    Notes: Abstract  In order to isolate genes whose expression is up-regulated after the initiation of meiosis, we screened a cDNA expression library of newt testes with antiserum against homogenates of testes derived from the spermatogonial and spermatocyte stages. We report the isolation of spermatocyte-specific cDNA clones encoding a newt homologue of the calcium-dependent phospholipid-binding protein, annexin V. Northern blot analysis showed that newt annexin V mRNA was 1.7 kb in length and was expressed strongly in testes, but weakly in other organs. In situ hybridization revealed that the expression of newt annexin mRNA was barely observed in spermatogonia, but increased significantly in leptotene-zygotene primary spermatocytes and reached a maximum level in pachytene spermatocytes and round spermatids. The newt annexin V cDNA predicted a 323-amino acid protein and had a 68% homology to human annexin V. The predicted amino acid sequence contained a conserved 4-fold internal repeat of approximately 70 residues like other annexin proteins. Immunoblot analysis using the monoclonal antibody against newt annexin V showed that the protein was expressed scarcely in spermatogonia but was abundantly expressed in stages from primary spermatocytes to spermatids; this pattern was consistent to that of the mRNA. Immunohistochemical analysis revealed that newt annexin V was localized in the cytoplasm of the spermatogenic cells, but not in somatic cells such as Sertoli cells or pericystic cells. These results indicate that the expression of newt annexin V is up-regulated in the spermatogenic cells after the initiation of meiosis and suggest that newt annexin V plays an important role in spermatogenesis.

    Notes: Abstract During the 6th August 1995, the CUTLASS Finland HF radar ran in a high time resolution mode, allowing measurements of line-of-sight convection velocities along a single beam with a temporal resolution of 14 s. Data from such scans, during the substorm expansion phase, revealed pulses of equatorward flow exceeding ∼600 m s−1 with a duration of ∼5 min and a repetition period of ∼8 min. Each pulse of enhanced equatorward flow was preceded by an interval of suppressed flow and enhanced ionospheric Hall conductance. These transient features, which propagate eastwards away from local midnight, have been interpreted as ionospheric current vortices associated with fieldaligned current pairs. The present study reveals that these ionospheric convection features appear to have an accompanying signature in the magnetosphere, comprising a dawnward perturbation and dipolarisation of the magnetic field and dawnward plasma flow, measured in the geomagnetic tail by the Geotail spacecraft, located at L = 10 and some four hours to the east, in the postmidnight sector. These signatures are suggested to be the consequence of the observation of the same field aligned currents in the magnetosphere. Their possible relationship with bursty Earthward plasma flow and magnetotail reconnection is discussed.

    Notes: Abstract We have demonstrated the presence of estrogen receptor mRNA and the mature protein in the cytoplasm and nucleus, respectively, of a 9,10-dimethyl-1,2-benzathracene-induced submandibular gland tumor in female rats. We have previously shown that progesterone receptors are also present in human salivary gland tumors. These results suggest that endocrine therapy may be effective in treatment of submandibular gland tumors.

    Notes: Abstract X-ray fluorescent spectroscope analysis easily finds ratios of geochemical elements in soil. Applying the method of ratio matching to measured ratios, the classification of geological layers, and the flow directions of groundwater can be determined. This method is applied to three hill slopes in Japan and the results are in good agreement with the observations and measurements of soil cores obtained by drilling. The classification of geological layers is found to be quantitatively connected with the rates of organic matter in soil cores.

    Notes: Abstract  X-ray fluorescent spectroscope-analysis and the method of ratio matching can determine mean directions of sand movement and geochemical classifications of sand grains. The result can also evaluate the physical condition of seabeds in a study area (erosion or accretion). This method is applied to a sedimentation problem in the fishing port of Tsurugiji in Japan and the result is in agreement with the comparison of the change of sea bottom topography measured in different years. The seasonal changes of sand movement on this coast and sedimentation in the port are also investigated.

    Notes: Abstract  The analysis of sand samples by X-ray fluorescent spectroscopy (XRF) gives the ratio of the geochemical elements to construct the sand samples. The application of the ratio matching to sand samples represents the correlation number between two sand samples with respect to geochemical elements. When the correlation number between two sand samples is low, the two samples are not geochemically similar each other. This denotes that the exchange of sand between two sampling points is scarce or the two samples are independent. When the correlation number between two sand samples is high, the two samples are geochemically similar, signifying that the exchange of sand between two sampling points is frequent or there is sand movement between two sampling points. If there exists prominent sand movement in the study area, the correlation number is almost 1 and kilo count number per second of each geochemical element per weight decreases along the flow direction. The decrease is caused by the reduction of sizes and the adherence of dirt on the surface of sand particles. Since the flow direction in rivers is usually the same as the direction of sand movement, it can be verified. This study obtains satisfactory results applying the method of prediction in sand movement to sediments in rivers.

    Notes: Abstract The analysis of sand samples by x-ray fluorescent spectroscopy (XRF) gives the ratio of the geochemical elements to construct the sand samples. The x-ray analysis therefore shows the geochemical characteristics of sand in the sampled area. In this study, sand is sampled on coasts and rivers of the Noto Peninsula to determine the geochemical elements and to show the geological characteristics that occur, especially iron and calcium. The experiments show the effect of rivers and Kotogahama beach on iron and calcium, respectively. Applying the method of ratio matching to the measured data of the geochemical elements, the direction of movement of sand on the coast is determined by considering the correlation matrix and the ratio of geochemical elements between sand samples at two locations. The predominant longshore direction movement of sand offshore in the study area is from south to north. Sand in rivers is not directly transported to adjacent beaches; however, offshore sand is transported to beaches. The estimated direction of movement of sand longshore near coastal structures agrees with observations. The proposed method to predict the direction of movement of sand gives the correct one in comparison with the observed data in rivers.

    Notes: Abstract  A diffusion layer that is likely to be formed at the interfaces of the multi-layered thin film would affect its overall mechanical properties; the thinner the thin film, the more significant would be the effect. We measure the distribution of atoms and estimate the thickness of the diffusion layer at the vicinity of the interfaces among thin films of Al and SiO2 and silicon wafer with the aid of Auger electron spectroscopy (AES). The effect of heat treatment after fabrication of the thin films on the diffusion is also investigated.

    Notes: Abstract Immunohistochemical methods were employed to investigate the cellular and ultrastructural localization of the gap junction protein connexin43 (Cx43) in rat pituitary. Western blots of pituitary homogenates probed with anti-Cx43 antibodies showed the presence of Cx43 in both anterior and posterior pituitary lobes. By light microscopy (LM), Cx43-immunoreactive (Cx43-IR) puncta were found in all areas of the posterior lobe, but at greater concentrations in peripheral regions of this structure. By electron microscopy (EM), immunogold labelling for Cx43 was seen at gap junctions between thin cytoplasmic processes of pituicytes. No immunoreactivity was detected in the intermediate lobe. The anterior lobe contained puncta similar to but more sparsely scattered than those in the posterior lobe, and by EM analysis these were demonstrated to correspond to labelled gap junctions between stellate cells. In addition, anti-Cx43 antibodies produced intracellular labelling in a small percentage of endocrine cells, which were distributed throughout the anterior lobe and determined by double immunostaining methods to be cells containing luteinizing hormone. By EM, labelling within these cells was associated with predominantly large secretory granules and other loosely organized organelles. The results indicate that gap junctions in the pituitary are composed of Cx43 and that this or a related protein may have a novel intracellular function within gonadotrophs.
