    Notizen: Summary Embryos of Drosophila melanogaster were irradiated in the presumptive head region with a UV-laser microbeam of 20 μm diameter at two developmental stages, the cellular blastoderm and the extended germ band. The ensuing defects were scored in the cuticle pattern of the head of the first-instar larva, which is described in detail in this paper. The defects caused by irradiating germ band embryos when morphologically recognisable lobes appear in the head region were used to establish the segmental origin of various head structures. This information enabled us to translate the spatial distribution of blastoderm defects into a fate map of segment anlagen. The gnathal segments derive from a region of the blastoderm between 60% and 70% egg length (EL) dorsally and 60% and 80% ventrally. The area anterior to the mandibular anlage and posterior to the stomodaeum is occupied by the small anlagen of the intercalary and antennal segments ventrally and dorsally, respectively. The labrum, which originates from a paired anlage dorsally at 90% EL, is separated from the remaining head segments by an area for which we did not observe cuticle defects following blastoderm irradiation, presumably because those cells give rise to the brain. The dorsal and lateral parts of the cephalo-pharyngeal skeleton appear to be the only cuticle derivatives of the non-segmental acron. These structures derive from a dorso-lateral area just behind the putative brain anlage and may overlap the latter. In addition to the segment anlagen, the regions of the presumptive dorsal pouch, anterior lobe and post-oral epithelium, whose morphogenetic movements during head involution result in the characteristic acephalic appearance of the larva, have been projected onto the blastoderm fate map. The results suggest that initially the head of the Drosophila embryo does not differ substantially from the generalised insect head as judged by comparison of fate map and segmental organisation.

    Notizen: Summary In this paper experiments concerning some aspects of the development of pole cells and midgut progenitors in Drosophila are reported. Cells were labelled by injecting horseradish-peroxidase (HRP) in embryos before pole bud formation and transplanted at different stages into unlabelled embryos, where the transplanted cells developed together with the unlabelled cells of the host. The hosts were then fixed and stained at different ages in order to demonstrate the presence of HRP in the progenies of transplanted cells. The main conlusions of the study are as follows. The gonads are the only organ to the formation of which pole cells normally contribute; those pole cells which do not participate in the formation of the gonads are finally eliminated or degenerate. Since the number of primordial germ cells in the gonads is the same irrespective of the number of pole cells present in the embryo, an (unknown) mechanism must exist regulating the final number of pole cells in each of the gonads. After their formation and before reaching the gonads, pole cells have been found to divide only up to two times. With respect to the midgut progenitors, the cells of both anlagen have been found to be committed to develop into midgut, although they behave as equivalent in that they do not apparently distinguish between the anterior and posterior anlage. Midgut progenitors have been found to divide a maximum of three times and to produce two different types of cells, epithelial cells of the midgut wall and spindle-like cells located internally in the gut.

    Notizen: Summary A comparison of pupation-temperature range was made in the laboratory on a temperature gradient (3–38°C) using 12 species ofDrosophila representing four species groups and four different ecological backgrounds (temperate-montane forest:virilis group; desert;repleta group; cosmopolitan:melanogaster group; tropical forest:willistoni group). Within groups, differences are found which usually reflect species' distributions. Comparisons of species' mating-, oviposition- and pupation-temperature ranges reveal that pupation most-often occurs at temperatures beyond those for mating and oviposition. Each species reflects a different combination of temperature effects. Individual species have different temperature-limits for mating, oviposition and pupation. Temperatures permissive for one response are not predictive of limits on other responses. Among species, temperature can affect a particular response differently. Within groups, species differences can be at high and/or low temperatures for any response, and temperature effects among closely related species can manifest themselves in one, or any combination of responses. One cannot predict which responses will be most and least limited, or at which end of the temperature scale a response will be most limited. Among groups,common, but notabsolute temperature ranges generally correspond to the geographic distributions and ecological backgrounds of the species triads. The evaluation of temperature effects on species, based on a single activity, may not be adequate for predicting adaptive strategies.

    Notizen: Summary Genetically marked maroon-like (mal) clones were induced by mitotic recombination with X-rays at the blastoderm stage in mal/mal + heterozygotes and were analysed in differentiated Malpighian tubules (MT). Marked cells were not confined to single anterior (MA) or posterior (MP) tubules, but were distributed among the four tubules. About 70% of the clones with two or more cells were fragmented, i.e. mal cells were separated by wild-type cells. Since the clones contain, on average, 6 cells and the differentiated MT consist of 484 cells (2 × 136 MA cells, 2 × 106 MP cells), we estimate that there are about 80 cells in the blastoderm anlage which on average pass through two to three mitoses. With increasing radiation doses (254 R, 635 R, 1270 R) a linear increase in clone frequency is observed. The mean sizes and size distributions of clones, however, remain unchanged. Since the increasing radiation dose also results in fewer differentiated Malpighi cells, we assume that regeneration does not occur. Therefore, size distributions of marked clones presumably represent real mitotic patterns in normogenesis. We suggest that essentially three successive mitoses take place, with a decreasing fraction of cells showing mitotic activity. Only a small fraction of cells goes through a fourth or even a fifth mitosis. Marked non-Minute clones induced in Minute heterozygotes are more frequent, but are not larger than non-Minute clones in wild-type background. Therefore, compartment boundaries cannot be recognized by this method. However, frequencies of marked cells found simultaneously in MA and MP pairs or in several single tubules of the same individuals are significantly higher than frequencies of multiple recombination events predicted by the Poisson distribution. From this, we conclude that neither the MA pair nor the MP pair nor single tubules represent compartments of the MT anlage.

    Notizen: Summary A method is presented which allows the study of the progeny of single cells during Drosophila embryogenesis. Cells from various larval anlagen of donor embryos labelled with a lineage tracer are individually transplanted from defined positions into similar, or different, positions in unlabelled hosts. The clones produced by these cells can be seen in whole mounts or in sections of fixed material, when using a histochemical marker (i.e. HRP), and/or in living embryos, when using fluorescent lineage tracers. The characteristics of the clones disclose lineage parameters, such as division patterns, morphogenetic movements and differentiation. The method is especially useful for testing the respective roles of positional information and cell lineage on the commitment of progenitor cells by transplanting these cells into heterotopic positions or into hosts of different genotypes.

    Notizen: Summary In this paper we describe a new allele of suppressor of forked, su(f) hd37, referred to as hd37, which was isolated in a hybrid dysgenesis mutation screen and is shown to be P induced by its high frequency of reversion in hybrid dysgenic crosses, and by in situ hybridization. hd37 suppresses forked and fails to complement the forked suppression of known su(f) alleles. However, it complements the recessive lethality of alleles in both of the su(f) lethal complementation groups. We also describe a new phenotypic effect of su(f) alleles, the enhancement of Minute(3)i 55. Recessive lethal alleles enhance the lethal effects of this Minute, but hd37 does not. The temperature sensitive period for forked bristle suppression by hd37 was found to be very narrow, consisting of a short interval (12–18 h) immediately before bristle formation. These results suggest that the several genetic functions associated with this locus may be genetically separable.

    Notizen: Summary Mutations previously known to affect early neurogenesis inDrosophila melanogaster have been found also to affect the development of the peripheral nervous system. Anti-HRP antibody staining has shown that larval epidermal sensilla of homozygous mutant embryos occur in increased numbers, which depend on the allele considered. This increase is apparently due to the development into sensory organs of cells which in the wild-type would have developed as non-sensory epidermis. Thus, neurogenic genes act whenever developing cells have to decide between neurogenic and epidermogenic fates, both in central and peripheral nervous systems. Different regions of the ectodermal germ layer are distinguished with respect to their neurogenic abilities.

    Notizen: Summary Mutations in seven different maternal-effect loci on the second chromosome of Drosophila melanogaster all cause alterations in the anterior-posterior pattern of the embryo. Mutations in torso (tor) and trunk (trk) delete the anterior- and posterior-most structures of the embryo. At the same time they shift cellular fates which are normally found in the subterminal regions of the embryo towards the poles. Mutations in vasa (vas), valois (vls), staufen (stau) and tudor (tud) cause two embryonic defects. For one they result in absence of polar plasm, polar granules and pole cells in all eggs produced by mutant females. Secondly, embryos developing inside such eggs show deletions of abdominal segments. In addition, embryos derived from staufen mothers lack anterior head structures, embryos derived from valois mothers frequently fail to cellularize properly. Mutations in exuperantia (exu) cause deletions of anterior head structures, similar to torso, trunk and staufen. However in exu, these head structures are replaced by an inverted posterior end which comprises posterior midgut, proctodeal region, and often malpighian tubules. The effects of all mutations can be traced back to the beginning stages of gastrulation, indicating that the alterations in cellular fates have probably taken place by that time. Analysis of embryos derived from double mutant mothers suggests that these three phenotypic groups of mutants interfere with three different, independent pathways. All three pathways seem to act additively on the system which specifies anterior-posterior cellular fates within the egg.

    Notizen: Summary The legs of flies from 16 different mutant strains ofDrosophila melanogaster were examined for abnormal cuticular polarities and extra joints. The strains were chosen for study because they manifest abnormal cuticular polarities in some parts of the body (10 strains) or because they have missing or defective tarsal joints (6 strains). All but three of the stocks were found to exhibit misorientations of either the bristles, hairs, or “bract-socket vectors” on the legs. The latter term denotes an imaginary vector pointing from a hairlike structure called a “bract” to the bristle socket with which it is associated. On the legs of wild-type flies nearly all such vectors point distally, as do the bristles and hairs. In the mutant flies, the most common vector misorientation is a 180° reversal. When the bract-socket vectors of adjacent bristle sites in the same bristle row point toward one another, the distance between the sites is frequently abnormally large, whereas when the vectors point in opposite directions, the interval is frequently abnormally small. This correlation is interpreted to mean that bristle cells actively repel one another via cytoplasmic extensions that are longer in the direction of the bract-socket vector than in the opposite direction. Repulsive forces of this kind may be responsible for “fine-tuning” the regularity of bristle spacing in wild-type flies. Extra tarsal joints were found in eight of the 16 strains. A ninth strain completely lacking tarsal joints appears in some cases to have an extra tibia-basitarsus joint in its tibia. Whereas the tarsi of wild-type flies contain four joints, the tarsi ofspiny legs mutant flies contain as many as eight joints. In this extreme extra-joint phenotype, four of the joints correspond to the normal wild-type joints, and there is an extra joint in every tarsal segment except the distal-most (fifth) segment. Nearly all such ectopic extra joints have inverted polarity. In other strains the extra tarsal joints are located mainly at the wild-type joint sites, and joints of this sort have wild-type polarity. The alternation of normal and inverted (extra) joints inspiny legs resembles the alternation of normal and inverted (extra) body segment boundaries in the embryonic-lethal mutantpatch, suggesting that tarsal and body segmentation may share a common patterning mechanism.

