    Notizen: Abstract Cyanobacterin is a secondary metabolite produced by the cyanobacterium, Scytonema hofmanni. Highly purified cyanobacterin was found to inhibit the growth of many cyanobacteria at a minimum effective dose of 2 μg/ml (4.6 μM). The antibiotic had no effect on eubacteria including the photosynthetic Rhodospirillum rubrum. The site of action of cyanobacterin was further investigated in the unicellular cyanobacterium, Synechococcus sp. Electron micrographs of antibiotic-treated Synechococcus cells indicated that cyanobacterin affects thylakoid membrane structure. The antibiotic also inhibited light-dependent oxygen evolution in Synechococcus cells and in spheroplasts. These data support our conclusion that cyanobacterin specifically inhibits photosynthetic electron transport. This activity is similar to herbicides such as 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU). The anhydro analog of cyanobacterin had no biological activity.

    Notizen: Abstract Electron microscopy of negatively stained isolated restriction enzyme EcoRI revealed particle projections with triangular or square outlines, indicating that the enzyme, in its tetrameric state, is tetrahedron-like. The two dimers making up the tetramer appear to be arranged in two planes orthogonal to each other. Complexes formed by EcoRI with the plasmids pBR322 or pGW10 were investigated by electron microscopic spreading techniques. In the presence of Mg2+, EcoRI was bound to the DNA molecules to form pearl necklace-like aggregates. The number of bound EcoRI particles was much higher as the sum of EcoRI-and 5′..AATT..3′ sites (with exceptions, the 5′..AATT..3′ sites may function as one type of EcoRI* sites) along the DNAs, indicating unspecific binding. In the absence of Mg2+, EcoRI was bound to the DNA only at the recognition site for EcoRI and the sites where the tetranucleotide sequence 5′..AATT..3′ was present. A direct correlation of the local concentrations of the bases A and T within the flanking sequences of the binding sites with the frequency of EcoRI to the DNA was observed. Dimers and tetramers of the enzyme was found to bind to the DNA. Tetramers occasionally exhibited two binding sites for DNA as indicated by the observation of DNA loops originating at the sites of bound tetrameric EcoRI particles.

    Notizen: Summary The structure of guppy (Poecilia reticulata) spermatogonia and spermatocytes has been studied using electron microscopy. The spermatogonia, situated at the apex of the seminiferous tubule, are almost all surrounded by a network of Sertoli cells; they have very diffuse chromatin and one or two large nucleoli. The cytoplasm contains relatively few organelles, although annulate lamellae are found. The mitochondria have few cristae and are concentrated at one pole of the cell; they are sometimes found with intermitochondrial cement. These spermatogonia are separated from each other, having no intercellular bridges or inclusion in Sertoli cells, and are relatively undifferentiated; they correspond to stem cells. The spermatogonia beneath the apex are organized into cysts. First-generation spermatogonia are more dense and heterogeneous, their nuclei becoming smaller and their chromatin becoming denser during successive generations. In spermatocytes, the synaptinemal complex exists as a modified form until metaphase. The concentration of organelles in the cytoplasm increases and the organelles become more diversified as spermatogenesis progresses. Many cytoplasmic bridges are observed (several per cell), indicating that the cells remain in contact after several divisions. These changes in germ cell structure have been related to some of the characteristic features of spermatogenesis in guppy, e.g. the large number of spermatogonial generations and the complexity of spermiogenesis.

    Notizen: Summary The ultrastructure of hepatic peroxisomes was investigated in Ichthyophis glutinosus (Amphibia: Gymnophiona), employing perfusion fixation and the diaminobenzidine (DAB) technique for the visualization of catalase. The majority of peroxisomes is circular or rod-shaped, although elongated particles occasionally occur. They contain a finely granular matrix, lightly stained after the DAB procedure. Their mean diameter is approximately 0.25 μm. Serial sections reveal that the circular and rod-shaped peroxisomal profiles are cross and oblique sections of highly tortuous, tubular organelles exceeding 2 μm in length. In addition to tubular profiles, elongated, rectangular particles, as well as straight dumbbell-shaped organelles with distinct marginal plates are observed. They range from 900 to 1650 nm in length (mean = 1200 nm). In the flattened, thin central portion of the dumbbell-shaped particle, the peroxisomal membranes form a cisterna enclosing one or two uniformly thick marginal plates, which display a definite substructure with a periodicity of 10 nm. These findings indicate that peroxisomes in the liver of Ichthyophis exhibit a complex organization. It is suggested that the organelles undergo a specific differentiation process, morphologically characterized by the formation of enlarged segments of unusual shape.

    Notizen: Summary Isolated ovaries from untreated, sexually mature rabbits were introduced into an in vitro perfusion system and perfused with a chemically defined medium containing albumin. The ovaries were perfused for up to 15 h (mean 11.5 h) and then processed for morphological investigation. Both at the light- and electron-microscopical levels, most of the ovaries exhibited a normal structure comparable with ovaries in situ. In two cases, however, marked accumulations of bacteria were found, although not inside the follicles. Since ovulation in the rabbit normally occurs between 9.5–13 h after mating or human chorionic gonadotrophin treatment, this model seems adequate for studies of ovulation in vitro. It is, however, important to study the ovaries microscopically after the perfusion to detect artifacts, e.g., bacterial infection, that may have influence on the process of ovulation.

    Notizen: Summary Spinal cord neurons from 9-day chick embryos were maintained in culture for up to 35 days and then fixed in 4% cacodylate-buffered glutaraldehyde containing 2% tannic acid. After about 15 days in culture a small percentage of the synaptic specializations present were characterized by striking electron-dense striations averaging 15 nm in width, oriented perpendicular to the postsynaptic membrane. These structures increased in frequency with time in culture (to a maximum of about 10% of all synapses in the oldest cultures); they were asymmetrical, protruding approximately 8 nm into the synaptic cleft, and more deeply (approximately 15–18 nm), into the postsynaptic cytoplasm. On the basis of earlier work by Sealock (1980) they are interpreted as concentrations of acetylcholine receptors. Similar membrane differentiations were also seen associated with active-zone areas of a few presynaptic membranes, and the possibility that these represent presynaptic acetylcholine receptors is discussed. Additional observations reported are (1) the presence of striations resembling those seen at the postsynaptic membrane in the membranes of some postsynaptic vesicles, and (2) filamentous links between the striations and cytoskeletal elements of the postsynaptic cell.

    Notizen: Summary The taste buds on the barbels in three species of teleosts (Cyprinus carpio, Misgurnus anguillicaudatus, Parasilurus asotus) were studied by means of fluorescence and electron microscopy. Intensely yellow-fluorescent cells, which are disk-shaped and located exclusively in a basal position, are observed in the barbel-buds of all fishes examined. The basal cells contain a large number of small clear vesicles approximately 40–60 nm in diameter, which show a tendency to aggregate in the cytoplasm facing the junction of the nerve terminals; chemically transmitting synapses are seen in the latter region. It is suggested from the present observations that the basal cells in the barbel-bud may originate from Schwann cells and have a dual function both as mechanoreceptors and paracrine elements. Since the administration of 5,6-DHT results in an appearance of small dense vesicles among the small clear vesicles, the possibility exists that the basal cell may be capable of taking up monoamines and storing them in the small clear vesicles.

    Notizen: Summary In an attempt to identify a cellular Ca2+-pool, from which calcium is released when secretagogues are applied, tissue fragments of the rat exocrine pancreas were incubated and fixed with glutaraldehyde in the presence of calcium. By means of this procedure electron-dense deposits were found on plasma membranes. X-ray microanalysis showed that these deposits contain calcium. Stimulation of tissue fragments with the use of the secretagogues carbachol or cholecystokinin reduced the number of deposits by about 80%. When the antagonist atropine was applied after carbachol stimulation, deposits reappeared on cell membranes, which then disappeared again after a second stimulation with cholecystokinin. In the presence of procaine, carbachol was inhibited and only slightly reduced the Ca2+-deposits on the plasma membranes. These results suggest that a calcium pool, from which calcium is released to induce enzyme secretion on stimulation, is located in the cell membrane

    Notizen: Summary Fathead minnows, Pimephales promelas, were exposed for 129 days to Lake Superior water acidified with sulfuric acid by means of a flow-through toxicant injection system. The effects of chronic acid stress (pH 6.5, 6.0, 5.5, 5.0) on gill histology were examined. Most of the histological effects were seen at pH 5.5 and 5.0 and were confined primarily to changes in numbers, distribution, and morphology of chloride cells. At low pH levels there tend to be more chloride cells in the gill epithelium and an increased percentage of these cells in the secondary lamellae. In contrast to normal chloride cells, chloride cells from fish exposed to low pH frequently had apical pits while some had bulbous apical evaginations. The occurrence of structural changes in chloride cells during exposure to acid water suggests that chloride cells may be involved in acclimation to acid stress.

    Notizen: Summary The size, cellular location, and identity of surface-associated components were determined for lipid droplets in lactating cells. Transmission electron-microscopic measurements were made involving 3801 droplets in approximately 211 cells from three rats and 1197 droplets in 66 cells from a mouse. For the purposes of droplet evaluation, cells were divided into seven locations ranging from basal to secreting positions. Droplets were also categorized with respect to contact with other droplets, basolateral plasma membrane, mitochondria, Golgi apparatus, secretory vesicles, and endoplasmic reticulum-cytoplasm (ERC). Data on droplet size showed that droplet growth occurs mainly in the secretory position, confirming previously published findings. Lipid droplets from mouse tissue, although somewhat smaller in size showed similar growth trends to those of the rat. Data on numbers of droplet contacts and percentages of droplet circumferences involved in associations with other cell components showed that the dominant interaction of lipid droplets was with the ERC. However, intimate association of droplets with mitochondria was noted in all cellular locations. In addition, nursed animals exhibited a greater proportion of droplet surface association with secretory vesicles and less in contact with mitochondria in comparison to those not nursed. The significance of these relationships to milk synthesis and secretion is discussed.

    Notizen: Summary The nucleoli of dictyate-stage growing oocytes in rat ovaries were examined both with routine electron microscopy and electron microscopy after silver nitrate and ammoniacal silver nitrate (Ag-AS) staining. The nucleoli of the unilaminar follicular oocytes consist of twisted strands of dense fibrillar components, aggregates of granular components, and small fibrillar centers. After Ag-AS staining, silver grains are numerous on the dense fibrillar strands, fewer on the fibrillar centers, and very sporadic on the granular aggregates. The same stainability of three nucleolar components with the Ag-AS method was also confirmed in the nucleoli segregated by actinomycin D. During the transition of growing oocytes from bilaminar to plurilaminar follicle stage, the nucleolar dense fibrillar strands gradually conglomerate and are transformed into large and compact spherules. The stainability of dense fibrillar components with the Ag-AS method was lost along with this nucleolar transformation. These results may provide some new clues on the functional significance of AgAS-positive proteins in the nucleoli.

    Notizen: Summary Stereology and semi-automatic image analysis were used with the aim of comparing the structure of parathyroid glands from untreated adult Mongolian gerbils fixed by immersion with those fixed by perfusion. Subclassification of the chief cells based upon the staining affinity or electron density of the cytoplasm was readily performed only in glands fixed by immersion, and so-called atrophic cells were observed only in these glands. The atrophic cells were often surrounded by “light” chief cells. In glands fixed by perfusion, “light” chief cells were only rarely encountered. A significant difference between glands fixed by immersion and those fixed by perfusion was found only with regard to the form of cells and nuclei, those fixed by perfusion being more spherical. When comparing individual cells within glands fixed by immersion, “light” chief cells were more spherical and had a significantly larger nuclear and cellular size, and a lower mitochondrial volume density than the “intermediate”/“dark” chief cells. Otherwise there were no significant differences in any of the parameters investigated. These data indicate that occurrence of socalled “light” chief cells and atrophic cells is a result of improper fixation. The results of this study do not favour the concept of a functional cycle with a simultaneous occurrence of active and inactive cells within parathyroid glands.

