    Description: Vitamin D receptor (VDR) mediates the biological function of the steroid hormone calcitriol, which is the metabolically active version of vitamin D. Calcitriol is important for a wide array of physiological functions, including calcium and phosphate homeostasis. In contrast to mammals, which harbor one VDR encoding gene, teleosts possess two orthologous vdr genes encoding Vdr alpha (Vdra) and Vdr beta (Vdrb). Genome mining identified the vdra and vdrb paralogs in the Atlantic cod (Gadus morhua) genome, which were further characterized regarding their phylogeny, tissue-specific expression, and transactivational properties induced by calcitriol. In addition, a selected set of polycyclic aromatic hydrocarbons (PAHs), including naphthalene, phenanthrene, fluorene, pyrene, chrysene, benzo[a]pyrene (BaP), and 7-methylbenzo[a]pyrene, were assessed for their ability to modulate the transcriptional activity of gmVdra and gmVdrb in vitro. Both gmVdra and gmVdrb were activated by calcitriol with similar potencies, but gmVdra produced significantly higher maximal fold activation. Notably, none of the tested PAHs showed agonistic properties towards the Atlantic cod Vdrs. However, binary exposures of calcitriol together with phenanthrene, fluorene, or pyrene, antagonized the activation of gmVdra, while chrysene and BaP significantly potentiated the calcitriol-mediated activity of both receptors. Homology modeling, solvent mapping, and docking analyses complemented the experimental data, and revealed a putative secondary binding site in addition to the canonical ligand-binding pocket (LBP). Calcitriol was predicted to interact with both binding sites, whereas PAHs docked primarily to the LBP. Importantly, our in vitro data suggest that PAHs can interact with the paralogous gmVdrs and interfere with their transcriptional activities, and thus potentially modulate the vitamin D signaling pathway and contribute to adverse effects of crude oil and PAH exposures on cardiac development and bone deformities in fish.

    Notes: Abstract At Malanjkhand, Central India, lode-type copper (-molybdenum) mineralization occurs within calcalkaline tonalite-granodiorite plutonic rocks of early Proterozoic age. The bulk of the mineralization occurs in sheeted quartz-sulfide veins, and K-silicate alteration assemblages, defined by alkali feldspar (K-feldspar ≫ albite) + dusty hematite in feldspar ± biotite ± muscovite, are prominent within the ore zone and the adjacent host rock. Weak propylitic alteration, defined by albite + biotite + epidote/zoisite, surrounds the K-silicate alteration zone. The mineralized zone is approximately 2 km in strike length, has a maximum thickness of 200 m and dips 65°–75°, along which low-grade mineralization has been traced up to a depth of about 1 km. The ore reserve has been conservatively estimated to be 92 million tonnes with an average Cu-content of 1.30%. Supergene oxidation, accompanied by limited copper enrichment, is observed down to a depth of 100m or more from the surface. Primary ores consist essentially of chalcopyrite and pyrite with minor magnetite and molybdenite. δ34S (‰) values in pyrite and chalcopyrite (−0.38 to +2.90) fall within the range characteristic of granitoid-hosted copper deposits. δ18O (‰) values for vein quartz (+ 6.99 to +8.80) suggest exclusive involvement of juvenile water. Annealed fabrics are common in the ore. The sequence of events that led to the present state of hypogene mineralization is suggested to be as follows: fracturing of the host rock, emplacement of barren vein quartz, pronounced wall-rock alteration accompanied by disseminated mineralization and the ultimate stage of intense silicification accompanied by copper mineralization. Fragments of vein quartz and altered wall rocks and striae in the ore suggest post-mineralization deformation. The recrystallization fabric, particularly in chalcopyrite and sphalerite, is a product of dynamic recrystallization associated with the post-mineralization shearing. The petrology of the host rocks, hydrothermal alteration assemblages, ore mineral associations, fluid inclusions and the sulfur and oxygen isotopes of ores are comparable to those in Phanerozoic (and reported Precambrian) porphyry-copper systems, and the Malanjkhand deposit has important implications for both metallogenic models for, and mineral exploration in, Precambrian terrains.

    Notes: Abstract Treatment of Vibrio cholerae el tor strain SLH22(J) with nitrofurantoin induced dose-dependent prophage ‘kappa’, the maximum induction being 6-fold the spontaneous induction level. UV-inactivated ‘kappa’ phages were Weigle reactivated, the maximum Weigle factor being 1.8 and 2.0 respectively in nitrofurantoin and UV pretreated el tor strain H218 Smr. Nitrofurantoin treatment also caused significant filamentation of the el tor strain H218 Smr and mutation of these cells from ampicillin sensitivity to ampicillin resistance. The levels of the four SOS-like responses induced by this drug were low but significant.

    Notes: Abstract Elementary processes in dusty, beam-driven plasma discharges are studied experimentally and theoretically for the first time. A theoretical model is constructed for a beam-driven plasma containing macroscopic particles. The effect of macroscopic particles on the electron energy distribution function is estimated assuming a Coulomb field for the particles. The resulting rate of electron-ion recombination on the macroscopic particles is compared with the electron loss constant calculated from the electron energy distribution function with an electron absorption constant in the orbital-motion approximation. This approximation, which is valid in the collisionless case, is found to work satisfactorily beyond its range of applicability. The distributions of the charged particles and electric fields created by macroscopic particles in a helium plasma are determined. The experimental data demonstrate the importance of secondary emission by high-energy electrons.