    Notizen: Summary Various types of sense organs are arranged in a highly reproducible pattern on the thoracic and abdominal segments ofDrosophila embryos and larvae. We describe this pattern and identify the neurons that innervate each sense organ. This identification is confirmed by the analysis of partial deficiencies for the scute region, which delete specifically some of the sense organs and their innervating neurons. Since our description of the sense organs accounts for all the sensory neurons that have been identified in the embryo, we believe that this description is accurate and complete, except in the terminal segment, where some sense organs remain to be identified.

    Notizen: Summary Some aspects of neural and epidermal cell lineages during embryogenesis of Drosophila melanogaster were studied by transplanting horseradish-peroxidase-(HRP-) labelled ectodermal cells from young gastrula donors into host embryos of similar ages. Heterotopic transplantations permitted us to assess the degree of commitment already attained by the transplanted cells. The resulting cell clones showed normal characteristics of cytodifferentiation and cell number. The results indicate that epidermal progenitors perform a maximum of three mitoses during embryonic development, whereas neuroblasts may perform more than ten mitoses. Clone size distribution is in both cases scattered, suggesting either a rather irregular mitotic pattern or cell death. As indicated by heterotopic transplantations, the neurogenic ectoderm for the ventral nervous system exhibits different neurogenic abilities in its different regions, decreasing from medial to lateral; we discuss the hypothesis that some medially located cells of the young gastrulating embryo could be committed towards the neural fate before segregating from the ectoderm. On the other hand, the cells of the dorsal ectodermal regions at the same stage seem to be indifferent with respect to commitment, for they are able to give rise to central neural lineages following their transplantation in the neurogenic region.

    Notizen: Summary The yeast communities from slime fluxes of three deciduous trees (Prosopis juliflora, Populus fremontii and Quercus emoryi) and the necroses of two cacti (Opuntia phaeacantha and Carnegiea gigantea) were surveyed in the region of Tucson, Arizona. In addition, the yeasts carried by dipterans associated with the fluxes or necroses (Drosophila carbonaria, D. brooksae, D. nigrospiracula, D. mettleri, and Aulacigaster leucopeza) were sampled. The results indicate that each host sampled had a distinct community of yeasts associated with it. The dipterans, which can act as vectors of the yeasts, deposited yeasts from other sources in addition to those found on their associated hosts. It is argued that host plant physiology is relatively more important than the activity of the vector in determining yeast community composition. Furthermore, the average number of yeast species per flux or necrosis is not different from the average number of yeast species per fly. It is hypothesized that the vector may affect the number of species per individual flux or not, and that the number is lower than the rot or necrosis could potentially support.

    Notizen: Abstract Extracts of mature grains of a large number of aneuploid derivatives of Triticum aestivum cv. Chinese Spring and of the members of five wheat-alien chromosome addition series were subjected to isoelectric focusing in polyacrylamide gels in order to study the genetic control of superoxide dismutase (SOD). Evidence was obtained that homologous structural genes for the mitochondrial form of SOD are located in the long arms of the homoeologous group 2 chromosomes of Chinese Spring and in chromosome 2R of Secale cereale cv. Imperial. The SOD gene loci located in chromosomes 2A, 2B, 2D, and 2R were designated Sod-A1, Sod-B1, Sod-D1, and Sod-R1, respectively. Chromosome-arm pairing data indicate that 2DL is not homoeologous to either 2AS or 2BL. The results of this study suggest, however, that 2BL is partially homoeologous to both 2AL and 2DL.

    Notizen: Abstract The aldox-2 locus in Drosophila melanogaster has been shown to affect differentially three molybdoenzymes, aldehyde oxidase, pyridoxal oxidase, and xanthine dehydrogenase. These effects are most obvious at times surrounding the pupal-adult boundary, when the normal organism accumulates large amounts of these enzymes in their active form. This locus has been more precisely mapped genetically to 2–82.9±2.1, with complete concordance between the effects of all recombinant chromosomes on all three enzymes. The cytogenetic location has also been determined to be between 52E and 54E8, with the likelihood that it lies within the region 54B1-54E8. The aldox-2 mutant allele has no visible phenotype and is completely recessive for enzyme effects at all stages tested. Segmental duplication of this region, including the aldox-2 + allele, has no apparent effect on the visible phenotype or the enzymatic activity. The mutant aldox-2 allele has no effect on the developmental expression of two unrelated enzymes, 6-phosphogluconate dehydrogenase and NADP+-dependent isocitrate dehydrogenase. The effects of this locus on aldehyde oxidase, xanthine dehydrogenase, and pyridoxal oxidase suggest that this locus may code for a product involved in the synthesis of the molybdenum cofactor common to these enzymes.

    Notizen: Abstract Aldox “null” alleles which were isolated from natural populations in Great Britain and North Carolina were analyzed for complementation. No complementation was observed between any combinations of “null” alleles for aldehyde oxidase (AO) specific activity in late third-instar larvae and newly emerged adults. AO immunologically cross-reacting material (AO-CRM) was quantitated in all homozygous stocks at both developmental stages as well as all allelic combinations in newly emerged adults. When the adult organism contains only Aldox n alleles, the polypeptides are not immunologically recognizable or may be rapidly degraded. Larvae and adults have different abilities to degrade mutationally altered enzymatically inactive AO polypeptide or synthesize them differentially. This is indicated by easily measurable AO-CRM levels in late third-instar larvae of Aldox n homozygotes, while newly emerged adult Aldox n homozygotes have very little, if any, AO-CRM. Newly emerged adult heterozygotes of Aldox n /Aldox + do have increased AO-CRM, indicating that the Aldox n alleles can code for a polypeptide which can be “rescued” if Aldox + gene product is present. Heterozygotes containing an Aldox + allele with a deficiency for the Aldox region produce 74.2% of the AO-CRM found in Aldox + homozygotes. This may indicate the presence of trans-acting factors which serve to activate gene expression in a system in which each gene copy is not maximally expressed.

Although temperature was found to have a significant effect on both kinetic and biochemical properties ofDrosophila ADH, no significant differential effect was observed between the major ADH allozymes. The results are discussed within the context of the selective maintenance ofAdh polymorphism in natural populations.

    Notizen: Abstract The role of sn-glycerol-3-phosphate oxidase (GPO; EC 1.1.99.5) in the variation of ethanol tolerance inDrosophila melanogaster was assessed in isofemale lines derived from individuals collected at the Chateau Tahbilk Winery and Wandin North Orchard of Victoria, Australia. When fed an undefined medium (semolina-treacle) with 6% ethanol (v/v), larvae of lines with high GPO activities survived better than did larvae of lines with low GPO activities. Although GPO was induced to higher activity levels by dietary ethanol in larvae of all the test lines, GPO activity was greater in lines representing the area outside the wine cellar. This implied that the cellar environment selected against individuals with high levels of GPO. These data do not explain the established difference in tolerance between cellar and outside populations. The GPO activities of lines were not dependent upon the activities of the lipogenic enzyme, glycerol-3-phosphate dehydrogenase; the major ethanol-degrading enzyme, alcohol dehydrogenase; or the citric acid cycle enzyme, fumarase. Thus, GPO activity is an important component of the metabolic mechanism of ethanol tolerance in larvae, but the mode of action of GPO has not been defined.

    Notizen: Abstract The mutualistic interactions of cactophilicDrosophila and their associated yeasts in the Sonoran Desert are studied as a system which has evolved within the framework of their host cactus stem chemistry. Because theDrosophila-yeast system is saphrophytic, their responses are not thought to directly influence the evolution of the host. Host cactus stem chemistry appears to play an important role in determining where cactophilicDrosophila breed and feed. Several chemicals have been identified as being important. These include sterols and alkaloids of senita as well as fatty acids and sterol diols of agria and organpipe cactus. Cactus chemistry appears to have a limited role in directly determining the distribution of cactus-specific yeasts. Those effects which are known are due to unusual lipids of organpipe cactus and triterpene glycosides of agria and organpipe cactus.Drosophilayeast interactions are viewed as mutualistic and can take the form of (1) benefits to theDrosophila by either direct nutritional gains or by detoxification of harmful chemicals produced during decay of the host stem tissue and (2) benefits to the yeast in the form of increased likelihood of transmission to new habitats. Experiments on yeast-yeast interactions in decaying agria cactus provide evidence that the yeast community is coadapted. This coadaptation among yeasts occurs in two manners: (1) mutualistic increases in growth rates (which are independent of the presence ofDrosophila larvae) and (2) stabilizing competitive interactions when growth reaches carrying capacity. This latter form is dependent on larval activity and results in benefits to the larvae present. In this sense, the coadapted yeast community is probably also coadapted with respect to itsDrosophila vector.

    Notizen: Abstract Microhabitat preferences ofDrosophila pseudoobscura strains were examined in a Waddington maze, with an emphasis on learning how early environment affected adult habitat choice. The genotypes were roughly those expected in a natural population; the environmental variables included light, temperature, and food. It was found that (1) the different genotypes chose habitats differently; (2) early experience affected subsequent habitat choice; and (3) the effect of early experience was complex, as preference for one niche dimension (temperature) was reinforced by experience with the generally preferred value, preference for another niche dimension (light) was weakened by experience with the generally preferred value, and preference for other niche dimensions (food) was generally unaffected by experience. In this study the contribution to the total chi square was about equal from genotype and from environment. The significance of these findings for studies of dispersal and population structure of natural populations is discussed.

