Notes: Abstract Seven congenic strains differing from C57BL/10Sn at theH-13 locus have been produced which define fourH-13 alleles. Isografting, exchanging of grafts between sublines, F1 testing, and linkage testing demonstrate the presence of additionalH genes in four of these strains. The medial survival times (MSTs) of skin grafts fromH-13a to unimmunizedH-13b recipients ranged from 69 to 83 days. Rejection across all other barriers was extremely weak with most MSTs being 〉 100 days. Preinjection of donor strain thymocytes caused accelerated rejection of skin grafts fromH-13a toH-13b mice, but had only minimal effect on skin grafts across other barriers. Rejection ofH-13 incompatible grafts was significantly stronger when the donor and host areH-3a than when they wereH-3b.

Notes: Abstract A new cell-membrane alloantigen determining locus, Ly-6, has recently been described, and the single specificity Ly-6.2 has been defined by the serum (BALB/c× A)F1 anti-CXBD. Using both fluorescence and cytotoxicity, we found this specificity predominantly on peripheral (extrathymic) T cells, as tissues react thus: thymus, 0–5 percent; spleen, 25 percent; lymph nodes, 69 percent; bone marrow, 15 percent. These reactions agree with the proportion of (Thy+, Ig−) cells present in these tissues. Cortisone-resistant thymus cells were positive. Absorption studies with thymus cells demonstrated the sparse or absent representation of Ly-6.2 on intrathymic T cells. Examination of spleen and lymph node cells from T cell-depleted C57BL/6 mice (after in vitro treatment with anti-Thy-1 serum or examination of tissues of C57BL/6-nu/nu mice) also showed a depletion of Ly-6.2+ cells. Conversely, removal of Ig+ B cells, which caused a relative increase in the number of T cells in the residual population, also increased the number of Ly-6.2+ cells. Additive effects of anti-Thy-1.2 and anti-Ly-6.2 could not be demonstrated, which suggests that the same population was Thy-1.2+, Ly-6.2+. However, additive effects could be shown with an anti-Ia serum and anti-Ly-6.2. The Ly-6.2 specificity is not found on red cells, liver, brain, or antibody-forming cells, but has been identified on a T-cell (but not B-cell) tumor and on kidney. Ly-6.2 can therefore be considered to be a marker for peripheral T cells, and it differs from the Thy-1 and the Ly-1,2,3, and 5 specificities in its relative absence from the thymus.

Notes: Abstract Thirteen new congenic lines have been produced which have chromosome-7 segments introduced from different strains onto the C57BL/10Sn background. Sublines B10.P(61NX)C,D, and E received chromosome-7 segments from P/J, B10.CE(62NX) from CE/J, B10.SEC(64NX)A,C,E, and F from SEC/1Re, B10.SM(65NX) from SM/J, B10.WB(66NX) from WB/Re, B10.A(67NX) from A/SnGrf, B10.AKR(68NX) from AKR/SnGrf, and B10.K(69NX) from C3H.K. Isograft testing indicated that three sublines, B10.P(61NX)D, B10.CE(62NX)B, and B10.WB(66NX)B are histoisogenic, i.e., histocompatible within each line. With the exception of B10.A(67NX), B10.AK(68NX), and B10.K(69NX), which have not been isografted, the remaining sublines showed residual heterozygosity on isografting. The three histoisogenic lines have undergone F1 testing and have been found to possess theH-4 a allele and new and distinct alleles at theH-1 locus. They have been designated B10.P(61NX)-H-4a H-1 d , B10.WB(66NX)-H-4a H-1 e , and B10.CE(62NX)-H-4a H-1 f . Direct exchange of grafts has indicated the following genotypes: B10.A(67NX)-H-4a H-1 b , B10.AK(68NX)-H-4a H-1 b , and B10.K(69NX)-F-4a H-1 b . The B10.SEC(64NX) and B10.SM(65NX) sublines have not been typed completely forH-4 andH-1. F 1 testing or direct exchange of skin grafts indicated that B10.P(61NX)-H-4a H-1 d , B10.WB(66NX)-H-4a H-1 e , B10.A(67NX)-H-4a H-1 b B10.AK(68NX)-H-4a H-1 b and B10.K(69NX)-H-4a H1 b possess nonon-H-1 histocompatibility differences from the G57BL/10 background.
