The procedure is carried out in its entirety within the culture flask, using only standard fixing and embedding reagents.     Notes: Abstract The purification of tryptophan decarboxylase from Catharanthus roseus (TDC, E.C.:4.1.1.27), to apparent homogeneity, is described. The enzyme represents a soluble protein with a molecular weight of 115 000±3 000, consisting of 2 identical subunits of 54 000±1 000. The pI was estimated to be 5.9 and the Km for L-tryptophan was found to be 7.5×10-5 M. Phenylalanine, tyrosine and DOPA were not decarboxylated by tryptophan decarboxylase from Catharanthus cells. Similar to the aromatic amino acid decarboxylase from hog kidney the enzyme does not appear to be obligatorily dependent on exogenously supplied pyridoxal phosphate, as it seems to contain a certain amount of this cofactor. The average percentage of TDC in the cells was found to be 0.002% in the growth medium while the level increased up to 0.03% when indole alkaloid biosynthesis was induced. The role of the protein as a bottleneck enzyme of indole alkaloid biosynthesis is discussed.

    Notes: Summary Ionic currents in cultured dorsal root ganglion (DRG) neurons fromadult guinea pigs were analyzed by voltage-clamp techniques. The Na+ inward current had a reversal potential at +33mV, and revealed activation and inactivation kinetics similar to those of squid giant axons. A typical value for the maximum Na+ conductance was 178 mS/cm2 and the peak current was 2.5mA/cm2. The delayed K+ outward current showed a fast and a slow phase of inactivation and was sensitive to tetraethylammonium (TEA; ∼130mm) and 4-aminopyridine (∼2mm). The maximum K+ conductance was 26±9 (mean±sd) mS/cm2. The slow Ca2+ inward current was identified in Na+-free, TEA-containing solution. Its peak value was increased by 1.7-fold when [Ca2+]o was increased from 5 to 10mm. The current was blocked by Co2+ but not by tetrodotoxin. Sr2+ and Ba2+ could substitute in carrying this current. The maximum peak of the Ca2+ current was 0.22±0.14mA/cm2. At potentials positive to 0mV, the Ca2+ current was often followed by a slowly developing outward current, which was also sensitive to Co2+, suggesting a Ca2+-activated outward current. It is concluded that the action potential of theadult guinea pig DRG neuron is mediated by Ca2+ as well as by Na+ and K+ currents. The current densities of these ionic channels are considered to be different fromembryonic neurons and from nodes of Ranvier.

    Notes: Summary Usin gintracellular microelectrode technique, the response of the voltageV across the plasma membrane of cultured bovine corneal endothelial cells to changes in sodium and bicarbonate concentrations was investigated. (1) The electrical response to changes in [HCO 3 − ] o (depolarization upon lowering and hyperpolarization upon raising [HCO 3 − ] o ) was dependent on sodium. Lithium could fairly well be substituted for sodium, whereas potassium or choline were much less effective. (2) Removal of external sodium caused a depolarization, while a readdition led to a hyperpolarization, which increased with time of preincubation in the sodium-depleted medium. (3) The response to changes in [Na+] o was dependent on bicarbonate. In a nominally bicarbonate-free medium, its amplitude was decreased or even reversed in sign. (4) Application of SITS or DIDS (10−3 m) had a similar effect on the response to sodium as bicarbonate-depleted medium. (5) At [Na+] o =151mm and [HCO 3 − ] o =46mm, the transients ofV depended, with 39.0±9.0 (sd) mV/decade, on bicarbonate and, with 15.3±5.8 (sd) mV/decade, on sodium. (6) After the preincubation of cells with lithium, replacement of Li by choline led to similar effects as the replacement of sodium by choline, though the response ofV was smaller with Li. This response could be reduced or reversed by the removal of bicarbonate or by the application of SITS. (7) Amiloride (10−3 m) caused a reversible hyperpolarization of the steady-state potential by 8.5±2.6 mV (sd). It did not affect the immediate response to changes in [Na+] o or [HCO 3 − ] o , but reduced the speed of regaining the steady-state potential after a change in [HCO 3 − ] o . (8) Ouabain (10−4 m) caused a fast depolarization of −6.8±1.1 (sd) mV, which was followed by a continuing slower depolarization. The effect was almost identical at 10−5 m. (9) It is suggested, that corneal endothelial cells possess a cotransport for sodium and bicarbonate, which transports net negative charage with these ions. It is inhibitable by stilbenes, but not directly affected by amiloride or ouabain. Lithium is a good substitute for sodium with respect to bicarbonate transport and is transported itself. In addition, the effect of amiloride provides indirect evidence for the existence of a Na+/H+-antiport. A model for the transepithelial transport of bicarbonate across the corneal endothelium is proposed.

