    Beschreibung: Drained peatlands are hotspots for carbon dioxide (CO2) and nitrous oxide (N2O) emissions. In Niedersachsen, 69 % of the peatland area is used for agriculture and is responsible for 11 % of the annual CO2 and N2O emissions. Raising peatland water tables to 0.30 m below surface could reduce emissions. In this study, three water regulation methods were tested to raise water table and to maintain grassland use on bog peatlands. In the first method, weirs were installed to block ditches and retain surface runoff within the blocking ditches. For the second method, ditch impoundment, additional water was pumped into the ditches creating constant ditch water levels. The third method, subsurface irrigation, further increased the water table within the grassland by allowing water to flow from the ditch into the bog area via subsurface pipes. Ditch impoundment and subsurface irrigation methods effectively raised the bog water tables. In the summer 2020, the mean water tables for ditch impoundment and subsurface irrigation reached 0.32 m and 0.23 m below surface, while the water table with blocking ditches was much lower with 0.61 m below surface. Overall, the subsurface irrigation method successfully raised the bog water tables and reduced subsidence. Negative aspects of this method are the costs for pipes and weirs, the reduced trafficability and the increased water demand.
    Beschreibung: Entwässerte Moore sind Hotspots für die Emission von Kohlendioxid (CO2) und Lachgas (N2O). In Niedersachsen werden 69 % der Moorflächen landwirtschaftlich genutzt und sind für 11 % der jährlichen CO2- und N2O-Emissionen verantwortlich. Eine Anhebung des Moorwasserstandes auf 0.30 m unter Geländeoberkante (GOK) könnte die Emissionen verringern. In dieser Studie wurden drei Methoden zur Wasserregulierung getestet, um Moorwasserstände anzuheben und die Grünlandnutzung in einem Hochmoor aufrechtzuerhalten. Für den Grabenanstau wurden Wehre installiert, um Gräben zu blockieren und den Oberflächenabfluss zurückzuhalten. Beim Grabeneinstau wird ein konstanter Wasserspiegel in den Gräben durch zugepumptes Wasser generiert. Bei der Unterflurbewässerung wird der Moorwasserstand weiter erhöht, in dem Wasser aus dem Graben über unterirdische Rohre in die Fläche geleitet wird. Mit dem Grabeneinstau und der Unterflurbewässerung konnten die Moorwasserstände erhöht werden. Im Sommer 2020 erreichten die mittleren Moorwasserstände beim Grabeneinstau und der Unterflurbewässerung 0.32 m und 0.23 m unter GOK, während der Moorwasserstand beim Grabenanstau mit 0.61 m unter GOK deutlich niedriger lag. Insgesamt konnten mit der Unterflurbewässerung Moorwasserstände erfolgreich angehoben und die Höhensackung der Mooroberfläche verringert werden. Negative Aspekte dieser Methode sind die Kosten für Rohre und Wehre sowie die Pumpe, die eingeschränkte Befahrbarkeit und ein erhöhter Wasserbedarf.

    Beschreibung: In Deutschland sind 92 % der Moore entwässert. Diese Flächen verursachen jedes Jahr etwa 53 Millionen Tonnen CO2-Äquivalente, was 6,7 % der gesamten Treibhausgasemissionen in Deutschland entspricht. Der überwiegende Teil (83 %) dieser Emissionen aus Moorböden resultiert aus landwirtschaftlichen Flächen. Ziel der 2021 veröffentlichten Nationalen Moorschutzstrategie ist es, die jährlichen Treibhausgasemissionen aus Moorböden bis 2030 um mindestens 5 Millionen Tonnen CO2-Äquivalente zu reduzieren. Um die Treibhausgas-Reduktionsziele zu erreichen, müssten in Deutschland, je nach Maßnahmen und Moortyp, rund 150.000 ha Moorflächen bis 2030 vernässt werden. Angesichts der langwierigen Prozesse der Projektentwicklung, der Genehmigungsverfahren und der technischen Umsetzung müsste aber umgehend und umfassend mit der Realisation begonnen werden. Dieses Ziel ist aufgrund der Freiwilligkeit der Maßnahmen derzeit nicht ansatzweise realistisch und erfordert massive, vor allem finanzielle Maßnahmen.
    Beschreibung: In Germany, 92 % of all peatlands are drained. These areas are responsible for approximately 53 million metric tons of CO2 equivalents each year, or 6.7 % of the total greenhouse gas emissions in Germany. The majority (83 %) of these emissions from peatlands result from agricultural used peatlands. The goal of the National Peatland Protection Strategy, published in 2021, is to reduce annual greenhouse gas emissions from peatland soils by at least 5 million metric tons of CO2 equivalents by 2030. To achieve the greenhouse gas reduction targets, around 150,000 hectares of peatland would need to be rewetted in Germany by 2030, depending on the rewetting technique and peatland type. However, in view of the long processes of project development, approval procedures and technical implementation, realization would have to begin immediately and comprehensively. Due to the voluntary nature of rewetting measures, this goal is not remotely realistic at present and requires massive measures, especially adequate funding.

