 1.7×10−12s.

    Notes: Abstract The characteristics of near uv dye lasers pumped with short duration, 308 nm pulses from a XeCl laser are described with particular emphasis on the parameters influencing high average power operation. The solvent is shown to effect the operation of all dye lasers studied, particularly the photochemical stability. Measurements of the efficiency, as a function of the integrated pump energy have been made for a series of dye-solvent combinations. With the best combinations, average output powers in excess of 1W and conversion efficiencies greater than 25% were obtained at a repetition rate of ∼100 Hz.

    Notes: Abstract Near-infrared laser action of atomic carbon was obtained using a hollow cathode discharge tube. Besides the known 1.4543 μm and 1.0691 μm laser lines, cw laser operation at 1.0683 μm, 0.9658 μm, and 0.9406 μm wavelength was observed for the first time. The source of carbon was due to dissociation of CO, CO2 or CH4 added to the He buffer gas.

    Notes: Abstract This work was an experimental investigation of a potential large-scale excimer laser system, in which a high-pressure KrF gas mix was pumped by the 4-MeV electron beam from an available linear accelerator. As a function of the mix and the pressure, fluorescence from KrF* and/or Kr2F* were produced with good efficiency. Comparing measured data with a kinetic model, it was found that the kinetics followed the “normal” patterns seen in low-energy electron pumping. The data fitting process also resulted in a new value for the three-body quenching rate constant for reactions involving Ar. A critical factor in the development of this laser will be an improvement in beam control over that afforded by a simple solenoidal magnet. The conditions necessary for lasing are predicted; no inherent limitations on scaling of the device were found.

    Notes: Abstract We investigated time-dependent effects related to the bistable operation of a CO2 laser with an intracavity nonlinear absorber (SF6). The response of the system to small perturbations was sensitively dependent on the operating laser line [P(12)→P(30)] of the 10.6 μm band. The narrow bistable region forP(12), due to a very narrow hysteresis loop, can be effectively utilized for optical switching, because of its pronounced sensitivity to variations in operational parameters. The maximum admissable beam blocking time (intracavity) for spontaneous return to lasing was measured forP(16), for a range of absorber pressures. This τm is related to the relaxation time of the upper level in the absorber.

    Notes: Abstract A tunable diode laser and a multipass optical cell are used to investigate the feasibility of performing radiocarbon dating by means of infrared spectroscopy. We show that the detection of14CO2 at concentrations of14C/12C≲10−12 is not limited by interferences from the background of normal CO2 molecules, provided the gas sample is cooled. Using short-time constants, the present sensitivity of the infrared technique is comparable to that of an ideal disintegration counter. The experimental results are extrapolated to long time constants to demonstrate that radiocarbon dating by infrared spectroscopy is feasible, and that carbon samples with a mass of less than 1 gm can be dated by this technique.

    Notes: Abstract Newly found reflection properties of the Fizeau wedge interferometer are shown to be of particular advantage in obtaining efficient multi-wavelength operation of dye lasers. Use is made of the fact that the reflecting Fizeau wedge can ensure at once a selective feedback for a given wavelength and quasi-total reflection in an other direction for all other wavelengths.     Notes: Abstract A synchronously mode-locked, cavity-dumped picosecond dye laser is described. The structure and intensity of the picosecond pulses measured under different conditions are reported. It was found that the structure of the pulses from the synchronously pumped dye laser depends critically on the length of the Ar+ laser pulses. At the shortest Ar+ laser pulses of about 70 ps the dye pulses are as short as 1.1 ps. With Ar+ laser pulses of 200 ps the dye laser pulses contains a broad satellite pulse which contains a large fraction of the total intensity. When a cavity dumper is added to the system one gets dye laser pulses 15–20 ps long with a substructure, which indicates incomplete mode-locking. Well mode-locked 1.5–2.0 ps pulses were obtained in the red part of the dye laser action spectrum, i.e. 620–650 nm for R6G, 595–608 nm for R 110 and 657–662 nm for RB, respectively. Addition of mode-locking dyes also improved the pulse quality at some wavelengths.

