    Description / Table of Contents: Résumé Du collagène d'os compact de mouton est préparé par décalcification dans I'EDTA et à partir de tendons de queux de rats, par extraction dans l'acide acétique et reconstitution dans NaCl. Le dépôt d'apatite dans le collagène osseux de mouton dans une solution de calcification métastable est étudié chimiquement et par microscopie électronique. Le collagène osseux est un bon catalyseur de nucléation pour le dépôt minéral, alors que le collagène de tendons de rat ne l'est pas. Le dépôt minéral du collagène osseux se produit en deux phases cinétiques séparées, une phase rapide de nucléation et une croissance cristalline, donnant naissance à de petits ilots calcifiés et une seconde phase lente de croissance dans des régions ne comportant pas de zones catalytiques. La seconde phase de dépôt minéral paraît être le résultat d'une diffusion inhibée d'ions à travers les fibrilles collagènes alignées, laissant de larges régions de collagène sans minéral, bien que le tampon reste hautement sursaturé. La microscopie électronique permet de penser que les zones de catalyse pourraient avoir un rapport avec la périodicité de 640 Å de collagène, mais l'importance d'un matériel noncollagènique, lié au collagène, n'est pas à exclure. L'activité catalytique faible du collagène reconstitué n'est pas liée à la présence d'inhibiteurs faiblement liés, bien que des inhibiteurs puissent être intimement liés à ce type de collagène, qui pourrait être absent du collagène osseux. La différence d'activité catalytique pourrait intervenir dans la calcification physiologique. Une hypothèse plus générale pour la nucléation de la phase minérale dans les systémes biologiques est nécessaire.
    Abstract: Zusammenfassung Kollagen wurde aus kompaktem Schafsknochen mittels EDTA-Entkalkung und aus Rattenschwanzsehnen durch Essigsäureextraktion und Rekonstitution mit NaCl gewonnen. Die Apatitablagerung aus einer metastabilen Verkalkungslösung auf Schafsknochenkollagen wurde chemisch und im Elektronenmikroskop untersucht. Es zeigte sich, daß das Knochenkollagen ein guter Nukleationskatalysator für die Mineralablagerung ist, was beim Rattenschwanzkollagen nicht zutraf. Im Knochenkollagen erfolgte die Mineralablagerung in zwei getrennten kinetischen Phasen: einer raschen Phase der Nukleation und des Kristallwachstums, welche kleine verkalkte Inseln entstehen läßt, und einer zweiten langsamen Phase, welcher das Wachstum in Bezierken, die keine katalytischaktiven Stellen einschließen, zuzuschreiben ist. Diese zweite Phase der Mineralablagerung wird als Resultat einer verminderten Ionendiffusion durch die enganeinanderliegenden Kollagenfibrillen angesehen, wodurch weite Kollagenbereiche ohne Mineral bleiben, obwohl der Puffer stark übersättigt ist. Elektronenmikrographien ließen vermuten, daß die katalytischaktiven Stellen in einem gewissen Verhältnis zur 640 Å-Periodizität des Kollagens stehen; es konnte jedoch nicht ausgeschlossen werden, daß nicht-kollagenhaltiges Material, welches an Kollagen gebunden ist, ebenfalls eine Rolle spielt. Die schlechte katalytische Aktivität des rekonstituierten Kollagens konnte nicht auf die Anwesenheit von schwachgebundenen Hemmstoffen zurückgeführt werden, obwohl Inhibitoren stark an dieses Kollagen gebunden sein könnten, die jedoch im Knochenkollagen nicht vorhanden sind. Die Unterschiede in der katalytischen Aktivität können mit der physiologischen Verkalkung in Beziehung stehen. Eine allgemeinere Hypothese für die Nukleation einer Mineralphase in biologischen Systemen wäre erforderlich.
    Notes: Abstract Collagen was prepared from compact sheep bone by decalcification with EDTA and from rat tail tendons by acetic acid extraction and reconstitution with NaCl. The deposition of apatite in sheep bone collagen in a metastable calcification solution was studied chemically and by electron microscopy. The bone collagen was shown to be a good nucleation catalyst for mineral deposition, while rat tail collagen was a poor catalyst. Mineral deposition in bone collagen occured in two separate kinetic phases, a rapid phase of nucleation and crystal growth, giving rise to small calcified islands, and a second slow phase, ascribed to growth in regions not involving the catalytic sites. This second phase of mineral deposition is considered to be the result of impaired ion diffusion through the closely-aligned collagen fibrils, thus leaving large areas of the collagen free of mineral even though the buffer remains highly supersaturated. Electron micrographs suggested that the catalytic sites might be in some relationship to the 640 Å periodicity of collagen, but a role for non-collagenous material bound to the collagen has not been excluded. The poor catalytic activity of reconstituted collagen was not due to the presence of loosely-bound inhibitors, although inhibitors could be strongly bound to this type of collagen and be absent from bone collagen. The differences in catalytic activity may have a bearing on physiological calcification. A more general hypothesis for nucleation of a mineral phase in biological systems is required.

    Description / Table of Contents: Résumé Un implant céramique non poreux est testé au niveau du fémur de rat en ce qui concerne son adhésivité à l'os. Un certain nombre de techniques morphologiques sont utilisées pour examiner le rapport entre l'implant et l'os néoformé. La microscopie électronique par transmission et la microscopie par fluorescence après marquage à la tétracycline ont donné les meilleurs résultats. Un rapport étroit entre l'os minéralisé et la céramique a été noté en microscopie électronique. Par marquage à la tétracycline, il semble que l'implant puisse stimuler la formation osseuse.
    Abstract: Zusammenfassung Ein unporöses keramisches Implantat in Rattenfemora wurde auf seine Fähigkeit geprüft, sich mit Knochen zu binden. Eine Anzahl morphologischer Techniken wurde verwendet, um die Beziehung zwischen den Oberflächen von Implantat und neuem Knochen zu untersuchen. Transmissions-Elektronenmikroskopie und Fluoreszenzmikroskopie nach Tetracyclinmarkierung waren die erfolgreichsten Techniken. Eine enge Beziehung zwischen mineralisiertem Knochen und dem Keramikimplantat konnte mit der Transmissions-Elektronenmikroskopie nachgewiesen werden. Das Aussehen der Tetracyclinmarkierung im keramischen Implantat deutet darauf hin, daß dieses wahrscheinlich die Fähighkeit hat, Knochenbildung zu erhöhen.
    Notes: Abstract A nonporous ceramic implant in rat femora was evaluated as to its ability to bond to bone. A number of morphologic techniques were utilized to examine the interfacial relationship of the implant to new bone. Transmission electron microscopy and fluorescence microscopy after tetracycline labelling were the most successful techniques. An intimate relationship between mineralized bone and the ceramic was demonstrated by transmission electron microscopy. The appearance of tetracycline labelling at the ceramic interface indicates that the implant may have capacity to enhance bone formation.

    Notes: Abstract Three-day-old rats were fixed by perfusion with glutaraldehyde and thin slices were cut of the first molar germs. The slices were treated with EDTA and “activated” with buffered solutions containing Mg2+, Ca2+ or Zn2+. Incubation was carried out in buffered solutions (pH 8.5) containing inorganic pyrophosphate and Pb2+. In the Mg2+-activated specimens incubation products were localized to the plasma membranes in the stratum intermedium and the subodontoblastic area. Lead deposits were found on the periphery of the dentinal globules. Incubation products were more randomly distributed in Ca2+-activated specimens whereas those activated with Zn2+ displayed a deposition of lead precipitates mainly corresponding to that seen after activation with Mg2+. The findings are discussed in reference to the localization of alkaline phosphatase in the dentin-producing tissues and it is proposed that the results are indicative of the presence of an inorganic pyrophosphatase in these tissues.

    Notes: Abstract Acid phosphatase activity was mainly localized in the lysosomes in all the regions of the outer epithelium. The transitional portion of the outer epithelium showed more intense activity than the other regions. During shell regeneration the activity of this portion decreased to a minimum level at 12 hours and was restored to normal at 72 hours. The other regions showed no change of activity during shell regeneration. It is postulated that the acid phosphatase in the transitional protion is responsible for conferring calcifiability to the organic matrix of the shell.

    Notes: Abstract Scanning and transmission electron microscopy showed that a ceiling quantity (1.56 mcg) of antifungal antibiotic Pyrrolnitrin caused heavy damage to dermathophyteMicrosporon audouinii Gruby CBS 313-54in vitro. Suitable preparation technique made it clear that the changes involved consisted of hyphal collapse on the edge of the culture, with loss of euplasmic organelles identity and cell autolysis. The cell wall, however, was apparently undamaged. These findings fit in with the suggestion that the mode of action of the antibiotic leads to generalised lipoproteic membranes damage. They must, however, be considered as representing the result of the terminal phase of cell distress.

    Notes: Summary Electron microscope studies of axons, distributing singly or in small bundles in the human ventricular and atrial myocardium, indicate a few per-cent of the axon profiles to be significantly large in diameter (1.5–3.0 μ). They are characteristically packed with a profuse number of mitochondria along with large granular vesicles, glycogen rosettes, lysosomic bodies; and some of them terminate on a “specific terminal cell” (Knoche and Schmitt). These mitochondria-rich, large axons are assumed to be terminal portions of the cardiac afferents. About half of the axons encountered in the ventricle and 2/3 in the atrium are non-vesiculated, usually less than 0.5 μ. in diameter. The varicosities containing numerous vesicles are mostly 0.5–1.5 μ in diameter and are assumed to be terminal portions of the cardiac efferents. The ratio between the number of axon profiles containing small granular vesicles and that of axon profiles containing agranular vesicles without small granular vesicles is 2∶1 in the ventricular myocardium and 1∶1.7 in the atrial myocardium.

    Notes: Summary Techniques of individual cell selection and precise ultramicrotomy have been employed to demonstrate that the contractile ring of cleaving HeLa cells is a transitory cytoplasmic organelle of distinctive fine structure and location. The contractile ring is an uninterrupted annulus encircling the equator of dividing cells exactly where the cleavage furrow forms. It is about 10 microns wide, up to 0.2 microns in thickness, and is composed exclusively of circumferentially aligned thin filaments 40–70 Å in diameter. Contractile ring filaments appear to be associated with the overlying plasma membrane. Controlled experiments with a mold metabolite (cytochalasin B) reveals that within a few minutes the drug abolishes the ability of HeLa cells to undergo cytokinesis. Cytochalasin B seems to decompose the contractile ring. It has no other clearly identifiable effects on other cell structures, notably the mitotic apparatus. Cytochalasin B is the only drug known which selectively inhibits cytokinesis in animal cells. In conclusion, the contractile ring is the most likely organelle responsible for cytokinesis in HeLa cells. Similar organelles probably occur in all cleaving animal cells. Successful cleavage depends upon the structural and functional integrity of the contractile ring.

    Notes: Summary The different patterns of keratin formation that have evolved in the class Reptilia are all variations of a common process. In Squamata (snakes and lizards), a sequence of layers composed of α or β keratin is formed periodically, after which the old epidermal generation is shed. In Chelonia (turtles and tortoises), the epidermis of the shell is composed of only β keratin, whereas the skin of the neck and leg is composed exclusively of α keratin. Molting in toto does not occur and shedding is a continuous process comparable to that in avian and mammalian epidermis. In Crocodilia (crocodiles, caimans, alligators) there is only a single layer of cornified cells, but the composition of the layer varies in different parts of the scale. The hinge regions have many of the morphological characteristics of α and β keratin whereas the center resembles β keratin. The living cells beneath contain accumulations of keratohyalin. There are four ultrastructural characteristics of a keratinized α layer: 1) cellular outlines remain distinct, 2) a thickened plasma membrane forms during keratinization, 3) 80 Å filaments embedded in an amorphous matrix can be seen, and 4) PAS-positive material accumulates in extracellular spaces between the desmosomes. The β layer exhibits none of these features. Instead the cells more or less (depending on species) coalesce into a compact layer which becomes attenuated in the hinge regions. A 30 Å filament pattern can be seen. The mesos layer of squamates resembles the hinge region of crocodilians, exhibiting a combination of the characteristics of both α and β keratin.

    Description / Table of Contents: Zusammenfassung Durch Vergleich fluoreszenz- und elektronenmikroskopischer Untersuchungen am zentralen Nervensystem von Planorbarius corneus L. wird nachgewiesen, daß in den Schlundringganglien Neurosekretzellen vorkommen (Nachweis mit Pseudoisocyaninchlorid), die mit Nervenzellen nicht identisch sind, die durch ihren hohen Gehalt an biogenen Aminen auffallen (Nachweis durch die Methode von Falck und Owman, 1965). Es können daher im Schlundring von Planorbarius corneus peptiderge und aminerge Neurosekretzellen unterschieden werden. Die PSC-positiven Neurosekretzellen enthalten elektronendichte Elementargrana und die aminergen Neurone „dense-core“ Vesikel. Der Nachweis biogener Amine in einigen Nervenzellen von Planorbarius corneus spicht für deren chemische Identität mit Transmittersubstanzen, ihre hohe Konzentration aber für eine Abgabe in die Körperflüssigkeit.
    Notes: Summary The neurosecretory system of the snail Planorbarius corneus has been investigated by fluorescence and electron microscopy. With the fluorochrome Pseudoisocyanin the established neurosecretory system in the cerebral ganglia and single neurosecretory cells in the other ganglia show an intensive yellow fluorescence. Electron micrographs reveal the presence of electron dense granules (elementary granules) in the pericarya and the axons of neurones which have the same localisation in the ganglia as the pseudoisocyanin-positive cells. The fluorescence technique for biogenic amines produces yellow and green fluorescence within neurons and in the neuropil and nerves. The fluorescence obtained in determinable areas and neurones correlates well with the electron microscopic location of “dense-core” vesicles within the pericarya and axons of cells in even the same areas. It is discussed, that in this animal both types of cells are so-called “neurosecretory cells”, because the high content of elementary granules in the “peptidergic neurosecretory cells” and of “dense-core” vesicles in the “aminergic neurosecretory cells” is an indication for secretion of these products in neurohaemal areas (circulatory channels).

