    Notes: Abstract Speed of nerve impulse conduction is greatly increased by myelin, a multi-layered membranous sheath surrounding axons. Myelinated axons are ubiquitous among the vertebrates, but relatively rare among invertebrates. Electron microscopy of calanoid copepods using rapid cryofixation techniques revealed the widespread presence of myelinated axons. Myelin sheaths of up to 60 layers were found around both sensory and motor axons of the first antenna and interneurons of the ventral nerve cord. Except at nodes, individual lamellae appeared to be continuous and circular, without seams, as opposed to the spiral structure of vertebrate and annelid myelin. The highly organized myelin was characterized by the complete exclusion of cytoplasm from the intracellular spaces of the cell generating it. In regions of compaction, extracytoplasmic space was also eliminated. Focal or fenestration nodes, rather than circumferential ones, were locally common. Myelin lamellae terminated in stepwise fashion at these nodes, appearing to fuse with the axolemma or adjacent myelin lamellae. As with vertebrate myelin, copepod sheaths are designed to minimize both resistive and capacitive current flow through the internodal membrane, greatly speeding nerve impulse conduction. Copepod myelin differs from that of any other group described, while sharing features of every group.

    Notes: Abstract The medullary pacemaker nucleus of Hypopomus triggers each electric organ discharge (EOD) by a single command pulse. It consists of electrotonically coupled ‘pacemaker’ cells, which generate the rhythm, and ‘relay’ cells, which follow the pacemaker cells and excite the spinal motoneurons of the electric organ. The pacemaker cells receive two inputs from the complex of the diencephalic prepacemaker nucleus (PPn), a GABA-ergic inhibition and a glutamatergic excitation. Relay cells, on the other hand, receive two glutamatergic inputs, one from a subnucleus of the PPn, the PPn-C, and a second from the sublemniscal prepacemaker nucleus (SPPn). We have labelled afferents to the pacemaker nucleus by injecting HRP to specific sites of the prepacemaker complex. By using immunogold-labelled antibodies and en-grid staining techniques, we demonstrated GABA and glutamate immunoreactivity in labelled synaptic profiles of ultra-thin sections of the pacemaker nucleus. The two types of synapses were interspersed on the surfaces of pacemaker cells, with GABA-immunoreactive synapses apparently representing the GABA-mediated input of the ‘PPn-I’, an inhibitory subdivision of the PPn, and glutamate-immunoreactive synapses representing the input of the ‘PPn-G’, an excitatory subdivision of the PPn. Only glutamate-immunoreactive synapses were found on relay cells.

    Notes: Summary The mantle edge of the freshwater pulmonate snailsLymnaea stagnalis andBiomphalaria pfeifferi was investigated with histochemical and ultrastructural methods. The mantle edge gland, which is involved in shell formation, consists of the periostracal groove and the belt. This belt appears to be composed of various regions. In the area of the periostracal groove a number of subepithelial gland cell types occur; these release their products into the groove. Between the groove cells ciliated free nerve endings terminate; the corresponding perikarya occur in the subepidermal connective tissue. Also in the posterior belt region free nerve endings were observed between the epithelial cells; in addition, a particular type of subepithelial gland cell was found in this area. The epithelial cells of this part of the belt have the ultrastructural characteristics of ion and water transporting cells; they are probably involved in calcium deposition and resorption. The possible role of the free nerve endings and of the subepithelial gland cells is discussed.

    Notes: Summary Using transmission and scanning electron microscopy, we have studied the ultrastructure of a number of urinary calculi, mainly composed of calcium phosphate. Three fundamental kinds of calcium phosphates were detected: nonstoichiometric carbonate apatite, nonhexagonal octacalcium phosphate, and calcium-magnesium whitlockite. The influence that the organic matter, substitutions in the phosphate lattice of CO3 and Mg, and apatitic stoichiometry have on the ultrastructure of the calcium phosphate calculi has been detailed. An originating apatitic unity named U2 is assumed to be the responsible for all the different structures of calcium apatites appearing in renal calculi. On the basis of our observations, a mechanism whereby apatites grow is postulated; magnesium functions as an inhibitor for the growing mechanism.

    Notes: Summary An original method for fractionating and preparing isolated crystals of homogeneous size was developed. It was demonstrated that enamel apatite crystals are at least 100 µm long. The flexibility of the very long crystallites was demonstrated. Crystal curvatures, accounting for the irregular course of the prisms through the enamel thickness, were visualized and measured. It was shown that in the deep forming enamel layer, lateral branches may grow out of the crystals and crystal fusing often occurs, inducing the crystallites to assume pyramidal shapes with their wide bases pointing toward the dentino-enamel junction and one or two tops toward Tomes' processes. During the maturation process, the two tops of the still immature crystals also fuse so that the mature crystals acquire a rodlike aspect, with parallel faces and steplike graduations along thec axis, allowing a close contact between the crystals. These results support the hypothesis that the crystallites would be continuous from the dentino-enamel junction to the surface.

    Description / Table of Contents: Résumé Une méthode nouvelle de décalcification histologique utilisant des solutions aqueuses de sulfate de chrome basique (III) est appliquée à des coupes fines de dentine humaine adulte. L'observation en microscopie électronique montre une bonne conservation structurale dans les régions extra-cellulaires. L'aspect de la matrice péricanaliculaire est particulièrement étudié. On y reconnait plusieurs couches bien individualisées. Une bonne concordance est notée entre les résultats obtenus par microscopie optique et électronique, permettant d'établir une hypothèse nouvelle sur la formation de la dentine péricanaliculaire.
    Abstract: Zusammenfassung Eine kürzlich entwickelte Methode zur histologischen Entkalkung mittels wäßriger Lösungen von basischem Chrom III-Sulfat wird an dünnen Schliffen von menschlichem Dentin Erwachsener angewandt. Darauffolgende Untersuchungen am Elektronenmikroskop zeigten eine sehr gute Wiedergabe der morphologischen Einzelheiten in den Extrazellulär-Regionen. Besonders beachtet wurde das Aussehen der peritubulären Matrix, innerhalb welcher gut differenzierte Schichten erkannt werden konnten. Eine wesentliche Übereinstimmung bestand zwischen Elektronenmikroskopie und kürzlich durchgeführten Untersuchungen an derselben Matric unter dem Mikroskop. Diese Methode ermöglicht es, eine neue Hypothese über die Bildung des peritubulären Dentins aufzustellen.
    Notes: Abstract A recently-developed method for histological decalcification using aqueous solutions of basic chromium (III) sulphate has been applied to thin sections of adult human dentine. Subsequent studies in the electron microscope show a very good retention of morphological detail in the extracellular regions. Special attention was given to the appearance of the peritubular matrix within which well-differentiated layers were recognised. There was substantial agreement between electron microscopy and recent studies of the same matric under the light microscope, allowing a novel hypothesis for the formation of the peritubular dentine.

Le matériel composant ce dernier provient de granules de sécrétion, élaborées au niveau de l'appareil de Golgi. Lorsque les granules golgiennes de la glande ventrale, contenant une substance, présentant une périodicité, déversent leur sécrétion en surface, en contact avec l'eau de mer, ce produit se disperse en particules, incluses dans un substrat. La formation du periostracum externe s'accompagne d'une réagrégation des particules sécrétoires golgiennes en une couche mince, présentant une structure périodique de 300 Å. En coupe transversale, le périostracum présente une structure régulièrement agencée, suggérant une nature cristalline. La couche externe atteint une épaisseur de 4–5 microns. La couche interne provient de granules de sécrétion de la glande dorsale. La formation de cette couche est identique à celle de la couche externe; cependant aucune périodicité n'y est visible. A l'état adulte, elle atteint une épaisseur de 0.4–0.5 micron. Outre son rôle de protection, le périostracum constitue une barrière entre l'eau de mer et l'espace pallial. Il est responsable, en outre, du dépôt et de l'orientation de cristaux inorganiques au niveau de la zone de développement de la carapace.
    Abstract: Zusammenfassung Eine dorsale und eine ventrale Drüse, die aus großen, kolbenförmigen Zellen bestehen und am Rande des Mantels vonLittorina gelagert sind, bewirken die Bildung der inneren und äußeren Schicht des Periostracums. Das entstandene Material, Periostracum inbegriffen, stammt von sekretorischen Granula, die vom Golgi-Apparat gebildet werden. Die Golgi-Granula bestehen aus einer Substanz, welche eine bestimmte Periodizität aufweist. Wenn nun die Golgi-Granula der ventralen Drüse an der Drüsenoberfläche erscheinen und mit Meerwasser in Kontakt kommen, sind sie weit verteilt und setzen sich aus Partikeln, die in ein Substrat eingebettet sind, zusammen. Die Bildung des äußeren Periostracums hat eine erneute Aggregation der sekretorischen Golgi-Partikeln zu einem dichten Blatt zur Folge, welches eine Periodizität von 300 Å zeigt. Betrachtet man das Periostracum in einem transversalen Schnitt, so findet man eine Gitterstruktur, die an eine kristalline Substanz denken läßt. Die äußere Schicht erreicht schließlich eine Dicke von 4–5 μ. Die innere Schicht entsteht durch die sekretorischen Granula der dorsalen Drüse. Die Bildung der inneren Schicht findet in ähnlicher Weise wie jene der äußeren statt, zeigt jedoch keine Periodizität. Im reifen Zustand erreicht sie eine Dicke von 0,4–0,5 μ. Zusätzlich zur Schutzfunktion bildet das Periostracum eine Schranke zwischen dem Meerwasser und dem Pallialraum; es reguliert zudem die Lage und die Anordnung der anorganischen Kristallbildung am Wachstumsrand der Muschel.
    Notes: Abstract A dorsal and ventral gland composed of large, flask-shaped cells located in the margin of the mantle ofLittorina give rise to the inner and outer layers of the periostracum respectively. The material comprising the periostracum is derived from secretory granules elaborated by the Golgi apparatus. When the Golgi granules of the ventral gland which consist of a substance exhibiting a definite periodicity, are discharged at the surface in contact with sea water, they are widely dispersed and consist of particles embedded in a substrate. Formation of the outer periostracum involves the re-aggregation of the Golgi secretory particles into a dense sheet which exhibits a periodicity of 300 Å. Viewed in transverse section the periostracum exhibits a lattice pattern suggestive of a crystalline substance. The outer layer eventually reaches a thickness of 4–5 μ. The inner layer is derived from the secretory granules of the dorsal gland. The formation of the inner layer occurs in a manner similar to that of the outer layer. It does not, however, exhibit a periodicity. In the mature state it attains a thickness of 0.4–0.5 μ. In addition to a protective function the periostracum provides a barrier between the sea water and the pallial space and also regulates the site and arrangement of mineral crystal formation at the growing margin of the shell.

La partie basale de la cellule présente de nombreux prolongements ramifiés. Ces prolongements contiennent de nombreuses mitochondries, ainsi que des invaginations de la membrane formant des vésicules qui migrent dans diverses parties de la cellule. Le noyau irrégulier est en position centrale. La région supranucléaire contient surtout de l'ergastoplasme, plusieurs appareils de Golgi et de nombreuses granules provenant de l'appareil de Golgi. Ces granules augmentent de taille et sont rejetées dans le sinus de la glande, où elles se modifient et se calcifient pour donner des sphérolithes. Des radioautographics glandulaires, obtenues 1/2 heure après injection de Ca45, montrent une localisation isotopique, située presque exclusivement dans les cellules; 4–24 heures après, l'isotope se localise dans les sphérolithes de la cavité glandulaire. Ces cellules semblent assurer deux fonctions liées à la production de sphérolithes: 1. absorption et transport de calcium de la région basale vers la surface cellulaire et 2. formation d'une matrice protéique qui se calcifiera.
    Abstract: Zusammenfassung Die aktivsezernierenden Zellen der calcifizierenden Drüsen vonLumbricus terrestris sind annähernd kuppelförmig gestaltet. Der untere Teil dieser Zellen besteht aus unzähligen ineinandergreifenden Zellfortsätzen. Diese Fortsätze enthalten zahlreiche Mitochondrien und membranöse Einstülpungen, welche Bläschen bilden, die zu den verschiedenen Regionen der Zelle wandern. Der Zellkern ist unregelmäßig und zentral angeordnet. Die supranukleäre Region ist zu einem großen Teil mit grobem endoplasmatischem Reticulum und mehreren Golgi-Komplexen sowie zahlreichen Granula, die von den Golgi-Komplexen stammen, ausgefüllt. Diese Granula vergrößern sich und werden in den Drüseninus ausgestoßen, wo sie schließlich reorganisiert und mineralisiert werden, um Sphärolithen zu bilden. 1/2 Std nach Injektion von Ca45 wurde die Drüse entfernt; eine Radioautographie zeigte, daß das Isotop beinahe ausschließlich in den Zellen lokalisiert war; 4–24 Std später befand sich das Isotopin den Sphärolithen der Drüsenhöhlung. Diese Zellen sind anscheinend an zwei für die Bildung von mineralisierten Sphärolithen notwendigen Funktionen beteiligt: 1. an der Calcium-Absorption und deren Transport von der Basalregion bis zur Zelloberfläche; 2. an der Bildung einer Proteinmatrix, in welcher die Mineralisation stattfindet.
    Notes: Abstract The actively-secreting cells of the calciferous glands ofLumbricus terrestris are roughly dome shaped. The basal part of the cell consists of numerous interdigitating cell processes. These processes contain numerous mitochondria and membranous infoldings, forming vesicles which migrate to various regions of the cell. The nucleus is irregular and centrally located. The supranuclear region is occupied to a great extent by the rough endoplasmic reticulum, several Golgi complexes and numerous granules derived from the Golgi complexes. These granules enlarge and are extruded into the gland sinus, where they eventually become reorganized and mineralized to give rise to spheroliths. Radioautographs of glands removed 30 min following injection of45Ca showed uptake of the isotope located almost exclusively in the cells; 4–24 h later the isotope was located in the spheroliths in the gland cavity. These cells apparently perform two functions necessary for the production of mineralized spheroliths, 1, absorption and transport of calcium from the basal region to the cell surface and 2, the elaboration of a protein matrix in which mineralization occurs.

