    Description: There is an urgent need for new drugs to treat malaria, with broad therapeutic potential and novel modes of action, to widen the scope of treatment and to overcome emerging drug resistance. Here we describe the discovery of DDD107498, a compound with a potent and novel spectrum of antimalarial activity against multiple life-cycle stages of the Plasmodium parasite, with good pharmacokinetic properties and an acceptable safety profile. DDD107498 demonstrates potential to address a variety of clinical needs, including single-dose treatment, transmission blocking and chemoprotection. DDD107498 was developed from a screening programme against blood-stage malaria parasites; its molecular target has been identified as translation elongation factor 2 (eEF2), which is responsible for the GTP-dependent translocation of the ribosome along messenger RNA, and is essential for protein synthesis.     Notes: In the title compound, C23H34O4, the two saturated six-membered rings have slightly flattened chair conformations and the unsaturated ring assumes a 8β,9α-half chair conformation distorted towards a 8β-sofa. The five-membered ring has an unusual conformation close to a 13β-envelope. The acetoxy and methyl ketone substituents are twisted with respect to the average molecular plane of the steroid nucleus. The molecules are hydrogen-bonded head-to-head via the hydroxy and methyl ketone groups forming dimers, which are stacked in planes perpendicular to the b axis.

    Notes: A theory for the order in σ phases based on the sphere-packing model of Wilson & Spooner [Acta Cryst. (1973), A29, 342-352] has been examined using a mathematical analysis. The analysis suggests that the prediction of order in these phases based on lattice-constant variations associated with variations in atomic diameter should be treated with caution, particularly where the results are at variance with experimentally determined results.