    Notes: Summary Micropuncture of cultured bovine corneal endothelial cells led to registrations stable for hours. Intracellular potentials were mainly in the range of −40 to −55 mV, average 46.3±0.6 mV (sem). Changes of extracellular [HCO 3 − ] led to voltage transients, their amplitude depending logarithmically on [HCO 3 − ] with a mean slope of 37.3±8.8 (sd) mV. After removal of bicarbonate/CO2, a steady-state depolarization was seen. This steady-state depolarization, but not the voltage transients, could be reduced by 1mm Ba++. After removal of bicarbonate, the voltage response to changes of extracellular potassium was reduced. Alteration of pH i induced by permeable buffers (butyrate, glycodiazine and ammonium) also resulted in voltage transients, internal acidification being correlated with a hyperpolarization, and internal alkalinization with a depolarization. Also changes of external pH caused voltage responses, alkalinization causing a hyperpolarization, acidification a depolarization. Methazolamide, an inhibitor of carbonic anhydrase, as well as stilbenes (SITS or DIDS) caused a reduction of the voltage response to HCO 3 − and pH. Their effects were additive. It is suggested that corneal endothelial cells possess one or two electrogenic transporters for HCO 3 − or related species, one of which is inhibitable by stilbenes.

    Notes: Abstract After disturbance of protein synthesis by puromycin and disturbance of transcription of chromosomal and ribosomal RNA by actinomycin D considerable changes were observed in the normal course of mitosis. An increase in the number of colchicine-like mitoses (c mitoses) sometimes accompanied, in particular, by segmentation of their cytoplasm with the formation of racemose structures, were observed. It is suggested that the development of c mitoses is associated not only with disturbances in the system of formation of the mitotic apparatus, but also with blocking of the synthesis of one of the chromosomal proteins that stabilize the spiralization of DNA strands. The other disturbance of division arising as a result of depression of metabolism, namely hollow metaphase, is associated not only with disturbances of the formation of the division spindle, but also with chromosomal changes. Selective depression of transcription of ribosomal RNA led to definite delay of anaphase and to coupling of the telomeric regions of the chromosomes, evidently on account of disturbance of the “protective membrane” of the chromosomes formed by RNA of the disintegrating nucleoli and RNA of the perichromatin granules.

    Notes: Abstract Two acid phosphatases isolated from culturedIpomoea (moring glory) cells were separated by column chromatography on DEAE-cellulose. The two acid phosphatases have different pH optima (pH 4.8–5.0 and 6.0) and do not require the presence of divalent ions. The enzymes possess high activity toward pyrophosphate,p-nitrophenylphosphate, nucleoside di- and triphosphates, and much less activity toward nucleoside monophosphates and sugar esters. The two phosphatases differ from each other in Michaelis constants, in the degree of inhibition by arsenate, fluoride and phosphate and have quantitative differences of substrate specificity. In addition, they also differ in their response to various ions.

    Notes: Summary The Chinese hamster ovary (CHO) cell/hypoxanthine guanine phosphoribosyl transferase (HGPRT) mutation assay developed by Hsie et al., was simplified by culturing the cells as unattached cultures, and also modified to include mutation at the Na+-K+ ATPase (ouabain resistance) gene locus. The cost and time involved were decreased by culturing the CHO cells unattached on nontissue culture plates during the expression period. The inclusion of a second gene locus ensures that mutagenicities observed were not due to the peculiar properties of a specific gene locus. These procedures are now used in our laboratory for routine testing of environmental chemicals and complex mixtures.

