    Description / Table of Contents: Résumé Une technique nouvelle, permettant l'analyse de la densité de faibles quantités de tissus calcifiés pulvérisés, est décrite. Cette technique utilise un analyseur électronique de taille de particules et de volume (»Compteur de Coulter«), pour déterminer les volumes relatifs des fractions pulvérisées, isolées par fractionnement densitométrique, après centrifugation dans des mélanges de solvants organiques. Certains des paramètres, responsables de la distribution densitométrique des poudres d'os, sont étudiés et la reproductibilité du fractionnement et de l'analyse est démontrée. L'application de cette méthode à l'os humain est illustrée par des résultats.
    Abstract: Zusammenfassung Es wird eine neue Technik zur Bestimmung der Dichteverteilung von kleinen Mengen pulverisiertem Knochengewebe beschrieben. Für diese Technik wird ein Apparat zur elektronischen Messung von Partikelgröße und-Volumen (Coulter Counter) verwendet; damit wird das totale relative Volumen der pulverisierten Fraktionen bestimmt, die durch Aufteilung der verschiedenen Dichten mittels Zentrifugieren in Mischungen von organischen Lösungsmitteln isoliert werden. Einige der Parameter, welche die Dichteverteilung von Knochenpulver beeinflussen, wurden untersucht und die Reproduzierbarkeit der Fraktionierung und der Analysen aufgezeigt. Die Anwendung dieser Technik auf Proben von Menschenknochen wird veranschaulicht und die Resultate werden besprochen.
    Notes: Abstract A new technique for the density distribution analysis of small quantities of powdered hard tissues is described. The technique uses an electronic particle size and volume analyser (“Coulter Counter”) to determine the total relative volumes of the powder fractions isolated by centrifugal density fractionation in organic solvent mixtures. Some of the parameters controlling the density distributions of bone powders are examined and the reproducibility of the fractionation and analysis demonstrated. The application of this technique to human bone samples is illustrated and the results discussed.

    Description / Table of Contents: Résumé Des expériences ont été faites sur le rat afin de tester les possiblités d'ingestion perorale simultanée de fluor (F) et de calcium (Ca) en proportions calculées pour traitement de certaines ostéopénies humaines. F sous forme de Na2PO3F et Ca sous forme de gluconate de calcium (CaGluc) n'influençaient pas l'un l'autre quant à l'utilisation par le squelette (fémur). Une viscosité élevée, produite par l'addition d'amidon ou de cellulose carboxyméthylée (CMC) à la solution ou dilution ingérée, augmentait l'utilisation du F même si CaGluc était remplacé par citrate de calcium, qui en soi avait un effet réducteur modéré sur l'utilisation du F. Le glycérophosphate de calcium réduisait fortement l'utilisation du F même en présence de CMC. L'utilisation du F comme NaF était fortement réduite par CaGluc, même en présence de CMC. Les concentrations testées de Na2PO3F, NaF ou CMC n'influençaient pas l'utilisation squelettique de Ca comme CaGluc.
    Abstract: Zusammenfassung Es wurden Rattenexperimente mit markierten Substanzen durchgeführt, um die Auswirkung simultaner peroraler Gaben von Fluor (F) und von Calcium (Ca) zu prüfen, und zwar in einem Verhältnis, das für die Behandlung gewisser menschlicher Osteopenien berechnet wurde. Fluor in Form von Na2PO3F und Ca in Form von Calciumgluconat (CaGluc) interferieren gegenseitig nicht bei der Verwertung durch das Skelet (Femur). Eine hohe Viscosität der eingegebenen Lösung oder der Aufschlämmung, die durch Zusatz von Stärke oder Carboxymethylcellulose (CMC) erzielt wurde, erhöhte die Verwertbarkeit von F sogar wenn CaGluc durch Calciumcitrat ersetzt wurde, welches die F-Verwertung leicht reduzierte. Calcium-glyzerophosphat verminderte die Fluoraufnahme in den Knochen stark, sogar in Anwesenheit von CMC. Die Verwertung von F als NaF war stark herabgesetzt durch CaGluc, selbst beim Vorhandensein von CMC. Die untersuchten Konzentrationen von Na2PO3F, NaF oder CMC hatten keinen Einfluß auf die Calciumaufnahme im Skelet in Form von CaGluc.
    Notes: Abstract Rat experiments with labelled compounds were carried out in order to test the possibilities of simultaneous peroral supply of fluorine (F) and calcium (Ca) in proportions calculated for treatment of certain human osteopenias. F in the form of Na2PO3F and Ca in the form of calcium gluconate (CaGluc) did not interfere with each other's utilisation by the skeleton (femur). A high viscosity produced by adding starch or carboxymethyl cellulose (CMC) to the ingested solution or slurry increased the utilisation of F even when CaGluc was replaced by calcium citrate, which moderately reduced F utilisation. Calcium glycerophosphate strongly reduced F utilisation even in the presence of CMC. The utilisation of F as NaF was strongly reduced by CaGluc, even in the presence of CMC. The tested concentrations of Na2PO3F, NaF or CMC did not influence the skeletal utilisation of Ca as CaGluc.

    Description / Table of Contents: Résumé La diffraction à rayons X, la diffraction électrique et le microscope électronique suggérent qu'en dehors des composants usuels de calcium phosphate (apatite), un nombre d'autres éléments et radicaux sont également présents dans l'arteriosclérose aortique, lesquels sont capables de jouer un rôle dans le développement de la plaque «calcifiée» et dans celui de la lésion anorganique, caractéristique pour l'artériosclerose juvenile. Selon les cas étudies, l'artériosclerose aortique peut se présenter déjà au cours de la troisième decade de la vie et sa composition anorganique est presq'identique à celle de la plaque développée.
    Abstract: Zusammenfassung Sowohl die Befunde der Röntgendiffraktion als auch jene der Elektronendiffraktion und der Elektronenmikroskopie lassen vermuten, daß bei der Aortenatherosklerose zusätzlich zu den üblichen Komponenten von Calciumphosphat (Apatit) eine Reihe anderer Elemente und Radikale vorhanden sind; diese können eine Rolle spielen bei der Entwicklung der “verkalkten” Plaque und bei anorganischen Läsionen juveniler Atherosklerosen. Bei den untersuchten Fällen kann die Aortenatherosklerose bereits im 30. Lebensjahr auftreten; die anorganische Zusammensetzung der Plaque ist nahezu identisch mit jener der bereits entwickelten.
    Notes: Abstract X-ray diffraction, electron diffraction and electron microscopy suggest that in aortic atherosclerosis, in addition to the usual components of calcium phosphate (apatite), a number of other elements and radicals are present which may play a role in the development of the “calcified” plaque and in juvenile atherosclerotic inorganic lesion. In the cases studied, aortic atherosclerosis may appear as early as in the third decade and its inorganic composition is almost identical with the developed plaque.

    Notes: Abstract Excessive administration of ammonium chloride to normal adult male rats receiving a diet adequate in vitamin D caused the development of osteoporosis. The osteoporosis was due to loss of bone substance and bone mineral associated with increased bone resorption.

    Description / Table of Contents: Résumé Après injection intraperitonéale de22Na, la rétention de l'isotope a été déterminée dans l'organisme entier de rasé et dans leur tissu osseux, pendant une période allant jusqu'à 650 jours. Contrairement au45Ca, qui est rapidement concentré dans l'os, puis lentement éliminé, la presque totalité du22Na absorbée par l'os le quitte avec une demie-période de 3–5.5 jours. Cependant, environ 5% de la charge squelettique présente une vitesse de mobilisation lente, avec une demie-période d'environ 700 jours. Etant donné que cette vitesse est comparable à celle de45Ca, il semble que la faible fraction de Na osseux fasse partie de la structure du cristal. Seuls deux des procédés expérimentaux utilisés ont un effet significatif sur la mobilisation du Na. Il s'agit de la consommation prolongée d'un régime pauvre en calcium et d'un changement de l'activité parathyroidienne. Une augmentation du Na alimentaire affecte l'augmentation de la vitesse de mobilisation du sodium dans son composé rapide, alors que le composé lent est peu ou pas affecté. Les calculs effectués, à partir de nos résultats, indiquent que, contrairement au calcium, le squelette ne sert pas de réservoir significatif pour le maintien du milieu liquide extracellulaire.
    Abstract: Zusammenfassung Ratten erhielten intraperitoneal22Na, und die Retention dieses Isotopes wurde während einer Zeitspanne bis zu 650 Tagen im ganzen Körper und in den Knochen bestimmt. Im Gegensatz zu45Ca, welches sehr rasch von den Knochen aufgenommen und dann nur langsam abgegeben wird, verschwindet der größte Teil des vom Knochen aufgenommenen22Na in einer Halbwertzeit von 3–5,5 Tagen. Jedoch zeigen etwa 5% des Knochengerüstes einen sehr langsamen Turnover mit einer Halbwertzeit von ungefähr 700 Tagen. Da dieser Anteil jenem von45Ca vergleichbar ist, kann daraus geschlossen werden, daß diese kleine Fraktion des Na vom Knochen einen integralen Teil der Kristallstruktur ausmacht. Nur zwei der verschiedenen experimentellen Anordnungen, welche ausprobiert wurden, ergaben eine signifikante Wirkung auf den Na-Turnover im Knochen. Diese bestanden einerseits aus einer langzeitigen Verfütterung von Ca-armer Diät, andererseits aus einer Veränderung der Parathyreoidea-Aktivität. Eine Erhöhung der Na in der Nahrung beeinflußte die schnelle Komponente des Na-Turnover im Knochen; sie zeigte aber wenig bis keine Wirkung auf die langsame Komponente. Berechnungen aus unseren Resultaten lassen vermuten, daß — im Gegensatz zum Ca — das Skelet keine signifikante Reservoir-Funktion zur Erhaltung des Na in der extracellulären Flüssigkeit ausübt.
    Notes: Abstract Rats were given22Na intraperitoneally and the retention of the isotope was determined in whole body and in bone for periods up to 650 days. In contrast to45Ca, which is rapidly taken up by bone and then very slowly released, most of the22Na taken up by bone leaves, with a halftime of 3–5.5 days. However, about 5% of the skeletal burden exhibits a very slow turnover, with a half-time of about 700 days. Since this rate is comparable to that of45Ca, it is concluded that this small fraction of bone Na is an integral part of the crystal structure. Only two of the several experimental procedures which were tried produced a significant effect on bone Na turnover. These were prolonged feeding of a low calcium diet and a change in parathyroid activity. An increase in dietary Na affected the fast component of bone Na turnover, but there was little if any effect on the slow component. Calculations from our data suggest that, in contrast to Ca, the skeleton does not serve a significant reservoir function for the support of extracellular fluid Na.

    Description / Table of Contents: Résumé L'effet du courant électrique sur la collagène soluble, en solution dans l'acide acétique dilué, a été étudié pour des voltages, variant de O à 25 volts. Au-dessus de 2,6 volts, la vormation des bandes collagéniques (définies dans ce travail) parait inversement proportionelle, dans le temps, au voltage appliqué. La formation des bandes parait liée au processus d'électrolyse. Les auteurs démontrent que les pH élevés se situent au niveau de la cathode et qu'ils sont suffisants pour induire une précipitation du collagène. Les résultats antérieurs, publiés dans la littérature, décrivant l'action de courant électrique implanté, sont interpretés en fonction du mécanisme étudié au cours de ce travail.
    Abstract: Zusammenfassung Die Wirkung eines elektrischen Stromes auf verdünnte essigsaure Lösungen von löslichem Kollagen wurde bei Spannungen zwischen O und 25 Volt untersucht. Über 2,6 Volt wurde die Bildung von Kollagenbanden (in der Arbeit näher beschrieben) beobachtet, und zwar nach Zeiten, die der angewandten Spannung entgegengesetzt proportional verliefen. Die Bandenbildung wird dem Elektrolyseprozeß zugeschrieben. Wir konnten zeigen, daß sich die hohen pH-Werte rund um die Kathode entwickelten und daß diese genügen, um die Kollagenfällung zu veranlassen. Die Natur dieses Vorganges ist solcher Art, daß erin vivo als Antwort auf durch Stress verursachte Biopotentiale nicht vorkommen kann. Der hier beschriebene Mechanismus erlaubt es, Literaturangaben über den Effekt von implantierten Spannungsquellen zu interpretieren.
    Notes: Abstract The effect of electric current on dilute acetic acid solutions of soluble collagen has been studied for impressed voltages of from 0 to 25 volts. Above 2.6 volts the formation of collagen bands (herein defined) were observed at times inversely proportional to the applied voltage. Band formation is attributed to the process of electrolysis. It has been shown that the high pH values are generated in the area of the cathode, and that they are sufficient to induce collagen to precipitate. The nature of the process is such that it cannot occurin vivo as a response to stress induced biopotentials. Reports in the literature describing the effect of implanted voltage sources are interpreted in terms of the mechanism described here.

    Description / Table of Contents: Résumé Un procédé de dissection a été mis au point pour permettre l'analyse zonale du cartilage de l'épiphyse des os de la jambe d'un foetus bovin. Des échantillons de tissu complet et lavé venant des différentes zones ont été analysés pour déterminer leur contenu en électrolyte et en constituants organiques, ainsi que pour leur densité, cendres et humidité. Les résultats ont montré que lorsque la quantité de cendres et la densité augmentaient, l'eau contenu dans le tissu diminuait. Les quantités de cendres dans les zones de cartilage en voie de calcification étaient plus grandes qu'il avait été. Quand elles étaient exprimées comme un pourcentage du poids sec, elles étaient les plus importantes dans le cartilage lavé calcifié que dans le autre zones. Au début de la minéralisation du cartilage, la quantité de Na (m moles/l de tissu frais) diminuait tandis que celles du Ca et du P inorganique augmentaient. Les niveaux de Mg augmentaient pendant que la calcification se poursuivait, mais seulement à une faction du taux du Ca et du P. Les rapports Ca/P inorganique étaient les plus grands dans le cartilage au repos (Cartilage non-différentié hyalin), suggérant un lien initiale entre Ca et les chrondromucoprotéines. Cependant, au début de la calcification, pendant la prolifération du cartilage les rapports Ca/P étaient beaucoup plus petits (ca. 1.50) mais augmentaient graduellement avec l'advancement de la minéralisation. Des changements importants survenaient dans la composition de la phase organique, pendant la calcification endochondrale. Comme il a été déterminé par l'analyse de l'hydroxyproline la quantité de collagéne diminuait progressivement pendant la calcification du cartilage, mais augmentait rapidement pendant la formation d'os. Comme il a été déterminé par l'analyse de l'héxosamine et du sulfute les chrondromucoprotéines étaient aux niveaux les plus éléves pendant la prolifération du cartilage et diminuaient constamment au cours de la calcification. Cependant, bien que la calcification était déja très avancée dans le cartilage hypertrophique, de grandes quantites de mucopolysaccharides étaient encore présentes. Les rapports sulfure/hhéxosamine montraient un léger déclin pendant les premiéres étapes de la calcification, mais augmentaient beaucoup pendant le cours de la minéralisation. Les quantités d'acide sialique étaient plus grandes dans le cartilage de l'épiphyse que dans le cartilage au repos ou dans l'os. Les lipides augmentaient rapidement pendant la calcification du cartilage, mais étaient très réduites dans l'os complètement formé. La signification de ces résultats est discutée.
    Abstract: Zusammenfassung Eine Seziermethode, die eine Schichten-Analyse der Beinepiphysenplatte von Rinderfeten erlaubt, wurde entwickelt. Proben vor und nach Waschen des Gewebes der verschiedenen Schichten werden untersucht in bezug auf Elektrolyte und organische Bestandteile, als auch in bezug auf Dichte, Aschengehalt und Feuchtigkeit. Die Resultate zeigten eine Zunahme des Aschengehaltes und der Dichte, während der Wassergehalt abnahm. Unerwartet hoch waren die Aschenwerte im in Verkalkung begriffenen Knorpel. Ausgedrückt in Prozent Trockengewicht, ergab gewaschener, verkalkter Knorpel den höchsten Wert aller Zonen. In den Frühstadien der Knorpelmineralisation nahm der Natriumgehalt (m Mol/l Frischgewebe) ab, während Ca und anorganischer P zunahmen. Mit fortschreitender Verkalkung erhöhte sich auch der Magnesium-Spiegel, allerdings nur zu einem Bruchteil des Ausmaßes, in welchem Ca und P zunahmen. Die höchsten Ca/P anorg. Verhältnisse wurden im Ruheknorpel (undifferenzierter hyaliner Knorpel) gefunden, was auf eine initiale Bindung von Ca durch Chondromucoproteine hinweist. Die Ca/P-Verhältnisse proliferierenden Knorpels waren jedoch bei Verkalkungsbeginn viel tiefer (ca. 1.50). Diese nahmen allerdings mit fortschreitender Mineralisierung stetig zu. In der endochondralen Verkalkungsphase fanden markante Veränderungen in der Zusammensetzung des organischen Anteils statt. Basierend auf der Hydroxyprolinanalyse nahm der Collagengehalt in der knorpeligen Verkalkungsperiode fortschreitend ab, während er jedoch bei der Knochenbildung rasch zunahm. Die an Hand von Hexosamin- und Schwefelanalysen bestimmten Chondromucoproteingehalte ergaben Höchstwerte im proliferierenden Knorpel und fielen stetig ab mit zunehmender Verkalkung. Trotz der im hypertrophischen Knorpel schon weit fortgeschrittenen Verkalkung waren immer noch große Mengen an Mucopolysacchariden vorhanden. Die Schwefel/Hexosamin-Verhältnisse zeigten eine minimale Abnahme in den frühen Verkalkungsphasen, nahmen jedoch markant zu bei fortschreitender Mineralisation. Der Sialinsäurespiegel war im Epiphysenknorpel, verglichen mit demjenigen des Ruheknorpels oder Knochens, erhöht. In der knorpeligen Verkalkungsphase nahmen die Lipide rasch zu, während jedoch die Werte des vollständig ausgebildeten Knochens stark vermindert waren. Die Bedeutung dieser Ergebnisse wird besprochen.
    Notes: Abstract A dissection procedure has been devised to permit zonal analysis of the epiphyseal plate of fetal calf leg bones. Samples of whole and washed tissue from the various zones were analyzed for their content of electrolyte and organic constituents, as well as for density, ash and moisture. Results showed that as ash content and density increased, water content decreased. Ash levels in calcifying cartilage zones were unexpectedly high. When expressed as a percentage of dry weight, washed calcified cartilage had the highest content of any zone. In the early stages of the mineralization of cartilage, Na content (mmoles/l of fresh tissue) decreased as Ca and inorganic P increased. Magnesium levels increased as calcification proceeded, but only at a fraction of the rate of Ca and P. Ratios of Ca/inorganic P were highest in resting cartilage (non-differentiated hyaline cartilage), suggesting an initial binding of Ca to chondromucoproteins. However, at the onset of calcification in proliferating cartilage, Ca/P ratios were much lower (ca. 1.50), but gradually increased with advancing mineralization. Marked changes occurred in the composition of the organic phase during endochondral calcification. As determined by hydroxyproline analysis, collagen content progressively decreased during cartilaginous calcification, but increased rapidly during bone formation. As determined by hexosamine and sulfur analysis, chondromucoproteins were at highest levels in proliferating cartilage and decreased steadily as calcification increased. However, although calcification was already well advanced in hypertrophic cartilage, large amounts of mucopolysaccharide still were present. Sulfur/hexosamine ratios showed a slight decline during the early stages of calcification, but increased markedly with further mineralization. Sialic acid levels were elevated in epiphyseal cartilage over those in resting cartilage or bone. Lipids increased rapidly during cartilaginous calcification, but were greatly reduced in fully-formed bone. The significance of these findings is discussed.

