    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Verglichen mit anderen Tiergruppen, scheint die artliche Bestandsaufnahme der rezenten Vögel nahezu abgeschlossen zu sein. Doch ist das System der Vögel weiterhin umstritten und mit vielen Neuerungen konfrontiert. Die Gründe dafür liegen hauptsächlich in neuen, vor allem molekularbiologischen Methoden und in den unerwartet reichen Fossilfunden der jüngsten Zeit. Als Beispiele werden Altgaumenvögel, Kranichvögel, Ibisse, Flamingos, Mausvögel, Hopfe und Sperlingsvögel kurz behandelt. Die hier erzielten Fortschritte lassen die Befürchtung Stresemanns, die Großsystematik der Vögel sei mit den vorhandenen Methoden phylogenetisch nicht interpretierbar, zunächst als unbergründet erscheinen. Doch erwachsen einer solchen Interpretation andere Hindernisse, deren Bedeutung bisher zu wenig beachtet wurde, nämlich Parallelentwicklungen, die viel verbreiteter sind als gemeinhin angenommen. Ihre Häufigkeit lässt sich sogar mit evolutionsbiologischen Argumenten begründen. Es ist deshalb nicht zu erwarten, dass die Diskussionen um das „richtige“ System bald verstummen. Um dennoch die Eindeutigkeit der Information in nicht-systematischen Veröffentlichungen zu wahren, wird empfohlen ein etabliertes Referenzsystem auf Zeit zu wählen.
    Notizen: Summary Unlike in most animal classes the inventory of extant species of the class Aves seems to be almost complete. Nevertheless avian systematics is challenged by many novelties and seems far from being settled. This is caused mainly by the application of novel methods of molecular analysis to phylogenetic problems and by the unexpectedly rich fossil record collected within the last 10–20 years. Examples from the Palaeognathae, Gruiformes, Threskiornithidae, Phoenicopteridae, Coliiformes, Upupiformes and Passeriformes are briefly treated. The progress in the field seems to disprove Stresemann's pessimistic view that the phylogeny of higher categories (orders) cannot be reconstructed by the available methods. However, phylogenetic interpretations are impeded by obstacles not considered by Stresemann and highly underestimated in most cases, namely by multiple independent developments leading to identical features. Frequent parallel developments are to be expected for theoretical evolutionary reasons. The diagnosis of such homoplasies can be extremely difficult or even impossible. Therefore we cannot expect the discussion about the “best” system of birds to end in the near future. Considering this dynamic situation in systematics, it is recommended to maintain unambiguousness of information in not strictly systematic publications by refering to a well established system as a temporally limited reference.

    Notizen: Abstract The peformance of the parasitoid Anagyrus kamali Moursi [Hymenoptera: Encyrtidae], as a function of host density, temperature, and photoperiod was investigated with the objective to optimize a mass-rearing system in the context of a biological control program. The number of hosts parasitized at densities varying from 2–100 hibiscus mealybug (HMB), Maconellicoccus hirsutus Green [Homoptera: Pseudococcidae], corresponded to a type II-III functional response in fixed-time conditions and a type III in variable-time conditions. Twenty-six percent of the oviposited eggs led to progeny emergence with a sex ratio of 0.49±0.102 (M/F), regardless of host density. Fecundity and oviposition period under six abiotic combinations (i.e., two temperatures (26±2 °C and 32±2 °C) and three photoperiods (L0:D24, L12:D12, L24:D0)) were measured. Lifetime fecundity and reproductive life were significantly affected by temperature and photoperiod conditions. Optimum female parasitoid lifetime fecundity was attained at 26±2 °C, L0:D24 with an average of 116.1±17.43 eggs. At 32±2 °C, L24:D0 and L12:D12, an average of 79.4±34.57 and 85.8±35.81 eggs were laid, respectively. Reproductive longevity was maximal at 26±2 °C, L0:D24 with 12±4.85 days of oviposition. Because the parasite A. kamali can be reared optimally without light, this may save tremendous energy costs.

    Notizen: Abstract Temperature dependencies were established for the egg-to-mummy and mummy-to-adult phases, for mummy mortality, and for parasitism of Aphidius ervi Haliday, Aphidius rhopalosiphi De Stefani-Perez, and Praon volucre (Haliday) (Hymenoptera, Aphidiidae), three parasitoids of Sitobion avenae (Fabricius) (Homoptera, Aphididae), at 8 °C, 12 °C, 16 °C, 20 °C, and 25 °C on winter wheat (cv. Haven). A physiological model described temperature-dependent development over the full temperature range, whereas a linear model was fitted for data above 8 °C and used to estimate the lower temperature thresholds and day-degrees (° D) required for development. The thresholds for A. ervi were 2.2 °C for egg-mummy development and 6.6 °C for mummy-adult development, those for A. rhopalosiphi were 4.5 °C and 7.2 °C, and those for P. volucre were 3.8 °C and 5.5 °C. The time to develop into mummies and adults differed significantly between the three species: A. ervi development into mummies required an average of 159 ° D, while development into adults took an average of 73 ° D. The corresponding average times required for A. rhopalosiphi and P. volucre to develop mummies were 124° D and 126° D, while their development into adults required an average of 70° D and 150° D, respectively. Mummy mortality was 25–35% at 8 °C and less at the higher temperatures tested, but began to increase again at 25 °C, showing a quadratic relationship between mortality and temperature. Parasitization was very low or, in the case of P. volucre, absent up to 12 °C and thereafter increased with increasing temperature. The relationship between parasitization, recorded as percent aphids mummified, and temperature was linear at the temperatures tested and depended on species. A. ervisuperparasitized 11.1% aphids at 20 °C and 16.6% aphids at 25 °C, whereas superparasitism was low in A. rhopalosiphi and absent in P. volucre. From 16 °C to 25 °C the P. volucre sex ratio increased. For A. ervi and A. rhopalosiphi there was no trend with temperature, but at 20 °C and 25 °C it was close to even. Field data for 1996 and 1997 allowed for a comparison of actual and expected emergence of overwintering mummies. In both years, parasitoids were predicted to have emerged from overwintering mummies well in advance of the onset of aphid infestation, and more than a month earlier than the first parasitized aphids were found in winter wheat. Observations from trap plants in other crops supported the predictions of the models. Other factors that can affect biological control by cereal aphid parasitoids are discussed.

    Notizen: Abstract Using computer simulations I studied the simultaneous effect of variable environments, mutation rates, ploidy, number of loci subject to evolution and random and assortative mating on various reproductive systems. The simulations showed that mutants for sex and recombination are evolutionarily stable, displacing alleles for monosexuality in diploid populations mating assortatively under variable selection pressure. Assortative mating reduced excessive allelic variance induced by recombination and sex, especially among diploids. Results suggest a novel adaptive value for sex and recombination. They show that the adaptive value of diploidy and that of the segregation of sexes is different to that of sex and recombination. The results suggest that the emergence of sex had to be preceded by the emergence of diploid monosexual organisms and provide an explanation for the emergence and maintenance of sex among diploids and for the scarcity of sex among haploid organisms.

    Notizen: Abstract Herbivorous and carnivorous arthropods use plant volatiles when foraging for food. In response to herbivory, plants emit a blend that may be quantitatively and qualitatively different from the blend emitted when intact. This induced volatile blend alters the interactions of the plant with its environment. We review recent developments regarding the induction mechanism as well as the ecological consequences in a multitrophic and evolutionary context. It has been well established that carnivores (predators and parasitoids) are attracted by the volatiles induced by their herbivorous victims. This concerns an active plant response. In the case of attraction of predators, this is likely to result in a fitness benefit to the plant, because through consumption a predator removes the herbivores from the plant. However, the benefit to the plant is less clear when parasitoids are attracted, because parasitisation does usually not result in an instantaneous or in a complete termination of consumption by the herbivore. Recently, empirical evidence has been obtained that shows that the plant's response can increase plant fitness, in terms of seed production, due to a reduced consumption rate of parasitized herbivores. However, apart from a benefit from attracting carnivores, the induced volatiles can have a serious cost because there is an increasing number of studies that show that herbivores can be attracted. However, this does not necessarily result in settlement of the herbivores on the emitting plant. The presence of cues from herbivores and/or carnivores that indicate that the plant is a competitor- and/or enemy-dense space, may lead to an avoidance response. Thus, the benefit of emission of induced volatiles is likely to depend on the prevailing faunal composition. Whether plants can adjust their response and influence the emission of the induced volatiles, taking the prevalent environmental conditions into account, is an interesting question that needs to be addressed. The induced volatiles may also affect interactions of the emitting plant with its neighbours, e.g., through altered competitive ability or by the neighbour exploiting the emitted information. Major questions to be addressed in this research field comprise mechanistic aspects, such as the identification of the minimally effective blend of volatiles that explains the attraction of carnivores to herbivore-infested plants, and evolutionary aspects such as the fitness consequences of induced volatiles. The elucidation of mechanistic aspects is important for addressing ecological and evolutionary questions. For instance, an important tool to address ecological and evolutionary aspects would be to have plant pairs that differ in only a single trait. Such plants are likely to become available in the near future as a result of mechanistic studies on signal-transduction pathways and an increased interest in molecular genetics.

    Notizen: Abstract Populations of the rapeseed pollen beetle Meligethes aeneus F. (Col., Nitidulidae) from areas with 0–16 years of history of intensive rapeseed growing were compared for key ecological characters. During the first 16 years of rapeseed cultivation the reproductive success of M. aeneus increased 200–300% over that of the beetles living on the natural host plants, cruciferous weeds. The increase was linear over time and statistically highly significant, and it did not appear to be related to food quality or to the size of the beetles. During the same period the tolerance to intraspecific competition decreased, possibly due to the relative absence of such competition on the new crop. Furthermore, the optimum population density for M. aeneus to maximize the size of its next generation on summer turnip rape was determined to be 0.5–1.0 beetles/plant, which is slightly below the economic threshold for chemical control (1 beetle/plant). Therefore the practical protection of the rapeseed yield also ensures the highest possible pest population size for the next year. These mechanisms may in part explain the particular noxiousness of the species as a pest all over Europe. In general these data show that after the introduction of a new crop plant into a region, significant changes during the recruitment process in a pestiferous insect may take place, contributing to the future pest status of the insect. It is suggested that such genetic and ecological changes in insects may be a more common mechanism than previously thought in initiating and sustaining pest outbreaks, and that conventional pest management methods may enhance that effect.

    Notizen: Abstract D. birchii and D. serrata, two endemic Australian Drosophila species, have a copulatory courtship. The males of these species begin to court the female after mounting her and often go on with the courtship after the copulation is over. In the present paper we have described behavioral interactions between the male and the female and analyzed acoustic signals produced by the flies during courtship. Species differences were more pronounced in female than in male behavior. Variation within the species was obvious in the relative proportions of time the flies spent in different behaviors. Even though courtship took place nearly solely during copulation, some remains of precopulatory courtship were observed in both species. It is suggested that copulatory courtship exhibited by D. birchii and D. serrata flies is a derived rather than a primitive character.

    Notizen: Abstract Adsorption refrigerators are a particular type of refrigerator in which compression is avoided, and in a sense replaced by adsorption. No mobile parts are needed; the energy input, instead of being mechanical, is thermal and is used to achieve desorption. Such machines have a cyclic operation, made of successive adsorption/evaporation and of desorption/condensation steps. The transient operation of adsorption refrigerators is a relatively recent subject of research. The modeling of the adsorber is the key point of such studies, because of the complex coupled heat and mass transfer phenomena that occur during the cycle. The present work therefore presents a study of an annular type adsorber which is intended to account for transient temperatures observed experimentally. The equipment in which the experiments were performed and which uses alcohol adsorption on activated carbon is briefly described, and its operating cycle described, along with typical experimental observations of pressure and temperature transients. A model of the adsorber unit is proposed which accounts for the coupling of adsorption and heat transfer, and describes mass-transfer in the annular adsorbent layer as a global diffusional mechanism with temperature dependent parameters. This model correctly predicts, qualitatively and semi-quantitatively, the observed trends of the temperature changes. Finally, various aspects of the performances are discussed.

