    Description: Tephra provides regional chronostratigraphical marker horizons that can link different climate archives with highly needed accuracy and precision. The results presented in this work exemplify, however, that the intermittent storage of tephra in ice sheets and during its subsequent iceberg transport, especially during glacial stages, constitutes a potential source of serious error for the application of tephrochronology to Nordic Seas and North Atlantic sediment archives. The peak shard concentration of the rhyolitic component of the North Atlantic Ash Zone II (NAAZ-II) tephra complex, often used to correlate marine and ice core records in Marine Isotope Stage (MIS) 3, is shown to lag the eruption event by ca. 100–400 years in some North Atlantic and Norwegian Sea cores. While still allowing for a correlation of archives on millennial timescales, this time delay in deposition is a major obstacle when addressing the lead–lag relationship on short timescales (years to centuries). A precise and accurate determination of lead–lag relationships between archives recording different parts of the climate system is crucial in order to test hypotheses about the processes leading to abrupt climate change and to evaluate results from climate models.     Description: Direct estimations of turbulent fluxes and atmospheric stability were performed from a sonic anemometer at 50 m height on a meteorological mast at the Horns Rev wind farm in the North Sea. The stability and flux estimations from the sonic measurements are compared with bulk results from a cup anemometer at 15 m height and potential temperature differences between the water and the air above. Surface flux estimations from the advanced weather research and forecast (WRF) model are also validated against the sonic and bulk data. The correlation between the sonic and bulk estimates of friction velocity is high and the highest among all velocity comparisons. From the sonic–bulk–WRF inter-comparison, it is found that the atmospheric stability measures at the sonic height tend to be closer to the neutral value than the WRF and bulk estimates, which are performed within an air layer closer to the surface, not only from a systematic bulk and WRF under-prediction of the friction velocity when compared with the sonic value but also because of the lower magnitude of the sonic heat flux compared with that from the WRF simulations. Although they are not measured but parameterized or estimated, the bulk–WRF comparisons of friction velocity and 10 m wind speed show good agreement. It is also shown that on a long-term basis, the WRF and bulk estimates of stability are nearly equal and that a correction towards a slightly stable atmospheric condition has to be applied to the long-term wind profile at Horns Rev and at other locations over the North Sea, the correction being larger for points close to the coast.

    Description: Metagenomic techniques are used to analyse bacterial communities allowing both culturable and unculturable species to be represented. However, the screening of oral metagenomic samples can be hindered by high animal host DNA content. This study evaluated methods for the reduction of human DNA concentrations within oral metagenomic samples. Plaque samples were collected from 27 patients presenting with periodontal disease and treated to remove human DNA using either selective lysis of eukaryotic cells at several buffer concentrations or differential centrifugation after treatment with trypsin and/or detergents. Human and bacterial DNA levels were determined by quantitative polymerase chain reaction (qPCR). The human DNA content of plaque extracts was significantly reduced by all treatments compared with an untreated control (P 〈 0.05). However, differential centrifugation simultaneously reduced the bacterial DNA content unless samples were pretreated with a detergent. Observations of Gram stained samples that were processed using differential centrifugation without detergent suggest that many bacteria remain adhered to human cells. An approach that uses differential centrifugation in parallel with selective lysis is recommended to fully represent the oral microbiota in metagenomic samples, including those tightly adhered to human cells and more delicate bacteria such as Mycoplasma.

    Description: Although it has been more than 30 years since the discovery of deep-sea hydrothermal vents, comprehending the interconnections between hydrothermal venting and microbial life remains a challenge. Here we investigate abiotic-biotic linkages in low-temperature hydrothermal biotopes at Desperate and Lilliput on the southern Mid-Atlantic Ridge. Both sites are basalt-hosted and fluids exhibit the expected chemical signatures. However, contrasting crustal permeabilities have been proposed, supporting pervasive mixing at Desperate but restricting circulation at Lilliput. In Desperate fluids, sulfide and O2 were readily available but H2 hardly detectable. Under incubation conditions (oxic unamended, sulfide-spiked, oxic and anoxic H2 -spiked at 18°C), only sulfide oxidation by Thiomicrospira fuelled biomass synthesis. Microbial phylogenies from Desperate incubation experiments resembled those of the natural samples suggesting that the incubation conditions mimicked the environment. In Lilliput fluids, O2 was limited, whereas sulfide and H2 were enriched. Autotrophy appeared to be stimulated by residual sulfide and by amended H2 . Yet, based on bacterial phylogenies only conditions in anoxic H2 -spiked Lilliput incubations appeared similar to parts of the Lilliput habitat. In anoxic H2 -spiked Lilliput enrichments Campylobacteraceae likely supported biomass production through H2 oxidation. We argue that the diverging circulation patterns arising from different subseafloor permeabilities act as major driving forces shaping these biotope structures.

    Description: This article presents a fundamental study on the role of particle breakage on the shear behavior of granular soils using the three-dimensional (3-D) discrete element method. The effects of particle breakage on the stress ratio, volumetric strain, plastic deformation, and shear failure behavior of dense crushable specimens undergoing plane strain shearing conditions are thoroughly investigated through a variety of micromechanical analyses and mechanism demonstrations. The simulation of a granular specimen is based on the effective modeling of realistic fracture behavior of single soil particles, which is demonstrated by the qualitative agreement between the results from platen compression simulations and those from physical laboratory tests. The simulation results show that the major effects of particle breakage include the reduction of volumetric dilation and peak stress ratio and more importantly the plastic deformation mechanisms and the shear failure modes vary as a function of soil crushability. Consistent macro- and micromechanical evidence demonstrates that shear banding and massive volumetric contraction depict the two end failure modes of a dense specimen, which is dominated by particle rearrangement–induced dilation and particle crushing–induced compression, respectively, with a more general case being the combination and competition of the two failure modes in the medium range of soil crushability and confining stress. However, it is further shown that a highly crushable specimen will eventually develop a shear band at a large strain because of the continuous decay of particle breakage.

    Description: Zonal wind anomalies in the western equatorial Atlantic during late boreal winter to early summer precondition boreal summer cold/warm events in the eastern equatorial Atlantic (EEA) that manifest in a strong interannual Atlantic cold tongue (ACT) variability. Local intraseasonal wind fluctuations, linked to the St. Helena anticyclone, contribute to the variability of cold tongue onset and strength, particularly during years with preconditioned shallow thermoclines. The impact of cold tongue sea surface temperature (SST) anomalies on the wind field in the Gulf of Guinea is assessed.     Description: Marine macroalgae are constantly exposed to epibacterial colonizers. The epiphytic bacterial patterns and their temporal and spatial variability on host algae are poorly understood. To investigate the interaction between marine macroalgae and epiphytic bacteria, this study tested if the composition of epibacterial communities on different macroalgae was specific and persisted under varying biotic and abiotic environmental conditions over a 2-year observation time frame. Epibacterial communities on the co-occurring macroalgae Fucus vesiculosus, Gracilaria vermiculophylla and Ulva intestinalis were repeatedly sampled in summer and winter of 2007 and 2008. The epibacterial community composition was analysed by denaturing gradient gel electrophoresis (DGGE) and 16S rRNA gene libraries. Epibacterial community profiles did not only differ significantly at each sampling interval among algal species, but also showed consistent seasonal differences on each algal species at a bacterial phylum level. These compositional patterns re-occurred at the same season of two consecutive years. Within replicates of the same algal species, the composition of bacterial phyla was subject to shifts at the bacterial species level, both within the same season but at different years and between different seasons. However, 7–16% of sequences were identified as species specific to the host alga. These findings demonstrate that marine macroalgae harbour species-specific and temporally adapted epiphytic bacterial biofilms on their surfaces. Since several algal host-specific bacteria were highly similar to other bacteria known to either avoid subsequent colonization by eukaryotic larvae or to exhibit potent antibacterial activities, algal host-specific bacterial associations are expected to play an important role for marine macroalgae.

    Description: A wind-driven meso-scale pattern of temperature, salinity and oxygen was found along a transect in the northern Bornholm Basin (southern Baltic Sea). Strong winds caused currents along this transect, which shifted cold intermediate water (minimum: 3.6C) towards the south. The transect was surveyed with a towed CTD-system and hydroacoustics in parallel to investigate the distribution of sprat, Sprattus sprattus balticus (Schn.) in relation to the observed meso-scale pattern. In those parts of the transect where the cold intermediate water was observed, sprat were restricted to water layers below the halocline. In other parts of the transect, sprat moved into higher water layers and occupied a wider depth range. The important factor was temperature, which set an upper limit to the vertical sprat distribution. The development of hydrography, as measured in the field, was evaluated with a hydrodynamic model.

    Description: Environmental perturbation, climate change and international commerce are important drivers for biological invasions. Climate anomalies can further increase levels of habitat disturbance and act synergistically to elevate invasion risk. Herein, we use a historical data set from the upper San Francisco Estuary to provide the first empirical evidence for facilitation of invasions by climate extremes. Invasive zooplankton species did not become established in this estuary until the 1970s when increasing propagule pressure from Asia coincided with extended drought periods. Hydrological management exacerbated the effects of post-1960 droughts and reduced freshwater inflow even further, increasing drought severity and allowing unusually extreme salinity intrusions. Native zooplankton experienced unprecedented conditions of high salinity and intensified benthic grazing, and life history attributes of invasive zooplankton were advantageous enough during droughts to outcompete native species and colonise the system. Extreme climatic events can therefore act synergistically with environmental perturbation to facilitate the establishment of invasive species.

    Description: Indoor mesocosms were used to study the combined effect of warming and of different densities of overwintering mesozooplankton (mainly copepods) on the spring development of phytoplankton in shallow, coastal waters. Similar to previous studies, warming accelerated the spring phytoplankton peak by ca. 1 day 1C1 whereas zooplankton did not significantly influence timing. Phytoplankton biomass during the experimental period decreased with warming and with higher densities of overwintering zooplankton. Similarly, average cell size and average effective particle size (here: colony size) decreased both with zooplankton density and warming. A decrease in phytoplankton particle size is generally considered at typical footprint of copepod grazing. We conclude that warming induced changes in the magnitude and structure of the phytoplankton spring bloom cannot be understood without considering grazing by overwintering zooplankton.

    Description: Recent studies suggest that cigarette smoking may trigger the development of psoriasis through oxidative, inflammatory and genetic mechanisms. Smoking initiates formation of free radicals that stimulate cell signalling pathways active in psoriasis including mitogen-activated protein kinase (MAPK), nuclear factor-κB (NF-κB) and Janus kinase/signal transducers and activators of transcription (JAK-STAT). Smoking damages the skin by increasing formation of reactive oxygen species and decreasing the gene expression of antioxidants. Nicotine also stimulates innate immune cells integral to the pathogenesis of psoriasis including dendritic cells, macrophages and keratinocytes. These cells release cytokines that activate T lymphocytes and perpetuate a cycle of chronic inflammation. Smoking also enhances expression of genes known to confer an increased risk of psoriasis, including HLA-Cw6, HLA-DQA1*0201 and CYP1A1. Improved understanding of the possible link between smoking and psoriasis pathogenesis may provide further insight into mechanisms underlying smoking, psoriasis and risk of subsequent cardiovascular disease.

    Description: Parameter estimation for stochastic dynamic systems is a core problem for the environmental and ecological sciences. This study considers parameter estimation for a simple nonlinear numerical model of marine biogeochemistry. We present a nonlinear stochastic differential equation based model for estimating parameters from non-Gaussian ocean measurements collected at a coastal ocean observatory. A sequential Monte Carlo procedure, or particle filter, provides for estimation of the time evolving state and also the basis for parameter estimation. Two approaches for estimating static parameters of the system are contrasted. The first is based on likelihood calculations, and the second on augmenting the system state with the static parameters. Sensitivity analysis identified two ecological parameters (in the differential equations model) and one statistical parameter (governing the level of dynamical noise) as candidates for estimation. Computed likelihood surfaces were found to be rough due to the sample based calculations; they also indicated the ubiquitous problem of ecological parameter dependence and identifiability. A modified state augmentation procedure, incorporating a smoothed bootstrap step, was used here for parameter estimation. Realizations for the parameter values provided by this method allowed for calculation of moments and density estimates that matched well the properties of the likelihood. Incorporation of prior information on the parameters was also considered within this context. It is concluded that such a modified state augmentation procedures provides a promising avenue in parameter estimation in numerical models

    Description: We present a 3-D joint inversion framework for seismic, magnetotelluric (MT) and scalar and tensorial gravity data. Using large-scale optimization methods, parallel forward solvers and a flexible implementation in terms of model parametrization allows us to investigate different coupling approaches for the various physical parameters involved in the joint inversion. Here we compare two different coupling approaches, direct parameter coupling where we calculate conductivities and densities from seismic slownesses and cross-gradient coupling, where each model cell has an independent value for each physical property and a structural similarity is enforced through a term in the objective function. For both types of approaches we see an improvement of the inversion results over single inversions when the inverted data sets are generated from compatible models. As expected the direct coupling approach results in a stronger interaction between the data sets and in this case better results compared to the cross-gradient coupling. In contrast, when the inverted MT data is generated from a model that violates the parameter relationship in some regions but conforms with the cross-gradient assumptions, we obtain good results with the cross-gradient approach, while the direct coupling approach results in spurious features. This makes the cross-gradient approach the first choice for regions were a direct relationship between the physical parameters is unclear.