    Description / Table of Contents: Résumé Les effects d'une dose de 1 mg de vitamine D3 (40 000 Unités,) administrée par voie intra-veineuse, ont été étudiés sur l'histologie osseuse et le métabolisme du phosphore chez 19 sujets contrôles dont l'histologie était normale et 28 malades présentant les caractères histologiques de l'ostéomalacie par carence vitaminique D. L'administration de la vitamine D n'a entraîné aucune modification histologique ou biologique significative chez les sujets contrôles. Mais chez les ostéomalaciques, il est apparu en moins de sept jours une augmentation très significative du front de calcification à l'interface tissue-ostéoïde-tissue-calcifié. Cette modification s'accompagnait d'une ascension progressive de la phosphatémie et de la réabsorption tubulaire du phosphore atteignant dans le même délai des valeurs normales.
    Abstract: Zusammenfassung Bei 19 Patienten mit normaler Knochenhistologie und bei 28 Patienten mit histologisch gesicherter Osteomalacie wurde die Wirkung von 1 mg Vitamin D3 (40000 Iv) i.v. auf die Knochenhistologie und den Phosphatmetabolismus untersucht. Bei den Kontrollpatienten konnten keine signifikanten Änderungen nach Vitamin D festgestellt werden, wogegen die Osteomalacie-patienten innerhalb von 7 Tagen eine deutliche Zunahme der Verkalkungszone an der Grenze zwischen Osteoid- und Knochengewebe zeigten. Diese Änderung war von einer fortschreitenden Zunahme des Serum-Phosphates, verbunden mit einer gesteigerten renalen tubulären Rückabsorption des Phosphates begleitet; beide kehrten anschließend gleichzeitig wieder zur Norm zurück.
    Notes: Abstract The effect of a 1 mg dose of intravenous Vitamin D3 (40,000 i.u.) on bone histology and phosphate metabolism was investigated in 19 patients with normal bone histology and 28 patients with histological evidence of osteomalacia. No significant changes occurred in the control patients after Vitamin D but the patients with osteomalacia showed a marked increase, within seven days, in the proportion of osteoid having a calcification front. This was accompanied by a progressive rise in the serum phosphate, which was associated with an increase in the renal tubular reabsorption of phosphate to normal.

L'explication pourrait être qu'une barrière cellulaire ôte le calcium par préférence au strontium du fluide d'os ou bien qu'une accentuation précoce de la proportion Sr/Ca dans l'os se réfléchit dans les fractions de densité supérieure.
    Abstract: Zusammenfassung Das Verhältnis Sr/Ca im Knochen ist annähernd dasselbe wie es im Serum vorliegt, während in einer physiologischen Lösung von pulverisiertem Hydroxyapatit das Sr zugunsten des Ca benachteiligt wird. Es wurde festgestellt, daß das Sr/Ca-Verhältnis von Fraktionen verschiedener Dichte von pulverisierten Rattentibiae mit zunehmender Dichte anstieg, und zwar bis zu einem Wert, der etwas höher lag als der im Serum beobachtete. Dies ließe sich so erklären, daß eine Zellbarriere beim Entzug aus der Knochenflüssigkeit Ca dem Sr vorzieht; oder aber daß sich eine frühzeitige Betonung des für den Knochen typischen Sr/Ca-Verhältnisses in den Fraktionen höherer Dichte widerspiegelt.
    Notes: Abstract The Sr/Ca ration in bone is approximately the same as in serum, whereas hydroxyapatite powders suspended in physiological solutions discriminate against strontium. It was found that the Sr/Ca ratio of various density fractions of powdered compact bone of rat tibia increased with increasing density to a value slightly higher than that observed in the serum. The explanation could be that a cellular barrier removes calcium preferentially to strontium from the bone fluid, or that early accentuation of the bone Sr/Ca ration is reflected in the higher density fractions.

Le rapport entre l'ingestion de calcium et la minéralisation vertébrale, mesurée par densitométrie radiographique quantitative, est fiable mais cependant significatif. Chez 53 personnes, atteintes d'ostéoporose, comparées à un nombre égal d'individus, d'âges similaires, les valeurs de la minéralisation vertébrale sont 60 pour cent plus faibles que celles du groupe témoin. L'ingestion totale moyenne de calcium est plus faible de 21 per cent chez les ostéoporotiques. Chez les sujets, à ingestion unique de calcium, les patients du groupe témoin ingèrent preque le double de calcium que le groupe des ostéoporotiques. Une diminution moyenne de l'ingestion calcique est notée avec l'âge. Il semble qu'il existe une diminution de l'absorption calcique avec l'âge, l'ostéoporose ou les deux, ainsi qu'un besoin plus grand en calcium chez les sujets âgés pour assurer l'équilibre en calcium. Indépendamment de l'homéostase calcique, un équilibre négatif en calcium augmente la résorption osseuse, et diminue l'ostéogenèse. De nombreux facteurs interviennent dans l'étiologie et la pathogénie de l'ostéoporose: les résultats de cette étude indique que la présence de calcium alimentaire est l'un de ces facteurs.
    Abstract: Zusammenfassung Die tägliche Calciumeinnahme wurde anhand einer Befragung von 398 Personen im Alter von 15–90 Jahren geschätzt. Die Korrelation der Calciumeinnahme und der Mineralisation der Wirbelsäule, wie sie röntgenologisch durch quantitative Messung der Dichte festgestellt wurde, war niedrig, aber durchwegs signifikant. Bei 53 Osteoporose-Patienten, die mit 53 Kontrollpersonen gleichen Alters verglichen wurden, waren die Mineralisationswerte der Wirbelsäule um 60% niedriger als jene der Kontrollgruppe, und die durchschnittlich geschätzte Gesamt-Calciumeinnahme der Osteoporotiker war um 21% tiefer. Von den Exploranden, die Angaben über eine gleichmäßige Calciumaufnahme machten, nahmen die Kontrollpersonen beinahe doppelt so viel Calcium ein als diejenigen mit Osteoporose. Im Durchschnitt wurde eine Verminderung der Calciumeinnahme mit fortschreitendem Alter festgestellt. Es ergibt sich deutlich, daß die Calciumabsorption mit dem Alter, bei Osteoporose oder in der Kombination dieser beiden Faktoren abnimmt, und daß eine größere Calciumeinnahme bei älteren Personen nötig wäre, um eine positive Calciumbilanz aufrechtzuerhalten. Ungeachtet der Komplexität der Calciumhomöostase, kann eine negative Calciumbilanz schließlich dazu führen, daß mehr Knochen resorbiert als gebildet wird. Demzufolge sollte bei erkanntem Bedürfnis eine adäquate Calciumeinnahme gesichert sein. Es ist eine Tatsache, daß viele Faktoren an der Ätiologie und Pathogenese der Osteoporose beteiligt sind; die Resultate dieses Berichtes unterstützen die Wahrscheinlichkeit, daß das Calciumangebot in der Nahrung einer dieser Faktoren ist.
    Notes: Abstract Lifetime daily calcium intake was estimated through interview of 398 individuals from 15 to 90 years of age. The correlation of calcium intake with vertebral mineralization as determined by quantitative radiographic densitometry was low but persistently significant. In 53 persons with osteoporosis matched by age with 53 individuals from the control group, vertebral mineralization values were 60% lower than those of the control group, and the mean estimated total calcium intake in osteoporotics was 21% lower. In those persons reporting a single lifetime calcium intake, the control patients ingested almost twice as much calcium as those with osteoporosis. A mean decrease in calcium intake with advancing years has been shown. Evidence points to a decrease in calcium absorption with age, osteoporosis, or both, as well as a greater need for calcium intake in the elderly to maintain a positive calcium balance. Regardless of the intricacies of calcium homeostasis, a negative calcium balance leads eventually to greater bone resorption than formation, hence the rationality of insuring an adequate calcium intake with recognized nutritional needs. Evidence suggests that many factors are involved in the etiology and pathogenesis of osteoporosis; the data in this report support the likelihood that availability of calcium in the diet is one of them.

    Description / Table of Contents: Résumé Une méthode quantitative pour étudier les effets précoces des hormones et autres agents sur la mobilisation du calcium osseux a été mise au point. Des moitiés de calottes craniennes de souris de 6 jours, ayant reçu du45Ca quatre jours auparavant, sont explantéesin vitro et placées dans des récipients séparés, en milieu liquide; une moitié sert de témoin, l'autre moitié est utilisée pour l'expérimentation. Ce mode d'action dans le temps, de chaque produit testé, est déterminé en prélevant aseptiquement de petits échantillons dans le milieu et en dosant l'isotope. L'hormone parathyroidienne et la vitamine A provoquent une résorption osseuse étendue ainsi qu'un passage augmenté du45Ca des os traités dans le milieu, au bout de 2 heures, si on les compare aux témoins. La calcitonine inhibe rapidement la mobilisation du45Ca des os résorbés: l'intervalle de temps est similaire à celui obtenuin vivo en abaissant le calcium sérique. Nos résultats indiquent que les agents, qui induisent la résorption osseuse, augmentent à la fois le nombre des ostéoclastes et l'efficacité des cellules existantes, en ce qui concerne la mobilisation du calcium. La calcitonine inhibe la libération du45Ca des explants vivants, maintenus dans un milieu contrôle. Cette réduction est attribuée à la suppression de la résorption endogène, en cours au moment de la transplantation: un échange isotopique est toujours observé, comme au niveau des explants sur vivants.
    Abstract: Zusammenfassung Eine quantitative Methode zur Bestimmung der Früheffekte von Hormonen oder andern Wirkstoffen auf die Wanderung von Calcium in den oder aus dem Knochen wurde entwickelt. Calvarien-Hälften von 6 Tagen alten, 4 Tage früher mit45Ca markierten Mäusen werden explantiert und in getrennten Schalen mit flüssiger Nährlösungin vitro belassen. Die eine Hälfte dient als Kontrolle, die andere wird für das Experiment eingesetzt. Anhand kleiner Proben, die den Medien aseptisch entnommen und auf ihren Isotopengehalt geprüft werden, kann die Wirkung der verschiedenen Agentien im Verlaufe der Zeit beobachtet werden. Sowohl Parahormon als auch Vitamin A verursachen eine ausgedehnte Knochenresorption und es wurde, im Vergleich zu den Kontrollen, innerhalb von 2 Std eine gesteigerte Abgabe von45Ca aus den behandelten Knochen in das Medium nachgewiesen, Die Freisetzung von45Ca aus resorbierenden Knochen wird durch Calcitonin rasch inhibiert. Wirkungsweise und Verlauf bestehen, wiein vivo, in einer Senkung des Serum-Calciums. Unsere Resultate zeigen, daß Wirkstoffe, welche eine Knochenresorption veranlassen, sowohl die Zahl der polynucleären Osteoklasten ansteigen lassen als auch die vorhandenen Zellen zur vermehrten Calcium-Mobilisation anregen. Calcitonin dagegen verhindert die Bildung neuer, polynucleärer Osteoklasten wie auch die Mobilisation von Knochenmineral durch die vorhandenen Osteoklasten. Calcitonin wirkt hemmend auf die Abgabe von45Ca von lebenden Explantaten, die in Kontrollmedium kultiviert werden. Diese Abnahme wird der Unterdrückung der endogenen Resorption zugeschrieben, welche im Moment der Knochenexplantation im Gange ist; der Austausch des Isotopes findet weiterhin statt, wie dies bei einem toten Explantat der Fall ist.
    Notes: Abstract A sensitive method has been developed for studying the early effects of hormones and other agents on the movement of calcium into and out of bone. Half-calvariae from 6-day-old mice that have been pulsed four days previously with45Ca, are explanted into separate dishes of liquid medium and maintainedin vitro; one half serves as control and the other for experimentation. The time course of action of any agent is followed by removing small samples aseptically from the media and analysing for isotope. Both parathyroid hormone and vitamin A cause extensive bone resorption, and as compared with controls an increased release of45Ca from the treated bones into the medium can be detected within 2 hours. Calcitonin rapidly inhibits the release of45Ca from resorbing bones; the time course is similar to that for its actionin vivo in lowering serum calcium. Our results indicate that agents that induce bone resorption increase both the number of multinucleate osteoclasts and the effectiveness of the existing cells in mobilising calcium. Calcitonin prevents the formation of new multinucleate osteoclasts, and also prevents existing osteoclasts from mobilising bone mineral. Calcitonin inhibits the release of45Ca from living explants maintained in control medium. This reduction is attributed to the suppression of the endogenous resorption that is in progress when the bones are explanted; exchange of isotope still occurs, as in a dead explant.

Fractionnement des extraits par chromatographie sur 2 p. 100 agarose ou par précipitation avec du NaCl (3M) donne une fraction constituée par du rRNA et characterisée par la composition nucléotidique. Cette fraction correspond à 70 p. 100 du RNA extrait à pH 7,6 et à 65–75 p. 100 du RNA extrait à pH 9,0 de l'os regénérant et à 85 p. 100 du RNA extrait de l'os normal. 10 p. 100 du RNA extrait de l'os regénérant et 5 p. 100 du RNA extrait de l'os normal sont de poids moleculaire plus haut que le rRNA. Les RNA de poids moleculaire plus bas que le rRNA représentent 20 p. 100 du RNA extrait à pH 7,6 et 25–35 p. 100 du RNA extrait à pH 9,0 de l'os regénérant et 10 p. 100 du RNA extrait de l'os normal. Les résultats de centrifugation en gradients de saccharose entre 5 et 20 p. 100 correspondent aux résultats des études de fractionnement.
    Abstract: Zusammenfassung Wird die RNS aus Kaninchenknochengewebe, das sich in der Regenerationsphase nach standardisierter Osteotomie befindet, mit wäßrigen Phenollösungen, extrahiert, so werden bei einem pH-Wert des Extraktes von 7,6 70–75% des totalen RNS-Gehaltes erhalten, gegenüber 10–15% bei einem, pH-Wert von 9,0. Von der normalen Kaninchenradiusdiaphyse ist die Ausbeute an RNS 50–70% bei einer Extraktion im Bereich von pH 7,6 und nur wenige Prozente bei pH 9,0. Eine Fraktionierung der Extrakte mittels Gelchromatographie auf 2% Agarose und durch Behandlung mit 3M Natriumchlorid ergab eine Fraktion, die der rRNS entspricht und durch ihre Nucleotidzusammensetzung charakterisiert ist. Diese Fraktion erreicht bei regenerierenden Knochen 70% der RNS in den bei pH 7,6 gewonnenen Extrakten, 65–75% in jenen bei pH 9,0. während bei normalen Knochen die Ausbeute an RNS 85% bei pH 7,6 betrug. Im Extrakt von pH 7,6 waren beim regenerierenden Knochen 10% und bei normalen Knochen 5% der RNS von höherem Molekulargewicht als rRNS. Kleiner molekular als rRNS waren 20% der RNS in den Extrakten des regenerierenden Knochens bei pH 7,6 und 25–35% bei pH 9,0, während die entsprechenden Extrakte beim normalen Knochen nur 10% dieser kleinmolekularen RNS enthielten. Das Sedimentationsbild mit 5–20% linearen Saccharosegradienten entsprach den Ergebnissen der Fraktionierungsversuche.
    Notes: Abstract Extraction of RNA with aqueous phenolic solutions of pH 7.6 and pH 9.0 yielded 70–75% of the total RNA content of rabbit bone tissue, regenerating after standardized osteotomies, in the pH 7.6 extract and 10–15% in the pH 9.0 extract. From normal rabbit radius diaphysis, 50–70% of the RNA was extracted at pH 7.6 and oly a few per cent at pH 9.0. Fractionation of the extracts by gel chromatography on 2% agarose and by treatment with 3M sodium chloride gave a, fraction corresponding to rRNA and characterized by base composition. This fraction amounted to 70% of the RNA in the pH 7.6 extracts, to 65–75% of the RNA in pH 9.0 extracts from regenerating bone and to 85% of the RNA in pH 7.6 extracts from normal bone. 10% of the RNA in pH 7.6 extracts from regenerating bone and 5% of that in extracts from normal bone were of larger size than rRNA. 20% of the RNA in pH 7.6 extracts and 25–35% in pH 9.0 extracts from regenerating bone and 10% in those from normal bone were of lower size than rRNA. The sedimentation pattern in 5–20% linear sucrose gradients corresponded to the results of the fractionation studies.

