    Notes: Serological and genetic analyses of H-2 antigens indicate that each K or D region allele controls two highly polymorphic antigenic sites, the alpha site corresponding to the private specificity and the gamma site corresponding to the long public specificities. Studies of cell surface distribution and biochemical characteristics of the alpha type specificity H-2.4 and gamma type specificity H-2.28 in the product of a D region allele, Dd, demonstrate that these specificities are carried on two different polypeptide chains. Accordingly, two distinct and polymorphic genes are postulated to code for the product controlled by the D region of H-2.

    Notes: Twenty-six combinations of C57BL/10 congenic mice involving incompatibilities in the I region and the H-2K or H-2D regions of the major histocompatibility complex were studied in a cell-mediated lympholysis assay (CML). All combinations were positive; however, six were unidirectional. Three of these were consistent with the mapping of an effector cell stimulating locus (ECS) between the IB and IC sub-regions. Possible reasons are given for the negativity of CML in I/H-2K or H-2D region incompatible combinations. A statistical method used to analyse the data is described.

    Notes: H-2 typing of two inbred mouse strains, GRS I/A and LIS/A, revealed that they both carry the same, previously unknown haplotype, designated H-2dx. The H-2dx haplotype has the K region allele identical or very similar to that of H-2d, however it differs from H-2d at the I region genes and has none of the D region alleles previously described in inbred mouse strains.When the H-2dx haplotype was used in F1 complementation tests, it failed to prevent rejection of B10.D2 skin grafts by (GR × B10.A)F1 and (LI × B10.A)F1 recipients, as well as rejection of R103 grafts by (GR × 2R)F1, (LI × 2R)F1, (GR × B10)F1, and (LI × B10)F1 recipients. In all these combinations, the donor and the recipients were matched at the K and D regions, but differed at the I region. The mean rejection time was 12 days (beginning) and 15.9 days (end of rejection).The second set skin grafts were rejected at approximately the same speed as the first set skin grafts, and after their rejection a cytotoxic antibody, most likely anti-Ia, was found in the serum of the recipient mice. The absence of accelerated second set skin graft rejection in the I region incompatibility observed in these experiments is in sharp contrast to the extremely fast rejection of K or D region incompatible second set skin grafts. This lack of demonstrable anamnestic response is either due to the presence of enhancing antibodies, or to an inherently inferior capacity of I region incompatibility to elicit, in vivo, an effective anamnestic response.

    Notes: The CBA/H mouse lymphocytes which divide when cultured together with allogeneic blood in mixed lymphocyte reactions (MLR) have been shown to be mainly of thymic origin with the aid of thymus grafted radiation chimaeras. By adaptation of a system used previously to quantity mitogen responsive cells, the relative number of cells in CBA/H mouse blood capable of responding in the MLR, and the effects of preimmunizing CBA/H mice with allogeneic cells have been quantified. In mixed lymphocyte reactions between parental and allogeneic F1 hybrid cells, substantial numbers of the latter cell type have been found in division. Attempts to elucidate the nature of these reacting F1 cells indicate that they are probably mainly of bone marrow origin and that they are responding in a non-specific manner.

    Notes: The immunogenicity of Thy-1 antigens was examined using the Thy-1 congenic mouse strains A and A/Thy-1.1. A disequilibrium of response was found between these strains. A strain mice rejected skin grafts and produced high titres of antibody to lymphoid cells from A/Thy-1.1 strain mice. However, in the reverse combination grafts were not rejected nor was there any significant antibody response. A/Thy-1.1 mice did not respond to Thy-1.2 antigen when it was presented on an H-2 allogeneic background (A·SW strain) but did respond to it when non-H-2 incompatibilities were involved (B10·A strain donor). We postulate the existence of Ir gene(s) for the response to Thy-1.2 antigens which are linked to H-2. The failure of A/Thy-1.1 strain mice to respond to Thy-1.2 antigens in the absence of carrier determinants is the result of a deletion (in the germ line) of the appropriate Ir gene itself resulting from the long association of the H-2a haplotype of the A mouse (donor of the genetic background of A/Thy-1.1 strain) with the Thy-1.2 gene.

    Notes: A group of Australian aborigines with a high frequency (15.5%) of asymptomatic Australia antigen (Au) carriers shows a statistically significant deficiency of W15 among Au carriers. The association was observed during an HL-A survey, and was confirmed by a further study designed to investigate the observed association. Further, the pooled data establish the association.The observed association can be explained satisfactorily in terms of an Au immune response locus closely linked with, and having alleles in linkage disequilibrium with alleles of, the Four locus of the HL-A system. Since a deficiency of W15 among Au carriers has been reported in a northern European study, though it has not been observed in others, it is argued that the association has not occurred by chance, but that selection involving both HL-A and immune response genes has been responsible for the observed association.A significantly high proportion of Au carriers were observed to be homozygous for antigens of the Four series. The data, and their interpretation, lend support to the proposition that asymptomatic Au carriers are homozygous for an inactive allele of the gene responsible for the immune response to Au.

    Notes: Studies of 521 sera from the Icelandic cousin marriage project were made to assess the incidence of various anti-tissue antibodies and the levels of immunoglobulins, as these were considered to be useful markers of the humoral immune response. Comparisons were made between these parameters and the HLA-A and B antigens, the blood groups, the immunoglobulin allotypes (Gm, Km and Am), the properdin factor (Bf), and other markers.These investigations offered another approach to the study of the sites of action of immune response genes in man. Because the immune response may be expected to differ for each individual and depend at least in part, on the degree of exposure to different antigens, no absolute correlation was expected. There was, however, a marked association between certain IgG anti-tissue antibodies and HLA antigens. This was most marked for HLA-A10, B18 and B27, but not for HLA-A1 or B8.The comparison of immunoglobulin levels with HLA antigens, was less striking, although HLA-A2 appeared to be associated with low levels of IgE. There were also some associations between immunoglobulin levels and ABO blood groups.

    Notes: The polymorphism of histocompatibility antigens is usually explained by classical Mendelian laws, which allow the inheritance of one allele per locus. Thus each individual would express two antigens for each locus of the pair of relevant homologous chromosomes, one from each parent.Another explanation is that the structural genes of all the different haplotypes are all in tandem. Regulatory genes would then determine the polymorphism by mimicking Mendelian inheritance–allowing only the relevant gene products for a particular haplotype to be expressed. If correct, one might expect the mechanism to fail sometimes, and silent genes of foreign haplotypes to be derepressed, allowing H-2 antigens of the wrong haplotype to become expressed.In the light of this hypothesis (Martin, 1975; Amos, 1971; Bodmer, 1975; Festenstein, 1978; Garrido et al., 1976a,b), the original findings in our laboratory of extra foreign ‘H-2-like’ determinants on tumour cells passaged in vitro and in vivo with and without virus are considered rather important (Festenstein, 1978; Garrido et al., 1976a,b); not only because they could lead to a better understanding of the genetic basis of MHS polymorphism, but also because of its implication for the surveillance of tumours (Garrido et al., 1976b; Gomard et al., 1977; Blank et al., 1976; Parmiani & Invernizzi, 1975) and microbially damaged cells (Doherty & Zinkernagel, 1975). These extra determinants were tested by various serological techniques and assays of cell mediated immunity.