    Description: he use and development of post-genomic tools naturally depends on large-scale genome sequencing projects. The usefulness of post-genomic applications is dependent on the accuracy of genome annotations, for which the correct identification of intron-exon borders in complex genomes of eukaryotic organisms is often an error-prone task. Although automated algorithms for predicting intron-exon structures are available, supporting exon evidence is necessary to achieve comprehensive genome annotation. Besides cDNA and EST support, peptides identified via MS/MS can be used as extrinsic evidence in a proteogenomic approach. We describe an improved version of the Genomic Peptide Finder (GPF), which aligns de novo predicted amino acid sequences to the genomic DNA sequence of an organism while correcting for peptide sequencing errors and accounting for the possibility of splicing. We have coupled GPF and the gene finding program AUGUSTUS in a way that provides automatic structural annotations of the Chlamydomonas reinhardtii genome, using highly unbiased GPF evidence. A comparison of the AUGUSTUS gene set incorporating GPF evidence to the standard JGI FM4 (Filtered Models 4) gene set reveals 932 GPF peptides that are not contained in the Filtered Models 4 gene set. Furthermore, the GPF evidence improved the AUGUSTUS gene models by altering 65 gene models and adding three previously unidentified genes.

    Description: The region offshore Eastern Java represents one of the few places where the early stage of oceanic plateau subduction is occurring. We study the little investigated Roo Rise oceanic plateau on the Indian plate, subducting beneath Eurasia. The presence of the abnormal bathymetric features entering the trench has a strong effect on the evolution of the subduction system, and causes additional challenges on the assessment of geohazard risks. We present integrated results of a refraction/wide-angle reflection tomography, gravity modelling, and multichannel reflection seismic imaging using data acquired in 2006 south of Java near 113°E. The composite structural model reveals the previously unresolved deep geometry of the oceanic plateau and the subduction zone. The oceanic plateau crust is on average 15 km thick and covers an area of about 100 000 km2. Within our profile the Roo Rise crustal thickness ranges between 18 and 12 km. The upper oceanic crust shows high degree of fracturing, suggesting heavy faulting. The forearc crust has an average thickness of 14 km, with a sharp increase to 33 km towards Java, as revealed by gravity modelling. The complex geometry of the backstop suggests two possible models for the structural formation within this segment of the margin: either accumulation of the Roo Rise crustal fragments above the backstop or alternatively uplift of the backstop caused by basal accumulation of crustal fragments. The subducting plateau is affecting the stress field within the accretionary complex and the backstop edge, which favours the initiation of large, potentially tsunamogenic earthquakes such as the 1994 Mw= 7.8 tsunamogenic event.

    Description: The Kumano fore-arc basin overlies the Nankai accretionary prism, formed by the subduction of the Philippine Sea Plate beneath the Eurasian plate offshore the Kii Peninsula, SW Honshu, Japan. Seismic surveys and boreholes within the framework of the Nankai Trough Seismogenic Zone Experiment (NanTroSEIZE) project show evidence of gas hydrates and free gas within the basin. Here we use high-quality borehole sonic data from Integrated Oceanic Drilling Program (IODP) Site C0009 to quantify the free gas distribution in the landward part of the basin. The Brie theory is used to quantify gas content from sonic logs, which are calibrated from laboratory measurements on drill cores. First, we show that the sonic data are mainly sensitive to the fluid phase filling the intergranular pores (effective porosity), rather than to the total porosity that includes water bound to clay minerals. We then compare the effective porosity to lithodensity-derived porosity that acts as a proxy for total porosity. The combination of these two data sets also allows assessment of clay mineralogy of the sediments. Second, we compute free gas saturation and find a gas-rich interval that is restricted to a lithological unit characterized by a high abundance of wood fragments and lignite. This unit, at the base of the fore-arc basin, is a hydrocarbon source that should be taken into account in models explaining gas distribution and the formation of the bottom-simulating reflector within the Kumano fore-arc basin.

    Description: The coccolithophore Calcidiscus leptoporus (strain RCC1135) was grown in dilute batch culture at CO2 levels ranging from similar to 200 to similar to 1600 mu atm. Increasing CO2 concentration led to an increased percentage of malformed coccoliths and eventually (at similar to 1500 mu atm CO2) to aggregation of cells. Carbonate chemistry of natural seawater was manipulated in three ways: first, addition of acid; second, addition of a IICO3-/CO32- solution; and third, addition of both acid and IICO3-/CO32- solution. The data set allowed the disentangling of putative effects of the different parameters of the carbonate system. It is concluded that CO2 is the parameter of the carbonate system which causes both aberrant coccolithogenesis and aggregation of cells.

    Description: Dependent on the ‘intrinsic’ effects on the crystal lattice of the rock constituents and the diminishing ‘extrinsic’ effects of pores and microcracks, elastic wave velocity versus pressure trends in cracked rocks are characterized by non-linear velocity increase at low pressure. At high pressure the ‘extrinsic’ influence vanishes and the velocity increase becomes approximately linear. Usually, the transition between non-linear and linear behaviour, the ‘crack closure pressure’, is not accessible in an experiment, because actual equipment is limited to lower pressure. For this reason, several model functions for describing velocity—pressure trends were proposed in the literature to extrapolate low-pressure P-wave velocity measurements to high pressures and, in part, to evaluate the ‘intrinsic’ velocity—pressure trend from low-pressure data. Knowing the ‘intrinsic’ velocity trend is of particular importance for the quantification of the crack influence at low pressure, at high pressure, the ‘intrinsic’ trend describes the velocity trend as a whole sufficiently well. Checking frequently used model functions for suitability led to the conclusion that all relations are unsuitable for the extrapolation and, if applicable, the estimation of the ‘intrinsic’ velocity trend. However, it can be shown that the ‘intrinsic’ parameters determined by means of a suitable model function, the zero pressure velocity and the pressure gradient depend on maximum experimental pressure in a non-linear way. Our approach intends to obtain better estimates of particular parameters from observed non-linear behaviour. A converging exponential function is used to approximate particular trends, assuming that the point of convergence of the function represents a better estimate of the zero pressure velocity and the pressure gradient, respectively. Whether the refined ‘intrinsic’ velocity trend meets the ‘true intrinsic’ velocity trend within acceptable errors cannot be proven directly due to missing experimental data at very high pressure. We, therefore, conclude that our approach cannot ensure absolutely certain ‘intrinsic’ velocity trends, however, it can be shown that the optimized trends approximate the ‘true intrinsic’ velocity trend better as all the other relations do.

    Description: The ultramafic-hosted Logatchev hydrothermal field (LHF) is characterized by vent fluids, which are enriched in dissolved hydrogen and methane compared with fluids from basalt-hosted systems. Thick sediment layers in LHF are partly covered by characteristic white mats. In this study, these sediments were investigated in order to determine biogeochemical processes and key organisms relevant for primary production. Temperature profiling at two mat-covered sites showed a conductive heating of the sediments. Elemental sulfur was detected in the overlying mat and metal-sulfides in the upper sediment layer. Micro-profiles revealed an intensive hydrogen sulfide flux from deeper sediment layers. Fluorescence in situ hybridization showed that filamentous and vibrioid, Arcobacter-related Epsilonproteobacteria dominated the overlying mats. This is in contrast to sulfidic sediments in basalt-hosted fields where mats of similar appearance are composed of large sulfur-oxidizing Gammaproteobacteria. Epsilonproteobacteria (7-21%) and Deltaproteobacteria (20-21%) were highly abundant in the surface sediment layer. The physiology of the closest cultivated relatives, revealed by comparative 16S rRNA sequence analysis, was characterized by the capability to metabolize sulfur components. High sulfate reduction rates as well as sulfide depleted in (34)S further confirmed the importance of the biogeochemical sulfur cycle. In contrast, methane was found to be of minor relevance for microbial life in mat-covered surface sediments. Our data indicate that in conductively heated surface sediments microbial sulfur cycling is the driving force for bacterial biomass production although ultramafichosted systems are characterized by fluids with high levels of dissolved methane and hydrogen

    Description: A typical marine controlled-source electromagnetic system consists of an electric dipole transmitter and one or more electric dipole receivers. The objective of a survey is to determine the seafloor resistivity by recording the electromagnetic transients, which diffuse through the earth from the transmitter to the receivers. Accurate knowledge of system geometry is crucial for proper interpretation; errors in the position and orientation of the transmitter and/or the receivers propagate into errors in the predicted seafloor resistivity. We show theoretically that for certain multireceiver set-ups and crustal electrical profiles that the geometry and the seafloor resistivity may be determined independently. A specific example is an experiment proposed in association with NEPTUNE Canada. Here, we have already deployed an electric dipole transmitter with a known orientation in a known location. A cabled streamer of receivers may be towed by a survey vessel in the vicinity of the transmitter on a known heading. For this configuration, an eigenparameter analysis of two seafloor models consisting of (1) a halfspace and (2) a resistive layer buried within a halfspace shows that the resistivity structure of the seafloor can be independently resolved from the cable location. Further studies of these two models also indicate that the position of the streamer must be roughly known in advance on the order of a hundred metres to be used as a suitable starting model in a non-linear inversion. The crucial information is contained in the parts of the pulse which travel through the seawater and which act as a calibration path. Such information is absent for a static DC method.

    Description: To understand the influence of changing surface ocean pH and carbonate chemistry on the coccolithophore Emiliania huxleyi, it is necessary to characterize mechanisms involved in pH homeostasis and ion transport. Here, we measured effects of changes in seawater carbonate chemistry on the fluorescence emission ratio of BCECF (2',7'-bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein) as a measure of intracellular pH (pH(i)). Out of equilibrium solutions were used to differentiate between membrane permeation pathways for H+, CO(2) and HCO(3)-. Changes in fluorescence ratio were calibrated in single cells, resulting in a ratio change of 0.78 per pH(i) unit. pH(i) acutely followed the pH of seawater (pH(e)) in a linear fashion between pH(e) values of 6.5 and 9 with a slope of 0.44 per pH(e) unit. pH(i) was nearly insensitive to changes in seawater CO(2) at constant pH(e) and HCO(3)-. An increase in extracellular HCO(3)- resulted in a slight intracellular acidification. In the presence of DIDS (4,4'-diisothiocyanatostilbene-2,2'-disulfonic acid), a broad-spectrum inhibitor of anion exchangers, E. huxleyi acidified irreversibly. DIDS slightly reduced the effect of pH(e) on pH(i). The data for the first time show the occurrence of a proton permeation pathway in E. huxleyi plasma membrane. pH(i) homeostasis involves a DIDS-sensitive mechanism.

    Description: The continental margin of Nicaragua and Costa Rica is characterized by significant lateral changes from north to south such as a decreasing dip of the slab, a decreasing magma production and a shift in the volcanic front. To investigate this transition, a joint on- and offshore local earthquake tomography was performed. Low P-wave velocities and high Vp/Vs ratios, indicative for hydration, were found in the upper-mantle and lowermost crust beneath the Sandino Basin. The mantle wedge hydration can be estimated to 2.5 wt. per cent beneath south Nicaragua. In contrast, the mantle wedge beneath north Costa Rica is weakly or not hydrated. The hydration leads to a local gap in the seismicity in Nicaragua. The lateral transition between the hydrated and non-hydrated areas occurs within a distance of about 10 km. This transition coincides with a change in the crustal thickness in the order of 5–10 km, thickening to the south, and in the tectonic regimes. The change in the tectonic regimes towards a stronger extension along the margin of Nicaragua could be the key for understanding the observations: the extension may support the opening of pathways for a wide zone of fluid migration and hydration through the overriding plate which are identified with areas of low Vp, high Vp/Vs and low seismicity.

    Description: The analysis of Cobalt (Co) at low pM concentrations in seawater with Adsorptive Cathodic Stripping Voltammetry involves high concentrations of sodium nitrite (NaNO2) to enhance the signal in an electrocatalytic reaction. In this study we found three substitutes for NaNO2 that critically affected the sensitivity. Optimisation of a method with potassium bromate (KBrO3) resulted in an excellent detection limit (0.9 pM) after a 90 s adsorption period. Reactant concentration and consumption were 10× reduced compared to protocols with NaNO2 and reagent blanks were lower. Accuracy and precision were verified with SAFe intercalibration standards and the method was applied using open ocean seawater samples. The reaction mechanism is discussed and differences to NaNO2 are shown.

