    Notes: Bank filtration and artificial ground water recharge are important, effective, and cheap techniques for surface water treatment and removal of microbes, as well as inorganic, and some organic, contaminants. Nevertheless, physical, chemical, and biological processes of the removal of impurities are not understood sufficiently. A research project titled Natural and Artificial Systems for Recharge and Infiltration attempts to provide more clarity in the processes affecting the removal of these contaminants. The project focuses on the fate and transport of selected emerging contaminants during bank filtration at two transects in Berlin, Germany. Several detections of pharmaceutically active compounds (PhACs) in ground water samples from bank filtration sites in Germany led to furthering research on the removal of these compounds during bank filtration. In this study, six PhACs including the analgesic drugs diclofenac and propyphenazone, the antiepileptic drugs carbamazepine and primidone, and the drug metabolites clofibric acid and 1-acetyl-1-methyl-2-dimethyl-oxamoyl-2-phenylhydrazide were found to leach from the contaminated streams and lakes into the ground water. These compounds were also detected at low concentrations in receiving public supply wells. Bank filtration either decreased the concentrations by dilution (e.g., for carbamazepine and primidone) and partial removal (e.g., for diclofenac), or totally removed PhACs (e.g., bezafibrate, indomethacine, antibiotics, and estrogens). Several PhACs, such as carbamazepine and especially primidone, were readily transported during bank filtration. They are thought to be good indicators for evaluating whether surface water is impacted by contamination from municipal sewage effluent or whether contamination associated with sewage effluent can be transported into ground water at ground water recharge sites.

    Notes: A reliable and sensitive analytical method for the determination of estrone (E1), 17β-estradiol (E2), and the synthetic hormone 17α-ethinylestradiol (EE2) has been established. Samples are concentrated using automated solid-phase extraction and analysis is performed by liquid chromatography with detection by tandem mass spectrometry. Recoveries of all analytes were between 93% and 107%, and limits of quantification (LOQs) between 0.1 and 0.4 ng/L for purified sewage, and surface, ground, and drinking water, and between 1 and 2 ng/L in case of raw sewage. For the investigation of estrogen release into the environment and its behavior during sewage and surface water treatment, and during ground water recharge, samples from municipal waste water treatment plants (WWTPs), a surface water treatment plant, a bank filtration site, and a ground water enrichment (GWE) pond were analyzed. El was found in the raw waste water with an average concentration of 157 ng/L, whereas E2 and EE2 were found with mean concentrations of only 18 and 9 ng/L, respectively. Sewage treatment by municipal WWTPs affected a removal of EE2 (76%), El (92%), and E2 (94%). In the investigated surface water of Berlin, only E1 could be detected at concentrations around or below 1 ng/L. E2 and EE2 were not present in the Berlin surface water above the LOQ of 0.2 ng/L, respectively. Surface water treatment also leads to a significant removal of E1 (〉 80%). In the ground water samples from the GWE site near Lake Tegel, only a few samples contained detectable concentrations of E1. These samples were collected from a shallow monitoring well located very close to the bank of the pond. Even a short distance between the bank and observation wells led to concentrations of El below the LOQ showing the potential of ground water recharge systems for the retention of estrogenic steroids depending on the environmental setting.