    Notes: Summary The purposes of this study were to determine whether periodontal ligament (PDL) cells are capable of producing mineralized nodules in vitro and to analyze ultrastructural features of the nodules. Rat PDL cells were obtained from coagulum in the socket at 2 days after tooth extraction and cultured at confluence in standard medium containing Dulbecco's Modified Eagle's Medium supplemented with 10% FBS and antibiotics. To test mineralized nodule formation, cells were further cultured for an additional 3 weeks in the standard medium containing (1) ascorbic acid (50 μg/ml) and sodium β-glycerophosphate (10 mM), (2) ascorbic acid, sodium β-glycerophosphate, and dexamethasone (5 μM), or (3) ascorbic acid alone. Cells were then fixed in 2.5% glutaraldehyde, postfixed in 1% OsO4, and prepared for light and electron microscopy. Threedimensional nodules containing mineralized matrices were formed only when the cells were cultured in the presence of ascorbic acid and dexamethasone. They were composed of multilayered fibroblasts (up to 13 layers), and highly organized collagen fibrils with 64 nm cross-banding patterns between the cell layers. The fibroblasts in the nodules exhibited an elongated shape with a high degree of cytoplasmic polarity throughout the nodule, and have the morphological features of PDL fibroblasts as seen in vivo. Mineral deposition with needle-like crystals was initiated on collagen fibrils located in intercellular spaces of the upper cell layers and became increasingly heavier towards the bottom half of the nodules. X-ray microanalysis and electron diffraction analysis confirmed that mineral deposition contained calcium and phosphate in the form of immature hydroxyapatite. These nodules contained neither osteoblasts nor osteocytes, and have their own morphological organization and characteristics which differ from those formed by bone cells in culture. Therefore, these data suggest that PDL cells are capable of forming mineralized tissue in vitro with the morphological characteristics different from bone mineralized nodules.

    Notes: Abstract Rat bone cells were cultured in the presence of bioactive glass-ceramic containing crystalline apatite and wollaston te. Scanning electron microscopy observations of the surface of the seeded ceramic disks revealed that cells attached, spread, and proliferated on the material surface. Soaking in cell-free culture medium showed that no change occurred in the surface structure. However, when cultured with bone cells and observed under a transmission electron microscope, an electron-dense layer was noted initially at the surface of the material, before bone formation occurred. In addition, energy-dispersive X-ray microanalysis demonstrated the presence of calcium and phosphorus in this layer. Progressively, during the following days of culture, active osteoblasts synthetized and laid down an osteoid matrix composed of numerous collagen fibrils arranged either parallel or perpendicularly to the first-formed electron-dense layer. Mineralization initiated on the ceramic surface dispersed then along the collagenous fibrils, leading to a mineralized matrix which surrounded the ceramic particles. These results demonstrate the capacity of apatite-wollastonite glass ceramic to initiate biomineralization in osteoblast cultures and to achieve a direct bond between the surface apatite layer of the bioactive glass-ceramic and the mineralized bone matrix.

    Description / Table of Contents: Résumé Des cals expérimentaux de neuf jours, formés au niveau de radius de jeunes rats, sont traités par la méthode calcium-cobalt de Gomori (1939) pour la mise en évidence ultrastructurale de la phosphatase alcaline afin d'étudier son rôle éventuel dans le dépôt du calcium. L'activité enzymatique apparait initialement sous forme de précipités globulaires en dehors de la membrane cellulaire de jeunes chondroblastes hypertrophiques. Ce précipité donne ensuite naissance à des corps sphériques de phosphatase alcaline qui se forme près de la cellule. Ces corps sphériques s'observent dans une zone intermédiaire plus éloignée. Une formation de cristaux en aiguilles (apparemment une calcification) se développe dans des corps isolés ou agrégés, laissant voir nettement leurs limites, même lorsque la calcification est plus avancée au point qu'on ne peut plus distinguer des cristaux individuels. Au niveau des coupes témoins, traitées de façon identique mais sans substrat ou avec de l'E.D.T.A., on n'observe ni précipité enzymatique ou corps sphériques. L'aspect des dépôts cristallins dans des corps qui contiennent de la phosphatase alcaline ne peut s'expliquer que par l'existence d'une association étroite entre enzymes et calcification.
    Abstract: Zusammenfassung Neun Tage alter experimenteller Kallus an Radii von jungen Ratten wurde mit Gomori's (1939) Calcium-Kobalt Methode untersucht, um die Verteilung der alkalischen Phosphatase und ihre Beziehung zur Calciumablagerung ultrastrukturell zu demonstrieren. Enzymaktivität zeigte sich zuerst als globulares Präzipitat außerhalb der Zellmembran von Knorpelzellen im Beginn der Hypertrophie. Aus dieser Präzipitatschicht entstanden dann gerundete Körperchen, die sich von der Zelle abtrennten. Solche Körperchen wurden auch in größerer Entfernung von der Zelle beobachtet, d.h. in einer Zwischenzone zwischen benachbarten Zellen. Nadelförmige Kristalle, wahrscheinlich von Calcium-Salzen, wurden in einzelnen oder aggregierten Körperchen beobachtet. Die äußere Zone der Körperchen blieb jedoch deutlich sichtbar, selbst dann, wenn der Calciumgehalt derart zugenommen hatte, daß einzelne Kristalle nicht länger erkennbar waren. In Kontrollen, die in gleicher Weise behandelt waren, aber ohne Substrat oder mit Zufügung von EDTA, wurden weder Präzipitate noch Körperchen beobachtet. Das Auftreten von Calciumablagerungen in alkalischer Phosphatase enthaltenden Körperchen scheint kaum anders erklärbar als durch eine enge funktionelle Verbindung zwischen Enzym und Calciumablagerung.
    Notes: Abstract Nine day old experimental calluses in radii of young rats were treated with Gomori's (1939) calcium-cobalt method to demonstrate ultrastructurally the presence of alkaline phosphatase in a search for its possible role in the desposition of calcium. Enzyme activity first appeared as globule-like precipitates outside the cell membrane of early hypertrophic cartilage cells. This precipitate layer then seemed to give rise to spherical bodies of alkaline phosphatase which occur at a slight distance from the cell. The spherical bodies were also observed further away from the cell in an intermediate zone between neighboring cells. Needle-like crystal formation, apparently calcification, occurred inside single or aggregated bodies, leaving their peripheral rim clearly visible, even when calcification had increased to such an extent that individual crystals could no longer be recognised. In controls, treated in the same way but without substrate, or with EDTA, no enzyme precipitate or spherical bodies were seen. The appearance of crystalline deposits in bodies which contain alkaline phosphatase seems difficult to explain on any other basis than that there is a close functional association between the enzyme and calcification.

    Notes: Summary The ultrastructural localization and gradient of activity of alkaline phosphatase were studied with respect to cell differentiation, matrix synthesis, and matrix mineralization in the incisor and molar teeth of 4-day-old Sprague-Dawley rats. The animals were perfused intracardially at room temperature with 2.5% glutaraldehyde in 0.1M sodium cacodylate (pH 7.4) with 3–4% sucrose. The jaws were dissected, immersion-fixed for 24 h, and the incisor and molar tooth germs removed. These were demineralized in 10% EDTA in NaOH (pH 7.4) with 7% sucrose. After reactivation of the enzyme with 0.1M MgCl in Tris-maleate buffer (pH 7.4) at 4°C, the teeth were incubated for alkaline phosphatase in a medium consisting of 6 ml 3% sodiumβ-glycerophosphate, 4 ml 0.2M Tris-HCl buffer (pH 9.2), 3 ml 1.6% MgSO4, 12 ml 0.5% lead citrate (pH⋍12), and 2.1 g sucrose. The pH was adjusted to 9.2 with 0.2M HCl, the volume made up to 30 ml, and the solution centrifuged for 10 min at 5000 rpm. Control teeth were incubated in medium minus the substrate. Finally, the specimens were routinely post-fixed and embedded for sectioning and examination with a Philips 300 electron microscope. A gradient of alkaline phosphatase activity was mapped along the developing teeth in the cells of the stratum intermedium, the proximal borders of the ameloblasts, the early dentine matrix, the predentine-dentine border, matrix vesicles, and the plasma membranes of odontoblasts and subodontoblast cells. The gradient of alkaline phosphatase activity was evident in the forming tooth from the cervical loop to the crown apex and was related to the cellular events, matrix synthesis, and matrix mineralization occurring during odontogenesis.

This technique revealed the presence of an organic ‘crystal ghost’ associated with each crystallite. The effectiveness of chromium sulphate as a demineralizing agent is discussed.

    Notes: Abstract Transmission electron micrographs of fully mineralized turkey leg tendon in cross-section show the ultrastructure to be more complex than has been previously described. The mineral is divided into two regions. Needlelike-appearing crystallites fill the extrafibrillar volume whereas only platelike crystallites are found within the fibrils. When the speciment is tilted through a large angle, some of the needlelike-appearing crystallites are replaced by platelets, suggesting that the needlelike crystallites are platelets viewed on edge. If so, these platelets have their broad face roughly parallel to the fibril surface and thereby the fibril axis, where the intrafibrillar platelets are steeply inclined to the fibril axis. The projection of the intrafibrillar platelets is perpendicular to the fibril axis. The extrafibrillar volume is at least 60% of the total, the fibrils occupying 40%. More of the mineral appears to be extrafibrillar than within the fibrils. Micrographs of the mineralized tendon in thickness show both needlelike-appearing and platelet crystallites. Stereoscopic views show that the needlelike-appearing crystallites do not have a preferred orientation. From the two-dimensional Fourier transform of a selected area of the cross-sectional image, the platelike crystallites have an average dimension of 58 nm. The needlelike-appearing crystallites have an average thickness of 7 nm. The maximum length is at least 90 nm. Atomic force microscopy (AFM) of unstained, unmineralized turkey leg tendon shows collagen fibrils very much like shadow replicas of collagen in electron micrographs. AFM images of the mineralized tendon show only an occasional fibril. Mineral crystallites are not visible. Because the collagen is within the fibrils, the extrafibrillar mineral must be embedded in noncollagenous organic matter. When the tissue is demineralized, the collagen fibrils are exposed. The structure as revealed by the two modalities is a composite material in which each component is itself a composite. Determination of the properties of the mineralized tendon from the properties of its elements is more difficult than considering the tendon to be just mineral-filled collagen.

Sie bestehen bei transversalen Schnitten aus toten Gängen sowie beim Lumen der Tubuli entweder aus ringförmigen oder ganzausfüllenden Ablagerungen. Im äußeren Dentin wurden keine Nervenendigungen beobachtet.
    Notes: Abstract An ultrastructural study of peripheral human coronal dentin in patients aged 11 to 75 years, has shown main aspects of the dentinal tubular content. In transverse sections, they consist of dead tracts and annular or solid content to the tubular lumen. No nerve endings were observed in the outer dentin.

Des témoins, ayant reçu une dose équivalente de liquide de dilution, sont sacrifiés à des intervalles de 1 heure à 6 jours, identiques aux temps de sacrifice des animaux injectés. Les cartilages épiphysaires supérieurs des tibias tibias (ECP) étudiés en microscopie électronique, montrent, dès trois heures après l'ionjection, une augmentation nette de 'activié sécrétoire, caractérisée, au niveau de la zone de sécrétion matricielle, par l'abondance dans les citernes golgiennes d'un matériel piqueté, constitué par des complexes protéino-polysaccharidiques. La désintégration de la membrane limitante de vésicules golgiennes individuelles est plus avancée après vingt quatre heures: après trois jours de traitement, seules quelques vésicules restent intactes et des plages d'un matériel initialement intravacuolaire sont visibles dans le cytoplasme. De longs filaments, rappelant les précurseurs ou les fibrilles primaires du collagène, sont visibles dans cette sécrétion. Après six jours, de grandes plages de cettre subestance remplissent les cellules de la couche pré-hypertrophieque, avec déplacement de l'ergastoplasme en périphérie. Des vacuoles cytoplasques, contenant un matériel semblable à celui qu'on retrouve dans la lacune, et présentant des filament finement moniliformes et disposés en rayons le long de la membrane limitante, sont visibles. Ces observations suggèrent une accélération initiale de l'activité sécrétoire chondrocytaire, suivie par un retard de transfert. La rétention consécutive et la polymérisation intracellulaire de produits précollagéniques accélèrent l'hypertrophie et favorisent ainsi la dégénérescence précoce des chondrocytes. Ces altérations ultrastructurales paraissent être spécifiques aux oestrog`enes.
    Abstract: Zusammenfassung Zehn männliche Hotlzmann-Ratten, die im Entwöhnungsstadium waren, erhielten täglich wässerige Oestradioldosen (Progynon, Schering) von 1 μ/g Körpergewicht i.p. Dann wurden sie gleichzeitig mit Kontrolltieren, welche die gleiche Menge Verdünnungsmittel erhalten hatten, in Intervallen von 1 Std bis zu 6 Tagen getötet. Platten des oberen tibialen Epiphysenknorples (ECP), welche für die Elektronenmikroskopie präpariert wurden, zeigtem, daß schon 3 Std nach der Injektion ein bemerkenswerte Erhöhung der sekretorischen Tätigkeit entsteht. Dies wurde in der Zone der Matrixausscheidung sichtbar, wo sich in den Golgi-Zisternen eine Anhäufung von punktiertem, aus Proteinpolysaccharid-Komplexen bestehendem Material zeigte. Der Zerfall der Membran, welche die einzelnen Golgi-Bläschen umgibt, nahm nach 24 Std zu; nach 3 Tagen Behandlung blieben nur wenige Gefäße intakt, und Ansammlungen von ursprünglich intravacuolörem Material konnten im Grundplasma beobachtet werden. Lange Fasern, welche auf primäre oder Prae-Kollagefibrillen hindeuteten, konnten in diesem Sekret gesehen werden. Nach 6 Tagen wurden die Zellen in der prähypertrophen Zone mit dieser Substanz richtiggehend überschwemmt, und das rauhe endoplasmatische Reticulum wurde anschließend gegen die Zellperipherie verlagert. Die oft beobachteten cytoplasmatischen Vacuolen enthielten ein Material, das dem in den Lacunen vorkommenden ähnlich ist und zeigten auf der ungebrenden Membran feinperlige, radial angeordnete Fasern. Unsere Beobachtungen deuten auf eine anfängliche Beschleuning der chondrocytischen sekretorischen Tätigkeit, mit nachfolgender Transportverlangsamung, hin. Die dadurch entstehende Retention und intrazelluläre Polymerisation von präkollagenen Produkten beschleunigt die Hypertrophie und begünstigt dadurch die frühe Degeneration von Chondrocyten. Diese ultrastrukturellen Veränderungen scheinen oestrogen-spezifisch zu sein.
    Notes: Abstract Ten male weanling Holtzman rats, injected intraperitoneally with aqueous estradiol (Progynon, Schering), in daily doses of 1 μg. per g body weight, were sacrificed, simultaneously with controls receiving an equivalent amount of diluent, at intervals ranging from one hour to six days. Upper tibial epiphyseal cartilage plates (ECP), procesed for electron microscopy, revealed, as early as three hours after injection, appreciable enhancement of secretory activity, evidenced, in the zone of matrix secretion, by the abundance in Golgi cisternae of stippled material representing proteinpolysaccharide complexes. Disintegration of the lining membrane of individual Golgi vesicles was advanced after twenty-four hours; following three days of treatment, few vesicles remained intact, and pools of initially intravacuolar material were observable in the gound plasm. Long filaments, suggestive of primary or precursor collagen fibrils were apparent in this secretion. After six days, virtual lakes of this substance filled cells in the zone of prehypertophy, with consequent displacement of the rough endoplasmic reticulum against the cell periphery. Cytoplasmic vacuoles, containing mateerial similar to that found in the lacunar moat, and displaying finely beaded, radially arrayed filaments on the lining membrane were frequently encountered. Our observations suggest an initial acclleration of chondrocytic secretory activity, with subsequent retardation of transport. The resultant retention and intracellular polymerization of precollagenous products accelerates hypertrophy, thereby promoting early degeneration of chondrocytes. These ultrastructural alterations are apparently estrogen-specific.