    Notes: The lymphocytes from 107 maternal-foetal pairs were examined for their in vitro responsiveness, as determined by the incorporation of tritiated thymidine following stimulation with phytohaemagglutinin (PHA), candida, varicella, mumps, streptokinase-streptodornase (SKSD) and tetanus toxiod. The data were collected and analysed in two sequential groups (forty-seven and sixty) in order to determine whether the results were reproducible. The variable chosen for analysis was the difference (d) between the square roots of the isotope incorporation in the stimulated and control cultures because it gave the most symmetrical distribution of the data. The experimental error in the determination of maternal lymphocyte stimulation was 1.4-8.6% and of the and of the foetal lymphocytes, 1.0-16.6% depending upon the antigen or mitogen and its concentration. The data in the two sets of patients were statistically the same in forty-eight of the fifty-six analyses (fourteen antigen or mitogen concentrations in autologous and AB plasma for maternal and foetal lymphocytes). The statistical limits of the distribution of responses for stimulation or suppression were set by an analysis of variance taking two standard deviations from the mean as the limits. When these limits were translated into stimulation indices, they varied for each antigen or mitogen and for different concentrations of the same antigen. Thus, a detailed statistical analysis of a large volume of lymphocyte transformation data indicates that the technique is reproducible and offers a reliable method for determining when significant differences from control values are present.

    Notes: An unexpected MLR reaction has been observed between three HLA-identical sibs; it consists of a bidirectional positive MLR between identical female twins and a sister. No argument for a lymphoid mosaic could be found, although twins were frequent in the family; similary no HLA-A/B or HLA-B/D recombinant could be demonstrated.The MLR, although weak, was highly reproducible. PLTs could be raised between the sibs, without an apparent segregation in this family nor in five other families, but such PLTs discriminated well between the positive and negative controls. In the absence of any proof that such a weak MLR locus could be on another chromosome than chromosome 6, two lines of argument are indirect evidences that such a locus could be indeed on chromosome 6: one of the sibs differs from the two others for two markers outside HLA-D-DR-Bf: glyoxalate (GLO) and red blood group P.

    Notes: It is well known that the major histocompatibility system (MHS) has unique properties pertaining to antigenic strength (Démant, 1973). In the 1973 and 1975 Workshops we investigated alloimmune interactions to try to find out which MHS genes are involved and how they confer antigenic strength. We found that in several models there are special antigenic strength conferring and/or controlling genes, that there are probably several such genes unique for each model, situated throughout the mouse MHS (H-2 complex). For example, the cell mediated lympholysis (CML) controlling genes, the Effector Cell Stimulating (ECS) genes, conferring strength against K and D region antigens are in the I region but separate from the I-A Lad genes (Festenstein et al., 1974) while the allograft controlling genes, for various rejection models, are in the D region as well as the I region (Festenstein et al., 1975). This is an attempt to bring together the data from these various models to formulate a functional concept of antigenic strength, in relation to its genetic basis.

    Notes: Further evidence supporting the hypothesis that the autoimmune disease of the NZB results from failure of T cell mediated autoimmune suppressor function has been obtained from ovum fusion-derived tetraparental NZB ? CFW chimaeras. Although the issue was complicated in three chimaeras by an acute interaction between the two parental strain cell populations, one animal remained well and no evidence of disease was found at postmortem. Electron microscopy demonstrated large numbers of C-type particles in all the chimaeras and renal changes of an acute type rather than chronic NZB disease in two of the animals. Erythrophagocytosis was observed in all the chimaeras and was probably initiated by an antiautoantibody receptor-determinant reaction. The phagocytosis of plasma cells suggested that the reaction was also directed against the clonal source of the autoantibody. A similar receptor-determinant reaction may be responsible for the ‘normal’ elimination of aberrant autoimmune clones.

    Notes: In a population of 164 very closely related members of a Swiss alpine village, the HL-A system and seven other immunogenetic systems were studied. The two main techniques of lymphocytotoxicity and leuco-agglutination were used in two different laboratories for the HL-A serology. The W4(4a) and W6(4b) antigens are especially emphasized, and discrepancies between the two techniques discussed. The use of large families proved valuable for the definition of several previously ill defined anitgens.

    Notes: An allotypic system of rabbit serum high density lipoprotein (HDL) has been reported previously from our laboratory. Homospecific rabbit sera revealed an antigen termed Hl 1, which is governed by autosomal dominant inheritance.An unexpected degree of cross-reactivity is described in this paper. Rabbit anti-Hl 1 sera were found to cross-react with a component present in pig serum. This component was found in all pig sera tested thus far, and there is therefore no evidence that it reflects an allotypic system in the pig.Fractionation in the preparative ultracentrifuge revealed that the component in pig serum resides in the HDL fraction. The antigenic resemblance between the component in pig serum and the rabbit Hl 1 antigen is considerable, as judged from experiments with antisera available at present. Immunological cross-reactivity of the described type may be more wide-spread than hitherto assumed.

    Notes: Experimental data are discussed which indicate that one alkali-labile tetrasaccharide present on each MN-glycoprotein subunit is part of the M- and N-antigen receptor site. It is concluded therefore that the difference between M- and N-antigens resides in the amino acid sequence of the glycoprotein. Based on the different topographical distribution of the MNSs-receptors within the membrane an explanation for the cross-reaction of anti-N reagents with ordinary M-erythrocytes is proposed.

    Notes: Loss of antigenicity in the mixed lymphocyte culture (MLC) reaction of lymphocytes precultured at 22°C for 7–10 days was accompanied by a decrease in bone-marrow-derived lymphocytes (B cells) from 22 ± 1% to 13 ± 1%, and an increase in thymus-derived lymphocytes (T cells) from 65 ± 2% to 83 ± 1% (P 〈 0.001). Depletion of B cells from a fresh lymphocyte suspension by either antihuman immunoglobulin-coated column fractionation or by sheep red blood cell (SRBC) rosette formation resulted in a significant reduction of the cell's ability to stimulate in MLC (P 〈 0.001). Coating of lymphocytes with rabbit antihuman brain serum abrogated their ability to respond but not the ability to stimulate in MLC.

