    Kurzfassung: This book presents the first comprehensive compilation of genome research on the Hevea brasiliensis rubber tree. The genomes of Hevea tree clones (cultivars) are described by three major international groups. Chapters on omics-driven investigations address a broad range of topics including genome annotation and utilisation, transcriptome and gene family analysis, genetic mapping, metabolic pathways in latex and molecular breeding. Additionally, an overview of fundamental rubber biology, especially on laticifers, provides a historical background that is relevant to rubber genome analysis. The book concludes with several perspectives on the future needs of rubber investigations and prospects of rubber genomics. Given the scope of topics, this book will appeal to researchers and university students working in genomics and biotechnology of the rubber tree, and to rubber breeders with an interest in non-conventional approaches to trait analysis, selection and breeding.

    Notizen: Summary The mean transmembrane potential of cultured osteoblastlike cells isolated from the cortical endosteal surface of rabbit long bones was −16.9±0.64 mV (n=335). Elevation of potassium concentration in medium caused a decrease in potential. As the external concentration of potassium reached 15 mmol/liter and above, there was a linear relationship between the potassium concentration in log scale and the membrane potential with a slope of −13 mV per 10-fold change in external potassium concentration. Dibutyryladenosine 3′,5′-cyclic monophosphate, parathyroid extract, hydrocortisone, and sodium fluoride all depolarized the membrane of osteoblast-like cells after both short (1–2 h) and long (24 h) exposures at suitable doses, whereas calcitonin and prostaglandin E2 hyperpolarized the membrane after long exposures. The Na+, K+ and Cl− concentrations of cultured osteoblastlike cells were 0.538, 0.984, and 0.358 mmol/g protein or 52.6, 96.3, and 35.0 mmol/liter cell water, respectively. The protein content of these cells was 8.18±0.6 g/100 g cells and the water content was 83.7 g/100 g cells. The above-mentioned chemical and hormonal preparations in doses that produced significant changes in the membrane potential of these cultured cells did not alter their electrolyte or protein contents 24 h after exposure. Intracellular pH of cultured osteoblastlike cells as determined by [14C]-dimethyloxazolidine-2,4-dione and3H2O averaged 7.03 ± 0.11 when the pH of culture medium was maintained at 7.4. Calculations based on the values for the membrane potential and the electrolyte concentrations observed in this study indicate that Na+, and H+, and Cl− are actively transported out of the cells and K+ into the cells.

    Notizen: This paper aims to study the effect of stress relieving on Limit Dome Height (LDH) ofTi-TWBs at elevated temperatures. This is achieved by developing a newly constructed heatingsystem. The elevated temperature of the system can be varied and monitored by a separately controlpanel. All Ti-TWBs were prepared and used to examine the LDHs under elevated temperatures.Selected specimens were heat-treated at 600°C within an hour before being formed by HILLEmachine. Meanwhile, the temperature of tool heating system was also adjusted from roomtemperature to 550℃. Specified tests were carried out to examine the stress relieving effects ofTi-TWBs on the LDHs with the temperature control panel. In addition, investigations were carriedout to ascertain whether the elevated temperatures of the critical tooling components, i.e. the die andthe blank holder, could result in any significant effects on LDHs of Ti-TWBs. The findings showthat LDHs of Ti-TWBs can be improved by stress relieving. The stress relieving condition can beobtained by nearly isothermal forming of specimens at a range of 550°C to 600°C

    Notizen: Laser Raman spectroscopy was used to monitor changes in the substitution pattern during the formation of a phenol-formaldehyde resin (resol). The substitution-sensitive bands were identified and their relative intensities related to molecular weight of the resol. Results of this study are similar to those obtained by previous workers in the field using paper chromatography. The absorbance ratio of 784-cm-1 to 998-cm-1 bands was found to be linearly related to the molecular weight of the resol.

    Beschreibung: Emerging evidence suggests that hepatocytes are primarily maintained by self-renewal during normal liver homeostasis, as well as in response to a wide variety of hepatic injuries. However, how hepatocytes in distinct anatomic locations within the liver lobule are replenished under homeostasis and injury-induced regeneration remains elusive. Using a newly developed bacterial artificial chromosome (BAC)-transgenic mouse model, we demonstrate that Lgr5 expression in the liver is restricted to a unique subset of hepatocytes most adjacent to the central veins. Genetic lineage tracing revealed that pericentral Lgr5+ hepatocytes have a long lifespan and mainly contribute to their own lineage maintenance during postnatal liver development and homeostasis. Remarkably, these hepatocytes also fuel the regeneration of their own lineage during the massive and rapid regeneration process following two-thirds partial hepatectomy. Moreover, Lgr5+ hepatocytes are found to be the main cellular origin of diethylnitrosamine (DEN)-induced hepatocellular carcinoma (HCC) and are highly susceptible to neoplastic transformation triggered by activation of Erbb pathway. Our findings establish an unexpected self-maintaining mode for a defined subset of hepatocytes during liver homeostasis and regeneration, and identify Lgr5+ pericentral hepatocytes as major cells of origin in HCC development.
