    Notes: Summary Maximization of microbial two-step conversion of 9α-fluorohydrocortisone to Δ1-dehydro-16α-hydroxy-9α-fluorohydrocortisone (triamcinolone) in a mixed culture of two microorganisms, Arthrobacter simplex and Streptomyces roseochromogenus, was attempted by a digital simulation, to optimize an operational parameter, pH, during batch cultivation. A mathematical model of the steroid transformation in the mixed culture was constructed with nine differential equations. The kinetic parameters other than the Michaelis-Menten constants in the mathematical model varied with pH of the culture medium. The mathematical model facilitated the simulation of the effect of pH on steroid conversion in the mixed culture. A modified Simplex method of direct search was applied to optimize the switching time and the combination of pH values in the batch culture with several step-changes of pH. Varied-pH processes showed much higher conversion yields than the constant pH process. A process of 3-phase pH change is expected to be advantageous in practical operation because of its lower susceptibility to pH perturbation, though the yield is almost the same as that of the 2-phase process.

    Notes: Summary Two alcohol dehydrogenases (ADHI and ADHII, EC 1.1.1.1) were purified to homogeneity from the cell extract of Zymomonas mobilis. The subunit molecular weights of ADHI and ADHII were 40,000 and 38,000, respectively, and both enzymes were homologous dimers. The optimal pHs of ADHI in ethanol oxidation and acetaldehyde reduction reactions were 9.5 and 4.5, and those of ADHII were 9.5 and 6.5, respectively. The optimal temperatures of ADHI and ADHII were 55° C and 45° C, respectively. ADHI was heat-inactivated at 65° C at a 10-fold higher rate than ADHII. ADHI and ADHII were inhibited by 4 μM and 1 mM p-chloromercuribenzoate, respectively, and the inhibitions were reversed by the addition of 70 mM 2-mercaptoethanol. ADHII activity was enhanced by 0.02 to 2 mM CoCl2 and inhibited by 0.4 mM o-phenanthroline; and the activity of inactivated ADHII was restored by addition of 1 mM CoCl2 or ZnCl2. ADHI was active on most primary alcohols but not secondary alcohols. ADHII was active on only ethanol, n-propanol, allylalcohol, and furfuryl alcohol. In the anaerobic culture of Z. mobilis, ADHII activity accounted for more than 80% of total alcohol dehydrogenase activity. In aerobic culture, ADHII was the main enzyme but was produced only in the early growth phase.

    Notes: Summary Two kinds of bacteria (DC33 and DC1115) were isolated from soil as biotransformers of dehydrocholic acid to 12-ketochenodeoxycholic acid, and identified to be Brevibacterium fuscum and Lactobacillus xylosus, respectively. Dehydrocholic acid was converted via 7,12-diketolithocholic acid to 12-ketochenodeoxycholic acid by both strains, and the product and the intermediate were isolated and chemically identified. By using a jar fermentor, 12-ketochenodeoxycholic acid was produced with a more than 50% yield after 52 h by Brevibacterium fuscum with aerobic growth and anaerobic conversion, and after 24 h by Lactobacillus xylosus under anaerobic conditions, respectively.

    Notes: Summary DL-seleno-methionine resistant mutants of Cephalosporium acremonium were isolated which have an enhanced capacity to utilized sulfate for the synthesis of cephalosporin C. Of these mutants, one designated as SMR-I3 produced three-fold more cephalosporin C from sulfate than its parent CW19. Mutant SMR-I3 required less dl-methionine for maximal synthesis of cephalosporin C, but an excess of dl-methionine inhibited the synthesis of the antibiotic. Furthermore, the mutant accumulated excessive methionine in the amino acid pool and possessed superior activity for sulfate uptake. These observations indicate that in the mutant SMR-I3, the biosynthesis of methionine from sulfate is very active and excess methionine becomes available for the synthesis of cephalosporin C.

    Notes: Summary A fungus identified as Cunninghamella blakesleeana (Lendner) can carry out 15β-hydroxylation of lithocholic acid to a new bile acid (3α,15β-dihydroxy-5β-cholanic acid). By optimizing the fermentation conditions, the amount of the product increased from 0.17 g/l to 1.2 g/l. Hydrophilicity measurements and in vitro cholesterol solubilization tests showed that 3α, 15β-dihydroxy-5β-cholanic acid was as effective as ursodeoxycholic acid in cholesterol solubilization.

    Notes: Summary The genepheA + coding chorismate niutase P-prephenate dehydratase, one of the regulatory enzymes of phenylalanine biosynthesis, was cloned into the down-stream of PR-PL tandem promoter. In this construction, both the native promoter-operator region and the attenuator region ofpheA + operon were eliminated so as to avoid the repression and attenuation ofpheA + gene expression. The expression ofpheA + gene was directed by PR-PL tandem promoter of bacteriophage lambda and controlled by a temperature sensitive repressor, cI857. It was shown that the expression as well as phenylalanine production was regulated by temperature. Maximum production of phenylalanine, 170 mg/l, was obtained at 40°C. The host strain, MC1065, produced a trace (4 mg/l) of phenylalanine at the same temperature.

    Notes: A new method was developed for estimating the volumetric oxygen transfer coefficient, kLa, in a fermentor. Various methods were investigated for the on-line estimation of kLa with an analog computer employing a steepest-descent calculation technique. The method by which kLa is estimated (by minimizing the error residue of the model) was found to be very applicable. A method for the simultaneous estimation of the volumetric oxygen transfer coefficient and respiration rate in biological systems is also presented.

    Notes: Power requirements in the agitation of non-Newtonian fermentation broths with and without aeration were measured by a strain gage-type dynamometer. Broth from the production of gluc-amylase by Endomyces species and carboxymethyl cellulose solutions were used as non-Newtonian fluids. In gas-liquid agitation systems, the correlation between Pg and P02 ND3/Q0.56 observed by Michel and Miller was found to be applicable to non-Newtonian fluids in laminar and transition regions. This was particularly true for fluids with apparent viscosities larger than 300 cp. The impeller diameter and impeller blade width had considerable effects on power consumption in a nongassed system. It was suggested, therefore, that Pg/P0 should be correlated by a dimensionless term involving some impeller-size factors.

    Notes: A modified simulation procedure based on a statistical approach was investigated. The procedure predicts the time course of fed-batch culture for glutamic acid production by a temperature-sensitive strain of Brevibacterium flavum. The statistical approach requires only a data base of state points obtained in experiments, and not perfect identification of fermentation models. The simulation procedure is based on regression analysis to estimate specific rate parameters of system equations using the data points selected with reference to the Euclid distance. It was modified in that the data selection procedure included the use of the Maharanobis distance as well as a modified distance defined in this study. Simulation results using the modified procedure allow reasonable prediction of the time course of fed-batch culture for glutamic acid compared to that involving the Euclid distance alone.

    Notes: In many cases of fermentation processes, it is difficult to measure all of the state variables needed to indicate the culture state and to predict state changes. We propose the use of time-series data of measurable state variables instead of unmeasurable to describe the fermenatation process. We also employ a discrimination method of independent variables of regression analysis using statistical measures, PSS and AIC. These statistical procedure were applied to fed batch culture for yeast production under aerobic conditions, and the effectiveness was ascertained experimentally.
