    Notes: Abstract Catechol 2,3-dioxygenase from the meta-cleavage pathway encoded on the TOL plasmid of Pseudomonas putida (pWWO) was investigated by electron microscopy. Negatively stained samples of the purified catechol 2,3-dioxygenase revealed that the enzyme consists of four subunits arranged in a tetrahedral conformation. Monoclonal antibodies raised against catechol 2,3-dioxygenase showed highly specific reactions and were used to localize the enzyme in Escherichia coli (pAW31) and P. putida (pWWO), using the protein A-gold technique carried out as a post-embedding immunoelectron microscopy procedure. Our in situ labeling studies revealed a cytoplasmic location of the catechol 2,3-dioxygenase in both cell types.

    Notes: Abstract  Analysis of electron-transfer (ET) kinetics data obtained from experiments on Ru-modified proteins (cytochrome c, azurin, myoglobin) reveals that distant donor-acceptor electronic couplings depend upon the secondary structure of the intervening polypeptide matrix. Rates of Fe2+→Ru3+ ET reactions in cytochrome c decay exponentially with tunneling-pathway length (decay constant 0.73 Å–1); these rates also decay exponentially with Ru-Fe distance (decay constant 1.1 Å–1). In azurin, a β-sheet protein, Cu+→Ru3+ rates exhibit an exponential Cu-Ru distance dependence with a decay constant of 1.1 Å–1. Comparison of distant couplings in azurin and myoglobin suggests that hydrogen bonds are better mediators across β sheets than through α helices.

    Notes: Abstract  Cyclic voltammetry has been used to determine the reduction potentials of blue (Pseudomonas aeruginosa azurin) and purple (Thermus thermophilus CuA domain) copper proteins unfolded by guanidine hydrochloride. These Cu(II/I) potentials [456 (azurin); 453 (CuA) mV vs., NHE] are higher than those of the folded proteins. The downshift of the potential in the folded state can be accounted for by assuming that rack-induced axial coordination stabilizes Cu(II) relative to Cu(I) in a protein-encapsulated active site.

    Notes: Abstract Polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), polychlorinated biphenyls (PCBs), and metals were monitored in the river sediments near the McCormick and Baxter (MCB) wood treatment facility, Stockton, CA. Transplanted clams and resident fish species were used to assess bioavailability. The highest PCDD and PCDF contamination in sediments were confined to an area next to the facility and an area in the nearby Stockton harbor (DK location). Pentachlorophenol (PCP) wood treatment at MCB was the most probable source of the contamination. PCBs contaminated a wider area of the Stockton Ship Channel and harbor. Metal concentrations were uniformly low except for the metalloid arsenic in the Old Mormon Slough and lead and zinc near boat docks in the Stockton harbor. Despite high mortality rates, clams (Corbicula fluminea) bioaccumulated PCBs, PCDDs, and PCDFs. In clams, PCBs and 2,3,7,8 TCDD were much closer to equilibrium with the sediments than were higher chlorinated PCDDs and PCDFs. All fish were at background levels for 2,3,7,8 TCDD. All fish had lower lipid adjusted PCDD/F and PCB concentrations in the skinned muscle than in the whole fish. PCBs in fish were above background levels for United States river systems. Although high contamination exists in the river near this superfund site, adverse effects on the aquatic community could not be demonstrated.

    Notes: Abstract The demography of two closely related alpine sedges, Carex curvula subsp. curvula and Carex curvula subsp. rosae (=C. curvula and C. rosae) has been investigated on their typical sites in the Central Alps. Both species proliferate vegetatively and develop dense tussocks but they show different dominance behaviours in their respective grasslands. It was hypothesized that this may be caused by different growth abilities. The main aim of the study was to compare the vegetative growth of the species under field conditions, under competition-free conditions and under changed soil conditions. An attempt was also made to clarify whether vegetative growth is density dependent. Permanent plots were established in the respective grasslands of the two species and the ramet density was counted over 3 years. Groups of 10 and of 30 ramets of each species were grown in pots with typical and with alien substrate and their growth was observed for 5 years at the field site. The grassland populations of both species were very stable and the overall ramet growth rate (λ) was close to 1.0. Within the pots, both species reached a high ramet number. Only the group of 30 ramets of C. curvula on alien soil could not recover from the transplantation shock. Within the pots, C. rosae showed a greater ramet turnover and a higher increase in ramets than C. curvula. On their native substrate, both species had a significantly higher ramet increase than on the alien substrate. Ramet growth was found to be density dependent for both species, the increases recorded for the groups of 10 being significantly greater than for the groups of 30. Although C. curvula produced fewer ramets than C. rosae, the aboveground dry weight of the former was significantly higher. This may be decisive for its greater competitive success in closed grasslands.

    Notes: Abstract. Laser-deposited metallic alloys and multilayers were studied in detail by a combination of high-resolution ex situ and time-resolved in situ experiments. The purpose of these experiments is to better understand the special properties of laser-deposited metallic films in comparison with conventionally prepared thin films. During deposition, thickness, resistance, and electron diffraction (THEED) experiments show that the film surface is resputtered, local mixing at the interfaces of multilayers on a nanometre scale occurs, and metastable phases up to large film thicknesses are formed. After deposition, a compressive stress of 1–2 GPa was measured using four-circle diffractometry, and growth defects were observed on an atomic scale by electron microscopy (HRTEM) and field ion microscopy (FIM). The obtained structural details of the metallic films can be explained by an implantation model for the laser deposition process.