    Beschreibung: Der Torfeinsatz in den Erden und Substraten des Gartenbaus ist mit CO2-Emissionen verbunden und eine Reduzierung durch den Ersatz mit nachwachsenden Rohstoffen daher spätestens seit der Klimaübereinkunft Paris 2015 eine gesellschaftliche Herausforderung. Torfmoose (Sphagnum spp.) stellen dabei einen der wenigen Ersatzstoffe dar, die die hohen qualitativen Anforderungen an Kultursubstrate erfüllen können. Mit der Einrichtung des Sphagnum farming Projektes „Barver“, Landkreis Diepholz, soll von 2018 bis 2021 die Torfmooskultivierung auf Hochmoorgrünland in der Praxis getestet werden. Voraussetzung zur Anlagenplanung war eine detaillierte Grundlagenerhebung des inhomogenen Torfkörpers, auf deren Basis der Oberbodenabtrag der Betriebsfläche (1 ha) und die Einrichtung des Bewässerungssystems erfolgte. Die Kulturfläche wurde abschließend manuell mit Torfmoosfragmenten von unterschiedlichen Spenderflächen beimpft. Für die erforderliche Polderbewässerung musste ein Kompromiss aus gering überstauten Schwarztorfflächen und flurnah bewässertem Weisstorfbereich akzeptiert werden. Schnell und flächig einsetzendes Wachstum der Torfmoosfragmente signalisiert, dass regionales Sphagnum farming auf degradiertem, drainierten Hochmoorgrünland bei funktionierender Wasserversorgung möglich ist. Die jetzt folgende Projektphase dient der Ermittlung der optimalen Bestandsentwicklung. Daneben sind technische und rahmenpolitische Fragen bis zu einer wirtschaftlich tragfähigen landwirtschaftlichen Torfmooskultivierung zu klären.
    Beschreibung: The use of peat in growing media for horticulture is associated with CO2 emissions and a reduction through replacement with renewable raw materials has therefore been a social challenge since Paris Agreement 2015 at the latest. Peat mosses are one of the few substitutes that can meet the high quality requirements for growing media. With the establishment of the Sphagnum farming project “Barver”, Diepholz district (Niedersachsen, Germany), peat moss cultivation on raised bog grassland is to be tested in practice from 2018 to 2021. A fundamental prerequisite of planning of the one hectare study site was an in-depth basic evaluation of its inhomogeneous peat body, being the base of site preparation and irrigation system. Finally, the study site was inoculated manually with fragmented peat mosses of different origins. The necessary watering of the polder had to be a trade-off between flood irrigation of a black peat area and subirrigation of the adjacent white peat zone. Rapidly and widespread growth of peat mosses indicate, that the regional cultivation of Sphagnum on degraded and drained bog grassland with a working irrigation system is feasible. The now starting phase is aiming on an optimal developed Sphagnum stock. Aside, there are technical challenges and administrative obstacles to be overcome before cultivation of peat mosses will give economically viable agricultural crops.

    Beschreibung: Die Untersuchungen zu Verbreitung und Standort der Blasenbinse (Scheuchzeria palustris) in Nordostdeutschland werden vorgestellt. In den Jahren 2000 bis 2005 sowie 2020 wurden 29 Moore (27 in Mecklenburg-Vorpommern, 2 in Brandenburg) mit nach 1950 gemeldeten Funden der Art untersucht. Einzugsgebiet, Moorfläche, Mooraufbau, hydrogenetischer Moortyp, Flora und Vegetation sowie Hydrologie der untersuchten Niedermoore werden präsentiert. Die Ursachen für den Rückgang der Vorkommen werden diskutiert. Die Untersuchungen haben ergeben, dass dem Wasserfaktor für die Erhaltung der Blasenbinsen-Vorkommen eine entscheidende Rolle zukommt. Wasserstandschwankungen müssen in Bezug zur Mooroberfläche mit 1 bis 2 dm sehr gering bleiben. Durch die Fähigkeit zur Oszillation der Mooroberfläche kann ein Überstau weitgehend vermieden werden. Voraussetzung hierfür ist ein mächtiger Moorkörper mit oberflächennah anstehenden wässrigen Torfmoos-Torfen.
    Beschreibung: In this paper studies on the distribution and habitat of Scheuchzeria palustris in Northeastern Germany are presented. Twenty-nine fens (27 in Mecklenburg-Vorpommern, 2 in Brandenburg) were surveyed between 2000 and 2005 and additionally in 2020. The surveyed sites were all sites where this species had been observed since 1950. The catchment area, fen area, stratigraphy, hydrogenetic mire types, flora and vegetation as well as the hydrology for each site are shown. The reasons for the decline in populations are discussed. This study shows that hydrology plays a decisive role in maintaining Scheuchzeria palustris populations. The water level should be stable in relation to the peat surface with fluctuations no larger than 1 to 2 dm. Due to the shrinking and swelling capacity of peat, flooding can be largely avoided. The prerequisite for this is a thick peat body with Sphagnum peat at the surface.

    Beschreibung: In den Jahren 2017 bis 2019 wurde das Flächennaturdenkmal „Rahmannsmoor“ in Krakow am See bezüglich der Lepidopteren, Ephemeropteren und Trichopteren untersucht. Parallel dazu sind moorstratigraphische Untersuchungen vorgenommen worden. Das in einer kuppigen Endmoränenlandschaft liegende Kesselmoor hat eine Moormächtigkeit von mehr als 5 Metern und ist über einem sandigen Untergrund aufgewachsen. Es befindet sich auf Grund einer längeren Trockenperiode in einer Phase der Stagnation, wobei mächtige, gering zersetzte Torfmoos-Torfe die Voraussetzungen für ein erneutes Moorwachstum geben. Vegetationskundlich wird das Rahmannsmoor von Torfmoosen, Wollgräsern, Moosbeeren und Rosmarinheiden geprägt. Es konnten 100 Taxa an Nachtfaltern, 13 Köcherfliegenarten und eine Eintagsfliegenart nachgewiesen werden. Tyrphobionte/-phile Faunenelemente sind bei den untersuchten Gruppen eher selten, wobei die „Restbiozönose“ noch zahlreiche eurytopere Arten der Moore beinhaltet. Das Moor soll entkusselt werden, um die baumfreien Flächen zu erweitern und die Verdunstung zu senken.
    Beschreibung: From 2017 to 2019, the natural monument “Rahmannsmoor” in Krakow am See was surveyed for lepidopterans, ephemeropterans and trichopterans. At the same time the stratigraphy was investigated. The kettle-hole mire, located in a dome-shaped end moraine landscape, is more than five meters deep and has a sandy subsoil. Peat growth is stagnated during dry years. Thick, slightly decomposed peat is a good prerequisite for renewed bog growth. In terms of vegetation, the Rahmannsmoor is characterised by Sphagnum mosses, cotton grasses, bog bilberry and rosemary heath. One hundred taxa of moths, 13 species of Trichoptera and one species of Ephemeroptera were recorded. Tyrphobiont and tyrphophilous faunal elements are rather rare in the studied groups, whereas the “residual biocoenosis” still contains numerous eurytope species of the mires. The shrubs will be removed from the peatland in order to enlarge the open areas and to reduce evaporation.