    Notes: The present study has shown that the intramuscular injection of human red cells into baboons can often produce long lasting antibodies of the IgG variety. The intravenous administration of human red cells into baboons sometimes fails to stimulate the production of antibodies to the first injection but after a second injection short duration IgG and IgM antibodies are generally observed. A similar pattern of antibody behaviour was also seen in a postnatal follow-up study of Rh immunized women. Of significance was the finding that some women were able to make long lasting high titred Rh antibodies whilst others frequently produced antibodies which decreased in intensity shortly after pregnancy. It is postulated that the formation of long lasting high titred Rh antibodies may be due to the intrauterine trapping of fetal Rh incompatible cells during parturition, while the formation of antibodies which show a marked decrease in intensity may result from the passage of fetal cells into the maternal blood circulation. Different in vivo pathways of antibody formation are indicated for the two modes of red cell antigen presentation.

    Notes: We are using the terms ‘low’ and ‘high’ expression when the H-Y antigen on thymocytes of mouse parental strains is, respectively, weakly and strongly immunogenic for F1 hybrids in a host-versus-graft popliteal lymph node enlargement assay. Using this assay we attempted to locate the genetic factor(s) responsible for the control of H-Y expression within chromosome 17 since our previous study indicated linkage with the H-2 complex. We produced to this aim recombinants from (B10.T x C3H/Di)F1 hybrids where crossing over took place between H-2 and T complexes. In this way we were able to locate a gene denoted Hye (for H-Y expression) whose ‘high’ and ‘low’ alleles originated, respectively, from the B10 and C3H/Di strains. In the recombinants, the expression did not follow the H-2 haplotype, but obviously depended on the position of crossing over between H-2 and T. This pointed to the Hye gene mapping proximally from the H-2 complex.

    Notes: We have initiated studies to determine whether the antibody and T-lymphocyte proliferative responses to lysozyme and its antigenic sites is genetically controlled in mice. Mice of the H-2f, H-2k and H-2p were high responders, while haplotypes H-2b, H-2d, H-2r and H-2s were low responders. Studies with recombinants indicated that the immune response is controlled by two H-2I region loci, one being in the I-A subregion and the other may be in the I-C subregions.

    Notes: The genetics of salivary secretion of ABH blood group substance was investigated. Eight secretor phenotypes were determined by Sephadex G-100 chromatography. Presence or absence of three different fractions were shown to be controlled by three different secretor gene loci. These loci are called Sec1, Sec2 and Sec3. Sec1 controls expression of the traditional secretor status. Sec2 and Sec3 are linked, but distinct, loci controlling the expression of low molecular weight ABH substance in human saliva. A dominant mode of inheritance for the Sec2 and Sec3 systems is described.

    Notes: MCG4 is a BALB/c sarcoma induced as a solid tumour with 0.2 mgs of methylcholantrene. The primary tumour was serially transplanted subcutaneously in syngeneic mice. The ascites form obtained was used to study the expression of H-2 antigeneic specificities in a postlabelling radioassay.MCG4 did not express H-2D.4 (private specificity of H-2d haplotypes) as well as H-2.3, H-2.8 and H-2.13 (public specificities). In addition it expressed H-2.5 (a public specificity not present in H-2d cells). These results were confirmed by quantitative absorption analysis using MCG4 and positive-negative normal lymphoid cells for a particular specificity.Results are discussed with regard to the control of expression of H-2 antigens by regulatory genes.

    Notes: The response of 105 maternal-foetal lymphocyte pairs to specific and non-specific stimulation were evaluated using a newly defined method of analysis. There were no significant differences in the responses of maternal or foetal lymphocytes to phytohaemogglutinin (PHA) or the various antigens as a function of concentration over the ranges tested. The maternal lymphocytes were stimulated by all of the antigens and responded to PHA three-five times more strongly than to the antigens. The foetal lymphocytes were stimulated by PHA and tetanus toxoid only and were suppressed by streptokinase-streptodornase (SKSD). They responded to stimulation by antigens at a lower level than did the maternal lymphocytes, but they responded at a much higher level to PHA. Unstimulated cultures of foetal lymphocytes incorporated more isotope than did those of maternal lymphocytes in both autologous and AB plasma.The data were cross-classified to determine whether the responses of the foetal lymphocytes varied concordantly with the responses of the maternal lymphocytes in both autologous and AB plasma by the Chi-square test for independence and by rank correlation analysis. There was no significant correlaiton in either plasma to stimulation with the antigens. Thus, the presence of antigen reactive lymphocytes in the circulation of the mother does not mean that the foetus is sensitized to that antigen.Comparison of the lymphocyte responses in autologous plasma with those in AB plasma provided evidence for the presence of circulating immunoregulatory substances. Autologous maternal plasma suppressed the lymphocyte responses to high concentrations of candida and SKSD and stimulated the response to mumps, varicella and tetanus toxoid. Autologous fetal plasma suppressed the lymphocyte responses to candida, varicell and SKSD and stimulated the response to PHA. The responsiveness of maternal lymphocytes to PHA was less in foetal plasma than in autologous maternal or AB plasma.

    Notes: Passive enhancement of H-2 incompatible skin allografts was studied in systems where the enhancing serum covered only a restricted number of Ia specificities involved in the incompatibility. Sera reacting with products of the I-A region were effective at prolonging survival, probably to the same extent as a serum covering all the specificities of the incompatibility. Such sera also inhibited MLR in the same strain combinations. Interpretation of these and other data suggests that products of the I-B, I-E and I-C regions play a subordinate role as target antigens for enhancement. It is concluded that either Ia antigens of different regions have different biological activities or enhancement is a function only of antibodies directed against antigens that stimulate in MLR.

    Notes: Thirty-one combinations of congenic and recombinant mouse strains were tested for heart and skin allograft survival times with and without ALS and the results compared with those obtained for MLR and CML. Although the genes for Lads, Ir and Ia specificities are all in the K end, on the average no greater strength of K end incompatibilities than D end were found. Some of the shortest survivals were D region only incompatible.We specifically wanted to find out if there were any special genes concerned with the control of antigenic strength within the H-2 complex and if so, did they correlate with those already described for CML—the effector cell stimulating or ECS genes. We found no evidence that the ECS genes in themselves were concerned with allograft strength but instead found evidence for certain genes in the I region and others outside the I region which could be acting as allograft potentiating genes. In some combinations there was an association with the ECS genes and the allograft potentiating genes, and we concluded that in these combinations the two types were closely linked but separate.We wanted to know if there was a specific lymphocyte subpopulation concerned with antigenic strength that was sensitive to ALS; the results of our experiments were consistent with this hypothesis but did not prove it.