Procedures adopted included use of leaf base explants, CS medium with 3 mg/l 2, 4-D and 0.25 mg/l kinetin for callus initiation and growth, MS medium with 0.5 mg/l IAA and 1 mg/l BAP for shoots, MS medium with 5 mg/l NAA and 7% (wt/vol) sucrose for rooting of shoots. Casein hydrolysate (N-Z amine) significantly shortened the lag period in the growth of sugarcane suspension cultures, but did not increase the rate of growth following the lag phase. Protoplasts isolated from two types of cultures could be grown to re-establish cell cultures but no plants have yet been regenerated derived from isolated protoplasts.

    Notes: Abstract An improved culture system for plant cells that employs filter paper resting on polyurethane saturated with liquid medium is described. It combines a simplified version of the system outlined by Weber and Lark [1979, Theor Appl Genet 55: 81–86] with the method of growth estimation described by Horsch et al. [1980, Can J Bot 58: 2402–2406]. The growth of plated cells or callus can be conveniently monitored through repeated non-destructive fresh weight measurements of the filter paper and adhering cells, thereby allowing the construction of a complete growth curve over the course of an experiment. Experiments with 3 Nicotiana genotypes (N. plumbaginifolia Viv., N. tabacum L. ‘SC 58’ and N. tabacum ‘WI 38’) and 3 Vitis vinifera L. genotypes (‘Chenin Blanc’, ‘Dogridge’ and ‘White Riesling’) clearly demonstrate higher growth rates of plated cells on polyurethane supports compared with agar. Further experiments with N. plumbaginifolia illustrate the use of polyurethane supports for culturing cells at low pH (4.0) and the recovery of spent medium for monitoring changes in pH. These features will greatly facilitate quantitative studies of mineral nutrition and metal toxicity in cultured cells. Polyurethane supports also allow the incorporation of conditioned medium or feeder cells to support the growth of cells at low densities and facilitate the rapid recovery of variant cells.

    Notes: Abstract Cell cultures of sweet potato grown in media containing sucrose, glucose, maltose, or starch secreted amylase into the growth medium. The growth rate of cells was not appreciably affected by the carbon source employed for growth, although cells grown on sucrose had a slightly longer lag period before exponential growth occurred. Amylase levels inside the cells were not affected by carbon source, but the amount of amylase released into the medium was drastically affected. Maltose-grown cells released the most amylase while sucrose-grown cells released the least. Cells grown in the light released about twice as much amylase as cells grown in the dark when grown on glucose, maltose, or starch. Three amylase electrophoretic forms were found in the storage root tissue from which all cultures were derived. Cells grown in culture exhibited either two or three amylase forms, depending on the carbon source. The slowest migrating root amylase was found only in cells grown on starch. The root amylase having intermediate mobility was present in all cultures, as was a form having higher mobility than the most mobile root form. The fastest migrating electrophoretic form from the root was not present in any of the cells.

    Notes: Abstract In cell suspension cultures of Catharanthus roseus a rapid accumulation of secondary compounds (tryptamine, indole alkaloids, phenolics) was observed after transfer of the cells into special ‘induction’-media devoid of phosphate and other essential growth factors [11, 14]. The increase of product levels was suppressed in the presence of phosphate which was almost completely taken up from the medium and accumulated by the cells within 48 h after inoculation. The activities of tryptophan decarboxylase (TDC), the first enzyme in indole alkaloid biosynthesis, and of phenyl-alanine ammonia-lyase (PAL), the key enzyme of phenylpropanoid biosynthesis, were influenced differently by phosphate. Whereas the accumulation of phenolics and PAL activity were similarly inhibited by low concentration of phosphate, the medium-induced enhanced activity of TDC was not affected although the product pools were considerably reduced. Some consequences for the regulation of secondary metabolism will be discussed.

