    Description: Streptococcosis is an acute infectious disease that causes mortality in marine and freshwater aquacultures. One of the most important hosts is susceptible to the disease of rainbow trout and during the past decade its industrial production has been growing in Iran. According to FAO, Iran is among the 10 countries producing fish in the world. The most important bacteria causing Strptococcosis includes Strptococcus, Vagococcus and Enterococcus genera. But in all cases, the clinical sings are the same. More than a decade has passed since the first report of this disease in Iran and unfortunately, this disease has become the most important problems of trout production in Iran. Therefor, 72 farms were selected in 8 provinces including Mazandaran, Gilan, Lorestan, CharmahalBakhtiyari, Fars, Kermanshah, Tehran and Kokiloye-Boyer Ahmad and a total 520 moribund and apparently healthy rainbow trout (weight 50 – 200gr) were collected during summer 2008 to 2009. Fish kidney, spleen and liver samples were culture aseptically and finally 206 isolates were identified as gram positive cocci. Using conventional biochemical test, S.uberis, S.agalactiae, S.dysgalactiae, S.faecium and S.inia. Additionally, 172 from 206 isolates were confirmed as S.uberis، S.agalactiae، S.dysgalactiae، S.faecium and S.inia using a PCR assay ased on the results, the most prevalent is belong to S.uberis (isolated from 5 of 7 province), S.dysgalactiae (isolated from 4 of 7 province), S.agalactiae (isolated from 2 of 7 province) and S.faecium and S.inia (from 1 of 7 province) respectively. It is necessary to mention, S.inia was identified just to Fars and any gram positive cocci was isolated from Tehran.

    Description: Study on Viral Nervouse necrosis (isolation, characterisation and pathogenesis) in Golden grey mullet in the Caspian Sea and study of pathogenecity and possibility of transmission to the other fish species (Sturgeon fishes, Rutilus frisii kutum and reared Rainbow trout and Carp) Viral Nervous Necrosis (VNN) is a worldwide disease affecting several species of cultured marine fish. For the past two decades, betanodavirus infections that cause Viral Nervous Necrosis (VNN) have emerged as major constraints on the culture and sea ranching of marine fish in almost all parts of the world. More than forty species mainly of marine origin have been so far affected and this number is likely to rise in future following the introduction of new species and the increase of aquaculture trade. Unknown acute mortality occurred in wild golden grey mullet Lisa auratus and Liza saliens in Iranian waters of Caspian Sea in recent years. In order to isolation and confirmation of causative agents of golden grey mullet mortality in the Caspian Sea, a complementary research investigation project was designed in 2005 and approved immediately in Iranian Fisheries Research Organization (IFRO). Many diagnostic aspects such as Virology (Cell culture and Elctereone Microscopy), Hemathology, Bacteriology, Histopathology, Molecular biology (NestedRT-PCR), Heavy metals measuerment and Serology (IFAT and IHC) were employed in mentioned multidiciplinary project. About 322 moribund fish samples which revealed skin darkening, erratic swimming behavior such as spiral and belly-up at rest and high distention of swimming bladder. Suspected samples were collected from coastal capture sites in iranian north proviences in 2006 till 2009. Targets tissue such as brain and eye were removed in strile condition and then kept in -80oC frezzer for cell culture and Nested-RT-PCR. Other tissue samples from liver, kidney, intestine, stomach, gill, skin and muscle, gall bladder and gonads were taken and fixed in 10% buffer formalin and same parts fixed in glutaraldehyde 3% for histopathology, IHC and EM respectively. Cytopathic effect (CPE) was observed in those cell cultures just six days after inoculation with the dilutions of the tested 312 homogenate supernatants. CPE in monolayers of cells cultured (SSN-1 cell line) was characterised by thin or rounded, refractile, granular cells with vacuoles. Nine samples were positive in virology assay. Nested- RT-PCR was done on suspected tissue samples and supernatant of CPE positive samples and 21 tissue samples and all CPE positive samples were positive. IFAT was selected as a confirmatory method for identifying viral strains replicating on cell cultures and carried out with rabbit anti-betanodavirus serum on suspected tissue samples and some smears of CPE positive samples. Some bright points approved betanodavirus antigen and confirmed cell culture and Nested-RT-PCR findings. In fixed tissue samples widespread and massive vacuolation were observed in brain, spinal cord, retina and optical nerve. In order to confirmation of diagnostic findings , IHC was done with