    Notes: The tetraparental model was used to investigate the mechanism of induction and maintenance of tolerance. Of eight C57BL?BALB/c, potential tetraparental mice, six were found to be chimaeric by coat colour and by one or more of the following parameters: g.p.i., γ2a allotype testing, lymphocytotoxicity testing and germ line. None of the mice were chimaeric by all of these tests but showed instead a variable spectrum of chimaerism. Tolerance was investigated by both in vivo and in vitro testing. When the mice were chimaeric by at least two criteria, they were unresponsive to skin allografting (five out of six mice) and their spleen cells were non-reactive against BALB/c × C57BL F1 s in the popliteal node GVH, test. In contrast, the MLRs were generally positive and could not be blocked by chimaera serum or suppressed by chimaera thymus suspensions. Thus while we cannot formally exclude a suppressive mechanism, our results are best explained by the classical theory of clonal elimination provided one accepts that clonal reactivity and elimination is heterogeneous. The very establishment of a successful chimaera may be dependent upon the elimination of clones causing lethal GVHR while those responsible for an MLR or other non-lethal parameters of chimaerism is retained.

    Notes: Monolayers of mouse peritoneal macrophages actively incorporated 14C-labelled uridine, an RNA precursor. This uptake did not seem to be due to in vitro activation or stimulation of the cells but rather to their high RNA metabolic activity. 14C-uridine uptake by the unstimulated adherent cells was a characteristic property of macrophages, because it was not reduced when the cells were (i) pretreated with trypsin (to detach remaining lymphocytes), (ii) precultured for several days (to allow PMN to die off) and (iii) pretreated with anti-mouse immunoglobulin or anti-θ serum plus complement.Pretreatment of macrophage monolayers with antibody plus complement, followed by a measurement of their 14C-uridine uptake could be used to test for anti-macrophage activity, for instance in certain anti-B cell or anti-T cell sera, or to detect cell surface antigens on macrophages. With this technique we demonstrated the presence of H-2 and Ia alloantigens on peritoneal macrophages. The presence of Ia antigens on macrophages may be of particular biological significance.

    Notes: With an improved method for extraction of HL-A soluble antigen from dried blood stains, eighteen HL-A antigens were tested in the microcytotoxicity test as described in the preceding paper (Rittner & Waiyawuth, 1974). With the antisera available, seven antigens could not be detected with sufficient confidence. As a serious handicap of the microcytotoxicity test, a selected panel of donor lymphocytes must be permanently available. A comparison between the microcytotoxicity and the microcomplement fixation test performed on platelet material recovered from the stains led to the conclusion that the microcomplement fixation test is better suited for a routine laboratory. Encouraging results were obtained in a larger series of microcomplement fixation tests with white blood cells from dried blood stains. Significant chi-squared values were calculated for fourteen of sixteen antigens tested. The possible causes of false positive and negative reactions are discussed. Anticomplementary reaction of anti-HL-A sera in microcomplement fixation tests should be taken into account.

    Notes: It is shown that S-s-U-erythrocytes have no detectable PAS-3, B- and C-glyco-protein bands which are demonstrable by SDS-polyacrylamide gel electrophoresis of red cell membranes and subsequent PAS-staining. PAS-1 and -2 are normal with respect to electrophoretic mobility and staining intensity. The staining intensity of PAS-3, B and C from heterozygous defective erythrocytes is about half of normal. One S-s-U+ blood sample did not differ from the S-s-U-bloods.

    Notes: A survey for constant region γ-chain allotypes (de locus) was undertaken in different lagomorph genera. As yet only Pronolagus rupestris, a paleolaginae, showed the presence of a determinant similar to rabbit d12 although it lacked the widespread e15 determinant.All seven individuals possessed the d12 like determinant which was studied by immunodiffusion, haemagglutination inhibition, radiobinding and binding inhibition assays. In addition, a new enzymic method for typing for d12, based on the presence of the asymmetric rabbit hing carbohydrate linked to the d12 characteristic threonine, is presented. This method suggests, however, that, unlike the d12 rabbit, Pronolagus does not seem to have a majority of IgG molecules with a glycosylated hinge.

    Notes: Chicken antibodies showing rigorous specificity for mouse immunoglobulins (Ig) allow the direct visualization of an Ig-like surface component on thymusderived lymphocytes (T-cells). These antibodies, therefore, provide a direct means of testing the hypothesis that the Ig-like molecules function in antigen recognition on these cells. In an attempt to determine if avian antisera to mouse Fab determinants recognize antigen-binding receptors, the surface distribution of three large fluorescent (red) protein antigens, keyhole limpet haemocyanin (KLH), horse spleen ferritin (HSF), and whale skeletal myoglobin (MYO) on BALB/c thymus antigen-binding lymphocytes (ABL), was compared with that of fluorescent (green) labelled chicken antibodies to mouse Fab fragments [CAM(Fab')2]. The distribution of antigen and antisera against the Thy-1 antigen and H-2d determinants was also compared.The frequency of ABL ranged from 0.2 to 1% and binding to these cells could be inhibited by the appropriate unlabelled antigen and CAM(Fab')2. Thymus ABL were 100% positive for Thy-1 antigen when doubly stained using fluorescent rabbit anti-mouse immunoglobulin (RAM Ig) in an indirect fluorescence assay, but only 0-10% of ABL were positive for binding of RAM Ig alone, indicating that nearly all ABL are T-cells.Coincidence studies of patches of antigen and the various ligands were carried out by comparing coded fluorescence photomicrographs of randomly selected thymus ABL. Of thirty-six ABL stained with anti-thy-1 antiserum, all but four cells had at least one patch of antigen which was not shared with antiserum. Of thirty-six ABL stained with anti-H-2d serum, none had completely shared patches of antigen and antiserum. Of the forty ABL stained with CAM(Fab')2, every cell had every patch of antigen also patched with the anti-Ig reagent. We intepret our results to indicate that the primary receptor for antigen on antigen-binding T-cells is either the Ig-like molecule on the T-cell surface or exists in close proximity to this molecule.

    Notes: A Japanese blood donor (H. T.) and his brother (M. S.) are the first homozygous MkMk individuals described; their red cells lack, as expected, known antigens of the MNSs blood group system and also have no demonstrable MN-active and Ss-active glycoproteins. Both MkMk individuals have a naturally occurring atypical antibody in their serum. The antibody in the serum of H. T. is inhibited by MNSs-active glycoprotein preparations from normal erythrocytes.

    Notes: The properties of five human alloantisera which inhibit the stimulating capacity of cells in an MLC reactions are described. All of the sera showed patterns of inhibition in families which did not correlate with HLA haplotypes. In a large proportion of the families investigated the sera did not react with parental cells but inhibited the cells from one or more of the children. Absorptions were performed with one of these sera using such non-reactive parental cells. Paternal cells absorbed out the inhibiting activity while maternal cells did not. This indicates that experession of the antigen with which the inhibiting antiserum reacts is insufficient for inhibition and other factors, either environmental or inherited from the mother, influence the capacity of the antiserum to cause inhibition. This antigen was most likely a non-HLA antigen.

