    Description: Antibodies play a central role in immunity by forming an interface with the innate immune system and, typically, mediate proinflammatory activity. We describe a novel posttranslational modification that leads to anti-inflammatory activity of antibodies of immunoglobulin G, isotype 4 (IgG4). IgG4 antibodies are dynamic molecules that exchange Fab arms by swapping a heavy chain and attached light chain (half-molecule) with a heavy-light chain pair from another molecule, which results in bispecific antibodies. Mutagenesis studies revealed that the third constant domain is critical for this activity. The impact of IgG4 Fab arm exchange was confirmed in vivo in a rhesus monkey model with experimental autoimmune myasthenia gravis.     Notes: Summary A simultaneous distillation-extraction apparatus proposed by Godefroot has been used for the GC study of volatile components of cheese. 5–10 g of cheese provide a fraction that can be directly injected into a GC or a GC/MS. Two capillary columns (SE-30 and SP-1000) were evaluated, the second one being adequate for quantitative determinations. Camphor was used as internal standard. The coefficients of variation ranged from 1.4 to 11.8. The technique can be used as a fast method for the characterization of cheeses from their volatile component concentrations.

    Notes: Summary A mixed-phase capillary GC column has been designed for the separation of the compounds commonly present in the volatile fraction of cheeses. The design includes the calculation of the optimum phase concentration and the operating conditions. The evaluation of the resulting column indicates that its performance in the qualitative and quantitative analysis of cheese volatile compounds is better than those of other columns coated with a single stationary phase.

    Notes: Summary The stability of atrazine, simazine, alachlor, metolachlor, and deethylatrazine on C18 Empore disks has been determined. Estuarine water (100 mL) spiked at 3 μg L−1 with the target pesticide mixture was preconcentrated on the disks; the disks were then stored at −20°C, 4°C, and at room temperature for periods up to three months and were analyzed by gas chromatography with nitrogen-phosphorus detection. Complete recovery was observed after storage at −20°C throughout the period of the study. Losses up to maximum of 10% were observed after storage at 4°C. Higher losses (up to 24% for alachlor) occurred only at room temperature; the coefficient of variation for these determinations (8–11%) was also higher than that for the others (3–5%). The stability of the pesticides was dependent on the water matrix, on storage temperature, and on properties such as vapor pressure and water solubility.

    Notes: Summary Tetramethylammonium hydroxide in methanol has been proposed as a catalyst for preparing methyl esters of both free fatty acids and glycerides of fats. Nevertheless, procedures normally recognized as suitable can be inadequate when the fat under study contains noticeable amounts of low relative molecular mass esters. The method using tetramethylammonium hydroxide has been examined and modified in order to analyze milk fats by capillary GC. The pyrolysis methylation of free fatty acids in capillary injectors has been also studied. The modified method was satisfactory for both neutral and acid milk fats.

    Notes: Abstract Equilibrium tautomers of aldopentoses (arabinose, ribose, xylose and lyxose) and aldohexones (allose, altrose, gulose, idose and talose) have been analyzed as their trimethyl-silyl ethers by capillary GC. Pentoses were separated on SE-54 and hexoses on OV-225. The four components of the tautomer equilibrium mixture have been identified using GC/MS and NMR data.

    Description: The first chordates appear in the fossil record at the time of the Cambrian explosion, nearly 550 million years ago. The modern ascidian tadpole represents a plausible approximation to these ancestral chordates. To illuminate the origins of chordate and vertebrates, we generated a draft of the protein-coding portion of the genome of the most studied ascidian, Ciona intestinalis. The Ciona genome contains approximately 16,000 protein-coding genes, similar to the number in other invertebrates, but only half that found in vertebrates. Vertebrate gene families are typically found in simplified form in Ciona, suggesting that ascidians contain the basic ancestral complement of genes involved in cell signaling and development.     Description: There is a long-standing controversy regarding the mechanisms that generate the functional subdivisions of the cerebral neocortex. One model proposes that thalamic axonal input specifies these subdivisions; the competing model postulates that patterning mechanisms intrinsic to the dorsal telencephalon generate neocortical regions. Gbx-2 mutant mice, whose thalamic differentiation is disrupted, were investigated. Despite the lack of cortical innervation by thalamic axons, neocortical region-specific gene expression (Cadherin-6, EphA-7, Id-2, and RZR-beta) developed normally.