    Notizen: Abstract The viability of irrigated systems in the Senegal River Valley is being brought into question today due to their under‐utilization. We assume that their viability depends largely on the way their different components behave and interact. We therefore sought to examine in greater depth today's knowledge of the structure of these systems and activities performed within them. This led to the development of a multi‐agent system model, a kind of virtual irrigated system, with a special focus on rules in use for access to credit, water allocation and cropping season assessment as well as organization and coordination of farmers. The purpose of this paper is to show how this kind of tool is relevant to the study of irrigated systems' viability. As an example it is used to examine the influence of existing social networks on the viability of irrigated systems.

    Notizen: Abstract The stability constants of thorium(IV) hydrolysis species have been measured at15, 25, and 35°C (in 1.0 mol dm−3 NaClO4) using both potentiometry and solventextraction. The results indicate the presence of the monomeric speciesTh(OH)3+, Th(OH)2+ 2, Th(OH)+ 3, and Th(OH)4, in addition to the polymericspecies Th4(OH)8+ 8 and Th6(OH)9+ 15. The polymeric species were found to beimportant, although the total thorium concentration was limited to 0.01–0.1mmol-dm−3. The solvent extraction measurements required the use of acetylacetone.As such, the stability constants of thorium(IV) with acetylacetone were alsomeasured using both potentiometry and solvent extraction. All logarithms of thestability constants were found to be linear functions of the reciprocal absolutetemperature indicating that ΔH o and ΔSo of reaction are both independent oftemperature (over the temperature range examined in the study).

    Notizen: Abstract A flow-through electrical conductance cell was assembled in order to measuremolar conductances of dilute aqueous electrolytes with a high degree of accuracyat high temperatures and pressures. The design of the cell is based on the conceptdeveloped at the University of Delaware and built in 1995, with modificationsthat will allow the cell to operate at much higher temperatures (to 600°C) andpressures (to 300 MPa). At present, the cell has been tested successfully bymeasuring aqueous (10−4-10−3 mol-kg−1) solutions of LiCl, NaCl, and KCl attemperatures 25–410°C and pressures 9.8–33 MPa. The results are in goodagreement with reported values, including those measured with the Delawareflow-through cell. These new results are also complementary to our previousresults, which were measured with a static high-pressure cell. Measurements attemperatures near the critical point of water (374°C, 22.1 MPa) require the useof lower solution concentrations that were unachievable in the past with thestatic cell.

    Notizen: Abstract This paper studies the impact of a reduction in hardware complexity on the supply-chain inventory against various customer on-time delivery alternatives and manufacturing environments. Different methods of reducing hardware complexity are proposed, and their impact on total supply-chain inventory and customer serviceability quantified. An analytical inventory optimization scheme taking into account multistage supply networks, product structure, forecast accuracy, lead-time variability, and supplier reliability is used to determine optimal inventory levels in a stochastic modeling environment. The analysis is based on a business case for an IBM midrange computer family consisting of more than 200 models and upgrades with hundreds of features. We investigate different hardware complexity reduction strategies, including low-usage feature reduction, low-volume feature reduction, and feature substitution, as well as quick response and postponement mechanisms. Our computational results show that, in a fabrication-fulfillment center environment, reducing hardware complexity results in significantly higher inventory savings than in an integrated manufacturing environment. The results presented in this paper were used to reduce hardware complexity in IBM's midrange computer division.

    Notizen: Abstract This paper concerns the relation between surface structure of crystalline vanadia-like active species on vanadia–tungsta catalyst and their activity in the selective reduction of NO by ammonia to nitrogen. The investigations were performed for Ti–Sn-rutile-supported isopropoxy-derived catalyst. The SCR activity and surface species structure were determined for the freshly prepared catalyst, for the catalyst previously used in NO reduction by ammonia (320 ppm NO, 335 ppm NH3 and 2.35 vol% O2) at 573 K as well as for the catalyst previously annealed at 573 K in helium stream containing 2.35 vol% O2. The crystalline islands, exposing main V2O5 surface, with some tungsten atoms substituted for V-ones, were found, with XPS and FT Raman spectroscopy, to be present at the surface of the freshly prepared catalyst. A profound evolution of the active species during the catalyst use at 573 K was observed. Dissociative water adsorption on V5+OW6+ sites is discussed as mainly responsible for the catalyst activity at 473 K and that on both V5+OW6+ and V4+OW6+ sites as determining the activity at 523 K.

    Notizen: Abstract An evolutionary point of view is proposed to make more appropriate distinctions between experience, awareness and consciousness. Experience can be defined as a characteristic linked closely to specific pattern matching, a characteristic already apparent at the molecular level at least. Awareness can be regarded as the special experience of one or more central, final modules in the animal neuronal brain. Awareness is what experience is to animals. Finally, consciousness could be defined as reflexive awareness. The ability for reflexive awareness is distinctly different from animal and human awareness and depends upon the availability of a separate frame of reference, as provided by symbolic language. As such, words have made reflexive awareness – a specific and infrequent form of awareness – possible. Conciousness might be defined as the experience evoked by considering, i.e. thinking about experiences themselves. If there is a hard problem of explaining consciousness, than this actually must be considered as the hard problem already met when trying to explain basic experience, since its nature remains elusive.

    Notizen: Abstract The philosophical consequences of synergetics, the interdisciplinary theory of evolution and self-organization of complex systems, are being drawn in the paper. The idea of discreteness of evolutionary paths is in the focus of attention. Although the future is open, and there are many alternative evolutionary paths for complex systems, not any arbitrary (either conceivable or desirable) evolutionary path is feasible in a given system. There are discrete spectra of possible evolutionary paths which are determined exclusively by inner properties of the corresponding systems. Synergetics allows us to reveal general laws of self-organization and, therefore, certain limits of arbitrariness of nature in choosing possible paths of evolution as well as in constructing of a complex evolutionary whole. A comparative analysis between the modern synergetic notions and a few ideas of the Western philosophy (F. Nietzsche, N. Hartmann, M. Heidegger) and of the Eastern teachings (Taoism, Buddhism) is made.

    Notizen: Abstract The ethical and moral behavior of Homo sapiens is no longer the exclusive domain of religion and philosophy because we recognize that such behavior affects the reproductive success of individuals within the species. We are a social species and therefore our survival is influenced by our capacity for cooperation and our willingness to take risks for kin. Emotions, some of which are found in other species, help to mediate our altruistic behavior. The reproductive benefits of helping kin, especially offspring, are readily seen. Helping non-kin can be beneficial if individuals can differentiate between ‘reciprocators’ and ‘non-reciprocators’ and direct altruistic behavior toward reciprocators. Also, if third parties are favorably impressed by observing altruistic behavior, the rewards need not come from the recipient of the altruistic behavior.

    Notizen: Abstract Fossil Trichoptera (caddisfly) remains have been identified and quantitatively recorded in the late-glacial and early-Holocene sediments from Kråkenes Lake, western Norway. The sediment sequence was deposited between 12,300 and 8850 14C BP, covering the Allerød, Younger Dryas, and early-Holocene periods. The first Trichoptera were recorded at 12,000 14C BP, and during the Allerod a diverse assemblage of Limnephilidae taxa developed in the lake. By about 11,400 14C BP the relatively thermophilous Polycentropus flavomaculatus and Limnephilus rhombicus were present, suggesting that the summer water temperature was at least 17 °C. This temperature fell by 5-8 °C at the start of the Younger Dryas, and the thermophilous taxa were replaced within 20-40 14C yrs by Apatania spp., including the arctic-alpine A. zonella, suggesting a maximum summer water temperature of 10-12 °C. The Trichoptera assemblage was impoverished in numbers and in diversity over the next 200 yrs as the severe conditions of the Younger Dryas developed. As soon as temperatures rose and glacial meltwater and silt input ended about 700 14C yrs later, the resident Apatania assemblage expanded immediately, within 10 yrs. About 130 yrs later, thermophilous taxa replaced Apatania, and a much more diverse assemblage than in the Allerod occupied the varied habitats made available by the development of the Holocene lake ecosystem. The 130 yr delay may have been caused by a gradual temperature increase crossing a critical threshold, or by the time taken for thermophilous taxa to migrate from their Younger Dryas refugia.

    Notizen: Abstract The discussion on climatic instability observed in Greenland ice cores during the Eemian period (substage 5e) resulted in discovery of a pronounced mid-Eemian cooling event. We report that the mid-Eemian cooling is found for the first time in the biogenic silica climatic record and microfossil abundance record of Lake Baikal. Timing of this event in Lake Baikal correlates well with timing of the European pollen records and marine sedimentary records. The presence of the mid-Eemian cooling signal in the Lake Baikal record suggests a much closer link between Asian climate influenced by strong pressure fields over the vast land masses and the climate-controlling processes in the North Atlantic during interglacial periods, than what was generally believed. Furthermore, the Lake Baikal record suggests that after the mid-Eemian cooling, the climatic conditions returned close to the warmth of the 5e optimum and thus argues that the warm conditions of the last interglacial persisted in Siberia throughout 5e, and did not end with the mid-Eemian cooling as suggested by several published marine records.

    Notizen: Abstract Although paleoclimatic research in the Arctic has most often focused on variations in temperature, the Arctic has also experienced changes in hydrologic balance. Changes in Arctic precipitation and evaporation rates affects soils, permafrost, lakes, wetlands, rivers, ice and vegetation. Changes in Arctic soils, permafrost, runoff, and vegetation can influence global climate by changing atmospheric methane and carbon dioxide concentrations, thermohaline circulation, and high latitude albedo. Documenting past variations in Arctic hydrological conditions is important for understanding Arctic climate and the potential response and role of the Arctic in regards to future climate change. Methods for reconstructing past changes in Arctic hydrology from the stratigraphic, isotopic, geochemical and fossil records of lake sediments are being developed, refined and applied in a number of regions. These records suggest that hydrological variations in the Arctic have been regionally asynchronous, reflecting the impacts of different forcing factors including orbitally controlled insolation changes, changes in geography related to coastal emergence, ocean currents, sea ice extent, and atmospheric circulation. Despite considerable progress, much work remains to be done on the development of paleohydrological proxies and their application to the Arctic.

    Notizen: Abstract The relationships between diatoms (Bacillariophyceae) in surface sediments of lakes and summer air temperature, pH and total organic carbon concentration (TOC) were explored along a steep climatic gradient in northern Sweden to provide a tool to infer past climate conditions from sediment cores. The study sites are in an area with low human impact and range from boreal forest to alpine tundra. Canonical correspondence analysis (CCA) constrained to mean July air temperature and pH clearly showed that diatom community composition was different between lakes situated in conifer-, mountain birch- and alpine-vegetation zones. As a consequence, diatoms and multivariate ordination methods can be used to infer past changes in treeline position and dominant forest type. Quantitative inference models were developed to estimate mean July air temperature, pH and TOC from sedimentary diatom assemblages using weighted averaging (WA) and weighted averaging partial least squares (WA-PLS) regression. Relationships between diatoms and mean July air temperature were independent of lake-water pH, TOC, alkalinity and maximum depth. The results demonstrated that diatoms in lake sediments can provide useful and independent quantitative information for estimating past changes in mean July air temperature (R2 jack = 0.62, RMSEP = 0.86 °C; R2 and root mean squared error of prediction (RMSEP) based on jack-knifing), pH (R2 jack = 0.61, RMSEP = 0.30) and TOC (R2 jack = 0.49, RMSEP = 1.33 mg l-1). The paper focuses mainly on the relationship between diatom community composition and mean July air temperature, but the relationships to pH and TOC are also discussed.

    Notizen: Abstract This paper reports a first estimate of the Holocene lake sediment carbon pool in Alberta, Canada. The organic matter content of lake sediment does not appear to depend strongly on lake size or other limnological parameters, allowing a simple first estimate in which we assume all Alberta lake sediment to have the same organic matter content. Alberta lake sediments sequester about 15 g C m-2 yr-1, for a provincial total of 0.23 Tg C yr-1, or 2.3 Pg C over the Holocene. Alberta lakes may represent as much as 1/1700 of total global, annual permanent carbon sequestration.

