    Notes: Metallocluster extrusion requirements, interspecies MoFe-protein primary sequence comparisons and comparison of the primary sequences of the MoFe-protein subunits with each other have been used to assign potential P-cluster (Fe-S cluster) domains within the MoFe protein. In each β unit of the MoFe protein, subunit domains, which include potential Fe-S cluster ligands Cys-62, His-83, Cys-88 and Cys-154, and β-subunit domains, which include potential Fe-S cluster ligands Cys-70, His-90, Cys-95 and Cys-153, are proposed to comprise nearly equivalent P-cluster environments located adjacent to each other in the native protein. As an approach to test this model and to probe the functional properties of the P clusters, amino acid residue substitutions were placed at the α- subunit Cys-62, His-83, Cys-88 and Cys-154 positions by site-directed mutagenesis of the Azotobacter vinelandii nifD gene. The diazotrophic growth rates. MoFe—protein acetylene-reduction activities, and whole-cell S 3/2 electron paramagnetic resonance spectra of these mutants were examined. Results of these experiments show that MoFe-protein α-submit residues, Cys-62 and Cys-154, are probably essential for MoFe-protein activity but that His-83 and Cys-88 residues are not. These results indicate either that His-83 and Cys-88 do not provide essential P-cluster ligand or that a new cluster-ligand arrangement is formed in their absence.

    Notes: The nucleotide sequence of the nifA gene from Azotobacter vinelandii was determined. This gene encodes an Mr= 58100 polypeptide that shares significant sequence identity when compared to nifA-encoded products from other organisms. Interspecies comparisons of nifA-encoded products reveal that they all have a consensus ATP binding site and a consensus DNA binding site in highly conserved regions of the respective polypeptides. The nifA gene immediately precedes the nifB-nifQ gene region but is unlinked to the major nif gene cluster from A. vinelandii. A potential regulatory gene precedes and is apparently cotranscribed with nifA. Mutant strains that have a deletion or a deletion plus an insertion within nifA are incapable of diazotrophic growth and they fail to accumulate nitrogenase structural gene products.

    Notes: Abstract The NifS and NifU nitrogen fixation-specific gene products are required for the full activation of both the Fe-protein and MoFe-protein of nitrogenase from Azotobacter vinelandii. Because the two nitrogenase component proteins both require the assembly of [Fe-S]-containing clusters for their activation, it has been suggested that NifS and NifU could have complementary functions in the mobilization of sulfur and iron necessary for nitrogenase-specific [Fe-S] cluster assembly. The NifS protein has been shown to have cysteine desulfurase activity and can be used to supply sulfide for the in vitro catalytic formation of [Fe-S] clusters. The NifU protein was previously purified and shown to be a homodimer with a [2Fe-2S] cluster in each subunit. In the present work, primary sequence comparisons, amino acid substitution experiments, and optical and resonance Raman spectroscopic characterization of recombinantly produced NifU and NifU fragments are used to show that NifU has a modular structure. One module is contained in approximately the N-terminal third of NifU and is shown to provide a labile rubredoxin-like ferric-binding site. Cysteine residues Cys35, Cys62, and Cys106 are necessary for binding iron in the rubredoxin-like mode and visible extinction coefficients indicate that up to one ferric ion can be bound per NifU monomer. The second module is contained in approximately the C-terminal half of NifU and provides the [2Fe-2S] cluster-binding site. Cysteine residues Cys137, Cys139, Cys172, and Cys175 provide ligands to the [2Fe-2S] cluster. The cysteines involved in ligating the mononuclear Fe in the rubredoxin-like site and those that provide the [2Fe-2S] cluster ligands are all required for the full physiological function of NifU. The only two other cysteines contained within NifU, Cys272 and Cys275, are not necessary for iron binding at either site, nor are they required for the full physiological function of NifU. The results provide the basis for a model where iron bound in labile rubredoxin-like sites within NifU is used for [Fe-S] cluster formation. The [2Fe-2S] clusters contained within NifU are proposed to have a redox function involving the release of Fe from bacterioferritin and/or the release of Fe or an [Fe-S] cluster precursor from the rubredoxin-like binding site.

    Notes: Abstract Using only the close connection between the bound-state properties of a potential and its scattering phase shifts, a limiting excitation energy of a finite nucleus is predicted. Calculations for208Pb give its value as 8.2 MeV/A. The excitation-energy dependence of the level-density parameter is also discussed and the connection with astrophysical calculations of stellar collapse rates is indicated.

    Notes: Abstract Deep-inelastic collisions are studied within a phenomenological approach based on a parametrized S matrix. The importance of a correct treatment of the Poisson sum is emphasized and the possibility of non-zero angular-momentum transfer is included. Statistical fluctuations are also considered. It is shown that the spin polarization is extremely sensitive to the treatment of the Poisson sum and that the spin alignment is independent of the scattering angle in the absence of statistical fluctuations.

    Notes: A high-torque rheometer was used to facilitate the polycondensation of 4-[5-amino-6-hydroxybenzoxazol-2-yl]benzoic acid (ABA) with trimesic acid and 1,3,5,7-tetrakis(4-carboxylatophenyl)adamantane to yield two- and three-dimensional benzobisoxazole polymers, respectively. Although the resultant polymer dopes exhibited improved homogeneity compared to polymer dopes previously prepared in glassware, improved polymer solution viscosities were not achieved. Fibers spun from the two- and three-dimensional polymers did not show a significant increase in compressive strength compared to fibers of the linear or one-dimensional benzobisoxazole polymer derived from the homopolymerization of ABA. Morphological studies of the polymer fibers and films by wide-angle X-ray scattering and scanning electron microscopy strongly indicated more lateral disorder and a more isotropic character for the three-dimensional structures compared to the one-dimensional structures. © 1997 John Wiley & Sons, Inc.     Notes: The conductivity study results of lithium-doped sulfonated PBI, a conjugated rigid rod polymer, poly[(1,7-dihydrobenzo[1,2-d:4,5-d′]dimidazole-2,6-diyl)-2-(2-sulfo)-p-phenylene], derivatized with pendants of propane sulfonate Li+ ionomer are reported. The room-temperature DC four-probe conductivity parallel to the surface of cast films was as large as 8.3 × 10-3 S/cm. Similar measurements with an eight-probe configuration showed no difference between bulk and surface conductivity. The ionic nature of the conductivity was indicated by constant voltage depletion experiments and by secondary ion mass spectroscopy measurements of the residues near the electrodes. The DC two-probe conductivity measured transverse to the sample surface was three to four orders of magnitude smaller than longitudinal conductivity, while the AC two-probe conductivity was even less. Electron microscopy indicated that the films had a layered structure parallel to the surfaces. This structural anisotropy was confirmed by refractive index values obtained from wave-guide experiments and by wide angle X-ray scattering. © 1997 John Wiley & Sons, Inc.