    Notes: Abstract Dissociated neonatal rat cerebellar cells were grown on medium supplemented with 10% horse serum (HS) and compared with those grown using a serum-free supplemented (SFS) medium, modified from Bottenstein and Sato (1979). containing insulin, transferrin, progesterone, putrescine, and selenium (after an initial 24 hr in 10% horse serum). Cells survived for several weeks using either medium. Cells grown in SFS had higher levels of GABA uptake than cells grown in HS. Cellular morphology and the proportion of neurons to glial cells were similar under the two conditions. Transferrin concentrations of 0.5, 10, and 100 µg/ml were tested. Neither neuronal nor glial cells were sensitive to this 200-fold variation. The SFS medium supports survival and maturation of both neurons and glial cells from rat cerebellum. However, the medium is not completely defined since (1) one day of serum is still required and (2) the heterogeneous cell population is undoubtedly conditioning the medium to some extent.

Examples of cells were found which were positive for one or the other neurotransmitter or for neither transmitter, but not for both. Furthermore, cells which appear similar by morphological criteria alone differed in transmitter specificity. 4. We conclude that the two transmitters tested are not localized within single cells and that morphology alone is inadequate to identify functional cell classes in this area.

    Notes: Summary 1. Asymmetric forms of AChE have generally not been detected in cultured chick skeletal muscle cells in the absence of cocultured neurons. To explore further neurotrophic effects of adult peripheral nerve extracts (NE) on musclein vitro, we reexamined the appearance of various molecular forms of AChE in cultured chick muscle cells in the presence of NE. 2. The various molecular forms of AChE were distinguished by sucrose gradient sedimentation and radioenzymatic techniques. 3. In the presence of NE, cells proliferated during the first 48 hr of culture, then fused and formed spontaneously contracting myotubes by 6–8 days in culture. 4. Total AChE, 5.4 S, and 11.5 S molecular forms reached activity plateaus by 8 days in culture which persisted until cultures were terminated at day 20. 5. Betweeen 1 and 6 days in culture, 19.5 S AChE (A12) was not detected. The A12 form was first observed at 7 days reaching a maximum of 11.3% of the total AChE at 14 days and then gradually declined to a level of 1% at day 20. 6. Since the A12 form declined in older cultures but comprised 25% in embryonic muscle tissue, we examined the possible requirement of neurons in culture to attain higher levels of A12 AChE. Spinal cord neurons were plated onto 6-day muscle cultures and AChE activities were measured between 8 and 20 days. 7. The results showed that 19.5 S AChE activity in the presence of both spinal cord neurons and NE was no greater than that found in the presence of NE alone. 8. To suppress spontaneous contraction, 0.6µM tetrodotoxin (TTX) or 15µM d-tubocurarine (dTC) were added to 5-day-old muscle cultures at a time when myotubes were differentiated but contractile activity had not begun. 9. TTX had cytotoxic effects and inhibited further development of myotubes. In contrast, dTC had no deleterious effect on morphological development, eliminated contraction, but did not interfere with the appearance of any forms of AChE including the A12 form. 10. These studies show that primary chick muscle cells are capable of producing the A12 form of AChE if cultured in NE-supplemented medium. In this culture system, production of the A12 form does not require activity or innervation.

In the two neuroblastoma lines a very high rate of synthesis of GM2 could be observed, whereas the glioma line C6 showed a high prevalence of synthesizing GM3. None of them produced GD1b or GT1b. Primary cultures appeared to be very similar to nervous tissuein vivo, containing essentially all the major brain gangliosides in comparable amounts. 3. Treatment of primary cultures with the neurotoxins kainic acid and hydroxydopamine showed a decrease in neurons as measured by125I-labeled tetanus toxin binding concomitant with a selective loss of GT1b, GD1b, GD1a, and GM1 but not GM3. 4. It is concluded that the ganglioside GM3 is confined mainly to nonneuronal cells, whereas the other four gangliosides mentioned are normal constituents of the neuronal membrane.