    Description: Latitudinal variation in thermal reaction norms of key fitness traits may inform about the response of populations to climate warming, yet their adaptive nature and evolutionary potential are poorly known. We assessed the contribution of quantitative genetic, neutral genetic and environmental effects to thermal reaction norms of growth rate for populations of the damselfly Ischnura elegans. Among populations, reaction norms differed primarily in elevation, suggesting that time constraints associated with shorter growth seasons in univoltine, high-latitude as well as multivoltine, low-latitude populations selected for faster growth rates. Phenotypic divergence among populations is consistent with selection rather than drift as QST was greater than FST in all cases. QST estimates increased with experimental temperature and were influenced by genotype by environment interactions. Substantial additive genetic variation for growth rate in all populations suggests that evolution of trait means in different environments is not constrained. Heritability of growth rates was higher at high temperature, driven by increased genetic rather than environmental variance. While environment-specific nonadditive effects also may contribute to heritability differences among temperatures, maternal effects did not play a significant role (where these could be accounted for). Genotype by environment interactions strongly influenced the adaptive potential of populations, and our results suggest the potential for microevolution of thermal reaction norms in each of the studied populations. In summary, the observed latitudinal pattern in growth rates is adaptive and results from a combination of latitudinal and voltinism compensation. Combined with the evolutionary potential of thermal reaction norms, this may affect populations’ ability to respond to future climate warming.

    Description: Biomineralization in the marine phytoplankton Emiliania huxleyi is a stringently controlled intracellular process. The molecular basis of coccolith production is still relatively unknown although its importance in global biogeochemical cycles and varying sensitivity to increased pCO2 levels has been well documented. This study looks into the role of several candidate Ca2+, H+ and inorganic carbon transport genes in E. huxleyi, using quantitative reverse transcriptase PCR. Differential gene expression analysis was investigated in two isogenic pairs of calcifying and non-calcifying strains of E. huxleyi and cultures grown at various Ca2+ concentrations to alter calcite production. We show that calcification correlated to the consistent upregulation of a putative HCO3- transporter belonging to the solute carrier 4 (SLC4) family, a Ca2+/H+ exchanger belonging to the CAX family of exchangers and a vacuolar H+-ATPase. We also show that the coccolith-associated protein, GPA is downregulated in calcifying cells. The data provide strong evidence that these genes play key roles in E. huxleyi biomineralization. Based on the gene expression data and the current literature a working model for biomineralization-related ion transport in coccolithophores is presented.

    Description: Detrital fission-track studies on sedimentary basins surrounding eroding mountain belts provide a powerful tool to reconstruct exhumation histories of the source area. However, examples from active arc-trench systems are sparse. In this study, we report detrital apatite fission-track (AFT) data from Holocene and Pleistocene turbiditic trench and modern river sediments at the Chilean margin (36°S-47°S). Sediment petrography and detrital AFT data point to different major sediment sources, underlining the need for multidisciplinary studies: whereas sediment petrography indicates the erosion of large volumes of volcanic detritus, no such volcanic signal is seen in the detrital age pattern. Areally subordinate plutonic units are identified as the main, often unique sources. This result has important implications for studies of fossil systems, where the feeder areas are eroded, and where the youngest age population is often interpreted to indicate active volcanism. For the southernmost part of the study area in the Patagonian Andes, where the source area is mainly composed of granitoids, the sediment is derived from only small portions along the main divide, pointing to focused glacial erosion there. Our detrital AFT data show no exhumational signal that could be related to the subduction of the actively spreading Chile Ridge at c. 47°S and to the opening of a slab window beneath the South American Plate.

    Description: An activity guided isolation of the H2O subextract of the crude extract of Melampyrum arvense L. afforded iridoid glucosides: aucubin (1), melampyroside (2), mussaenoside (3), mussaenosidic acid (4), 8-epi-loganin (5); flavonoids: apigenin (6), luteolin (7), luteolin 7-O-β-glucopyranoside (8); a lignan glycoside dehydrodiconiferyl alcohol 9-O-β-glucopyranoside (9); and benzoic acid (10). β-Sitosterol (11) and a fatty acid mixture (12) were identified as the active principles of the CHCl3 subextract. The structures of the isolates were elucidated by spectroscopic methods, while the composition of 12 was identified by GC-MS after methylation. Luteolin (7) appeared as the most active compound against Trypanosoma brucei rhodesiense and Leishmania donovani (IC50 values 3.8 and 3.0 μg/mL). Luteolin 7-O-β-glucopyranoside (8) displayed the best antiplasmodial activity against Plasmodium falciparum (IC50 value 2.9 μg/mL). This is the first detailed phytochemical study on Turkish M. arvense and the first report of the antiprotozoal effect of Melampyrum species and its constituents

    Description: As part of our continuing research on seaweeds, crude MeOH extracts of two green, three brown and six red algae collected from Marmara, Black, Aegean and Mediterranean Seas were screened. Four parasitic protozoa, i.e. Plasmodium falciparum, Trypanosoma brucei rhodesiense, T. cruzi, Leishmania donovani and the tubercle bacillus Mycobacterium tuberculosis were used as test organisms for the in vitro assays. The selective toxicity of the extracts was also determined against mammalian L6 cells. All seaweed extracts were active against T. brucei rhodesiense; the Dasya pedicellata extract was the most potent (IC50 value 0.37 µg/mL). The same extract also weakly inhibited the growth of T. cruzi (IC50 62.02 µg/mL). All seaweed extracts also showed leishmanicidal activity (IC50 values 16.76–69.98 µg/mL). The majority of the extracts also exhibited antiplasmodial potential and the most potent extracts were those from D. pedicellata (IC50 0.38 µg/mL), Codium bursa (IC50 1.38 µg/mL) and Caulerpa rasemosa (IC50 3.12 µg/mL). One brown and two red algal extracts showed some weak activity against Mycobacterium tuberculosis (MIC values 125–256 µg/mL). Except for the extract of Dasya pedicellata, none of the extracts displayed any cytotoxicity. This is the second study investigating the antiprotozoal activities of Turkish marine algae and identifies Dasya pedicellata, an understudied algal species, as a candidate for further studies.

    Description: Aim  Many aquatic invertebrates produce dormant life-history stages as a means to endure inhospitable environments and to facilitate natural long-distance dispersal, yet we have little understanding of the role of dormant stages as a mechanism for human-mediated introductions of non-indigenous species. We explore the survival of invertebrate dormant eggs in collected ships’ ballast sediment over a 1-year period to determine relative invasion potential across taxa (i.e. rotifers, copepods, cladocerans and bryozoans) and different habitats (freshwater, marine). Location  Canadian Atlantic and Pacific coasts and Laurentian Great Lakes. Methods  During 2007 and 2008, 19 ballast samples were collected as a part of a larger study. The degradation rate of dormant eggs was assessed by enumerating dormant eggs and by conducting viability hatching experiments. Results  Taxa examined included rotifers, copepods, anomopods, onychopods and bryozoans. Dormant eggs of rotifers degraded at the highest rate of all taxa examined, with no viable eggs remaining within 10 months. Copepods showed a less rapid degradation rate than rotifers. The degradation rate of anomopod dormant eggs was significantly slower than that of both rotifers and copepods. Onychopods and bryozoans did not visibly degrade at all over 12 months. Viability hatching experiments were successful for rotifers, copepods, and anomopods. Onychopods and bryozoans did not hatch during any of the three hatching trials. Main conclusions  Dormancy is not equally beneficial to all invertebrate taxa. Our results indicate that dormant eggs of rotifers and copepods degrade at a rapid rate and may not pose high invasion risk. In contrast, the slow degradation rate of anomopod dormant eggs and the lack of degradation of onychopod and bryozoan dormant eggs could result in high invasion risk because of their accumulation in ballast tanks. Species having resistant dormant eggs mostly belong to freshwater taxa making freshwater habitats at higher invasion risk by dormant invertebrates than marine habitats.

    Description: As sessile filter feeders, sponges rely on a highly efficient fluid transport system. Their physiology depends on efficient water exchange, which is performed by the aquiferous system. This prominent poriferan anatomical character represents a dense network of incurrent and excurrent canals on which we lack detailed 3D models. To overcome this, we investigated the complex leucon‐type architecture in the demosponge Tethya wilhelma using corrosion casting, microtomography, and 3D reconstructions. Our integrative qualitative and quantitative approach allowed us to create, for the first time, high‐resolution 3D representations of entire canal systems which were used for detailed geometric and morphometric measurements. Canal diameters lack distinct size classes, and bifurcations are non‐uniformly ramified. A relatively high number of bifurcations show previously unknown and atypical cross‐sectional area ratios. Scaling properties and topological patterns of the canals indicate a more complex overall architecture than previously assumed. As a consequence, it might be more convenient to group canals into functional units rather than hierarchical clusters. Our data qualify the leucon canal system architecture of T. wilhelma as a highly efficient fluid transport system adapted toward minimal flow resistance. Our results and approach are relevant for a better understanding of sponge biology and cultivation techniques.

    Description: Cephalopods are extraordinary molluscs equipped with vertebrate-like intelligence and a unique buoyancy system for locomotion. A growing body of evidence from the fossil record, embryology and Bayesian molecular divergence estimations provides a comprehensive picture of their origins and evolution. Cephalopods evolved during the Cambrian (∼530 Ma) from a monoplacophoran-like mollusc in which the conical, external shell was modified into a chambered buoyancy apparatus. During the mid-Palaeozoic (∼416 Ma) cephalopods diverged into nautiloids and the presently dominant coleoids. Coleoids (i.e. squids, cuttlefish and octopods) internalised their shells and, in the late Palaeozoic (∼276 Ma), diverged into Vampyropoda and the Decabrachia. This shell internalisation appears to be a unique evolutionary event. In contrast, the loss of a mineralised shell has occurred several times in distinct coleoid lineages. The general tendency of shell reduction reflects a trend towards active modes of life and much more complex behaviour.

    Description: The increasing market demand for cephalopods and the experiences obtained with different species has boosted the interest in developing their culture in Latin America. In 2008, an international workshop was held in Puerto Montt, Chile, with 14 experts in experimental cephalopods aquaculture from Brazil, Chile, Spain, and Mexico. Several topics were approached within the holobenthic species Octopus maya and the merobenthic species Enteroctopus megalocyathus, Octopus vulgaris, and Robsonella fontaniana. Part of the conclusions demonstrated that the two greatest difficulties for their production were survival of paralarvae for merobenthic species, and survival of early juveniles for holobenthic species. Besides, there is a need to study the endogenous and exogenous factors affecting health and nutritional status of embryos, paralarvae, and juveniles. These stages, which may limit the culture, should be extensively studied in order to develop the appropriate environmental conditions and culture systems for the physiological and behavioral requirements, from egg incubation up to juveniles to reach a grow-out phase.

    Description: Stable isotope analysis has emerged as one of the primary means for examining the structure and dynamics of food webs, and numerous analytical approaches are now commonly used in the field. Techniques range from simple, qualitative inferences based on the isotopic niche, to Bayesian mixing models that can be used to characterize food‐web structure at multiple hierarchical levels. We provide a comprehensive review of these techniques, and thus a single reference source to help identify the most useful approaches to apply to a given data set. We structure the review around four general questions: (1) what is the trophic position of an organism in a food web?; (2) which resource pools support consumers?; (3) what additional information does relative position of consumers in isotopic space reveal about food‐web structure?; and (4) what is the degree of trophic variability at the intrapopulation level? For each general question, we detail different approaches that have been applied, discussing the strengths and weaknesses of each. We conclude with a set of suggestions that transcend individual analytical approaches, and provide guidance for future applications in the field.

    Description: Tomography is like a photograph that was taken by a camera with blurred and defective lenses that deform the shapes and colours of objects. Reporting quantitative parameters derived from tomographic inversion is not always adequate because tomographic results are often strongly biased. To quantify the results of tomographic inversion, we propose a forward modelling and tomographic inversion (FM&TI) approach that aims to find a more realistic solution than conventional tomographic inversion. The FM&TI scheme is based on the assumption that if two tomograms derived from the inversion of observed and synthetic data are identical, the synthetic structure may appear to be closer to the real unknown structure in the ground than the inversion result. However, the manual design of the synthetic velocity distribution is usually time-consuming and ambiguous. In this study, we propose an approach that automatically searches for a probabilistic model. In this approach, a synthetic model is iteratively updated while taking into account the bias of the model in previous stages of the FM&TI performance. Here, we present an example of synthetic modelling and real data processing for an active source refraction data set corresponding to a marine profile across the subduction zone in Chile at about 32°S latitude. A key feature of the model is a low-velocity channel above the subducted oceanic crust, which was defined in the synthetic model and expected in the real case. The conventional first arrival traveltime tomography was barely able to resolve this channel. However, after several iterations of the FM&TI modelling, we succeeded in reconstructing this channel clearly. In the paper, we briefly discuss the nature of this low-velocity subduction channel, and we compare the results with other studies.