    Notizen: Abstract As part of a series of papers describing the use of a simulation model to extrapolate experimental measurements of methane (CH4) emissions from rice fields in Asia and to evaluate the large-scale effect of various mitigation strategies, the collation and derivation of the spatial databases used are described. Daily weather data, including solar radiation, minimum and maximum temperatures, and rainfall were collated from 46 weather stations from the five countries in the study, namely China, India, Indonesia, Philippines, and Thailand. Quantitative soil data relevant to the input requirements of the model were derived by combining data from the World Inventory of Soil Emissions (WISE) database, the ISIS database, and the FAO Digital Soil Map of the World (FAO-DSMW). These data included soil pH; organic carbon content; sand, silt, and clay fractions; and iron content for top and subsoil layers, and average values of bulk density and available water capacity for the whole profile. Data on the areas allocated to irrigated, rainfed, upland, and deepwater rice at the province or district level were derived from the Huke & Huke (1997) database developed at IRRI. Using a geographical information system (GIS), a series of georeferenced data sets on climate, soils, and land use were derived for each country, at the province or district level. A summary of the soil-related derived databases is presented and their applicationn for use in global change modeling discussed.

    Notizen: Abstract Ecotones are inherent features of landscapes, transitional zones, and play more than one functional role in ecosystem dynamics. The delineation of ecotones and environmental boundaries is therefore an important step in land-use management planning. The delineation of ecotones depends on the phenomenon of interest and the statistical methods used as well as the associated spatial and temporal resolution of the data available. In the context of delineating wetland and riparian ecosystems, various data types (field data, remotely sensed data) can be used to delineate ecotones. Methodological issues related to their detection need to be addressed, however, so that their management and monitoring can yield useful information about their dynamics and functional roles in ecosystems. The aim of this paper is to review boundary detection methods. Because the most appropriate methods to detect and characterize boundaries depend of the spatial resolution and the measurement type of the data, a wide range of approaches are presented: GIS, remote sensing and statistical ones.

    Notizen: Abstract The evolution, taxonomy and conservation of the genus Equuswere investigated by examining the mitochondrial DNA sequences of thecontrol region and 12S rRNA gene. The phylogenetic analysis of thesesequences provides further evidence that the deepest node in thephylogeny of the extant species is a divergence between twolineages; one leading to the ancestor of modern horses (E.ferus, domestic and przewalskii) and the other to thezebra and ass ancestor, with the later speciation events of the zebrasand asses occurring either as one or more rapid radiations, or withextensive secondary contact after speciation. Examination of the geneticdiversity within species suggested that two of the E. hemionussubspecies (E. h. onager and E. h. kulan) onlyrecently diverged, and perhaps, are insufficiently different to beclassified as separate subspecies. The genetic divergence betweendomestic and wild forms of E. ferus (horse) and E.africanus (African ass) was no greater than expected within anequid species. In E. burchelli (plains zebra) there was anindication of mtDNA divergence between populations increasing withdistance. The implications of these results for equid conservation arediscussed and recommendations are made for conservation action.

    Notizen: Abstract In the period 1875–1920, a debate about the generality and applicability of evolutionary theory to all organisms was motivated by work on unicellular ciliates like Paramecium because of their peculiar nuclear dualism and life cycles. The French cytologist Emile Maupas and the German zoologist August Weismann argued in the 1880s about the evolutionary origins and functions of sex (which in the ciliates is not linked to reproduction), and death (which appeared to be the inevitable fate of lineages denied sexual conjugation), an argument rooted in the question of whether the ciliates and their processes where homologous to other cellular organisms. In the beginning of the twentieth century, this question of homology came to be less important as the ciliates were used by the British protozoologist Clifford Dobell and the American zoologist Herbert Spencer Jennings to study evolutionary processes in general rather than problems of development and cytology. For them, homology mattered less than analogy. This story illustrates two partially distinct problems in evolutionary biology: first, the question of whether all living things have common features and origins; and second, whether their history and current nature can be described by identical mechanisms. Where Maupas (contra Weismann) made the ciliates qualitatively the same as all other organisms in order to create a cohesive evolutionary theory for biology, Jennings and Dobell made them qualitatively different in order to achieve the same end.

    Notizen: Abstract A fiber optic low-coherence sensor based on the spectral shift of tantalum pentoxide thin films for absolute temperature sensing up to 650°C is described. A tantalum pentoxide single layer was deposited directly onto the cleaved end-face of a single mode optical fibre and was illuminated with an super luminescence diode (SLD) source through a directional coupler. Interference fringes of the film on reflection were obtained within the optical bandwidth of the SLD using an optical spectrum analyser. The spectral shift versus temperature rise showed no turning points and the output was unambiguous, linear, monotonic and gave about 0.016 nm wavelength shift in the spectrum per°C. A semi-empirical calibration procedure based on the refractive index (n) and thickness (l) of the tantalum pentoxide film for absolute thermometric measurements is described.

    Notizen: Abstract A new method of simultaneous remote sensing of atmospheric wind and temperature by a ultraviolet Rayleigh lidar is described. This technique uses two narrowband filters located at either side of the wings of the Rayleigh backscatter spectrum to analyze Rayleigh backscattering signals. These filters are selected to be greatly sensitive to both velocity and temperature. By measuring the ratio and the sum of the two normalized filtered signals, the line-of-sight wind velocity and temperature profiles can be retrieved. A lidar system is proposed for the wind velocity and temperature measurements in the middle atmosphere, and the simulation results show that the accuracies of velocity and temperature are about 1 m/s and 2 K at the height of 30 km, respectively. The influence of aerosol component has been estimated for clear weather conditions, and with an uncertainty of aerosol component of 15% the errors are about 0.1 m/s and 2 K above the troposphere, respectively.

    Notizen: Abstract Comparative gene expression studies are often limited by low availability of tissue and poor quality of extractable mRNA. Collective PCR amplification of minute quantities of mRNA has great potential for overcoming these limitations. However, there remains significant concern about the effects of amplification on the absolute and relative abundance of individual mRNAs that could complicate subsequent gene expression studies. To address this problem, we systematically compared the relative abundance of many specific mRNAs from complex cDNA preparations (from tissue and cultured cells) both before and after amplification by PCR. Our results demonstrated that, as expected, the absolute abundance of different mRNAs in a cDNA library is altered in an unpredictable manner by PCR amplification. However, we found that the concentration ratios of specific mRNAs among different cDNA preparations were routinely well conserved after PCR amplification. Thus, for the purpose of comparative expression studies for specific mRNAs in two (or more) complex cDNAs, PCR-amplified cDNA is equally useful as unamplified cDNA. These results provide a rigorous experimental validation and offer a theoretical treatment to support the utility of PCR amplified cDNA for differential gene expression studies. We conclude that the inherent difficulties in performing differential screening studies such as gene chip and array analyses on limited amounts of biological materials can be overcome by a PCR amplification step without compromising data quality.

    Notizen: Abstract The application of non-linear heating program to a heat-flux DSC apparatus has attracted much attention. From thermodynamics viewpoint, it is shown that the variation of enthalpy of a sample changing with temperature change is due, to both the true heat capacity of the sample and the enthalpy of some transformations occurring in the sample, characterized by its degree of advance. Using the simple assumption that the rate of the transformation is proportional to the distance from the thermodynamic equilibrium, an electrical model of the thermal event is given. Using the coupled cell model of the DSC apparatus, we show how to obtain the rate of transformation of the sample and heat capacity, which is directly related to the base line of the experiment.

    Notizen: Abstract Results are presented of optical emission spectroscopy (OES) application asa control tool to improve fly-ash plasma vitrification. A twin-torch plasmasystem has been used for the fly-ash processing, and a new OES method hasexamined metallic vapors above the melt. The method allows the study ofnonhomogeneous optically thin plasmas exhibiting a symmetry plane withoutsophisticated tomographic systems. The dc arc torches are mounted above acold crucible filled with a synthetic glass. The arc intensity is from200 to 400 Å. Argon is introduced into the torches along the cathodeand the anode, while argon, oxygen or hydrogen are injected through thelance between the torches. Local plasma temperatures above the melt havebeen evaluated using measured relative intensities of spectral lines ofthe plasma-forming gas. Metallic vapor concentration in the plasma isdeduced from the intensity ratio of the metal–gas spectral lines. Leadoxide has been used to study heavy-metal behavior at the fly-ash plasmavitrification. Distribution of the lead along the crucible surface,depending on the plasma-forming gas composition as well as the concentrationevolution with time, have been examined. The elemental analysis of theresultant glass has been measured by scanning electron microscopy (SEM)with energy-dispersive spectrometry (EDS). A predictive model has beenadapted to simulate the noncongruent vaporization of heavy metals from themelt. According to the data obtained, steep variations of the volatility ofthe elements depend strongly on reducing properties of gases controllingthe plasma composition near the melted surface. In addition, the melttemperature and the redox potential of the gas phase are found to be themost critical parameters.

    Notizen: Abstract The volatilization of quartz in a transferred arc plasma followed byquench and recondensation is a promising route to the production offumed silica. In this work, an existing model of a transferred arcwas modified and combined with a newly developed model of a moltensilica anode to predict the behavior of a transferred arc evaporatoras a function of current and plasma gas flow rate. The model predictstemperature, current, and flow fields in both the plasma and anode aswell as evaporation rates. Although quantitative agreement withexperimental results was not possible because of insufficient propertydata for silica at high temperature, the results were within an orderof magnitude of those measured experimentally. The model developed isuseful for the design and scaleup of this type of reactor.

    Notizen: Abstract A high-pressure and high-temperature phase of TiO2 : TiO2 II is formed transiently during room-temperature high-energy ball milling of anatase TiO2 : TiO2 anatase → TiO2 II → TiO2 rutile. Rutile is the only phase present after prolonged ball milling. The present paper focuses on the influences of physical and chemical processing conditions on the transformation kinetics. The effects of two milling parameters on the kinetics of phase transformation of anatase TiO2 were investigated: the nature of milling tools and the powder-to-ball weight ratio R. Granulometric characterizations and TEM observations have demonstrated that the transformation of TiO2 anatase into TiO2 II occurs without fracturing of particles and that TiO2 II nanograins form at the surface of anatase particles. The parameter R affects only the transformation rate. For a given R, the transformation rate is the largest with alumina grinding tools, intermediate with zirconia tools, and the smallest with steel tools. The parameters involved in current models of the mechanical alloying process do not suffice to explain the differences in transformation rates observed here. A parameter, which takes into account the influence of the mechanical properties of grinding materials, is considered.

    Notizen: Abstract The stresses generated in alumina scales during different oxidation stepscontrol the protective character of such scales and their lifetime. Laboratoryand industrial Fe–Cr–Al alloys, differing by their impuritycontent and preparation, and for which the oxidation kinetics werepreviously characterized, were studied here to experimentally determine ortheoretically calculate the stress level in the alumina scales. Theexperiments were performed by X-ray diffraction (XRD) at room temperature,using the sin2psi method. The oxide-stress level wascorrelated with many parameters, such as the scale microstructure andmorphology, the nature of the substrate, the substrate and oxide thickness,the oxide grain size, the cooling rate, etc. A numerical elasto-viscoplasticmodel was developed to calculate the thermal stresses and to point out theeffect of the relaxation phenomena either during cooling or duringisothermal treatment after oxidation. The results obtained by theelasto-viscoplastic model are in good agreement with the experimental ones.

    Notizen: Abstract Cardiac hypertrophy has been observed in newborn infants treated with dexamethasone (DEX). This study was undertaken to examine whether DEX-induced hypertrophy in newborn rats is associated with redistribution of cardiac myosin heavy chain (MHC) isoforms and if so, the effects involve transcriptional regulation. Newborn rats were injected with either DEX (1 mg/kg/day; s.c.) or equivalent volume normal saline for 1, 3, 5, 7 or 9 days. Hypertrophy was quantified by heart dry/wet wt ratios, heart/body wt ratios, and total protein content of the myocardium. Changes in the expression of cardiac MHC mRNA were characterized by northern blot and slot blot analyses, using isoform specific probes for a- and β-MHC genes. DEX effect on α-MHC gene transcription was analyzed by transiently transfecting various α-MHC promoter/CAT reporter constructs into primary cultures of cardiac myocytes derived from one day old rat pups. DEX administration into newborn rats produced significant cardiac hypertrophy ranging from 23% at day 1 to 59% at 9 days. The hypertrophy was accompanied by immediate increase (83%) in steady state level of the α-MHC mRNA within one day and a maximum increase (148%) at 7 days of treatment. The steady state level of β-MHC mRNA declined by 25% at day 1 and a maximum decrease of 54% at day 7 of DEX treatment. The changes in MHC mRNA were also reflected in their protein levels as determined by V1 and V3 isozyme analysis. DEX treatment of primary cultures of cardiomyocytes following transfection with a-MHC promoter/CAT reporter constructs resulted in increased CAT expression in a dose dependent manner. The minimum α-MHC gene sequences responding to DEX treatment were located between the -200 to -74-bp region of the gene, resulting in 2-fold and 6-fold activation of CAT reporter after 0.05 and 0.1 mM doses of DEX, respectively. Our data indicate that DEX induced cardiac hypertrophy in newborn rats is accompanied by increased expression of α-MHC and decreased expression of β-MHC. The α-MHC effects are mediated in part through transcriptional mechanisms.

