    Notes: Abstract An alternative to open heart surgery in treating arterial diseases causing restricted blood flow is the implantation of intracoronary metallic stents. In spite of the advances in implantation and in spite of the excellent mechanical properties of metallic stents, there are still limitations because of the thrombogenicity of the metal. We have, hence, directed our attention to the coating of metallic stents with an ultrathin polymer layer by chemical vapor deposition (CVD) polymerization of 2-chloroparacyclophan. In a second step of surface modification the poly(2-chloroparaxylylene) layer is modified by treatment with a sulfur dioxide plasma in order to obtain a more hydrophilic surface with new functional groups. The results demonstrate the stable polymer coating of the stents and the improvement of haemocompatibility after treatment with sulfur dioxide plasma. Platelet adhesion is decreased from 85% for the metal surface to 20% for the CVD-coated and sulfur-dioxide-plasma treated surface. © 1999 Kluwer Academic Publishers

    Notes: Abstract  Using RFLP markers, QTLs for tuber starch-content and tuber yield were mapped in two F1 populations derived from crossing non-inbred di-haploid potato breeding lines. QTLs were identified and mapped, based on both single-marker tests and interval analyses. A model specifically developed for interval QTL analysis in non-inbred plant species was successfully applied for the first time to experimental data. Results of both methods of QTL analysis were similar but not identical. QTLs for tuber starch-content and tuber yield were analysed in segregating populations K31 and LH in five and two environments, respectively. Population K31 was fully genotyped whereas population LH was selectively genotyped according to high and low tuber-starch content. Eighteen putative QTLs for tuber starch-content were identified on all 12 potato linkage groups and eight putative QTLs for tuber yield were identified on eight linkage groups. Twenty of twenty six putative QTLs were reproducibly detected in at least two environments and/or mapping populations. Few major QTLs for tuber starch-content were highly stable across environments but were detected in only one of the two mapping populations analysed. Most QTLs for tuber yield were linked with QTLs for tuber starch-content suggesting that the effects on both traits are controlled by the same genetic factors. The results are discussed with respect to marker-assisted selection in potato.

    Notes: Abstract  A yeast artificial chromosome (YAC) library was constructed from high-molecular-weight DNA of potato (Solanum tuberosum). Potato DNA fragments obtained after complete digestion with four different rare-cutter restriction enzymes were cloned using the pYAC-RC vector. The library consists of 21 408 YAC clones with an average insertion size of 140 kb. The frequency of YAC clones having insertions of chloroplast or mitochondrial DNA was estimated to be 0.5% and 0.3%, respectively. The YAC library was screened by PCR with 11 DNA markers detecting single genes or small gene families in the potato genome. YACs for 8 of the 11 markers were detected in the library. Using 2 markers that are linked to the resistance genes R1 and Gro1 of potato, we isolated two individual YAC clones. One of these YAC clones was found to harbour one member of a small family of candidate genes for the nematode resistance gene Gro1.

    Notes: Abstract Applying Green's continuum theory of a slender body, the process of liquid jet break-up is analysed for a viscoelastic upper-convected Jeffreys fluid. In contrast to a Newtonian liquid an enforced growth of the perturbation is received from a linear analysis. A non-linear numerical analysis shows the viscosity-dependent filament formation between growing droplets of the viscoelastic liquid. The radius of these filaments decreases in an uniaxial extensional flow.

    Notes: Abstract The aim of our investigations was to elucidate the effects of acid precipitation on some enzymes of the primary metabolism of ectomycorrhizas. Mycorrhizas of the type of Piceirhiza nigra Gronbach and of Russula ochroleuca (Pers.) Fr. and Tuber puberulum Berk. and Br. were collected from a stand of Norway spruce (Picea abies [L.] Karst.) during the growing seasons of 1991 and 1992. The experimental plots had been limed (Ca: 22 kmol ha-1, Mg: 20 kmol ha-1) in 1984 and exposed to acid irrigation (pH 2.7–2.8, H2SO4: 2 kmol ha-1 a-1) from 1984 to 1990. Crude extracts of mycorrhizas were assayed for the activities of glucose-6-phosphate dehydrogenase (G6P-DH, EC 1.1.1.49), 6-phosphogluconate dehydrogenase (EC 1.1.1.44), NADP-dependent isocitrate dehydrogenase (EC 1.1.1.42) and NAD-dependent glutamate dehydrogenase (EC 1.4.1.2). The influence of the experimental treatments on these enzyme activities of the primary metabolism was generally low. For P. nigra, the activity of G6P-DH was decreased on the irrigated plot (photometric determinations). This seems to be a selective effect on the fungal partner, since quantitative enzyme electrophoresis revealed a decrease in the percentage of the fungal enzyme activity in relation to the total enzyme activity, whereas the content of the fungal compound ergosterol was not affected. A decrease in the fungal G6P-DH activity could also be detected in mycorrhizas of Tuber puberulum. There was also a seasonal variation in the proportion of fungal activity of G6P-DH in relation to the total G6P-DH activity. In the photometric assay (total activity) the effect was not discernible. This is indicative of a degree of regulation between the two partners, which could only be detected by quantitative enzyme electrophoresis. In addition, it could be deduced from the electrophoretograms, that in the case of G6P-DH and 6PG-DH the fungal enzyme activity was dominating in all mycorrhizas studied whereas in the case of ICA-DH the fungal band varied from being conspicuous to absent in different species of mycorrhizas. The banding pattern of G6P-DH was reproducibly different for all investigated species of mycorrhizas.

The disadvantage of PES-membranes is their hydrophobic character, which in contact with protein containing solutions leads to high protein adsorption and as a consequence to deterioration of membrane properties. In this report the surface modification of PES by means of glow discharge induced grafting of 2-hydroxyethyl methacrylate (HEMA) is described. Graft polymerisation creates a largely wettable layer of poly(2-hydroxyethyl methacrylate) (PHEMA) on the surface of PES. This has been shown by contact angle measurements using the Wilhelmy plate method. Chemical characterization is carried out by means of X-ray photoelectron spectroscopy (XPS) and infrared spectroscopy/attentuated total reflection (FTIR-ATR). The influence of storage conditions on the surface properties of modified PES samples has been investigated after storage in vacuum, water, and air.