Les protéoglycanes, résistants à l'extraction, sont visibles sous forme de granules concentrés dans les régions périlacunaires. Les granulations plus importants des protéoglycanes semblent venir du chondrocyte. Dans la matrice, éloignée des chondrocytes, ces granules deviennent plus étroites. Un composant non granulaire “amorphe” masque les fibres de collagène, de telle sorte qu'elles sont difficilement visibles dans le cartilage intact.
    Abstract: Zusammenfassung Die löslichen Proteoglycane wurden mittels Extraktion in 4 M Guanidinhydrochlorid aus Rippenknorpelschnitten des Rindes entfernt. Dies erlaubte die Sichtbarmachung von großen Mengen von verstreuten und auseinandergerissenen Collagen in der Matrix. Die Protoglycane, welche sich nicht extrahieren lassen, erscheinen als kleine, in den perilacunären Regionen konzentrierte Körnchen. Die großen Proteoglycan-Körner scheinen in den Chondrocyten zu entstehen. Sobald sie sich in die Matrix, außerhalb der Chondrocyten, verlagern, werden die Körner kleiner. Ein nicht-granulärer, „amorpher” Bestandteil verhüllt die Collagenfasern, so daß diese im intakten Knorpel nicht deutlich gesehen werden können.
    Notes: Abstract Removal of the soluble proteoglycans from slices of bovine costal cartilage by extraction in 4 M guanidinium hydrochloride permitted the visualization of abundant amounts of dispersed and disaggregated collagen in the matrix. Proteoglycans which are resistant to extraction are seen as small granules which are concentrated in the perilacunar regions. Large proteoglycan granules appear to originate in the chondrocyte. As they come to occupy positions in the matrix distant from the chondrocyte, the granules become smaller. A non-granular, “amorphous” component masks the collagen fibers so that they cannot be readily seen in the intact cartilage.

    Notes: Summary The potassium pyroantimonate technique was utilized for the selective subcellular localization of calcium in the mandibular condylar cartilage of 1-day-old rats. Electron dense calcium pyroantimonate precipitates were localized principally in mitochondria and at the cell membrane of the chondrocytes. In addition, small intracellular vesicles 0.1–0.2µm in diameter were observed in proximity to the cell membrane of chondrocytes of the mid-hypertrophic zone. The results suggest that these vesicles were being extruded from the cell into the extracellular matrix. Energy-dispersive analysis by X-rays confirmed that calcium is the principal cation of the electron-dense precipitates.

    Notes: Summary Diaphyseal tibial bone of 12.5 – 13-day and 19-day-old embryos and 20-day-old hatched chicks infected with retrovirus MAV.2-O were examined by transmission electron microscopy. The viruses were associated with lining osteoblasts and osteocytes. Whereas the infection of the osteoblast layer seemed to be a transient stage, virus association with osteocytes was a constant and main ultrastructural feature. The viruses were found either in the osteoid or in the periosteocytic space of the bone lacunae. They arose from dense cytoplasmic areas located near the cell plasmalemma via a budding process. The newly budded virus particles often had a large tail or a fine stalk-like process lost in the extracellular space. The viruses underwent calcification by deposition of inorganic material and were incorporated in the bone trabeculae. No production of virus was observed in typical osteoclasts with well-differentiated ruffled borders. The viral-induced avian osteopetrosis seemed to result from increased bone deposition through stimulation of osteoblast and osteocyte activities, whereas osteoclastic bone resorption seemed to be undisturbed.

    Description / Table of Contents: Résumé La diffraction à rayons X, la diffraction électrique et le microscope électronique suggérent qu'en dehors des composants usuels de calcium phosphate (apatite), un nombre d'autres éléments et radicaux sont également présents dans l'arteriosclérose aortique, lesquels sont capables de jouer un rôle dans le développement de la plaque «calcifiée» et dans celui de la lésion anorganique, caractéristique pour l'artériosclerose juvenile. Selon les cas étudies, l'artériosclerose aortique peut se présenter déjà au cours de la troisième decade de la vie et sa composition anorganique est presq'identique à celle de la plaque développée.
    Abstract: Zusammenfassung Sowohl die Befunde der Röntgendiffraktion als auch jene der Elektronendiffraktion und der Elektronenmikroskopie lassen vermuten, daß bei der Aortenatherosklerose zusätzlich zu den üblichen Komponenten von Calciumphosphat (Apatit) eine Reihe anderer Elemente und Radikale vorhanden sind; diese können eine Rolle spielen bei der Entwicklung der “verkalkten” Plaque und bei anorganischen Läsionen juveniler Atherosklerosen. Bei den untersuchten Fällen kann die Aortenatherosklerose bereits im 30. Lebensjahr auftreten; die anorganische Zusammensetzung der Plaque ist nahezu identisch mit jener der bereits entwickelten.
    Notes: Abstract X-ray diffraction, electron diffraction and electron microscopy suggest that in aortic atherosclerosis, in addition to the usual components of calcium phosphate (apatite), a number of other elements and radicals are present which may play a role in the development of the “calcified” plaque and in juvenile atherosclerotic inorganic lesion. In the cases studied, aortic atherosclerosis may appear as early as in the third decade and its inorganic composition is almost identical with the developed plaque.

    Notes: Abstract A technique to correlate the ultrastructural distribution of mineral with its organic material in identical sections of mineralized turkey leg tendon (MTLT) and human bone was developed. Osmium or ethanol fixed tissues were processed for transmission electron microscopy (TEM). The mineralized tissues were photographed at high, intermediate, and low magnifications, making note of section features such as fibril geometry, colloidal gold distribution, or section artifacts for subsequent specimen realignment after demineralization. The specimen holder was removed from the microscope, the tissue section demineralized in situ with a drop of 1 N HCl, then stained with 2% aqueous vanadyl sulfate. The specimen holder was reinserted into the microscope, realigned with the aid of the section features previously noted, and rephotographed at identical magnification used for the mineralized sections. A one to one correspondence was apparent between the mineral and its demineralized crystal “ghost” in both MTLT and bone. The fine structural periodic banding seen in unmineralized collagen was not observed in areas that were fully mineralized before demineralization, indicating that the axial arrangement of the collagen molecules is altered significantly during mineralization. Regions that had contained extrafibrillar crystallites stained more intensely than the intrafibrillar regions, indicating that the noncollagenous material surrounded the collagen fibrils. The methodology described here may have utility in determining the spatial distribution of the noncollagenous proteins in bone.

    Description / Table of Contents: Résumé Une nouvelle méthode d'isolement d'ostéoclastes est mise au point pour des analyses biochimiques et de microscopie électronique. Pour isoler les cellules par microdissection, des empreintes d'os métaphysaire sont utilisées. Cette méthode, supérieure aux coupes d'os, permet une meilleure préservation cytologique et enzymatique et permet d'obtenir des cellules totales plus faciles à manipuler, avec des résultats plus reproductibles. Par analyse planimètrique de cellules isolées, colorées histochimiquement, il apparait que les ostéoclastes constituent plus de 90% de la masse de l'échantillon. Les concentrations de la phosphatase acide et de certaines enzymes, liées au nucléotide pyridinique, entrant dans le métabolisme de l'acide citrique, sont déterminées dans des échantillons d'ostéoclastes, pesant de 0,2 à 2,0 μg, isolés à partir de rats normaux et parathyroidectomisés. L'activité en aconitase, mesurée en direction de la transformation de citrate en isocitrate, est de 0,5–0,8 M/Kd/H, la plus faible des activités étudiées. Les activités en GDH et NADP-ICDH sont 5 à 10 fois supérieures que celle de l'aconitase, mais seulement un dixième à un tiers de celle de la phosphatase acide, de la déshydrogénase lactique ou malique.
    Abstract: Zusammenfassung Es wird eine neue Technik beschrieben, welche die Isolierung von Osteoklasten für biochemische und elektronenmikroskopische Untersuchungen ermöglicht. Als Ausgangsmaterial zur Zellisolierung wurden Abstriche von Metaphysenknochen benützt. Die Verwendung von Abstrichen bietet gegenüber Knochenschnitten wichtige Vorteile, wie z.B. eine bessere Erhaltung der cytologischen und enzymatischen Eigenschaften sowie die Gewinnung von unverletzten Zellen, welche leichter verarbeitet werden können und besser reproduzierbare Daten ergeben. Durch planimetrische Analyse der histochemisch gefärbten Ausstriche von isolierten Zellen konnte nachgewiesen werden, daß die Osteoklasten über 90% des gesamten Probenmaterials ausmachen. Die Mengen verschiedener Enzyme, welche an Pyridinnukleotid gebunden und am Citronensäuremetabolismus beteiligt sind, sowie der sauren Phosphatase wurden in Osteoklastenproben bestimmt, welche ein Gewicht von 0,2–2,0 μg hatten und aus Knochen von normalen und mit Parathyroidextrakten behandelten Ratten isoliert worden waren. Die Aktivität der Aconitase, welche in der Richtung von Citrat zu Isocitrat gemessen wurde, war mit 0,5–0,8 M/Kd/H die niedrigste aller untersuchten Aktivitäten. Die Aktivitäten der GDH und der NADP-ICDH waren 5–10mal höher als jene der Aconitase, entsprachen jedoch nur einem Zehntel bis einem Drittel derjenigen der sauren Phosphatase, der Laktat- oder der Malatdehydrogenase.
    Notes: Abstract A new method is described by which osteoclasts can be isolated for biochemical and electron microscopic analyses. As a source of cells for isolation by microdissection, imprints of metaphyseal bone were used. The use of imprints provides important advantages over bone sections, including a higher degree of cytologic and enzymatic preservation, and the delivery of whole cells which are more readily manipulated and which yield data that are more readily reproduced. By planimetric analysis of the histochemically-stained isolated cell samples, it was shown that osteoclasts represent over 90% of the sample mass. The levels of several of the pyridine nucleotide-linked enzymes involved in citric acid metabolism, as well as acid phosphatase, were determined in osteoclast samples weighing 0.2 to 2.0 μg isolated from normal and parathyroid-treated rats. Aconitase activity measured in the direction of citrate to isocitrate was 0.5–0.8 M/Kd/H, the lowest of the activities studied. The activities of GDH and NADP-ICDH were 5 to 10 times higher than that of aconitase but only a tenth to a third that of acid phosphatase, lactic or malic dehydrogenase.

Au cours de cette étude, des rats máles de 200 g sont injectés à l'aide de doses supra-physiologiques de testostérone pour observer les effects sub-microscopiques sur les diverses zones du cartilage épiphysaire. Au niveau de la zone de division cellulaire, on note une augmentation des cellules en division. Les cellules, en voie de maturation, présentent plus de produits de sécrétion, à un stade plus précoce de leur cycle d'évolution, et semblent subir une hypertrophie plus rapide. Dans la zone pré-hypertrophique, la matrice intercellulaire présente des foyers de calcification précoce, ainsi que des fibres collagènes plus longues et plus épaisses que chez les témoins. Il apparait que, chez l'animal entier, des doses même élevées de testostérone provoquent initialement une stimulation de la prolifération chondrocytaire, avant de favoriser les processus de maturation.
    Abstract: Zusammenfassung Obwohl experimentelle und klinische Erfahrung darauf hinweisen, daß hohe Dosen von Testosteron zu einem frühzeitigen Wachstumsabschluß führen, sind der genaue Mechanismus und der eigentliche Wirkungsort dieses Hormons im Wachstumsapparat der Röhrenknochen unbekannt geblieben. In diesem Experiment wurden 200 g schweren männlichen Ratten supraphysiologische Testosterondosen injiziert, um die submikroskopischen Auswirkungen auf die verschiedenen Zonen des Epiphysenknorpels zu beobachten. In der Zone der Zellmitosen fand sich eine erhöhte Anzahl von sich teilenden Zellen. Die reifenden Zellen häuften im Frühstadium ihres Lebenscyclus größere Mengen von Sekretionsprodukten an und schienen eine abruptere Hypertrophie durchzumachen. In der prähypertrophen Zone enthielt die interterritoriale Matrix Herde von früher und verfrühter Verkalkung, sowie dickere und längere Kollagenfasern als vergleichsweise in Kontrolltieren. Daraus wird geschlossen, daß bei unbehandelten Tieren sogar große Testosterondosen anfänglich eine Stimulation der Chondrocytenproliferation verursachen, bevor sie die Reifungsprozesse veranlassen.
    Notes: Abstract Although experimental and clinical experience indicates that large doses of testosterone lead to premature cessation of growth, the exact mechanism and precise site of action of this hormone on the growth apparatus of long bones remain unknown. In this study, plateaued male rats were injected with supraphysiologic doses of testosterone to observe the submicroscopic effects on the various zones of the epiphyseal cartilage. In the zone of cell division there were increased numbers of dividing cells. The maturing cells accumulated larger amounts of secretory products at earlier stages of their life cycle, and appeared to undergo a more abrupt hypertrophy. In the zone of prehypertrophy, the interterritorial matrix contained foci of early and premature calcification and thicker and longer collagen fibers than at comparable levels in controls. It is concluded that in intact animals, even large doses of testosterone initially cause a stimulation of chondrocyte proliferation, prior to promoting maturation processes.