The radiated wavefields are obtained by point source superposition, making use of an analytical relationship for the wave propagation direction as a function of the spatial coordinates. Approximate formulae are given for the radiation characteristics of circular and rectangular transducers in the far-field, which are in a form similar to the isotropic material case. Focusing on transversely isotropic weld metal specimens and unidirectional composites, numerical evaluation is performed for normal-incidence probes as well as for electromagnetic acoustic transducers.

    Notizen: Abstract Previously, chick heme oxygenase-1 (cHO-1) gene was cloned by us and two regions important for induction by sodium arsenite were identified. These two regions were found to contain consensus sequences of an AP-1 (-1580 to -1573) and a MRE/cMyc complex (-52 to -41). In the current study, the roles of these two elements in mediating the sodium arsenite or cobalt chloride dependent induction of cHO-1 were investigated further. DNA binding studies and site-directed mutagenesis studies indicated that both the AP-1 and MRE/cMyc elements are important for the sodium arsenite induction, while cobalt chloride induction involves only the AP-1 element. Electrophoretic mobility shift assays showed that nuclear proteins binding to the AP-1 element was increased by both sodium arsenite or cobalt chloride treatment, whereas the binding of proteins to the MRE/cMyc element showed a high basal expression in untreated cells and the binding activity was only slightly increased by sodium arsenite treatment. Site-directed mutagenesis studies showed that, to completely abolish sodium arsenite induction, both the AP-1 and MRE/cMyc elements must be mutated; mutation of either element alone resulted in only a partial effect. In contrast, a single mutation at AP-1 element was sufficient to reduce the cobalt chloride induction almost completely. The MRE/cMyc complex plays a major role in the basal level expression, and shares some similarities to the upstream stimulatory factor element (USF) identified in the promoter regions of mammalian HO-1 genes and other stress regulated genes. Because sodium arsenite is known to cause oxidative stress and because activation of AP-1 proteins has been shown to be a key step in the oxidative stress response pathway, we also explored the possibility that the induction of the cHO-1 gene by sodium arsenite is mediated through oxidative stress pathway(s) by activation of AP-1 proteins. We found that pretreatment with antioxidants (N-acetyl cysteine or quercetin) reduced the induction of the endogenous cHO-1 message or cHO-1 reporter construct activities induced by sodium arsenite or cobalt chloride. These antioxidants also reduced the protein binding activities to the AP-1 element in the electrophoretic mobility shift assays. In summary, induction of the cHO-1 gene by sodium arsenite or cobalt chloride is mediated by activation of the AP-1 element located at -1,573 to -1,580 of the 5′ UTR.

    Notizen: Abstract The evolution of grain size, grain-size distribution, morphological and crystallographic texture, surface roughness, and the contribution of various surface facets to the growth of polycrystalline diamond films is performed by carrying out a series of two-dimensional computer simulations. The films are assumed to grow from a set of randomly oriented, {100}- and {111}-faceted nuclei by the motion of their vertices (the points where the adjoining facets of the same or neighboring grains meet). The vertex velocities are found to be a function of the orientation and the growth rate of the adjoining facets. To quantify the latter, a {100} to {111} growth-rate parameter is used. The results show that the evolution of the grain size and its distribution, surface roughness, morphological and crystallographic texture, and the portion of the film grown from different surface facets are all mutually linked and governed by the magnitude of the growth-rate parameter. The latter can be controlled by the CVD processing conditions, such as the substrate temperature, reactor pressure, mole fraction of carbon-source gas (e.g., CH4, C2H2).

    Notizen: Abstract Reactive oxygen species (ROS; O2-, H2O2, and OH·), normal by-products of cellular metabolic processes, are kept in control by antioxidant enzymes, such as catalase, glutathione peroxidase (GPX) and superoxide dismutases (SODs). To understand the role of antioxidant enzymatic defenses against ROS injury following ischemia-reperfusion, we examined the effect on kidney exposed to varying periods (30, 60 or 90 min) of ischemia followed by different periods of reperfusion. The enzymatic activities and protein levels of catalase, GPX, CuZnSOD and MnSOD were relatively unaffected at 30 min of ischemia followed by 0, 2 or 24 h reperfusion. However, 60 or 90 min of ischemia followed by 0, 2 or 24 h of reperfusion resulted in a decrease in activities and protein levels which paralleled the duration of ischemic injury. MnSOD activity tended to recover towards normal during reperfusion. Examination of the mRNA levels of these antioxidant enzymes demonstrated a severe decrease in mRNA levels of catalase and GPX at a time point of minimal ischemic injury (30 min of ischemia followed by reperfusion) suggesting that loss of mRNA of catalase and GPX may be the first markers of alterations in cellular redox in ischemia-reperfusion injury. Greater loss of mRNA for catalase, GPX and CuZnSOD were observed following longer periods (60 or 90 min) of ischemia. The mRNA for MnSOD was upregulated at all time points of ischemia-reperfusion injury. Actually, the greater decrease in mRNAs for catalase, GPX and CuZnSOD in the acute phase (within 24 h) subsequently showed a further decrease in these enzyme activities in the subacute phase (72 or 120 h after ischemia). These enzyme activities in the 30 min ischemia group, but not in the 90 min group, already showed tendencies for normalization at 120 h after ischemia. To understand the molecular basis of the loss of mRNA of these antioxidant enzymes during ischemia-reperfusion injury, we examined the rate of transcription by nuclear run-on assays. The similar rates of transcription in control and kidney exposed to ischemia-reperfusion indicates that the loss of mRNA for catalase, GPX and CuZnSOD are possibly due to the increased rate of turnover of their mRNAs. These studies suggest that expression of antioxidant genes during ischemia-reperfusion are not coordinately expressed and the differential loss of antioxidant enzymes may be the contributing factor(s) towards the heterogeneous renal tissue damage as a result of ischemia-reperfusion induced oxidative stress.

    Notizen: Abstract Interleukin-1β (IL-1) is a potent inducer of cyclooxygenase-2 (COX-2) and prostaglandin biosynthesis in many types of cells, yet little is known about the molecular mechanisms regulating IL-1 mediated prostanoid biosynthesis in the endothelium of the microvasculature. Therefore, we examined the cis- and trans-acting factors regulating IL-1-induced COX-2 expression in the human microvascular endothelial cell line, HMEC-1. IL-1 enhanced steady state levels of COX-2 protein and mRNA synthesis by ≈ 2-fold which preceded a 2-fold increase in PGFα biosynthesis. Expression of a series of COX-2 promoter-luciferase constructs in IL-1 treated HMEC-1 cells revealed that the 'full length' (-1432/+59 bp) promoter was 10 times more active than the SV-40 promoter/enhancer and that it could be further activated by IL-1. Surprisingly however, all except for the shortest COX-2 promoter construct retained the ability to respond to IL-1 and luciferase activity driven by -191/+59 bp COX-2 promoter was as responsive to IL-1 as the full-length promoter. Moreover, site-directed promoter mutagenesis and electophoretic mobility shift assays (EMSA) indicate that the combinatorial actions of AP2, NF-IL6, and CRE elements are critical for both constitutive and IL-1-inducible COX-2 promoter activity. Understanding the mechanism(s) regulating COX-2 gene expression and prostaglandin biosynthesis in the microvasculature has important implications with regard to inflammation and angiogenesis in vivo.

    Notizen: Abstract Expression of the gene coding for thymosin β-4 (Tβ-4), the major G-actin sequestering peptide in the cell, is regulated mainly at the level of transcription. In this study, we examined the nucleotide sequence of the 5′-flanking region (from - 2202 to - 881) of the mouse Tβ-4 gene, and demonstrated that the DNA fragment from -278 to +410 of this gene was capable of directing the expression of a chloramphenicol acetyltransferase reporter gene in NIH3T3 cells. However, expression of the reporter gene in cells cannot be induced by interferon-a treatment even though a rapid activation of endogenous Tβ-4 gene by this cytokine was observed. These results suggest that the projected interferon-stimulated response element (ISRE) might reside in other parts of the mouse Tβ-4 gene.

    Notizen: Abstract The effect of thioacetamide (TA), an hepatotoxic and hepatocarcinogenic compound, on the expression and activity of the cytosolic enzyme glutathione-S-transferase (GST) was studied in rat liver. Four h following the administration of 14C-labeled thioacetamide (10 mg/Kg), several subunits of GST were found to be radioactively labeled. A single sublethal dose of TA (250 mg/Kg) decreased by three-fold the expression of classα GST at 24-48 h of treatment, but did not significantly affect the transcription of class μ GST. The activity of the enzyme toward 1-chloro-2,4-dinitrobenzene was mildly inhibited (66% of the control) by a 24 h TA treatment and gradually increased thereafter. It is proposed that the covalent binding of TA or its derivative to the GST subunits does not affect the activity of the enzyme. Nevertheless, GST activity inhibition is due to the deleterious effect of TA on GST transcription.

    Notizen: Abstract Within the CLEAR project a new approach to integrated assessment modelling has been developed for the participatory integrated assessment of regional climate change involving citizens' focus groups. The climate change decision problem was structured by focusing separately on climate impacts and mitigation options. The attempt was made to link the different scales of the problem from the individual to the global level. The abstract topic of climate change was related to options on the level of a citizen's individual lifestyle. The option of a low energy society was emphasised in order to embed the climate change decision problem in a wider range of societal concerns. Special emphasis was given to the characterisation and communication of uncertainties. The chosen approach allows different kinds of uncertainties in one framework to be addressed.     Notizen: Abstract The potential ecological impact of ongoing climate change has been much discussed. High mountain ecosystems were identified early on as potentially very sensitive areas. Scenarios of upward species movement and vegetation shift are commonly discussed in the literature. Mountains being characteristically conic in shape, impact scenarios usually assume that a smaller surface area will be available as species move up. However, as the frequency distribution of additional physiographic factors (e.g., slope angle) changes with increasing elevation (e.g., with few gentle slopes available at higher elevation), species migrating upslope may encounter increasingly unsuitable conditions. As a result, many species could suffer severe reduction of their habitat surface, which could in turn affect patterns of biodiversity. In this paper, results from static plant distribution modeling are used to derive climate change impact scenarios in a high mountain environment. Models are adjusted with presence/absence of species. Environmental predictors used are: annual mean air temperature, slope, indices of topographic position, geology, rock cover, modeled permafrost and several indices of solar radiation and snow cover duration. Potential Habitat Distribution maps were drawn for 62 higher plant species, from which three separate climate change impact scenarios were derived. These scenarios show a great range of response, depending on the species and the degree of warming. Alpine species would be at greatest risk of local extinction, whereas species with a large elevation range would run the lowest risk. Limitations of the models and scenarios are further discussed.

    Notizen: Abstract In order to elucidate the effects of temperature and photoperiod on the quality and quantity of plant root exudates, a Japanese cucumber (Cucumis sativus, cv. Shougoin-Aonaga-Fushinari) was grown hydroponically in growth chambers under controlled temperature and photoperiod conditions with or without the addition of activated charcoal (AC) to the nutrient solutions. Fresh AC was used to trap the organic compounds exuded from cucumber roots every two weeks. Cucumber plants without AC were severely retarded in root growth and in the accumulation of dry matter, especially at high temperature and long photoperiod, compared to those with AC. The growth inhibitors, adsorbed on the AC or accumulated in the nutrient solution without AC, were extracted by organic solvents and analyzed by GC-MS. Benzoic acid and its derivatives, cinnamic acid derivatives, and fatty acids were identified. The rate of root exudation in vegetative and reproductive stages for some of these organic acids increased with the elevation of temperature and the elongation of photoperiod, and the mean rate was two or more times higher than the minimum exudation at low temperature with short photoperiod. Some of the identified compounds significantly inhibited the germination and/or root growth of lettuce and cucumber.