    Beschreibung: Die Lehrausbildung zum Forstwirt im Fachbereich „Naturschutz/Landschaftspflege“ war 2018 bis 2021 auf die Umsetzung der Moorrevitalisierung im Fauna-Flora-Habitat-Gebiet „Moorwaldgebiet Großdittmannsdorf“ gerichtet. Erreicht wurde eine lokale Verbesserung des Waldinnenklimas (Luftfeuchtigkeit, Kühlung), der Grundwasserneubildung und Stabilisierung des Bodenwasserhaushaltes mit vorteilhaften Wirkungen auf die biologische Vielfalt (Feuchtbiotope, Moorbiozönose).
    Beschreibung: From 2018 to 2021, the apprenticeship training for foresters in the field of “nature conservation/landscape management” was focused on the implementation of measures for peatland revitalisation in the Habitats Directive Site “Moorwaldgebiet Großdittmannsdorf ”. A local improvement of the internal forest climate (humidity, cooling), groundwater recharge and stabilisation of the soil water balance with beneficial effects on biodiversity (wetland biotopes, bog biocoenosis) was achieved.

    Beschreibung: Entlang der Donau zwischen Ulm und Ingolstadt liegen verschiedene Flusstal-Niedermoore, die sich während und nach der letzten Eiszeit entwickelt haben. Nahezu alle Moorböden sind in den vergangenen 200 Jahren entwässert und immer intensiver genutzt worden, weshalb fortwährend erhebliche Mengen an Klimagasen entweichen. Am Beispiel von acht Niedermoorrenaturierungen werden aus 30-jähriger Erfahrung die unterschiedlichen Hemmnisse und Lösungsansätze dargestellt. Daraus gewonnene Erkenntnisse könnten auch für andere Regionen nützlich sein. Es wird aufgezeigt, wie klimawirksamer Moorschutz rascher und effektiver gelingen kann, und welche Voraussetzungen dafür geschaffen werden müssen. Dass man beim Klimaschutz auch durch Moorbodenschutz schneller zu Ergebnissen kommen muss, zeigt die 26. Weltklimakonferenz 2021 (COP26) in Glasgow.
    Beschreibung: Along the Danube between Ulm and Ingolstadt, various river valley fens developed after the last ice age. Almost all peatlands have been drained over the past 200 years and intensively used resulting in significant greenhouse gase emissions. Due to the example of eight peatland restoration projects, obstacles and solutions are presented from over 30 years of experience that could be useful for other regions. Conclusions are drawn as to how peatlands can be protected more quickly and effectively to lower greenhouse gas emissions. Additionally, the conditions that need to be created for peatland protection are identified. The 26th United Nations Climate Change Conference (COP26) in Glasgow demonstrated the need to achieve quick results in climate protection through peatland protection to avoiding further global warming.

    Beschreibung: Canada has undertaken commitments to recognize the rights of Indigenous Peoples in fisheries through policies and agreements, including Integrated Fishery Management Plans, the Reconciliation Strategy, and Land Claim Agreements (LCAs). In addition to recognizing rights, these commitments were intended to respect geographic adjacency principles, to enhance the economic viability of Indigenous communities, and to be reflective of community dependence on marine resources. We examined the determinants of quota allocations in commercial fisheries involving Nunatsiavut, Northern Labrador, the first self-governing region for the Inuit peoples in Canada. It has been argued that current fishery allocations for Nunatsiavut Inuit have not satisfied federal commitments to recognize Indigenous rights. Indicators that measure equity in commercial allocations for the turbot or Greenland halibut (Reinhardtius hippoglossoides) and northern shrimp (Pandalus borealis) fisheries were identified and assessed. In these two cases, historical allocations continue to predominate for allocations based upon equity or other social or economic considerations. We illustrate equity-enhancing changes in the quota distribution under scenarios of different levels of inequality aversion, and we make qualitative assessments of the effects of these allocations to Nunatsiavut for socioeconomic welfare. This approach could benefit fisheries governance in Northern Labrador, where federal commitments to equity objectives continue to be endorsed but have not yet been integrated fully into quota allocations.

