    Description: Some specific and non-specific immune variables of grass carp against Aeromonas hydropila and in the present of organophosphate, diazinon was studied at 18-20?C. Also, the toxicity of this toxicant was assessed in this species. Isolation and characterization of the fish IgM showed that fish possesses IgM with molecular weight of 480-640 KD having 15-20% of total serum protein. The LC50 96 hours of the toxicant was 15.13 mg/L and the survival fish showed erythropenia, leucopenia and a decrease in levels of blood enzymes, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase and total protein, while level of glucose increased. When fish were exposed to sublethal concentrations of diazinon (1, 2 and 4 mg/L) for 12 hours large change were then observed in hematological and biochemical variables of blood feaures plus a suppression of chemiluminuscence (CL) response by blood leucocytes during 45 days post-expposure. When fish were intraperitoneally immunized with Aeromonas hydrophila whole cell antigens and exposed to 2 mg/L of diazinon, the immunological responses consisting of lysozyme content, CL response, antibody titre were suppressed during one moth post-exposure. Also, light and electron microscopic examinations of tissues of kidney, liver, gill, spleen and nostril of fish exposed to acute and sublethal concentration of the toxicant revealded remarkable changes including hyperemia, heamorrhage, necrosis, damaging of reticulum endoplasmic of epithelial cells and vacuolation of club cell of nostril, and degeneration of mithochondria in kidney cells, reduction in excretion of amorphous proteinous materials and vesicle numbers on the cell surface and also blocking of cell surface cannals of nostril cells.

    Description: In this survey the Physical and Chemical factors, heavy metals,agricultural pesticides and microbial contaminations were studied in mohammadiye channel extending Eslamshahr, Robat karim and Shahriar cities of Tehran Province during the 2011.Specimens were obtained from 5 different stations .The measured factors were temperature,DO, Cl,HCO3,COD,BOD,CO2,Mg,……. The analysis of Physical and Chemical factors of Mohammadiye channel water showed that except water temperature,all factors in comparison to the standards were at the standard level,So they are suitable for aquaculture. The result of measurements of heavy metals including lead, copper,Iron,Cadmium, Zinc and mercury in spring, summer and fall in comparison to existing standards showed that unfortunately except copper the amount of other heavy metals are higher than standard levels in the most of stations , Lead amounts at stations 2,3,4 and 5 . zinc at stations 1 and 5 , Iron at stations 1,3 and 5 , cadmium and mercury in all stations were higher than the standards for fish culture. The measured amounts of organophosphate and organochlorine pesticides and result of microbial contaminations culture were less than standard levels. Due to mean of water temperature that is high from middle of Spring to the end of summer , It is suitable for culture of warmwater fishes and because of low water temperature in the station No1 It is recommended for culture of coldwater fishes by using of 50-70gr of young fishes .

The main stages for establishment of the system consisted of: 1-Conducting a gap analysis to compare the present state of the laboratories with ISO/IEC 17025 Training 2 General requirements for the competence of testing and calibration laboratories Validation of methods Estimation of uncertainty Internal audits 3- Performing of technical and management requirements 4-Submit of quality manual to Iran Accreditation System (IAS) in order to accredit In January of 2010 the laboratories were accredited by Iran Accreditation System (IAS). The main results were including: 1-Increase the accuracy of measurement 2-Improvement of the Repeatability and Reproducibility of the test methods 3-Traceability and standardization of test methods 4- Calibration of measurement instruments 6- Updating of test methods 7-Standardization of physical condition of the laboratories 8- Getting the certification from Iran Accreditation System (IAS).

    Description: In this study, 11 male of Caspian trout (Salmo truta caspius) (with mean length and weight 37/8 ± 5/3 cm and 523/3 ± 24/7 respectively) and 23 male of Caspian kutum (Rutilus frisii kutum) (with mean length and weight 36/1 ± 7/1 cm and 631/3 ± 21/6 g respectively) were evaluated. All the fish were good at the initial examination of sexual maturity. After sperm sampling, their quality were tested. In this step, the parameters such as motility, duration of mobility, density, pH and osmolality were measured. After this stage, the sperm samples of Caspian trout in the ratio 1: 3 were diluted with the aqueous solution containing compounds (0.3M Glucose, 10% Methanol, 10% egg yolk) and the freezing process was done manually and the sperm was frozen in liquid nitrogen. The sperm samples of Caspian kutums were diluted (ratio of 1: 3) with two soluble diluent containing compounds (350 mM glucose, 30 mM Tris and 4% Polyethylene glycol) and (350 mM glucose, 30 mM Tris and 2% Glycerol) and were frizzed automatically by Planner Kryo instrument and placed in liquid nitrogen. The sperm samples were thawed 1 to 3 months after the date of first freezing and their quality were assessed by measuring percent and timing motility. The results showed that the obtained semen volume of Caspian trout was more than Caspian kutum. Moreover, percentage of motile sperm, timing motility and sperm density of Caspian trout were higher than those of Caspian kutum but osmolality and pH of Caspian trout were lower than those of Caspian kutum. Over time, the percentage of sperm motility and mobility for both species declined compared with fresh samples. After thawing, percentage of motile sperm and timing motility of Caspian kutum were lower than those factors Caspian trout. The results showed that the sample of Caspian kutum sperm that were diluted by ethylene glycol after thawing and were immotile ll of them. However, the samples were diluted by glycerol, after thawing, were alive and motile. According to the results, it seems very important species differences that must be fully considered in the process of freezing sperm. The use of a single protocol would not be successful in cryopreservation because the reaction of sperm against to chemical agents is variable. Therefore, it is essential to get the right information to protect valuable Caspian fish by using cryopreservation. Further studies on the characteristics of each species, as well as the freezing process take appropriate diluent.
