    Notes: Summary Fragments of human breast epithelium, devoid of all stromal and basal lamina components, which maintain their in vivo topological organisation can be cultured for up to 28 days within a reconstituted rat-tail-derived collagen matrix. These organoids initially undergo a loss of structural and 3-dimensional organisation, typified by loss of lumina formed by epithelial cells, and myosin from myoepithelial cells. Their subsequent reorganisation is dependent on the presence of serum, insulin, hydrocortisone, and cholera toxin in tissue culture medium. After this preliminary phase, a reduction in the concentration of serum, insulin, hydrocortisone, and cholera toxin is necessary to allow the structural differentiation of epithelial and myoepithelial cells. The myoepithelial cells also regain their ability to produce the basal lamina component laminin. The use of bovine-dermal collagen as the matrix, rather than rat-tail-derived collagen is shown to result in more stable organisation and differentiation of the organoids. The successful use of single-cell pellets (derived by trypsinisation of the organoids) in place of organoids in such cultures illustrates that there is no requirement for pre-existing cell/ cell contact or topological organisation of cells prior to embedding within the collagen matrix.

    Notes: Abstract  Immunolocalisation of type XIV collagen/undulin in the human mammary gland revealed greater deposition in the interlobular stroma than in the intralobular stroma. The interlobular stroma is located between the breast lobules and their associated intralobular stroma. Fibroblasts isolated from the interlobular stroma synthesised 3- to 5-fold more type XIV collagen/undulin than intralobular fibroblasts, but synthesised type I and type IV collagens in similar amounts. The differential expression of type XIV collagen/undulin was maintained with passage in culture. The results suggest a role for type XIV collagen/undulin in stabilising dense collagen fibrils. The maintenance of two types of structurally distinct stromas may be important during developmental processes in the mammary gland.

    Notes: Abstract The thermal properties (heat capacity, thermal diffusivity, and electrical resistivity) of a Cu + 10 wt% Sn alloy in both solid and liquid phases have been reported. Using these values it was confirmed that the Lorenz relation is suitable for obtaining thermal conductivity from electrical resistivity in the liquid phase of this alloy. Also, the temperature differential (dλ/dT) obtained from such an approach was in excellent agreement with the thermal conductivity values calculated from thermal diffusivity.

    Notes: Abstract A four-probe dc technique for measuring liquid-metal electrical resistivities has been developed, and liquid-resistivity measurements on gallium, tin, lead, copper, a lead tin alloy, a copper tin alloy, and a zinc alloy have been presented. The Lorenz relation has been used to calculate thermal conductivities from these resistivity measurements. The thermal conductivities calculated from these resistivity measurements are (where data are available) in good agreement with recommended liquid-metal thermal conductivities.

    Notes: The role of the cytoskeleton and cell attachments in the alignment of baby hamster kidney fibroblasts to ridge and groove substratum topography was investigated using confocal scanning microscopy. This was carried out with normal cells and cells treated with the cytoskeleton modifiers cytochalasin D, colcemid, and taxol. Actin was localised with fluorescent phalloidin. Tubulin, Vinculin, and intracellular adhesion molecule-1 were visualised by indirect immunofluoresence. The spreading, elongation, and orientation of the cells after 24 h of culture in these conditions were measured on grooves of 5, 10, and 25 μm width and 0.5, 1, 2, and 5 μm depth. We have also observed events over the first 30 min of cell attachment. Five minutes after cell attachment, F-actin condensations were seen close to the intersection of groove wall and ridge top, that is, at a topographic discontinuity. The condensations were often at right angles to the groove edge and showed a periodicity of 0.6 μm. Vinculin arrangement at the early stages of cell spreading was similar to that of actin. Organisation of the microtubule system followed later, becoming obvious at about 30 min after cell plating. The Curtis and Clark theory (that cell react to topography primarily at lines of discontinuity in the substratum by actin nucleation) is supported by these results. The use of cytoskeletal poisons did not entirely abolish cell reaction to grooves. Colocemid increased cell spreading and reduced cell orientation and elongation. Cytochalasin D reduced cell spreading, orientation, and elongation. Taxol reduced cell elongation but did not affect cell spreading and orientation. We conclude that the aggregation of actin along groove/ridge boundaries is a primary driving event in determining fibroblast orientation on microgrooved substrata.

    Notes: Fragmentation pathways of the synthetic pyrethroid cypermethrin and four structurally related insecticides were investigated using a tandem quadrupole mass spectrometer incorporating a hexapole collision cell under positive-ion electron impact ionization conditions. Conventional mass spectrometry using the first quadrupole analyser only and tandem mass spectrometry on selected precursor ions and product ions, and also constant neutral loss scan experiments, were used. Mechanisms and fragmentation pathways are proposed to explain the inherent stability of ions associated with the benzylphenoxy portion of this class of insecticide.

    Notes: The introduction of more rigorous regulations aimed at controlling the use of sulphonated dyestuffs and dyestuff intermediates has pointed to the need for screening methods for the presence of these involatile compounds. Although much work has been caried out recently on the mass spectrometric behaviour of these compounds, it has been concerned principally with the production of molecular weight information using novel ionization methods rather than with any attempt to obtain structurally diagnostic fragment ions. In this mass spectrometric investigation of a range of naphthalene sulphonic acid salts, positive-ion and negative-ion fast-atom bombardment (FAB) mass spectra have been obtained and FAB-tandem mass spectrometry (MS/MS) has been used to acquire spectra (daugher-ion, parent-ion and neutral-loss) in an attempt to develop a useful screening method. On known compounds, best results were found when using a negative-ion neutral-loss scan to detect the loss of 64 mass units (corresponding to loss of SO2). This was confirmed by successful use of this scan to screen a sample of sulphonate residues.

    Notes: The design and use of an RF-only collision-cell ramp generator is described. The generatio controls the collision energy voltage applied to the RF-only collision cell in a hybrid mass spectrometer of BEqQ geometry. The generator enables the collision-cell voltage to be increased or decreased in a linear ramp, over the full 500 V available. The start/stop functions of the ramp are synchronized under data system control to the scan of the quadrupole mass analyser. The collision-induced dissociation spectra so produced show an increase in the product ion abundance.