    Notes: The enhancement of L-tyrosine aminotransferase activity by dexamethasone, an exclusive function of the liver, was serially measured at different passages of eight rat liver epithelial cell lines initiated and continuously grown in either a serum-supplemented medium or a serum-free medium. The enzyme basal activity was found to be 5.4 ± 1.8 mU for cell lines in serum and 6.8 ± 3.4 mU for cell lines without serum. Under the influence of dexamethasone (10−6 mol/l for 5 hours) this basal level could be increased up to 2.9 fold in the presence of serum and 2.5 fold in its absence when investigations were carried out at early passages. During the following subcultures the induction ratio gradually declined and scarcely any induction could be detected after the 15th passage for cells grown in serum and after the 25th passage for cell lines grown without serum.

Activity is depressed in mouse neuroblastoma cells (NIE-115) grown in synthetic N2 medium lacking riboflavin. MAO activity in depleted cells is stimulated by added riboflavin, and this recovery is blocked by inhibitors of RNA and protein synthesis, and not by an inhibitor of protein glycosylation. Recovery from riboflavin depletion appears to depend upon new RNA and protein synthesis, and not on the addition of FAD cofactor to an inactive MAO precursor.

    Notes: Cultured rat aortic smooth muscle cells (SMC) were examined by electron microscopy and found to contain polygonal networks of 75 A° thin myofilament bundles. The cells also had large bundles of longitudinally aligned thin myofilaments with periodically spaced dense bodies. Abundant plasmalemmal vesicles were present at the cell periphery, and the cells were connected by desmosomes. Intercellular spaces contained sparse amounts of elastic fibers, a material generally present in SMC cultures. Analyses of amino acids by automated column-chromatography showed that isodesmosine and desmosine, two amino acid residues unique for elastin, were present. Accordingly, it was concluded that polygonal networks, previously detected solely in cultured nonmuscle cells, were present in SMC.Other findings suggest (1) a change in myofilament arrangement takes place during cell migration, and (2) rat aortic SMC grown in tissue culture flasks is an important experimental tool in the study of cell motility since such myofilament rearrangements were observed to occur up to fourteen days in first passage.

    Notes: This paper describes an extension of an earlier study on oscillations which can develop on distillation trays. The oscillations take the form of violent lateral movements of the gas/liquid biphase, and they are generally undesirable for good column performance. It has been found that lower density gases need higher velocities to initiate oscillation, and vice versa for higher densities. The predictive method for the onset of oscillations proposed previously appears to remain valid for the range of gas density covered, and for small hole size trays. The presence of oscillations has been shown to increase weeping significantly.

    Notes: The numerical method of orthogonal collocation is used within a thermodynamically consistent framework to calculate equilibrium vapor compositions from P-T-x data for binary systems. The method may be used for isothermal or isobaric data in both the normal liquid and in the critical region, and it applies to any choice of standard states.To use the method it is necessary to have experimental or estimated values of vapor phase fugacity coefficients for both components in the mixture and liquid molar volumes (isothermal case) or heats of mixing (isobaric case). If vapor composition data are available, each set of P-T-x-y data may be checked for thermodynamic consistency by comparing y (experimental) with y (calculated). Illustrative calculations are given. The method is shown to provide an excellent procedure for obtaining Henry's constants from P-x data for systems with one noncondensable component.

In each case the problem can still be solved as a linear program. In the first two cases, the problem size is doubled, while for the row dependent case a finitely terminating cutting plane algorithm is constructed.

The results are derived using the model of the linear program with flexibility previously introduced for the inequality constrained case. However, because a feasible region common to all perturbed constraint sets does not exist in the equality constrained case, a flexibility set which intersects all perturbed sets individually rather than jointly is defined. The flexibility set is constructed by identifying a finite subset of all perturbed constraint sets which need to be investigated. Three cases for the equality constrained problem are considered: independent variations in the array elements, column dependent variations, and row dependent variations. In each case the problem is solved using a possibly large but decomposable linear program. In the first two cases, this program needs to be solved only once; while in the row dependent case an iterative but finite solution procedure is required.