    Notes: Summary The fine structure of amniotic and amniotic-plaque epithelia has been studied from normal term pregnancies. The columnar/cuboidal amniotic epithelial cells usually have apical or central nuclei, some free ribosomes, patches of granular endoplasmic reticulum, juxtanuclear Golgi complexes, rod-shaped mitochondria, lipid droplets and some glycogen granules. They have short, blunt microvilli which frequently branch and bathe in the amniotic fluid. The lateral plasma membranes enclose tortuous intercellular spaces which are always interrupted by variously folded processes and desmosomes. The epithelial cells rest on a basal lamina and exhibit highly folded basal processes. The amniotic epithelial cells are neither distinctly Golgi and fibrillar types nor “light” and “dark” in appearance. Amnion from near the umbilical cord contains many microscopic and several large plaques. Similar structures are not found on the reflected amnion. The microscopic plaques are whitish and translucent, whereas the large ones are opaque. The large plaques vary between 1–3 mm in diameter, and are over 15 cell layers thick. Each large plaque has a main central region and edges continuous with either the microscopic plaque or the simple amniotic epithelium. The main region shows four zones, namely, stratum basale, stratum spinosum, stratum granulosum and stratum corneum. Such zones are not distinct at the edges. The fine structure of basal cells compares with the amniotic epithelial cells, but the cells of spinosum and granulosum layers possess variable amounts of tonofibrils, keratohyalin granules, free ribosomes and other cytoplasmic organelles and inclusions. The corneum cells are keratinized and are frequently separated by intercellular spaces. They slough into the amniotic cavity singly or as a sheet, and contribute towards the composition of the amniotic fluid. The plaques are of amniotic origin, and are not formed by adhesion of either squamous cells or fetal skin cells (masses of keratinized squames). The present observations suggest that the occurrence of amniotic plaques is normal. The presence of plaques may not be necessarily associated with fetal abnormality. However, increase in numbers of plaques may be caused by conditions of fluid imbalance. The homology and significance of plaques in eutherian mammals have been discussed.

    Description / Table of Contents: Résumé L'ultrastructure des cellules interstitielles du testicule de Lacerta vivipara a été étudiée entre le printemps et l'automne pendant deux années. Le retioulum endoplasmique lisse, et les mitochondries à crêtes tabulaires sont les organites les plus remarquables comme dans les autres cellules productrices de stéroïdes, mais les liposomes et l'appareil de Golgi sont bien représentés aussi. Les variations ultrastructurales les plus significatives apparaissent entre le printemps et le début de l'été. Au printemps, alors que les caractères sexuels secondaires sont hypertrophiés, un système remarquable de vésicules et de vacuoles se développe à partir du reticulum et probablement aussi du Golgi. Au début de l'été, lorsque les caractères sexuels secondaires sont atrophiés, les vacuoles sont moins nombreuses et le reticulum forme un réseau dense de tubules typiques, souvent étroitement associés aux liposomes; les crêtes mitochondriales sont gonflées. Ces images sont discutées en fonction de l'activité saisonnière d'élaboration d'hormones. L'hypertrophie des systèmes membranaires au printemps correspond probablement à la production ou (et) à l'excrétion des hormones androgènes. Au début de l'été, la cellule n'élabore pas d'androgènes, mais n'est peut-être pas complètement inactive: elle pourrait stocker des précurseurs hormonaux.
    Notes: Summary Interstitial cells of the testis of Lacerta vivipara have been studied electronmicroscopically in animals obtained between spring and autumn. Smooth endoplasmic reticulum and mitochondria with tubular cristae are the most prominent organels, lipid droplets and Golgi apparatus being also well developed. The most significant ultrastructural changes occur between spring and the beginning of summer. In spring, during the hypertrophy of secondary sexual characters, a conspicuous system of vesicles and vacuoles originates from the smooth endoplasmic reticulum and probably also from the Golgi apparatus. At the beginning of summer, when secondary sexual characters are atrophied, vacuoles are less prominent and the smooth endoplasmic reticulum consists of a dense network of typical tubules, often closely associated with the lipid droplets; the cristae of the mitochondria are swollen. These ultrastructural findings are discussed in relation to the production of hormones. The hypertrophy of membrane systems in spring corresponds presumably to production or (and) release of androgen hormones. In the beginning of summer the cell does not produce androgens, but probably is not completely inactive: it may store precursors of hormones.

    Description / Table of Contents: Zusammenfassung Normale und hypoxische Herzmuskelzellen aus der Wand des linken Ventrikels der Ratte wurden quantitativ-morphologisch anhand von elektronenmikroskopischen Längsschnitten nach Perfusionsfixierung untersucht. In normalen Zellen waren alle Myofibrillen relaxiert, die mittlere Sarcomerlänge betrug 2,2 μm. Die Schnittfläche wurde zu 55% von Myofibrillen, zu 27% von Mitochondrien und zu 18% von Grundplasma und Reticulum eingenommen. Die zwischen den Myofibrillen liegenden Mitochondrien waren längsoval und im Mittel 2,3mal so lang wie breit. Es bestand kein Unterschied zwischen subendokardial und subepikardial gelegenen Zellen. 10 min nach Erstickung der Tiere waren in den sonst unauffälligen Muskelzellen die Glycogengranula vermindert. Nach 20 min führte die Hypoxie zu einer Zunahme der relativen Schnittfläche der Mitochondrien um etwa 16% und zu einer beginnenden Kontraktur der Myofibrillen (Sarcomerlänge 2,0 μm). 20 min Hypoxie in Hypothermie (25–30°C intrathorakal) veränderte die normale Zellstruktur dagegen kaum. Wenn die Herzen während der 20 min dauernden Hypoxie in Normothermie mit einer procainhaltigen sauerstoff- und glucosefreien Blutersatzlösung durchspült wurden, waren die Myofibrillen relaxiert, die Schwellung der Mitochondrien dagegen wurde nicht reduziert. 30 min nach Erstickung wurde die Kontraktur stärker (Sarcomerlänge 1,7 μm). Nach 60 min bildeten sich Superkontraktionsknoten, einzelne Myofibrillen waren in Höhe der I-Bänder unterbrochen. Die Cristae der Mitochondrien wichen auseinander, die Schnittfläche der Mitochondrien hatte um 27% zugenommen. Während in Normotherapie eine Asphyxie des Tieres bereits nach 10 min die Herzmuskelzellen funktionell schwer schädigt, ist die Schädigung morphologisch erst nach 20 min eindeutig. Das bedeutet, daß für die elektronenmikroskopische Präparation eine Hypoxie von unter 10 min bedeutungslos ist. Hinsichtlich der morphologischen Manifestationszeit für die Unterbrechung der Sauerstoffversorgung stimmen unsere Befunde an Herzmuskelzellen gut mit vergleichbaren Angaben an Leberzellen überein.
    Notes: Summary In heart muscle cells of the left ventricle of rats the distribution of cell organelles and their reaction to hypoxia were investigated by electron microscopy. In normal hearts fixed by perfusion with aldehydes, the mean sarcomere length was 2.2 μm. 27% of the longitudinal sectional area was occupied by mitochondria, 55% by myofibrils and 18% by sarcoplasmic reticulum and ground plasm. The mitochondria situated in rows between the fibrils were oval and measured 2.3 times more in length than in width. There was no difference between cells from subendocardial and subepicardial regions. 10 min hypoxia (complete occlusion of the trachea) did not affect the appearance of muscle cells but diminished the number of glycogen granules. After 20 minutes the area occupied by mitochondria was increased by 16%, the mitochondria between the myofibrils were more spherical and only 1.5 times longer than wide. The sarcomeres shortened to 2.0 μm. With hypothermia (25–30°C) hypoxia of 20 minutes duration did not affect the cell structure. Perfusion of the heart by a saline solution, which contained procaine but neither oxygen nor glucose, for 20 minutes prevented shortening of the sarcomeres but not swelling of the mitochondria. 30 minutes after occlusion of the trachea the myofibrils shortened to a sarcomere length of 1.7 μm. After 60 minutes irregularly and excessively contracted myofibrils appeared and some sarcomeres were interrupted at the level of the I-bands. In some of the swollen mitochondria the cristae were widely separated. The increase of the area occupied by mitochondria was 27%. Asphyxia affects heart muscle cells severely with respect to function within 10 min, but morphologically it takes 20 min before a definite effect can be noticed. As to the time after which lack of oxygen is manifested morphologically, our results are consistent with findings in liver cells.

    Notes: Summary Thymic small lymphocytes of dd-mice were qualitatively and quantitatively studied by electron microscopy. Differences in fine structure were revealed between cortical and medullary small lymphocytes. Cortical small lymphocytes are rounded in cell outline with a round nucleus. The cytoplasm surrounding the nucleus as a narrow rim is scanty and appears relatively dense due to an abundance of free ribosomes. The cell organelles are not well developed. On the other hand, medullary small lymphocytes are more irregular in shape with uneven cell membranes. Their nuclei are also more irregular in outline with frequent infoldings of the nuclear membrane. The cytoplasm is more abundant and paler with less numerous ribosomes. The cell organelles are better developed. Quantitative analysis was made of both cortical and medullary small lymphocytes by means of the point counting method. The nuclei of both cortical and medullary small lymphocytes are almost the same in size (a diameter of 4.9 μ). The cell sizes are different between cortical and medullary lymphocytes: cortical small lymphocytes with a diameter of 5.5 μ were smaller than medullary ones with a diameter of 6.4 μ. Cortical small lymphocytes are very sensitive to the destructive effects of hydrocortisone, whereas the medullary ones are resistant. Periarteriolar lymphoid sheath in the splenic white pulp, which is known to be a thymus-dependent area, contains small lymphocytes that were similar in cytological details to medullary small lymphocytes of the thymus. In the light of the recent knowledge about a recirculating long-lived small lymphocyte pool, it appears probable that medullary small lymphocytes represent a contribution to the pool and that small lymphocytes with a long life span can be cytologically identified by electron microscopy.

Im Bereich von Abzweigungen kleiner Nerven bildet das Perineurium ein gekammertes, stark kollagenfibrillenhaltiges Hüllensystem. Die Endigung der einschichtigen Perineuralhülle hat die Form einer in das Interstitium geöffneten Röhre. Im ansatznahen Drittel des Mesenteriums und in der Schleimhaut der Trachea wurden ausschließlich perineuriumfreie Nerven gefunden. Es liegt nahe, diese perineuriumfreien Bezirke zu funktionellen Endräumen zusammenzufassen. In perineuriumfreien Bezirken wird das gehäufte Auftreten von Nervenauftreibungen mit Vesikeln, die Transmittersubstanz enthalten, und Mitochondrion beobachtet. Die erhobenen Befunde werden mit den an dickem Perineurium gewonnenen Ergebnissen verglichen und diskutiert.
    Notes: Summary The perineurial sheath of small tracheal and mesenteric nerves consists of a layer of flat, basement membrane coated cells. Its ramifications form a complicated layered system, interspersed with collagen fibrils. The tube formed by the thin perineurial sheath ends openly, which affords communication between the respective interstitial spaces. The mesenteric nerves close to the duodenum as well as the nerves in the vicinity of the tracheal epithelium show no perineurial sheath. Peripheral nerves lacking a perineurial layer seem to be close to their terminations. Their fibers show varicosities containing transmitter vesicles and mitochondria with longitudinally orientated cristae. Our results are compared with those found in thick, multilayered perineurial sheaths and functional differences are discussed.

    Notes: Summary The morphology of postnatal differentiation of the Golgi apparatus, the nucleus, the perikaryon, and the dendrites was studied in Purkinje cells of the rat cerebellum for 30 days after birth using histochemical, histological, and electron microscopic methods. The Golgi apparatus during differentiation undergoes morphological and positional changes. From the 1st to 7th postnatal day, the Golgi apparatus is found in a supranuclear position, and is connected with the axes of differentiating primary dendrites by beam-like processes. From days 8 to 11 this connection disappears, and most of the Golgi apparatus assumes a lateronuclear and infranuclear position. After the 11th or 12th day, the Golgi apparatus is found in perinuclear and peripheral cytoplasmic positions. The formation of granular endoplasmic reticulum occurs in the vicinity of the perinuclear Golgi apparatus. The differentiation of cell and nuclear forms requires approximately 20 days. The morphological changes of differentiation are discussed in relation to the participation of the Golgi apparatus in the differentiation of dendrites and in the formation of the granular endoplasmic reticulum.

    Notes: Summary The structure of the cerebral ganglion of the shore crab, Carcinus maenas, is investigated by conventional electron miscroscope techniques, with particular emphasis on the relation of intracerebral blood vessels to other elements in the brain. The ganglion is permeated by a continuous network of channels which may be interpreted as invaginations of the ganglion surface. The afferent vessel (cerebral artery) is of mesodermal origin, but apparently terminates as an open-ended vessel soon after entering the brain, where it runs within the invaginated channels. The greater part of the cerebral vasculature, therefore, has no mesodermal endothelial lining. Tissue components in the diffusion path between blood and brain which could conceivably restrict diffusion, are the thick glial basement membrane, junctions between perivascular and between interstitial glia, and polymeric material in the extracellular space. However, apart from a barrier to large colloidal particles at the basement membrane, the present EM observations do not decisively pinpoint sites of diffusional restriction, nor can they be interpreted as evidence that such restriction exists.