    Notizen: Abstract The levels of plasma apolipoprotein (apo) E, an anti-atherogenic protein involved in mammalian cholesterol transport, were found to be 2-3 fold lower in mice over-expressing human apoA-I gene. ApoE is mainly associated with VLDL and HDL-size particles, but in mice the majority of the apoE is associated with the HDL particles. Over-expression of the human apoA-I in mice increases the levels of human apoA-I-rich HDL particles by displacing mouse apoA-I from HDL. This results in lowering of plasma levels of mouse apoA-I. Since plasma levels of apoE also decreased in the apoA-I transgenic mice, the mechanism of apoE lowering was investigated. Although plasma levels of apoE decreased by 2-3 fold, apoB levels remained unchanged. As expected, the plasma levels of human apoA-I were almost 5-fold higher in the apoAI-Tg mice compared to mouse apoA-I in WT mice. If the over-expression of human apoA-I caused displacement of apoE from the HDL, the levels of hepatic apoE mRNA should remain the same in WT and the apoAI-Tg mice. However, the measurements of apoE mRNA in the liver showed 3-fold decreases of apoE mRNA in apoAI-Tg mice as compared to WT mice, suggesting that the decreased apoE mRNA expression, but not the displacement of the apoE from HDL, resulted in the lowering of plasma apoE in apoAI-Tg mice. As expected, the levels of hepatic apoA-I mRNA (transgene) were 5-fold higher in the apoAI-Tg mice. ApoE synthesis measured in hepatocytes also showed lower synthesis of apoE in the apoAI-Tg mice. These studies suggest that the integration of human apoA-I transgene in mouse genome occurred at a site that affected apoE gene expression. Identification of this locus may provide further understanding of the apoE gene expression.

    Notizen: Abstract Nucleic acid molecules with high affinities for a target transcription factor can be introduced into cells as decoy cis-elements to bind these factors and alter gene expression. This review discusses a synthetic single-stranded palindromic oligonucleotide, which self-hybridizes to form a duplex/hairpin and competes with cAMP response element (CRE) enhancers for binding transcription factors. This oligonucleotide inhibits CRE- and Ap-1-directed gene transcription and promotes growth inhibition in vitro and in vivo in a broad spectrum of cancer cells, without adversely affecting normal cell growth. Evidence presented here suggests that the CRE-decoy oligonucleotide can provide a powerful new means of combating cancers, viral diseases, and other pathological conditions by regulating the expression of cAMP-responsive genes.

    Notizen: Abstract To investigate the molecular mechanism(s) of action of catecholamines on the expression of the angiotensinogen (ANG) gene in kidney proximal tubular cells, we used opossum kidney (OK) cells with a fusion gene containing the 5′-flanking regulatory sequence of the rat ANG gene fused with a human growth hormone (hGH) gene as a reporter, pOGH (rANG N-1498/+18), permanently integrated into their genomes. The level of expression of the ANG-GH fusion gene was quantified by the amount of immunoreactive-hGH (IR-hGH) secreted into the medium. The addition of norepinephrine (NE), isoproterenol (a β1/β2-adrenergic receptor (AR) agonist) and iodoclonidine (an α2-AR agonist) stimulated the expression of the ANG-GH fusion gene in a dose-dependent manner, whereas the addition of epinephrine and phenylephrine (α1-AR agonist) had no effect. The stimulatory effect of NE was blocked by the presence of propranolol (β-AR blocker), atenolol (β1-AR blocker), yohimbine (α2-AR blocker), Rp-cAMP (an inhibitor of cAMP-dependent protein kinase AI & AII) and staurosporine (an inhibitor of protein kinase C), but was not blocked by ICI 118, 551 (β2-AR blocker) and prazosin (α1-AR blocker). The addition of a combination of isoproterenol and iodoclonidine or a combination of 8-Bromo-cAMP (8-Br-cAMP) and phorbol 12-myristate (PMA) synergistically stimulated the expression of the ANG-GH fusion gene as compared to the addition of isoproterenol, iodoclonidine, 8-Br-cAMP or PMA alone. Furthermore, the addition of NE, 8-Br-cAMP or PMA stimulated the expression of pOGH (rANG N-806/-779/-53/+18), a fusion gene containing the putative cAMP responsive element (CRE, ANG N-806/-779) upstream of the ANG promoter (ANG N-53/+18) in OK cells, but had no effect on the expression of fusion genes containing the mutant of the CRE. Gel mobility shift assays revealed that the ANG-CRE binds with the DNA-binding domain (bZIP 254-327) of the cAMP-responsive binding protein (CREB). The binding of the labeled ANG-CRE to CREB (bZIP254-327) was displaced by unlabeled ANG-CRE and the CRE of the somatostatin gene but not by the mutants of the ANG-CRE. Finally, NE stimulated the phosphorylation of CREB in OK cells. These studies demonstrate that the molecular mechanism(s) of NE action on the expression of the ANG gene in OK cells may be mediated via both the PKA and PKC signalling pathways and via the phosphorylation of CREB. The phosphorylated CREB then interacts with the CRE in the 5′-flanking region of the ANG gene and subsequently stimulates the gene expression.

    Notizen: Abstract Pressure overload of the heart is associated with a perturbed gene expression of the cardiomyocyte leading to an impaired pump function. The ensuing neuro-endocrine activation results in disordered influences of angiotensin II and catecholamines on gene expression. To assess whether angiotensin II type 1 receptor inhibition can also counteract a raised sympathetic nervous system activity, spontaneously hypertensive rats fed a hypercaloric diet were treated with eprosartan (daily 90 mg/kg body wt) and cardiovascular parameters were monitored with implanted radiotelemetry pressure transducers. Both, blood pressure and heart rate were increased (p 〈 0.05) by the hypercaloric diet. Although eprosartan reduced (p 〈 0.05) the raised systolic and diastolic blood pressure, the diet-induced rise in heart rate was blunted only partially. In addition to drugs interfering with the enhanced catecholamine influence, compounds should be considered that selectively affect cardiomyocyte gene expression via 'metabolic' signals.

    Notizen: Abstract Angiotensinogen (AGT) is a unique substrate of the renin-angiotensin system and fibronectin (FN) is an important component of the extracellular matrix. These play critical roles in the pathophysiological changes including cardiovascular remodeling and hypertrophy in response to hypertension. This study was performed to examine the regulation of AGT and FN gene in cardiac myocytes (CMs) and vascular smooth muscle cells (VSMCs) in response to mechanical stretch. Mechanical stretch significantly increased the AGT mRNA expression in CMs, while these stimuli did not affect FN mRNA levels. On the other hand, Mechanical stretch upregulated FN mRNA levels in VSMCs, whereas no increase in AGT mRNA levels was observed in response to stretch stimuli. An angiotensin II type 1 (AT1) receptor antagonist (CV11974) significantly decreased these stretch-mediated increases in mRNA level and promoter activity of the AGT and FN gene, whereas angiotensin II type 2 (AT2) receptor antagonist (PD123319) did not affect the induction. These results indicate that mechanical stretch activates transcription of the AGT and FN gene mainly via AT1 receptor-pathway in CMs and VSMCs. Furthermore, mechanisms regulating AGT and FN gene seem to be different between CMs and VSMCs.

    Notizen: Abstract Angiotensin II (Ang II) is an important regulator of the function of medullary thick ascending limb of loop of Henle (MTAL). Recent studies showed that changes in Ang II receptor expression occur and underlie changes in the function of proximal tubules during altered sodium intake. The present experiment was designed to determine (1) whether expression of the type 1 Ang II (AT1) receptor in the MTAL is regulated by altered sodium intake, and (2) the specific pathway(s) mediating sodium-induced AT1 expression in the MTAL. Wistar rats were fed a normal sodium (0.5%, NS), low sodium (0.07%, LS), or high sodium (4%, HS) diet for 2 weeks. Northern blot analysis and radioligand binding showed that in rats fed a normal sodium diet the rank of order for both AT1 mRNA expression and receptor density was outer medulla 〉 cortex 〉 inner medulla. Sodium restriction significantly increased both AT1 mRNA expression and receptor density in the outer medulla. In contrast, neither AT1 mRNA expression nor receptor density in the outer medulla was altered by sodium loading. Losartan treatment (3 mg/kg/per day by oral gavage for 2 weeks) prevented low sodium-induced upregulation of the AT1 receptor in the outer medulla, but it had no effect on AT1 expression in the outer medulla of rats fed a normal sodium diet. Highly purified suspensions of MTAL were isolated from rats fed a normal or low sodium diet. Low sodium intake significantly increased AT1 mRNA level by 184% and AT1 receptor density by 58% in MTALs. Primary cultures of MTAL cells were treated with PBS, Ang II (10-8 M), and Ang II + 17 octadecynoic (17 ODYA, 10 μM). Ang II caused about 2-fold increase in AT1 mRNA levels, and this increase was diminished by about 30% by the addition of 17 ODYA. We conclude that (1) sodium restriction but not sodium loading increases AT1 receptor expression in the MTAL, (2) low sodium-induced upregulation of the AT1 receptor in the MTAL is Ang II-dependent, and (3) Ang II-induced upregulation of the AT1 receptor in the MTAL is mediated, at least in part, by cytochrome P450 pathways.

    Notizen: Abstract The aim of this study was to explore the possible participation of cardiac renin-angiotensin system (RAS) in the ischemia-reperfusion induced changes in heart function as well as Ca2+-handling activities and gene expression of cardiac sarcoplasmic reticulum (SR) proteins. The isolated rat hearts, treated for 10 min without and with 30 μM captopril or 100 μM losartan, were subjected to 30 min ischemia followed by reperfusion for 60 min and processed for the measurement of SR function and gene expression. Attenuated recovery of the left ventricular developed pressure (LVDP) upon reperfusion of the ischemic heart was accompanied by a marked reduction in SR Ca2+-pump ATPase, Ca2+-uptake and Ca2+-release activities. Northern blot analysis revealed that mRNA levels for SR Ca2+-handling proteins such as Ca2+-pump ATPase (SERCA2a), ryanodine receptor, calsequestrin and phospholamban were decreased in the ischemia-reperfused heart as compared with the non-ischemic control. Treatment with captopril improved the recovery of LVDP as well as SR Ca2+-pump ATPase and Ca2+-uptake activities in the postischemic hearts but had no effect on changes in Ca2+-release activity due to ischemic-reperfusion. Losartan neither affected the changes in contractile function nor modified alterations in SR Ca2+-handling activities. The ischemia-reperfusion induced decrease in mRNA levels for SR Ca2+-handling proteins were not affected by treatment with captopril or losartan. The results suggest that the improvement of cardiac function in the ischemic-reperfused heart by captopril is associated with the preservation of SR Ca2+-pump activities; however, it is unlikely that this action of captopril is mediated through the modification of cardiac RAS. Furthermore, cardiac RAS does not appear to contribute towards the ischemia-reperfusion induced changes in gene expression for SR Ca2+-handling proteins.

    Notizen: Abstract The effects of propranolol and verapamil on contractile dysfunction, subcellular remodeling and changes in gene expression in cardiac hypertrophy due to pressure overload were examined. Rats were subjected to banding of the abdominal aorta and then treated with either propranolol (10 mg/kg daily), verapamil (5 mg/kg daily) or vehicle for 8 weeks after the surgery. Depression of the left ventricular function in the hypertrophied heart was associated with decreases in myofibrillar and myosin CA2+ ATPase activities as well as Ca2+-pump and Ca2+-release activities of the sarcoplasmic reticulum (SR). The level of a-myosin heavy chain (α-MHC) mRNA was decreased while that of β-MHC mRNA was increased in the pressure-overloaded heart. The level of SR Ca2+-pump ATPase (SERCA2) mRNA and protein content for SERCA2 were decreased in the pressure overloaded heart. Treatment of the hypertrophied animals with propranolol or verapamil resulted in preservation of the left ventricular function and prevention of the subcellular alterations. Shift in the α- and β-MHC mRNA levels and changes in the expression in SERCA2 mRNA level and protein content were also attenuated by these treatments. The results suggest that blockade of β-adrenoceptors or voltage-dependent calcium channels normalizes the cardiac gene expression, prevents subcellular remodeling and thus attenuates heart dysfunction in rats with cardiac hypertrophy. Furthermore, both cardiac β-adrenoceptors and L-type Ca2+-channels may be involved in the genesis of cardiac hypertrophy due to pressure overload.

