    Notes: Summary Intracellular ATP-dependent Ca2+ sequestration mechanisms were studied in isolated dispersed rat pancreatic acini following treatment with saponin or digitonin to disrupt their plasma membranes. In the presence of45Ca2+ concentrations 〈10−6 mol/liter, addition of 5 mmol/liter ATP caused a rapid increase in45Ca2+ uptake exceeding the control by fivefold. ADP mimicked the ATP effect by 50 to 60%, whereas other nucleotides such as AMP-PNP, AMP-PCP, CTP, UTP, ITP, GTP, cAMP and cGMP did not. Maximal ATP-promoted Ca2+ uptake was obtained at 10−5 mol/liter Ca2+ uptake by mitochondrial inhibitors was dependent on the Ca2+ concentration, indicating the presence of different Ca2+ storage systems. Whereas the apparent half-saturation constant found for mitochondrial Ca2+ uptake was ∼4.5×10−7 mol/liter, in the presence of antimycin and oligomycin (nonmitochondrial uptake) it was ∼1.4×10−8 mol/liter. In the absence of Mg2+ both ATP- and ADP-promoted Ca2+ uptake was nearly abolished. The Ca2+ ionophore and mersalyl blocked Ca2+ uptake. Electron microscopy showed electrondense precipitates in the rough endoplasmic reticulum of saponintreated cells in the presence of Ca2+, oxalate and ATP, which were absent in intact cells and in saponin-cells without ATP or pretreated with A23187. The data suggest the presence of mitochondrial and nonmitochondrial ATP-dependent Ca2+ storage systems in pancreatic acini. The latter is likely to be located in the rough endoplasmic reticulum.

    Notes: Summary We have previously shown that inositol-1,4,5-trisphosphate (IP3) releases Ca2+ from an intracellular calcium store in permeabilized acinar cells of rat pancreas (H. Streb et al., 1983,Nature (London) 306:67–69). This observation suggests that IP3 might provide the missing link between activation of the muscarinic receptor and Ca2+ release from intracellular stores during stimulation. In order to localize the intracellular IP3-sensitive calcium pool, IP3-induced Ca2+ release was measured in isolated subcellular fractions. A total homogenate was prepared from acinar cells which had been isolated by a collagenase digestion method. Endoplasmic reticulum was separated from mitochondria, zymogen granules and nuclei by differential centrifugation. Plasma membranes and endoplasmic reticulum were separated by centrifugation on a sucrose step gradient or by precipitation with high concentrations of MgCl2. IP3-induced Ca2+ release per mg protein in the total homogenate was the same as in leaky cells and was sufficiently stable to make short separation procedures possible. In fractions obtained by either differential centrifugation at 7000×g, sucrose-density centrifugation, or MgCl2 precipitation there was a close correlation of IP3-induced Ca2+ release with the endoplasmic reticulum markers ribonucleic acid (r=0.96, 1.00, 0.91, respectively) and NADPH cytochromec reductase (r=0.63, 0.98, 090, respectively). In contrast, there was a clear negative correlation with the mitochondrial markers cytochromec oxidase (r=−0.64) and glutamate dehydrogenase (r=−0.75) and with the plasma membrane markers (Na++K+)-ATPase (r=−0.81) and alkaline phosphatase (r=−0.77) in all fractions analyzed. IP3-induced Ca2+ release was distributed independently of zymogen granule or nuclei content of the fractions as assessed by electron microscopy. The data suggest that inositol-1,4,5-trisphosphate releases Ca2+ from endoplasmic reticulum in pancreatic acinar cells.

