    Notes: Summary Following 2/3 hepatectomy + CCl4, the labelling- (3H-thymidine) and mitosis-index of liver epithelia increases 8 h later than after an unique 2/3 hepatectomy, and the peak of compensatory regeneration is reached 16 h later.     Notes: Summary Direct intranuclear inclusions appearing in convoluted tubules in kidneys of white rats after a chronic lead intoxication do not participate in synthesis or migration of RNA and protein, which was demonstrated by autoradiography following an injection of3H-cytidine or3H-l-phenylalanine. The synthetic activity for RNA and protein increases impressively in the residual karyoplasm. Intranuclear inclusions neither disturb modus and velocity of RNA migration from nucleolus and karyoplasm into cytoplasm, nor impair cytoplasmic protein metabolism.

    Notes: Summary The percentage of DNA-synthesising heart muscle nuclei of white rats (3H-thymidine, autoradiography) decreases rapidly during the first month of life reaching a minimum at the fourth month. From then up to 27 months the number of labelled muscle nuclei remains rather constant. Also in nuclei of connective tissue the3H-thymidine labelling index decreases as a function of the rats age. The number of mitotic figures is always about 10 times lower than labelled nuclei. Mostly in heart muscle cells they occur as collapsed mitoses never leading to cytoplasma division14.

    Notes: Summary Autoradiographic studies following a single i.p. injection of3H-thymidine were performed in liver and kidney parenchyma of new-born and young adult rats at different ages (1, 2, 4, 7, 12, 18, 24, 30, 60 and 120 days). In 1-day-old animals the tritium index (i.e. percentage of DNA synthesizing nuclei) of both organs is lower than in the rat embryo. From 1–4 days fluctuations occur, then the tritium index rises at 7 and 12 days. But now an exponential decrease is observed up to day 120. This depression and fluctuation of the tritium index probably depends on a postpartal functional transposition and especially in the liver on a structural transformation. The mean grain density in the labelled nuclei increases from 1 up to 24 days, which is likely connected with a very high amount of endogenous thymidine in new-born rats. From day 24 the mean grain density can be taken as a relative measure for the rate of DNA synthesis. The reduction in the rate of DNA synthesis from 24–120 days is explained as a consequence of the commutation from the rapid to the slow mode in cellular proliferation.

    Notes: Summary During the first wave of parenchymal liver regeneration in adult rats after partial hepatectomy, the cellular synthesis and migration of RNA and the metabolism of protein were studied by autoradiography following an injection of3H-cytidine or3H-l-phenylalanine and double injections of 1 of these precursors +3H-thymidine. The following results were obtained: the synthesis and migration of RNA and the metabolism of protein are enhanced under these conditions of proliferation. In spite of this, the relation of metabolic activity in nucleolus, karyoplasm and cytoplasm remains constant. By double injection techniques it is proved that no differences exist in migration of RNA into the cytoplasm and cytoplasmic protein synthesis between cells with or without DNA synthesizing nuclei.

    Notes: Summary By continuous infusion of3H-thymidine, autoradiographic studies were performed to compare cellular proliferation in the compensatory regenerating kidney of rats after contralateral nephrectomy or contralateral temporal (1 h) ischaemia with physiological cell renewal in controls. Under each experimental condition, the highest labelling index is observed within the stratum labyrinthicum and then a gradual decrease occurs from there to the stratum subcorticale, zona intermedia and basalis. As to the topic of DNA synthesizing cells in the different zones of the kidney, there are only quantitative differences between physiological cell renewal of the controls and compensatory regeneration following contralateral nephrectomy and contralateral ischaemia.

Similar effects are noted in the cytoplasma of B-cells. Die Schnitte wurden mit einem autoradiographischen Stripping Film überzogen, exponiert, entwickelt und fixiert. Dabei treten Silberkörner auf, die über DNS-Farbstoffkomplexen liegen. Ihre Entstehung wird auf eine chemische Reduktion von Ag+-Ionen im Film durch die Orcein-Färbung vor der autoradiographischen Technik zurückgeführt. Die Lokalisation derartig induzierter Silberkörner über karyoplasmatischen Strukturen wird mit solchen Silberkörnern verglichen, die im typischen3H-Thymidin-Autoradiogramm an Hämatoxilin-Eosin nachgefärbten Quetschpräparaten vom Leberparenchym junger Mäuse auftreten.
    Notes: Summary Squash preparations of liver parenchymal cells from mice, which did not receive any radioactive precursor, had been produced by the orcein sqash technique. The slides were then coated with an autoradiographic stripping film, exposed, developed and fixed. Silver grains were observed lying upon DNA stain complexes. They had been induced by a chemical reduction of Ag+ ions in the film as a consequence of the orcein prestaining prior to the autoradiographic procedure. The localisation of these silver grains over caryoplasmatic structures were compared with those of a typical3H-thymidine autoradiogram in squash preparations of young mice, poststained with haematoxilin eosin.