    Notizen: Summary Electron-microscopic studies of lymphoid tissues from bovine fetuses and from calves disclosed a non-lymphoid cell type in the thymus-dependent zones of secondary lymphoid tissues and in the thymus that is distinguishable from reticulum cells, epithelial and endothelial cells, and macrophages. Based on morphological and topographical criteria, the cell is identified as the interdigitating cell. In addition, studies of the tissues of normal and virus-challenged fetuses, and of conventionally reared calves, indicated that the interdigitating cells originate from monocytoid cells, which undergo differentiation in the thymus-dependent zones during an immune response.

    Notizen: Summary Gap junctions exist in the septa between the segments of the lateral giant axons in the ventral nerve cord of the crayfish Procambarus. A large increase in the resistance (uncoupling) of these gap junctions was brought about by mechanical injury to the axonal segments. Both thin sections and freeze-fracture preparations were used to monitor the morphological changes which occurred up to 45 min after injury. There was no apparent change in the organization (a loose polygonal array) of the intramembrane particles which make up the junctional complex up to 45 min after injury. In some instances, however, the intramembrane particles appeared to have moved away from the junctional area. Other junctional regions were internalized and appeared similar to what have been called annular gap junctions. Also at this time (20–25 min after injury), a dense cytoplasmic plug formed in uninjured axon near the junctional region. It is concluded that the gap junctions that exhibit a loose polygonal organization of the intramembrane particles may be either in a state of low resistance (coupled) or a state of high resistance (uncoupled).

    Notizen: Summary Ovulation was induced in rabbits by intravenous administration of human chorionic gonadotrophin (HCG), and 4–5 h later the ovaries were isolated and introduced into an in-vitro perfusion system containing synthetic medium with albumin. Rupture of follicles occurred in vitro within the physiological time range (mean 11.3 h after injection of HCG), although with a reduced frequency. Preovulatory and ruptured follicles were studied in detail by light and electron microscopy. In the granulosa layer of ruptured or preovulatory follicles cytoplasmic blebbing activity, disappearance of CallExner bodies and differentiation toward luteinized cells were found. Perhaps the most important sign of normal preovulatory development in vitro was that the basement membrane surrounding the granulosa layer was penetrated by projections of granulosa cells. In the absence of this penetration phenomenon the granulosa layer prolapsed out of the follicle. Immediately before rupture, follicles showed marked degeneration, restricted to the outer layers of the apical wall, which is compatible with the hypothesis that degradative enzymes are released close to the surface of preovulatory follicles. Although the majority of follicles that ovulated under in-vitro conditions showed the same kind of morphological alterations as can be seen in vivo, occasional atypical ruptures occurred without any overt signs during perfusion. Also technical manipulations of the perfusion system, e.g., nonphysiological increase of perfusion pressure, could force follicles to rupture. This illustrates the importance of careful morphological study of all ovaries perfused in vitro before conclusions are drawn.

    Notizen: Summary In the pars distalis of the hypophysis of adult Rana temporaria, three types of nerve-fiber profiles were found at two distinct sites, in both lateral parts of the bordering regions of the anterior lobe with the intermediate lobe of the hypophysis. The first type of nerve-fiber profile consists of bundles of very fine axonal elements (diameter: 〈0.7 μm). The second type is formed by larger nerve fibers (diameter up to 4 μm) containing a few neurosecretory granules of approximately 100 nm. The third type of nervefiber profile resembles the second type but these nerve fibers make synaptoid contacts on at least two different types of glandular cells. The possible functional significance of these nerve fibers in the pars distalis is discussed. No nerve fibers were found (1) in the central part of the bordering region of the pars distalis with the intermediate lobe, (2) at the bordering region with the median eminence and (3) with the neurohypophysial stalk, and (4) in all other parts of the pars distalis.

Vascular injections with microfil were cleared and dissected; corrosion casts were studied under the scanning electron microscope (SEM). Perfusion-fixed tissue was used for SEM and transmission electron microscope (TEM) studies, including freeze-fracture specimens. The branches of one mesonephric artery carry up to 15 glomeruli. Several glomeruli occupy the same arterial branch, with very short afferent arterioles proper. The efferent vessels, frequently 2–5, leave the extensive vascular pole opposite the entering arteriole and split into peritubular capillaries radiating towards the superficial veins. These capillaries form vascular regions in the shape of flattened pyramids. Along its course, one nephron is supplied by vessels derived from 4–7 glomeruli. The nephrons have less vascular contact than in the definitive kidney. The ultrastructure of the single mesonephric vessels matches the metanephric counterparts. Epithelioid cells with renin granules are common in afferent arterioles, larger arteries, and efferent vessels. The lobulated glomeruli are up to 750 μm long and flattened, showing the usual features of podocytes, mesangial cells, and an attenuated endothelium with fenestrations between 50 and 250 μm. It partially retains its own basement membrane. There is no proximal mesangium.

    Notizen: Summary The ultrastructure of Pacinian corpuscles of the cat located in the crural region and innervated by the interosseous nerve was studied 1 to 14 months after denervation. Both the Pacinian inner core and capsule remained well preserved one month after denervation. However, the denervated inner cores underwent progressive atrophy and wasting, which resulted in a gradual reduction of the amount of inner-core cells and lamellae, widening of interlamellar clefts, formation of empty spaces in the axial region and a considerable increase in the number of collagen fibrils. In spite of the wasting, the inner core still survived 14 months after denervation, but at least half of its volume became occupied by collagen fibrils which surrounded the remaining inner-core cells and lamellae. Collagen fibrils assembled in the denervated core were markedly thinner than those found in the capsule, as is also the case in normal Pacinian corpuscles. In the capsule, discrete focal degeneration, occasional pyknosis of the innermost capsular cells and macrophage infiltration were observed from the first month after nerve section onward, but the number of capsular layers remained within the normal range (30–40) up to 14 months after denervation.

    Notizen: Summary Addition of certain heterocyclic nitrogen-carbon compounds to standard osmium tetroxide solutions used as secondary fixative resulted in an enhanced general membrane contrast in cells of the marine algaEmiliania huxleyi (Lohmann) Kamptner. Ultrastructural cell morphology and the contrast distribution were compared between cells treated according to a standard secondary fixation procedure and cells post-fixed when above mentioned heterocyclic compounds were introduced; in both cases some of the ultrathin sections were post-stained. Different compounds were tested: 1,2,4-triazole (TRA), 3-amino-1,2,4-triazole (A-TRA), 5-amino-tetrazole (A-TEA) and 2,4,6-tri-amino-1,3,5-triazine (melamine). The results were interpreted to indicate the possible bonding types arising from interaction of the heterocyclic compounds with osmium tetroxide and with membrane constituents. Interpretations were partly inspired by considerations from coordination chemistry. All above tests which did not include post-staining of thin sections could be performed at alkaline pH, and consequently calcified structures were preserved. The enhanced osmium accumulation at membranes was verified with X-ray microanalysis, which also showed that in the cases where membranes were visibly contrasted, localization of probable sites of intracellular non-crystalline calcium was facilitated.

    Notizen: Summary There is an immense literature on biological and biochemical analyses of soils. Such analyses have revealed the enormous richness of species in soil and their vast range of metabolic potentials and ecological diversity. Accordingly, the approaches used to investigate the soil biota and its biochemistry usually have to be modified or adapted depending upon the purpose of the investigation. Studies of micro-organisms in the soil environment, are complicated because microbial cells are commonly attached to surfaces where they live side-by-side with other populations in consortia usually containing different morphological and physiological types. Such assemblages of organisms cannot be described quantitatively using cultural techniques, such as plate counts, which underestimate both cell numbers and viable biomass. The development of more powerful observational and staining techniques has improved our knowledge of the diverse morphological and biochemical composition of soil micro-communities. Such findings have been amplified at a grosser level by laboratory studies with multi-component systems (microcosms) to mimic field situations and to assess the range of biochemical potentials of microbial consortia. But despite notable advances in analytical methods we are still, with a few exceptions, unable to detect or identify those microorganisms which carry out specific biochemical transformations or determine whether particular cells are alive, dormant or dead at the time of observation. Considerable work has been done to define some of the fundamental ecological attributes of microbial assemblages in soil. Productive work on the metabolic activities of the soil microbiota, specially geochemical transformations of C, N, S and P, has been under way for more than a century. But only in more recent years have more sensitive and reproducible analytical methods become available to measure viable biomass in soil. This will enable some insight to be gained into the role that microbial biomass plays as a labile source and sink for plant nutrients.

    Notizen: Summary The stability of pores and particles is essential for optimum growth of plants. Two categories of aggregates macro- (〉 250 μm) and micro- (〈250 μm) depend on organic matter for stability against disruptive forces caused by rapid wetting. Dispersion of clay particles from microaggregates is promoted by adsorption of complexing organic acids which increase the negative charge on clays. The acids are produced by plants, bacteria and fungi. However, the dispersibility of clay in microaggregates is offset by the binding action of polysaccharides, mainly mucilages produced by bacteria, but also by plant roots and fungal hyphae. The stability of microaggregates is also enhanced by multivalent cations which act as bridges between organic colloids and clays. Macroaggregates are enmeshed by plant roots, both living and decomposing, and are thus sensitive to management, and increase in number when grasses are grown and the soil is not disturbed. Lack of root growth,i.e. fallow, has the opposite effect. Various implications for management of soil structure are discussed.

    Notizen: Abstract Recently published results for solution crystallized PE single crystals have shown, that the experimental heat of fusionΔH * is higher, if the solvent is exchanged to silicon oil (oil suspension samples) as compared with dried mats. This has been interpreted by the collapse of the original hollow pyramids during drying, inducing lateral defects within the lamellae. The present investigation does not confirm this unexpected result.ΔH * of dried mats (T c 66 to 91 °C) and of the corresponding oil suspension samples agree within the rather small limits of experimental error. The crystallinities as derived fromΔH *, density or WAXS are in excellent agreement. SEM micrographs of cold fractured dried mats show their spongy macromorphology, but TEM micrographs of stained ultra-thin sections reveal the lamellar morphology of the walls, consisting of curved lamellae and stacked hollow pyramides. If a dried mat is sintered at room temperature, a dense transparent film is obtained with a rather regular stacked morphology of large flat lamellae.ΔH * of these films agrees with that of the original mat.

They elute at the same location on chromatography with Ac 22. The fatty acid composition of AcA and the bone resorptive factor is the same. Some differences in carbohydrate composition are observed. TheActinomyces factor does not affect calcium influx or cyclic AMP in isolated bone cells. Therefore it is concluded that AcA stimulates resorption either by gaining entrance into bone cells or by way of a yet undetermined second messenger.

The relationship between enzymatical digestibility and structure or composition ofChlorella cell walls were studied by electron microscopy and staining techniques with some specific dyes. The cell wall structures of the 12Chlorella strains were grouped into three types: (1) with a trilaminar outer layer, (2) with a thin outer monolayer, and (3) without an outer layer. Protoplasts were formed only from the strains with a cell wall of Type 2. In the strains with a cell wall of Type 1, the outer layer protected the inner major microfibrillar layer against enzymatic digestion. The cell wall of Type 3 was totally resistant to the enzymes; the chemical composition of the cell wall would be somewhat different from that of other types.

