    Notes: Abstract Methanosarcina thermophila TM-1 produced hydrogen during growth on acetate, maintaining a concentration of approx. 0.3 μM in the culture, corresponding to a hydrogen partial pressure of approx. 40 Pa. Increasing the partial pressure of hydrogen to 250 Pa and more led to a gradually increasing inhibition of acetate metabolism. No growth was observed when the gas phase contained 2000 Pa or more and acetate metabolism did not occur even after prolonged incubation (more than 2 weeks). M. thermophila was capable of limited consumption of hydrogen. Consumption of low concentrations of hydrogen proceeded simultaneously with acetate utilization. The affinity for hydrogen (K m=5 μM) was within the range normally found for hydrogenutilizing methanogens, while the corresponding V max (1.2 μmol hydrogen consumed mg-1 · cells h-1) was orders of magnitude lower.

    Notes: Abstract A thermophilic acetate-decarboxylating methanogenic bacterium was isolated from a laboratory-scale 60°C sludge digestor. Cells form straight filaments with flat to blunted ends normally consisting of 2–3 cells held together by a sheath-like outer cell wall. The organism uses acetate, H2-CO2 and formate for methanogenesis and growth. With acetate as the sole methanogenic substrate, almost all of the radioactivity from methyl-labelled acetate appeared as methane. Acetate was converted to methane in equimolar amounts with a doubling time of 3 days.

    Notes: Abstract The physiology is described of a thermophilic acetate-decarboxylating methanogenic bacterium grown with acetate as sole energy and carbon source. The organism had a optimal pH range of 7.3–7.5 and a temperature optimum for methanogenesis near 60°C; no growth occurred above 75°C. Addition of penicillin, d-cycloserine, vancomycin and ampicillin produced no inhibition of methanogenesis at low concentrations 10–100 mg/l). No requirement for growth factors was observed during exponential growth on acetate, but sulfide was found to be necessary for optimal growth. Growth yield was 1.7–2.3 g dry weight per mol of acetate utilized. The apparent Ks obtained for methanogenesis from acetate was 0.8 m mol/l.

    Notes: A hot spring in the solfataric field of Pisciarelli (Naples – Italy) was analysed for Archaeal diversity. Total DNA was extracted from the environment, archaeal 16S rRNA genes were amplified with Archaea specific primers, and a clone library consisting of 201 clones was established. The clones were grouped in 10 different groups each representing a specific band pattern using restriction fragment length polymorphism (RFLP). Members of all 10 groups were sequenced and phylogenetically analyzed. Surprisingly, a high abundance of clones belonging to non-thermophilic Crenarchaeal clusters were detected together with the thermophilic archaeon Acidianus infernus in this thermophilic environment. Neither Sulfolobus species nor other hyperthermophilic Crenarchaeota were detected in the clone library. The relative abundance of the sequenced clones was confirmed by terminal restriction fragment analyses. Amplification of 16S rRNA genes from Archaea transferred from the surrounding environment was considered negligible because DNA from non-thermophilic Crenarchaeota incubated under conditions similar to the solfatara could not be PCR amplified after 5 min.

    Notes: One hundred and sixty-nine bacterial strains were isolated from activated sludge from a waste water treatment basin operating under alternating aerobic/anaerobic conditions. Sixteen strains from a subsample of 23 nitrogen oxide reducers were true respiratory denitrifiers, and all denitrified under both anaerobic and aerobic conditions. REP-PCR band analysis showed different patterns for all strains. One strain (strain 1) produced large amounts of N2O and was studied in detail. Nitrous oxide was the major end product of denitrification by this strain, and NO−2 was reduced more efficiently than NO−3. The aerobic denitrification was most pronounced with NO−2 as electron acceptor, and the reduction of NO−2 was not coupled to NH+4 oxidation.

    Notes: Abstract Increasing the incubation temperature of two swamp slurries from 2°C to37°C resulted in a 8- to 18-fold increase in the H2 partial pressure. The concentration of volatile fatty acids remained fairly constant except for butyrate, which decreased with increasing temperature. Calculation of Gibbs free energies of syntrophic degradation of butyrate and propionate, and of methanogenesis from acetate and H2 revealed that these reactions were exergonic after the slurries had stabilized at the incubation temperatures. The changes in H2 partial pressure and butyrate concentration with temperature were found important to render the processes exergonic within the tested temperature range.

