    Notes: A new liquid membrane separation technique for gas mixtures has been developed: feed and sweep gases flow through the lumen of two different sets of hydrophobic microporous hollow fibers while a liquid on the shell side acts as the membrane. This membrane is identified as the contained liquid membrane (CLM). Major shortcomings of immobilized liquid membrane techniques are eliminated. The novel separation device is identified as the hollow fiber contained liquid membrane (HFCLM) permeator. Experimental studies are made with different CO2-N2 feed mixtures and a pure helium sweep stream, with special emphasis on model landfill gas purification. Either pure water or an aqueous solution of K2CO3 is used as membrane. The effects of several variables on the performance of the HFCLM permeator have been studied. A three-component permeation model, incorporating axial pressure drop, is developed for binary mixture separation in a HFCLM permeator having a sweep gas stream. The experimental data show good agreement with the theoretical predictions.

    Notes: A novel gas purification technique called rapid pressure swing absorption (RAPSAB) was developed by integrating the best features of membrane contacting, gas-liquid absorption, and pressure swing adsorption (PSA). In this cyclic separation process, a well-packed microporous hydrophobic hollow-fiber module was used to achieve nondispersive gas absorption from a high-pressure feed gas into a stationary absorbent liquid on the module shell side during a certain part of the cycle followed by desorption of absorbed gases from the liquid in the rest of the cycle. The total cycle time varies between 20 s and upwards. Separation of mixtures of N2 and CO2 (around 10%) where CO2 is the impurity to be removed was studied using absorbent liquids such as pure water and a 19.5% aqueous solution of diethanolamine (DEA). Three RAPSAB cycles studied differ in the absorption part. Virtually pure N2 streams were obtained with DEA as absorbent demonstrating the capability of bulk separation to very high levels of purification. Numerical models developed predict the extent of purification for pure water and the DEA solution for one of the simpler cycles. Model simulations describe the observed behavior well.

    Notes: Conventional membrane permeation of gases employ a non-step process between the feed and the product on two sides of a membrane. By applying the overall driving force change through a two-step process using two membranes in one device, we illustrate theoretically the possibility of achieving a much higher enrichment without any extra energy consumption. Such an internally staged permeation is experimentally carried out in a hollow-fiber permeator with two sets of asymmetric cellulose acetate hollow fibers for the systems of O2—N2 (air) and CO2—N2. The superior enriching performance of the internally staged permeator over the same permeator operated in the conventional mode is demonstrated experimentally. A cocurrent flow pattern is found to be superior to a countercurrent flow pattern for the conditions employed. The simulation model incorporating fiber lumen pressure drop predicts the experimental performance quite well. A further improvement in performance can be achieved by recycling the intermediate pressure shell reject stream to the feed stream. The energy requirement in an internally staged permeator without recycle is less than that of a conventional permeator with permeate recycle.

    Notes: Estimates for the thickness of the interface between poly (2,6-dimethyl-1,4-phenylene oxide) (PPO) and copolymers of styrene-acrylonitrile (SAN) and styrene-maleic anhydride (SMA) based on the theory of Helfand and Tagami are compared to neutron reflectivity (NR) measurements. Good agreement is found between the NR measurements and theoretical predictions that make use of a mean field binary interaction model and previously reported binary interaction energies.     Notes: Differentiation of the megakaryocytic leukemia cells, CMK, was induced by long-term (12 day) treatment with the combination of IL-3 and the nucleoside analogue ribavirin (RV), which reduces cellular GTP levels. In a previous report we demonstrated the induction of early messages and antigens, as well as the formation of giant polyploid cells in the cultures (Majumdar et al., 1994, J. Cell. Physiol., 160:29-39). Here we show high level induction of messages for the late markers, Platelet Factor 4, GMP140 (P-Selectin), thrombospondin, and beta thromboglobulin. The induced cells are also positive for these antigens by immunocytochemical analysis. The high level message induction resulted from synergy between the inducers. Pretreatment of the cells with IL-3 could accelerate the rise in message seen with the inducer combination. The increase in differentiation markers was accompanied by a reduction of the proliferative capacity of the cells. Riboguanosine, which has anti differentiation activity, blocked the induction of early and late antigens by the inducer combination, and also by IL-3 acting alone, but did not block the reduction in proliferative competence. In this model of megakaryocytic differentiation IL-3 treatment yields and initial stimulation of growth followed by growth supperssion, and is the principal driver of the differentiation process. RV functions primarily as a stimulator of message and protein expression in synergy with IL-3. © 1995 Wiley-Liss Inc.

    Notes: Megakaryocyte differentiation is a lengthy process with cells moving through a continuum delineated by the sequential expression of specific gene products. The limited number of primary cells available from marrow for analysis has brought attention to some leukemic cell lines which show enhanced megakaryocyte marker expression following incubation with inducing agents, the most common of which is phorbol myristate acetate (PMA). We developed an alternative induction protocol for the megakaryocytic leukemic cell line CMK, which involved incubation of the cells with IL-3 and the nucleoside analog, ribavirin, for 1-2 weeks. This treatment was neither toxic nor cytostatic and yielded increased levels of the surface glycoproteins GPIIb/IIIA and GPIb-IX. Levels of some megakaryocytic messages (GPIIIa, GPIX) showed a marked rise by 12 days of incubation in the inducer combination. This was due to a synergistic interaction between IL-3 and ribavirin which influenced both transcriptional and posttranscriptional events. Light and electron microscopy demonstrated the presence of large polyploid cells, with morphological features similar to those of megakaryocytes, in the induced cultures. Analysis of the heterogeneity of response in the cell population to the induction regimen after several days of treatment suggested that cells which failed to display surface markers had been stimulated by the inducers but did not have sufficient time to complete expression of that marker. The results were consistent with the view that the cells in the starting population were distributed along a temporal expression pathway, and those which were first to express the earliest marker would also lead in the expression of a later marker. The order of expression was the same as that during normal megakaryocyte development. © 1994 Wiley-Liss, Inc.

    Notes: The conformation of numerous chemical compounds is strongly influenced by solvents. Knowledge of their structure in solution is necessary, especially for a discussion of the biological and pharmacological activity of the molecules. The neurotransmitters and their agonists and antagonists are known to be flexible molecules that interact with quite distinct receptors. The conformational properties of several GABA (γ-aminobutyric acid) mediators have been studied by the MNDO technique. The optimized geometries of the molecules have been used to study the solvent effects on their conformational properties considering the supermolecule approach for their first hydration shell. A conjectural pharmacophoric pattern for several GABA inhibitors has already been suggested from the molecular electrostatic potentials (MEP) of several molecules using a localized bond orbital technique. In the present work, MEP calculations have been carried out considering a solvent effect on the MNDO optimized geometries to investigate any deviation from the earlier results.