These peripherally located thylakoid centers are cylinders 30 nm wide by 320 nm long, consisting of globular subunits oriented in nonparallel stacked arrays. Thylakoids are attached to the outer surface of the cylinder along its longitudinal axis.     Notizen: Abstract In contrast to former findings lysozyme was able to attack the cell walls ofStaphylococcus aureus under acid conditions. However, experiments with14C-labelled cell walls and ribonuclease indicated that, under these conditions, lysozyme acted less as an muralytic enzyme but more as an activator of pre-existing autolytic wall enzymes. Electron microscopic studies showed that under these acid conditions the cell walls were degraded by a new mechanism (i.e. “attack from the inside”). This attack on the cell wall started asymmetrically within the region of the cross wall and induced the formation of periodically arranged lytic sites between the cytoplasmic membrane and the cell wall proper. Subsequently, a gap between the cell wall and the cytoplasmic membrane resulted and large cell wall segments became detached and suspended in the medium. The sequence of lytic events corresponded to processes known to take place during wall regeneration and wall formation. In the final stage of lysozyme action at pH 5 no cell debris but “stabilized protoplasts” were to be seen without detectable alterations of the primary shape of the cells. At the same time long extended ribbon-like structures appeared outside the bacteria. The origin as well as the chemical nature of this material is discussed. Furthermore, immunological implications are considered.

    Notizen: Summary By the use of thin sections and freeze-fracture replicas the glomerular and tubular structures of the kidney of the frog (Rana esculenta) were studied with special reference to intercellular junctions. In the glomerulus the filtration barrier is of very variable thickness, and frequent tight and gap junctional contacts occur between podocyte processes. Although structurally less elaborate, the proximal tubule resembles its mammalian counterpart. In the initial part the tight junctions are relatively shallow but become very broad in the mid and distal portions of the proximal tubule. The proximal tubular cells are extensively linked by gap junctions. In some animals the shapes of the cells in the proximal and distal portions of the proximal tubule were markedly different. The distal tubule consists of two segments which differ mainly in the pattern of interdigitations and the structure of the zonulae occludentes. Similarities with the tight junctional morphology of the mammalian distal tubule are striking. In the first part of the distal tubule (diluting segment) a narrow band of parallel tight junctions is found closely resembling that found in the mammalian straight distal tubule; in the more distal part of the distal tubule, however, a broad band of anastomosing tight junctional strands exists, like the zonula occludens of the mammalian convoluted distal tubule. The connecting tubule displays cellular dimorphism: its wall contains a mixture of light and dark (flask) cells. The luminal and basolateral membranes of the flask cells are covered with numerous rod-shaped particles. The tight junctions of the connecting tubule are broad and increase in depth and number of strands along its length; they are typical of a very tight epithelium. In spite of several dissimilarities with phylogenetically younger kidneys our findings suggest that many structural principles of the mammalian kidney are also represented in the kidneys of amphibians. The structural-functional relationships are discussed.

    Notizen: Summary In the gastric mucosa of two teleost species, the perch (Perca fluviatilis) and the catfish (Ameiurus nebulosus) three endocrine cell types were found, located predominantly between the mucoid cells of the gastric mucosa. A fourth cell type is present in the gastric glands of catfish. Each cell type was defined by its characteristic secretory granules. Type-I cells were predominant in both fish. These cells contained round or oval granules with a pleomorphic core. The average diameter of granules was 400 nm for the perch and 270 nm for the catfish. Type-II cells of both species displayed small, highly osmiophilic granules about 100 nm in diameter. The secretory granules of type-III cells (260 nm in the perch and 190 nm in the catfish) were round or slightly oval in shape and were filled with a finely particulate electron-dense material. Type-IV cells of the catfish were found in the gastric glands only. Their cytoplasm was filled with homogeneous, moderately electron-dense granules averaging 340 nm in diameter. The physiological significance of these different morphological types of gastric endocrine cells requires further investigation.

    Notizen: Summary Immuno-electron microscopy was performed on live, cultured, early postnatal cerebellar and retinal cells of the mouse to identify A2B5 antigenbearing elements. In cerebellar cultures, granule cells, some immature oligodendroglia, and astroblasts express A2B5 antigen on their cell surfaces. The typical features of astroblasts include large cisternae of the endoplasmic reticulum and a mixed population of intermediate-sized filaments and microtubules. Immature oligodendroglia cells express the antigen on their cell bodies and on procecesses filled with cytoplasm. Cytoplasm-free membranous whorls, however, are devoid of A2B5 antigen, but not of 0 or NS-1 antigens. In retinal cultures, A2B5 antigen is observed on differentiating neurons with the exception of photoreceptor cells as identified by ribbon synapses.

    Notizen: Summary The ultrastructure of Pacinian corpuscles located on the crural interosseous membrane was studied in adult rats 6 h to 10 months after transection of the right sciatic nerve. Axon terminals degenerated one day after transection and were engulfed and resorbed by cells of the inner core within one week. The axial space left after removal of the axonal debris was closed by the lamellae of the inner core. The main structural features of the inner core and capsule remained preserved after denervation throughout the period of study. The denervated inner cores, however, became atrophic 10 months after neurotomy, their mean diameter being reduced by 17.5% compared with that of contralateral control corpuscles. The number of capsular lamellae was unaltered, and perineurial pathways of the peripheral nerve stump remained preserved. Schwann cells proliferated and formed Büngner bands during the first month after denervation, but retracted their processes and became atrophic at later stages after neurotomy. Survival of Pacinian corpuscles after long-term denervation in adult rats is in contrast to their rapid degeneration within several days after nerve section in neonates.

    Notizen: Summary Electron microscopy reveals that sarcolemmata of adjacent muscle cells form pentalaminar junctions by fusion of apposed trilaminar double leaflet membranes. These junctions appear to be candidates for low resistance pathways between muscle fibers. The muscles depolarize slowly when bathed in solutions containing elevated concentrations of KCl, and the sucrose gap method can then be used to measure the potential difference between polarized and depolarized regions.     Notizen: Summary A peritrophic membrane (PM) has been demonstrated in the gut of feeding larvae, nymphs, and adults of the tick Ixodes dammini. This is the first report of a PM in ticks. This temporary structure divides the lumen of the gut into two compartments, an endoperitrophic space, the lumen proper, and an ectoperitrophic space located between the PM and the epithelial cells of the gut wall. The PM is a mechanical barrier and even such small particles as ribosomes derived from ingested reticulocytes are retained in the lumen proper; they are never found in the ectoperitrophic compartment. In Ixodes dammini fed on hamsters infected with Babesia microti some of the parasites are found in the ectoperitrophic space. This passage is accomplished by a highly specialized organelle, the arrowhead, which develops in some Babesia during their metamorphosis in the gut of the vector. The arrowhead, while passing through the PM, changes its fine structure and loses its internal organization as if releasing some of its contents. Its disintegration continues and it disappears shortly after the Babesia have entered the epithelial cells. Only Babesia equipped with the arrowhead structure are able to cross the PM. This is the first documented case of a parasite traversing a solidified PM.

    Notizen: Summary Scanning electron microscopy of various regions of the body of the marine gastropod Pleurobranchaea californica (McFarland) has revealed a characteristic cell type that bears cilia with dilated discoid-shaped tips. The tips of the cilia consist of an expansion of the ciliary membrane around a looped distal extension of the axoneme. These kinocilia have been observed in numerous other marine invertebrates and are generally referred to as paddle cilia (Tamarin et al. 1974) or discocilia (Heimler 1978). Although many functions have been proposed for paddle cilia, little empirical evidence supports any of the proposals. In Pleurobranchaea we have found that the distribution of this ciliated cell type corresponds exactly to areas of the body known from behavioral studies (Lee et al. 1974; Davis and Matera 1981) to mediate chemoreception. Transmission electron microscopy of the epithelium lining the rhinophores and tentacles of Pleurobranchaea revealed details of the ultrastructure of these ciliated cells and showed that they are primary receptors. These ciliated receptors lie in a yellow-brown pseudostratified columnar epithelium that superficially resembles the olfactory mucosa of vertebrates.

    Notizen: Summary The possible biogenesis of two pigment granule types present in the monochromatic, brown chromatosomes enveloping the ventral nerve chord of the freshwater palaemonid shrimps Macrobrachium acanthurus, M. heterochirus and M. olfersii is examined by transmission electron microscopy in thin section and freeze fracture replicas. Prominent, membrane limited granules are suggested to have their origin in a complex, juxtanuclear, smooth endoplasmic reticulum labyrinth, continuous with the nuclear envelope. Amembranous, lipocarotenoid granules possibly derive from the external surface of the smooth endoplasmic reticulum. Nuclear envelope and SER membranes contain numerous 11 nm diameter intramembranous particles while pigment granule membranes exhibit fewer particles. A dictyosomal origin for the lipocarotenoid granules is discounted. Granulogenesis is suggested to be a continuous process in crustacean chromatophores.

    Notizen: Summary Ultrastructural and stereological assessment of the mature avian anterior latissimus dorsi (ALD) muscle showed that it contains two kinds of extrafusal fibers. This fine structural dichotomy of fiber types in the ALD correlated well with their previously reported histochemical duality. Distinct differences occur in sarcomere banding, myofibrillar area, sarcotubular and mitochondrial density, and in morphology of motor-nerve terminals. Both myofiber types in this muscle were interpreted as representing varieties of “slow” or tonic muscle fibers. Both fibers contain myofibrils that, despite differences in cross-sectional area, were large, irregular, and ribbon-shaped, typical of the “Felderstruktur” appearance of true “slow” fibers. Whereas the majority of fibers (type-1) are devoid of well-defined M-bands, the minor fiber population (type-2) exhibit prominent M-bands in the center of each sarcomere. In addition, type-1 tonic fibers contain a significantly lower mitochondrial and sarcotubular volume than the tonic fibers of type-2. While both fiber types exhibit motor-nerve terminals that are small, smooth and punctate in appearance, those on the type2 fibers often had a number of shallow postjunctional folds. Whether or not these two classes of extrafusal fiber in this muscle represent two separate and distinct types of motor units remains to be determined functionally.

    Notizen: Summary Normal diploid cells require serum to proliferate in culture. Platelet-derived growth factor has been identified as the main serum component responsible for this effect. Here, smooth muscle cells were isolated enzymatically from the aorta of 5-day-old rats and cultured in the presence of 10% whole blood serum (WBS) or plasma-derived serum (PDS), i.e. with or without platelet factor, and studied by transmission electron microscopy. The cells proliferated actively in WBS-medium but remained quiescent in PDS-medium. Fine structurally, cells from WBS-cultures demonstrated numerous mitochondria, an extensive rough endoplasmic reticulum (RER), a large Golgi complex, a few lysosomes, and microfilaments arranged in parallel bundles. After transfer to PDS-medium, the RER- and Golgi cisternae were markedly dilated and the number of membrane-associated ribosomes decreased. Segregation of fragments of cytoplasm within autophagosomes was frequently observed and the number of lysosomes increased. Lipid droplets were more abundant and often gathered in the Golgi area. Moreover, the cells had become more irregular in shape and showed many bleb-like processes at their surface. Microfilament bundles had also become more prominent and crossed each other in different directions. These observations show that the removal of platelet factor from the medium clearly modifies the fine structure of cultured smooth muscle cells. The findings are in good agreement with the concept that platelet factor not only supports the proliferation of cultured cells but also stimulates their secretory activity.