Milkfish larvae could be reared successfully without the use of trochophore larvae of oysters as feed during the first few days.     Beschreibung: Round scad exploration by purse seine in the waters of western Philippines was conducted from April 22 to May 7, 1998 for a period of five (5) fishing days with a total catch of 7.3 tons and an average of 1.5 tons per setting. Dominant species caught were Decapterus spp. having 70.09% of the total catch, followed by Selar spp. at 12.66% and Rastrelliger spp. 10.70%. Among the Decapterus spp. caught, D. macrosoma attained the highest total catch composition by species having 68.81% followed by D. kurroides and D.russelli with 0.31% and 1.14% respectively. The round scad fishery stock was composed mainly of juvenile fish (less than 13 cm) and Age group II (13 cm to 14 cm). Few large round scad at Age group IV and V (20 cm to 28 cm) stayed at the fishery. Other fishes caught were: Auxis rochei (0.85%), A. thazard (0.12%), Caranx spp. (0.45%), Emmilichthys nitidus (0.58%), Euthynnus affinis (0.42%), Leiognathus ruconius (0.58%), Loligo sp. (0.31%), Megalaspis cordyla (0.09%), Rastrelliger spp. (10.70%), Sardinella longiceps (0.03%), Scomberoides lysan (0.24%), Selar spp. (12.66%), Sphyraena spp. (0.90%), Thunnus albacares (0.96%) and others (1.02%). Tuna and tuna like fishes such as yellowfin tuna, eastern little tuna, bullet tuna, frigate tuna and oceanic squid are distributed in the upper latitudes of the survey area. On the other hand, round scads, big-eyed scads and Indian mackerels are dominantly present in the lower latitudes of the survey area.

    Beschreibung: An exploratory tuna longline fishing survey was conducted using the research and training vessels of the Southeast Asian Fisheries Development Center, the 1,178 GT MV SEAFDEC and the Bureau of Fisheries and Aquatic Resources, the 165 GT MV MAYA-MAYA in the South China Sea Waters, West of the Philippines from April to May, 1998. A total of 3,796 hooks was set in sixteen (16) fishing stations. There were no tuna caught during the entire survey but only minor and irrelevant species like the Pacific lancetfish, sharks and an opah species, Lampris guttatus. The important fishing and oceanographic factors during the survey and other research results on longline are described and analyzed. Additional longline studies within and near the Philippines territorial waters are also presented to substantiate the research results of the joint SEAFDEC/BFAR resource exploratory.

    Beschreibung: Cholera is an acute specific infection caused by the organism, Vibrio cholera. Diagnosis may be confirmed by the presence of large numbers of the comma-shaped bacilli on direct microscopic examination of a fecal or vomitus smear, and by the isolation of the organism on culture. Fish and shellfish have been identified as vehicles of cholera. Large numbers of V. cholera must usually be ingested to cause cholera. Thus problems often occur when poor handling and inadequate refrigeration have allowed the organism to multiply. Presented in the paper is the methodology of determining the presence of Vibrio cholera in fish and fishery products.

    Beschreibung: Fishery statistics in Brunei Darussalam have been collected by the Department of Fisheries since the 1940s. In the early 1980s, a number of statistics were collected, such as catch and effort data of artisanal fishermen, aquaculture statistics, processing statistics as well as data from major wet markets focusing on the amount, prices of marketed fish either from the local fishermen or imported. In 1984, the collection of statistics on commercial fishing was started. The statistics have been used in the formulation of fishery management and development policies as well as for sectoral development. Following a brief account of the responsibility and statistic collections of the Department of Fisheries, an examination is made of employment in the fishery sector, fisheries production, fish marketed and the GDP for the fishery industry. Fisheries in Brunei Darussalam is a very healthy industry, where production is well below the maximum allowable harvestable limit of 20,000 tons at 30%. The Department is embarking to increase production from the capture fishery to reduce the country's dependence on imported fish; the same applies for aquaculture, in order to complement production from the capture fisheries. The processing sector is also increasing in importance, especially with the increase in the number of capture fishery licenses, and the demand for quality and value-added products.

The paper provides the methodology in the determination of the amount of sodium chloride in fish jelly products. Instructions for sample preparation and the reagents needed are presented. Detailed procedures in the determination of the salt in a sample are provided.

    Beschreibung: Food poisoning due to Vibrio parahaemolyticus is a food-borne infection resulting from the ingestion of a large number of this organism (about 106-109 viable cells). The major symptoms are diarrhea and abdominal pain with headache, fever, and vomiting also occurring. The organisms are excreted during the acute stage of the illness after which they decrease rapidly. The differentiation of V. parahaemolyticus from other pathogenic species of Vibrio is based mainly on salt tolerance, Voges-Proskauer reaction, fermentation of sucrose, and growth at 43°C. Presented in the paper is the methodology of determining the presence of Vibrio cholera in fish and fishery products.

    Beschreibung: Zur Wiedervernässung von Hochmooren wird ein Staukörper aus gewachsenem Hochmoortorf gefordert. Mit repräsentativen Werten der gesättigten Wasserdurchlässigkeit wurden für statische Bedingungen Berechnungen der Sickerwasserverluste in Abhängigkeit unterschiedlicher Staukörpermächtigkeiten vorgenommen. Folgende Ergebnisse lassen sich aus diesen Modellen ableiten: 1. Staukörpermächtigkeiten 〉 0,5 m reduzieren die Sickerwasserverluste nur noch unwesentlich. 2. Erst Humositätsgrade H ≥ 7 nach v. POST bewirken ausreichenden Wasserstau. 3. Schrumpfrisse im ausgetrockneten Staukörper aus stark zersetztem Hochmoortorf können die Stauwirkung beeinträchtigen. Eine Staukörpermächtigkeit von 0.5 m sollte daher nicht unterschritten werden. Als Verdunstungsschutz werden mindestens 0,3 m Bunkerde bzw. Weißtorf über dem Staukörper gefordert. 4. Unter klimatisch ungünstigen Bedingungen ist ein Grundwasseranschluß des Staukörpers erforderlich.
    Beschreibung: Rewetting requires an impermeable layer of undisturbed raised-bog peat. For static conditions calculations of seepage dependent on the thickness of the impermeable layer have been carried out by means of representative data of saturated water permeability. The results of these model calculations are: 1. An increase of the impermeable ayer over 0,5 m insignificantly reduces the seepage. 2. Only a degree of decomposition H 2:7 according to v. POST causes a sufficient water rise. 3. An impermeable layer of highly decomposed raised-bog peat can be destroyed by crack formation after drying up. Therefore, the thickness of this layer should not be less than 0,5 m. A top spit or white peat layer of at least 0,3 m is required to prevent evaporation. 4. Under unfavourable climatic conditions contact of the impermeable layer with the ground water is necessary.