    Description: In diverse animal species, from insects to mammals, females display a more efficient immune defence than males. Bateman’s principle posits that males maximize their fitness by increasing mating frequency whereas females gain fitness benefits by maximizing their lifespan. As a longer lifespan requires a more efficient immune system, these implications of Bateman’s principle may explain widespread immune dimorphism among animals. Because in most extant animals, the provisioning of eggs and a higher parental investment are attributes of the female sex, sex-role reversed species provide a unique opportunity to assess whether or not immune dimorphism depends on life history and not on sex per se. In the broad-nosed pipefish Syngnathus typhle, males brood and nourish the eggs in a ventral pouch and thus invest more into reproduction than females. We found males to have a more active immune response both in field data from four populations and also in an experiment under controlled laboratory conditions. This applied to different measures of immunocompetence using innate as well as adaptive immune system traits. We further determined the specificity of immune response initiation after a fully factorial primary and secondary exposure to a common marine pathogen Vibrio spp. Males not only had a more active but also a more specific immune defence than females. Our results thus indeed suggest that the sex that invests more into the offspring has the stronger immune defence.

    Description: Host–parasite coevolution can lead to a variety of outcomes, but whereas experimental studies on clonal populations have taken prominence over the last years, experimental studies on obligately out-crossing organisms are virtually absent so far. Therefore, we set up a coevolution experiment using four genetically distinct lines of Tribolium castaneum and its natural obligately killing microsporidian parasite, Nosema whitei. After 13 generations of experimental coevolution, we employed a time-shift experiment infecting hosts from the current generation with parasites from nine different time points in coevolutionary history. Although initially parasite-induced mortality showed synchronized fluctuations across lines, a general decrease over time was observed, potentially reflecting evolution towards optimal levels of virulence or a failure to adapt to coevolving sexual hosts.

    Description: The protist Labyrinthula zosterae (Phylum Bigyra, sensu Tsui et al. 2009) has been identified as a causative agent of wasting disease in eelgrass (Zostera marina), of which the most intense outbreak led to the destruction of 90% of eelgrass beds in eastern North America and western Europe in the 1930s. Outbreaks still occur today, albeit at a smaller scale. Traditionally, L. zosterae has been quantified by measuring the necrotic area of Z. marina leaf tissue. This indirect method can however only lead to a very rough estimate of pathogen load. Here, we present a quantitative real-time polymerase chain reaction (qPCR) approach to directly detect and quantify L. zosterae in eelgrass tissue. Based on the internal transcribed spacer (ITS) sequences of rRNA genes, species-specific primers were designed. Using our qPCR, we were able to quantify accurately and specifically L. zosterae load both from culture and eelgrass leaves using material from Europe and North America. Our detection limit was less than one L. zosterae cell. Our results demonstrate the potential of this qPCR assay to provide rapid, accurate and sensitive molecular identification and quantification of L. zosterae. In view of declining seagrass populations worldwide, this method will provide a valuable tool for seagrass ecologists and conservation projects.

    Description: We explore the attenuation in the export ratio of jelly-POM (particulate organic matter) with depth as a function of the decay rate, temperature, and sedimentation rate. Using data from the Vertical Transport In the Global Ocean project, we compare ratios computed with the Martin-curve, with a particle-based parameterization, and with sediment-trap data. Owing to the temperature dependence of the decay rate (Q10 5 4.28), the jelly-POM export ratio below 500m is 2045% larger in subpolar and temperate areas than in the tropics. Vertical migration of gelatinous zooplankton leads to a variable starting depth of a jelly fall (death depth), which governs the start of remineralization, and the fate of the biomass. Owing to the absence of observations, we employ a sinking speed matrix ranging from 100 m d21 to 1500 m d21 to represent slow- and fast-sinking carcasses. The assumption of a constant decay rate k independent of temperature in other particle-based models may not be appropriate. These results provide information for including jelly-POM in global biogeochemical model formulations.

    Description: Fucoidan, a sulfated polysaccharide in brown seaweed, has various biological activities including anti-tumor activity. We investigated the effects of fucoidan on the apoptosis of human promyeloid leukemic cells and fucoidan-mediated signaling pathways. Fucoidan induced apoptosis of HL-60, NB4, and THP-1 cells, but not K562 cells. Fucoidan treatment of HL-60 cells induced activation of caspases-8, -9, and -3, the cleavage of Bid, and changed mitochondrial membrane permeability. Fucoidan-induced apoptosis, cleavage of procaspases, and changes in the mitochondrial membrane permeability were efficiently blocked by depletion of mitogen-activated protein kinase (MAPK) kinase kinase 1 (MEKK1), and inhibitors of MAPK kinase 1 (MEK1) and c Jun NH2-terminal kinase (JNK). The phosphorylation of extracellular signal-regulated kinase 1/2 (ERK1/2) and JNK was increased in fucoidan-treated HL-60, NB4, and THP-1 cells, but not K562 cells. ERK1/2 activation occurred at earlier times than JNK activation and JNK activation was blocked by MEK1 inhibitor. In addition, fucoidan-induced apoptosis was inhibited by addition of glutathione and/or L-NAME, and fucoidan decreased intracellular glutathione concentrations and stimulated nitric oxide (NO) production. Buthionine-[R,S]-sulfoximine rendered HL-60 cells more sensitive to fucoidan. Depletion of MEKK1 and inhibition of MEK1 restored the intracellular glutathione content and abrogated NO production, whereas inhibition of JNK activation by SP600125 restored intracellular glutathione content but failed to inhibit NO production in fucoidan-treated HL-60 cells. These results suggest that activation of MEKK1, MEK1, ERK1/2, and JNK, depletion of glutathione, and production of NO are important mediators in fucoidan-induced apoptosis of human leukemic cells

    Description: Mutations or environmental factors that result in reversal of conspicuous left–right asymmetries provide an opportunity to study developmental mechanisms. They may also provide insight into evolutionary changes in asymmetry states within and between species. King crabs (family Lithodidae) have a larger right claw and females typically exhibit a dextrally offset abdomen. Nevertheless, I observed a high incidence of left handedness in laboratory reared box crabs (Lopholithodes foraminatus) and captured the first known egg-bearing female lithodid to exhibit reversed asymmetry. This provided a unique opportunity to characterize the reversed phenotype and to compare the incidence of reversed asymmetry in the offspring of normal and reversed females. Asymmetry of the chelae became apparent in the first postzoeal stage (glaucothoe) and handedness was maintained through subsequent instars. Females with larger left claws developed reversed abdominal asymmetry by the fourth crab stage. No reversed asymmetry was observed in the mandibles of zoea larvae or juveniles of either handedness. The incidence of reversed asymmetry in glaucothoe reared from one reversed and three normal females was high (between 20% and 30%), and independent of maternity (P=0.67). Removal of the right cheliped of fourth stage zoeae, and the major cheliped of glaucothoe, did not reverse the direction of asymmetry. Elevated larval rearing temperature also did not affect the frequency of reversed individuals. This lack of evidence for either heritability or induction of handedness is enigmatic. Further investigation of reversed asymmetry in lithodid crabs may provide valuable insights into the development and evolution of bilateral asymmetries.

    Description: Transfer of the small gaseous molecules CO2 and NH3 across biological membranes, long thought to occur solely by simple diffusion, is now known to be facilitated by members of two multigene families: aquaporins (AQP) and rhesus (Rh) proteins. Although it is accepted that AQP1 and Rh proteins are involved in CO2 and NH3 transfer, respectively, the idea that a single channel can exhibit selectivity for both gases is controversial. Indeed, studies using the same in vitro model (human red blood cells) have provided evidence both for and against a role for Rh proteins as CO2 channels. Thus, this study was initiated to provide in vivo evidence for a dual function of Rh proteins as ammonia and CO2 channels. Here, we show that in zebrafish (Danio rerio), direct ammonia–CO2 competition experiments in adults or translational knockdowns of Rh proteins in larvae affects both ammonia and CO2 excretion. These results suggest that Rh proteins in zebrafish may be common pathways for transport of ammonia and CO2.

    Description: The evolution of internal structure plays a pivotal role in the macroscopic response of granular materials to applied loads. A case in point is the so‐called ‘stress–dilatancy relation’, a cornerstone of Soil Mechanics. Numerous attempts have been made to unravel the connection between stress–dilatancy and the evolution of fabric and contact forces in a deforming granular medium. We re‐examine this connection in light of the recent findings on force chain evolution, in particular, that of collective force chain failure by buckling. This study is focussed on two‐dimensional deformation of dense granular assemblies. Analysis of individual and collective force chain bucklings is undertaken using data from a discrete element simulation. It is shown that the kinematics of force chain buckling lead to significant levels of local dilatation being developed in the buckling force chain particles and their confining first‐ring neighbors. Findings from the simulation are used to guide the development of a lattice model of collective, localized force chain buckling. Consideration of the statics and kinematics of this process yields a new stress–dilatancy relation. The physics of buckling, even at its simplest form, introduces a richness into the stress–dilatancy formulation in a way that preserves the essential aspects of fabric evolution, specifically the buckling mode.

    Description: Cormorants, Phalacocorax spp., have great potential to affect recreational fisheries and have increased substantially in abundance over the last decades in UK inland waters. Fabricated refuges provide a potential means of reducing fish losses, yet the benefits of such structures may be marginal if natural shelters are abundant and favoured, or if strong density-dependence limits refuge use. This study examined the efficacy of artificial shelters in mesocosm enclosures that allowed standardised and replicated observations of roach, Rutilus rutilus (L.), distribution. When given a choice between occupying open water, simulated reedbeds and artificial brushwood shelters, roach used brushwood shelters extensively across a range of fish densities. When fish had a choice of occupying open water or reedbeds offering no overhead cover, they actively avoided reedbeds and used open water almost exclusively. Occupation of reedbeds was positively related to the amount of overhead cover they provided. When artificial brushwood shelters and reedbeds offered complete overhead cover, brushwood shelters were occupied twice as much as reedbeds. Artificial shelters may therefore have wide application in stillwaters with abundant reedbeds unless the reedbeds are coupled with extensive overhead cover.

    Description: The Swedish UNESCO World Cultural Heritage site of the Birka and Hovgården Iron Age settlements is well suited for the testing of high-resolution archaeological prospection methods. In May 2006 ground-penetrating radar (GPR) and magnetometer test measurements were conducted at Birka, resulting in data of outstanding quality and new archaeological discoveries, but also demonstrating the need for increased spatial sampling regarding GPR prospection at complex Scandinavian sites. Therefore Birka was selected as a testing ground for a pilot study investigating the suitability of the novel multichannel GPR array system MIRA (MALÅ Imaging Radar Array) for efficient, large-scale GPR surveys with very dense spatial sampling. The study was conducted in May 2008 by MALÅ Geoscience AB in collaboration with the archaeological prospection unit of the Swedish National Heritage Board. The very high-resolution three-dimensional GPR pilot survey demonstrated that it is possible to survey 1 ha and more per day with 8 cm cross-line spacing, mapping archaeological structures in unprecedented resolution, such as postholes of only 25 cm diameter. This paper describes the tested technology and methodology as well as the fieldwork and the results of the study.

    Description: In many geotechnical systems, it is not uncommon to observe failure in zones of high localized strain called shear bands. The existing models predict the existence and the extent of these localizations, but provide little insight into the micromechanics within the shear bands. This research captures and compares the variation in microstructure both inside and outside of shear bands that formed in physical laboratory plane strain and companion numerical two-dimensional discrete element method (DEM) biaxial compression experiments. Unsheared and sheared laboratory specimens of Ottawa 20–30 sand of varying dilatancy were solidified using a two-stage resin impregnation procedure. The solidified specimens were sectioned and the resulting surfaces were prepared for microstructure observation using optical bright-field microscopy and stereological analysis. Statistical properties of microstructural parameters for sub-regions in a grid pattern and along predefined inclined zones were determined. Similar measurements were performed on 2D DEM simulation specimens at varying strain levels to characterize the evolution of microstructure with increasing strain. The results showed how differences evolved in the mean, standard deviation, and entropy of void distributions with increasing global strain levels. The results indicate how disorder increases and that the material within the shear band does not adhere to the classical concept of critical state, but reaches a terminal void ratio that is largely a function of initial void ratio. Furthermore, there appears to be a transition zone between the far field and the fully formed shear block, as opposed to an abrupt delineation as is traditionally inferred.