    Notizen: Abstract At a moderate concentration (2.5%, v/v) dietary ethanol reduced the chain length of total fatty acids (FA) and increased the desaturation of short-chain FA in Drosophila melanogaster larvae with a functional alcohol dehydrogenase (ADH). The changes in length in total FA were postulated to be due to the modulation of the termination specificity of fatty acid synthetase. Because the ethanol-stimulated reduction in the length of unsaturated FA was blocked by linoleic acid, it was thought to reflect the properties of FA 9-desaturase. Although the ethanol-stimulated reduction in chain length of unsaturated FA was also observed in ADH-null larvae, ethanol promoted an increase in the length of total FA of the mutant larvae. Thus, the ethanolstimulated change in FA length was ADH dependent but the ethanol effect on FA desaturation was not. Ethanol also stimulated a decrease in the relative amount of phosphatidylcholine and an increase in phosphatidylethanolamine. Because similar ethanol-induced changes have been found in membrane lipids of other animals, ethanol may alter the properties of membranes in larvae. It is proposed that ethanol tolerance in D. melanogaster may be dependent on genes that specify lipids that are resistant to the detrimental effects of ethanol.

    Notizen: Abstract Measurements of wing-beat frequency (WBF) have been used to characterize flight muscle metabolic rate in Drosophila melanogaster during tethered flight. Progeny of crosses between 17 X-chromosome substitution lines and three null-activity stocks have been studied in order to determine the effect on flight metabolism of sharply reduced activity of α-glycerophosphate dehydrogenase (αGPDH). It was found that flies with an approximate 50% reduction in αGPDH activity have a metabolic rate that is, in most cases, indistinguishable from that of wild-type flies and, in the most extreme cases, reduced by only 4%. These results demonstrate that αGpdh is not a “major gene” for flight metabolism, in the quantitative genetic sense of the term. These results are in agreement with the Kacser and Burns (1973, 1979, 1981) theory of flux, which postulates that the activity of an enzyme embedded in a multienzyme pathway can sometimes vary from wild-type to very low levels (perhaps 5–10% wild type) with no significant effect on flux through the total pathway.

    Notizen: Summary The behaviour of plastids and mitochondria during the formation and development of the male gametophyte of Chlorophytum comosum has been investigated using electron microscopy. During first pollen mitosis an intracellular polarization of plastids occurs in that the plastids are clustered in the centre of the microspore. The originating generative cell normally lacks plastids. Only in a small number of microspores have plastids been observed near the dividing nucleus of the microspore and later on in the generative cell. These observations agree with the genetic investigations of Collins (1922) on the mode of plastid inheritance which demonstrated a small amount of biparental plastid inheritance in Chlorophytum. The cytological mechanisms underlying plastid polarization during the first pollen mitosis are discussed.

    Notizen: Summary A 5BS/5RmS translocation chromosome spontaneously recovered from a ‘Chinese Spring’ — Secale montanum wheat-rye telocentric 5RmS addition line has been identified and cytologically studied using C-banding in somatic and meiotic cells. Analysis of the translocated chromosome showed that a terminal segment of the short arm of 5B had been replaced by a short terminal region of chromosome arm 5RmS. The translocation led to the deletion of the genetic system promoting pairing located in 5BS, which is slightly compensated for when doses of 5RmS are increased, indicating homoeology to wheat chromosome 5BS. The α-amylase phenotype in 5B/5Rm translocated material was studied and found to be identical to that of ditelocentric line 5BL of ‘Chinese Spring’. An effect on the α-amylase activity was detected as a result of the removal of the terminal region of 5BS, perhaps as a consequence of variation in dormancy period duration.

The observations, compared to the euploid control and relative to previous data found on 7-day-old etiolated seedlings (G7 stage) of the same lines lead to the following statements: 1) the structural genes of 24 spots can be assigned to 12 chromosome arms; 2) regulatory effects are completely different between the 1F and the G7 stages which may indicate that the regulation of protein amounts is often stage-specific; 3) no case of complete gene dosage compensation is observed among 4 groups of hypothesized homoeoallelic products; 4) homoeologous DT lines do not manifest similar effects which suggest the absence of homoeology for the detected regulatory effects.

    Notizen: Summary Intergeneric hybrids were produced between common wheat, Triticum aestivum (2n=6x=42, AABBDD) and wheatgrass, Etymus caninus (Agropyron caninum) (2n=4x=28, SSHH) — the first successful report of this cross. Reciprocal crosses and genotypes differed for percent seed set, seed development and F1 hybrid plant production. With E. caninus as the pollen parent, there was no hybrid seed set. In the reciprocal cross, seed set was 23.1–25.4% depending upon wheat genotype used. Hybrid plants were produced only by rescuing embryos 12–13 days post pollination with cv ‘Chinese Spring’ as the wheat parent. Kinetin in the medium facilitated embryo germination but inhibited root development and seedling growth. The hybrids were vigorous, self sterile, and intermediate between parents. These had expected chromosome number (2n=5x=35, ABDSH), very little chromosome pairing (0.51 II, 0.04 III) and some secondary associations. The hybrids were successfully backcrossed with wheat. Chromosome number in the BC1 derivatives varied 54–58 with 56 as the modal class. The BC1 derivatives showed unusually high number of rod bivalents or reduced pairing of wheat homologues. These were sterile and BC2 seed was produced using wheat pollen.

    Notizen: Summary Fertile r0 plants of the winter wheat line ND7532 (Triticum aestivum L.) were regenerated from callus tissue after 60–190 days in culture. Seeds produced from these self-pollinated plants were planted in the field. Of the 5586 R1 plants, 32 differed for one or more agronomic traits from plants not passed through tissue culture process. Gliadin electrophoregrams were prepared from bulk samples of R2 seed from these 32 plants. Four of the 32 produced gliadin patterns different from controls, so 12 seeds of each of these four lines were examined individually. Three of the four mutant lines were fixed for the presence of a mutant protein of 50 relative mobility units (RMU) and the corresponding loss of a parental protein of 26 RMU. The remaining line segregated for the presence/absence of band 50 and the corresponding loss/retention of band 26. The mutant protein of 50 RMU was never seen in control plants. This indicated that either band 50 was coded for by a mutant gene allelic to the gene that coded for band 26 or that bands 26 and 50 were coded for by two different structural alleles under the control of a common regulatory locus. Each of the 12 seeds from the four mutant lines contained a prominent protein band at 30 (RMU), which was only observed as a faint band in one control seed. The types of variation in gliadin patterns observed in somaclones of ND7532 were similar to those reported for the line ‘Yaqui 50E’, except that, gliadin changes occurred less frequently in ND7532.

    Notizen: Summary The effects of different doses of various steroids on growth, and on costal and epiphyseal chondrocytes, have been studied in prenatal, immature, and adult Long-Evans rats using histochemical techniques, and both light and electron microscopy. Both prenatal and postnatal treatments have been employed. The steroids used were cortisone (CA), betamethasome (BM), and, in the prenatal group only, dexamethasone (DM). Body weight is reduced in all treated rats (except the low dose of CA) by day 17 of gestation, with greater weight reductions occurring in rats receiving the higher dose level of each steroid. In rats treated prenatally or neonatally, and sacrificed postnatally on days 39–43 or days 116–127, body weights, and tibial and tail lengths, are less than in correspondingly aged controls, thus showing a persistence of the effects of treatment. Costal and epiphyseal cartilages in prenatal rats show cellular, synthetic, and ultrastructural alterations induced by treatment with glucocorticoids but the responses are not necessarily comparable. Except for the low dose of DM, the higher doses of each steroid are more effective in inhibiting, or altering, growth and cellular differentiation in the developing fetuses. Surprisingly, a low dose of DM has a more devastating effect on the cells and extracellular matrix of both costal and epiphyseal cartilage, than do higher dose-levels of the various steroids. Low doses of CA and BM are also effective in inhibiting or altering growth and cellular differentiation, but their effectiveness is largely limited to 17 days of gestation. The order of effect of the various doses of the different steroids on fetal cartilage, listed in decreasing order of severity, is as follows: 0.12 DM, 0.24 DM, 0.42 BM, 50 CA, with 25 CA and 0.18 BM being approximately equal and only slightly different from control cartilages. The effect of prenatal or neonatal glucocorticoid treatment on chondrocytes is minimal in the 30–43 day, or 116–127 day, postnatal groups. In immature and adult rats, cortisone affects the chondrocytes more deleteriously than does betamethasone, and a 5.0 mg dose of CA seems to affect chondrocytes, body weight, and tibial and tail lengths more than 0.2 or 7.5 mg doses.

    Notizen: Summary The ultrastructure of microtriches of the rat tapeworm, Hymenolepis diminuta, was examined with a number of electron-microscopic techniques. Fixatives containing different buffers, non-ionic detergents, chelators, tannic acid and various concentrations of aldehydes were tested for ability to stabilize cytoskeletal components while extracting background material. These methods revealed features unique to these specialized microvilli, and permitted construction of a detailed model of microthrix architecture. The microtriches of H. diminuta are comprised of a microfilament-containing base, a dense cap and a complex junctional region between the base and cap. The microfilaments of the base are contiguous distally with a tubular structure (the junctional tubule) within the junctional region; proximally, the microfilaments end abruptly: a terminal web appears to be absent. A beveled bilayered cylinder of dense material (the core tunic) encircles the microfilamentous core. The core tunics and junctional tubules of the microtriches are specifically and uniformly aligned along the strobila. Microtriches therefore can be distinguished from other microvilli (e.g., those of enterocyte brush borders) by their complex ultrastructure and precise orientation upon the cytoplasmic surface.

    Notizen: Summary Human duodenal endocrine cells reactive with antibodies to cholecystokinin (CCK) 33 (10–20) and/or gastrin 34 (1–15) were studied by a combination of immunohistochemical and electron-microscopic methods. By immunohistochemistry, three types of endocrine cells were distinguished in human duodenal mucosa, i.e., those only positive for only CCK, those positive for both CCK and gastrin and those only positive for only gastrin. Ultrastructurally, the first cell type is characterized by many secretory granules with an eccentric dense core (mean diameter; 271+-74 nm). The second cell type, which was less frequent than the other two, has ultrastructural features that resemble type-I cells. The last cell type was composed of two types of cells containing small secretory granules identical to those of IG cells (mean diameter; 171+-31 nm) or large secretory granules indistinguishable from those of I cells (mean diameter; 286+-50 nm).

    Notizen: Summary The ultimobranchial glands of the chicken were examined by electron microscopy and immunocytochemistry using a calcitonin antiserum. Electron microscopy confirmed the presence of C-cells, containing numerous secretory granules storing calcitonin, in the luminal lining of cyst-like structures found in these glands. These cells were furnished with prominent microvillar projections at their luminal surface, and the cytoplasm of the apical region was filled with fibril material. Furthermore, the cells contained prominent junctional complexes and desmosomes at their apico-lateral surfaces. In these C-cells, secretory granules were concentrated near the lumen and some were attached to the apical cell membrane. The luminal content of the cysts had a colloid-like and flocculent appearance, and was frequently seen attached to the cytoplasmic projections or apical cell membrane of the C-cells. Since the cysts progressively increase in volume and number with age, it is suggested that they may partly play a role in the storage of excess or unneeded hormonal products.