    Notes: Summary Compared to wild type cells, strains bearing the pleiotropic regulatory mutations cgr4 or cas1 synthesize apocatalase T at a high rate when grown on high glucose. Like heme-deficient ole3 single mutants, ole3 cgr4 and ole3 cas1 double mutants accumulate no catalase T protein in vivo. This defect introduced by the ole3 mutation is cured by the addition of ALA.     Notes: Summary Two vectors, termed pG63.11 (7.6 Kb) and pHCG3 (9.6 Kb), suitable for yeast transformation have been constructed. The pHCG3 vector has cosmid properties. Both vectors contain a single 3.3 Kb EcoRI-HindIII fragment of yeast origin which carries the yeast URA3 gene (1.1 Kb) and the origin of replication of the 2 µm plasmid (2.2 Kb). They confer ampicillin resistance and they contain 5 unique EcoRI,HpaI,HindIII,BamHI and SalI restriction sites. Cosmid pHCG3 was used to clone the nitrogen fixation (nif) gene cluster of Klebsiella pneumoniae carried by twoHindIII fragments of 17 and 26 Kb, respectively. The resulting cosmid, termed pGPC875 (53 Kb) which conferred a Nif+ phenotype to Escherichia coli, was introduced in yeast by transformation. No acetylene reduction activity was detectable in the transformants. However it was shown that the entire information for nitrogen fixation can be replicated and maintained intact in yeast for more than 50 generations of growth.

    Notes: Summary We have studied the organization and expression of a group of tRNA genes located on a 2,700 base pair portion of the yeast mitochondrial genome between the genetic markers cap (chloramphenicol resistance) and oxil (cytochrome oxidase subunit II). This region is spanned by mitochondrial DNA inserts of two recombinant plasmids, pYm162 and pYm267, which have been extensively mapped and sequenced. This tRNA group is composed of six tRNA genes, coding for tRNA AAR Lys , tRNA AGR Arg , tRNA GGN Gly , tRNA GAY Asp , tRNA AGY Ser , and tRNA CGN Arg . We report the sequence for the majority of the 2,700 base pair region including the genes for all six tRNAs. All six genes are oriented in the same direction and are, therefore, transcribed from the same DNA strand. Further, a comparison of the organization of this region with the analogous region of a related wild type strain shows that the tRNA gene order in the two strains is the same. Five of the six tRNA genes have corresponding transcripts in wild type RNA. Although a potential structural gene for tRNA CGN Arg is present, we do not detect a tRNA CGN Arg gene transcript.

    Notes: Abstract Borrelidin is a specific inhibitor of the threonyl-tRNA-synthethase. A class of dominant borrelidin resistant mutants (BOR1) of Saccharomyces cerevisiae was biochemically characterized. The mutants possess an altered aspartokinase which is insensitive to threonine inhibition. The threonine and homoserine pools in these mutants are 22 times larger than in the wild type. By feeding aspartate under a variety of conditions the homoserine pool is increased even 57 times whilst the threonine pool is reduced.

    Notes: Abstract The effect of the tryptophan analogue 4-methyltryptophan in Saccharomyces cerevisiae has been investigated. 4-Methyltryptophan inhibits the aminoacylation of tryptophan specific transfer ribonucleic acid (tRNATrp). The mode of inhibition is competitive and the analogue is not charged onto tRNATrp. Thus 4-methyltryptophan application depletes the cells from charged tRNATrp. As a consequence cell growth and protein synthesis are strongly reduced. 4-Methyltryptophan is degraded efficiently in culture media inoculated with the wild type strain; the effects of 4-methyltryptophan were therefore found to be transient.