    Description: Gas hydrate-related bottom-simulating reflectors mark the phase boundary between hydrate and free gas in the subsurface, and therefore may be used to estimate geothermal gradients and hence heat flow. The depth and temperature of the phase boundary depend on the composition of the hydrateforming gas and of the pore fluid. In the absence of direct sampling, these compositions remain unknown. We develop an alternative approach that is less sensitive to compositional uncertainties and can be applied when the bottom-simulating reflector is densely sampled in a region with significant seabed relief. We apply this approach to a three-dimensional seismic dataset from the eastern Black Sea.

    Description: Summary 1. Parasitized females in mammals, fish and birds can enhance the immune defence of their offspring by transferring specific antibodies for the embryo. Likewise, social insect mothers transfer immunity despite the fact that invertebrates lack antibodies. 2.  Female trans-generational immune priming is consistent with parental investment theory, because mothers invest more into rearing their offspring than fathers. However, when immune priming is not directly linked to parental care, as is often the case in insects that abandon their eggs after oviposition, both sexes might benefit from protecting their offspring. 3. Using the red flour beetle, Tribolium castaneum, we show that after parental exposure to heat-killed bacteria, trans-generational immune priming occurs through fathers as well as mothers. 4. This novel finding challenges the traditional view that males provide only genes to their offspring in species without paternal care, and raises the possibility of a division of tasks with respect to immune protection between parents.

    Description: During the last decade, global surface temperatures did not increase as rapidly as in the preceding decades. Although relatively small compared to the observed centennial scale global warming, it has renewed interest in understanding and even predicting climate on time scales of decades, and sparked a community initiative on near‐term prediction that will feature in the fifth intergovernmental panel on climate change assessment report. Decadal prediction, however, is in its infancy, with only a few publications existing. This article has three aims. The first is to make the case for decadal prediction. Decadal fluctuations in global climate similar to that of recent decades were observed during the past century. Associated with large regional changes in precipitation and climate extremes, they are of socioeconomic importance. Climate models, which capture some aspects of observed decadal variability, indicate that such variations might be partly predictable. The second aim is to describe the major challenges to skilful decadal climate prediction. One is poor understanding of mechanisms of decadal climate variability, with climate models showing little agreement. Sparse observations in the past, particularly in the ocean, are also a limiting factor to developing and testing of initialization and prediction systems. The third aim is to stress that despite promising initial results, decadal prediction is in a highly experimental stage, and care is needed in interpreting results and utilizing data from such experiments. In the long‐term, decadal prediction has the potential to improve models, reduce uncertainties in climate change projections, and be of socioeconomic benefit. Copyright © 2010 John Wiley & Sons, Ltd.

    Description: Recent seismic evidence suggested that most oceanic plate hydration is associated with trench-outer rise faulting prior to subduction. Hydration at trenches may have a significant impact on the subduction zone water cycle. Previous seismic experiments conducted to the northwest of Nicoya Peninsula, Northern Costa Rica, have shown that the subducting Cocos lithosphere is pervasively altered, which was interpreted to be due to both hydration (serpentinization) and fracturing of the crustal and upper-mantle rocks. New seismic wide-angle reflection and refraction data were collected along two profiles, running parallel to the Middle American trench axis offshore of central Nicaragua, revealing lateral changes of the seismic properties of the subducting lithosphere. Seismic structure along both profiles is characterized by low velocities both in the crust and upper mantle. Velocities in the uppermost mantle are found to be in the range 7.3–7.5 km s−1; thus are 8–10 per cent lower than velocities typical for unaltered peridotites and hence confirm the assumption that serpentinization is a common process at the trench-outer rise area offshore of Nicaragua. In addition, a prominent velocity anomaly occurred within the crust beneath two seamounts. Here, velocity reduction may indicate increased porosity and perhaps permeability, supporting the idea that seamounts serve as sites for water percolation and circulation.

    Description: Wide-angle seismic data have been used to determine the velocity and density structure of the crust and uppermost mantle beneath the Cape Verdes mid-plate swell. Seismic modelling reveals a ‘standard’ oceanic crust, ∼8 km in thickness, with no direct evidence for low-density bodies at the base of the crust. Gravity anomaly modelling within the constraints and resolution provided by the seismic model, does not preclude, however, a layer of crustal underplate up to 3 km thick beneath the swell crest. The modelling shows that while the seismically constrained crustal structure accounts for the short-wavelength free-air gravity anomaly, it fails to fully explain the long-wavelength anomaly. The main discrepancy is over the swell crest where the gravity anomaly, after correcting for crustal structure, is higher by about 30 mGal than it is over its flanks. The higher gravity can be explained if the top 100 km of the mantle beneath the swell crest is less dense than its surroundings by 30 kg m−3. The lack of evidence for low densities and velocities in the uppermost mantle, and high densities and velocities in the lower crust, suggests that neither a depleted swell root or crustal underplate are the origin of the observed shallower-than-predicted bathymetry and that, instead, the swell is most likely supported by dynamic uplift associated with an anomalously low density asthenospheric mantle.

    Description: Aim: We investigate the relationship between local and regional richness in marine fouling assemblages using an expanded and globally replicated approach by incorporating two dimensions of diversity (taxonomic and functional) and different successional stages. Location: Global. Methods: In eight different biogeographic regions (Australia, Brazil, Chile, England, Italy, Japan, Portugal and Sweden) 68 polyvinylchloride (PVC) panels (15 × 15 × 0.3 cm) were deployed for colonization. Communities colonizing panels were analysed by measuring percentage cover at each of four different successional ages: 2, 4, 6 and 8 months. Local richness was assessed as the average number of species and functional groups (FGs) per panel and regional richness was evaluated as the estimated (Jack2) asymptote of the sample-accumulation curves for species and FG on experimental panels. Results: We found that the shape of the relationship between local and regional richness depended on successional stage and the type of richness considered, i.e. taxonomic or functional richness. Hardly any relationship was detectable between local taxonomic richness and regional taxonomic richness at any successional stage. In contrast, the relation between local functional and regional functional richness shows a unimodal pattern of change during succession, passing through the stages ‘independent’, ‘unsaturated rising’, ‘saturated rising’ and once again ‘independent’. Main conclusions: The relationship between local and regional richness, whether taxonomic or functional, frequently displays independence of the two scales, particularly in early and late phases of the successional process.

    Description: Novel high-pressure biotechnical systems that were developed and applied for the study of anaerobic oxidation of methane (AOM) are described. The systems, referred to as high-pressure continuous incubation system (HP-CI system) and high-pressure manifold-incubation system (HP-MI system), allow for batch, fed-batch, and continuous gas-phase free incubation at high concentrations of dissolved methane and were designed to meet specific demands for studying environmental regulation and kinetics as well as for enriching microbial biomass in long-term incubation. Anoxic medium is saturated with methane in the first technical stage, and the saturated medium is supplied for biomass incubation in the second stage. Methane can be provided in continuous operation up to 20 MPa and the incubation systems can be operated during constant supply of gas-enriched medium at a hydrostatic pressure up to 45 MPa. To validate the suitability of the high-pressure systems, we present data from continuous and fed-batch incubation of highly active samples prepared from microbial mats from the Black Sea collected at a water depth of 213 m. In continuous operation in the HP-CI system initial methane-dependent sulfide production was enhanced 10- to 15-fold after increasing the methane partial pressure from near ambient pressure of 0.2 to 10.0 MPa at a hydrostatic pressure of 16.0 MPa in the incubation stage. With a hydraulic retention time of 14 h a stable effluent sulfide concentration was reached within less than 3 days and a continuing increase of the volumetric AOM rate from 1.2 to 1.7 mmol L−1 day−1 was observed over 14 days. In fed-batch incubation the AOM rate increased from 1.5 to 2.7 and 3.6 mmol L−1 day−1 when the concentration of aqueous methane was stepwise increased from 5 to 15 mmol L−1 and 45 mmol L−1. A methane partial pressure of 6 MPa and a hydrostatic pressure of 12 MPa in manifold fed-batch incubation in the HP-MI system yielded a sixfold increase in the volumetric AOM rate. Over subsequent incubation periods AOM rates increased from 0.6 to 1.2 mmol L−1 day−1 within 26 days of incubation. No inhibition of biomass activity was observed in all continuous and fed-batch incubation experiments. The organisms were able to tolerate high sulfide concentrations and extended starvation periods.

    Description: In this overview, we consider the rationale and practice of invertebrate introductions into Lithuanian waters, the current distribution of introduced species, test the theoretical expectations of introduction of peracaridan species into lakes, and summarize the large-scale consequences of these translocations. After some early attempts to transfer locally available species within the country, peracaridan species from the Ponto-Caspian region were introduced into Lithuanian waters during the 1960s (Pontogammarus robustoides, Obesogammarus crassus, Chaetogammarus warpachowskyi, Paramysis lacustris, Limnomysis benedeni and Hemimysis anomala). After their establishment at the site of first introduction in a newly constructed water reservoir on the Nemunas River, further deliberate introductions into Lithuanian lakes and water reservoirs, as well as into more distant areas, were undertaken. These introduced species soon contributed to fish diet and a subsequent increase in the production of commercially important fish was envisaged. However, our collation of available data does not support the enhancement of fish production in Lithuanian lakes. Although perch (selected as a model fish species) assimilated the introduced species into diet and sometimes in large quantities, there was no subsequent influence on somatic growth rates when compared with perch from lakes devoid of alien peracaridans. Comparisons of littoral fish communities of different lakes, and commercial catches in the lake with the most numerous populations of introduced species also indicate no significant effect on fish production. Negative consequences of introduction are more than obvious in contrast. To date, most of the introduced Ponto-Caspian amphipod and mysid species occur in different sites of the Baltic Sea basin outside Lithuania, and their further dispersal may be expected. These species, especially amphipods, have proved to be highly ecologically aggressive, inducing change in resident macroinvertebrate communities and causing increase of biological contamination (i.e. detract from naturalness). Thus, the introduction of alien fodder species was ill advised from an economic, and even adverse from an environmental perspective.

    Description: Whole genome amplification (WGA) approaches provide genomic information on single microbial cells and hold great promise for the field of environmental microbiology. Here, the microbial consortia of the marine sponge Aplysina aerophoba were sorted by fluorescence-activated cell sorting (FACS) and then subjected to WGA. A cosmid library was constructed from the WGA product of a sample containing two bacterial cells, one a member of the candidate phylum Poribacteria and one of a sponge-specific clade of Chloroflexi. Library screening led to the genomic characterization of three cosmid clones, encoding a polyketide synthase (PKS), a non-ribosomal peptide synthetase (NRPS) and the Chloroflexi 16S rRNA gene. PCR screening of WGA products from additional, FACS-sorted single bacterial symbiont cells supports the assignment of the Sup-PKS gene to the Poribacteria and the novel NRPS gene to the Chloroflexi. This promising single-cell genomics approach has permitted cloning of entire gene clusters from single microbial cells of known phylogenetic origin and thus provides a sought-after link between phylogeny and function.

    Description: A brief description of the Deterministic Modelling Hydrological System (DMHS) ‘Runoff–Erosion–Pollution’ proposed by the first author is presented. This system is being developed with the aim of giving it a universal character so that it can be applied in mountainous and flat terrain, and in basins of different natural climatic zones regardless of their size. The main feature of the model is its independence of the scaling problem. The basis of our approach consists of a simple theory of runoff elements. This is different from the generally accepted use of partial differential equations such as the Saint Venant equation for surface and channel flow and the Boussinesq equation for underground waters describing the water movement from runoff formation origins to the basin outlet. The results of runoff simulation for six mountainous watersheds of different sizes across Eastern Siberia within the Lena River basin and their statistical evaluation are presented. The selected river basins ranged in size from about 40 km2 (small scale) to the entire Lena River basin (2·4 million km2), classified as a large-scale basin.

    Description: Many marine sponges contain massive numbers of largely uncultivated, phylogenetically diverse bacteria that seem to be important contributors to the chemistry of these animals. Insights into the diversity, origin, distribution, and function of their metabolic gene communities are crucial to dissect the chemical ecology and biotechnological potential of sponge symbionts. This study reveals a sharp dichotomy between high and low microbial abundance sponges with respect to polyketide synthase (PKS) gene content, the presence of methyl-branched fatty acids, and the presence of members of the symbiotic candidate phylum "Poribacteria". For the symbiont-rich sponge Cacospongia mycofijiensis, a source of the tubulin-inhibiting fijianolides (=laulimalides), near-exhaustive large-scale sequencing of PKS gene-derived PCR amplicons was conducted. Although these amplicons exhibit high diversity at the sequence level, almost all of them belong to a single, architecturally unique group of PKSs present in "Poribacteria" and are proposed to synthesize methyl-branched fatty acids. Three components of this PKS were studied in vitro, providing initial insight into its biochemistry.