    Notes: The problem of synthesizing an optimal low risk reaction network from unreliable reactions is formulated. An expected cost criterion function for decision-making is derived both for the case of a continuously operating network of reactions and for the case of reaction paths that involve batch reactions. It is shown that in general the decision space has a nonserial structure and the search for the optimal path will involve use of network search methods. An example is given where the optimal reaction path is synthesized for deoxyribonucleic acid. This example demonstrates the decision-making strategy for a class of bath reaction systems.

    Notes: The transformation of the energy equation from the mass average to the volume average reference frame exposes a pseudo Dufour effect. The theoretical model for absorption into a stagnant liquid including this effect, the heat of absorption, variable density, an interfacial velocity, and an arbitrary equilibrium condition at the interface is able to predict experimental data over a wide range of conditions. The data consisted of liquid temperature profiles and mass transfer rates into a stagnant liquid. The ammonia-water system exhibited interfacial temperature rises from 6.9°K to 18.2°K. The interfacial temperature rise for the propane-decane system was 1.3°K. Simpler theoretical models were able to predict the mass transfer rates or the temperature profiles.

    Notes: Bubble size is one of the most important parameters in the design and simulation of a fluidized-bed reactor.A correlation of the bubble size and growth in fluidized beds of various diameters is developed. A maximum bubble diameter determined from the bubble coalescence is incorporated in the correlation to relate the effect of the bed diameter on the bubble size.Experimental data of bubble size reported are used to develop and test the validity of the correlation. The bubble diameters calculated using this correlation show good agreement with the observed bubble diameters.

    Notes: Experimental data have been obtained using an Analytical Ultracentrifuge, in regions much closer to the interface in immiscible liquid/liquid systems than have hitherto been reported. The water/toluene system was investigated with acetic and propionic acids as the transfer solutes. The direction of mass transfer was from the water to the toluene phase. The data derived from the study indicates that the maximum mass transfer flux in the water/toluene system occurs, not at the interface, but at regions further away from it and that the water/toluene system offers significant interfacial resistance of about 104 s/cm to the transfer of acetic and propionic acids. The water/n-butanol system was also studied and found to offer negligible resistance to the transfer of acetic acid across the interface.Diffusion coefficients for ethylene glycol, glycerol, and n-butanol in water were also determined with the Ultracentrifuge. The values derived in this study were reproducible to within 5% and were in close agreement with those derived from classical diffusion experiments.

    Notes: The population balances describing the time dependence of the size distribution can, under some conditions, be trasformed by means of a similarity transformation into an ordinary integro-differential equation containing two instead of three variables. If there is compatibility between the transformed equation and the constraints given by the total mass conservation equation and the equation for the total number of particles, a self-preserving spectrum of the first kind can be obtaiened. There are, however, many situations such as the sintering controlled aging of supported metal catalysts, coagulation of colloidal particles in laminar shear flow, and coagulation of colloidal particles in a turbulent flow when the particles are smaller than the size of the smallest eddy for which, although a similarity transformation is possible, the transformed equation has no solution because of incompatibility with the above mentioned constraints. A second kind of self-preserving spectrum is suggested for these situations. The new variables are induced from a particular case for which an analytical result is available. A detailed presentation of the sintering controlled aging of supported metal catalysts is presented.

    Notes: To obtain a semi-theoretical equation for the excess Gibbs energy of a liquid mixture, Guggenheim's quasi-chemical analysis is generalized through introduction of the local area fraction as the primary concentration variable. The resulting universal quasi-chemical (UNIQUAC) equation uses only two adjustable parameters per binary. Extension to multicomponent systems requires no ternary (or higher) parameters.The UNIQUAC equation gives good representation of both vapor-liquid and liquid-liquid equilibria for binary and multicomponent mixtures containing a variety of nonelectrolyte components such as hydrocarbons, ketones, esters, amines, alcohols, nitriles, etc., and water. When well-defined simplifying assumptions are introduced into the generalized quasi-chemical treatment, the UNIQUAC equation reduces to any one of several well-known equations for the excess Gibbs energy, including the Wilson, Margules, van Laar, and NRTL equations.The effects of molecular size and shape are introduced through structural parameters obtained from pure-component data and through use of Staverman's combinatorial entropy as a boundary condition for athermal mixtures. The UNIQUAC equation, therefore, is applicable also to polymer solutions.