    Notes: We synthesized on Tn erythrocytes with human sera, UDP-Gal, and activators T-specific haptenic structures in satisfactory yield. The specificity of this biosynthesis was ascertained by agglutination with human and animal anti-T, by specific absorption of human anti-T as well as by agglutination inhibition assays.With isolated human erythrocyte T antigen as substrate we synthesized N- and M-specific structures with sera from individual human donors in presence of CMP-sialic acid by incubation for 24 hr at 37°C. Serology on the recovered product was carried out with nineteen monospecific human and animal sera under strictly standardized and controlled conditions with the mandatory tube assay. All M- as well as N-derived T antigens tested acquired N specificity with all transferase sera of all MN types. In contrast, M-activation of M- and N-derived T antigens occurred only if the transferase donor had the M gene. The nine M transferase sera used all gave M-activation of MM- and NN-derived T antigens. None of twelve transferase sera from NN donors M-activated any T antigen. NN antigen was transformed to a M-specific one by all transferase sera from MM donors but by none from NN donors.We have not yet established the biochemical-genetic relation of M to N; N may be the immediate precursor of M or M may originate directly from T. The sialytransferase responsible for M activation may be a N transferase ‘modified’ by the M gene product or an entirely different sialyltransferase.

    Notes: The ability of mouse amniotic fluid (MAF), alpha-foetoprotein (AFP) and MAF depleted of AFP (MAF - AFP) to suppress primary one-way MLR's was investigated. It was found that MAF, AFP and MAF - AFP were all suppressive of MLR's specific for MHC, K, D or I + S determinants. Suppression was observed when either lymph node or spleen cells were used as the responder cells. Nylon wool column passage of these cells did not significantly affect the immunosuppressive action of these substances. In contrast, MLR's specific for non-MHC/M-locus determinants demonstrated either diminished suppression or augmentation of the response, compared with the MHC stimulated MLR's. Our results show a differential effect of whole MAF and its fractions on the proliferative responses induced by various allogeneic stimuli and suggest that suppression is not due to a non-specific effect on proliferation regardless of the stimulus or cell type involved.

Response to GT, however, differed, with the B2B2 genotype being the only responder. No real genotype differences in immune response to DNP-conjugates and sheep red blood cells (SRBC) could be detected.

    Notes: Antisera to hepatitis B surface (HBs) antigen, both of human and rabbit origin, have been examined. The anti-HBs serum derived from a multiply transfused patient did, in addition, contain antibodies directed against the inherited β-lipoprotein antigen Ag(x), whereas one of the rabbit immune sera also contained antibody to the inherited Lp(a) antigen. Thus, in human, as well as in animal anti-HBs sera, antibodies to inherited normal serum antigens may cause false positive reactions. This problem may be overcome by absorption procedures if appropriate control systems are avialable. False positive reactions caused by the Ag(x) antigen may also be avoided by use of agarose as supporting medium for the test, as Ag(x) precipitin lines do not appear in this medium. An undialysable high molecular weight component may be obtained from Oxoid ‘Ionagar’ by washing. When this added to agarose the Ag(x) antigen reaction appears also in this medium.

    Notes: The H-2 restriction phenomenon was evaluated with regard to the immune response to Moloney sarcoma virus (M-MSV)-induced tumours in the mouse. Using an in vitro51Cr release assay the lytic activity of lymphocytes from M-MSV immune strains of mice, including several H-2 recombinant strains, was determined on leukaemic cell lines originally induced in mice of different strains by neconatal infection with Moloney leukaemia virus. In analogy with other experimental models, it was observed that a compatibility between effector and target cells at the K and/or D regions is generally necessary and sufficient to obtain the cytotoxic effect. However, for the H-2d and H-2b haplotypes, identity at the K or D region respectively was not sufficient for lysis to occur. Attempts to demonstrate that this lack of activity might be associated with the absence of H-2-linked Ir responder genes, were not successful.

    Notes: The role of H-2 and non-H-2 gene products was studied in a model of non-immune cell-cell interactions which underlie the homing affinity to lymph nodes of injected 51Cr-labelled lymph node cells. The effect of various donor-recipient combinations was tested by comparing the allogeneic/syngeneic ratio of radioactivity recovered from the lymph nodes. The homing affinity was reduced to about 50% when the donor-recipient incompatibility extended over the whole H-2 chromosome. When confined to a single region (H-2K, I or D) H-2 incompatibility caused no significant allogeneic inhibition of the homing affinity. The cumulative effect of two partial incompatibilities (K + I or D + I) was, however, reflected in a significant allogeneic inhibition. Non-H-2 incompatibilities had, as a rule, a weak effect; the non-H-2 gene products may not be directly involved in the cell interactions under test.

    Notes: The concentration of cAMP was measured in palatal shelves and tongues of 14.5-day old foetuses, 14.5-day old foetuses from steroid treated mothers, and 15.5-day old foetuses from four inbred lines of mice which represent the four possible combinations of two H-2 alleles and two residual genetic backgrounds. The incidence of spontaneous and steroid-induced cleft palate in these four strains was also determined. Analyses of variance of the cAMP data reveal that both the H-2 region and residual genetic background determine cAMP concentrations in both tissues and on both days of development. Similar analyses of cAMP concentrations after steroid treatments of the mother indicate that the interaction between H-2 and residual genetic background is significantly different in the injected than in the uninjected mice in both palatal shelves and tongues. The incidence of steroid-induced cleft palate parallels the palatal shelf concentration of cAMP before steroid treatment of the mother with one exception. These data suggest that a portion of the H-2 controlled component of susceptibility to steroid-induced cleft palate is mediated through alterations in the metabolism of cAMP.

    Notes: Descendants of Dutch colonists, who emigrated to Surinam in the last century and survived epidemics of typhoid and yellow fever with a total mortality of about 60%, were tested for twenty-six polymorphisms. The gene frequencies were compared with those of a large Dutch control sample. An analysis of drift indicated that the variations in gene frequencies observed for C3, Gm, HLA-B, and GLO were unlikely to be due to drift. Therefore these data might indicate selection through genetic control of survival in these epidemics.