    Notes: Summary Studies of cardiogenesis in the chick embryo focus attention upon the intercellular junctions of epicardial, myocardial, and endocardial cells, and the role they play in diffusion across the cardiac wall. Cell membranes of apposed epicardial cells approach as close together as 40 Å; those of the endocardium additionally form focal tight junctions. In the myocardium focal tight junctions are restricted to the apposed membranes of the superficial layer of cells. The majority of close appositions in all parts of the myocardium are 40 Å gap junctions. Desmosomes and fascia adherens are distributed throughout the myocardium. Diffusion of horseradish peroxidase through the epicardium and endocardium occurs primarily through the intercellular junctions. The width of the cleft between cells, 200–300 Å, also permits the diffusion between cells of the larger ferritin particles. Pinocytotic activity, responsible for ferritin transfer across mesothelial and endothelial cells in the adult, is not significant. Tracers injected into the pericardial cavity or vasculature can be observed passing through the heart in the direction of their respective diffusion gradients. Unlike the apical junctions of epithelial cells, to which they have been compared, membrane specializations of the superficial myocytes do not form a seal separating the pericardial cavity, or subepicardial space, from the extracellular spaces of the myocardium.

    Notes: Summary The ultimobranchial body of fresh-water turtles,Pseudemys scripta andChrysemys picta, ultrastructurally and histochemically resembles the gland of other vertebrate groups and the homologous thyroid parafollicular cells of mammals. Characteristic features of all of these tissues are secretory granules measuring approximately 150–250 mμ, a distended endoplasmic reticulum, prominent Golgi regions and large numbers of free ribosomes. Unusual features of the turtle ultimobranchial body are an abundance of large cytoplasmic bodies measuring 800–1000 mμ and a dense, homogenous material within the lumina of the ultimobranchial follicles. The large cytoplasmic bodies usually occur near the luminal portion of the cells and are of similar electron density to the luminal contents, suggesting a possible functional relationship of these two glandular components.

Chez le Pigeon entier, l'Eminence Médiane Caudale (E.M.C.) se distingue de l'Eminence Médiane Rostrale (E.M.R.) essentiellement par l'absence dans les deux couches les plus externes (couches palissadique et superficielle) de l'E.M.C. de granules de gros calibres (1600 à 1900 Å), la rareté de granules de diamètre moyen (1200–1400 Å) et la prédominance de petites vésicules à cœur dense de 600–800 Å. 2. La préhypophysectomie entraine: a) dans l'E.M.R. la quasi disparition de granulations dans les deux couches externes; b) dans l'E.M.C. la ≪vidange≫ de nombreux axones, mais un enrichissement relatif, parmi les granulations restantes, des granulations de gros calibre (1600–1900 Å) aux dépens des granules de plus petit calibre. 3. Un shock insulinique entraine des modifications du même ordre: a) déplétion des granules denses, limitée dans ce cas à la portion la plus antérieure des deux couches externes de l'E.M.R.; b) enrichissement relatif des granulations de moyen (1200–1400 Å) et de gros (1600–1900 Å) calibre dans l'E.M.C. avec, en plus dans l'E.M.C., un enrichissement en vésicules de type synaptique. 4. Un traitement à la métopirone produit un accroissement du nombre des granulations de moyen (1200–1400 Å) calibre dans les couches externes de l'E.M.R. et de l'E.M.C., et un enrichissement important de l'E.M.C. en vésicules de type synaptique. 5. Le traitement à la prednisolone conduit à un enrichissement très marqué des couches externes de l'E.M.R. en grains de 1200–1400 Å, et à un enrichissement des couches externes de l'E.M.C. en granulations de 1000 Å. Ces résultats sont discutés dans la perspective des régulations hypothalamo-corticotropes, particulièrement en ce qui concerne les granules de 1200–1400 Å.
    Notes: Summary The effects of adenohypophysectomy, and of several experimental interventions on the hypothalamo-pituitary-adrenal cortical axis have been studied in relation to the fine structure of the median eminence in the pigeon. 1. In control animals, the following morphological features of the caudal median eminence (C.M.E.) distinguish it from the rostral median eminence (R.M.E.): a) the absence in both external layers of the C.M.E. of large (1,600–1,900 Å) electron-dense granules, b) the presence in the C.M.E. of a small number of medium-size (1,200–1,400 Å) granules, and c) the predominance in the C.M.E. of small (600–800 Å) dense-core vesicles. 2. Adenohypophysectomy leads to: a) almost complete disappearance of electron-dense granules in both external layers of the R.M.E., and b) “emptying” of numerous axons and a relative increase in the number of large (1,600–1,900 Å) granules in the C.M.E. 3. Insulin shock produces modifications similar to those of adenohypophysectomy. The depletion of electron-dense granules from the axons is, however, restricted to the most anterior part of the R.M.E., and, in the C.M.E., the relative increase in the number of larger granules affects the 1,200–1,400 Å and the 1,600–1,900 Å size granules. 4. Metopirone enhances the number of medium-size (1,200–1,400 Å) granules in the external layers of both the R.M.E. and the C.M.E. and causes a significant increase in the number of synaptic-like vesicles in the C.M.E. 5. Prednisolone treatment leads to a marked enrichment of the external layers of the R.M.E. with 1,200–1,400 Å granules, and of the external layers of the C.M.E. with 1,000 Å granules. These results have been discussed with special reference to the hypothalamic control of the adrenocorticotropic function, especially reviewing the role of the 1,200–1,400 Å granules.

Elektronenmikroskopische Untersuchungen an der Neurohypophyse von Ratte und Forelle ergeben, daß sich eine Exocytose von Elementargranula an den Endigungen der neurosekretorischen Fasern nur selten abspielt. Es wird daher angenommen, daß die Abgabe von Hormonen in der Neurohypophyse in der Regel nach dem Muster des „membrane-release“ abläuft. 2. Die Exocytose wird nicht durch eine unmittelbare tangentiale Fusion der Membran des Elementargranulums mit dem Plasmalemm der Nervenendigung (Axolemm) eingeleitet. Vor allem bei Anwendung eines Goniometertisches wird erkennbar, daß vor der Exocytose zwischen Axolemm und Membran des Granulums eine Verbindung in Gestalt eines Stieles entsteht. Die Länge dieses Verbindungsstückes entspricht etwa 2 Axolemmdicken. An der Basis des Stiels im Axolemm tritt das Stoma auf, durch das der Inhalt des Granulums bzw. dieses selbst das Axonende verläßt. 3. Die Herkunft kleiner membrannaher Vesikel (Durchmesser 500 Å) in den Endigungen neurosekretorischer Nervenfasern in der Neurohypophyse konnte nicht geklärt werden. Anzeichen einer kompensatorischen Endocytose im Sinne von Nagasawa, Douglas und Schulz (1970) wurden nicht beobachtet.
    Notes: Summary 1. Electron microscopical investigations of the neurohypophysis in rat and trout reveal that exocytosis of neurosecretory elementary granules from the nerve endings occurs only rarely. The authors are of the opinion that hormone release in the neural lobe follows mainly the “membrane-release” pattern. 2. Exocytosis is not performed by tangential fusion of the elementary granule membrane and the plasmalemma of the nerve ending (axolemma). Administering the goniometer technique one can observe the appearance of a stalk-like structure connecting the two membranes. The basis of the stalk in the axolemma corresponds to the site of the stoma through which the core of the vesicle leaves the nerve ending. 3. The mechanism of the origin of small clear vesicles (diameter 500 Å approx.) near the axolemma of the neurosecretory terminal has not been elucidated. The authors did not observe equivalents of a compensatory endocytosis in the vicinity of granules released by exocytosis.

    Notes: Summary The organization of the cerebral ganglion of the shore crab Carcinus maenas, is investigated by conventional histological and electronmicroscopic techniques. This study forms part of a comprehensive survey of the blood-brain interface, particularly interesting in this group, as decapod Crustacea are unusual among invertebrates in possessing an intracerebral blood supply. Apart from the intracerebral blood vessels, tissue organization is closely similar to that observed in insect central neural ganglia. The ganglion is surrounded by the neural lamella, an acellular connective tissue sheath, probably containing mucopolysaccharide and collagen. A layer of specialised glia, the perineurium, immediately underlies the neural lamella, and appears to contribute to its formation. Large glia occupying a conspicuous cortical zone below the perineurium may be involved in glycogen metabolism and storage. Further morphologically distinct glial types are observed associated with neurones and blood vessels, but all neuroglia within the ganglion are probably of common origin. Neurone cell bodies are generally situated peripherally in groups, and send axons into neuropil (synaptic) areas in the ganglion core. Large lacunae in the cortical region and narrower 20 nm clefts deeper in the ganglion, constitute the interstitial space, and contain deposits of fibrillar material. Possible physiological implications are discussed.

    Notes: Summary The developmental changes in the neutrophilic myelocyte from normal human bone marrow have been analyzed by means of phase contrast and electron microscopy. This developmental stage is characterized principally by the elaboration of secondary (specific) granules. In addition, there is a modest decrease in cell size, a decrease in the number and mean size of primary (azurophil) granules, a decrease in the number of polysomes, free ribosomes and mitochondria, a depletion of rough endoplasmic reticulum, an increase in cytoplasmic glycogen, an increase in chromatin aggregations and a loss of nucleoli, and the formation of a markedly indented nucleus. The myelocyte stage has been subdivided into three arbitrary phases based upon morphological and functional characteristics which relate to the onset, active production and cessation of secondary granulogenesis.

    Notes: Summary Two types of nerve cells, namely, neurosensory and neurosecretory cells have been identified and described in the gastrodermis of Hydra pseudoligactis. The morphological criteria used for the identification of gastrodermal nerves are based on those presented previously for epidermal nerves. The third type of nerve cell in the epidermis, ganglionic cells, was not observed in these studies. The distribution, function and origin of gastrodermal nerve cells are discussed briefly.

    Notes: Summary The fine structure of the pterin layer was investigated in both wild type Rana pipiens and Rana pipiens homozygous for the speckle mutant gene. No difference in morphology of the layer was noted between the wild type and mutant. The layer lines the outer surface of the stratum compactum of the dermis and separates this stratum from the stratum spongiosum. The pterin layer consists of extra-cellular material and contains membrane-bounded granules filled with fine spicules. Many of the spicules are somewhat similar in appearance to the initial calcification loci present in developing membrane bone. The layer first appears in the tadpole at approximately stage 14 (Taylor and Kollros, 1946); subsequent developmental stages are described.

Sie kommen wahrscheinlich schon normalerweise bei verschiedenen Tierarten in bestimmten Regionen des ZNS vor.
    Notes: Summary The electron microscopic study of a rat's striatum has revealed peculiar parallel arrays of membrane-bound cisternae with a strikingly dense intercisternal cytoplasmic matrix in the perikarya of a few neurons. The finding corresponds exactly to the unique lamellar configurations recently described in nerve cells of the entopeduncular nucleus and orbitofrontal cortex of the cat. These “dark cisternal fields” are regarded as distinct districts of the endoplasmic reticulum in a special functional state. They seem to occur normally in certain regions of the CNS in different animal species.

    Notes: Summary The cerebral ganglion and the ventral nerve cord of Lumbricus terrestris have been studied with the electron microscope. The results are as follows: In the neuropile small granular vesicles (300 to 500 Å) occur in some varicose nerve fibres after fixation with potassium permanganate. This indicates the presence of noradrenaline. Sometimes only a few of the vesicles produce a positive reaction. After incubation with α-methyl-noradrenaline the numbers of nerve terminals with small granular vesicles greatly increase, indicating the presence of dopamine and/or 5-hydroxytryptamine. In this case the reaction is now complete. The number of small granular vesicles is largest in the terminal swellings. These findings are consistent with histofluorescence, chemical, and microspectrofluorometric analyses, which have demonstrated noradrenaline, dopamine, and 5-hydroxytryptamine in neurones in the central nervous system. Large granular vesicles (600 to 900 Å) are to be found in some perikarya, not identical with neurosecretory cell bodies. In this case the granular vesicles in the axon are smaller and fewer. This indicates a simultaneous proximo-distal transport and gradual decrease in size of the granular vesicles. The intraneuronal distribution of the vesicles is in agreement with the distribution of the fluorophores in the fluorescent neurones. Neurosecretory neurones are found most likely not to contain monoamines.

    Notes: Summary The arcuate nucleus, median eminence, and the lateral preoptic area from the brains of aldehyde-perfused male and female rats were examined by electron microscopy. In the lateral preoptic area, three neuronal types are described: a small light neuron, a larger light one, and a dark neuron resembling the larger light one in size and nuclear shape. Many myelinated axons are interposed among single neurons or neuronal pairs. The relationship of structures to each other is discussed. Several observations not previously reported are illustrated from tissue of the arcuate nucleus and median eminence.

    Notes: Summary In Drosophila melanogaster, the cyst cell that surrounds the head region of sperm bundle becomes spheroidal or ellipsoidal and is trapped by the terminal epithelium of the testicular wall during the synchronous coiling of sperm. Extensions of this cyst cell are projected caudally into the interspaces between sperm heads probably to anchor the heads. Coiling of sperm tails is initiated at the head region and proceeds by the progressive retraction of the linear portion from the apical testicular region into the coiled portion at the base. The addition of one turn of coil is accompanied by one full rotation of the sperm bundle. When coiled, normal tails are tightly packed into a hexagonal lattice, and minute tubular structures of about 150 Å in diameter occupy the space between them. Sperm with abnormal tails are separated from those with normal ones and isolated into a separate part of the cyst lumen. Acid phosphatase is involved in the dissolution of the minute tubules for the liberation of sperm from the cyst. Sperm are released leaving the major portion of the cyst cells intact. This portion contains degenerating abnormal tails and the waste products of the individualization process. This detritus is ingested by the terminal epithelium and eventually degenerates.