Le premier stade de formation d'un prisme est identique à celui observé dans la formation du nacre, à savoir l'élaboration d'une lamelle dense aux électrons qui sert de limite interne au futur prisme. Les fragments de lamelles se détachent et migrent vers un espace bordé extérieurement par le periostracum. Ces fragments lamellaires forment des enveloppes, au niveau desquelles on observe le dépôt initial et la croissance des cristaux. En même temps, on voit apparaitre des parois interprismatiques nettes, qui dérivent aussi des lamelles. La croissance de nouveaux cristaux et d'éléments organiques donne finalement un prisme adulte allongé. La croissance de la coquille se fait en périphérie, surtout par formation de nouveaux prismes. En outre, un environnement modifié, qui consiste en un dédoublement du periostracum au niveau de la surface distale, donne naissance à des ilôts étroits, contenant des prismes, qui se forment sur les bords de l'espace produit par la courbe du periostracum.
    Abstract: Zusammenfassung Die Prismenbildung beginnt in der proximalen Region der äußeren Oberfläche der äußeren Mantelfalte in Pallialraum, der gegen außen durch das Periostracum begrenzt wird. Der erste Schritt bei einer Prismenbildung verläuft gleich, wie dies bei der Perlmutterbildung beobachtet werden kann, nämlich in Form der Ausarbeitung einer elektronenoptisch dichten Lamelle, welche als innere Begrenzung des zukünftigen Prismas dient. Fragmente der Lamelle werden abgetrennt und wandern zu einem Zwischenraum, der gegen außen durch das Periostracum abgeschlossen wird. Diese Lamellenfragmente bilden Hüllen, innerhalb welcher der Kristall entsteht und sein Wachstum stattfindet. Gleichzeitig bilden sich dicke, zwischen den Prismen liegende Wände, die ebenfalls von den Lamellen abstammen. Das aus der Bildung zusätzlicher Kristalle bestehende Wachstum, zusammen mit den organischen Komponenten, läßt schließlich das reife längliche Prisma entstehen. Das Wachstum der Muschel spielt sich am Rande hauptsächlich durch Bildung neuer Prismen ab. Durch eine Veränderung der Umgebung, bestehend aus einer Verdoppelung des Periostracums an der distalen Oberfläche, entstehen zusätzlich dünne, prismenhaltige Sporne, welche innerhalb des begrenzten Raumes vorkommen, der sich durch das Überschlagen des Periostracums bildet.
    Notes: Abstract The initiation of prisms occurs in the proximal region of the outer surface of the outer mantle fold in the pallial space bounded externally by the periostracum. The first step in the formation of a prism is similar to that observed in the formation of nacre, namely, the elaboration of an electron-dense lamella that serves as the internal boundary of the future prism. Fragments of the lamella become detached and migrate to a chamber bounded externally by the periostracum. These lamellar fragments form envelopes within which crystal initiation and growth oocur. At the same time stout interprismatic walls appear. They are also derived from the lamellae. Growth consisting of the formation of additional crystals and the organic components finally give rise to the mature elongated prism. Growth of the shell occurs at the margin chiefly by formation of new prisms in this area. In addition a modified environment consisting of duplicature of the periostracum on the distal surface results in the formation of thin spurs containing prisms that occur within the confines of the space created by the periostracal loop.

Les rapports entre phases inorganique et organique sont discutés.
    Abstract: Zusammenfassung Die organische Phase (oder Kristallit-Schatten), die zu jedem Kristallit gehört, sowie das Hintergrundmaterial, das zu jeder Kristallitgruppe gehört, wurden in calcifiziertem Knorpel sichtbar gemacht. Zu diesem Zweck wurde basisches Chromsulfat als ein kombiniertes Fixierungs-, Färbe- und Demineralisierungsmittel verwendet. Nachfolgende Behandlung des Gewebes mit Papain oder Hyaluronidase läßt vermuten, daß die Kristallitschatten einen Proteinpolysaccharidkomplex darstellen und daß das Hintergrundmaterial hauptsächlich aus Protein mit einigen sauren Polysacchariden besteht. Die Beziehung zwischen anorganischen und organischen Phasen wird diskutiert.
    Notes: Abstract The organic phase (or crystallite ghost) associated with each crystallite, together with the background material associated with each crystallite cluster, was demonstrated in calcified cartilage using basic chromium sulphate as a combined fixative, stain, and demineralizing agent. Subsequent treatment of the tissue with papain, or with hyaluronidase, suggests that the crystallite ghosts represented a protein-polysaccharide complex and that the background material was principally protein together with some acid polysaccharide. The relationship between inorganic and organic phases is discussed.

    Notes: Summary Cytodifferentiation of inner enamel epithelium and the adjacent connective tissue from the tip of the cervical loop to the initiation of enamel elaboration in twoMacaca species was examined. Ten- to twelve-month-old specimens were fixed by perfusion and the permanent tooth buds were prepared for transmission electron microscopy. At the cervical loop proper, inner enamel epithelium cells have lobed nuclei, a paucity of cytoplasm, and wide extracellular spaces; the basal lamina facing the dental papilla is straight. With increasing distance from the tip of the cervical loop, the following changes occur gradually: (a) preameloblasts elongate from 15 to 45 µm, and their organelles, particularly mitochondria and profiles of rough endoplasmic reticulum, become more numerous; (b) extracellular spaces decrease between preameloblasts starting at the basal (infranuclear) end; (c) the basement membrane becomes convoluted and associated with aperiodic fibers; (d) preodontoblast projections penetrate the aperiodic fibers; (e) collagen fibers subjacent to the basement membrane increase in density, with particularly thick fibers paralleling the aperiodic fibers. These modifications occur within three-fourths of the distance from the tip of the cervical loop to the mineralization front. The condensation of preodontoblasts is followed immediately by predentin synthesis. Concomitantly, the basement membrane breaks down and the aperiodic fibers are engulfed by preameloblasts. Preameloblast projections penetrate junctional predentin, contact mineralized dentin, and enamel synthesis ensues. At this stage the ameloblast is 45 µm long, the nucleus is central or basal, the Golgi apparatus has migrated apically, but the Tomes' process has not yet formed. The results indicate that odontogenesis inMacaca monkeys more closely resembles human odontogenesis than does that in the murine rodents.

    Abstract: Zusammenfassung Die Beschießung mit Argonionen wurde angewendet, um die Dicke von Calciumcarbonat-Schalen der zweischaligen MuschelnMytilus undMercenaria zu reduzieren. Diese Technik erlaubte die Ausführung von Transmissions-Elektronenmikroskopie und Elektronendiffraktion, wobei gleiche Proben nach einer bereits bestehenden Methode vorbereitet und als Kontrollen herangezogen wurden. Es wurden zusätzliche Resultate zu den Muschelstruktur-studien erhalten, welche früher publizierte Arbeiten unterstützen, die mit der Abklatschmethode und der Raster-Elektronenmikroskopie ausgeführt worden waren. Zusätzlich wurde eine „schaumartige” Struktur der Muschelaragoniten, besonders im Perlmutter vonMytilus, beobachtet. Da es sich um ein durch Hitze verursachtes Artefakt handelt, wird diese Struktur als Einschlüsse von Wasser und organischem Material interpretiert, was den Befunden von verschiedenen veröffentlichten chemischen und elektronenmikroskopischen Arbeiten entspricht.
    Notes: Abstract Use is made of the argon ion-bombardment technique to reduce the thickness of calcium carbonate shells of the bivalvesMytilus andMercenaria for transmission electron microscopy and electron diffraction, with comparison of single-stage replicas of similar specimens serving as controls. As an additional approach to shell structure studies, it gives results which support earlier published work with both replicas and scanning microscopy. In addition, a “frothy” structure is detected in the shell aragonites, especially inMytilus nacre. As a heat-induced artifact, it is interpreted as representing trapped water and organic material inclusions, an interpretation consistent with several published chemical and electron microscope studies.

Die Absorption einer exogenen, elektronisch dichten Verbindung, Thoriumdioxyd, erfolgt durch diese Kanäle, offenbar durch einen „Membranfluß”. Der Inhalt der Kanäle wird zu den Lysosomen („spezifische Granula”) geführt, welche unter dem gekrausten Rand liegen. In „Übergangsgebieten”, welche sich neben dem gekrausten Rand befinden, scheint die Aufnahme der Thoriumdioxydpartikel durch „Bläschenbildung in der Membran” (konventionelle Endocytose) stattzufinden.
    Notes: Abstract The ruffled border of osteoclasts in the fracture callus of rat consists of complex infoldings of the plasma membrane forming narrow channels. Absorption of an exogenous, electron-dense compound, thorium dioxide, has been shown to take place via these channels, apparently through “membrane flow”. The contents of the channels are transferred to lysosomes (“specific granules”) located subjacent to the ruffled border. In “transitional regions” adjacent to the ruffled border, uptake of thorium dioxide particles appeared to occur through “membrane vesiculation” (conventional endocytosis).

    Notes: Summary The egg-shell of Japanese quail was studied by several techniques. Semithin sections (1μm thick) of non-decalcified shell were observed by normal and polarized light microscopy. Thin sections of non-decalcified shell, examined by transmission electron microscopy, permitted us to observe the forms and dimensions of crystals of calcite within different layers of the shell: mammilary layer, layer of cones, palissade layer and surface crystal layer. There appears to be two distinct zones in the layer of cones as well as in the superficial crystal layer. Electron microdiffraction revealed the orientation of calcite crystals in the columns. Some crystal defects (twins?) were described and the possibility of their artefactual formation during ultramicrotomy is discussed. Localization of Ca, Mg, P and S were made by X-ray microanalysis of semithin sections. This technique shows that shell membranes, and chiefly the true cuticle, are also mineralized but, in these layers, minerals are not crystallized. Otherwise the distribution of Mg is not uniform throughout the shell thickness; it is less concentrated in the external zone of the layer of cones. These results together with observation of developing shells by scanning electron microscopy allowed us to propose a scheme for shell organization of the quail egg. This organization was related with decalcification which occurs during hatching.

    Notes: Summary Newly formed osteodentin obtained from the anterior extremities of fetal or young rat incisors was observed by means of electron microscopy and electron probe X-ray microanalysis. Cells related to osteodentin formation frequently showed membrane bound intracellular bodies containing varying amounts of fine, needle-shaped crystals, which were identified as apatite. The intracellular clusters of apatite crystals were extruded from the cells through membrane fusion or cellular degeneration. These extracellular clusters seemed to be gradually incorporated into the mineralizing collagenous matrix, which developed around them. Frequent occurrence of dense, dotshaped or filamentous profiles suggested that the dense bodies seen in the perinuclear regions or in the Golgi area were the sites of crystal formation. Energy dispersive X-ray point analysis showed that the intracellular or extracellular apatite clusters contained sulfur in a concentration higher than was present in the mineralizing collagenous matrix. Furthermore, wave dispersive X-ray line analysis showed that the concentration of sulfur was higher in the osteodentin matrix than in the dentin matrix. The sulfur detected is presumed to be contained in acid mucopolysaccharides, which were distributed more heavily in the osteodentin matrix than in the dentin matrix. On the basis of these data, it was concluded that the unique chemical and structural characteristics of the osteodentin result primarily from the incorporation of apatite clusters of intracellular origin and associated acid mucopolysaccharides.

In osteocytic bone the osteoblast secreted a collagenous preosseous matrix in which it became enmeshed and then was termed a preosteocyte. When the preosseous matrix mineralized, the preosteocyte was termed an osteocyte and was completely surrounded by bone. In anosteocytic bone the osteoblasts receded from the mineralizing front and never became trapped as osteocytes. During resorption, types A and B resorptive cells, present in both bone types, invaded the matrix and demineralized the osseous zone. These cells were characterized by large amounts of granular endoplasmic reticulum and intracellular inclusions containing crystal-like material. Although functionally similar to mammalian osteoclasts, these cells lacked a characteristic ruffled border and were not multinucleated. The osteocytes of cellular bone did not appear to be involved during demineralization.

Die oberen Enden der Zellen sind von Microvilli eingefaßt. Einige der Zellen haben zudem apikale Zilien. Die Zellen enthalten Golgi-Apparate im apikalen Cytoplasma. Eine große Anzahl von Mitochondrien sind über das, ganze Cytoplasma verteilt. Die mögliche Funktion dieser mineralhaltigen Zellen als Aufbewahrungsorte für Calcium und/oder Phosphor wird besprochen.
    Notes: Abstract A small patch of cells in the epithelium of the anterior surface of the collar of the serpulidPomatoceros caeruleus contains membrane-bound vacuoles filled with crystalline material. The crystals have rhomboidal or rectangular profiles and have been shown by electron diffraction analysis to be composed of hydroxyapatite and calcium magnesium phosphate. The apices of the cells are bordered by microvilli. Some cells also have apical cilia. The cells contain Golgi complexes in the apical cytoplasm. Large numbers of mitochondria are distributed thoughout the cytoplasm. The possible function of these mineral-containing cells as sites for storage of calcium and/or phosphorus is discussed.

    Notes: Abstract This investigation sought to determine whether splenic lymphocytes obtained from Balb/C mice exposed to aflatoxin B1 (AFB1) showed any ultrastructural changes which could account for the immunodysfunction attributable to aflatoxins. Lymphocytes obtained from Balb/C mice administered aflatoxin B1 in olive oil daily for three weeks were studied using both transmission and scanning electron microscopy. The lymphocytes demonstrated ultrastructural changes primarily in the mitochondria where marked internal dissociation of the cristae was revealed by transmission electron microscopy. All other cellular organelles were unaffected. No significant alterations in external structure were observed under scanning electron microscopy. The findings of this study indicate that AFB1 administration does not affect the surface topography of lymphocytes, but AFB1, by causing extensive mitochondrial damage, may affect the way in which these cells function. This could be a possible explanation for the immunodysfunction associated with AFB1.

