    Description: Separation and purification are critical industrial processes for separating components of chemical mixtures, and these processes account for about half of industrial energy usage (1). Gas mixtures of compounds with very similar physical properties are particularly difficult to separate. On pages 137 and 141 of this issue, Cadiau et al. (2) and Cui et al. (3), respectively, show that microporous materials can be designed to have high adsorption capacity and selectivity for particular hydrocarbons, enabling energy-efficient separation. Author: Jerry Y.     Description: Biomass-degrading microorganisms use lytic polysaccharide monooxygenase (LPMO) enzymes to help digest cellulose, chitin, and starch. By cleaving otherwise inaccessible crystalline cellulose chains, these enzymes provide access to hydrolytic enzymes. LPMOs are of interest to biotechnology because efficient depolymerization of cellulose is a major bottleneck for the production of biologically based chemicals and fuels. On page 1098 of this issue, Kracher et al. (1) compare LPMO-reducing substrates in fungi from different taxonomic groups and lifestyles, based on both biochemical and genomic evidence. The results provide insights into reductive activation of LPMO that are important for developing more efficient industrial enzymes for lignocellulose biorefineries. Author: Angel T. Martínez

    Description: Water plays a central role in scientific disciplines ranging from geology to astronomy to biology. Yet it is an extraordinarily dif cult liquid to understand because of its complex, ever-changing patterns of hydrogen bonds. Studies of small water clusters have provided important insights into the concerted hydrogen-bond motions that can occur in water. These studies are also crucial for developing an accurate potential function for simulating the properties of liquid water and ice (1). On page 1310 of this issue, Richardson et al. (2) provide evidence for a concerted type of motion in which two hydrogen bonds in a water cluster are broken simultaneously (see the figure). The results have implications for many areas of scientific study, including the chemistry of polar solvents, the conformations of proteins, and the dissolution of ions in minerals. Author: David C.     Description: Lower olefins, particularly ethylene (C2H4), propylene (C3H6), and butylene (C4H8), are important intermediates in the manufacture of products such as plastics, solvents, paints, and medicines. They are produced worldwide in amounts exceeding 200 million tons per year (see the photo) (1), mostly from crude oil. More recent approaches use methanol or synthesis gas (syngas; a mixture of carbon monoxide and hydrogen) as feedstocks, but capital investments are high and/or selectivities to lower olefins limited. A bifunctional catalyst reported by Jiao et al. on page 1065 of this issue (2) enables the direct conversion of synthesis gas to lower olefins with a surprisingly high selectivity.     Description: Effective differentiation of prochiral carbon–hydrogen (C–H) bonds on a single methylene carbon via asymmetric metal insertion remains a challenge. Here, we report the discovery of chiral acetyl-protected aminoethyl quinoline ligands that enable asymmetric palladium insertion into prochiral C–H bonds on a single methylene carbon center. We apply these palladium complexes to catalytic enantioselective functionalization of β-methylene C–H bonds in aliphatic amides. Using bidentate ligands to accelerate C–H activation of otherwise unreactive monodentate substrates is crucial for outcompeting the background reaction driven by substrate-directed cyclopalladation, thereby avoiding erosion of enantioselectivity. The potential of ligand acceleration in C–H activation is also demonstrated by enantioselective β-C–H arylation of simple carboxylic acids without installing directing groups.

