    Notes: Summary With a self-neutralizing, rocket-borne parachute aspiration probe concentration and mobility measurements of positively and negatively charged carriers were made between 72 and 40 km over Sardinia. Between 72 km and 60 km a ‘heavy’ and a ‘light’ group of positively charged particles were found. The ‘heavy’ group was more abundant than the ‘hight’ group in the greater heights. For the negatively charged particles only one mobility group was found which roughly corresponded to the mobility of the ‘light’ group of the positively charged carriers. The probe was designed so that no electrons could enter the analyzer section of the aspiration system. By this the electron concentration was given by the difference in concentration between the positively and negatively charged particles.

    Notes: Abstract In order to investigate linkage, we used serum allotypes of the two rabbit C κ isotypes and restriction fragment length polymorphisms (RFLPs) of the genes for V κ, C κ, and T-cell receptor C β . The inheritance of these genetic markers was studied through backcross and F2 matings. Southern analysis and hybridization of genomic DNA with a Cκ probe detected a 5 kb Pst I fragment linked to expression of the K2bas1 allotype and the presence of the κ1b bas gene and a 6.6 kb Pst I fragment linked to the expression of the K1b9 allotype, the presence of the κ2 bas2 gene and lack of expression of the K2bas1 allotype. A Vκ probe detected a 1.3 kb Eco RI fragment linked to the presence of the κ1b bas gene and expression of the K2bas1 allotype. In contrast, the 9 or 14 kb Eco RI RFLP (C β a or C β b) detected with a Tcr β chain probe segregated independently from Cκ allotypes and RFLPs. It has previously been found that Cκ and C β are also unlinked in man, whereas in the mouse they are linked at a distance of ≈8 centimorgans.

    Notes: Abstract We report the sequence of a cDNA encoding a rabbit immunoglobulin γ heavy chain of d12 and e14 allotypes with high homology to partial cDNA sequences from rabbits of d11 and e15 allotypes. The encoded rabbit protein shows homologies with human (68–70%) and mouse (60–63%) γ chains. The nucleotide sequence homologies of the CH domains range from 76–84% with human and 64–76% with mouse sequences. Comparison of the portion of VH encoding amino acid positions 34–112 with a previously determined VH sequence of the same allotype shows high conservation of sequences in the second and third framework segments but more marked differences both in length and encoded amino acids of the second and third complementarity-determining regions (CDRs). We also found a high degree of homology with a human genomic V-region, VH26 (77%) and a remarkable similarity between rabbit and human second CDR sequences and human genomic D minigenes. These results provide additional evidence that D minigene sequences share information with the CDR2 portion of VH regions.

    Notes: Abstract In contrast to rabbits of b4, b5, b6, and b9 allotypes whose serum immunoglobulins (Igs) are predominantly composed of kappa-type light chains, rabbits of the mutant Basilea strain have serum Igs that are largely of lambda type. We prepared several antisera that recognized a minor K2 (bas) light chain that is produced by Basilea rabbits. With these antisera we identified the K2 (bas) isotype in the serum of the original b 9/b 9 male rabbit whose offspring displayed the Basilea mutant phenotype. It was present in one half of his nonmutant offspring which inherited b9 from him and another b allotype from their mothers. Breeding was conducted both in Basel and at the NIH to develop and maintain colonies of mutant Basilea strain rabbits. The data obtained during colony development confirm that the trait of expression of the bas allotype maps to the same genetic region (b locus) that is known to control the allelic b allotypes b4, b5, b6 and b9. Homozygotes or heterozygotes of b4, b5 or b6 allotype (b b /b b ) were mated with homozygous b bas /b bas rabbits to produce F1s, and then F2s as well as progeny of backcrosses to both homozygous parental types (b b /b b and b bas /b bas ) were produced. The bas allotype segregates as an allele (or pseudoallele) at the b locus although there was a deficiency in recovery of homozygous bas offspring in both the F2 and backcross matings to b bas /b bas parental type in the NIH colony. This selective deficiency may reflect a deleterious effect on survival of homozygous bas progeny.