    Notizen: Summary Alterations of the retinula cells in the retina of the light-adapted crayfish in response to hyper- and hypoosmotic van Harreveld solutions was examined by transmission electron microscopy. Increased osmolarity of the bathing medium to twice that of the physiological value leads to shrinkage of the retinula-cell somata. Microvilli, on the other hand, do not undergo shrinkage. Some other characteristic irreversible changes do, however, take place, including detachment of microvilli from the soma, showing a two- or threefold increase in diameter, and a concomitant decrease in number, probably due to fusion of microvilli. Prolonged incubation or higher osmolarities (5 isosmol) cause alteration of the microvillar membranes to whorls. Structural changes are often restricted to microvillar stacks evaginating from certain individual retinular cells. The number of affected stacks increases depending on the duration of incubation or the osmotic pressure. Hypoosmotic solution (0.5 isosmol) also induces an increase of microvillar diameters and a concomitant reduction in number of microvilli per stack. Exposure to a 20% solution of glycerol causes destruction of the rhabdom structure and the formation of whorls from microvillar membranes. The present findings suggest that the structure of the microvilli is stabilized by an axial cytoskeleton.

    Notizen: Summary Phagocytic activity and population development of medullary interdigitating cells (IDC) and cortical macrophages have been studied in the rat thymus after irradiation-induced thymocyte necrosis. IDC clearly demonstrate phagocytic activity, particularly in the 16h stage after irradiation. At this stage the number of necrotic thymocytes is maximal and the total number of phagocytic cells is insufficient to phagocytize all necrotic material. IDC increase in number slightly and are predominantly phagocytizing in the corticomedullary region (CMR), where they appear to develop from macrophage-like cells. The results indicate that medullary IDC can be phagocytic but have a different developmental pathway than cortical macrophages. Cortical macrophages greatly increase in number and acquire the appearance of tingible body macrophages by phagocytizing many necrotic thymocytes. They seem to develop from monocytes that normally enter the thymus at the CMR. During the acute involution macrophages probably also enter the cortex via the connective tissue capsule. It is suggested that thymus medullary IDC probably belong to the mononuclear phagocytes, as do the cortical macrophages. In the specific medullary environment IDC gradually develop their characteristic ultrastructure for an apparently other than phagocytic function. The similarities between IDC and epidermal Langerhans cells are discussed.

Within a few minutes of ovulation, the stratum granulosum and theca interna disappear completely in the middle part of the stigma. By 30 min before ovulation, the rough endoplasmic reticulum in the fibroblasts of the theca externa develops conspicuously. Then a few minutes before ovulation, the theca externa becomes much thinner with a conspicuous disintegration of the collagenous fibers into individual fibrils. These structural changes may contribute to the fragility of the stigma tissue and induce rupture of the follicle.

    Notizen: Summary Intercellular junctions in the young mouse ovary were examined by electron microscopy utilizing freeze-fracture and thin-sectioning techniques. Projections from the granulosa cells adjacent to the oocyte (GI) traverse the zona pellucida and form small gap junctions on the oocyte surface. On the P-face of these cells, the junctional aggregations are occasionally associated with linear strands of particles. In contrast, large gap junctional areas are frequently observed between the more peripherally located granulosa cells (GE) and are also present in the theca interna (TI) cell layer surrounding the follicles. Three types of tight junctional strands are discernible on the P-face of theca externa cells (TE): angularly zigzag strands consisting of intermittently distributed intramembranous particles on wide ridges, intermediate zigzag strands consisting of more continuously distributed particles, and wavy strands consisting of rather fused particles. Tight junctional strands are also present in the middle of grooves on the E-face of endothelial cells of blood vessels. In the germinal epithelial cell layer, tight junctional strands appear to be discrete and form a less anastomosing network.

    Notizen: Summary Using light and electron microscopy, the morphogenesis of the pars intermedia of the sheep pituitary gland was examined in developing lambs between 26 days of gestation and the newborn stage. Following the establishment of contact between the anterior and posterior lobe primordia seen at 26 days, the connection with the pharyngeal roof disappeared by 31 days. The lumen of Rathke's pouch, which was a prominent cavity at the earlier stages, became inconspicuous by 40 days but progressively increased in size during gestation and, in some newborn animals, contained colloid material. At 40 days, the pars intermedia consisted of a uniform population of undifferentiated cells. Cells with cytoplasmic granules were first identified at 50 days. The cytological appearance of granular cells at 70 days indicated increased synthetic activity and by 80 days they closely resembled adult glandular cells. At 100 days, membrane activity suggestive of exocytosis was first observed in granular cells; fenestrated capillaries were present, and early follicle formation between adjacent non-granular cells was seen. This apparent exocytotic release of granules was observed much more frequently between 100 days of gestation and the newborn stage than in adult pars intermedia cells. These findings indicate that glandular cells of the developing pars intermedia are actively engaged in synthesis, storage and secretion from an early stage.

    Notizen: Summary The early uterine response to transplanted, delayed and estrogenactivated blastocysts was studied ultrastructurally and compared with that induced by intrauterine instillations of deciduogenic agents (arachis oil, air). The uterine responses to delayed and activated blastocysts showed no ultrastructural or temporal differences. Already within 4 h after transfer to a sensitized uterus, the delayed blastocysts exhibited signs of activation, and both types of blastocysts had started to attach onto an undamaged epithelial lining. Signs of stromal cell differentiation into decidual cells were also seen as early as 4 h after transfer, while the Pontamine-blue reaction did not appear until after 8 h. The results therefore indicate that the transplanted blastocysts induced decidualization atraumatically and that the delayed blastocysts were either deciduogenic already before transfer or rapidly acquired deciduogenic properties after transfer. Artificial decidual induction with oil and air led to damage or death of a large number of cells in the uterine luminal epithelium. Within only 15 min after instillation pronounced signs of cell damage were seen, and later numerous cells were extruded from the epithelial lining. In the stroma ultrastructural signs of decidual cell differentiation and a Pontamine-blue reaction were observed as early as 4 h after induction. It is therefore suggested that oil and air induce decidualization via the epithelium by means of trauma.

    Notizen: Summary Ultrastructural studies of the ventricular surface of the frog cerebellum showed regional differences. In the midline region of the adult cerebellum was found a band of profusely ciliated squamous ependymal cells. In the rest of the cerebellum the ependymal cells were columnar and each had a single cilium. In the cerebellum of the premetamorphic tadpole, the squamous ependymal cells of the midline region also were monociliated. During metamorphosis they gradually became multiciliated. Additionally, supraependymal cells and synaptic elements were present on the ventricular surface of the cerebellum of adult frogs as well as in late metamorphic tadpoles. In contrast, supraependymal cells were rarely observed in premetamorphic tadpoles, and it was concluded that the supraependymal system develops during metamorphosis. It is postulated that the band of cilia may be associated with the circulation of cerebrospinal fluid, and supraependymal synaptic elements function in neuroendocrine regulation.

    Notizen: Summary The intramembrane structures of the pleated septate junction which occur in the junctional complex of the intestine of the chaetognath Sagitta setosa have been investigated. The pleated septate junction is made up of linear rows of irregularly shaped and sized particles, often fused into short rods, and pits which can be fused into furrows. The distribution of these structures on E and P faces depends upon the preparative methods used. Many of the morphological characteristics are the same as those of the “lower invertebrate pleated septate junction type” defined by Green (1981a). The physiological significance of this junction is obscure. On the basis of the presence of septate junctions (both of the paired septate junction and pleated septate junction types) which have mainly morphological characteristics of the “lower invertebrate pleated septate junction” we can add to the hypothesis that chaetognaths are not related to the molluscs and arthropods.

    Notizen: Summary Four immunoreactive endocrine cell types can be distinguished in the pancreatic islets of B. conchonius: insulin-producing B cells, somatostatin-producing A1 (= D) cells, glucagon-producing A2 cells and pancreatic poly-peptide-producing PP cells. The principal islet of this species contains only a few PP cells, while many PP cells are present in the smaller islets. Except for the B cell all pancreatic endocrine cell types are also present in the pancreatic duct. At least six enteroendocrine cell types are present in the gut of B. conchonius: 1. a cell type (I) with small secretory granules, present throughout the intestine, and possibly involved in the regulation of gut motility; 2. a C-terminal gastrin immunoreactive cell, probably producing a caerulein-like peptide; these cells are located at the upper parts of the folds, especially in the proximal part of the intestinal bulb; 3. a met-enkephalin-immunoreactive cell, present throughout the first segment; 4. a glucagon-immunoreactive cell, which is rare in the first segment; 5. a PP-immunoreactive cell, mainly present in the first half of the first segment; 6. an immunoreactive cell, which cannot at present be specified, located in the intestinal bulb. The latter four cell types are mostly located in the basal parts of the folds, although some PP-immunoreactive cells can also be found in the upper parts. Most if not all enteroendocrine cells are of the open type. The possible functions of all enteroendocrine cell types are discussed.

    Notizen: Summary An indirect immunofluorescence study of the frozen sections of the spleen of an anuran amphibian, Xenopus laevis, showed that lymphocytes bearing a small amount of immunoglobulin (Ig) were localized mostly in the white pulp of non-immunized toads. There were fewer fluorescent cells in the red pulp. In the toads hyperimmunized with human gamma globulin (HGG), cells with strong cytoplasmic fluorescence increased significantly in the outer part of the white pulp. Electron microscopy of spleens from these toads showed that plasma cells at different stages of maturation were abundant in the white pulp, whereas in the red pulp, a smaller number of maturer plasma cells were observed. These results indicate that, in contrast with its mammalian counterpart, the splenic white pulp of this anuran is the site where thymusin-dependent lymphocytes commence blast formation and transformation into plasma cells.

    Notizen: Abstract The effects of disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) on bone and serum chemistry were investigated in adult rabbits. EHDP was administered by subcutaneous injection at doses of 0.25, 2.5 and 10 mg/kg body weight/day for of 28 days. Blood samples were obtained weekly from each rabbit and serum levels of total calcium, ionized calcium, inorganic phosphate and alkaline phosphatase were determined. At the end of the treatment period all rabbits were sacrificed and the tibiae removed for chemical analysis and histological evaluation. The effect of EHDP administration on serum chemistry was both dose- and time-related. The highest of the three doses, 10 mg/kg/day, resulted in a time-related decrease in total serum calcium. This dose also caused a rapid but transient reduction in serum ionized calcium. The effect of EHDP on serum inorganic phosphate was biphasic. Administration of 2.5 mg/kg/day resulted in a time-related elevation in this parameter, whereas the 10 mg/kg/day dose resulted in a time-related hypophosphatemic response. There were no significant drug-related changes in tibial fat-free dry weight, ash weight, total calcium or total phosphorus values. However, administration of 2.5 and 10 mg/kg/day EHDP resulted in increased osteoid tissue as measured histologically. These results are compared with data from other EHDP studies, and discussed in relation to the maturity and growth-state of the experimental animals.

    Notizen: Abstract Scanning and transmission electron microscopy showed that a ceiling quantity (1.56 mcg) of antifungal antibiotic Pyrrolnitrin caused heavy damage to dermathophyteMicrosporon audouinii Gruby CBS 313-54in vitro. Suitable preparation technique made it clear that the changes involved consisted of hyphal collapse on the edge of the culture, with loss of euplasmic organelles identity and cell autolysis. The cell wall, however, was apparently undamaged. These findings fit in with the suggestion that the mode of action of the antibiotic leads to generalised lipoproteic membranes damage. They must, however, be considered as representing the result of the terminal phase of cell distress.