    Notizen: Summary The ultrastructure of Leydig cells in a seasonally breeding rodent, Rattus fuscipes, was studied in the breeding and non-breeding season and compared with Leydig cell morphology after suppression of gonadotrophin secretion induced by hypophysectomy or chronic administration of testosterone. Serum luteinizing hormone (LH) and testosterone (T) were measured and in-vitro T production by testes was assessed by stimulation with human chorionic gonadotrophin (hCG). In non-breeding wild-trapped rats and rats with experimental suppression of gonadotrophins, the Leydig cells were atrophied and exhibited variable amounts of cytoplasmic lipid and crystalloid inclusions, the latter commonly dominating the cytoplasmic area. Compared with fertile rats, serum LH and hCG-stimulated T production of experimentally regressed rats was significantly reduced, confirming structural features indicative of Leydig cell inactivity. Atrophy of Leydig cell nuclei was accompanied by the formation of unusual intranuclear vesicles sometimes containing small crystalloids. Ultrastructural analysis suggested transfer of the vesicles to the cytoplasm where their unification gave rise to much larger crystalloid bodies. Crystalloids occurred when serum LH was depressed and with either full (T treatment) or arrested spermatogenesis (hypophysectomy) suggesting that their formation is governed by pituitary function and is not dependent upon the degree of spermatogenic activity.

    Notizen: Summary The temporal sequence of ultrastructural changes induced in the rat rod photoreceptor by 80 lux light-stress has been studied. The changes seen were compared with those produced by a much dimmer (3 lux) illumination. Some of the early signs of abnormality were (1) degradation of some disk membranes at the tips of outer segments, (2) disaggregation and detachment of ribosomes, (3) lighter matrices in swollen mitochondria, (4) disappearance of the Golgi apparatus, (5) proliferation of autophagic bodies in the inner segments, and (6) appearance of perimitochondrial membrane whorls in the synaptic terminals. No single change could be identified that would inexorably lead to cell death. The overall picture, however, suggested that an inability of the cell to maintain its anabolic balance is responsible for the pyknosis that occurs when the 80 lux exposure exceeds 12–15 h. All changes were reversible when exposure duration did not exceed 12 h, the normal length of the light cycle for these rats.

    Notizen: Summary In numerous investigations using cultured smooth-muscle cells, investigators have consistently added 10–20% fetal calf serum (FCS) to the medium to maintain viable cells. In the present study we utilized an optical technique to investigate whether smooth-muscle cells, cultured with or without FCS, maintain their contractile activity in vitro. With such optical measurement, we were able to detect signals due to spontaneous contractions, in muscle cells cultured in FCS-free medium for up to 8 days, and, for the first time, were also able to observe the conduction of these cell contractions. The ultrastructural characteristics of cultured smooth-muscle cells during contractile activity, were also examined by electron microscopy. The cells were mature and well-differentiated, and were packed with numerous myofilaments. They had developed long cell processes, and were linked to one another by gap junctions. These observations indicated that the smooth-muscle cells, cultured without FCS for 7 to 8 days, were morphologically mature and maintained their contractile activity, whereas the cells cultured in FCS-containing medium showed no detectable signs of contractile activity.

    Notizen: Summary The topography, external structure and ultrastructure of a cephalic sense organ, described for the first time, were studied by light and electron microscopy in the parasitic copepod Pachypygus gibber. This species is unusual in that it has three reproductive sexual forms (two males, one female).The cephalic organ, present only in the atypical male, is made up of numerous functional units, each composed of 4 cells: two sensory cells, one basal enveloping cell and one apical canal-forming cell opening outside via a pore. Many hundred pores are situated within the cuticle of the ventral pleural borders. Through each pore protrude two ciliary endings. An interesting feature is that the ciliary ends are without cuticular cover and thus, directly exposed to the surroundings, a situation unique in arthropods. The structural characteristics of this sense organ and the particular mode of life of the atypical male (with an additional free planktonic phase), lead to the hypothesis that its function is linked to chemical reception in the complex behavioural patterns such as host and sex recognition, during the free life.

    Notizen: Summary The ultrastructure of immunoreactive somatotrophs and lactotrophs in pituitaries of fetal rats at 19, 20 and 21 days of gestation and on the day of birth was studied. Somatotrophs, first detectable at 19 days of gestation, undergo only minor modifications before reaching the structure described for adults. In particular there is an increase in the endoplasmic reticulum and Golgi apparatus. Lactotrophs, first identifiable in newborn rats, are very different in ultrastructure from adult cells, because the secretory granules are generally small, but variable in shape and size, and the Golgi complex is prominent.

    Notizen: Summary Molar tooth germs from three-day-old rats were cultured successfully for fourteen days, permitting the study of the development in vitro of both extracellular matrix and cellular elements such as odontoblasts and ameloblasts. The ultrastructure of the cultured tooth germs was compared with the ultrastructure of tooth germs in vivo at a comparable developmental stage. Progenitor cells of odontoblasts and ameloblasts were found to differentiate in vitro. Odontoblasts seemed to contain more lysosome-like bodies and fewer secretory granules than in vivo. They formed normally mineralizing dentine or a thick layer of dense, unmineralized predentine with incidentally some amorphous, extracellular material. Enamel was exclusively present opposite well developed dentine. It was often hyperor hypomineralized and enamel rods were not as regularly shaped as in vivo. In places where no enamel formation had taken place, large amounts of amorphous extracellular material were sometimes seen. From these observations it can be concluded that cellular development in cultured tooth germs appeared more or less normal, but extracellular matrix formation and mineralization were sometimes disturbed.

    Notizen: Summary Reflecting chromatophores in the dermis of the skin of a freshwater goby, Odontobutis obscura, are of an iridophore type. These chromatophores contain numerous reflecting platelets, which are similar to those in iridophores of other fish and amphibian species. It was found that these iridophores are motile, i.e., these cells respond to certain stimuli with translocation of the platelets within the cells. K+ ions induced dispersion of the platelets in excised scale preparations, but not in excised scales from chemically denervated fish. Norepinephrine and melatonin also induced dispersion of the platelets. Alpha-MSH was effective in aggregating these organelles into the centrospheres of the cells. The conclusions reached are: (1) iridophores of O. obscura are motile; (2) the movement of the iridophores is under nervous and hormonal control.

    Notizen: Summary The cytotoxic effects of ethane dimethanesulphonate upon rat Leydig cells were examined ultrastructurally up to 3 days after treatment and related to changes in serum levels of gonadotrophins and testosterone. Six hours after administration of ethane dimethanesulphonate the usual tubulo-vesicular morphology of Leydig-cell smooth endoplasmic reticulum was converted to small vesicles and the Golgi apparatus showed focal hypertrophy into anastomosing tubules. These changes became more marked by 12 h with many Leydig cells exhibiting karyopyknosis and hyperchromatism. Necrotic Leydig cells were often engulfed by macrophages, the latter containing pyknotic fragments of Leydig cells within their cytoplasm. One day after administration, advanced necrosis of Leydig cells occurred, many of which were phagocytosed by macrophages, and on day 3, destruction of Leydig cells was complete resulting in their elimination from the interstitial tissue, which contained only loose connective tissue and macrophages. Structural alterations to the Leydig cells from 6–24 h was reflected by a significant reduction in serum testosterone levels which further declined to the limits of detection accompanying the abolition of Leydig cells on day 3. These changes were paralleled by a significant elevation of serum LH and FSH levels suggesting diminished feedback regulation of pituitary gonadotrophin secretion. The results indicate that ethane dimethanesulphonate is a rapidly acting Leydig cell toxin which may be a useful experimental tool in further studies of spermatogenic function mediated via Sertoli cell-Leydig cell interaction.

    Notizen: Summary A new endothelial cell structure, named the endothelial pocket, has been found by combined transmission and scanning electron microscopic studies of renal peritubular capillaries. Transmission EM observations made on these and other fenestrated capillaries demonstrated that each pocket consists of an attenuated fold of fenestrated endothelium that projects 200 nm into the lumen above the rest of the endothelial surface. Beneath this luminal fold, there is a space and then another layer of fenestrated endothelium which abuts the basal lamina. The linear density of endothelial pockets was measured in the capillaries of the kidney cortex, intestinal mucosa and exocrine pancreas in mice and determined to be 0.067, 0.017 and 0.007 pockets·μm-1 respectively. Cationic ferritin decoration of the anionic sites on the luminal surface of the endothelium in these capillary beds revealed that both unlabelled and labelled diaphragms are clustered. In such specimens, the majority of the luminal diaphragms on endothelial pockets did not have cationic ferritin binding sites detectable by either scanning or transmission EM. On this account as well as on account of their general morphology, endothelial pockets appear to be multifold versions of the simple transendothelial channel.

    Notizen: Summary Light- and urethane-induced retinopathies in rats are characterized by loss of photoreceptors. Retinal capillaries subsequently become incorporated into the normally avascular retinal pigment epithelium. These models provided an opportunity to study the response of epithelial cells to closely apposed capillaries, in order to determine if capillaries contribute to the polar organization of epithelial cells. Pigment epithelial cells reorganized their lateral plasma membrane where the latter faced intraepithelial capillaries. This normally flat, undifferentiated membrane developed attachment sites, folds and intracytoplasmic tubules, and exhibited endocytosis and putative basal lamina secretion. These structural and functional specializations are normally restricted to the basal plasma membrane — the normal vascular front of the cell facing the dense meshwork of capillaries constituting the choriocapillaris. We conclude that RPE cells, and perhaps epithelia in general, polarize in response to an adjacent capillary bed.