    Notizen: Abstract The TIS11 gene is an immediate early gene that is induced rapidly and transiently by phorbol 12-myristate 13-acetate and various growth factors. To study transcriptional regulation of the gene, a genomic clone of rat TIS11 was isolated, and the organization of exon-intron structure and transcriptional initiation site were determined. The rat TIS11 gene consisted of 2 exons spanning approximately 2.5 kb. Several canonical sequences for binding of transcriptional factors were found in the 5′-flanking region. The 5.3 kb of the 5′-flanking region fused to a luciferase reporter gene showed promoter activity when introduced into rat pheochromocytoma PC12 cells. Analyses with serial 5′-deletion mutants suggested that the major positive regulatory region is located at the region of -241 to -76, and that the minimum promoter region is within the 76-bp upstream of the transcriptional initiation site. Gel mobility shift assays revealed that PC12 cell nuclear proteins specifically bind to the major positive regulatory region of the TIS11 gene. The identified nuclear protein components may act as the positive trans-acting factors in the basal expression of the TIS11 gene in PC12 cells.

    Notizen: Abstract In order to rationalize the physicochemical properties of human serum-transferrin (STf) and the STf-receptor (TfR) recognition process, we have tried to predict the 3D structures of apo- and iron-loaded STf using a homology modeling technique to study the changes in the structural characteristics that take place upon the uptake of iron by STf in solution. The crystal structures of both forms for ovotransferrin were used as templates for the STf modeling. The modeled structure of STf gave a satisfactory interpretation for the typical physicochemical properties such that (1) STf has a negative electrophoretic mobility and its value increases with iron uptake, and (2) the radius of gyration Rg of Tf decreases with iron uptake. It was found that upon iron binding, interdomain closures take place with large movements of the NII and CII subdomains comprising the N- and C-lobes in STf through a hinge-bending motion, accompanied by the opening of the bridge region with a displacement of more than 15 Å. Moreover, in view of the findings from our capillary electrophoresis experiments that the electrostatic interactions significantly contribute to a specific binding of Fe2-STf with TfR, it is inferred that the connecting (bridge) and its neighboring region associated with a surface exposure of negative charge play an important role in the STf-receptor recognition process.

The intact provirus is near 6 kb in length and shows a genomic organization of 5" LTR, gag, pol, env, and 3" LTR. The primer binding site (PBS) is that of a tRNAgly. The lack of functional open reading frames and occurrence of significant gaps in most, if not all, members of this group show it to be ancient. Our estimate of copy number per haploid genome is 30+. Members of this group have been isolated from Mus musculus domesticus, M. m. casteneus, M. m. hortulanus, M. caroli, and M. spretus. The occurrence of these sequences throughout such diverse members of the genus Mus may indicate that the date of the original infection predated the divergence of the extant Mus lineages at around 2.5 million years ago. Analysis of gap (deletion/insertion) patterns indicates that these sequences may have proliferated within the Mus genome by a mechanism of reverse transcriptase-mediated transposition. As yet, there are no closely related murine retroviruses described. The closest mammalian retrovirus based on sequence similarity is from the miniature swine (Sus scrofa).

    Notizen: Abstract Moderate temperatures or low concentrations of denaturants diminish the catalytic activity of some enzymes before spectroscopic methods indicate protein unfolding. To discriminate between possible reasons for the inactivation of ribonuclease A, we investigated the influence of temperature and guanidine hydrochloride on its proteolytic susceptibility to proteinase K by determining the proteolytic rate constants and fragment patterns. The results were related to changes of activity and spectroscopic properties of ribonuclease A. With thermal denaturation, the changes in activity and in the rate constants of proteolytic degradation coincide and occur slightly before the spectroscopically observable transition. In the case of guanidine hydrochloride-induced denaturation, however, proteolytic resistance of ribonuclease A initially increases accompanied by a drastic activity decrease far before unfolding of the protein is detected by spectroscopy or proteolysis. In addition to ionic effects, a tightening of the protein structure at low guanidine hydrochloride concentrations is suggested to be responsible for ribonuclease A inactivation.

    Notizen: Summary The expression of antioxidant genes has been analyzed in a potato plant and during tuber dormancy. Manganese superoxide dismutase (MnSOD), cytosolic copper and zinc superoide dismutase (Cu/ZnSOD), catalase class II, cytosolic ascorbate peroxidase (APX) and glutathione peroxidase (GPX) are expressed at the RNA level in all the contexts analyzed. By contrast, the expression of the iron superoxide dismutase (FeSOD) and plastidic Cu/ZnSOD seems to be limited to green tissues, as shown by northern blots and native gels. A complex DAB-peroxidase isozyme pattern (using diaminobenzidine as substrate) has been observed in different developmental contexts analyzed, but hardly observed in tubers. During tuber dormancy, MnSOD and cytosolic Cu/ZnSOD activity was relatively constant in both Désirée and Bintje varieties while catalase activity decreases. Moreover, tuber dormancy breakage did not involve significant changes in the activity of these enzymes. On the basis of these results, the possible link between active oxygen species (AOS) metabolism and dormancy is discussed.

    Notizen: Summary The SWACROP root-water uptake function for potatoes was determined in the 1994–95 crop season on a Ferralsol in Havana province, west Cuba. The maximum pressure-heads under which water is optimally extracted by potato roots were very similar to the original Feddes function. Nevertheless, the anaerobiosis point and the wilting point pressure-head in both functions were not in agreement. However, the simulated values of soil water contents in the 0–40 cm soil depth by performing SWACROP runs in the 1995–96 potato crop season using both functions were very similar. The original Feddes function seems to be good enough for predicting soil water contents in the tropics and no specific other functions need to be developed for any particular soil or climate condition.

    Notizen: Summary Potato tuber life-cycle is composed of many individual developmental stages including tuber formation, tuber development, dormancy and sprouting. We have used cDNA-AFLP fingerprinting to analyse gene expression in 24 individual stages of development, over the period from stolon formation through sprouting. In addition to these developmental stages, different tissues were analysed to assess tissue specificity and various controls were incorporated to determine process specificity. In total around 18000 transcript derived cDNA fragments (TDFs) were visualised from which circa 2600 were included in a statistical analysis allowing general conclusions about gene expression during development. More than 200 process specific TDFs were isolated and sequenced throughout the potato tuber life-cycle. The sequence similarities of these TDFs to known genes give an insight into the kinds of processes occurring during tuberisation, dormancy and sprouting.

    Notizen: Abstract Phylogenetic relationships within theGibberella fujikuroi species complex were extended to newly discovered strains using nucleotide characters obtained by sequencing polymerase chain reaction (PCR) amplified DNA from 4 loci used in a previous study [nuclear large subunit 28S rDNA, nuclear ribosomal internal transcribed spacer (ITS) region, mitochondriaal small subunit (mtSSU) ribosomal DNA, and β-tubulin] together with two newly sampled protein-encoding nuclear genes, translation elongation factor EF-1α and calmodulin. Sequences from the ribosomal ITS region were analyzed separately and found to contain of two highly divergent, nonorthologous ITS2 types. Phylogenetic analysis of the individual and combined datasets identified 10 new phylogenetically distinct species distributed among the following three areas: 2 within Asia and 4 within both Africa and South America. Hypotheses of the monophyly ofFusarium subglutinans and its two formae speciales, f. sp.pini and f. sp.ananas, were strongly rejected by a likelihood analysis. Maximum parsimony results further indicate that the protein-encoding nuclear genes provide considerably more phylogenetic signal that the ribosomal genes sequenced. Relative apparent synapomorphy analysis was used to detect long-branch attraction taxa and to obtain a statistical measure of phylogenetic signal in the individual and combined datasets.

    Notizen: Abstract To investigate the systematic position of the unitunicate pyremomycetePapulosa amerospora, we performed phylogenetic analyses of SSU rDNA sequences from 37 ascomycetes. Among these sequences were some new ones from taxa that might be related toPapulosa: Hyponectriaceae (Hyponectria buxi, Monographella nivalis), Phyllachorales (Phyllachora graminis), and Xylariales (Barrmaelia melanotes, Poronia punctata). Our results showed 100% bootstrap support for a clade of all unitunicate pyrenomycetes, the class Sordariomycetes. We also found strong support for recognizing the subclasses Hypocreomycetidae and Xylariomycetidae. The remaining taxa, belonging to subclass Sordariomycetidae, appeared as a polyphyletic group in one analysis, but was monophyletic when shorter SSU sequences were used.Barrmaelia melanotes, Poronia punctata, Hyponectria buxi, andMonographella nivalis are members of Xylariomycetidae, but we could not determine whetherMonographella should be included in Hyponectriaceae. The new family Papulosaceae is erected forPapulosa on molecular and morphological bases, but the exact systematic position ofPapulosa within subclass Sordariomycetidae is still uncertain, since the genus did not cluster consistently with any of the included taxa. Phyllachorales are not closely related to Diaporthales, as previously suggested.

    Notizen: Abstract A comparative investigation of the intracellular content of poly-β-hydroxybutyric acid showed that Yersinia pseudotuberculosis strains accumulated, on the average, lower amounts of this reserve substance than Listeria monocytogenes strains. The intracellular pool of poly-β-hydroxybutyric acid was responsible for the growth of the bacteria at low temperatures (4–6°C) in the absence of any exogenous carbon and energy source.

    Notizen: Abstract The effects of light and temperature conditions on the maintenance of some red microalgae (order Porphyridiales) in collections were studied. Periodical subculturing on solid and liquid media was used for maintenance. Halophilic species P. aeruginosa, P. cruentum, and P. sordidum were lost in 2.5 months if kept in darkness. The viability of algae in the dim light slightly declined in 4–5 months and was species-dependent. The results were compared with earlier obtained data on mesophilic Chlorococcales. For the green algae, darkness proved to be the optimal condition, while their viability markedly decreased at light. It was suggested that this discrepancy was caused by the presence of phycobiliproteins in the cells of red algae. Dim light conditions are optimal for the synthesis of phycobiliproteins, which are rapidly destroyed in darkness, thus leading to cell death.

    Notizen: Abstract Field and laboratory studies were carried out between 1995 and 1997 on four populations of Diaptomus leptopus found in seasonally temporary, occasionally temporary, and a permanent environment to assess the relative importance of photoperiod and temperature regimes versus other proximate local cues in inducing diapause egg production. Patterns of diapausing and subitaneous egg production were determined by observation of individual females bearing clutches that were produced in the field. A laboratory common-garden experiment was performed to assess the effects of four different regimes of temperature and photoperiod on the induction of diapause. Patterns of diapausing egg production differed among ponds: diapause occurred early in the seasonally temporary environment and occurred rarely in the permanent environment. In the common-garden experiment, populations exhibited substantial changes in the onset of diapause when compared to patterns found under field conditions. These results provide indirect evidence that the different populations respond to available cues of environmental change in different ways in nature, or that environmental cues vary among habitats.

    Notizen: Abstract The seasonal variation in length of the most abundant copepod species in the lagoon of Venice were compared and the relative influence of temperature, chlorophyll concentration and salinity examined. Temperature seems to be the primary factor influencing copepod body length in the lagoon of Venice, but the different species vary markedly in their response. Calanoid copepods (Acartia clausi, A. tonsa, Paracalanus parvus and Centropages ponticus) showed a more definite trend of size variation with temperature than the cyclopoids (Oithona nana and O. similis) and the harpacticoid Euterpina acutifrons. The size of the poecilostomatoids Oncaea media and O. subtilis was not affected by temperature, and was almost constant over time. Differences in the metabolic rates, longevity and specialization of calanoid, cyclopoid and harpacticoid copepods could explain their different responses to temperature.

