    Description / Table of Contents: Zusammenfassung Wir führen ein Konzept für Näherungslösungen in der Vektoroptimierung ein und vergleichen dieses mit einem neuen Konzept aus [8]. Weiterhin untersuchen wir Beziehungen zwischen der Menge der Näherungslösungen eines Vektoroptimierungsproblems und den Näherungslösungen eines entsprechenden parametrischen Ersatzproblems. Schließlich beweisen wir Stabilitätseigenschaften des skalaren Ersatzproblems.
    Notes: Abstract We introduce a concept for approximately efficient solutions in vector optimization and compare it with another recent concept given in [8]. Further, we study relations between the set of approximately efficient solutions of a vector optimization problem and the approximate solutions of a corresponding parametric surrogate optimization problem. Finally, we prove stability properties for the scalar surrogate problem.

    Notes: Abstract Mortality of the coffee berry borer was studied under controlled laboratory conditions in Tapachula, Mexico. For adult female borers subjected to a range of relative humidities (RH) without food at 25°C, the longest mean survival time (20 days) was obtained at 93.5% RH. Adult borer survival was also studied at a range of temperatures for a fixed relative humidity (93.5% RH); at 20°C mean survival time was 28 days. Fecundity and mortality of borer stages in berries was studied for a range of humidities at 25°C. Maximum fecundity was obtained at 90 and 93.5% RH. Immature stages were ejected from the berry at 84% RH and above, which is interpreted as a form of brood hygiene.

    Notes: Abstract In short-term field trials at combinations of ambient temperature (°C) and insolation (W·m−2), larval Colorado potato beetles (Leptinotarsa decemlineata [Say] [Coleoptera: Chrysomelidae]) were observed after their release on the adaxial surface of leaflets on potato plants (Solanum tuberosum L. Solanaceae). The larvae either began feeding or moved under the leaflet; mean interval from release to expression of these behaviors (2.9±0.05 min [n =358]) was independent of air temperature and insolation. Proportion of larvae moving under the leaflet increased logistically with both air temperature and insolation. A 1 W·m−2 change in insolation (P) evoked the same effect on this proportion as a 0.0838 °C change in air temperature (T a ), so the two quantities were combined as T*=T a +P·0.0838 °C/(W·m−2), which has units of °C. The proportion of larvae moving under the leaflet increased logistically with T*. In 1-day field trials we monitored air temperature, insolation and proportion of larvae under the leaflet, and compared the latter to predictions from the logistic regression derived from the short-term trials. Consistently more larvae occurred under leaflets than predicted from the logistic regression; this bias diminished as T* increased until at T*≥40 °C, observed and predicted proportions were equal. This pattern of deviation from the predictions of the logistic regression is consistent with a thermoregulatory strategy in which larvae move away from hostile conditions, rather than seek optimal conditions.

    Notes: Abstract Choice and no-choice studies were conducted to determine how the glandular trichomes of the wild potato,Solanum berthaultii Hawkes, affect host preference of the Colorado potato beetle,Leptinotarsa decemlineata (Say). Given a feeding choice betweenS. tuberosum andS. berthaultii, larvae and adults preferred the foliage ofS. tuberosum, but adults were more discriminating. When foliage ofS. berthaultii was appressed toS. tuberosum leaflets, fewer adults fed on the appressed leaflets. When given a choice between ‘trichome-intact’ and ‘trichome-removed’S. berthaultii foliage, adults preferred to feed on the latter. The preference for ‘trichome-removed’ foliage and the percent of adults initiating feeding, increased with the degree of trichome removal. These studies provide evidence that the resistance ofS. berthaultii is associated with feeding deterrents localized in the glandular trichomes, thatS. berthaultii possesses more than one mechanism of resistance to the Colorado potato beetle, and that the expression of resistance is dependent on the developmental stage of the insect.

Il s’avère que les mâles sont plus actifs que les femelles et queS. zeamais possède une meilleure aptitude au vol queS. oryzae. De plus, dans les deux espèces élevées sur blé, seuls les charançons normalement symbiotiques sont capables de voler, tandis que les aposymbiotiques ont perdu ce pouvoir. En complémentant la nourriture avec des vitamines (acide pantothénique et riboflavine) et certains acides aminés (phénylalanine et proline), cette activité a pu être restaurée en partie. L’influence de la symbiose sur l’aptitude au vol du charançon du genreSitophilus est commentée en fonction du métabolisme énergétique (coenzymes, proline) et du rôle joué par la symbiose dans ce comportement qui permet d’augmenter la dissémination de l’espèce.
    Notes: Abstract The flight activity (take-off) of various strains ofSitophilus oryzae L. andS. zeamais Mots. (Coleoptera, Curculionidae) have been compared on wheat. It was shown that males are more active than females and thatS. zeamais, is more active thanS. oryzae. Furthermore, in the two species, when reared on wheat kernels, only symbiotic weevils were able to fly, while aposymbiotic insects did not. By supplementation of the diet with vitamins (pantothenic acid, riboflavin) and amino acids (phenylalanine and proline), this activity could be restored in part. The influence of symbiosis on the flight ability ofSitophilus weevil is discussed in relation to the energy metabolism (coenzymes, proline), the role played by symbiosis in this behaviour and its possible effect on the dissemination of the species.

    Notes: Abstract Overwintering Colorado potato beetles (Leptinotarsa decemlineata (Say)) were concentrated primarily within woody borders, and mortality was lower in borders than in potato fields. After overwintering, only 15–44% of live beetles were in the potato fields. In experiments with small plots, colonization of fields from woody borders was reduced ∼60% by a trap crop, either treated with adulticide or with beetles collected daily. Such trap crops, or simply pitfall traps to prevent colonization from woody borders, could significantly reduce early-season adult numbers and subsequent larval populations. However, success is dependent on the local densities of overwintered beetles, and the prevalence of arrestment behavior in the case of trap crops.

    Notes: Abstract Thirty cohorts of the Colorado potato beetle (CPB) were reared for ten consecutive generations on the cultivated potato,Solanum tuberosum, and on two accessions (PI 473331 and PI 473334) of the resistant wild species,S. berthaultii. At each generation, selection was imposed for increased survival, decreased development time, and for increased fecundity. Although several parameters responded to selection, net replacement rate (R0) onS. berthaultii was consistently less than 1/3 that on cultivated potato. At the tenth generation, eachS. tuberosum strain female replaced herself with 110 daughters, whereas females from strains reared on PI 473334 and PI 473331 produced an average of 30 and 7 daughters, respectively. Slopes of the R0 regression lines for populations reared onS. tuberosum andS. berthaultii did not differ from zero, indicating no significant response to selection. Our findings suggest that populations of the Colorado potato beetle may have genetic variability in some performance traits for adaptation toS. berthaultii, but that adaptation is unlikely to occur as rapidly as previously reported.

    Notes: Abstract FemalePhoturis versicolor fireflies attempt to capture males by responding to heterospecific flash patterns. A mating-dependent switch occurs which affects response timing and frequency of female flashes. We examined the switch using females of known age, mating status, and flash experience to assess how accurate mimicry is, what factors influence it, and what mechanism produces it. Presentations of simulated male flash patterns before and after mating revealed elements of an entrainment mechanism controlling female responsiveness. Unmated females preferentially answered conspecific patterns with variable latencies, averaging 1 s. Mating induced changes in both response frequency and response latency: Females answered heterospecific patterns more frequently, and latencies elicited by conspecific patterns shifted away from the unmated range. Heterogeneity in mean and variance of response latency among individuals indicates that females do not share a discrete reply to a given pattern. Little correspondence exists between latencies of sympatric species andP. versicolor females, suggesting that the flash response mechanism produces entriainment to any rhythmic pattern, not a one-to-one matching between prey and predator latencies. Different selective scenarios underlie strict mimicry versus entrainment mimicry.

    Notes: Abstract The kinetics of the solvolysis of [Co(CN)5Cl]3− have been investigated in water +2-methoxyethanol and water + diethylene glycol mixtures. Although the addition of these linear hydrophilic cosolvent molecules to water produces curvature in the variation of log(rate constant) with the reciprocal of the dielectric constant, their effect on the enthalpy and entropy of activation is minimal, unlike the effect of hydrophobic cosolvents. The application of a Gibbs energy cycle to the solvolysis in water and in the mixtures using either solvent-sorting or TATB values for the Gibbs energy of transfer of the chloride ion between water and the mixture shows that the relative stability of the emergent solvated Co(III) ion in the transition state compared to that of Co(CN)5Cl3− in the initial state increases with increasing content of cosolvent in the mixture. By comparing the effects of other cosolvents on the solvolysis, this differential increase in the relative stabilities of the two species increases with the degree of hydrophobicity of the cosolvent.