monoclonal antibody antibetanodavirus and some red-brown points were observed. Theses findings revealed expected viral antigens and confirmed previous results. Moreover, virus particles with 25-30 nm in diameter were visualized in infected brain and retina using positive staining in TEM. Also pathogenicity test was employed to confirm the obtained results. So Guppy fish Poecilia reticulata and sturgeon fry were used instead of the experimental host due to ease of handling and susceptibility. After 15 days post infection, guppy bathed in VNN-infected tissue culture with 104 TCID50 showed clinical signs similar to naturally infected Golden grey mullet, and the mortality rate reached up to 100% in 75 dpi. When target organs were examined by cell culture isolation, serology, and histopathology, all revealed the presence of virus in the Guppy. Suspected supernatant injected to sturgeon fry through intravitreous injection and widespread vacuolation were observed in brain and spinal cord buy IHC and Real time PCR were negative. In conclusion, with attntion to obtained results in this investigation such as ecological factors, clinical signs, histopathological, virological and bacteriological results, molecular analysis, (IHC, IFAT, PCR), TEM demonstration, serological and hematological findings, it could be confirmed that VNNV was the main causative agent for disease outbreak in Golden grey mullet in Southern coastline of Caspian Sea.

    Description: In this survey the Physical and Chemical factors, heavy metals, agricultural pesticides and microbial contaminations were studied in mohammadiye channel extending Eslamshahr, Robat karim and Shahriar cities of Tehran Province during the 2011.Specimens were obtained from 5 different stations .The measured factors were temperature, DO, Cl, HCO_3, COD, BOD, CO_2, Mg,... The analysis of Physical and Chemical factors of Mohammadiye channel water showed that except water temperature, all factors in comparison to the standards were at the standard level, So they are suitable for aquaculture. The result of measurements of heavy metals including lead, copper, Iron, Cadmium, Zinc and mercury in spring, summer and fall in comparison to existing standards showed that unfortunately except copper the amount of other heavy metals are higher than standard levels in the most of stations , Lead amounts at stations 2,3,4 and 5 . zinc at stations 1 and 5 , Iron at stations 1,3 and 5 , cadmium and mercury in all stations were higher than the standards for fish culture. The measured amounts of organophosphate and organochlorine pesticides and result of microbial contaminations culture were less than standard levels. Due to mean of water temperature that is high from middle of Spring to the end of summer, It is suitable for culture of warmwater fishes and because of low water temperature in the station No1 It is recommended for culture of coldwater fishes by using of 50-70gr of young fishes .

    Description: This study was conducted for the first time and the effects of probiotic bacteria isolated from the gut of Rainbow trout (Oncorhynchus mykiss). In the first step, after the purification of the isolated bacteria, probiotic properties in vitro was assessed with injection to healthy fish and challenge with three species of pathogenic bacteria in culture media. The second phase of the bacteria isolated in 5 treatments (log 7, 8 and 9 of the lactic bacteria (LAB), Vibrio sp and Pseudomonas sp) and a control treatment on growth and survival, hematological parameters (RBC and WBC cells), immunology and physiology parameters tested during the 60 days of the above parameters after 30 and 60 days and finally at the end, the final assessment was Streptococcosis resistance in fish. The results showed that the isolated bacteria were able to enhance the growth parameters (weight, feed conversion, feed fat, protein efficiency ratio and specific growth rate) and survival. The results showed that log 8 LAB was significant difference with other treatments and control. When using log 8 LAB and Vibrio, the MCV, MCH and MCHC were decreased with no significant difference. The log 8 has the greatest effect on the amount of liver enzyme (AST), IgM and complement component C3, and had significant difference with other treatments. The challenge examination to Streptococcus iniae showed the highest survival in treatments with log 8 (96.66%), and Vibrio (93.33%) and then subjected to other treatments and control are also the least survival (25.38%). The conclusion of that study is the first probiotic properties of bacteria isolated from trout to changes in quality indicators are in particular the LAB bacteria and the second log 8 of LAB had significant positive of changes development and safety, and the fish are resistanted against Streptococcosis.