    Notes: Abstract The intracellular levels of adenosine triphosphate and several glycolytic intermediates were determined in Saccharomyces cerevisiae in relation to the presence of the metabolically antagonistic enzymes phosphofructokinase and fructose-1,6-bisphosphatase. Phosphofructokinase is synthesized constitutively in cells grown in the presence of glucose and fructose-1,6-bisphosphatase derepression occurs upon the exhaustion of glucose from the growth medium. Transcriptional regulation of fructose-1,6-bisphosphatase was suggested based on experiments with wild type cells using 8-hydroxyquinoline, a known inhibitor of nuclear transcription, and with the S. cerevisiae mutant strain A364A (ts-136) blocked in the transport of nuclear RNA at non-permissive temperature. The level of phosphofructokinase was reduced more than 25-fold under conditions of high citrate accumulation in an aconitaseless, glutamate requiring mutant strain, MO-1-9B. There was a rapid decrease in the levels of adenosine triphosphate and fructose-1,6-bisphosphate at the end of log-phase of culture growth when both fructose-1,6-bisphosphatase and phosphofructokinase were present in the cells simultaneously. The changes in the levels of key glycolytic intermediates, but not the changes in adenosine triphosphate, during the simultaneous presence of these two enzymes, can be explained without involving any futile cycling.

    Notes: Abstract The yeast, Hansenula wingei has two mating types designated 5 and 21. Cells of each mating type were found to produce mating type-specific sex pheromone which induces sexual agglutinability of the opposite mating type. Crude fractions of these pheromones were prepared by using an Amberlite CG 50 (H+ type) column. The agglutinability-inducing action of the pheromones required glucose as carbon source, but no external nitrogen source. The action of the pheromones was inhibited by 5 μg/ml cycloheximide. The optimum pH for the pheromone action was 4.0. α Pheromones of Saccharomyces cerevisiae and Saccharomyces kluyveri induced sexual agglutinability of 5 mating type cells but did not that of 21 mating type cells. a Pheromones of the Saccharomyces yeasts had no effect on both 5 and 21 mating type cells. The sex pheromones of H. wingei had no effect on the sexual agglutinability of inducible a cells of S. cerevisiae. From the experimental results obtained so far, we propose to call 5 and 21 mating types in H. wingei a and α mating types, respectively.

    Notes: Abstract Scar rings (SR) from scarless cells at the early stages of budding and mature bud scars from Saccharomyces cerevisiae isolated by both chemical and enzymic treatment of cell walls were observed by selected-area electron diffraction (SAED), X-ray diffraction and electron microscopy with simultaneous physico-chemical characterization (including molar mass, intrinsic viscosity and crystallite size) of α-chitin and glucan. The SR, composed of glucan with strong 0.608; 0.397 and 0.293 nm X-ray reflections, was formed at the start of budding. The SAED patterns of α-chitin both in the adjacent circular zone and in the parts of newly formed primary septum (PS), observed when the development of the PS started, did not differ from those of α-chitin in the single mature bud scar. The bud scar consisted of α-chitin, glucan and mannan and their content, as well as the crystallite size of chitin, depended on the mode of preparation.