    Notes: Abstract Amplitudes of chemical relaxation signals can provide useful information as to the thermodynamics of coupled chemical reactions. The temperature-jump technique has been used to investigate the thermodynamic behavior of the Nickel(II)-3,5-dinitrosalicylate system in buffer solution, where complex formation steps are coupled to proton transfer steps. The analysis of the relaxation curves is based on the transformation of a set of coupled elementary reactions into a set of uncoupled ‘normal reactions.’ By analogy with classical titrations, the experiments have been performed by changing the metal ion concentration at constant ligand concentration and pH. Each measured amplitude is associated in this way to a point of a ‘dynamic titration’ and a procedure is formulated by which the values of the equilibrium constants and enthalpies of the normal reactions are simultaneously obtained by simple linear plots. From the dependence of these parameters on suitable functions of the concentrations of the reactants the values of ΔG° and ΔH° of the individual steps are derived. It is shown that the addition of a buffer (instead of an indicator) influences the stoichiometric coefficients of the normal reaction in such a way that measurable amplitudes are produced in systems that, as the presently investigated, in unbuffered solution would remain insensitive to the external perturbation. The circumstances under which the dynamic method offers advantages over the classical techniques are discussed.

    Notes: Abstract A strain of Saccharomyces cerevisiae has been adapted to increasing concentrations of copper at two different pH values. The growth curve at pH 5.5 is characterized by a time generation increasing with the amount of added copper. A significant decrease of cell volume as compared with the control is also observed. At pH 3 the cells grow faster than at pH 5.5 and resist higher copper concentrations (3.8 against 1.2 mm). Experimental evidence indicates that, after copper treatment, the metal is not bound to the cell wall, but is localized intracellularly. A significant precipitation of copper salts in the medium was observed only at pH 5.5. Increased levels of superoxide dismutase (SOD) activity were observed in copper-treated cells and which persisted after 20 subsequent inocula in a medium without added metal. On the contrary, catalase activity was not stimulated by copper treatment and, hence, not correlated with SOD levels. The mechanism of copper resistance, therefore, probably involves a persistent induction of SOD, but not of catalase, and it is strongly pH-dependent.

    Notes: Abstract A critical review is given of the degradation of cellulose in the low-temperature region (below about 300°C) of power transformer operation. The large number of kinetic studies, under a variety of environmental conditions from Kraft paper in insulating oil, to cotton and paper in oxygen, are considered in terms of a first-order polymer chain scission model. In many cases, the data are replotted to suit the model. A common activation energy of 111±6 kjmol−1 is calculated and it is shown that the pre-exponential factor, rather than the activation energy, is sensitive to the oxidizing nature of the environment and the susceptibility to degradation of the material. The chemical mechanisms of degradation are reviewed, and conclusions and recommendations are made regarding chemical condition monitoring and life prediction of electrical insulation.

    Notes: Abstract Different mechanisms of atomic carbon and oxygen recombination on a rhodium surface are studied with Auger electron spectroscopy (AES) and X-ray photoelectron spectroscopy (XPS). The kinetics of adsorbed carbidic carbon oxidation (carbon coverageθ c ≈ 0.1–0.3 ML) by gas-phase oxygen that proceeds by a Langmuir-Hinshelwood reaction mechanism, provides the value of the activation energy for recombination (E rec ⇂t ≈ 170±20 kJ/mol).E rec ⇂t depends slightly on the carbon coverage. An Eley-Rideal type of reaction was observed for adsorbed oxygen and atomic gas-phase carbon recombination which occurs in a dynamic regime. The low value found for the activation energy (near zero) is consistent with the mechanism that this exothermic reaction is too fast for energy dissipation into the substrate; the energy is mainly transferred into translational, vibrational and rotational energy of CO.

    Notes: Abstract Symmetry properties of positive solutions of a Dirichlet problem for a strongly nonlinear parabolic partial differential equation in a symmetric domainD ⊂ R n are considered. It is assumed that the domainD and the equation are invariant with respect to a group {Q} of transformations ofD. In examples {Q} consists of reflections or rotations. The main result of the paper is the theorem which states that any compact inC(D) negatively invariant set which consists of positive functions consists ofQ-symmetric functions. Examples of negatively invariant sets are (in autonomous case) equilibrium points, omega-limit sets, alpha-limit sets, unstable sets of invariant sets, and global attractors. Application of the main theorem to equilibrium points gives the Gidas-Ni-Nirenberg theorem. Applying the theorem to omega-limit sets, we obtain the asymptotical symmetrization property. That means that a bounded solutionu(t) asr→∞ approaches subspace of symmetric functions. One more result concerns properties of eigenfunctions of linearizations of the equations at positive equilibrium points. It is proved that all unstable eigenfunctions are symmetric.

    Notes: Abstract This paper deals with the stability of periodic polling models with a mixture of service policies. Customers arrive according to independent Poisson processes. The service times and the switchover times are independent with general distributions. The necessary and sufficient condition for the stability of such polling systems is established. The proof is based on the stochastic monotonicity of the state process at the polling instants. The stability of only a subset of the queues is also analyzed and, in case of heavy traffic, the order of explosion of the queues is given. The results are valid for a model with set-up times, and also when there is a local priority rule at the queues.

    Notes: Abstract Consider a queueing system where customers arrive at a circle according to a homogeneous Poisson process. After choosing their positions on the circle, according to a uniform distribution, they wait for a single server who travels on the circle. The server's movement is modelled by a Brownian motion with drift. Whenever the server encounters a customer, he stops and serves this customer. The service times are independent, but arbitrarily distributed. The model generalizes the continuous cyclic polling system (the diffusion coefficient of the Brownian motion is zero in this case) and can be interpreted as a continuous version of a Markov polling system. Using Tweedie's lemma for positive recurrence of Markov chains with general state space, we show that the system is stable if and only if the traffic intensity is less than one. Moreover, we derive a stochastic decomposition result which leads to equilibrium equations for the stationary configuration of customers on the circle. Steady-state performance characteristics are determined, in particular the expected number of customers in the system as seen by a travelling server and at an arbitrary point in time.

    Notes: Abstract The non-linear stability of the libration pointL 4 in the restricted problem has been studied when there are perturbations in the potentials between the bodies. It is seen that the pointL 4 is stable for all mass ratios in the range of linear stability except for three mass ratios depending upon the perturbing functions. The theory is applied to the following four cases: (i) There are no perturbations in the potentials (classical problem). (ii) Only the bigger primary is an oblate spheroid whose axis of symmetry is perpendicular to the plane of relative motion (circular) of the primaries. (iii) Both the primaries are oblate spheroids whose axes of symmetry are perpendicular to the plane of relative motion (circular) of the primaries. (iv) The primaries are spherical in shape and the bigger is a source of radiation.

    Notes: Abstract In this paper, we analyze a simple free boundary model associated with solid combustion and some phase transition processes. There is strong evidence that this “one-phase” model captures all major features of dynamical behavior of more realistic (and complicated) combustion and phase transition models. The principal results concern the dynamical behavior of the model as a bifurcation parameter (which is related to the activation energy in the case of combustion) varies. We prove that the basic uniform front propagation is asymptotically stable against perturbations for the bifurcation parameter above the instability threshold and that a Hopf bifurcation takes place at the threshold value. Results of numerical simulations are presented which confirm that both supercritical and subcritical Hofp bifurcation may occur for physically reasonable nonlinear kinetic functions.

    Notes: Abstract It is known that an abridged case of the averaged planar general three-body problem, at first-order resonance, is analytically integrated, using an expansion of the disturbing function linear in the eccentricities. There exist different methods with the help of which the integration can be performed. For the first time Sessin and Ferraz-Mello in the years 1981–88 (Sessin, 1981, 1983; Ferraz-Mello and Sessin, 1984; Ferraz-Mello, 1987, 1988) did an analytic integration for the restricted elliptic three-body problem, in terms of the variablesK andH (K=ΣD j e j cos (ψ1−π j ),H=ΣD j e j sin (ψ1−π j ),D j = const, wheree j and π j are, respectively, the eccentricity and the longitude of the periapsis of thei-th planet, ψ1 is the Delaunay's anomaly), which is inconvenient for the analytical investigation of the evolution of the major semi-axesa j , the eccentricitiese j and the resonance phases ϕ j =ψ1−π j . Later, a different method for the analytical integration of the general three-body problem, in the variablesa j ,e j and ϕ j , was considered by the author (Shinkin, 1993). A disadvantage of both methods is the fact that they use non-canonical changes of variables. But there exists a third very beautiful canonical method of analytical integration of the general planetary problem, which is briefly considered in the present paper and allows us to describe the bifurcations of separatrices (i.e. appearance, disappearance, splitting and confluence of separatrices) separating the domains of libration and circulation of the resonance phase on the phase plane in the averaged planar general three-body problem at first-order resonance. The bifurcation parameter is analytically found and plays an important role in a qualitative description of all kinds of motion in the examined problem.