    Notes: Abstract The three-dimensional ultrastructure ofCryptococcus neoformans was studied by quick-freezing and deep-etching (QF-DE) method.C. neoformans, strain CDC551, was cultured on agar. The viable yeast cells (107 cells) were inoculated into each mouse from the tail vein. Three weeks after the inoculation, the brains of the mice were perfused with fixatives, quickly frozen, freeze-fractured, deeply etched and rotary shadowed with platinum and carbon. In addition, the viable cells ofC. neoformans on agar were picked up and quickly frozen, and replica membranes were prepared as described above. The ultrastructure ofC. neoformans was three-dimensionally demonstrated by the QF-DE method. The capsule was composed of fine meshworks of microfibrils (10–13 nm in diameter), which were directly attached to the cell walls. The capsule of the in vivo yeasts (yeast cells in the brain lesion) was thicker than that of the in vitro yeasts (yeast cells on agar culture). At the outer part of the cell wall, a particle-accumulating layer was observed. This layer in vivo was thicker than that in vitro. Occasionally, the yeast cells were ingested by phagocytes in the mouse brain. Although the cytoplasm of such yeast cells was destroyed, the capsular meshworks were well preserved. The ultrastructure of the capsule was the same both in cultured and phagocytized yeasts in the cystic lesions of the brains. This lack of morphological changes of the capsular meshworks suggests that they are resistant to the digestion by phagocytes. This stability of capsular structures may provide one of the important pathogenic factors in cystic lesions byC. neoformans.

    Notes: Abstract Mature maize (Zea mays L.) embryos were exposed to aflatoxin B1 (AFB1) concentrations ranging from 0.1 to 25 µg/ml for 9 days. With increasing toxin concentration above 2 µg/ml, primary root elongation of germinated embryos was progressively inhibited, to reach a maximum value of 81% at 25 µ/ml toxin. An ultrastructural investigation of the subcellular alterations induced following toxin exposure provided evidence of deteriorative changes in several compartments of the plant cell. Alteration in membrane integrity (e.g., the tonoplast, plasmalemma and inner mitochondrial membrane) was a frequent feature of many cells. Apparent fusion of vacuoles, incorporation of cytoplasmic components into vacuoles and intravacuolar membrane whorls might be interpreted as deteriorative alterations. The results are discussed in the light of ultrastructural findings for other plant systems exposed to similar AFB1 concentrations, as well as findings for animal systems.

    Notes: Abstract The ultrastructural organization of the spores of the sporocarp of Endogone flammicorona was studied. Two types of organization are described. Initially the spore possessed a vacuolate protoplasm and was bound by two cell wall layers. The spore was surrounded by a hyphal mantle formed of a sheet of vacuolized hyphae with uniformly thin walls. Secondly, although the ultrastructural features of the spore appeared the same, it was now surrounded by a hyphal mantle with unevenly thickened walls (i. e., the so-called flaming crown) due to the gradual and irregular deposition of granules and lamellae. This crown gives the spore its most commonly observed morphological feature and is the preminent character employed taxonomically to speciate Endogone flammicorona Trappe & Gerdemann.

    Notes: Summary The ultrastructure of glycerol extracted thrombocyte models was studied before and after incubation with ATP and under the influence of Salyrgan as inhibitor of contraction. The contractile system of the thrombocytes—thrombosthenin—consists of a spatial network of 50 Å wide thrombosthenin A and 100–120 Å wide thrombosthenin M filaments. At rest, the contractile system is arranged in a marginal zone. The ATP-induced contraction led to a concentric condensation of the network which resulted in a central cluster of cell organelles and at a later stage, disruption of the plasma membrane and release of the thrombocyte granules. All these changes which also occur during the formation of the hemostatic plug in vivo are therefore attributed to the reaction of the contractile protein.

    Notes: Summary In the mouse, the rostral zone of the pars intermedia is almost exclusively composed of typical corticotrophic cells. They are located around and even within the neural stalk, at the level of transition between stalk and neural lobe. In the rat, the corticotrophic cells of the rostral zone are found in scattered islets among the MSH producing cells, and also in the neural lobe. In both the rat and mouse, these cells are in direct contact with various types of nerve terminals. Synaptoid contacts with aminergic and neurosecretory nerve fibers are observed. Furthermore they are also closely related to the hypophysial portal vessels. Following adrenalectomy, the cells located in the neurohypophysis always react more intensely than tose in the rostral zone. The functional significance of these corticotrophic cells which are subject to both humoral and neural regulation remains as yet hypothetical. Their participation in neurogenic stress response seems probable.

    Notes: Summary In order to characterize different sub-zones in the palisade zone of the rat median eminence, quantitative ultrastructural parameters were applied to brains fixed in aldehyde-osmium tetroxide. The palisade zone has been subdivided in 4 successive sub-zones. Increasing numbers of granular vesicles (GV), especially those smaller than 110 nm, are observed from dorsal to ventral sub-zones. There also are more GV per unit area of nervous tissue in the perivascular than in the more dorsal sub-zones. The individual nerve profiles exhibit a larger size in the perivascular layer than in the more dorsal areas, whereas the number and size of nerve profiles devoid of vesicles diminish from dorsal to ventral. As a consequence more GV occur in the perivascular nerve profiles. In the GV containing nerve profiles the concentration of GV is, however, constant in the different sub-zones. A fluctuating size of the preterminal and terminal parts of the nerve fibres is suggested.

    Notes: Summary The ultrastructure of Cyclidium, including the cilia, kinetosomes, pellicle, microtubules and kinetodesmal fibers is similar to that recorded for other ciliates. Of special interest is the attachment of rod-shaped bacteria within the longitudinally directed shallow surface folds of the protozoan. Both the bacteria and the surface of Cyclidium seem to possess an outer coating of a sticky substance which upon contact holds the bacteria to the protozoan. The bacteria appear to be attached by only a relatively small area of their surfaces. A dense substance appears within the alveolus of the pellicle at the regions of the attachment of the bacteria. The association of the organisms is probably a temporary one, and it is unknown whether either organism is benefited or harmed by the association. The position of the mitochondria in Cyclidium is unusual in that they all lie flattened against the inner membrane of the pellicle, usually in a position directly opposite to that of the attachment of the bacteria to the surface, thus being separated from the bacteria by only the outer cell membrane and the pellicle. Whether or not this close topographical relationship is of significance is unknown.

    Notes: Summary Acclimation to low salinity induces changes in the ultrastructure of the gill cells of the marine euryhaline amphipod, Gammarus oceanicus. The gills are composed of a single cell type. In 100% artificial sea water, these cells contain moderate numbers of mitochondria which are randomly distributed in the cytoplasm. The plasma membrane is extensively invaginated at the apical, lateral, and basal surfaces. Acclimation to 20% artificial sea water induces a further invagination at the apical cell membrane to form an elaborate apical labyrinth. The extracellular spaces between the folds in the basal cell membrane dilate to 1500 Å or more. Mitochondria are more abundant and in many cells they undergo a change in conformation. The mitochondria are crowded into thin leaflets of cytoplasm between the dilated basal invaginations or into the narrow space between apical and basal cell membranes. Consequently, they lie in close contact with the plasma membrane over much of their surface.

Die paarigen lateralen Riesenzellen (LRZ) sind im Gegensatz zu der unpaaren medianen Riesenzelle (MRZ) häufig multipolare Neuronen, die bis zu vier Faserfortsätze aufweisen können. Das perikaryale Cytoplasma zeigt helle und dunkle Areale. Organellen und andere Zelleinschlüsse finden sich ganz überwiegend in den dunklen osmiophilen Zonen und lassen regelmäßig eine bestimmte Anordnung erkennen. So ist häufig eine ringförmige Verteilung von Golgi-Feldern um den gelappten Kern vorhanden. Mitochondrien sind häufig in einem typischen „Axonhügel-Komplex“ angeordnet. Freie Ribosomen dominieren gegenüber membrangebundenen. Diese Neuronen sind reich an Glykogen, das oftmals ausgedehnte Felder einnimmt. Vereinzelt weisen die LRZ Faser-Soma-Synapsen auf.
    Notes: Summary The structure of the segmentally arranged somata of the dorsal giant fibres Günther, 1969) in the ventral nerve cord of the earthworms Lumbricus terrestris L., is described. These neurones are compared with other neurones of invertebrates and vertebrates. The paired lateral giant cells (LGC) are often multipolar in contrast to the single unipolar median giant cell (MGC). Up to four fibres originate from each LGC. The dotted appearance of both these neurones (clear and dark perikaryal areas) is due to the regular distribution of osmiophilic strands of cytoplasm containing the majority of organells and other inclusions (e.g. endoplasmatic reticulum and ribosomes). The majority of ribosomes are not associated with membranes but are found scattered freely in the cytoplasm. A typical “axon hillock” with an accumulation of mitochondria is often found. The cells show large deposits of glycogen. Some of the LGC-somata have synaptic contacts.

    Notes: Summary A subectodermal nerve-net is demonstrated in ten species of Ctenophores by means of either Methylene Blue vital staining or silver impregnation. There is no evidence of subendodermal nerve-net. The ectodermal nerve-net displays a characteristic polygonal pattern which is thought to result from morphogenetic events. Both bipolar and tripolar neurones occur. Three types of concentrations of nervous tissue are described: a) a high accumulation of nervous perikarya at the aboral sensory pole; b) a noticeable densification of the net along the eight meridional ciliary strands of every species, and around the lips of Beroidea; c) in the Cydippids Pleurobrachia and Hormiphora two thick strands of fibers and neurones interconnecting the aboral organ and the tentacles: the tentacular nerves. Ultrastructural evidence is given for the presence of nervous elements under and amidst ectodermal epithelial cells. Three features allow the recognition and characterization of neurites and neurones: a) numerous clear and/or granulated vesicles; b) microtubules in variable amounts; c) frequent and highly differentiated synaptic contacts. The very peculiar arrangement of the presynaptic elements is observed in all the species of Ctenophores so far studied. These synaptic contacts suggest chemical transmission in the nerve-net. On the basis of ultrastructural evidence the author refutes the nervous nature previously attributed to the ciliated cells of the meridional grooves. A parallel is drawn between nervous systems of Cnidaria and Ctenophora.

    Notes: Summary Five categories of granulated cells were distinguished by their ultrastructural features, and quantitative analyses were made of the pars distalis cells in normal and castrated lizards. The gonadotropin-producing cell was identified on the basis of its uniform distribution in the gland as well as from cytological changes resulting from castration. The secretory granules of the gonadotropic cell vary in size (100–500 mμ) and density, and lipid bodies are commonly present. Following castration, the endoplasmic reticulum proliferates, forming many small, rough-surfaced, dilated cisternae which do not coalesce greatly as in other vertebrate species. Degranulation is accompanied by hypertrophy and hyperplasia of the mitochondria and by the appearance in the cytoplasm of conspicuous clusters of microfilaments. The designated gonadotropic cell was the only class of secretory cell showing consistent changes following three weeks of castration. In addition to the uniformly distributed gonadotrope cell, two secretory cells occur mainly in the rostral half of the gland, and two in the caudal half. Tentative identification of the cell types is discussed in the light of available information on the localization of the hormones in the pars distalis of this species.

    Notes: Summary Smooth muscle-like cells are present in thecae externae, corpora lutea, and interstitial tissue of hamsters and gerbils. The smooth muscle-like cells, as examined by electron microscopy, are fusiform with central nuclei; the cytoplasm contains numerous myofilaments, dense bodies, micropinocytotic vesicles, and dense accumulations of glycogen-like particles. In addition to the smooth muscle-like cells, fibroblasts and cells that have characteristics of both fibroblasts and smooth muscle are located in the thecae externae of both species. There is no ultrastructural evidence of innervation in the theca folliculi, corpora lutea, or interstitial tissue of either species. A possible function for the smooth muscle-like cells is discussed.

Un nouveau type de structure s'associe dans le foie aux membranes plasmatiques: il s'agit des saccules ergastoplasmatiques ou subsurface cisternae. 2.Associations desmosomes-organites. Dans ces complexes, les tonofilaments desmosomiques entrent étroitement en contact avec la paroi des organites cellulaires associés: mitochondrie ou peroxysome. Déjà présents au 13ème jour de la gestation, ces structures s'observent plus fréquemment à partir du ler jour post-natal. La signification de ces associations reste à déterminer, d'autant plus que leur présence a déjà été signalée dans de trés nombreux tissus. Quelques hypothèses sont présentées.
    Notes: Summary During the transition between the fetal and postnatal periods associations between cell junctions and cell organelles appear in the liver of the mouse. These associations are of two types: desmosome-organelle and nexus-organelle. 1.Nexus-Organelles Association. Unilaterally or bilaterally, various organelles — mitochondria, cysternae, microbodies or lysosomes — adhere tightly along the gap. These associations appear only between the 21st day of gestation and the first postnatal day. Thereafter they gradually disappear and are replaced by desmosome-organelle associations. Another type of structure — ergastoplasmic saccules or cisternae of rough endoplasmic reticulum — become associated with the cell membrane. 2.Desmosome-Organelle Associations. In these complexes the desmosomal microfilaments are in close contact with membranes of the associated organelles — mitochondria or peroxysomes. The associations, which exist as early as the 13th day of gestation increase after the first postnatal day. The significance of these associations remains to be ascertained, especially since they occur in many other tissues. A few hypotheses are presented.

    Notes: Summary The fat body of the adult housefly is composed of two types of cells, the lipid-and glycogen-rich fat body cells and the oenocytes. A comparison of the fine structure of the abdominal fat body in 4-day old and 31–35 day old male houseflies indicated an increase in lipid and a decrease in glycogen content in the fat body cells of old flies. Oenocytes of old flies exhibit deteriorative alterations with an accumulation of secondary lysosomes. Both fat body cells and oenocytes in senile flies are ingested by hemocytes.