    Notizen: Abstract Three-day-old rats were fixed by perfusion with glutaraldehyde and thin slices were cut of the first molar germs. The slices were treated with EDTA and “activated” with buffered solutions containing Mg2+, Ca2+ or Zn2+. Incubation was carried out in buffered solutions (pH 8.5) containing inorganic pyrophosphate and Pb2+. In the Mg2+-activated specimens incubation products were localized to the plasma membranes in the stratum intermedium and the subodontoblastic area. Lead deposits were found on the periphery of the dentinal globules. Incubation products were more randomly distributed in Ca2+-activated specimens whereas those activated with Zn2+ displayed a deposition of lead precipitates mainly corresponding to that seen after activation with Mg2+. The findings are discussed in reference to the localization of alkaline phosphatase in the dentin-producing tissues and it is proposed that the results are indicative of the presence of an inorganic pyrophosphatase in these tissues.

Des conditions chimiques précises sont nécessaires pour obtenir de l'apatite, en tant que seule phase solide en équilibre dans la solution. Les résultats de diagramme de phase d'équilibre sont comparés avec ceux obtenus dans des milieux synthétiques.
    Kurzfassung: Zusammenfassung Es wurden Gleichgewichts-Phasendiagramme für das System CaO-P2O5-H2O bestimmt, indem hydrothermale Synthese-Techniken im Temperaturbereich von 300–600° und bei einem Druck von 2 Kb H2O verwendet wurden. Es wurde gut-kristallisiertes Hydroxyapatit erzeugt und charakterisiert. Es wurden geringe Unterschiede in den Parametern der Zelleinheiten festgestellt, welche von der angewandten Temperatur und der Zusammensetzung des Startmaterials abhingen. Es waren genaue chemische Bedingungen nötig, um Apatit als die einzige feste Phase im Gleichgewicht mit der Lösung zu erhalten. Die Resultate der Gleichgewichts-Phasendiagramme werden mit früheren Untersuchungen mit der Synthesetechnik verglichen.
    Notizen: Abstract Equilibrium phase diagrams have been determined for the system CaO-P2O5-H2 using hydrothermal synthesis techniques in the temperature range 300–600° and 2 Kb H2O pressure. Well-crystallized hydroxyapatite has been produced and characterized. Small variations in unit cell parameters dependent on temperature of synthesis and bulk composition of the starting materials have been determined. Precise chemical conditions were required to obtain apatite as the only solid phase in equilibrium with solution. Equilibrium phase diagram results are compared with previous synthetic investigations.

    Notizen: Abstract Acid phosphatase activity was mainly localized in the lysosomes in all the regions of the outer epithelium. The transitional portion of the outer epithelium showed more intense activity than the other regions. During shell regeneration the activity of this portion decreased to a minimum level at 12 hours and was restored to normal at 72 hours. The other regions showed no change of activity during shell regeneration. It is postulated that the acid phosphatase in the transitional protion is responsible for conferring calcifiability to the organic matrix of the shell.

    Notizen: Summary The urethra of the rat was examined by transmission and scanning electron microscopy. Under a transmission electron microscope flask-shaped chromaffin cells containing membrane-bound osmiophilic granules were seen to possess microvilli at their apical surfaces. The microvilli projected into large extracellular spaces which were apparently in continuity with the lumen of the urethra. Using scanning electron microscopy, a surface view of the lumen of the urethra was obtained. It showed a gently undulating surface with distinct intercellular boundaries. Scattered over the surface were numerous deep depressions between individual cells. These were thought to correspond with the large extracellular spaces into which microvilli had been seen to project. It is suggested that urethral chromaffin cells may “trigger” the afferent part of a reflex causing contraction of the urethral longitudinal muscle.

    Notizen: Summary The ultrastructure and protein content of the five subfractions of the crude mitochondrial fraction from the brain stem of the 1-day old and adult rat was examined. The morphological composition of the subfractions after fixation in glutaraldehyde and osmiumtetroxide in the adult rat brain stem resembled that previously reported for the whole brain; synaptosomes sedimented in a sucrose gradient in subfractions C and D. In the 1-day old rat, mature synaptosomes were found in subfractions A, B, C and D; E contained mainly free mitochondria. 80–95% of the processes in the adult and 10–30% in the 1-day old rat contained synaptic vesicles which were of four types: (1) small agranular vesicles (2) large dense core vesicles (3) large agranular vesicles (4) coated vesicles. Pre- and postsynaptic membrane thickenings were demonstrated in many nerve-ending particles. In the subfractions of the 1-day old rat the protein content was one half and the distribution resembled that in the adult. Evidently nerve endings develop faster in the brain stem than in cortical areas; a serotoninor adrenergic origin of the early synaptosomes is suggested.

    Notizen: Summary The paranodal zone of myelinated nerve fibers from the CNS of cats has been studied by means of the lanthanum technique and after freeze-etching. Special interest was given to the axonal-glial zones of contact in this region. Examination of the membranes of the paranodal pockets and the axolemma revealed that the membrane thickenings which occur between the adjacent membranes actually consist of bands. The bands show a specific pattern of diagonal running subunits 10 nm apart. These substructures of the axonal side of the membrane contacts protrude outwards, i.e. towards the glial membrane, where they fit into depressions on the glial membrane surface. This arrangement in form of a “zip-fastener” makes the whole structure very flexible. The subdivision of the bands was identified with both techniques. The different membrane aspects obtained by the freeze-etching technique are discussed in respect to the splitting theory, and a hypothetical model of this membrane specialization is presented.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Standardgerinnsel aus Rinderfibrin wurden in der Kammer einer Versuchsanordnung einem mit Wasser nahezu gesättigten Luftstrom ausgesetzt. Die Gerinnsel kollabierten, und ihr Restwasser wurde in Zeitabständen durch Hitzetrocknung bei 105°C bestimmt. Es resultierte eine zeitabhängige Kurve, die aus 3 Phasen besteht. Elektronen-mikroskopische Aufnahmen zeigen, daß infolge eines Verlustes des freien Wassers während der ersten Phase das Pibringerüst des Gerinnsels kollabiert. Während der dritten Phase kollabiert das Proteingerüst der Fibrinmoleküle infolge des Verlustes von Strukturwasser. Während der zweiten Phase besteht das Gerinnsel aus raumfüllend gepackten Fibrinmolekülen. Das Strukturwasser beträgt dann etwa 5,25 g/g Trockengewicht; die Höhe ist etwa 180 μm und ergibt den wahrscheinlichsten Wert des Strukturwassers, 6,25 g/g Protein. Wenn dieser experimentelle Wert mit entsprechenden, theoretisch zu erwartenden Werten verglichen wird, die von elektronenmikroskopischen Molekularmodellen für Fibrin hergeleitet wurden, so ergibt das Gerinnsel, das aus raumfüllend gepackten Pentagondodekaeder-Modellen besteht, mit etwa 8 g Strukturwasser/g Fibrin die beste Näherung an den im Experiment erhaltenen Wert. Dieses Modell kann — im Gegensatz zu anderen Modellen — Massen-, Volumens- und Morphologieverhältnisse des Gerinnsels und des Moleküls erklären. Die verbleibende Differenz zwischen wahrscheinlichstem experimentellem und theoretischem Strukturwasser-Gehalt ist leicht durch experimentelle Ungenauigkeiten und Anwesenheit von Nicht-Fibrin im Molekularvolumen erklärbar. Mit an Sicherheit grenzender Wahrscheinlichkeit repräsentiert deshalb das Pentagondodekaeder-Modell die Allgemeinstruktur des Fibrin- und Fibrinogenmoleküls.
    Notizen: Summary Standard clots of bovine fibrin collapsed in the highly humid air stream of a laboratory device. Their residual water, determined by heat drying at 105°C, resulted in a time dependent curve composed of 3 distinct phases. Electron micrographs show that during the first phase free water is released causing a collapse of the fibrous framework of the clot, during the third phase structural water is lost causing a breakdown of the molecular protein. The clot of the second phase consists of space fillingly packed fibrin molecules, whose directly determined structural water is approximately 5.25 g/g dry wt., but whose height (approximately 180 μm) results in the most probable value 6.25 g structural water/g protein. If this amount is compared with respective theoretical values of electron microscopic molecular models of fibrin, the clot consisting of space fillingly packed pentagon dodecahedron models yields approximately 8 g structural water/g fibrin, closely to the experimental value. In contrast to other models, this model explains simultaneously the shape of the curve, mass and volume relationships and morphology of clot and molecule. The remainding difference between the most probable experimental and this theoretical value can easily be attributed to experimental errors and impurities contained in the volume of the molecule. Consequently, the general structure of the fibrinogen and fibrin molecule is represented by the pentagon dodecahedron model.

    Notizen: Summary The distribution of monoamines in the brain of Bufo poweri was investigated with the Falck-Hillarp method. Green fluorescence was observed in neurons of the diencephalic preoptic recess organ (PRO) and its so-called additional cells, in the paraventricular organ (PVO) and its so-called accompanying cells, in the nucleus infundibularis dorsalis (NID) and in the mesencephalic nucleus reticularis mesencephali (NRM). Yellow fluorescence was found in some cells of the PVO and NID and in neurons of the caudal part of the NRM. Extensive tracts of aminergic fibres are described running caudo-rostrally and mainly terminating in the septa and striata of the telencephalon. It is suggested that the aminergic nuclei mentioned above partly contribute to the ascending tracts. Other fibres, originating in the PVO and NID are curving centro-caudally towards the median eminence. Although with the electron microscope some apparently aminergic nerve fibres were detected in the pars intermedia, such fibres have not been observed fluorescence microscopically. The ultrastructural study of the PRO has shown the presence of two types of aminergic liquor-contacting neurons. Among the distal cells of the PRO, aminergic and peptidergic neurons were distinguished. The presence of PRO additional cells, which are considered as a rostral extension of the PRO, is discussed with regard to the terrestrial habitat of the species.

    Notizen: Summary Electron microscopy and autoradiography (H3-proline) were used to determine the cell(s) responsible for collagen synthesis and lamination in the mesoglea of the sea anemone, Aiptasia diaphana. Mesogleal collagen is synthesized by the epidermal epitheliomuscular cells which contain much rough endoplasmic reticulum, and secretory vesicles attached to microtubules which cross the basal plasmalemma and anchor in the basement membrane; these cells incorporate large amounts of H3-proline. The mesogleal collagen fibers are non-striated, have clear centers, and a diameter of 200–260 Å; their walls are composed of 65 Å diameter subunit fibrils which appear to be helically oriented. Epitheliomuscular cells rest upon a subepidermal basement membrane which is composed of mesogleal collagen fibers, 65 Å subunit fibrils, and dense granules. This subepidermal basement membrane labels definitively with H3-proline, and is the region where soluble collagen precursors apparently form subunit fibrils which associate to yield mesogleal fibers. The columnar mesogleal collagen fibers are arranged to form layers: the fibers of each layer have the same longitudinal orientation, while those of adjacent layers display an approximate orthogonal arrangement. It is felt that the subepidermal basement membrane is responsible for this organization of mesogleal fibers. Mesogleal amoebocytes do not label with H3-proline and show no ultrastructural evidence of collagen secretion.

Electron microscopic examination of the hippocampus, mid-brain reticular formation and the area postrema revealed marked diminution in the electron density of the pigment granules and vacuolization. This type of lipofuscin was detected in some phagocytic cells and in the capillary endothelium. Conspicuous vacuolization of the capillary wall was discernible. These changes were not observed in the “control group” of animals.