    Notes: The effect of different cadmium (Cd) concentrations (5, 50 and 500 μM) on growth, Cd accumulation and antioxidative systems was studied in Paxillus involutus, grown in liquid medium. Cd was rapidly accumulated by P. involutus and resulted in growth inhibition within 24 h. Antioxidative enzymes (superoxide dismutase (SOD), EC 1.15.1.1; catalase (CAT), EC 1.11.1.6; monodehydroascorbate radical reductase (MDAR), EC 1.6.5.4; dehydroascorbate reductase (DAR) glutathione reductase (GR), EC 1.8.1.7 and glutathione-dependent peroxidase (GPx), EC 1.11.1.9) were active in the investigated fungus. Furthermore, high concentrations of glutathione but no ascorbate were detected. Cd exposure resulted in a significant induction of SOD activity. However, activities of enzymes responsible for the detoxification of H2O2 showed no Cd-dependent increase or were only transiently induced (CAT, GPx) and no accumulation of H2O2 was detected. Exposure to low Cd concentrations (5 and 50 μM) caused an increase in GR, while 500 μM Cd led to an inhibition of GR and CAT. Increased glutathione concentrations were observed as a consequence of all Cd treatments. These results suggest that the antioxidative protection of the investigated strain of P. involutus was sufficient to avoid Cd-mediated oxidative stress. It is likely that this strain was able to detoxify high concentrations of Cd by transport of Cd into the vacuole because a high correlation between Cd and sulphur in the vacuole was detected by EDX.

    Notes: Carbon catabolite repression (CCR) in bacteria is generally regarded as a regulatory mechanism to ensure sequential utilization of carbohydrates. Selection of the carbon sources is mainly made at the level of carbohydrate-specific induction. Since virtually all carbohydrate catabolic genes or operons are regulated by specific control proteins and require inducers for high level expression, direct control of the activity of regulators or control of inducer formation is an efficient measure to keep them silent. By these mechanisms, bacteria are able to establish a hierarchy of sugar utilization. In addition to the control of induction processes by CCR, bacteria have developed global transcriptional regulation circuits, in which pleiotropic regulators are activated. These global control proteins, the catabolite gene activator protein (CAP), also known as cAMP receptor protein, in Escherichia coli or the catabolite control protein (CcpA) in Gram-positive bacteria with low GC content, act upon a large number of catabolic genes/operons. Since practically any carbon source is able to trigger global transcriptional control, expression of sugar utilization genes is restricted even in the sole presence of their cognate substrates. Consequently, CAP- or CcpA-dependent catabolite repression serves as an autoregulatory device to keep sugar utilization at a certain level rather than to establish preferential utilization of certain carbon sources. Together with other autoregulatory mechanisms that are not acting at the gene expression level, CCR helps bacteria to adjust sugar utilization to their metabolic capacities. Therefore, catabolic/metabolic balance would perhaps better describe the physiological role of this regulatory network than the term catabolite repression.

    Notes: . Besnoitia besnoiti is an economically important tissue cyst-forming apicomplexan of cattle in Africa and Israel. Tissue cysts and bradyzoites of B. besnoiti from the skin of a naturally infected bull were studied by transmission electron microscopy. Tissue cysts enclosed host cell and bradyzoites. Bradyzoites were 6–7. 5 × 1. 9–2. 3 μm in size and contained organelles found in coccidian merozoites including numerous micronemes, rhoptries, amylopectin granules, and a posteriorly located nucleus. Enigmatic bodies, characteristically found in Besnoitia sp. bradyzoites, were not observed. Therefore, enigmatic bodies should be removed as a generic character of the bradyzoites of Besnoitia species.

    Notes: Neisseria gonorrhoeae (gonococci, GC) are phagocytosed by neutrophils through the interaction between opacity proteins (Opa) and the CEA (CD66) family of antigens. In order to study this interaction, we used the human myeloid leukemia HL60 cell line, which differentiates into granulocyte-like cells upon treatment with dimethylsulfoxide (DMSO) or retinoic acid (RA). We found that RA-, but not DMSO- or untreated-HL60 cells, can phagocytose OpaI-expressing gonococci as well as Escherichia coli. The interaction of OpaI E. coli with RA-treated HL60 cells was inhibited by antibodies against CEACAM1. Phagocytosis of OpaI E. coli was found to be a result of the expression of CEACAM1 in RA-treated HL60 cells. Our results indicate that the level of expression of CEACAM1 in HL60 cells can be regulated by treatment with RA in a differentiation-dependent manner, and that this is important for phagocytosis of OpaI-expressing gonococci or E. coli.