    Description / Table of Contents: Résumé L'objet de cette étude a été de déterminer l'effet du mode de préparation (par meulage) sur la largeur des raies de diffraction de l'émail. La préparation d'émail, par meulage, en utilisant divers procédés ainsi qu'une pièce à main dentaire conventionnelle provoque un élargissement des pics obtenus (002, 211, 200 et 202) lorsqu'on la compare avec de la poudre d'émail, obtenue par meulage à l'aide de billes. L'élargissement des raies n'est pas observé lorqu'un monocristal d'hydroxylapatite est meulé à l'aide d'un diamant fin. En général, l'élargissement est moins important, lorsque le meulage est effectué à l'aide de turbines dentaires. L'importance du meulage dépend de façon variable d'un ou plusieurs des facteurs suivants: rugosité des instruments coupants, vitesse de meulage, direction de meulage, et la présence ou l'absence d'eau. Le meulage prolongé par billes de l'émail provoque aussi un élargissement dans les mêmes conditions, cependant, l'os n'est pas endommagé. Ces résultats indiquent que l'émail est plus sensible que l'hydroxylapatite et l'os. L'élargissement de raies peut être dû soit à une déformation de la maille cristalline, soit à une diminution de taille des cristaux.
    Abstract: Zusammenfassung Es wurde eine Untersuchung durchgeführt, um den Einfluß der Probenvorbereitung (Zerreibungsmethode) auf die Breite des Linienprofil-Querschnittes von Zahnschmelz zu bestimmen. Gewinnung von Zahnschmelz mit den verschiedenen Schneidinstrumenten einer konventionellen Bohrmaschine verursachte eine Verbreiterung aller untersuchten Peaks (002, 211, 200 und 202) im Vergleich zum gleichen Schmelz, der mit dem Rosenbohrer zerrieben wurde. Eine Verbreiterung der Linie konnte nicht beobachtet werden, wenn ein einzelner Kristall von Hydroxyapatit mit einem ganz feinen Diamanten zerrieben wurde. Im allgemeinen war die Verbreiterung weniger ausgesprochen, wenn die hochtourige Bohrtechnik zur Anwendung kam. Das Ausmaß der Verbreiterung, das durch Zahnbohrer verursacht wurde, war abhängig von einem oder mehreren der folgenden Faktoren: Rauheit des Schneidinstrumentes, Zerreibungsgeschwindigkeit, Zerreibungsrichtung und das Vorhandensein oder Fehlen von Wasser. Verlängerte Zerreibung von Schmelz mit dem Rosenbohrer verursachte ebenfalls eine Verbreiterung. Unter identischen Bedingungen blieb der ausgeglühte Knochen jedoch unversehrt. Diese Beobachtungen zeigen, daß Schmelz für Zerreibungsschäden anfälliger ist, als Hydroxyapatitkristalle oder ausgeglühter Knochen. Die eigentliche Ursache der Linienverbreiterung kann entweder eine Schädigung infolge Distortion des Gitters oder eino Reduktion der Größe der individuellen Kristalle sein.
    Notes: Abstract A study was conducted to determine the effect of sample preparation (grinding method) upon breadth of the diffraction profile of enamel. Collecting enamel by grinding with various cuttin tools in the low-speed dental handpiece caused broadening of all peaks (002, 211, 300 and 202) examined, compared to ball, ground, counter-part enamel. Line broadening was not observed when a single crystal of mineral hydroxyapatite was ground with a very fine diamond. In general, broadening was less pronounced with the high-speed air turbine technique. The amount of broadening caused by dental burs depended upon one or more of the following factors: coarseness of cutting instrument, grinding speed, grinding direction, and the presence or absence of water. Prolonged ball grinding of enamel also caused broadening; under identical conditions, however, annealed bone remained undamaged. These findings indicate that enamel is more sensitive to grinding damage than the mineral hydroxyapatite crystal or annealed bone. The actual cause of line broadening may be either strain due to lattice distortions or a reduction in size of individual crystallites.

Le nombre d'ostéoclastes dans l'os médullaire différait peu des valuers témoin pendant les périodes de déficience et de réplétion subséquente, sauf pour une augmentation significative au premier jour de déplétion. L'effect histologique le plus important dans l'os médullaire était une augmentation marquée en nombre d'ostéoblastes aux troisième, cinquième, et un peu moins au septième jours de déplétion. Le nombre d'ostéoblastes était en corrélation positive avec la teneur de l'os médullaire en ostéoide et négative avec son degré de calcification. L'activité de l'os médullaire en phosphatase alcaline augmentait avec la longueur de la déficience calcique. Un jour après le retour des pondeuses à une diète contenant 3,2% de calcium, la calcification de l'os médullaire avait augmenté de façon significative, le nombre d'ostéoblastes avait diminué au niveau ou au-dessous du niveau de contrôle et l'activité de la phosphatase alcaline avait baissé considérablement. L'importance de ces résultats est discutée par rapport au controle des populations des cellules dan l'os et au rôle de l'os médullaire.
    Abstract: Zusammenfassung Die Zahl der Knochenzellen und die Zusammensetzung des Femurs von Legehennen wurden während einer siebentägigen Calciumentzugsperiode (Calciumgehalt des Futters 0,13%) und einer siebentägigen Ersatzperiode (Calciumgehalt des Futters 3,2%) untersucht. Histologisch zeigte nur die Cortex eindeutige Knochenresorption und osteoklastische Aktivität. Abgesehen von einer signifikanten Zunahme am 1. Tag des Calciumentzuges, variierte die Zahl der Osteoklasten im Markknochen sowohl während der Entzugs- als auch während der nachfolgenden Ersatzperiode wenig. Die wichtigste histologische Änderung im Markknochen bestand in einer starken Zunahme in der Zahl der Osteoblasten am 3., 5. und etwas weniger am 7. Tag der Entzugsperiode. Die Zahl der Osteoblasten zeigte eine positive Korrelation mit dem Osteoidgehalt des Markknochens und eine negative mit dem Grade seiner Verkalkung. Die Aktivität der alkalischen Phosphatase im Markknochen war desto größer je länger den Hennen die calciumarme Ration verfüttert worden war. Die Wiederverabreichung der Ration, welche 3,2% Calcium enthielt, verursachte innerhalb eines Tages eine signifikante Zunahme in der Verkalkung des Markknochens, ein Absinken der Osteoblastzahl auf die Kontrollwerte oder unter sie und eine drastische Verringerung der alkalischen Phosphataseaktivität. Die Bedeutung dieser Ergebnisse in bezug auf die Kontrolle des Knochenzellenbestandes und auf die Funktion des Markknochens wird diskutiert.
    Notes: Abstract Bone cell activity and the composition of the femur of laying hens were studied during 7 days of calcium depletion on a 0.13% calcium diet and 7 days of calcium repletion on a 3.2% calcium diet. Histologically, only cortical bone showed clear signs of bone resorption and osteoclastic activity during the depletion period. The number of osteoclasts in medullary bone varied little from control values throughout both calcium depletion and repletion, except for a significant increase on the first day of depletion. The major histologicalchange in medullary bone was a marked increase in the number of osteoblasts on the third, fifth and, to a lesser extent, seventh, day of depletion. The number of osteoblasts in medullary bone was positively correlated with its osteoid content and negatively correlated with its degree of calcification. Alkaline phosphatase activity of medullary bone increased with the time the hens had been on the calcium-deficient diet. Returning the hens to the 3.2% calcium ration caused, within one day, a significant increase in medullary bone calcification, a decrease of osteoblast numbers to, or below, control levels, and a drastic reduction in alkaline phosphatase activity of medullary bone. The significance of these findings in relation to the control of bone cell populations and the functions of medullary bone is discussed.

    Abstract: Zusammenfassung Die Resultate einer Verfütterung von hohen stabilen Strontiumdosen an Hühnern werden erläutert. Strontiumzusätze zur Nahrung in der Höhe von 3000–50000 p.p.m. führten zu einer signifikanten Zunahme des Strontiumgehaltes der Tibia und verursachten keine wesentlichen Änderungen des Calciumgehaltes. Die Calciumkonzentration im Plasma verminderte sich, wenn der Nahrung ansteigende Strontiummengen zugegeben wurden. Mit zunehmendem Strontiumzusatz zur Nahrung zeigte der Calciumgehalt der Eischale eine fortlaufende Abnahme, während sich der Strontiumgehalt entsprechend erhöhte. Durch Röntgendiffraktionsanalysen der stark Strontium-haltigen Knochen und Eischalen konnte nicht festgestellt werden, in welcher Form das Strontium abgelagert wurde.
    Notes: Abstract The results of feeding high dietary levels of stable strontium to hens are reported. Dietary levels of strontium from 3,000 p.p.m. to 50,000 p.p.m. showed a significant increase in strontium content of the tibia bone and essentially no change in the calcium content. Plasma calcium concentration was shown to decrease with increasing dietary strontium treatment. Egg shell calcium showed a progressive decrease with increasing dietary strontium treatment, whereas the strontium content has a corresponding increase. X-ray diffraction analyses of bones and shells containing large amounts of strontium were unsuccessful in evaluating the from in which strontium was deposited.

Du Triton a servi pour le scintillant liquide dans le compteur de scintillation liquide. Un procédé de détermination des deux isotopes dans le meme echantillon a été décrit. Différents critéres on été appliques a chacque étape afin d'établir la validité et les limites d'application de la méthode.
    Abstract: Zusammenfassung Eine Methode ist beschrieben, die die quantitative und radioaktive Bestimmung von45Ca und L-[-L14C] Ascorbinsäure in Knochen von Tieren, denen die beiden Isotopen verabreicht worden sind, erlaubt. Calcium und Ascorbinsäure werden quantitativ mit 6% Trichloressigsäure ausgezogen, wobei die Ascorbinsäure erhalten bleibt. Quantitative Analysen für Calcium und Ascorbinsäure werden nach bekannten Methoden in einem Teil des Extraktes bestimmt und die Bestimmung der Radioaktivität in einem anderen Anteil mit Hilfe eines Flüssigkeits-Szintillations-Zählers durchgeführt. Dem flüssigen Szintillanten wird Triton zugesetzt. Diese Versuchsanordnung ermöglicht außerdem die gleichzeitige Bestimmung der beiden Isotopen im Knochen-Extrakt. Die Methode wurde sorgfältig auf ihre Verwendbarkeit ausgewertet.
    Notes: Abstract Methods were developed for the determination of45Ca plus L-[1-14C] asorbic acid in bones of animals following administration of both isotopes. Asorbic acid and calcium were extracted quantitatively with 6% trichloracetic acid from fresh bone under such mild conditions that asorbic acid was not destroyed. The chemical determinations of calcium and ascorbic acid according to standard procedures were performed in aliquots of the same extract. The determination of the radioactivity of calcium and ascorbic acid were then carried out in aliquots of the same extract. A Triton-containing liquid scintillation fluid was employed for the radioassay of45Ca and L-[1-14C] ascorbic acid in the liquid scintillation spectrometer. A procedure allowing the determination of both isotopes in the same extract is described. A number of criteria were applied to each step, in order to determine the validity and limitations of the procedure.

    Description / Table of Contents: Résumé Une méthode nouvelle de décalcification histologique utilisant des solutions aqueuses de sulfate de chrome basique (III) est appliquée à des coupes fines de dentine humaine adulte. L'observation en microscopie électronique montre une bonne conservation structurale dans les régions extra-cellulaires. L'aspect de la matrice péricanaliculaire est particulièrement étudié. On y reconnait plusieurs couches bien individualisées. Une bonne concordance est notée entre les résultats obtenus par microscopie optique et électronique, permettant d'établir une hypothèse nouvelle sur la formation de la dentine péricanaliculaire.
    Abstract: Zusammenfassung Eine kürzlich entwickelte Methode zur histologischen Entkalkung mittels wäßriger Lösungen von basischem Chrom III-Sulfat wird an dünnen Schliffen von menschlichem Dentin Erwachsener angewandt. Darauffolgende Untersuchungen am Elektronenmikroskop zeigten eine sehr gute Wiedergabe der morphologischen Einzelheiten in den Extrazellulär-Regionen. Besonders beachtet wurde das Aussehen der peritubulären Matrix, innerhalb welcher gut differenzierte Schichten erkannt werden konnten. Eine wesentliche Übereinstimmung bestand zwischen Elektronenmikroskopie und kürzlich durchgeführten Untersuchungen an derselben Matric unter dem Mikroskop. Diese Methode ermöglicht es, eine neue Hypothese über die Bildung des peritubulären Dentins aufzustellen.
    Notes: Abstract A recently-developed method for histological decalcification using aqueous solutions of basic chromium (III) sulphate has been applied to thin sections of adult human dentine. Subsequent studies in the electron microscope show a very good retention of morphological detail in the extracellular regions. Special attention was given to the appearance of the peritubular matrix within which well-differentiated layers were recognised. There was substantial agreement between electron microscopy and recent studies of the same matric under the light microscope, allowing a novel hypothesis for the formation of the peritubular dentine.

