    Description: As a component of a three-year cooperative effort of the Washington State Department of Ecology and the National Oceanic and Atmospheric Administration, surficial sediment samples from 100 locations in southern Puget Sound were collected in 1999 to determine their relative quality based on measures of toxicity, chemical contamination, and benthic infaunal assemblage structure. The surveyencompassed an area of approximately 858 km2, ranging from East and Colvos Passages south to Oakland Bay, and including Hood Canal. Toxic responses were most severe in some of the industrialized waterways of Tacoma’s Commencement Bay. Other industrialized harbors in whichsediments induced toxic responses on smaller scales included the Port of Olympia, Oakland Bay at Shelton, Gig Harbor, Port Ludlow, and Port Gamble. Based on the methods selected for this survey, the spatial extent of toxicity for the southern Puget Sound survey area was 0% of the total survey area for amphipod survival, 5.7% for urchin fertilization, 0.2% for microbial bioluminescence, and 5-38% with the cytochrome P450 HRGS assay. Measurements of trace metals, PAHs, PCBs, chlorinated pesticides, other organic chemicals, and other characteristics of the sediments, indicated that 20 of the 100 samples collected had one or more chemical concentrations that exceededapplicable, effects-based sediment guidelines and/or Washington State standards. Chemical contamination was highest in eight samples collected in or near the industrialized waterways of Commencement Bay. Samples from the Thea Foss and Middle Waterways were primarilycontaminated with a mixture of PAHs and trace metals, whereas those from Hylebos Waterway were contaminated with chlorinated organic hydrocarbons. The remaining 12 samples with elevated chemical concentrations primarily had high levels of other chemicals, including bis(2-ethylhexyl)phthalate, benzoic acid, benzyl alcohol, and phenol. The characteristics of benthic infaunal assemblages in south Puget Sound differed considerably among locations and habitat types throughout the study area. In general, many of the small embayments and inlets throughout the studyarea had infaunal assemblages with relatively low total abundance, taxa richness, evenness, and dominance values, although total abundance values were very high in some cases, typically due to high abundance of one organism such as the polychaete Aphelochaeta sp. N1. The majority of thesamples collected from passages, outer embayments, and larger bodies of water tended to have infaunal assemblages with higher total abundance, taxa richness, evenness, and dominance values. Two samples collected in the Port of Olympia near a superfund cleanup site had no living organisms in them. A weight-of-evidence approach used to simultaneously examine all three “sediment qualitytriad” parameters, identified 11 stations (representing 4.4 km2, 0.5% of the total study area) with sediment toxicity, chemical contamination, and altered benthos (i.e., degraded sediment quality), 36 stations (493.5 km2, 57.5% total study area) with no toxicity or chemical contamination (i.e., high sediment quality), 35 stations (274.1 km2, 32.0% total study area) with one impaired sediment triadparameter (i.e., intermediate/high sediment quality), and 18 stations (85.7km2, 10.0% total study area) with two impaired sediment parameters (i.e., intermediate/degraded quality sediments). Generally, upon comparison, the number of stations with degraded sediments based upon the sediment quality triad of data was slightly greater in the central Puget Sound than in the northern and southern Puget Sound study areas, with the percent of the total study area degraded in each region decreasing from central to north to south (2.8, 1.3 and 0.5%, respectively). Overall, the sediments collected in Puget Sound during the combined 1997-1999 surveys were among the least contaminated relative to other marine bays and estuaries studied by NOAA using equivalent methods.     Description: Water tracks are zones of high soil moisture that route water downslope over the ice table in polar environments. We present physical, hydrological, and geochemical evidence collected in Taylor Valley, McMurdo Dry Valleys, Antarctica, which suggests that previously unexplored water tracks are a significant component of this cold desert land system and constitute the major flow path in a cryptic hydrological system. Geological, geochemical, and hydrological analyses show that the water tracks are generated by a combination of infiltration from melting snowpacks, melting of pore ice at the ice table beneath the water tracks, and melting of buried segregation ice formed during winter freezing. The water tracks are enriched in solutes derived from chemical weathering of sediments as well as from dissolution of soil salts. The water tracks empty into ice-covered lakes, such as Lake Hoare, resulting in the interfingering of shallow groundwater solutions and glacier-derived stream water, adding complexity to the geochemical profile. Approximately four orders of magnitude less water is delivered to Lake Hoare by any given water track than is delivered by surface runoff from stream flow; however, the solute delivery to Lake Hoare by water tracks equals or may exceed the mass of solutes delivered from stream flow, making water tracks significant geochemical pathways. Additionally, solute transport is two orders of magnitude faster in water tracks than in adjacent dry or damp soil, making water tracks "salt superhighways" in the Antarctic cold desert. Accordingly, water tracks represent a new geological pathway that distributes water, energy, and nutrients in Antarctic Dry Valley, cold desert, soil ecosystems, providing hydrological and geochemical connectivity at the hillslope scale.