    Notes: Abstract The kinetics of a catalytic reaction is frequently formulated in terms of simple concepts of the Langmuir type. Apart from limitations arising from the non-uniformity of the catalyst's surface and from the coverage dependence of the rate “constants”, several other complications may come into play. These may arise on the “quantum level” where energy flow between the various degrees of freedom may cause failure of simple transition state theory, as well as on the “continuum level” where formulation of rate equations in terms of coupled non-linear differential equations may give rise to a rich scenario of spatio-temporal self-organization, including kinetic oscillations, chaos, and formation of concentration patterns. Several of these phenomena are illustrated by selected examples.

    Notes: Abstract We study the structure of chaos in a simple Hamiltonian system that does no have an escape energy. This system has 5 main periodic orbits that are represented on the surface of section $$(y,\dot y)$$ by the points (1)O(0,0), (2)C 1,C 2(±y c, 0), (3)B 1,B 2(O,±1) and (4) the boundary $$y^2 + \dot y^2 = 1$$ . The periodic orbits (1) and (4) have infinite transitions from stability (S) to instability (U) and vice-versa; the transition values of ε are given by simple approximate formulae. At every transitionS →U a set of 4 asymptotic curves is formed atO. For larger ε the size and the oscillations of these curves grow until they destroy the closed invariant curves that surroundO, and they intersect the asymptotic curves of the orbitsC 1,C 2 at infinite heteroclinic points. At every transitionU →S these asymptotic curves are duplicated and they start at two unstable invariant points bifurcating fromO. At the transition itself the asymptotic curves fromO are tangent to each other. The areas of the lobes fromO increase with ε; these lobes increase even afterO becomes stable again. The asymptotic curves of the unstable periodic orbits follow certain rules. Whenever there are heteroclinic points the asymptotic curves of one unstable orbit approach the asymptotic curves of another unstable orbit in a definite way. Finally we study the tangencies and the spirals formed by the asymptotic curves of the orbitsB 1,B 2. We find indications that the number of spiral rotations tends to infinity as ε → ∞. Therefore new tangencies between the asymptotic curves appear for arbitrarily large ε. As a consequence there are infinite new families of stable periodic orbits that appear for arbitrarily large ε.

    Notes: Abstract In this paper we deal with the circular Sitnikov problem as a subsystem of the three-dimensional circular restricted three-body problem. It has a first analytical part where by using elliptic functions we give the analytical expressions for the solutions of the circular Sitnikov problem and for the period function of its family of periodic orbits. We also analyze the qualitative and quantitative behavior of the period function. In the second numerical part, we study the linear stability of the family of periodic orbits of the Sitnikov problem, and of the families of periodic orbits of the three-dimensional circular restricted three-body problem which bifurcate from them; and we follow these bifurcated families until they end in families of periodic orbits of the planar circular restricted three-body problem. We compare our results with the previous ones of other authors on this problem. Finally, the characteristic curves of some bifurcated families obtained for the mass parameter close to 1/2 are also described.

    Notes: Abstract A new approach to the integration of vertex singularities is described. This approach is based on a non-uniform subdivision of the region of integration and the technique fits well to the subdivision strategy used in many adaptive algorithms. A nice feature with this approach is that it can be used in any dimension and on any region of integration which can be subdivided into subregions of the same form. The strategy can be applied both to vertex singularities and internal point singularities. In the latter case this can be done without an initial subdivision of the region in order to put the singular point in a vertex. It turns out that the technique has excellent numerical stability properties.

    Notes: Abstract Earlier studies have shown that sulfur dioxide and metal-support interaction can strongly influence propane oxidation over platinum. In particular, oxidation activity is enhanced when platinum is supported on sulfated γ-alumina or zirconia compared to γ-alumina. Therefore, it is of interest to compare the performance of palladium under the same experimental conditions. Four model catalysts were examined: Pt/γ-alumina, Pt/zirconia, Pd/γ-alumina and Pd/zirconia. The metal loading was kept at or below 0.05 wt% to emphasize changes in activity attributable to metal-support interaction. Reaction rates were measured with and without sulfur dioxide. Surface sulfation was analyzed by measuring acid strength and evaluating spectra obtained by Fourier-transform infrared spectroscopy. In contrast to platinum, sulfation does not promote propane oxidation on Pd/γ-alumina, and Pd/zirconia is less active than Pd/γ-alumina.

    Notes: Abstract We compared the nucleotide substitution pattern over the entire genome of two unique variants of the 6,300-bp selfish DNA (2 μm) plasmid in Saccharomyces cerevisiae. The DNA sequence of the left-unique region is identical among 2-μm variants, while the right-unique region shows substantial divergence. This chimeric pattern cannot be explained by neutral or Darwinian selection models. We propose that horizontal transmission of the 2-μm plasmid coupled with a directed, polarized gene conversion maintains the DNA sequence of the left-unique region, whereas the right-unique region is subject to random drift and Darwinian selection.

Es ist zu erwarten, daß die Stabilitätsregionen mit wachsendem Zeithorizont schrumpfen und daß sie in diesem Sinne monoton sind. In der vorliegenden Arbeit werden verschiedene hinreichende Bedingungen für diese Monotonie untersucht. Die Bedingungen setzen unter anderem die Existenz von Planungs- und Vorhersage-Horizonten voraus und verallgemeinern so Ergebnisse einer früheren Arbeit, in der Aussagen für gewöhnliche Planungs-Horizonte vorgestellt wurden.
    Notes: Abstract The set of cost inputs for which an optimal solution of the dynamic lot size model remains valid is called stability region. The size of this region may be viewed as a measure of robustness of a solution. It is an expectation that the stability regions shrink with growing time horizons and that they are monotonous in this sense. In the present paper several sufficient conditions implying monotonicity will be studied. The conditions cover the existence of planning and forecast horizons and generalize the results of a previous paper in wich monotonicity results were presented for the case of ordinary planning horizons.

    Notes: Abstract 5,10,15,20-tetrakis(3-chloro-4-sulfophenyl)porphine (m-Cl-TPPS4) was synthesized and used for the Spectrophotometric determination of mercury by flow injection analysis. A pseudo-first-order reaction kinetic mechanism was proposed with a rate constant of 0.8 min−1 for Hg(II) withm-Cl-TPPS4 in the presence of 8-hydroxyquinoline in a medium of 1.0M acetic acid and sodium acetate buffer solution (pH 6.22). In the optimum conditions of reaction temperature (85 ° C), stopped-flow time (60 s) and sampling volume (100 μl), the method's relative standard deviation was 0.82% (n = 12) at 5.0 μg ml−1 mercury, with a linear range of 0–12.0 μg ml−1 and an analytical frequency of 60h−1. The detection limit (3σ) was 0.025 μg ml−1. Interference studies showed that most metal ions co-existing with Hg2+ could be tolerated at 100-fold excess levels, but Zn2+, Cu2+ and Mn2+ needed to be masked. The method has been applied to the analysis of water samples with satisfactory results.

The influence of resonant phenomena on the motion of the shortened (linearized) system is considered.

    Notes: Abstract Individual granules within a given granule population of a slow-release fertilizer (SRF) have a different release pattern. The populations studied differed both in relation to the time delay before the start of the release process and to the duration of the release. An association between a short delay period and a rapid release was found. The random reease distribution can be approximated using first-order rate equations. In cases, a term describing a “lag” period should be added. The distribution of release timing among the fertilizer granules may allow a long-lasting nutrient supply to the plant, as long as there are enough granules within the root zone to allow a uniform supply pattern.

    Notes: Summary Acanthoscelides obtectus (Say) is one of the most damaging pests of kidney beans,Phaseolus vulgaris L. worldwide. However, aromatic plants from the families Lamiaceae, Lauraceae, Myrtaceae and Poaceae can protect kidney beans by direct or delayed insecticidal effect, through increased adult mortality and inhibition of beetle reproduction (both oviposition and adult emergence). The efficiency of hydrodistillated and intact plants fromThymus vulgaris andT. serpyllum, Mentha piperata, Rosmarinus officinalis, Satureia hortensis, Eucalyptus globulus, Laurus nobilis, Origanum vulgare, andCymbopogon nardus was compared. For both extracts,Origanum vulgare had the best effect. The insecticidal effect was induced by more than the essential oils because no significant difference was noticed between distilled and intact plants extract. Inhibition of reproduction was particularly important. These results suggest that lipidic as well as non lipidic, allelochemicals, such as phenolics, or non-protein amino-acids, or flavonoids may be involved in the toxicity of aromatic plants to this beetle.

