    Notes: Summary Segments of mitochondrial DNA (mtDNA) carrying the gene for the α-subunit of F1-ATPase (atpA) were detected by Southern hybridization with atpA from pea as probe. In the case of Nicotiana langsdorffii, we identified four fragments that are derived from combinations of two different 5′ and two different 3′ flanking regions of atpA. All four types share the coding region, suggesting that they result from homologous recombination in the coding region of atpA. By contrast, N. glauca generated only one analogous fragment, which indicated the existence of only a single type of atpA in N. glauca. In the case of somatic hybrids obtained by fusion between protoplasts from N. langsdorffii and N. glauca, analysis with EcoRI or HindIII detected three new fragments in addition to the parental fragments. These new fragments can be explained by homologous recombination within the coding region of atpA. Our results show that the coding region of atpA is involved not only in intragenomic homologous recombination but can also be involved in homologous recombination between two parental mitochondrial genomes of somatic hybrids.

    Notes: Abstract Concentrations of four essential elements (Fe, Mn, Zn, and Cu) and two toxic metals (Cd and Hg) were measured in selected tissues of 19 pelagic seabird species collected in the North Pacific and neighboring waters. Essential metal concentrations were generally highest in the liver and less variable than toxic metals among species and also within each species. Fe concentrations in the muscle were higher in Alcidae than in the other families, whereas the opposite trend was found for Fe and Mn in the liver. Zn concentrations varied among species, depending on the Cd concentrations. On the other hand, toxic metal concentrations were highest in the liver or kidney and varied widely among species, greatly depending on differences in the diet among species. Extraordinarily high Hg concentrations were found in Black-footed Albatrosses,Diomedea nigripes, exceeding 300 μg/g wet weight in some, and seemed to be due to constraints on the elimination of Hg. Also, some geographical differences in Cd and Hg concentrations of the seabirds were observed. The concentrations of Cd and Hg reported here, however, seem to be natural rather than due to environmental pollution.

    Notes: Summary In acetic acid fermentation, the number of viable cells decrease as the acetic acid concentration increases to more than about 40 g/l, which means that the productivity attainable by conventional fed-batch and repeated fed-batch operations using one fermentor is limited. In this paper, based on a fed-batch experiment using Acetobacter aceti 2096, a mathematical model was developed. The optimization carried out showed the superiority of repeated fed-batch operation using two fermentors. The performance evaluation was made with respect to productivity and product concentration. It was shown to be attractive in practice to use multiple fermentors, in particular for high product concentrations. Experiments were then conducted to ascertain the simulation results.

    Notes: Abstract Intracellular 2,4-dichlorophenoxyacetic acid (2,4-D) concentration profiles in rice (Oryza sativa L.) callus were measured for various subculture periods. 2,4-D was taken up into the callus by uphill transportation from the medium. The intracellular 2,4-D taken up was metabolized gradually during cultivation. The intracellular 2,4-D concentration markedly affected the redifferentiation process after subculture in such a way that the higher its concentration was, the more strongly the redifferentiation process was suppressed, and the lesser was the browning of the callus.

    Notes: Abstract To produce PreS2 antigen of human hepatitis B virus extra-cellularly in Bacillus subtilis, it was fused with the COOH-termini of B. subtilis α-amylases of 522 (Amy+), 467 (Amy+) and 443 (Amy−) amino acids, Among them, α-amylase-A467, which has 467 amino acids, was a relatively stable carrier when the cells were cultured in Luria-Bertani (LB) medium. The α-amylase-A443-PreS2 hybrid protein (Amy−) was quickly degraded. The α-amylase-A522-PreS2 hybrid was most efficiently produced when a B. subtilis transformant of a protease-super-deficient mutant was cultured in the presence of 0.5 m sodium sulphate. The production of A522-PreS2 hybrid protein under such conditions reached 5–10 mg/l and was eight, and 200–500 times higher than those obtained by the transformants of an alkaline/neutral protease-deficient mutant of B. subtilis, and a wild-type strain, in LB medium, respectively.

    Notes: Summary We have studied the pharmacokinetics of methylphenidate enantiomers after the oral administration of different doses of racemic methylphenidate to one patient with narcolepsy and to four healthy volunteers. The plasma concentrations of (+)-methylphenidate were much higher than those of (−)-methylphenidate after each dose in all subjects. In the patient the oral clearance (CL/f) of (+)-methylphenidate fell 3-fold and the area under the concentration-time curve (AUC) rose 7-fold when the dose was increased from 20 to 40 mg (from 0.27 to 0.53 mg·kg−1), in spite of the relatively constant terminal half-life of 2.6–2.7 h. Similar dose-dependency was also observed in the healthy volunteers in the dose range of 10–60 mg (0.12–0.77 mg·kg−1). The mean value of CL/f for the 40 mg dose was significantly lower than that for the 20 mg dose. The mean AUC of the (+)-isomer corrected to a dose of 10 mg increased significantly between the 20 mg and 40 mg doses. In the urine (+)- and (−)-ritalinic acid were excreted for 48 h after each dose as 32–37% and 34–40% of the dose respectively. The mean total recoveries (sum of enantiomers of methylphenidate and its metabolite, ritalinic acid) in the urine were relatively constant (63–78% of the doses), suggesting that the changes in AUC with dose may not be due to a change in the intestinal absorption of racemic methylphenidate. We conclude that the nonlinear kinetics of (+)-methylphenidate may be due to saturation of its presystemic elimination.

    Notes: Summary The effect of several forms of suspension polymerized poly(vinyl chloride) particles on mechanochemical block copolymerization in the solid poly(vinyl chloride)-styrene-sodium dodecyl sulfate solutions has been studied by ultrasonic irradiation at 60 °C. The block copolymerization of styrene was initiated by free radicals produced from the poly(vinyl chloride) particles by ultrasonic waves. The rates of copolymerization increased with increasing the additional amount of the solid poly(vinyl chloride), the porosity, and the average diameter of the grain particles. In particular, the influence of the average diameter was much larger than that of the porosity. When the porosity and the average diameter were increased, the rates of decrease in the viscosity-average degree of polymerization of the degraded poly(vinyl chloride) were much increased. In addition, the changes in the composition of the block copolymer and homopolymers in the reaction products were obtained.