    Description: Over the past few years, the syndrome of streptococcosis has been associated with outbreaks in rainbow trout (Oncorhynchus mykiss, Walbaum) and caused significant economic losses in the aquaculture industry in Iran. The main purpose of this work was molecular identification of some causative agents of streptococcosis in rainbow trout. A total of 520 samples were collected from the head kidney of diseased fish (weight, 50_200g) in 72 farms of 8 provinces in Iran, during 2008 to 2009. Bacterial isolates representing morphology and biochemical profiles of Streptococcus spp. were further confirmed by polymerase chain reaction (PCR). DNA extraction was carried out from a single colony by using the extraction promega kit following the conditions described by the supplier. The PCR assay was developed based on the 16S rRNA and glucose kinase genes of Streptococcus spp. for the rapid and specific detection and identification of this pathogen from different sources. Approximately 40% of specimens were infected to Streptococcus spp. Consequently, five pathogenic species have been identified, including S. iniae in Fars province, S. faecium in Mazandaran province, S. agalactiae in Gilan and Mazandaran provinces, S. dysgalatiae in Lorestan, Kohgiluyeh and Boyerahmad, Gilan and Kermanshah provinces and S. uberis, which was common in all provinces (except Mazandaran and Lorestan). The dominant species (based on important species index) were S. uberis, S. dysgalactiae and S. agalactiae, respectively.

    Description: Mnemiopsis leidyi (Comb Jelly) is an exotic species which has not been reported from the Caspian Sea till 1999, but it was observed and reported from the southern coasts of Caspian Sea in 1999. Invasion of the Mnemiopsis leidyi to the Caspian Sea pelagic ecosystem has been one of the main difficulties in the past decade. Mnemiopsis leidyi has been brought from the Black Sea to the Caspian Sea by ballast water of ships and it has settled in the southern Caspian Sea through the time. There is a little information regarding the microbial flora in comb jelly but a large number of comb jelly carry natural flora and pathogens located in the coastal waters such as Aeromonas and Vibrio. However, it might have been entered into the Caspian Sea many years ago, but it has recently caused excessive reduction in kilka fish stocks because it is considered as a food competitors for them. The comb jelly may also carry some new microorganisms like parasites and microbial flora to the Caspian Sea. This study was carried out to investigate the parasites and bacterial flora of B. ovata and M. leidyi as probable dangers for the Caspian Sea ecosystem.

    Description: Study of survey health management and diseases in hatcheries and fish farms can help us to knowledge and application control methods such as: prevention, treatment and increase high levels of production in hatchery and farms, finally. This survey carried out from 2005 to 2008 for 4 years in sturgeon hatcheries and farms of Golestan province. Sturgeon fishes include Huso Huso, Ship sturgeon, Acipenser persicus collected and for virology, bacteriology, fungius and hematology examined. Also, physicochemical parameters measured and recorded in different stages of culture. Results of this study showed that all of samples in virology was negative and did not observe any doubetful causes. In bacteriology CFU was variation from 3/9 ×105 to 6/9×10. The most parasites that detected in this survey was Cocolanus espherolanus, Sceria binopsulus semiarmatus and Amphilina fuliacea that separates from Acipenser Percicus, especially. The results about hematology parameters some important hematological indices of ship sturgeon include: The total RBC for female and mail specimens measured as 5.3±1.5 ×10^5, 4.8±0.5×10^5 per mm^3 respectively. The amount of haematocrit and hemoglobin for female and mail determined: 34.3±2.8, 35±1.4 percent and 10.3±0.9, 8.9±0.8 gr/dl .The MCV: 216.3± 96.2, 736.5± 102.5, MCH: 720.2±309.5, 186±0.7 and MCHC: 30±0.8, 25.5±3.4 percent respectively.The total WBC were (female, male): 21320±1054, 20580±777 per mm^3 and neutrophil: 16.4±2.5, 17±1.4 percent and lymphocyte: 74.4±2.4, 73.5± 0.7 percent and eosinophil: 6±1.4, 6.4±0.5 percent, monocyte: 2.8±0.8, 3.5±0.7 percent. There was not any significant differences (p〉0.05) between mentioned parameters in male and female (students t-test). Also evaluation of hematological parameters in bluga ( Huso huso) include: total RBC were (male , female) 5±0.3 ×105 , 4.9±0.6 ×105 per mm^3 ,respectively and hematocrit: 33.2±6.7 , 35.4±3.4 percent and hemoglobin: 11.2±1.5 , 12.2±1gr/dl and MCV: 669.9±172.2, 723.9±982.4 and MCH: 226.2±42.5, 249.5±35.4 and MCHC: 34.1±2.4, 34.6±3.6 percent respectively. The totals WBC were (male, female): 24800±707.1, 23042±1375.4 per mm^3 and neutrophil: 18.5±0.7, 21.4±1.1 percent and lymphocyte: 73.5±1.4, 68.4±1.1 percent and eosinophil: 5±2.8, 7±1.2 percent and monocyte: 3.5±3.5, 3.2±0.8 percent. According to statistically study the count of lymphocyte had significant difference between male and female fish and this count in male was higher than female. (p≥0.05).