Le taux d' incorporation de la cystéine S35 dans les cellules B est identique chez toutes les femelles et il est légèrement supérieur à celui des neurones banaux. Ces résultats confirment l'inactivité sécrétoire protéique des cellules B. Les cellules C incorporent 3 à 5 fois plus de cystéine S35 que les neurones banaux. Elles synthétisent donc une ou plusieurs protéines contenant de la cystéine ce qui réaffirme leur activité neurosécrétrice chez Locusta. Les cellules A possèdent le taux d'incorporation de cystéine S35 le plus élevé: 5 à 8 fois celui des neurones banaux. Chez toutes les femelles, les cellules A synthétisent plus de neurosécrétion et en éliminent proportionellement plus que les cellules C. La neurosécrétion A est élaborée sous sa forme figurée plus rapidement (30 min) que la neurosécrétion C (60 min). Le renouvellement de la neurosécrétion A est donc quantitativement plus important et plus rapide que celui de la neurosécrétion C. Chez les femelles immatures, les cellules A et C synthétisent plus de matériel et en éliminent proportionnellement plus que chez les femelles mûres. Le temps nécessaire à l'élaboration et à la vidange des grains de neurosécrétion A est identique chez toutes les femelles. Il en est de même pour le matériel C. Le renouvellement des neurosécrétions A et C est donc plus important chez les femelles immatures que chez les femelles mûres mais il n'est pas plus rapide. L'accumulation du matériel fuchsinophile dans les cellules A et C lors de la maturation ovarienne correspond à une réduction de leur fonction neurosécrétrice: elle résulte d'une diminution de l'activité d'élimination des cellules neurosécrétrices A et C supérieure à l'affaiblissement de leur activité de synthèse.
    Notes: Summary After injection of 35S-cysteine, the A, B, C cells and the ordinary neurones of pars intercerebralis in Locusta — immature and mature females — are radioactive. The rate of incorporation of 35S-cysteine into the B cells is the same for all the females, and it is slightly higher than the rate of incorporation of 35S-cysteine into the ordinary neurones. These results demonstrate the proteinic secretory inactivity of the B cells. The C cells incorporate 3 to 5 times more 35S-cysteine than ordinary neurones. Thus, the C cells synthesize one protein or several proteins with cysteine; this observation confirms their neurosecretory activity in Locusta. The A cells have the highest rate of incorporation of 35S-cysteine: 5 to 8 times the one of ordinary neurones. In all the females, the A cells produce and release proportionally more neurosecretion than the C cells. The production of granules is faster in A cells (30 min) than in C cells (60 min). The turnover of the A neurosecretion is consequently higher and quicker than the C neurosecretion. In immature females, the A and C cells synthesize and release proportionally more material than in mature females. The time necessary for production and release of the A neurosecretion is the same for all the females. It is so for the C material. The turnover of the A and C neurosecretions is thus more important in immature females than in mature females but it is not more rapid. The accumulation of stainable neurosecretory material in A and C cells at the time of ovarian maturation is associated with a reduction of their neurosecretory activity: it is due to a decrease of the rate of release of the A and C cells being more important than their rate of production.

    Notes: Summary As described in other invertebrate muscles, most thick myofilaments of the dorsal longitudinal muscle of Sabellastarte magnifica appear to be obliquely arranged with respect to the longitudinal axis of the myofibrils. Some sections, however, show long bundles of myofilaments parallel to the longitudinal axis of the myofibrils. Since the oblique striation concept cannot account for such images, a different (longitudinal) arrangement is proposed for Sabellastarte which can account for all observed images. A wax and threads model built according to this arrangement has been used to demonstrate that by oblique sectioning the longitudinal model can generate a false appearance of filament obliquity.

    Notes: Summary The tentacle of Rhabdopleura compacta (Hemichordata) consists of two layers of cells surrounding a central coelomic cavity. The two layers of cells are separated by a cell free basement lamella. The tentacles on the arms of Rhabdopleura bear three longitudinal rows of cilia. The ciliated cells are closely associated with bundles of nerve fibres, and between some of the cells and nerve fibres there are synapses. The peripheral regions of the ciliated cells are joined to one another by desmosomes. Tonofibrils join some of these desmosomes to the kinetosomes of the cilia. The nerve fibres are confined to the ectodermal layer and the muscle cells to the layer of cells within the basement lamella. In the ectodermal layer besides ciliated cells there are mucus cells, densely pigmented cells, and green bodies. The function of these last two types of cells is secretory. Most of the epithelial cells have microvilli upon their free borders.

    Notes: Summary The ultrastructural alterations in the anterior midgut epithelium of the fly Calliphora were investigated by electron microscopy. Nine developmental stages, from late larval to late adult, were studied. During metamorphosis, a tube is formed by proliferation of undifferentiated cells residing in cell nests in the basal part of the larval epithelium. The process of microvillus formation is described and compared with that in the embryonic stage. Differences between larval and imaginal cells were found in the rate of microvillus formation and in the occurrence of dense vesicles. The formation of a basal labyrinth coincides roughly with microvillus formation. Glycogen deposits occur mainly in the pupal stage and are almost always absent after emergence. Epithelial crypts are formed in late pupae by modification of the existing layer of cells together with a localized development of muscle fibres. In the adult stage changes may take place in the form of the cell apex, which generally assumes a more pointed shape, and in the amount of osmiophilic material, aggregating almost exclusively in the supranuclear part of the cell.

Les cellules de ce réseau se caractérisent à la fois par la multiplicité de leurs prolongements et par leur organisation autour de cavités pour la plupart submicroscopiques, «les follicules». Les cellules folliculo-stellaires correspondent à une partie des cellules chromophobes de la microscopie photonique. La fente hypophysaire, par de nombreux caractères, semble faire partie de ce système folliculo-stellaire, dont elle pourrait représenter une région privilégiée par sa situation, ses dimensions et son contenu. L'existence des cellules folliculo-stellaires dans un grand nombre d'espèces, leurs caractères cytologiques hautement différenciés, leurs relations étroites avec le milieu intérieur interstitiel et leurs modifications en fonction de l'âge et des conditions expérimentales permettent de penser qu'elles sont étroitement attachées à la fonction glandulaire.
    Notes: Summary Electron microscopic investigation of the rat anterior pituitary reveals, throughout the whole pars distalis, a cellular network composed of agranular cells. These cells, the stellate and folliculo-stellate cells, are satellites of the granulated secretory cells. The cells of the network are characterized both by their multiple cytoplasmic processes and by their organization around cavities, most of them submicroscopic, the “follicles”. Those cells probably represent a large part of the “chromophobe” cells previously observed by light microscopy. On account of several characters, the pituitary cleft seems to be related to the “réseau folliculo-stellaire” and could be a peculiar part of this system because of its localization, size and contents. The “réseau folliculo-stellaire” is present in numerous species, showing a typical and differentiated cytology, and close relationships with the interstitial medium. Owing to its morphology and to modifications according to age and experimental conditions, this network seems to be a system intimately involved in the glandular function.

    Notes: Summary The ascidian myocardium is composed of small striated myoepithelial cells. The sarcoplasmic reticulum (SR) of these cells was reconstructed from serial sections. T-tubules are absent, but subsarcolemmal cisternae of the SR, that may be the counterpart of terminal cisternae, form couplings with the sarcolemma. Longitudinal SR tubules, parallel to the myofilaments, are interconnected near the middle of the A-band and form a transverse collar. Cinematographic photography of spontaneous contractions in fresh myocardial preparations produced records that could be analyzed frame by frame. Contractions are typically limited to parts of the myofilament field of single cells. They are locally symmetrical with respect to Z-bands; either both A-bands on each side of a Z-band converge on it (contractions), or neither A-band moves with respect to the Z-band. It is suggested that the spontaneous contractions are the result of local Ca++ release from randomly distributed subsarcolemmal cisternae. It is proposed that the symmetry of contraction is due to a rapid sequestering of Ca++ by the SR collars at the middle of the A-bands and a possible diffusion barrier at that level.

Ähnliche Befunde anderer Autoren und die eigenen Ergebnisse sprechen dafür, daß es sich um das Substrat einer weitverbreiteten Zellreaktion handelt, die u.a. das morphologische Äquivalent der Sequestrierung eines z.B. im Zuge der Zelldifferenzierung überflüssig gewordenen Zytoplasmaanteils darstellt.
    Notes: Summary In the endothelial, the media and mesenchymal cells of the vessels in the chicken's allantoic and vitelline membranes “multivesicular structure” regularly occur. They consist of aggregated vesicles and/or vacuoles (diameters ranging from 800 Å to 5000 Å) and seem to present a rather common and widespread cellular reaction, that might serve as a mechanism to sequester those parts of the cytoplasm which became superfluous e.g. during cell differentiation.

    Notes: Summary In the transitional zone of the enamel organ (rat) some of the amelocytes perish. Their debris is phagocytized and digested by stratum intermedium cells and macrophages. These two cell types also seem to remove cytosegresomes expelled from those amelocytes which survive and redifferentiate into transporting amelocytes. Digestion of the amelocyte debris in the stratum intermedium cells is effected rapidly and completely. Degeneration of stratum intermedium cells was not observed in the transitional zone.

    Notes: Summary Ultrastructural observations of the giant axon of Myxicola infundibulum reveal that the axoplasm contains neurofilaments, a few neurotubules and mitochondria. Finger-like projections issuing from the glial cells of the sheath encircle the giant axon at various angles. The space between the axolemma and sheath is 125 Å. Branches of the giant axon are also surrounded by a glial sheath as they course through the neuropil. Some branches of the giant axon seem to fuse with certain neurons, creating a syncytial arrangement between the giant axon and these neurons. Many small nerve fibers course longitudinally in the neuropil of the nerve cord. Most of these axons are separated from each other by a space of 200 Å without intervening glial processes. Synapses in the neuropil have both clear 600 Å vesicles and larger dense core vesicles suggesting chemical transmission. Some, but not all, of the synaptic areas show thickened membranes and dense material in the synaptic cleft.

Leur origine est parfois délicate à définir. Certains dictyosomes restent en relation constante avec les granules péricentriolaires et les deux anneaux différents, même lorsque ces derniers quittent la région acrosomienne. L'élaboration du cou enfoncé dans la profonde fossette nucléaire ne semble pas se faire à partir des centrioles. L'origine nucléaire de certaines protéines basiques qui le composent est envisagée en fonction de la concordance étroite entre sa formation et le remplacement des histones riches en lysine par les histones enrichies en arginine. Les différentes preuves morphologiques de passage au niveau de l'enveloppe nucléaire ont été recherchées. Les voies d'un éventuel transfert n'ont pu être clairement définies. La zone cervicale de la spermatide de pleurodèle est comparée à la région équivalente du spermatozoïde d'insecte et de mammifère. Certains éléments de la spermatide jeune présentent des caractères communs avec le corps chromatoïde des mammifères. Leur mode d'évolution est relativement semblable. Les spermatides d'urodèles semblent donc posséder un corps chromatoïde comme celles de la plupart des autres vertébrés.
    Notes: Summary The development of the different components of thePleurodeles spermatozoon neck region have been investigated and described from the early beginning spermiogenesis process. In some cases their origine is not well defined. Some dictyosomes remain in a constant relationship with the pericentriolar granule and the two different rings, even after they had left the acrosomal region. The development of the neck in a deep postnuclear niche, does not seem related with the centrioles. The nuclear origine of some basic proteins of the neck is discussed in relation to the close correspondence between their appearance and the substitution of the lysine rich histone by the arginine rich histone in the nuclear. The various morphologic signs of transit at the nuclear envelope level have been investigated. The ways along which some materials is eventually transferred out of the nucleus remained an open question. ThePleurodeles spermatid neck region is compared with the same region of the insect and mammalian sperm. Some components of thePleurodeles young spermatids display common characters with the mammalian chromatoid body. Their development are fairly similar. Thus the Urodele spermatid seems to posses a chromatoîd body like most other vertebrate spermatids.

    Notes: Summary Receptor cells in the epithelium and the basiepithelial nerve net of the prostomium of Lumbricus terrestris were investigated with electron microscope with special regard to the presence of monoamines. The receptor cells are found in groups of about 40 intermingled with supportive cells. After pretreatment with α-methyl-noradrenaline and fixation with potassium permanganate a few receptor cells in each group and some nerve fibres in the basiepithelial nerve net contain small granular vesicles (about400 Å) characteristic for monoaminergic neurons. The distribution and relative number of these receptor cells and nerve fibres coincide well with previous reports on fluorescent receptor cells and varicose fibres. That the monoamine-storing small granular vesicles not are visualized until pretreatment with α-methyl-noradrenaline is in accordance with recent microspectrofluorometric analysis, which shows that dopamine is the only primary monoamine present in the epithelium. In the epithelium there are occasional receptor cells and nerve fibres containing large vesicles (1000–1800 Å) which resemble the neurosecretory vesicles in the central nervous system. Photoreceptor cells having an intracellular cavity with microvilli and cilia have infrequently been observed at the base of the epithelium. No synapses on the mucous cells have been noticed. Nor have any synaptic specializations been observed in the basiepithelial nerve net. The morphological conditions necessary for the existence of possible axo-axonal synapses are briefly discussed.

    Notes: Summary This study is concerned with the organization of smooth surfaced canaliculi in fibers of regenerating sciatic nerves in a toad. Immediately proximal to the lesion the axons appeared crowded with widened canaliculi and vesicles. The derivation of these components was established through the analysis of isolated canaliculi in PTA stained preparations. In nerve sections 0.5 mm above the lesion most canaliculi are confined to the subsurface axoplasm. The amount of this subsurface reticulum appeares considerably reduced in axons profiles located 1 mm proximal to the lesion. Above this zone only small bundles of subsurface and axial canaliculi are found to occur in the regenerating fibers. The observations sustain the opinion that the accumulation of canaliculi at the end of the interrupted fibers is due to transport of this component by axoplasmic flow.