    Notes: Summary We have carried out experiments aimed at explaining the observed variations in transformation frequencies when Saccharomyces cerevisiae or Saccharomyces carlbergensis are transformed with chimeric plasmids that contain one of 4 possible EcoRI fragments of the yeast 2-μm circle. These plasmids fall into 2 classes when used to transform 2 different yeast his3 auxotrophs, one (strain LL20) harbours indigenous 2-μm circle, and the other (strain YF233) is devoid of this plasmid. Hybrid plasmids containing either the 2.4 mega-dalton (mD) R-form EcoRI fragment (pYF88) or the l.4 mD L-form EcoRI fragment (pYF177) of 2-μm circle transform either of the two hosts at a high frequency (50,000 colonies per Mg in LL20 and 10,000 colonies per μg in YF233). Hybrid plasmids containing the 1.5 mD R-form EcoRI fragment (pYF87) or the 2.5 mD L-form EcoRI fragment (pYF178) of the 2-μm circle transform LL20 at a reduced frequency (6,000–16,000 colonies per μg) and YF233 at extremely low frequencies (1–5 colonies per μg). All plasmids retrieved from strain YF233 that had been transformed with pYF88 or pYF177 were identical to the original transforming plasmid. Of the plasmids retrieved from strain LL20 that had been transformed with pYF87 and pYF178, approximately half had acquired an extra copy of the 2-μm circle. Of the plasmids retrieved from strain LL20 that had been transformed with pYF88 and pYF177, an average of only approximately 13% had acquired an extra copy of 2-μm circle. Taken together, these observations indicate that the transformation of yeast by a plasmid lacking the ability to replicate (pYF87 and pYF1780) occurs by the recombinational acquisition of 1 copy of the host 2-μm circle, which serves to supply the incoming plasmid with missing essential sequences. A comparison of 2-μm circle DNA fragments carried by pYF88 and pYF177 indicates that the region of 2-μm circle required for high frequency transformation is a 1.2 mD segment that is common to the 2.4 mD R-form and 1.4 ml) L-form EcoRI fragments. This region extends from the EcoRI cut site adjacent to the PstI site, through to the end of the inverted repeat. However, the inverted repeat sequence alone is not sufficient to bestow high frequency transformation of yeast.

The mutation named gal11 was recessive, non-allelic to GAL4, GAL80, GAL2, or GAL3, and unlinked to the gene cluster of GAL1, GAL10, and GAL7. It caused a ‘coordinate’ reduction of galactokinase, galactose-1-P uridylyl transferase, and UDP-glucose 4-epimerase by a factor of more than 5, rendering the mutant cells galactose-nonfermenting. The effect of the mutation was manifested not only in cells grown on galactose but also in cells constitutively synthesizing the galactose-metabolizing enzymes.

    Notes: Summary The size of the transcriptional unit of the structural genes for three galactose-metabolizing enzymes which form a cluster on chromosome II in Saccharomyces cerevisiae was studied by the ultraviolet light (UV)-mapping technique. Thus the size of the primary transcripts of GAL7 for galactose-1-phosphate uridylyl transferase, GAL10 for uridine diphosphoglucose 4-epimerase, or GAL1 for galactokinase were estimated to be 0.81 x 106, 1.1 x 106, or 1.3 x 106 respectively. In the light of these data together with the known directions of transcription of the genes, we concluded that each of three genes was transcribed from its own promoter.

    Notes: Abstract As an outcome of a systematic study of excitation kinetics and fluorescence spectra concerning theD′(3π2u )→A′(3π2g ) transitions in homonuclear diatomic halogens and corresponding transitions in interhalogen molecules [1], this paper describes, for the first time, TEA laser action in the three halogen monofluorides ClF, BrF, and IF. Output energies in the mJ range were demonstrated in a fast TEA discharge arrangement at the wavelengths of 284.4, 354.5, and 490.8 nm for ClF, BrF, and IF, respectively. The results suggest parameter scalability and wavelength tunability.

    Notes: Abstract We report the observation that the helical TE N2 laser can effectively be simplified by giving up the use of decoupling elements as well as by abolishing the segmentation of the electrode structure. Although, as a consequence of this simplification, the operating pressure range was slightly decreased, the output power could be improved by roughly 30%, a result which is attributed to the new electrode geometry exhibiting lower inductance and lower damping losses.

    Notes: Abstract The operation of an electrically excited gasdynamic CO laser with closed-cycle gas flow is described. Details of the gasdynamics, the recompression, and the cooling of the flowing gas are given. The electrical discharge produces contaminating gas compounds—in particular CO2—which accumulate in the recirculating gas. The resulting transient behavior of the laser gas composition, the discharge characteristics, and the laser performance are studied and compared with the results obtained with the same CO laser in an open-flow system. It is found that the closed-cycle system may be operated for considerable time if the CO2 contamination is removed from the laser gas.

