    Notes: Abstract The NifS and NifU nitrogen fixation-specific gene products are required for the full activation of both the Fe-protein and MoFe-protein of nitrogenase from Azotobacter vinelandii. Because the two nitrogenase component proteins both require the assembly of [Fe-S]-containing clusters for their activation, it has been suggested that NifS and NifU could have complementary functions in the mobilization of sulfur and iron necessary for nitrogenase-specific [Fe-S] cluster assembly. The NifS protein has been shown to have cysteine desulfurase activity and can be used to supply sulfide for the in vitro catalytic formation of [Fe-S] clusters. The NifU protein was previously purified and shown to be a homodimer with a [2Fe-2S] cluster in each subunit. In the present work, primary sequence comparisons, amino acid substitution experiments, and optical and resonance Raman spectroscopic characterization of recombinantly produced NifU and NifU fragments are used to show that NifU has a modular structure. One module is contained in approximately the N-terminal third of NifU and is shown to provide a labile rubredoxin-like ferric-binding site. Cysteine residues Cys35, Cys62, and Cys106 are necessary for binding iron in the rubredoxin-like mode and visible extinction coefficients indicate that up to one ferric ion can be bound per NifU monomer. The second module is contained in approximately the C-terminal half of NifU and provides the [2Fe-2S] cluster-binding site. Cysteine residues Cys137, Cys139, Cys172, and Cys175 provide ligands to the [2Fe-2S] cluster. The cysteines involved in ligating the mononuclear Fe in the rubredoxin-like site and those that provide the [2Fe-2S] cluster ligands are all required for the full physiological function of NifU. The only two other cysteines contained within NifU, Cys272 and Cys275, are not necessary for iron binding at either site, nor are they required for the full physiological function of NifU. The results provide the basis for a model where iron bound in labile rubredoxin-like sites within NifU is used for [Fe-S] cluster formation. The [2Fe-2S] clusters contained within NifU are proposed to have a redox function involving the release of Fe from bacterioferritin and/or the release of Fe or an [Fe-S] cluster precursor from the rubredoxin-like binding site.

    Description: The Earth Resources Observation System (EROS) Program was established by the U.S. Department of the Interior in 1966 under the administration of the Geological Survey. It is primarily concerned with the application of remote sensing techniques for the management of natural resources. The retrieval system employed to search the EROS database is called INORAC (Inquiry, Ordering, and Accounting). A description is given of the types of images identified in EROS, taking into account Landsat imagery, Skylab images, Gemini/Apollo photography, and NASA aerial photography. Attention is given to retrieval commands, geographic coordinate searching, refinement techniques, various online functions, and questions regarding the access to the EROS Main Image File.

    Description: It is pointed out that there are many factors which affect the decision-making process in determining which databases should be selected for conducting the online search on a given topic. In many cases, however, the major consideration will be related to cost. The present investigation is concerned with a comparison of the costs involved in making use of DIALOG and the European Space Agency's Information Retrieval Service (ESA/IRS). The two services are very comparable in many respects. Attention is given to pricing structure, telecommunications, the number of databases, prints, time requirements, a table listing online costs for DIALOG and ESA/IRS, and differences in mounting databases. It is found that ESA/IRS is competitively priced when compared to DIALOG, and, despite occasionally higher telecommunications costs, may be even more economical to use in some cases.

    Description: Numerous public administration studies have indicated that local government agencies for a variety of reasons lack access to comprehensive information resources; furthermore, such entities are often unwilling or unable to share information regarding their own problem-solving innovations. The NASA/University of Kentucky Technology Applications Program devotes a considerable effort to providing scientific and technical information and assistance to local agencies, relying on its access to over 500 distinct online databases offered by 20 hosts. The author presents a subjective assessment, based on his own experiences, of several databases which may prove useful in obtaining information for this particular end-user community.

    Description: It is proposed that the different styles of online searching can be described as either formal (highly precise) or informal with the needs of the client dictating which is most applicable at a particular moment. The background and personality of the searcher also come into play. Particular attention is focused on meatball searching which is a form of online searching characterized by deliberate vagueness. It requires generally comprehensive searches, often on unusual topics and with tight deadlines. It is most likely to occur in search centers serving many different disciplines and levels of client information sophistication. Various information needs are outlined as well as the laws of meatball searching and the adversarial approach. Traits and characteristics important to sucessful searching include: (1) concept analysis, (2) flexibility of thinking, (3) ability to think in synonyms and (4) anticipation of variant word forms and spellings.

    Description: In connection with the utilization of databases, problems can arise with respect to the understanding or evaluation of the provided information. Database guide sheets can be an important aid for overcoming these problems. A number of systems are already sufficiently documented by guide sheets, while other systems do not have guide sheets with adequate descriptive information. Details regarding database guide sheets are discussed, taking into account the type of descriptive information provided, the sample online record, and access points and other related functions for searching the database. Attention is given to cases when a guide sheet should be made, details regarding the preparation of such a sheet, the importance of neatness, and important points which should be kept in mind when guide sheets are being prepared.