    Notizen: Summary The development of the submandibular gland of the mouse was studied by means of electron microscopy, from the 14th day of gestation up to birth. In the first two days the acini are solid and their cells contain polyribosomes and a few lamellae of endoplasmic reticulum. Beginning with the 16th day secretory granules appear and rapidly fill an increasing number of cells. The different electron density of the granules makes it possible to distinguish 1. two types of granules, dense and pale, and sometimes intermediate ones, 2. “polymorphic” granules. The latter consists of electron dense and electron pale parts combined in different configurations. The possible significance of the various types of granules is discussed.

    Notizen: Summary Two distinct types of neuron in the cat trochlear nucleus (one large, one small) are described, the α- and γ-motoneurons, respectively. Four types of terminals are observed which establish axo-dendritic synapses. Two of them (Types I and II) perform axo-somatic synapses as well. Terminals en passant (Types I and II) are predominant. The Type I terminal is long and slender with a characteristic distribution of the axoplasmic organelles and the unique feature of a relative narrowing of the synaptic cleft as compared to the width of the neighboring extracellular space. Its vesicle population is pleomorphic and a conspicuous glial barrier surrounds the synaptic zones. The Type II terminal differs slightly from Type I, revealing a wider synaptic cleft and lacking a characteristic distribution of the axoplasmic organelles. The type III terminal is rarely observed performing axo-somatic synapses, but is a common finding in the neuropil. Post-junctional dense bodies are often present in its axodendritic synapses. The Type IV nerve terminal performs axo-dendritic synapses and is characterized by a rich content of large granulated vesicles. Axo-axonal synapses are observed only very rarely. The synaptic organization of the feline trochlear nucleus is compared with the synaptic morphology of the oculomotor nuclei of inframammalian species (Waxman and Pappas, 1971). In addition to certain similarities (e.g., richness of synapses en passant), significant differences are encountered: the present study provides no morphological evidence for electrotonic transmission in the trochlear nucleus of cat.

The intrafusal fibres of rabbit and guinea pig spindles have been studied using quantitative stereological techniques at the ultrastructural level. The crosssectional areas occupied by myofilaments have been measured in the polar and equatorial regions of both types of intrafusal fibre. There are considerably fewer myofilaments in the equatorial regions of both types of fibre compared with their polar regions.

    Notizen: Summary The effects of a chronic ACTH administration (up to 12 consecutive days) on the 3H-thymidine uptake by the mitochondrial compartment of rat adrenal zona fasciculata were investigated by high resolution autoradiography, and compared with the changes in volume and number per cell of these organelles induced by the hormonal treatment. Up to the 9th day of treatment there is a significant increase in the tracer incorporation into adrenocortical mitochondria which is coupled with a significant increase in the volume of the organelles. After 12 days of hormone administration a significant decrease in the 3H-thymidine mitochondrial uptake is found, which is associated with a conspicuous increase in the number of mitochondria per cell and a net decrease in their average volume. The data are discussed in the light of evidence indicating that mitochondria possess a genetic apparatus largely independent of nuclear control. It is hypothesized that ACTH controls the growth and proliferation of adrenocortical mitochondria and that the mechanism of this action of ACTH involves stimulation of the mitochondrial DNA synthesis.

    Notizen: Summary Two intracellular phospho-hydrolases are revealed in cells of Trichomonas vaginalis. One of the enzymes is an acid phosphatase and the other a nucleoside triphosphatase (ATP-ase). Both enzymes are found to be present in the Golgi saceuli and vesicles of the cells. The ATP-ase activity has prevalence to the more superficially positioned saceuli of the Golgi region and the acid phosphatase to the more deeply positioned saceuli. Both enzymes are also found in the phagolysosomes but only acid phosphatase could be demonstrated in the small primary lysosomal structures of the cell cytoplasm. The findings indicate that both enzymes are active in the hydrolytic processes associated with endocytosis of foreign matter.

    Notizen: Summary Freeze-fracture studies were conducted on the membranes of normal cockroach hemocytes. The plasmalemma is asymmetric with the A fracture face containing 80–100 Å membrane intercalated particles at a concentration of ∼2500/μ2. The B fracture face contains 120–150 Å particles with a relatively low density (∼800/μ2). The nuclear envelope displays an asymmetry with the A fracture face containing ∼1500 particles/μ2 and the B face containing ∼300/μ 2. No significant particle size differences were observed in nuclear envelope fracture faces. Two types of symmetric membranes were also found in these cells. Both A and B fracture faces of the membrane surrounding the numerous cytoplasmic inclusion bodies contain particle sizes and concentrations similar to the B face of the plasmalemma. A second type of symmetry was observed in cells apparently engaged in “exocytosis”. Vesicles (∼0.1 μD) from this process were completely particle free on both fracture faces. Such particle free vesicles could be found in the cytoplasm, attached to the plasmalemma, or completely separated from the cell.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Die perinatale Entwicklung des juxtaglomerulären Apparates, insbesondere das Verhalten der epitheloiden Zellen wurde an 18 trächtigen Ratten and 65 Embryonen bzw. Jungtieren elektronenmikroskopisch untersucht. Parallel dazu wurde die Reninkonzentration bzw. der Reningehalt bestimmt. Spezifische Sekretgranula finden sich erstmals am 18. Tag der Fetalzeit, und zwar in den epitheloiden Zellen des juxtaglomerulären Apparates juxtamedullärer Nephrone. Die Granula machen einen Struktur- und wahrscheinlich auch Funktionswandel durch, der eine Einteilung in drei verschiedene Granulatypen nahelegt. Diesen drei Typen spezifischer (reninhaltiger) Sekretgranula werden die Lysosomen als unspezifische Einschlüsse der epitheloiden Zellen gegenübergestellt. Im einzelnen kommt es während der perinatalen Phase in den fetalen Epitheloidzellen zuerst, d.h. pränatal, zu einer Hypertrophie des rauhwandigen endoplasmatischen Retikulums und des Golgi-Apparates. Hierin sehen die Autoren einen Hinweis auf eine erhöhte Proteinsynthese mit entsprechender Zunahme der Transport- und Konzentrationsvorgänge. Nach der Geburt findet sich dann auch eine deutliche Zunahme der Sekretgranula vom Typ III, die als reife Reningranula (mature granules) angesehen werden. Die bisher noch nicht beschriebenen kleinen Sekretgranula vom Typ I dagegen kommen in weniger entwickelten Epitheloidzellen häufiger vor. Bestimmungen der Reninverteilung zwischen Zytoplasma und den durch Differential-zentrifugation gewonnenen Granula-Fraktionen ergeben auffallende Parallelen zu den morphologischen Befunden. Diese sprechen im allgemeinen dafür, daß “freies Renin” im Ergasto-plasma und “gebundenes Renin” in den Sekretgranula vorliegt. Mit zunehmendem Alter scheint es außerdem — parallel zur relativen Abnahme des Ergastoplasmas und Zunahme der reifen Reningranula — zu einer Verminderung des extragranulären zugunsten des granulär gebundenen Renins zu kommen.
    Notizen: Summary The perinatal development of the Juxtaglomerular apparatus (JGA), with particular reference to the epithelioid cells, was studied. Electron microscopic studies were carried out on 18 gravid rats and 65 embryos with corresponding determinations of renin concentration and the renin contents by bio-assay. Specific secretory granules of the epithelioid cells of the JGA of juxtamedullary nephrons are first observed on the 18th day of gestation. The classification into three types of secretory granules suggests that they undergo a structural and probably also a functional transformation. All three types of secretory granule may be regarded as specific, renin-containing, cytoplasmic organelles. During the last days of gestation, the rough-surfaced endoplasmic reticulum and the Golgi apparatus become hypertrophied in epithelioid cells, indicating an increased protein synthesis as well as transport and concentration processes. After birth, there is a definite increase in the number of Type III secretory granules-which may be regarded as mature granules. The small Type I secretory granulesnot previously described-are found more frequently in the immature epithelioid cells. Determinations of the renin distribution in the plasma and in fractions of cytoplasmic constituents (total homogenate, secretory granules) prepared by differential centrifugation showed striking congruence with the morphological findings. It is generally assumed that “soluble renin” occurs in the endoplasmic reticulum and “bound renin” in secretory granules. A shift from extragranular to granular (bound) renin, occurs with increasing age, having its morphological equivalent in the relative decrease of the endoplasmic reticulum and increase in mature secretory granules.

A morphometric analysis of the cell types in the pituitary gland of the adult, winter, fresh-water stickleback, Gasterosteus aculeatus form leiurus, reveals differences between both the relative and absolute volumes of the various organelles in different cell types. The morphometric data on the relative volumes of the organelles, together with section profile diameters of the secretory granules and information on the surface area: volume ratio of the nuclei are then used to build “reconstruction drawings” of “average” cells. A distinction is made between the ultrastructural description and identification of cell types.

    Notizen: Summary Lateral line organs in young salamanders of the species Ambystoma mexicanum were investigated with scanning and transmission electron microscopy. They were found to differ from the lateral line organs in adult animals (1) by being lower, having short hair cells and supporting cells, (2) by the hair cells having areas of lateral contact, (3) by the occasional presence, at the edge of the organ, of hair cells at an early developmental stage. Two types of nerve endings are seen: (1) afferent, and (2) less commonly, efferent vesiculated ones. Synaptic bodies have been seen in the cytoplasm without association to afferent synapses.

    Notizen: Summary Qualitative and quantitative studies were made to determine the amount of nerve fiber supplying corpora lutea (CL) of rats during the oestrous cycle and pregnancy and sow CL during days 4–6 after ovulation. Fluorescence microscopy of freeze-dried, paraformaldehyde treated (Falck-Hillarp method) rat ovaries reveals adrenergic nerve fibers which run along with vessels and form a network among interstitial gland cells. Nerve fibers do not enter the granulosa cell layer in follicles or CL. In the CL circumference both vascular and non-vascular nerves occur the latter being related to the fibromuscular layer and probably innervating smooth muscle cells. No striking differences exist between the innervation of the ovary in non-pregnant and pregnant rats. Bodian and methylene blue staining did not contribute to a more detailed knowledge of rat ovary nerve supply. Electron microscopic quantitative analysis of rat and pig CL (rat: day 18 of pregnancy; pig: day 4–6 after ovulation) revealed no axon profiles in 2.000 grid squares (one square measuring 2.25×10-2 mm2) of randomly taken CL sections. Thus it was possible to calculate an upper limit of 133 μm of nerve fibers per 1 mm3 CL tissue, in case there were any at all.

    Notizen: Summary The interaction between the trophoblast and the maternal epithelium at early implantation was studied by means of light and electron microscopy. The uterine horns were fixed in situ and a double-embedding method was used to locate implantation sites. Observations were made on mice killed at 2 hour intervals 90–116 h. post coitum which covered the following stages: pre-attachment (i) with zona pellucida intact (ii) with zona pellucida in dissolution (iii) after loss of the zona; attachment; adherence; and invasion. The intact zona pellucida was electron opaque and of uniform density. In the stage of apparent dissolution it became electron dense and was trapped between trophoblast and epithelium. At preattachment the trophoblast cells were round. Subsequently they became long and attenuated, often with lysosomes in the cytoplasm proximal to the epithelial layer. Epithelial cells, which could be seen in various stages of degeneration were apparently phagocytosed by the trophoblast. Occasional pyknotic epithelial cells were seen, as well as some apparently normal ones which contained cytosegresomes. The possible reasons for their presence are discussed. The microvilli of the epithelial cells changed from regular and pointed at preattachment to short and irregular at adherence and invasion.