Chaque tentacule est innervé par quatre faisceaux nerveux, dépourvus d'envelope gliale, qui courent entre l'épithélium et l'assise collagène qui délimite le canal tentaculaire interne; trois sont rassemblés sous les trois rangées cellulaires épithéliales de l'arête orale du tentacule et le quatrième est médian-dorsal. Les cellules épithéliales orales-latérales, par leur forme pédonculée, par la densité de leur cytoplasme, par leur cil unique et par leur relation topographique et cytologique avec les nerfs tentaculaires sous-jacents, présentent des adaptations structurales telles qu'il parait probable qu'elles assurent une fonction tactile. L'examen du collier péripharyngien montre son caractère organisé et la complexité des connexions qui coordonnent l'ensemble des tentacules et relient le lophophore à d'autres secteurs de l'innervation.
    Notes: Summary The pathway of the tentacle nerves and the organisation of the peripharyngial nervous belt from which they arise, have been studied in Electra pilosa by silver stained whole mounts (Bielschowsky technique), and by ultrastructural investigation. Each tentacle is innervated by four nervous fascicles without any differentiated glial sheath, running between the epithelium and the collagen layer which surrounds the muscles and the peritoneal cells in the internal tentacle canal. Three nerves are gathered underneath the three rows of epithelial cells forming the oral edge of the tentacle. The fourth nerve is in medio-dorsal location. The oral epithelial cells show such ultrastructural adaptations in their general shape, in the density of their cytoplasm, in their ciliary apparatus reduced to a single cilium and in their close topographical and cytological relationship with the underlying tentacle nerves that it seems most probable they have a tactile function. The analysis of the pattern of the peripharyngial nervous belt shows a precise organisation and the intrication of the connections which coordinate the tentacles and link the lophophore set to other pathways of general innervation.

    Notes: Summary The response of somatotrophs to large doses of purified bovine growth hormone was examined in uninephrectomized rats drinking 1% sodium chloride in an attempt to prolong the action of the hormone. Growth hormone produced a triphasic response in the ultrastructure of somatotrophs. Uninephrectomy and salt alone induced no changes in somatotrophs as compared to unoperated animals consuming tap water. In the first phase (3 days) the cross sectional area of the cell increased, the size of the largest granule per cell as well as the number of granules per cell increased significantly. All these parameters then reached control level after 7 days of injections during a second phase. Suppression of somatotrophs by exogenous growth hormone was evident by 16 days (third phase) when the area of the cell, the size of the largest secretory granule as well as the number of granules per cell all decreased significantly. Somatotrophs apparently did not react to suppression by a lysosomal mechanism inasmuch as no increase in the number of lysosome-derived structures was seen even at 16 days.

    Notes: Summary The ultrastructure of the secretory cells of the adenohypophysis of juvenile sockeye salmon was investigated. Pituitary glands were collected from immature fish transferred experimentally to sea water and subsequently returned to fresh water. The rostral pars distalis contained three cell types: ACTH cells, prolactin cells, and non-secretory cells. The prolactin and non-secretory cells were joined together in the form of follicles by desmosomes and they both had cilia and microvilli projecting into the follicle lumen. Various follicular structures such as lumen, multivesicular structures, and peripheral basement membrane are discussed as possible sites of prolactin cell granule release. The columnar ACTH cells were found at the junction of the rostral pars distalis and the neurohypophysis. The cytoplasmic granules in these cells were characteristically separated from their limiting membrane by a clear space. Multivesicular structures were also found in association with this cell type. The caudal pars distalis also contained three cell types: one acidophil (putative somatotrop) and two basophils (putative thyrotrops and gonadotrops), all of which were similar to those described in adult fish. The pars intermedia contained only one cell type. They appeared to be active cells and were characterized by containing membrane-bounded granules similar to those found in the ACTH cells. Changes in ambient salinity had no apparent effect on any cell type described.

    Notes: Summary Experiments were performed to compare the permeability of capillaries supplying the endoneurial environment, which is invested by perineurium, with vascular permeability in the pulp where perineurium is absent. Anaesthetised rats were perfused through the aorta with physiological solutions containing lanthanum nitrate at 37° C. Pieces of inferior alveolar nerve and segments of mandibular incisors were immersion-fixed and transverse sections were examined electron microscopically for the distribution of lanthanum. In the pulp the nerve fibres pass between lanthanum-impermeable arterioles and venules en route to the incisal end. In the peripheral pulp a few capillaries were permeable but the most permeable capillaries lay between the odontoblasts. Pulpal capillary permeability was attributed to the fenestrated endothelium and contrasted with the unfenestrated endoneurial capillaries which were impermeable to lanthanum. Whereas the tight junctions of endoneurial capillaries are known to prevent certain blood-borne substances from entering the endoneurium, it was not clear whether the permeability of the pulpal capillaries, which are distant from the nerve fibres, could affect the nerve fibre environment. No extravasated lanthanum reached the pulpal nerve fibres suggesting that they are not affected. Technically it was not possible to examine the incisal third of the tooth where the situation could be different because the volume of the pulp decreases and capillaries lie closer to the nerve fibres.

    Notes: Summary The effects of FSH on the testicular interstitial tissue of immature hypophysectomized rats were studied by comparing morphological changes in Leydig cells with quantitative changes in interstitial tissue histology using morphometric analysis. Three groups of rats received subcutaneous injections of 0.5 ml saline vehicle or 10 μg rFSH or 20 ng oLH (equivalent to the amount of LH known to contaminate the FSH), twice daily for 7 days. Administration of FSH significantly increased testis weight and stimulated more advanced spermatogenesis compared to saline or LH. Morphometric analysis of testes of LH-treated rats showed a small but significant increase in total interstitial cell volume compared to saline treatment. FSH caused much greater increases in the total volume of interstitial tissue and interstitial cells than either saline or LH and significantly increased the total volume of interstitial fluid by comparison with the other groups. FSH but not saline or LH treatment resulted in a striking hypertrophy of Leydig cells, to produce cells ultrastructurally identical to Leydig cells from adults. Since the target tissue of FSH is the seminiferous epithelium, the observed effects on Leydig cells by FSH treatment suggest that the secretion of factors by the seminiferous tubules may mediate the maturation of Leydig cells.

    Notes: Summary Employing electron-microscopic methods that help retain polyanionic materials, we describe the extracellular coverings of a sea urchin (Lytechinus variegatus) throughout ontogeny. The surface of the embryo is covered by a two-layered cuticle (commonly called the hyaline layer), which in turn is covered by a granular layer. The granular layer is retained after addition of alcian blue to the fixative solutions, and has not been previously described for any sea urchin. After hatching, the granular layer disappears, but the hyaline layer continues to cover most of the larval surface until settlement and metamorphosis. A few days before metamorphosis, the hyaline layer lining the vestibular invagination of the competent pluteus larva is replaced by a three-layered cuticle resembling that of the adult sea urchin. The hyaline layer covering the rest of the larva is evidently lost at metamorphosis during the involution of the general epidermis.

    Notes: Summary The ultrastructure of nerve cells and the finestructural organization of synaptic contacts have been investigated in the intestinal nerve in the snail Helix pomatia. Three types of nerve cells, occurring singly or in groups, can be distinguished on the basis of the ultrastructure of their perikaryon and content of granules. The peripheral output of these nerve cells has been verified by retrograde CoCl2 and NiCl2 staining. Both axosomatic and axo-axonic specialized synaptic contacts occur in the intestinal nerve. Presynaptic elements of these synaptic contacts contain 100–120 nm granular vesicles or 120–200 nm neurosecretory-like granules. Following intracellular horseradish peroxidase (HRP) labelling of identified central neurons responsible for peripheral regulatory processes, several labelled axons running toward the periphery can be followed throughout the branches of the intestinal nerve. These labelled axon processes (either primary axon or small collaterals) form specialized synaptic contacts, inside the intestinal nerve, and are always in a postsynaptic position. The occurrence of peripheral axo-somatic and axo-axonic synapses provides a morphological basis for integrative processes taking place in the intestinal nerve (peripheral nervous system) of Helix pomatia.

    Notes: Summary The eyespots of tornariae of enteropneusts (Ptychodera flava from Hawaiian waters and an unknown species from southern California) were studied by electron microscopy. An ocellus is composed of two types of cells: sensory and supportive. The former is characterized by a bulbous cilium (with 9+2 axoneme) at its distal end, one or sometimes two arrays of microvilli from its sides below the cilium, and a basal axon. The latter features large, clear vesicles which presumably contained the reddish-orange pigment seen in the ocellus of a living larva. Five-day old tornariae of P. flava are positively phototactic. Both cilium and microvilli may function as photoreceptors. The tornarian ocellus studied is compared with eyespots of other invertebrates, and the evolutionary significance of its putative photoreceptors is discussed.

    Notes: Summary The early spermatid possesses a round nucleus and a diplosome which is close to the cellular membrane and with which paracentriolar formations are associated; a sole mitochondrion is located between the diplosome and the nucleus. During the spermatid evolution, the nucleus is laterally elongated, its main axis being perpendicular to the flagellar axis; the mitochondrion migrates towards the lateral end of the nucleus; the distal centriole is extended along the nucleus, then leaves the cell as a pseudoflagellum. This centriolar extension is made up of regular triplets and of a particular type of doublets that we have called “A–C doublets“. The authors call attention to the similarity between the spermatozoa of this Elopiforme and the spermatozoa of the Anguilliformes. This observation favours Greenwood et al.'s proposition, to place these teleost into the super-order of Elopomorpha.

Die Mitochondrien sind rund und enthalten tubuläre Innenstrukturen, die Lipoidtropfen sind elektronenoptisch dicht. Während der Rückbildung des Corpus luteum setzt sich das endoplasmatische Retikulum aus ungeordneten Vesikeln und Tubuli zusammen, die Mitochondrien sind länglich oder verzweigt, die Lipoidtropfen elektronenoptisch hell. Die Bedeutung dieser feinstrukturellen Veränderungen in der Luteinzelle wird diskutiert.
    Notes: Summary The fine structure of the lutein cells in guinea pigs corpora lutea of pregnancy (15th, 35th, 45th, 50th, 55th, 63th day and 2 days after birth) and during oestrous cycle (9, 14, 16, 20 days after ovulation) is described. During the active phase of the corpus luteum the formation of concentric whorls of agranular endoplasmic reticulum around lipid droplets is observed, but later the granular endoplasmic reticulum increases. In this stadium the mitochondria are round with tubulous cristae and the lipid droplets are increased in density. During the involution of the corpus luteum the endoplasmic reticulum does not form concentric whorls, the mitochondria are elongated, polymorphic, the lipid droplets have lower electron density. These ultrastructural changes in the lutein cells are discussed concerning the role of the cell organelles in the steroid synthesis.

In der frühpostnatalen Proliferationsphase wandeln sich spindelförmige Zellen in polygonale Leydigzellen um. Dabei beginnt an einem Zellpol die Ausbildung des glatten endoplasmatischen Retikulums. Dieses Organell nimmt in der weiteren Entwicklung einen großen Teil des Zelleibes ein und bildet unregelmäßige Vesikel und konzentrisch geschichtete Zisternen. Es erfolgt eine Vermehrung der Mitochondrien, in ihnen entwickeln sich Tubuli. In der Phase der präpubertären Hypertrophie erreichen die Leydigzellen ihre größten Durchmesser (ca. 30 μm) und sind möglicherweise zu einer Steroidhormonproduktion befähigt. Da das glatte endoplasmatische Retikulum vornehmlich in der Zellperipherie, die übrigen Organellen bevorzugt im Zentrum liegen, sind die Zwischenzellen deutlich zoniert. Die Phase der präpubertären Regression ist durch ausgeprägte Zelldegeneration und Größenabnahme der überlebenden Zellen gekennzeichnet. In der Phase der pubertären Proliferation nehmen die Zwischenzellen erneut an Größe zu. Parallel hierzu vermehren sich Mitochondrien und glattes endoplasmatisches Retikulum. Beide Proliferationsphasen innerhalb der postnatalen Ontogenese werden durch das Auftreten zahlreicher Cytoplasmafilamente eingeleitet, deren Menge sich danach wieder verringert. Unabhängig von dem wellenförmigen Verlauf der Zwischenzellentwicklung ist die Ausbildung von Lysosomen, Microbodies, Mikrovilli, typischen Zellhaften und in der Zellperipherie gelegenen Polyribosomen. Die Leydigzellen sind von einer basalmembranähnlichen Struktur umgeben. Die Mitochondrien enthalten zu allen Zeiten durch elektronendichtes Material verkittete Tubuli.
    Notes: Summary The postnatal development of porcine testicular interstitial cells proceeds in a wave-like manner and may be divided into four periods. In the period of early postnatal proliferation fusiform intertubular cells change into polygonal Leydig cells. This process starts with formation of smooth endoplasmic reticulum in one of the cell poles. Later the smooth endoplasmic reticulum is abundant and consists of irregular vesicles and concentrically arranged cisterns. Mitochondria increase in number and develop tubular cristae. In the period of prepubertal hypertrophy Leydig cells reach maximal size (approx. 30 μm) and possibly may be capable of androgen biosynthesis. The cells of this period exhibit two different zones, since the smooth endoplasmic reticulum prefers the periphery, the other organelles, however, occupy the central region of the cells. The period of prepubertal regression is characterized by cellular degeneration and diminuation of the surviving interstitial cells. In the period of pubertal proliferation an increase of Leydig cell size is accompanied by multiplication of mitochondria and development of smooth endoplasmic reticulum. Both periods of proliferation are initiated by appearance of numerous cytoplasmic filaments which subsequently decrease in number. Interstitial cells of all periods studied exhibit the following features: They possess lysosomes, microbodies, polyribosomes, microvilli and are surrounded by a kind of basement membrane. Cell connections appear as tight junctions and desmosome-like structures. The mitochondria contain closely packed tubules joined by electron dense material.

Among those selectively stained sites, our studies have been focussed on: (1) The dense synaptic material consisting of several presynaptic clumps, termed projections, an intrasynaptic dense line and a subsynaptic web from which fine fibrillar wisps extend into the surrounding ground substance; (2) Neurofilaments and neurotubules, the surface of which is bristled by numerous side-arms; (3) A microfilamentous network intertwines the neurotubules, the neurofilaments and the mitochondria in the dendrites and axon, and is also connected to the E-PTA dense undercoating delineating the inner aspect of the plasma membrane and to the fine wisps emanating from the subsynaptic web. A three-dimensional microfilamentous latticework is thus formed in the nerve cell processes; (4) Dense cytoplasmic inclusions, termed nematosomes, which are usually located in the ground substance of the perikaryon among or in the vicinity of clusters of ribosomes. Tiny microfilaments emanate from the peripheral strands of these bodies. The presence of basic residues in the chains of structural proteins of which consist the subsynaptic web and the nematosome is plausible, since the specificity of the E-PTA staining procedure for the detection of basic residues has previously been put forth. The occurrence of a three-dimensional microfilamentous network in the nerve cell processes led us to hypothesize that it plays a role in translocation of materials. It may provide the motive force for the axoplasmic transport, for instance, with the neurotubules, as well as, plausibly, with the neurofilaments, serving as attachment sites and guideways.