    Notizen: Abstract This review covers discoveries made over the past 30–35 years that were important to our understanding of the synthetic pathway required for initiation of the antennae or branches on complex N-glycans and O-glycans. The review deals primarily with the author's contributions but the relevant work of other laboratories is also discussed. The focus of the review is almost entirely on the glycosyltransferases involved in the process. The following topics are discussed. (1) The localization of the synthesis of complex N-glycan antennae to the Golgi apparatus. (2) The “evolutionary boundary” at the stage in N-glycan processing where there is a change from oligomannose to complex N-glycans; this switch correlates with the appearance of multicellular organisms. (3) The discovery of the three enzymes which play a key role in this switch, N-acetylglucosaminyltransferases I and II and mannosidase II. (4) The “yellow brick road” which leads from oligomannose to highly branched complex N-glycans with emphasis on the enzymes involved in the process and the factors which control the routes of synthesis. (5) A short discussion of the characteristics of the enzymes involved and of the genes that encode them. (6) The role of complex N-glycans in mammalian and Caenorhabditis elegans development. (7) The crystal structure of N-acetylglucosaminyltransferase I. (8) The discovery of the enzymes which synthesize O-glycan cores 1, 2, 3 and 4 and their elongation.

    Notizen: Abstract Several evolutionary mechanisms have been identified in the literature that would generate altruism in humans. The most powerful (except for kin selection) and most controversial is group selection, as recently analyzed by Sober & D.S. Wilson. I do not take a stand on the issue of the existence of group selection. Instead, I examine the level of human altruism that could exist if group selection were an engine of human evolution. For the Sober & Wilson mechanism to work, groups practicing altruism must grow faster than other groups. I call altruistic behavior that would lead to faster growth ‘efficient altruism’. This often consists of cooperation in a prisoner's dilemma. ltruistic acts such as helping a temporarily hungry or injured person would qualify as efficient altruism. Efficient altruism would also require monitoring recipients to avoid shirking. Utilitarianism would be an ethical system consistent with efficient altruism, but Marxism or the Rawlsian system would not. Discussions of efficient altruism also help understand intuitions about fairness. We perceive those behaviors as ‘fair’ that are consistent with efficient altruism. It is important to understand that, even if humans are selected to be altruistic, the forms of altruism that might exist must be carefully considered and ircumscribed.

Amodel, as the product of modeling, might bevalued according to its correspondence to reality. Yet Levins' emphasis on provisionality and changeredirects attention to the processes ofmodeling, through which scientists select and generatetheir problems, define their categories, collect theirdata, compare competing models, and present theirfindings. I identify several points where decisionsare required that are not determined by nature. Thisinvites examination of the social considerationsmodelers are reacting to at the “sites of sociality”.Modelers must weave “socio-ecological webs” so thatthe models can be seen to represent their subjectmatter at the same time as the modelers secure thesupport of colleagues, collaborators and institutions,and enjoin others to act upon their conclusions. Notonly do theory justification and theory generationmerge, but the joint project becomes simultaneouslyphilosophical and sociological.

    Notizen: Abstract One of the principal difficulties in assessing Science as aProcess (Hull 1988) is determining the relationship between the various elements of Hull's theory. In particular, it is hard to understand precisely how conceptual selection is related to Hull's account of the social dynamics of science. This essay aims to clarify the relation between these aspects of his theory by examining his discussion of the``demic structure'' of science. I conclude that the social account cando significant explanatory work independently of the selectionistaccount. Further, I maintain that Hull's treatment of the demicstructure of science points us toward an important set of issues insocial epistemology. If my reading of Science as a Process iscorrect, then most of Hull's critics (e.g., those who focus solelyon his account of conceptual selection) have ignored promisingaspects of his theory.

Large-scale trends in functional complexity andcorrelations between functional complexity and other variables, such assize, have been proposed. However, the notion of number of functions hasalso been operationally intractable, in that no method has been developedfor counting functions in an organism in a systematic and reliable way.Thus, studies have had to rely on the largely unsupported assumption thatnumber of functions can be measured indirectly, by using number ofmorphological, physiological, and behavioral “parts” as a proxy. Here, amodel is developed that supports this assumption. Specifically, the modelpredicts that few parts will have many functions overlapping in them, andtherefore the variance in number of functions per part will be low. If so,then number of parts is expected to be well correlated with number offunctions, and we can use part counts as proxies for function counts incomparative studies of organisms, even when part counts are low. Alsodiscussed briefly is a strategy for identifying certain kinds of parts inorganisms in a systematic way.

    Notizen: Abstract Suppose that the human tendency to think of certain actions andomissions as morally required – a notion that surely lies at the heart of moral discourse – is a trait that has been naturallyselected for. Many have thought that from this premise we canjustify or vindicate moral concepts. I argue that this is mistaken, and defend Michael Ruse's view that the moreplausible implication is an error theory – the idea thatmorality is an illusion foisted upon us by evolution. Thenaturalistic fallacy is a red herring in this debate,since there is really nothing that counts as a ‘fallacy’ at all. If morality is an illusion, it appears to followthat we should, upon discovering this, abolish moraldiscourse on pain of irrationality. I argue that thisconclusion is too hasty, and that we may be able usefullyto employ a moral discourse, warts and all, withoutbelieving in it.

    Notizen: Abstract Phylogenetic relationships of Erysiphales within Ascomycota were inferred from the newly determined sequences of the 18S rDNA and partial sequences of the 28S rDNA including the D1 and D2 regions of 10 Erysiphales taxa. Phylogenetic analyses revealed that the Erysiphales form a distinct clade among ascomycetous fungi suggesting that the Erysiphales diverged from a single ancestral taxon. The Myxotrichaceae of the Onygenales was distantly related to the other onygenalean families and was the sister group to the Erysiphales calde, with which it combined to form a clade. The Erysiphales/Myxotrichaceae clade was also closely related to some discomycetous fungi (Leotiales, Cyttariales and Thelebolaceae) including taxa that form cleistothecial ascomata. The present molecular analyses as well as previously reported morphological observations suggest the possible existence of a novel evolutionary pathway from cleistothecial discomycetous fungi to Erysiphales and Myxotrichaceae. However, since most of these fungi, except for the Erysiphales, are saprophytic on dung and/or plant materials, the questions of how and why an obligate biotroph like the Erysiphales radiated from the saprophytic fungi remain to be addressed. We also estimated the radiation time of the Erysiphales using the 18S rDNA sequences and the two molecular clockes that have been previously reported. The calculation showed that the Erysiphales split from the Myxotrichaceae 190–127 myr ago. Since the radiation time of the Erysiphales does not exceed 230 myr ago, even when allowance is made for the uncertainty of the molecular clocks, it is possible to consider that the Erysiphales evolved after the radiation of angiosperms. The results of our calculation also showed that the first radiation within the Erysiphales (138–92 myr ago) coincided with the date of a major diversification of angiosperms (130–90 myr ago). These results may support our early assumption that the radiation of the Erysiphales coincided with the evolution of angiosperm plants.

    Notizen: Abstract The response of selected rotifers and cladocerans to Microcystis aeruginosa, offered as colonies and single cells, was compared to that on a diet of Chlorella vulgaris using the life table demography approach. The test zooplankton species were Simocephalus vetulus, Daphnia carinata, Moina macrocopa, Scapholeberis kingi, Ceriodaphnia cornuta, Brachionus calyciflorus and Hexarthra mira. To detect the development of resistance to toxins from Microcystis in zooplankton, in addition to the laboratory cultured strain of Ceriodaphnia cornuta, another strain of the same species was also used (designated as C. cornuta 2, this was collected from a pond containing Microcystis and cultured in the laboratory on Chlorella for a few weeks prior to experimentation). Experiments were conducted at 20 °C and 30 °C. Survivorship was high on Chlorella in most species but low on diets of Microcystis. Except for C. cornuta 2, S. kingi and S. vetulus, all other test species were adversely affected by Microcystis. The ability to utilise Microcystis improved at 30 °C in M. macrocopa, D. carinata and H. mira. The longest mean lifespan was recorded for C. cornuta 2 (25.3 ± 4.86 d) and the lowest for B. calyciflorus (0.58 ± 0.05 d). The highest net reproductive rate was observed for C. cornuta 1 (44.9 ± 4.88) and the longest generation time of 26.6 ± 2.13 d for S. vetulus. Among the cladocerans that showed positive values of population growth rate (r), M. macrocopa had the highest of 0.96 ± 0.04 per day.

    Notizen: Abstract A stromatolitic microbial mat extensively covers La Solana streambed, a calcareous Mediterranean stream. This stromatolite shows remarkable biological and physiological diversity. It is mainly composed by cyanobacteria, with Rivularia and Schizothrix as the most abundant taxa. The stromatolite is photosynthetically adaptated to the high irradiances reaching the streambed. Photosynthetically active chlorophyll is present even in the lowest layers of the stromatolite, indicating the presence of well-preserved cyanobacteria in that part. Diffusion of gases and nutrients within the stromatolite can be possible because of the high porosity of the crust. It has been experimentally established that the stromatolite recovers heterotrophic and autotrophic activities in a few hours, after being desiccated for long periods. Recovery after desiccation is indicative of the high resilience of this community to environmental extremes, which are common in Mediterranean climatic regimes. The stromatolitic community is adapted to nutrient limitation, both to low availability of inorganic phosphorus and nitrogen (that constrain growth of primary producers), and to low dissolved organic carbon (mainly affecting heterotrophs). Stromatolitic heterotrophs mainly rely on the organic carbon stored in the crust as the main organic carbon source. These strategies are the direct response of the stromatolite to oligotrophy, and justify the restricted occurrence in stream systems affected by organic pollution.

    Notizen: Abstract Three Calluna vulgaris (L.) Hull heathlands (1100, 1500 and 1780 m altitude) are studied in order to establish and estimate parameters which will be integrated in a discrete event simulation of heathland functioning in the case of a 2 °C climate warming. The sites, situated in the Chaîne des Puys and the Massif du Sancy (France), present similar conditions for Calluna growth: bedrock (trachyt), exposure and slope, so that they can be compared on a climatic basis. Main parameters sampled are: age distributions, life expectancies, layering probabilities, annual growths, biomasses (standing crop), C/N rates and potential respiratory of soils. In addition, experiments were carried out on germination, hypocotyle elongation and growth of mycorrhizal fungi in order to estimate the inhibitory-to-growth action against the substitution process. On one stand (exhibiting gaps) the patch structure of Calluna plants was recorded and mapped. The rising of biomass expected is calculated according to the Aerts's model, as a function of nitrogen availability which would increase under a scenario of climate warming. From a similar point of view, layering probabilities, life expectancies and inhibition of seedlings survival modelling – under a linear model assumption – are proposed.

    Notizen: Abstract Large-scale steady-state groundwater flow in atoll carbonate platforms results from temperature and salinity-induced density gradients. Atolls are built on top of a basaltic substrate that provides geothermal heating from beneath. Moreover, they are immersed in the tropical ocean where temperature decreases rapidly with depth. Groundwater circulation in these platforms has long been associated with the geothermal heat flux because it is capable of generating inward and upward flow of oceanic origin water by buoyancy effects. This study shows that hydraulic circulation occurs even in the absence of a geothermal flux because the combination of the cold subsurface ocean waters with the warm surface conditions is sufficient to maintain a convection cell within the carbonate platform. Using a one-dimensional analytical model, validated by more sophisticated two-dimensional simulations, we can investigate the interaction between these two driving forces. The flow rate inside the platform is, in fact, a function of the ratio of the geothermal flux to the temperature gradient in the ocean. It increases with the geothermal flux but decreases with the oceanic temperature gradient. This one-dimensional model also shows that taking salinity effects on density into account increases the flow rates transiting through the platform by a third.

    Notizen: Abstract We have been interested in Automatic Guided Vehicles (AGV) for several years. In this paper, we synthesize controllers for AGV applications using monocular vision. In particular, we are interested in road following and direction change tasks, and in analyzing the influence of extrinsic camera parameter perturbations on vehicle behavior. We use the bicycle as the kinematic vehicle model, and we choose the position of the white band on the road as the sensor signal. We define an interaction between the camera, which is mounted inside the vehicle, and the white band detected in the image space. Using this kind of interaction, we present how to use a pole assignment technique to solve the servoing task. We show the simulation and experimental results (1/10 scale demonstrator) with and without perturbations. We then investigate the use of a robust controller to slow down the effect of perturbations on the behavior of the vehicle.