    Description / Table of Contents: Résumé Une méthode quantitativein vitro est utilisée pour étudier l'efficacité relative des préparations de calcitonine, provenant de diverses espèces. La méthode est basée sur le fait que la calcitonine inhibe de façon nette la libération de45Ca de moitiés de calottes craniennes de souris, âgées de 6 jours, ayant été marquées auparavant; cet effect est lié à la suppression de la résorption endogène, qui se déroule, lorsque les os sont explantés et qui se continuein vitro en l'absence de calcitonine. Des différences nettes de l'effect d'inhibition des préparations de calcitonine ont été trouvées, qu'elles proviennent de corps ultimobranchiaux ou de thyroides de mammifères. La calcitonine humaine et la calcitonine porcine, standard A, se comportent de façon identique et sont environ dix fois plus actifs que la calcitonine porcinek standard B, en se basant sur les critéres unitaires utilisés au cours de cette étude chez le rat. Les calcitonines de corps ultimobranchiaux sont plus efficaces à doses faibles, pendant des périodes plus longues, que les préparations thyroidiennes et elles sont au moins dix fois plus efficaces que la calcitonine porcine standard A et cent fois plus actives que la calcitonine porcine standard B. Les explants traités par la calcitonine libère l'isotope dans le milieu d'une manière identique à celle de l'os mort, tant que la calcitonine reste active: pour la calcitonine de saumon, on observe pendant 20 heures un échange d'isotope entre l'os vivant, traité avec ce produit et le calcium du milieu, ainsi que cela se produit pour des fragments d'os congelé. Ces résultats sont utiles pour des études quantitatives, devant déterminer le mode d'action d'autres produits influençant la résorption et l'apposition osseuse. L'adjonction de calcitonine permet, en effet, de mesurer l'échange d'isotope libéré de l'os, ainsi traité à tous les stades, sans tuer les tissus ou sans avoir recours à des inhibiteurs spécifiques du métabolisme.
    Abstract: Zusammenfassung Ein quantitativerin vitro-Versuch wurde angewandt, um die relative Wirksamkeit von Calcitonin-Präparaten aus verschiedenen Spezies zu untersuchen. Der Versuch beruht auf der Feststellung, daß Calcitonin einen ausgeprägten Hemmeffekt auf die Freisetzung von45Ca aus markierten Calvarien-Hälften von 6tägigen Mäusen hat. Dieser Effekt kommt wegen der Unterdrückung der endogenen Resorption zustande, die im Moment der Knochenexplantation im Gange ist, undin vitrobei Abwesenheit von Calcitonin weiter fortschreitet. Auffallende Unterschiede des Hemmeffektes konnten bei Calcitonin-Präparaten sowohl von Ultimobranchialkörpern als auch von Säuger-Thyreoideae festgestellt werden. Menschliches Calcitonin sowie Standard A Schweine-Calcitonin verhalten sich gleich und sind ungefähr 10mal wirksamer als Standard B Schweine-Calcitonin, ausgedrückt in Einheiten bezogen auf den Rattenversuch. Die von Ultimobranchialkörpern stammenden Calcitonin-Präparate sind wirksamer in niedrigerer Dosierung und während einer längeren Zeitspanne als die Thyreoidea-Präparate. Sie wirken zudem mindestens 10mal stärker als Standard A Schweine-Calcitonin und 100mal stärker als Standard B Schweine-Calcitonin. Explantate, welche mit Calcitonin behandelt wurden, geben das Isotop auf die gleiche Weise wie totes Knochengewebe in das Medium ab, solange das Calcitonin wirksam ist. Für Calcitonin vom Lachs besteht eine Zeitpsanne von 20 Std, während welcher die lebenden, mit diesem Präparat behandelten Knochen das Isotop mit dem Calcium des Medium austauschen, in gleicher Weise wie durch Einfrieren und Auftauen getöteten Knochens. Diese Methode wird vermutlich sehr nützlich sein für quantitative Untersuchungen über den Aktionsmodus anderer Stoffe, welche Knochenresorption und-wachstum beeinflussen, da der Calcitonin-Zusatz es ermöglicht, die Austauschkomponente des aus dem behandelten Knochen freigesetzten Isotopes jederzeit zu messen, ohne daß dabei Gewebe zerstört oder weniger spezifische Inhibitoren des Metabolismus herangezogen werden müssen.
    Notes: Abstract Anin vitro method has been used to investigate the relative effectiveness of calcitonin preparations from different species. It is based on the finding that calcitonin has a marked inhibitory effect on the release of45Ca from prelabelled half-calvariae from 6-day-old mice; this effect is due to the suppression of the endogeneous resorption that is in progress when the bones are explanted, and which continuesin vitro in the absence of calcitonin. Striking differences have been found in the inhibitory effect of calcitonin preparations from either ultimobranchial bodies or from mammalian thyroids. Human calcitonin and porcine standard A calcitonin behave similarly and are about ten times more potent than porcine standard B calcitonin in terms of units based on the rat assay. The calcitonins of ultimobranchial origin are more effective at lower doses for a longer period of time than the thyroid preparations, and are at least ten times more potent than porcine standard A calcitonin or 100 times more potent than porcine standard B. Explants treated with calcitonin release isotope into the medium in a similar manner to that of dead bones, as long as the calcitonin remains effective; for salmon calcitonin there is a twenty-hour period in which the release of isotope from living bones treated with this preparation exchange isotope with the calcium in the medium like frozen-thawed bone. This finding should be very useful in quantitative studies on the mode of action of other compounds that influence bone resorption and accretion, since the addition of calcitonin will enable the exchange component of the isotope released from treated bones, to be measured at any stage without killing the tissue or resorting to less specific inhibitors of metabolism.

    Description / Table of Contents: Résumé L'os médullaire des poules au moment de la ponte ainsi que l'os néoformé de coqs soumis à des substances oestrogéniques, ont été examinés par les méthodes de l'histologie, de l'histochimie, de la microradiographie et de l'alpharadiographie afin de comparer la résorption naturelle à celle produite par la parathormone. Chez les pondeuses, la résorption a été provoquée par la formation de la coquille; chez les coqs, elle s'est produite à la suite de la diminution du taux des oestrogènes. La résorption naturelle de l'os médullaire a été marquée par un accroissement de la basophilie, de l'azurophilie et de la metachromasie, par la diminution graduelle de la densité organique et minérale. Ces modifications ont été observées d'abord dans la portion distale de l'os médullaire; elles se sont progressivement propagées à la région sous corticale. L'extrait parathyroidien semble avoir favorisé tous ces phénomènes, en stimulant l'ostéolyse ostéocytaire et l'ostéoclasie. Il ne nous a pas été possible cependant de nous rendre compte si les ostéoclastes ne se sont attaqués qu'aux portions des travées déja modifiées par l'ostéolyse. Ces résultats concordent avec l'idée que la résorption normale de l'os médullaire de la poule au temps où la coquille de l'oeuf se dépose, est déclanchée par la parathormone. Il en est de même chez le coq à la suite du retrait des oestrogènes.
    Abstract: Zusammenfassung Die Substantia spongiosa der Femora von Legehennen und von Hähnen die mit Östrogen behandelt waren, wurde während spontaner und künstlicher, mit Extrakten von Epithelkörperchen induzierter, Resorption einer histologischen, histochemischen, alpharadiographischen und mikroröntgenographische Untersuchung unterworfen. Die natürliche Resorption wurde bei den Hennen durch die Eischalenproduktion und bei den Hähnen durch Östrogenentzug hervorgerufen. Die natürliche Resorption war durch erhöhte Basophilie, Azurphilie und Metachromasie, sowohl als auch durch verminderter alpharadiographischer und mikroröntgenographischer Dichte der Trabeculae der Substantia spongiosa charakterisiert. Diese Veränderungen wurden zuerst in den zentralen, dem Cavum medullare nächstgelegenen, Teilen der Spongiosa wahrgenommen. Später waren auch die peripheren Teile, mit geringerer Osteocytenkonzentration, betroffen. Diese Veränderungen konnten durch Gaben von Epithelkörperchenextrakten verstärkt werden. Die Resorption der Spongiosa wird durch osteocytische Osteolyse und Osteoklasie bewirkt. Es konnte aber nicht entschieden werden ob diese beiden Prozesse gleichzeitig stattfinden, oder ob die Osteoklasten die Trabeculae erst angreifen, nachdem sie bereits teilweise durch Osteolyse abgebaut worden sind. Diese Resultate stimmen mit der Hypothese überein, daß die natürliche Resorption von Spongiosa während der Eischalenproduktion bei Hennen und nach Entzug von Östrogen bei Hähnen, durch Epithelkörperchenhormonen bedingt ist.
    Notes: Abstract The medullary bone in the femora of laying hens and of oestrogen-treated cocks has been examined by histological, histochemical, alpharadiographic and microroentgenographic techniques while undergoing both natural resorption and resorption induced by injection of parathyroid extract. In the hens, natural resorption was brought about by egg-shell formation and in the cocks by withdrawal of oestrogen. Natural resorption was accompained by increasing basophilia, azurophilia and metachromasia and by decreasing alpharadiographic and microradiographic density of the trabeculae of the medullary bone. These changes were observed initially in the central region (towards the marrow cavity) and subsequently in the peripheral regions also, where the number of osteocytes per unit area of bone was less. Parathyroid extract enhanced all these effects. It appears that resorption of medullary bone was brought about by osteocytic osteolysis and by osteoclasis, but it was not possible to determine whether both processes occurred concurrently or whether the osteoclasts attacked the trabeculae only after they had been partially degraded by osteolysis. These results are consistent with the hypothesis that the natural resorption of medullary bone during egg-shell formation in hens and following the withdrawal of oestrogen in cocks is induced by parathyroid hormone.

La partie basale de la cellule présente de nombreux prolongements ramifiés. Ces prolongements contiennent de nombreuses mitochondries, ainsi que des invaginations de la membrane formant des vésicules qui migrent dans diverses parties de la cellule. Le noyau irrégulier est en position centrale. La région supranucléaire contient surtout de l'ergastoplasme, plusieurs appareils de Golgi et de nombreuses granules provenant de l'appareil de Golgi. Ces granules augmentent de taille et sont rejetées dans le sinus de la glande, où elles se modifient et se calcifient pour donner des sphérolithes. Des radioautographics glandulaires, obtenues 1/2 heure après injection de Ca45, montrent une localisation isotopique, située presque exclusivement dans les cellules; 4–24 heures après, l'isotope se localise dans les sphérolithes de la cavité glandulaire. Ces cellules semblent assurer deux fonctions liées à la production de sphérolithes: 1. absorption et transport de calcium de la région basale vers la surface cellulaire et 2. formation d'une matrice protéique qui se calcifiera.
    Abstract: Zusammenfassung Die aktivsezernierenden Zellen der calcifizierenden Drüsen vonLumbricus terrestris sind annähernd kuppelförmig gestaltet. Der untere Teil dieser Zellen besteht aus unzähligen ineinandergreifenden Zellfortsätzen. Diese Fortsätze enthalten zahlreiche Mitochondrien und membranöse Einstülpungen, welche Bläschen bilden, die zu den verschiedenen Regionen der Zelle wandern. Der Zellkern ist unregelmäßig und zentral angeordnet. Die supranukleäre Region ist zu einem großen Teil mit grobem endoplasmatischem Reticulum und mehreren Golgi-Komplexen sowie zahlreichen Granula, die von den Golgi-Komplexen stammen, ausgefüllt. Diese Granula vergrößern sich und werden in den Drüseninus ausgestoßen, wo sie schließlich reorganisiert und mineralisiert werden, um Sphärolithen zu bilden. 1/2 Std nach Injektion von Ca45 wurde die Drüse entfernt; eine Radioautographie zeigte, daß das Isotop beinahe ausschließlich in den Zellen lokalisiert war; 4–24 Std später befand sich das Isotopin den Sphärolithen der Drüsenhöhlung.     Notes: Abstract The actively-secreting cells of the calciferous glands ofLumbricus terrestris are roughly dome shaped. The basal part of the cell consists of numerous interdigitating cell processes. These processes contain numerous mitochondria and membranous infoldings, forming vesicles which migrate to various regions of the cell. The nucleus is irregular and centrally located. The supranuclear region is occupied to a great extent by the rough endoplasmic reticulum, several Golgi complexes and numerous granules derived from the Golgi complexes. These granules enlarge and are extruded into the gland sinus, where they eventually become reorganized and mineralized to give rise to spheroliths. Radioautographs of glands removed 30 min following injection of45Ca showed uptake of the isotope located almost exclusively in the cells; 4–24 h later the isotope was located in the spheroliths in the gland cavity. These cells apparently perform two functions necessary for the production of mineralized spheroliths, 1, absorption and transport of calcium from the basal region to the cell surface and 2, the elaboration of a protein matrix in which mineralization occurs.

    Description / Table of Contents: Résumé Divers agents chimiques, colorés, fluorescents et se localisant dans les os, à savoir la tétracycline, l'alazazine rouge S et le DCAF, ont été administré en série à des rats sevrés et on a mesuré le taux de la formation et de la résorption osseuse sur les coupes transversales du tiers supérieur de la diaphyse. Ici la formation osseuse périostale s'effectue progressivement avec peu de changement endostéal. Avec alimentation carnée, la croissance des rats est significativement restreinte pendant la première semaine, mais se rétablit ensuite. Bien qu'il y ait croissance des os, ceux-ci ne se minéralisent pas normalement et ils deviennent rapidement fragiles et amincis. La résorption osseuse est lente d'abord, puis s'accelère pendant 2–3 semaines pour atteindre un taux de 15μ par jour, après quoi elle se ralentit de nouveau. Bien que le taux de formation osseuse soit réduit, en comparaison avec celui des os normaux, la résorption s'effectue environ deux à trois fois plus rapidement que la croissance osseuse. Des études microradiographiques sur des rats à régime carné mais carencés en calcium ont permis la constatation suivante: tandis que la résorption s'effectue à la marge endostéale et que la formation osseuse a lieu sur l'aspect périostéal, la matière osseuse nouvellement formée est moins calcifiéc que chez les témoins.
    Abstract: Zusammenfassung Einige farbige, fluoreszierende, knochensuchende Chemikalien, z. B. Tetracyclin, Alazarin-Rot S und CDAF, wurden nacheinander an entwöhnten Ratten verabreicht, worauf man die Knochenbildungs- und Knochenresorptionswerte an hartgeschliffenen Schnitten des oberen Drittels der Diaphyse gemessen hat. Hier findet fortschreitend periostale Knochenbildung statt, mit geringer Veränderung des Endosteums. Bei Fleischdiät wird das Körperwachstum während der ersten Woche erheblich beschränkt; danach aber normalisiert es sich wieder. Obwohl die Knochen noch wachsen, zeigen sie keine normale Mineralisierung und werden schnell zerbrechlich und dünn. Die Knochenresorption ist anfangs langsam, dann beschleunigt sie sich während einer Zeitspanne von 2–3 Wochen bis auf 15 μ pro Tag, um sich dann wieder zu verlangsamen. Während die Knochenbildungsgeschwindigkeit relativ zum Normalwert heruntergesetzt wird, verläuft die Resorption ungefähr 2–3mal so schnell wie die Knochenbildung. Mikroradiographische Untersuchungen an mit Fleisch ernährten Ca-armen Ratten haben bestätigt, daß während die Resorption am Endosteumrande stattfindet und sich die Knochenbildung an der Periostenfläche fortsetzt, die neugebildete Knochensubstanz weniger kalzifiziert ist, als die der Kontrolltiere.
    Notes: Abstract Different coloured, fluorescent bone-seeking chemicals, viz., tetracycline, Alizarin Red S, and DCAF, have been administered sequentially to weanling rats and the rate of formation and resorption of bone measured from hard-ground cross sections of the upper third of the diaphysis of the femur. On a meat diet, bodily growth is significantly restricted for the first week and then recovery occurs. While bones grow they fail to mineralize normally and rapidly become fragile and rarefied. Resorption of bone is at first slow, then accelerates for a period of 2–3 weeks to about 15μ/day and then slows again. While the rate of bone formation is reduced relative to normal bone, resorption proceeds at approximately two to three times the rate of bone growth. Microradiographic studies confirm tht while resorption occurs on the endosteal margin and formation proceeds on the periosteal aspect of meat fed Ca-deficient rats, new bone is less calcified than that in control animals.

Le degré de calcification au quatrième jour de culture measuré par la relation calcium/hydroxyproline, suit un fonction lineáire avec le logarithme des doses de sulfate de dehydroepiandrostérone employées (0.5, 1,0 et 2,0 mM). Les renseignements obtenus indiquent que les frontaux de 13 jours, cultivés “in vitro” constituent modeles experimentaux appropriés pour étudier l'effet des androgénes sur le tissue osseux.
    Abstract: Zusammenfassung Stirnbeine von Hühnerembryonen an ihrem 12. und 13. Entwicklungstag entnommen und in vitro kultiviert zeigen verschiedene Arten der Glucoseverwertung der Periostzellendichte, des Calcium- und Hydroxyprolingehaltes. Wird Dehydroepiandrosteronsulfat dem Medium in einer 1 mM-Konzentration zugegeben, so beteiligen sich die 12tägigen Stirnbeine vorwiegend an der Knochengewebesynthese, während die 13tägigen signifikant stärker verkalken als die Kontrollen. Gemessen an der Calcium/hydroxyprolin Ratio bildet die Verkalkung der 13tägigen Stirnbeine eine lineare Funktion mit den Logarithmen der verwendeten Dosen von Dehydroepiandrosteronsulfat (0,5, 1,0 und 2,0 mM). Das in vitro kultivierte 13tägige Stirnbein schein ein geeignetes Experimentiermodell zur Studie der Dehydroepiandrosteronsulfatwirkung auf das Knochengewebe zu sein, weil es das grundlegende Phänomen (erhöhte Verkalkung) wiedergibt, welches man auch bei mit Androgenen behandelten Menschen und Tieren beobachtet.
    Notes: Abstract Chick embryo frontal bones at 12 and 13 days of development cultivatedin vitro exhibit different patterns of glucose utilization, periosteal cellular density and calcium and hydroxyproline content. When dehydroepiandrosterone sulfate is added to the medium at a concentration 1 mM, 12-day frontals engage primarily in osteoid tissue synthesis while 13-day frontals calcify at a significantly greater rate than controls. Measured with the ratio calcium/hydroxyproline, the calcification of 13-day frontals follows a linear function with the logarithm of the doses of dehydroepiandrosterone sulfate employed (0.5, 1.0 and 2.0 mM). The 13-day frontal bone cultivatedin vitro seems to be an adequate experimental model for the study of the effects of dehydroepiandrosterone sulfate on bone tissue because it reproduces the basic phenomenon (increased calcification) observed in man and animals treated with androgens.