    Notes: Summary Oogenesis was studied in adult Triturus vulgaris (Urodela) with the electron microscope. The oocytes investigated ranged between 50 μm and 1600 μm in diameter. Two types of yolk platelet formation were found. Since both types involve the incorporation of high numbers of pinocytotic vesicles they are believed to be of an extraoocytic origin. On the basis of the order of their appearance they were named primary and secondary yolk. Five different types of vesicles were found, which participate in a variety of activities, such as yolk formation and the formation of the Golgi apparatus. They originate from four different sources, namely the nuclear membrane, the cytoplasm in connection with ribosome-like particles, the Golgi apparatus and the plasma membrane through pinocytosis. The results obtained were discussed especially with respect to differences found between the anura and the urodela, such as the presence or absence of cortical granules or equivalent structures.

    Notes: Summary The retina of nudibranch eyes contains two types of large cells; pigment cells which comprise about two-thirds of the total, with unpigmented sensory cells making up the remainder. Both pigment and receptor cells carry microvilli on their distal borders, but no traces of cilia were observed among them. The cornea of the eyes of aeolid and dendronotid nudibranchs is composed of a single layer of small cells, unlike the dorids where the cornea is made up of one of more large cells. The latter contain nuclei comparable in size with those of the pigment cells in the retina, but are themselves unpigmented. The elliptical eyes ofAplysia contain three types of retinal cell; the pigment cells and two kinds of receptor cells. The “ciliary” receptor cells bear equal numbers of cilia (9+2) and microvilli, while the “microvillous” receptor cells carry long tufts of microvilli with only an occasional cilium among them. The proximal cytoplasm of the receptor cells inAplysia and the nudibranchs contains large quantities of the small spherical vesicles (averaging 660 Å in diameter) which appear to be characteristic of gastropod eyes.

    Notes: Summary 1. The ultrastructure of the corpora allata of last larval instars and adults of Oncopeltus was studied. The unpaired gland undergoes submicroscopic alterations and shows signs of degradation in old animals. The organ is partly covered and penetrated by corpus cardiacum tissue. Axons with different types of neurosecretory granules form synaptoid contacts with the corpus allatum cells. 2. “Dark” and “light” gland cells can be differentiated on account of the degree of electron density. The former predominate during the last larval stage and in the young imago, the latter in mature males and females. It is highly probable that the “light” cells are the active (i.e. hormone producing) ones and the “dark” cells the inactive ones. 3. The active cells are characterized by rough endoplasmatic reticulum (often in whorls), small amounts of smooth endoplasmatic reticulum and many multivesicular bodies. Abundant free ribosomes, a not particularly well developed Golgi apparatus, dense bodies, and cytolysomes are present in active and inactive cells. 4. The nuclei contain one to four prominent and variously shaped nucleoli, which show differences between adult males and females with respect to their location in the nucleus. 5. The corpus allatum cells of Oncopeltus are obviously engaged in extensive protein synthesis. Tangible structural indications for the manufacture of juvenile hormone were not observed. Possible sites of hormone release are discussed.

    Notes: Summary Changes of muscle weights, fiber diameters and ultrastructure were studied in the slow anterior latissimus dorsi (ALD) and in the fast posterior latissimus dorsi (PLD) of the chick three weeks after denervation and tenotomy, and after combined denervation and tenotomy of the two muscles. The slow ALD muscle becomes hypertrophic after denervation (Feng, Jung and Wu, 1962). Three weeks after nerve section, wet weights of ALD muscles are increased by 60% and fiber diameters become by 30% larger than those of contralateral control muscles. In spite of this hypertrophy, degenerative changes are seen in the ultrastructure, similar to those described in denervated atrophic muscles. Areas of dedifferentiation with autophagic vacuoles and aggregates of tubules are found in superficial layers of some fibers. Disintegration of Z lines and filaments along one or two sarcomeres occurs in a number of myofibrils, especially in muscles of young animals. In contrast to denervation alone, simultaneous denervation and tenotomy of the ALD muscles results in atrophy. Decrease of muscle weights and reduction of fiber diameters are similar as after tenotomy; in both cases muscle fibers waste by degeneration and atrophy of myofibrils. The fast PLD muscles underwent extensive atrophy in all three series of experiments. Corresponding atrophic and degenerative changes of ultrastructure were found in all instances.

Special attention was paid to the different types of boutons, and the mode of termination of the striatonigral connection was examined. Two different types of nigra neurons with deep nuclear membrane indentations were observed. The synaptic organization of rat substantia nigra is very complex. Six types of boutons, with many characteristics, could be differentiated: pleomorphicvesicle boutons (40%), elongated-vesicle boutons (10%), small-round-vesicle boutons (10%), large-round-vesicle boutons (20%), “clear-terminal” (15%) and dense-core-vesicle boutons (about 5%). Vesicle-containing dendrites were observed entering dendro-dendritic synapses. One to four days after large unilateral striatal lesions were made, a great number of degenerating boutons were seen in the ipsilateral substantia nigra, while the contra-lateral substantia nigra was entirely free of degeneration. The newly defined pleomorphic-vesicle boutons were found almost exclusively in the process of degeneration. The neuronal origin of the other bouton types is discussed.

    Notes: Summary The pecten oculi of the sparrow consists of capillaries, pigment cells and a superficial membrane. Because of the loose structure of the first two components broad intercellular spaces occur in the pecten. The capillary wall consists of endothelial cells and a perivascular membrane. The bodies of the endothelial cells are flattened, while the plasmalemma of both their surfaces (basal and luminal) is strongly folded and forms numerous microfolds with an average thickness of 700 Å. The height of the inner microfolds is 1.4–1.8 μm, the outer microfolds measure 1.3–1.6 μm. They lie densely packed side by side and are separated by recesses of the capillary lumen ca. 500 Å wide. Due to this the surface of the endothelial cell is increased by approximately 20-fold. The adjoining endothelial cells abut or overlap with margins, and are joined by the zonulae adherentes. Pigment cells form numerous processes and microvilli. Some rest on the capillary walls, while others penetrate the superficial membrane of the pecten or fill the intercellular spaces.

    Notes: Summary Vasa deferentia of rats chronically treated with high doses of guanethidine sulphate (30 or 60 mg/Kg/day i.p.) were examined using electron microscopic, fluorescence histochemical and pharmacological techniques. Counts of the axon population in segments of the proximal (urethral) end of the vas deferens showed a reduction to approximately 55% and 35% in the number of axon profiles after treatment for one week with the two dose levels respectively. In the same period only a few cell bodies in the hypogastric ganglion (from which most of the adrenergic innervation of the vas deferens arises) reached the stage of terminal degeneration. Although many axons showed some abnormalities, the number of axons observed in terminal stages of degeneration in treated tissue did not exceed, at any stage examined, the very small numbers observed in control tissue. Organ bath studies showed that the contractile response to transmural stimulation was lost fastest at the distal (epididymal) end of the treated vas deferens. These results have led to the conclusion that, in contrast to the degeneration of adrenergic axons produced by surgery or 6-hydroxydopamine, the sympathectomy produced by guanethidinein vivo involves theretraction of adrenergic axons prior to complete degeneration of the cell bodies.

    Notes: Summary Neuromuscular junctions, muscle fibers, and intramuscular nerve branches in crayfish opener muscles denervated for periods up to 368 days were examined with the electron microscope. The majority of the denervated muscles responded to stimulation of the isolated distal part of the motor axon with electrical activity and contractions. The neuromuscular junctions and muscle fibers appeared normal in such preparations: synapses and synaptic vesicles were present in the nerve terminals, and the organization of filaments and tubules in the muscle fibers was not distinguishable from that in normal muscles. In some preparations denervated for a long time, and in others denervated by tying the motor axon close to the muscle, the motor axons showed physiological and morphological evidence of degeneration. Synapses, when present in such preparations, showed few synaptic vesicles, which were often clumped. Muscle fibers showed signs of degeneration. These observations suggest that isolated distal motor axon segments can remain alive for at least a year, retaining normal structure. As long as the motor axon is viable, the muscle remains normal in appearance, but if the axon degenerates, the muscle becomes abnormal. A trophic interdependence between nerve and muscle is probably involved.

    Notes: Summary An electron microscopic investigation of the lutein cells of the blue fox was undertaken, based on the hypothesis that differences in plasma progesterone levels at differing stages of pregnancy might be reflected in the ultrastructural organization. Comparisons were made between corpora lutea taken from animals mated 1, 2, 5, 14, 18, 20, 28, 33, 36, 39 and 45 days after the estimated time of ovulation. Measurements of progesterone on plasma samples were performed by a rapid competitive protein-binding assay. During the period with increasing and/or high plasma progesterone levels, (i.e. 1 to 14 days after ovulation) the lutein cells are characterized by evenly distributed cisternal agranular ER, mitochondria with both tubular and lamellar cristae, and electron-dense lipid droplets. The abundant agranular ER is closely associated with the lipid droplets and mitochondria. During the period with declining plasma progesterone levels, the lutein cells present a different morphological picture: the agranular ER assumes the form of bundles of parallel tubules disposed in several planes. During the latest stages observed, these “bundles” are disrupted and most of the agranular ER become arranged in smaller concentric whorls. Both kinds of whorls regularly enclose lipid droplets, dense bodies and mitochondria. The regions between the whorls contain scattered cisternae of endoplasmic reticulum, ribosomes, mitochondria and lysosome-like dense bodies.

    Notes: Summary The paraganglionic plates of the harvestman, Trogulus, are neurohemal organs composed of a thick neural lamella facing a blood sinus, basal glial cells, and seven distinct types of axon terminals (A1, A2, B1, B2, B3, B4 and C) identified on the basis of elementary granule size, electron density, and structure of contents. The release of A1 type granules occurs by exocytosis into the extraaxonal space below the acellular neural lamella. The release of the content of C granules occurs after fragmentation into small electron dense vesicles. Clusters of these vesicles intermixed with clear vesicles pressing against the axolemma beneath the neural lamella, apparantly correspond to the sites of release.

    Notes: Summary Proximal to an experimental stenosis of the small intestine of rats and guinea-pigs a remarkable hypertrophy of the muscle coat develops 3–5 weeks after the operation. There is no increase in the length of the intestine but an overall increase in volume of the muscularis externa up to 10 times. This increase is accounted for by an increase in size and in number (by mitosis) of smooth muscle cells of both the longitudinal and circular layers. Bundles of newly-formed smooth cells appear in the serosa and are circularly arranged. In the hypertrophic smooth muscle cells of the circular layer the ratio of surface to volume is 0.80 (0.80 μm2 of cell surface for every μm3 of cell volume) as against 1.4 in the control muscle. The hypertrophic muscle cells have a highly developed sarcoplasmic reticulum and show a large number of nexuses. The density of innervation (number of axons per given number of smooth muscle cells) is smaller than in controls. Few collagen fibrils are visible in the extracellular space.

The characteristics of these are described and their development traced. The distribution of nerve terminals and neurons within the PI is documented. Electron microscopy shows the superficial layer of the epithelium to consist mainly of cells bearing microvilli. Below this, often very deeply, lie very many large cells, which bear numerous cilia. These cells open to the surface via narrow processes running between the epithelial cells. Although details of their innervation have yet to be established the large cells are assumed to be receptors and these findings are interpreted as evidence that this organ is indeed chemosensory.

    Notes: Summary Ultrastructural features of the subcommissural organ (SCO) cells in larvae (stages 56–58, according to Nieuwkoop and Faber, 1956), toadlets (3 months after metamorphosis) and older toads (2-year old) of Xenopus laevis are described. Several age-related morphological differences in the endoplasmic reticulum (ER) of the SCO cells have been found. In old toads the rough ER assumes a special “ladder-like” membrane configuration in its cisternal lumen. By means of the periodic acid-chromic acid-silver methenamine (PA-CrA-SM) method, complex carbohydrates are detected electron microscopically in the SCO cells. Positive reactions take place in the cell adhesive apparatus, the secretory granules, part of the Golgi complex, and the intracisternal “ladder-like” structure. Passing through the Golgi complex, the secretory products mature into the secretory granules by association of their proteinaceous component with polysaccharides. The majority of the secretory granules are released from the apical cell surface by means of reverse pinocytosis, while the rest are released through the basal process into the blood circulation.

    Notes: Summary The innermost layer of the circular musculature of the ileum of the guinea-pig, mouse, rat, rabbit, cat and dog is made of special small and dark muscle cells (sd cells), which can be identified by light and electron microscopy. These cells have a higher surface to volume ratio than ordinary muscle cells. They show all the organelles of smooth muscle cells and are in close relationship with a great number of extrinsic and intrinsic nerve fibres. They send numerous thin cytoplasmic processes towards the bulk of the circular layer, but nexuses between the sd cells and ordinary muscle cells are not found. It is suggested that some of the nerve fibres in this part of the circular layer are motor, setting the sd cells to a given length, and some are afferent in luminal pressure.

    Description / Table of Contents: Zusammenfassung Im Auge von Ephestia kommen Kontakte zwischen den Zellen benachbarter Retinulae vor. Diese Kontakte finden sich am häufigsten in der Kernregion der Retinulazellen und können 2 oder mehr Retinulae miteinander verbinden. Da keine besonderen Membranstrukturen in der Kontaktzone beobachtet wurden und da andererseits die Retinulazellen auf diesem Niveau und weiter distal Pigmentgranula enthalten, scheinen diese Kontakte weder die Zellen chemisch bzw. elektrisch zu koppeln noch Reizlicht zu übertragen. Vielmehr wird angenommen, daß die Verlagerungen des Cytoplasmas bei Hell-Dunkeladaptation in benachbarten Retinulae durch die gegenseitige Berührung koordiniert werden.
    Notes: Summary Connections were found between retinula cells of adjacent retinula cell columns in the eye of Ephestia. The connections occur most frequently at the level of the retinula cell nuclei and may involve two or more retinula columns simultaneously. The absence of specialized structural modifications of the membranes and the presence of pigment granules at the level of the connections or distal to them indicates that these connections are probably not involved in selective chemical or electrical communication nor in light transmission. It is suggested that the connections may serve tactily to coordinate cytoplasmic movement in adjacent retinula columns during light-dark adaptation.