    Beschreibung: Es wird über 5 Jahrzehnte Erfahrungen mit deutschem Sandmischkulturboden − einem anthropogenen Bodentyp − berichtet. Aus dem Ertragsverlauf werden die bodenbildenden Prozesse − Setzung, Homogenisierung, Humifizierung − deutlich. Der jeweilige Grad der Bodenentwicklung − von der rohen Mischung aus Torf und Sand zum ± humosen Sand − kann durch bodenchemische Untersuchungen z.B. Schweretrennung in CCl4‚C/N-Verhältnis‚ N-Fraktionierung, aktuelle/potentielle KAK dargestellt werden. Solche Untersuchungen können der objektiven Bewertung von Neukulturen und zur Steuerung ihrer Bodenentwicklung dienen.
    Beschreibung: It is reported on experiences with German sand-mix-culture soils within 50 years. The soil-forming processes − subsidence, homogenization, humification − are correlated with the development of the yields. The corresponding degrees of soil development − from a rough mixture of peat and sand to a ± humic sandy soil − can be shown by soil-chemical analyses like separation of particles with different densities in CCl4, C/N-ratio, N-fractionation, actual/potential CEC. Such investigations can be useful for the actual estimation of new cultures and the control of their development.

    Beschreibung: In den Sanduntergrund einschneidende Entwässerungsgräben behindern eine Wiedervernässung von teilabgetorften Hochmooren. In Modellversuchen wurden verschiedene Materialien (Schwarztorf, Bunkerde, Bentonit) auf ihre Eignung als Material zur Grabenabdichtung überprüft. Dabei wurden verschiedene Abdichtungsprozesse - Verdichtung des Abdichtmaterials, - SenkundäreAbdichtumgdes Sanduntergrundes durch infiltriertes Material (mechanische Filterwirkung) untersucht. In den Modellversuchen erwiesen sich die Varianten "Schwarztorf, verdichtet" und "Sand/Bentonit-Gemisch" als praktikable Verfahren zur Grabenabdichtung. Sekundäre Abdichtungsprozesse konnten bei einigen Materialien (Schwarztorf-Suspension; Bunkerde) beobachtet werden, sie erwiesen sich bei kurzfristiger Betrachtung (10 Jahre) jedoch als zu uneffektiv.
    Beschreibung: Ditches cut into the sandy subsoil impede a rewetting of cutover peatlands. Different materials (black peat, top spit, bentonite)were tested for their suitability of sealing these ditches. Different sealing processes were examined - compaction of the material, - secondary sealing of the sandy subsoil by clogging with infiltrated material. The treatments "black peat-compacted" and "sand/bentonite-mixture"showed the best sealing properties. Secondary sealing by mechanical filtering was also observed for some other materials ("black peat-suspension", "top spit"), however the process of mechanical filtering is too ineffective in the short-term view (~10 years).