    Notes: Summary The defensive mechanisms which protect ladybird beetles (Coccinellidae) against predators are reviewed. Besides behavioural mechanisms, such as thanatosis and reflex bleeding, chemical defence mechanisms are playing a prevalent role. Indeed, ladybirds are protected not only by their smell, but also by repulsive alkaloids, most of which are considered to be of autogenous origin. In a few cases, dietarily-acquired substances are also involved. Particular emphasis is laid on the repellent alkaloids which are contained in the haemolymph of many species. The structures of 34 nitrogen-containing compounds isolated so far are presented, and their distribution within the family is discussed in the light of the most widely accepted classification of these beetles. To conclude, the mode of release of the alkaloids, their variation through the life cycle and their repellent and toxic properties are discussed, as well as the few biosynthetic data yet available.

    Notes: Summary The exocrine glandular secretions of larvae of the subfamily Chrysomelinae are known to repel conspecific adults, other competitive phytophagous insects and natural enemies. InPhaedon cochleariae, the intraspecific activity of tlc fractions of the larval secretion was tested in order to examine the ecological significance of two fractions containing minor components and a fraction containing the major compound, the cyclopentanoid monoterpene (epi)chrysomelidial. InChrysomela lapponica, the defensive activity of the larval secretion against ants is known from specimens feeding upon willow or birch. The feeding preferences of larvae and adults ofC. lapponica from a Finnish and a Czech population were tested. The Finnish individuals significantly preferred feeding uponSalix borealis, whereas they hardly fed upon birch. The Czech specimens clearly preferred birch (Betula pubescens) to willow species. Application of salicin onto leaves of a willow species free of this phenolglycoside revealed that the Finnish individuals preferred feeding upon leaves with salicin. On the other hand, the Czech individuals avoided feeding upon leaves ofB. pubescens treated with salicin. The chemical composition of the glandular secretion of the Finnish larvae differed from the one of the Czech larvae. GC-MS-analyses of the secretions revealed that salicylaldehyde was the only major component of the secretion of Finnish larvae feeding upon the salicin-containing willowS. borealis. The glandular secretion of the Czech larvae feeding upon birch contained numerous esters of isobutyric acid and 2-methylbutyric acid. When Czech larvae had fed upon a salicin-containing willow (S. fragilis), the major compounds of their secretion were benzoic acid, salicylalcohol and benzoic acid esters; salicylaldehyde was only detected in traces. Thus,C. lapponica individuals from the Finland population adapted so closely to a salicincontaining willow that they clearly prefer this plant for food and that they obviously derive their main larval defensive compound (salicylaldehyde) from their host-plant.

    Notes: Abstract The lipid fraction of the defensive secretion of adultGastrophysa viridula was analyzed by gas-liquid chromatography and gas chromatography-mass spectrometry. Three long chain acetates and two methyl-branched hydrocarbons were identified and their amounts in the secretion of single field-collected and laboratory-reared beetles determined. The composition of the secretion was affected by the sex, reproductive status, and age of the beetle. The ratio of (Z)-11-eicosenyl acetate to (Z)-13-docosenyl acetate ranged from 0.30 to 0.78 in male beetles and from 1.05 to 2.06 in female beetles. The biological significance of this sexual dimorphism is discussed.

    Notes: Abstract The first known long-range female-released sex pheromone for the family Cerambycidae is reported fromMigdolus fryanus, a sugarcane pest in South America. Although two female-specific compounds, namely, N-(2′S)-methylbutanoyl 2-methylbutylamine and N-formyll-isoleucine methyl esters were identified, field tests with synthetic chemicals revealed that only the amide was active and that the amino acid derivative neither increased or decreased trap catches by the amide. This is the first identification of amide as a sex pheromone.

    Notes: Abstract Missense errors in the translation of mRNAs in Saccharomyces cerevisiae were screened by looking for charge heterogeneity of proteins on two-dimensional gels resulting from the substitution of charged and neutral amino acids. No such mistranslation was detected in wild-type yeast strains grown in the presence of the translational error-inducing antibiotic paromomycin. However, paromomycin-induced mistranslation of a heterologous mRNA, encoding human phosphoglycerate kinase expressed in yeast, was seen. We suggest that the combination of error-prone translation of a heterologous mRNA, and growth in the presence of paromomycin, leads to an accumulation of mistranslated proteins that can be detected by two-dimensional gel electrophoresis.

    Notes: Abstract A multidrug resistance gene, YDR1, of Saccharomyces cerevisiae, which encodes a 170-kDa protein of a member of the ABC superfamily, was identified. Disruption of YDR1 resulted in hypersensitivity to cycloheximide, cerulenin, compactin, staurosporine and fluphenazine, indicating that YDR1 is an important determinant of cross resistance to apparently-unrelated drugs. The Ydr1 protein bears the highest similarity to the S. cerevisiae Snq2 protein required for resistance to the mutagen 4-NQO. The drug-specificity analysis of YDR1 and SNQ2 by gene disruption, and its phenotypic suppression by the overexpressed genes, revealed overlapping, yet distinct, specificities. YDR1 was responsible for cycloheximide, cerulenin and compactin resistance, whereas, SNQ2 was responsible for 4-NQO resistance. The two genes had overlapping specificities toward staurosporine and fluphenazine. The transcription of YDR1 and SNQ2 was induced by various drugs, both relevant and irrelevant to the resistance caused by the gene, suggesting that drug specificity can be mainly attributed to the functional difference of the putative transporters. The transcription of these genes was also increased by heat shock. The yeast drug-resistance system provides a novel model for mammalian multidrug resistance.

The mutant showed higher sensitivity to photoactivated bifunctional psoralen (8-MOP+UVA) during all stages of the meiotic cycle. Mutation induction by 3-CPs+UVA or 8-MOP+UVA in meiosis-committed cells revealed no significant differences between wild-type and thepso3-1 mutant. The status of thePSO3 gene has no influence on the kinetics of induction of gene conversion and crossing-over after 3-CPs+UVA treatment in meiosis-committed cells: gene conversion was blocked while recombination was induced. After treatment with 8-MOP+UVA gene conversion was also blocked in both strains while crossing-over could only be observed in meiosis-committed wild-type cells.

    Notes: Abstract We have devised a new strategy to clone DNA sequences from an yeast autonomously-propagating plasmid into a non-autonomous integrative vector by in-vivo recombination. The method consists of a first step in which the replicative plasmid carrying the DNA fragment of interest forms a co-integrate with the non-replicative plasmid by an induced in-vivo reciprocal exchange accompanied by gene conversion. The dimeric plasmid obtained is then purified and cut with an appropriate restriction enzyme and ligated independently to obtain the two intact monomeric plasmids, the original autonomous plasmid plus the new non-autonomous plasmid carrying the subcloned DNA fragment. The dimeric co-integrate can also serve as substrate for a second in-vivo reciprocal exchange that produces new autonomous plasmids carrying the desired DNA fragment. The technique considerably expands the applications of in-vivo cloning in yeast by complementing three important characteristics of previously published methods: (1) it can be used to clone into non-propagating vectors; (2) co-transformation experiments are not required; and (3) the intermediate co-integrate can be used to generate new types of autonomously-propagating plasmids directly. These characteristics are independent of whether the DNA insert is flanked by appropriate restriction sites or whether it does, or does not, express a detectable phenotype in yeast. The method is particularly useful for the cloning of large DNA fragments and can be used for plasmids from organisms other than yeasts.

    Notes: Abstract In this paper we examine the expression of the Saccharomyces cerevisiae CYT2 gene, which encodes cytochrome c 1 heme lyase. This enzyme is required for covalent attachment of heme to apocytochrome c 1, a subunit of the mitochondrial respiratory chain. Transcription of the 1-kb CYT2 mRNA initiates at four prominent sites at a distance of 52–225 bp in front of the AUG start codon. The level of CYT2 mRNA is not influenced by the presence or absence of oxygen or of heme, but it is subject to carbonsource control. The concentration of the CYT2 mRNA is significantly reduced in glucose-grown cells as compared to cells grown under non-repressing conditions. Neither the HAPp activator proteins nor MIG1p, a repressor protein involved in glucose repression, seem to mediate this effect.

    Notes: Abstract The effect of the Escherichia coli RecA protein on mitotic recombination in the diploid D7 strain of Saccharomyces cerevisiae damaged by UV radiation was investigated. The D7 strain was transformed by two modified versions of the pNF2 plasmid: one, containing the ADH-1 promoter, and the other containing the recA gene tandemly arranged behind the ADH-1 promoter region. Immunological analysis proved the presence of the 38-kDa RecA protein in D7/pNF2ADHrecA transformants. We observed a positive effect of recA gene expression on mitotic gene conversion, mainly at higher doses of UV radiation. The results indicate that a RecA-like activity could participate in steps preceeding mitotic conversion events in yeast.