    Notes: Summary As revealed by light microscopical investigations the human Sertoli cell presents different appearances according to the pattern of infranuclear cytoplasmic inclusions. Although two or three stages of spermatogenesis are seen in a single cross section of a seminiferous tubule the Sertoli cells all show virtually the same features in such a cross sectioned tubule. The different appearances are also evident under the electron microscope. Although no obvious correlation was found with the stages of spermatogenesis in the seminiferous epithelium, the Sertoli cell appearances described here may be assumed to represent different metabolic situations. Other features of Sertoli cell ultrastructure are discussed such as the presence of residual bodies in the apical cytoplasm, glycogen-rich areas protruding towards the tubular lumen or the extracellular space, and membrane bound, round structures, found between the membranes of the smooth endoplasmic reticulum and resembling the “microbodies” of steroid producing cells.

    Notes: Summary The effect of bile duct ligation on the intercellular junctions of hepatocytes was investigated. The features and the arrangement of the bile canaliculi and the zonulae occludentes alter concomitant to the increase of the intracanalicular pressure. The lumen of the bile canaliculi enlarges and the microvilli disappear. The array of the zonulae occludentes becomes irregularly shaped, the number of strands diminishes and interruptions of the strands occur. With peroxidase a leakage in the bile-blood barrier is detected. Furthermore a disappearance of gap junctions between the hepatocytes after bile duct ligation is observed. The present investigation shows that the zonulae occludentes are mobile structures which are changed by increased unilateral pressure. Après la mue imaginale, les cellules glandulaires ont toujours une activité sécrétrice. De plus, chez les soldats femelles et les reines, elles contiennent de nombreuses formations cristallines d'origine mitochondriale. Le rôle de ces glandes (sécrétion de salive ou de phéromone) est discuté.
    Notes: Summary The mandibular glands of Kalotermes were examined in different castes. They showed sexual dimorphism in the soldiers and primary reproductives. Moreover, in female soldiers and queens, mandibular gland cells contained numerous crystalline structures of mitochondrial origin. The role of these glands (secretion of saliva or pheromone) is discussed.

    Notes: Summary Nerve profiles in the basal part of the palisade zone of rat median eminence have been divided into 4 categories on the basis of their vesicular content: profiles a with agranular vesicles of about 50 nm, b with similar agranular vesicles and also with granular vesicles of mainly 70–140 nm, c with granular vesicles alone and d without vesicles. After intravenous injection of exogenous peroxidase, castrated male rats showed, in comparison to sham-operated controls, a 3–5 fold increase in the percentage of nerve profiles of category a which contained peroxidase loaded vesicles of about 50 nm. In contrast, no significant change was observed for nerve profiles of other categories. It is concluded that exogenous peroxidase may be used to demonstrate changes in the activity of tuberoinfundibular nerve terminals at the ultrastructural level.

    Notes: Summary This report describes histochemical and ultrastructural studies of tail muscles in tadpoles of Rana japonica and Rana catesbeiana during metamorphosis, this process being accompanied by degeneration of the tail. Degeneration of individual tail muscles does not occur at the same time; this is true for both the red and white muscle fibres. The initial phase of degeneration showed mesenchymal macrophages first invading the muscle fibres and then sending out many long cytoplasmic processes which split the fibres apart. The disappearance of myofibrils during degeneration proceeds along at least two different mechanisms even within a single muscle fibre. In one type, the Z-band becomes diffuse and then disappears, resulting in fragmentation of the myofibrils at the sites previously occupied by the Z-bands. The second pattern of degeneration is characterized by disappearance of the Z-band followed by a fanning out of the myofilaments not associated with fragmentation of myofibrils. As atrophy of muscle fibres proceeds, acid phosphatase activity is localized in the perinuclear sarcoplasm. Macrophages show more intense acid phosphatase activity than do the muscle fibres. The formation of autophagic vacuoles is described and discussed.

    Notes: Summary Five-week old Japanese quail held under short daylengths (8L/16D) were injected daily for two weeks with 20 μg/day of purified chicken luteinizing hormone (LH) or with one of two chicken follicle-stimulating hormone (FSH) fractions. Both intact and hypophysectomized birds were used. After killing, the testes were weighed and fixed for light- and electron-microscopy. Following hypophysectomy testicular weights fell significantly below those of saline-injected controls, indicating that short day conditions are not equivalent to “physiological hypophysectomy”, the pituitary-gonadal axis continuing to function at a low level. Chicken LH stimulated differentiation of the interstitium, producing mature Leydig cells containing the full complement of organelles typical of steroid-secretion. In addition it induced some testicular growth caused by spermatogonial division and partial differentiation of the Sertoli cells. This effect was assumed to be a secondary response to androgen secretion but a direct action of chicken LH on the tubules is not dismissed. LH given to intact quail caused greater testicular growth, presumably due to synergism with endogenous hormones. Both chicken FSH fractions induced testicular growth and development; in intact birds the effects were similar to those observed in intact LH-treated birds. In hypophysectomized quail given FSH the testicular weights were greater than in hypophysectomized LH-treated birds and the seminiferous tubules were larger. Sertoli cells were stimulated and pachytene spermatocytes were numerous. Leydig cell differentiation was minimal in all FSH-treated birds, being less pronounced in hypophysectomized than in intact birds.

    Notes: Summary An attempt has been made to discriminate additional types of synapses than have been previously described in the nucleus accumbens septi of the cat, which can, according to Brockhaus (1942), justifiably be termed the fundus striati due to the fact that it possesses all of the morphological and some of the neurochemical features of the striatum. This was undertaken in order to correlate at least one type of synapse with each different afferent pathway. Nine distinct types of synapses could be differentiated electron microscopically: Type I: axo-spinous synapses with sparse, small, round vesicles which seemed to be the nigro-striatal endings (35%). Type II: axo-somatic or axo-dendritic en passant synapses containing small, round vesicles (3%). Type III: axo-spinous synapses filled with densely-packed, small, round vesicles displaying strong postsynaptic thickenings which seem to be corticostriatal (17%). Type IV: large axo-spinous synapses with densely-arranged, small, round vesicles contacting larger spines branching off a pedicle (9%). Type V: axo-somatic or axo-dendritic synapses containing large pleomorphic vesicles, probably axon collaterals (1%). Type VI: axo-somatic or axo-dendritic synapses with elongated small vesicles (20×45 nm) (3%). Type VII: large axo-somatic or axo-dendritic synapses filled by denselypacked, small, round vesicles (11%). Type VIII: large axo-somatic or axo-dendritic synapses containing loosely-arranged, small, round vesicles (8%). Type IX: axo-somatic or axo-dendritic synapses containing large, round vesicles in a translucent axoplasm (13%).

    Notes: Summary The adrenal glands of decapitated and encephalectomized fetal rats were investigated electron microscopically and compared to those of normal intact fetal rats. Although the adrenal cortices did not show three zones (zona glomerulosa, fasciculata, and reticularis) on the 16.5th day of gestation when the decapitation or encephalectomy was carried out in utero, the zonation was recognized in fetuses operated on the 21.5th day of gestation. The same was true for normal control fetuses. However, cytoplasmic characteristics suggesting steroidogenesis in the cortical cells were reduced to various degrees in the encephalectomized or decapitated fetuses, especially in the latter ones. The change in cytoplasmic appearance was more conspicuous in the inner portion of the cortex. This result suggests that for the maintenance of normal adrenocortical function the hypothalamus may be indispensable even during the prenatal life of rats.

    Notes: Summary To study the penetration of protein from the blood into and through the pars intermedia of anurans, the electron-dense tracer, horseradish peroxidase (HRP), was injected into the vascular system of the frog, Rana pipiens. Pinocytotic vesicles were seen along plasma membranes of the stellate cells abutting the pericapillary spaces between the pars intermedia and pars nervosa. The HRP was seen within both the pericapillary spaces and these pinocytotic vesicles. In addition, the HRP was very prominent in the extracellular spaces between adjacent stellate and/or MSH cells and around nerve endings and thus, appeared to move from the vascular spaces and into the interior of the pars intermedia. Therefore, the poorly vascularized pars intermedia may have such a route for transfer of materials throughout the gland.

    Notes: Summary The ultrastructure of the corneal nerves of the rat was studied in tissue fixed by immersion in and by perfusion with glutaraldehyde-containing fixatives. Of the four types of axonal terminal identified in the nerves, those with the features of adrenergic and cholinergic terminals were confined to the nerves at the limbus and were concentrated in the perivascular plexuses. The remaining two types of terminal were found on axons located in all parts of the cornea and on both intraepithelial axons and axons in the stromal nerves. Of these, one contained the numerous mitochondria which occur in the terminals of axons associated with known mechanoreceptors and the second contained variable and often small numbers of both clear and large dense-cored vesicles. While most of the mitochondria-containing terminals were seen in nerves located near the periphery, vesicle-containing terminals were numerous in all of the nerves, and especially in those in the avascular cornea. In material fixed by immersion in glutaraldehyde-paraformaldehyde, the vesicle-containing terminals appeared to be dilated, but in material fixed by perfusion there was little evidence of any increase in the diameter of the axons in the terminal regions. The structure of the terminals was compared with that of the terminals of axons identified in the nerves of the skin and the urinary tract and the differences in the vesicle content of the terminals to those reported in other studies of the corneal nerves was related to the use of different fixation procedures. The possibility that axons possessing such terminals are identical with the beaded axons and both the cholinesterase-positive and fluorescent axons demonstrated in light microscopical studies of the corneal nerves is discussed, and the widespread distribution of the axons in the cornea is equated with the hypothesis that they are afferent in nature and represent the peripheral receptors for pain impulses.

    Notes: Summary The fine structure of the corpus epididymidis of the rabbit has been studied following organ culture. Various modifications of tissue preparation and culture conditions were examined to obtain good maintenance of cellular integrity as well as to preserve sperm fertilizing ability. After 5 to 7 days in culture in the absence of hormonal support, the epididymal epithelium showed signs indicative of cellular regression. Such changes included shrinkage of the cells, loss of the border of stereocilia, decrease in smooth endoplasmic reticulum, and an increase in autophagic vacuoles. The presence of androgens in culture media prevented cellular regression to varying degrees, depending on the hormone utilized. With regard to maintenance of cellular integrity, potency of the androgens tested was as follows: 5α-dihydrotestosterone 〉= 3α-androstanediol 〉 testosterone 〉 3β-androstanediol. Addition of insulin to dihydrotestosterone-containing cultures resulted in no improvement in maintenance. Phagocytosis of spermatozoa by epithelial cells was observed in cultured tubules and the degree of spermiophagy was inversely proportional to successful maintenance of fine structural characteristics of epithelial cells. The morphological findings reported here correlate well with the fertilizing ability of spermatozoa from cultured epididymis as reported in an accompanying communication.

    Notes: Summary The neurosecretory system of the freshwater snail Bulinus truncatus was investigated. With the Alcian blue-Alcian yellow (AB/AY) staining method at least 10 different types of neurosecretory cells (NSC) were distinguished in the ganglia of the central nervous system. The differences in staining properties of the NSC — with AB/AY the cells take on different shades of green and yellow — are borne out at the ultrastructural level: the NSC types contain different types of neurosecretory elementary granules. The neurosecretory system of B. truncatus is compared to that of Lymnaea stagnalis, the species which has received the most attention among the pulmonates. It appears from the comparison that the systems of both species show many similarities, although some differences are also apparent.

    Notes: Summary Electron microscopic examination of normal human testicular tissue revealed annulate lamellae (AL) in the cytoplasm of primary spermatocytes and spermatids. AL of primary spermatocytes are encountered in the perinuclear region, parallel to the nuclear envelope and form single or multiple membranous profiles containing numerous annuli (500–600 Å in diameter) frequently associated with a fibrillar electron dense material. Spermatids contain numerous layers of AL either continuous with the nuclear envelope and caudal to the acrosome or peripherally positioned in the cytoplasm. Individual lamellae possess terminal dilations and display continuities with the endoplasmic reticulum. The interlamellar space in spermatid AL is entirely filled with a fine granular electron dense material. Additionally, the break-down of AL in spermatozoan residual bodies is indicated by a dilation of AL cisternae to form vacuoles following the dissolution of pore complexes.

    Notes: Summary Blue-green fluorescent subependymal cells with intraventricular processes were shown by the fluorescent histochemical method to be distributed from the preoptic recess to the infundibular recess of the frog hypothalamus. Electron microscopy revealed at least two types of CSF-contacting subependymal cells, type 1 containing large dense granules (about 100–200 nm in diameter) and type 2 containing small dense core vesicles (about 60–100 nm in diameter). Subsequent to fixation in permanganate solution, the small dense core vesicles in type 2 cells reacted with the fixative and consistently showed a dense content. However, the large granules in type 1 cells were mostly pale or less dense after this fixation. Two hours after intraventricular injection of 3H-dopamine, a large number of silver grains appeared only in the cytoplasm of intraventricular processes possessing dense core vesicles (type 2 cells). A few grains were also found in the perikarya. It is concluded that type 2 cells are catecholamine-storing cells. It is suggested that type 1 cells in the infundibular recess are peptidergic neurons which may secrete some hypothalamic regulating hormones of the anterior pituitary. Most of these cells in the preoptic recess belong to the neurosecretory cells of the preoptic nucleus, while some cells probably function similarly to those in the infundibular recess.

    Notes: Summary In addition to a submuscular and subepithelial nerve plexus an infra-epithelial nerve-net also occurs in Notoplana acticola. It contains naked nerve cells which lie between the bases of epithelial cells. Individual neurites occur separately or in small tracts. Synapses contain clear spherical vesicles and are polarized. Post-synaptic neurites tend to be flat and ribbon-like. Rhabdite and mucus secreting cells in the epithelium have synapses associated with them. Neurites ending on the rhabdites appear to originate in the subepithelial plexus. The “sensory-free nerve endings” of earlier workers are reinterpreted as being motor terminals on epithelial secretory cells.
