    Notes: During screening for cyclodextrin-forming microorganisms, an alkalophilic Bacillus sp, which produced high activity of cyclodextrin glycosyltransferase, was isolated and identified as Bacillus firmus. The crude enzyme transformed starch to mainly β-and γ-cyclodextrin. The purified enzyme had an optimum pH of 7.5–8.5 and its optimum temperature was 65°C, which is the highest optimum temperature as compared to other cyclodextrin glycosyltransferases except that produced by Bacillus amyloliquefaciens.

    Notes: Abstract Archived data, together with field observations collected between 1980 and 1982 off the west coast of Korea, suggest that the onset of winter monsoon winds in late fall initiates a residual southward flow that potentially carries large volumes of resuspended bottom sediments into the Korea Strait. During the calm conditions of summer, muds are replenished by high river discharges and reform the band of soft material, which characteristically occurs as a series of mudflats near the coast. Thus, these mudflats serve as a temporary storage facility during summer accumulation and as a source during winter erosion.

    Notes: Abstract Clay mineral studies by XRD analyses of eleven surficial mud samples from the central and southeastern Yellow Sea reveal that clay minerals in all sediment samples are quite similar, except for the presence of smectite in the central Yellow Sea samples. Smectite, thus, is suggested to the significant clay mineral in distinguishing the finegrained sediment sources: the Huanghe River-derived muds contain smectite whereas smectite hardly exists in those from the Keum River.

    Notes: Abstract The human granulocyte colony-stimulating factor (hG-CSF) was efficiently secreted at high levels in fed-batch cultures of recombinant Saccharomyces cerevisiae. However, the secreted recombinant hG-CSF (rhG-CSF) was shown to exist as large multimers in the culture broth due to strong hydrophobic interaction. It was hardly monomerized even by urea at high concentration. This multimer has been reported to diminish specific receptor-binding activity of hG-CSF and causes undesirable problems in the downstream process. When the rhG-CSF was secreted to extracellular broth in the presence of a non-ionic surfactant (Tween 80) in the culture media, the multimerization of the secreted rhG-CSF was efficiently prevented in the fed-batch cultures. Also, the monomer fraction and secretion efficiency of rhG-CSF were significantly increased at the higher culture pH (6.5). Without using any denaturing agents, the secreted rhG-CSF monomer was easily purified with high recovery yield and purity via a simple purification process under acidic conditions, consisting of diafiltration, cation exchange, and gel filtration chromatography. A lyophilization process devoid of intermonomer aggregation was also designed using effective stabilizing agents.

    Notes: Abstract A strain of Aspergillus niger isolated from sugarcane fields, produced an extracellular transfructosylase in the culture medium. Sucrose and raffinose induced the production to the enzyme, which was purified by 138-fold. The optimum pH for activity and stability were 5.5 and 6.5, respectively. Its optimum temperature was 55°C. The enzyme hydrolysed sucrose rapidly and simultaneously formed fructooligosaccharides by transfructosylation.

    Notes: We investigated the influence of inorganic phosphate concentration on the production of curdlan by Agrobacterium species. A two-step culture method was employed where cells were first cultured, followed by curdlan production under nitrogen-limiting conditions. In the curdlan production step, cells did not grow but metabolized sugar into curdlan. Shake-flask experiments showed that the optimal phosphate concentration for curdlan production was in the range of 0.1–0.3 g l−1. As the cell concentration increased from 0.42 to 1.68 g l−1 in shake-flask cultures, curdlan production increased from 0.44 to 2.80 g l−1. However, the optimal phosphate concentration range was not dependent upon cell concentration. The specific production rate was about 70 mg curdlan g-cell−1 h−1 irrespective of cell concentration. When the phosphate concentration was maintained at 0.5 g l−1 under nitrogen-limiting conditions, as high as 65 g l−1 of curdlan was obtained in 120 h.     Notes: Maltose and sucrose were efficient carbon sources for the production of curdlan by a strain of Agrobacterium sp. A two-step, fed-batch operation was designed in which biomass was first produced, followed by curdlan production which was stimulated by nitrogen limitation. There exists an optimal timing for nitrogen limitation for curdlan production in the two-step, fed-batch operation. Maximum curdlan production (60 g L−1) was obtained from sucrose with a productivity of 0.2 g L−1 h−1 when nitrogen was limited at a cell concentration of 16.0 g L−1. It was also noted that the curdlan yield from sucrose was as high as 0.45 g curdlan g−1 sucrose, and the highest specific production rate was 1.0 g curdlan g−1 cells h−1 right after nitrogen limitation. Of particular importance was the use of molasses as a cheap carbon source to produce curdlan in the two-step, fed-batch cultivation. As high as 42 g L−1 of curdlan with a yield of 0.35 g curdlan g−1 total sugar was obtained after 120 h of fed-batch cultivation.

    Notes: Synthesis of human mini-proinsulin precursors was investigated in controlled fed-batch cultures at high cell concentrations of recombinant Escherichia coli. Transcription of the recombinant gene was controlled by a T7 promoter system. The human mini-proinsulin was prepared by substituting a C-chain peptide of natural proinsulin with a peptide sequence of only nine amino acids. The reduced size of fusion proinsulin and hence the increased purity of human insulin in the recombinant product may contribute to increasing the fermentation yield of human insulin. Three precursors (T1-, T2-, and T3-M2PI) were constructed by utilizing the N-terminus residues of human tumor necrosis factor α as fusion partners. The T2 precursor was most soluble in the cytoplasm, and exerted the most inhibitory effect on recombinant cell growth. In the production of T2-M2PI, significant amounts of undesirable metabolic by-products (acetate and ammonia) accumulated in the culture broth even at very low specific cell growth rate. The major portion of all synthesized precursors aggregated to insoluble inclusion bodies but the protein aggregates were easily converted to monomers in the presence of the anionic detergent (SDS) without using any reducing agent. With the expression of T1-M2PI, growth inhibition was minimal, and the maximum volumetric yield of mini-proinsulin (M2PI) in fermentation cultures was at the highest level among the synthesized precursors.