    Notes: Abstract The solubility of several dopants (Sb, Ga, Bi, In) in laser treated silicon has been investigated. The dopants were introduced by vacuum deposition followed by ruby laser irradiation. Their solubility was determined by Rutherford backscattering spectroscopy measurements in channelling and random conditions. In all cases, a maximum solubilityC S * , much higher than the equilibrium solubility limitC S 0 and independent of the pulsed laser energy density, was found. The values obtained are in good agreement with those calculated from a simple model based on phase diagram considerations, using the relationship: $$C_S^* = \frac{{C_S^0 }}{{k_0 }}k^* ,$$ wherek 0 andk * are the equilibrium and effective distribution coefficients. Finally, the existence of a new solubility limit for a laser treatment is discussed.

    Notes: Abstract A method is described for detection of CO2-laser power variations which requires no optical detector. It is based on plasma-tube impedance changes induced by the laser power flow within the cavity. A constant-current high-voltage power supply and a special equalizer are necessary to obtain a voltage proportional to the laser power. Details for the design of the equalizer are given. The detection scheme works for frequencies up to about 5 kHz and can be used for laser frequency stabilization or signature analysis.

    Notes: Abstract The stability of parallel pin electrode CO2-laser discharges, coupled by carrier diffusion only, against concentration of current in one column and subsequent transition into an arc, is investigated theoretically. A small signal description of the time dependent behaviour of a column with falling characteristic caused by a wall sheath of considerable width is used. It is shown that an inductance in series to each discharge is not able to avoid glow to arc transition, though enlarges the duration of the glow.

    Notes: Abstract Absorption and fluorescence spectra of rarefied vapors ofp-terphenyl, 1,4-bis-[2(5-phenyloxazolyl)]-benzene, 2,5-diphenyl-1,3,4-oxadiazole, 2,5-diphenyl-1,3-oxazole, 2,5-diphenylfuran are measured. Extinction coefficients, quantum yields of fluorescence and one-photon decomposition of molecules are determined for vapors excited by pulsed laser radiation at 266 and 355 nm in the temperature range from 200 to 400°C. It is shown that for all cases studied the one-photon decomposition is only a small part of the total quantum yield of the radiationless deactivation of the fluorescent state. Laser action has, for the first time, been obtained in 2,5-diphenylfuran vapor at 356 nm.

    Notes: Abstract A previous analysis that described loss of gain in a dye solution as a result of photochemical decomposition by pump photons is refined and is extended to the situation where impurities absorb at the pump wavelength. A limitation in the first paper is that it described only a single absorption depth in the medium at a constant pump fluxI p. The present examination includes the fall-off ofI p and integrates the total signal output with depth for a single-pass amplifier operated in the fully saturated gain condition. The equations are simpler, but as before loss of output, expressed in terms of a quantum yield of laser decayQ L, is minimized for a photochemically bleachable dye, and for one with high gain. Pumping in the vicinity of the absorption maximum is also advantageous.

    Notes: Abstract Fluorescence spectra in the near infrared are observed of the dye Eastman 9860 and of new, faster mode-locking dyes. The quantum efficiencies are determined to be small with η≃10−3. The short fluorescence lifetimes are deduced from optical gain measurements. The fluorescence time-constants are found to have the same value as the corresponding absorption recovery times. Even laser action is observed in these fast saturable absorbers previously belieyed to be non-fluorescent.

    Notes: Abstract A laser system capable of generating picosecond pulses of 2 ps duration and peak powers of up to 50 MW, tunable throughout the emission spectrum of the lasing medium is described. The system consists of a cw dye laser which is tuned and is actively mode-locked by means of a novel interferometer. Single-pulse amplification is achieved via a two-stage N2-laser-pumped amplifier arrangement. One of the most attractive features of the system here described is its inherent versatile broad tunability, which should allow, with different lasing media and pump sources, ps pulse generation from 400 to 700 nm using the same interferometer device.