    Beschreibung: The use of molecular biological approaches has defined new mechanisms that store information in the mammalian nervous system. Environmental stimuli alter steady-state levels of messenger RNA species encoding neurotransmitters, thereby altering synaptic, neuronal, and network function over time. External or internal stimuli alter impulse activity, which alters membrane depolarization and selectively changes the expression of specific transmitter genes. These processes occur in diverse peripheral and central neurons, suggesting that information storage is widespread in the neuraxis. The temporal profile of any particular molecular mnemonic process is determined by specific kinetics of turnover and by the geometry of the neuron resulting in axonal transport of molecules to different synaptic arrays at different times.     Beschreibung: The long-term effects of excitotoxic lesions in the nucleus basalis magnocellularis of the rat were found to mimic several neuropathological and chemical changes associated with Alzheimer's disease. Neuritic plaque-like structures, neurofibrillary changes, and neuronal atrophy or loss were observed in the frontoparietal cortex, hippocampus, amygdala, and entorhinal cortex 14 months after the lesions were made. Cholinergic markers in neocortex were reduced, while catecholamine and indoleamine metabolism was largely unaffected at this time.     Beschreibung: A mutation in the white gene of Drosophila mauritiana that results from insertion of the transposable element mariner is genetically unstable in both germ cells and somatic cells. Somatic instability is indicated by the occurrence of animals having mosaic eyes with patches of pigmented cells on a peach-colored background. Normally uncommon, the frequency of mosaicism is so greatly enhanced in a particular mutant strain that virtually every animal in the strain is an eye-color mosaic. The molecular basis of the mosaicism is the excision of the mariner element from its location in the DNA of the white gene in somatic cells. The phenomenon results from a single dominant genetic factor located in chromosome 3.     Beschreibung: Thirty years have elapsed since Wald and his colleagues showed that 11-cis retinal was isomerized to all-trans when rhodopsin was bleached, yet little has been understood about the reverse process that generates 11-cis retinal for rhodopsin regeneration. It is not known whether the isomerization is enzyme-mediated, whether it occurs in the pigment epithelium or in the retina, or whether retinal, retinol, or a retinyl ester is the vitamin A compound that is isomerized. Radiolabeled all-trans retinol and high-performance liquid chromatography have now been used to demonstrate the existence of an eye-specific, membrane-bound enzyme (retinol isomerase) that converts all-trans to 11-cis retinol in the dark. By means of expression cloning in Escherichia coli, fusion proteins were derived from specific fragments of a complementary DNA encoding the mouse alpha-chain, allowing the mapping of the toxin-binding site to residues 160-216 and the main immunogenic region to residues 6-85.     Beschreibung: A new class of excitatory premotor interneurons that are important in the generation of locomotion in the lamprey has now been described. In the isolated spinal cord, these neurons act simultaneously with their postsynaptic motoneurons during fictive swimming. They are small and numerous, and they monosynaptically excite both motoneurons and inhibitory premotor interneurons. The excitatory postsynaptic potentials are depressed by an antagonist of excitatory amino acids. These interneurons receive reticulospinal input from the brain stem and polysynaptic input form skin afferents.     Beschreibung: Recordings have been made of changes in intracellular calcium ion concentration ([Ca2+]i) that can be attributed to the operation of an electrogenic, voltage-dependent sodium-calcium (Na-Ca) exchanger in mammalian heart cells. Guinea pig ventricular myocytes under voltage clamp were perfused internally with fura-2, a fluorescent Ca2+-indicator, and changes in [Ca2+]i and membrane current that resulted from Na-Ca exchange were identified through the use of various organic channel blockers and impermeant ions. Depolarization of cells elicited slow increases in [Ca2+]i, with the maximum increase depending on internal [Na+], external [Ca2+], and membrane voltage. Repolarization was associated with net Ca2+ efflux and a decline in the inward current that developed instantaneously upon repolarization.     Beschreibung: Among the major parasitic infections, schistosomiasis may be the most promising candidate for human vaccination. Information about mechanisms of immunity, gained mainly from experimental models but likely to be relevant to human infection, indicates a dynamic balance between protective and regulatory (blocking) mechanisms. Besides cell-mediated responses leading to macrophage activation, antibody-dependent cell-mediated cytotoxicity systems involving precise antibody isotypes and nonlymphoid cells (mononuclear phagocytes, eosinophils, and platelets) appear to be essential effectors of immune attack. The slow development of immunity in humans seems related to the production of antibodies that cross-react with schistosomulum surface antigen and block the binding of antibodies of the effector isotype. Schistosomes that survive in the bloodstream and produce chronic infections may evade the immune system as a result of intrinsic changes in membrane susceptibility and of transient expression of target antigens; at other stages of the parasite life cycle, cross-reactive molecules may be secreted that play an essential role in the induction of immunity. Several schistosome proteins have been characterized as candidates for vaccination.     Beschreibung: The functional organization of the cerebral cortex is modified dramatically by sensory experience during early postnatal life. The basis for these modifications is a type of synaptic plasticity that may also contribute to some forms of adult learning. The question of how synapses modify according to experience has been approached by determining theoretically what is required of a modification mechanism to account for the available experimental data in the developing visual cortex.     Beschreibung: Expression of c-myb proto-oncogene messenger RNA (mRNA) and protein has been detected principally in tumors and in normal tissue of hematopoietic origin. In each hematopoietic lineage examined, expression of the c-myb gene is markedly downregulated during hematopoietic maturation. However, the mechanism by which differential expression of the c-myb gene is regulated is not known. In murine B-lymphoid tumor cell lines, the amount of steady-state c-myb mRNA is 10 to more than 100 times greater in pre-B cell lymphomas than in B cell lymphomas and plasmacytomas. The downregulation of c-myb mRNA correlates with events at the pre-B cell-B cell junction. Differential expression of c-myb mRNA levels detected between a pre-B cell lymphoma and a mature B cell lymphoma is now shown to be mediated by a block to transcription elongation in the first intron of the c-myb locus. In addition, this developmentally regulated difference in transcriptional activity is correlated with alterations in higher order chromatin structure as reflected by changes in the patterns of hypersensitivity to deoxyribonuclease I at the 5' end of the c-myb transcription unit.     