Ces changements qui se manifestent par des changements de la biréfrigence extrinsique du tissu osseux, ont été suivis à l'aide du microscope polarisant et aussi du microscope électronique. De même l'entrée de45Ca dans les cristaux a été mesurée dans certains cas. Les cristaux osseux sont hétérogènes quant à leur solubilité. Le produit de solubilité du minéral osseux, exprimé par la relationp [Ca]+p[P], est de 6.51±0.07 (σ) mesuré par la microscopie polarisante à l'instant où les changements furent les plus petits. Ce chiffre s'accorde avec les résultats des autres chercheurs qui ont mesuré la solubilité de la poudre osseuse par des autres méthodes. La solubilité des cristaux dans notre système varie inversement avec les concentrations des ions hydroxyliques ou fluors, mais varie directement avec la concentration des ions citriques.
    Abstract: Zusammenfassung Ein Systemin vitro ermöglichte es durch Veränderung der Zusammensetzung der umgebenden Lösung das Wachstum und die Auflösung von Knochenkristallen herbeizuführen. Die hervorgebrachten Veränderungen konnten dank der doppelbrechenden Eigenschaften des Knochengewebes mit dem Polarisationsmikroskop festgestellt werden. Diese Beobachtungen wurden durch Untersuchungen am Elektronenmikroskop und Bestimmungen der45Ca-Aufnahme gestützt. Die Knochenkristalle erwiesen sich als heterogen in bezug auf ihre Löslichkeit. Wenn die hervorgerufene Kristallveränderung im Polarisationsmikroskop minimal war, so entsprach das Gesamtlöslichkeitsprodukt des Knochenminerals, ausgedrückt als p[Ca]+p[P] 6,51±0,07 (S.D.). Diese Resultate bestätigten die Befunde anderer Forscher, welche die Knochenpulverlöslichkeit mittels konventioneller Methoden bestimmten. Die Löslichkeit der Kristalle bei diesem Systemin vitro variierte entgegengesetzt zur Hydroxyl- und Fluoridionenkonzentration, aber im gleichen Sinne wie die Citrationenkonzentration.
    Notes: Abstract Anin vitro system was used to induce growth and dissolution of bone crystals by manipulating the composition of their fluid environment. The induced changes could be detected with the polarizing microscope because of the extrinsic birefringence of the bone tissue. Supporting observations were made with the electron microscope and by determining45Ca uptake. The bone crystals were found to be heterogeneous with regard to their solubility. When induced crystal changes were minimal by polarization microscopy the overall solubility product of the bone mineral expressed asp [Ca]+p[P] was 6.51±0.07 (S.D.). This result corroborated the findings of other investigators who determined bone powder solubility by conventional methods. Solubility of the crystals in thein vitro system varied inversely with hydroxyl ion and fluoride ion concentration, but directly with citrate ion concentration.

    Notes: Summary After cutting a neck connective of Schistocerca gregaria, only 2% of the axons on each side of the lesion degenerate. The remainder show reactive changes, which last for approximately one week at 28° C. There is no morphological change in either of the pro/mesothoracic connectives after injury to the neck connective. Phagocytes invade the stumps, but attack only degenerating cells, and are absent by Day 7. Regeneration from the connective stumps begins a week after injury; a functional link may be formed by Day 10, but by Day 23 the new connective cannot function adequately for the locust's survival, if the undamaged connective is then cut. The chief morphological changes in the reactive axoplasm are increases in the number of mitochondria, neurotubules, vesicles and vacuoles. These changes appear to be a local response, and not to be influenced by the neuron cell bodies. Some glial cytoplasm (presumably enucleated), degenerates rapidly after injury, and replacement begins by Day 5. Tracheoles, never seen in normal connectives appear in the reactive connective from Days 3–8, this is interpreted as a migration from the ganglion in response to oxygen deficiency in the connective. The results are discussed in relationship to previous work.

    Notes: Summary The free or “non-glandular” pseudobranch of the flounder Paralichthys lethostigma was studied with the electron microscope. Cells typical of glandular type pseudobranchs are found. This indicates that, at least in the flounder, the free pseudobranch should be called “glandular”. In addition, the chloride-type cells, commonly found in the gill, buccal epithelium, and surface epithelium of other fish, have been found in the pseudobranch, where they have not been described previously. The fine structure of both the chloride-type and the pseudobranch-type cell has been characterized and contrasted. We have not been able to confirm previous reports that the specific cells in both pseudobranch and gill are identical in the flounder.

    Description / Table of Contents: Zusammenfassung Die ableitenden Lymphgefäße im Mesenterium des Dünndarms männlicher weißer Ratten wurden elektronenmikroskopisch und histochemisch untersucht. Elektronenmikroskopisch findet sich ein lückenloses Endothel mit reichlich pinocytotischen Vesikeln. Eine Basalmembran ist im Klappenbereich durchgehend, sonst nur bruchstückweise vorhanden. Die darunter gelegene Bindegewebsschicht (Lamina propria interna) ist sehr unterschiedlich ausgeprägt. Die lichtoptische Einteilung in muskelstarke und muskelschwache (klappennahe) Abschnitte des Lymphangions läßt sich elektronenmikroskopisch bestätigen. Die zahlreichen membrannahen Vesikel der Muskelfasern sind bei Kontraktion in zahnartigen Fortsätzen angereichert. Auch beim Normaltier treten sog. „aktivierte“ Muskelzellen auf. Osmiophiles Material ist nach Fettfütterung zwischen den Muskelzellen und in der Lamina propria interna der Gefäßwand vorhanden, nicht dagegen in den Klappen. Der Kontakt zwischen den Muskelzellen erfolgt mittels fingerartiger Fortsätze. Endothel und Klappen sind frei von nervösen Elementen. Nervenendigungen und Axone sind zwischen den Muskelfasern nicht zu beobachten, hingegen ganz selten Ganglienzellen. Histochemisch sind im Endothel starke Aktivitäten der sauren Phosphatase und der Monoaminooxydase nachzuweisen, auch unspezifische Esterasen, ATP-ase sowie verschiedene Dehydrogenasen sind vorhanden. Cholinesterase, Cholindehydrogenase und alkalische Phosphatase fehlen. In der Media sind unspezifische Cholinesterase, unspezifische Esterasen, ATPase, Monoaminooxydase, Succinodehydrogenase und NAD-Diaphorase nachweisbar. Im einzelnen kann die Stärke der Aktivität einem bestimmten Abschnitt (muskelstark/muskelschwach) des Lymphangions zugeordnet werden. Damit ist eine Einteilung der Lymphangione in klappentragende und muskelmanschettenhaltige Anteile auch histochemisch möglich. Im Gegensatz zur Adventitia der Blutgefäße gibt die der Lymphgefäße keine Reaktion auf alkalische Phosphatase. Die dort reichlich vorhandenen Mastzellen sind Naphthol-AS-D-chloracetat-esterase positiv.
    Notes: Summary The large lymphatic vessels in the mesentery of male white rats were investigated with ultrastructural and histochemical methods. Electron micrographs show an uninterrupted simple endothelial layer, the individual cells of which are rich in pinocytotic vesicles. A distinct continous basement membrane is to be found only in the region of the valves, otherwise it is lacking over wide parts of the vessel wall. The so-called lamina propria interna differs greatly in its extent. Electron microscopy confirms the subdivision of the lymphangion into segments rich in muscle cells and others (in the area of the valves) which are less rich in them. Muscle cells contain numerous vesicles in close connection with the cell membrane which in contracted cells are to be found predominantly in toothlike projections. Also in the normal rat, so-called “activated” muscle cells are to be found. After a meal rich in fat, osmiophilic material can be seen between the muscle cells and in the lamina propria interna, however, not in the valves. Muscle cells are in contact with each other by finger-like processes. The endothelial layer and the valves are lacking nervous elements. There are no nerve endings and no axons between the muscle cells, but very rarely ganglion cells can be found. Histochemically, strong activities of acid phosphatase and MAO have been demonstrated in the endothelial layer. Unspecific esterases, ATP-ase and various dehydrogenases are also present, while cholinesterase, cholindehydrogenase and alkaline phosphatase are lacking. In the media, unspecific esterases, unspecific cholinesterase, MAO, succinodehydrogenase and Naddiaphorase can be found. The intensity of enzymatic activities can be correlated to specific segments of the lymphangion. It is, therefore, possible to distinguish by histochemical methods the valve areas, which are poor in muscle cells, from the muscle-cuffs. In contrast to the blood vessels, the adventitia of the large lymphatics is alkaline-phosphatase negative. The mast cells, abundant in the adventitial layer, show α-Naphthol-AS-D-chloroacetate-esterase activity.

Die Zellen des Sacculus sind typische Podocyten, an denen eine Ultrafiltration ablaufen kann. Möglicherweise wird die Filtration durch einen den Sacculus umgebenden Muskel unterstützt. Die Zellen des Tubulus zeigen basale Einfaltungen und im proximalen Teil auch Mikrovilli. Sowohl im Tubulus als auch im Ausführgang, dessen Zellen ebenfalls basale Einfaltungen aufweisen, werden Reabsorptionsprozesse vermutet.
    Notes: Summary The fine structure of the maxillary kidney of Scutigerella immaculata Newport (Symphyla) has been investigated. It may be compared with segmental organs of other Arthropoda having an end-sac which forms a primary urine by ultrafiltration. The filtration may be supported by a muscle surrounding the end-sac. The tubular part of the nephridium and the efferent duct show structures which may be involved in reabsorption.

    Notes: Summary For a limited period during the oogenesis of Protopterus, blebs of the perinuclear cistern contain, in addition to other inclusions, a special kind of microtubular elements. Most of these blebs face parts of multiple nucleolar bodies that extend toward and make contact with the inner nuclear membrane. The microtubular lumen contains a finely dispersed material of moderate electron density which seems to be in contact with this nucleolar material. Aside from these intracisternal structures there are, within both the perinuclear cytoplasm and the nucleoplasm, similar microtubular arrays without apparent connection with the nuclear envelope. These are either enclosed by membranes derived from those of the envelope or unconfined, having escaped through breaks in their respective bounding membranes. Extracisternal tubules are presumed to have passed their period of putative functional activity and to be undergoing a process of regression and subsequent disintegration. Among possible roles attributable to the intracisternal microtubular apparatus are the following: (1) It may serve for the transport of special nucleolar components to the cytoplasm, possibly to be incorporated in the matrix of developing perinuclear mitochondria; (2) it may provide openings in the nuclear membranes for the direct passage of particulate elements between nucleus and cytoplasm; (3) it may be instrumental in the breakdown of parts of the nuclear envelope prior to its restitution during the subsequent phase of oogenesis.

    Notes: Summary Studies of possible neuroendocrine structures in the pulmonate gastropod Helisoma tenue show that cerebral fuchsinophilic neurons with electron-dense granules (mean diameter 1,500 Å) apparently release their secretory content in an “intercerebral commissure neurohemal area” near the mediodorsal bodies (MDB) or in the “median labial nerve neurohemal area”. The MDB cells have axon-like processes which branch and end at the cerebral surface, separated by a thin capsule from the cerebral fuchsinophilic cells. The presence of granules (mean diameter 800 Å) in the terminals of the MDB cells suggests cell body origin, distal transport and release of the granular secretory material. The secretory product may have an influence on cerebral fuchsinophilic neurons. Electron microscopy reveals the presence of granules of different sizes and densities in expanded neurites at the periphery of the intestinal nerve of the visceral ganglion which may indicate the presence of a neurohemal area. However, the granules in the intestinal nerve neurites and in the visceral ganglion fuchsinophilic cells are similar to granules found in the heart which also suggests that the granules may carry a neurotransmitter.

La cuticule du cadre est formée d'une masse compacte et amorphe, sclérifiée, surmontée d'une cuticuline de structure normale. Les deux autres types de cuticules possèdent une structure plus classique: épicuticule comprenant cuticuline et zone dense, et endocuticule. La cuticule de l'épithélium rectal a toujours une structure comparable à celle d'une cuticule périphérique non sclérifiée. La cuticule des papilles est plus variable, et toujours différente de celle de l'épithélium rectal. Ces différences portent sur la cuticuline, la zone dense, les filaments épicuticulaires; l'endocuticule sur les papilles montre une organisation fibrillaire moins nette, qui paraît corrélative d'une réaction positive à l'APS. Des canaux poraires n'ont jamais été observés dans la cuticule des papilles; il en existe quelquefois dans la cuticule de l'épithélium rectal. Une sous-cuticule, contenant des mucopolysaccharides acides, est présente dans certains cas dans les deux types de cuticule. Dans les espèces ne possédant pas de papilles rectales, il n'existe qu'un seul type de cuticule rectale, variable suivant l'espèce. La cuticule des papilles montre, dans certaines espèces, des dépressions superficielles, d'un diamètre voisin de 0,2 μ, réparties uniformément mais sans ordre défini (≪dépressions épicuticulaires≫). De telles dépressions s'observent aussi dans quelques espèces dépourvues de papilles (Tenebrio). Au niveau de ces dépressions, la cuticuline a une structure particulière et la zone dense est réduite. Les rapports entre structure et perméabilité de la cuticule sont discutés. On envisage en particulier les rôles respectifs de la cuticuline, de la zone dense, des filaments épicuticulaires et des dépressions épicuticulaires.
    Notes: Summary The ultrastructure of the rectal cuticle was studied in eleven species of Insects from nine orders. Special attention was given to those having rectal papillae. In such species, three types of cuticule were observed: 1) the cuticle of the rectal epithelium, 2) the cuticle of the papillae, 3) the cuticle of the border of the papillae. The cuticle of the border is made of a compact amorphous substance, which is sclerotized and covered by a typical cuticulin layer. The two other types of cuticle exhibit a more conventional organization, namely the epicuticle comprising both the cuticulin and the dense layer, and the endocuticle. The cuticle of the rectal epithelium is similar to unsclerotized external cuticle. The cuticle of the papillae is more variable and always differs from the cuticle of the rectal epithelium. Variations occur with regards to the cuticulin, the dense layer and the epicuticular filaments. The endocuticle of the papillae has a less conspicuous fibrillar structure which we believe is correlated with the PAS-positive reaction found in this endocuticle. Pore canal sometimes are observed in the cuticle of the rectal epithelium but never above the papillae. In some cases, a sub-cuticular layer containing acid mucopolysaccharids is present in the two types of cuticle. In species lacking rectal papillae, the rectal wall is covered by only one type of cuticle, the features of which vary according to species. In some Insects, the cuticle of papillae shows numerous superficial depressions, each of about 0.2 μ in diameter. These “epicuticular depressions” are uniformly distributed throughout the papillae but are not arranged in any particular pattern. We have also found such depressions in other Insects lacking rectal papillae (e.g. Tenebrio). In the epicuticular depressions, the cuticulin exhibits a modified organization and the dense layer is reduced. The relations between ultrastructure and cuticular permeability are discussed. The possible roles of the cuticulin, the dense layer, the epicuticular filaments and the epicuticular depressions are examined.

Die Lage der Ableitungspunkte wurde durch den histologischen Nachweis der elektrolytisch gesetzten Eisenmarken bestimmt. Diese Ableitungsstellen befinden sich im Epiphysenparenchym. Ein stärkeres versilberbares Nervenbündel verläuft im Stiel der Epiphyse. Adrenerge Nervenfasern, die grün fluoreszieren, begleiten die Kapselgefäße und dringen mit diesen in das follikuläre Epiphysengewebe ein. Das Epiphysenparenchym zeigt nach Falck-Hillarp eine Gelbfluoreszenz mit umschriebenen apikalen oder basalen Anreicherungen der fluoreszierenden Substanz. Mit Reserpin kann nur ein Teil dieses Materials entspeichert werden. Elektronenmikroskopisch lassen sich in der Taubenepiphyse keine typischen pinealen Rezeptorzellen nachweisen. Regelmäßig lamellierte Außenglieder fehlen vollständig. Es finden sich lediglich cytoplasmareiche bulböse Zilien und zirkuläre Lamellenkomplexe, die in Tubuli übergehen. Die letzteren können sich mit einem dichten körnigen Material füllen. An einigen Stellen haben solche Lamellenverbände einen Zusammenhang mit keulenförmig verdickten Zilien. Diese Zone ist aber anders strukturiert als das für retinale und pineale Lichtsinneszellen charakteristische Verbindungsstück. Die Zeichen der sekretorischen Aktivität der Taubenepiphyse sind überzeugender als die für einen lichtempfindlichen Sinnesapparat sprechenden Strukturmerkmale. Ein bestimmter Typ der Pinealocyten ist reich an 800–1200 Å großen granulierten Vesikeln, die sowohl am apikalen als auch am basalen, auf die Blutgefäße ausgerichteten Zellpol angehäuft sein können. An den manchmal gelappten basalen Endfüßen sind Zeichen der Ausschleusung des Vesikelinhalts bzw. eines Materialschwundes aus dem granulierten Vesikelkern zu erkennen. Die sekretorische Aktivität der Taubenepiphyse wird mit Hinweis auf die in ihr reichlich vorkommenden 5-Hydroxyindole (Quay) und das Problem der Melatonin-Bildung diskutiert. Erörtert wird auch die Möglichkeit einer Stimulierung dieser Sekretion über den Sympathikus.
    Notes: Summary Direct illumination or stimulation with light of the lateral eyes induced no electrical activity in the central part of the pineal organ in the pigeon. In the pineal stalk, however, a train of impulses was observed that failed to respond to changes of illumination. The position of the tip of the recording electrode (iron deposit), was histologically clearly determined to be in the pineal parenchyma. A distinct nerve bundle could be traced down the pineal stalk. Adrenergic nerve fibers showing a green fluorescence accompanied pineal capsular vessels into pineal follicular tissue. Using the method of Falck-Hillarp, the pineal parenchyma showed high concentrations of yellow fluorescent material in the apical or basal parts of the pinealocytes. Only a part of the fluorescent material could be dispersed with reserpine. In the electron microscope, no typical photoreceptor cells were identified in the pineal organ. Only large bulbous cilia, and circular lamellar or tubular complexes, were observed; they sometimes contained a dense granular material. Occasionally, the lamellar complexes had a direct relation to cilia of club-shaped appearance. Signs of secretory activity were more definitive in the pigeon pineal organ than sensory structures. One type of pinealocytes contained many 800–1,200 Å diameter granular vesicles concentrated in the apical or basal (apposed to blood vessels) cell parts. At the occasionally lobulated basal end-feet, a disappearance of the content of the granular vesicles indicated a release mechanism. This secretory activity was discussed in relation to the 5-hydroxyindole content of the pigeon pineal organ (Quay) and the general problem of melatonin synthesis; also, the possibility was considered that the secretory activity may be stimulated by sympathetic nerve fibers.

