    Description: Microsatellite enriched library of Hyriopsis cumingii was constructed according to the strong affinity between biotin and streptavidin. One hundred clones with foreign inserts were sequenced and 65 clones were screened for usable microsatellites, of which 36 were deemed unique, of sufficient length (more than 8 repeats) and possessed adequate flanking regions for primer designment. Among 36 primer pairs designed, 25 yielded scorable amplification products. Upon testing 30 individuals were sampled using the 25 pair primers from Dongting Lake of Hunan Province, China. Then thirteen polymorphic microsatellite markers were isolated and characterized. These loci exhibited high levels of genetic polymorphism, so the observed number of alleles per locus ranged from 4 to 9. The ranges of observed and expected heterozygosity were 0.2543 to 0.8913 and 0.3629 to 0.8217, respectively, and the average polymorphic information content was 0.5198. Two microsatellite loci were significantly deviated from Hardy-Weinberg equilibrium due to the presence of null alleles, and no linkage disequilibrium found. These microsatellite loci will be useful for assessing the genetic diversity and population structure of H. cumingii.

    Notes: Abstract This paper presents some test and analysis results for a spot welded joint subjected to tensile and alternate load. The effect of sheet rigidity on the tensile strength and fatigue life of the spot welded joint is studied by using the stress intensity factorsK I,K II,K III and an “effective stress intensity factor” Kϕθmax calculated by the finite element method for crack around the nugget. The results show that the “effective stress intensity factor” Kϕθmax is an essential parameter for estimating the fatigue life of the spot welded joint.

    Notes: Abstract Hepatitis B viral core protein is present in the nucleus and cytoplasm of infected hepatocytes. There is a strong correlation between the intrahepatic distribution of core protein and the viral replication state and disease activity in patients with chronic hepatitis. To understand the role of core protein in the pathogenesis of HBV, we used a yeast two-hybrid system to search for cellular proteins interacting with the carboxyl terminus of core protein, as this region is involved in a number of important functions in the viral replication cycle including RNA packaging and DNA synthesis. A cDNA encoding the extreme C-terminal region of human actin-binding protein, ABP-276/278, was identified. This interaction was further confirmed both in vitro and in vivo. In addition, the extreme C-terminal region of ABP-276/278 interacted with the nearly full-length HBV core protein. Since this region is present in both the core and the precore proteins, it is likely that both core and precore proteins of HBV can interact with the C-terminal region of ABP-276/278. The minimal region of ABP-276/278 which interacted with the HBV core protein was the C-terminal 199 amino acid residues which correspond to part of the 23rd repeat, the entire 24th repeat and the intervening hinge II region in ABPs. The potential functional outcome of ABP interaction in HBV replication and its contribution to the pathological changes seen in patients with chronic HBV infection are discussed.

    Notes: Abstract In this paper a group of stress functions has been proposed for the calculation of a crack emanating from a hole with different shape (including circular, elliptical, rectangular, or rhombic hole) by boundary collocation method. The calculation results show that they coincide very well with the existing solutions by other methods for a circular or elliptical hole with a crack in an infinite plate. At the smae time, a series of results for different holes in a finite plate has also been obtained in this paper. The proposed functions and calculation procedure can be used for a plate of a crack emanating from an arbitrary hole.

    Notes: Abstract In this paper, Muskhelishvili complex function theory and boundary collocation method are used to calculate the stress intensity factors (SIF) of a plate with two cracks emanating from an arbitrary hole. The calculated examples include a circular, elliptical, rectangular, or rhombic hole in a plate. The principle and procedure by the method is not only rather simple, but also has good accuracy. The SIF values calculated compare very favorably with the existing solutions. At the same time,the method can be used for different finite plate with two cracks emanating from a hole with more complex geometrical and loading conditions. It is an effective unified approach for this kind of fracture problems.

    Notes: A 1H NMR study of methyl, ethyl, n-propyl, isopropyl and allyl alcohols adsorbed on charcoals with various pore structures showed that the adsorption mechanism is by capillary condensation in pores with radii smaller than 30 Å, and by mono-/multi-layer adsorption on the solid surface of pores with larger radii. This mechanism is identical with that of adsorbed hydrocarbons. The 13C NMR parameters suggest that the adsorbed alcohols are preferentially oriented in the monolayer, with the methyl groups bonded to the solid surface. Longer chain alcohols are bent in capillaries with radii smaller than 10 Å. The broadening of the 13C resonances was partly eliminated in magic angle spinning experiments by using a spinning rate of 1.5-1.8 kHz.