    Notes: Summary ATP-dependent Ca2+ uptake into isolated pancreatic acinar cells with permeabilized plasma membranes, as well as into isolated endoplasmic reticulum prepared from these cells, was measured using a Ca2+-specific electrode and45Ca2+. Endoplasmic reticulum was purified on an isopycnic Percoll gradient and characterized by marker enzyme distribution. When compared to the total homogenate, the typical marker for the rough endoplasmic reticulum RNA was enriched threefold and the typical marker for the plasma membrane Na+,K+(Mg2+)ATPase was decreased 20-fold. When different fractions of the Percoll gradient were compared,45Ca2+ uptake correlated with the RNA content and not with the Na+,K+(Mg2+)ATPase activity. The characteristics of nonmitochondrial Ca2+ uptake into leaky isolated cells and45Ca2+ uptake into isolated endoplasmic reticulum were very similar: Calcium uptake was maximal at 0.3 and 0.2 mmol/liter free Mg2+, at 1 and 1 mmol/liter ATP, at pH 6.0 and 6.5, and free Ca2+ concentration of 2 and 2 μmol/liter, respectively. Calcium uptake decreased at higher free Ca2+ concentration.45Ca2+ uptake was dependent on monovalent cations (Rb+〉K+〉Na+〉Li+〉choline+) and different anions (Cl−〉Br−〉SO 4 2− 〉NO 3 − 〉I−〉cyclamate−〉SCN−) in both preparations. Twenty mmol/liter oxalate enhanced45Ca2+ uptake in permeabilized cells 10-fold and in vesicles of endoplasmic reticulum, fivefold. Calcium oxalate precipitates in the endoplasmic reticulum of both preparations could be demonstrated by electron microscopy. The nonmitochondrial Ca2+ pool in permeabilized cells characterized in this study has been previously shown to regulate the cytosolic free Ca2+ concentration to 0.4 μmol/liter. Our results provide firm evidence that the endoplasmic reticulum plays an important role in the regulation of the cytosolic free Ca2+ concentration in pancreatic acinar cells.

    Description / Table of Contents: Zusammenfassung Es wurden die Genauigkeiten der Röntgenfluorescenzanalyse von Sinterproben der Bleiverhüttung nach Herstellung der Untersuchungsproben durch Schmelzaufschluß mit Kaliumdisulfat und nachfolgendem Tablettieren des gemahlenen Schmelzproduktes sowie durch direktes Tablettieren des feinstgemahlenen Probengutes untersucht. Dabei wurde festgestellt, daß beide Probenpräparationsverfahren etwa die gleiche Genauigkeit aufweisen, gute Ergebnisse mit relativen Standardabweichungen kleiner als 1% für die Bestandteile außer für Schwefel aber nur erhalten werden, wenn eine rechnerische Matrixkorrektur vorgenommen wird. Da der Zeitaufwand für das Pulverpreßverfahren nur halb so hoch ist wie für das Schmelzaufschlußverfahren, ist dieses für die Betriebsanalyse besonders geeignet.
    Notes: Summary The accuracy of X-ray fluorence analysis of sinters for lead smelting has been investigated. The samples were prepared by smelting the sample with potassium disulphate and pelleting the smelt, and by direct pelleting the pulverized test material. It was found that both preparation procedures showed nearly the same accuracy. But good results with relative standard deviations smaller than 1% for all elements except sulphur were only achieved by calculated matrix correction. Because the time needed for direct pelleting is only half as high as for the smelting reaction, the former method is most suitable for process control.

    Notes: Summary In an attempt to identify a cellular Ca2+-pool, from which calcium is released when secretagogues are applied, tissue fragments of the rat exocrine pancreas were incubated and fixed with glutaraldehyde in the presence of calcium. By means of this procedure electron-dense deposits were found on plasma membranes. X-ray microanalysis showed that these deposits contain calcium. Stimulation of tissue fragments with the use of the secretagogues carbachol or cholecystokinin reduced the number of deposits by about 80%. When the antagonist atropine was applied after carbachol stimulation, deposits reappeared on cell membranes, which then disappeared again after a second stimulation with cholecystokinin. In the presence of procaine, carbachol was inhibited and only slightly reduced the Ca2+-deposits on the plasma membranes. These results suggest that a calcium pool, from which calcium is released to induce enzyme secretion on stimulation, is located in the cell membrane