    Notizen: Abstract In standard neuro-evolution, a population of networks is evolved in a task, and the network that best solves the task is found. This network is then fixed and used to solve future instances of the problem. Networks evolved in this way do not handle real-time interaction very well. It is hard to evolve a solution ahead of time that can cope effectively with all the possible environments that might arise in the future and with all the possible ways someone may interact with it. This paper proposes evolving feedforward neural networks online to create agents that improve their performance through real-time interaction. This approach is demonstrated in a game world where neural-network-controlled individuals play against humans. Through evolution, these individuals learn to react to varying opponents while appropriately taking into account conflicting goals. After initial evaluation offline, the population is allowed to evolve online, and its performance improves considerably. The population not only adapts to novel situations brought about by changing strategies in the opponent and the game layout, but it also improves its performance in situations that it has already seen in offline training. This paper will describe an implementation of online evolution and shows that it is a practical method that exceeds the performance of offline evolution alone.

    Notizen: Abstract Hydro power schemes operating in a free electricity market seek to maximize profits by differing generation rates to take best advantage of fluctuating selling prices, subject to the constraints of keeping storage lakes within their operational bounds and avoiding spillage losses. Various computer algorithms can be used in place of manual scheme operation to aid this maximization process, so it is desirable to quantify any profit gained from a given algorithm. A standard approach involves applying the algorithm to a period of past river flow records to see how much additional scheme income might have been obtained. This process requires the use of a hydro power scheme model, which inevitably can only approximate operational details, so the anticipated income gains are likely to be biased estimates of actual income gained from implementation of the algorithm. In addition to preliminary algorithm evaluation, it is desirable that hydro scheme managers have methodology to confirm anticipated income gain. Such confirmation can be difficult because true income gains are typically in the order of a few percentage and may not be easily distinguishable from background noise. We develop an approach, which allows estimation of true income gain for the situation where a change is made from manual to computer control of hydro power scheme operations, or upgrading from one maximization algorithm to another. The method uses a regression model to describe the former period of scheme operation. Postimplementation residuals from the regression predictions then provide estimates of actual income gain. The method can be sensitive to small but consistent income gains. Also, there is no requirement to construct any hydro scheme simulation model so bias effects should be considerably reduced. The approach was developed in the context of evaluating an income-maximization algorithm applied to a small hydro power scheme in the Kaimai Ranges of New Zealand. However, the methodology seems sufficiently simple and general to be applicable, with modification, to other power schemes moving toward increasing income through operational changes.

    Notizen: Abstract Variations in growth and survival of hatchery-reared post-metamorphicjuveniles of great scallop Pecten maximus prompted anexamination of settlement and postlarval development. The effects ofseawater flow and temperature on great scallop metamorphosis andpostlarvae were studied over a 4–5 week period. In allexperiments, and regardless of environmental conditions, great scallopmetamorphosed after a 2–3 week period with values of 35 to70%. Subsequently, spat numbers increased slightly. Spatmortality generally occurred from the third week onward and reachedlevels as high as 30% by the fifth week under standardconditions. At 20 °C, however, 60% mortality levels wererecorded. Differences in spat growth rate, ranging from 37 to 45 μmday−1, were noticed at different seawater flow ratesbut no clear tendency could be discerned. Temperature affected spatgrowth with an increase in size from 24 μm day−1 at15 °C to 35 μm day−1 at 18 °C. Conversely,growth was suppressed at 20 °C (14 μm day−1).For optimal metamorphosis and postlarval development in great scallop, aseawater flow of 4.3 L h−1 per sieve and a temperatureof 15 °C are recommended.

    Notizen: Abstract The recently developed natural radon tracer method has potential as a rapid, low‐cost, nondestructive, and noninvasive method for quantifying NAPL contamination. In the subsurface, radon‐222 (radon) is produced by the decay of naturally occurring radium‐226 contained in the mineral fraction of aquifer solids. In groundwater radon occurs as a dissolved gas, with a half‐life of 3.83 days. In the absence of NAPL, the radon concentration in groundwater quickly reaches a maximum value that is determined by the mineral composition of the aquifer solids, which controls the rate of radon emanation. In the presence of NAPL, however, the radon concentration in the groundwater is substantially reduced due to the preferential partitioning of radon into the organic NAPL phase. A simple equilibrium model and supporting laboratory studies show the reduction in radon concentration can be quantitatively correlated with residual NAPL saturation. Thus, by measuring the spatial distribution in radon it may be possible to identify locations where residual NAPL is present and to quantify the NAPL saturation. When the basic processes of partitioning, radon emanation from the aquifer solids, and first‐order decay are incorporated into an advective/dispersive transport model, good agreement is obtained with the results of laboratory and field experiments. Model sensitivity analyses shows many factors can contribute to the radon concentration response, including the length of the NAPL zone, NAPL saturation, groundwater velocity, porosity, and radon emanation. Thus, care must be taken when applying the radon method to locate and quantify NAPL contamination in the subsurface.

    Notizen: Abstract Peat is an alternative filter medium for the treatment of various waste streams including septic tank effluent. The water holding capacity and adsorption capacity of peat make it a favorable filter medium over sand or gravel which are commonly used as the filter medium for the drainage field of septic systems. This paper presents the results of a field study to evaluate the hydraulics of a peat filter used to treat the septic tank effluent from a public school west of Ottawa, Canada. An experimental box was placed within the filter during its construction to provide access to the vertical profile of the peat layer. The filter is periodically pulsed with septic tank effluent, which is distributed over the top of the filter and flows vertically through the peat. The filter was instrumented with tensiometers and transducers to monitor the pore water pressures in response to a pulse of septic tank effluent. An in-depth study of the hydraulics of the system was completed. The soil moisture retention curve and the hydraulic conductivity as a function of density were determined in laboratory experiments. A one-dimensional unsaturated flow model was used to predict the pressure response due to a pulse. A comparison of the field and model results illustrates the impact of the density variations, and the corresponding hydraulic conductivity variations, on the model predictions. The compaction of the peat is an important design consideration for the filter since it directly impacts the flow characteristics and the hydraulic retention time within the filter.

    Notizen: Abstract Ecosystem Management (EM) is a new holistic approach to the management of natural resources. It integrates the scientificknowledge on ecological interrelationships and the complex socioeconomic and political frame of values, aiming at the long-term sustainability of a region, i.e., the ecosystem. Itincludes the human, biological and natural dimensions, and canregulate the internal structures and functions of the ecosystem,and the inputs and outputs from it, so that solutions to development and protection problems can be achieved and be socially well-accepted. It has been given attention since 1991,mainly in the U.S.A., where the existing legal framework formeda basis for EM. The former experience of programs based on the traditional methods of management, the goal of sustainable development stated by many governmental, public or private agencies, and the advent of scientific knowledge, permitted the development of a theoretical basis for EM. Today, a significant number of EM programs have been established and are in progressin the U.S.A. Here the concept, general principles and methodology of EM are reviewed, and the EM program in the Stateof Florida in the U.S.A. is presented, which is considered a successful case study. In addition, the examples of managementof two very important ecosystems in Northern Greece are critically presented for comparison, in order to evaluate if they have EM characteristics.

    Notizen: Abstract Two-dimensional, vertically-averaged hydrodynamic and sediment transport models were developed and applied as part of a PCB fate and transport modeling study of Thompson Island Pool (TIP), Upper Hudson River. Mechanistic formulations were used to simulate cohesive and non-cohesive suspended load transport; site-specific data were extensively used to determine model inputs. This modeling approach is compared and contrasted to non-mechanistic solids transport sub-models used in other contaminant fate studies. A minimum number of model parameters were adjusted to calibrate the sediment transport model using data collected during the 1994 spring flood. The model was validated during the 1997 spring flood and for a 22-year (1977–1998) period. Successful calibration and validation of the model showed that: (1) deposition and resuspension processes were realistically and accurately formulated in the model; (2) the model is an effective diagnostic tool for quantitatively evaluating net deposition and erosion from various areas of TIP; and (3) sediment transport results can be coupled with a PCB fate model with a high degree of confidence.

    Notizen: Abstract The development, testing and application of a dynamic two-dimensional (longitudinal-vertical) mass balance model for dissolved oxygen (DO) and chlorophyll (Chl) for rivers is documented that for the first time accommodates both the oxygen demand and filtering effects of zebra mussels. The test system is a phytoplankton-rich section (∼ 15 km long) of the Seneca River, NY, that is believed to represent an upper bound of the impact of this exotic invader. Changes in common measures of water quality of the river brought about by the zebra mussel invasion are reviewed and related longitudinal patterns in DO, Chl, and Secchi disc transparency are documented. Model testing is supported by comprehensive measurements of DO, Chl, and various forcing conditions over a three-month period, and independent determinations of several model coefficients. Wide variations in the areal consumption rate of DO (ZOD; g·m−2·d−1) and filtering rate (m3·m−2·d−1) of zebra mussels, as determined through model calibration, occurred over the study period. Values of ZOD in areas with dense zebra mussel populations at times (e.g., 〉 50 g·m−2·d−1) were an order of magnitude greater than the sediment oxygen demand associated with organically enriched deposits. The value of determinations of these fluxes from model calibration procedures is evaluated within the context of the limitations of protocols presently available to support independent specification of these rates. Model analyses are conducted to evaluate the relative magnitude of source and sink processes for DO and Chl, the potential operation and implications of feedback from low DO levels on oxygen consumption by zebra mussels, and the sensitivity of model simulations to selected sources of uncertainty and variability. Model projections of oxygen resources of the river are presented in a probabilistic format in evaluating reductions in zebra mussel biomass that would be necessary to eliminate violations of standards and regain assimilative capacity.

Perceptions and activities related topast and future dry periods was investigated throughinterviews with managers at the ten major English andWelsh water supply/sewerage companies, and severalsmaller, water-only companies. Several of thecompanies report that they have observed a trendtowards drier summers in their regions, and a number of companies say that they have observed a changetowards more intensive rainfall of shorter duration. Recent supply measures in a number of regions havebeen aimed at improving storage and distributionrelated to the perceived change in rainfall intensity.A new requirement to incorporate regional climatechange scenarios in future supply assumptions appearsto have had little impact on planning in the region todate. Many water resource planners believe that thescenarios generated are too aggregated and do notencourage a precautionary approach to planning. Somemanagers believe that records of historical droughtconditions, such as experienced in 1933/1934 or 1995/1996,as worst-case scenarios provide a better basis for planning.

    Notizen: Abstract There has been a great deal of interest in determining phylogenetic relationships within the family Dasyuridae due to the widespread distribution, ecological diversity, and relative plesiomorphy of this taxon within the Australasian marsupial radiation. In the past, it has been extremely problematic to determine the phylogenetic relationships among species within Dasyurus, with numerous studies using both morphological and molecular characters providing different topologies. Here, the mitochondrial DNA (mtDNA) control region is used as a novel set of characters in an attempt to identify relationships among the six closely related extant species. Sequences were obtained from multiple individuals representing all extant species of quolls including, when possible, individuals from different geographical regions. Sequences were analyzed using both parsimony criteria and neighbor-joining methods. Results presented here concur with those of Krajewski et al. (1997) in (1) placing D. geoffroii in a highly supported clade with D. spartacus, (2) resolving a monophyletic group of D. albopunctatus + D. geoffroii + D. spartacus, and (3) placing D. hallucatus as the sister taxon to all other species of quolls. Results also show two highly supported and geographically distinct clades of D. maculatus (Tasmanian and mainland) that do not correspond to the currently used subspecific nomenclature. Preliminary results also indicate that there are different clades among geographic groups of D. hallucatus that warrant further investigation. The mtDNA control region is a highly variable locus and may be used in forensic tests for species identification in this genus.

    Notizen: Abstract The Oryza officinalis complex is a genetically diverse, tertiary genepool of rice. We analyzed part of the primary structure of the integrase coding domain (ICD) of a gypsy-like retrotransposon from species of the O. officinalis species complex. PCR was performed with degenerate primers that hybridized to conserved sequences in the integrase genes of gypsy-type retrotransposons, using total DNA from different species of the O. officinalis complex as templates. Cloning and sequencing of the PCR products showed that the amplified fragments are highly homologous to each other (75–90%) and belong to one family of retrotransposons that is related to the previously studied RIRE-2 element from rice. Two main subfamilies of 292 and 351 bp were distinguished. Analysis of primary sequence data supports previous reports that sequence divergence during vertical transmission has been the major influence on the evolution of gypsy-type retrotransposons in Oryza species. Based on sequence data phylogenetic relationships among species of the O. officinalis complex were estimated. The data suggests that O. eichingeri is more closely related to the ancestral species of the complex.
