    Notes: Abstract The human T-cell receptor gamma (TCRG) locus comprises 14 variable genes (TRGV), five joining segments (TRGJ), and two constant region genes (TRGC). The constant gamma 1 gene, TRGC1, consists of three exons, whereas the TRGC2 gene contains four or five exons due to the duplication or triplication of exon 2 and spans 9.5 kb or 12 kb, respectively. In this paper, we define the alleles of the T-cell receptor gamma J2 and C2 genes, and we show that two Hind III allelic fragments, 5.4 kb and 8 kb, characterize unambiguously the C2 gene with duplication or triplication of exon 2. We show also that the cDNA of the HPB-MLT cell line results from the transcription of an allelic TRGC2 gene with duplicated exon 2. We propose a model involving unequal crossing-overs to explain the organization and the evolution of the TRGC locus. Moreover, we analyze the TCRG haplotypes in four different populations (French, Lebanese, Tunisian, and Black African) to underline their interest for population genetics.

    Notes: Abstract  The V10 variable gene of the human T-cell receptor gamma locus (TCRG-V10), the only member of the subgroup III, has a structural defect which inhibits the splicing of the leader intron. We show that there is a single point mutation in the V10 leader donor splice site responsible for this situation and that this mutation is found in the different populations tested, indicating that V10 corresponds to a pseudogene in humans. We restored the splice site by mutagenesis and obtained correct splicing in vitro. Analysis of the V10 germline gene in different primates reveals functional splice sites in the closest human apes, the chimpanzee and the gorilla. The splice competence of TCRG-V10 in higher primates was addressed in peripheral blood lymphocytes from chimpanzee by specific cDNA amplification, and correct splicing of the TCRG-V10 leader intron was found as well as a majority of in frame rearrangements involving only the TCRG-J1 or J2 segments. These results suggest that V10+γ /δ T cells may represent an important subset in the non-human higher primates, contrary to the situation observed in the human.

    Notes: Abstract The human T-cell receptor gamma region spans 160 kb of genomic DNA and is densely populated by coding sequences. Restriction fragment length polymorphisms have been previously documented for the constant region genes, the joining segments, and the variable genes belonging to subgroups I and IV. Here were further define the polymorphism of theV gamma I subgroup genes, based on complete mapping of theEco RI andTaq I allelic restriction fragments. We describe seven haplotypes; five result from polymorphic restriction sites, the sixth corresponds to a deletion of about 10 kb encompassingV4 andV5, and the seventh results from an insertion of an additional gene,V3P, betweenV3 andV4. As a consequence of the deletion or insertion polymorphism, the number ofV gamma I subgroup genes vary from seven in haplotypeTRGVI *3 to ten in haplotypeTRGVI *4, whereas the most common haplotype,TRGVI *1, has nineV genes, five of them being functional. Frequencies of the differentTRGVI haplotypes in French, Lebanese, Tunisian, and Black African populations are given.

    Notes: Abstract The five humanIGHG genes consist of three constant domain exons plus one of or four hinge exon(s), the quadruplicated hinge region being characteristic of theIGHG3 gene. Besides this structural difference, theIGHG genes are polymorphic, as demonstrated by the restriction fragment length polymorphism and, at the protein level, by the Gm allotypic antigenic determinants. In this paper, we report the sequence of theG3m(b0, b1, c3, c5, u) IGHG3 allele, typical of the Black African populations and of populations with Negroid admixture, found in a homozygous Tunisian designated as LAT. We demonstrate that thisG3 allele contains only three hinge exons instead of four (the probable result of an unequal crossing over) and thatIGHG3 genes with triplicated hinge exons (and therefore encoding shorter γ 3 chains) are present in healthy individuals from different populations. Moreover, we show that the LAT G3m (b0, b1, c3, c5, u) coding sequence results from the conversion, in the CH3 exon, of theG3m (b0, b1, b3, b4, b5, u, v) allele, the most frequentIGHG3 gene in the Negroid populations, by the homologous region of aIGHG4 gene. The structural features of theLAT IGHG3 allele, which are the lack of one hinge exon and its conversion by theIGHG4 gene, demonstrate that both crossing-over and gene conversion events occur in the evolution of the humanIGHG genes.

    Notes: Inspired by Bolton and Zwick (1995) we study four different games, namely the ultimatum and dictator game as well as the two games in which a veto (non-acceptance) of the responder implies that only one of the two players does not receive the proposed payoff. In the experiment the participants had to play either as the proposer or as the responder in all four games. Only the proposer was privately informed about the actual cake size which could be either small or large. The main results were that the offers were more generous when the responder could only veto the proposer's payoff than in ultimatum bargaining which, in turn, induced more generous offers than dictatorship. The worst proposals were observed when the responder could only reject his own payoff. A similar tendency was also observed for the minimal offers required for acceptance although many acceptance strategies were non-monotonic. Allocators with large cakes did not hide behind the small cake by offering an equal share of the small cake. A pre-experimental questionnaire designed to trigger considerations of backward induction led to more generosity.

    Notes: Defines the purpose of this research as to design a coverall for fire fighting, with special emphasis on sizing and fit to maximize wearer comfort and mobility, as well as incorporate design features which would improve the functionality of the garment. Uses a structured functional apparel design approach to develop garment specifications and an interaction matrix. Using a literature review, user input, and market survey, details the development of a prototype fire fighting coverall. Evaluates the prototype coverall using objective measurements of range of motion for ten selected body movements and subjectively by having wearers fill out a wearer acceptability scale to determine their perceptions of fit and comfort in both their current and prototype coveralls. Results show that for all measurements, the prototype coveralls were preferable to the current coveralls worn, indicating the potential for further development of this design.

    Notes: Protective clothing should ideally provide maximum comfort and protection for the wearer. The design and fit of a garment are factors which can affect both the protective aspects of a garment as well as its comfort. Proper garment fit depends on the relationship of the size of the garment compared with the size of the wearer. Garment ease (where the garment is larger than the wearer) should allow for comfort and mobility; both too much or too little ease can result in a garment that is uncomfortable and restrictive to movement. The purpose of this study was to explore a research technique to isolate the effects of garment ease in one area of a garment while ease in all other garment areas was controlled, and to determine a design that would maximize wearer mobility. Using five male subjects, protective overalls with differing amounts and garment location of crotch ease were evaluated for their effects on mobility and wearer acceptance. Range of motion measurements for selected joints were evaluated using a Leighton Flexometer. Subjects completed a subjective evaluation scale after performing an exercise protocol while wearing the overalls. Results indicated that a specific amount of ease in the crotch length of overalls may be appropriate. Additionally, an overall design that had all needed crotch ease in the back waist area of the garment may be desirable over the more conventional method of adding ease evenly between the front and back sections of the garment. The methodology used in this study provides a means of evaluating not only the potential for design variations in protective clothing, but provides a means to evaluate the dynamic aspects of fit of clothing.