    Notes: Abstract In former studies it was found that the ERV1 gene is essential for cell viability and for the biogenesis of functional mitochondria. A temperature-sensitive nuclear mutant exhibits a severe reduction in all the mitochondrial transcripts. Elimination of the gene leads to growth arrest after a few cell divisions. The putative gene product bears the characteristics of a regulatory factor since it has low expression rate and a high content of charged amino acids. In this study it is further verified that the ERV1 gene alone is responsible for the observed cellular and mitochondrial defects. The 5′ region of the gene is analysed by DNA deletions and complementation studies. Expression of the gene under the control of the GAL1-10 promoter in a disruption strain of ERV1 allows a more detailed specification of its influence on mitochondrial and cellular functions. Immediate and complete loss of mitochondrial genomes is observed after the promoter has been shut off, whereas the yeast cells are still able to grow for a limited time under these conditions. Analysis of the cells by in-vivo DNA flurorescence demonstrates a specific arrest in the cell-division cycle as the terminal phenotype. To further characterize the temperature-sensitive allele of ERV1 the mutated gene has been isolated and sequenced. A single point mutation which leads to the exchange of a single amino acid is found in the reading frame.

    Notes: Abstract The nuclear gene MRP-L13 of Saccharomyces cerevisiae, which codes for the mitochondrial ribosomal protein YmL13, has been cloned and characterized. It is a single-copy gene residing on chromosome XI. Its nucleotide sequence was found to be identical to that of the previously reported ORF YK105. A comparison of the predicted protein sequence of the MRP-L13 gene product and the actual N-terminal amino-acid sequence of the isolated YmL13 protein indicated that the mature protein is preceded by a mitochondrial signal peptide of 86 amino-acid residues, which is the longest among all known mitochondrial ribosomal proteins of S. cerevisiae. No sequence similarity was found to any other ribosomal protein in the current databases. The transcription of MRP-L13 was found to be repressed in the presence of glucose. Its protein product is not strictly essential for mitochondrial functions, but disruption of the gene by insertion of LEU2 noticeably affected cellular growth on non-fermentable carbon sources.

    Notes: Abstract We used a genetic approach to study the nuclear factors involved in the biogenesis of mitochondrial tRNAs. A point mutation in the mitochondrial tRNAAsp gene of Saccharomyces cerevisiae had previously been shown to result in a temperature-sensitive respiratory-deficient phenotype as a result of the absence of 3′ end-processing of the tRNAAsp. Analysis of mitochondrial revertants has shown that all revertants sequenced have a G-A compensatory change at position 53, which restores the hydrogen-bond with the mutated nucleotide. We then searched for nuclear suppressors to identify the nuclear gene(s) involved in mitochondrial tRNA 3′ end-processing. One such suppressor mutation was further characterized: it restores tRNAAsp maturation and growth at 36°C on glycerol medium in heterozygous diploids, but leads to a defective growth phenotype in haploids.

    Notes: Abstract We have constructed a gene coding for the 12-kDa intermediate form of the 2s methionine-rich protein from Bertholletia excelsa seeds. This protein, expressed intracellularly in yeast, is characterised by a 20-min balf-life. By adding 11 amino acids corresponding to the peroxisome-targeting sequence (PTSc) of luciferase, we have significantly increased its half-life. This stabilization allowed accumulation of the BZN protein into the peroxisome as judged by cell fractionation. Accumulation of the 12-kDa protein results in a significant increase of the total methionine content in yeast cells (30%) indicating that such a microorganism could represent a practicable protected shuttl for an animal-feed additive.

    Notes: Abstract Three respiratory-deficient mutants of cytochrome oxidase subunit I in the yeast mitochondrion have been sequenced. They are located in, or near, transmembrane segment VI, the catalytic core of the enzyme. Respiratory-competent revertants have been selected and studied. The mutant V244M was found to revert at the same site in valine (wild-type), isoleucine or threonine. The revertants of the mutant G251R were of three types: glycine (wild-type), serine and threonine at position 251. A search for second-site mutations was carried out but none were found. Among 60 revertants tested, the mutant K265M was found to revert only to the wild-type allele.

    Notes: Summary After a review of the existing state of affairs, an improvement is made in the stability theory for solitary-wave solutions of evolution equations of Korteweg-de Vries-type modelling the propagation of small-amplitude long waves. It is shown that the bulk of the solution emerging from initial data that is a small perturbation of an exact solitary wave travels at a speed close to that of the unperturbed solitary wave. This not unexpected result lends credibility to the presumption that the solution emanating from a perturbed solitary wave consists mainly of a nearby solitary wave. The result makes use of the existing stability theory together with certain small refinements, coupled with a new expression for the speed of propagation of the disturbance. The idea behind our result is also shown to be effective in the context of one-dimensional regularized long-wave equations and multidimensional nonlinear Schrödinger equations.

    Notes: Abstract Rylux BSU, a new fluorescent brightener from the family of 4,4′-diaminostilbene-2,2′disulfonic acid derivatives, inhibited growth and cytokinesis of the yeast Saccharomyces cerevisiae. In the presence of 0.1–1 mg/ml Rylux BSU the cells grew in clumps, had irregular shape and were larger than controls. They formed apparently normal primary septa but their secondary septa and lateral cell walls, especially those in older cells, were abnormally thick with large deposits of amorphous wall material in the periplasmic spaces all over the cell surface. Chitin content in the cell walls of cells grown in the presence of Rylux BSU was increased 2 to 5 times in comparison to that of the controls and glucan content was reduced by up to 30%. In the in vitro assays with particulate membrane fractions, Rylux BSU acted as a non-competitive inhibitor of β-1,3-glucan synthase with inhibitory constant K i=1.75 mg/ml whereas the chitin synthase was inhibited to a much lesser extent. From the difference of the effects of Rylux BSU on the synthesis of chitin in vivo and in vitro it is concluded that the brightener interacts with chitin synthase only indirectly, possibly by influencing the properties of integral plasma membrane.

    Notes: Summary Saccharomyces cerevisiae in the form of baker's yeast, cells cultivated on a yeast extract-peptone-glucose medium, as well as cells immobilized in 18% (w/v) polyacrylamide gel showed the ability to hydrolyze 1.727 mM sodium phytate solution at 45°C, pH 4.6, in a stirred tank reactor. Seventy percent yield of dephosphorylation was observed after 2 h using a baker's yeast concentration of 5.8 g dry matter per 100 ml. Hydrolytic activity at 1.8–2.0 μM Pi min−1 was observed between 1st and 3rd h of the reaction in cells cultured 24 or 48 h. No inhibition by the substrate was found at sodium phytate concentrations of 0.587–1.727 mM. After 1.5 h of hydrolysis a single, well distinguished peak ofmyo-inositol-triphosphate was the main product found. By means of immobilization the stability of the biocatalyst was enhanced 3.3-fold and reached its half-life at 64 ninety-minute runs.

    Notes: Summary The evolution of the cell and must contents of three short-chain fatty acids (C6, C8 and C10) and their ethyl esters during fermentations withSaccharomyces cerevisiae racescerevisiae, bayanus andcapensis were studied. The former is a fermentative yeast and the last two are ‘flor’ film yeasts. The acid concentrations in the musts increased throughout the alcoholic fermentations, and maximum cell concentrations of the fatty acids were reached after 48 h of fermentation. Maximum ester concentrations in the cells were attained after 48–72 h of fermentation. In the musts, ethyl octanoate and ethyl decanoate reached a peak also at this point, and ethyl hexanoate after 10 days. After 134 days,S. cerevisiae racecapensis formed a thick ‘flor’ film whileS. cerevisiae racebayanus developed a thin film andS. cerevisiae racecerevisiae formed no film. At this point, acid contents remained constant in the wines produced byS. cerevisiae racescerevisiae andbayanus, and decreased in those obtained with racecapensis. The ethyl ester contents tended to decrease with the exception of ethyl decanoate in the fermentations carried out byS. cerevisiae racescerevisiae andbayanus.

    Notes: Abstract A system of suspending traps from a movable balloon was developed and tested on pollinating and wood boring Coleoptera in a tropical rainforest canopy in Pasoh Forest Reserve, Peninsular Malaysia. The balloons, 2.5 m in diameter, were filled with helium gas for levitation and moored in different directions by three pieces of thin rope. Traps in which benzyl acetate and ethanol were used as attractants for the pollinators and wood borers, respectively, were suspended from the balloon at different heights. The trapping results show that the balloon system can be used for studying canopy insects and their vertical distribution on a spatial and temporal scale.

The superior self-assembling properties of the amphiphiles are advantageous for forming surface monolayer and bilayer membranes (BLMs).     Notes: Abstract The renin-angiotensin system (RAS) is the most important regulator of electrolyte homeostasis and blood pressure. Our recently generated transgenic mice carrying either the human renin (hREN) or human angiotensinogen (hANG) genes did not develop hypertension but dual gene strains obtained by cross-mating separate lines of mice exhibited a chronically sustained increase in blood pressure, suggesting the presence of species-specific reactivity between renin and angiotensinogen. In order to examine this specificity, the present study was designed to perform a strictly comparative study on hydrolysis of hANG by hREN and mouse submandibular renin (mREN)in vitro by using pure proteins. The recombinant hANG (rhANG) and the synthetic human-type tridecapeptide (hTDP), Asp-Arg-Val-Tyr-Ile-His-Pro-Phe-His-Leu-Val-Ile-His, corresponding to the N-terminal sequences of hANG, were used to determine the species specificity of recombinant hREN (rhREN) and mREN. While hTDP was cleaved by both rhREN with similar Km and with the same order of kcat, rhANG was cleaved by mREN with 16.7-fold higher Km and with 28.2-fold lower kcat than by rhREN. These results showed that kcat/Km value of mREN for rhANG was 468-fold lower than that for rhREN acting on rhANG.