    Notes: Summary Electron opaque cross-bridge structures span the inter- and intracisternal spaces and provide membrane-to-membrane connections between adjacent cisternae of dictyosomes of pollen tubes ofClivia andLilium. Additionally, the classic intercisternal rods, characteristic of intercisternal regions near the maturing face of dictyosomes, are connected with the adjacent membranes through similar cross-bridge elements. We suggest that these structural links are responsible for maintaining the flattened appearance of the central parts of Golgi apparatus cisternac as well as for the coherence of cisternae within the stack. Observations on other plant (e.g. microsporocytes ofCanna) and animal cells (e.g. rodent liver and hepatoma cells, newt spermatocytes) show that such an array of membrane cross-links is a universal feature of Golgi apparatus architecture. The cross-bridges appear as part of the complex “zone of exclusion” which surrounds dictyosomes, entire Golgi apparatus and Golgi apparatus equivalents in a variety of cell types.

Die Pigmentzellen enthalten dicht gepackte Pigmentkörner, einen gelappten Kern, Mitochondrien vom Cristatyp, wenig endoplasmatisches Retikulum, Mikrotubuli, lamellated und vesicular bodies. Gegenüber den rhahdombildenden Anteilen der Sehzellen ist ihre Zelloberfläche glatt, im übrigen bilden sie lange fingerförmige Ausstülpungen. 3. Artemia salina besitzt zwei Arten von Frontalorganen. Die „dorsalen PO“ sind möglicherweise neurosekretorische X-Organe. Das ventrale FO wird als Sehorgan gedeutet. Es besteht aus zwei Gruppen von Sehzellen ventral vom MA, die eigene Nervenfortsätze zum Protocerebrum senden. Die optische Achse ist der des ventralen Augenbechers entgegengesetzt. Zwischen MA und FO verläuft ein Nerv, der wahrscheinlich dem MA angehört. 4. Übereinstimmungen der Feinstruktur von MA, FO und den Retinulazellen der Komplexaugen betreffen insbesondere die Binnenstruktur der Rhabdommikrovilli und Äquivalente unterschiedlicher Funktionszustände (Hell-Dunkel-Adaptation) hinsichtlich der Ausbildung von perirhabdomalen Vakuolen und des Ausmaßes von pinocytotischen Vorgängen an der Basis der Rhabdome.
    Notes: Summary The median eye (MA) and the ventral frontal organ (FO) of Artemia salina L. (adult specimens) have been investigated with the electron microscope. 1. The MA is an inverse cup-shaped eye. A pigment cup, consisting of two pigment cells, surrounds three groups of photosensory cells, which form ramified rhabdoms of the closed type. Their cytoplasm contains numerous vesicles, tubular mitochondria, small Golgi fields, microtubules, variable multivesicular and lamellated bodies and lipid inclusions, which are surrounded by spirals of endoplasmic reticulum. 2. The pigment cells contain densely packed pigment granules, an indented nucleus, crested mitochondria, small amounts of endoplasmic reticulum, microtubules, lamellated and vesicular bodies. Opposite the rhabdomeric surface of the visual cells their cellular surface is smooth, otherwise it bears long fingershaped projections. 3. Artemia salina possesses two types of frontal organs. The “dorsal FOs” are possibly neurosecretory X-organs. The ventral FO is interpreted to represent a photosensory organ. It consists of two groups of sensory cells located ventrally of the MA, which possess own nerve-processes leading to the Protocerebrum. Their optical axis is opposite the one of the ventral eye cup. Between MA and FO a nerve occurs, which presumably belongs to the MA. 4. Considerable finestructural similarities between MA, PO and the retinula cells of the compound eyes exist as far as the internal structure of the rhabdomeric microvilli and the equivalents of different functional stages (Light-Dark-Adaption) are concerned, namely perirhabdomeric vacuoles and the degree of pinocytotic processes at the base of the rhabdoms.

    Notes: Summary This is an electron microscopic study of non-fixed and non-dehydrated normal human stratum corneum from the lumbar region. Non-stained sections have a low contrast. In sections examined 3 days after skin biopsy the cytoplasm of the cells shows a uniform contrast or exhibits dark and light areas. A single layer delimits the cytoplasm from the intercellular space. The latter is partly filled out with substance. In sections stained 2 to 4 days after skin biopsy the fibrils are distinct. On the basis of the variations in their opacity and ultrastructure three types of horny cells are clearly distinguishable. In cells of type 1 intensely stained keratohyalin and less opaque fibrillar substance occur. A distinct keratin pattern is not found. In cells of type 2 the fibrils show areas with distinct kerytohyalin and keratin pattern and transitional phases between these two stages of fibrillar differentiation. The keratin pattern representing the final stage of the fibrillar differentiation process is visualized through a successive “discoloration” of the filaments, whereas the interfilamentous substance retains the opacity of the keratohyalin. In cells of type 3 the entire fibrillar substance exhibits a keratin pattern. This consists of less opaque filaments with a diameter of 74 Å. The septa representing the interfilamentous substance are estimated as 30 Å at their thinnest points. These observations of the fibrils are completely comparable to the findings in fixed and dehydrated normal human stratum corneum. In sections stained particularly more than 18 days after skin biopsy the fibrils exhibit pronounced changes in their staining properties with concomitant decrease in distinctness or a complete extinction of the keratin pattern. The observations of the modified plasma membrane and the intercellular space in stained sections correspond to the findings in fixed and dehydrated normal human stratum corneum. The modified plasma membrane and the structures in the intercellular space appear with equal distinctness, whether the sections are stained 2 to 4, 6 to 12 or 14 to 21 days after skin biopsy.

La porphyrinogénèse se développe dans des vacuoles et organites d'origine golgienne qui participent également à la mélanogénèse.
    Notes: Summary In Lineus ruber the biochemical differentiation of the new pigmentary cells is sequential during the reparative regeneration of the eyes. The enzymatic pathways for porphyrin and melanin biosynthesis appear successively. Ultrastructural and high resolution radioautography studies, show that melanization occurs in specialized organelles called premelanosomes, melanosomes and melanin granules. Prophyrogenesis occurs in Golgi vesicles which are also involved in melanogenesis.

Notre travail comporte une étude descriptive de ces cellules en microscopie optique et électronique, et une étude minéralogique abordée par des techniques de coloration, de dissolution, de polarisation de la lumière et de diffraction électronique. Les résultats obtenus montrent que les sphérules, par leur aspect, leur nombre, leur volume et leur état de cristallisation, présentent des caractères différents selon la saison, ce qui permet d'établir une relation entre l'état des nodules calcaires et les variations du métabolisme des individus.
    Notes: Summary In Ferrissia wautieri calcium is stored in the connective tissue in intracellular calcareous spherules. The cells have been studied light- and electronmicroscopically and a mineralogical analysis of the spherules was done by selective staining, dissolution, polarisation and electron diffraction. The results of these studies show that the aspect, the number, the volume and the rate of cristallisation of the calcospherites are different according to the seasons. Therefore it may be assumed that the characteristics of the spherules are connected with metabolic variations in these animals.

In conditions when host puparia are available, the calyx cells resemble typical secretory cells with rough endoplasmic reticulum, Golgi complex and mitochondria. After periods of host deprivation the calyx cells increase in size, the organelles change and become reorientated and cytolysomes appear producing a configuration of cells undergoing autophagy. 3. When host puparia become available again, the cells show an ability to recover and recommence production of secretory droplets.

    Notes: Summary The fine structure of the neural sheath, glial cells and nerve cells in the brain of adult male houseflies is described. The neural sheath is composed of neural lamella and perineurium. The neural lamella consists of an external lamina and collagen-like fibrils which are embedded in an amorphous matrix. The perineurial cells form a continuous layer around the brain. On their inner surface, perineurial cells form junctional complexes with glial cell processes. A cortical cellular layer composed of neurons and glial cells surrounds the centrally located neuropil. Three types of glial cells are identified. Glial cells differ in size and in relative development and distribution of organelles. Thin processes of glioplasm completely surround the cell bodies of the neurons. Five types of neurons are described. Most of the neurons are monopolar, a few are bipolar.

    Notes: Summary A morphologic and histochemical study was carried out on the liver of larval and adult lampreys at the optical and electron microscopic level. In the larva the liver is composed of blind ending single cell thick tubules of hepatocytes. The tubular lumina provided with microvilli are morphologically comparable with the canalicular lumens of the higher species of animals. The cytoplasm of the hepatocytes contains numerous inclusions with heterogeneous appearance and crystalline material. The biliary system is composed of numerous bile ductules and ducts. In the adult lamprey, the biliary system has disappeared. The hepatocytes loose their tubular arrangement and the characteristic differentiation of their biliary pole. In contrast to previous reports in the literature the presence of bile pigment in the adult lamprey liver could not be demonstrated with any histochemical technique.

Bei beiden Tierarten beträgt der Septalspalt nur 65 Å (E.f.) bzw. 75 Å (L.t.) und ist deshalb als „gap junction“ anzusprechen. Daneben fallen folgende Differenzierungen am Septum auf: „septate junctions” (L.t. und E.f.), „intermediate junctions“ (L.t. und E.f.), beidseitig am Septum gelegene und nicht von einer Membran umschlossene Membranappositionen (E.f.), sowie einseitig am Septum gelegene „dense projections“ (L.t.). Bei L.t. kommen außerdem auf beiden Seiten des Septums Vesikel vor, die von einer Elementarmembran umschlossen sind und nach Größe (φ ca. 575 Å), Lage, Haufenbildung und elektronenoptischem Habitus den Vesikeln chemisch übertragender Synapsen gleichen. Die Befunde werden hinsichtlich ihrer möglichen Bedeutung bei der elektrischen Übertragung und als Indizien für das Vorliegen einer gemischten Synapse diskutiert.
    Notes: Summary Electronmicroscopical examination of the dorsal giant fibers in the earthworms Lumbricus terrestris (L.t.) and Eisenia foetida (E.f.) reveals that their segmental septa are structures of so far unknown complexity. In both species the extracellular cleft between the two axon membranes of the septum amounts to only 65 Å (E.f.) and 75 Å (L.t.) respectively and is therefore regarded as “gap junction”. The following other structural differentiations of the septum were observed: “septate junctions” (E.f. and L.t.), “intermediate junetions” (E.f. and L.t.), densities apposed to both sides of the septum and not surrounded by a membrane (E.f.), and densities resembling “dense projections” on one side of the septum only (L.t.). In addition the septa of L.t. show vesicles on both sides which are bounded by a unit membrane and resemble the vesicles of chemically transmitting synapses in size (φ ca. 575 Å), location, accumulation, and electronoptical habitus. The significance of the findings in regard to electrotonic transmission and the possible existence of a mixed synapse is discussed.

    Notes: Summary Pictures, in front view, are presented of the nuclear pores from the oocytes of the newt Taricha granulosa. Negative staining is used. It is directly visible, on a substantial proportion of the pores, that the number of subunits in the annulus is 8. This conclusion had been reached earlier by other writers, who had used the rotation technique to ascertain the radial symmetry. Die Abgrenzung der Acinusspitze gegen das interacinäre Gewebe hin bildet die Basalmembran des wandständigen Begleitzellepithels. Die Oocyten werden follikelartig von den ineinander verzahnten und durch Desmosomen verknüpften Begleitzellen umgeben. Nur in der Acinuskuppe liegen sie der hier stark verdickten Basalmembran unmittelbar auf. Die Spermatiden sitzen nur mit ihrem anterioren Zellpol den Begleitzellen apikal auf und sind durch Desmosomen mit ihnen verknüpft. Veränderungen der Ultrastruktur der Spermatiden während der Spermiohistogenese werden an drei gegeneinander abgrenzbaren Spermiohistogenesestadien aufgezeigt. Dabei finden die Kernstruktur, das Auftreten von Tubulikörpern und das Abstreifen des Restplasmas vom Mittelstück besondere Beachtung. Den recht uneinheitlich strukturierten Begleitzellen kommen für Oocyten und Spermatiden Ernährungs- und Transportfunktionen zu. Sie phagocytieren überfällige Geschlechtszellen. Es können jedoch trotz ihrer heteromorphen Struktur keine prinzipiell verschiedenen Begleitzelltypen mit jeweils nur einer spezifischen Funktion unterschieden werden. Das in früheren lichtmikroskopischen Arbeiten als Begleitzellprodukt beschriebene „Kinoplasma“ erweist sich als kernwärts wanderndes Restplasma der Spermatiden.
    Notes: Summary Three species of cells always coexist in the tips of hermaphroditic gonad-acini of fertile Planorbarius corneus: oocytes, spermatids and auxiliary cells. The basement membrane of the auxiliary cell epithelium separates the acinus tips from the interacinary tissue. Like follicles the oocytes are enclosed by interlocked and desmosomically attached auxiliary cells. Only in the utmost tips of the acinus the oocytes are in direct contact with the here dilated basement membrane. The spermatids are attached to the auxiliary cells only with their anterior cell-pole and connected with these by desmosomes. Alterations of the spermatid-ultrastructure during the spermiohistogenesis can be studied in three separate stages of the spermiohistogenesis. Particular attention is given to the nuclear structure, the tubular bodies and the shedding of residual plasma from the middle-piece. The rather irregularly structured cells serve oocytes and spermatids as mediators for nutrition and transport. Occasionally occurs phagocytosis of germ-cells. Basically, even though their structures vary, auxiliary cells are not restricted to one specific function.     Notes: Summary The adenohypophysis of the newt Triturus cristatus carnifex Laur. shows three types of cells: 1) cells with granules of about 350–550 mμ in diameter, 2) cells with small granules of 200–250 mμ in diameter and globules with a cristal-like arrangement containing cylinders with a diameter of about 960 Å and 3) cells containing small granules only. The AA. discuss the ultrastructural changes of the gland and the modifications of sexual secondary characters (S.S.C.) in animals given Cyproterone acetate (1/2 mg every three days). The animals have been treated for a period of time varying between 3 and 5 months, starting in October-November, when S.S.C. begin to develop again. At the end of the treatment the newths showed a loss of S.S.C., and the ultrastructure of the adenohypophysis resembled that of castrated animals, i.e.: great swelling of R.E.R. and partial degranulation of glycoprotein secreting cells which contain the 200 mμ granules and the globules. The S.E.R. showed also swelling and hyperactivity.