    Description: Comb jelly Mnemiopsis leidyi investigation was conducted in the southern region of the Iranian coast of the Caspian Sea during the 2010 in the seasons of spring, summer, autumn and winter at 8 transect with depths of 5, 10, 20, 50 and 100m. Sampling was done at 0–5, 0-10, 0-20, 20-50 and 50–1000 m using a 500 µ and 50 cm mouth opening area. During 2010, a total of 757 samples of comb jelly were collected by the size length of smaller than 5 mm to a maximum of 35 mm. Mean annually Mnemiopsis abundance and biomass was 11±2 ind.m3 and 0.6±0.1 g.m^3 . Maximum abundance was recorded in autumn with 25.7±4.8 ind.m3 and maximum biomass was observed in summer of 1.2±0.1 g.m^3. Minimum abundance was found in spring with 2.3±0.4 ind.m3 and minimum biomass was in winter of 0.1±0.3 g.m^3. Statistical comparison showed that the density of comb jelly were not significantly different in spring and summer (P〉0.05, Tukey test), and winter and summer with the same density, while having the highest density of autumn compared to other seasons which showed a significant difference (P〈0.01, Tukey test). Comb jelly biomass showed summer and spring with higher than winter and autumn, compared to the statistical comparison showed no significant difference in biomass between seasons of summer- spring and fall – winter (P〉0.05). Fluctuations of comb jelly abundance and biomass in different seasons showed that the water level of 20 m had a higher value than other layers. In the spring, more than 98 percent of the comb jelly was recorded in the upper layer (less than 20 m), and only 2 percent of the animals living in layers of 20-50 m. In the summer and autumn, about 93 percent of Mnemiopsis was found in the layers above 20 m and 7 percent of those in the lower layers (maximum 50 meters). In the winter, more comb jelly were observed in layers from 20 to 50 m (about 14 percent) and about 87 percent of them were collected in layers less than 20 meters. As a whole, the total maximum annual average abundance was recorded in the autumn of layer 0-5 m with 94.0± 33.8 ind.m3 and the highest average biomass in the 0-5 m layer of summer with 3.1±1.0 g.m^3 . Frequency of group length (size) of the population M. leidyi showed that comb jellies of less than 15 mm (prior to adult) consisted of 85.8%, in which at less than 20 m, group length (size) of 15 mm included of 82-92% and at the depth of more than 50 m, these group was 8-18%. Also comb jelly adults (greater than 16 mm) in the surface layers (less than 20 m) were included of approximately 73 to 100% and below 20 m, 0-27 % of the population. Data of the 8 transect showed that the highest density and biomass of Mnemiopsis were recorded in Torkaman with 16.4± 6.5 ind.m^3 , and 0.8±0.2 g.m^3 , respectively. Statistical comparison of the mean abundance and biomass of comb jelly showed that there were a significant differences between Turkmen and Amirabad transect compared to other transects (P〈0.05). Regional distribution of M. leidyi in the southern Caspian Sea showed aggressive area in terms of density and biomass are areas with significant differences (P〈0.05). Comb jelly population showed that both West and East in terms of abundance and biomass were similar but different with the central region. Eastern and western regions compared to the central region have the higher abundance. Based on the study looks at the rise and persistence parameters of comb jelly, it seems two main factors such as water temperature to the desired value, especially in summer (22-30°C), and food availability (zooplankton) have more roles in the southern of the Caspian Sea. After the invasion of M. leidyi into the Caspian Sea, not only the abundance and biomass of zooplankton decreased but also the species diversity decreased from 36 to 15 species. Hence, it seems the decline in the South Caspian comb jelly was due to decreased of fecundity for lack of zooplankton feeding resources.