    Description: The effects of different phases of semidiurnal and spring-neap tidal cycles on phytoplankton and environmental drivers were evaluated in a tidal, freshwater location of a mesotidal estuary (Guadiana estuary, SW Iberia). An Eulerian approach was used and sampling covered different seasons during 2008. Samples were collected during spring and neap tides, at high tide, mid-ebb, low tide and mid-flood. Several physical-chemical variables were measured, as well as phytoplankton abundance and biomass. Salinity was higher at high-tide and suspended particulate matter was higher during spring tides and flood, due to higher vertical mixing and resuspension of bottom sediments. Chlorophyll a concentration during winter and summer neap tides was higher than during spring tides, whilst the abundance of pennate diatoms was higher during winter and Spring spring tides than during neap tides, probably reflecting differences in river discharge. Overall, tidally-induced differences detected in the freshwater tidal reaches of the Guadiana estuary were not as considerable as those observed in the lower estuary. However, the occurrence of tidally-induced variability in some seasons reflects that thorough sampling programs to study estuarine tidal dynamics should be conducted throughout the year.     Description: The Baltic Sea is characterised by a heterogeneous oceanographic environment. The deep water layers forming the habitat of Baltic cod (Gadus morhua callarias L.) are subjected to frequently occurring pronounced anoxic conditions. Adverse oxygen conditions result in physiological stress for organisms living under these conditions. For cod e.g. a direct relationship between oxygen availability and food intake with a decreasing ingestion rate at hypoxia could be revealed. In the present study, the effects of oxygen deficiency on consumption rates were investigated and how these translate to stock size estimates in multi-species models. Based on results from laboratory experiments, a model was fitted to evacuation rates at different oxygen levels and integrated into the existing consumption model for Baltic cod. Individual mean oxygen corrected consumption rates were 0.1–10.9% lower than the uncorrected ones. At the currently low predator stock size, however, the effect of oxygen-reduced consumption on the total amount of eaten prey biomass and thus predation mortalities was only marginal. But should successful management lead to higher cod stock sizes in the future, then total predation mortalities will greatly increase and thus improved precision of these estimates would be valuable for the assessment of prey stocks.

    Description: Parasitism is a common form of life and represents a strong selective pressure for host organisms. In response to this evolutionary pressure, vertebrates have developed genetically coded defences such as the major histocompatibility complex (MHC). Mechanisms of parasite-mediated selection not only maintain outstanding polymorphism in these genes but have also been proposed to further promote host population divergence and ultimately speciation because it can drive evolution of local adaptation in which MHC genes play a crucial role. This review first highlights the dynamics and complexity of parasite-mediated selection in natural systems, which not only depends on dominating parasite strategies and on the taxonomic diversity of the parasite community but also includes the differences in parasite communities between habitats and niches, creating divergent selection on locally adapted populations. Then the different ways in which MHC genes potentially allow vertebrates to respond to these dynamics and to adapt locally are outlined. Finally, it is proposed that varying selection strength in time and space may lead to variation in the strength of precopulatory reproductive isolation which has evolved to maintain local adaptation.

    Description: Summer heat waves have already resulted in mortality of coastal communities, including ecologically important seagrass meadows. Gene expression studies from controlled experiments can provide important insight as to how species/genotypes react to extreme events that will increase under global warming. In a common stress garden, we exposed three populations of eelgrass, Zostera marina, to extreme sea surface temperatures, simulating the 2003-European heat wave. Populations came from locations widely differing in their thermal regime, two northern European locations [Ebeltoft (Kattegat), Doverodde (Limfjord, Baltic Sea)], and one southern population from Gabicce Mare (Adriatic Sea), allowing to test for population specificity in the response to a realistic heat stress event. Eelgrass survival and growth as well as the expression of 12 stress associated candidate genes were assessed during and after the heat wave. Contrary to expectations, all populations suffered equally from 3 weeks of heat stress in terms of shoot loss. In contrast, populations markedly differed in multivariate measures of gene expression. While the gene expression profiles converged to pre-stress values directly after the heat wave, stress correlated genes were upregulated again 4 weeks later, in line with the observed delay in shoot loss. Target genes had to be selected based on functional knowledge in terrestrial plants, nevertheless, 10/12 genes were induced relative to the control treatment at least once during the heat wave in the fully marine plant Z. marina. This study underlines the importance of realistic stress and recovery scenarios in studying the impact of predicted climate change.

    Description: We examined the influence of forecasted changes in global temperatures and pCO2 on N2 fixation and assimilation in the ecologically important cyanobacterium Trichodesmium spp. Changes of mRNA transcripts (nifH, glnA, hetR, psbA, psaB), protein (nitrogenase, glutamine synthetase) pools and enzymatic activity (nitrogenase) were measured under varying pCO2 and temperatures. High pCO2 shifted transcript patterns of all genes, resulting in a more synchronized diel expression. Under the same conditions, we did not observe any significant changes in the protein pools or in total cellular allocations of carbon and nitrogen (i.e. C : N ratio remained stable). Independently of temperature, high pCO2 (900 µatm) elevated N2 fixation rates. Levels of the key enzymes, nitrogenase and glutamine synthetase that mediate nitrogen assimilation did not increase, implying that the high pCO2 allowed higher reaction turnover rates through these key enzymes. Moreover, increased temperatures and high pCO2 resulted in higher C : P ratios. The plasticity in phosphorous stoichiometry combined with higher enzymatic efficiencies lead to higher growth rates. In cyanobacteria photosynthesis, carbon uptake, respiration, N2 fixation and nitrogen assimilation share cellular components. We propose that shifted cellular resource and energy allocation among those components will enable Trichodesmium grown at elevated temperatures and pCO2 to extend its niche in the future ocean, through both tolerance of a broader temperature range and higher P plasticity.

    Description: Plankton sampling was conducted in the Baltic to obtain sprat larvae. Their individual drift patterns were back-calculated using a hydrodynamic model. The modelled positions along the individual drift trajectories were subsequently used to provide insight into the environmental conditions experienced by the larvae. Autocorrelation analysis revealed that successive otolith increment widths of individual larvae were not independent. Otolith increment width was then modelled using two different generalized additive model (GAM) analyses (with and without autocorrelation), using environmental variables determined for each modelled individual larval position as explanatory variables. The results indicate that otolith growth was not only influenced by the density of potential prey but was controlled by a number of simultaneously acting environmental factors. The final model, not considering autocorrelation, explained more than 80% of the variance of otolith growth, with larval age as a factor variable showing the strongest significant impact on otolith growth. Otolith growth was further explained by statistically significant ambient environmental factors such as temperature, bottom depth, prey density and turbulence. The GAM analysis, taking autocorrelation into account, explained almost 98% of the variability, with the previous otolith increment showing the strongest significant effect. Larval age as well as ambient temperature and prey abundance also had a significant effect. An alternative approach applied individual-based model (IBM) simulations on larval drift, feeding, growth and survival starting as exogenously feeding larvae at the backcalculated positions. The IBM results revealed optimal growth conditions for more than 97% of the larvae, with a tendency for our IBM to slightly overestimate larval growth.

    Description: We examined the diel variation in nitrogen and carbon metabolism in Crocosphaera watsonii WH8501 at the physiological and gene expression level in order to determine the temporal constraints for N2 fixation and photosynthesis. N2 fixation and photosynthesis were restricted to the dark and light periods, respectively, during a 24 h light–dark cycle. All genes studied here except one (psbA2) showed diel variations in their expression levels. The highest variation was seen in nifH and nifX relative transcript abundance with a factor of 3–5 × 103 between light and dark periods. Photosynthesis genes showed less variation with a maximum factor of about 500 and always had high relative transcript abundances relative to other genes. At the protein level, the photosystems appeared more stable than the nitrogenase complex over a 24 h light–dark cycle, suggesting that C. watsonii retains the ability to photosynthesize during the dark period of the diel cycle. In contrast, nitrogenase is synthesized daily and exhibits peak abundance during the dark period. Our results have implications for field studies with respect to the interpretation of environmental gene expression data.

    Description: Global change drivers create new environmental scenarios and selective pressures, affecting plant species in various interacting ways. Plants respond with changes in phenology, physiology, and reproduction, with consequences for biotic interactions and community composition. We review information on phenotypic plasticity, a primary means by which plants cope with global change scenarios, recommending promising approaches for investigating the evolution of plasticity and describing constraints to its evolution. We discuss the important but largely ignored role of phenotypic plasticity in range shifts and review the extensive literature on invasive species as models of evolutionary change in novel environments. Plasticity can play a role both in the short-term response of plant populations to global change as well as in their long-term fate through the maintenance of genetic variation. In new environmental conditions, plasticity of certain functional traits may be beneficial (i.e., the plastic response is accompanied by a fitness advantage) and thus selected for. Plasticity can also be relevant in the establishment and persistence of plants in novel environments that are crucial for populations at the colonizing edge in range shifts induced by climate change. Experimental studies show taxonomically widespread plastic responses to global change drivers in many functional traits, though there is a lack of empirical support for many theoretical models on the evolution of phenotypic plasticity. Future studies should assess the adaptive value and evolutionary potential of plasticity under complex, realistic global change scenarios. Promising tools include resurrection protocols and artificial selection experiments.

    Description: Gracilaria vermiculophylla (Ohmi) Papenf., an agar-producing red alga introduced from northeast Asia to Europe and North America, is often highly abundant in invaded areas. To assay its genetic diversity and identify the putative source of invasive populations, we analyzed the mitochondrial cytochrome c oxidase subunit I (cox1) gene from 312 individuals of G. vermiculophylla collected in 37 native and 32 introduced locations. A total of 19 haplotypes were detected: 17 in northeast Asia and three in Europe and eastern and western North America, with only one shared among all regions. The shared haplotype was present in all introduced populations and in ∼99% of individuals in the introduced areas. This haplotype was also found at three native locations in east Korea, west Japan, and eastern Russia. Both haplotype and nucleotide diversities were extremely low in Europe and North America compared to northeast Asia. Our study indicates that the East Sea/Sea of Japan is a likely donor region of the invasive populations of G. vermiculophylla in the east and west Atlantic and the east Pacific.

    Description: Combined phylogenetic, physiological, and biochemical approaches revealed that differences in defense-related responses among 17 species belonging to the Gracilariaceae were consistent with their evolutionary history. An oxidative burst response resulting from activation of NADPH oxidase was always observed in two of the subgenera of Gracilaria sensu lato (Gracilaria, Hydropuntia), but not in Gracilariopsis and in species related to Gracilaria chilensis (“chilensis” clade). On the other hand, all species examined except Gracilaria tenuistipitata var. liui and Gracilariopsis longissima responded with up-regulation of agar oligosaccharide oxidase to an challenge with agar oligosaccharides. As indicated by pharmacological experiments conducted with Gracilaria chilensis and Gracilaria sp. “dura,” the up-regulation of agar oligosaccharide oxidase involved an NAD(P)H-dependent signaling pathway, but not kinase activity. By contrast, the activation of NADPH oxidase requires protein phosphorylation. Both responses are therefore independent, and the agar oligosaccharide-activated oxidative burst evolved after the capacity to oxidize agar oligosaccharide, probably providing additional defensive capacity to the most recently differentiated clades of Gracilariaceae. As demonstrated with Gracilaria gracilis, Gracilaria dura, and Gracilariopsis longissima, the different responses to agar oligosaccharides allow for a fast and nondestructive distinction among different clades of gracilarioids that are morphologically convergent. Based upon sequences of the chloroplast-encoded rbcL gene, this study suggests that at least some of the samples from NW America recorded as Gs. lemanaeiformis are probably Gs. chorda. Moreover, previous records of Gracilaria conferta from Israel are shown to be based upon misidentification of Gracilaria sp. “dura,” a species that belongs to the Hydropuntia subgenus.

    Description: Two new brominated compounds, subereaphenol K (2) and 2-(3,5-dibromo-1-ethoxy-4-oxocyclohexa-2,5-dien-1-yl)acetamide (3), together with subereaphenol B (methyl 2-(2,4-dibromo-3,6-dihydroxyphenyl)acetate; 1) with a revised structure, and five dibromotyrosine-derived metabolites, 4–8, were isolated from the sponge Suberea sp. and characterized by 1D- and 2D-NMR spectroscopic and HR-MS spectrometric data. Compounds 1, 2, 6, and 8 exhibited various weak or moderate bioactivities, including antimicrobial and cytotoxic activities. Furthermore, compounds 1 and 2 inhibited human recombinant phosphodiesterase 4 (PDE4) with IC50 values of 2 μM, whereas compounds 6 and 8 were less active.