    Description: Several compounds currently in development for the treatment of thrombotic disorders demonstrate high levels of specificity for single targets of the blood coagulation cascade such as factor Xa and thrombin. However, development of a single molecule dual inhibitor against factor Xa and thrombin may expand the efficacy to safety ratio of treatment options for arterial and venous thrombosis. The objective of this study was to determine if simultaneous administration of PD 0313052, a selective Xa inhibitor and argatroban, a direct thrombin inhibitor, would lead to a synergistic antithrombotic effect in a rabbit AV shunt model of thrombosis. Intravenous administration of PD 0313052 alone at doses of 0.1, 0.3, and 1.0 mg/kg/min resulted in thrombus weight (TW) reductions of 11±3, 25±10 and 67±7 % compared to the vehicle group. Argatroban at 1, 3 and 10 mg/kg/min reduced TW 16±13, 47±10 and 75±6 %. When PD 0313052 was administered at 0.1 mg/kg/min in combination with argatroban at 1, 3 or 10 mg/kg/min TW was reduced 50±7, 60±7 and 82±9 %. Likewise, argatroban at 1 mg/kg/min combined with 0.1, 0.3 or 1mg/kg/min of PD 0313052 resulted in TW reductions of 56±9, 60±9 and 84±5 %, respectively. At the lowest combined doses of PD 0313052 and argatroban there was no change in bleeding time relative to the additive fold-increases from each drug alone. The EC50 of intravenously administered PD 0313052 and argatroban was 67±23 and 178±58 ng/ml, respectively. When the drugs were combined the EC50 was reduced to 12±6 ng/ml with the PD 0313052/argatroban combination and to 83±29 ng/ml with the argatroban/PD 0313052 combination. A synergistic effect was also observed in an ex vivo assay of thrombin generation (TG). Predicted additive inhibition of TG based on the individual effects of each compound was −9±7, 9±2 and 29±7 % compared to 10±5, 32±5 and 55±3 % with the 313052/argatroban combination. The predicted effects of the argatroban/PD 0313052 combination was −9±7, 1±7 and 16±9 % compared to the actual inhibition of 5±3, 14±5 and 31±7 %. These results demonstrate a significant synergistic antithrombotic effect by combining low doses of a factor Xa and a thrombin inhibitor and support the hypothesis that development of a single molecule inhibitor against different hemostatic targets may offer greater efficacy in the prevention and treatment of venous and arterial thrombosis.

    Description: Water tracks are zones of high soil moisture that route shallow groundwater down-slope, through the active layer and above the ice table. A water track in Taylor Valley, McMurdo Dry Valleys, was analysed for surface hydrogeological, geochemical, and biological characteristics in order to test the hypothesis that water tracks provide spatial structure to Antarctic soil ecosystems by changing the physical conditions in the soil environment within the water tracks from those outside the water tracks. The presence of the water track significantly affected the distribution of biotic and abiotic ecosystem parameters: increasing soil moisture, soil salinity, and soil organic matter within the water track relative to soils outside the water track, and reducing soil phosphate, soil pH, and the population of nematodes and other invertebrates in water track soils relative to off track soils. These results suggest that water tracks are distinct and extreme ecological zones in Taylor Valley that provide long-range (kilometre to multi- kilometre) structure to Antarctic hillslope ecosystems through physical control on soil moisture and solute content. Contrary to expectations, these high soil-moisture sites are not hotspots for faunal biological activity because high soil salinity makes them suitable habitats for only the most halo-tolerant organisms.