    Notes: Summary The effects of different concentrations of Mg2+, Ca2+, or Na+ on the morphology and growth of Methanosarcina thermophila TM-1 growing on acetate at concentrations comparable with those found in anaerobic digestors was studied. At 30 mm Mg2+ or less, M. thermophila grew as large aggregates that settled rapidly. At 100 mm Mg2+ or more, the bacteria grew as single cells or a mixture of single cells and small aggregates is suspended culture. Mg2+ was necessary for growth and could not be substituted by addition of either Ca2+ or Na+. The optimal Mg2+ concentration was 30 mm and no growth was observed at 400 mm Mg2+. Cultures could be adapted to 300 mm Mg2+ without a change in growth rate. Added Ca2+ was not required for growth and had no effect on cell morphology. Inhibition by Na+ was directly related to the Mg2+ concentration. When the Mg2+ was 0.05 mm or less, 0.35 m Na+ completely inhibited growth. However, more Na+ was required for inhibition at higher Mg2+ concentrations. The same inhibitory effect of Na+ was observed when the temperature was 52°C or 45°C. The potential for disaggregation of Methanosarcina aggregates in anaerobic digestor environments was discussed.

    Notes: Abstract The effects of Cd, Cu, and Ni on pure cultures of thermophilic methanogenic bacteria were studied. The bacteria used wereMethanobacterium thermoautotrophicum and TAM, a thermophilic, acetate-decarboxylating, methanogenic bacterium. Much lower concentrations of heavy metals were needed to cause initial inhibition of TAM (1 mg/liter Cu and Cd; 5 mg/liter Ni) compared withM. thermoautotrophicum (10 mg/liter Cu and Cd; and 100 mg/liter Ni). No growth of TAM occurred at 5 mg/liter Cu and 25 mg/liter Ni, while the corresponding values forM. thermoautotrophicum were 50 mg/liter Cu and 200 mg/liter Ni. Cd (50 mg/liter) was totally inhibitory toM. thermoautotrophicum but allowed minimal growth of TAM. Ni stimulated both organisms at an optimal concentration of 5 mg/liter forM. thermoautotrophicum and 1 mg/liter for TAM. The toxicity of Cd and Cu was found to depend upon the presence of Ni in the medium.

    Description / Table of Contents: Der Zweck der vorliegenden Arbeit war, eine Entscheidung zwischen den «Mainz»- und «Keele-Theorien» über die Polymerisation der 1,3-Dioxacycloalkane durch wasserfreie Perchlorsäure in Methylendichlorid herbeizuführen. Mittels der Jaacks'schen Methode zur Bestimmung von tertiären Oxoniumionen entdeckten wir, daß deren Konzentration verschwindend klein ist, wenn unter peinlichstem Ausschluß von Feuchtigkeit polymerisiert wird. Wir erzielten diese Resultate mit 1,3-Dioxolan (1), 4-Methyl-1,3-dioxolan (4) und 1.3-Dioxepan (5). Sie wurden durch unsere Bestimmungen des Gehaltes an Hydroxy-Gruppen unterstützt, die an Polymeren vorgenomrnen wurden, welche mit verschiedenen Redgenzien neutralisiert worden waren.Es folgt hieraus. dall bei eincm Wassergehalt der Reaktionsgemische von weniger als ca. 10-4M die Fortpflanzung überwiegend durch sekundäre Oxoniumionen stattfindet. so daß sie nach dem Plesch-Westerrmann'schen Ringerweiterungsmechanismus verlaufen muß. Da Wasser die Bildung von tertiären Oxoniumionen verursacht. läßt es sich erklären, wieso andere Forscher und auch wir, zuvor in weniger sorgfältig durchgeführten Ansätzen, diese Ionen (manchmal sogar in überwiegenden Mengen) nachweisen konnten.Unsere Befunde beinhalten auch. daß Penczeks Theorie, welche coordinierte Oxy-Carbe-niumionen als die wachsenden Zentren betrachtet, auf die hier diskutierten Systeme nicht anwendbar ist.Der heuristische Wert der Kingerweiterungstheorie wird hier kurz durch einen Hinweis auf eine daraus entstandene neue Methode zur Synthese von «Kronen-Athern» aus 1,3-Dioxacykloalkanen veranschaulicht.
    Notes: The object of the work described was to discriminate between the two principal rival theories concerning the polymerisation of 1,3-dioxacycloalkanes by anhydrous perchloric acid, the “Mainz theory” and the “Keele theory”. By means of Jaacks's method for determining tertiary oxonium ions we found that in polymerisations under the driest conditions the concentration of these is negligibly small. This was done with 1,3-dioxolane (1), 4-methyl-1,3-dioxolane (4), and 1,3-dioxepane (5), and the findings are supported by determinations of the content of hydroxy groups of polymers prepared and “killed” under different conditions.It follows that when the water content of the reaction mixtures is less than ca. 10-4M the propagation is principally by secondary oxonium ions, and that it must therefore go by the ring-expansion mechanism of Plesch and Westermann.Water causes the formation of tertiary oxonium ions and this probably explains why previously other workers and ourselves had reported their presence, and sometimes their dominance, in reaction mixtures prepared under much less stringent conditions.The recent theory of Penczek, which involves co-ordinated oxy-carbenium ions, is also shown not to be applicable to the systems considered here. -  The heuristic value of the ring-expansion theory is illustrated briefly by reference to a new method of synthesising crown ethers from 1,3-dioxacycloalkanes, which arose from it.
