Whole cells of Arthrobacter simplex (A.T.C.C. 6946) were entrapped in poly(vinyl alcohol) which was previously esterified with 3-mercaptopropionic acid.The entrapped microorganisms showed steroid-Δl-dehydrogenase activity and the transformation of hydrocortisone to prednisolone was investigated. The steroid-Δl-dehydrogenase activity of the immobilized cells increased in the presence of an inducer such as hydrocortisone. The effect of cell-, substrat-and cofactor-concentration, pH, and temperature was determined. The half-live was analyzed with and without nutrients.The activity of the immobilized Arthrobacter simplex product was 200 μmol. g-1 h-1

Polyvinylalkohol wurde durch polymeranaloge Reaktion mit 3-Mercaptopropionsäure zu einem zum Einschluß von Mikroorganismen geeigneten Polymeren derivatisiert.Die Eignung des Polyvinylalkohol-Derivats zur Immobilisierung von Mikroorganismen wurde mit Corynebakterium simplex (A.T.C.C. 6946) überprüft. Mit den Immobilisaten, die Steroid-Δl-Dehydrogenase-Aktivität zeigten, wurde Hydrocortison zu Prednisolon dehydriert.Die Δl-Dehydrogenase-Aktivität der immobilisierten Zellen erhöhte sich durch Enzyminduktion mit Hydrocortison.Die Aktivität der Immobilisate wurde in Abhängigkeit von der Zell-, Substrat- und Cofaktorkonzentration, des pH-Wertes und der Temperatur untersucht.Die Halbwertszeit der Immobilisate wurde mit und ohne Nährmedium bestimmt. Die Aktivität der Corynebakterium simplex-Immobilisate betrug 200 μmol · g-1 h-1.