The structure of the oxidized specimens was examined. TGA, XRD, EPMA, SEM, metallographic analysis, and microhardness measurements were carried out. Impurity elements were found to increase the oxidation rate of technical zirconium. The mechanism of the effect of impurity elements on zirconium oxidation was shown to differ for the α- and β-phases. Activation energies were calculated for the parabolic and linear stages of oxidation.

Oxidation-reaction products are HfO2(moncl) and α-solid solution of oxygen in hafnium. Anomalous oxidation behavior of hafnium having a high impurity content was found in the range 800–950°C. Activation energies for the parabolic and linear oxidation stages were calculated.

    Notes: Abstract In this minireview, we summarize recent advances on the enzymology of ADP-ribose polymer synthesis. First, a short discussion of the primary structure and cloning of poly(ADP-ribose) polymerase (PARP) [EC 2.4.2.30], the enzyme that catalyzes, the synthesis of poly(ADP-ribose), is presented. A catalytic distinction between the multiple enzymatic activities of PARP is established. The direction of ADP-ribose chain growth as well as the molecular mechanism of the automodification reaction catalyzed by PARP are described. Current approaches to dissect ADP-ribose polymer synthesis into individual reactions of initiation, elongation and branching, as well as a partial mechanistic characterization of the ADP-ribose elongation reaction at he chemical level are also presented. Finally, recent developments in the catalytic characterization of PARP by site-directed mutagensis are also briefly summarized.

    Notes: Abstract TheMaladera matrida beetle (Coleoptera, Scarabaeidae, Melolonthinae), a relatively new species to science, was first identified in Israel in 1983. In the course of field observations it was found that adultM. matrida beetles emerged from the soil at sunset to feed and mate. During the first 20 min of flight, most of the beetles were males. The females emerged shortly afterwards, and aggregations numbering 20–30 individuals with equal proportions of males and females were eventually formed on peanut plants. Laboratory olfactometer bioassays showed that peanut leaves (food) attracted both males and females. Field-trapping experiments and olfactometer studies showed thatM. matrida beetles were highly attracted by live virgin females in the presence of food (cut-up peanut leaves). Another set of field trapping experiments indicated that airborne volatiles produced by live virgin females plus food had the same attracting ability as live virgin females plus food. The attraction exerted by the combination of live virgin females and peanut leave volatiles suggests a synergism effect. Accordingly, we propose a two-stage mechanism of chemical communication in theM. matrida beetles: first, the males cause mechanical damage to the host plant to attract both sexes; later, the females emit attractants (sex pheromone) while eating or shortly thereafter.

    Notes: Abstract AdultMaladera matrida Argaman (Coleoptera: Scarabaeidae: Melolonthinae) males emerge from soil for an active period at dusk, a few minutes before the females. Adults are found during most of the active hours on the foliage in aggregations composed of an equal sex ratio. The mechanism of aggregation behavior ofM. matrida beetles was studied in a Y-shaped olfactometer. No evidence was found for the existence of an aggregation pheromone released either by males or by females, but behavior tests indicate that adultM. matrida beetles, males as well as females, are attracted to volatiles of an injured host plant. The following scenario is suggested: Males emerge daily from soil at dusk, a few minutes before the females, and immediately start feeding. Additional males are attracted to the injured host's volatiles and form aggregations. When females emerge from soil, the attractant volatiles are concentrated in spots, and the females join the aggregations, forming an equal sex ratio.

    Notes: Abstract Larvae and adults of the Colorado potato beetle,Leptinotarsa decemlineata (Say), are shown to have galeal gustatory cells that are highly sensitive to distillate of potato leaf extracts, (E)-2-hexen-1-ol, (E)-2-hexenal, and other saturated and unsaturated six-carbon alcohols. In larvae and adults, the sensory response patterns elicited by leaf homogenate, leaf distillate and a mixture of these two extracts differ in subtle ways. Beetle larvae feed most readily on Millipore disks treated with leaf homogenate and the mixture, but they did not feed on disks treated with leaf distillate. The differences in behavioral response and sensory input are used to derive a potential gustatory code that may stimulate different levels of feeding. This code may be disrupted by compounds present in nonhost leaves, thus leading to reduced feeding. Possible interactions of sapid leaf volatiles, amino acids, sugars, and potentially deterrent plant compounds are discussed.

    Notes: Abstract Bolts of Scots pine,Pinus sylvestris L., attacked by the bark beetleTomicus piniperda (L.) were baited with ethanol and α-pinene to attract antagonistic insects and thereby enhance their detrimental effects on the production of bark beetle progeny. Unbaited and caged bolts were included in the experiments as controls. Attraction of beetles to the bolts and subsequent emergence were estimated using traps. Six phloem-feeding species (potential competitors ofT. piniperda) and four predatory species were caught in significantly higher numbers at the baited bolts than at the unbaited ones. The number of offspring and the productivity ofT. piniperda were four to seven times higher in unbaited bolts than in baited bolts. Exclusion of other insects, by using cages, resulted in a nine-fold increase in the number ofT. piniperda offspring per square meter and productivity (offspring per egg gallery) compared with unbaited, exposed bolts.Hylurgops palliatus (Gyll.) (Scolytidae) andRhagium inquisitor (L.) (Cerambycidae) attacked both the baited and unbaited bolts, whereasAcanthocinus aedilis (L.) (Cerambycidae) andPytho depressus (L.) (Pythidae) reproduced almost exclusively in the baited ones. Large numbers of larvae ofThanasimus (Cleridae) andRhizophagus (Rhizophagidae) emerged from both the baited and unbaited bolts. Adults ofPlegaderus vulneratus (Panzer) andCylister linearis (Er.) (Histeridae) emerged almost exclusively from the baited bolts. The low progeny production ofT. piniperda in the baited bolts was attributed largely to the influence of adults ofRhizophagus andEpuraea (Nitidulidae), and larvae ofThanasimus andA. aedilis.

    Notes: Abstract Males ofCarpophilus obsoletus Erichson produce an aggregation pheromone to which both sexes respond. The pheromone was identified by GC-MS as (2E,4E,6E,8E)-3,5,7-trimethyl-2,4,6,8-undecatetraene (1), which is also a minor constituent of the pheromone blends ofC. hemipterus (L.),C. freemani Dobson, andC. lugubris Murray. The pheromone was synergized in wind-tunnel bioassays by propyl acetate, a “host-type” coattractant. In a dose-response study, 50 pg of1, plus propyl acetate, was significantly more attractive than just propyl acetate. Pheromone emission from groups of 65 males, feeding on artificial diet, averaged 2.2 ng/male/day. Emissions from individual males were larger, averaging 72 ng/day and ranging as high as 388 ng/day. Synthetic1 was tested in a date garden in southern California (500 µg/rubber septum), using fermenting whole-wheat bread dough as the coattractant. The pheromone plus dough attracted significantly more beetles than dough alone (means were 4.2 and 0.0 beetles per week per trap). Captured beetles were 54% females. Field trap catches were highest during the months of July and August.

    Notes: Abstract Females of the Oriental beetle,Anomala orientalis (Waterhouse), release a sex pheromone composed of a 9:1 blend of (Z)- and (E)-7-tetradecen-2-one. The double-bond position of the pheromone was determined by DMDS derivatization and interpretation of the fragmentation patterns produced by monounsaturated ketones. In a sustained-flight tunnel, males responded by flying toward female beetles and attempting to copulate with them. Both effluvium and whole-body extracts of OB females were analyzed, and the activity was found only in the airborne extracts. Flight-tunnel bioassays also showed that a synthetic 90:10Z/E blend on a rubber septum was attractive and that the responses of males to this blend were equivalent toZ isomer alone, but much better than to the singleE isomer.

    Notes: Abstract A new means for generating silica gel has been developed. Simple two component systems comprising tetraalkoxysilanes and strong carboxylic acids such as formic acid can react rapidly to give transparent monolithic gels. The acid serves as solvent, water source, and catalyst for both hydrolysis and condensation. Water need not be present as an initial reactant; it is generated in situ during the reaction. The gelation reaction is at least two orders of magnitude faster than those conducted in conventional acid-catalyzed aqueous systems at comparable pH*. Kinetic evidence indicates a lowering of the activation energy of condensation reactions, believed to be associated with reaction of silyl carboxylates and silanol groups. Physical properties of the dry gels such as bulk density more closely resemble conventional acid-catalyzed gels than those associated with other rapidly gelling systems. Dry gels often exhibit porosity so fine that nitrogen (at 77°K) is not absorbed at significant rates. Independent evidence of porosity arises from comparison of skeletal and bulk densities, sample immersion in water and adsorption isotherms of CO2.