    Description: The meaning of language is represented in regions of the cerebral cortex collectively known as the 'semantic system'. However, little of the semantic system has been mapped comprehensively, and the semantic selectivity of most regions is unknown. Here we systematically map semantic selectivity across the cortex using voxel-wise modelling of functional MRI (fMRI) data collected while subjects listened to hours of narrative stories. We show that the semantic system is organized into intricate patterns that seem to be consistent across individuals. We then use a novel generative model to create a detailed semantic atlas. Our results suggest that most areas within the semantic system represent information about specific semantic domains, or groups of related concepts, and our atlas shows which domains are represented in each area.     Description: CRISPR-Cas9 nucleases are widely used for genome editing but can induce unwanted off-target mutations. Existing strategies for reducing genome-wide off-target effects of the widely used Streptococcus pyogenes Cas9 (SpCas9) are imperfect, possessing only partial or unproven efficacies and other limitations that constrain their use. Here we describe SpCas9-HF1, a high-fidelity variant harbouring alterations designed to reduce non-specific DNA contacts. SpCas9-HF1 retains on-target activities comparable to wild-type SpCas9 with 〉85% of single-guide RNAs (sgRNAs) tested in human cells. Notably, with sgRNAs targeted to standard non-repetitive sequences, SpCas9-HF1 rendered all or nearly all off-target events undetectable by genome-wide break capture and targeted sequencing methods. Even for atypical, repetitive target sites, the vast majority of off-target mutations induced by wild-type SpCas9 were not detected with SpCas9-HF1. With its exceptional precision, SpCas9-HF1 provides an alternative to wild-type SpCas9 for research and therapeutic applications.     Description: Transected axons fail to regrow in the mature central nervous system. Astrocytic scars are widely regarded as causal in this failure. Here, using three genetically targeted loss-of-function manipulations in adult mice, we show that preventing astrocyte scar formation, attenuating scar-forming astrocytes, or ablating chronic astrocytic scars all failed to result in spontaneous regrowth of transected corticospinal, sensory or serotonergic axons through severe spinal cord injury (SCI) lesions. By contrast, sustained local delivery via hydrogel depots of required axon-specific growth factors not present in SCI lesions, plus growth-activating priming injuries, stimulated robust, laminin-dependent sensory axon regrowth past scar-forming astrocytes and inhibitory molecules in SCI lesions. Preventing astrocytic scar formation significantly reduced this stimulated axon regrowth. RNA sequencing revealed that astrocytes and non-astrocyte cells in SCI lesions express multiple axon-growth-supporting molecules.     Description: Wearable sensor technologies are essential to the realization of personalized medicine through continuously monitoring an individual's state of health. Sampling human sweat, which is rich in physiological information, could enable non-invasive monitoring. Previously reported sweat-based and other non-invasive biosensors either can only monitor a single analyte at a time or lack on-site signal processing circuitry and sensor calibration mechanisms for accurate analysis of the physiological state. Given the complexity of sweat secretion, simultaneous and multiplexed screening of target biomarkers is critical and requires full system integration to ensure the accuracy of measurements. Here we present a mechanically flexible and fully integrated (that is, no external analysis is needed) sensor array for multiplexed in situ perspiration analysis, which simultaneously and selectively measures sweat metabolites (such as glucose and lactate) and electrolytes (such as sodium and potassium ions), as well as the skin temperature (to calibrate the response of the sensors). Our work bridges the technological gap between signal transduction, conditioning (amplification and filtering), processing and wireless transmission in wearable biosensors by merging plastic-based sensors that interface with the skin with silicon integrated circuits consolidated on a flexible circuit board for complex signal processing. This application could not have been realized using either of these technologies alone owing to their respective inherent limitations. The wearable system is used to measure the detailed sweat profile of human subjects engaged in prolonged indoor and outdoor physical activities, and to make a real-time assessment of the physiological state of the subjects.     Description: Medulloblastoma is a highly malignant paediatric brain tumour, often inflicting devastating consequences on the developing child. Genomic studies have revealed four distinct molecular subgroups with divergent biology and clinical behaviour. An understanding of the regulatory circuitry governing the transcriptional landscapes of medulloblastoma subgroups, and how this relates to their respective developmental origins, is lacking. Here, using H3K27ac and BRD4 chromatin immunoprecipitation followed by sequencing (ChIP-seq) coupled with tissue-matched DNA methylation and transcriptome data, we describe the active cis-regulatory landscape across 28 primary medulloblastoma specimens. Analysis of differentially regulated enhancers and super-enhancers reinforced inter-subgroup heterogeneity and revealed novel, clinically relevant insights into medulloblastoma biology. Computational reconstruction of core regulatory circuitry identified a master set of transcription factors, validated by ChIP-seq, that is responsible for subgroup divergence, and implicates candidate cells of origin for Group 4.     Description: Although ammonia (NH3) is made on a vast scale for use in fertilizers, its use as a chemical feedstock or as an energy carrier is much more limited. Many reactions that occur easily with its substitution products (amines) are sluggish for NH3, in part because of the difficulty of activating the N-H bond. For fuel cells, NH3 is attractive because it does not generate greenhouse gases, as do methanol and methane (1), and is more easily stored than hydrogen (H2). Amine-containing organic molecules are used in pharmaceutical and materials applications, and accessing these structures directly from ammonia could limit the generation of by-products during their synthesis (2). Bringing NH3 up to speed for these applications will require both the development of catalysts that can activate the strong N–H bond of ammonia and a fundamental understanding of the N–H bond cleavage step. On page 730 of this issue, Bezdek et al. (3) report a molybdenum complex capable of weakening the N–H bond of NH3 and releasing a H atom to generate H2 under mild conditions. Author: Jessica Hoover