    Notes: Summary The vitreous body of the rat eye was studied by scanning electron microscopy. The freeze-dried specimens were either unfixed, freshly frozen or fixed and rinsed before being frozen. The vitreous body is surrounded by a coat of intercellular material covering a thin sheath of chiefly parallel fibers. The latter forms the base for a regularly organized three-dimensional network of fibers. The interfibrillar spaces are filled with ground substance and also contain a few cells. The shape of the cells varies from bipolar to polyhedronal. Their interconnected processes are usually undivided. The functional importance of this complex regular organization of the fibers and cells in the vitreous body is stressed.

    Notes: Summary The sarcolemma and T system and the sarcoplasmic reticulum were studied in a fast acting muscle, the extraocular muscle of Fundulus grandis. The predominant morphological fiber type contained abundant glycogen, a highly developed T system with triads at the A-I junction and a well developed sarcoplasmic reticulum. The numerous tubular profiles of T system and sarcoplasmic reticulum with their characteristic contents made this fiber especially suitable for cytochemical investigation. Revel's thorium dioxide technique for the ultrastructural localization of tissue polyanions was applied to thin sections of extraocular muscle. The results confirmed an earlier report by Goldstein and Philpott on fish branchial muscle. The polyanionic material was associated with the sarcolemma, the T system and the sarcoplasmic reticulum of these fibers. Both the distribution and the amount of polyanionic substance associated with these physiologically important membrane systems support the idea that the reactive material serves as an anionic substrate for binding cationic calcium.

    Notes: The venom system of Nasonia vitripennis is well-developed and composed of an unbranched acid gland and associated reservoir. Fine-structural, histochemical and electrophoretic studies indicate that the venom is produced by two protein-secreting epithelia. The bulk of the venom is synthesised in the columnar cells of the acid gland and discharged via “vesicular organelles” and the efferent ductular system into the lumen of the reservoir. The acid gland also contains squamous chitogenous cells, situated either around the central lumen or interposed between the bases of the columnar cells. Once within the reservoir, the venom is probably activated by enzymatic secretions from the reservoir secretory cells. Each of these cells has a “vesicular organelle” but, in contrast to the columnar cells of the acid gland, the cytoplasm contains a preponderance of free ribosomes, and protein segregation apparently occurs outside the Golgi complexes.The venom is expelled through the efferent discharge duct by muscular contractions, which open the duct lumen and bring it into contact with the funnel of the ovipositor. Excessive distortion of the duct is prevented by a massive ventral ligament.

    Notes: There are eight retinula cells in the ommatidium of the compound eye of the toadbug (Gelastocoris oculatus), two of which are central in position. Along the axial sides of the six peripheral retinula cells expand six cytoplasmic processes from the apical crystalline cone cells. These processes, which contain longitudinally-oriented microtubules, are associated with all eight retinula cells by means of desmosomal junctions. In addition to providing structural support, the possibility is set forth that the interconnecting cone processes might also serve to functionally integrate the retinula cells of an ommatidium. The eight retinula cells possess microvillus surfaces, which are especially prominent in the six peripheral cells, where they extend into the lumen of the ommatidium. There is evidence of pinocytotic activity at the bases of microvilli. Multivesicular bodies are present in the cytoplasm of retinula cells, and the means by which these bodies might be elaborated are discussed.

    Notes: The mechanism of respiration in the bullfrog has been analyzed by means of pressure recordings from the buccal cavity, the lungs and the abdominal cavity, by cinematography and cinefluorography, and by electromyography of buccal, laryngeal and abdominal muscles. Gas flow was investigated by putting frogs in atmospheres of changing argon and nitrogen content and monitoring the concentration of the nostril efflux.Three kinds of cyclical phenomena were found. (1) Oscillatory cycles consist of rhythmical raising and lowering of the floor of the mouth, with open nares. They have a definite respiratory function in introducing fresh air into the buccal cavity. (2) Ventilatory cycles involve opening and closing of the glottis and nares and renewal of a portion of the pulmonary gas. More muscles are involved and the pattern of muscular activity is more complex than in the oscillatory cycles. (3) Inflation cycles consist of a series of ventilation cycles, interrupted by an apneic pause. The intensity of the ventilatory cycles increases before this pause and decreases immediately thereafter. This results in a stepwise increase in pulmonary pressure, to a plateau (coincident with the pause) followed by a sudden or stepwise decrease.The respiratory mechanism depends on the activity of a buccal force pump, which determines pulmonary pressure whose level is always slightly less than the peak pressure values of the ventilation cycles. The elevated pulmonary pressure is responsible for the expulsion of pulmonary gas during the second phase of the next ventilation cycle. This pressure is maintained by the elastic fibers (and the smooth masculature) of the lungs.

    Notes: The complete regeneration of a new oral-disc and tentacles has been observed and described for Aiptasia diaphana. These structures are regenerated quite rapidly: seven to ten days at 20°C. At three days post-amputation, the new primary, secondary, and tertiary tentacle buds begin to develop in direct association with the underlying primary, secondary, and tertiary septae (respectively) of the column, suggesting that the latter organize the form of the regenerating oral-disc. Two days after amputation, the zooxanthellae of the presumptive oral disc arrange themselves into a ring which quite precisely delimits the area from which the tentacle buds will form. In spite of its suggestive proximity, this accumulation of algae plays no role in the induction of tentacle buds as was shown by studying regeneration in anemones which essentially lacked large quantities of these symbiotic algae.Cuts perpendicular to the longitudinal axis of the column result in an equal rate of tentacular regeneration around the entire circumference of the presumptive oral disc. Oblique amputations foster an asynchronous regeneration: the tentacle buds of the distal-most area of the severed column are larger and regenerate much sooner than those of the proximal region. Similar results were obtained by studying anemones which were cut perpendicular to their longitudinal axes at different levels along the column. The data suggest that an oral-aboral gradient exists concerning the time required for the initiation of tentacle budding and the rate of tentacle regeneration.

    Notes: A structure for a generalized insect epidermal cell during the formation of the epicuticle is proposed, based on studies of several different epidermal cell types. The protein epicuticle is defined as the dense homogeneous layer below the cuticulin. The formation of the protein epicuticle involves secretory vesicles arising in Golgi complexes, and marks an interlude in the involvement in cuticle formation of plasma membrane plaques. The plaques are concerned in cuticulin formation before and in fibrous cuticle formation after the deposition of the protein epicuticle.The epidermis is characterized by the possession of a cytoskeleton of microtubules and a matrix of microfibers. In the elongated cells forming bristles and spines, the microfibers are often oriented in bundles with an axial banding which repeats every 120 Å. The microtubules are also arranged in columns with a trigonal packing and center to center spacing of about 800 Å. These cytoskeletal structures separate the other organelles into channels which may restrict the pathways open for the movement of secretory and pinocytotic vesicles. The protein epicuticle arises from the secretory vesicles which discharge at the apical surface. The contents disperse and reaggregate below the cuticulin. The Golgi complexes in the basal and central regions have many secretory vesicles and a small saccular component, differing from those nearer the apex which are smaller and have fenestrated saccules. The small coated vesicles (800 Å in diameter) associated with both sorts of complex, probably move to the apical and basal faces of the cell where they may give rise to the large coated vesicles (2000 Å in diameter) inserted in the plasma membrane. Pinocytosis occurs from both apical and basal faces but most lytic activity is in the apical region. Plant peroxidase injected into the haemocoel is taken up basally and transported to the apical MVBs. The large coated vesicles on the apical face may be concerned in the control of the extracellular subcuticular environment. They appear to fill up and detach, fusing to become the apical MVBs.

    Notes: Submandibular glands of the opossum have been studied by light and electron microscopy and compared with other mammalian salivary glands. The glands have four parenchymal segments which connect in the order named below to convey saliva toward the oral cavity. 1Secretory units are elongated branching tubules exhibiting mucous and special serous cell types. Mucous cells predominate and resemble those in other salivary glands. Special serous cells differ from “typical” serous cells. They contain a preponderance of tubular or vesicular endoplasmic reticulum and secretory granules which vary from electron lucent to electron opaque.2Intercalated ducts are short segments lined by nonsecretory, cuboidal cells.3Striated ducts are numerous and lie in the center of the lobule. The duct epithelium has four cell types, designated light cells, dark cells, Type I basal cells, and Type II basal cells. Light cells possess basal infoldings associated with mitochondria, but the other cell types lack this characteristic.4Excretory ducts are also lined by four cell types which bear the same names as those of striated ducts. Three of the four cell types are virtually identical to those of striated ducts, but light cells differ. They do not always contain basal infoldings and the supranuclear cytoplasm lacks distinct inner and outer zones.The glands resemble salivary glands of higher mammals in many respects while possessing certain unique cytological features which may reflect the secretory needs of the organism.

    Notes: Several biological distances based on cranial and mandibular variation among breeding groups of white-tailed deer were calculated and compared with geographic distances among the groups. Distances based on epigenetic variation among ten groups were calculated using 16 non-metric variants of the cranium and mandible. Penrose's size and shape distances and Mahalanobis' D2 distance were calculated for 11 groups; the calculations were based on seven skeletal and seven dental metric variables of the mandible.The biological distances were correlated with geographic distance as follows: the epigenetic distance, 0.74; Penrose's shape distance, 0.71; Penrose's size distance, 0.45; and Mahalanobis' distance, 0.37. All correlations were significant at the 0.01 level. The epigenetic and Penrose shape correlations were significantly higher than the Mahalanobis correlation.Because of the conditions under which the breeding groups were selected, it was assumed that genetic affinites among the groups would be a function of geographic distance. The results suggest that the epigenetic distance and Penrose's shape distance reflect genetic affinities among groups better than do the Penrose size and Mahalanobis distances.

    Notes: The mechanism of lung ventilation in chelonians has been much debated. Electromyographic studies show that the basic mechanism in the snapping turtle, Chelydra serpentina, is dependent on the activities of four major respiratory muscles that are capable of varying the volume of the visceral cavity. The precise mechanism utilized varies in response to environmental factors, especially the depth to which the animal is submerged. Chelydra tends to reduce muscular activity to a minimum, and hydrostatic pressure or gravity replaces muscular effort whenever possible. The response is subject to hysteresis. Both the mechanics and pattern of ventilation in Chelydra differ from those of Testudo. The differences appear to be attributable in part to Chelydra's markedly reduced plastron and more extensive respiratory musculature and in part to the different habitats occupied by the two species.

    Notes: The purported “neoblasts” of the serpulid Spirorbis have been studied in Spirorbis (Paradexiospira) vitreus and Spirorbis (Laeospira) borealis at both the light and electron microscopic levels. These perivasal cells occur in greatest abundance around the ventral blood vessel of the achaetous region. In light microscope preparations, the perivasal cells are intensely basophilic, containing basally situated nuclei, and relatively large nucleoli. The fine structure of the perivasal cells reveals that they contain an abundance of rough endoplasmic reticulum, well-developed Golgi complex, heterogeneous dense bodies, and cytolysomes. The respiratory pigment chlorocruorin, which has a diameter of about 230 Å and is believed to be composed of two superimposed hexagonal components, has been localized within: cisternae of the rough endoplasmic reticulum, elements of the Golgi complex, and membrane-bounded vesicles at the base of the perivasal cells. Evidence is advanced which strongly suggests that molecules of chlorocruorin are transported from the perivasal cells into the lumen of the vessel by reverse pinocytosis. It is concluded that whatever other functional role(s) the perivasal cells of Spirorbis may have, a major function is the synthesis and secretion of chlorocruorin. Whether the perivasal cells can be considered to be pluripotent is discussed.

    Notes: The concept of functional components was initially proposed by van der Klaauw ('45, '52) to indicate overlap of functional influences particularly in mammalian skulls; his analysis marked a departure from the study of single characters to that of function-modified systems. A very similar set of terms is now coming into vogue to describe the mechanically separable components of highly kinetic fish, amphibian and reptilian skulls. In these cases the term functional unit often pertains only to the musculo-skeletal system and is utilized during the process of description; it is often applied before a complete functional analysis has been carried out.Yet, any structure tends to be affected by the influence of multiple functions, and any function will almost certainly affect multiple characteristics of the animal. Since functional components overlap, the term should not be used to label an essentially topographical dissection of the animal. It cannot be expected that each loosely connected component of a kinetic skull subserves as a single “function,” and that this function does not overlap onto other units.It is suggested that the term mechanical unit be substituted as a label for the mechanical sub-divisions often utilized to organize descriptions. The concept of functional units in the original sense then remains available as an analytical tool.

The follicle cells synthesize the egg coverings, the vitelline and chorionic membranes, and elaborate the micropyle and dorsal chorionic appendages. The changing structure of the nurse cell nucleus and changes in organelle composition of its cytoplasm are described. The nurse cells synthesize ribosomes, lipid droplets, and mitochondria. These components pass through the ring canal system into the oocyte, which increases in volume some 200,000 times during its 78 hours of development.

    Notes: Each muscle of the third metasomal segment of the male of Nomia melanderi Ckll. is described in detail. The points of attachment of each muscle are compared with their homologs in other pregenital segments and with their homologs in the female. The function desgnated for each muscle describes its action alone or in conjunction with other muscle(s). New names are given to genital muscles by referring in the name to their points of attachment. Each intratergal muscle has homologous points of attachment in the pregenital segments of both sexes. The median tergo-dorsoplical muscle of the seventh segment and the oblique tergo-dorsoplical muscle of the eighth segment have changed their points of attachment. The intrasternal muscles are modified to suit the needs of courtship and mating, thus they are different from their homologs in the female. The spiracular muscles are well developed in all segments except the eighth, where the sterno-spiracular muscle is absent. The extrinsic genital muscles are derived from the intrasternal muscles of the eighth and ninth segments. The parameral and volsellar muscles are reduced in number. The aedeagal muscles, except the aedeago-phallic, have retained similar points of attachment to those found in primitive Hymenoptera. The topography of the metasomal nervous system is reported in detail by following each nerve and nervule to its termination. The study shows that (at least in Nomia) the criterion of nerve-concentration should not be used alone to indicate evolutionary levels. To accommodate the morphological changes in the terminal segments the Anterior and Posterior Lateral Nerves have migrated to new locations. The pattern of nerve topography (even at the nervule level) is homologous both in the different pregenital segments and between the sexes. The fact that homology does not exist between the external genitalia of the male and the modified ovipositor of the female supports the thesis that the male genital capsule is of phallic rather than prephallic origin. A pair of intersegmental membrane glands located between the seventh and eighth sterna is described. These glands may be the source of a pheromon responsible for gregariousness among “sleeping” males.

    Notes: Mature myoid cells in the parenchyma of reptilian thymus contain all the organelles typical of striated muscle. The presence of both immature and degenerating stages indicates a turnover of myoid cells in the adult thymus. In the earlier stages of differentiation myoid cells resemble thymic epithelial cells. A close parallel exists between developing myoid cells, skeletal muscle differentiating in vitro, rhabdomyoma and rhabdomyosarcoma. Elaborate lattice-like structures are formed by transverse tubules. These structures are compared with similar configurations which have been described in muscle and mitochondrial cristae.

    Notes: The hatching threads of praying mantis embryos are silk-like in appearance, but cellular in origin. Their development can be divided into four phases. In phase 1 each embryonic cercus produces a hollow column of cells which is pushed out dorso-laterally, on either side of the embryo, between the epidermis and the chorion. In phase 2 each column becomes transformed into an unbroken helical cellular filament. The terminal five or six cells at the distal end of each filament become permanently attached to the inner surface of the chorion. The cellular arrangement of the filament is superseded by an apparent syncytial condition. In phase 3 the extensive proximal parts of the two filaments become folded into a compact space, flanked by the cerci and styli, at the tip of the abdomen. Throughout phase 4 the filaments remain in two tightly coiled groups, connected to the chorionic attachments by loosely coiled distal regions. Progressive secretion of chitin by, and around, each filament forms a sheath, 1-2 μ thick, which provides the tensile component of the hatching threads. During emergence these threads become unravelled to form a double thread which allows the insect to hang from the ootheca and complete its first ecdysis.