    Notizen: Summary The tunica albuginea in ovaries of sexually mature rabbits is only a few cell layers thick but is much more densely packed with collagen bundles than the broad theca externa of the Graafian follicles. Smooth muscle cells were demonstrated in theca externa around the whole follicle; ultrastructural evidence of smooth muscles in the apical region of theca externa in rabbit follicles has not been presented previously. At 8 hrs after injection of an ovulatory dose of HCG oedema was observed below the germinal epithelium covering preovulatory follicles. In the oedematous area, dead or degenerating cells could be found, and the underlying tunica albuginea contained degenerated fibroblasts and showed dissociation and fragmentation of the collagen. The alterations gradually proceeded inwards, and, at 9.5 hrs and later, marked changes were obvious even in the innermost portion of theca externa. The observation that the changes started, and were most pronounced, in the collagen-rich tunica albuginea and gradually proceeded inwards, supports the hypothesis that lytic substances emanating from the surface epithelium covering preovulatory follicles play a central role in effecting follicle rupture.

    Notizen: Summary The possibility of a direct retinohypothalamic projection was reinvestigated in Passer domesticus by electron microscopy following left unilateral retinectomy. To avoid misinterpretation of non-specific degeneration, the course of degenerative changes was observed at intervals of 6, 12, 24, 48 and 96 hours after operation. Of the hypothalamic areas examined in only one, the contralateral suprachiasmatic nucleus, was it possible to identify reliable indications of secondary anterograde degeneration comparable to those observed in the contralateral optic tectum. Single dark profiles within the supraoptic nucleus and the basal infundibular (tuberal) nucleus showed neither internal changes in structure nor an increase in number per unit area in retinectomized birds. Since photoperiodically induced gonadal growth occurs in totally blinded birds the functional significance of the retinohypothalamic projection to the suprachiasmatic nucleus is open to discussion.

    Notizen: Summary Kölliker's and Hatschek's pit and the wheel organ in the head region of Amphioxus have been studied with the electron microscope. Kölliker's pit is lined by ciliated cells lacking specific ultrastructural characteristics which might suggest an olfactory function. No nerve terminals have been found near this structure. The wheel-organ consists of tall ciliated cells, which occasionally have been found to be innervated and which are clearly marked off the rest of the epithelial lining of the oral cavity. The epithelium of Hatschek's pit again is ciliated and possesses markedly euchromatin rich nuclei. In the area of Hatschek's pit numerous epithelial cells have been observed which possess accumulations of small granules and vesicles in their basal cytoplasm. Such basally granulated cells occur in the immediate neighbourhood of blood spaces of the glomus and other vessels; the connective tissue between blood and epithelium is loosened up or has completely disappeared. The question of homology of the structures investigated with the olfactory groove and hypophysis of vertebrates is briefly discussed.

    Notizen: Summary The elastica interna was studied in the notochord sheath of the caudal one third of the tail of Rana rugosa tadpoles. Dense amorphous bodies are present in depressions at the surface of superficial notochord cells. These bodies are intimately associated with microfibrils. The notochord basement membrane tends to cover the surface of the dense amorphous bodies but often appears to fuse with them and disappear. There is a striking close resemblance between the amorphous dense bodies and the elastica interna. These observations, together with the observation of dilated cisterns of granular endoplasmic reticulum with dense contents in the cytoplasm of superficial notochord cells, suggest that the amorphous dense bodies and the elastica interna are produced by superficial notochord cells.

    Notizen: Summary The fine structure of the juxtaglomerular apparatus in the toad, Bufo bufo was investigated.—It is suggested that the granules in the media cells of the afferent arteriole are formed from the Golgi apparatus. Many granules have a content of lamellar material. The media cells do not show the ultrastructural features of active secretory cells. In the media cells, the myofilaments are situated near the vascular lumen. The other cell organelles, including the granules, are preferentially located at the opposite pole of the cell in the neighbourhood of the macula densa cells and the adventitial nerve fibres. In these regions the media cells show many pinocytotic vesicles. The nerve fibres innervating the juxtaglomerular cells are non-myelinated and their varicosities contain dense core vesicles. The basement membranes of media cells and adjacent macula densa cells occasionally fuse, which may indicate a functional relationship between these cells.

    Notizen: Summary The capacity of certain B-lymphocytes to bind complement (demonstrated by erythrocyte-antibody-complement complexes, EAC) was used as a marker of the B-lymphocyte regions of the human white splenic pulp. This was carried out on cryostat sections in order to correlate enzyme histochemical findings (5-nucleotidase, ATPase, acid phosphatase, nonspecific esterase, alkaline phosphatase) to immunological functions. EAC were typically found in the follicle centers and marginal zone, whereas periarteriolar lymphocyte sheaths remained negative. The enzyme histochemical patterns of lymphocytes and reticulum cells allowed a clear distinction between areas with and those without EAC-binding. This was shown most clearly when the following enzymes were demonstrated in combination: 5-nucleotidase (5-Nase) + alkaline phosphatase, ATPase + acid phosphatase, and nonspecific esterase + acid phosphatase. 5-Nase correlated best to EAC-positive areas with a positive reaction in follicle wall lymphocytes and dendritic reticulum cells, whereas periarteriolar sheaths contained no 5-Nase-positive structures. Reticulum cells around periarteriolar lymphocyte sheaths at the margin of the red splenic pulp showed a particularly strong alkaline phosphatase reaction. The reticulum cells of the areas containing B-lymphocytes and those of the EAC-negative periarteriolar regions, which probably contain T-lymphocytes, were specifically labeled for different sets of enzymes. These findings suggest that specialized, morphologically different reticulum cells may be the “guide rails” for the different freely circulating lymphocyte populations.

    Notizen: Summary Rodlet cells from many species of fresh water fish were examined with the electron microscope. Observations of these cells in the epithelia of intestine, gill, and other organs favor the interpretation that rodlet cells are secretory cells rather than protozoan parasites (Rhabdospora thelohani). Evidence is presented that rodlet cells develop from undifferentiated cells near the epithelial basement membranes and migrate“upward” to secrete their contents in a holocrine manner at the epithelial surface. The major secretory product is packaged in unique club-shaped sacs which contain a central core of highly electron dense material with less dense material surrounding it. Secretion often appears to be accomplished by active contraction of a 0.5 μm fibrillar border resembling smooth muscle which lies just inside of the plasma membrane. Some rodlet cells, however, merely break apart and release their contents.

The ultrastructural localization of reaction products from ruthenium red and tricomplex flocculation staining indicates that the epithelial surface is coated by saturated phospholipids and mucopolysaccharides. Utilization of various chemical markers demonstrates the epithelial cell to be physiologically active in the transport of certain substances. Evidence supporting the concept that the lamellar inclusion bodies are responsible for the surfactant lining is presented.

    Notizen: Summary Arterio-venous anastomoses (AVA) in gill filaments of Tilapia mossambica exhibit a distinct polarity. Two different types of highly specialized endothelial cells, both of epitheloid shape, line these vessels: Type I cells, contacting the arterial lumen, are elongated and about two to three times as large as type II endothelial cells. Their surface is increased by tentacular protrusions which reach far into the arterial lumen. Filament whorls forming tubelike structures with centrally located glycogen granules are abundant in these cells. Type II endothelial cells are located proximal to the central venous sinus (CVS). Their less abundant and more electron dense cytoplasm is free of filament whorls. There are also intermediate cell forms at approximately the middle of each anastomosis. Short cell processes protrude from all endothelial cell types into the AVA lumen. Outside the indistinct vascular basement lamina, a layer of cover cells tightly envelopes the AVA. These cells are, however, absent around the part of the AVA adjacent to the CVS. Here the endothelial cells are in immediate contact with the interstitium. Endothelial cells sheathed by cover cells reach the interstitium through basal foot processes. Nerve fibre bundles regularly come into close contact with the AVAs. Possible functions of the AVAs, including osmoreception are discussed.

    Notizen: Summary Electron microscopy has been utilized to determine fates of sacs of ribosomes (variously termed “transosomes”, “unique organelles”, or “lining bodies”) formed in the follicular cells of avian follicles and subsequently entering-or being taken in-by the oocytes. Small follicles (0.5 mm diameter) of laying hens, a hen afflicted with Marek's disease and prelaying pullets were examined in this regard. In the case of the hen with Marek's disease and the prelaying pullets, sacs of ribosomes were found to be present within the oocyte but those present in the interior of the cell were in the form of digestive vacuoles. In a fourth group of hens, receiving actinomycin D, larger oocytes (2.0 mm diameter) exhibited breakdown of the membranous vesicles in which the sacs of ribosomes entered the oocyte, as well as dissolution of membranes surrounding the forming yolk granules and the membranes of coated vesicles. Annulate lamellae were present in oocytes of the hen afflicted with Marek's disease and appeared within at least 24 hours in oocytes from hens receiving actinomycin D.

    Notizen: Summary Palisade-shaped nerve endings of the small normal hairs of the rat snout were examined with the electron microscope (fixation by perfusion). The terminals are located inside the ‘glassy membrane’ in the area of the neck of the hair root. The 10–20 radially arranged terminal axons are in direct contact with the basement membrane of the epithelium of the external root sheath. The axons are surrounded on all sides by leaf-shaped processes of the Schwann cells. The surfaces of these cell processes are marked by numerous vesicle-like invaginations (approx. 1000 Å dia.). Transverse sections from several areas of the palisadeshaped nerve endings are compared with longitudinal sections. In the upper area ‘empty’ vesicles (approx. 500–600 Å in diameter) occur, along with electron-dense vesicles (approx. 800–1100 Å in diameter); in the middle area, the axons are distended and contain accumulations of mitochondria.

    Notizen: Summary The excretory organs of Amphioxus occur as segmentally arranged structures throughout the pharyngeal region and may be divided into three components: the solenocytes, the renal tubule, and the renal glomerulus. The solenocytes possess foot processes that rest upon the coelomic surface of the ligamentum denticulatum. The tubular apparatus of the solenocytes consists of ten triangular rods surrounding a central flagellum. The distal end of the tubular apparatus enters branches of the renal tubule. The renal tubule eventually opens into the atrial cavity of Amphioxus. The renal glomerulus is a sinus within the connective tissue of the ligamentum dentieculatum where it connects elements of the branchial circulation with the dorsal aorta. The renal glomerulus, like other blood vessels of Amphioxus, lacks an endothelial lining. If Amphioxus is adapted to artificial sea water at different concentrations there is no change in kidney morphology suggesting that Amphioxus is either is osmotic with its environment or is osmoregulating with other organs.

    Notizen: Summary The dorsal lobe and the coagulating gland of the rat prostate were studied after administration of the anti-androgenic compound Cyp A for varying periods (7–20 days). There was a general involution of the organs with macroscopic atrophy and reduced amount of secretory material. Ultrastructurally the cells were pale, contained less organelles as compared with the controls, loss of cytoplasm and reduction both of cell height and width. The major changes also included reduction of the Golgi area, the rough endoplasmic reticulum and the number of free ribosomes. Within the dorsal lobe prominent nuclear changes were found, comprising different types of intranuclear inclusions of membranous configurations. The alterations observed in the present study are similar to the changes which were found in castrated animals and are consistent with atrophy. The results are discussed in relation to the available biochemical and autoradiographic data on the different lobes.