    Notizen: Summary VIP-like immunoreactivity was revealed in a few chromaffin cells, medullary ganglion cells and a plexus of varicose nerve fibers in the superficial cortex and single varicose fibers in the juxtamedullary cortex and the medulla of the rat adrenal gland. VIP-like immunoreactive chromaffin cells were polygonal in shape without any distinct cytoplasmic processes and they appeared solitarily. Their cytoplasm contained abundant granular vesicles having a round core and the immunoreactive material was localized to the granular core. VIP-immunoreactive ganglion cells were multipolar and had large intracytoplasmic vacuoles. The immunoreactive material was localized not only in a few granular vesicles but also diffusely throughout the axoplasm. VIP-immunoreactive varicose nerve fibers in the superficial cortex were characterized by abundant small clear vesicles and some large granular vesicles, while those in the juxtamedullary cortex and medulla and the ganglionic processes were characterized by abundant large clear vesicles, as well as the same vesicular elements as contained in the nerves in the superficial cortex. The immunoreactive material was localized on the granular cores and diffusely in the axoplasm in both nerves. Based on the similarity and difference in the composition of the vesicles contained in individual nerves, it is likely that the VIP-immunoreactive nerve fibers in the medulla and the juxtamedullary cortex are derived from the medullary VIP-ganglion cells, while those in the superficial cortex are of extrinsic origin. The immunoreactive nerve fibers in both the cortex and the medulla were often in direct contact with cortical cells and chromaffin cells, where no membrane specializations were formed. The immunoreactive nerve fibers were sometimes associated with the smooth muscle cells and pericytes of small blood vessels in the superficial cortex. In addition they were often seen in close apposition to the fenestrated endothelial cells in the cortex and the medulla, only a common basal lamina intervening. Several possible mechanisms by which VIP may exert its effect in the adrenal gland are discussed.

    Notizen: Summary Of all avian species the emperor penguin is the best adapted bird to attain the greatest diving depths and diving durations. Therefore the lung of this bird was investigated with electron-microscopic, i.e., freeze-fracture and thin-section methods. The parabronchi are surrounded by bundles of smooth muscle cells innervated by varicosities of autonomic nerves. The parabronchial epithelium is flat, bears a few microvilli and does not show any conspicuous ultrastructural specializations; only individual cells contain secretory granules. The atrial epithelial cells bear apical microvilli and are interconnected by adhering and tight junctions (5–10 sealing strands), the latter presumably forming an effective barrier against paracellular fluid movements. The cells contain lamellar inclusions of two types: (i) round membrane-bounded granules, the lamellar content of which is fixation-labile, and (ii) large polymorphic compact deposits of well-preserved lamellae. In both types of inclusions the individual lamellae can be of trilaminar appearance, whereas their fracture faces are smooth. Lamellar material also covers the epithelium of atria, infundibula and air capillaries. In thin areas the diameter of the morphological blood-air barrier measures 220–330 nm. Usually the endothelium of the blood capillaries is thicker (40–180 nm) than the air capillary epithelium (25–150 nm). Both epithelium and endothelium are interconnected by tight junctions, which seem to be more extensive and presumably tighter in the epithelium than in the endothelium. Frequently the common basal lamina is the thickest individual component of the blood-air barrier, measuring between 170–230 nm. Often collagen fibrils occur in this area of the barrier. In comparison with that of other birds the entire blood-air barrier of the emperor penguin is relatively thick, probably owing to an adaptation of the lung tissue which must resist high hydrostatic pressure during diving excursions.

    Notizen: Summary Electron-microscopic immunocytochemistry for the demonstration of parathormone in parathyroid chief cells was performed in adult male rats, gerbils, mice, and dogs, using the protein A-gold technique. Protein A-gold particles were detected over both large and small secretory granules in all the animals examined. In the former, they were concentrated not only over type-I granules with a large core, but also over type-II granules with a small core. They were also located over atypical granules, including heterogeneously dense granules, granules having vesicles in a finely particulate core, and distorted granules. All labelled secretory granules were characterized by the presence of a clear halo of varying width around the core. Occasionally, Golgi cisternae as well as Golgi vacuoles with a finely particular content were also labelled. The labelling of the secretory granules was strong in dogs, moderate in rats and gerbils, and weak in mice. In addition, it was more intense in the non-osmicated preparations than in the osmicated preparations. The frequency of both types of large granules showed species differences. The possible factors involved in these differences are discussed.

    Notizen: Summary Peripolar cells are granulated glomerular epithelial cells that form a cuff around the vascular pole of the glomerulus. Quantitation of these cells in 17 species of mammals (including man, several laboratory animals and a variety of other species) indicated that they were detectable by light microscopy in all but one of the mammals that were examined (the Australian hopping mouse). In adult mammals with detectable peripolar cells, the “peripolar cell index” (the percentage of randomly sectioned glomeruli that displayed peripolar cells in histological sections of kidney) ranged from 0.15 (for echidna) to 11.86 (for sheep). Newborn lambs and rats showed strikingly high values (23.30 and 10.76, respectively) compared with their adult counterparts. Using electron microscopy, peripolar cells were observed in all species that were examined, including the Australian hopping mouse. Morphologically, peripolar cells were similar in all species although their size and granule population varied. They showed a predominantly outer cortical glomerular distribution and a close anatomical relationship with the renin-containing myoepithelioid cells. These findings indicate that peripolar cells are present in a wide variety of species and support the view that such cells may play a significant role in the regulation of normal renal function.

    Notizen: Summary The ultrastructure of the endometrial epithelium of the pig was studied during the estrous cycle and early pregnancy up to implantation. Special attention was given to the luminal epithelium and morphological indications of protein synthesis. Although the general morphology of the luminal and glandular epithelia is similar (both tissues consist of secretory cells and ciliated cells at all the stages studied), it appears that the two epithelia should be considered as two functionally different units in the pre-implantation period. Morphological evidence suggests the presence of at least three different secretory products within luminal epithelial cells; they are released at different times, i.e. at estrus, between day 8 and 10 and after day 11. The glandular epithelium shows release of secretory products from day 10–11. Increasing amounts of glycogen were found within epithelial cells, especially in pregnant gilts from day 12. The possible significance of secretory activity of the epithelium is discussed in relation to the development of the embryos.

    Notizen: Summary We have continued the transcriptional analysis of the region of cytological locus 67B that contains the four small heat shock genes and other genes. Transcription from one of the heat shock genes in the region, hsp 26, takes place during high temperature treatment and at certain developmental stages, without heat shock, in several tissues, such as imaginal discs and adult ovaries. Observations of unexpected products after nuclease protection experiments periments provided the first indication of what genomic blot experiments showed to be small deletions. The alleles containing the deletion are expressed at the same level as the wild type allele. The deletion shortens the protein product, implying that it is in the coding region. Furthermore, flies homozygous for one of the deletion alleles are viable.

    Notizen: Summary The female-sterile mutants fs(1) 1163 of Drosophila melanogaster described by Gans et al. (1975) has been characterised as a yolk protein 1 (YP1) secretion mutant (Bownes and Hames 1978b; Bownes and Hodson 1980). We have cloned and sequenced the YP1 gene from this strain, and the strain in which the mutant was induced. One amino acid substitution was found in the predicted polypeptide sequence, an isoleucine to asparagine change at position 92. The sequence of the leader peptide was identical to previously published YP1 sequences. The possible effects of the amino acid change were investigated by computer analysis, which suggests there is no major alteration of secondary structure, but that a hydrophobic region in YP1 is lost in the mutant. This may affect higher order structure.

    Notizen: Summary Mitosis in the marine red algaLomentaria baileyana (Rhodymeniales, Rhodophyta) was studied with the electron microscope. Nucleus associated organelles known as polar rings (PRs) migrate to establish the division poles at prophase. At prometaphase, shallow invaginations in the nuclear envelope (NE) form on two sides of each PR and soon rupture. The gaps that are consequently formed contain several small fragments of NE. A larger region of NE remains intact between the two gaps. By metaphase several cisternae of perinuclear endoplasmic reticulum (PER) have enclosed most of the nucleus but remain absent from the polar regions. The nucleolus disperses partially and a typical metaphase plate of chromosomes is formed. Each PR has disjoined into separate proximal and distal portions. MTs converge widely on all regions of the polar area, but do not extend into the cytoplasm. Some MTs end near or at the chromosomes while others extend slightly farther past the chromosomes or diagonally to the NE. As chromosomes move to opposite poles at anaphase, they are accompanied by nucleolar material. An interzonal midpiece (IZM) is created as the pole to pole distance increases and the NE remains intact except for the polar gaps. Following detachment from the IZM, the daughter nuclei are separated by a large central vacuole as a cleavage furrow develops and eventually constricts to form two cells following pit connection formation. It is suggested that mitosis inLomentaria represents an evolutionary intermediate between that seen in the higher and lower groups of red algae. This conclusion is in agreement with conventional morphological and light microscopic criteria used to placeLomentaria in theRhodymeniales, which is considered to be the next to most advanced order in theRhodophyta.

    Notizen: Summary We analyzed transformation, recrystallization, splitting and dispersion of prolamellar bodies during chloroplast development in pea seedlings illuminated by white, red and blue light of low intensity. With the help of a stereometric method we determined that there was a significant increase of prolamellar body number and a sharp decrease of their volume in differentiating chloroplast even in the first 2 hours of illumination. Decrease of prolamellar body dimensions was due both to gradual dispersion of its elements into primary thylakoids (indicated by the decrease of total volume of prolamellar bodies in plastid) and to splitting of prolamellar bodies (indicated by the increase of number of promellar bodies in plastid). Red light was more effective in transformation, splitting and dispersion of prolamellar bodies than blue light during the first 8–12 hours. Longer treatment with blue light had a stronger influence on these processes and on complete recrystallization than other light treatments.

    Notizen: Summary Ultrastructural observations on the principal endomembranes (endoplasmic reticulum, Golgi apparatus) of synchronously growing of wild type and mutant (CW 2, CW 15) strains ofChlamydomonas reinhardii have been carried out. The dictyosomes of the Golgi apparatus in all three cases are highly polar in morphology but lack intercisternal filaments. A clear spatial relationship between dictyosomes and endoplasmic reticulum is seen and a transfer of vesicles from the latter to the former is easily visualized. Coated vesicles invariably appear to be restricted to the trans-pole of the dictyosomes. The endoplasmic reticulum adjacent to the cis pole of dictyosomes is considerably hypertrophied in the case of the wild type, only partially so in the mutant CW 2 but not at all in the mutant CW 15. In the wild type this swelling is most extreme during the period of wall deposition and for several hours afterwards. The results are discussed in relation to the biosynthesis and intracellular transport of, particularly O-glycosidically linked, glycoproteins.