    Description: Direct methods for stereoselective functionalization of sp 3 -hybridized carbon–hydrogen [C(sp 3 )–H] bonds in complex organic molecules could facilitate much more efficient preparation of therapeutics and agrochemicals. Here, we report a copper-catalyzed radical relay pathway for enantioselective conversion of benzylic C–H bonds into benzylic nitriles. Hydrogen-atom abstraction affords an achiral benzylic radical that undergoes asymmetric C(sp 3 )–CN bond formation upon reaction with a chiral copper catalyst. The reactions proceed efficiently at room temperature with the benzylic substrate as limiting reagent, exhibit broad substrate scope with high enantioselectivity (typically 90 to 99% enantiomeric excess), and afford products that are key precursors to important bioactive molecules. Mechanistic studies provide evidence for diffusible organic radicals and highlight the difference between these reactions and C–H oxidations mediated by enzymes and other catalysts that operate via radical rebound pathways.

    Description: Oxygen electrochemistry plays a key role in renewable energy technologies such as fuel cells and electrolyzers, but the slow kinetics of the oxygen evolution reaction (OER) limit the performance and commercialization of such devices. Here we report an iridium oxide/strontium iridium oxide (IrO x /SrIrO 3 ) catalyst formed during electrochemical testing by strontium leaching from surface layers of thin films of SrIrO 3 . This catalyst has demonstrated specific activity at 10 milliamps per square centimeter of oxide catalyst (OER current normalized to catalyst surface area), with only 270 to 290 millivolts of overpotential for 30 hours of continuous testing in acidic electrolyte. Density functional theory calculations suggest the formation of highly active surface layers during strontium leaching with IrO 3 or anatase IrO 2 motifs. The IrO x /SrIrO 3 catalyst outperforms known IrO x and ruthenium oxide (RuO x ) systems, the only other OER catalysts that have reasonable activity in acidic electrolyte.

    Notes: Es wurde der Zusammenhang der Zeitdauer des Haltepunktes auf der Abkühlungskurve mit der Schmelzwärme von Metallen und ihren Verbindungen ermittelt. Mit Hilfe dieser Feststellung konnten dann die Schmelzwärmen der Metallverbindungen, sowie von Mg und A1, bestimmt werden. Zur Kontrolle wurden dieselben nochmals nach dem kalorimetrischen Verfahren bestimmt.

Die Atomgewichte der ersten 27 Elemente, beginnend mit Helium, sind nicht Vielfache des Atomgewichts von Wasserstoff mit einer ganzen Zahl, wie es sein müßte, wenn PROUTS ursprüngliche Hypothese in ihrer numerischen Form richtig wäre. Dies kann ausgedrückt werden durch die Feststellung, daß die Atomgewichte, bezogen auf Wasserstoff, nicht ganze Zahlen sind. Wenn man jedoch diese Atomgewichte kritisch prüft, so findet man, daß sie von den entsprechenden ganzen Zahlen um einen fast konstanten Prozentsatz abweichen, daß die Abweichung negativ ist und im Mittel den Wert -0.77%, besitzt.