    Notes: The effects of thymocyte interaction modulation factor on the adhesion of mouse allogeneic thymocytes and B-cells are reported. This glycoprotein, produced by short term cultures of thymocytes, has already been described as reducing the adhesion of syngeneic B-cells, leucocytes and macrophages. Adhesion was measured in suspension culture using the collision efficiency method. This paper reports that:1. In addition to the syngeneic effect of thymocyte IMF in reducing adhesion of certain unlike cell types there is also an allogeneic effect in which an allogeneic T-IMF will diminish the adhesion of a thymocyte population, or still further reduce the adhesion of a B-cell population than would a syngeneic T-IMF.2. Thymocyte IMFs were prepared from different congenic strains and tested on the adhesion of syngeneic and allogeneic thymocytes. When factor and cells were syngeneic or metched at any H-2 locus except H-2 D there was no effect on adhesion since it remained at the same value as in controls in their own IMF. But whenever factor and cells were mismatched at H-2 D there was a marked diminution in the adhesion of the cells.3. Antibodies raised against specific thymocyte IMFs could be used to detect the presence of T-IMF binding to the surface of cells by immunofluorescence or immune cytolysis. These systems show that the antibodies against thymocyte IMF can be used to type the H-2 D type of a cell and that these factors are present at the surface of thymocytes and certain other cell types. They confirm that the thymocyte IMF is either in H-2 D product or is closely associated with H-2 D locus in its binding and action. They also show that the T-IMF antigen on non-lymphocytic types is produced by T-cells or thymocytes.4. The general relevance of these results is discussed in relation to cell recognition phenomena.

    Notes: Mice of two congenic inbred strains C3H/Sn (H-2k) and C3H. B10 (H-2b were infected with 100 Schistosoma mansoni cercariae. After the infection, the following parameters for the immunologica response were studied: worm burden, mortality, antibody titre, spleen index, eosinophilia, delayed type of hypersensitivity and in vitro response to three S. Mansoni antigen preparations.No difference in the worm burden and in the in vitro response to the antigen preparations of adult worm antigen, soluble egg antigens and the egg antilgen MSA1, was found. The C3H.B10 mice showed a singificantly higher mortality, antibody titre and delayed type of hyprsensitivity while the C3H/Sn mice showed a significantly higher spleen index and eosinophilia. This indicates that the H-2 region influences the course of an acute S. mansoni infection, whereas the susceptibility to the infection seems not to be influenced, as is shown by the worm burden

Gross, histopathological and chemical examinations demonstrated that prevalence and severity of nephrocalcinosis increased with increasing CO2 concentrations.     Notes: Abstract. The histopathology of red-sore disease, caused by the gram-negative bacterium, Aeromonas hydrophila, is described for largemouth bass, Micropterus salmoides. Externally, lesions range from those affecting a few scales (pin-point), to those associated with extensive chronic ulcerations; there is focal haemorrhage, oedema and dermal necrosis which exposes underlying muscles producing infiltration of mononuclear and granulocytic inflammatory cells. Internally, the liver and kidneys are foci for toxic products produced by A. hydrophila with, in the most severe cases, complete destruction of the structural integrity of both organs. Pathological changes were not serious in either the spleen or heart, even in cases with massive damage in the liver and kidney. Internal and external lesions were similar in both natural and experimentally induced infections. The pathobiology of red-sore disease in bass is postulated to be linked to elevated water temperature stimulating increased metabolism, decreased body condition and stress, leading to the increased production of corticosteroids and the concomitant rise in susceptibility to infection.

    Notes: Abstract. The occurrence of cysts of Henneguya psomspermica on a population of perch was studied over a periot of 19 months. Cysts were restricted to the gill filaments although no attempt was made to assess the occurrence of earlier developnental stages of the parasite. Cysts were not recorded from a small sample of perch fry (aged 4–6 weeks). There was a peak of prevalence, and intensity of the infection in juvenile perch (aged 8–9 months), while the prevalence and intensity in adult perch (aged 1–1 and over) was low. In adult perch there was a seasonal November-February.

    Notes: Abstract. Papillomatosis of the European eel Anguilla anguilla L. occurs predominantly in waters of low to medium salinity (3–20%). Tumour-bearing eels were kept under laboratory conditions for several weeks in salt concentrations of 8, 15 and 30%. As opposed to those kept in freshwater, the experimental animals showed significant decreases in tumour growth rate. At the same time the tumour tissue underwent a marked redifferentiation, i.e. the originally poorly differentiated neoplastic tissue developed numerous mucous and club cells. The characteristic intercellular spaces of tumorous tissue disappeared with the formation of regular membrane interdigitations between adjacent cells. The nucleus to cytoplasm ratios of the neoplastic cells decreased to one-third of those of the untreated tumour. These changes resulted in a reversion of the neoplastic tissue to a structural condition essentially similar to that of normal epidermis. After a period of 2 to 3 months an adaptation to the altered salinity of the external medium was evident in that the tumorous tissue began to return to its original state.

Sheepshead minnows, Cyprinodon variegatus Lacépède, exposed to 5.5 to 31 μg/1 of the herbicide trifluralin, throughout their first 28 days of life, developed a heretofore undescribed vertebral dysplasia. This dysplasia consisted of semi-symmetrical hypertrophy of vertebrae (three to 20 times normal), characterized by foci of osteoblast and fibroblasts actively laying down bone and bone precursors. Effects of the abnormal vertebral development were dorsal vertebral growth into the neural canal, ventral compression of renal ducts, and longitudinal fusion of vertebrae. Pish, exposed for 51 days to 16.6 μg/1 trifluralin and thereafter depurated for 41 days, showed no increase in vertebral dysplasia during depuration; however, residual spinal column damage was evident. Serum calcium concentrations were elevated in adult fish exposed for 4 days to 16.6 μg/1 trifluralin. Fluorosis or mimicry of hypervitaminosis A are considered possible mechanisms for the osseous effect, but are not considered to be the only possible causes. The highly predictable nature of this disorder in experimental exposures strengthens the probability that young fish may serve as experimental models for determining effects of chemicals on early vertebrate ontogeny, particularly in regard to skeletal development.

    Notes: Abstract. Rainbow trout were fed diets contaminated with either PCB (Aroclor 1254, 500 μg/g dry food) or Mirex (500 μg/g dry food). There was no apparent effect of PCB on thyroid histology or on serum thyroid hormone levels. Mire effected significant reductions in serum T4 (P〈0.05) and T3 levels (P〈0.01) without apparent change om the histological appearance of the thyroi. Liver weights and total liver lipid content were significantly (P〈0.001) greater in PCB fed fish compared with controls as was the carcass lipid content (P〈0.01). Mirex had no apparent effect on liver size or liver lipid content but there was a decline in carcass lipid content (P〈0.02) which persisted 25 days after returning Mirex-fed fish to the control diet (P〈0.05).

Examination of the digestive tract of rainbow trout fry infected with Hexamita salmonis revealed no pathological changes in the epithelial cells of the pyloric caeca or upper intestine even where the parasites were closely apposed to the brush border of these cells. No invasion of the epithelium by the parasites was observed, although examples of shrinkage necrosis (apoptotic bodies) of epithelial cells were often found. The ultrastructure of the parasite resembled that of the closely related Giardia lamblia. Despite the lack of pathological changes to the intestinal epithelium, treatment of the fish to remove the parasites greatly reduced mortalities.