    Notizen: Summary Seasonal changes in the mitochondrial ultrastructure were examined in palisade parenchyma cells of a tuft-formingDiapensia lapponica L. collected at monthly intervals in Northern Finland. Quantitative analyses to measure volume and surface densities were conducted during different periods of growth (stages of growth, acclimation, winter period and deacclimation) in the annual cycle. The volume density was highest in the summer and lowest in the spring; the difference was significant with both fixatives used GA and GA/FA. The largest membrane area (the mitochondrial outer membrane and the cristal membranes together) was observed in the summer and autumn, and was significantly less in the winter and spring. This correlated with fewer mitochondria in the spring and a smaller number of cristae in the winter and spring. In the material fixed in GA/FA the distribution of length/width ratios of mitochondria was relatively uniform in all seasons. However, the mitochondrial ultrastructure had the most varied appearance during the winter. Hypertonie GA/FA solution did not cause significant differences either in the ultrastructure or the volume and surface densities of the mitochondria.

During germination these organelles migrate into a narrow germ tube which subsequently expands to form the gun cell initial. The extracellular components of the attack apparatus (i.e. missile and injection tube) are formed entirely in the developing gun cell; indirect evidence suggests that both the Golgi complex and r-ER are involved in their synthesis. The intra-cellular component of the attack apparatus comprises the posterior, anterior and apical vacuoles. The posterior vacuole forms by fusion and expansion of the original cyst provacuoles; the formation of the anterior and apical vacuoles occurs late in gun cell differentiation and involves fusion of Golgi-derived vesicles.

The purity of the isolated cell walls was judged biochemically by the lack of activity of cytoplasmic marker enzymes and morphologically by examination at both the light and electron microscope levels. The purified cell walls were extracted with various salt treatments and the molecular weight range of most of the extracted proteins was between 14 and 31 kDa. The salt extracted hydrolytic enzymes were basic in nature (pI〉7.0) compared to their cytosolic counterparts (pI〈7.0). Some enzymes were readily extracted from cell walls (β-glucosidase and β-NAcglucosaminidase) with high salt treatment while most of the α-mannosidase activity associated with purified cell walls could not be removed even with sequential high salt treatments.

    Notizen: Summary Populations ofPhytophthora palmivora zoospores induced to undergo synchronous differentiation by the addition of pectin showed an ordered sequence of surface changes. Within 10 seconds “blebs” and “microvilli” appeared on the cell and flagellar surfaces. These projections gave the zoospore a highly irregular and even convoluted surface for up to 30 seconds poststimulation. Between 30 and 40 seconds, pits appeared on the surface, which began to assume a smoother texture. Zoospores then rounded up and by 4 minutes had assumed an almost spherical form. At this time the surface became wrinkled as though shrinkage had occurred. Germ tubes could be seen emerging from almost all cells by 40 minutes, and as germ tubes enlarged, the cell surface became smooth again but with a fibrous texture. Many zoospores retained their flagella through to the germling stage. When cells were stimulated in iso-osmotic rather than hypo-osmotic media, germ tubes in most cases emerged at or very near the site of flagellar attachment. Differentiation in iso-osmotic media showed the same sequence except that the cells did not assume the spherical shape normally characteristic of cysts and germlings differentiating under hypo-osmotic conditions. Strontium-induced differentiation again followed the same general sequence as desribed above, although the cell surface was noticeably more convoluted and irregular prior to the stage of the formation of pits, and flagella were always shed between 40 seconds and 1 minute. Calcium addition up to 20 seconds after the initiation of differentiation with pectin prevented a high proportion of zoospores from encysting, the cells remained motile, and although the frequency of surface blebs and microvilli increased, pits did not appear. These surface changes are consistent with those expected during stimulus-mediated secretion, and it appears that calcium is able to inhibit differentiation only before the secretory step (pit formation).

    Notizen: Summary Root-nodules of soybean plants dark-stressed for 8 days and then allowed to recover for up to 17 days were examined by transmission electron microscopy. Control nodules possessed all the ultrastructural features characteristic of infected and uninfected nodule cells. Minimal changes in the appearance of host cells and bacteroids occurred during the first four days of dark stress. After 8 days of dark stress, damage was observed in the cellular and organelle membranes; however, very few changes were observed in the bacteroids. Nodule structure continued to degrade during the first two days of recovery after which time nodules either recovered or completely degraded. In the former case, structural integrity returned to all nodule cells. In the latter case all structural integrity of the host cell disappeared; however, bacteroids appeared intact suggesting that they remained viable.

    Notizen: Summary The effect of 2,6-dichlorobenzonitrile on cytokinesis of meristematic cells of onion root during both treatment and recovery has been studied by electron microscopic techniques. 2,6-dichlorobenzonitrile interferes with cell plate formation in such a way that Golgi apparatus vesicles of treated cells appear to be different than controls and seem to coalesce as anomalous partial cell plates. During recovery, an apparently normal progression of cytokinesis is observed and abnormal portions of the cell plate are retained. Nuclear constrictions are observed frequently during recovery as a result of temporal alterations in cytokinesis. Our results show that 2,6-dichlorobenzo-nitrile induces anomalous and/or incomplete cell plates, which might be caused by an altered function of Golgi apparatus.

    Notizen: Abstract The hypothesis that negative assortative mating occurs as a mechanism limiting inbreeding between genetically related individuals ofDrosophila melanogaster was tested. In order to avoid bias linked to using inbred lines, experiments made use of the F1 hybrid progeny between lines rendered homozygous on chromosomes 1, 2, and 3. No negative assortative mating was found, but significant additive variation was observed between lines for orientation, vibration, copulation latencies, and copulation duration. There was no consistency of results, either among parameters or between sexes from the same line. It is therefore unlikely that the variations observed are due merely to quantitative differences in “vigor”. Since all lines originated from the same wild population, these differences are a possible estimate of natural variation in sexual behavior.

    Notizen: Abstract A comparison of pupation height in light and dark was made using 12 species ofDrosophila, representing four species groups and four different ecological backgrounds (temperate-montane forest,virilis group desert,replate group; cosmopolitanmelanogaster group; tropical forest,willistoni group). Light condition has a significant effect on pupation height in only two of the species. In the light,D. montana stays close to the food surface, whileD. melanogaster pupates higher in light than in dark. Light-dependent patterns of pupation response do not correspond to those previously reported for the light-dependent mating response. Considerable interspecific variation exists for pupation height in each species triad, some of which could provide a basis for larval niche separation. Patterns of species differences in the desertrepleta triad are the same in light and in darkness.

    Notizen: Summary The yolk proteins stored in Drosophila, oocytes for utilisation during embryogenesis are an ideal system for studying the regulation of gene expression during development. The 3 major polypeptides found in yolk in D. melanogaster are synthesised in the fat body and ovarian follicle cells and selectively accumulated by the oocyte during vitellogenesis. In order to understand more about their regulation and the mechanism of uptake, studies on other species are necessary. Three yolk polypeptides have previously been identified in the D. melanogaster sibling species (D. melanogaster, D. simulans, D. mauritiana, D. erecta, D. teissieri, D. orena and D. yakuba). In D. melanogaster three genes located on the X chromosome are known to code for these yolk polypeptides. in this study genomic Southern transfers and in situ hybridisation experiments were carried out on the sibling species. Using the three cloned yolk protein genes from D. melanogaster, homologous sequences could be detected in the sibling species. It is suggested that three yolk protein genes occur in each of these species, all being located on the X chromosome, and that two of the genes are very closely linked in these same species. Yolk protein gene-homologous DNA sequences have also been identified in two more distantly related species D. funebris and D. virilis.

    Notizen: Summary T(Y;2) translocations were used to cytologically localise the wingless locus of Drosophila melanogaster. We found that an existing T(Y;2), which is an insertion of a segment of 2L into the Y chromosome, has wg + within this insert. This Y chromosome was used to generate an attached XY chromosome containing wg +. The mutation claret-nondisjunctional (ca nd) was used to induce the loss of this XY chromosome and thus generate gynandromorphs with wg 1/wg 1 male tissue and wg +/wg 1/wg 1 female tissue. Analysis of these gynanders demonstrated that a genotypically wingless mutant hemithorax is usually also phenotypically mutant in these half body mosaics; thus wg 1 is discautonomous. This observation is of interest as it is known that wg is not cell autonomous.

    Notizen: Summary In order to correlate the synthesis of a previously described set of follicel cell (Fc) proteins with a known mutation that affects female fertility, three female sterile mutations, fs(1)384, fs(1)508 and fs(1)1501, mapping in the same region as the Fc locus (7C1-9), were analysed with respect to Fc synthesis. The fs(1)508 strain displayed a normal Fc protein pattern, while in fs(1)384 no Fc protein synthesis could be detected. The fs(1)1501 pattern of Fc polypeptide synthesis was totally different from that of any previously analysed strain, displaying a set of proteins that were much larger than the standard Fc variant form. Two of the female sterile mutations, fs(1)384 and fs(1)1501, were combined in rans with two wild-type strains displaying two different electrophoretic variant forms of the Fc proteins. The combinations were then analysed for Fc protein synthesis, using the fact that females heterozygous for two of the Fc variant forms display both parental forms. The results indicate that the fs(1)384 mutation is directly involved in the synthesis of the Fc proteins, as the trans heterozygotes only synthesize the Fc form derived from the wild-type parent. We also suggest that the large proteins synthesized by the fs(1)1501 mutant are a defective Fc variant form. The nature of the two mutations is also discussed.

    Notizen: Summary An attempt was made to produce doubled haploids on 16 winter wheat and six spring and winter triticale genotypes thought to carry genes for interspecific incompatibility. The potential for haploid production was maximized by the use of Hordeum bulbosum genotypes selected for high crossability on crossable wheat genotypes, the use of two post-pollination applications of gibberellic acid and by the pollination of immature florets. A low frequency of seed was set on both the wheat and the triticale genotypes, having mean seed sets of 0.20 per cent and 0.27 per cent respectively. Although the frequency of embryos (‘seed quality’) was high, doubled haploid production was further limited by poor embryo differentiation and regeneration. Haploid plantlets were obtained from the wheat cultivars Moulin and Renard, although successful chromosome doubling and doubled haploid production was achieved in Moulin only.

    Notizen: Summary Two transformations commonly used in the analysis of multilocation variety trials are the logarithmic transformation and the site mean transformation, in which each observation at a site is divided by the site mean yield prior to analysis. Expressions are derived for differences between the yields of varieties after each of these transformations, in terms of the original yields. Similar expressions are also determined for the regression mean method of estimation, which is based on linear regressions of variety means on site means. It is shown that the two transformations produce almost identical analyses of variance, and that relative yields are very similar on the three scales of measurement provided that the harmonic mean of site means is used as the predictor value in the regression method.