Beschreibung: Cleavage of the peptide bonds of preprosomatostatin at basic residues near the carboxyl terminus yields somatostatin-14, somatostatin-28, and somatostatin-28 (1-12). However, little is known about the molecular forms derived from the amino terminal portion of the precursor, even though this part of the prohormone is highly conserved through evolution. By using an antibody against the amino terminus of prosomatostatin, a decapeptide with the structure Ala-Pro-Ser-Asp-Pro-Arg-Leu-Arg-Gln-Phe, corresponding to preprosomatostatin (25-34), was isolated from the endocrine portion of the rat stomach, the gastric antrum. The antral decapeptide may represent a bioactive product generated from prosomatostatin after a monobasic cleavage similar to that involved in the formation of somatostatin-28.     Beschreibung: Intraperitoneal administration of human recombinant interleukin-1 (IL-1) to rats can increase blood levels of corticosterone and adrenocorticotropic hormone (ACTH). The route by which IL-1 affects pituitary-adrenal activity is unknown. That the IL-1-induced pituitary-adrenal activation involves an increased secretion of corticotropin-releasing factor (CRF) is indicated by three lines of evidence. First, immunoneutralization of CRF markedly attenuated the IL-1-induced increase of ACTH blood levels. Second, after blockade of fast axonal transport in hypothalamic neurons by colchicine, IL-1 administration decreased the CRF immunostaining in the median eminence, indicating an enhanced release of CRF in response to IL-1. Third, IL-1 did not stimulate ACTH release from primary cultures of anterior pituitary cells.     Beschreibung: Exposure to bacterial endotoxins has long been known to stimulate the release of anterior pituitary hormones; administration of endotoxin was at one time a common clinical test of anterior pituitary function. Endotoxin is a potent stimulus for production of the endogenous pyrogenic protein, interleukin-1 (IL-1), by macrophages and monocytes. The possibility that IL-1 has a direct effect on the secretion of hormones by rat pituitary cells in a monolayer culture was investigated. Recombinant human IL-1 beta stimulated the secretion of adrenocorticotropic hormone, luteinizing hormone, growth hormone, and thyroid-stimulating hormone. Increased hormone secretion into culture supernatants was found with IL-1 concentrations ranging from 10(-9) M to 10(-12) M. Prolactin secretion by the monolayers was inhibited by similar doses. These concentrations of IL-1 are within the range reported for IL-1 in serum, suggesting that IL-1 generated peripherally by mononuclear immune cells may act directly on anterior pituitary cells to modulate hormone secretion in vivo. Incubation of IL-1 solutions with antibody to IL-1 neutralized these actions.     Beschreibung: Elevation of glucose transport is an alteration common to most virally induced tumors. Rat fibroblasts transformed with wild-type or a temperature-sensitive Fujinami sarcoma virus (FSV) were studied in order to determine the mechanisms underlying the increased transport. Five- to tenfold increases in total cellular glucose transporter protein in response to transformation were accompanied by similar increases in transporter messenger RNA levels. This, in turn, was preceded by an absolute increase in the rate of glucose transporter gene transcription within 30 minutes after shift of the temperature-sensitive FSV-transformed cells to the permissive temperature. The transporter messenger RNA levels in transformed fibroblasts were higher than those found in proliferating cells maintained at the nonpermissive temperature.     Beschreibung: Li-Fraumeni syndrome is manifested in a variety of neoplasms that are transmitted in a dominantly inherited pattern. The noncancerous skin fibroblasts of family members exhibit a unique characteristic of being resistant to the killing effect of ionizing radiation. A three- to eightfold elevation in expression of c-myc and an apparent activation of c-raf-1 gene have been observed in these noncancerous skin fibroblasts.     Beschreibung: The major late transcription factor (MLTF) is a 46-kilodalton polypeptide that specifically binds to and activates transcription from the major late promoter of adenovirus. The presence of this promoter-specific transcription factor in uninfected HeLa cell extracts suggests that MLTF is also involved in the transcription of cellular genes. This report demonstrates that MLTF specifically stimulates transcription of the rat gamma-fibrinogen gene through a high-affinity binding site. Stimulation of transcription by MLTF was not dependent on the exact position of the MLTF binding site with respect either to the transcription initiation site or to adjacent promoter elements.     Beschreibung: A wide variety of human tumors contain an amplified or overexpressed erbB-2 gene, which encodes a growth factor receptor-like protein. When erbB-2 complementary DNA was expressed in NIH/3T3 cells under the control of the SV40 promoter, the gene lacked transforming activity despite expression of detectable levels of the erbB-2 protein. A further five- to tenfold increase in its expression under influence of the long terminal repeat of Moloney murine leukemia virus was associated with activation of erbB-2 as a potent oncogene. The high levels of the erbB-2 product associated with malignant transformation of NIH/3T3 cells were observed in human mammary tumor cells that overexpressed this gene.     Beschreibung: Oncogenes encoding serine/threonine or tyrosine kinases were introduced into the established rodent fibroblast cell line NIH 3T3 and tested for tumorigenic and metastatic behavior in T cell-deficient nude mice. Transforming oncogenes of the ras family were capable of converting fibroblast cell lines to fully metastatic tumors. Cell lines transformed by the kinase oncogenes mos, raf, src, fes, and fms formed experimental metastases and (in some cases) these genes were more efficient at metastatic conversion than a mutant ras gene. In contrast, cells transformed by either of two nuclear oncogenes, myc or p53, were tumorigenic when injected subcutaneously but were virtually nonmetastatic after intravenous injection.     Beschreibung: The body plan of Drosophila is determined to a large extent by homeotic genes, which specify the identity and spatial arrangement of the body segments. Homeotic genes share a characteristic DNA segment, the homeo box, which encodes a defined domain of the homeotic proteins. The homeo domain seems to mediate the binding to specific DNA sequences, whereby the homeotic proteins exert a gene regulatory function. By isolating the normal Antennapedia gene, fusing its protein-coding sequences to an inducible promoter, and reintroducing this fusion gene into the germline of flies, it has been possible to transform head structures into thoracic structures and to alter the body plan in a predicted way.     