    Description: The aim of this study was comparison the effect of cow dung as organic fertilizer and chemical fertilizer on microbial flora, fish and human pathogenic bacteria and physico-chemical parameters of cyprinus fish pond water. The water samples were collected from two ponds, pond 1 was fertilized by cow dung and pond 2 was fertilized by chemical fertilizer, respectively from May to October 2011. . The aerobic and anaerobic bacteria were enumerated in TSA by serial dilution of the sample, followed by conventional pure plate method. Coliforms bacteria were similarly isolated on Chrom agarTM ECC. Moreover, the water samples were used to analysis of BOD, dissolved oxygen, conductivity, transparency, total alkalinity, total dissolved solid, total hardness, total nitrogen and total phosphorous. Temperature and pH were measured by thermometer and portable pH meter in place of ponds. For isolating of fish and human pathogenic bacteria, thirty six fish were collected from pond 1 from July to October and then sampled from their liver and kidney and inoculated in blood agar. The mean of total count of bacteria and total coliform count in cow dung sample were 4 × 104 ± 1200 and 4700 ± 300 CFUmL^-1 respectively. The mean of water total count bacteria of pond 1 were significantly higher than the water of pond 2. The water total count bacteria of pond 1 were significantly increased in August, but water total count of pond 2 was not shown significant variation from May to October. The results showed that Pseudomonas, Klebsiella, Enterobacter, Yersinia and E.coli were isolated from water of pond 1 but just Pseudomonas and E.coli were isolated from water of pond 2. Moreover, the human pathogenic bacteria shuc as Shigella and Salmonella were not isolated from of water and fish samples from pond 1. In this study, the bacteria growths were negative in all of the inoculated plate from fish liver and kidney. The total hardness, total phosphorus, total dissolved solid and conductivity of the pond 1 water were significantly higher than the pond 2 water. But the BOD, dissolved oxygen and transparency of the pond 2 were significantly higher than the pond 1 water. There were no significant differences in the total nitrogen, total alkalinity and pH between water of pond 1 and 2. In recent years, the chemical fertilizer has been used in cyprinus fish pond but many studies have showed that they were high risked as environmental pollution. Thus, the use of livestock manure especially cow dung could be a suitable alternative. The results showed the use of cow dung increased the bacterial population and diversity. Increasing of bacteria population caused increased total phosphorus that plays an important role in bloom of plankton because we found decreasing transparency and planktons are an important food for cyprinus fish. In despite of isolation of fish and human pathogenic bacteria from water, the bacteria growths were negative in all of the inoculated plate from fish liver and kidney. In summary, it seems the cow dung is a suitable alternative for fertilizing of cyprinus fish pond water if a microbial and physico-chemical monitoring of fish and pond water were continuously arranged.

