    Notes: Summary Several sweet potato genotypes were found to lack completely or to have only traces ofβ-amylase in their storage roots. Such genotypes do not increase in sweetness during cooking because, without a sufficient amount ofβ-amylase, the normal hydrolysis of starch to maltose does not occur in the cooking process. In order to study the inheritance of this biochemical variant in the genotype, 41 families were generated. The following conclusions were drawn from analyzing these families. (1) This trait is controlled by one recessive allele (designatedβ-amy) (2) It is inherited in a hexasomic or tetradisomic manner, but not disomically or tetrasomically. This conclusion supports previous cytological data that sweet potato is an autohexaploid or has two identical genomes plus one genome which is somewhat different. (3) Theβ-amy allele appears to exist at a high frequency in cultivated germplasm. (4) Breeding sweet potato for lowβ-amylase activity is relatively easy. New types of sweet potato without normalβ-amylase activity have great potential for processing and as a staple food.

    Notes: Abstract. Although the suppressed instability of supersonic free shear layer flow has been documented by many investigators, the underlying physics are still ambiguous. In the present study, numerical simulations were performed to cast physical insight into the two-dimensional large-scale structure which is organized in a supersonic free shear layer. It is found that an acoustic interaction of the disturbed shear layer with a flow channel wall or another shear layer is indispensable to generate the organized structure. The undisturbed vorticity layer is deformed into a traveling wavy one. As the flow Mach number increases, its degree of deformation decreases so that the formation of shock waves embedded in the structure is avoided.

    Notes: Abstract The concentration dependences of the Fourier transform infrared spectra of aqueous solutions of sodiumn-pentanesulfonate,n-hexanesulfonate,n-heptanesulfonate, andn-octanesulfonate were studied in concentration ranges encompassing the critical micellization concentrations (c.m.c). Changes in wavenumber, bandwidth, and intensity of infrared absorption bands were used to monitor the changes in molecular association with concentration. The premicellar aggregation below the c.m.c. may accompany the association of the alkyl chain, but not the counter-ion binding with the SO 3 − group. Above the c.m.c, the front location of the counter-ion against the SO 3 − group at the micelle surface is demonstrated.

    Notes: Summary In our laboratory, EAPc-7 a strain having higher aspartase activity was derived from Escherichia coli ATCC 11303. For the improvement of l-aspartic acid productivity using EAPc-7 cells immobilized in χ-carrageenan, it was necessary to eliminate the fumarase activity which converts fumaric acid to l-malic acid. Several treatments for specifically eliminating fumarase activity from EAPc-7 cells were tested and it was found that when EAPc-7 cells were treated in a culture broth (pH 4.9) containing 50 mM l-aspartic acid at 45° C for 1 h, fumarase activity was almost completely eliminated without inactivation of the aspartase. The treated cells, immobilized in χ-carrageenan, were used for continuous production of l-aspartic acid from ammonium fumarate. The formation of l-malic acid was negligible and the half-life of the immobilized preparation was 126 days. Productivity of immobilized preparation of treated EAPc-7 cells in l-aspartic acid production was six times of that of the parent cell preparation.

    Notes: Summary The regulatory mechanism of ATP regeneration by the glycolytic pathway in Hansenula jadinii cells was investigated by analyzing the initial stage of CDP-choline fermentation. As a result, the “on-off” of ATP regeneration was found to be determined by the ATP concentration overcoming the inhibitory effect of phosphate buffer on hexokinase activity. The concentration of ATP at the initial stage of fermentation was greatly influenced by the kinds and amounts of glycogen in cells. Based on these results, the regulatory mechanism of ATP regeneration by the glycolytic pathway is discussed in detail.

    Notes: Summary To develop a practical process for d-alanine production from dl-alanine, we screened 107 yeasts for their asymmetric degrading activity against dl-alanine. Candida maltosa JCM1504 degraded the l-isomer ten times more rapidly than the d-isomer. The cells of this strain were used as a biocatalyst for eliminating the l-isomer. However, when the degradation reaction was conducted in the presence of a high concentration of dl-alanine, the pH of the reaction mixture was rapidly increased by the liberation of ammonia from l-alanine, and consequently the reaction stopped. This hindrance was overcome by controlling the pH value at 6.0 with H2SO4 during the reaction. Additionally, we found that the maximum rate of l-isomer degradation was obtained at 30° C and pH 6.0 under conditions of high aeration (1.0 vvm) and agitation (1200 rpm). Under the optimal conditions, the l-isomer of 200 g dl-alanine/l was completely degraded within 40 h and 90 g d-alanine/l remained in the reaction mixture. d-Alanine was easily isolated from the reaction mixture. The chemical and optical purity of the d-isomer product so obtained was 99.0% and 99.9% enantiomeric excess, respectively.

    Notes: Summary Enzyme-catalysed hydrolysis of esters of 4-hydroxy-3-methyl-2-(2-propynyl)-cyclopent-2-enone (HMPC) was examined for the preparation of the optically pure alcohol moiety of synthetic pyrethroids. Among microorganisms and lipases tested, some bacterial lipases hydrolysed the ester of HMPC with high enantioselectivity and high reaction rate. Arthrobacter lipase gave the optically pure (R)-HMPC at 50% hydrolysis in a two-liquid phase reaction system of water and the insoluble substrate. The hydrolysis proceeded even at a substrate concentration of 80w/v%. The enantioselectivity was not changed with the chain length of the acid moiety of the esters. By combination of the enzymatic resolution with a chemical inversion of the (R)-alcohol, an efficient proess was developed for the total conversion of racemic HMPC to (S)-HMPC, which is an important alcohol for preparation of an insecticidallyactive synthetic pyrethroid.

    Notes: Summary For the efficient production of l-alanine from ammonium fumarate using the aspartase activity of immobilized Escherichia coli cells and l-aspartate β-decarboxylase activity of immobilized Pseudomonas dacunhae cells, alanine racemase and fumarase activities should be eliminated. We investigated various procedures to eliminate these side reactions, and found that both activities of intact E. coli cells could be eliminated by treating the culture broth at pH 5.0 and 45° C for 1 h, and those of intact P. dacunhae cells could be eliminated by treating the culture broth at pH 4.75 and 30° C for 1 h. Further, it was confirmed that l-alanine was efficiently produced using these two immobilized pH-treated microorganisms.