    Description: We present results of a seismic refraction experiment which determines the crustal and upper-mantle structure of an oceanic core complex (OCC) and its conjugate side located south of the 5°S ridge–transform intersection at the Mid-Atlantic Ridge. The core complex with a corrugated surface has been split by a change in location of active seafloor spreading, resulting in two massifs on either side of the current spreading axis. We applied a joint tomographic inversion of wide-angle reflected and refracted phases for five intersecting seismic profiles. The obtained velocity models are used to constrain the magmatic evolution of the core complex from the analysis of seismic layer 3 and crustal thickness. An abrupt increase of crustal velocities at shallow depth coincides with the onset of the seafloor corrugations at the exposed footwall. The observed velocity structure is consistent with the presence of gabbros directly beneath the corrugated fault surface. The thickness of the high-velocity body is constrained by PmP reflections to vary along and across axis between 〈3 and 5 km. The thickest crust is associated with the central phase of detachment faulting at the higher-elevated northern portion of the massif. Beneath the breakaway of the OCC the crust is 2.5 km thick and reveals significantly lower velocities. This implies that the fault initially exhumed low-velocity material overlying the gabbro plutons. In contrast, crust formed at the conjugate side during OCC formation is characterized by an up to 2-km-thick seismic layer 2 overlying a 1.7-km-thick seismic layer 3. Obtained upper-mantle velocities range from 7.3 to 7.9 km s−1 and seem to increase with distance from the median valley. However, velocities of 7.3–7.5 km s−1 beneath the older portions of the OCC may derive from deep fluid circulation and related hydrothermal alteration, which may likely be facilitated by the subsequent rifting. Our velocity models reveal a strongly asymmetric velocity structure across the ridge axis, associated with the accretion of gabbros into the footwall of the detachment fault and upper-crustal portions concentrated at the conjugate side. Our results do not support a substantial increase in the axial ridge's melt supply related to the final phase of detachment faulting. Hence, the footwall rifting at 5°S may be a generic mechanism of detachment termination under very low melt conditions, as predicted by recent numerical models of Tucholke et al.

    Description: Sex differences in behavior, morphology, and physiology are common in animals. In many bird species, differences in the feather colors of the sexes are apparent when judged by human observers and using physical measures of plumage reflectance, cryptic (to human) plumage dichromatism has also been detected in several additional avian lineages. However, it remains to be confirmed in almost all species whether sexual dichromatism is perceivable by individuals of the studied species. This latter step is essential because it allows the evaluation of alternative hypotheses regarding the signaling and communication functions of plumage variation. We applied perceptual modeling of the avian visual system for the first time to an endemic New Zealand bird to provide evidence of subtle but consistent sexual dichromatism in the whitehead, Mohoua albicilla. Molecular sexing techniques were also used in this species to confirm the extent of the sexual size dimorphism in plumage and body mass. Despite the small sample sizes, we now validate previous reports based on human perception that in male whiteheads head and chest feathers are physically brighter than in females. We further suggest that the extent of sexual plumage dichromatism is pronounced and can be perceived by these birds.     Description: The coccolithophore Emiliania huxleyi (Lohmann) W. W. Hay et H. Mohler was cultured in natural seawater with the addition of either the microtubule-inhibitor colchicine, the actin-inhibitor cytochalasin B, or the photosynthesis inhibitor 3-(3,4 dichlorophenyl)-1,1-dimethyl-urea (DCMU). Additionally, E. huxleyi was cultured at different light intensities and temperatures. Growth rate was monitored, and coccolith morphology analyzed. While every treatment affected growth rate, the percentage of malformed coccoliths increased with colchicine, cytochalasin B, and at higher than optimal temperature. These results represent the first experimental evidence for the role of microtubules and actin microfilaments in coccolith morphogenesis.

    Description: The availability and composition of dissolved nitrogen in ocean waters are factors that influence species composition in natural phytoplankton communities. The same factors affect the ratio of organic to inorganic carbon incorporation in calcifying species, such as the coccolithophore Emiliania huxleyi (Lohman) W. W. Hay et H. Mohler. E. huxleyi has been shown to thrive on various nitrogen sources, including dissolved organic nitrogen. Nevertheless, assimilation of dissolved nitrogen under nitrogen-replete and -limited conditions is not well understood in this ecologically important species. In this study, the complete amino acid sequences for three functional genes involved in nitrogen metabolism in E. huxleyi were identified: a putative formamidase, a glutamine synthetase (GSII family), and assimilatory nitrate reductase. Expression patterns of the three enzymes in cells grown on inorganic as well as organic nitrogen sources indicated reduced expression levels of nitrate reductase when cells were grown on NH(4)+ and a reduced expression level of the putative formamidase when growth was on NO(3)-. The data reported here suggest the presence of a nitrogen preference hierarchy in E. huxleyi. In addition, the gene encoding for a phosphate repressible phosphate permease was more highly expressed in cells growing on formamide than in cells growing on inorganic nitrogen sources. This finding suggests a coupling between phosphate and nitrogen metabolism, which might give this species a competitive advantage in nutrient-depleted environments. The potential of using expression of genes investigated here as indicators of specific nitrogen-metabolism strategies of E. huxleyi in natural populations of phytoplankton is discussed.

    Description: Great subduction earthquakes exhibit segmentation both within the rupture of individual events and in the long term history of the margin. The 2004 December 26 Aceh-Andaman and 2005 March 28 Nias event in northern Sumatra are two of the largest earthquakes in recent years, with both co- and post-seismic displacements constrained in unprecedented detail. Using aftershock locations from a temporary seismic array in the boundary region between both events and waveform modelling of large aftershocks, we demonstrate that the vast majority of aftershocks in the study region occur on the plate interface within a narrow band ( 20 km) seaward of the outer arc high. Comparing the seismicity distribution to the co- and post-seismic displacements, we infer that the seismic band marks the transition between the seismogenic zone and stable sliding. The location of the band and therefore the transition appears to be correlated with the ∼500 m bathymetry contour. This close correspondence is disrupted at the boundary between the two great earthquakes, where the transition to seismogenic behaviour occurs further landward by ∼25 km. To the west of Simeulue, where seafloor bathymetry throughout the forearc is deeper than 500 m, the seismic band terminates abruptly and the focus of aftershock activity is found near the trench. The seismic efficiency of afterslip varies dramatically along strike: the segment below the Banyak islands, in the gap between the two main asperities of the Nias earthquake, accommodates a much larger proportion of afterslip seismically.

    Description: The structure and seismicity of the subduction zone of centralCosta Rica have been investigated with local earthquake tomography down to ca. 50 km depth. Seismic traveltime data sets of three on- and offshore seismic networks were combined for a simultaneous inversion of hypocentre locations, 3-D structure of P-wave velocity and Vp/Vs ratio using about 2000 highquality events. The seismicity and slab geometry as well as Vp and Vp/Vs show significant lateral variation along the subduction zone corresponding to the changes of the incoming plate which consists of serpentinized oceanic lithosphere in the northwest, a seamount province in the centre and the subducting Cocos Ridge in the southeast of the investigation area. Three prominent features can be identified in the Vp and Vp/Vs tomograms: a high-velocity zone with a perturbation of 4–10 per cent representing the subducting slab, a low-velocity zone (10–20 per cent) in the forearc crust probably caused by deformation, fluid release and hydration and a low-velocity zone below the volcanic arc related to upwelling fluids and magma. Unlike previously suggested, the dip of the subducting slab does not decrease to the south. Instead, an average steepening of the plate interface from 30◦ to 45◦ is observed from north to south and a transition from a plane to a step-shaped plate interface. This is connected with a change in the deformation style of the overriding plate where roughly planar, partly conjugated, clusters of seismicity of regionally varying dip are observed. It can be shown that the central Costa Rica Deformation Belt represents a deep crustal transition zone extending from the surface down to 40 km depth. This transition zone indicates the lateral termination of the active part of the volcanic chain and seems to be related to the changing structure of the incoming plate as well.

    Description: Marine invasions are taking place at an increasing rate. When occurring in blooms, zooplanktivorous comb jellies of the genus Mnemiopsis are able to cause pelagic regime shifts in coastal areas and may cause the collapse of commercially important fish populations. Using microsatellites, developed for the first time in the phylum Ctenophora, we show that Mnemiopsis leidyi has colonized Eurasia from two source regions. Our preliminary data set included four sites within the putative source region (US East Coast and Gulf of Mexico) and 10 invaded locations in Eurasian waters. Bayesian clustering and phylogeographic approaches revealed the origin of earlier invasions of the Black and Caspian Sea in the 1980s/1990s within or close to the Gulf of Mexico, while the 2006 invasion of the North and Baltic Seas can be directly traced to New England (pairwise FST = 0). We found no evidence for mixing among both gene pools in the invaded areas. While the genetic diversity (allelic richness) remained similar in the Baltic Sea compared to the source region New England, it was reduced in the North Sea, supporting the view of an initial invasion of Northern Europe to a Baltic Sea port. In Black and Caspian Sea samples, we found a gradual decline in allelic richness compared to the Gulf of Mexico region, supporting a stepping-stone model of colonization with two sequential genetic founder events. Our data also suggest that current practices of ballast water treatment are insufficient to prevent repeated invasions of gelatinous zooplankton.

    Description: Increased importance of genetic drift and selection for stress resistance have been predicted to lead to a reduction in the degree of phenotypic plasticity in populations at margins of a species' geographical range, relative to those in the centre. We examined the effect of population positioning within the species range on degree of active morphological plasticity to vegetation shade. Importantly, we discriminated between active, size-independent morphological adjustments in response to shade and passive changes in morphology caused by the dependence of morphological traits on plant size, as only the former are considered to be adaptive. Two closely related and ecologically similar Agrimonia species were examined in the same geographical location, where one species reaches the edge of its distribution (Agrimonia pilosa) and the other does not (A. eupatoria). Plasticity to light availability is likely to be advantageous for both species as they occupy habitats with variable light conditions. However, we hypothesised that high levels of environmental stress should lead to reduced active plasticity in marginal compared with more central populations. Agrimonia eupatoria exhibited active adjustments in leaf morphology in response to tree shade, and in elongation of stems and inflorescences in response to herbaceous shade. In contrast, A. pilosa exhibited very limited active plasticity. High active plasticity allowed A. eupatoria to retain constant shoot growth in a wide range of light conditions, while the lack of active plasticity in A. pilosa resulted in a strong dependence of shoot growth on light availability. We propose that high levels of environmental stress in marginal areas of a species' range may lead to a significant reduction in the degree of active plasticity. Our results clearly indicate that discrimination between active and passive plasticity is crucial for reaching valid conclusions about differences in adaptive plasticity between marginal and non-marginal populations.

    Description: Robust, scalable simultaneous localization and mapping (SLAM) algorithms support the successful deployment of robots in real-world applications. In many cases these platforms deliver vast amounts of sensor data from large-scale, unstructured environments. These data may be difficult to interpret by end users without further processing and suitable visualization tools. We present a robust, automated system for large-scale three-dimensional (3D) reconstruction and visualization that takes stereo imagery from an autonomous underwater vehicle (AUV) and SLAM-based vehicle poses to deliver detailed 3D models of the seafloor in the form of textured polygonal meshes. Our system must cope with thousands of images, lighting conditions that create visual seams when texturing, and possible inconsistencies between stereo meshes arising from errors in calibration, triangulation, and navigation. Our approach breaks down the problem into manageable stages by first estimating local structure and then combining these estimates to recover a composite georeferenced structure using SLAM-based vehicle pose estimates. A texture-mapped surface at multiple scales is then generated that is interactively presented to the user through a visualization engine. We adapt established solutions when possible, with an emphasis on quickly delivering approximate yet visually consistent reconstructions on standard computing hardware. This allows scientists on a research cruise to use our system to design follow-up deployments of the AUV and complementary instruments. To date, this system has been tested on several research cruises in Australian waters and has been used to reliably generate and visualize reconstructions for more than 60 dives covering diverse habitats and representing hundreds of linear kilometers of survey.