    Notizen: Summary Mast cells in the tongue and lamina propria of the duodenal mucosa, representing two cell types with different morphological, histochemical and functional properties have been studied under the electron microscope. Both cell types were found to contain similar moderately electron dense and homogeneous granules surrounded by a tight membrane constituting the basic ultrastructural characteristics of mast cells. The mucosal mast cells, however, contained fewer granules per cell and the individual granules often exhibited considerable variation in size. In addition the mucosal mast cells, unlike those of the tongue, peritoneum and skin, had a smooth plasma membrane lacking microvilli and possessed irregularly shaped or lobulated nuclei. In the tongue mast cells, administration of Compound 48/80 caused the formation of vacuoles around the granules, expulsion of granules from the cells and disintegration of the granular matrix, but no changes that could be attributed to 48/80 could be found in the mucosal mast cells.

    Notizen: Summary The effects of a chronic administration of ACTH (up to 36 consecutive days) on the mitochondria of the zona fasciculata of the rat adrenal cortex were investigated by stereologic techniques. It was found that, while the volume of the mitochondrial compartment significantly increases in relation to the duration of treatment, the size and number of mitochondria display a different pattern. Up to the 9th day of hormone treatment mitochondria significantly increase in volume, whereas their number per cell is only slightly increased. After 12 days of ACTH-treatment there is a tremendous increase in the number of organelles per cell, resulting in a significant decrease in their average volume. After 24 and 36 consecutive days of treatment the number of mitochondria per cell as well as their average volume both show a slight but significantly constant increase. The hypothesis that ACTH controls the processes of growth and division of adrenal mitochondria is discussed in the light of evidence indicating that mitochondria contain a complete genetic apparatus largely independent of nuclear control.

    Notizen: Summary The action of luteinizing releasing hormone (LH-RH) on FSH and LH-gonadotrophs from the anterior pituitary glands of normal rats has been studied with electron microscopy. After 15 min. incubation with LH-RH, LH cells showed secretory granules undergoing exocytosis, but FSH did not appear to release secretory products. On the other hand, incubations with the releasing hormone for 3 and 6 hr stimulated exocytosis from both FSH and LH-gonadotrophs. Of more interest was the finding that FSH cells appeared as typical “signet cells” of castration when glands were incubated for 3 and 6 hr with the releasing hormone. The appearance of such “signet cells” was prevented if puromycin was added to the medium together with LH-RH.

    Notizen: Summary Major ultrastructural changes in neurons were studied during sequential periods of hypostomal regeneration in Hydra. Some neurons remain unaffected except that at certain periods following amputation, they become more active in neurosecretory production. Other neurons in various stages of differentiation were also observed. Most emphasis was placed on degenerating neurons showing a loss of organelles and destruction of the perikarya. Certain large, membrane-bounded structures (up to 1.7 μ in diameter) suggested tentatively to be of a lysosomal-like nature, may be partly responsible for the degenerative process. The neurites of these cells first assume a beaded appearance and/or contain bulbous endings. The eventual isolated fragments of neurites contain typical membrane-bounded neurosecretory droplets (850/1700 Å in diameter) which disintegrate forming particulate materials (350 Å in diameter). Following complete disruption of the neurites, some of the granules accumulate in the extracellular spaces before they are disintegrated. From the data presented in this and the preceding paper, it is suggested that these particles, derived from neurosecretory droplets, may be responsible for the stimulation of: 1) interstitial cell differentiation into neurons, 2) the increased activity in neurosecretory production in normal cells, 3) the conspicuous increase in neurite length and consequently the exaggerated degree of anastomosis, and 4) the possible capacity of neurons to regenerate neurites.

    Notizen: Summary The ultrastructure of the lamina propria in the ovine seminiferous tubule was studied from 54 days (postcoitum) fetuses up to sexual maturity. In young animals (aged about two months) the effect of hypophysectomy and/or hormones was also analyzed. Initially, large areas of the lamina propria already show signs of morphological differentiation. The non cellular component is a single or sometimes double lamella. Outwardly, the cellular component is represented by rounded connective tissue cells or by elongated cells, exhibiting 80 Å filaments in the cytoplasm and arranged in 3 to 4 layers. By the first week after birth, the non cellular component is formed by 8 to 10 lamellae and the cellular component shows typical contractile cells, the fibroblasts being now located only in the periphery. Basically, the same aspect is found in the adult ram, thus differing from the more extensively studied prototypes of lamina propria architecture of rodents or primates. In as much as the sheep lamina propria differentiates precociously, probably induced by fetal testicular hormones, it is relatively insensitive to hypophysectomy or hormonal substitution. Hypophysectomy and cyproterone acetate administration cause intracytoplasmic lipid deposition in the contractile cells, the other components being not changed. Some questions are raised concerning the role of this peculiar lamina propria in the function of the blood-testis barrier.

    Notizen: Summary Neural elements within the parenchyma of the sebaceous gland have not been reported previously. Nerve endings have been observed only in the connective tissue surrounding the gland or in close association with the undifferentiated basal cells. In this study, electron microscopy revealed the possible presence of nerve endings (or terminal portions of neural elements) in the suprabasal level of functional sebaceous glands of pinnae of white rats. Morphologically, there are two distinct types of nerve endings. Type 1 is bordered by a membrane of relatively irregular contour and contains a single mitochondrion, various-sized vesicles, numerous microtubules, fine neurofilament-like fibrils, and occasional ribosome-like granules. Type II is also bordered by a membrane, but its contour was relatively smooth and rounded. Moreover, Type II contains many mitochondria, varying in size, density, and the arrangement of cristae. While ribosome-like granules are scattered throughout the structure in relative abundance, there are scarcely any fine neurofilament-like fibrils or microtubules. Whether these two structures are sensory or autonomic fibers could not be determined by electron microscopic examination.

    Notizen: Summary The ultrastructure of the pineal gland of 18 human fetuses (crown-rump lengths 30–178 mm) was investigated. The pineal gland exhibits a pyramidal shape and consists of an anterior and posterior lobe. Only one parenchymal cell type, the pinealocyte, was observed. Few neuroblasts were seen between the pinealocytes and in the extended perivascular space. The pinealocytes possess all the organelles necessary for hormone synthesis. No specific secretory granule could be observed. The organ is abundantly vascularized and richly innervated. The morphology of the capillaries indicates the existence of a blood-brain barrier. The ultrastructure of the human fetal pineal gland suggests that the gland has a secretory function in early intrauterine life.

    Notizen: Summary An ultrastructural analysis of myelin in the ventral habenular and in the interpeduncular nuclei of the adult frog Rana esculenta has been carried out. In the ventral habenular nuclei, in addition to normally myelinated fibres, some myelin irregularities have been observed. They consist of myelin enwrapping the perikarya of some neurons and of isolated flaps of collapsed myelin. In the interpeduncular nucleus numerous myelinated fibres occur but few redundant myelin irregularities have been noticed. The morphological data suggest that myelination of fibres in these sites is due to the spiral wrapping mechanism from a glial cell process while the myelin irregularities described in the ventral habenular nuclei are probably due to membrane synthesis within the cytoplasm of the myelinated neurons and of the oligodendrocytes which are sometimes observed in contact with the ensheathed neurons. In the interpeduncular nucleus myelinated fibres indenting astrocytes and oligodendrocytes have been observed.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Die Ultrastruktur des Mitteldarmes von Aedes aegypti Weibchen wurde morphometrisch untersucht. Besonderes Interesse lag dabei auf quantitativen Unterschieden im Bau der Epithelzellen eines vorderen (A) und eines hinteren (P) Abschnittes des Mitteldarmes, um jedem dieser beiden Abschnitte bestimmte Funktionen zuordnen zu können. 5 physiologisch verschiedene Stadien wurden verglichen: 1 = unreife Weibchen; 2 = zum 1. Blutmahl (B.M.) bereit; 3 = Verdauung des B.M.; 4 = zum 2. B.M. bereit; 5 = alternde Weibchen. Wenn der Mitteldarm noch nicht funktionsbereit ist (Stadium 1) gleichen sich die Epithelzellen von A und P quantitativ ultrastrukturell. In den folgenden Stadien hingegen werden deutliche Unterschiede sichtbar. Das Zellvolumen und die Kern-Cytoplasma-Relation werden in P generell größer als in A. Parallel dazu nehmen der Mitochondriengehalt und die Oberfläche des rauhen endoplasmatischen Retikulums in P gegenüber A zu. In A entwickeln sich mehr Microvilli, ein extensiveres basales Labyrinth und ein deutliches glattes endoplasmatisches Retikulum. Vergrößerte Cytoplasmavolumina vor allem in den P-Zellen werden 24 Std. nach dem 1. B.M. gefunden (Stadium 3). Parallel dazu erreichen die Volumina der Organellen und/oder ihre Membranoberflächen ebenfalls Maximalwerte. Diese Vergrößerung des gesamten Zell-apparates läßt den Schluß zu, daß während der Blutverdauung die Funktionskapazität des Mitteldarmes erhöht ist. Basierend auf den quantitativ ultrastrukturellen Unterschieden zwischen den Zellen von A und P wurde postuliert, daß dem vorderen Abschnitt des Mitteldarms verstärkte Absorptionsfunktionen zukommen. Dem hinteren Abschnitt hingegen wurden gesteigerte Protein-syntheseleistungen im Zusammenhang mit der Bildung von Verdauungsenzymen zugeordnet. Die vorliegenden Resultate wurden an einem, in Beziehung auf seine Ernährung, spezialisierten Insekt gewonnen (Blutverdauung). Trotzdem scheint die postulierte Zuordnung von Funktionen zu bestimmten Mitteldarmzonen mit dem “secretion—absorption”-Modell (Berridge, 1970) übereinzustimmen.
    Notizen: Summary The ultrastructure of the midgut of Aedes aegypti females was investigated by means of morphometric techniques. The interest was focussed on quantitative differences in the composition of epithelial cells between the anterior (A) and posterior (P) part of the midgut. The aim was to attribute defined functions to each of these 2 parts. 5 different physiological stages were compared: 1 = immature female; 2 = ready for first blood meal (b.m.); 3 = digestion of b.m.; 4 = ready for 2nd b.m.; 5 = aging female. Whereas in stage 1, when the gut has not yet reached a functional state the epithelial cells of A and P resemble each other, distinct differences develop in subsequent stages. Generally the cell volume and the nuclear-cytoplasmic ratio become higher in P than in A. Concomitantly the mitochondrial content and the surface area of the rough endoplasmic reticulum are more prominent in P than in A. On the other hand more microvilli, a more extensive basal labyrinth and a well developed smooth endoplasmic reticulum are present in A. Enlarged cytoplasmic volumes, especially high in P, are found 24 h after the first b.m. (stage 3). In accordance with this increase the volume of organelles and/or their membrane surface areas also reach maximal values. This augmentation of the metabolic cell apparatus suggests that during blood digestion the whole organ develops an increased functional capacity. From the quantitative structural differences in A and P we concluded that the A-part of the midgut is especially involved in absorption. To the P-part more protein synthesis may be attributed, thus indicating an enhanced formation of digestive enzymes. Although this study deals with a rather specialised insect (blood digestion), it appears that our hypothesis attributing functions to defined midgut zones, agrees to a certain extent with the “secretion-absorption-model” of Berridge (1970).

    Notizen: Summary The larynx musculature of female tree frogs is very poorly developed compared with that of males. This is probably due to the fact that they are less used; female tree frogs do not produce mating calls. The larynx muscles of the female are composed of fibers with small, medium, and large cross-sections. The distribution of the motor end plates varies correspondingly: in regions with fibers of very small diameter the density of synapses is about 33 times greater than in regions with large fibers. There is also a difference in fine structure between male and female laryngeal muscles. The muscles of females comprise red fibers, white fibers, and various transitional types. The arrangement of the triads and their number per sarcomere is the same in all fiber types. All fibers contain about the same amounts of glycogen, but the fat content varies widely among the different types.
