    Notizen: A series of laboratory and modelling experiments on the oxidation of propene in the gas phase has been undertaken to determine conditions which give high yields of propene oxide. The conditions under which the experiments were conducted were 505-549 K and up to 4 bar pressure. It is proposed that propene oxide is formed from propene by reaction with several peroxy radicals including HO2 and CH3CO3. However, one of the more important radicals is hydroxypropylperoxy. Its reaction with propene, under these conditions is more important than concerted decomposition to formaldehyde and acetaldehyde. © 1995 John Wiley & Sons, Inc.

    Notizen: Saccharomyces cerevisiae possesses a single essential gene (GLC7) encoding protein phosphatase 1 (PP1). Elevated expression of this gene from the GAL1 promoter is highly detrimental to the cell, causing a growth defect and aberrant bud morphology, which leads to cells exhibiting long, extended buds. By comparison, expression of GLC7 from the weaker MET3 promoter was without significant effect on either growth or morphology. However, repression of GLC7 expression from the MET3 promoter in cells where the MET3-GLC7 fusion was the sole source of PP1 resulted in a mitotic delay. Such cultures showed a massive decrease in the rate of proliferation in conjunction with a significant increase in the proportion of large, budded cells. 4′,6-diamidino-2-phenylindole dihydrochloride (DAPI) staining and anti-tubulin immunofluorescence analysis of these cells revealed that many were blocked in mitosis, with a short spindle and DAPI-stained material stretched between the mother and daughter cell within the bud neck. These results support a role for PP1 in the completion of mitosis in S. cerevisiae.

    Notizen: Since the isolation of the first yeast protein phosphatase genes in 1989, much progress has been made in understanding this important group of proteins. Yeast contain genes encoding all the major types of protein phosphatase found in higher eukaryotes and the ability to use powerful genetic approaches will complement the wealth of biochemical information available from other systems. This review will summarize recent progress in understanding the structure, function and regulation of the PPP family of protein serine-threonine phosphatases, concentrating on the budding yeast Saccharomyces cerevisiae.

    Notizen: A theory is presented for fully anisotropic overall molecular tumbling with restricted group internal rotation. This formalism is used with 13C NMR relaxation data and results from molecular mechanics to compute the rotational diffusion rates and methyl group internal rotation rates for sodium taurocholate and ginsenoside-Re in organic solvents. Although existing theories for steroid derivatives describe methyl internal rotation in terms of free isotropic motion, methyl rotation in ginsenoside-Re is found to be both sluggish and spatially restricted. The energetic barrier to methyl internal rotation increases significantly with the bulkiness of flexible side-chains in a series of steroid natural products.

    Notizen: The KlCMD1 gene was isolated from a Kluyveromyces lactis genomic library as a suppressor of the Saccharomyces cerevisiae temperature-sensitive mutant spc110-124, an allele previously shown to be suppressed by elevated copy number of the S. cerevisiae calmodulin gene CMD1. The KlCMD1 gene encodes a polypeptide which is 95% identical to S. cerevisiae calmodulin and 55% identical to calmodulin from Schizosaccharomyces pombe.Complementation of a S. cerevisiae cmd1 deletion mutant by KlCMD1 demonstrates that this gene encodes a functional calmodulin homologue. Multiple sequence alignment of calmodulins from yeast and multicellular eukaryotes shows that the K. lactis and S. cerevisiae calmodulins are considerably more closely related to each other than to other calmodulins, most of which have four functional Ca2+-binding EF hand domains. Thus like its S. cerevisiae counterpart Cmd1p, the KlCMD1 product is predicted to form only three Ca2+-binding motifs. The KlCMD1 sequence has been assigned Accession Number AJ002021 in the EMBL/GenBank database. © 1998 John Wiley & Sons, Ltd.

    Notizen: A numerical technique is presented for the analysis of surface displacements of a non-homogeneous elastic half-space subjected to vertical and/or horizontal surface loads uniformly distributed over an arbitrarily shaped area. The non-homogeneity considered is a particular form of power variation of Young's modulus with depth. Since the exponent which determines the degree of non-homogeneity may vary from zero to unity, both the homogeneous half-space and the Gibson soil may be included as limiting cases in a single numerical scheme. In order to account for the arbitrary shape of the loading, the boundary of the loaded area is linearized piecemeal. This enables the modeling of any load pattern according to the desired degree of accuracy. Special attention is focused on the integration scheme, since the singularity associated with the Green's function becomes progressively more pronounced the greater the non-homogeneity parameter gets. The performance of the numerical procedure is studied using analytical solutions for rectangular shaped areas. Further comparisons with well-known solutions based on integral transform techniques for a uniformly distributed load acting on a circular area of the non-homogeneous soil mass show excellent agreement as well. © 1997 John Wiley & Sons, Ltd.

    Notizen: An alternative approach for calculating the surface displacements of a non-homogeneous half-space acted upon by a vertical and/or horizontal load, uniformly distributed over a rectangular area, is presented in this part of the paper. The procedure proposed proves to be extremely efficient since the displacements can be found without numerical integration for this special loading pattern. Comparisons with solutions for rectangular loaded areas on Boussinesq- and Gibson-type soil show perfect agreement. In the case of the non-homogeneous half-space the procedure was checked using the method outlined in the first part of this paper revealing that both approaches come up with identical answers. Some results of a parametric study are presented for the surface displacements of a non-homogeneous half-space subject to vertical and horizontal loading. In this study both the material properties of the soil mass, i.e. Young's modulus and Poisson's ratio and the aspect ratio of the loading are varied. These results are presented in the form of influence charts which may readily be used in hand calculations for estimating the displacements of footings on a non-homogeneous soil. © 1997 John Wiley & Sons, Ltd.

    Notizen: Permittivity ∊′ and dielectric loss ∊″ of novolac have been determined in the temperature interval from  - 150 to 150°C and in the frequency interval from 102.5 to 106 Hz. Novolac exhibits a main and a secondary relaxation process. For the main relaxation a master curve of ∊″ was calculated, the shape of which was described by the Havriliak - Negami function (HN). The parameters of the HN fit were interpreted by a model that relates the values to the polymer structure. Furthermore, the temperature dependence of the dielectric relaxation times was analyzed by a WLF approach for the main relaxation and an Arrhenius approach for the secondary relaxation. The dielectric relaxation times of the main relaxation were correlated with the electrical conductivity and the melt viscosity.