    Notes: A method has been developed for selection of the set of torn streams in a process simulation that leads to convergence of a direct substitution calculation in the minimum number of iterations. It is based on the establishment of families of decompositions, the members of each of which have identical convergence behavior. Explicit criteria for choice among families have been found that lead, in almost every case, to a single optimum decomposition family. These results have been verified by application to several chemical process simulation examples.

    Notes: Equations are proposed to predict the three-dimensional motion of droplets in a spray dryer, based on a knowledge of the characteristics of the atomizing device and of the gas flow patterns in the drying chamber. If the droplet size distribution produced by the atomizing device is known or can be assumed, the trajectories of the droplets can be calculated throughout the drying process and hence the evaporative capacity and thermal efficiency of the spray dryer can be predicted.Experimental verification of this theoretical approach was obtained from a study of the drying of calcium lignosulfonate solutions of various concentrations in a 122-cm diam. × 183-cm high laboratory circular concurrent chamber with a conical bottom where the drying air was introduced tangentially near the top.An experimental study of the effects of a number of operating variables on the capacity and the efficiency of the spray dryer was also carried out. These effects were interpreted in terms of the droplet trajectories obtained in each case.

    Notes: Measurements of water flux, solute flux, and electrochemical potential were made simultaneously in a membrane transport system designed for the studies in anomalous osmosis and thermoosmosis using a series of cations with a common anion. The cation exchange membrane employed was dimensionally stable. The anomalous osmosis appeared to be composed of two components, the normal osmosis and a facilitated osmosis; whereas the thermoosmosis was composed solely of a facilitated component. The anomalous water flux did not correlate with the solute flux or the electro-chemical potential. Anomalous osmosis depended strongly upon the cation species.

    Notes: A comprehensive investigation has been made of the boundaries of the regime of bubbling aggregative fluidization. Experiments were done with sand, glass beads, clover seed, and iron shot fluidized with helium, air, and freon-12 in columns 2.5, 5, 10, and 21 cm in diameter. Bed heights ranged from 1 to 60 column diameters, particle diameters from 0.07 to 1.1 mm, particle densities from 1300 to 7600 kg/m3, and gas densities from 0.17 to 5.2 kg/m3. Correlations are presented for the void fraction at the minimum bubbling point and for the superficial fluid velocity at the points of minimum fluidization, minimum bubbling, and minimum slugging.

    Notes: A flash photomicrographic method and modified dye-light transmittance technique are employed to measure size distributions and mixing frequencies in a turbulently agitated flow vessel. This liquid-liquid dispersion is nearly spatially homogenous. Mixing frequencies are strongly dependent on impeller speed and moderately dependent on holdup fraction.

    Notes: Experimental heat transfer correlations were determined for natural convection in mercury in a uniformly heated vertical channel with width-to-height ratios varying from 0.25 to infinity (the single plate limit). Local modified Grashof numbers varied between 108 and 1011. Temperature and velocity profiles were obtained for these same channel configurations and local Grashof numbers. Instantaneous velocity plots indicate the range of conditions studied to proceed from unstable laminar flow well into the transition regime. Heat transfer results are in good agreement with earlier results reported for stable laminar flow which were limited to values of modified Grashof number less than 109.

    Notes: The composite algorithm derived in Part I is used to construct models from pulse responses of lumped and distributed systems, as well as an empirically measured residence time density. Along the way, salient points in the numerical implementation of the algorithm are indicated. In the present recursive procedure, the initial transient is predicted accurately by lower-order partial realizations; successive partial realizations involve progressively longer tails of the pulse response. The zeroth moment of the impulse response is used to advantage when realizations with no zeros are appropriate.