Besides DNA polymerase, RNA polymerase, primase, 3′→5′ exonuclease and an ATPase activities were found to be associated with it. The presence of some of these enzymes were confirmed by Western blot. This high molecular weight multienzyme complex containing DNA has most of the attributes of a putative replisome.

    Notes: Abstract A cDNA encoding farnesyl diphosphate synthase, an enzyme that synthesizes C15 isoprenoid diphosphate from isopentenyl diphosphate and dimethylallyl diphosphate, was cloned from an Arabidopsis thaliana cDNA library by complementation of a mutant of Saccharomyces cerevisiae deficient in this enzyme. The A. thaliana cDNA was also able to complement the lethal phenotype of the erg20 deletion yeast mutant. As deduced from the full-length 1.22 kb cDNA nucleotide sequence, the polypeptide contains 343 amino acids and has a relative molecular mass of 39689. The predicted amino acid sequence presents about 50% identity with the yeast, rat and human FPP synthases. Southern blot analyses indicate that A. thaliana probably contains a single gene for farnesyl diphosphate synthase.

However, it is evident that either Ser or Thr at position 102 are of a size able to accommodate a wide variety of substrates in the active site and substrate specificity appears to rely largely on size discrimination in these mutants. The Q102F and Q102Y mutant enzymes have low catalytic efficiency and do not show this relaxed substrate specificity. However, their activities are restored by the presence of an aromatic substrate. All of the enzymes have a very low catalytic efficiency with branched chain aliphatic substrates.

    Notes: Abstract The interaction between the fluorescent ouabain derivative DEDO and purified renal Na,K-ATPase (isolated from different animal species) is investigated. Equilibrium binding studies provide a pK value of about 7.5 and a stoichoimetric coefficient of 1. Nonmodified ouabain exhibits the same affinity to the rabbit enzyme; the enzyme originating from the other sources binds DEDO 10 times less strongly than ouabain. Kinetic studies indicate that this is the consequence of a 10 times higher dissociation rate constant of the complexes formed with DEDO. The fluorescence emission intensity of DEDO is enhanced, being dependent on the enzyme source. The single decay time of DEDO is 3 ns in the absence and 21 ns in the presence of the rabbit enzyme and 14 ns in the presence of the pig renal enzyme. This result suggests that the fluorophore of DEDO is bound to a very hydrophobic environment of the enzyme. Further characterization of the static fluorescence spectra provides evidence for energy transfer between Trp residues of the enzyme and DEDO. Distance estimations suggest that one or two Trp residues are likely to be located in the proximity of the fluorophore.

    Notes: Abstract Cytochrome b5 binds spontaneously to lipid vescles and also self-associates in aqueous solution. Two mutant proteins have been generated, one has a self-association constant which is less than that of the native protein, while the other has a larger self-association constant. All three proteins have Trp in the membrane-binding domain but as aqueous solutions of these proteins contain differing amounts of monomeric protein, the kinetics of fluorescence enhancement, when the proteins are mixed with lipid vesicles, are complex. Similar complex kinetics are seen when the Trp are quenched by the addition of bromolipid vesicles. The mutant which has Trp 108 and 112 both replaced by Leu does not self-associate and shows monoexponential stopped-flow fluorescence kinetics. Identical rate constants are seen with this mutant for fluorescence enhancement by POPC and fluorescence quenching by three bromolipids with bromines at the 6,7-, 9,10-, and 11,12-positions of thesn-2 acyl chain. This rate constant is only 1% of the calculated collisional rate constant and it is suggested that the reduced rate is caused by a reduction in the number of productive collisions rather than by a slow rate of penetration of the membrane-binding domain into the bilayer.

    Notes: Abstract The zinc-binding long-chain alcohol dehydrogenases from plants and animals exhibit a considerable level of amino acid sequence conservation. While the functional importance of many of the conserved residues is known, the role of others has not yet been determined. We have identified a naturally occurring Adh-1 allele in the legume Phaseolus acutifolius with several unusual characteristics. Individuals homozygous for this allele, Adh-1CN, possess a single isozyme starch gel electrophoretic pattern suggestive of a null allele, and exhibit ADH enzyme activity levels ca. 60% lower than the standard wild-type Adh-1F line. Interestingly, analysis of Adh-1CN homozygotes on an alternative gel system indicates that Adh-1CN does encode a polypeptide capable of forming functional homo- and heterodimers. However, the levels of ADH activity displayed by these isozymes are far lower than those observed for the corresponding wild type ADH-1F isozymes. Dialysis experiments indicate that isozymes containing the ADH-1CN polypeptide are inactivated by slightly acidic conditions, which may explain the apparent null phenotype on starch gels. Elevated temperatures cause a similar loss of enzyme activity. The deduced amino acid sequences of ADH-1CN and ADH-1F were obtained from their corresponding cDNA clones, and the only significant difference detected between the two is a single amino acid replacement substitution. Residue 144 is occupied by phenylalanine in the ADH-1F polypeptide, whereas serine occupies this position in the ADH-1CN polypeptide. The proximity of residue 144 to the catalytic zinc in the substrate-binding pocket, coupled with the fact that it is integral to a defined hydrophobic core of the ADH polypeptide, may explain the observed disruptive effect that the serine substitution has on both the activity and stability of the ADH-1CN polypeptide. It also provides an explanation for the maintenance of phenylalanine or the structurally similar tyrosine at this residue in Zn-binding long-chain ADHs.

    Notes: Abstract The deterministic dynamics of the classical single-locus multiple-allele model of gametophytic incompatibility is analyzed with the intention to prove the conjecture that the symmetric state (uniform distribution of genotypes) is the only polymorphic equilibrium and that this equilibrium is globally asymptotically stable in the interior of the frequency simplex. It is shown that the minimum allelic frequency increases strictly over the generations as long as a uniform allelic distribution is not realized. Hence, the minimum allelic frequency is a Ljapunov function for the invariant set of genotypic frequencies characterized by a uniform allelic distribution. Within this set, the uniform genotypic distribution is approached in an exponential fashion, which proves the assertion. An evolutionary optimization rule associated with the global convergence to the symmetric state is implied by the fact that at this state the overall amount of pollen elimination resulting from incompatible crosses is minimized.

    Notes: Abstract Steric structures of dysprosium(III) aminobenzoate complexes with the 1∶1 and 1∶2 molar ratio in aqueous solutions were determined on the basis of pH-metric and paramagnetic birefringence data. An increase in conjugation observed for the series of the acids,viz., benzoic,meta-, ortho-, andpara-aminobenzoic acids, results in the increased stability of the complexes with the 1∶1 and 1∶2 composition. In the case ofpara-aminobenzoic acid, the polyhedra [DyL(H2O)6]2+ and [DyL2(H2O)4]+ are cubes with the ligands coordinated to one and two edges, respectively. In the case ofmeta-aminobenzoic acid, the polyhedra [DyL(H2O)6]2+ and [DyL2(H2O)4]+ are a dodecahedron with the ligand coordinated to one edge and a square anti-prism with the ligands coordinated to two edges, respectively. In the case ofortho-aminobenzoic acid, both the 1∶1 and 1∶2 complexes have structures that are intermediate between the structures ofmeta- andpara-aminobenzoic acids.

    Notes: Abstract The results of kinetic studies on ligand substitution in [M3(CO)11X]− complexes (M = Ru, Os; X = Cl, Br, I) are summarized. The [Os3(CO)11X]− complexes react with PPh3 under mild conditions to initially yield monosubstituted products [Os3(CO)10(PPh3)X]−. The rate of CO substitution obeys a first-order equation with respect to the concentration of the complex and does not depend on the ligand concentration. The rates of the reactions decrease in the order Cl 〉 Br 〉 I withΔH≠ values increasing from 15 to 18 kcal mol−1 and ΔS ≠ values varying from −19 to −13 cal mol−1 K−1. The enhanced reactivities of these complexes as well as the low activation energies and negative activation entropies are discussed in terms of the effects of μ-X bridge formation on the transition state of the reaction. Reactions of PPN[Ru3(CO)11−x (Cl)] (PPN is the bis(triphenylphosphine)iminium cation;x=0, 1) and PPN[Ru3(CO)9(μ3-I)] with alkynes are also reported. The reactivities of alkynes follow the order BuC≡CH ≥ PhC≡CH ≥ EtC≡CEt ≥ PhC≡CPh. The higher rates of the reactions of monosubstituted acetylenes compared with those of their disubstituted analogs are explained by agostic interaction between the metal atom and the C-H bond in the reaction transition state and by steric effects. The results obtained attest that the reaction with alkynes occursvia intermediates containing halide bridges and that μ3-halide complexes are more reactive than μ2-halide complexes.