Der Grundgedanke dieser zweiten Mitteilung uber die Atomstruktur ist folgender: Das Gesetz, welches bei den Beziehungen von Atomgewicht und Valenz zwischen Gliedern einer jeden radioaktiven Reihe Anwendung findet, trifft auch für die leichteren Atome zu. Bei einer radioaktiven Reihe bedingt der Verlust eines Teilchens mit einer Masse von 4 eine Abnahme der Wertigkeit um 2 und verschiebt so das Element um 2 Gruppen des periodischen Systems nach links, während gleichzeitig die Atomzahl um 2 vermindert wird. Wenn dies auch für die leichteren Elemente, beginnend mit Helium, zutrifft, so müßte die Zunahme um das Gewicht eines Heliumatome für jeden Anwachs um 2 in der Atomzahl die Atomgewichte der Elemente liefern, die zu den Gruppen mit geraden Zahlen gehören. Die nach diesem Verfahren abgeleiteten Atomgewichte stimmen im ganzen mit den experimentell festgestellten Werten überein, woraus hervorgeht, daß die Theorie den Tatsachen entspricht.

Die in der vorliegenden Untersuchung ausgearbeitete Lösungsmethode zur relativen Atomgewichtbestimmung isotoper Bleiarten beruht auf der Dichtebestimmung gesättigter Nitratlösung der betreffenden Bleiarten. Die theoretische Grundlage der Methode ist auf folgenden zwei Voraussetzungen aufgebaut: a)daß die molare Zusammensetzung der gesättigten Lösungen d. h. das Verhältnis Mole Salz/Mole Wasser, für die Nitrate verschiedener Bleiarten gleich ist;b)daß die in Mol/Liter ausgedrückte Löslichkeit für die Nitrate verschiedener Bleiarten den gleichen Wert hat.Die zweite Voraussetzung folgt aus der ersten, wenn die Molekularvolumina der Nitrate der verschiedenen Bleie gleich sind und die Volumänderung beim Lösungsvorgange bei allen den gleichen Wert hat.

Der Unterschied beträgt bei der besten Probe nur 0.2%.3Versuche, diese Fehlergrenze möglichst klein zu halten, bzw. ganz aufzuheben, scheiterten daran, daß infolge Verflüchtigung von Rutheniumtetroxyd der Endpunkt der Oxydation bei weiter fortgesetztem Glühen nicht festgestellt werden konnte.4Ein Einfluß der Glühtemperatur auf die gesamte Sauerstoffaufnahme scheint nicht zu bestehen, denn man erhält bei allen Temperaturen von 700 bis 1000° als Reaktionsprodukt dieselbe Verbindung, Rutheniumdioxyd.5Die Geschwindigkeit der Oxydation ist von der Temperatur in starkem Maße abhängig.6Die Verflüchtigung von Ruthenium in Form des Tetroxyds beginnt bei Temperaturen über 600° und steigert sich beträchtlich mit Erhöhung der Temperatur, so daß sie bei 1200° ungefähr 4000 mal so groß ist als bei 700°.7Teile des flüchtigen Tetroxyds zerfallen an den kälteren Stellen des Apparates. Man beobachtet in den Glühröhren da, wo eine Temperatur von ungefähr 500° herrscht, die ringförmige Abscheidung von blauen Dioxydkriställchen und an noch kühleren Stellen die Abscheidung von amorpher, schwarzer Substanz. Unter besonderen Versuchsbedingungen werden selbst in der Vorlage kleine, blaue, dünne Gebilde von Rutheniumdioxyd abgeschieden, die vollkommen einheitlich sind und rauhe Oberfläche zeigen.8Metallisches Ruthenium wird durch Glühen bei ungefähr 800° weitgehend verändert, verliert seine feinblätterige Form und wird kompakt und matt.9Das bei bestimmten, hohen Temperaturen geglühte Ruthenium hat nach der Reduktion im Wasserstoffstrome bei Wiederholung der Oxydation unter den gleichen Verhältnissen eine geringere Oxydationsgeschwindigkeit als zuvor.