    Description: The aim of the study was to determine if soil salt deliquescence and brine hydration can occur under laboratory conditions using natural McMurdo Dry Valleys soils. The experiment was a laboratory analogue for the formation of isolated patches of hypersaline, damp soil, referred to as ‘wet patches’. Soils were oven dried and then hydrated in one of two humidity chambers: one at 100% relative humidity and the second at 75% relative humidity. Soil hydration is highly variable, and over the course of 20 days of hydration, ranged from increases in water content by mass from 0–16% for 122 soil samples from Taylor Valley. The rate and absolute amount of soil hydration correlates well with the soluble salt content of the soils but not with grain size distribution. This suggests that the formation of bulk pore waters in these soils is a consequence of salt deliquescence and hydration of the brine from atmospheric water vapour.

    Description: Cryoconite holes are near-vertical tubes that form in the surface of glaciers when solar-heated debris melts into the ice. Those that form in the McMurdo Dry Valleys of Antarctica are distinctive, in that they have ice lids and are closed to the atmosphere for periods of years to decades. Photoautotrophs and heterotrophs grow within this closed environment, perturbing the poorly buffered water chemistry, yet maintaining the potential for photosynthesis. Microbial excretion and decomposition of organic matter produces dissolved organic carbon (DOC): dissolved inorganic carbon ratios of ∼1:2. Much of the dissolved nitrogen pool (80-100%) exists as dissolved organic nitrogen (DON). The DON : DOC ratio is ∼1 : 11 (mol/mol), typical of organic particulate material at the Earth's surface. The combination of photoautotrophy, heterotrophy and weak chemical buffering within these microcosms promotes values of pH, pCO2, O2 saturation and percentage total dissolved nitrogen as DON that reach 10·99, 10-7·6 atm, 160% and 100% respectively, which are a unique combination among the surface waters on Earth. These ice-sealed cryoconite holes could be important analogues of refugia on Snowball Earth and other icy planets. © 2004 John Wiley and Sons, Ltd.

    Description: The distribution of streamwater within ice-covered lakes influences sub-ice currents, biological activity and shoreline morphology. Perennially ice-covered lakes in the McMurdo Dry Valleys, Antarctica, provide an excellent natural laboratory to study hydrologic-limnologic interactions under ice cover. For a 2h period on 17 December 2012, we injected a lithium chloride tracer into Andersen Creek, a pro-glacial stream flowing into Lake Hoare. Over 4h, we collected 182 water samples from five stream sites and 15 ice boreholes. Geochemical data showed that interflow travelled West of the stream mouth along the shoreline and did not flow towards the lake interior. The chemistry of water from Andersen Creek was similar to the chemistry of water below shoreline ice. Additional evidence for Westward flow included the morphology of channels on the ice surface, the orientation of ripple marks in lake sediments at the stream mouth and equivalent temperatures between Andersen Creek and water below shoreline ice. Streamwater deflected to the right of the mouth of the stream, in the opposite direction predicted by the Coriolis force. Deflection of interflow was probably caused by the diurnal addition of glacial runoff and stream discharge to the Eastern edge of the lake, which created a strong pressure gradient sloping to the West. This flow directed stream momentum away from the lake interior, minimizing the impact of stream momentum on sub-ice currents. It also transported dissolved nutrients and suspended sediments to the shoreline region instead of the lake interior, potentially affecting biological productivity and bedform development. © 2014 John Wiley & Sons, Ltd.