    Notizen: Summary During test baking, the wheat line QT2870, bred from a rye-derived parent, Kavkaz, and having the pedigree Kavkaz/Timgalen//3*Oxley, showed excessive dough stickiness when slightly overmixed. Conventional quality tests on QT2870 and three commercial varieties did not show any major differences which could explain this dough stickiness. However, resistogram data for QT2870 were considerably different from those for the other cultivars. The curves for QT2870 had lower breaking points and sharper curve angles, indicating that it had a lower tolerance to high speed mixing than the other varieties. Dough stickiness and a lack of mixing tolerance are likely to be major problems limiting the use of rye-derived parents in Australian bread wheat breeding programmes.

    Notizen: Summary Twelve spring wheat cultivars were grown as isolated plants in the field and their pre-anthesis growth was interpreted in terms of plant growth analysis. Relative growth rate (RGR) decreased steadily with time due to a decline of leaf area per unit plant weight (LAR), which could be explained by the reduction of the portion of leaf weight in total plant weight (LWR). Growth per unit leaf area (NAR) and leaf area per unit leaf weight (SLA) changed only little with time. Differences between cultivars for NAR and LAR were of similar magnitude: both 8% when measured by the genetic coefficient of variation. Because both quantities were negatively correlated, the genetic variation of RGR was only 5%. Genetic variation for LWR and SLA were also of similar size, both about 4%. Estimates of genetic variances and covariances based on cultivar means appeared to be biased strongly when the error variation of the means was neglected. Special attention is paid to the methodology of plant growth analysis.

    Notizen: Summary Seven genes, viz. Sr5, Sr6, Sr7a, Sr8a, Sr9b, Sr12 and Sr17 were associated with seedling resistance to Puccinia graminis tritici in Kenya Plume wheat. The predominant field cultures were avirulent on seedlings with Sr7a, but possessed virulence for the other six genes. However, Sr7a did not confer adult-plant resistance when present on its own. Adult-plant resistance was attributed to Sr2 and possibly also to the interaction of Sr7a and Sr12. Two genes, Lr13 and Lr14a, were identified in seedling tests with various cultures of Puccinia recondita tritici. Lr13 conferred adult-plant resistance to the predominant field strains. Genetic recombination between Lr13 and Sr9b was estimated at 17.6±3.1%.

    Notizen: Summary Seedling and field reactions to four European races of stripe rust were determined for 254 wheat cultivars, mostly from Australia and New Zealand. Numerical analyses of the data employed a divisive classification procedure with termal reallocation to form 10 groups which were then further classified, ordinated and diagnosed by several procedures. The groups formed ranged from one comprising four wheats with low seedling reactions to all four rust strains and mean field rust of only 1.0%, to one comprising 105 wheats with high seedling reactions and mean field rust of 64.7%. Groups of cultivars with intermediate levels of rust resistance were classified both on relative level of overall rust and on differential reactions to rust strains. One group of 13 cultivars had moderately high seedling reactions but averaged only 1.5% rust in the field. Seedling response to gibberellic acid (GA) was also measured and diagnosed as an external attribute, along with grain type and region of origin. Principal co-ordinate analysis revealed that greater resistance to stripe rust was associated with the insensitivity to GA typical of semidwarf wheats and with an origin in northern Australia, where semidwarf wheats have been most used in the breeding program. In terms of numerical analysis of disease data in plant breeding programs, the study highlighted the advantages of using divisive classification with terminal reallocation of group members.

    Notizen: Summary Genetic studies were conducted to determine the inheritance of biotype E greenbug resistance in CI 17882 (CI 15092/T. speltoides//Fletcher/3/4* Centurk), a wheat germplasm line previously released as resistant to wheat streak mosaic virus (WSMV). In addition, the association of greenbug and WSMV resistance in CI 17882 was examined. Results indicated that biotype E greenbug resistance in CI 17882 is conditioned by a single dominant gene that is not linked with the WSMV resistance gene.

    Notizen: Summary Because copper is extremely important to the development of normal polllen, an attempt was made to induce male sterility in wheat by applying specific copper-binding ligands to wheat plants. Four different chelates were used at two rates in three methods of application. All four chelates, cupferron, neocuproine, benzotriazole and cuprizone, reduced grain yield at high concentration applied to the soil at sowing but benzotriazole was most effective, even when applied at late tillering to either soil or foliage, and it also reduced yield to a lesser extent when applied at low concentration. At high concentration of benzotriazole (50 mg kg-1 of dry soil) the percentage of pollen staining with I2/KI was very low (0–7%) depending on method of chelate application), and this soil treatment resulted in complete male sterility. The appearance of the pollen, anthers, grain, ears and leaves in many cases mimicked that of normal copper deficiency, and also that caused by other recognised gametocides. These results raise the question of whether binding of copper or some other disturbance of copper metabolism may be the mechanism by which andro-gametocidal chemicals work and if so, dictate a theoretical basis for selecting such chemicals for testing.

The standard formula for the expected gain from selection is based on the assumption that the character value is the sum of two independent normal variables, one genetic and the other environmental. If the genetic variable is normal, but the environmental variable has a positively skewed distribution, then upwards selection will give a lower gain than that expected if both distributions are normal. If the distribution of the environmental variable is negatively skewed, then upwards selection will give a larger than expected gain. Of course, with downwards selection, the results will be reversed. This leads to asymmetry of response if both upwards and downwards selection are used. In cases where unexpected responses to selection are obtained and normality of the character distribution has been assumed, skewness of the data should be checked as possible cause.

    Notizen: Summary Ten to 20 spring wheat (Triticum aestivum L.) cultivars of Israeli origin were grown in three winter (normal) and two summer (abnormal) growing seasons. During the period of emergence to anthesis mean daily temperature was on the average 12°C higher and photoperiod was about 3 h longer in the summer than in the winter. Data was collected on the durations of the periods from emergence to double-ridge (GS1), double ridge to anthesis (GS2) and anthesis to grain maturation (GS3), as well as on yield and yield components. The duration of all developmental stages was reduced by high temperature. While the duration of GS2 was the most thermo-sensitive, it may also have been reduced by the longer summer photoperiod. The effect of photoperiod on GS2 could not be isolated, but the results were interpreted to show that the effect of photoperiod on the duration of GS2 was relatively small. The most heat-affected yield component was number of grains per spikelet and the least affected component was the number of spikes per plant. High temperature reduced grain weight via reduced grain growth duration and not grain growth rate. A general linear regression model of yield on its components revealed that while variation for number of spikes per plant had the greatest effect on yield variation among cultivars in the winter, variation for number of grains per spikelet and spikelets per spike were by far the most important in the summer. Grain weight was the least important component, in this respect, in all seasons. Varieties which sustained the highest yield in hot environments were able to maintain the longest duration of GS2 and the highest number of grain per spike.

    Notizen: Abstract Seven specimens of Rhynchodemus sylvaticus (Leidy) collected from a variety of localities in the US and having variously developed copulatory organs are believed to represent stages in the development of the copulatory apparatus. Four specimens were juveniles with under-developed male components, one specimen had a well-developed female atrium and small male component, and two specimens were mature with a male organ twice the size of the female part. In early stages, the male component had sperm ducts, seminal vesicle, and narrow atrium; more mature stages had a considerable elongated atrium with thick folds in its muscularized wall, a massive muscular bulbus; and a sigmoid ejaculatory duct opening into the large bulbar cavity. Morphological features of mature male copulatory organs in all species of the genus Rhynchodemus are basically similar whereas external body features (color and number of dorsal stripes) of these same species differ.

    Notizen: Abstract The ultrastructure of male copulatory organs having a stylet has been studied in some genera of the Proseriata. Within the Monocelididae there was a variety of stylet-like hard structures. The stylet in Monocelis fusca was a differentiation of the basement membrane of the epithelium lining a penis-like muscular papilla. The penis papilla in Ectocotyla consisted of circular muscles surrounded by a thickened basement membrane and an epithelium. Archilopsis sp. and Archilina sp. with a duplex copulatory bulb, had a stylet within a spiny cirrus. The stylet in Archilopsis sp. was a cylindrical muscular protrusion with a thickened basement lamina that lined the cirrus lumen. The stylet structure in Archilina sp. was composed of four long spines which were derivatives of the basement membrane. In Ectocotyla multitesticulata and Dupliminona corsicana, the accessory prostatoid organ was provided with a hook-shaped stylet that was differentiated in the basement membrane and of which the material was continuous with the fibrous matrix between the muscles of the prostatic bulb. The stylet and needles in the Archimonocelis species were intracellular differentiations. The copulatory organ in Carenscoilia biforamen consisted of a tubiform stylet and four needles, all of which were also intracellular specializations. I consider copulatory hard structures in the Turbellaria to be taxonomically significant in terms of structure, differentiation, and location (whether subcellular, in the basement membrane, or intracellular).

    Notizen: Abstract Using transmission electron microscopy and serial sections with light-microscopic autoradiography, I have investigated the ultrastructure of wound healing, the distribution of cells preparing for proliferation, and the fates of cells labelled with exogenous tritiated thymidine ([3H]T) in Microstomum lineare undergoing wound healing and regeneration. Immediately after decapitation the open wound was reduced to a minimum by strong contraction of circular muscle fibers. The wound epidermis was cellular, consisting of thin parts of epidermal cells from the epidermis around the wound. These epidermal cells maintained close adhesive contact with one another through zonulae adherentes and septate junctions. No proliferating cells were found in the old epidermis. The only cells taking up [3H]T were mesenchymal and gastrodermal neoblasts which proliferated and migrated towards the surface. The final epidermis was formed by conjunction of the wound epidermis and newly differentiated epidermal cells. Regeneration in Microstomum, in contrast to that of planarians, occurs mainly by morphallaxis, without the formation of a regeneration blastema, but also through continuous cell proliferation, migration, and differentiation.