    Notes: The present investigation was undertaken in an attempt to determine the role played by the nerve in the regeneration of the lower jaw of the adult newt, Triturus viridescens. The results indicated that the number of nerve fibers normally available at the amputation surface was very low compared with that of the newt forelimb. Furthermore, denervation of the lower jaw reduced the number of nerve fibers available to an extremely low level and maintained the number at a low level for up to four weeks without intervening redenervations. The regenerative events in the denervated and amputated lower jaws were indistinguishable histologically from those in amputated jaws having normal innervation. This presented an apparent exception to the general rule that regeneration of external body parts is dependent on the nerve. Several possible explanations are proposed by which this apparent exception might be explained. The process following amputation might be an exaggerated form of wound healing and tissue regeneration which can occur in the absence of nerves. The tissues of the lower jaw might be more sensitive to the influence of those nerve fibers present. The nerve fibers themselves might be qualitatively different and thus exert a greater influence on the tissues.

The median cord arises independently from the lateral cords and is histologically distinguishable from the latter throughout development. Intrasegmentally, median cord nuclei possess prominent nucleoli and many small chromatin granules surrounded by clear nuclear sap. This region forms what appear to be glial elements at the midline of the neuropile. Intersegmentally, a spherical clump of eight to twelve large nuclei develops surrounded by dark-staining granular cytoplasm. Each intersegmental clump migrates anteriorly into the preceding ganglionic region but degenerates soon after katatrepsis.

    Notes: The order of ossification of bones in the skeleton of Rana pipiens during larval growth and metamorphosis has been determined from observations on specimens fixed in 70% alcohol and stained with alizarin red S. The axial skeleton ossifies in a generally cephalo-caudal sequence, beginning with the parasphenoid bone at Taylor-Kollros stages IV-IX, followed by vertebrae (V-IX) and then the urostyle (IX-XIV). Exoccipitals (VII-IX), frontoparietals (XI-XII) and prootics (XIII-XVII) are additional cranial bones which successively ossify before metamorphosis. With the onset of metamorphosis at stage XVIII jawbones and rostral bones of the skull ossify in the following succession: premaxilla, maxilla, septomaxilla, nasal, dentary, angular, squamosal, pterygoid, prevomer, mentomeckelian, quadratojugal, palatine, columella, posteromedial process of “hyoid.” The sphenethmoid does not ossify until after metamorphosis.Ossification of limbbones begins with the femur or humerus at stages X-XII and progresses proximo-distally to the phalanges by stages XIII-XV. Carpals, however, do not ossify until stage XXV or after metamorphosis. The ilium of the pelvic girdle begins to ossify at stages X-XII, but the ischium is delayed until stages XX-XXIII. Scapula and coracoid of the pectoral girdle undergo initial ossification at stages XII-XIV, suprascapula and clavicle at stages XIII-XV. The sternum does not begin to ossify until stage XXIV. The possible role of thyroid hormones in stimulating osteogenesis is discussed.

    Notes: Using the Colcemid technique, the mitotic incidence (MI) was measured in the epidermis, lung, spleen, liver, kidney and ovarian follicular cells of metamorphosed, immature Xenopus laevis laevis. The MI was higher at 25°C than at 20°C, and there was a significant ranking correlation between organs in respect of the MI in different animals. With the exception of the liver and kidney, organ cultures showed good preservation for up to six days in vitro using a medium supplemented with 10% fetal calf serum, and values for MI comparable with or even higher than in vivo were obtained.

    Notes: Bovine parotid glands exhibit outstanding structural differences when compared with those of non-ruminant mammals. The acini are tortuous, branched and lined with cells of different heights, imparting a scalloped appearance to acinar lumina. Numerous microvilli, ca. 1.5 μ in length, extend into the lumina and intercellular canaliculi. Intercellular canaliculi measure ca. 3 μ in diameter and interweave in close association with intercellular tissue spaces. Intercellular tissue spaces are separated from the extraacinar spaces across a basal lamina only, whereas junctional complexes guard canaliculi from direct continuity with tissue spaces and/or extraacinar spaces. Flattened cytoplasmic lamellae extend from adjacent acinar cells and loosely interdigitate with one another across the tissue spaces. Acinar cells contain more mitochondria and less granular endoplasmic reticulum than parotid glands of non-ruminant mammals. Two types of secretory material, in the form of inclusions which vary in size and electron density, are present in the acinar cells. Intercalated ducts connect acini with striated ducts which in turn, empty into collecting ducts located between gland lobules. In terms of frequency of “basal infoldings” and numbers of mitochondria, striated ducts of calf parotid glands are not as well developed as those of certain other salivary glands. Myoepithelial cells are most often present at junctions of acini and intercalated ducts where they may attach to both acinar and ductal epithelium. Nerve “terminals” were not observed on the epithelial side of basement membranes in relation to the secretory cells.

    Notes: Development of the adult fly foot falls into clearly defined phases of cell division, growth, cuticle secretion and cell death. The pulvillus is composed dorsally of two giant cells and ventrally of thousands of minute tenent cells; the former produce the dorsal footpad cuticle and the latter the thousands of tenent hairs. Cell divisions are still occurring in future tenent cells when increase in size of the cells and in polyteny of the chromosomes is already occurring in the two dorsal cells. Also cell death occurs considerably earlier in the tenent cells, yet the sequential secretion of some six cuticular layers takes place at comparable times in dorsal and ventral cuticles. The cuticular layers formed are, in their order of secretion: ecdysial membrane, cuticulin of the epicuticle, dense exocuticle, homogeneous exocuticle, an intermediate layer, wax of the epicuticle, and an extensive mass of endocuticle. The ecdysial membrane seems to perform an important mechanical role in maintaining the shape of the delicate cytoplasmic projections of the tenent cells, before and during cuticle secretion, and in establishing the cuticular pattern of ridges in the dorsal cuticle. Comparisons are made with trichogen cell cuticle development and with tracheal cuticle. Tracheal, trichogen and dorsal footpad cuticle patterns are compared.Details of giant cell activity provide a working basis for studies of nuclear-cytoplasmic interactions, and the whole system raises many unsolved problems in the general field of cell differentiation and pattern formation.

    Notes: Dense bodies in the heart muscle of Venus mercenaria exist in two forms, free and attached. Free dense bodies morphologically consist of fascicles of thin filaments in parallel array and bound together by a dense, amorphous proteinaceous material. The binding of dense bodies to the cell membrane is effected via connecting filaments of the amorphous material of the dense body which join a condensation of morphologically similar material attached to the inner osmiophilic layer of the unit membrane. This composite of dense body, connecting filaments, membrane condensation and unit cell membrane has been termed collectively the attachment plaque. The attachment plaque is part of an extensive network on the cell surface which obligates that surface to a role in the contractile process. Moreover, this set of attachment plaques imposes an organization and an orientation to most thin filaments of the cell and preserves the contractile axis of the cell.

    Notes: Large quantities of colloidal particles were rapidly transported around the junctional complex into the lateral intercellular spaces by flounder renal epithelial cells. Large invaginations containing particles developed in the apical cytoplasm of cells when tracer particles were injected into the tubular lumens. Some membranebounded profiles containing particles appeared close to the lateral intercellular spaces. Particles were then found in the lateral intercellular spaces, between the basal plasmalemma and the basement membrane, and within the basement membrane. It is suggested that this transport might operate in situ and provide a morphological mechanism to explain a type of protein transport noted in the renal tubules of another flounder species by Maack and Kinter ('67). It is interesting to consider that perhaps a similar mechanism for the transport of intact proteins might also operate in mammalian nephrons as well.

    Notes: The ventral lobe of the adenohypophysis of the smooth dogfish, Mustelus canis, a viviparous elasmobranch, has been found to possess distinctive cells identified as basophils on the basis of staining properties. At maximum size, such a cell consists of a distended vesicle containing PAS-positive, AF-negative material surrounded by a thin envelope of cytoplasm and an eccentric nucleus. In earlier stages of these cells, vesicles are small or absent and granules in the more abundant cytoplasms are AF or Alcian-positive.Basophil numbers are high in pre-ovulation and mid-ovulation females, decrease markedly after the end of ovulation until embryos are about 1 cm long then increase greatly during August and September while embryos grow to 8 cm in length. Early high counts, if these basophils are gonadotropes, may be correlated with stimulation of the ovary and ovulation; reduced numbers suggest inhibition, possibly by ovarian hormones for a period, while subsequent increase may indicate indirect involvement in uterine conditions in this viviparous species. Conclusion are, admittedly, tentative as specimens were available during only a fraction of the ten month gestation period.

    Notes: The serigenous glands of a number of different sawfly larvae have been examined. Silk is secreted by pear-shaped cells which may be fused together in pairs or triplets, or exist simply as free, single cells. The cells are arranged in numerous groups attached to a pair of wide silk reservoirs by means of short canals. Each gland cell contains a large, irregular, ramifying nucleus and an intracellular duct which receives droplets of synthesised silk protein. Two modifications of this basic arrangement are described. It is suggested that the secretory cells are dermal gland cells, and that the intracellular duct is a rudimentary end-apparatus. A comparison is made between these and some other types of dermal gland cell found in insects.

    Notes: The structure of human labial salivary gland acini was studied by light and electron microscopy. Contrary to previous reports, these glands were pure mucous in nature; no serous elements were present. The acinar cells were found in all stages of maturation. Immature cells were characterized by an extensive and highly organized rough-surfaced endoplasmic reticulum. The Golgi complex was extremely prominent, consisting of stacks of flattened cisternae and swarms of small vesicles. Mucous droplets were almost completely absent. As secretory activity progressed, the endoplasmic reticulum involuted, while the Golgi cisternae became distended and formed many vacuoles. In mature mucous cells, the apical cytoplasm was filled with membrane-bounded mucous droplets, and the nucleus was displaced basally. The droplets frequently showed great variation in density from cell to cell, and even within the same cell they sometimes were quite heterogeneous. They were liberated from the acinar cells by an apocrine process, so that droplets with intact limiting membranes were often observed in the acinar lumen. These droplets soon lysed, their contents fusing into streams of mucus. Occasionally during apocrine secretion a mucous cell failed to reconstitute its apical surface, and its entire contents spilled into the acinar lumen.Unusual cytoplasmic inclusions were present in many of the acinar cells. These inclusions, which were surrounded by a single membrane, consisted of lipid droplets closely associated with bundles of fine filaments.

    Notes: Oogenesis and embryonic development in the marine sponge, Haliclona ecbasis, were studied using standard histological procedures.When the oocytes reach a diameter of about 30 μ, nurse cells begin to aggregate around them. Then when the oocytes are about 36 μ in diameter, they begin to engulf the associated nurse cells. Whole nurse cells are engulfed; and although the nucleus of the nurse cells disappears either as or soon after the cells are engulfed, the cytoplasm remains essentially unchanged. The accumulation of these cells within the oocytes most of the cytoplasm is nurse cell cytoplasm.During cleavage of the egg, the engulfed nurse cells are gradually fragmented, but otherwise appear unchanged. At the same time the cytoplasm of the nurse cells is progressively incorporated into that of the blastomeres by what appears to be fusion process. When the latter process is complete, the embryo develops into a typical parenchymula larva.

    Notes: The hands of the Hominoidea evidence four adaptive modes which distinguish the lesse apes (Hylobatidae), the orangutan (Pongo), the African apes (Pan), and man (Homo) from one another. The hands of the apes consist of compromises between manipulatory and locomotor functions because selection has operated for precision of grip as well as for special locomotor mechanisms. The human hand is almost totally devoted to manipulation. The hands of gibbons, orangutans and the African apes differ in many features that may be correlated with locomotion. The gibbons and siamang are specially adapted for ricochetal arm-swinging. The great apes possess morphological adaptations for arboreal foraging and climbing distinct from those of the hylobatids. In addition, the African apes have become secondarily adapted for terrestrial quadrupedal locomotion. Many features that distinguish the hands of chimpanzees and gorillas may be associated with the development of efficient knuckele-walking propulsive and support mechanisms.

    Notes: Ampullary organs of the transparent catfish, Kryptopterus bicirrhus, are present in large numbers on the head and in a regular pattern of lines on the body and fins. The organs lie in the epidermis, and have a pore that opens to the surface. Flattened cells form a roof and walls. On the floor of the organ there are a “sensory hillock,” composed of spherical receptor cells and columnar supporting cells, and a “secretory hillock” composed of columnar secretory cells. The receptor cells are nonciliated and have only afferent innervation. The organ cavity is filled with jelly. The organs are compared with ampullary organs of the weakly electric fish Eigenmannia, ampullae of Lorenzini of Raja, and small pit organs of Amiurus. Structural characteristics of the ampullary organs of Kryptopterus make them especially suitable for electrophysiological studies.

    Notes: A detailed topography of adrenergic innervation in invertebrates (lobster), low vertebrates (fish, amphibians, reptiles, birds), and nine species of mammals is presented. Flack and Hillarp's specific fluorescent histochemical method using freeze-dried material was used. Phylogenetically, adrenergic innervation appeared earlier under the ciliary epithelium and in the muscle than surrounding the vessels, and in all species many fibers were without any connection to the vessel walls. Adrenergic innervation was very rich in the dilator muscle extending toward the epithelium of the posterior chamber; a surprisingly rich network was found in the sphincter muscle and also in ciliary spaces of some species. Numerous fluorescent mast cells were visualized in the pecten of the bird eye and in the ciliary tissue of the sheep and cow.

    Notes: The cecropia moth oocyte accumulates proteins from the blood during vitellogenesis; the proteins reach the oocyte by an intercellular route, are taken in by pinocytosis, and become components of the protein yolk spheres. Different proteins vary greatly in the extent to which they are incorporated into the yolk spheres. One objective of the work described in this paper was to determine where the selectivity of protein uptake occurs. An autoradiographic analysis of the uptake of tritiated blood proteins injected into the hemocoel indicated that there are at least two sites of selectivity - one between the hemocoel and the intercellular spaces of the follicular epithelium that surrounds each oocyte, and another between the intercellular spaces and the yolk spheres. Another objective was to determine if only proteins from the blood are deposited in the protein yolk spheres. Studies of the incorporation of tritiated leucine provided evidence that the ovary itself synthesizes proteins that are deposited in the yolk spheres along with the blood proteins. Finally, evidence is presented that the various regions of the oocyte cortex are not equally active in the deposition of yolk.

    Notes: The mechanisms of development of posterior levels of neural tubes of chick embryos were analyzed by study of serial cross-sections of a continuous series of normal embryos between 40 to 72 hours of incubation. Two extirpation experiments were performed in ovo on other embryos of the same stages. Descriptive studies revealed the presence of an overlap zone in which two types of neural tube formation occurred. Open neural tube formation (by fusion of neural folds) occurred dorsally in this region; closed neural tube formation (by canalization of solid medullary cord tissue) occurred ventrally. Extirpation of the posterior end of the neural plate produced defects within the lumbosacral region, indicating that the posterior neural plate participates in the formation of the lumbosacrum, and that the overlap zone is therefore in the lumbosacral region. Extirpation of the prospective neural tissue in the anterior end of the tail bud indicated that only the most posterior levels of the neural tube originate exclusively by cavitation of the tail bud. In both extirpation experiments a neural tube formed independently within the tail bud tissue, indicating that formation of the neural tube in this region is not dependent upon direct continuity with neural tissue anteriorly.

    Notes: The interstitial cells of Pennaria tiarella differentiate exclusively from the central endoderm of the planula. Shortly after their appearance, most of the interstitial cells become cnidoblasts. Subsequently, as the larva transforms into a polyp, both cnidoblasts and interstitial cells migrate from the endoderm, through endoblast and mesoglea, into the ectoderm. It is suggested that some interstitial cells remain in the endoderm and differentiate into the gland and mucous cells of the polyp gastroderm.

    Notes: A series of dimensions of the shoulder girdle of primates has previously been chosen as being related to function in that anatomical region. Their examination by canonical analysis suggests that they do indeed reflect aspects of the use of the shoulder in locomotion in the different primates.Further analysis is here performed using the technique of neighborhood limited classification and this confirms the basic picture presented by the previous analysis. The new method also gives more detailed information about the grouping of the specimens; thus it endorses the reality of functional divisions that appear to exist in the data. And in addition the groupings reflect differences in the structure of the shoulder that correlate well with certain taxonomic subdivisions of the order. The method maintains contact with individual specimens throughout the analysis and is capable of placing them within groups, at the boundaries of groups, within the interfaces between groups, or as satellites to groups.The new method appears to have a part to play in the description of the relationships between biological objects that is complementary to that of canonical analysis. As the mathematical concepts upon which the two techniques are based differ totally, the risk that the results might be inherent in statistical assumptions is thus averted.

    Notes: In the annelid Enchytraeus albidus the ovary is composed of packets containing eight synchronously developing oocytes. Each oocyte in the packet is connected, via a bridge, to a common cytoplasmic mass. Developmental synchrony of oocytes within individual packets is probably related to the ooplasmic continuity.The young previtellogenic oocyte contains many polysomes, a few cisternae of smooth and rough endoplasmic reticulum, small Golgi complexes, and mitochondria. Many of the mitochondria are dumbbell-shaped and may thus represent division stages.Vitellogenesis is marked by the appearance of peripherally located lipid yolk and small, densely staining granules scattered throughout the ooplasm. There is an increase of smooth endoplasmic reticulum, mitochondria, and enlarged Golgi elements. Small multivesicular-like bodies, the early stages of developing yolk, are derived from the Golgi complex. The mature yolk sphere is bipartite and consists of (a) a variable number of dense spheres, the core bodies, which are produced in the ooplasm by the Golgi complex and which become embedded in (b) a dense matrix. The electron opaque tracer, horseradish peroxidase is incorporated into the oocyte and deposited in the matrix suggesting that this component of the yolk sphere is obtained by micropinocytosis. Enzyme digestions and various cytochemical techniques suggest that the core bodies are rich in carbohydrate, probably as glyco- or mucoproteins, and that the matrix is rich in lipid.

