    Notes: Experiments were performed in a pilot scale 0.30 m3 conventional stirred-tank fermentor using water, air/water, and air/K2SO4 solutions. Both single- and two-stage impeller systems were investigated. Overall and tank-side coefficients for heat transfer from a 0.012 m diameter coil were measured for a range of impeller speeds and superficial gas velocities. Power input, bubble size, and gas hold-up were also determined. An analysis of the experimental results indicates that previously published correlations for single-phase heat transfer in stirred tanks (of the type: Nu = C(Re)α(Pr)β) are not applicable for single- or multiimpeller gas/liquid systems. The introduction of air alters the mixing pattern significantly, affecting both average and local tank-side heat transfer coefficients. Power input and gas hold-up are suggested as the major correlating parameters for the determination of tank-side heat transfer coefficients.

    Notes: Various aspects of process water recycle in a continuous flow fermentation process are analyzed. Simple mass balance equations in terms of product and feed components for a single-stage reactor producing biomass are developed. Constraints on the recycle ratio, imposed by the efficiency of the dewatering stage, are examined. The recycle analysis is extended using a kinetic growth model incorporating water soluble product formation and growth inhibition. The potential effect of recycle on substrate conversion and product accumulation is also examined and the concept of a critical recycle ratio in fermentation processes is developed.

    Notes: The effect of regularly varying the input nutrient concentration on the growth and composition (elemental and macromolecular) of Escherichia coli ML30 grown in chemostat culture is reported. After each change in substrate concentration a lag between the stimulus and organism response occurred, the magnitude of which was dependent upon the frequency of the perturbations. Both the growth yield and macromolecular composition of the cells were found to vary with the frequency of nutrient cycling: the yield decreased by some 10% as the cycle-times decreased from 6.0 to 0.25 hr. It is suggested that maintaining microorganisms under controlled transient conditions might be useful in increasing yields of specific microbial products (e.g., single-cell protein).

    Notes: The effect of regular variations in the input limiting nutrient concentration at different cycle amplitudes on the growth and composition (elemental and macromolecular) of Escherichia coli ML30 in chemostat culture is described. The cycle time was maintained constant at 2.0 hr. The basal level of nutrient fed to cultures was also maintained constant but the concentration amplitude was varied over a range from 0.2 to 5.0 g/liter. A lag between the stimulus and organisms response was observed and found to be independent of cycle amplitude over most of the range examined. Increasing the cycle amplitude did not effect the cell yield from glucose or the elemental composition of the organisms. However, large variations in the macromolecular composition of the cells (on a dry weight basis) were found; at 4.0 g/liter amplitude the cells contained some 82% protein. The relevance of this type of reactor operation in large-scale fermentations is discussed.

    Notes: The growth of a mixed methane-utilizing culture in a continuous flow fermenter has been studied under both methane and oxygen limitation. Small additions of methanol have been shown to inhibit the methane-utilizing moiety in the culture and it has been shown that the Hyphomicrobium sp. in the mixed culture removes any inhibitory methanol. The interaction between the methane-utilizing Pseudomonas sp., and the Hyphomicrobium sp. has been explained and a model of the continuous mixed culture under oxygen limitation has been formulated. Qualitative predictions of transient phenomena by the model have been verified experimentally.