During an extended survey aimed at investigating the occurrence of neoplasms in the common demersal fish species inhabiting zones of Port Phillip Bay, Victoria, Australia, exposed to domestic and industrial contamination, superficial lesions resembling malignant growths were found in high prevalence (20% and 28% respectively) in two species of leatherjacket (Family Balistidae), Navodon multi-radiatus and Scobinichthys granulatus, and infrequently (0.3%) in Atopomycterus nichthemerus (Diodontidae). The leatherjacket lesions took three forms, all being histologically consistent with long-standing granulomatous tissue associated with varying degrees of mononuclear cell infiltration. The A. nichthemerus lesions were histologically similar but located beneath an intact epidermis. In some of the lesions, profiles of parasitic worms (either solitary or several) were found after examination of multiple sections suggesting that the ‘tumours’ may have represented chronic inflammatory tissue reactions to superficial parasite infection (xenomas).The resemblance to fish neoplasia of both the macroscopic appearance of these lesions, particularly those affecting leatherjacket species, and of certain features of their microscopic structure is discussed. The necessity for critical evaluation of the range of proliferative tissue reactions in fish is emphasized, in order to permit accurate distinction between non-neoplastic conditions and true neoplasia in epidemiological surveys using fish species as indicators of environmental contamination with specific biological hazards.

    Notes: Abstract. The occurrence of Neoechinorhynchus rutili (Müller, 1780) in the white-fish Coregonus nasus (Pallas) sensu Svardson from the Bay of Bothnia was investigated in 772 fish obtained as monthly or bimonthly samples during the period July 1975-January 1977.In both years most parasites were found only in November and December. All of the 402 parasites found were immature and very small. Most of them were in the first third of the intestine, and only a few were attached to the mucosa. No encysted parasites were found in other any tissues. It is thus suggested that the whitefish of the Bay of Bothnia is only an accidental host for N. rutili.A clear correlation was found between the importance of ostracods in the diet and the occurrence of N. rutili. The first intermediate host in the Bay of Bothnia was shown to be an ostracod, Candona neglecta (Sars.).

    Notes: Abstract. Two cell lines that are persistently infected with IPN virus have been established. Viral persistence in the cultures was demonstrated by the detection of infectious virus (102-105,5 TCID/ml50) in the culture fluids, by specific immunofluorescence, and by their resistance to superinfection by homologous virus. The growth, rate of these cells was indistinguishable from normal, uninfected cells. Virus stocks harvested from persistently infected cultures contained a greater proportion of defective, interfering particles than those from lytically infected cells which suggests that these particles may be responsible for the maintenance of these carrier cultures.

An ulcerative dermal necrosis-like salmon disease appeared in Laukaa Central Fish Culture Station in Middle-Finland in autumn 1976 showing lesions in different sites on diseased landlocked salmon Salmo salar L., sea trout Salmo trutta trutta L. and brown trout Salmo trutta lacustris L. Lesions from the diseased landlocked salmon were studied by transmission and scanning electron microscopy.Both aquatic bacteria and fungi grew in abundance in the lesions, changing the fine structure of the epidermis.In all of the lesions studied virus-like particles 30–33 nm in diameter were found. The relationship of these particles to the disease is still unresolved.

    Notes: Abstract. A virulent, 2,3-butanediol positive culture of Aeromonas hydrophila, subspecies hydrophila, was fed or injected intraperitoneally into channel catfish. The positive infection and tissue involvement were monitored by the 2,3-butanediol test with tissue homogenate enriched with a buffered nutrient broth containing dextrose. The results revealed that the fish infected orally showed very little or no spleen involvement. Fish infected intraperitoneally, however, showed spleen involvement in a very short time, accompanied by the rapid onset of an acute systemic infection.Light and electron microscope studies of the spleen revealed that the pathogenic bacteria were mainly located within the reticular sheath of the ellipsoids and only sparsely in the surrounding regions, and that intense phagocytic activity of macro-phages had taken place in the ellipsoids. It was also observed that the phagocytized bacteria divide extracellularly, and probably intracellularly, resulting in the destruction of the endothelial and reticular cells of the ellipsoids. This finding substantiates the cytolytic and fibrolytic capacities of A. hydrophila for destroying host defence systems and may contribute to its invasive powers. Other cytological changes in cells in spleen after infection are described, and the resistance of A. hydrophila to intracellular enzymatic digestion by the macrophages is discussed.

    Notes: Abstract. Chronic mortalities associated with meningeal and pancreatic lesions are described, from two trout farms in Scotland. Histopathological and microbiological studies suggested that bacterial infection with strains of Pseudomonas fluorescens of low pathogenicity was developing in fish already affected by the lesions of chronic infectious pancreatic necrosis virus infection (IPN). Antibiotic therapy reduced losses temporarily; eradication of the virus resulted in complete elimination of the condition.

    Notes: Abstract. Two entirely separate outbreaks of PKD are described in an attempt to properly define the condition and enable it to be differentiated from other diseases of a similar nature. Losses of up to 75% of fingerling rainbow trout were recorded in one outbreak, illustrating the potentially serious nature of the disease. Although the kidneys showed the most obvious pathological changes, other organs were also affected. The host response was granulomatous in type, the foci of each cellular reaction being 5-20 μm eosinophilic cells considered to be parasitic. The cytoplasm of these cells contained numerous electron dense bodies some with a tail, but each with an electron lucent bar always orientated at right angles to the plasmalemma whenever they were near it. The most obvious cytoplasmic inclusion of these ‘parasites’ were other cells numbering up to five or six, each with its own nucleus and nucleolus, but devoid of any of the small electron dense bodies. The significance of either of these inclusions is unknown.A limited differential diagnosis is given, and the argument presented that the condition PKD is the result of infection by a parasite, possibly an amoeba, in combination with poor quality water.

    Notes: Abstract. The spleen, kidney and liver melano-macrophage centres of 14 species of clinically normal teleost fish were examined histochemically for ferric iron. Iron was present in varying amounts within the splenic centres of most specimens, but by contrast it was rarely found in the centres of the kidney and the liver.Under conditions of starvation and in diseased fish, a markedly increased deposition of ferric iron occurred in the splenic centres of nearly all fish examined. By comparison, the iron content in the kidney and liver centres was generally still very low. These results suggest that there exist important functional differences between the pigment centres of different haemopoietic tissues of teleosts.

Fifty-four per cent of the specimens in a sample of 147 lake whitefish (Coregonus clupeaformis) from northern Quebec exhibited a pronounced lack of bilateral symmetry in the distribution of metacercariae of the genus Diplostomum. This asymmetry was shown not to be a product of chance. A comparable degree of asymmetry was generated by a computer simulation incorporating a positive feedback mechanism. The simulation and the biological significance of such asymmetry are discussed.