    Description: The national research plan with title of "Study on health status of Iran Coldwater hatchery and rearing farms" was done in order to identification and tracking of main causative agents of recent mortality in Iran Coldwater hatchery and rearing farms and recognize of infected zones and design of landscape distribution of Epizootic Coldwater fish diseases in the country. This plan was conducted parallel and in same time in some polar provinces of Coldwater fish production such as Mazandaran, Gilan, Ardebil, West Azerbaijan, East Azerbaijan and Kurdistan in three years from 2006-2008. In this regard, about 23,21 and 20 farms in Mazandaran province, 30 farms in West and East Azerbaijan and Kurdistan provinces and 19 farms in Gilan and Ardebil were selected as randomly and tracking were done yearly from 2006-2008. This research was conducted according to regular planning consists of farms visit that was done according to statistical plan and completion of Questionnaires and sampling. The findings in Mazandaran province revealed that fish infections background and infectious diseases were increased in period of 2006-2008. In fact, 56% of all fish farms in 2006, 71% in 2007 and 85% in 2008 were recorded as infected farms. In other side, average weight of fish final products was more 500gr in consume market. So, regarding to long period of fish culture in mentioned fish occurrence of more morbidity could be expected. The streptococcus infections were most important fish bacterial diseases that have more incidence and pathogenicity in collected questionnaires. Also it revealed more occurrences in summer season in above 15oC temperature in affected fish farms. Also, Enteric Redmouth Disease (ERM) and Saprolegniasis were reported as second and third degree in examined farms. Meanwhile, findings of control, prevention and treatment of our survey revealed that using of antibiotics and detergent materials were increased in mentioned province. Indeed, 34.7% of all fish farms in 2006, 71.4% in 2007 and 75% in 2008 have applied treatment operation and using of detergent materials in affected farms. In other side, frequent using of Erythromycin antibiotic was reported several times in 2008 so fish farmer faced to Bacterial resistance and should be using from Florfenicol as alternative antibacterial. Also, in feed sampling from all fish farms about 3 fish farms in 2006 and 5 fish farms in 2008 were faced with food poisoning that originated from high TVN and peroxidase in fish food consumed. These farms revealed mass mortality that stopped after change feeding regime. In conclusion absence of fish health management could be considered as main agent of mentioned mortality in Mazandaran province. The findings of our survey in West and East Azerbaijan and Kurdistan revealed that absence of sufficient experiences in fish farmers and their neglect from water quality concepts, farm cleanness, using of suitable detergent materials for fish ponds and instruments and disability in continuous monitoring of physic-chemical factors of consumed water could be considered as most important problems. So, fish morbidity and mortality, growth decrement and low production rate were expected. Unfortunately, despite the occurrence of infectious and non- infectious diseases in examined fish farms, there were no documents and information about diseases history, clinical signs in dead fish, feeding regime, fish mortality (rate of daily mortality, age and weight of moribund fish), treatment operation, drugs usage, growth rate, physico-chemical factors and fish density. So planning for control and prevention of mentioned diseases were unsuccessful in affected fish farms. Also, occurrence of some epizootic fish viral diseases such as VHS, IPN and IHN were observed frequently with mass mortality about 40100% in some examined fish farms that produced some tragic economic lost in the mentioned provinces. Similarly, some bacterial, fungal and parasitic diseases were observed repeatedly in some inspected farms such as Flavobacterium, streptococcus infections. Also, some unknown causative agents in parasitic diseases were observed several times such as Dactylogyrus, Gyrodactylus, Ichthyophthirius multifiliis, Trichodina, Chilodonella, Diplostomum and fungal diseases such as Saprolegniasis. In conclusion it seems that absence of fish health management, inadequate rate of hygienic technical services, absence of quarantine programs for transfer of new eyed-eggs, juvenile and live broodstocks packages without health certificates from authorized organizations, uncontrolled entry of foreign eyed-eggs (France, Denmark, Australia and Armenia) and neglect in using of suitable detergent materials for fish ponds, instruments, consumed water and infected eggs, not screening of broodstocks in hatcheries could be considered as most important causative agents in occurrence of infectious diseases and main problems in affected farms in mentioned provinces. Similarly, our findings in Gilan and Ardebil provinces revealed that their problems were similar to other provinces. In fact, neglect in establish of necessary Infrastructures in fish farms and dereliction in health management concepts could be considered as main reasons of occurrence of fish infectious and non-infectious diseases in studied fish farms in mentioned provinces. In fact, often fish farms visited hadn't suitable structures without control and prevention approach. There were not observed detergent using, quarantine programs and prevention methods. Entrance of unknown persons, birds and wild animals were ordinary in mentioned farms. Unfortunately level of farmer’s knowledge was low and no training courses and extension programs were planned by authorized organizations. Meanwhile, food storages were unsuitable and food packages were stored in inappropriate situation so poisoning conditions were increased in examined farms. In conclusion it could be finalized that Coldwater fish farms in Gilan and Ardebil provinces situated in insufficient position and correction of current situation are needed urgently. In final elicitation, it would be mentioned that lack of basic infrastructures could be introduced as most important reasonfor spread of diseases, mortality and related economic losses in studied provinces. Therefore, attention to environmental affairs and access to Sustainable Development are recommended. Also consideration of biosecurity regulations and health management concepts would be important requirements for modification and reformation of Coldwater fish farms in examined provinces towards A Better Tomorrow.
