    Description: This study presents a novel multi-scale view-planning algorithm for automated targeted inspection using unmanned aircraft systems (UAS). In industrial inspection, it is important to collect the most relevant data to keep processing demands, both human and computational, to a minimum. This study investigates the viability of automated targeted multi-scale image acquisition for Structure from Motion (SfM)-based infrastructure modeling. A traditional view-planning approach for SfM is extended to a multi-scale approach, planning for targeted regions of high, medium, and low priority. The unmanned aerial vehicle (UAV) can traverse the entire aerial space and facilitates collection of an optimized set of views, both close to and far away from areas of interest. The test case for field validation is the Tibble Fork Dam in Utah. Using the targeted multi-scale flight planning, a UAV automatically flies a tiered inspection using less than 25% of the number of photos needed to model the entire dam at high-priority level. This results in approximately 75% reduced flight time and model processing load, while still maintaining high model accuracy where needed. Models display stepped improvement in visual clarity and SfM reconstruction integrity by priority level, with the higher priority regions more accurately modeling smaller and finer features. A resolution map of the final tiered model is included. While this study focuses on multi-scale view planning for optical sensors, the methods potentially extend to other remote sensors, such as aerial LiDAR.

    Description: The quaternary structure and functional properties of synaptophysin, a major integral membrane protein of small presynaptic vesicles, were investigated. Cross-linking and sedimentation studies indicate that synaptophysin is a hexameric homo-oligomer, which in electron micrographs exhibits structural features common to channel-forming proteins. On reconstitution into planar lipid bilayers, purified synaptophysin displays voltage-sensitive channel activity with an average conductance of about 150 picosiemens.     Notes: Zusammenfassung 1. Eine Reihe von Aspergillus niger-Stämmen bildet auf den Nährmedien von Czapek-Dox oder D. Müller Glucoseoxydase, nicht dagegen auf Molliard-Nährlösung. Submerszüchtung liefert im allgemeinen etwas aktiveres Mycelmaterial als Oberflächenkultur. 2. In Extrakten aus glucoseoxydase-haltigem Mycel durchschnittlicher Aktivität (Q O 2 25–150) its die Aktivität von typischen Enzymen des Embden-Meyerhof-Abbaus (Hexokinase, Aldolase, Triosephosphatdehydrogenase) auf einen Bruchteil (2/5 bis 1/6) gegenüber glucosseoxydasefrei gezüchtetem Mycel reduziert. Weniger ausgeprägt ist eine entsprechende Reduktion (etwa 30%) in der Aktivität von typischen Enzymen des Warburg-Dickens-Abbaus (Glucose-6-phosphat-und 6-Phosphogluconat-dehydrogenase). Wird in einer Müller-Nährlösung die Zuckerkonzentration von (normal) 5% auf 30% gesteigert, dann erhöht sich nur die Glucoseoxydase-Aktivität ums Mehrfache, die Aktivität der Enzyme des phosphorylierenden Abbaus fällt durchweg erheblich ab. 3. Während die Aktivität der Glucoseoxydase in Müller-Mycel zwischen 2. und 3. Züchtungstage aufs Mehrfache ansteigt, um dann sehr langsam abzufallen, sinkt die Aktivität der Enzyme des phosphorylierenden Zuckerabbaus schon vom 2. oder 3. Tage an ab und ist am 4. Tage praktisch verschwunden. 4. Während Glucoseoxydase ein ziemlich breites Optimalgebiet zwischen pH 4,5 und 6,5 aufweist und selbst bei pH 3 noch rund 1/3 der optimalen Aktivität zeigt, haben alle untersuchten Enzyme des phosphorylierenden Zuckerabbaus steilere Aktivitäts-pH-Kurven mit ausgeprägten Optima zwischen pH 7,3 und 8,6; nur Triosephosphat-dehydrogenase zeigt bei pH 5 noch merkliche Aktivität. 5. Die unter 2. bis 4. angeführten Befunde machen das Zustandekommen der Gluconsäure-Gärung auf Kosten der phosphorylierenden Abbauformen, insbesondere des sonst überwiegenden Embden-Meyerhof-Abbaus, durch entsprechend „geprägtes” Mycel enzymatisch verständlich.