The specificity and kinetics of the immune response of rainbow trout (Salmo gairdneri) to single injections of an O-antigen extracted from the bacterial pathogen Yersinia ruckeri, which causes enteric redmouth in fish, were investigated by the passive haemolytic plaque assay and serum antibody quantitation. Doses ranging from 5 ng to 500 mg in 10-fold increments were injected intraperitoneally into groups of trout held at 17 × 1°5°C. The occurrence of plaque forming cells (PFC) and humoral antibody was followed for 35 days after injection. Trout gave an immune response to doses of 500 ng and above. Seven days after injection no humoral antibody was detected, but PFC were found in the spleen. The maximum PFC numbers occurred 11 days after injection. On day 21, few PFC were found, whereas serum antibody titres were highest. The antibody from immunized trout showed little or no cross-reactions with sheep red blood cells passively labelled With antigens from other fish pathogens.

    Notes: Abstract. Fungal infection of both wild and hatchery-reared sexually mature brown trout, Salmo trutta L., was caused by Saprolegnia diclina Humphrey Type 1. The major ions in the serum of infected fish were all significantly reduced in concentration and it is suggested that the primary cause of death was osmoregulatory breakdown resulting in a lethal haemodilution. Severe hypoproteinaemia and a significant reduction in the albumin/globulin ratio were reflected in the electro-phoretogram of the serum proteins from infected fish.

    Notes: Abstract. Lymphocystis disease is reported from seven fish species in 48 of Wisconsin's 72 counties (67%). It was previously known from only two fish species in five counties. The record from northern pike, Esox Indus L. is new. Five other host records are new for Wisconsin. A relationship between the disease distribution in new waters and walleye, Stizostedion vitreum (Mitchill), stocking sources is noted. Histologlcal observations from black crappie, Pomoxis nigromaculatus (Lesueur) and walleye are similar to previously published accounts. Events in hyaline capsule degeneration are described for the first time. In Wisconsin, the disease was most commonly observed in the spring, particularly in walleye, but was consistently evident throughout the rest of the year.

    Notes: Abstract. An unidentified mycoplasma was isolated from cultures of Atlantic salmon (AS) cells and implicated as the cause of cytopathic effects (CPE). The agent did not visibly affect RTG-2 cells. Experimental infection of RTG-2 cells with Acholeplasma laidlawii resulted in increased cellular granularity and destruction. Scanning electron microscopic (SEM) examination of infected RTG-2 and AS cultures revealed typical mycoplasmas distributed on cell surfaces and a marked effect on the cell surface topography, characterized by a loss of microvilli and cellular processes. SEM also revealed mycoplasmal contamination that was not detected by culturing. Comparisons of contaminated and uncontaminated RTG-2 cells showed enhanced (2–100 fold) replication of infectious pancreatic necrosis virus (IPNV) and infectious haematopoietic necrosis virus (IHNV) in the presence of A, laidlawii.

    Notes: Abstract. Blood samples from 961 estuarine and marine fish comprising 44 species from the northern Gulf of Mexico were examined for haematozoa. Trypanosoma mugilicola n. sp., characterized by an anteriorly situated nucleus (NI = 3.8), is described from the striped mullet (Mugil cephalus); it was also found in the hog-choker (Trinectes maculatus). Unidentified trypanosomes occurred in the hogchoker and bay whiff (Citharichthys spilopterus). Trypanoplasma bullocki is recorded for the first time from both the Gulf of Mexico, and from the southern flounder (Para-lichthys lethostigma) and Atlantic croaker (Micropogonias undulatus). Three species of haemogregarines are identified for the first time from the Gulf of Mexico, Haemo-gregarina platessae in the southern flounder, hogchoker, lined sole (Achirus lineatus) and blackcheek tonguefish (Symphurus plagiusa); H. mugili in the striped mullet; and H. delagei in the clearnose skate (Raja eglantera). A few piscivorous leeches from the region are evaluated as potential vectors.

    Notes: Abstract. The prevalence and intensity of Ichtyobodo necator infestations on farmed salmonids was investigated over a 7-month period. The infestations were found to be markedly age dependent. Ichtyobodo infestations peaked at 4 weeks after commencement of first feeding and mortalities peaked at 4 6% per week at 8 weeks in the 0+ fish. Both infestations and mortalities showed a marked decline to zero shortly after these periods with no chemotherapy. No Ichtyobodo were seen on the 1+ fish until late in the study after a drop in temperature to below 10°C, when they reappeared on the 0+fish also. Ichtyobodo were found at temperatures between 3–5°C and 16°C and in the 0+fish appeared irrespective of the condition of the fish.It is suggested that some form of host defence mechanism is operating which limits the Ichtyobodo infestations in farmed salmonids.

    Notes: Abstract. An outbreak of infectious haematopoietic necrosis causing high mortality in rainbow trout fry occurred in New York State. Despite the fact that the hatchery effluent was recycled, spread of viral infection or of clinical disease was not demonstrated.Endocardial and glomerular endothelial necrosis and fibrinous peritonitis were among the histopathologic changes seen. Many of the signs and lesions were similar to those seen in infectious pancreatic necrosis.

A previous paper has revealed that experimental streptococcal infection was associated with a rapid growth of the intestinal bacteria, suggesting association of the condition with an exotoxin. The present study was undertaken to see the effect of the exotoxin on the streptococcal infection by administering cell free culture before the bacterial challenge. Cell free culture of Streptococcus sp. strain YT-3 was inoculated intramuscularly at a dosage of 0–5 ml per 100 g body weight 1 h prior to bacterial challenge. Three fish were killed at intervals after challenge for viable counting of bacteria in the viscera and blood. Intramuscular inoculation with either low virulent or virulent bacteria alone at dosages of 106 to 107 cells did not produce the disease in the fish, with almost complete clearance of the bacteria from the viscera and blood within 120 h. When the exotoxin was inoculated intramuscularly prior to the bacterial challenge, however, either low virulent or virulent bacteria at 106 to 107 cells could produce fatal infection with prominent streptococcal clinical signs.

    Notes: Abstract. An in vitro technique was developed to screen drugs for toxicity to laboratory cultured protozoans. Drugs were diluted using standard microtitration equipment. An easily cultured protozoan, Tetrahymena pyriformis, was added to the dilutions of drugs and viability was checked daily for 10 days. Drugs used as a model system to test the technique were formalin and malachite green. Effective concentrations of formalin determined by this in vitro technique were close to recommended therapeutic treatment levels for Ichthyophthirius. Malachite green was effective at concentrations in excess of recommended therapeutic treatment levels for Ichthyophthirius. The synergistic effect of formalin and malachite green was demonstrated. This technique can he of use in determining effectiveness of new drugs as parasiticides.