Beschreibung: The earliest known response of eggs to sperm in many species is a change in egg membrane potential. However, for no species is it known what components of the sperm cause the opening of the egg plasma membrane channels. Protein isolated from sperm acrosomal granules of the marine worm Urechis caused electrical responses in oocytes with the same form, amplitude, and ion dependence as the fertilization potentials induced by living sperm. Sperm initiated fertilization potentials in oocytes when sperm-oocyte fusion, but not binding, was inhibited by clamping oocyte membrane potentials to positive values. Acrosomal protein also initiated electrical responses in clamped oocytes. The corresponding synthetic peptide (Arg19-Phe-Ala-Arg-Lys-Gly-Ala25-Leu-Arg-Gln-Lys-Asn-Val-His -Glu-Val-Lys-Asn36) acts as a potent substrate antagonist with an inhibitory constant of 147 +/- 9 nM. It is a specific inhibitor of protein kinase C and inhibits both autophosphorylation and protein substrate phosphorylation. Substitution of Ala25 with serine transforms the pseudosubstrate into a potent substrate.     Beschreibung: X-ray holography offers the possibility of three-dimensional microscopy with resolution higher than that of the light microscope and with contrast based on x-ray edges. In principle, the method is especially advantageous for biological samples if x-rays in the wavelength region between the carbon and oxygen K edges are used. However, until now the achieved resolution has not exceeded that of the light microscope because of the poor coherence properties of the x-ray sources and the low resolution of the detectors that were available. With a recently developed x-ray source based on an undulator on an electron storage ring, and high resolution x-ray resist, a hologram has been recorded at about 400-angstrom resolution.     Beschreibung: The calcium-calmodulin-dependent protein kinase II is a major component of brain synaptic junctions and has been proposed to play a variety of important roles in brain function. A complementary DNA representing a portion of the smaller 50-kilodalton subunit of the rat brain enzyme has been cloned and sequenced. The calmodulin-binding region has been identified and a synthetic analog prepared that binds calmodulin with high affinity in the presence of calcium. Like the 50-kilodalton kinase polypeptide, the concentration of the messenger RNA varies both neuroanatomically and during postnatal development of the brain.     Beschreibung: A human B cell subpopulation identifiable by the expression of the cell surface antigen Leu-1 (CD5) is responsible for most of the immunoglobulin M rheumatoid factor secreted in vitro after the cells are stimulated with Staphylococcus aureus. The ability of B cells bearing the Leu-1 marker (Leu-1+) to secrete rheumatoid factor is present early in development and extends to adulthood, since Leu-1+ B cells from cord blood and from peripheral blood lymphocytes of both normal adults and patients with certain autoimmune conditions secrete rheumatoid factor in comparable amounts.     Beschreibung: The mammalian visual system has a hierarchic structure with extensive reciprocal connections. A model is proposed in which the feedback pathways serve to modify afferent sensory stimuli in ways that enhance and complete sensory input patterns, suppress irrelevant features, and generate quasi-sensory patterns when afferent stimulation is weak or absent. Such inversion of sensory coding and feature extraction can be achieved by optimization processes in which scalar responses derived from high-level neural analyzers are used as cost functions to modify the filter properties of more peripheral sensory relays. An optimization algorithm, Alopex, which is used in the model, is readily implemented with known neural circuitry.     Beschreibung: The cerebellar cortex is perhaps the best characterized structure in the mammalian central nervous system. Although the major cerebellar cell classes are well known, a new class of cerebellar cortical neuron has now been identified with a monoclonal antibody (Mab) generated by a procedure for rapid immunization and selective immunosuppression of antibody responses. This procedure generates a high frequency of immunoglobulin G-class antibodies of desired specificity, and has allowed the generation of two antibodies that recognize subsets of cerebellar cortical neurons. One of these antibodies defines a previously unrecognized class of cerebellar neuron.     Beschreibung: A portion of the Duchenne muscular dystrophy (DMD) gene transcript from human fetal skeletal muscle and mouse adult heart was sequenced, representing approximately 25 percent of the total, 14-kb DMD transcript. The nucleic acid and predicted amino acid sequences from the two species are nearly 90 percent homologous.     Beschreibung: In order to identify the genetic factors associated with the radiation-resistant human laryngeal carcinoma cell line (SQ-20B), tumor cell DNA was transfected into NIH/3T3 cells. A high incidence (six out of six) of raf sequences was found in transfected NIH/3T3 clones and the tumorigenic potential of SQ-20B DNA could be linked to genomic fragments that represent most of the kinase domain of human c-raf-1. An apparently unaltered 3.5-kilobase pair (kb) human c-raf transcript was identified in SQ-20B cells but was not observed in the transfected NIH/3T3 cell clones. Two new transcripts (4.2 kb and 2.6 kb) were found in tumorigenic clones; the large transcript was missing in a very poorly tumorigenic clone. Cytogenetic analysis indicated that the normal autosomes of chromosome 3 were absent in SQ-20B karyotypes and had formed apparently stable marker chromosomes. Unlike the recipient NIH/3T3 cell line, 30 percent of the transformed clone-1 metaphases had minute and double-minute chromosomes representative of amplified DNA sequences.     Beschreibung: In order to study the rate and form of tubulin transport in cultured neuronal cells, the fluorescence recovery after the photobleaching of a fluorescent tubulin analog has been followed within the neuritic processes of differentiated PC12 cells. In these cells, as in peripheral axons, tubulin is transported in coherent, nondiffusing waves at two different slow rates that are within the range of the slow components a and b of axonal transport measured in vivo. Finally, it appears that most, if not all, of the tubulin analog is moving out these processes.     Beschreibung: The molecular basis for the marked difference between primate and rodent cells in sensitivity to the cardiac glycoside ouabain has been established by genetic techniques. A complementary DNA encoding the entire alpha 1 subunit of the mouse Na+- and K+-dependent adenosine triphosphatase (ATPase) was inserted into the expression vector pSV2. This engineered DNA molecule confers resistance against 10(-4) M ouabain to monkey CV-1 cells. Deletion of sequences encoding the carboxyl terminus of the alpha 1 subunit abolish the activity of the complementary DNA. The ability to assay the biological activity of this ATPase in a transfection protocol permits the application of molecular genetic techniques to the analysis of structure-function relationships for the enzyme that establishes the internal Na+/K+ environment of most animal cells.     Beschreibung: Neoplasms produce substances that induce blood vessel formation (angiogenesis). Fractions from ethanol extracts of the Walker 256 carcinoma were isolated by silica column chromatography and C18 reversed-phase high-performance liquid chromatography. Two of the isolated fractions induced neovascularization when tested in the rabbit corneal micropocket assay. One of the fractions was identified as nicotinamide by desorption-electron impact mass spectrometry, nuclear magnetic resonance spectroscopy, and gas chromatography-mass spectrometry. The second active fraction contained nicotinamide as part of a more complex, as yet unidentified, molecular arrangement.