    Notes: Summary It has been shown in previous studies that the endocrine control of molting is blocked by selective X-irradiation of the hemocytopoietic tissue of young last instar larvae of Locusta migratoria, the treated animals surviving as larvae for months without ever initiating a molt. The aim of the present study is to determine, on both a morphological and a physiological basis, if the block is related to a precocious functional involution of the prothoracic glands, known to elaborate a molting hormone. The prothoracic glands of normal animals clearly show morphological signs of functional involution during the late fifth (last) larval instar and degenerate completely some days after the imaginal molt. In the prothoracic glands of X-ray treated animals (selective irradiation of the hemocytopoietic tissue, the prothoracic glands being shielded), the signs of intense secretory activity persist up to 40 days following the irradiation. No signs of involution could ever be noticed in the prothoracic glands of such larvae. The irradiation of the hemocytopoietic tissue thus not only blocks molting, but also prevents the normal involution of the prothoracic glands. Extirpation of the prothoracic glands in young larvae of Locusta is known to result in the inhibition of the subsequent molt. Implantation of prothoracic glands from normal donors of the same age restores molting in fifth instar larvae previously deprived of their own glands. Using these results in physiological assays for the implanted prothoracic glands, we chose as donors irradiated animals of different ages and young normally developing adults. The prothoracic glands of irradiated animals fully retain their physiological activity after the X-ray treatment and are able to induce molting in larvae deprived of their own glands. It appears that the radiosensitive cells of the hemocytopoietic tissue of Locusta produce a substance which, while having no overt influence on the functioning of the prothoracic glands, is necessary for molting and probably acts in synergy with the prothoracic factor (molting hormone).

So läßt sich unter anderem durch Modifizierung der Fixationsmedien (Glutaraldehyd, Osmiumsäure in Kombination mit verschiedenen Puffern) eine optimale Differenzierung der zellulären Strukturelemente von Glomuszellen erreichen. An den osmiophilen Vesikeln fällt eine starke Variabilität ihres Erscheinungsbildes auf. Feinstrukturelle Unterschiede werden in Elektronendichte des Vesikelinhaltes, Größe und Ausbildung des Hofes sowie in Aufbau und Verlauf der Membran deutlich. Auf Grund dieser Kriterien lassen sich im wesentlichen 4 Haupttypen der Granula von seltener vorkommenden Sonderformen abgrenzen.
    Notes: Summary The organospecific structures of the carotid body and the adrenal medulla show a different appearance under various experimental conditions. Thus, it is possible to obtain an optimal differentiation of the cellular structure elements of the carotid body cells by modifying the fixatives (glutaraldehyde, osmic acid, in combination with various buffer systems). Especially the osmiophilic vesicles show a great variability in their appearance. Ultrastructural differences become evident in the electron-density of the vesicle content, in the size of the electron-lucent space (halo of the dense core), as well as in the structure and conduct of the membrane. Based on these criteria four main types of granules can be said to occur in the carotid body. These can be distinguished from more uncommon special types.

    Notes: Summary Cytoplasmic filaments have been observed in the cells of normal and pathological kidneys. These filaments are usually grouped into bundles anchored to electron dense bodies underlying the cell membrane. In the embryonic human metanephros the filaments are found within the cells of different portions of the nephron at various stages of development. They appear first in the podocytes, almost simultaneously in the Bowman's capsule and tubular cells, then in the “mesangial cells”, and finally in the cells of the media of the afferent glomerular and interlobular arterioles. The presence of filaments and their attachment bodies in the mammalian nephron suggests that the podocytes and the so-called mesangial cells have a contractile activity, thus representing an intraglomerular apparatus which regulates the intravascular pressure, blood flow and filtration rate in the glomerular capillaries, whilst the contractile activity of the Bowman's capsule and proximal, distal, and collecting tubules, could facilitate the progression of the filtrate. The increase in number of the filaments in some pathological conditions is probably related to the functional changes of the intraluminal pressure in the glomerular capillaries, in the Bowman's space, and in the tubular lumen.

    Notes: Summary Umbilical vessels of guinea-pig fetuses were studied shortly before birth. In all umbilical cords investigated an innervation of the umbilical vessels is lacking. The intrafetal parts of the umbilical vessels on the other hand are richly innervated. A marked difference in the amount of nerve fibres and the pattern of innervation is found between artery and vein. The artery is supplied by a dense nerve plexus which spins around the media and which originates from nerve bundles within the outer adventitial layers. The comparatively scanty innervation of the vein exhibits a more coarsely meshed net pattern. The nerve bundles in the vein exhibit a close affinity to the vasa vasorum. Number and type of the close contacts between the muscle cells are different in the various sections of the umbilical vessels. Similar to the distribution of nerves they are almost absent in the vessels of the umbilical cord, numerously, however, in the intrafetal parts. Contrary to the innervation, the close contacts in the vein are developed more numerously and more broadly than in the corresponding artery.

    Notes: Summary Nuclear bag and nuclear chain intrafusal fibres are present in guinea-pig muscle spindles. Unlike muscle spindles in other species two types of nuclear chain fibre seem to be present. The electron microscopical appearance of one type of nuclear chain fibre is similar to that of nuclear bag fibres. It is suggested that under tension the nuclei of small nuclear bag fibres become sufficiently displaced to form nuclear chain-like fibres. The frequent occurrence of fibres which combine some of the properties of both nuclear bag and nuclear chain fibres indicates the possible occurrence of a third type of intrafusal fibre. The sensory innervation of guinea-pig muscle spindles is similar to that of the cat and the rat. Three types of motor nerve ending which could be classified according to the complexity of their subneural apparatus were seen.

Untersucht wurden Tiere vom 12.–90. Lebenstag. Folgende Ergebnisse sind neu: Weder bei jungen noch bei erwachsenen Ratten kommt im Hypoglossuskern Mikroglia vor. Etwa ab. 3. Tag nach Axotomie kann bei erwachsenen Tieren Mikroglia nachgewiesen werden. Sie entsteht aus aktivierten und proliferierten Oligodendrocyten und pericytenähnlichen Bindegewebszellen. Die Mikroglia umhüllt die Nervenzellen. Eine Aktivierung und Hypertrophie von Astrocyten erfolgt etwa ab 8. Tag nach Axotomie. Von diesem Tage an drängen sich Astrocytenfortsätze zwischen Mikroglia und Nervenzelloberfläche. Die Mikroglia beginnt zu degenerieren. Bei Jungtieren bewirkt die Axotomie nur geringgradige Veränderungen. Schon beim Normaltier findet man aktive Oligodendroglia und Astrocyten sowie häufig pericytenähnliche Bindegewebszellen.
    Notes: Summary The ultrastructure of the Nucleus hypoglossi in normal albino rats and following axotomy is described with special reference to the glia. Animals 12 to 90 days old were investigated. The following results are new: Normally in young and adult rats microglia is absent in the nucleus. Following axotomy from the 3rd day no microglia can be demonstrated in adult rats. It originates from activated and proliferated oligodendrocytes and pericyte-like cells of the vascular connective tissue. These cells surround the neurons. An activation and hypertrophy of astrocytes is visible from the 8th postoperative day on. Astrocyte processes make contacts with the perikarya and separate the microglia from the neurons. Microglia cells then show signs of degeneration. In young animals alterations following axotomy are only sparse. Already normally activated oligodendrocytes and astrocytes as well as pericyte-like connective tissue cells can be seen.

    Notes: Summary Vinblastine sulfate was administered to adult rats by intravenous injections. Kidney cortex was fixed after 1, 2, or 5 hours of treatment and studied by routine transmission electron microscopy. In control animals, cells of distal convoluted tubules possessed numerous microtubules with an average diameter of 280 Å. In treated animals, the microtubules of these cells were reduced in number, and paracrystalline inclusions characteristic of vinblastine treatment were common. Macrotubules (570 Å average diameter) were also present and often were seen close to, or in apparent continuity with, paracrystals. Since the work of others indicates that vinblastine-induced paracrystals contain microtubular protein (tubulin), observation of continuities between paracrystals and macrotubules is interpreted as evidence that macrotubules are also composed of tubulin and that macrotubules may become incorporated into paracrystals. Unlike the ordinary microtubules of cells of the distal tubules, vinblastine-induced macrotubules exhibited cross-striations in longitudinal view and subunit structure in cross section. Macrotubules and paracrystals were also observed in cells of the proximal convoluted tubule, mesangium, glomerular endothelium, parietal epithelium of Bowman's capsule, and visceral epithelium of Bowman's capsule. Continuities between macrotubules and paracrystals, although relatively common in occurrence in distal tubule cells, were only rarely seen in the other kinds of cells examined.

    Notes: Summary Morphological evidence is presented supporting the possibility of basal secretion into hypendymal capillaries of the adult rabbit subcommissural organ (SCO). The synthetic apparatus of the SCO cell is described as well as the heterogeneous granules and vesicles which are concentrated in the basal processes bordering a widened perivascular space. The origin of the electron dense granules, of which two fairly distinct subgroups are found, is discussed. A binding of secretory sacs to the lateral plasma membrane is seen. The possibility of a lateral secretion is supported by the presence of a system of extracellular channels between SCO cells which are filled with a flocculent material resembling that of the secretory sacs. Nerve perikarya which are separated from the SCO by only a few glial fibers are demonstrated. Synapses are described in nerve fascicles bordering on the hypendymal capillaries. The possibility of an innervation of the hypendymal region is discussed as well as possible nervous connections with the pineal gland.

Die Zellen werden nicht durch desmosomale Bildungen untereinander verbunden. In den oft geweiteten Interzellularlakunen findet sich feinstflockiges Material. Spezialzellen normal ernährter Schnecken besitzen ein außerordentlich reiches, geordnetes, rauhes endoplasmatisches Retikulum und einen großen Kern mit großem Nucleolus. Golgi- und Vakuolenapparat der Zellen bleiben klein. Mit zunehmender Dauer des Fastens wird das rauhe ER durch ungeordnetes, glattes endoplasmatisches Retikulum ersetzt, wächst der Golgiapparat und nimmt die Zahl der Autolysosomen zu. Die feinstrukturellen Unterschiede zwischen Spezialzellen, Blasenzellen und Nervenzellen werden beschrieben, die Funktion der Spezialzellen abschließend diskutiert.
    Notes: Summary “Cellules spéciales” of Aelidoidea have been investigated in the cerata of Eubranchus farrani and Trinchesia granosa. All groups of “cellules spéciales” are limited by a basal lamina. Opposing membranes are not connected by specialized structures as desmosomes etc. The intercellular cleft often is widened and contains a fine floccular substance. In normally nourished animals the “cellules spéciales” show a rich, highly organized, rough surfaced reticulum, while there are only a few vacuoles and a small Golgi Complex. In starved animals mainly smooth reticulum is found, but the Golgi Apparatus has enlarged and autolysosomes are abundant. The fine structural differences of “cellules spéciales”, globular cells and nerve cells are described, the function of the “cellules spéciales” is discussed.

In den Monaten November bis Februar kommen im Zytoplasma der Leydigzellen kleine Mitochondrien des tubulären Typs, Vesikel des glatten endoplasmatischen Retikulum und freie Ribosomen vor, Fettvakuolen treten nur vereinzelt auf, der Golgikomplex ist gering entwickelt. In der Zeit von März bis Juni nimmt die Zahl der Zellorganellen zu. Es erscheinen in Gruppen angeordnete Lysosomen und außer den kleinen Mitochondrien Riesenmitochondrien, des Golgifeld ist größer, die Fettvakuolen sind vermehrt. Sowohl die kleinen als auch die Riesenmitochondrien enthalten Kristalle, die aus osmiophilen globulären Einschlußkörpern hervorzugehen scheinen. In den Monaten Juni/Juli ist fast der ganze Zelleib einer Leydigzelle von Fettvakuolen ausgefüllt. Die Zahl der Riesenmitochondrien und Lysosomen verringert sich, das Golgifeld ist wieder unauffällig. Eine Beziehung zwischen der Vermehrung und Entfaltung der Zellorganellen und der in der Literatur angegebenen Phase der gesteigerten Steroidproduktion konnte festgestellt werden. Das Auftreten der Riesenmitochondrien und Lysosomen und ihre Bedeutung für die saisonbedingte Entwicklung und Involution der Leydigzellen wird diskutiert.
    Notes: Summary Seasonal changes in the structure of the testicular interstitial cells in Rana esculenta were investigated. Throughout November–February, small mitochondria of the tubular type, visicles of the smooth endoplasmic reticulum and free ribosomes predominate and the cytoplasm contains few lipid droplets and a poorly developed Golgi apparatus. During the period from March to June the cell organelles increase in number. There are numerous aggregated lysosomes and conspicuous accumulations of lipid droplets. Besides small mitochondria increasing numbers of giant mitochondria occur and the Golgi complex is enlarged. The small and giant mitochondria contain crystals which seem to originate from osmiophilic globular inclusion bodies. During June and July the whole cytoplasm may be filled with lipid droplets. Concomitantly there is a decrease in the number of lysosomes and giant mitochondria and the Golgi apparatus is in a state of involution. Possible correlations between the development of the cell organelles and the period of increased steroid production are pointed out. The appearance of giant mitochondria and lysosomes and their importance for seasonal Leydig cell development and involution are discussed.

    Notes: Summary Numerous morphological continuities between the cisternae of the convex face of the Golgi apparatus and the granular endoplasmic reticulum (GER) were observed in post-division amebae that had divided following enucleation and renucleation. Electron microscopic radioautography with the use of 3H-uridine as a tracer indicated that perhaps the Golgi apparatus is derived from the GER. The possibility of the connections between GER and Golgi apparatus facilitating transport of materials between the two is also discussed.