    Description: To improve our mechanistic understanding and predictive capacities with respect to climate change effects on the spring phytoplankton bloom in temperate marine systems, we used a process‐driven dynamical model to disentangle the impact of potentially relevant factors which are often correlated in the field. The model was based on comprehensive indoor mesocosm experiments run at four temperature and three light regimes. It was driven by time‐series of water temperature and irradiance, considered edible and less edible phytoplankton separately, and accounted for density‐dependent grazing losses. It successfully reproduced the observed dynamics of well edible phytoplankton in the different temperature and light treatments. Four major factors influenced spring phytoplankton dynamics: temperature, light (cloudiness), grazing, and the success of overwintering phyto‐ and zooplankton providing the starting biomasses for spring growth. Our study predicts that increasing cloudiness as anticipated for warmer winters for the Baltic Sea region will retard phytoplankton net growth and reduce peak heights. Light had a strong direct effect in contrast to temperature. However, edible phytoplankton was indirectly strongly temperature‐sensitive via grazing which was already important in early spring at moderately high algal biomasses and counter‐intuitively provoked lower and later algal peaks at higher temperatures. Initial phyto‐ and zooplankton composition and biomass also had a strong effect on spring algal dynamics indicating a memory effect via the broadly under‐sampled overwintering plankton community. Unexpectedly, increased initial phytoplankton biomass did not necessarily lead to earlier or higher spring blooms since the effect was counteracted by subsequently enhanced grazing. Increasing temperature will likely exhibit complex indirect effects via changes in overwintering phytoplankton and grazer biomasses and current grazing pressure. Additionally, effects on the phytoplankton composition due to the species‐specific susceptibility to grazing are expected. Hence, we need to consider not only direct but also indirect effects, e.g. biotic interactions, when addressing climate change impacts.

    Description: In the continuation of our research on seaweeds, crude extracts of 21 brown algae collected from the south coast of England and the west coast of Ireland were screened for in vitro trypanocidal, leishmanicidal and antimycobacterial activities. Mammalian stages of a small set of parasitic protozoa; i.e. Trypanosoma brucei rhodesiense, T. cruzi and Leishmania donovani, and the tubercle bacillus Mycobacterium tuberculosis were used as test organisms. The extracts were also evaluated for selectivity by testing on a mammalian cell line (L6 cells). Only four extracts were moderately active against T. cruzi, whereas all algal extracts showed significant activity against T. brucei rhodesiense, with Halidrys siliquosa and Bifurcaria bifurcata (Sargassaceae) being the most potent (IC50 values 1.2 and 1.9 μg/mL). All algal extracts also displayed leishmanicidal activity, with H. siliquosa and B. bifurcata again being the most active (IC50s 6.4 and 8.6 μg/mL). When tested against M. tuberculosis, only the B. bifurcata extract was found to have some antitubercular potential (MIC value 64.0 μg/mL). Only three seaweed extracts, i.e. H. siliquosa, B. bifurcata and Cystoseira tamariscifolia showed some cytotoxicity. To our knowledge, this is the first study on the antiprotozoal and antimycobacterial activity of brown algae from British and Irish waters

    Description: In the continuation of our search for natural sources for antiprotozoal and antitubercular molecules, we have screened the crude extracts of four green marine algae (Cladophora rupestris, Codium fragile ssp. tomentosoides, Ulva intestinalis and Ulva lactuca) collected from the Dorset area of England. Trypanosoma brucei rhodesiense, Trypanosoma cruzi, Leishmania donovani and Mycobacterium tuberculosis were used as test organisms in the in vitro assays. The selective toxicity of the extracts was also determined toward mammalian skeletal myoblast (L6) cells. The crude seaweed extracts had no activity against M. tuberculosis, but showed antiprotozoal activity against at least two protozoan species. All algal extracts were active against T. brucei rhodesiense, with C. rupestris being the most potent one (IC50 value 3.7 μg/ml), whilst only C. rupestris and U. lactuca had moderate trypanocidal activity against T. cruzi (IC50 values 80.8 and 34.9 μg/ml). Again, all four extracts showed leishmanicidal activity with IC50 values ranging between 12.0 and 20.2 μg/ml. None of the extracts showed cytotoxicity toward L6 cells, indicating that their antiprotozoal activity is specific.     Description: As part of our continuing research on seaweeds, we have screened the crude extracts of 23 red marine algae collected from England and Ireland. The clinically important blood-stage life forms of Trypanosoma brucei rhodesiense, T. cruzi, Leishmania donovani and Mycobacterium tuberculosis were used as test organisms in the in vitro assays. The selectivity of the extracts was determined by using mammalian skeletal myoblast (L6) cells. All algal extracts showed activity against T. brucei rhodesiense, with Corallina officinalis and Ceramium virgatum being the most potent (IC50 values 4.8 and 5.4 μg/ml), whilst none of the algal extracts inhibited the growth of T. cruzi. Except for Porphyra leucosticta, extracts from all seaweeds also showed leishmanicidal activity with IC50 values ranging from 16.5 to 85.6 μg/ml. Only the crude extract of Calliblepharis jubata showed some weak activity against Mycobacterium tuberculosis (MIC value 256 μg/ml), while the others were inactive at this concentration. Corallina officinalis was the only seaweed that displayed some marginal cytotoxicity (IC50 value 88.6 μg/ml), and all remaining extracts were non-toxic towards L6 cells at 90 μg/ml concentration.     Description: 1. Mid-ocean exchange and saltwater flushing were implemented as management practices to reduce the likelihood of new biological invasions in the Laurentian Great Lakes associated with ships’ ballast water and sediments. Despite this, there has been no formal assessment of the efficacy of these procedures. Here, we conduct a comparative analysis of community composition of dormant taxa transported by ballast sediment before and after regulations came into effect in 2006. 2. Ballast sediment samples were collected from 17 ships during the post-regulation interval of 2007 and 2008. Invertebrate eggs were counted, hatched and species identified in the laboratory. Results were compared to similar samples collected from 39 ships between 2000 and 2002, prior to implementation of saltwater flushing regulations. 3. The estimated amount of residual ballast sediment transported by vessels was significantly lower during the post-regulation period, ranging from 〈1 to 45 tonnes per ship, with an average of 5 tonnes. Mean density and number of dormant viable eggs per ship declined 91 and 81%, respectively. 4. Community composition also changed through time, with Rotifera accounting for 78% of taxa transported prior to regulation, whereas Cladocera and Copepoda each accounted for 38% of abundance post-regulation. Although the number of non-indigenous species (NIS) declined 73% per ship after 2006, the reduction was not statistically significant; however, the number of freshwater NIS – which pose the greatest risk of invasion for the Great Lakes – was significantly lowered. 5. Our comparative analysis suggests that ballast management regulations enacted in 2006 markedly reduced the probability of introduction of NIS via dormant eggs carried in ballast sediments.

    Description: Tissue TG (transglutaminase) or TG2 is the most ubiquitously expressed member of the large TG family that catalyses deamidation of a glutamine residue, formation of an N ε(γ‐glutamyl)‐lysine cross‐linking between lysine and glutamine residues and/or covalent incorporation of polyamines into a glutamine residue, exerting a number of physiological and/or pathological functions. Extracellular TG2 contributes to wound healing and exacerbation of liver fibrosis through a role in extracellular matrix assembly and cell adhesion. Intracellular TG2 acts as a GTPase in normal cells when the intracellular Ca2+ concentration is as low as 10–20 nM, participating in the transmembrane signalling of phospholipase Cδ as a component of α1‐adrenergic receptor complexes, and thereby supporting the growth of hepatic cells. When cells are injured and the intracellular Ca2+ concentration rises to more than 700–800 nM, TG2 dramatically alters its structure and transforms into a cross‐linking enzyme. TG2 primarily exists in the cytosol in normal cells, but is distributed among multiple intracellular milieus during tissue injury or apoptosis. In particular, TG2 has been shown to be abundant in the nuclei of cells undergoing apoptosis, although its role in the nucleus and the underlying mechanisms remain unresolved. Recently, three findings in the study of alcoholic steatohepatitis have shed light on these issues. Omary's group disclosed that TG2‐mediated cross‐linking of keratin 8 is essential for the formation of Mallory–Denk bodies. We have demonstrated that in both mouse models of alcoholic steatohepatitis and human patients with alcoholic steatohepatitis, TG2 translocates into the nucleus and provokes hepatocyte death via cross‐linking and inactivation of a transcription factor, Sp1, leading to down‐regulation of the hepatocyte growth factor receptor, c‐Met. Furthermore, Giebeler et al. has reported that down‐regulation of c‐Met is associated with liver fibrosis. In the present review article, we introduce these recent advances in knowledge with regard to the the roles of TG2 in alcoholic steatohepatitis.

    Description: Cephalopods are a key component of marine food webs, providing sustenance for myriad marine species. Cephalopods are also of increasing economic importance as evidenced by the rapid rise in their global landings over recent decades. If fisheries continue on this trajectory, conflicts may transpire among cephalopod and finfish fisheries, particularly in ecosystems where cephalopods are highly valuable both directly as a landed commodity and indirectly as prey for other harvested species. We provide the first measure of the ecosystem services that cephalopods contribute to fisheries in 28 marine ecosystems, both as a commodity and an ecological support service. We also evaluate how current demands on cephalopods compare to mid-20th century conditions. We find that cephalopod contributions to fisheries vary widely, but are substantial in many ecosystems. Commodity and supportive services provided by cephalopods contributed as much as 55% of fishery landings (tonnes) and 70% of landed values ($USD). The contribution of cephalopods as a commodity was generally greatest in the coastal ecosystems, whereas their contribution as a supportive service was highest in open ocean systems. Further, the commodity and supportive services provided by cephalopods to fisheries landings increased in most of the coastal ecosystems between the mid-20th century (years 1960–70) and contemporary periods (years 1990–2004), indicating the rising demand for cephalopods. Current demands have no historical precedent and ecosystems in which cephalopods are highly exploited as a targeted resource and as an ecological support service should be further evaluated to prevent the unsustainable development of marine fisheries within them.

    Description: Objectives Carotenoids and retinoic acid derivatives are topically applied for sun‐protective and whitening purposes. Fucoxanthin is a carotenoid derived from edible sea algae, but its effect on melanogenesis has not been established. Therefore, we examined the effect of fucoxanthin on melanogenesis. Methods Inhibitory effects on tyrosinase activity, melanin formation in B16 melanoma and skin pigmentation in UVB‐irradiated guinea‐pigs were evaluated. To elucidate the action of fucoxanthin on melanogenesis, its effect on skin melanogenic mRNA expression was evaluated in UVB‐irradiated mice. Fucoxanthin was given topically or orally to mice once a day and UVB irradiation was applied for 14 days. The effect of fucoxanthin on skin melanogenic mRNA expression was evaluated by real time reverse transcription polymerase chain reaction. Key findings Fucoxanthin inhibited tyrosinase activity, melanogenesis in melanoma and UVB‐induced skin pigmentation. Topical application of fucoxanthin (1%) significantly suppressed mRNA expression of cyclooxygenase (COX)‐2, endothelin receptor A, p75 neurotrophin receptor (NTR), prostaglandin E receptor 1 (EP1), melanocortin 1 receptor (MC1R) and tyrosinase‐related protein 1. The suppression of p75NTR, EP1 and MC1R expressions was observed at 0.01% application. Also, oral application of fucoxanthin (10 mg/kg) significantly suppressed expression of COX‐2, p75NTR, EP1 and MC1R. Conclusions These results suggest that fucoxanthin exhibits anti‐pigmentary activity by topical or oral application in UVB‐induced melanogenesis. This effect of fucoxanthin may be due to suppression of prostaglandin (PG) E2 synthesis and melanogenic stimulant receptors (neurotrophin, PGE2 and melanocyte stimulating hormone expression).

    Description: A 415 cm thick permafrost peat section from the Verkhoyansk Mountains was radiocarbon-dated and studied using palaeobotanical and sedimentological approaches. Accumulation of organic-rich sediment commenced in a former oxbow lake, detached from a Dyanushka River meander during the Younger Dryas stadial, at ∼12.5 kyr BP. Pollen data indicate that larch trees, shrub alder and dwarf birch were abundant in the vegetation at that time. Local presence of larch during the Younger Dryas is documented by well-preserved and radiocarbon-dated needles and cones. The early Holocene pollen assemblages reveal high percentages of Artemisia pollen, suggesting the presence of steppe-like communities around the site, possibly in response to a relatively warm and dry climate ∼11.4–11.2 kyr BP. Both pollen and plant macrofossil data demonstrate that larch woods were common in the river valley. Remains of charcoal and pollen of Epilobium indicate fire events and mark a hiatus ∼11.0–8.7 kyr BP. Changes in peat properties, C31/C27 alkane ratios and radiocarbon dates suggest that two other hiatuses occurred ∼8.2–6.9 and ∼6.7–0.6 kyr BP. Prior to 0.6 kyr BP, a major fire destroyed the mire surface. The upper 60 cm of the studied section is composed of aeolian sands modified in the uppermost part by the modern soil formation. For the first time, local growth of larch during the Younger Dryas has been verified in the western foreland of the Verkhoyansk Mountains (∼170 km south of the Arctic Circle), thus increasing our understanding of the quick reforestation of northern Eurasia by the early Holocene.