    Notes: The fine structure of the thyroid of the newt, Notophthalmus viridescens, was studied with the electron microscope. Specimens were injected I.P. with 30 μc of I131 and sample thryoids were examined at 12 hour intervals thereafter. The ultrastructure of the normal thyroid gland is described, and compared with that of the irradiated glands. The first visible ultrastructural change observed after injection of the radioiodin was a striking alteration of nuclear morphology. This effect was followed by an increase in the frequency of whorled lamellar structures, a decrease in the number of microvilli, and degeneration of the Golgi apparatus and endoplasmic reticulum. Further effects observed included an increase in the number of large cytoplasmic granules and a decrease in the number of smaller ones, the presence of autophagic vacuoles, and finally, an increase in the number of degenerated mitochondria.

    Notes: In order to obtain more direct evidence for the occurrence of myoblast fusion in the developing amphibian embryo, the following transplantations were performed in vitro. The nuclei of early embryos. Ambystoma tigrinum and A. maculatum, were labeled with tritiated thymidine. Portions of prospective somite areas from these labeled donors were grafted homoplastically and orthotopically into unlabeled hosts of the same, or nearly the same, stage. The stages employed were: neurula, early tail bud, and late tail bud. Hosts were raised until they had developed into more advanced larval forms, fixed, sectioned, and prepared for radioautographic processing according to the customary procedures.The histological preparations contained varying numbers of multinucleate myotubes of a “composite” nature: that is, individual myotubes contained labeled nuclei of the donor, side by side with unlabeled nuclei of the host. There was no doubt that the mononucleate myoblasts of the grafts had fused with those of the host species to form the mutlinucleate composite myotubes.In addition to the above determinations, the method of thymidine labeling has proven to be a satisfactory method of tracing, in the context of the intact organism, somitic cell derivatives up to the feeding larval stage. Mesenchymal cells from the grafted labeled somitic tissues were consequently found in: dermatomic, sclerotomic and intermyotomic locations; the matrix of the dorsal fin; the limb bud; the abdominal muscles.

    Notes: Implants were made into forelimbs of Triturus viridescens using fresh, frozen and boiled kidney and liver of T. viridescens and R. pipiens. Limbs were recovered at intervals up to 70 days post-implantation.Kidney implants from Wisconsin R. pipiens gave twice as many extensive accessory structures as did Vermont frog kidney. Total induction percentages, however, were similar.Quantitative and qualitative parameters for implant-induction of accessory structures were investigated. The decrease in antigenicity and increased rate of cytolysis of frozen implants resulted in increased similarity between frog and newt kidney in rate and pattern of breakdown and in rates of induction. Modification of rate and duration of the release of the stimulating factor from the implant did not result in induction by liver implants.No evidence was found for any increase in innervation prior to or coincident with blastema formation. Implantation and implant cytolysis may cause hypersensitivity of limb tissues to the normal innervation pattern or trophic stimuli from the implant may act with those from the injured limb tissues to produce growth.The general pattern of host reaction to the implanted material was studied and described.

    Notes: The genetics and anatomy of the homoeotic mutant labiopedia (lp) of Tribolium confusum is described. The mutant is the second known among the insects to affect the mouthparts. The two-segmented labial palps of the larva are completely replaced by leg-like appendages resembling the prothoracic legs and exhibiting the apex of the trochanter and all parts distal to the trochanter. The labial palps of the pupa and adult are likewise replaced by legs. The three-segmented palps of the adult are replaced by appendages closely similar to the prothoracic legs in many characters and exhibiting the apex of the coxa and all other parts of a normal walking leg. The legs have never been seen to move although they are supplied with labial nerves and an almost complete, though highly reduced, set of muscles. The labial appendages are invariably leg-like and well-developed, failing to show the range of variability which is commonly observed in homoeotic mutants. The leg-like form of the heteromorphic organ is in striking conformity with the appendicular origin of the palps.The lp gene is recessive and sex-linked, with lethal to semilethal effects. It is the third sex-linked gene discovered in Tribolium confusum and the first sex-linked homoeotic mutant known among the insects. Since the inheritance of lp is entirely in the manner of a sex-linked gene, it most probably is located on the original X chromosome, unless the translocated autosomal portion attached to the Y has become inert.

    Notes: Light and electron microscope studies of the digital lamellar setae of geckos and anoline lizards demonstrate that the free ends of the setae consist of flattened spatulas of less than 1 μ in width. The anoline setae are simple structures usually less than 30 μ in length and with a single terminal spatula to each seta. In contrast the setae of geckos are complex structures of about 100 μ in length, with numerous branchings, and having many spatulas per seta. The spatulas of Gekko and Aristelliger were smaller (0.2-0.4 μ in width) than the spatulas of Anolis (0.8 μ in width). The electron microscope studies indicate that the scales of lizards appear to be covered with small epidermal spines (1.5 μ long). The setae of anoles and geckos are considered to have evolved independently from these more primitive epidermal spines. It is further suggested that the mechanism that allows the lamellae to adhere to the substratum is a surface phenomenon. The spatulas provide a large surface that is in contact with the substratum and thus produces a large total frictional force. The α layer of the lizard stratum corneum can be readily identified in the lamellae. However, the structure of the β layer is not easily interpreted and there is evidence of a fibrous layer between the Oberhautchen and the α layer in the skin of the outer lamellar surface.

    Notes: The rat submaxillary salivary gland has five distinct parenchymal zones.1Acini consist of secretory and myoepithelial cells. An extensive network of canaliculi connect the many cells within an acinus to the main lumen. The fine structure of acinar secretory cells suggests that they are capable of great synthetic capacity; each cell having a large amount of ergastoplasm, many Golgi zones, and a great amount of secretory material. It is proposed that these cells are of the continually secreting type.2Intercalated ducts consist of cuboidal cells and myoepithelium. This segment connects the acini to the main conduit system of the gland. The fine structure of the cuboidal cells indicates that they are essentially nonsecretory.3The granular duct consists of three types of columnar cells; (a) dark narrow cells which contain many free ribosomes but no ergastoplasm or granules, (b) light granular cells which have varying amounts of ergastoplasm and granules, (c) dark granular cells which are full of granules while the other cell constituents including the nucleus, occupy a basal position. It is proposed that these three cells represent different secretory stages of the same cell type. This supports the interpretation that secretion in these cells is not continuous, but cyclic in nature.4The striated duct forms a small portion of the total gland parenchyma and consists of tall columnar cells with extensive infolding of the basal plasma membrane, relatively little ergastoplasm and very few granules. It seems likely that ion and water metabolism is a specialized function of this segment.5The excretory duct consists of three cell types: (a) tall columnar light cells, (b) dark columnar vesiculated cells and (c) small basal cells. The basal infoldings of these cells and the arrangement of many capillaries around these ducts suggests that this segment is primarily concerned with water transport.

    Notes: It was demonstrated that the germinal (surface) epithelium around the mouse ovary consists of simple squamous, cuboidal and columnar arrangements of cells. With the electron microscope it was shown that these cells have the usual cytoplasmic constituents such as the Golgi complex, mitochondria, ergastoplasm, and free ribosomes. Their nuclei are surrounded by a bilamellar envelope with coarse granular masses of nuclear material being distributed along its inner surface. From the surface of these cells extend numerous microprojections which usually are irregular in squamous cells, but are villus-like on the cuboidal and columnar forms. These microvilli appear to be simply extensions of the cell surface and apparently reflect the capacity of mesothelium for such specialization at its free surface. The intercellular boundaries are seen to consist of wavy, downward extensions of the plasma membrane or a complex irregular interdigitation of coarse lateral processes. The latter form is especially prominent between squamous cells. A membrane is present at the basal cell surface. The observations concerning the presence of microprojections and the nature of the intercellular boundaries were discussed in the light of the controversies on these subjects which appear in the early literature. The possible permeability of the germinal epithelium and its functional significance during maturation of the ovary were also discussed.

    Notes: Pre-copulatory behavior of male Aedes involves pursuit, tarsal contact, ventral orientation, and terminalial contact. Copulation itself involves seizure of the female's cerci by the male's claspers, extension of the male's paraprocts, erection of the aedeagus, and ejaculation. The male's ability to copulate is prevented by damaging or removing his seventh abdominal segment. If this segment is left intact, his freshly removed abdomen can copulate with a freshly isolated female's abdomen when their terminalia are rubbed together appropriately. The male's genital apparatus accurately “recognizes” the terminalium of an inseminated female, and forced-copulation cannot be induced. The claws at the ends of the male's claspers are inserted into the bases of the female's cerci. The thumbs of the apical paraprocts of the male fit into a temporary coital cavity within the upper vagina. The hooks of the male's apical paraprocts are inserted into the female's cloacal hollow. The teeth on the distal end of the aedeagus ratchet into the teeth on the dorsal vaginal valve and evert the upper vagina. The posterior ends of the seminal vesicles and accessory glands open and contractions of these organs release seminal material only into the bursa of the female.

    Notes: The fine structure of the mid-gut epithelium and regenerative cells of larvae of a moth (Ephestia kühniella) is described. Particular attention is paid to the absorptive and goblet cells and their lateral junctions; these features are discussed in terms of the digestive and regulatory functions of the epithelium. One digestive pathway has been investigated with the aid of ingested ferritin; intake of this marker by endocytosis and the evident involvement of Golgi vesicles in the transformation of endocytic vesicles into multivesicular bodies, together with the fate of the latter, are discussed in terms of the digestive function of this part of the alimentary tract and of the lysosome concept.

    Notes: The endoskeletal structure supporting the gill-books of Limulus polyphemus has been investigated by means of light and electron microscopy, chemical analysis and x-ray diffraction. This tissue is a cartilage which has significant correspondences with both vertebrate cartilage and plant tissues. Morphologically, the Limulus cartilage resembles certain cellular vertebrate cartilages with relatively scant matrix, and also certain plant parenchyme, collenchyme and sclerenchyme tissues. Of particular interest, was the observation that during cytoplasmic division, a phragmasome-like structure appears between the daughter cells of the dividing gill cartilage cells. This phragmasome-like structure appears to be a precursor of new matrix (cell-wall) formation between the young chondrocytes, in much the same fashion as its counterpart in plant tissues. Perichondrial cells and underlying chondrocytes contain lipid droplets, abundant glycogen and ribosomes, as do corresponding vertebrate cartilage cells. In some of the Limulus cells, glycogen and ribosomes appear to be admixed with lipid, forming aggregates in which all three materials are in intimate intraparticulate relationship. During molting, the number of ribosomes seen in chondrocytes increases. The tissue contains both hydroxyproline and hydroxylysine, and gives a weak x-ray diffraction pattern.

    Notes: A principal component analysis revealed that the two major components of mandibular shape variation among individuals within breeding groups of white-tailed deer in Canada and the United States involve contrasts between the mandible and the dentition and between the premolars and the molars. Size variation appeared to account for 34% of the total variation within the groups, and the two major shape variations accounted for 23% and 8% respectively.A canonical analysis was used to identify the major components of mandibular variation among the breeding groups and to provide measures of the proportion of the total variation accounted for by each component. Among male groups, size variation was associated with latitude, and the major shape variation was closely associated with longitude, so a bivariate plot of the first two canonical variates reflected the general geographic orientation of the populations.The mandibular size in a Tennessee population that descended from Wisconsin and Michigan ancestors appears to have not decreased appreciably in the more southerly habitat after introduction more than 20 years ago. Changes in range conditions in eastern Upper Michigan over the past 30 years have not influenced local mandibular morphology as reflected by the first two canonical variates. Regardless of general smallness of individuals, the mandibular morphology of the deer from the Cross Timbers area in Kansas appears to approximate more closely that of northern populations than that of the more southerly populations from Oklahoma and Texas.Sexual dimorphism on the first two canonical axes was observed.

    Notes: Structural and functional changes have been correlated during metamorphic degeneration of a single muscle fiber, the plantar retractor of G. mellonella, its axon, and their junctions to determine which features persist as long as muscle contractility. Changes commence simultaneously in muscle and nerve near cuticular attachments, and spread towards the center. Alterations associated with the muscle, including appearance of collapsed tracheoles and mitochondria with dense bodies, begin late in the last larval instar. Within 12 hours after pupal ecdysis some tracheolar withdrawal occurs, sarcoplasmic reticulum becomes reduced, and many mitochondria have dense bodies, dense membranes, or are enlarged. By 17-19 hours primary myofilaments and striations begin to disappear, microtubules and autophagic vacuole-like bodies appear, and phagocytes invade the muscle. It remains partially contractile upon electrically stimulating its nerve, the ventral nerve, until these changes spread throughout the fiber.Neuromuscular junction changes, including appearance of dense mitochondria and isolation bodies, begin late in the last larval instar. Junctions become fewer, and none remain in those muscle areas where tracheoles, sarcoplasmic reticulum, and primary myofilaments have disappeared.Preliminary studies on nerve discharge activity to the muscle suggest that nerve silence occurs at approximately the time when the muscle loses all contractility. In some axons isolation bodies appear and neurotubules are lost, other axons remain unchanged, and new ones develop later in the pupal state. Phagocytes invade the neural lamella and it disappears in the late pupa, but it reappears in the adult.The adult ventral nerve has over three times more axons and a thinner layer of glial cells than the larval nerve.

    Notes: Ampullary receptor organs of the South American weakly electric gymnotid fish Eigenmannia virescens consist of a pore at the surface of the skin, a canal through the epidermis, and the expanded basal end of the canal in the corium. The cavity of the organ contains a jelly that is filled with fine fibers. The canal wall consists of three to six layers of flattened cells that appear to be derived from the adjacent skin. Along the lumen of the organ the cells are joined by tight junctions. Usually there are four spherical receptor cells in the base of the organ. They are innervated by single neural terminals. These organs are compared to tuberous receptor organs found in the same species, and the functional significance of the fine structure observed in these cells is discussed.

    Notes: The campaniform sensilla on halteres of Drosophila were studied by electron microscopy in order to establish the relationships of functional elements in the sensory system. The surface of the sensillum consists of an oval cuticular cap membrane which may contain resilin, the rubberlike protein. A border of denser cuticle rings the cap membrane, and extending down around the neural process is a third type of cuticle filled with a fourth light fibrous type. The four cuticular components form a system for displacement of the neural process. The neural process is differentiated into a terminal fan-shaped structure projecting from a bulbous dilatation which tapers to a neck region ending proximally with two basal bodies. The neural process is packed with microtubules. Surrounding the dendrite is an inner enveloping cell, attached to the basal body region by septate desmosomes and by desmosomes to which microtubules of the enveloping cell are applied. An outer enveloping cell surrounds the inner one. The tip of the neural process is covered with a dense secretion which is tightly bound to the cap membrane. The dense secretion is surrounded by an extracellular fluid which might be compressed hydraulically by the cuticular system. The stimulus of cuticular distortion could thus be transmitted to the neural process which may be displaced between its fixed ends.

    Notes: The heteropteran cibarium forms a sucking pump which conveys fluid foods into the pharynx. The food pumps of Hydrocorisae have the additional function of grinding or filtering particulate matter; they contain ridges, hairs, and sclerotized processes which have probably evolved at least twice among the hydrocorisine families. Aphelocheirus, like the Naucoridae, possesses a modified anteclypeus and a tripartite type of food pump. The main sucking action occurs in the pump's anterior and posterior regions, while the middle region is specialized for grinding and filtering. The anteclypeus has broadened and fused with other parts of the cranium, and is thus braced against the pull of the powerful cibarial dilator muscles. In the Naucoridae the three regions of the pump have the same functions as those of Aphelocheirus. The pumps of the five naucorid genera here studied are structurally very similar to each other but differ considerably from that of Aphelocheirus. Cibarial morphology, as well as respiratory differences, thus supports the contention that Aphelocheirus is not a member of the Naucoridae but should be placed in a separate family.

    Notes: Drosophila melanogaster homozygous for the second chromosomal, recessive lethal gene 1(2)gl form puparia much later than wild type (+) and are unable to metamorphose. Implantation of + ring glands accelerates puparium formation by 1(2)gl hosts. The ring gland is a compound structure containing the prothoracic glands (pg), corpus allatum (ca), and corpus cardiacum (cc). An electron microscopic study demonstrates that in both the pg and ca the most common subcellular component is smooth surfaced endoplasmic reticulum (ser). In + flies the amount of ser per pg increases by ten times during the four hour prepupal period. In 1(2)gl flies which have spent two days in the prepupal period the pg looks juvenile and contains only 1% the amount of ser per cell found in the + prepupal pg. The ca cells look alike in both + and 1(2)gl individuals, and the cortical cells of the cc in both contain abundant neurosecretory spheres. We suggest that the + allele of 1(2)gl indirectly influences the synthesis of ser by the pg and that this is the site where dietary cholesterol is transformed to ecdysone.