    Notizen: Abstract Autoradiography has been applied to two acoel turbellarians, Convoluta convoluta and Oxyposthia praedator, to determine the distribution and fate of proliferative cells. In C. convoluta, mitotic figures and nuclei that labelled with [3H]thymidine could be observed in the peripheral parenchyma but not in the middle zone of the central parenchyma. The time required for regeneration of physiologically competent digestive cells was about 10–15 days. In O. praedator, mitotic figures (in metaphase and telophase) were observed in the peripheral parenchyma while none were found in the epidermis either in untreated animals or after treatment with colchicine. Mitotic figures were found only rarely in the central parenchyma and only in its marginal zone. Autoradiographs of O. praedator demonstrated [3H]thymidine incorporation into both the nuclei and the cytoplasm of peripheral parenchymal cells. In the central parenchyma, no nuclei with primary labelling were observed. The digestive parenchyma of the acoels is regarded as a unique histological system involving both specialized cells of the central parenchyma and stem cells located in the peripheral parenchyma.

    Notizen: Abstract The planarian Dugesia lugubris is a balanced hermaphrodite, meaning that male genetic factors are in equilibrium with female factors. Differentiation of the gonads is controlled by the region in which they develop. According to the classical theory of germ cell formation, these cells stem from neoblasts that are induced to differentiate by factors specific to the gonadal regions, factors presumably due to gradients formed by neurosecretory activity of the cephalic ganglia and longitudinal nerve cords. A more recently proposed theory holds that germ cells in regenerates originate not from neoblasts but from dedifferentiated cells and that characteristics of the gonadal regions are determined by direct interactions of cells here. Results of our experiments with homo- and autoplastic grafst support the classical theory. Prepharyngeal portions grafted onto posterior body portions retained their ability to maintain or induce development of ovaries. Postpharyngeal portions grafted onto anterior portions produced only testes even though the brain developed normally in these regenerates. Under these experimental conditions, gonad regeneration took longer than it does in normal regeneration (i.e., that in which body regions are not displaced).

    Notizen: Abstract The earliest detectable change during regeneration of the gastrodermis in Dugesia japonica was an aggregation of regenerative cells underneath the gastrodermis remaining at the wound margin. The gastrodermal cells in experimental regenerates retained some of their original characters and presented no indication of cell dedifferentiation. The regenerative cells came into contact with the basal surface of gastrodermal cells, forming stratified cell layers. Differentiation of these cells into gastrodermal cells was initiated by the development of synthetic organelles within their cytoplasm. These differentiating cells gave rise to two different types of gastrodermal cells, namely phagocytic cells and sphere cells. In later stages, there was an apparent movement of differentiated gastrodermal cells towards the parenchyma.

    Notizen: Abstract The ontogenetic changes in the distribution, abundance, and size of rhabdoids were examined in an undisturbed laboratory population of Dugesia polychroa. Irrespective of triclad age, rhabdoids in the epidermis and parenchyma were more abundant on the dorsal than the ventral side of the body. No significant differences were found in the abundance of epidermal or parenchymal rhabdoids among the anterior, medial, and posterior regions of the body. Rhabdoid number and size changed significantly with triclad age, with a marked depression coinciding with the onset of cocoon production. Rhabdoid discharge was correlated with physical and/or physiological disturbance and occurred in the absence of any overt environmental disturbance. Simple allometric relationships were observed between rhabdoid size and number on one hand and body plan area on the other. Different allometric trends were observed from field-collected individuals compared to the undisturbed laboratory population. The potential function of rhabdoids in the Tricladida is discussed in light of these findings.

    Notizen: Abstract The tolerance of adult specimens of Dugesia lugubris and D. polychroa for 13 different chlorinities ranging from 15.0–3.8‰ and for two temperatures, viz. 4 and 23 °C, was tested. At chlorinities of 7.5‰ and lower, the survival time of both species was considerably longer than at higher chlorinities (a few hours at 7.5‰, one to several days at 6.6‰ and lower concentrations). It is assumed that this is determined by the osmoregulatory capacity of the planarians. It was found that at low chlorinities combined with a high temperature D. polychroa survived longer than D. lugubris, while at the same chlorinities the opposite was true for a low temperature. The effect of temperature on survival at low chlorinities was more drastic for D. lugubris than for D. polychroa. The results correlate with data on the distribution of both species in The Netherlands. Outside areas with an average chlorinity below 2‰ the two species were rarely found.

    Notizen: Abstract The most profound structural variety in morphofunctional systems and morphogenetic mechanisms, i.e. the highest morphological diversity, is observed in those groups where these systems and mechanisms are evolutionarily most primitive. Here, such variety can involve the basic body plan of a given phylum and the types of morphogenesis characteristic of it. This correlation provides a new criterion of evolutionary primitiveness, namely, the criterion of initial morphological diversity. The highest morphological diversity among turbellarian groups is observed in the order Acoela. Acoel turbellarians are archaic in most of their features, apparently being a group near the base of the turbellarian phylogenetic tree. Among other turbellarians there are a few groups that also are archaic in some few features (above all, the Catenulida), although on the whole they are more advanced than the Acoela. The Turbellaria as a whole is notable for its morphological diversity in comparison with other classes of the Scolecida.

    Notizen: Abstract The copulatory organ of Haplopharynx quadristimulus Ax, 1971 (Carolina form, Rieger, 1977) consists of a proximal prostatic vesicle and a distal stylet apparatus comprising a central tubular stylet and four to five peripheral accessory spines. By electron microscopy it could be seen that the stylet and spines were intracellular specializations. The copulatory organ can be interpreted as a specialization of an epithelial canal extending from the testes to the body wall. In the complex stylet apparatus, the epithelium was differentiated into six cell types. The stylet, which was formed in a matrix syncytium next to the prostatic vesicle, extended into the lumen of the stylet canal. The interior of the stylet apparatus contained one group of cells that had thick ciliary rootlets and another that had rootlet-like ribbons. The cells that contain the rootlets enveloped bundles of longitudinally arranged muscles. The accessory spines were formed in cells which lay peripheral to the muscle bundles. The spines, stylet, rootlet-like ribbons, and rootlets had similar patterns of periodic cross striations. The similarity in striation patterns suggests that the accessory spines and stylet are composed of modified ciliary rootlets.

    Notizen: Abstract The phylogenetic significance of flatworm sperm morphology is discussed against the background of general spermatology. The modified type of spermatozoon of the Nemertodermatida, a group of primitive flatworms, indicates that the Platyhelminthes evolved from forms characterized by the primitive type of metazoan sperm and by the primitive mode of fertilization, implying the release of sperm freely into sea water. The occurrence of aberrant types of spermatozoa in most platyhelminths is obviously a consequence of early evolution of the internal mode of fertilization, which characterizes all true members of this group. It can be concluded, from the ultrastructure of these aberrant spermatozoa that ‘higher’ metazoans cannot have evolved from ‘seriated’ flatworms related to the recent Seriata (Proseriata and Tricladida). Even the seemingly primitive Acoela have such aberrant spermatozoa that evolution of ‘higher’ metazoans from acoels related to the recent Acoela seems highly improbable. The ultrastructure of the spermatozoa of the parasitic groups of flatworms (Monogenea, Digenea, Cestoda) is very similar to that found in the Kalyptorhynchia, a further indication that the parasitic groups are related to the rhabdocoel turbellarians.

    Notizen: Abstract The nervous system of the polyclad turbellarian Notoplana acticola consists of a series of nerve plexuses and a central ganglion, the brain. The brain contains a variety of cell types including multipolar heteropolar and bipolar neurons. These cell types are rare in other invertebrate ganglia. Individual neurons also contain a variety of different ion channels. both spiking and nonspiking neurons are found. Some neurons are multimodal interneurons. Habituation appears to be a postsynaptic phenomenon. Sensitization and long-term potentiation have not been demonstrated. Polyclads appear to represent a stage in the evolution of centralized nervous systems where much of the neuronal machinery underlying behavior occurs in the peripheral nervous system and the brain's main functions are the coordination and sequencing of peripherally placed reflexes. Even at this stage, however, the brain already contains cells that seem as advanced as those found in higher organisms.

    Notizen: Abstract Using characters discernible through electron microscopy, we redefine the organ traditionally identified as the frontal organ in acoelomorph turbellarians as being a collection of two to several large mucus-secreting glands whose necks emerge together through a frontal pore at the exact apical pole of the body, i.e. at the point where the pattern of epidermal ciliary rootlets converges. Representatives that we have studied of each of the acoel families Paratomellidae, Diopisthoporidae, Solenofilomorphidae, Convolutidae, Otocelidae, and Mecynostomidae, as well as a representative of the Nemertodermatida, have such glands. Up to five additional types of glands that open anteriorly outside of the frontal pore, some of which are indistinguishable from glands of the general body wall, could be seen in the nemertodermatid, in Hesiolicium inops (Paratomellidae), and in representatives of the latter four acoel families. In Paratomella, three different types of glands open in diffuse fashion in a frontal glandular complex reminiscent of that in the Macrostomida. Sensory elements near the frontal pore appear to be independent of the gland necks, and so the organ cannot be considered a sensory organ. The frontal organ, as described above, appears very likely to be homologous within the Acoelomorpha, and represents another strong (although unrooted) autapomorphy for this line of turbellarian evolution.

    Notizen: Abstract The nervous systems of the turbellarians Microstomum lineare and Polycelis nigra and of the cestodes Diphyllobothrium dendriticum and Schistocephalus solidus were studied by means of the peroxidase-antiperoxidase (PAP) immunocytochemical method, with the use of antisera to the neuropeptides FMRF-amide, vasotocin, leu-enkephalin, met-enkephalin, neurotensin, somatostatin, and VIP, and to the bioamine serotonin. Anti-FMRF-amide positive perikarya and fibers occurred in all species, while the occurrence of the vertebrate brain-gut peptides and serotonin varied between the species. Anti-somatostatin and anti-VIP gave a negative result. Anti-FMRF-amide and anti-vasotocin positive immunoreactivity was found in the brain and gut of M. lineare, and in the CNS and the peripheral nerve net of the cestodes. We suggest that the brain-gut peptides of free-living flatworms act on the subtegumental region in the cestodes, which lack a gut but absorb their nutrients directly through the tegument.

    Notizen: Abstract Photoreceptor structure and function in the Platyhelminthes has traditionally been treated separately in the Turbellaria on one hand and the conventional parasitic classes on the other. In this paper, an attempt is made to bring together data from the literature and to highlight deficiencies and areas where a more integrated approach would be beneficial. This is done with particular reference to the endosymbiont genus Paravortex which belongs to the Turbellaria but which functionally has more similarities to the parasitic platyhelminths especially with regard to the host-finding requirement of the larval stages.