    Notes: Summary In the enamel organ of rat incisors macrophages are present in the zone of matrix formation, the transitional zone, the enamel maturation and pigmentation zone. The macrophages accumulate adjacent to redifferentiating amelocytes in the transitional zone. The macrophages phagocytize fragments of disintegrating amelocytes. In addition to the well known complement of organelles the macrophages present an elaborated microtubular system, scattered, thick filaments, a cortical feltwork of thin filaments, and spherical nuclear bodies. The microtubules emanate from “attached” and free pericentriolar satellites and radiate aster-like towards the cell surface or into pseudopods or curve along the nuclear surface for long distances, often related to nuclear constrictions. It is suggested that the microtubular system plays a prominent role in directional movement of the macrophages. The cortical filaments, if contractile, may create the cytoplasmic flow necessary for the cell motility.

    Description / Table of Contents: Zusammenfassung Speicheldrüsen von Mückenlarven (Chironomus Thummi) haben eine ausgedehnte interzelluläre Kommunikation. In elektronenmikroskopischen Bildern der Speicheldrüsen wurden zwei Arten von interzellulären Verbindungen gefunden, die für die Zellkopplung verantwortlich sein könnten: septate junctions und close membrane junctions. Da die räumliche Ausdehnung der septate junctions viel größer zu sein scheint als die der close junctions, wurden erstere als wahrscheinliche Kopplungsstrukturen angesehen. Es gibt jedoch Hinweise, daß die Strukturen, welche die Zellkopplung bewirken, sehr labil sind. Unter den Faktoren, die zu einer Unterbrechung der zellulären Kommunikationen führen können, sind auch osmotische Effekte. Um mögliche Einflüsse dieser Art auf die Ultrastruktur zu verhindern, wurden die Drüsen für die mikroskopische Inspektion in isoosmotischen Lösungen fixiert. Unter diesen Bedingungen lassen sich ausgedehnte Membrankontakte vom nexus-Typ zwischen den Drüsenzellen erkennen. Ihre Ausdehnung scheint ebenso groß zu sein wie die der septate junctions. Es besteht nach diesen Befunden die Möglichkeit, daß wie in anderen kommunizierenden Zellsystemen, so auch in Speicheldrüsen die interzelluläre Kommunikation durch nexus bewirkt wird.
    Notes: Summary Cells ofChironomus salivary glands communicate through intercellular connections of high permeability. Electron micrographs of salivary glands show two kinds of junctions between the membranes of adjacent cells, which may be responsible for cell coupling: septate junctions and close membrane junctions. A large fraction of lateral cell surfaces is occupied by septate junctions, while the area of close membrane junctions appears to be very small. Consequently septate junctions have been considered as likely sites for intercellular coupling. There are however some indications that intercellular communication is provided by structures which seem to be unstable. As osmotic effects are among the factors which can disrupt cellular communications, we have tried to eliminate possible effects of the fixing solutions on the ultrastructure of intercellular connections by using isoosmotic fixatives. Under these conditions large regions of close membrane junctions of the nexus kind have been observed to occur between gland cells. They are of similar size as septate junctions.     Notes: Summary Four types of striated muscle fibers with distinctive ultrastructure were defined in the Atlantic hagfish (Myxine glutinosa, L.): “white”, “intermediate”, and “red” fibers of m. parietalis, and “red” fibers of m. craniovelaris. “White” fibers are thick, contain very few mitochondria and fat vacuoles, and possess distinct and separate myofibrils with thin Z-disks and distinct M-lines. “Intermediate” fibers are thinner, possess largely similar myofibrils that often are even better separated due to a higher content of fat vacuoles and especially mitochondria and glycogen granules. “Red” fibers of m. parietalis contain large amounts of mitochondria, fat vacuoles, and glycogen granules. Their myofibrils possess M-lines, and although branching more, the myofibrils of “red” fibers conform with a “Fibrillenstruktur” pattern like those of “white” and “intermediate” fibers. “Red” fibers of m. craniovelaris are very thin, possess many smaller fat vacuoles, and large amounts of mitochondria and glycogen granules. The myofibrils are significantly thinner than in m. parietalis fibers, run as quite independent well separated units, possess thicker Z-disks, and lack M-lines. Large amounts of myosatellite cells are associated with these “red” fibers. Triads are located near A/I-junctions in all four fiber types and occur irregularly, the density of triads being different in the various fiber types.

Diese Myofibrillenabschnitte weisen unterschiedliche Längen auf. Am kleinsten terminalen Myofibrillenabschnitt ist die Ribosomenkonzentration am höchsten. Mit zunehmender Verlängerung des Abschnittes erkennt man Aktinfilamente, die zwischen den dicht gelagerten Ribosomen liegen. Erreicht der terminale Myofibrillenabschnitt etwa die Länge einer Sarkomere, hat die Ribosomenkonzentration abgenommen, die Filamentanzahl zugenommen. In diesem Stadium treten zwischen den Aktinfilamenten erste Myosinfilamente auf. Eine neue Z-Scheibe entsteht in engem Kontakt mit dem Sarkolemm. Letztere löst sich nach und nach über ihre ganze Länge von der stets schräg ansetzenden Zellmembran. Die Frage nach der Spannungsübertragung an undifferenzierten terminalen Myofibrillenabschnitten wird diskutiert.
    Notes: Summary Longitudinal growth of myofibrils in the skeletal muscle of Macropodus opercularis appears to take place at their terminal parts. Since the Z-disks are arranged in lines and perpendicularly to the longitudinal axis, there are repeated terminal myofibril regions of different lengths following the last complete sarcomere. At the shortest terminal myofibril region, which is apparently the youngest one, ribosome concentration is very high. In the adjacent terminal regions of greater length, which probably represent older ones, actin filaments can be detected among the ribosomes. As soon as the terminal myofibril region approaches the full sarcomere length, the concentration of ribosomes is found to be reduced and the number of filaments increased. At this stage the first myosin filaments are clearly observed among actin filaments. Therefore, during longitudinal growth myosin filaments morphologically appear after the formation of actin filaments, whereas during myofibrillogenesis in the same muscle both types appear simultaneously in the differentiating sarcomere. After the arrangement of actin and myosin filaments into the A- and I-bands, a new Z-disk is formed in close contact with the sarcolemma and gradually detached over its entire length from the inclined cell membrane. The problem of tension transmission via the undifferentiated terminal myofibril regions is discussed in relation to these findings.

    Notes: Summary The planula larva of the solitary coral Balanophyllia regia has an ectoderm of flagellate, diplosomal collar cells. The collar of these cells is composed of a ring of microvilli linked with mucus strands. Four types of flagellate gland cells, three types of nematocyst and spirocysts are present in the planula ectoderm. The function of these ectoderm cells is discussed. The mesogloeal muscular and packing tissues of the planula are briefly described. The tentacle of the adult coral, examined for comparison, has an ectoderm of flattened flagellate cells with a shallow collar. Collar cells similar to those of the planula are occasionally found on the tentacle and their function is not known. Independent sensory cells built on a modified collar cell plan with collar of thickened microvilli are common in the tentacle. These are quite separate from the three kinds of tentacular nematocyte. Distended glandular areas occur in the tentacle ectoderm. The flagellate tentacle gastrodermis, muscle and mesogloeal region are briefly described. The evolutionary significance of collar cell ectoderm in a planula is discussed and the occurrence of collar cells throughout the animal kingdom, reviewed.

    Notes: Summary Osteoclasts from the tibial metaphyses of young rats treated with porcine calcitonin were studied by electron microscopy. The animals were sacrificed 1 1/2, 4, 8 or 12 hours after injection of the hormone. In survey sections examined by light microscopy the osteoclasts appeared smaller than in control animals. At the ultrastructural level the osteoclasts showed the following alterations: 1) The typical ruffled border was absent. 2) Acid phosphatase was not present in the extracellular space between cell and bone. 3) The number of large vacuoles was decreased and there was no local accumulation of vacuoles in the cytoplasm. 4) The vacuoles did not contain bone crystals. 5) Vacuoles with cell organelles were increased in number. The majority of these vacuoles were identified as autolysosomes because they contained acid phosphatase and the enclosed cell organelles were partially digested. The above changes were present at all time intervals studied. The findings suggest that calcitonin decreases or inhibits bone resorption by osteoclasts. A decreased function of the osteoclasts may contribute to the hypocalcemic effect of the hormone. The increased number of autolysosomes is evidence of an enhanced autophagocytosis. Possible origins of the autolysosomes in osteoclasts are discussed.

Vergleichend-anatomisch gehören die fluoreszierenden Areale des Rückenmarks zu den autonomen Zentren. Es wurde versucht, mit Pharmaka, die das autonome Nervensystem beeinflussen oder Krämpfe erzeugen, das Glykogen des Lumbalwulstes zu mobilisieren (Quantitative Glykogenbestimmung nach Krisman, 1962). Besondere Beachtung verdient die elektronenmikroskopische Feinstruktur der Zwickelräume zwischen den großen Glykogenkörperzellen, wo Ausläufer dieser Zellen und Nervenfasern eng beieinander liegen. Die Kapillaren des Glykogenkörpers werden fast vollständig von Ausläufern seiner Zellen umscheidet.
    Notes: Summary With the method of Falck-Hillarp bundles of fluorescent nerve fibers were observed in the vicinity of the avian lumbar glycogen body. They encompass the glycogen body laterally. Numerous fluorescent fibers penetrate from this border zone into the glycogen. The fluorescent regions of the avian lumbar spinal cord belong to the autonomic centers. Attempts were made to decrease the high glycogen content of the glycogen body by administering drugs which act on the autonomic system, and also by using convulsant drugs. Glycogen was estimated quantitatively by the Krisman method (1962). Electron micrographs show that the triangular spaces between the enlarged cells of the glycogen body are occupied by the processes of these cells and by nerve fibers. The capillaries of the glycogen body are nearly completely ensheathed by the processes of the glycogen-body cells.

    Notes: Summary Colchicine was intraperitoneally administered chronically to adult male Sprague-Dawley rats. The ultrastructural study of hind-limb muscles revealed that myofilament desorientation resulted. Bundles of myofilaments were found coursing perpendicular or oblique to the longitudinal axis of the muscle fiber. It is concluded that a colchicine-sensitive factor is involved in maintaining normal orientation of myofibrils in mature muscle. Also found in the sarcoplasm of the colchicine treated animals were complex spheromembranous bodies. These bodies enveloped mitochondria or other organelles and appeared to be derived from the sarcoplasmic reticulum. The lysosomal nature of these bodies is indicated by the localization of acid phosphatase activity in them. Acid phosphatase activity was also displayed in the sarcoplasmic reticulum. The spheromembranous bodies seem to be part of a sarcotubulo-lysosomal system in skeletal muscle.

    Notes: Summary In the tortoise Testudo graeca, the lizards Lacerta dugesi and Lacerta pityusensis, and the snake Natrix natrix, the innervation of the testicular interstitial tissue was studied by light and electron microscopy, the acetylcholinesterase (ache) technique, the Falck-Hillarp method for the detection of catecholamines, and the application of 6-hydroxydopamine. The intertubular spaces of the reptilian testes studied contain adrenergic nerve fibers the amount and distribution of which varies considerably both in various species and in various stages of the reproduction cycle. Nerve fibers do not enter the seminiferous epithelium. Fluorescence microscopy of the lizard testis reveals catecholaminergic varicosities which are mainly arranged around blood vessels, but do not show obvious connexions to Leydig cells. Ache-positive fibers are equally distributed in lizard testes surrounding each seminiferous tubule. In Natrix natrix ache-positive fibers are irregularly spread among groups of tubules, without showing a definite relation to Leydig cells either. By electron microscopy bundles of unmyelinated axons and axon terminals can be more easily detected in the testes of immature animals than in adult. Terminals of nerve fibers containing small (400–500 Å in diameter) and large (800–1400 Å) dense-cored vesicles and sometimes small clear vesicles establish contacts with Leydig cells. Three types of contact are described. 1. “Contacts” par distance at a distance of about 2000 Å and basal lamina interposed; 2. membranous contacts having a 200 Å gap only between axolemma and Leydig cell plasmalemma; 3. invaginations of terminals into Leydig cell perikarya. The latter may exhibit surface specialisations, which strongly resemble postsynaptic membrane thickenings. Experiments using 6-hydroxydopamine underline the adrenergic character of testicular nerve fibers, which can be regarded as another example of non-cholinergic, ache-positive neurons. In the testis of the immature tortoise profiles of axons occur which probably represent purinergic, ache-positive neurons.

    Notes: Summary The principles of stereology have been applied to a stereometric analysis of the ultrastructural composition of normal rat peritoneal macrophages and of cells stimulated five days previously by a single intraperitoneal dose of Freund's adjuvants. Material obtained by a systematic random sampling regime was analysed and the data groups for various morphological parameters were compared. Estimates were made of cell numbers. From electron micrographs the volume proportions of nuclei, mitochondria, heterogeneous granules and rough endoplasmic reticulum were determined. The average numbers and dimensions of mitochondria, granules and free ribosomes were also evaluated. The volume-to-surface ratios of cells and their nuclei were computed. By the independent measurement of cellular volumes it was possible to obtain estimates of nuclear and cytoplasmic volumes, to evaluate the membrane surface areas of the average cells and their nuclei, and to calculate numbers of organelles per average cell. The structural alterations induced by the complete adjuvant were similar to those evoked by the incomplete adjuvant. Stimulation was characterized by a cellular hypertrophy accompanied by an increase in mean granule size and in the number of mitochondria per average cell. At the same time, there was a substantial decrease in the number of granules and a considerable depletion of both nuclear and plasma membrane surface areas.