    Notes: Abstract Cerulenin, an antifungal antibiotic produced by Cephalosporium caerulens, is a potent inhibitor of fatty acid synthase in various organisms, including Saccharomyces cerevisiae. The antibiotic inhibits the enzyme by binding covalently to the active center cysteine of the condensing enzyme domain. We isolated 12 cerulenin-resistant mutants of S. cerevisiae following treatment with ethyl methanesulfonate. The mechanism of cerulenin resistance in one of the mutants, KNCR-1, was studied. Growth of the mutant was over 20 times more resistant to cerulenin than that of the wild-type strain. Tetrad analysis suggested that all mutants mapped at the same locus, FAS2, the gene encoding the α subunit of the fatty acid synthase. The isolated fatty acid synthase, purified from the mutant KNCR-1, was highly resistant to cerulenin. The cerulenin concentration causing 50% inhibition (IC50) of the enzyme activity was measured to be 400 μM, whereas the IC50 value was 15 μM for the enzyme isolated from the wild-type strain, indicating a 30-fold increase in resistance to cerulenin. The FAS2 gene was cloned from the mutant. Sequence replacement experiments suggested that an 0.8 kb EcoRV-HindIII fragment closely correlated with cerulenin resistance. Sequence analysis of this region revealed that the GGT codon encoding Gly-1257 of the FAS2 gene was altered to AGT in the mutant, resulting in the codon for Ser. Furthermore, a recombinant FAS2 gene, in which the 0.8 Kb EcoRV-HindIII fragment of the wild-type FAS2 gene was replaced with the same region from the mutant, when introduced into FAS2-defective S. cerevisiae complemented the FAS2 pheno-type and showed cerulenin resistance. These data indicate that one amino acid substitution (Gly → Ser) in the α subunit of fatty acid synthase is responsible for the cerulenin resistance of the mutant KNCR-1.

    Notes: Abstract GAL11 was first identified as a gene required for full expression of some galactose-inducible genes that are activated by GAL4, and it was subsequently shown to be necessary for full expression of another set of genes activated by RAP1/GRFl/TUF. Genetic analysis suggests that GAL11 functions as a coactivator, mediating the interaction of sequence-specific activators with basal transcription factors. To test this hypothesis, we first tried to identify functional domains by deletion analysis and found that the 866–910 region is indispensable for function. Using reporters bearing various upstream activating sequences (UAS) and different core promoter structures, we show that the involvement of GAL11 in transcriptional activation varies with the target promoter and the particular combination of cis elements. Gel electrophoresis in the presence of chloroquine shows that GAL11 affects the chromatin structure of a circular plasmid. Based on these findings, the role of GAL 11 in regulation of transcription, including an alteration in chromatin structure, is discussed.

    Notes: Abstract The post yield behavior of rigid-perfectly plastic solids at the collapse load is studied based on the finite deformation theory. By using the general duality theory developed by Gao-Strang (1989), a global stability criteria is proposed and a pair of dual extremum principles, expressed in terms of displacements, displacement rates and the Kirchhoff stresses are established for plastic collapse analysis. It is proved that under large deformations, the existence of the plastic limit state at the collapse load depends on the directional derivative of a so-called complementary gap function. The application to the nonlinear plastic collapse theory yields a pair of dual bounding theorems for limit loading factor associated with any transient displacement of the deformed body when the global extremum criteria are satisfied.

    Notes: Summary The aim of this study was to design a parenteral dosage form for the investigational cytotoxic drag carzelesin. A stable formulation in PET (Polyethylene glycol 400/absolute ethanol/Tween 80, 6:3:1, v/v/v) was developed. The prototype, containing 0.50 mg carzelesin in 2.0 ml PET formulation, was found to be the optimal formulation in terms of solubility, stability and dosage requirements in phase I clinical trials. Quality control of the formulation showed that the pharmaceutical preparation of carzelesin in PET is not negatively influenced by the manufacturing process. Shelf life studies demonstrated that the formulation is stable for at least 1 year, when stored at −30°C in the dark. In addition, the stability of carzelesin in the PET formulation is discussed as a function of temperature, additives and after dilution in infusion fluids.

    Notes: Abstract A side-by-side diffusion cell setup has been used to determine the pH 7.4 buffer-octanol interfacial transfer constants for twelve compounds. The compounds are a diverse selection of pharmaceuticals, amino acids, and small organics which covers a log distribution coefficient (K) range of −3 to 2. A model based on the resistance summation approach, which deals explicitly with the various barriers involves in the transport process, was used to derive transfer constants from the transport data for each compound. The model gave constants that were well behaved in a published model describing the correlation of the forward and reverse transfer constants to equilibrium K values (r2 = 0.999). These studies demonstrate the utility of side-by-side diffusion cells for the determination of interfacial transfer constants. This type of setup offers the advantages of controlled interfacial area, measurable hydrodynamic effects, and a commercially available apparatus.

    Notes: Abstract Evaluation of entrapping efficiency is difficult for an o/w emulsion formulation containing a lipophilic oily drug, isocarbacyclin methyl ester (TEI-9090), by commonly employed techniques (dialysis, ultrafiltration, or gel filtration), because of its adsorption to the system materials. Employing this characteristic of TEI-9090, we developed an adsorption technique with polydimethylsiloxane-coated glass beads (PDMS-GB). The assay is based on the quantitative adsorption of unentrapped TEI-9090 to the PDMS-GB. The entrapping efficiency of a 10% soybean oil emulsion containing [3H]TEI-9090 (1 µg/mL) assayed by this method approached 100%. The PDMS-GB assay was performed for the emulsion diluted 100 times with physiological saline at different time intervals after dilution over a period of 24 hr. A plot of [3H]TEI-9090 in the emulsion particles versus time showed rapid release within 1 hr, followed by very slow release, reaching equilibrium. Applying first-order kinetics, the data were found to fit to a biexponential equation over the first hour of release. The terminal release resembled the first-order release of the drug from the phospholipid-rich infranatant, which was separated from the creamy layer by ultracentrifugation of the emulsion and contained 35% [3H]TEI-9090. These results suggest that the drug is released from two components in the emulsion.

    Notes: Abstract Recombinant human interleukin 11 (rhIL-11) is a multispectrum cytokine that plays an important role in megakaryocytopoiesis and platelet production. Probing rhIL-11 chemical reactivity in aqueous solution is an important initial step in developing a dosage form for rhIL-11 clinical trials. This report documents rhIL-11 degradation kinetics at 50°C in solutions adjusted to pH 3.0 to 9.5. Stressed samples were analyzed by reverse-phase HPLC and degradation product peaks were isolated for structural characterization. The results show maximal stability in the region pH 6.5 to 7.0. Degradation product identification shows that the major reaction pathway in acidic solution involves peptide cleavage at aspartate133–proline134. In alkaline solution, protein disappearance proceeds via nonspecific loss to container surfaces. Degradation products at alkaline pH have not been identified.

    Notes: Abstract Chimeric L6 is a mouse–human monoclonal antibody specific for tumor cell-associated antigens. The factors affecting the physical and chemical stability of chimeric L6 were assessed at elevated temperatures (30–60°C) and by multiple freezing and thawing. Three routes of degradation were observed: chemical degradation to smaller molecular weight species, irreversible aggregation, and formation of a reversible dimer. The specific pathway depended on the stress condition applied and the pH, with maximal overall stability to both thermal stress and multiple freezing/thawing observed at about pH 5.5. Other factors including antibody concentration, buffer concentration, NaCl concentration, and agitation had minimal influence on the stability. Commonly used sugars, polyhydric alcohols, and amino acids effectively prevented freeze/thaw-induced aggregation.

    Notes: Abstract In aqueous solutions, dalvastatin (1) undergoes epimerization as well as hydrolysis. The transformation of the drug was studied as a function of pH at 25°C in aqueous solutions containing 20% acetonitrile. At all pH values, first-order plots for the conversion are biphasic, indicating rapid equilibration of 1 with its epimer (2) and slower hydrolysis of 1 to the corresponding β-hydroxy acid (3). Apparent first-order rate constants for the biexponential equation are given as a function of pH. The alkyl–oxygen cleavage of the lactone ring results in the epimerization of 1 to 2, whereas the acyl–oxygen cleavage results in the hydrolysis of 1 to 3. The epimerization is an SN1 reaction reaching an equilibrium of [l] eq/[2] eq = 1.27. The epimerization rate is increased with an increase in the water content of the solvent. The hydrolysis of 1 to 3 is acid and base catalyzed. The hydrolysis is reversible in acidic media and irreversible in neutral and basic media. At pH values greater than 9, the hydrolysis reaction proceeds more rapidly than the epimerization.