    Notes: Abstract We previously reported that some rabbits have three different copies of T-cell receptor b (Tcrb) constant region genes unlike man, mice, and rats who generally have two copies. Two of theseCß genes were found on an ≈ 14 kilobase (kb) and one on an ≈ 6 kbEco RI fragment. The gene on the 6 kb fragment is of ß2 type. A previously described portion of the 14 kb fragment appeared to have sequences characteristic ofCß1. We have now shown that the 6 kb fragment is adjacent to and 3′ of the 14 kb fragment. Furthermore, the second linked sequence ofCß gene present on the 14 kb fragment resembles to a large extent theCß2 gene present on the 6 kb fragment. Moreover, this secondCß gene has a 5′ cluster ofJß sequences resemblingJß2 of other species. However, exon 4 and the 3′ unranslated region (3′UT) are of the ß1 type. Mapping studies using southern analyses of both genomic DNA and the 14 kb clone have identified another cluster ofJß2 sequences 5′ of the third tandemCß2 gene present on the 6 kbEco RI fragment. Thus, the second gene on the 14 kb fragment appears to be a chimeric genomicTcrb gene that may have arisen by an unequal crossing-over event analogous to that which may have deletedCß1,Dß2, andJß2 in NZW mice.

    Notes: Abstract To identify possible genetic factors affecting human longevity we compared allele pools at two candidate loci for longevity between a sample of 143 centenarians (S) and a control sample of 158 individuals (C). The candidate loci were APOB and TPO, which code for apolipoprotein B and thyroid peroxidase, respectively. Both restriction fragment length (RFL) (XbaI2488 and EcoRI4154) and variable number of tandem repeat (VNTR) (3′APOB-VNTR) polymorphisms were analysed at the APOB locus; the TPO-VNTR polymorphism (intron 10) was analysed at the TPO locus. The main result of the investigation was that there is an association between the APOB locus and longevity that is revealed only when multiallelic polymorphisms are considered. In particular: (i) the frequency of 3′APOB-VNTR alleles with fewer than 35 repeats is significantly lower in cases than in controls; (ii) the linkage disequilibrium between the XbaI-RFLP and the EcoRI-RFLP is significantly different from 0 in cases but not in controls; (iii) the EcoRI-RFLP and XbaI-RFLP allele frequencies do not discriminate between cases and controls. The differences observed between case and control allele pools are specific to the APOB locus, since no significant difference was observed at the TPO locus.

    Notes: Abstract This report summarizes our current understanding of the heavy chain haplotypes found in our laboratories' rabbits. Independently derived data from several laboratories have been synthesized into a consistent picture of the linked inheritance of allotypic markers found on the different heavy chain classes and subclasses of rabbit immunoglobulins in pedigreed rabbits, including the families of three apparentVH-CH recombinants. In one recombinant, the entire group ofCH markers (Cμ, Cγ, and Cα) recombined with the set ofVH. Although in the other two recombinants all CH markers may also have recombined as a group, in one of these only IgG and IgACH genes were informative; in the other recombinant, only the IgG allotypes were informative. Some allotypic determinants found on IgM molecules (“conformational”) appear only when a specific variable region allotype (VHa) is combined with a specific μ constant region allotype (Cμ). New combinations ofVHa and Cμ allotypes were generated in two of the genetic recombinants and led to new “conformational” determinants. The gains and losses observed lend support to the hypothesis that the determinants result from conformations generated by the combination of allotype-specificVH and Cμ protein sequences. Conceivably, DNA events that joinVH to diversity (D)- and joining (J)-coding sequences or mRNA processing events that splice J to Cμ could be involved in generating the sequences that form allotype-specific determinants.

    Description / Table of Contents: Zusammenfassung Um einen natürlichen, von Kodierungen unabhängigen Vergleich zwischen primitiv-rekursiven Funktionen und Automatentransduktionen zu ermöglichen, werden die Grzegorczyk-Hierarchie, die Rekursionszahl-Hierarchie und die Loop-Hierarchie von arithmetischen auf Wortfunktionen verallgemeinert. Dabei ergeben sich einige Unterschiede zum arithmetischen Fall. Unter Benutzung von Turingmaschinen und verallgemeinerten endlichen Automaten werden alle Inklusionsprobleme der Funktionenklassen dieser Hierarchien gelöst. Von Automaten definierte Funktions- und Sprachklassen werden innerhalb dieser Hierarchien klassifiziert. Außerdem werden primitiv-rekursive Transformationen zwischen verschiedenen Monoiden behandelt.
    Notes: Abstract In order to compare primitive recursive functions and transductions defined by automata in a natural way independent of encodings, we generalize the Grzegorczyk hierarchy, the recursion number hierarchy and the loop hierarchy from arithmetical to wordfunctions. We observe several differences between the arithmetical and the non-arithmetical theory. By means of turingmachines and generalized sequential machines all inclusion problems for the function classes of these hierarchies are solved. Transductions and languages defined by automata are classified within these hierarchies. Moreover, we introduce and study primitive recursive transformations between different monoids.