    Notes: Summary The excretory organs of Amphioxus occur as segmentally arranged structures throughout the pharyngeal region and may be divided into three components: the solenocytes, the renal tubule, and the renal glomerulus. The solenocytes possess foot processes that rest upon the coelomic surface of the ligamentum denticulatum. The tubular apparatus of the solenocytes consists of ten triangular rods surrounding a central flagellum. The distal end of the tubular apparatus enters branches of the renal tubule. The renal tubule eventually opens into the atrial cavity of Amphioxus. The renal glomerulus is a sinus within the connective tissue of the ligamentum dentieculatum where it connects elements of the branchial circulation with the dorsal aorta. The renal glomerulus, like other blood vessels of Amphioxus, lacks an endothelial lining. If Amphioxus is adapted to artificial sea water at different concentrations there is no change in kidney morphology suggesting that Amphioxus is either is osmotic with its environment or is osmoregulating with other organs.

Ultrastructurally the cells were pale, contained less organelles as compared with the controls, loss of cytoplasm and reduction both of cell height and width. The major changes also included reduction of the Golgi area, the rough endoplasmic reticulum and the number of free ribosomes. Within the dorsal lobe prominent nuclear changes were found, comprising different types of intranuclear inclusions of membranous configurations. The alterations observed in the present study are similar to the changes which were found in castrated animals and are consistent with atrophy. The results are discussed in relation to the available biochemical and autoradiographic data on the different lobes.

    Notes: Summary The effects of penicillin and chlortetracyline HCl on the fine structure of the intracellular symbiotes of the pea aphid were studied in an attempt to remove the symbiote population. High penicillin concentrations, 1% and 0.1%, caused symbiote breakdown but were toxic and/or repellent to the aphids; at 0.1% specific effects were observed on the symbiotes' cell walls. After the use of 0.01% penicillin in the aphid diet, the symbiotes had abnormal cell walls and were abnormally dilated; however, symbiote division and transmission from one aphid generation to the next seemed unaffected and the aphids appeared normal. Aphids fed 0.1% chlortetracycline failed to reproduce. After 7 days, their symbiotes were found to break down at a high rate but aphid mitochondria were also adversely affected at this stage. Following 0.002% chlortetracycline, the aphids produced aposymbiotic progeny with apparently normal mitochondrial populations; these larvae failed to develop.

    Notes: Summary Electron microscopy and autoradiography (H3-proline) were used to determine the cell(s) responsible for collagen synthesis and lamination in the mesoglea of the sea anemone, Aiptasia diaphana. Mesogleal collagen is synthesized by the epidermal epitheliomuscular cells which contain much rough endoplasmic reticulum, and secretory vesicles attached to microtubules which cross the basal plasmalemma and anchor in the basement membrane; these cells incorporate large amounts of H3-proline. The mesogleal collagen fibers are non-striated, have clear centers, and a diameter of 200–260 Å; their walls are composed of 65 Å diameter subunit fibrils which appear to be helically oriented. Epitheliomuscular cells rest upon a subepidermal basement membrane which is composed of mesogleal collagen fibers, 65 Å subunit fibrils, and dense granules. This subepidermal basement membrane labels definitively with H3-proline, and is the region where soluble collagen precursors apparently form subunit fibrils which associate to yield mesogleal fibers. The columnar mesogleal collagen fibers are arranged to form layers: the fibers of each layer have the same longitudinal orientation, while those of adjacent layers display an approximate orthogonal arrangement. It is felt that the subepidermal basement membrane is responsible for this organization of mesogleal fibers. Mesogleal amoebocytes do not label with H3-proline and show no ultrastructural evidence of collagen secretion.

    Notes: Summary The spermatids of Campodea C. remyi Denis are clumped in small bundles of 236 to 650 elements during spermiogenesis, their acrosomes being embedded within an homogeneous electron dense material. The spermatid comprises an acrosomal region and a postacrosomal region within which, the two mitochondrial derivatives and the nucleus are arranged in parallel. The acrosome is rather long, it is reinforced by seven to ten microtubules. The axoneme is of the 9+9+ pattern. The mitochondria are of almost equal length. The shorter nucleus shows a very electron dense chromatin. At mid-nuclear level the cytoplasmic membrane is folded up to form a sort of collar opening backwards. Distal to the nucleus, superimposed lamellar structures are found located in an expanded part of the spermatid.

    Notes: Summary The ultrastructure of the nucleus of human Sertoli cells has been studied in testicular samples from normal adults and patients with hypospermatogenesis or absence of the germinal cell line (Sertoli cell only syndrome). In all cases the ultrastructure of the nuclear structures was alike. The nucleus is irregularly shaped and its membrane presents numerous pores. The inner aspect of the nuclear membrane is electron dense due to the presence of a fibrous lamina. The outer aspect presents a concentric arrangement of 50 Å cytoplasmic filaments. The chromatin is rather dispersed. Some perichromatin granules are found occasionally. The nucleolus is characteristic of human Sertoli cells and displays three clear components. In addition, sphaeridia of at least two principal morphological types were found. Their significance in the cell metabolism as well as relationship to either the nucleolus or the nuclear DNA are discussed. The need for further investigation on the functional meaning of the nuclear organelles of human Sertoli cells is stressed.

Sie haben überwiegend kugelige Gestalt und liegen kernnah zwischen Kern und apikalem Plasmalemm. Feinstrukturell sind sie erstaunlicherweise nicht einheitlich aufgebaut, sondern treten in zwei Typen auf: Typ I besteht aus einer relativ kompakt erscheinenden kugeligen Ansammlung von Mitochondrien. Die Mitochondrien liegen in einer dichten, teils granulären, teils fibrillären intermitochondrialen Substanz eingebettet. Typ II besteht aus einer ebenfalls kugeligen Ansammlung vieler kleiner radiär angeordneter Golgi-Stapel, welche in der gleichen dichten, teils granulären, teils fibrillären Substanz liegen wie die Mitochondrien von Typ I. Die Funktion beider Typen von Dotterkernen wird diskutiert. Sicherlich haben sie nichts mehr mit der Dottersynthese zu tun wie möglicherweise die Dotterkerne der Oocyten, doch gibt es bisher auch keine Anzeichen dafür, daß sie bevorzugt am Dotter- oder Lipidabbau beteiligt sind.
    Notes: Summary In all blastomeres of Nassarius from 4- to 16-cell stage yolk nuclei occur. Most of them are spherical bodies, lying juxtanuclearly between the nucleus and the apical plasmalemma. Strangely they are not ultrastructurally uniform but fall into two categories (Fig. 5): Type I is a massive spherical accumulation of mitochondria embedded in a dense intermitochondrial substance, which appears to contain both granules and filaments. Type II is a ball of radially arranged small Golgi stacks clustered around a centre of Golgi vesicles and other organelles embedded in a ground cytoplasm structurally similar to the intermitochondrial substance of type I. The function of both types of yolk nuclei is unknown. These segmentation yolk nuclei have nothing to do with yolk synthesis any more. On the other hand there are no indications that yolk nuclei occurrence is correlated with the break-down of yolk because neither lipid droplets nor protein yolk granules are observed in or beside the yolk nuclei.

    Notes: Summary The possibility of a direct retinohypothalamic projection was reinvestigated in Passer domesticus by electron microscopy following left unilateral retinectomy. To avoid misinterpretation of non-specific degeneration, the course of degenerative changes was observed at intervals of 6, 12, 24, 48 and 96 hours after operation. Of the hypothalamic areas examined in only one, the contralateral suprachiasmatic nucleus, was it possible to identify reliable indications of secondary anterograde degeneration comparable to those observed in the contralateral optic tectum. Single dark profiles within the supraoptic nucleus and the basal infundibular (tuberal) nucleus showed neither internal changes in structure nor an increase in number per unit area in retinectomized birds. Since photoperiodically induced gonadal growth occurs in totally blinded birds the functional significance of the retinohypothalamic projection to the suprachiasmatic nucleus is open to discussion.

    Notes: Summary Major ultrastructural changes in neurons were studied during sequential periods of hypostomal regeneration in Hydra. Some neurons remain unaffected except that at certain periods following amputation, they become more active in neurosecretory production. Other neurons in various stages of differentiation were also observed. Most emphasis was placed on degenerating neurons showing a loss of organelles and destruction of the perikarya. Certain large, membrane-bounded structures (up to 1.7 μ in diameter) suggested tentatively to be of a lysosomal-like nature, may be partly responsible for the degenerative process. The neurites of these cells first assume a beaded appearance and/or contain bulbous endings. The eventual isolated fragments of neurites contain typical membrane-bounded neurosecretory droplets (850/1700 Å in diameter) which disintegrate forming particulate materials (350 Å in diameter). Following complete disruption of the neurites, some of the granules accumulate in the extracellular spaces before they are disintegrated. From the data presented in this and the preceding paper, it is suggested that these particles, derived from neurosecretory droplets, may be responsible for the stimulation of: 1) interstitial cell differentiation into neurons, 2) the increased activity in neurosecretory production in normal cells, 3) the conspicuous increase in neurite length and consequently the exaggerated degree of anastomosis, and 4) the possible capacity of neurons to regenerate neurites.

    Notes: Summary Dimethylaminoethyl p-chlorophenoxy acetate (80 mg/kg body weight) was administered (i. m.) to guinea pigs for 30 to 56 days. Electron microscopic examination of the hippocampus, mid-brain reticular formation and the area postrema revealed marked diminution in the electron density of the pigment granules and vacuolization. This type of lipofuscin was detected in some phagocytic cells and in the capillary endothelium. Conspicuous vacuolization of the capillary wall was discernible.     Notes: Summary The caudal musculature of the free-swimming tadpole of the ascidian, B. schlosseri consists of cylindrical mononucleated cells connected in longitudinal rows flanking the axial notochord. During resorption of the larval tail, which is apparently induced by the contraction of the epidermis, muscle cells are dissociated and pushed into the body cavity where most of them are rapidly engulfed by phagocytes. In the initial stages of tail withdrawal muscle cells display surface alterations due to the disruption of intercellular junctions and disarrangement of myofibrils. Extensive degenerative changes, with shrinkage of mitochondria and disintegration of the contractile material are subsequently observed. Lysosomes and autophagic vacuoles are rarely seen and appear to play a secondary role in the degradation of the muscle cells, which occurs predominantly within the phagocytes. Myofilaments and myofibrils have never been observed within autophagic vacuoles. Clumps of muscle fragments and degenerated phagocytes undergo eventual dissolution in the blood lacunae, concomitantly with the differentiation of the young oozooid.

    Description / Table of Contents: Zusammenfassung Die quantitativ-morphologische Analyse des juxtaglomerulären Apparates nach DOC-Behandlung zeigte, daß mit der Hemmung der Reninbildung eine Hypoplasie der epitheloiden Zellen des juxtaglomerulären Apparates einhergeht. Der Granulagehalt der juxtaglomerulären Zellen nahm ab; die Kern-Plasma-Relation änderte sich und die Leiomyofibrillen in den Epitheloidzellen nahmen zu. An den übrigen Anteilen des juxtaglomerulären Apparates, insbesondere der Macula densa, wurden keine morphologischen Veränderungen beobachtet. Die DOC-Behandlung führte darüber hinaus zu einer Bildung von Einschlüssen in den Podocyten, den Mesangiumzellen und den Zellen der Bowman-Membran. Die verschiedenen Möglichkeiten, über welche Faktoren die juxtaglomerulären Zellen nach DOC-Behandlung degranuliert werden, werden diskutiert. Das ultrastrukturelle Bild der Epitheloidzellen nach DOC-Behandlung entspricht deutlich einer gehemmten Sekretion, da das Ergastoplasma und der Golgi-Apparat ganz besonders stark zurückgebildet werden.
    Notes: Summary Quantitative morphological analysis of the Juxtaglomerular apparatus of rats treated with deoxycorticosterone (DOC) and salt revealed hypoplasia of the epithelioid cells as a concomitant of the inhibition of renin synthesis. The granular content of the Juxtaglomerular cells was significantly decreased, while the nucleus-to-cytoplasm ratio and the volume density of leiomyofilaments in the epithelioid cells increased. No morphological alterations were observed in the remaining constituents of the Juxtaglomerular apparatus, including the macula densa. DOC-treatment also induced the appearance of inclusions in the podocytes in the cells of the mesangium, and in the cells of Bowman's capsule. The possible mechanisms of degranulation of the Juxtaglomerular cells after treatment with DOC are discussed. The ultrastructural appearance of the epithelioid cells after treatment with DOC corresponds to an inhibition of secretion, since the rough endoplasmic reticulum and the Golgi apparatus undergo marked regressive changes.

    Notes: Summary A major function of the larval salivary glands of Drosophila melanogaster is known to be the production of a mucopolysaccharide that serves as an adhesive during puparium formation. In order to localize the mucosubstances during development substrate histochemical methods were used, and the site of acid phosphatase was demonstrated by the ultrahistochemical lead-salt method. It could be shown that the “glue”-granules in the corpus cells of larval salivary glands as well as the large secretion vacuoles in the prepupal corpus cells give a positive β-amylase-resistent PAS-reaction, which indicates neutral mucosubstances. Granular PAS-positive deposits in the larval and prepupal collum cells were reduced after preincubation with β-amylase and may represent glycogen, which has also been seen in electron micrographs of these cells. The Hale-reaction gave a weak indication that acid mucosubstances are present in the larval “glue” granules and in the large prepupal secretory vacuoles. After digestion of sialic acid with α-neuraminidase the weak indication was absent showing that the acid mucosubstances had been sialomucines. Ultrahistochemical demonstration of acid phosphatase indicated the presence of this enzyme in Golgi fields and lysosomal structures. Acid phosphatase seems to be missing in the large secretion vacuoles of the prepupal salivary gland. It is concluded, that the large vacuoles in the corpus cells of prepupal salivary glands represent a secretion product, obviously a mucosubstance. The lysosomal structures, containing acid phosphatase, may be accumulated in preparation for the autolysis of the gland which begins about two hours after the pupal moult, i.e. 15 hours after puparium formation.