    Notes: Abstract A saturated absorption technique, using an actively stabilized cw dye laser has been developed to measure the saturation intensities of hyperfine transitions in a gaseous medium. Hyperfine lines belonging to theB-X system of molecular iodine have been investigated. The experimental method proposed, provides a means for measurement of the absolute value of the saturation parameter of the transition with an accuracy better than 10%. The behavior of the saturation intensity of some transitions with iodine pressure has been investigated in the range 30 m Torr→90mTorr.

    Notes: Abstract A simple single particle model of a free-electron laser (FEL) amplifier has been used in a computer simulation to determine the maximum fractional conversion of electron kinetic energy to laser energy. The simulation results can be represented by a single universal curve. A simple scaling relationship for the length of the optimized constant period helix together with the universal curve permit one to predict maximum fractional energy conversion for any set of values of initial electron energy, initial laser intensity, magnetic field amplitude, and magnet period.

    Notes: Abstract The spatial coherence function |Γ(s)| of ruby laser pulses has been measured by self-diffraction at a laser-induced grating depending on the distances between two points in the beam profile. The spatial coherence of the laser beam was changed by operating the laser in the fundamental transverse mode, in a transverse multimode and by applying different pump powers. The degree of spatial coherence was experimentally found to be a Gaussian, as described by the Collett-Wolf model. The new method for measuring the spatial coherence function |Γ(s)| is described and possible improvements are discussed.

    Notes: Abstract Tibi grains consist of a unique and very stable symbiotic association of Lactobacillus brevis, Streptococcus lactis and Saccharomyces cerevisiae embedded in a dextran matrix. The structural organization of the grain was examined by light, scanning and transmission electron microscopy. The grain was constituted of an outer compact layer and an inner spongy structure. The outer layer was more densely populated by the microorganisms than the inner layer but dextran, stained on frozen thin sections with fluorescein-conjugated concanavalin A, was more abundant in the inner layer.

    Notes: Abstract After a short period of tolerance, living cells of Saccharomyces cerevisiae were irreversibly damaged by low concentrations of sulfite. The length of the period of tolerance and the rate of the damaging effect depended on the concentration on sulfite, pH-value, temperature, the physiological state of the cells, and incubation time. Inhibitors of protein synthesis and mitochondrial ATP synthesis did not alter the deleterious effect of sulfite on living cells. Furthermore, cell damage leading to inhibition of colony formation occured under aerobic as well as under anaerobic conditions. Prior to cell inactivation sulfite induced the formation of respiratory deficient cells. The active agent was shown to be SO2.

Protein synthesis, fermentative and respiratory activity and catabolite repression were not affected. Elimination of catabolite inactivation by the addition of PMSF revealed that catabolite repression started at different times for different enzyme.

    Notes: Abstract The resistance of exponentially growing yeast cells to killing by exposure to 52°C increased markedly as the growth temperature was increased. Identical killing curves were obtained for cells suspended in growth medium or in 0.9% saline. Cells resistant to killing at 52°C were quite sensitive to killing at slightly higher temperatures. These results suggest a primary role for membrane damage in the mechanism of heat killing.

    Notes: Summary Effect of yeast autolyzate on the growth of vineless pea plant was investigated. The yield was increased remarkably when yeast autolyzate was added, as compared with the control area in which the plants received only mineral fertilizer solution. The yield stimulation was the highest where 0.1 ppm of the autolyzate was added, the yield increasing by more than 80%. The reason why the yields were lower in the areas where the amount of yeast autolyzate added was incrased to 1 ppm and to 10 ppm, was presumed to be due to the fact that the substances may have been absorbed, decomposed, and utilized by soil microorganisms.
