    Description: A challenge for HIV-1 immunogen design is the difficulty of inducing neutralizing antibodies (NAbs) against neutralization-resistant (tier 2) viruses that dominate human transmissions. We show that a soluble recombinant HIV-1 envelope glycoprotein trimer that adopts a native conformation, BG505 SOSIP.664, induced NAbs potently against the sequence-matched tier 2 virus in rabbits and similar but weaker responses in macaques. The trimer also consistently induced cross-reactive NAbs against more sensitive (tier 1) viruses. Tier 2 NAbs recognized conformational epitopes that differed between animals and in some cases overlapped with those recognized by broadly neutralizing antibodies (bNAbs), whereas tier 1 responses targeted linear V3 epitopes. A second trimer, B41 SOSIP.664, also induced a strong autologous tier 2 NAb response in rabbits.     Notes: Abstract Foliar analysis was conducted to determine the differences between macronutrient concentrations and contents of current year's fascicles of loblolly pine (Pinus taeda L.), pitch pine (P. rigida Mill.), and first (F1) and second (F2) generation plantations of the pitch × loblolly hybrid (P. rigida × taeda). No significant differences in foliar P, K, Ca, and Mg concentrations were found between any of the species or hybrid families tested. Foliar N concentration differed among F1 families, but not among the F2 families. Foliar N concentrations of loblolly and pitch pine in the F1 plantation were not significantly different. Fascicles of pitch pine tended to be shorter and lighter than those of loblolly pine and the hybrids, and nutrient contents of pitch pine foliage tended to be slightly less. An assessment of site effects on tree growth and nitrogen nutrition in the F1 plantation revealed that the hybrids respond differently to an enriched site. Six of the eight species and hybrids had higher foliar N contents on the better quality site, while four had increased stem volumes.

    Notes: Summary A greenhouse study was conducted to evaluate the effect of ectomycorrhizae on loblolly pine (Pinus taeda L.) growing in a Piedmont soil. Pine seedlings were inoculated with one of four species of fungi (Scleroderma aurantium, Pisolithus tinctorius, Thelophora terrestris, andRhizopogon roseolus). The seedlings were grown in pots containing a Cecil sandy clay loam amended to create a gradient of extractable P ranging from 5.9 to 52.5 μg/g. After ten months, all colonized seedlings were significantly larger than control seedlings. However, of the four fungi,Scleroderma aurantium mediated a far superior shoot growth response to increasing levels of soil P; the seedlings were significantly larger than those colonized by any other fungus and also had the largest root systems and greatest degree of mycorrhizal colonization.

    Notes: A procedure is described in which a wide bore glass capillary column is used as an alternative to the more traditional packed column in the analysis of amino acid levels in plasma. The coefficients of variation for all amino acids (with the exception of aspartic acid) were better than 11% with recoveries ranging from 81% to 122%. The data are compared with the corresponding results obtained using a packed column and show significant differences (p 〈 0.005) between values for glycine, serine, isoleucine, proline, methionine, aspartic acid, phenylalanine, and lysine. A similar comparison between results from the wide bore and the fused-silica open tubular (FSOT) column shows better agreement. Adjustment of chromatographic conditions for the wide bore analysis yields results in good agreement with those from FSOT analysis but which still differe significantly from the backed column data.

    Notes: A procedure is described in which a wide bore capillary column is used as an alternative to the more traditional packed column for the quantitative analysis of amino acids as their N-heptafluorobutyryl isobutyl ester (HBB) derivatives. The column, installed in a gas chromatograph previously configured for use with a packed column, is shown to give good reproducibility by repeated determination of amino acid response factors (RSD values for all amino acids are below 3%). A number of problems, encountered during the use of this column, are discussed and suitable techniques to overcome them are reported.

    Notes: In the present study, the use of gas chromatography (GC) for the determination of amino acids in human cerebrospinal fluid (CSF) is described. Although some amino acids may be determined using a packed column following the removal of glucose, the major interfering component, the inadequate resolution of other amino acids from remaining unidentified components results in poor quantitation. The use of wide bore columns improves reproducibility considerably, but still it does not provide sufficient resolution to enable quantitative determination of all amino acids in human CSF. Good reproducibility data, with CV values for all amino acids of 7% or less and recoveries generally between 80% and 100%, can only be obtained using the fused silica open tubular (FSOT) column. Normal amino acid levels are presented for 10 samples of human CSF, which compare well with data previously reported in the literature.
